However, the signaling cascade of costimulatory molecules has remained elusive. Here, I provide evidence that CD44 supports proliferation as well as apoptosis mainly, if not exclusively, by enhancing signal transduction via the TCR/CD3 complex. Blockade of CD44 interferes with mounting of an immune response. This has been demonstrated by the significantly decreased IL-2 production of a T helper line, when stimulated in the presence of a competing CD44 receptor globulin. To evaluate the underlying mechanism, CD44 was cross-linked by an immobilized antibody (IM7). Cross-linking of CD44 induces proliferation of peripheral T cells and apoptosis of thymocytes and a T helper line in the presence of subthreshold levels of anti-CD3. CD44-induced proliferation was accompanied by an upregulation of the activation markers CD25 and CD69 and an increased cytokine production. TCR-mediated apoptosis was accompanied by an upregulation of CD95 ligand and CD95 receptor, which could be greatly enhanced by costimulation via CD44. On the level of signal transduction, coligation of CD44 with CD3 resulted in a strong and sustained increase of early tyrosine phosphorylation events and upregulated downstream signal transduction pathways, such as the ras/ERK and the JNK signaling cascades. These pleiotropic effects of CD44 are due to its involvement in the most proximal events in TCR signaling, as demonstrated by a strong increase in the phosphorylation of the TCR z-chain and ZAP-70. Notably, cross-linking of CD44 was binding-site dependent and was only effective when supporting colocalization of the TCR/CD3 complex and CD44. Cross-linking of CD44 via immobilized IM7 also induced profound changes in cell morphology, characterized by strong adhesion, spreading and development of surface extensions, which were dependent on a functional tubulin and actin cytoskeleton. These cytoskeletal rearrangements were mediated by rac1, a small GTPase of the rho subfamily, and src-family kinases, two of which, fyn and lck, were found to be associated with CD44. By cross-linkage of CD44 these kinases were redistributed into so called lipid rafts. It is supposed that for T cell activation a relocation of the TCR/CD3 complex into the same membrane microdomains is required. The data are interpreted in the sense that the costimulatory function of CD44 relies on its cooperativity with the TCR. Most likely by recruitment of phosphokinases CD44 significantly lowers the threshold for the initiation of signaling via the TCR. The requirement for immobilized anti-CD44, the necessity for neighbouring anti-CD3 and the dependence on the binding site of CD44 strongly suggest that the costimulatory mechanism involves cytoskeletal rearrangements, which facilitate recruitment and redirection of src-family protein kinases in glycolipid enriched membrane microdomains. N2 - Es ist bekannt, dass die Aktivierung von T-Zellen zwei Signale erfordert, die Bindung des T-Zell-Rezeptors (TZR) an den Peptid-Haupthistokompatibilitätskomplex und die Bindung sogenannter kostimulatorischer Moleküle an ihren Liganden. Über diese Rezeptor-Liganden-Interaktionen wird eine Kettenreaktion von Signalen ausgelöst, die die Aktivierung einer ganzen Reihe von Genen bewirken. Die Signale die über die Liganden Bindung des TZR in Gang gesetzt werden, sind weitgehend bekannt. Hingegen ist unser Wissen über das Funktionsprinzip kostimulatorischer Moleküle äußerst lückenhaft. In der vorliegenden Arbeit wird aufgezeigt, dass und auf welche Weise CD44, eines dieser kostimulatorischen Moleküle, maßgeblich zur Verstärkung der über den TZR transduzierten Signale beiträgt. Ausgangspunkt der vorgelegten Studie war der Befund, dass eine Blockade von CD44, z.B. mittels eines kompetitiven löslichen CD44 Moleküls, die Antigen-induzierte Aktivierung einer T-Zelllinie blockieren kann. Zur Klärung des zugrunde liegenden Mechanismus wurde CD44 mittels immobilisierter Antikörper quervernetzt. Bei gleichzeitiger, suboptimaler Stimulation des T-Zell-Rezeptors induzierte die Quervernetzung von CD44 zum einen die Proliferation reifer T-Zellen, zum anderen den apoptotischen Zelltod von Thymozyten beziehungsweise einer T-Helferzelllinie. Erwartungsgemäß war die CD44-induzierte Proliferation mit der Expression sogenannter Aktivierungsmarker wie CD25 und CD69 und einer erhöhten Zytokinproduktion verbunden. Dementsprechend wurde bei CD44-induzierter Apoptose eine deutlich gesteigerte Expression des CD95 Moleküls und seines Liganden beobachtet. Die kostimulatorische Aktivität von CD44 war mit einer gesteigerten Tyrosinphosphorylierung und einer Hochregulation der ras/ERK und JNK Signalkaskaden verbunden. Da von allen beobachteten Veränderungen bekannt ist, dass sie auch über die Besetzung des TZR mit einem geeigneten Stimulus ausgelöst werden können, war es naheliegend anzunehmen, dass sich die kostimulatorische Funktion von CD44 auf eine Verstärkung der TZR-vermittelten Signale zurückführen läßt. Der Befund einer gesteigerten Phosphorylierung der TZR-z-Kette und der ZAP-70 Kinase, über die die Aktivierung von T-Zellen eingeleitet wird, unterstützt nachhaltig diese Hypothese. Hinzu kommen zwei weitere Befunde: 1. Die Quervernetzung von CD44 führt nur unter der Voraussetzung einer räumlichen Annäherung von CD44 und dem TZR zu einer Unterstützung der T-Zellaktivierung. Diese findet in sogenannten "Rafts" statt; 2. Die Quervernetzung von CD44 führt zu Adhäsion und Zellspreitzung, bedingt durch ein Rearrangement des Zytoskeletts an dem die GTPase rac1 und die src-Kinasen fyn und lck beteiligt sind. Diese beiden Kinasen sind konstitutiv mit CD44 assoziiert. Bei Quervernetzung des Moleküls und Reorganisation des Zytoskeletts wird eine deutliche Anreicherung von fyn und lck in den Rafts, also in direkter Nachbarschaft zum TZR, beobachtet. Diese Daten unterstützen nachhaltig meine Arbeitshypothese, dass sich die kostimulatorische Funktion von CD44 auf eine kooperative Aktivität mit dem TZR zurückführen läßt. Es ist wahrscheinlich, dass durch Rekrutierung der Kinasen lck und fyn der Schwellenwert für die Initiierung von Signalen über den TZR signifikant erniedrigt wird. Die Rekrutierung dieser Kinasen wird durch eine Reorganisation des Zytoskeletts, die mit der Einbringung dieser Kinasen in den Bereich der Rafts einhergeht, ermöglicht. Inwieweit die erhobenen Befunde zur kostimulatorischen Aktivität des Adhäsionsmoleküls CD44 im Sinne eines Verstärkungs-mechanismus eingeleitet durch räumliche Annäherung generelle Gültigkeit haben, kann noch nicht abschließend beantwortet werden. Die Mehrheit der in jüngster Zeit erhobenen Befunde zur Klärung des Funktionsprinzips kostimulatorischer Moleküle bestätigt jedoch das Prinzip nachbarschaftlicher Kooperation. Doch weist dieses Behandlungskonzept weiterhin einen wesentlichen Nachteil auf: noch immer wird der langfristige Erfolg der Therapie zu oft durch die so genannte Transplantatabstoßung gefährdet. Hierbei handelt sich um eine vom Organtransplantat ausgelöste Immunantwort, die zu dessen Zerstörung führt. Die derzeit einzige Möglichkeit eine Abstoßung zu verhindern, ist die Unterdrückung des Immunsystems mit so genannten Immunsuppressiva. Auch wenn diese erstmals in den 60er Jahren des 20. Jahrhunderts erfolgreich eingesetzten Medikamente ständig verbessert werden, bleiben die von ihnen ausgelösten Nebenwirkungen weiterhin ein ernstzunehmendes Problem. Sie unterdrücken die gesamte körpereigene Abwehr, was zum Schutz der Organtransplantate vor Abstoßung gewünscht ist, doch fördern sie hierdurch die Entstehung von Tumoren und Infektionen. Bei der Transplantatabstoßung handelt es sich um eine von CD4+ T-Lymphozyten ausgelöste Immunantwort. Diese Lymphozyten werden von allogenen Peptiden, die von Spender-MHC-Molekülen stammen, über den indirekten Weg der Alloantigenerkennung aktiviert. An der Transplantatabstoßung ist zwar eine Vielzahl von Alloantigenen beteiligt, doch ist es möglich, Peptidantigene zu identifizieren, die einen nachweisbaren Effekt auf die Transplantatabstoßung ausüben. So wurde in der eigenen Arbeitsgruppe die für die Abstoßung allogener Organtransplantate beteiligten MHC (RT1u)-Peptidantigene charakterisiert. Insbesondere die Bedeutung des aus 19 Aminosäuren bestehenden allogenen Peptids P1 für die Alloimmunantwort wurde intensiv untersucht. So weisen P1-spezifische T-Lymphozyten ein ausgeprägtes Th1-Cytokin-Muster auf und beschleunigen die Abstoßung von Wistar-Furth-Organtransplantaten in Lewis-Ratten. Das Ziel dieser Arbeit war die Charakterisierung des T-Zellrezeptor Vb-Repertoires P1-spezifischer T-Lymphozyten mit der Methode des PCR-ELISA. In einem ersten Schritt wurden Thymozyten und T-Lymphozyten unterschiedlicher Lymphknotenstationen untersucht. Thymozyten exprimierten alle 22 TCR Vb-Elemente und einzig TCR Vb14 war überrepräsentiert. Die T-Lymphozyten der cervikalen, mesenterialen, iliakalen und poplitealen Lymphknoten zeigten ebenfalls eine charakteristische Überexpression bestimmter TCR Vb-Elemente. So exprimierten cervikale T-Lymphozyten bevorzugt die TCR Vb-Elemente 2, 6, 8.3 und 16, mesenteriale T-Lymphozyten die TCR Vb-Elemente 2, 4, und 8.1, illiakale T-Lymphozyten die TCR Vb-Elemente 2 und 6 und popliteale T-Lymphozyten die TCR Vb-Elemente 2, 4 und 9. Die Immunisierung mit dem nicht-immunogenen Kontrollpeptid (Autoantigen) Ac führte zu einer leichten Veränderung des T-Zellrezeptor-Repertoires, bei der die TCR Vb-Elemente 14 und 16 überexprimiert waren. Das Adjuvant TiterMax beeinflusste kaum das TCR Vb-Repertoire. Die Immunisierung mit dem allogenen Peptid P1 führte zu einer eindeutigen Beeinflussung des T-Zellrezeptor-Repertoires. Popliteale T-Lymphozyten, die 7 Tage nach Immunisierung analysiert wurden, zeigten ein Repertoire, bei dem die TCR Vb-Elemente 15, 16, 17 und 20 überexprimiert waren. Dieses Repertoire war am Tag 3 nach Immunisierung noch nicht so ausgebildet. Wurden die antigenspezifischen T-Lymphozyten nach ihrer Isolierung mit P1 in vitro restimuliert, so waren in diesem T-Zellrezeptor-Repertoire die TCR Vb-Elemente 8.3, 15, 16 und 20 überexprimiert. Zum Vergleich: in naiven T-Lymphozyten waren die Vb-Elemente 2, 4 und 9 überexprimiert. Damit war es zu einer deutlichen Verschiebung im T-Zellrezeptor-Repertoire antigenspezifischer T-Lymphozyten gekommen, die auf das Peptidantigen P1 zurückzuführen ist. Mit der Methode des PCR-ELISA wurde das T-Zellrezeptor-Repertoire antigenspezifischer T-Lymphozyten bestimmt. Hiermit sind wesentliche Voraussetzungen geschaffen worden, um T-Zellklone zu etablieren und ihre Bedeutung für die Transplantatabstoßung genauer zu untersuchen. N2 - Organ transplantation is an important medical therapy for patients with irreversibly damaged organs. However, this therapeutic intervention has a great disadvantage because the long-term success of organ allografts is still too often endangered by the so called allograft rejection, an immune response directed to the allograft and leading to its destruction. The major approach for the prevention and management of allograft rejection is to suppress the immune system with immunosuppressive agents. These agents were introduced into transplantation medicine in the 1960s and since that time they have been continually improved. However, their side effects remain a seriousness problem because the suppression of the immune defence inhibits the allograft rejection on the one hand but causes infections and increases tumour incidence on the other hand. The allograft rejection is mediated by CD4+ T lymphocytes and the responsible antigens recognized by them are allogenic peptides processed from donor MHC molecules. Although a large number of allgenenic peptides are involved in allograft rejection, it is possible to identify certain peptide antigens involved in allograft rejection. In our group allogeneic peptides from MHC class I molecules from Wistar Furth rats were investigated. The significance of the allogeneic peptide P1, consisting of 19 amino acids, in inducing allograft rejection was analysed in detail. P1-specific T lymphocytes demonstrated a Th1-cytokine dominated profile and accelerated the rejection of allografts in Lewis rats donated by Wistar Furth rats. The aim of this study was to characterise the T cell receptor (TCR) Vb repertoire of P1-specific T lymphocytes with the PCR-ELISA technique. First, thymocytes and T lymphocytes from different lymph nodes were analysed. Thymocytes expressed all 22 TCR Vb elements and only TCR Vb14 was overrepresented. The T lymphocytes of cervical, mesenteric, iliacal und popliteal lymph nodes also demonstrated a certain repertoire of overrepresented TCR Vb elements. The adjuvant TiterMax did not influence the TCR Vb repertoire. In contrast, the immunisation with the allogeneic peptide P1 clearly influenced the T cell receptor repertoire. Popliteal T lymphocytes analysed 7 days after immunisation demonstrated a TCR Vb repertoire where the TCR Vb elements 15, 16, 17 und 20 were overrepresented. On day 3 after immunisation this repertoire was not yet clearly developed. In P1-specific T lymphocytes restimulated in vitro the expression levels of the TCR Vb elements 8.3, 15, 16 and 20 were increased. In comparison, in naïve T lymphocytes the TCR Vb elements 2, 4 and 9 were overrepresented. These results underline that the immunisation with peptide P1 induces a characteristic TCR Vb repertoire. The TCR Vb repertoire of P1-specific T lymphocytes was analysed with the PCR-ELISA technique. The determination of the T cell receptor repertoire is a prerequisite to establish T cell clones and to analyse their involvement in allograft rejection. Trotz Fortschritten in den Behandlungsmöglichkeiten und der medikamentöser Prophylaxe bleibt die Sterblichkeitsrate bei invasiven Erkrankungen hoch. Aus diesem Grund ist die Entwicklung von spezifischeren Immuntherapien von Nöten. Ein Ansatz ist die genetische Modifikation von T Zellen, durch den Transfer von A. fumigatus spezifischen T Zell Rezeptoren (TCRs), für eine adoptive Therapie. Um dieses Konzept zu evaluieren wurden TCRs, die für die extrazellulären Zellwandglykonase Crf1 (Crf1/p41) spezifisch sind, auf primäre T Zellen transferiert und die Effektor-Funktion analysiert. Das Crf1/p41 Epitop induziert bei gesunden Spendern eine funktionelle TH1 Immunantwort gegen A. fumigatus und führt zur Produktion hoher Mengen von Interferon γ (IFN-γ). Für die Identifikation von A. fumigatus spezifischen TCRs wurden siebenunddreißig Crf1/p41 spezifische T Zellklone von drei HLA DRB1*04 Spendern generiert. Anschließend wurden die TCR β Ketten über die sehr variable komplementaritätsbestimmende Region 3 (CDR3) bestimmt. Es konnten zwölf unterschiedliche TCRs ermittelt werden, von denen vor allem die variablen β (Vβ) Kette 18 sehr dominant, während die Vβ Ketten 1 und 6 nur in wenigen Klonen vertreten waren. Zur weiteren Charakterisierung der Crf1/p41 spezifischen TCRs wurden die variablen α (Vα) Ketten bestimmt (Vα 3, Vα 15 und Vα 26). Somit liegt eine polyklonale T Zell Immunantwort vor. Anschließend wurden die Crf1/p41 spezifischen TCRs in den retroviralen Vektor pMP71 kloniert und auf Jurkat 76 Zellen, welche keinen endogenen TCR exprimieren, und auf primäre CD4+ T Zellen transferiert. Die Expression von Crf1/p41 spezifischen TCRs, transduziert in CD4+ T Zellen, zeigten spenderspezifische Unterschiede und die Expression war niedriger im Vergleich zu den transduzierten Jurkat 76 Zellen. Daher wurde auf Optimierungsstrategien zurückgegriffen, die für den adoptiven Transfer mit TCR-modifizierten T Zellen zur Behandlung von Krebs entwickelt wurden. Angewandt wurden die Codonoptimierung der TCR codierenden Sequenz, Murinisierung der TCR konstanter Ketten, Induktion einer weiteren Disulfidbrücke. Ebenfalls wurde das Vektorsystem optimiert. Der Optimierungsprozess der Crf1/p41 spezifischen TCR 1 führte zu einer erhöhten Oberflächenexpression des TCR sowohl in Jurkat 76 (3 bis 5fach) als auch in primären CD4+ T Zellen (2fach). In funktionellen Analysen wurde die Proliferationsfähigkeit und IFN-γ Produktion, durch die Stimulation von transduzierten CD4+ T Zellen (TCR 1 optimiert) mit Crf1/p41 beladenen dendritischen Zellen (DCs), bestätigt. Diese Ergebnisse weisen darauf hin, dass der Transfer von A. fumigatus spezifischen TCRs eine protektive anti-fungale Immunantwort fördern könnte. Demzufolge auch als ein geeignetes Mittel in einer potentiellen Immuntherapie gegen A. fumigatus Infektionen in immunsupprimierten Patienten, eingesetzt werden könnte. N2 - The human pathogenic fungus Aspergillus fumigatus (A. fumigatus) can cause severe invasive infections in immunosuppressed patients. Despite progresses in the treatment and prophylaxis of invasive infections the mortality rate remains high. On that account the development of a more specific immune therapy seems necessary. One approach is genetic modification of T cells by transfer of A. fumigatus specific T cell receptors (TCR) for an adoptive therapy. To evaluate this concept, TCRs specific for a peptide derived from extracellular cell wall glucanase Crf1 (Crf1/p41) were transferred to primary T cells. Than their effector function was analyzed. In healthy donors the epitope Crf1/p41 induces a functional TH1 immune response towards A. fumigatus combined with a high Interferon-γ (IFN-γ) production. To identify A. fumigatus specific TCRs, thirty-seven Crf1/p41 specific T cell clones were generated from three HLA-DRB1*04 positive healthy donors. Afterwards, the TCR β chains were analyzed by sequencing the most variable complementarity determining region 3 (CDR3). Twelve different TCRs were detected whereas variable β (Vβ) 18 was very dominant, and Vβ 1 and Vβ 6 were present only in some clones. For further characterization of Crf1/p41 specific TCRs, variable α (Vα) chains were identified (Vα 3, Vα 15 and Vα 26). Thus, the T cell response is polyclonal. Subsequently Crf1/p41 specific TCRs were cloned into the retroviral vector pMP71 and transduced into Jurkat 76 cells that lack the expression of endogenous TCR, and into primary CD4+ T cells. The expression of Crf1/p41 specific TCR transduced in CD4+ T cells was donor-dependent and the expression was lower compared to transduced Jurkat 76 cells. Consequently, optimizing strategies engineered for TCR-modified adoptive T cell transfer in cancer therapy were used to induce a higher TCR expression on T cells. Those strategies include codon-optimization of the TCR coding sequence, murinization of constant chains of TCR, induction of an additional disulfide bond. Additionally the vector system was optimized. The optimization process of Crf1/p41 TCR 1 led to a higher surface expression in Jurkat 76 (3-5x) and primary CD4+ T cells (2x). Functional analyses revealed proliferation and IFN-γ production after the stimulation of transduced CD4+ T cells (TCR 1 optimiert) with Crf1/p41 pulsed mature dendritic cells (mDC). These data suggest that the transfer of A. fumigatus specific TCRs might foster protective anti-fungal immune responses. Therefore this might be a suitable tool for immunotherapeutic use against A. fumigatus infections in immunosuppressed patients. Ihr Hauptmerkmal ist die Expression eines semi-invarianten T Zellrezeptors (TCR), der die Bindung von CD1d:Glycolipid Komplexen ermöglicht, wohingegen ‚klassische‘ T Zellen an Komplexe aus MHC (Haupthistokompatibilitätskomplex) Molekülen und Peptiden binden. Die während der Reifung im Thymus durch Transkriptionsfaktoren festgelegte Voraktivierung der iNKT Zellen ermöglicht das unmittelbare Freisetzen von Cytokinen bei Antigenkontakt, wodurch iNKT Zellen die adaptive Immunantwort stark beeinflussen können: Sie tragen sowohl zur Regulation von Autoimmunerkrankungen als auch der Bekämpfung von Krebs und Infektionen bei. Der iNKT TCR setzt sich aus einer invarianten a-Kette (AV14/AJ18 in der Maus bzw. AV24/AJ18 im Menschen) und einer charakteristischen Auswahl an b-Ketten (vorwiegend BV8S2, BV7 und BV2 in der Maus und BV11 im Menschen) zusammen. Das Cerebrosid a-Galactosylceramid (aGC, KRN7000) stellt eines der potentesten Antigene für iNKT Zellen dar. Die Präsentation dieser Antigenklasse erfolgt durch CD1d Moleküle, die, abgesehen von tiefen hydrophoben Bindungstaschen, strukturell MHC I Molekülen ähneln, jedoch nicht polymorph sind und außerhalb des MHC Locus codiert sind. Die, zwischen Maus und Mensch hochkon- servierte, Interaktion von iNKT TCR und CD1d:aGC Komplex zeichnet sich bei potenten Antigenen durch die eingeschränkte Nutzung der Antigenspezifität bestimmenden Regionen aus: CDR1a, CDR3a und CDR2b. Die den CDR3b definierende V-D-J Umlagerung der b-Kette stellt im iNKT TCR den Bereich der höchsten Variabilität dar, beeinflusst jedoch nur die Bindung schwächerer Antigene. Natürlich auftretende Variabilität innerhalb der a-Kette kann durch Abweichungen von der kanonischen V-J Umlagerung am Beginn des CDR3a entstehen und beeinflusst ebenfalls die Bindung des iNKT TCR. Die iNKT Zellpopulation in F344 Ratten ähnelt in Frequenz und Korezepotorexpression derjenigen des Menschen. Ratten besitzen ein CD1D Gen, welches hoch homolog zu denen der Maus ist und zwei dem BV8S2 Gensegment der Maus homologe BV Segmente (BV8S2 und BV8S4), die in F344 Ratten beide funktionell sind. Eine Besonderheit der Ratte ist jedoch das Auftreten einer AV14 Multigenfamilie von bis zu zehn Gensegmenten. Diese unterscheiden sich neben dem HV4 vor allem in ihren CDR2 Sequenzen und werden anhand dieser Unterschiede in zwei Gruppen (Typ 1 und 2) eingeteilt. Zusätzlich wurde in der iNKT Zellpopulation eine hohe Frequenz an natürlich auftretenden A93G Substitutionen in der TCR↵ Kette beschrieben und es wurde gezeigt, dass, im Gegensatz zur Kreuzreaktivität zwischen iNKT TCR und CD1d von Maus und Mensch, iNKT Zellen der Ratte nicht an Maus CD1d binden. Die Besonderheiten des Ratten iNKT TCR und deren Auswirkungen auf die TCR Expression und Ligandenbindung der Ratten iNKT Zellpopulation wurden in der vorliegenden Arbeit untersucht. Durch in dieser Arbeit durchgeführte in vitro Mutagenesestudien konnten Position 68 in der vierten Hypervariablen Schleife (HV4↵) und Position 93 zu Be- ginn des CDR3↵ als entscheidende Modulatoren der CD1d Bindung im iNKT TCR von Ratte und Maus identifiziert werden, wobei auch speziesspezifische Unterschiede aufgedeckt werden konnten. Die Spezieskreuzreaktivität des Ratten iNKT TCR selbst hing stark von einer A93G Substitution im TCRa ab. Bei Untersuchungen der b-Kette zeigte sich, dass sowohl BV Segmente als auch CDR3b Region die Ligandenbindung in differenziellem Zusammenspiel beeinflussen, was bei Paarung mit unterschiedlichen AV14 Segmenten verschieden ausgeprägt sein konnte. Weiterhin wurden humane CD1d Dimere generiert und zum ersten Mal die Bindung von Ratten CD1d an humane iNKT TCR gezeigt. Weiterhin wurde in dieser Arbeit das TCR Repertoire von iNKT Zellen der F344 Ratte und deren CD1d Bindungseigenschaften charakterisiert. Hierzu wurde die bereits etablierte Methode der in vitro Expansion von iNKT Zellen aus der Rattenmilz weiterentwickelt, was die Langzeitkultur und -expansion der sortierten iNKT Zellpopulation ermöglichte. Bei Untersuchung der TCR Expression konnte gezeigt werden, dass die Auswahl der im Ratten iNKT TCR genutzten BV Gensegmente ähnlich limitiert ist wie in der Maus. Neben der dominanten Nutzung der BV8S4 und BV8S2 Gensegmente wurden hauptsächlich BV8S1, BV14 und BV7 gefunden. Bei Untersuchungen der CD1d Dimerbindung der iNKT Zellpopulation konnte der Einfluss der na- türlich auftretenden A93G Substitution in der iNKT TCRa Kette bestätigt werden. Außerdem zeigte sich hier ebenfalls der Einfluss des BV Gensegments auf die Ligandenbindung, wobei BV8S4 negative Zellen im Vergleich zu BV8S4 positiven Zellen eine stärkere Ratten CD1d Dimerbindung zeigten. N2 - Type 1 NKT cells, also called iNKT cells (invariant Natural Killer T cells), are a subpopulation of abT cells with characteristic features. CD1d molecules resemble MHC I structurally but possess deep hydrophobic pockets as antigen binding grooves. The iNKT TCR interaction with CD1d:aGC complexes is highly conserved between mice and humans. Its binding mode is characteristic for the restricted CDR (complementarity determining region) usage and relies mostly on CDR1a, CDR3a and CDR2b. The only highly variable region within the iNKT TCR, the CDR3b defining diverse V-D-J rearrangement of the TCRb chain, has been shown to only influence binding to less potent antigens. Natural variability within the TCRa chain is usually only possible due to non canonical AV14/AJ18 rearrangements at the beginning of CDR3a and has been shown to indirectly influence ligand binding. The iNKT cells in F344 rats resemble human iNKT cells in terms of frequency and coreceptor expression profile. Rats have one CD1D gene, highly homologous to those found in mice, and two BV8S2 homologs (BV8S2 and BV8S4 ), which are both functional in F344 rats. Peculiar for the rat is the presence of an AV14 gene family with up to ten members, which have been grouped by their CDR2↵ sequences into Type 1 and 2, and a high frequency of A93G substitutions within the iNKT TCRa chains. Additionally, in contrast to the species cross-reactive interaction of iNKT TCR and CD1d from mice and humans, a lack of mouse CD1d binding by the rat iNKT TCR has been demonstrated. Those characteristic features of the rat iNKT TCR and its influence on TCR expression and ligand binding of rat iNKT cells were investigated in this study. Due to in vitro mutagenesis studies conducted in this thesis, position 68 within the fourth hypervariable loop of the TCRa chain (HV4a) as well as position 93 at the start of CDR3a could be identified as strong modulators of CD1d binding within the iNKT TCR of rat and mouse with specific features for both species. Additionally, the TCR repertoire of F344 rat iNKT cells and its ligand binding has been characterized in this study. For this purpose, the already established in vitro expansion of rat iNKT cell from splenocytes was further developed, allowing long term culture and expansion of purified iNKT cell populations. Analysis of the TCR expression did reveal a restricted BV segment usage, similar to mouse iNKT cells. Besides the predominant usage of BV8S2 and BV8S4 gene sements, mainly BV8S1 BV14 and BV7 were found. In these models, a preferential activation of regulatory T cells was observed by in vivo administration of CD28SA. In stark contrast, human volunteers receiving TGN1412, a humanized CD28-specific mAb, experienced a life-threatening cytokine release syndrome during the first-in-man trial. Preclinical tests employing human peripheral blood mononuclear cells (PBMC) failed to announce the rapid cytokine release measured in the human volunteers in response to TGN1412. The aim of this thesis project was to find an explanation of why standard PBMC assays failed to predict the unexpected TGN1412-induced "cytokine storm" observed in human volunteers. CD28 superagonists can activate T cells without T cell receptor (TCR) ligation. They do depend, however, on “tonic” TCR signals received by MHC scanning, signals that they amplify. PBMC do not receive these signals in the circulation. Short-term in vitro preculture of human PBMC at a high cell density (HDC) resulted in massive cytokine release during subsequent TGN1412 stimulation. Restoration of reactivity was cell-contact dependent, associated with TCR polarization and tyrosine-phosphorylation, and blocked by HLA-specific mAb. In HDC, both CD4 T cells and monocytes functionally mature in a mutually dependent fashion. However, only CD4 memory T-cells proliferate upon TGN1412 stimulation, and were identified as the main source of pro-inflammatory cytokines. Importantly, responses to other T-cell activating agents were also enhanced if PBMC were first allowed to interact under tissue-like conditions. A new in vitro protocol is provided that returns circulating T-cells to a tissue-like status where they respond to TGN1412 stimulation, and it might represent a more reliable preclinical in vitro test for both activating and inhibitory immunomodulatory drugs. Finally, the surprising observation was made that the IgG1 “sibling” of TGN1412, which is of the poorly Fc receptor-binding IgG4 isotype, has a much lower stimulatory activity. We could exclude steric hindrance as an explanation and provide evidence for removal of TGN1112 from the T-cell surface by trans-endocytosis. In diesen Modellen konnte nachgewiesen werden, dass CD28SA-Injektionen zu einer verstärkten Aktivierung regulatorischer T-Zellen in führen. Entgegen diesen Beobachtungen im Tiermodell reagierten die Teilnehmer einer ersten klinischen Studie auf die Administration des humanisierten CD28SA TGN1412 mit einer akut lebensbedrohlichen systemischen Zytokinausschüttung. Vorklinische Studien an humanen mononukleären Zellen des Blutes (PBMC) hatten keinen Hinweis auf eine mögliche plötzliche Zytokinausschüttungen als Reaktion auf TGN1412 gegeben. In der vorliegenden Arbeit wurde versucht eine Erklärung zu finden, warum PBMC-basierte Tests, wie sie vorklinischen Studien als Standard eingesetzt werden, nicht auf den unerwarteten TGN1412-induzierten „Zytokinsturm“ der Probanden hinwiesen. CD28SA aktivieren T-Zellen ohne Ligation des T-Zell Rezeptors (TCR). Jedoch werden zur CD28SA-abhängigen Aktivierung von T-Zellen „tonische“ TCR Signale benötigt, die durch MHC Scanning der T-Zellen an der Oberfläche anderer Zellen erzeugt werden. PBMC, welche sich in der Zirkulation befinden, erhalten diese „tonischen“ TCR Signale nicht. Kurzzeitige Vorkultur humaner PBMC bei hoher Zelldichte (high-density culture, HDC) führte zu einer starken Zytokinantwort bei nachfolgender TGN1412 Stimulation. Diese wiedererlangte Reaktivität gegenüber TGN1412 ging mit Tyrosin-Phospholrylierung sowie der Polarisierung von TCR Molekülen einher, war abhängig von Zellkontakten und konnte durch HLA-spezifische mAbs geblockt werden. Während der HDC durchlaufen sowohl CD4 T-Gedächtniszellen, als auch Monozyten eine voneinander abhängige funktionelle Reifung. TGN1412-induzierte Zellproliferation beschränkt sich jedoch auf CD4 T-Gedächtniszellen, die auch die Hauptquelle der proinflammatorische Zytokine sind. Antworten auf weitere T-Zell aktivierende Agenzien waren ebenfalls erhöht, wenn PBMC zunächst auf gewebeartige Bedingungen zurückgesetzt wurden. Die vorliegende Arbeit beschreibt damit ein neuartiges in vitro Protokoll für humane PBMC, welches T-Zellen der Zirkulation in einen gewebeartigen funktionellen Status versetzt, in welchem sie auf TGN1412 antworten. Die Fähigkeit, Toleranz für sich selbst und auch für mittransplantierte Organe über die MHC Barriere hinweg zu induzieren, ist einzigartig. Dies steht im Gegensatz zum zentralen Dogma der Transplantationsimmunologie, wonach MHC differente Organe nach Transplantation ohne Immunsuppression irreversibel zerstört werden. Ziel der Arbeit war, die verschiedenen intrahepatischen CD4+ T-Lymphozyten durchflusszytometrisch zu charakterisieren und eine in vitro Aktivierung naiver Leukozyten mit der in vivo zu vergleichen. Dies wurde mit der Fragestellung verknüpft, ob es einen möglichen Phänotyp regulatorischer intrahepatischer T-Lymphozyten gibt, der für die Induktion von Spontantoleranz verantwortlich ist. Auch wurde das Zytokinmuster dieser intrahepatischen T-Lymphozyten bestimmt, um eine mögliche funktionelle Aussage treffen zu können. Für die Versuche wurden Lebertransplantationen von Lewis-Spendertieren (LEW) auf Dark Agouti (DA) Empfänger durchgeführt. Die Transplantate wurden dauerhaft (> 300 Tage p.op.) spontan, d.h. ohne Immunsuppression toleriert. Andere vaskularisierte Organe, wie z.B. Herzen, wurden von DA-Empfängern abgestoßen. Die intrahepatischen und Milz T-Lymphoyzten wurden an den Tagen 3, 30 und 100 nach Transplantation untersucht. Unmittelbar nach Lebertransplantation kam es zu einem massiven Anstieg der intrahepatischen Leukozyten. Das Leberinfiltrat wurde dabei von aktivierten CD4+ T-Lymphozyten dominiert (ca. 23 Prozent CD4+CD45RCneg T-Lymphozyten), von denen ca. 50 Prozent den IL-2 Rezeptor exprimierten. Als Zeichen einer intrahepatischen Immunantwort wurde gewertet, dass das Verhältnis aktivierter CD4+CD45RCneg zu ruhenden CD4+CD45RCpos T-Lymphozyten dynamisch war. Zur funktionellen Charakterisierung der verschiedenen Populationen an CD4+ T-Lymphozyten wurden die Zytokinmuster dieser Zellen mit zwei verschiedenen Verfahren bestimmt. Zum einen wurden die sezernierten Zytokine INF-gamma, TNF-alpha, IL-10 und IL-13, zum anderen deren spezifische mRNAs semiquantitativ nachgewiesen. Für die sezernierten Zytokine waren keine wesentlichen Unterschiede zwischen den beiden Subpopulationen nachzuweisen. Bei der Analyse der mRNA-Transkription zeigte sich jedoch, dass die CD4+CD45RCneg T-Lymphozyten stärker die Th2-Zytokine IL-10 und IL-13 exprimierten, während die CD4+CD45RCpos T-Lymphozyten stärker die Th1-Zytokine INF-gamma, TNF-alpha exprimierten. In dieser Arbeit wurde zudem gezeigt, dass die CD4+CD45RCpos und CD4+CD45RCneg T-Lymphozyten jeweils heterogene Zellpopulationen sind, die noch in CD4posCD45RCpos, CD4hochposCD45RChochpos, CD4posCD45RCneg und CD4hochposCD45RCneg T-Lymphozyten differenziert werden können. Die CD4hochposCD45RChochpos repräsentieren dabei mit großer Wahrscheinlichkeit naive T-Lymphozyten, wohingegen die CD4hochposCD45RCneg aktivierte T-Lymphozyten darstellen. Wir gehen davon aus, dass die CD4posCD45RCpos T-Lymphozyten auch ruhende Gedächtniszellen und die CD4posCD45RCneg T-Lymphozyten auch aktivierte Gedächtniszellen enthalten. Der Nachweis intrahepatischer Gedächtnis T-Lymphoyzten zeigt, dass die Leber immunologisch ein überaus interessantes Organ repräsentiert. Furthermore, liver grafts may induce immunological tolerance to a subsequent heart, kidney or skin graft from the same donor. Without liver grafts these organs are destroyed by the recipient’s immune system. The survival of liver grafts is, however, not related to a failure of the recipient to mount an immunological response against donor MHC molecules. An immune-mediated damage of the transplanted livers with an accompanying infiltrate and up-regulation of pro-inflammatory cytokines is temporarily observed. This is in contrast to the well-established maxim of transplantation immunology that rejection will occur when donor and recipient transplantation antigens are mismatched, especially those encoded by MHC. The liver contains a large population of resident and migratory T lymphocytes. Since T lymphocytes mediate both rejection and tolerance, the aim of the study was to phenotype intrahepatic CD4+ T lymphocytes by flow cytometry. In addition, secreted cytokines as well as cytokine-specific mRNA were analysed. With these investigations we wanted to identify an intrahepatic T cell subpopulation with regulatory properties. For the orthotopic liver transplantation we used rats of the inbred strains Lewis, as donors, and Dark Agouti, as recipients. The liver grafts in this strain combination survived permanently (>300 days), although heart grafts from the same donor were rejected. T lymphocytes isolated from liver grafts and spleens were analysed on days 3, 30, and 100 after transplantation. The amount of immune cells from the recipient increased dramatically in liver allografts within 3 days after transplantation. With approximately 23% activated CD4+ CD45RCneg dominated this infiltrate and the ratio of activated CD4+CD45RCneg and rested CD4+CD45RCpos increased. This observation was interpreted as an intrahepatic immune response mediated by infiltrated immune cells. The presence of the cytokines INF-gamma, TNF-alpha, IL-10 und IL-13 was analysed in the supernatant of isolated intrahepatic T incubated for 3 days in vitro. Cytokine-specific mRNA was semiquantified by ELISA-PCR. The supernatants did not demonstrate any differences between both subpopulations. In contrast, the results of the ELISA-PCR revealed increased mRNA levels of the Th2 cytokines IL-10 and IL-13 for CD4+CD45RCneg T lymphocytes and increased levels of the Th1 cytokines INF-gamma and TNF-alpha for CD4+CD45RCpos T lymphocytes. In the present study it was also shown that both populations of CD4+CD45RCpos (rested) and CD4+CD45RCneg (activated) T lymphocytes were heterogeneous and divisible into CD4posCD45RCpos, CD4strong pos CD45RCstrong pos and CD4pos CD45RCneg, CD4strong posCD45RCneg T cells by flow cytometry. The CD4strong pos CD45RCstrong pos T cells may also represent naïve T cells, whereas the CD4strong pos CD45RCneg may be a certain type of activated T cells. We have evidence that rested memory T lymphocytes are located within the population of CD4pos CD45RCpos T lymphocytes and activated memory T lymphocytes are present within the population of CD4pos CD45RCneg T lymphocytes. The proof of intrahepatic memory T cells indicates that the liver represents an immunologically important organ. Its ability to induce spontaneous tolerance in many fully mismatched donor recipient combinations is unique and additional studies are necessary to further explore the true potential of intrahepatic T cells for tolerance induction. Die Pathogenese dieser Krankheiten ist bis jetzt noch nicht bekannt. Ziel dieser Arbeit war es, die ins Zielgewebe eingedrungenen autoaggressiven T-Lymphozyten aus gefrorenem Biopsiegewebe zu isolieren, ihren ab-T-Zell-Rezeptor zu analysieren und diesen rekombinant zu exprimieren. Folgeversuche mit den TZR-Transfektanten sollten erste Hinweise auf ein mögliches Antigen liefern. Um autoaggressive T-Zellen von irrelevanten zu unterscheiden, konzentrierten wir uns auf Zellen, die zum einen im Zielgewebe klonal expandiert vorlagen, zum anderen einen direkten morphologischen Kontakt mit der Zielzelle aufwiesen. Wir untersuchten das T-Zell-Repertoire verschiedener Patienten auf klonal expandierte Populationen durch CDR3-Spektratyping und isolierten positiv gefärbte Kandidatenzellen durch Lasermikrodissektion aus dem Gewebe. Anschließend wurden die T-Zell-Rezeptoren mit einer speziellen, im Rahmen der Arbeit entwickelten Einzelzell-Muliplex-RT-PCR analysiert. Bisher war es nicht möglich, die a- und b-Kette desselben T-Zell-Rezeptors aus Biopsiematerial zu identifizieren. Dies lag an der geringen Anzahl verfügbarer anti-a-Ketten-Antikörper sowie an der wesentlich höheren Variabilität der a-Ketten-Gene. Aus dem Biopsiegewebe eines Patienten isolierten wir 23 ab-TZR-Pärchen, welche alle die identische expandierte TZR-b-Kette zeigten. 20 der 23 Zellen wiesen eine identische a-Kette auf. Die Dominanz des TZR ist ein Hinweis auf die pathogene Relevanz dieses T-Zell-Klons. Die anderen drei Zellen zeigten drei unterschiedliche funktionelle a-Ketten. Aus der Biopsie eines zweiten Patienten isolierten wir zwei weitere ab-TZR-Pärchen. Die vier Rezeptoren des ersten Patienten wurden in einer T-Zell Hybridom Zellinie exprimiert. Vorläufige Versuche, ein mögliches Antigen zu detektieren, zeigten, dass es sich wahrscheinlich weder um ein muskelspezifisches Antigen noch um ein ubiquitär exprimiertes Selbst-Antigen handelt. Die hier beschriebene Methode der Isolierung und Analyse von autoaggressiven T-Lymphozyten kann man nicht nur zur Untersuchung des TZR-ab-Repertoires von Myositis- oder MS-Patienten einsetzen. Sie ist ebenfalls geeignet, andere Autoimmunerkrankungen sowie Tumor- oder Infektionserkrankungen zu untersuchen. N2 - Multiple sclerosis and the inflammatory muscle diseases polymyositis and inclusion body myositis are autoimmune diseases. T cells invade and destroy brain tissue or muscle fibers, respectively. The pathogenesis of these diseases is still unknown. The aim of this thesis was to isolate tissue invading autoaggressive T-lymphocytes from frozen biopsy specimens, to analyse their T cell receptor a-and b-chains and to express the receptors in a recombinant expression system. This may provide a tool to search for possible antigens. To distinguish pathogenic from irrelevant bystander cells we used two criteria: the clonal expansion in the target tissue triggered by antigen induced proliferation and the direct morphological contact with a target cell. We analysed the T cell repertoire of different patients by CDR3-spectratyping to find clonal expanded populations and isolated immunohistochemically stained candidate cells by laser microdissection. Then we identified coexpressed pairs of a- and b-chains by a newly developed multiplex PCR protocol. The detection of matching a- and b-chains in histological sections has not been achieved until now. The a-chain analysis has been a great problem because of the paucity of available a-chain antibodies and the great variability of the a-chain genes. From the tissue of one patient we isolated 23 ab-T cell receptor pairs which expressed the same expanded T cell receptor b-chain. 20 of these 23 a-chains were identical, suggesting a pathogenic relevance of this T cell clone. The a-chains of the three other T cells were different. We characterized two further clones from another patient. The receptors from the first patient were expressed in a T hybridoma cell line. Initial studies to identify possible antigens revealed that the T cell receptors did not recognize a muscle specific antigen nor a widely expressed self-antigen. This method for the analysis and reconstruction of tissue-infiltrating ab-T cells can be applied to many other autoimmune diseases as well as tumors or infectious diseases. The knowledge of individual dose is a prerequisite for planning a medical treatment and for health risk evaluations. In the present work two biodosimetrical assays were calibrated in young patients who were treated with radioiodine for thyroid cancer. Patients were from Belarus. They suffered from radiation induced thyroid cancer as a consequence of the Chernobyl reactor accident. In radioiodine therapy (RIT) bone marrow and lymphatic organs are exposed to ionizing radiation at doses of 0.1 to 0.75 Sv within about 2 days. Since several RIT have to be applied with interval between each of them from 6 months up to approximately 1 year, total dose can be up to 2 Sv within 2 to 3 years. The dose for thyroid tissue is approximately 1000 times higher. The dose-response relationship was measured by the T-cell receptor test (TCR test) in T4 lymphocytes with and without in vitro incubation or by the micronucleus assay in transferrin receptor positive reticulocytes (MN-Tf-Ret test). In all these assays, the frequency of radiation-induced mutants of blood cells is measured using flow cytometry. The TCR test is a cumulative biodosimeter, which measures the total radiation dose within the last 5 to 10 years, whereas the result of the MN-Tf-Ret test reflects the radiation dose of approximately 24 hours interval. It takes 8 hours and 3 days to perform TCR and MN-Tf-Ret tests respectively. Calibration curves based on radioiodine treated patients can be used for dose estimation in humans, if the radiation conditions correspond to those in RIT. This limits their applicability to low dose-rate β- and γ-irradiation and to doses per session not higher than about 0.5 Sv. If higher doses or dose-rates as well as the other types of ionizing radiation are involved, calibration curves in animals are indispensable. In the case MN-Tf-Ret test mouse models are established and may be used. The TCR assay was performed in 72 thyroid cancer patients aged between 14 and 25. T-cell mutant frequency (Mf) reaches its maximum only after half a year following the RIT. Then it declines exponentially. This decline could be described by the 3 parameter single exponential decay function. Based on this equation, the radiation dose could be calculated when the Mf and the time interval since exposure are known. Furthermore, the experimentally measured Mf value, which significantly exceeds the corresponding calculated Mf value would indicate an individual with higher radiosensitivity. However, among our patients there were none. The reticulocytes micronuclei test (MN-Tf-Ret) was performed in 46 radioiodine treated patients. When measuring the MN frequency (f(MN-Tf-Ret)) the measured cell fraction should be limited only to the youngest cohort of reticulocytes, because all the micronucleated erythrocytes are quickly removed from the peripheral blood by spleen. Thus, the MN test was performed only in CD71 positive (having transferring receptor) reticulocytes. These reticulocytes just entered the peripheral blood flow from red marrow. The MN frequency was measured before the therapy and then every day after the irradiation until day 7. MN frequency curve has typical shape with latent period for days 0 to 3. Then there is a sharp increase in MN frequency which lasts for 24 hours and could start between days 3 and 4. In the following days the MN frequency is dropping to its base level that equals the one before the treatment. The decay of MN frequency is depending on the half-life of radioiodine in the patient organism. If the half-life is low, then the increased f(MN-Tf-Ret) lasts shorter and vice versa. It was shown that the MN frequency curve could be described by the model where all the micronuclei arise only through the last mitosis of erythroblasts in the red marrow and the MN frequency is proportional to the radiation dose in the last cell cycle. The shape of this curve depends on the cell kinetics of erythropoiesis on one side and the exponential decay of radioiodine activity on the other. To the best of our knowledge, the MN-Tf-Ret test was applied in the present study for the first time in biological dosimetry. N2 - Einleitung: Eine nicht adäquate Funktion des Immunsystems ist ein großes klinisches Problem, welches Chemotherapien durch schwere bakterielle oder mykotische Infektionen komplikationsreich macht. Während einer Immunantwort spielen Zytokine, die von T-Zellen produziert werden, eine wichtige Rolle für die Effektivität der Antwort. Um zu verstehen, welche Veränderungen während der Therapie im Immunsystem auftreten, untersuchten wir die Subpopulationen und Funktionen (Zytokine , Immunglobuline) der Lymphozyten. Patienten: 23 Kinder (medianes Alter 5y; 2m-14y; 15 weiblich, 8 männlich) mit B-ALL behandelt nach dem ALL-BFM 2000-Protokoll. Blutproben wurden gesammelt bei Diagnosestellung und an den Tagen 8, 15, 33, 64 sowie vor Protokoll M, vor Protokoll 2 und während der Erhaltungstherapie. Methoden: Wir analysierten die Lymphozyten-Subpopulation mittels Durchflußzytometrie. Die Bestimmung intrazelluläre Zytokine (IFNγ, IL2, TNFα, IL4, IL5, and IL10) erfolgte durch FACS-Analysen nach in vitro Stimulation mit PMA, Ionomycin und Brefeldin für 24h. Zusätzlich untersuchten wir verschiedene Zytokine im unstimulierten Serum mittels ELISA und studierten TRECs und Spectratypes sowie die Immunglobulinlevels. Ergebnisse: Die B-Zellen verringerten sich schnell von einem Medianen Wert von 301+120/mm³ vor Chemotherapie auf 85+138/mm³ am Tag 33 und stiegen nicht mehr bis zum Ende der Therapie an. Die T-Zellzahl fiel zu Beginn der Therapie ab, allerdings konnten wir partielle Erholungen, die proportional zur Therapieintensität waren detektieren. Zudem fiel eine Verschiebung zugunsten der CD8+-Zellen auf. NK-Zellen zeigten keine signifikanten Veränderungen. Bei den von CD3+ -Zellen produzierten Zytokinen fiel eine Expressionssteigerung von IFNγ auf. Wir konnten eine Korrelation zwischen γδ-TCR und IFNγ -Produktion(FACS) sowie IFNγ -Werte(ELISA) und hohe Anzahl von Gedächtniszellen finden. Außerdem korrelieren CD45RA+Zellen mit CD4IL2+Zellen. TGFβ im unstimulierten Sera korrelierte signifikant (p<0,01) mit CD19+Zellen. TGFβ wurde auch von Blasten exprimiert. Wir stellten Unterschiede im Zytokinprofil zwischen dem mit Dexamethason bzw Prednison behandelten Patienten fest; insbesondere die IFNγ-Sekretion war sehr viel größer unter Prednisonbehandlung (p<0,01). TREC-Werte waren höher unter Dexamethason, aber das mag mit beeinflusst sein durch das Alter, da jüngere Kinder signifikant höhere TREC-Werte haben. Bei der Analyse des TZR-Repertoire zeigte sich eine höhere Komplexität im Prednisonzweig. Wir konnten Vβ-Genfamilien mit höherer Komplexität (BV22, BV23) und mit niedrigerer Komplexität (BV13b, BV6a) detektieren. Die Komplexität des TZR korrelierte positiv mit der Anzahl der naiven T-Zellen und dem Alter(p<0,01). Bis d15 nahm die Komplexität des Repertoires ab und erholte sich langsam im Therapieverlauf. Zusammenfassung: Wir detektierten eine Verschiebung zu Gunsten der TH1-Zytokine. B-Zellen wurden durch die Therapie ausgelöscht. Dieses Ergebnis mag eine klinische Relevanz haben in der prophylaktischen Gabe von Immunglobulinen, um schwere Infektionen durch das Fehlen der B-Zellen zu vermeiden. N2 - Multidrug chemotherapy is a highly effective treatment for paediatric acute lymphoblastic leukaemia (ALL), but at the same time compromises immunity of patients. Immune function in a homogenous cohort of 23 children with standard- and intermediate-risk ALL was analysed by immunophenotyping, intracellular cytokine staining, assessment of serum cytokine concentrations, T-cell receptor (TCR) repertoire diversity and thymic function. B-cells were most severely affected by chemotherapy, rapidly declined under induction and did not recover until the cessation of maintenance therapy. This recovery was paralleled by a relative increase in naive IgM(+)IgD(+)CD27(-) B-cells, indicating de novo B-cell generation as the major pathway for B-cell reconstitution. T- and Natural Killer-cells were less severely affected. Although numerically diminished by chemotherapy, they had partially recovered at the end of induction. Interestingly, CD4:CD8 ratio, distribution of naive versus memory T-cells, cytokine production, TCR-repertoire complexity and thymic function were all only marginally affected by chemotherapy. Patients receiving dexamethasone had significantly less IFNgamma(+) T-cells than those receiving prednisone. Our data show that during chemotherapy in standard- and intermediate-risk paediatric ALL patients the T-cell system remains relatively well preserved. Future studies will show if this effect can be exploited for inclusion of immunotherapy in standard ALL treatment protocols.
