Schutz bieten Ballaststoffe, die zu kurzkettigen Fettsäuren, wie zum Beispiel Butyrat, fermentiert werden. Wir konnten mittels Western Blot eine Verringerung der Genexpression der Proteine Integrin-ß4 (Zelladhäsionsfaktor), Interleukin-1ß (Entzündungsfaktor) und HMGA (Transkriptionsfaktor) ab 48 Stunden Inkubationszeit mit 2mmol/l Butyrat in der gut-differenzierten HT-29-, der entdifferenzierten SW-480-Kolonadenokarzinomzelllinie sowie deren Metastasenzelllinie SW-620 zeigen. Unsere Ergebnisse weisen darauf hin, dass Butyrat die Zelladhäsion, die Progression, das metastatische Potential, die entzündliche Komponente und die Transkriptionsrate kolorektaler Karzinomzellen vermindert. Die erhöhte Expression der Proteine Integrin-ß4 und HMGA ist ein frühes Ereignis im Übergang zu malignen Formen. N2 - Colorectal cancer is one of the most frequent cancers in our industrialized world. Butyrate production from dietary fibers by fermentation is protective against it. By western blot we could demonstrate the diminution of gene expression of the proteins integrin-ß4 (cell adhesion factor), interleukin-1ß (inflammatory factor) and HMGA (transcription factor) after 48 hours of incubation with 2mmol/l butyrate in well-differentiated HT-29-, not-differentiated SW-480-colonic adeno carcinoma cell lines and its metastatic cell line SW-620. Our examinations show that butyrate diminishes the cell adhesion, progression, metastatic potential, inflammatory component and rate of transcription in colonic carcinoma cells. The increased expression of the proteins integrin-ß4 and HMGA is an early event in the transition to malignant forms. Thus they could serve as diagnostic and prognostic markers and as a target for anticancer drugs like butyrate. Hierzu erfolgte eine Unteruchung von Angiogenesefaktoren und deren Beeinflussung durch die kurzkettige Fettsäure Butyrat. Die untersuchten Faktoren umfassten VEGF, als potentes angiogenetisches Agens und dessen zugehörigen Rezeptor FLT-1. Zudem wurde das Tumorsuppressorgen p53, ein wichtiger Modulator der Tumorentstehung und Tumorprogression, mituntersucht. Ziel war es, die positiven Effekte im Rahmen der Prävention auf die metastasierten kolorektalen Karzinome bzw. derer Zelllinien aufzuzeigen. Die verwendeten Methoden stützten sich einerseits auf die Analyse der mRNA und der Proteine. Hierfür wurden die Zellkultur, die RNA-Isolation, ein RNase Protection Essay (RPA), der Westernblot und ein Enzyme-Linked Immunosorbent Assay (ELISA) durchgeführt. Die Analysen zeigten einen positiven Effekt auf die Supprimierbarkeit des Wachstumsfaktors VEGF und dessen Rezeptor FLT-1, sowie auch auf das Tumorsuppressorgen p53mut- in seiner mutierten Variante. Butyrat senkte somit die Expression von VEGF und FLT-1 sowohl auf RNA- als auch auf Proteinebene. Bezüglich der Expression von p53 konnte gezeigt werden, dass die RNA-Expression durch Butyrat ebenfalls eine Hemmung erfuhr. Ein weiteres Ergebnis der Arbeit bestand darin, den Stress, welcher sowohl die Zugabe von Butyrat als auch eine Episode mit Serumentzug auf das Verhalten des Wachstumsfaktors VEGF hatte. Hierbei zeigte sich eine sehr rasche Zunahme der Expression. Diese normalisierte sich im weiteren Verlauf und hatte somit keinen längerfristigen Einfluß auf die Ergebnisse. Zusammenfassend zeigt sich eine deutliche Beeinflussung der Angiogenese durch Butyrat, welche auch auf dem Hintergrund der aktuellen Studienlage bezüglich VEGF Antikörpern einen adjuvanten Behandlungsweg darstellen könnte. Kritisch anzumerken bleiben die Probleme der Applikation von Butyrat. N2 - Angiogenesis is a critical factor in growth and metastasis of colon tumors. Neovascularisation is necessary for tumor growth and spread. Malignant cells are capable to activate dormant endothelial cells via overexpression of pro-angiogentic factors such as the vascular endothelial growth factor (VEGF) and the simultaneous inhibition of the expression anti-angiogentic factors such as angiostatin. Of these angiogenic factors, VEGF plays a major role. In tumor cells the expression of VEGF is modulated by growth factors, genetic aberrations, and hypoxia. Among those factors, the product of the mutated form of the p53 tumor suppressor gene, which occurs frequently in human colon carcinoma, have been show to have a stimulatory effect on VEGF expression. Two tyrosine kinase VEGF-receptors have been described, which bind VEGF with high affinity: KDR (Kinase domain region) and flt-1 (fms-like tyrosine kinase 1), which are both predominantly expressed on vascular endothelial cells. Binding of VEGF to its receptor results in receptor dimerization and activation of the intrinsic kinase followed by receptor autophosphorylation and signal transduction. The role of VEGF and its receptors in malignant disease is characterized by increased expression of both VEGF and its receptors in tumors. This often correlates with a higher microvessel density and poor prognosis. Considerable effort has been generated to develop VEGF/VEGF receptor (VEGFR) antagonists. Inactivation of VEGF with neutralizing antibodies or expression of a dominant-negative mutant of flk-1 results in an effective inhibition of tumor angiogenesis and tumor growth in vivo. VEGF is an important angiogenic factor in primary and metastatic human colon cancer with may have a prognostic value. Patients suffering form hepatic metastasis expressed higher levels of VEGF mRNA than those without. The expression of the VEGF189 mRNA isoforms is correlated with liver metastasis and poor prognosis in colon cancer. Epidemiologic, animal and intervention studies support the overall conclusion that an inverse association between dietary fiber intake and colorectal cancer risk does exist. Fermentation of dietary fiber by anaerobic bacteria within the colon generates an array of short chain fatty acids (SCFAs) such as butyrate, propionate, valerate and others. Of these SCFAs, butyrate appears to mediate the most profound protective effects of a high-fiber diet. In vitro, butyrate has been shown to modulate cell proliferation, differentiation, motility, adhesion, and apoptosis. These effects may constitute the underlying mechanism of butyrate’s potential to protect against colon carcinogenesis. Zu diesen zählt auch Butyrat, welches den Kolonepithelien als Hauptenergieträger dient. Mehrere klinische Studien konnten zeigen, daß eine lokale Therapie der Colitis ulcerosa (CU) mit Butyrat zu einer deutlichen Abnahme der Entzündungsaktivität führt. In nahezu sämtlichen entzündlichen Prozessen ist der nukleäre Faktor kappa B (NF-kappa B) an der transkriptionellen Regulation proinflammatorisch wirkender Genprodukte beteiligt. Durch Mediatoren wie TNF alpha oder IL-1 beta wird eine nukleäre Translokation von normalerweise im Zytoplasma an inhibitorische I kappa B-Proteine gebundenem NF-kappa B hervorgerufen. Im Rahmen der CU ist NF-kappa B u. a. in Lamina propria-Makrophagen (LPMNC) und intestinalen Epithelzellen (IEC) aktiviert. Zielsetzung: In Zellkulturversuchen mit Adenokarzinomzelllinien (HeLa 229, SW480, SW620) sowie an Biopsien von Patienten mit einer distalen CU sollte aufgezeigt werden, inwieweit Butyrat die Aktivierung von NF-kappa B zu inhibieren vermag und welcher Mechanismus hier möglicherweise zugrunde liegt. Ergebnisse: Nach Stimulation von HeLa 229 mit TNF alpha oder IL-1 beta kommt es in ca. 90 % der Zellen zu einer nukleären Translokation von NF-kappa B, welche mittels einer Immunfluoreszenzmarkierung nachgewiesen wurde. Durch Vorinkubation mit 2 bzw. 4 mM Butyrat über 12, 24, 36 und 48 Stunden ließ sich sowohl eine zeit- als auch dosisabhängige Inhibition dieser Translokation erzielen. Bei Anwendung von 4 mM Butyrat war, verglichen mit unbehandelten Zellen, eine signifikante Reduktion des Anteils von nukleärem NF-kappa B bereits nach einer 24stündigen Inkubation (TNF alpha; IL-1 beta: 36 h), bei 2 mM Butyrat erst nach 36 h (TNF alpha; IL-1 beta: 48 h) festzustellen. Im direkten Vergleich von 2 und 4 mM Butyrat zeigte sich nach TNF alpha-Stimulation bereits nach 24 h ein signifikanter Vorteil der höheren Dosis, bei Verwendung von IL-1 beta erst nach 36 h. Die qualitativen Ergebnisse von HeLa 229 ließen sich ebenfalls an den Zelllinien SW480 und SW620 nachweisen. Bei Untersuchung des inhibitorischen I kappa B alpha-Proteins mittels Western Blot konnte für die o. g. Zelllinien eine TNF alpha-induzierte Phosphorylierung von I kappa B alpha aufgezeigt werden. Durch eine 24stündige Präinkubation mit 4 mM Butyrat war diese effektiv hemmbar. Nachweise von Ikappa B alpha an IL-1 beta-stimulierten HeLa 229-Zellen erbrachten, gemeinsam mit den Ergebnissen zur NF-kappa B-Translokation, Hinweise auf mögliche alternative I kappa B alpha-unabhängige Wege der NF-kappa B-Aktivierung. Hinsichtlich der klinischen Anwendung von Butyrat wurden LPMNC in Biopsien von Patienten mit einer distalen CU durch eine immunhistochemische Doppelfärbung von NF-kappa B und CD68 untersucht. Bei unbehandelten Erkrankten ließ sich in nahezu 80 % der LPMNC eine nukleäre Translokation von NF-kappa B nachweisen. Eine topische Behandlung mit 100 mM Butyrat (Klysmen) führte nach 4 Wochen zu einer signifikanten Reduktion der Anzahl von aktivierten LPMNC sowohl innerhalb der Butyrat- als auch im Vergleich mit der Placebogruppe. Dieser Effekt war auch nach 8 Wochen noch nachweisbar. Zur weiteren Objektivierung der Befunde wurden die Biopsien ebenfalls histologisch untersucht. Die Dichte der neutrophilen Granulozyten im Krypten- und Oberflächenepithel wurde durch Butyrat gegenüber Placebo signifikant reduziert; alle übrigen morphologischen Entzündungsparameter änderten sich mitunter zwar deutlich, jedoch wurde hier nicht das Signifikanzniveau erreicht. Klinisch konnte unter Butyratbehandlung bereits nach 4 Wochen eine signifikante Abnahme des Disease Activity Index (DAI) festgestellt werden, nach 8 Wochen auch im Vergleich mit der Placebogruppe. Signifikante endoskopische Veränderungen ergaben sich nach einer Behandlungsdauer von 8 Wochen nur innerhalb der Butyratgruppe. Schlussfolgerung: Was die Anwendung von Butyrat in der Behandlung der CU angeht, so konnte in der vorliegenden Arbeit gezeigt werden, daß mit dieser KKFS sowohl in vitro (zytokinstimulierte Adenokarzinomzelllinien als Entzündungsmodell) als auch in vivo ein potenter und gleichzeitig kostengünstiger Inhibitor der NF-kappa B-Aktivierung zur Verfügung steht. So korreliert die im klinischen Teil dieser Untersuchung nachweisbare Reduktion der Entzündungsaktivität (DAI, Endoskopie-Score) mit einer signifikanten Hemmung der NF-kappa B-Translokation in LPMNC. Dennoch erscheinen, auch vor dem Hintergrund der geringen Fallzahlen, weitere Untersuchungen zur Anwendung von Butyrat bei Patienten mit einer CU sinnvoll. N2 - Background: The short chain fatty acid (SCFA) butyrate as main energy source for colonocytes is produced by anaerobic bacterial fermentation of resistant starch within the colonic lumen. Several clinical trials suggest that topical treatment of ulcerative colitis (UC) with butyrate can be effective in ameliorating symptoms and controlling chronic inflammation of the intestinal mucosa. Nuclear factor kappa B (NF-kappa B) is a central mediator of inflammation and enhances the transcriptional regulation of several proinflammatory genes. In most unstimulated cells NF-kappa B is confined to the cytoplasm bound to its inhibitor I kappa B alpha. Stimuli such as TNF alpha or IL-1 beta lead to release of NF-kappa B from I kappa B alpha and nuclear translocation. In UC activated NF-kappa B was demonstated in lamina propria macrophages (LPMNC) and intestinal epithelial cells (IEC). Aims: The intention was to show if butyrate is able to inhibit NF-kappa B-activation and by which mechanism this might be achieved. Cultured adenocarcinoma cells (HeLa 229, SW480, SW620) and biopsy specimens of patients suffering from distal UC were investigated. Results: Stimulation of HeLa 229 with TNF alpha or IL-1 beta caused nuclear translocation of NF-kappa B in 90 % of the cells, detected by immunofluorescence staining. Preincubation with 2 and 4 mM butyrate for 12, 24, 36 and 48 hours lead to a time and dose dependent inhibition of NF-kappa B translocation. Using 4 mM of butyrate caused a significant decrease of nuclear NF-kappa B after 24 hours of pretreatment (TNF alpha; IL-1 beta: 36 h), using 2 mM of butyrate only after 36 hours (TNF alpha; IL-1beta: 48 h) compared to untreated cells. Direct comparison of both 2 and 4 mM butyrate treatment followed by a stimulation with TNF alpha showed a significant advantage of the higher dose after 24 hours, using IL-1beta this effect was observable only after 36 hours. Qualitive results of HeLa 229 could also be achieved in SW480 and SW620 cells. The investigation of inhibitory I kappa B alpha protein in the above mentioned cells by using a western blot system showed a TNF alpha-induced phosphorylation of I kappa B alpha. A 24 hour preincubation with 4 mM butyrate achieved an actual inhibition of I kappa B alpha-phosphorylation. Investigating IL-1 beta-stimulated HeLa 229 cells revealed an alternative and I kappa B alpha-independent way of NF-kappa B activation especially in correlation to the results of NF-kappa B -translocation experiments. Regarding the clinical application of butyrate an immunohistochemical double-staining of NF-kappa B and CD68 in LPMNC was established. In untreated patients nearly 80 % of the LPMNC showed a nuclear translocation of NF-kappa B. Topical treatment with 100 mM butyrate (klysma) for 4 weeks lead to a significant reduction of activated LPMNC in the verum-group as well as compared to placebo. This effect was still detectable after 8 weeks. In addition a detailed analysis of histological changes was performed. After treatment with butyrate a significant decrease in both the number of neutrophils in crypt and surface epithelia could be observed compared to placebo; however other butyrate-related morphological changes were clearly remarkable but not significantly different to those found in placebo-treated patients. Regarding the clinical course the comparison of DAI scores after 4 weeks already revealed a significant decrease in the butyrate-treated group, as well as after 8 weeks by between-group comparison. After 8 weeks a significant reduction of the endoscopic score of mucosal inflammation could be noticed in the butyrate-treated patients as compared to entry. Conclusions: Applying butyrate to treat UC, the presented work gives evidence for this SCFA to act in vitro (cytokine stimulated adenocarcinoma cells as a model for inflammation) and in vivo as a potent and inexpensive inhibitor of NF-kappa B activation. According to these data the clinically observed reduction of inflammation (DAI, endoscopic score) is significantly correlated to inhibition of NF-kappa B translocation in LPMNC. However the clinical results are preliminary and - especially regarding the case numbers - confirmation by a larger trial on the application of butyrate to patients with UC is necessary. Die kurzkettige Fettsäure Butyrat ist ein wichtiges Produkt bakterieller Fermentation von Ballaststoffen bzw. von unverdaubaren Kohlenhydraten im Kolon. Butyrat hat paradoxe Effekte auf Epithelzellen des Kolons: Haupternergieträger und Wachstumsstimulator normaler Mukosa einerseits, Proliferationshemmer und Apoptoseinduktor kolorektaler Karzinomzellen in vitro andererseits. Auch für NSAID wie Aspirin belegen epidemiologische Studien einen chemoprotektiven Effekt gegenüber dem Kolonkarzinom. Für das Zytokin TNFalpha werden einerseits apoptoseinduzierende Effekte für kolorektale Karzinomzellen in vitro beschrieben, jedoch gelten einige Kolonkarzinomzellinien als resistent gegen TNFalpha. Andererseits besitzt TNFalpha auch proinflammatorische und antiapoptotische Wirkung über Aktivierung des nukleären Faktors kappa B (NF-kappaB). In der vorliegenden Arbeit wurden die Einflüsse sowohl von Aspirin als auch von TNFalpha auf die durch Butyrat induzierte Apoptose an humanen kolorektalen Karzinomzellinien untersucht. Zu diesem Zweck wurde ein durchflußzytometrischer Annexin V – Propidiumjodid – Assay etabliert. Mit Hilfe dieses Assays konnte gezeigt werden, daß der Apoptose induzierende Effekt sich sowohl durch eine Kombination mit Aspirin als auch durch eine Kombination mit TNFalpha im Sinne einer additiven Wirkung steigern läßt. Der Einfluß von Butyrat auf die antiapoptotische Wirkung von TNFalpha über Modulation von NF-kappaB wurde in einem Electophoretic Mobility Shift Assay (EMSA) untersucht. Die Verstärkung der Butyrat-induzierten Apoptose durch eine Kombination mit TNFalpha ist mit einer Hemmung der TNFalpha induzierten Aktivierung von NF-kappaB assoziiert. In einem RNase Protection Assay war auf mRNA-Ebene keine Beeinflussung der NF-kappaB abhängigen antiapoptotischer Faktoren (TRAF-1 und -2, c-IAP1 und 2 und XIAP) durch Butyrat nachweisbar. Die Verstärkung der Apoptose durch TNFalpha zeigt, daß Butyrat in seiner protektiven Wirkung in der Lage ist, neben einer direkten Beeinflussung der Kolonozyten auch auf körpereigene Signalwege zu wirken. Die Untersuchungen dieser Arbeit leisten einen Beitrag zur weiteren Klärung der molekularen Grundlagen der Butyratwirkung auf Kolonepithelzellen. N2 - Epidemiological studies suggest a protective effect of a high fibre enriched diet against the development of colorectal cancer. The short chain fatty acid butyrate is an important product of bacterial fermentation of fibres, respectively of undigested carbohydrates within the colon. Butyrate has paradoxical effects on colonic epithelial cells: main energy source and growth stimulus for normal mucosa on the one hand, inhibition of proliferation induction of apoptosis of colorectal carcinoma cells in vitro on the other hand. Also for NSAID like aspirin by epidemiological studies there is body of evidence for a chemo protective effect against colon cancer. For the cytokine TNFalpha apoptosis inducing effects on colorectal carcinoma cells in vitro are also described, but some colorectal carcinoma cell lines are considered to be reisstent to TNFalpha induced apoptosis. Furthermore, TNFalpha also has pro-inflammatory and anti-apoptotic properties by activating the nuclear factor kappa B. Aim of this work was to investigate the effects of aspirin and TNFalpha on the butyrate induced apoptosis in human colorectal carcinoma cell lines. For this reason a flow cytometric annexin V – propidium iodine – assay was established. It was possible to show by this assay that the apoptosis inducing effect of butyrate was enhanced to an additive effect by combining as well with aspirin as well as with TNFalpha. To further investigate whether butyrate influences the anti-apoptotic effect of TNFalpha by modulating NF-kappaB, an electrophoretic mobility shift assay (EMSA) was performed. The enhancement of butyrate induced apoptosis by combination with TNFalpha is associated with an inhibition of TNFalpha induced NF-kappaB activation. A RNase protection assay did show no influence of on the mRNA expression of NF-kappaB dependent factors (TRAF-1 and -2, c-IAP1 and 2, and XIAP). The enhancement of TNFalpha induced apoptosis demonstrates that the protective effects of butyrate not only are a result of direct influencing colonocytes but also by modulating signal cascades within the body. The results of this study support understanding the molecular effects of butyrate on colorectal epithelial cells. Maybe, in future there will be possibilities to use butyrate as an adjuvant therapeutic agent by prevention and therapy of colorectal carcinomas. Die genauen Mechanismen hinter der Wirkung beider Substanzen sind ungeklärt. Um die Effekte von Butyrat und Acetylsalicylsäure, sowie deren Kombination, auf das Wachstum, die Differenzierung und Apoptose von Kolonkarzinomzellen zu untersuchen, wurden Zellkulturexperimente und Western Blot Analysen durchgeführt. Unter Butyratinkubation kam es zu einer Steigerung der Differenzierung von HT-29-Kolonkarzinomzellen, jedoch nicht von Caco-2 Zellen. Acetylsalicylsäure allein hatte keinen relevanten Effekt auf die Zelldifferenzierung und in Kombination konnte Acetylsalicylsäure die differenzierende Wirkung des Butyrats nicht verstärken. Butyrat und Acetylsalicylsäure hemmten die Proliferation der untersuchten kolorektalen Karzinomzellen. Bei Inkubation mit einer Kombination von Butyrat und Acetylsalicylsäure wurde der antiproliferative Effekt der Einzelsubstanzen deutlich verstärkt. Im Western-Blot konnte diese Beobachtung durch Untersuchung des Proliferationsmarkers PCNA bestätigt werden. Gleichzeitig veränderte sich das Expressionsmuster zellzyklusregulierender Proteine in den untersuchten Zellen: Die Cyclin D3-Expression wurde in Caco-2 Zellen sowie in HT-29 Zellen durch Butyratinkubation erhöht. In Kombination mit Acetylsalicylsäure verstärkte sich die Wirkung von Butyrat auf die Expression von p21Waf1/Cip1 und cdk2: Acetylsalicylsäure und Butyrat in höheren Konzentrationen reduzierten die Expression von cdk2 in HT-29 Zellen, in niedrigen Konzentrationen führte nur die Acetylsalicylsäure-Butyrat-Kombination zu einem Abfall des cdk2-Proteingehalts. Butyrat induzierte die Expression des cdk-Inhibitors p21Waf1/Cip1 in beiden Kolonkarzinomzellinien. Die Wirkung auf p21Waf1/Cip1 war hierbei p53-unabhängig, da es gleichzeitig zu einem Abfall der Konzentration an p53-Protein in HT-29 Zellen kam und in Caco-2 Zellen kein p53 nachweisbar war. Acetylsalicylsäure hatte erst in höherer Konzentration einen Einfluss auf die p21Waf1/Cip1 Proteinexpression in HT-29 Zellen. Die Kombination von Butyrat und Acetylsalicylsäure übertraf in ihrer Wirkung die des Butyrats schon bei Konzentrationen, bei denen die alleinige Butyratinkubation ohne Effekt blieb. Weiterhin induzierte Butyrat, über einen Anstieg der Expression des pro-apoptotischen Faktors Bak, Apoptose in den untersuchten Kolonkarzinomzellen, bei gleichzeitigem Abfall des anti-apoptotischen Bcl-XL. Bei Inkubation mit der Acetylsalicylsäure-Butyrat-Kombination kam es zu einer deutlichen Verstärkung der Apoptoseinduktion, wobei Acetylsalicylsäure die Wirkung des Butyrats auf Bak und Bcl-XL jedoch nicht steigerte. Zudem konnte gezeigt werden, dass der Effekt des Butyrats mit einer Inhibition der Phosphorylierung und Degradation von IkBa, und damit einer Verhinderung der Aktivierung des Transkriptionsfaktors NFkB einhergeht, was die Wirkung von apoptose-induzierenden Substanzen verstärken kann. Zusammenfassend zeigte sich bei Kombination der Einzelsubstanzen Butyrat und Acetylsalicylsäure eine Verstärkung ihrer Wirkung auf die Zellproliferation, sowie die Apoptoseinduktion bei Kolonkarzinomzellen. N2 - Numerous experimental and clinical study data support the view of a preventive effect of butyrate and aspirin on the incidence of colorectal carcinomas. The exact underlying mechanisms however remain unclear. In order to examine the effects of butyrate, aspirin and their combination on proliferation, differentiation and apoptosis in colorectal carcinoma cells, cell culture experiments and Western blot analyses were performed. Butyrate-incubation induced differentiation in HT-29 colon carcinoma cells, but not in Caco-2 cells. Aspirin did not exert relevant effects on cell differentiation and did not support the butyrate effects. Both butyrate and aspirin decreased the proliferation of the investigated cell lines. Coincubation with both substrates enhanced the antiproliferative effects of the single substances. In Western blot analyses this observation was confirmed by decreased levels of PCNA, a marker for cell proliferation. At the same time the expression pattern of cell cycle regulating factors was changed: Cyclin D3 expression was induced by butyrate in Caco-2 and HT-29 cells. In combination with aspirin the effect of butyrate on p21Waf1/Cip1 und cdk2 expression was enhanced: Aspirin and butyrate in high concentrations decreased the expression of cdk2 in HT-29 cells whereas in low concentrations only the aspirin-butyrate-combination decreased cdk2 protein. Butyrate induced the expression of the cdk-inhibitor p21Waf1/Cip1 in both cell lines. Butyrate induced p21Waf1/Cip1 independently from p53 as butyrate decreased p53 levels at the same time. Aspirin induced p21Waf1/Cip1 expression when applied at high concentrations. In combination butyrate and aspirin had a stronger effect on p21Waf1/Cip1 expression already at concentrations at which butyrate alone did not have an effect. Also butyrate induced apoptosis by inducing pro-apoptotic Bak and decreasing anti-apoptotic Bcl-XL in the investigated cell lines. When butyrate and aspirin were applied in combination the pro-apoptotic effects of butyrate were enhanced. However aspirin did not have an effect on Bak and Bcl-XL expression. Furthermore it could be shown that the butyrate effects are associated with an inhibition of phosphorylation and subsequent degradation of IkBa. This was followed by an inhibition of the activation of the transcription factor NFkB, which can support the effect of apoptosis-inducing factors. Taken together an enhanced effect of the aspirin-butyrate combination on cell proliferation and apoptosis in colorectal cancer cells was observed. Dabei moduliert Butyrat die Expression von verschiedenen Zellzyklus- und Apoptoseregulatoren. Wie Butyrat seine Wirkungen auf chromosomaler Ebene vermittelt, ist bisher nicht ausreichend geklärt. Bekannt ist, dass Butyrat Histondeacetylasen nichtkompetetiv hemmt und durch Hyperacetylierung von nukleären Histonproteinen die Chromatinstruktur moduliert. Die „high-mobility-group“ (HMG) Proteine sind neben Histon-Proteinen strukturelle Bestandteile des Chromatins. Die vermehrte Expression des HMGA1-Proteins ist eine Gemeinsamkeit vieler humaner Malignome und die HMGA1-Proteinfamilie wurde selbst als neue Gruppe von Onkogenen erkannt. Die HMGA1 Proteine beeinflussen die Expression zahlreicher Gene und stehen selbst wiederum unter der Kontrolle von Onkogenen, wie z.B. dem c-myc. Im Rahmen dieser Arbeit konnte gezeigt werden, dass Butyrat und der Histon-deacetylase-Inhibitor Trichostatin A die Expression von HMGA1 in humanen Adenokarzinomzellen des Gastrointestinaltraktes modulieren. Butyrat-Konzentrationen von 2mM und 4mM führten erstmals nach 24-stündiger Inkubation zu einer deutlichen Reduktion der mRNA-Expression von HMGA1. In der Magenkarzinomzelllinie 23132/87 und der Kolonkarzinomzelllinie Sw-480 wird die HMGA1 mRNA Expression durch Butyrat zeit- und dosisabhängig reduziert. In der Kolonkarzinomzelllinie Sw-620 konnte für die untersuchten Konzentrationen keine Dosisabhängigkeit festgestellt werden. Auch Trichostatin A bewirkt eine Reduktion der mRNA-Expression von HMGA1 in den untersuchten Zelllinien. Wie Butyrat die Expression von HMGA1 moduliert, ist nicht geklärt. Denkbar ist, dass Butyrat über Modulation vorgeschalteter Signalkaskaden, wie z.B. im Falle des c-myc, die HMGA1-Expression beeinflusst. Aber auch über eine Modulation der Chromatinstruktur durch Hemmung der Histondeacetylasen mit nachfolgenden Veränderungen der Expression verschiedener Gene könnte Butyrat die Expression von HMGA1 beeinflussen. Da die gleiche Wirkung auf die HMGA1-Expression durch den spezifischen Histondeacetylase-Inhibitor Trichostatin A vermittelt wird, könnte die Hemmung dieser Enzyme der Mechanismus sein, über den Butyrat seine Wirkungen ausübt. In vitro- und in vivo- Untersuchungen zeigen, dass eine Hemmung der HMGA1-Proteinsynthese die maligne Transformation verhindern, das metastatische Potential verringern und sogar das Wachstum bereits existenter Tumoren verlangsamen kann. Der protektive Effekt von Butyrat hinsichtlich der malignen Transformation und die Wirkung von Butyrat auf bereits maligne transformierte Zellen könnten zumindest teilweise dadurch erklärt werden, dass Butyrat die Expression von HMGA1 in Tumorzellen reduziert und damit dessen malignes Potential vermindert. Auch die Nicht-Histon-Proteine HMGN1 und HMGN2 stellen wichtige strukturelle Elemente des Chromatins dar. Sie werden sowohl in Normalgewebe als auch in maligne transformierten Zellen exprimiert. Auch die untersuchten Zelllinien Sw-480, Sw-620 und 23132/87 exprimieren HMGN1 und HMGN2. Signifikante Unterschiede zwischen der Primärtumorzelllinie Sw-480 und der Metastasenzelllinie Sw-620 des Sw-480 Kolonkarzinoms waren nicht feststellbar. Sowohl Butyrat als auch Trichostatin A senken in den untersuchten Karzinomzelllinien die Expression von HMGN1- bzw. HMGN2-mRNA, wobei dieser Effekt teilweise zeit- und dosisabhängig ist. Da die gleiche Wirkung auf die Expression von HMGN1 und HMGN2 durch den spezifischen Histondeacetylase-Inhibitor Trichostatin A vermittelt wird, könnte die Hemmung der Histondeacetylasen auch in diesem Fall der Mechanismus sein, über den Butyrat seine Wirkungen ausübt. Es gibt zunehmend Hinweise, dass HMGN1 und HMGN2 und die Modulation ihrer Expression eine bedeutende Rolle bei der Regulation zellulärer Vorgänge, wie z.B. Transkription und Replikation, haben. Nachdem in den letzten Jahren gezeigt wurde, welche Bedeutung Veränderungen der Chromatinstruktur durch Modifikation der Histonproteine für die Karzinogenese haben, ist zu vermuten, dass auch die Modulation der Chromatinbestandteile HMGN1 und HMGN2 in diesem Zusammenhang von Bedeutung sein könnte. Über die Bedeutung der HMGN-Expression, deren Modulation bzw. die der posttranslationalen Modifikation in humanen Karzinomen ist bis jetzt wenig bekannt. Von Bedeutung könnte dabei z.B. die kürzlich nachgewiesene Butyrat-induzierte Hyperacetylierung von HMGN2 sein. Unklar ist noch das unterschiedliche Ausmaß der Acetylierung der HMGN-Proteine in verschieden differenzierten Karzinomen, der Vergleich zwischen Normalgewebe und maligne transformierten Zellen und die Dynamik der Acetylierung in der Tumorprogression. In human carcinoma cells butyrate causes apoptosis, differentiation and growth inhibition. The molecular mechanisms by which butyrate mediates its effects are not yet fully understood. The family of non-histone nuclear proteins (high-mobility-group (HMG) proteins) is divided into three subfamilies, HMG-B, HMG-A and HMG-N, respectively. HMG-N proteins bind to chromatin and induce structural changes that affect DNA-dependent activities, such as transcription and replication. The HMG-A proteins are involved in gene expression during development and immune response. In normal epithelial cells HMG-A protein expression is low, however, HMG-A is overexpressed in several human carcinomas. In this study we examined the effects of butyrate on HMG protein expression in human gastrointestinal carcinoma cells. Treatment of human gastrointestinal carcinoma cells with butyrate and Trichostatine A led to a significant reduction of HMG-mRNA levels. The effect was observed in all examined cell lines, independent of origin or degree of differentiation. Butyrate mediates a wide range of effects in vitro and in vivo, such as inhibition of colon carcinogenesis and induction of apoptosis and differentiation. Modulation of the expression of transcription factors as well as the induction of changes within chromatin structure and composition are potential pathways of butyrate action. Butyrate is known to induce acetylation of core histones and non-histone nuclear proteins. Alterations of the chromatin structure caused by histone hyperacetylation or deacetylation may play an important role in changes of gene expression patterns and may consequently promote cancerogenesis. Modulation of HMG-mRNA expression constitutes an additional step in the modification of the chromatin structure. Therefore, it may at least in part be responsible for the wide range of butyrate associated effects.
