Das Krankheitsbild reicht von leichten Hautinfektionen bis hin zu lebensbedrohlichen Infektionen wie Endokarditis, Sepsis oder Pneumonien. S. aureus ist ein Haupterreger nosokomialer Infektionen. Besonders die Antibiotikaresistenzentwicklung von S. aureus–Stämmen ist problematisch. Als wirksame Antibiotika können zur Zeit oft nur noch Vancomycin, Synercid oder Linezolid zur Therapie eingesetzt werden. Die alarmierende Resistenzentwicklung in S. aureus verdeutlicht, dass die Entwicklung neuer Antibiotika und die Identifizierung neuer bakterieller Angriffsstrukturen dringend erforderlich ist. Gängige antiinfektive Therapeutika sind gegen die bakterielle Zellwandsynthese, den DNA- und RNA-Stoffwechsel oder die Proteinbiosynthese gerichtet. In dieser Arbeit sollten Virulenz-relevante Zielstrukturen für die Entwicklung neuer Antibiotika untersucht werden. Insgesamt wurden sieben Gene analysiert, von denen vier zu Anfang dieser Arbeit in S. aureus noch nicht charakterisiert waren. Die Zielgene (clpP, purH, ssrA und smpB) in S. aureus sollten deletiert werden, um ihre Überlebensnotwendigkeit in vitro- und in vivo zu überprüfen. Eine Deletion gelang bei den Genen clpP und purH, die somit als nicht essenziell in S. aureus zu betrachten sind. Die bereits zuvor als nicht-essenziell charakterisierten Gene arlR, arlS und putP wurden deletiert und die Mutanten dclpP, darlR, darlS, dpurH und dputP wurden phänotypisch in Hinsicht auf ihren Einfluss auf die Pathogenität in S. aureus analysiert. Die differenzielle Genexpression der Mutanten dclpP und darlR wurde mit Hilfe von Microarray-Hybridisierungsexperimenten untersucht. Die ∆clpP-Mutante zeigte einen starken Wachstumsdefekt bei verschiedenen Temperaturen (30, 37, 42°C) und war nicht mehr in der Lage bei 20°C zu wachsen. Ebenso war das Wachstum unter anaeroben Bedingungen stark beeinträchtigt. Der Stamm dclpP wies eine verringerte hämolytische Aktivität sowie eine verminderte Adhärenz an Polystyren auf. Außerdem konnte eine stark erhöhte autolytische Aktivität in einem Triton X-100-Assay beobachtet werden. In einem Invasions-Zellkulturassay mit 293T-Epithelzellen konnte eine ~10-fach erhöhte Invasivität im Vergleich zu dem isogenen Wildtyp festgestellt werden. Die Komplementierung der ∆clpP-Mutante durch Einführung eines clpP-Expressionsvektors führte nahezu bei allen getesteten Bedingungen zur Wiederherstellung des wildtypischen Phänotyps. Die Transkriptomanalyse der dclpP-Mutante ergab eine deutliche Veränderung in der Genexpression (15 % aller Gene). Eine computerunterstützte Analyse der Upstreambereiche der deregulierten Gene führte zu der Identifizierung verschiedener Regulons, die bei der bakteriellen Antwort auf verschiedene Stressbedingungen eine Rolle spielen. Die clpP-Deletion betrifft besonders Regulatoren, deren Aktivität in Abhängigkeit zu veränderten Redox-Bedingungen reguliert wird, wie z. B. verschiedenen Stressbedingungen und Anaerobiose. Die Konstruktion der darlR- und darlS-Mutanten führte zu einer gesteigerten hämolytischen Aktivität, einer erhöhten Adhärenz an Polystyren sowie einer erhöhten autolytischen Aktivität in Triton X-100-Assays. Die Internalisierungsrate durch 293T-Epithelzellen war vermindert. Die darlR-Mutante wurde in einem Katheter-assoziierten Infektionsmodell in Ratten eingesetzt. Die kompetitive Infektion mit Mutante und Wildtyp ergab einen deutlichen Nachteil bei der Etablierung einer Infektion durch die Mutante. Die Transkriptomanalyse der 8325darlR-Mutante in der exponenziellen und in der stationären Phase unterstreicht den großen Einfluss des ArlRS-Zwei-Komponenten-Systems auf die Regulation der Genexpression in S. aureus. In der exponenziellen Phase wurden insgesamt 5 % und in der stationären Phase 15 % der Gene differenziell exprimiert. dpurH- und dputP-Mutanten wiesen in vitro keine Veränderungen im Wachstums-verhalten, der Biofilmbildung oder hämolytischen Aktivität auf. In einem Infektionsmodell in Ratten führte die Deletion von purH in dem S. aureus-Stamm MA12 zu einer signifikanten Verminderung der Virulenz. Die Herstellung von smpB- und ssrA-Deletionsmutanten verlief ohne Erfolg. Es wurde versucht, einen direkten Nachweis für den essenziellen Charakter dieser Gene durch den Einsatz konditional letaler Expressionssysteme zu erbringen. Weder der Austausch des wildtypischen durch einen regulierbaren Promotor noch eine Antisense-RNA-Strategie war für eine eindeutige Klärung dieser Frage ausreichend. Es konnte durch diese Arbeit jedoch gezeigt werden, dass die Antisense-RNA-Strategie eine Beeinträchtigung des Wachstums von S. aureus bewirkt. N2 - Staphylococcus aureus is an important opportunistic pathogen that can cause a wide range of diseases in humans and animals. The outcome of an infection can range from mild skin infections to life-threatening infections, like endocarditis, bacteraemia and pneumonia. Currently, methicillin-resistant S. aureus strains (MRSA) that have acquired numerous additional resistances towards several antibiotics have established in the hospital-setting. Therefore, the treatment of hospital-acquired infections with S. aureus in a rising number of patients is becoming increasingly complicated. The remaining effective antibiotics to treat infections with multi-resistant MRSA are vancomycin, synercid, and linezolid. However, MRSA isolates with intermediate and high resistance towards vancomycin have been described recently, as well as synercid- and linezolid-resistant strains. The emergence and spreading of multi-resistant S. aureus is alarming and emphasises the need to develop new antibiotics. Currently, well-established therapeutics targeting processes of cell wall biosynthesis, DNA and RNA metabolism, and protein biosynthesis. This study comprises the investigation of putative targets of novel antibiotics. Therefore, seven target genes were selected and functionally characterised in vitro and in vivo. Four of these genes have not been characterised in S. aureus at the beginnig of this study. A deletion mutagenesis approach for these genes (clpP, purH, ssrA and smpB) should provide an indication of their essientiallity for the organism. clpP and purH could be successfully inactivated, supporting their non-essential role in S. aureus. The non-essential genes arlR, arlS and putP were inactivated and different phenotypic studies of the S. aureus deletion mutants ∆clpP, ∆arlR, ∆arlS, ∆purH and ∆putP were performed. Whole genome microarray analysis was applied on ∆clpP and ∆arlR mutants to study the manifold transcriptional changes by the inactivation of these two regulators in S. aureus. The growth rate of ∆clpP was strongly reduced at different temperatures (30°C, 37°C, 42°C) and completely inhibited at 20°C. Growth was highly impaired under anaerobic conditions. Moreover, ClpP deficiency resulted in a reduced hemolytic activity and diminished adherence to polystyren as well as a strongly enhanced autolytic activity. The invasiveness of ∆clpP was significantly elevated (~10-fold) compared to the isogenic wildtype as shown in an invasion assay with the 293T epithelial cell line. All these effects could be successfully reverted by complementation of the mutant strain with a vector expressing the native clpP sequence. The microarray analysis of ∆clpP revaeled a strong shift of the bacterial transcriptome (15 % of all genes were differentially regulated). Especially regulators and genes which reply to varying redox conditions, for example as a result of different stresses or anaerobiosis, were shown to be affected at the transcriptional level. Deletion mutants of the virulence-associated two component system genes arlR and arlS caused a higher hemolytical activity, enhanced adherence to polystyrene and an enhanced autolytic activity in a Triton X-100 assay. Furthermore, internalisation of arlRS bacteria by 293T epithelial cells was diminished, and ∆arlR was significantly less infectious than the wildtype in a catheter-related infection model in rats. The microarray analysis of ∆arlR in the exponential and stationary growth phase revealed a strong influence of the ArlRS-system on gene expression in S. aureus. In the exponential phase a total of 5 %, and in the stationary phase 15 % of all genes were differentially regulated. ∆purH and ∆putP showed no alterations of growth, biofilm formation or hemolytic properties compared to the wild type strain. Inactivation of purH, however, caused a significant attenuation of S. aureus MA12 in a rat-infection model. Since the construction of ssrA and smpB deletion mutants have failed, conditional lethal expression systems should be utilised to address the question of essentiality of these genes. It could be shown by this study that neither the genetic exchange of the wildtype promoter with an in vitro adjustable promoter nor an antisense RNA-based approach were sufficient to clearify the status of ssrA and smpB as essential genes in S. aureus.
