N2  - Ziel dieser Arbeit war es, den in der Literatur bei Karzinomen häufig von Deletionen betroffenen kurzen Arm von Chromosom 8 unter Verwendung von Mikrosatellitenmarkern zur LOH-Analytik bezüglich der häufigsten Deletionsregion genauer zu kartieren und zu untersuchen, ob das Kandidaten-Tumorsuppressorgen MTUS 1 in diesem Bereich lokalisiert ist. Dabei wurden ausgehend vom zentral gelegenen Marker D8S254, der in der Literatur als prognostisch relevant eingestuft wurde, vier weitere Marker (D8S542, D8S549, D8S261, D8S1734) eingesetzt, um die minimale Verlustregion auf dem Chromosom 8p22 definieren zu können. Als Untersuchungskollektiv wurden 48 DNA-Probenpaare jeweils von korrespondierendem Tumor- und Normalschleimhautgewebe herangezogen. Dabei stellte sich der Bereich zwischen den Markern D8S254 und D8S261 als die minimale Verlustregion heraus. Die detektierte minimale Verlustregion beinhaltet das Kandidaten-Tumorsuppressorgen MTUS 1. Die Ergebnisse dieser Arbeit können deshalb als weiterer Hinweis auf seine vermutete Rolle als Tumorsuppressorgen gewertet werden.
N2  - LOH of the microsatellite marker D8S254, wich is located on chromosome 8p22, is correlated with a poor prognosis in several carcinomas, including the colorectal cancer. This is considered to be a hint for the localisation of a tumorsuppressor gene in this region. Different genes there, including the MTUS1, are candidate tumorsuppressor genes. The purpose of this study was to localize a minimal regional loss on 8p22. Four further microsatellite markers (D8S542, D8S549, D8S261, D8S1734) beside the marker D8S254 were used. 48 colorectal cancers were tested. In result - the minimal regional loss could be located between the markers D8S254 and D8S261. This region is next to the localisation of the MTUS1-gene. So the assumption of its role could be confirmed in this study.
