TY - THES A1 - Ruile, Philipp T1 - Einfluss des ACE-Hemmers Quinapril auf das Kapillarwachstum - morphologische Charakterisierung am chronisch ischämischen Rattenherzen T1 - Impact of the ACE inhibitor Quinapril on the capillary growth - Morphological characterisation on the chronic ischemic rat heart N2 - Zielsetzung der vorliegenden Arbeit war es, das in unserem Labor etablierte Modell der chronischen Koronarstenose der Ratte morphologisch zu charakterisieren. Besonderes Augenmerk galt hierbei dem mikrovaskulären Remodeling zwei Wochen nach myokardialer Ischämie. Darüber hinaus sollten Effekte des ACE-Hemmers Quinapril auf das mikrovaskuläre Remodeling unter chronischer Ischämie untersucht werden. Zur Kapillardichtebestimmung wurde eine neue immunhistochemische Färbung, die Lectinfärbung, in unserem Labor etabliert. Sie erlaubt eine selektive Anfärbung des Kapillarendothels und damit eine exakte Kapillardichtebestimmung im Querschnitt. Zusätzlich kam die Orientatormethode zum Einsatz, mit deren Hilfe Aussagen über dreidimensionale Strukturen aus der zweidimensionalen Ebene möglich sind. So lassen sich Längenveränderungen der myokardialen Kapillaren als weiterer Hinweis eines angiogenen Geschehens untersuchen. In früheren Untersuchungen unserer Arbeitsgruppe konnten Hypertrophiereaktionen und relative Kapillardichteänderungen am chronischen Infarktmodell nachgewiesen werden. Die vorliegende Arbeit zeigte jedoch, dass eine chronische Koronarstenosierung bis 2 Wochen nach Operation trotz eines deutlichen Zelluntergangs zu keinem signifikantem mikrovaskulären und myozytären Remodeling führt. Die in anderen Arbeiten gefundenen angiogenen Effekte von Quinapril auf die ischämische Skelettmuskulatur konnten für den Herzmuskel der Ratte 2 Wochen nach Operation nicht gefunden werden. Möglicherweise sind unsere Ergebnisse auf die korrekte Bestimmung der Hypertrophie-unabhängigen, absoluten Kapillardichte-bestimmung zurückzuführen, die in Arbeiten anderer Gruppen nicht berücksichtigt wurden. N2 - The aim of this dissertation was the morphological characterisation of the model of the chronic coronary stenosis of the rat, established in our laboratory. The point of interest was the microvascular remodelling two weeks after myocardial ischemia. In addition to this the effects of the ACE inhibitor Quinapril on the microvascular remodelling during chronic ischemia were examined. For the measurement of the capillary density a new immunohistochemical stain, the lectin stain, was established in our laboratory. This stain allows a selective coloration of the capillary endothel and so an exact measurement of the capillary density in the cross sectional area. Additionally the orientator method was applied, which allows statements on three-dimensional structures from the two-dimensional level. Thereby length changes of the myocardial capillaries as an further indication of angiogenesis can be examined. Former studies of our study group proved hypertrophic reactions and relative changes of capillary density on the chronic infarct model. But this dissertation proved that a chronic coronary stenosis up to two weeks after surgery does not result in a significant microvascular and myocyte remodelling, despite of a remarkable necrosis. The effects of Quinapril on the angiogenesis of the ischemic skeletal muscles, found in earlier studies, could not be proved for the heart muscle of the rat two weeks after surgery. Probably our results are traced back to our correct measurements of the hypertrophic independent, absolute measurement of capillary density, that were not considered by other studies.
