We can choose between a variety of possibilities, from simply pointing at the correct object as we say its name to lengthy definitions in encyclopaedias, which can sometimes fill multiple pages. Although the former approach is pretty straightforward and is also very important for first language acquisition, it is obviously not a practical solution for defining the semantics of the whole lexicon. The latter approach is more widely accepted in this context, but it turns out that defining dictionary and encyclopaedia entries is not an easy task. In order to simplify the challenge of defining the meaning of words, it is of great advantage to organize the lexicon in a way that the structure in which the words are integrated gives us information about the meaning of the words by showing their relation to other words. These semantic relations are the focal point of this paper. In the first chapter, different ways to describe meaning will be discussed. It will become obvious why semantic relations are a very good instrument to organizing the lexicon. The second chapter deals with WordNet, an electronic lexical database which follows precisely this approach. We will examine the semantic relations which are used in WordNet and we will study the distinct characteristics of each of them. Furthermore, we will see which contribution is made by which relation to the organization of the lexicon. Finally, we will look at the downside of the fact that WordNet is a manually engineered network by examining the shortcomings of WordNet. In the third chapter, an alternative approach to linguistics is introduced. We will discuss the principles of corpus linguistics and, using the example of the British National Corpus, we will consider possibilities to extract semantic relations from language corpora which could help to overcome the deficiencies of the knowledge based approach. In the fourth chapter, I will describe a project the goal of which is to extend WordNet by findings from cognitive linguistics. Therefore, I will discuss the development process of a piece of software that has been programmed in the course of this thesis. The technique is used for characterizing the structural properties of multi-protein complexes. The high-resolution fluorescence microscopy and AFM are combined (FIONA-AFM) to allow for the identification of individual proteins in such complexes. This is achieved by labeling single proteins with fluorescent dyes and determining the positions of these fluorophores with high precision in an optical image. The same area of the sample is subsequently scanned by AFM. Finally, the two images are aligned and the positions of the fluorophores are displayed on top of the topographical data. Using quantum dots as fiducial markers in addition to fluorescently labeled proteins, fluorescence and AFM information can be aligned with an accuracy better than 10 nm, which is sufficient to identify single fluorescently labeled proteins in most multi-protein complexes. The limitations of localization precision and accuracy in fluorescence and AFM images are investigated, including their effects on the overall registration accuracy of FIONA-AFM hybrid images. This combination of the two complementary techniques opens a wide spectrum of possible applications to the study of protein interactions, because AFM can yield high resolution (5-10 nm) information about the conformational properties of multi-protein complexes while the fluorescence can indicate spatial relationships of the proteins within the complexes. Upon prolonged binding of an agonist, receptor internalization usually takes place, leading to the termination of this downstream signaling pathway and activation of alternative ones. However, a set of recent studies have shown that at least some GPCRs (e.g. thyroid stimulating hormone receptor) continue signaling to adenylyl cyclase after internalization. In this study, I aimed to investigate canonical signaling by internalized μ opioid receptors (MORs), which are Gi-coupled receptors, using a fluorescence resonance energy transfer (FRET) sensor for cyclic AMP (cAMP) known as Epac1-camps. My results show that the cyclic AMP inhibition signal induced by the binding of DAMGO, a MOR agonist, persists after agonist washout. We hypothesized that this persistent signal might come from internalized DAMGO-bound receptors located in the endosomal compartment. To test this hypothesis, I used dynasore and Dyngo 4a, two dynamin inhibitors that are known to prevent clathrin-mediated endocytosis. Interestingly, dynasore but not Dyngo 4a pretreatment largely blunted the response to MOR activation as well as to adenylyl cyclase activation with Forskolin (FSK). In addition, DAMGO-induced cAMP signal remained persistent even in the presence of 30 M Dyngo 4a. These results might point to a complex interplay between clathrin-mediated internalization and MOR signaling. The completely exposed Dwyka succession is interbedded with thin bentonites, altered distal pyroclastic deposits, which were derived from the magmatic arc at the southern rim of Gondwana. Dropstone-bearing and dropstonefree sequences intercalate with four diamictites, of which the two lowest were certainly recognised as tillites. Four events of deglaciation were proven at Zwartbas and thus consist with correlative deposits in southern Africa. Numerous fossilised fishes, trace fossils, and plant fragments appear frequently within the lower half of the Dwyka succession whereas trace fossils were principally found in the complete succession. Although the environmental determination is quite problematic, the fossil assemblage rather implies proximal, shallow water conditions with temporary restricted oxygenation. The hinterland was covered with considerable vegetation, which points to a moderate climate. Water salinity determinations based on shale geochemistry rectify contrary palaeontological results and point to rather brackish or non-marine conditions in comparison to present-day salinites. Geochemical analyses of the bentonites relate the pyroclastic deposits with acid to intermediate source magmas, as they are known from the magmatic arc in present-day Patagonia. Tectono-magmatic comparisons furthermore emphasise a syn-collision or volcanic-arc situation of the magma source. However, significant cyclicity in the production of the pyroclastic deposits was not observed. During the compilation of this paper doubts arose about the correct classification of the Nama rocks as it is found in literature. Since no certain clues were found to revise the classification of the Nama rocks, the original classification remains still valid. Thus the Kuibis and Schwarzrand Subgroup constitute the Nama succession and date it to Vendian age. A glacial unconformity represents a hiatus for about 260 Ma. This is covered by sediments of the Karoo Supergroup. Late Carboniferous and early Permian glacial deposits of diamictitic shale of the Dwyka and shales of the Ecca Group overlie the unconformity. The shales of the Dwyka Group contain fossiliferous units and volcanic ash-layers. A sill of the Jurassic Tandjiesberg Dolerite Complex (also Karoo Supergroup) intruded rocks at the Dwyka-Ecca-boundary. Through qualitative interviews with eco citizens, this study offers an insight into life in an eco city and how aware residents are of their new home's targets and incentives. The thesis investigates, how the eco city encourages residents, who have often never received adequate environmental education, to recycle and adapt their purchasing behaviour. The findings reveal the existence of several technical measures, practical incentive schemes and dissemination techniques that encourage residents to act according to environmental considerations. However, residents are often unaware of the numerical targets or do not make the connection between incentive and intended behaviour. The study highlights possibilities for improvement, in order to enhance residents' understanding of the eco city's objectives. Advantages of SMLM imply its superior specificity due to targeted molecular fluorescence labeling and its enhanced tissue preservation compared to electron microscopy, while reaching similar resolution. To reveal the molecular organization of protein structures in brain tissue, SMLM moves to the forefront: Instead of investigating brain slices with a thickness of a few µm, measurements of intact neuronal assemblies (up to 100 µm in each dimension) are required. As proteins are distributed in the whole brain volume and can move along synapses in all directions, this method is promising in revealing arrangements of neuronal protein markers. However, diffraction-limited imaging still required for the localization of the fluorophores is prevented by sample-induced distortion of emission pattern due to optical aberrations in tissue slices from non-superficial planes. In particular, the sample causes wavefront dephasing, which can be described as a summation of Zernike polynomials. To recover an optimal point spread function (PSF), active shaping can be performed by the use of adaptive optics. The aim of this thesis is to establish a setup using a deformable mirror and a wavefront sensor to actively shape the PSF to correct the wavefront phases in a super-resolution microscope setup. With the ability to address local needs more precisely in their unique geographic, social and economic contexts, cities play a vital role in implementing overall climate mitigation goals on the local level. In China, many renewable energy cities have emerged as well. However, official documents suggest that Chinese government authorities establish such renewable energy cities strategically, which leads to the assumption that the impulse to become renewable is different from other countries, where bottom-up initiatives are more common. Hence, this thesis explores answer to the question why and how the Chinese government promotes the energy transition of Chinese cities and regions. To explore the dynamics of local energy transition projects, this thesis adopts two frameworks from the field of sustainability transitions, the multi-level perspective and strategic niche management, and applies them to seven European and two Chinese case studies. The European sample includes the cities Graz, G{\"u}ssing, Freiburg, and Helsinki as well as the communities Feldheim, J{\"u}hnde and Murau. The Chinese sample consists of the bottom-up initiative Shaanxi Sunflower Project and the demonstration project Tongli New Energy Town. A comparative analysis evaluates in how far the cases correspond to the multi-level perspective or strategic niche management. The comparison of the case studies reveals that the development of renewable energy cities in China goes beyond a top-down vs. bottom-up logic. In the Chinese context, strategic niche management should be understood as experimentation under hierarchy, which serves at pretesting different approaches before rolling them out nationwide. In addition, the analysis shows that both the multi-level perspective and strategic niche management have their advantages and disadvantages for niche development. Niches following the logic of the multi-level perspective may result in higher stakeholder acceptance, whereas strategic niche management can in turn accelerate niche development. However, since natural niche development cannot be steered intentionally, decision-makers who intend to induce local renewable energy projects have no other option but to resort to strategic niche management. To increase stakeholder acceptance and thus to improve the project outcome, decision-makers are advised to accommodate sufficient room for stakeholder participation in the project design.}, subject = {China}, language = {en} }
