Unter ihnen ist Malaria neben der Immunschwächekrankheit AIDS und Tuberkulose am weitesten verbreitet. Laut WHO erkrankten allein im Jahr 2016 rund 216 Millionen Menschen an Malaria und weltweit verstarben 445.000 Menschen an den Folgen dieser Infektion. Solange die Wirksamkeit des Impfstoffs RTS,S/AS01 gegen Malaria noch Schwachstellen aufweist und andere Impfstoff-Kanditaten sich noch in präklinischen Testphasen befinden, ist vor allem die Entwicklung neuer Wirkstoffe, auch im Hinblick auf die rasante Ausbreitung von Resistenzen gegen herkömmliche Medikamente, weiterhin eine dringende Aufgabe. Eine vielversprechende Wirkstoffklasse mit interessanten strukturellen Eigenschaften und einer ungewöhnlichen Biosynthese aus Acetat-Einheiten sind die Naphthylisochinolin-Alkaloide aus den beiden paläotropischen Pflanzenfamilien der Dioncophyllaceae und Ancistrocladaceae. Die Naphthylisochinolin-Alkaloide sind hervozuheben aufgrund ihrer exzellenten Aktivität gegen Plasmodium falciparum, den Erreger der Malaria tropica, sowie wegen ihrer Wirksamkeit gegen Erreger weiterer Krankheiten wie beispielsweise AIDS, Leishmaniose und Afrikanische Trypanosomiasis. Auch im Kampf gegen maligne Erkrankungen wie Leukämie und pankreatischen Krebs werden sie aufgrund ihrer cytotoxischen Eigenschaften als vielversprechende Leitstrukturen betrachtet. Die strukturell beeindruckendsten Naphthylisochinolin-Alkaloide sind die dimeren Mbandakamine, die von unserer Arbeitsgruppe vor einiger Zeit aus einer kongolesischen Ancistrocladus-Liane isoliert wurden. Sie besitzen sieben stereogene Elemente und sind die ersten natürlich vorkommenden Dimere mit einer höchst unsymmetrischen 6',1''-gekuppelten zentralen Biarylachse. Diese impliziert eine außergewöhnlich hohe sterische Hinderung an der zentralen Achse, wie sie noch in keinem anderen dimeren Naphthylisochinolin-Alkaloid gefunden wurde. Verbunden mit ihren bemerkenswerten und vielseitigen pharmakologischen Wirkeigenschaften sind sie ausgesprochen interessante Moleküle für eine synthetische Erschließung. Ziel dieser Arbeit war die erstmalige Totalsynthese von Mbandakamin A und B sowie die Synthese ihrer monomeren Hälften 5-epi-Korupensamin E und 8-O-Methylkorupensamin A. Zudem sollten weitere Naphthylisochinolin-Dimere, die bei der Synthese der Mbandakamine anfallen, isoliert und charakterisiert werden. Alle neuen mono- und dimeren Naphthylisochinoline sollten abschließend am Schweizerischen Tropen- und Public-Health-Institut auf ihre biologische Aktivität getestet werden. Zusätzlich gelang im Rahmen eines Kooperationsprojekts erstmals die stereochemische Charakterisierung des strukturell ganz neuartigen, inhärent chiralen Wolframbiscorrols durch online HPLC-ECD-Analyse in Kombination mit quantenchemischen Rechnungen. N2 - Tropical infectious diseases are still the leading cause of death in developing countries. Among them, malaria is the most widespread one, besides the immunodeficiency syndrome AIDS and tuberculosis. According to the WHO about 216 million people were diagnosed with malaria in 2016, and it was estimated that 445 000 deaths due to malaria occurred globally. As long as vaccinations still show weaknesses in their efficacy or are only in the pre-clinical phase, the development of new drugs is inevitable because of the rapid spread of drug resistance. A promising class of agents with interesting structural properties and an unusual biosynthesis are the naphthylisoquinoline alkaloids from the two palaeotropic plant families of Dioncophyllaceae and Ancistrocladaceae, because not only the naphthalene parts but also the isoquinoline moieties originate from acetate- units. The naphthylisoquinoline alkaloids have attracted attention due to their high activities against Plasmodium falciparum, the pathogen causing malaria tropica, and for their effectiveness against the pathogens of further tropical diseases such as AIDS, leishmaniasis, schistosomiasis and trypanasomiasis. In the fight against malignant diseases such as leukemia and pancreatic cancer, they are also considered as promising lead structures, because of their cytotoxic properties. Structurally most impressive naphthylisoquinoline alkaloids are the dimeric mbandakamines, recently isolated by our group from a Congolese Ancistrocladus liana. The mbandakamines possess seven stereogenic elements and are the first naturally occurring dimers with a highly unsymmetric 6',1''-coupled central biaryl axis. This implies an exceptionally high steric hindrance that has not been found in any other dimeric naphthylisoquinoline alkaloid so far. In combination with their remarkable pharmacological activities, they are extremely interesting synthetic goals. The aim of of the present work was the first synthesis of 5-epi-korupensamine E and 8-O- methylkorupensamine A, which are the two molecular halves of mbandakamines A and B, followed by their phenol-oxidative coupling to give rise to the mbandakamines. Additionally, further mono- and dimeric naphthylisoquinolines obtained en route to the mbandakamines were to be isolated and characterized. Moreover, all new mono- and dimeric naphthylisoquinolines were to be tested for their antiprotozoal activities at the Swiss Tropical and Public Health Institute. In addition,in a cooperation project the stereochemical characterization of an inherently chiral tungsten biscorrole by online-HPLC-ECD analysis in combination with quantum-chemical calculations was achieved for the very first time. Ihr einzigartiges pharmakologisches Potenzial wird durch die selektive Interaktion mit einer Vielzahl von Zielmolekülen hervorgerufen. Neben ausgeprägten In-vitro-Aktivitäten gegen protozoische Erreger wie Leishmanien, Plasmodien und Trypanosomen besitzen die Vertreter dieser einzigartigen Naturstoffklasse nach neuesten Untersuchungen auch vielversprechende antitumorale Aktivitäten. Für deren Weiterentwicklung zu möglichen Arzneistoffen ist es daher unabdingbar, ihr pharmakologisches Potenzial tiefergehend zu untersuchen. Owing to the central role of natural products in the discovery and development of new drugs, the isolation, structural elucidation, the total synthesis and derivatization, the identification of target molecules, and investigations on the mode of action of these natural compounds are still essential. The small, yet fascinating class of naphthylisoquinoline alkaloids, which are exclusively obtained from the two plant families of Dioncophyllaceae and Ancistrocladaceae, does not only stand out due to their structural diversity with more than 200 representatives, but also shows pharmacologically interesting activities. In addition to pronounced in vitro activities against protozoan pathogens such as leishmania, plasmodia and trypanosoma, the representatives of this unique class of natural products also show promising antitumoral activities. For their further development as possible drugs, their pharmacological potential has to be investigated in more detail. The aim of the present thesis was the development of total synthetic approaches to biologically interesting naphthylisoquinoline alkaloids by means of different strategies. Um den Einfluss der verschiedenen Reagenzien auf die exzitonischen Eigenschaften und die PL-Emission zu quantifizieren, wurden die Dispergierreagenzien ausgetauscht, die Temperaturabhängigkeit bestimmt und die Konzentration der Reagenzien variiert. Die Dispergierreagenzien eines immobilisierten, SC-stabilisierten (6,5)-SWNT-Ensembles wurden im Mikrofluidikkanal ausgetauscht. Wird der Kanal mit Wasser gespült, verringern sich die PL-Intensität und die Emissionsenergie, da der Wasserfluss die Tensidmoleküle von der Oberfläche entfernt. Beim Austausch einer DOC-Umgebung gegen Wasser nimmt die PL-Intensität ebenfalls ab und die PL-Emissionsenergie verringert sich. Die Austauschexperimente verlaufen reversibel und der instantane Anstieg der Emissionsenergie bei der Tensidadsorption weist auf eine kooperative Anlagerung hin. Deshalb ist anzunehmen, dass sich Tensid-SWNT-Heteromizellen ausbilden. Anschließend werden die Emissionsenergie und die PL-Intensität in verschiedenen Dispergierreagenzien und in Wasser verglichen. Die größte Emissionsenergie und PL-Intensität werden während des Wechsels von einer SDS- zu einer (GT)16-Lösung gemessen. Dies kann auf die lückenlose Bedeckung der SWNT-Oberfläche mit einer heterogenen Schicht aus SDS-Molekülen und (GT)16-Strängen zurückgeführt werden. In reiner SDS-Umgebung emittieren die Nanoröhren Licht mit der zweithöchsten Energie, aber die PL-Intensität liegt unter der in einer SC-Umgebung. Die Emissionsenergie in der SC-Umgebung ist geringer und davon abhängig, ob die SWNTs bereits mit (GT)16-Strängen stabilisiert waren, da dies eine permanente Rotverschiebung der Emissionsenergie in der SC-Umgebung sowie eine verringerte PL-Intensität verursacht. In wässriger Umgebung verringert sich nach erfolgtem (GT)16-Kontakt die PL-Intensität dauerhaft. Danach wurde die Anlagerung von Tensidmolekülen an die (6,5)-SWNT-Oberfläche in Suspensionen mit der Temperatursprungmethode untersucht. Die Temperatur im Mikrofluidikkanal wurde anhand der linearen Abnahme der Emissionsenergie SC- und DOC-stabilisierter SWNTs mit steigender Temperatur bestimmt. Die Suspensionstemperatur ist in den verschiedenen Temperatursprungexperimenten unabhängig von der Messposition im Mikrofluidikkanal und wird durch die absolute Position auf den Peltier-Elementen bestimmt. Zudem stimmen die im Kanal gemessenen Temperaturen für SC- und DOC-stabilisierte (6,5)-SWNTs überein, weshalb in diesem Experiment nicht die erwartete Einstellung eines Gleichgewichts wie in einem Temperatursprungexperiment der Fall, sondern die Momentantemperatur gemessen wird. Die schnelle Gleichgewichtseinstellung zwischen freien und auf der SWNT-Oberfläche adsorbierten Tensidmolekülen beim Temperatursprung zeigt, dass die SC- und DOC-(6,5)-SWNT-Suspensionen thermochrome Farbstoffe sind. Wegen der Temperaturabhängigkeit der Emissionsenergie ist es bei wissenschaftlichen Arbeiten wichtig, neben dem verwendeten Dispergierreagenz auch die Temperatur der SWNT-Suspension anzugeben. Abschließend wurden die kritischen Mizellenkonzentrationen von Tensid-SWNT-Suspensionen in Verdünnungsexperimenten und daraus die thermodynamischen Bildungsparameter der Tensid-SWNT-Heteromizellen ermittelt. In der temperaturabhängigen Analyse der SC-SWNT-Mizellenbildung wird ein konstanter Hill-Koeffizient erhalten, der die Mizellenbildung als positiv kooperativ klassifiziert. Für die Bestimmung der Freien Mizellierungsenthalpie wurden nur die CMCs aus den Verdünnungsexperimenten verwendet, da die Mizellenbildung bei der Aufkonzentration teils kinetisch gehemmt ist. Da die Freie Mizellierungsenthalpie bei allen Temperaturen negativ ist, stabilisiert die Bildung der Heteromizellen das System. Die Triebkraft für die Mizellenbildung ist über 322 K die Enthalpie, während unterhalb von 316 K der Entropiegewinn dominiert. Die Verdünnung einer DOC-SWNT-Suspension zeigt keine Änderung der Emissionsenergie, obwohl dabei sowohl die primäre als auch die sekundäre CMC von DOC unterschritten werden. Zuletzt wurden die Verdünnungsexperimente mit einer SDS-SWNT-Suspension durchgeführt und die thermodynamischen Parameter der Mizellenbildung bestimmt. Da auf die Auflösung der Mizellenstruktur direkt die Aggregation der SWNTs folgt, wurde für die Ermittlung der CMC näherungsweise die Konzentration am Maximum der Emissionsenergie verwendet. Daraus ergibt sich bei jeder Temperatur eine negative Freie Mizellierungsenthalpie, deren Beiträge analog zu SC bei kleineren Temperaturen als 323 K entropisch und bei höheren Temperaturen enthalpisch dominiert werden. Somit ermöglichen die Experimente mit SC- und SDS-SWNT-Suspensionen die temperaturabhängige Bestimmung der CMC und damit die Berechnung der Freien Mizellierungsenthalpie sowie der zugehörigen enthalpischen und entropischen Beiträge. N2 - The goal of this thesis is to study the interplay between the SWNT surface and various dispersion agents. The influence of different surfactants on photoluminescence (PL) intensity and emission energy of (6,5)-SWNTs was quantified experimentally. The experiments are separated into three categories: surfactant exchange, determination of the temperature dependency of the PL, and the variaton of surfactant concentration. The dispersion agents of immobilized (6,5)-SWNT ensembles, that are placed in a microfluidic channel, are exchanged. Purging water through the microfluidic channel diminishes the PL intensity and red shifts PL emission energy, as the surfactants either SC or DOC are removed from the SWNT surface. The decrease of the PL intensity and emission energy are also observed if solution is exchanged for water. The surfactant exchange is reversible and the surfactant adsorption shows cooperativity features, which provide evidence for the formation of a surfactant SWNT hetero micelle. Afterwards the PL intensity and PL emission energy of the SWNTs are compared in different dispersing agents and water. The peak values for PL emission energy and PL intensity are measured when exchanging the SDS surfactant for (GT)16 ssDNA. They are caused by a complete coverage of the SWNT surface with a hetero-layer consisting of both species. The second highest PL emission energy is emitted in pure SDS environment followed by SC environment. Regarding the PL intensity the opposite effect is observed. A SC shell yields higher PL intensities than the SDS shell. If the SWNTs had previously been in contact with the (GT)16 ssDNA, the PL intensity was permanently reduced in the SC environment and was accompanied by a permanent red shift of the PL emission energy. The decreased PL intensity renders the water environment indistinguishable from the (GT)16 ssDNA covered SWNTs because they now show the same emission energy and equally low intensities. Subsequently the attachment of surfactant molecules to the surface of colloidally stabilized SWNTs is analyzed by using a temperature jump method. The temperature in the microfluidic channel was measured using the linear relationship between the temperature increase and the decrease of the PL emission energy of SC- and DOC-SWNT solutions. The solution temperature in the temperature jump experiments is independent of the channel position but determined by the absolute position on the Peltier element. In addition, the use of DOC instead of SC as surfactant showed no difference in temperature development, which leads to the conclusion that instead of the equilibration process, the actual liquid temperature is measured since the former is too fast for this experimental approach. The instantaneous equilibration of free and adsorbed surfactant molecules allows to deduce the solution temperature. Thus, SC- and DOC-SWNT solutions can be used as thermochromic dyes. More importantly, the correlation of PL intensity and PL emission energy with temperature emphasizes the importance of providing solution temperatures alongside the dispersing agents and their respective concentrations in scientific studies. Critical micelle concentrations (CMC) of surfactant SWNT solutions are determined in dilution experiments. Afterwards, thermodynamic parameters of the SWNT surfactant heteromicelle formation process are calculated. Hill analysis was used to identify the CMC at the inflection point of the sigmoidal PL emission energy decrease and to determine the Hill coefficient, classifying the micelle formation to be cooperative. Only the CMCs of the dilution experiments were used to calculate the micellar Gibbs energies since the micelle formation in the titration experiments is kinetically hindered at lower temperatures. Since the micellar Gibbs energy is negative at all temperatures, the micelle formation stabilizes the system. At temperatures above 322 K the micellar Gibbs Energy is dominated by the enthalpic contribution while the micelle formation is driven entropically below 316 K. The same experiment was conducted using DOC as dispersion agent, but showed no difference in the PL emission energy passing both the primary and the secondary CMCs of a pure DOC solution. Further experiments feature the dilution of SDS-SWNT solutions and the calculation of the thermodynamic parameters. As the breakup of the micellar structure is directly followed by SWNT aggregation, the CMC is approximately defined as concentration at the PL emission energy maximum. As the micellar Gibbs energy is always negative, the micelle formation provides a stabilization of the SDS-SWNT System, which is driven entropically below 323 K and dominated by enthalpy at higher temperatures. In conclusion, dilution experiments allow for the temperature dependent determination of CMCs and calculation of micellar Gibbs Energy with corresponding entropic and enthalpic contributions. Diese Sekundärmetabolite können oft medikamentös gegen Pathogene eingesetzt werden, die den Menschen befallen und Krankheiten verursachen. Die Herausforderung besteht dabei in der selektiven und sensitiven Detektion, der schonenden Isolierung und der richtigen und kompletten Strukturaufklärung dieser Moleküle, sowie der eventuellen synthetischen Modifikation, um eine bessere Verträglichkeit oder Wirkung für den menschlichen Körper zu erreichen. Leistungsfähige chromatographische Instrumente zur Trennung wie HPLC und CZE, emfindliche Detektoren wie UV- und Massenspektrometer, sowie aussagekräftige Messverfahren zur Charakterisierung struktureller Merkmale wie NMR- und CD-Spektroskopie und quantenchemische Rechnungen sind dabei von essentieller Bedeutung. Mit diesen – und weiteren – Methoden gelang in der vorliegenden Arbeit die Detektion, Isolierung und Strukturaufklärung neuer Naphthylisochinolin-Alkaloide aus zwei tropischen Ancistrocladus-Lianen, die Charakterisierung von bekannten und neuen Polyketiden aus einem Pilz der Gattung Streptomyces, sowie die Analyse von Glucosinolaten im Phloemsaft der Modellpflanze Arabidopsis thaliana. N2 - Plants and microorganisms produce a plethora of substrates that ensure immediate and reliable defense and signal transmission processes, to survive in a mostly hostile and rival-dominated enviroment. These compounds often show pharmacological properties which are beneficial for the human health and/or can help to heal and prevent human diseases that are caused by pathogens. To profit from the favorable effects of the metabolites, it is essential to have a high sensitivity and selectivity for the detection, a smooth isolation procedure, and effective methods that lead to the unequivocal elucidation of the absolute stereostructure, which then can be synthetically modified in order to increase the pharmacological impact and/or the tolerance in the human body. Powerful tools to overcome these challenges are chromatographic instruments for the separation like HPLC and CZE, sensitive detection devices like UV and mass spectrometer, and specialized characterization methods like NMR- and CD spectroscopy and quantum-chemical calculations. Diese spezielle Klasse acetogeniner Sekundärmetabolite weist durch die verschiedenen Kupplungspositionen der beiden namensgebenden Molekülbausteine eine breite strukturelle Diversität auf und zeichnet sich durch vielfältige pharmakologische Wirksamkeiten, z.B. antiplasmodiale, antileishmaniale oder antitrypanosomale Aktivitäten, aus. Zur Synthese dieser Naturstoffe wurde im Arbeitskreis Bringmann eigens eine Methodik entwickelt, das Lacton -Konzept. Diese Methode erlaubt durch eine Vorfixierung der beiden Molekülhälften durch eine Esterbrücke, anschließender intramolekularer Kupplungsreaktion und der stereoselektiven Öffnung des erhaltenen Lactons den atropselektiven Aufbau der Naphthylisochinoline. Als Ziele dieser Arbeit ergaben sich somit die Synthese pharmakologisch und strukturell interessanter Naphthylisochinolin-Alkaloide mittels des Lacton-Konzepts sowie die Isolierung und Strukturaufklärung weiterer Sekundärmetabolite aus Triphyophyllum peltatum (Dioncophyllaceae), welche anschließend auf ihre Bioaktivität hin untersucht werden sollten, um potenziell neue Leitstrukturen für neue Wirkstoffe zu finden. N2 - The paleotropical plant families of the Dioncophyllaceae and Ancitrocladaceae are the only known producers of naphthylisoquinoline alkaloids. This special class of acetogenic secondary metabolites shows a broad structural diversity - due to the different coupling sites of their molecular building blocks - and displays multifaceted pharmacological properties, e.g. antiplasmodial, antileishmanial and antitrypanosomal activities. For the synthesis of these natural products, a special method was elaborated in the bringmann working group, the lactone concept. This method permits the atroposelective synthesis of naphthylisoquinolines by prefixation of the two molecular halves via an ester bridge, subsequent intramolecular coupling, and stereoselective opening of the resulting lactone. Therefore, the main goals of this thesis were the synthesis of pharmacologically and structurally interesting naphthylisoquinoline alkaloids by using the lactone concept as well as the isolation and structural elucidation of further secondary metabolites from Triphyophyllum peltatum (Dioncophyllaceae) and the investigation of their bioactivities to possibly find new lead structures for new active agents. These secondary metabolites are often indispensable for life of the respective organisms and are produced in an efficient and highly stereo selective way. The efficiency and selectivity of 'nature' is a great challenge for chemists in the laboratory. Although a great variety of methods have been developed to mimic nature, in many cases the chemical synthesis is less stereo selective and/or yields are far below that of the biosynthetic pathway. The development and enhancement of methods is thus a great intention of modern natural chemistry. Beside the total synthesis, the investigation of biosynthetic pathways and the identification of the respective enzymes is a highly interesting field of research. Learning from nature can help to develop new synthetic strategies and methods. Many of the isolated compounds do not only possess the intended biological activity, but have also outstanding effects against pathogens of human diseases. Die Vielfalt an zellulären Prozessen, die durch BMPs reguliert werden, von der Knochenentwicklung und Organhomöostase bis hin zur Neurogenese, erstaunt – und wirft angesichts von teils redundanten, teils spezifischen Funktionen der BMPs Fragen zu den Mechanismen ihrer Signalübermittlung auf. Die Signaltransduktion von BMPs erfolgt wie bei den strukturell verwandten TGF-βs und Activinen durch die ligandeninduzierte Oligomerisierung von transmembranen Serin/Threonin-Kinaserezeptoren, von denen zwei Typen – Typ I und Typ II – existieren. Somit stellt sich die Frage, wie die Bildung von heteromeren Ligand-Rezeptor-Komplexen und die Aktivierung definierter intrazellulärer Signalkaskaden zusammenhängen und wie letztlich ein bestimmtes BMP-Signal durch einen „Flaschenhals“, repräsentiert durch die begrenzte Anzahl an Rezeptorsubtypen, in das Zellinnere übermittelt wird. Die Interaktionen zwischen BMP-2 / GDF-5 und den Typ I Rezeptoren BMPR-IA / BMPR-IB sind ein Paradebeispiel für Bindungspromiskuität und -spezifität. Während BMP-2 beide Rezeptoren BMPR-IA und BMPR-IB mit gleicher Bindungsaffinität bindet („promiske Interaktion“), zeigt GDF-5 eine 15-20fach höhere Bindungsaffinität zu BMPR-IB („spezifische“ Interaktion). Dieser Unterschied ist scheinbar gering, aber physiologisch überaus relevant. Um Einblick in die Mechanismen der molekularen Erkennung zwischen den Bindungspartnern zu gewinnen, wurden binäre und ternäre Komplexe aus den Liganden BMP-2 oder GDF-5, den extrazellulären Domänen der Typ I Rezeptoren BMPR-IA oder BMPR-IB sowie der extrazellulären Domäne des Typ II Rezeptors ActR-IIB untersucht. Die hier vorliegende Arbeit beschreibt die strukturelle und funktionelle Analyse dieser Ligand-Rezeptor-Komplexe. Um den Einfluss struktureller Flexibilität auf die BMP Typ I Rezeptor Erkennung näher zu analysieren, wurde zudem die Struktur von BMPRIA in freiem Zustand mittels NMR-Spektroskopie aufgeklärt. Aus Mutagenesedaten und der Kristallstruktur des GDF-5•BMPR-IB-Komplexes lassen sich im Vergleich zu bekannten Kristallstrukturen Merkmale ableiten, mit denen die Ligand-Rezeptor-Bindung und -Erkennung charakterisiert werden kann: (1) Die Hauptbindungsdeterminanten in Komplexen von BMPR-IA und BMPR-IB mit ihren Liganden sind unterschiedlich. Während in Komplexen mit BMPR-IB ein hydrophobes Motiv die Bindungsaffinität bestimmt, trägt in Komplexen mit BMPR-IA eine polare Interaktion signifikant zur Bindungsenergie bei. Ein Vergleich der Strukturen von freien und gebundenen Liganden und Typ I Rezeptoren zeigt, dass interessanterweise diese Hauptbindemotive erst bei der Ligand-Rezeptor-Interaktion entstehen, sodass ein „induced fit“ vorliegt und die Moleküle entsprechend „aufeinander falten“. (2) Die Bindungsspezifität wird durch periphere Schleifen in den Typ I Rezeptoren bestimmt. Wie Untersuchungen von Punktmutationen in BMPR-IA zeigen, die einer krebsartigen Darmerkrankung (Juvenile Polyposis) zugrunde liegen, führt erst die „richtige“ Kombination aus Flexibilität in den Schleifen und Rigidität des Rezeptorgrundgerüsts zu signalaktiven Typ I Rezeptoren mit einer potentiell den Liganden komplementären Oberfläche. Die mangelnde sterische Komplementarität von Ligand- und Rezeptoroberflächen führt zu der niedrigeren Bindungsaffinität von GDF-5 zu BMPR-IA im Vergleich zu BMPR-IB. Interessanterweise zeigen die hier vorgestellten, hochaufgelösten Strukturdaten, dass die Orientierungen/Positionen der Typ I Rezeptoren BMPR-IA und BMPR-IB in den Bindeepitopen der Liganden BMP-2 und GDF-5 variieren. Unter der Voraussetzung, dass die extrazelluläre Domäne, das Transmembransegment und die intrazelluläre Domäne der Typ I Rezeptoren ein starres Element bilden, sollte sich die unterschiedliche Orientierung der extrazellulären Domänen der Typ I Rezeptoren in der Anordnung der Kinasedomänen widerspiegeln und sich auf die Signaltransduktion auswirken. Möglicherweise ist eine bestimmte Anordnung der Kinasedomänen der Typ I und Typ II Rezeptoren für eine effiziente Phosphorylierung bzw. Signaltransduktion erforderlich. Der Vergleich mehrerer Ligand-Typ I Rezeptor-Komplexe zeigt, dass die unterschiedliche Orientierung dieser Rezeptoren möglicherweise vom Liganden abhängt. Angesichts der Bindungspromiskuität unter BMP-Liganden und -Rezeptoren könnten so spezifische Signale übermittelt und spezifische biologische Funktionen reguliert werden. Die in dieser Arbeit vorgestellten Erkenntnisse tragen wesentlich zur strukturellen Charakterisierung der Ligand-Rezeptor-Erkennung in der BMP-Familie bei. N2 - Bone morphogenetic proteins (BMPs) are ubiquitous, secreted cytokines involved in a manifold of biological functions. The diversity of cellular processes regulated by BMPs, from bone development to tissue homeostasis and neuronal processes, is amazing – and raises questions about the mechanisms of signal transduction in the light of redundant functions on the one hand, and specific functions on the other hand. Similar to structurally related activins and TGF-βs, the signal transduction of BMPs is accomplished by ligand-induced oligomerization of transmembrane BMP type I and type II serine/threonine receptor kinases. According to current knowledge, only four type I and three type II receptor subtypes are available for BMP signal transduction, facing a multitude of more than 18 BMP ligands. Binding of BMP ligands to their receptors can be highly specific meaning that only one specific receptor of either subtype is used for signaling. In contrast, many BMP ligands can recruit more than one receptor subtype, which results in binding promiscuity. However, even though receptor subtypes are bound in a promiscuous manner, only certain biological functions are triggered. Dependent on the BMP ligand, specific cellular processes are activated and regulated. This discrepancy between unspecific binding and specific signaling events and the biological response raises the question how the formation of heteromeric ligand-receptor complexes is linked to the activation of defined intracellular signaling cascades, and finally, how a certain BMP signal is transduced into the interior of the cell through a „bottleneck“ represented by the limited number of receptor subtypes. The interaction between BMP-2 / GDF-5 and the BMP type I receptors BMPR-IA / BMPR-IB is a prime example for binding promiscuity and binding specificity. BMP-2 binds BMPR-IA and BMPRIB with almost equal binding affinity („promiscuous interaction“) while GDF-5 exhibits a 15-20fold higher binding affinity to BMPR-IB („specific interaction“). Although this difference is seemingly small, it is however of considerable relevance for the physiological role of these ligands. To gain insight into the mechanisms of molecular recognition between the binding partners, binary and ternary ligand-receptor complexes consisting of BMP-2 or GDF-5, the extracellular domains of the type I receptors BMPR-IA or BMPR-IB, and the extracellular domain of the type II receptor ActRIIB were investigated. The thesis presented here describes the structural and functional analysis of these ligand-receptor complexes. To analyse the effect of structural flexibility on BMP type I receptor recognition in more detail, the structure of free BMPR-IA was determined using NMR spectroscopy. Based on data from a limited mutagenesis and the crystal structure of the GDF-5•BMPR-IB complex several characteristics concerning ligand-receptor binding and recognition can be deduced: (1) The main binding determinants in complexes of BMPR-IA and BMPR-IB with their ligands BMP-2 and GDF-5 differ. A hydrophobic binding motif determines binding affinity in complexes of BMPR-IB, whereas a polar interaction significantly contributes to binding energy in complexes of BMPR-IA. These main binding motifs are only formed during complex formation as demonstrated by a comparison between structures of free and bound ligands as well as type I receptors. Both ligand and receptor fold „onto each other“ which suggests an induced fit mechanism. (2) Binding specificity is encoded on loops at the periphery of the binding epitope of the type I receptors. Only the „appropriate“ combination between structural flexibility in the receptor loops and structural rigidity of the receptor backbone results in signal active type I receptors, as shown by analysis of single polymorphisms in BMPR-IA causing juvenile polyposis syndrome, a cancerous disease of the intestine. A lack of steric surface complementarity between GDF-5 and BMPR-IA, that cannot be overcome by structural flexibility, leads to the lower binding affinity in comparison to BMPR-IB. Interestingly, the high resolution structure of the GDF-5•BMPR-IB complex shows that the orientations/positions of BMPR-IA in the binding epitope of BMP-2 and of BMPR-IB in the binding epitope of GDF-5 vary. Assuming that the extracellular domain, the transmembrane segment, and the intracellular domain of the type I receptors form a rigid element, the different orientations of the extracellular domains should also reflect the assembly of the kinase domains and therefore, affect signal transduction. One can assume that a defined arrangement of the kinase domains of type I and type II receptors is required to allow for efficient phosphorylation and signal transduction, respectively. The comparison of several BMP ligand-type I receptor complexes suggests that the different orientations of these receptors are likely dependent on the ligand. Considering the binding promiscuity among BMP ligands and receptors such a mechanism would represent a possible way for the transmission of specific signals and regulation of specific biological functions. The insights into molecular structure and function of BMP ligands and receptors presented in this thesis contribute significantly to a more detailed understanding of their binding properties. The question why the interaction of BMP ligands and receptors is promiscuous on the one hand and specific on the other hand in spite of structurally highly homologous molecules can now be answered for BMP-2 and GDF-5 as well as BMPR-IA and BMPR-IB. The pharmaceutical and crop protection companies still generate a great deal of lead structures based on polyketides. In particular the biosynthesis of aromatic polyketides and the enzymes involved therein, the polyketide synthases (PKSs), have been recognized by researchers as an ideal model system for the study of structure-function relationships in multienzyme complexes. Since 2001, a classification of fused-ring aromatic polyketides at a metabolic level exists: the so-called F/S-mode-system. LEONARD (Ancistrocladaceae) wurde sieben Metabolite isoliert: Amyrin, 3,3-Di-O-methylellagsäure, zwei bisher unbekannte Naphthylisochinoline, Ancistroealain A und B, sowie drei Naphthoesäuren, die hier erstmals beschriebenen Ancistronaphthoesäuren A und B, sowie die bisher nur als Syntheseprodukt bekannte Eleutherolsäure. Ausgehend von Ancistroealain A gelang die stereoselektive Partialsynthese des bekannten Ancistrobertsonin C. Ancistroealain A zeigte in-vitro eine zehnfach höhere Aktivität gegen Leishmania donovani, dem Erreger der visceralen Leishmaniose, als das derzeit bei der Behandlung eingesetzte Pentostam. Um für In-vivo-Untersuchungen genug Material zur Verfügung stellen zu können, wurde ein totalsynthetischer Zugang etabliert. Die Suzuki-Kupplung eines geeigneten Isochinolin-Bausteines (zehn Stufen ausgehend von 3,5-Dimethoxybenzoesäure) mit einer Naphthalin-Boronsäure (acht Stufen ausgehend von 3-Methoxybenzaldehyd) führte in einer Gesamtausbeute von 9.2 % bzw. 6.2 % zu dem Naturstoff. Ancistroealain A und sein Atropdiastereomer Ancitrotanzanine B, die an einer chiralen HPLC-Phase getrennt werden konnten, entstanden aufgrund der asymmetrischen Induktion durch das stereogene Zentrum C-3 in einem Verhältnis von 45:55. Der Ansatz einer atropselektiven Suzuki-Kupplung mit chiralem Katalysator führte zu Diastereomerenüberschüssen bis zu 75:25. Aus Pavetta crassipes K. SCHUMANN (Rubiaceae) konnte das Phythosterol Ursolsäure isoliert werden, während aus Rothmannia urcelliformis (HIERN) BULLOCK (Rubiaceae) 1-epi-Geniposid und Gardenamid A isoliert wurde. Im Rahmen einer Kooperation mit H. Rischer gelang die Isolierung von Plumbagin, Plumbasid A und Rossolisid aus der in Neu Guinea beheimateten tropischen Kannenpflanze Nepenthis insignis DANSER. Bei Verfütterungsexperimenten wurde (L)-[13C3,15N]-Alanin in die Kannen von sterilen Pflanzen eingebracht und ein Einbau in Plumbagin beobachtet. Die Pflanze verstoffwechselt die Aminosäuren auf den üblichen Abbauwegen und erlaubt so die Verfütterung von Alanin als ‚maskiertes’ Acetat. Das beobachtete Einbaumuster bewies die polyketidische Biosynthese von Plumbagin. In einer Kooperation mit Prof. Fiedler (Tübingen) wurden Streptomyceten aus extremen Habitaten auf die Produktion interessanter Sekundärmetabolite untersucht und z.B. bekannte Verbindungen wie Sulfomycin I, Benzoesäure, p-(Dimethylamino)-benzoesäure, Juliochrome Q3-3 und Dehydrorabelomycin nachgewiesen. Der alkalophile Stamm AK 409 produzierte Pyrrol-2-carbonsäure und Pyrocoll, das im Rahmen dieser Arbeit erstmals als Naturstoff auftrat. Besonderes Interesse erregten die Antitumor-Eigenschaften von Pyrocoll. Die durchgeführte ‚biomimetische’ Synthese von Pyrocoll ausgehend von Pyrrol-2-carbonsäure ermöglichte es uns, die für die In-vivo-Biotests nötigen Substanzmengen darzustellen. Aus dem Streptomyceten AK 671 wurden eine bekannte Anthrachinoncarbonsäure und ein als Naturstoff neuartiges Diketonaphthalinglucuronid isoliert. Eine enzymatische Hydrolyse führte zu dem Harris-Franck-Keton, das in dem Kulturfiltrat erstmals als Naturstoff nachgewiesen werden konnte. Das bei Verfütterungsexperimenten mit [13C2]-Acetat von uns beobachtete Einbaumuster in das Glucuronid erlaubte die Aufklärung der Schlüsselschritte der Biogenese. Bei der Synthese von Naphthylisochinolinen besteht die zentrale Aufgabe in dem Aufbau der Biarylachse. Bei der Synthese von Ancistrobertsonin A nach dem ‚Lacton-Konzept’ wird ein Naphtalin-Baustein mit einer zusätzlichen C1-Einheit für die Esterbrücke benötigt, die nach der Kupplung entfernt werden muß. Hierzu bewährte sich bei Versuchen an einem Modelsystem die Reaktionssequenz Baeyer-Villiger-Oxidation, Triflierung und reduktive Eliminierung. Der für die Synthese von Ancistrobertsonin A benötigte Naphthalin-Bausteines wurde in neun Stufen (Gesamtausbeute: 37 % bzw. 13%) dargestellt. Die Synthese des Isochinolin-Bausteines gelang in zwölf Stufen (9.4 %). Der Abschluß dieser Synthese ist in zukünftigen Arbeiten geplant. N2 - The natural product chemistry is a important part of chemistry because natural products, with there broad variety of structural features, are new leads for the development of specific pharmaceuticals and pesticides. Plants and microorganisms are excellent sources for new active compounds in pest control and pharmacy. Ancistrocladus ealaensis J. Léonard (Ancistrocladaceae), a tropical liana indigenous to Central Africa, belongs to the small monogeneric family of the Ancistrocladaceae. Seven metabolites were isolated: the well-known phytosterol -amyrin , 3,3- di-O-methylellagic acid, two new 5,8’-coupled naphthylisoquinoline alkaloids, ancistroealaines A and B , and three naphthoic acids, the synthetically known eleutherolic acid and the two new naphthoic acids ancistronaphthoic acids A and B. Ancistrobertsonine C was synthesized by stereoselective partial-synthetic preparation starting from ancistroealaine A. Against Leishmania donovani, the pathogen of visceral Leishmaniasis, we found excellent activities of ancistroealaine A, ten times more active than the standard pentostam. The synthesis of ancistroealaine A was established to allow further investigations on the in vivo activities. Starting from 3-methoxybenzaldehyde, the naphthalene moiety was prepared in eight steps. The isoquinoline part was synthesized starting from 3,5-dimethoxybenzoic acid in ten steps. The concluding Suzuki coupling resulted in the natural product with an overall yield of 9.2 % or 6.2 % and a diastereomeric ratio of 45:55 induced by the chiral center C-3. The atropoisomeres were separated by chromatography on a chiral phase. The application of a catalytic atroposelective Suzuki coupling gave diastereomeric ratios up to 75:25 Ursolic acid was found in Pavetta crassipes K. SCHUMANN (Rubiaceae), a shrub indigenous to tropical Africa, whereas 1-epi-geniposide and gardenamide A were isolated from Rothmannia urcelliformis (HIERN) BULLOCK, a small tree, which is widespread in the forests of East Africa. In this work, the absolute configuration of gardenamide A was established. From Nepenthes insignis Danser, a species occurring in the lowlands of New Guinea, three metabolites were isolated: Plumbagin, plumbaside A and rossoliside. Feeding experiments with (L)-[13C3,15N]-alanine revealed the acetogenic origin of plumbagin. This work showed that alanine is transformed into acetyl-CoA and can be used as a ‘masked’ precursor. None of the two glycosides were labelled after any of the feeding experiments. They probably constitute storage forms of the respective naphthoquinones. In cooperation with Prof. Fiedler et al. Streptomyces strains, living under extreme conditions, were screened for secondary metabolites. A number of known compounds such as sulfomycine I, benzoic acid, p-(dimethylamino)-benzoic acid, juliochrome Q3-3 and dehydrorabelomycine were found. The alcalophilic strain AK 409 became attractive due to the isolation of two metabolites, pyrrol-2-carboxylic acid and pyrocoll, the latter found as a natural product for the first time. Pyrocoll exhibited in vitro high anticancer activities. ‘Biomimetic’ synthesis of pyrocoll, starting from pyrrol-2-carboxylic acid led to the desired material and showed a better yield (91 %) than all synthetic pathways previously known. From the strain AK671 two natural products were isolated: a known anthraquinone carboxylic acid and a new diketonaphthalene glucuronide. Enzymatic hydrolysis gave the Harris-Franck ketone, a known synthetic compound, which was found as natural product in the culture for the first time. Even though the enzymes that are responsible for the biosynthetic construction of anthraquinones are well known, intermediates as such are isolated only very rarely. Feeding experiments with [13C2]-acetate resolved the biosynthesis of the Harris-Franck ketone and proofed it to be an intermediate in the synthesis of anthraquinones. For the synthesis of ancistrobertsonine A the ‘lactone concept’ was used. Therefore a naphthalene building block with an additional C1-unit next to the axis was needed. The necessary removal of this group (Baeyer-Villiger oxidation, conversion to the triflate and reductive elimination) after the coupling was tested on a model system. The synthesis of ancistrobertsonine A was developed up to the esterification of the naphthalene building block (synthesised in nine steps) and the isoquinoline moiety (synthesised, starting from 3,5-dimethoxybenzoic acid in twelve steps). The synthesis will be finished in future work. Weiterhin gelang eine erste, zudem atropselektive Totalsynthese von Knipholon und verwandten Phenylanthrachinonen, die den ersten stereoselektive Zugang zu Biarylanthrachinonen im allgemeinen darstellt. Schließlich wurden neuartige natürlich vorkommende Phenylanthrachinone entdeckt und deren Strukturen aufgeklärt. N2 - During this thesis using the 'lacton concept' a stereoselective approach to the AB biaryl fragment of vancomycin was established requiring the study of different protective group strategies.
