Methods and results The frequency of 12 comorbidities and their impact on NYHA class and SF-36 physical functioning score (SF-36 PF) were evaluated in 1,294 patients with HFpEF and 2,785 with HFrEF. HFpEF patients had lower NYHA class (2.0 ± 0.6 vs. 2.4 ± 0.6, p < 0.001) and higher SF-36 PF score (54.4 ± 28.3 vs. 54.4 ± 27.7, p < 0.001). All comorbidities were significantly (p < 0.05) more frequent in HFrEF, except hypertension and obesity, which were more frequent in HFpEF (p < 0.001). Adjusting for age and gender, COPD, anemia, hyperuricemia, atrial fibrillation, renal dysfunction, cerebrovascular disease and diabetes had a similar (p for interaction > 0.05) negative effect in both groups. Obesity, coronary artery disease and peripheral arterial occlusive disease exerted a significantly (p < 0.05) more adverse effect in HFpEF, while hypertension and hyperlipidemia were associated with fewer (p < 0.05) symptoms in HFrEF only. The total impact of comorbidities on NYHA (AUC for prediction of NYHA III/IV vs. I/II) and SF-36 PF (r 2) in multivariate analyses was approximately 1.5-fold higher in HFpEF, and also much stronger than the impact of a 10\% decrease in ejection fraction in HFrEF or a 5 mm decrease in left ventricular end-diastolic diameter in HFpEF. Conclusion The impact of comorbidities on physical impairment is higher in HFpEF than in HFrEF. For this purpose, the effects of PS on cognition, anxiety-and depression-related behavior were examined using a maternal restraint stress paradigm of PS in C57BL6 wild-type (WT) and heterozygous 5-Htt deficient (5-Htt +/-) mice. Additionally, in female offspring, a genome-wide hippocampal gene expression profiling was performed using the Affymetrix GeneChip (R) Mouse Genome 430 2.0 Array. 5-Htt +/- offspring showed enhanced memory performance and signs of reduced anxiety as compared to WT offspring. In contrast, exposure of 5-Htt +/- mice to PS was associated with increased depressive-like behavior, an effect that tended to be more pronounced in female offspring. Further, 5-Htt genotype, PS and their interaction differentially affected the expression of numerous genes and related pathways within the female hippocampus. Specifically, MAPK and neurotrophin signaling were regulated by both the 5-Htt +/- genotype and PS exposure, whereas cytokine and Wnt signaling were affected in a 5-Htt genotypexPS manner, indicating a genexenvironment interaction at the molecular level. We were able to differentiate untreated neuroblastoma patients from untreated Wilms tumor patients with an accuracy of 86.8\%, a sensitivity of 87.0\% and a specificity of 86.7\%. The separation of treated neuroblastoma patients from treated Wilms tumor patients' yielded comparable results with an accuracy of 83.8\%. We furthermore identified the antigens that contribute most to the differentiation between both tumor types. The analysis of these antigens revealed that neuroblastoma was considerably more immunogenic than Wilms tumor. The reported antigens have not been found to be relevant for comparative analyses between other tumors and controls. Results Data on dosimetry and biokinetics can be difficult to find, are scattered in various journals and, especially in paediatric nuclear medicine, are very scarce. The data collection and calculation methods vary with respect to the time-points, bladder voiding, dose assessment after the last data point and the way the effective dose was calculated. In many studies the number of subjects included for obtaining biokinetic and dosimetry data was fewer than ten, and some of the biokinetic data were acquired more than 20 years ago. Conclusion It would be of interest to generate new data on biokinetics and dosimetry in diagnostic nuclear medicine using state-of-the-art equipment and more uniform dosimetry protocols. We show that EcN affects the initial Salmonella invasion steps by modulating Salmonella virulence gene regulation and Salmonella SiiE-mediated adhesion, but not extra-and intracellular Salmonella growth. However, the inhibitory activity of EcN against Salmonella invasion always correlated with EcN adhesion capacities. EcN mutants defective in the expression of F1C fimbriae and flagellae were less adherent and less inhibitory toward Salmonella invasion. Another E. coli strain expressing F1C fimbriae was also adherent to IPEC-J2 cells, and was similarly inhibitory against Salmonella invasion like EcN. Conclusions: We propose that EcN affects Salmonella adhesion through secretory components. A further aim was to offer recommendations to control neuropathic pain. Results: We describe the neuropathy in Fabry disease, focusing on peripheral small fiber dysfunction - the hallmark of early neurologic involvement in this disorder. The clinical course of peripheral pain is summarized, and the importance of medical history-taking, including family history, is highlighted. A thorough physical examination (e. g., angiokeratoma, corneal opacities) and simple non-invasive sensory perception tests could provide clues to the diagnosis of Fabry disease. Reported early clinical benefits of enzyme replacement therapy include reduction of neuropathic pain, and adequate management of residual pain to a tolerable and functional level can substantially improve the quality of life for patients. Conclusions: Our recommendations can assist in diagnosing Fabry small fiber neuropathy early, and offer clinicians guidance in controlling peripheral pain. By viral proliferation assays and cell survival tests, we demonstrated that GLV-1h68 efficiently colonized, replicated in, and did lyse these cancer cells in culture. Experiments with HuH7 and PLC xenografts have revealed that a single intravenous injection (i.v.) of mice with GLV-1h68 resulted in a significant reduction of primary tumor sizes compared to uninjected controls. In addition, replication of GLV-1h68 in tumor cells led to strong inflammatory and oncolytic effects resulting in intense infiltration of MHC class II-positive cells like neutrophils, macrophages, B cells and dendritic cells and in up-regulation of 13 pro-inflammatory cytokines. Furthermore, GLV-1h68 infection of PLC tumors inhibited the formation of hemorrhagic structures which occur naturally in PLC tumors. Interestingly, we found a strongly reduced vascular density in infected PLC tumors only, but not in the non-hemorrhagic HuH7 tumor model. We observed that SOX11 expression is associated with unmethylated DNA and presence of activating histone marks (H3K9/14Ac and H3K4me3) in embryonic stem cells and some aggressive B-cell neoplasms. In contrast, adult stem cells, normal hematopoietic cells and other lymphoid neoplasms do not express SOX11. Such repression was associated with silencing histone marks H3K9me2 and H3K27me3. The SOX11 promoter of non-malignant cells was consistently unmethylated whereas lymphoid neoplasms with silenced SOX11 tended to acquire DNA hypermethylation. SOX11 silencing in cell lines was reversed by the histone deacetylase inhibitor SAHA but not by the DNA methyltransferase inhibitor AZA. These data indicate that, although DNA hypermethylation of SOX11 is frequent in lymphoid neoplasms, it seems to be functionally inert, as SOX11 is already silenced in the hematopoietic system. However, recent findings demonstrate that Trk receptors can be transactivated independently of their neurotrophin ligands. Methodology: In this study we examined the role of BDNF, TrkB kinase activity and monoamine reuptake in the AD-induced TrkB activation in vivo and in vitro by employing several transgenic mouse models, cultured neurons and TrkB-expressing cell lines. Principal Findings: Using a chemical-genetic TrkB(F616A) mutant and TrkB overexpressing mice, we demonstrate that ADs specifically activate both the maturely and immaturely glycosylated forms of TrkB receptors in the brain in a TrkB kinase dependent manner. However, the tricyclic AD imipramine readily induced the phosphorylation of TrkB receptors in conditional bdnf(-/-) knock-out mice (132.4+/-8.5\% of control; P = 0.01), indicating that BDNF is not required for the TrkB activation. Moreover, using serotonin transporter (SERT) deficient mice and chemical lesions of monoaminergic neurons we show that neither a functional SERT nor monoamines are required for the TrkB phosphorylation response induced by the serotonin selective reuptake inhibitors fluoxetine or citalopram, or norepinephrine selective reuptake inhibitor reboxetine. However, neither ADs nor monoamine transmitters activated TrkB in cultured neurons or cell lines expressing TrkB receptors, arguing that ADs do not directly bind to TrkB. We thus investigated the association of these loci with incident CKD in 26,308 individuals of European ancestry free of CKD at baseline drawn from eight population-based cohorts followed for a median of 7.2 years (including 2,122 incident CKD cases defined as eGFR < 60ml/min/1.73m(2) at follow-up) and with ESRD in four case-control studies in subjects of European ancestry (3,775 cases, 4,577 controls). SNPs at 11 of the 16 loci (UMOD, PRKAG2, ANXA9, DAB2, SHROOM3, DACH1, STC1, SLC34A1, ALMS1/NAT8, UBE2Q2, and GCKR) were associated with incident CKD; p-values ranged from p = 4.1e-9 in UMOD to p = 0.03 in GCKR. After adjusting for baseline eGFR, six of these loci remained significantly associated with incident CKD (UMOD, PRKAG2, ANXA9, DAB2, DACH1, and STC1). SNPs in UMOD (OR = 0.92, p = 0.04) and GCKR (OR = 0.93, p = 0.03) were nominally associated with ESRD. Considering that adaptive immune system is well developed in older children, and that T cells were shown to be involved in tumor escape and progression of cancers, we sought to determine whether HR patients may tend to show a signature of adaptive immune responses compared to LR patients who tend to have diminished T-cell responses but an intact innate immune response. Methods: We performed microarray analysis of RNA extracted from the tumor specimens of HR and LR patients. Flow cytometry was performed to determine the cellular constituents in the blood while multiplex cytokine array was used to detect the cytokine profile in patients' sera. A HR tumor cell line, SK-N-SH, was also used for detecting the response to IL-1 beta, a cytokines which is involved in the innate immune responses. Results: Distinct patterns of gene expression were detected in HR and LR patients indicating an active T-cell response and a diminished adaptive immune response, respectively. A diminished adaptive immune response in LR patients was evident by higher levels of IL-10 in the sera. In addition, HR patients had lower levels of circulating myeloid derived suppressor cells (MDSC) compared with a control LR patient. LR patients showed slightly higher levels of cytokines of the innate immune responses. Treatment of the HR tumor line with IL-1b induced expression of cytokines of the innate immune responses. The stimulus material consisted of true and false negated sentences (sentence level) and prime-target expressions (word level). Stimuli were presented acoustically and no overt behavioral response of the participants was required. Event-related brain potentials to target words preceded by true and false negated expressions were analyzed both within group and at the single subject level. Across the different processing conditions (word pairs and sentences), target words elicited a frontal negativity and a late positivity in the time window from 600-1000 msec post target word onset. Amplitudes of both brain potentials varied as a function of the truth value of the negated expressions. Results were confirmed at the single-subject level. In sum, our results support recent suggestions according to which evaluation of the truth value of a negated expression is a time-and cognitively demanding process that cannot be solved automatically, and thus requires conscious processing. Our paradigm provides insight into higher-order processing related to language comprehension and reasoning in healthy subjects. As predicted, the view-fearful condition led to higher arousal ratings than the view-neutral condition with the reappraise-fearful condition in between. For the fNIRS results the induction condition showed an activation of the bilateral PFC compared to the baseline condition (viewing neutral). The regulation condition showed an activation only of the left PFC compared to the baseline condition, although the direct comparison between induction and regulation condition revealed no significant difference in brain activation. In dem ersten Teil dieser Arbeit konnte gezeigt werden, dass die Behandlung von M{\"a}usen mit dem neu generierten monoklonalen Antik{\"o}rper INU1, der gegen murines CLEC-2 gerichtet ist, zu dem vollst{\"a}ndigen und hochspezifischen Verlust des Rezeptors in zirkulierenden Thrombozyten f{\"u}hrte, ein Prozess, der als „Immundepletion" bezeichnet wird. Die CLEC-2-defizienten Thrombozyten waren nicht mehr durch den CLEC-2-spezifischen Agonisten Rhodozytin aktivierbar, w{\"a}hrend die Aktivierung durch alle anderen getesteten Agonisten nicht beeintr{\"a}chtigt war. Dieser selektive Defekt f{\"u}hrte unter Flussbedingungen ex vivo zu stark verminderter Aggregatbildung der Thrombozyten. Außerdem zeigten in vivo-Thrombosestudien, dass die gebildeten Thromben instabil waren und vermehrt embolisierten. Infolgedessen war die CLEC-2 Defizienz mit einem deutlichen Schutz vor arterieller Thrombose verbunden. Außerdem ließ die in INU1-behandelten M{\"a}usen beobachtete variable Verl{\"a}ngerung der Blutungszeit auf einen moderaten h{\"a}mostatischen Defekt schließen. Diese Ergebnisse zeigen zum ersten Mal, dass CLEC-2 in vitro und in vivo signifikant zur Thrombusstabilit{\"a}t beitr{\"a}gt und eine essentielle Rolle in der H{\"a}mostase und arteriellen Thrombose spielt. Daher stellt CLEC-2 eine potentiell neue antithrombotische Zielstruktur dar, die in vivo inaktiviert werden kann. Diese in vivo-Herabregulierung von Thrombozytenoberfl{\"a}chenrezeptoren k{\"o}nnte einen vielversprechenden Ansatz f{\"u}r zuk{\"u}nftige antithrombotische Therapien darstellen. Der zweite Teil dieser Arbeit behandelte den Effekt einer Doppelimmundepletion der immunoreceptor tyrosine-based activation motiv (ITAM)- und hemITAM-gekoppelten Rezeptoren Glykoprotein (GP) VI und CLEC-2 auf H{\"a}mostase und Thrombose mittels einer Kombination der GPVI- beziehungsweise CLEC-2-spezifischen Antik{\"o}rper JAQ1 und INU1. Eine Einzeldepletion von GPVI oder CLEC-2 in vivo beeintr{\"a}chtigte nicht die Expression und Funktion des jeweils anderen Rezeptors. Eine gleichzeitige Behandlung mit beiden Antik{\"o}rpern f{\"u}hrte jedoch zu dem nachhaltigen Verlust der GPVI- und CLEC-2-vermittelten Signale in Thrombozyten, w{\"a}hrend andere Signalwege nicht betroffen waren. Im Gegensatz zu den Einzeldefizienzen, wiesen die GPVI/CLEC-2 doppeldefizienten M{\"a}use einen schwerwiegenden Blutungsph{\"a}notyp auf. Außerdem f{\"u}hrte die Behandlung zu einer starken Beeintr{\"a}chtigung der arteriellen Thrombusbildung, die die Effekte der Einzeldefizienzen weit {\"u}bertraf. Von Bedeutung ist auch, dass gleiche Ergebnisse in Gp6-/- M{\"a}usen gefunden wurden, die mittels INU1-Behandlung CLEC-2-depletiert wurden. Dies veranschaulicht, dass der Blutungsph{\"a}notyp nicht durch Sekund{\"a}reffekte der kombinierten Antik{\"o}rperbehandlung hervorgerufen wurde. Die vorliegende Arbeit besch{\"a}ftigt sich mit der Etablierung und Validierung von LTP in hippocampalen Hirnschnitten der Ratte um ged{\"a}chtnissteigernde Substanzen zur potentiellen Behandlung der Alzheimer'schen Erkrankung zu charakterisieren. Dazu wurde zun{\"a}chst ein Messsystem zur parallelen Charakterisierung mehrerer Schnitte aufgebaut, das Messungen bis zu sieben Stunden erlaubt (Kapitel 2). Dann wurden unterschiedliche Protokolle etabliert um Fr{\"u}h- und Sp{\"a}tphasen-LTP zu generieren. Dabei w{\"u}rde Fr{\"u}hphasen-LTP konzeptionell eher mit dem Kurzzeitged{\"a}chtnis einhergehen, w{\"a}hrend Sp{\"a}tphasen-LTP dem Langzeitged{\"a}chtnis gleichkommen w{\"u}rde (Kapitel 3). Da in Alzheimer-Patienten haupts{\"a}chlich ein Defizit cholinerger und glutamaterger Neurone vorliegt, wurden die validierten LTP Formen benutzt, um solche Substanzen zu analysieren, die potentiell cholinerge und/oder glutamaterge neuronale Funktion erh{\"o}hen. Die Effekte zweier ausschließlich cholinerge Funktion erh{\"o}hender Substanzen wurden analysiert: Der α4β2 nicotinische Acetylcholin-Rezeptor Agonist TC-1827 (Kapitel 4) und der Acetylcholinesterase-Inhibitor Donepezil (Kapitel 5). Beide Substanzen erh{\"o}hten Fr{\"u}hphasen-LTP, aber hatten keinen Effekt auf Sp{\"a}tphasen-LTP. ADX-47273 erh{\"o}hte Sp{\"a}tphasen-LTP, aber hatte keinen Effekt auf Fr{\"u}hphasen-LTP, wohingegen BAY 73-6691 eine erh{\"o}hende Wirkung auf beide LTP Formen aufwies und sogar Fr{\"u}h- in Sp{\"a}tphasen-LTP umwandelte. Die gleichen Effekte, wie bei dem PDE9A-Inhibitor, konnten auch mit dem partiellen α7 nicotinische Acetylcholin-Rezeptor Agonisten SSR180711 (Kapitel 4) demonstriert werden. SSR180711 wirkt sowohl auf cholinerge, als auch auf glutamaterge neuronale Funktion. Dann wurde die F{\"a}higkeit der Substanzen {\"u}berpr{\"u}ft, durch l{\"o}sliche Aβ Oligomere verschlechtertes LTP zu verbessern (Kapitel 6). L{\"o}sliche Aβ Oligomere, auch als amyloid-β derived diffusible ligands (ADDLs) bezeichnet, werden zurzeit als eine mutmaßliche Ursache der Alzheimer'schen Erkrankung angesehen. In der vorliegenden Arbeit wurde gezeigt, dass ADDLs Fr{\"u}h- und Sp{\"a}tphasen-LTP in verschiedenem Ausmaß vermindern. Donepezil und TC-1827 konnten die durch ADDLs induzierten Defizite bei Fr{\"u}hphasen-LTP geringf{\"u}gig wiederherstellen, aber sie hatten keinen Einfluss auf das durch ADDLs verschlechterte Sp{\"a}tphasen-LTP. Im Gegensatz dazu, konnten sowohl SSR180711 als auch BAY 73-6691 ein durch ADDLs verschlechtertes Fr{\"u}h- und Sp{\"a}tphasen-LTP komplett wiederherstellen. ADX-47273 hatte keinen positiven Effekt auf Fr{\"u}hphasen-LTP, welches durch ADDLs verschlechtert worden war, konnte aber ein durch ADDLs verschlechtertes Sp{\"a}tphasen-LTP teilweise wiederherstellen. Somit wurde der vorherige Befund der Arbeit best{\"a}tigt: Substanzen, welche die glutamaterge Funktion verbessern, scheinen nicht nur wirksamer im Bezug auf LTP-Erh{\"o}hung zu sein als Substanzen die ausschließlich cholinerge Funktion erh{\"o}hen, sondern sie sind auch in der Lage, durch l{\"o}sliche Aβ Oligomere verursachte Defizite bei LTP zu verbessern. Aus einem pr{\"a}klinischen Blickwinkel und basierend auf den Ergebnissen der vorliegenden Arbeit weisen demnach Substanzen, die glutamaterge Funktionen verbessern, ein hohes therapeutisches Potential als alternative Ans{\"a}tze bez{\"u}glich kognitiver Defizite auf. The indocyanine green plasma disappearance rate, serum bilirubin, aspartate aminotransferase, alanine aminotransferase, γ-glutamyltransferase, alkaline phosphatase, glutamate dehydrogenase, lactate dehydrogenase and creatine kinase were determined repeatedly. Finally, liver neutrophils were counted and liver cell apoptosis was assessed by terminal deoxynucleotidyl transferase nick end labeling (TUNEL). Results: Aspartate aminotransferase increased in the PCV group about three-fold and in the HFOV+ECLA group five-fold (p\<0.001). The second essay extends the model to a two-country world consisting of the capital-rich "North" and the capital-poor "South". The North has trade unions that set high OHS standards. There are no unions in the South and OHS standards are low. Trade between these two countries can imply a reduction in safety standards in the North, lowering the positive welfare effects of trade. Moreover, when trade unions are also established in the South, northern OHS standards might be further reduced. The third essay studies the impact of unions on OHS from an empirical perspective. It focuses on one component of OHS: occupational injuries. A literature summary including 25 empirical studies shows that most studies associate unions with less fatal occupational injuries. This is in perfect line with the anecdotal evidence and the basic model from the first essay. However, the literature summary also shows that most empirical studies associate unions with more nonfatal occupational injuries. This puzzling result has been explained in the literature by (1) lower underreporting in unionized workplaces, (2) unions being more able to organize hazardous workplaces, and (3) unionized workers preferring higher wages at the expense of better working conditions. Using individual-level panel data, this essay presents evidence against all these three explanations. However, it cannot reject the hypothesis that workers reduce their precautionary behaviour when they join a trade union. Hence, the puzzle seems to be due to a strong moral hazard effect. The electronic disorder is characterized by statistically distributed and localized transport sites which were shown to form a Gaussian density of states. In this electronic environment the charge transport occurs via thermally activated hopping between the localized states and therefore depends on the temperature and the local electric field. Particularly, a dependence of the carrier mobility on the charge carrier concentration is observed due to filling of tail states. Inorganic nanocrystalline semiconductors, however, are expected to present a different electronic structure: Within the volume of a nanocrystallite the semiconductor is assumed to reflect the electronic properties of the crystalline bulk material. However, the outer shell is characterized by a relatively large density of surface states and correspondingly bending of the energy bands, which creates an energetic barrier between the adjacent particles. In a nanocrystalline thin film this characteristic can be rate-limiting for the inter-particle carrier transport as reflected by reduced charge carrier mobility. The effective barrier height can be reduced by controlled doping of the nanocrystals which results in improved majority carrier transfer rates across the barrier. However, doping results in the simultaneous increase of the defect density and consequently to enhanced limitation of the mobility due to charge carrier scattering. Die von BMP induziete breite Palette von biologischen Reaktionen konvergiert auf drei eng verwandten Smad Proteine. Sie vermitteln intrazellul{\"a}re Signale von BMP-Rezeptoren in den Zellkern. Die Spezifit{\"a}t des BMP-Signalwegs wurde intensiv auf der Ebene der Ligand-Rezeptor-Wechselwirkungen erforscht, aber, wie die verschiedenen Smad Proteine die durch BMPs hervorgerufen differenziellen Signale beitragen, bleibt unklar. In dieser Arbeit haben wir die BMP / Smad Signalweg in verschiedenen Aspektenuntersucht. Auf der Suche nach einem geeigneten Fluoreszenz-Reporter im Zebrafisch, verglichen wir verschiedene photo-schaltbaren Proteine und fand EosFP der beste Kandidat f{\"u}r diesen Modellorganismus im Bezug auf seine schnelle Reifung und Fluoreszenz-Intensit{\"a}t. Wir haben durch molekulare Modifizierung geeignete Vektoren erstellt, die Tol2-Transposon basieren trangenesis im Zebrafisch zu erm{\"o}glichen. Damit wurden schließlich transgenzebrafisch-Linien erzeugt. Wir kombinierten Fluoreszenz-Protein-Tagging mit hochaufl{\"o}sender Mikroskopie und untersuchten die Dynamik der Smad-Proteine in Modellsystem Zebrafisch. Es wurde beobachteten, dass Smad5 Kern-Translokation erf{\"a}hrt, als BMP Signalgeber bei Zebrafisch Gastrulation. Wir erkundeten die Beteiligung der Smad Proteine w{\"a}hrend der Myogenese-zu-Osteogenese Umwandlung von C2C12 Zelllinie, die durch BMP4 induziert wurde. Mit siRNA versuchten wir die endogene Smad Proteine niederzuschlagen, wobei die Auswirkungen auf diesen gekoppelten noch unterschiedlichen Verfahren durch quantitative real-time PCR und Terminal-Marker F{\"a}rbung ausgewertet. Adhesion to the extracellular matrix occurs at specialized plasma membrane domains where transmembrane adhesion receptors, signaling proteins such as kinases and phosphatases, and a large number of adaptor proteins interact with the cytoskeleton in a tightly regulated and synchronized fashion. Whereas altered cell adhesion and migration are known to be important in cardiovascular disease and malignant tumors, the target proteins and molecular interactions that regulate these complex processes still remain incompletely understood. Whereas numerous kinases are known to regulate cell adhesion dynamics, information about the involved protein phosphatases is still very limited. A newly emerging phosphatase family contains the unconventional active site sequence DXDX(T/V) and belongs to the haloacid dehalogenase (HAD) superfamily of hydrolases. Our laboratory has recently discovered AUM, a novel phosphatase that belongs to this poorly characterized enzyme family. Initial findings pointed toward a potential involvement of AUM in the regulation of cell adhesion to the extracellular matrix. The objective of the present study was to study the potential role of AUM in cell adhesion. We could show that cells stably depleted of AUM are characterized by accelerated adhesion on immobilized fibronectin. To confirm these findings, we used an siRNA-based approach for the acute depletion of AUM and observed a similar phenomenon. Rescue experiments were performed with stably AUM-depleted cells to ensure that the above mentioned effects are indeed AUM specific. We observed that the re-addition of AUM normalizes cellular adhesion kinetics on fibronectin. These results clearly show that AUM exerts important functions in cell-matrix adhesion. To investigate the molecular basis of these effects, we have characterized integrin expression patterns using flow cytometry. Interestingly, fibronectin-stimulated AUM-depleted cells are characterized by an increase in the cell surface expression of conformationally active 1-integrins. Consistent with the important role of 1-integrins in the regulation of RhoA activity, we also observed a specific increase in RhoA-GTP, but not Rac1-GTP-levels during cell adhesion to fibronectin. Consistent with these findings and with the important role of RhoA for focal adhesion maturation, AUM depleted cells showed more elongated and more centripetally oriented focal adhesions as compared to control cells when spread on fibronectin. Taken together, this study has revealed an important role of AUM for cell-matrix adhesion. We analyzed the production of cAMP after aab-mediated β1-AR activation in vitro using a fluorescence resonance energy transfer (FRET) assay. This assay is based on HEK293 cells stably expressing human β1-AR as well as the cAMP-sensor Epac1-camps. The assay showed a concentration-dependent increase in intracellular cAMP upon stimulation with the full agonist (-) isoproterenol. This response was comparable to results obtained in isolated adult murine cardiomyocytes and was partially blockable by a selective β1-AR antagonist. In the same assay poly- and monoclonal anti-β1ECII-abs (induced in different animals) could activate the adrenergic signaling cascade, whereas isotypic control abs had no effect on intracellular cAMP levels. Using the same method, we were able to detect functionally activating aabs in the serum of heart failure patients with ischemic and hypertensive heart disease as well as patients with DCM, but not in sera of healthy control subjects. In patients with DCM we observed an inverse correlation between the stimulatory potential of anti-β1-aabs and left ventricular pump function. To adopt this assay for the detection of functionally activating anti-β1ECII-aabs in clinical routine we attempted to establish an automated large-scale approach. Neither flow cytometry nor FRET detection with a fluorescence plate reader provided an acceptable signal-to-noise ratio. It was possible to detect (-) isoproterenol in a concentration-dependent manner using two different FRET multiwell microscopes. However, due to focus problems large-scale detection of activating anti-β1ECII-abs could not be implemented. Neutralization of anti-β1-aabs with the corresponding epitope-mimicking peptides is a possible therapeutic approach to treat aab-associated autoimmune DCM. Using our FRET assay we could demonstrate a reduction in the stimulatory potential of anti-β1ECII-abs after in vitro incubation with β1ECII-mimicking peptides. Cyclic (and to a lesser extent linear) peptides in 40-fold molar excess acted as efficient ab-scavengers in vitro. Intravenously injected cyclic peptides in a rat model of DCM also neutralized functionally active anti-β1ECII-abs efficiently in vivo. For a detailed analysis of the receptor-epitope targeted by anti-β1ECII-abs we used sequentially alanine-mutated β1ECII-mimicking cyclic peptides. Our data revealed that the disulfide bridge between the cysteine residues C209 and C215 of the human β1-AR appears essential for the formation of the ab-epitope. Substitution of further amino acids relevant for ab-binding in the cyclic scavenger peptide by alanine reduced its affinity to the ab and the receptor-activating potential was blocked less efficiently. In contrast, the non-mutant cyclic peptide almost completely blocked ab-induced receptor activation. Using this ala-scan approach we were able to identify a "NDPK"-epitope as essential for ab binding to the β1ECII. The aim of this thesis was to investigate if the immunostimulatory properties of Toll-like receptor (TLR) agonists can be utilized to develop novel therapeutic intervention strategies for the treatment of asthma using murine models of allergic inflammation. For this purpose five different TLR agonists were tested in preclinical mouse models of acute and chronic asthma, both in preventive and therapeutic settings. Firstly, TLR-2, 3, 4, 7/8 and 9 agonists were delivered intratracheally at different doses before pulmonary allergen exposure in the asthma model of acute inflammation. TLR9 agonist CpG-containing oligodeoxynucleotides (CpG) > TLR7 agonist Resiquimod (R848) > TLR3 agonists poly(I:C) strongly reduced allergen induced airway eosinophilia and IL-4 levels in a dose-dependent manner. All TLR agonists increased neutrophil numbers, TLR4 agonist lipopolysaccharide (LPS) > TLR2 agonist lipoteichonic acid (LTA) > poly(I:C) > CpG > R848 and, with the exception of R848, the amount of pro-inflammatory cytokines in the airways. Suppressive effects were not dependent upon IFN-γ and IL-10 or associated with increased numbers of regulatory T cells in the airways. All TLR agonists, except LTA, similarly reduced airway eosinophilia and IL-4 levels when applied therapeutically after allergen challenge. These results show that the TLR agonists have different suppressive effects on TH2 responses in the airways which further depend on the dose and the experimental setup in which they were tested. Interestingly, all agonists induced airway neutrophilia, albeit to different degrees, raising the question if TLR ligands are safe for human use when applied directly into the lung. Different TLR agonists are also being developed for human use as adjuvants combined with allergen in specific immunotherapy. Recent clinical data suggest that this may be achieved by induction of allergen-specific TH1 responses. For this reason, the ability of different TLR agonists to induce allergen-specific TH1 and suppress allergen-specific TH2 responses in a preclinical setting was investigated in this thesis. Different doses of the TLR agonists were applied together with allergen, then mice were exposed to allergen aerosol. CpG > LPS >LTA dose-dependently strongly suppressed the development of airway eosinophilia with poly(I:C) and R848 having no effect. The decrease in eosinophilic numbers was associated withincreased neutrophils present in the airways. IL-4 and IL-5 levels in the bronchoalveolar lavage fluid were also decreased when poly(I:C), LPS, and CpG were used. All TLR agonists increased allergen-specific IgG2a, and with the exception of poly(I:C), reduced allergen-specific IgE levels in the serum. Cutaneous anaphylaxis to allergen was completely prevented when LPS or CpG were given as adjuvant. The strongest TH1 responses were induced by CpG and poly(I:C), characterized by the presence of IFN-γ in the bronchoalveolar lavage and the highest allergen-specific IgG2a levels in the serum. This data supports approaches to use TLR9 or TLR4 agonists for human therapy as adjuvant in combination with allergen in novel specific immunotherapy formulations. In the last part of the thesis, it was investigated if TLR activation can also affect the pathology of severe chronic asthma. Therapeutic administration of R848 or CpG reduced features of inflammation and remodeling. Both agonists showed superior effects to dexamethasone, with CpG being more efficient than R848. This result again supports a TLR9-based therapy as a viable option for the treatment of severe chronic asthma which may present a potential alternative for anti-inflammatory therapy with steroids. Taken together, the results of this thesis support the use of TLR agonists to treat asthma. The most favorable efficacy/safety ratio is to be expected from TLR-based therapies combining TLR4 or TLR9 agonists with allergen in specific immunotherapy. With the help of genetically modified mice and the use of specific protein inhibitors and receptordepleting antibodies, the work presented in this thesis identified novel mechanisms underlying thrombus formation in hemostasis and thrombosis. In the first part of the study, it was shown that von Willebrand Factor (vWF) binding to glycoprotein (GP)Iba is critical for the formation of stable pathological thrombi at high shear rates, suggesting GPIba as an attractive pharmacological target for antithrombotic therapy. The subsequent analysis of recently generated phospholipase (PL)D1-deficient mice identified this enzyme, whose role in platelet function had been largely unknown, as a potential target protein downstream of GPIba. This was based on the finding that PLD1- deficient mice displayed severely defective GPIba-dependent thrombus stabilization under high shear conditions in vitro and in vivo without affecting normal hemostasis. The second part of the thesis characterizes the functional relevance of the immunoreceptor tyrosine-based activation motif (ITAM)-bearing collagen receptor GPVI and the recently identified hemITAM-coupled C-type lectin-like receptor 2 (CLEC-2) for in vivo thrombus formation. Genetic- and antibody-induced GPVI deficiency was found to similarly protect mice from arterial vessel occlusion in three different thrombosis models. These results confirmed GPVI as a promising antithrombotic target and revealed that antibody-treatment had no obvious off-target effects on platelet function. Similarly, immunodepletion of CLEC-2 by treating mice with the specific antibody INU1 resulted in markedly impaired thrombus growth and stabilization under flow in vitro and in vivo. Furthermore, it could be demonstrated that double-immunodepletion of GPVI and CLEC-2 resulted in severely decreased arterial thrombus formation accompanied by dramatically prolonged bleeding times. These data revealed an unexpected redundant function of the two receptors for in vivo thrombus formation and might have important implications for the potential development of anti-GPVI and anti-CLEC-2 antithrombotic agents. The third part of the thesis provides the first functional analysis of megakaryocyte- and platelet-specific RhoA knockout mice. RhoA-deficient mice displayed a defined signaling defect in platelet activation, leading to a profound protection from arterial thrombosis andand ischemic brain infarction, but at the same time also strongly increased bleeding times. It is responsible for sleeping sickness in sub-saharan Africa. The parasite cycle involves more than nine developmental stages that can be clearly distinguished by their general morphology, their metabolism and the relative positioning of their DNA-containing organelles. During their development, trypanosomes remain exclusively extracellular and encounter changing environments with different physico-chemical properties (nutritional availability, viscosity, temperature, etc.). It has been proposed that trypanosomes use their flagellum as a sensing organelle, in agreement with the established role of structurally-related cilia in metazoa and ciliates. Recognition of environmental triggers is presumed to be at the initiation of differentiation events, leading to the parasite stage that is the best suited to the new environment. These changes are achieved by the modification of gene expression programmes, mostly underlying post-transcriptional control of mRNA transcripts. We first demonstrate that the RNA-binding proteins ALBA3/4 are involved in specific differentiation processes during the parasite development in the fly. They are cytosolic and expressed throughout the parasite cycle with the exception of the stages found in the tsetse fly proventriculus, as shown by both immunofluorescence and live cell analysis upon endogenous tagging with YFP. Knock-down of both proteins in the developmental stage preceding these forms leads to striking modifications: cell elongation, cell cycle arrest and relocalization of the nucleus in a posterior position, all typical of processes acting in parasites found in the proventriculus region. When ALBA3 is over-expressed from an exogenous copy during infection, it interferes with the relocalization of the nucleus in proventricular parasites. This is not observed for ALBA4 over-expression that does not visibly impede differentiation. Both ALBA3/4 proteins react to starvation conditions by accumulating in cytoplasmic stress granules together with DHH1, a recognized RNA-binding protein. ALBA3/4 proteins also partially colocalize with granules formed by polyA+ RNA in these conditions. We propose that ALBA are involved in trypanosome differentiation processes where they control a subset of developmentally regulated transcripts. These processes involving ALBA3/4 are likely to result from the specific activation of sensing pathways. In the second part of the thesis, we identify novel flagellar proteins that could act in sensing mechanisms. Several protein candidates were selected from a proteomic analysis of intact flagella performed in the host laboratory. This work validates their flagellar localization with high success (85\% of the proteins examined) and defines multiple different patterns of protein distribution in the flagellum. Two proteins are analyzed during development, one of them showing down-regulation in proventricular stages. The functional analysis of one novel flagellar membrane protein reveals its rapid dynamics within the flagellum but does not yield a visible phenotype in culture. This is coherent with sensory function that might not be needed in stable culture conditions, but could be required in natural conditions during development. Especially the Port Jackson willow (Acacia saligna) extensively suppresses the indigenous Fynbos vegetation with its rapid growth. The main objective of this study was to investigate indicators for a quick and early prediction of the health of the remaining Fynbos fragments in the City of Cape Town with help of remote sensing. First, the productivity of the vegetation in response to rainfall was determined. For this purpose, the Enhanced Vegetation Index (EVI), derived from Terra MODIS data with a spatial resolution of 250m, and precipitation data of 19 rainfall stations for the period from 2000 till 2008 were used. Within the scope of a flexible regression between the EVI data and the precipitation data, different lags of the vegetation response to rainfall were analysed. Furthermore, residual trends (RESTREND) were calculated, which result from the difference between observed EVI and the one predicted by precipitation. Negative trends may suggest a degradation of the habitats. In addition, the so-called Rain-use Efficiency (RUE) was tested in this context. It is defined as the ratio between net primary production (NPP) - represented by the annual sum of EVI - and the annual rainfall sum. These indicators were analysed for their suitability to determine the health of the indigenous Fynbos vegetation. Furthermore, the degree of dispersal of invasive species especially the Acacia saligna was investigated. With the specific characteristics of the tested indicators and the spectral signature of Acacia saligna, i.e. its unique reflectance over the course of the year, the dispersal was estimated. Since the growth of invasive species dramatically reduces the biodiversity of the fragments, their presence is an important factor for the condition of ecosystem health. This work focused on 11 test sites with an average size of 200ha, distributed over the whole area of the City of Cape Town. Five of these fragments are under conservation and the others shall be protected in the near future, too, which makes them of special interest. In January 2010, fieldwork was undertaken in order to investigate the state and composition of the local vegetation. The results show promising indicators for the assessment of ecosystem health. The coefficients of determination of the EVI-rainfall regression for Fynbos are minor, because the reaction of this vegetation type to rainfall is considerably lower than the one of the invasive species. Thus, a good distinction between indigenous and alien vegetation is possible on the basis of this regression. On the other hand, the RESTREND method, for which the regression forms the basis, is only of limited use, since the significance of these trends is not given for Fynbos vegetation. Furthermore, the RUE has considerable potential for the assessment of ecosystem health in the study area. The Port Jackson willow has an explicitly higher EVI than the Fynbos vegetation and thus its RUE is more efficient for a similar amount of rainfall. However, it has to be used with caution, because local and temporal variability cannot be extinguished in the study area over the rather short MODIS time series. These results display that the interpretation of the indicators has to be conducted differently from the literature, because the element of invasive species was not considered in most of the previous papers. An increase in productivity is not necessarily equivalent with an improvement in health of the fragment, but can indicate a dispersal of Acacia saligna. This shows the general problem of the term 'degradation' which in most publications so far is only measured by productivity and other factors like invasive species are disregarded. On the basis of the EVI-rainfall regression and statistical measures of the EVI, the distribution of invasive species could be delineated. Generally, a strong invasion of the Port Jackson willow was discovered on the test sites. The results display that a reasoned and sustainable management of the fragments is essential in order to prevent the suppression of the indigenous Fynbos vegetation by Acacia saligna. Die Tatsache, dass Cataglyphis eine hohe Verhaltensflexibilit{\"a}t aufweist, welche mit sich drastisch {\"a}ndernden sensorischen Anforderungen verbunden ist, macht diese Ameisen zu besonders interessanten Studienobjekten bei der Erforschung synaptischer Plastizit{\"a}t visueller und olfaktorischer Gehirnzentren. Diese Arbeit fokussiert auf plastische {\"A}nderungen in den Pilzk{\"o}rpern (PK) - sensorischen Integrationszentren, die mutmaßlich an Lern- und Erinnerungsprozessen, und auch vermutlich am Prozess des Landmarkenlernens beteiligt sind - und auf plastische {\"A}nderungen in den synaptischen Komplexen des Lateralen Akzessorischen Lobus (LAL) - einer bekannten Relaisstation in der Polarisations-Leitungsbahn. Um die strukturelle synaptische Plastizit{\"a}t der PK in C. fortis zu quantifizieren, wurden mithilfe immunozytochemischer F{\"a}rbungen die pr{\"a}- und postsynaptischen Profile klar ausgepr{\"a}gter synaptischer Komplexe (Mikroglomeruli, MG) der visuellen Region (Kragen) und der olfaktorischen Region (Lippe) der PK-Kelche visualisiert. Erfahrene Fourageure wurden in Dunkelheit zur{\"u}ckgef{\"u}hrt, um zu untersuchen, ob die lichtausgel{\"o}ste synaptische Umstrukturierung reversibel ist, doch ihre PK zeigen nur einige die Zur{\"u}ckf{\"u}hrung widerspiegelnde Plastizit{\"a}tsauspr{\"a}gungen, besonders eine {\"A}nderung der pr{\"a}synaptischen Synapsinexprimierung. Mithilfe immunozytochemischer F{\"a}rbungen, konfokaler Mikroskopie und 3D-Rekonstruktionen wurden die pr{\"a}- und postsynaptischen Strukturen synaptischer Komplexe des LAL in C. fortis analysiert und potentielle strukturelle {\"A}nderungen bei Innendiensttieren und Fourageuren quantifiziert. Die Ergebnisse zeigen, dass diese Komplexe aus postsynaptischen, in einer zentralen Region angeordneten Forts{\"a}tzen bestehen, welche umringt sind von einem pr{\"a}synaptischen kelchartigen Profil. Eingehende und ausgehende Trakte wurden durch Farbstoffinjektionen identifiziert: Projektionsneurone des Anterioren Optischen Tuberkels kontaktieren Neurone, welche in den Zentralkomplex ziehen. Der Verhaltens{\"u}bergang wird von einer Zunahme an synaptischen Komplexen um ~13\% begleitet. Dieser Zuwachs suggeriert eine Art Kalibrierungsprozess in diesen potentiell kr{\"a}ftigen synaptischen Kontakten, welche vermutlich eine schnelle und belastbare Signal{\"u}bertragung in der Polarisationsbahn liefern. Die Analyse von im Freiland aufgenommener Verhaltenweisen von C. fortis enth{\"u}llen, dass die Ameisen, bevor sie mit ihrer Fouragiert{\"a}tigkeit anfangen, bis zu zwei Tage lang in unmittelbarer N{\"a}he des Nestes Entdeckungsl{\"a}ufe unternehmen, welche Pirouetten {\"a}hnliche Drehungen beinhalten. W{\"a}hrend dieser Entdeckungsl{\"a}ufe sammeln die Ameisen Lichterfahrung und assoziieren m{\"o}glicherweise den Nesteingang mit spezifischen Landmarken oder werden anderen visuellen Informationen, wie denen des Polarisationsmusters, ausgesetzt und adaptieren begleitend ihre neuronalen Netzwerke an die bevorstehende Herausforderung. Dar{\"u}ber hinaus k{\"o}nnten die Pirouetten einer Stimulation der an der Polarisationsbahn beteiligten neuronalen Netzwerke dienen. Videoanalysen legen dar, dass Lichtaussetzung nach drei Tagen die Bewegungsaktivit{\"a}t der Ameisen heraufsetzt. Die Tatsache, dass die neuronale Umstrukturierung in visuellen Zentren wie auch die Ver{\"a}nderungen im Verhalten im selben Zeitrahmen ablaufen, deutet darauf hin, dass ein Zusammenhang zwischen struktureller synaptischer Plastizit{\"a}t und dem Verhaltens{\"u}bergang von der Innendienst- zur Fouragierphase bestehen k{\"o}nnte. Cataglyphis besitzen hervorragende visuelle Navigationsf{\"a}higkeiten, doch sie nutzen zudem olfaktorische Signale, um das Nest oder die Futterquelle aufzusp{\"u}ren. Mithilfe konfokaler Mikroskopie und 3D-Rekonstruktionen wurden potentielle Anpassungen der prim{\"a}ren olfaktorischen Gehirnzentren untersucht, indem die Anzahl, Gr{\"o}ße und r{\"a}umliche Anordnung olfaktorischer Glomeruli im Antennallobus von C. fortis, C. albicans, C. bicolor, C. rubra, und C. noda verglichen wurde. Arbeiterinnen aller Cataglyphis-Arten haben eine geringere Glomeruli-Anzahl im Vergleich zu denen der mehr olfaktorisch-orientierten Formica Arten - einer Gattung nah verwandt mit Cataglyphis - und denen schon bekannter olfaktorisch-orientierter Ameisenarten. C. fortis hat die geringste Anzahl an Glomeruli im Vergleich zu allen anderen Cataglyphis-Arten und besitzt einen vergr{\"o}ßerten Glomerulus, der nahe dem Eingang des Antennennerves lokalisiert ist. C. fortis M{\"a}nnchen besitzen eine signifikant geringere Glomeruli-Anzahl im Vergleich zu Arbeiterinnen und K{\"o}niginnen und haben einen hervorstechenden M{\"a}nnchen-spezifischen Makroglomerulus, welcher wahrscheinlich an der Pheromon-Kommunikation beteiligt ist. Die Verhaltensrelevanz des vergr{\"o}ßerten Glomerulus der Arbeiterinnen bleibt schwer fassbar. Die Tatsache, dass C. fortis Mikrohabitate bewohnt, welche von allen anderen Cataglyphis Arten gemieden werden, legt nahe, dass extreme {\"o}kologische Bedingungen nicht nur zu Anpassungen der visuellen F{\"a}higkeiten, sondern auch des olfaktorischen Systems gef{\"u}hrt haben. On the basis of the frequent involvement of the Ras-RAF-MEK-ERK signaling pathway in human non-small cell lung carcinoma (NSCLC), we have explored the target cell availability, reversibility, and cell type specificity of transformation by oncogenic C-RAF. Targeting expression to alveolar type II cells or to Clara cells, the two likely precursors of human NSCLC, revealed differential tumorigenicity between these cells. Whereas expression of oncogenic C-RAF in alveolar type II cells readily induced multifocal macroscopic lung tumors independent of the developmental state, few tumors with type II pneumocytes features and incomplete penetrance were found when targeted to Clara cells. Induced tumors did not progress and were strictly dependent on the initiating oncogene. Deinduction of mice resulted in tumor regression due to autophagy rather than apoptosis.
