With the retreat of the flood 'Whedos' usually become infested with numerous hydato-and tenagophytes, while the presence and density of the free-floating macrophytes were positively related to the nutrient content of the 'Whedo'. Extensive plant infestation also affects water quality through the decomposition of organic material and its accumulation in thick mud layers on the pond bottom as well as through the nocturnal oxygen consumption. Unfavourable water quality, especially low dissolved oxygen as well as high conductivity and nitrite levels, was identified to be the main factor determining which fish species were able to survive the harsh conditions prevailing in the 'Whedos' during the dry season. With the deteriorating water quality with advancing dry season, fish diversity decreased significantly leaving only species that are highly adapted to such unfavourable conditions. The most abundant species were Clarias gariepinus, Heterotis niloticus, Oreochromis niloticus L., Hemichromis c.f. letourneauxi, Polypterus senegalus and Epiplatys spilargyreius. Besides, the investigations also concentrated on the fish diversity of the rivers Niger and Sota with the results that for three species distribution gaps could be closed and for further three species their already known distribution could be expanded. But otherwise it could also be detected that some economically important species that were abundant in the past. In regard to the 'Whedo'-management, the investigations showed that the owners lack most of the knowledge on appropriate management strategies, e.g. the feeding and stocking regime. The exploitation period depends on the extent of the previous annual flood and the location of the 'Whedo' within the floodplain, but the main season is from February to April. The biomass harvested on a hectare basis separated for each of the 'Whedos' averaged 17 tons/ha in 2008 and 8.6 tons/ha in 2009. However, 72 percent of the total biomass of Clarias only had an average weight of 40 grams. Therefore, two separated feeding trials were conducted and in total 6 supplementary feeds were tested on Clarias gariepinus. Groundnut cake, fish trash, rice bran, blood meal and azolla meal were used in different combination and rations to formulate the experimental diets. Diet containing 19 percent blood meal resulted in the best economical benefits showing that the use of high quality feed ingredients such as groundnut cake is not recommendable because local fish prices are too low to compensate the additional feeding costs. Instead of high quality feed farmers should focus on ingredients that are free of charge and easy to process. The supplementation based on 19 percent blood meal resulted in the doubling of the net profit compared to the income based on feeding only rice bran, thus provided higher additional income, enhancing the livelihood of the fish farmers. Concluding, the 'Whedo'-aquaculture system is still in its infancy but nevertheless is an attractive system for the rural population because of existing knowledge of post-flood wetland fisheries as well as the low investment needed for its installation. Additionally, the local fish supply will increase and hence not only contribute to a better provision of protein-rich food and reduced pressure on the wild fish stocks but might also prevent fish prices to increase in a way that the poor won't be able anymore to afford their most important source of animal protein. But fish farmers need more knowledge on appropriate management strategies and thus should be provided with technical support to guarantee a successful development and not to discourage the owners as a consequence of avoidable failures. A C-terminal modified FYVE zinc finger motif is necessary to target Spir proteins towards intracellular membranes. The function and regulation of the Spir actin organizers at vesicular membranes is almost unknown. Live cell imaging analyses performed in this study show that Spir-2 is localized at tubular vesicles. Cytoplasmic Spir-2-associated vesicles branch and form protrusions, which can make contacts to the microtubule network, where the Spir-2 vesicles stretch and slide along the microtubule filaments. The analysis of living HeLa cells expressing eGFP-tagged Spir-2, Spir-2-ΔKIND and Spir-2-ΔKW (lacking the 4 WH2 domains and the KIND domain) showed Spir-2-associated tubular structures which differ in their length and motility. Throughout the course of that study it could be shown that the tail domain of the actin motor protein myosin Vb, as a force-generating molecule, is colocalizing and co-immunoprecipitating with Spir-2-ΔKW. By using the tail domain of myosin Vb as a dominant negative mutant for myosin Vb-dependent vesicle transport processes it could be shown that Spir-2-ΔKW/MyoVb-cc-tail- associated vesicles exhibit an increased elongation. Moreover, using the microtubule depolymerizing drug nocodazole it could be shown that the elongation and the motility of Spir-2-ΔKW-associated vesicles depends on an intact microtubule cytoskeleton. Motility and morphological dynamics of Spir-2-associated vesicles is therefore dependent on actin, actin motorproteins and microtubule filaments. These results propose a model in which myosin/F-actin forces mediate vesicle branching, allowing the vesicles to move to and in between the microtubule filaments and thereby providing a new degree of freedom in vesicular motility. To determine the exact subcellular localization of Spir-2, colocalization studies were performed. It could be shown that Spir-2 shows a partial colocalization to Rab11a-positive compartments. Furthermore, Spir-2 exhibits an almost identical localization to Arf1 and the Arf1 small G protein but not Rab11a could be immunoprecipitated with Spir-2-ΔKW. This suggests, that Arf1 recruits Spir-2 to Arf1/Rab11a-positive membranes. Another important function of the Spir-2 C-terminus is the membrane targeting by the FYVE domain. By performing a protein-lipid overlay assay, it has been shown that purified GST- and 6xHis-tagged Spir-2-ΔKW bind phosphatidic acid suggesting a mechanism in which Spir-2 is recruited to phosphatidic acid-enriched membranes. Its native range is unknown, and there is little information concerning its social structure as a potential mechanism facilitating invasion as well as its ecology in one of the putative native ranges, South-East Asia. Using mitochondrial DNA sequences, I demonstrated that the majority of the current Indopacific colonies were likely introduced from South-East Asian populations, which in turn may have been introduced much earlier from a yet unidentified native range. By conducting behavioral, genetic and chemical analyses, I found that A. gracilipes supercolonies contain closely related individuals, thus resembling enlarged versions of monogynous, polydomous colonies of other ant species. Furthermore, mutually aggressive A. gracilipes supercolonies were highly differentiated both genetically and chemically, suggesting limited or even absent gene flow between supercolonies. Intranidal mating and colony-budding are most likely the predominant, if not the exclusive mode of reproduction and dispersal strategy of A. gracilipes. Consequently, a positive feedback between genetic, chemical and behavioral traits may further enhance supercolony differentiation though genetic drift and neutral evolution. This potential scenario led to the hypothesis that absent gene flow between different A. gracilipes supercolonies may drive them towards different evolutionary pathways, possibly including speciation. Thus, I examined one potential way by which gene flow between supercolonies of an ant species without nuptial flights may be maintained, i.e. the immigration of sexuals into foreign supercolonies. The results suggest that this option of maintaining gene flow between different supercolonies is likely impaired by severe aggression of workers towards allocolonial sexuals. Moreover, breeding experiments involving males and queens from different supercolonies suggest that A. gracilipes supercolonies may already be on the verge of reproductive isolation, which might lead to the diversification of A. gracilipes into different species. Regarding the ecological consequences of its potential introduction to NE-Borneo, I could show that A. gracilipes supercolonies may affect the local ant fauna. The ant community within supercolonies was less diverse and differed in species composition from areas outside supercolonies. My data suggest that the ecological dominance of A. gracilipes within local ant communities was facilitated by monopolization of food sources within its supercolony territory, achieved by a combination of rapid recruitment, numerical dominance and pronounced interspecific aggression. A. gracilipes' distribution is almost exclusively limited to anthropogenically altered habitat, such as residential and agricultural areas. The rate at which habitat conversion takes place in NE-Borneo will provide A. gracilipes with a rapidly increasing abundance of suitable habitats, thus potentially entailing significant population growth. An potentially increasing population size and ecological dominance, however, are not features that are limited to invasive alien species, but may also occur in native species that become 'pests' in an increasing abundance of anthropogenically altered habitat. Lastly, I detected several ant guests in supercolonies of A. gracilipes. I subsequently describe the relationship between one of them (the cricket Myrmecophilus pallidithorax) and its ant host. By conducting behavioral bioassays and analyses of cuticular hydrocarbon (CHC) profiles, I revealed that although M. pallidithorax is attacked and consumed by A. gracilipes whenever possible, it may evade aggression from its host by a combination of supreme agility and, possibly, chemical deception. This thesis adds to our general understanding of biological invasions by contributing species-specific data on a previously understudied invasive organism, the Yellow Crazy Ant Anoplolepis gracilipes. Introductions which may have occurred a long time ago may make it difficult to determine whether a given species is an introduced invader or a native pest species, as both may have pronounced ecological effects in native species communities. Because of its huge socioeconomic burden absorbing 6\% of all health care budgets and with the fact that life expectancy increases globally, one can assume that stroke is already, and will continue to be, the most challenging disease. Ischemic stroke accounts for approximately 80\% of all strokes and results from a thrombotic or embolic occlusion of a major cerebral artery (most often the middle cerebral artery, MCA) or its branches Following acute ischemic stroke, the most worrisome outcome is the rapidly increasing intra-cranial pressure due to the formation of space-occupying vasogenic oedema which can have lethal consequences. Permeability changes at the Blood-Brain Barrier (BBB) usually accompanies the oedematous development and their time course can provide invaluable insight into the nature of the insult, activation of compensatory mechanisms followed by long term repair. Rodent models of focal cerebral ischemia have been developed and optimized to mimic human stroke conditions and serve as indispensable tools in the field of stroke research. The presented work constituting of three separate but complete works by themselves are sequential, where, the first part was dedicated to the establishment of non-invasive small animal imaging strategies on a 3 tesla clinical magnetic resonance scanner. This facilitated the longitudinal monitoring of pathological outcomes following stroke where identical animals can serve as its own control. Tissue relaxometric estimations were carried out initially to derive the transverse (T2), longitudinal (T1) and the transverse relaxation time due to magnetic susceptibility effects (T2*) at the cortical and striatal regions of the rodent brain. Statistically significant differences in T2*-values could be found between the cortex and striatal regions of the rodent brain. The derived tissue relaxation values were considered to modify the existing imaging protocols to facilitate the study of the rodent model of ischemic stroke. The modified sequence protocols adequately characterized all the clinically relevant sequels following acute ischemic stroke, like, the altered perfusion and diffusion characteristics. Subsequent to this, serial magnetic resonance imaging was performed to investigate the temporal and spatial relationship between the biphasic nature of BBB opening and, in parallel, the oedema formation after I/R injury in rats. T2-relaxometry for oedema assessment was performed at 1 h after ischemia, immediately following reperfusion, and at 4, 24 and 48 hours post reperfusion. Post-contrast T1-weighted imaging was performed at the last three time points to assess BBB integrity. The biphasic course of BBB opening with significant reduction in BBB permeability at 24 hours after reperfusion was associated with a progressive expansion of leaky BBB volume, accompanied by a peak ipsilateral oedema formation. At 48 hours, the reduction in T2-value indicated oedema resorption accompanied by a second phase of BBB opening. In addition, at 4 hours after reperfusion, oedema formation could also be detected at the contralateral striatum which persisted to varying degrees throughout the study, indicative of widespread effects of I/R injury. The observations of this study may indicate a dynamic temporal shift in the mechanisms responsible for biphasic BBB permeability changes, with non-linear relations to oedema formation. Two growth factor peptides namely pigment epithelium derived factor (PEDF) and epidermal growth factor (EGF) with widely different trophic properties were considered for their beneficial effects, if any, in the established rodent model of I/R injury and studied up to one week employing magnetic resonance imaging. Both the selected, trophic factors demonstrated significant neuroprotection as demonstrated by a reduction in infarct volume, even though PEDF was found to be the most potent one. PEDF also demonstrated significant attenuation of oedema formation in comparison to both the control and EGF groups, even though EGF could also demonstrate oedema suppression. In the present work, we noticed that interventions with macromolecule protein/peptides by itself could mediate remote oedema at distant sites even though the significance of such an observation is not clear at present. Susceptibility (T2*) weighted tissue relaxometric estimations were considered at the infarct region to detect any metabolic changes arising out of any neuroprotection and/or cellular proliferation / neurogenesis. PEDF group demonstrated a striking reduction of the T2*-values, which is indicative of an increased metabolic activity. Moreover, all the groups (Control, EGF and PEDF) demonstrated significantly elevated T2*-values at the contralateral striatum, which is indicative of widespread metabolic suppression usually associated with a variety of traumatic brain conditions. Moreover, as expected from the properties of PEDF, it demonstrated an extended BBB permeability suppression throughout the duration of the study. This study underlines the merits of considering non-invasive imaging strategies without which it was not possible to study the required parameters in a longitudinal fashion. All the observations are adequately supported by reasonably well defined mechanisms and needs to be further verified and confirmed by an immunohistochemical study. These results also need to be complemented by a functional study to evaluate the behavioural outcome of animals following these treatments. NFATc1 nimmt innerhalb dieser Familie eine Sonderrolle ein, da dessen Aktivit{\"a}t auch durch eine stark induzierbare Expression gesteigert werden kann. Dies ist insbesondere f{\"u}r die Differenzierung und Funktion von T- und B-Lymphozyten von Bedeutung. Weiterhin ist NFATc1 f{\"u}r die Muskel- oder Herzentwicklung notwendig. Eine Reihe von Arbeiten belegen dar{\"u}ber hinaus eine Beteiligung dieses Transkriptionsfaktors an der Entstehung von Leuk{\"a}mien und Lymphomen. W{\"a}hrend klassische Hodgkin-Lymphome allerdings durch eine abgeschaltete NFATc1-Expression gekennzeichnet sind, wird f{\"u}r T-ALL (Akute Lymphatische Leuk{\"a}mie der T-Zelle) eine {\"U}berexpression beschrieben. Inwiefern die Calcineurin-abh{\"a}ngige Kerntranslokalisation den einzigen Aktivierungsmechanismus f{\"u}r NFAT-Faktoren darstellt, ist noch nicht eindeutig gekl{\"a}rt. Nach optimaler Aktivierung von T- bzw. B-Zellen ist die kurze, induzierbare Isoform NFATc1/A das Hauptprodukt des NFATc1-Gens. In dieser Arbeit wurden f{\"u}r die gezielte Deletion des NFATc1-Gens in der Maus zwei verschiedene konditionelle Systeme verwandt. Hierzu wurden Tiere, die ein mit „flox"-Sequenzen versehenes drittes Exon des NFATc1-Gens in der Keimbahn tragen, mit verschiedenen Cre-Rekombinase expremierenden Linien verkreuzt. Der Verlust funktionellen NFATc1-Proteins erfolgt dann fr{\"u}h in der B-Zell-Differenzierung im Knochenmark (Cd79a/mb-1-cre x Nfatc1flx/fl) bzw. in reifen B-Zellen (Cd23-cre x Nfatc1flx/flx). W{\"a}hrend in keiner dieser Linien signifikante Defekte in der Differenzierung "konventioneller B2" B-Lymphozyten beobachtet wurden, hatte die fr{\"u}he Inaktivierung des NFATc1-Gens im Knochenmark den Verlust der B1a-Zell-Population im Peritoneum zur Folge. In vitro zeigten NFATc1-/--B-Zellen aus der Milz nach Aktivierung {\"u}ber den B-Zell-Rezeptor deutliche Defekte in der Zellteilung bei einer gleichzeitigen Zunahme des aktivierungsinduzierten Zelltodes (AICD, activation induced cell death). Die vergleichende Transkriptomanalyse identifizierte wichtige Gene des Ca++/Calcineurin-Signalweges als NFATc1-Zielgene und mitverantwortlich f{\"u}r die Proliferationsdefekte. In NFATc1-defizienten B-Zellen konnte Re-Expression von NFATc1 in geringer Konzentration den aktivierten Zelltod inhibieren, wohingegen hohe Konzentrationen diesen noch weiter f{\"o}rderten. Zusammengenommen l{\"a}sst sich daher schließen, dass NFATc1 entscheidend an der Kontrolle von Proliferation und Zelltod peripherer B-Lymphozyten beteiligt ist. Eine weitere wichtige Funktion kommt NFATc1 beim Klassenwechsel im Immunglobulin-Lokus zu. In den untersuchten M{\"a}usen war die IgG3-Produktion nach Immunisierung mit NP-Ficoll (einem T-Zell-unabh{\"a}ngigen Antigen des Typs II) deutlich reduziert, wenn das NFATc1-Gen in den B-Lymphozyten funktionslos war. Auch die Bildung von IgG3+-Plasmablasten war gehemmt. Zu {\"a}hnlichen Ergebnissen f{\"u}hrten Untersuchungen an isolierten B-Lymphozyten in einem in vitro Klassenwechsel-Modell. Demgegen{\"u}ber zeigten Immunisierungen der Tiere mit NP-KLH (einem T-Zell-abh{\"a}ngigen Antigen) keine signifikanten Abweichungen im Klassenwechsel. Furthermore, the trap states in the blend were compared with these of the pure materials. Concerning the lifetime of organic solar cells the influence of oxygen on P3HT and P3HT:PC61BM blends was studied. The experimental techniques used to investigate the trap states in the organic semiconductors were (fractional) thermally stimulated current (TSC) and current based deep level transient spectroscopy (Q-DLTS). Fractional TSC measurements on P3HT diodes revealed a quasi-continuous trap distribution. The distribution suggested two different traps in P3HT with approximately Gaussian energy distributions and maxima at about 50 meV and 105 meV. Thereby, the former was attributed to the tail states within the regular Gaussian density of states due to the low activation energy. The latter, deeper traps, however, exhibited a strong dependence on oxygen. Exposure of the P3HT diodes to oxygen, ambient air and synthetic (dry) air all revealed an increase of the deeper traps density with exposure time in the same manner. While the lower limit of the trap density in non aged P3HT samples was in the range of (1.0 - 1.2)×10^22 m^-3, it was more than doubled after an exposure of 50 h to air. An increase of the trap density with oxygen exposure time was also seen in the Q-DLTS measurements accompanied with an increase of the temperature dependence of the emission rates, indicating an enhanced formation of deeper traps. Due to the raise in density of the deeper traps, the charge carrier mobility in P3HT significantly decreased, as revealed by photo-CELIV measurements, resulting in a loss in mobility of about two orders of magnitude after 100 h exposure to synthetic air. The increased trap density was attributed to p-doping of P3HT by the transfer of an electron to adsorbed oxygen. This effect was partially reversible by applying vacuum to the sample for several hours or, more significantly, by a thermal treatment of the devices in nitrogen atmosphere. The trap states in the methanofullerenes PC61BM, bisPC61BM and PC71BM were investigated by TSC measurements. PC61BM yielded a broad quasi-continuous trap distribution with the maximum of the distribution at about 75 meV. The comparison of the TSC spectra of the three methanofullerenes exhibited significant differences in the trap states with higher activation energies of the most prominent traps in bisPC61BM and PC71BM compared to PC61BM. This probably originates from the different isomers bisPC61BM and PC71BM consist of. Each of the isomers yields different LUMO energies, where the lower ones can act as traps. The lower limit of the trap density of all of the three investigated fullerene derivatives exhibited values in the order of 10^22 m^-3, with the highest for bisPC61BM and the lowest for PC61BM. By applying fractional TSC measurements on P3HT:PC61BM solar cells, it was shown that the trap distribution in the blend is a superposition of the traps in pure P3HT and PC61BM and additional deeper traps in the range of about 250 meV to 400 meV. The origin of these additional traps, which can not be related to the pure materials, was attributed to a higher disorder in the blend and P3HT/PC61BM interfaces. This conclusion was supported by standard TSC and Q-DLTS measurements performed on pristine and annealed P3HT:PC61BM blends, exhibiting a higher ratio of the deep traps in the pristine samples. The lower limit of the trap density of the investigated annealed solar cells was in the range of (6-8)×10^22 m^-3, which was considerably higher than in the pure materials. The influence of oxygen on P3HT:PC61BM solar cells was investigated by exposure of the devices to synthetic air under specific conditions. Exposure of the solar cells to oxygen in the dark resulted in a strong decrease in the power conversion efficiency of 60 \% within 120 h, which was only caused by a loss in short-circuit current. Simultaneous illumination of the solar cells during oxygen exposure strongly accelerated the degradation, resulting in an efficiency loss of 30 \% within only 3 h. Thereby, short-circuit current, open-circuit voltage and fill factor all decreased in the same manner. TSC measurements revealed an increase of the density of deeper traps for both degradation conditions, which resulted in a decrease of the mobility, as investigated by CELIV measurements. However, these effects were less pronounced than in pure P3HT. Absch{\"a}tzungen {\"u}ber die Art und St{\"a}rke des Klimawandels auf gr{\"o}ßeren, planungsrelevanten Massst{\"a}ben sind jedoch nach wie vor mit großen Unsicherheiten verbunden. F{\"u}r eine detailliertere Erforschung der Auswirkungen der globalen Erw{\"a}rmung auf regionaler oder gar lokaler Ebene besteht daher noch großer Forschungsbedarf. In dieser Dissertation wird zu diesem Zweck ein statistischer Ansatz verfolgt. Dieser erlaubt die Identifikation systematischer Unterschiede zwischen den Auspr{\"a}gungen klimatologischer Feldgr{\"o}ßen (bodennahe Lufttemperatur und Niederschlag) wie sie von sogenannten General Circulation Models (GCMs) simuliert werden im Vergleich zu den betreffenden Parametern aus Beobachtungsdaten. Als Beobachtungsdaten werden die NCEP Reanalysen, die statistisch interpolierten Datens{\"a}tze der CRU sowie Stationsdaten aus Vietnam verwendet. Hierbei zeigt sich, dass die aktuellen Klimamodelle die r{\"a}umlichen Muster der betrachteten Variablen in befriedigender Weise reproduzieren. Die Analyse des regionalen Klimawandels in S{\"u}dost-Asien erfolgt durch die Auswertung von Klimamodellrechnungen. Diese wurden von verschiedenen GCMs durchgef{\"u}hrt, wobei unterschiedliche Annahmen {\"u}ber die zuk{\"u}nftigen Treibhausgasemissionen ber{\"u}cksichtigt wurden. Der Fokus dieser Dissertation ist die Analyse der projizierten zeitlichen Entwicklung von bodennaher Temperatur und Niederschlag im 21. Jahrhundert. Hierbei werden sowohl j{\"a}hrliche als auch saisonale Mittelwerte bzw. Summen ber{\"u}cksichtigt. Neben diesen rein physikalisch-klimatologischen Betrachtungen behandelt diese Dissertation auch einen angewandten Aspekt, n{\"a}mlich den Impakt des Klimawandels auf die Landwirtschaft, exemplarisch untersucht am Beispiel Vietnams. F{\"u}r die Absch{\"a}tzung der Vulnerabilit{\"a}t dieses essentiellen Wirtschaftsbereiches wird ein statistisches Modell entwickelt in das an klimatischen Parametern die bodennahe temperatur sowie der Niederschlag einfliessen. Diese Untersuchung leistet damit einen wichtigen Beitrag zum Wissenstand {\"u}ber die Auswirkungen des Klimawandels in den niederen Breiten. Die sozio-{\"o}konomische Entwicklung jedes Staates der Erde wird von den Folgen des Klimawandels beeinflusst, allerdings variiert der Grad der Beeintr{\"a}chtigung erheblich. Vermutlich werden Entwicklungsl{\"a}nder wie Vietnam die Auswirkungen des Klimawandels besonders stark zu sp{\"u}ren bekommen. Die Ursachen f{\"u}r diese hohe Vulnerabilit{\"a}t liegen unter anderem in der Wirtschaftsstruktur: der allgemein hohe Stellenwert nat{\"u}rlicher Ressourcen und eine geringe Diversit{\"a}t verringern hier die M{\"o}glichkeiten zur Adaption an die beobachteten und projizierten Ver{\"a}nderungen. Die vorliegende Dissertation gliedert sich wie folgt: In Kapitel 1 stellt eine allgemeine Einf{\"u}hrung zur Thematik dar. The optical, electrochemical and spectroelectrochemical properties of these hybrid materials are furthermore studied. The work therefore is divided into two parts. The first part deals with triarylamine and PCTM-radical functionalized gold nanoparticles. The focus thereby was on the synthesis and on the investigations of chromophore-chromophore interactions and gold core-chromophore interactions. The chromopores, especially triarylamines, were attached to the gold core via different bridging units and were studied with optical and electrochemical methods. The purity and dimensions of the nanoparticles was determined by 1H-NMR spectroscopy, diffusion ordered NMR spectroscopy (DOSY), TGA, XPS and STEM. Furthermore a cyclic voltammetry technique was used to determine the composition of the particles via the Randles-Sevcik equation. An analysis of these parameters led to a model of a sea urchin-shaped nanoparticle. Optical measurements of the particles revealed an anisotropic absorption behavior of the triarylamine units due to gold core-chromophore interaction. However this behavior depends strongly on the relative orientation of the transition dipole moment of the chromophore to the gold surface and the distance of the chromophore to the surface. Hence, the anisotropic behavior was exclusively detected in the spectra of the Au-Tara1 particles. The short and rigid pi-conjugated bridging unit thereby facilitates this gold core-chromophore interaction. It was shown from electrochemical investigations that the triarylamine units can be chemically reversibly oxidized to the triarylamine monoradical cation. Furthermore, the measurements revealed a strong interligand triarylamine-triarylamine interaction which was only seen for the Au-Tara1 particles. The long pi-conjugated bridging units of the Au-Tara2 and Au-Tara3 particles as well as the aliphatic bridging unit of Au-Tara4 prevent any detectable interligand interactions. One may conclude that both the gold core-chromophore and the interligand triarylamine-triarylamine interaction depend on the length and the rigidity of the bridging unit. The electron transfer behavior of the triarylamine units adsorbed onto the gold core was additionally studied via spectroelectrochemical (SEC) measurements which are able to reveal weaker interactions. The investigations of Au-Tara1 and Au-Tara2 revealed a significant strong coupling between neighboring triarylamine units which is due to through-space intervalence interactions. This behavior was not detected for Au-Tara3 or for Au-Tara4. The SEC analysis also revealed that these observed interligand interactions depend on the length and the rigidity of the bridging unit. Thus, the systematic variation of the bridging unit gave a basic insight in the optical and electrochemical properties of triarylamines, located in the vicinity of a gold nanoparticle. The second part of this work aimed at the synthesis of new molecules, denoted as SERS-markers, for immuno SERS applications. For this purpose, the SERS-markers were designed to have a Raman-active unit and a thiol group for chemisorptions to Au/Ag nanoshells. In cooperation with the group of Schl{\"u}cker (University of Osnabr{\"u}ck) the SERS-markers were absorbed onto Au/Ag nanoshells, denoted as SERS-labels, and characterized. The SERS spectra of the SERS-labels exhibited intense and characteristic SERS-signals for each marker. For immuno SERS investigations SEMA3 was functionalized with a hydrophilic end unit. This marker was adsorbed onto an Au/Ag nanoshell and encapsulated with silica. An anti-p63 antibody was bound to the silica surface in order to generate a SERS-labeled antibody for the detection of the tumor suppressor p63 in benign prostate. Immuno-SERS imaging of prostate tissue incubated with SERS-labeled anti-p63 antibodies demonstrated the selective detection of p63 in the basal epithelium. It is known to act as a potent hepatotoxin and liver carcinogen in rodents. In a 2-year bioassay, chronic furan administration to rats was shown to cause hepatocellular adenomas and carcinomas and very high incidences of cholangiocarcinomas even at the lowest furan dose tested (2.0 mg/kg bw). However, the mechanisms of furan-induced tumor formation are poorly understood. Furan is metabolized by cytochrome P450 (CYP) enzymes, predominantly CYP2E1, to its major metabolite cis-2-butene-1,4-dial (BDA). BDA is thought to be the key mediator of furan toxicity and carcinogenicity and was shown to react with cellular nucleophiles such as nucleosides and amino acid residues in vitro. It is well known that covalent protein binding may lead to cytotoxicity, but the cellular mechanisms involved remain to be elucidated. Since covalent binding of reactive intermediates to a target protein may result in loss of protein function and subsequent damage to the cell, the aim of this study was to identify furan target proteins to establish their role in the pathogenesis of furan-associated liver toxicity and carcinogenicity. In order to identify target proteins of furan reactive metabolites, male F344/N rats were administered [3,4-14C]-furan. Liquid scintillation counting of protein extracts revealed a dose-dependent increase of radioactivity covalently bound to liver proteins. After separation of the liver protein extracts by two-dimensional gel electrophoresis and subsequent detection of radioactive spots by fluorography, target proteins of reactive furan intermediates were identified by mass spectrometry and database search via Mascot. A total of 61 putative target proteins were consistently found to be adducted in 3 furan-treated rats. The identified proteins represent - among others - enzymes, transport proteins, structural proteins and chaperones. Pathway mapping tools revealed that target proteins are predominantly located in the cytosol and mitochondria and participate in glucose metabolism, mitochondrial β-oxidation of fatty acids, and amino acid degradation. These findings together with the fact that ATP synthase β subunit was also identified as a putative target protein strongly suggest that binding of furan reactive metabolites to proteins may result in mitochondrial injury, impaired cellular energy production, and altered redox state, which may contribute to cell death. Moreover, several proteins involved in the regulation of redox homeostasis represent putative furan target proteins. Loss of function of these proteins by covalent binding of furan reactive metabolites may impair cellular defense mechanisms against oxidative stress, which may also result in cell death. Besides the potential malfunction of whole pathways due to loss of functions of several participating proteins, loss of function of individual proteins which are involved in various cellular processes such as transport processes across the mitochondrial membranes, cell signaling, DNA methylation, blood coagulation, and bile acid transport may also contribute to furan-induced cytotoxicity and carcinogenicity. Covalent binding of reactive metabolites to cellular proteins may result in accumulation of high amounts of unfolded or damaged proteins in the endoplasmic reticulum (ER). In response to this ER stress, the cell can activate the unfolded protein response (UPR) to repair or degrade damaged proteins. To address whether binding of furan reactive metabolites to cellular proteins triggers activation of the UPR, semiquantitative PCR and TaqMan® real-time PCR were performed. In the case of UPR activation, semiquantitative PCR should show enhanced splicing of X-box binding protein-1 (XBP1) mRNA (transcription factor and key regulator of the UPR) and TaqMan® real-time PCR should determine an increased expression of UPR target genes. However, our data showed no evidence for activation of the UPR in the livers of rats treated either with a single hepatotoxic dose or with a known carcinogenic dose for 4 weeks. This suggests either that furan administration does not induce ER stress through accumulation of damaged proteins or that activation of the UPR is disrupted. Consistent with the latter, glucose-regulated protein 78 (GRP78), identified as a target protein in our study, represents an important mediator involved in activation of the UPR whose inhibition was shown to impair induction of the UPR. Thus, adduct formation and inactivation of GRP78 by furan metabolites may disturb activation of the UPR. In addition to impaired activation of UPR, protein repair and degradation functions may be altered, because several proteins involved in these processes also represent target proteins of furan and thus may show impaired functionality. Although much attention has been devoted to the identification of phosphorylation sites in murine BAD (mBAD), little data are available with respect to phosphorylation of human BAD (hBAD) protein. In this work, we investigated the quantitative contribution of BAD targeting kinases in phosphorylating serines 75, 99 and 118 of hBAD (Chapter 3.1). Our results indicate that RAF kinases phosphorylate hBAD in vivo at these established serine residues. RAF-induced phosphorylation of hBAD was not prevented by MEK inhibitors but could be reduced to control levels by use of the RAF inhibitor Sorafenib (BAY 43-9006). Consistently, expression of active RAF suppressed apoptosis induced by hBAD and the inhibition of colony formation caused by hBAD could be prevented by RAF. In addition, using surface plasmon resonance technique we analyzed the direct consequences of hBAD phosphorylation by RAF with respect to complex formation of BAD with 14-3-3 proteins and Bcl-XL. Phosphorylation of hBAD by active RAF promotes 14-3-3 protein association, whereby the phosphoserine 99 represents the major binding site. Furthermore, we demonstrate in this work that hBAD forms channels in planar bilayer membranes in vitro. This pore-forming capacity is dependent on phosphorylation status and interaction with 14-3-3 proteins. Additionally, we show that hBAD pores possess a funnel-shaped geometry that can be entered by ions and non-charged molecules up to 200 Da (Chapter 3.2). Since both lipid binding domains of hBAD (LBD1 and LBD2) are located within the C-terminal region, we investigated this part of the protein with respect to its structural properties (Chapter 3.3). Our results demonstrate that the C-terminus of hBAD possesses an ordered β-sheet structure in aqueous solution that adopts helical disposition upon interaction with lipid membranes. Additionally, we show that the interaction of the C-terminal segment of hBAD with the BH3 domain results in the formation of permanently open pores, whereby the phosphorylation of serine 118 proved to be necessary for effective pore-formation. In contrast, phosphorylation of serine 99 in combination with 14-3-3 association suppresses formation of channels. These results indicate that the C-terminal part of hBAD controls hBAD function by structural transitions, lipid binding and phosphorylation. Using mass spectrometry we identified in this work, besides the established in vivo phosphorylation sites at serines 75, 99 and 118, several novel hBAD phosphorylation sites (serines 25, 32/34, 97, 124 and 134, Chapter 3.1). To further analyze the regulation of hBAD function, we investigated the role of these newly identified phosphorylation sites on BAD-mediated apoptosis. We found that in contrast to the N-terminal phosphorylation sites, the C-terminal serines 124 and 134 act in an anti-apoptotic manner (Chapter 3.4). Our results further indicate that RAF kinases and PAK1 effectively phosphorylate BAD at serine 134. Notably, in the presence of wild type hBAD, co-expression of survival kinases, such as RAF and PAK1, leads to a strongly increased proliferation, whereas substitution of serine 134 by alanine abolishes this process. Furthermore, we identified hBAD serine 134 to be strongly involved in survival signaling in B-RAF-V600E containing tumor cells and found phosphorylation of this residue to be crucial for efficient proliferation in these cells.
