Für diese Arbeit wurde untersucht, ob sich der Effekt ischämischer Präkonditionierung durch verlängerte oder repetitive Applikation der Ischämie verstärken lässt. Desweiteren wurde untersucht, ob sich die Präkonditionierung mit Desfluran durch Verabreichung einer höheren Konzentration oder verlängerte bzw. repetitive Applikation verstärken lässt. Methodik: In einem akuten in vivo Herzinfarktmodell des weißen Neuseelandkaninchens wurden folgenden Experimente durchgeführt. Alle Versuchstiere durchliefen eine Koronararterienokklusion von 30 min mit anschließender Reperfusion von 180 min. Zur kontinuierlichen ischämischen Präkondtionierung erhielten die Interventionsgruppen zuvor 2 min, 3 min, 5 min bzw. 15 min Ischämie. Zur repetitiven ischämischen Präkonditionierung erhielten sie zwei bzw. drei einminütige oder drei fünfminütige Zyklen Ischämie getrennt von ebenso langen Reperfusionsphasen. Zur anästhetikainduzierten Präkonditionierung erhielten die Versuchstieren vor Ischämie und Reperfusion Desfluran. Entweder kontinuierlich über 30 min oder 90 min oder repetitiv über drei zehnminütige Zyklen getrennt von ebenso langen Abflutungsphasen jeweils in Konzentrationen von 0,5, 1,0 und 1,5 MAC. Im Anschluss an die Experimente wurden die Infarktgrößen gemessen und als prozentualer Anteil des ischämischen Risikoareals dargestellt. Ergebnisse: In der Kontrollgruppe betrug die Infarktgröße 61%. 5 min Ischämie konnten eine Präkonditonierung bewirken (23%). Weder die Behandlung mit 15 min (27%) Ischämie noch mit drei fünfminütigen Zyklen (12%) waren signifikant effektiver als die einfache fünfminütige. Kontinuierliche Ischämien von 2 min (49%) bzw. 3 min (47%) senkten die Infarktgröße nicht. Zwei bzw. drei einminütigen Zyklen wirkten dagegen präkonditionierend (jew. 34%). 1,0 MAC Desfluran über 30min verabreicht senkte die Infarktgröße (35%). Weder eine höhere Konzentration von 1,5 MAC (40%) noch deren Verabreichnung über 90 min (32%) waren signifikant effektiver als 1,0 MAC. 0,5 MAC wirkte weder über 30 min (52%) noch über 90 min (56%) verabreicht präkonditionierend. Eine repetitive Verabreichung über drei zehnminütige Zyklen bewirkte dagegen Präkonditionierung (36%). Zusammnefassung: Die kardioprotektiven Effekte von kontinuierlicher ischämischer bzw. anästhetikainduzierter Präkonditionierung lassen sich oberhalb ihrer jeweiligen Reizschwelle nicht mehr verstärken. Dagegen lassen sich an sich unterschwellige Reize (Ischämie < 5 min bzw. 0,5 MAC Desfluran) durch repetitive Applikation über die Reizschwelle heben und wirken dann präkonditionierend. N2 - Objective: Ischemic and anesthetic preconditioning of the myocardium induce a higher resistance to subsequent ischemia and reperfusion. For this study it was examined, whether longer or repetitive application of ischemia enhances the effects of ischemic preconditioning. It was also examined, whether the effects of Desflurane-induced preconditioning are enhanced by applicating it in higher concentrations, for a longer period of time or in a repetitive manner. Methods: The following experiments were conducted in an in vivo myocardial infarction model of New Zealand White rabbits. All animals underwent 30 min of coronary artery occlusion followed by 180 min of reperfusion. For continuous preconditioning some animals were subjected to 2 min, 3 min, 5 min or 15 min of ischemia afore. For repetitive peconditioning some recieved two or three 1-min-cycles or three 5-min-cycles of ischemia seperated by reperfusion-cycles of the same time. For anesthetic preconditioning animals recieved Desflurane prior to ischemia and reperfusion. Some recieved it continuously over 30 min or 90 min, others repetitivly over three 10-min-cycles seperated by wash-out-periods of the same time. Desflurane concentrations of 0.5, 1.0 and 1.5 MAC were used. After experiments were finished infarct size was messured and expressed as percentage of the ischemic area at risk. Results: Infarct size was 61% in control group. 5 min of ischemic preconditioning lowered infarct size significantly (23%). Neither preconditioning with 15 min (27%) of ischemia was more effective than with 5 min, nor were three 5-min-cycles (12%). Neither 2 min (49%) nor 3 min (47%) peroids of continuous ischemia achieved preconditioning. But so did two and three 1-min-cycles of ischemia (both 34%). 1.0 MAC Desfluran given over 30 min lowered infarct size (35%). A higher concentration of 1.5 MAC (40%) was not significantly more protective than 1.0 MAC, even when it was applied over a period of 90 min (32%). 0.5 MAC could not achieve preconditioning, neither applied over 30 min (52%), nor over 90 min (56%). But given over three 10-min-cycles 0.5 MAC Desfluran was protective (36%). Conclusion: Cardioprotective effects of continuous ischemic and anesthetic preconditioning can not be enhanced as soon as the underlying stimulus has risen above its threshold for preconditioning. But stimuli that usually are beneath the threshold for preconditioning (ischemia < 5 min, 0.5 MAC Desflurane) can be enhanced by repetitive application and can achieve cardioprotection like this. Ein spezifisches Virulenzgen, welches die Schwere der Meningokokkenerkrankung bestimmt, konnte bisher nicht definiert werden. Vorangegangene Studien zeigen eine Korrelation der Letalität mit der Bakterienlast, Unterschiede bezüglich der Letalität je nach Serogruppe, eine erhöhte Letalität bei Infektionen mit sogenannten hyperletalen Feintypen (bisher nicht veröffentlichte Daten des NRZMHi) sowie einen Unterschied in der maximal in Flüssigkultur erreichten Konzentration der Bakterien zwischen invasiven Stämmen und Trägerstämmen. In dieser Arbeit wurden mögliche Gründe für die Hyperletalität bestimmter Meningokokkentypen experimentell untersucht. Insbesondere wird die Frage analysiert, ob die hyperletalen Meningokokkentypen mit einer höheren bakteriellen Last im Blut assoziiert sind und ob sie andere Wachstumscharakteristiken im Vergleich zu ihren Kontrollstämmen in vitro zeigen. Hierzu erfolgte mittels quantitativer Echtzeit-Polymerase-Kettenreaktion die Bestimmung der bakteriellen Last in 62 Blutproben von Patienten mit bestätigter invasiver Meningokokkenerkrankung über den Nachweis des ctrA-Gens. Darunter waren elf Proben des hyperletalen Feintyps B:P1.7-2,4:F1-5 und fünf Proben des hyperletalen Feintyps C:P1.5,2:F3-3. Die Wachstumsversuche wurden mit 30 zufällig gewählten Stämmen der hyperletalen Feintypen B:P1.7-2,4:F1-5, C:P1.5-1,10-8:F3-6 und C:P1.5,2:F3-3 mit ihren jeweiligen nach Alter und Geschlecht abgeglichenen nicht zu der Gruppe der hyperletalen Feintypen gehörenden Kontrollstämmen in dem Medium PPM+ durchgeführt. Die Wachstumsgeschwindigkeit μ sowie die Kapazität A (maximale Konzentrationszunahme als Logarithmus der gemessenen OD im Verhältnis zur Ausgangsdichte ODT0) wurden durch nicht-lineare Regression anhand der modifizierten Gompertz-Funktion ermittelt. Die Messung der optischen Dichte erfolgte alle 30 Minuten über 16 Stunden bei 620nm durch das Gerät TECAN Infinite 200 Pro (Tecan Group Ltd., Männedorf / Schweiz). Die Methode wurde anhand einer publizierten Studie zwischen Trägerstämmen und invasiven Stämmen (Schoen et al., 2014) validiert und bestätigte einen marginalen Unterschied in der optischen Dichte (p=0,057, Wilcoxon-Test) zwischen den Gruppen. Es zeigte sich kein Unterschied in der Wachstumsgeschwindigkeit. Aus den Ergebnissen dieser Arbeit können drei wesentliche Schlussfolgerungen gezogen werden: 1.) Die Bakterienlast in dieser Stichprobe ist, entgegen der Literatur, nicht abhängig von der Serogruppe und dem Feintyp, jedoch von der Krankheitsmanifestation. 2.) Die Kapazität A ist in der Gruppe der „hyperletalen“ Typen im Vergleich zu den Kontrollstämmen möglicherweise höher. 3.) Größere Stichproben (Nativmaterial, Stämme) sind erforderlich, um die Beobachtungen dieser Studie zu bestätigen. N2 - The invasive meningococcal disease still has a case fatality rate of 5 to 10 percent despite antibiotic treatment. A specific virulence gene, which determines disease severity could not be determined so far. Earlier studies have shown a correlation between lethality and bacterial load, differences in lethality between serogroups, a disproportionately high lethality of infections with distinct meningococcal finetypes (data not yet published by the NRZMHi) as well as a difference in the maximum concentration of bacteria in fluid culture between invasive and carrier strains. This dissertation analyses possible causes for the higher fatality rate of certain meningococcal finetypes. It tries to answer the question whether the so called highly lethal meningococcal finetypes are associated with a higher bacterial load in blood and whether they show different growth characteristics in comparison to their control strains in vitro. Bacterial loads in 62 blood samples from patients with confirmed invasive meningococcal disease were measured by real-time polymerase chain reaction targeting the ctrA gene. The samples contained eleven samples of the highly lethal meningococcal finetype B:P1.7-2,4:F1-5 and five samples of the highly lethal meningococcal finetype C:P1.5,2:F3-3. Growth characteristics of 30 randomly selected strains pertaining to the highly lethal finetypes B:P1.7-2,4:F1-5, C:P1.5-1,10-8:F3-6 and C:P1.5,2:F3-3 were compared to strains from age- and sex- matched controls in proteose-peptone-medium (PPM+). Growth rates (μ) and maximum densities (A) were estimated by non-linear regression on the basis of the modified Gompertz function. Optical density at 620nm was measured every 30 minutes over 16 hours using a heatable plate reader (Tecan, Maennedorf, Switzerland). The method was validated on a published sample of invasive and carrier strains (Schoen et al., 2014) confirming marginally significant differences in maximal densities (p=0,057, Wilcoxon test), yet not growth rates, between groups. In summary we conclude: 1. The bacterial load in this sampling is not dependent on serogroup and finetype, but on the clinical picture. This finding contrasts the current literature. 2. The maximum density A is possibly higher in the group of highly lethal finetypes in comparison to the control strains. 3. Larger samples of both patient material and bacterial strains are required to confirm the observations of this study. Lm repliziert nach Befreiung aus dem primären phagosomalen Kompartiment sehr effizient und schnell im Zytosol sehr vieler nicht-phagozytischer Wirtszellen als auch in professionellen antigenpräsentierenden Zellen (APC). Diese Fähigkeit in relevante immunologische APCs einzudringen und zu replizieren, erlaubt die zielgerichtete Übertragung heterologer Antigene in die MHC-Klasse-I- und MHC-Klasse-II-Präsentationswege, um so eine effektive zelluläre Immunität zu etablieren. In der vorliegenden Arbeit wurden die in vivo Effizienzen der Aktivierung von antigen-spezifischen CD8+ and CD4+ T-Zellen miteinander verglichen, sobald das plasmidkodierte Proteinantigen Ovalbumin (OVA) in Form von bakteriell exprimierten und exportierten Proteinen, von cDNA oder mRNA durch die jeweiligen virulenzattenuierten Lm trpS Stämme in das Zytosol von antigenpräsentierenden Zellen freigesetzt wurde. Die Freisetzung wurde durch ein Listeria-spezifisches Phagenlysin, welches von den Bakterien vorwiegend im Zytosol der Wirtszelle exprimiert wird, unterstützt. Die Übertragung dieser unterschiedlichen biologisch-aktiven Moleküle durch die autolysierenden Listerien führte im Falle des Proteins und der mRNA erfolgreich zu einer MHC-Klasse-I-Präsentation eines Ovalbumin-Peptides (SIINFEKL), welche letztendlich eine adaptive zelluläre Immunität unter Beteiligung von T-Gedächtniszellen induzierte. Dabei stellte sich die Übertragung des Proteins durch Lm als die effizienteste Strategie im Induzieren einer zellulären adaptiven Immunantwort mit gegen Ovalbumin gerichteten CD8 und CD4 T-Gedächtniszellen heraus. Autolysierende Listerien, welche die plasmidkodierende OVA-DNA übertrugen, lösten dagegen keine OVA-spezifische T-Zellantwort aus. Da sich der Trägerstamm Lm trpS aufgrund der Autolysiskassette zwar als virulenzattenuiert herausgestellt hatte, jedoch bei höher Applikationsdosis dieses Stammes es nur zu einer unvollständigen Lysis kam, wurden die jeweiligen Effizienzen weiterer noch stärker attenuierter autolysierender Lm Stämme als Überträger des Ovalbumins (Lm Mutanten trpS hlyW491A und (trpS aroA aroB)) bestimmt. Beide ermöglichten die OVA-Präsentation über MHC-Klasse-I-Moleküle mit nachfolgender klonaler Expansion spezifischer CD8+ T-Zellen in vergleichbaren signifikanten Werten zum WT Stamm trpS. Ferner wurde zum ersten Mal eine signifikante Präsentation des OVAs über MHC-Klasse-I-Moleküle durch die autolysierende Mutante trpS hlyW491A, welche die plasmidkodierende DNA freisetzte, nachgewiesen. Die autolysierende Lm (trpS aroA aroB) Mutante in hoher CFU (5107) stellte sich dabei als ein sehr vielversprechender Träger des heterologen Proteinantigens heraus, da sie im Gegensatz zum autolysierenden Stamm Lm trpS eine sehr geringe Leberschädigung hervorrief. In diesem Zusammenhang wurde festgestellt, das durch Freisetzung von OVA Antigenen in das Zytosol oder ins Phagosom von APCs, welche von den jeweiligen Lm (trpS aroA aroB) Stämmen als exportiertes, zellwandverankertes oder intrazellulär verbleibendes Protein exprimiert wurden, vergleichbare Häufigkeiten an proliferierten OVA-spezifischen CD8+ T-Zellen induzierten werden konnten. Es zeigten sich jedoch deutliche Unterschiede in der Aktivierung antigen-spezifischer CD4+ T-Zellen durch diese Lm (trpS aroA aroB) OVA-Trägerstämme. Die Strategie der Übertragung exportierter Proteine ins Phagosom oder ins Zytosol antigenpräsentierender Zellen war die wirkungsvollste, um gleichzeitig effiziente MHC-Klasse-I- und MHC-Klasse-II-restringierte Antigenpräsentationen in vivo zu induzieren. Es wurden alternative plasmidkodierende Lysiskassetten für die Freisetzung von DNA-Vakzinen (Baktofektion) aus den Bakterien konstruiert, die alle aus Lyseproteinen eines Listeria-spezifischen Phagens und einem vorangestellten zytosolischen listeriellen Promotor bestehen. Diese wurden in ihrer Effizienz mit der ursprünglich eingesetzten Lysiskassette PactA-ply118 verglichen. Dabei wurde beobachtet, dass zwei von den vier neukonstruierten Lysiskassetten in einige Zellinien vergleichbare Baktofektionsraten erzielten. Jedoch ist die ursprüngliche Phagenlysin-Kassette PactA-ply118 für die Übertragung von Plasmid-DNA in das Zytosol von Wirtszellen die wirksamste, da diese in vivo zu einer besonders hohen Attenuation der Bakterien führte. N2 - Virulence-attenuated strains of the Gram positive pathogen Listeria monocytogenes (Lm) are optimal candidates for the delivery of heterologous antigens into mammalian hosts. Lm escapes the phagosome, replicates efficiently within the cytosol of many host cells including appropriate cells of the immune system like macrophages and dendritic cells (antigen presenting cells, APCs). These natural biological properties of Lm to enter and replicate in relevant immunological APCs allow the targeted delivery of heterologous antigens into the antigen-processing pathway of MHC class I and MHC class II molecules leading to a strong cellular immunity. In this work, the in vivo efficiences of activation of antigen-specific CD8 and CD4 T cells were compared when the plasmid-encoded protein antigen ovalbumin was secreted by the bacteria as a protein or delivered as cDNA or as mRNA. Delivery was supported also by a specific endolysin produced by the bacteria. Listeriae harbouring this transcriptional unit undergo lysis when they reach the cytosol. In the case of OVA as protein or as mRNA, delivery of these different biological active molecules by autolysing Listeria resulted in significant OVA peptide (SIINFEKL) presentation in the context of MHC class I molecules, which also induced an adaptive cellular immunity with memory T cells. Secretion of OVA by the carrier bacteria yielded the strongest immune response involving OVA-specific memory CD8 and CD4 T cells. In contrast, infection with autolysing Listeria delivering OVA-encoded DNA failed to generate OVA-specific T cells. Due to its harbouring of the autolysing cassette, carrier strain Lm trpS was virulence-attenuated, but infection with 5107 bacteria of this strain resulted in a partial lysis. Therefore investigation of other attenuated and also autolysing strains (Lm mutants trpS hlyW491A and (trpS aroA aroB)) was necessary. Both these strains facilitated OVA presentation via MHC-class-I molecules resulting in clonal expansion of specific CD8+ T cells in comparable frequencies as the wild-type strain. Furthermore, autolysing Lm trpS hlyW491A delivering OVA-encoded DNA led to specific presentation of OVA in the context of MHC class I molecules. Additionally, autolysing Lm (trpS aroA aroB) strain with high infection dose (5107) is a promising carrier for heterologous protein antigens because this mutant exhibited only marginal liver toxicity in contrast to the strain Lm trpS. In this regard, comparable frequencies of proliferated OVA-specific CD8+ T cells were induced through the delivery of OVA antigens into phagosome or cytosol of APCs that were expressed as secreted proteins, as anchored-proteins or as intracellular proteins by the respective carrier Lm strains (trpS aroA aroB). Activation of antigen-specific CD4+ T cells by these OVA-carrier Lm (trpS aroA aroB) was different compared to activation of antigen-specific CD8+ T cells. Secretion of OVA by the carrier bacteria yielded the strongest activation of both MHC-class I and II restricted antigen presentation in vivo. Different lysis cassettes were tested consisting of Listeria-specific phage lysis proteins and an intracellular promoter of Listeria for the delivery of DNA vaccines (bactofection). The efficiency of these cassettes was compared to the original cassette (PactA-ply118). Two of the four new cassettes achieved comparable bactofection rates in some cell lines as the originally used cassette. However, the Lm strain containing the cassette PactA-ply118 was found to be the most effective due to high attenuation in mice. Disease progression, drug discovery, and basic host-pathogen interactions are studied using different approaches, which rely on models ranging from 2D cell culture to complex 3D tissues and animals. In this review, we discuss the models used in N. gonorrhoeae research. We address both in vivo (animal) and in vitro cell culture models, discussing the pros and cons of each and outlining the recent advancements in the field of three-dimensional tissue models. From simple 2D monoculture to complex advanced 3D tissue models, we provide an overview of the relevant methodology and its application. Finally, we discuss future directions in the exciting field of 3D tissue models and how they can be applied for studying the interaction of N. gonorrhoeae with host cells under conditions closely resembling those found at the native sites of infection. Their target spectrum ranges from a single RNA pair, in the case of plasmid-encoded FinO, to global RNA regulons, as with enterobacterial ProQ. To assess whether the FinO domain itself is intrinsically selective or promiscuous, we determine in vivo targets of Neisseria meningitidis, which consists of solely a FinO domain. UV-CLIP-seq identifies associations with 16 small non-coding sRNAs and 166 mRNAs. Meningococcal ProQ predominantly binds to highly structured regions and generally acts to stabilize its RNA targets. Loss of ProQ alters transcript levels of >250 genes, demonstrating that this minimal ProQ protein impacts gene expression globally. Phenotypic analyses indicate that ProQ promotes oxidative stress resistance and DNA damage repair. We conclude that FinO domain proteins recognize some abundant type of RNA shape and evolve RNA binding selectivity through acquisition of additional regions that constrain target recognition. FinO-domain proteins are bacterial RNA-binding proteins with a wide range of target specificities. Here, the authors employ UV CLIP-seq and show that minimal ProQ protein of Neisseria meningitidis binds to various small non-coding RNAs and mRNAs involved in virulence. Here, a qPCR-based method to determine the amount of human DNA in mouse DNA was validated according to the guidelines of the European Medicines Agency and the International Council for Harmonisation of Technical Requirements for Pharmaceuticals for Human Use. Furthermore, a preclinical worst-case scenario study was performed in which this method was applied to investigate the biodistribution of 2 x 10\(^6\) intravenously administered, genetically modified, blood outgrowth endothelial cells from hemophilia A patients after 24 h and 7 days. The validation of the qPCR method demonstrated high accuracy, precision, and linearity for the concentration interval of 1:1 x 10\(^3\) to 1:1 x 10\(^6\) human to mouse DNA. The application of this method in the biodistribution study resulted in the detection of human genomes in four out of the eight investigated organs after 24 h. After 7 days, no human DNA was detected in the eight organs analyzed. This biodistribution study provides mandatory data on the toxicokinetic safety profile of an actual candidate cell-based medicinal product. The extensive evaluation of the required validation parameters confirms the applicability of the qPCR method for non-clinical biodistribution studies. In sensory neurons, cGMP permits signal modulation, amplification and encoding, before depolarization. Here we implement a guanylyl cyclase rhodopsin from Blastocladiella emersonii as a new optogenetic tool (BeCyclOp), enabling rapid light-triggered cGMP increase in heterologous cells (Xenopus oocytes, HEK293T cells) and in Caenorhabditis elegans. Among five different fungal CyclOps, exhibiting unusual eight transmembrane topologies and cytosolic N-termini, BeCyclOp is the superior optogenetic tool (light/dark activity ratio: 5,000; no cAMP production; turnover (20 °C) ~17 cGMPs\(^{-1}\)). Via co-expressed CNG channels (OLF in oocytes, TAX-2/4 in C. elegans muscle), BeCyclOp photoactivation induces a rapid conductance increase and depolarization at very low light intensities. In O\(_2\)/CO\(_2\) sensory neurons of C. elegans, BeCyclOp activation evokes behavioural responses consistent with their normal sensory function. BeCyclOp therefore enables precise and rapid optogenetic manipulation of cGMP levels in cells and animals. At 300 ppm in the diet, the rate of 3.5 X 10\(^{-5}\)(8/236 300) was not significantly increased over background. NOS enzymes in general catalyse NO production. However, under conditions of substrate and cofactor deficiency, the enzyme directly catalyse superoxide formation. Considering this alternative chemistry, the effects of NOS on key events in spontaneous hyperlipidemia driven atherosclerosis have not been investigated yet. Here, we evaluate how endothelial nitric oxide synthase (eNOS) modulates leukocyte/endothelial-(L/E) and platelet/endothelial-(P/E) interactions in atherosclerosis and the production of nitric oxide (NO) and superoxide by the enzyme. Principal Findings: Intravital microscopy (IVM) of carotid arteries revealed significantly increased L/E-interactions in apolipoproteinE/eNOS double knockout mice (apoE\(^{-/-}\)/eNOS\(^{-/-}\)), while P/E-interactions did not differ, compared to apoE\(^{-/-}\). eNOS deficiency increased macrophage infiltration in carotid arteries and vascular cell adhesion molecule-1 (VCAM-1) expression, both in endothelial and smooth muscle cells. Despite the expression of other NOS isoforms (inducible NOS, iNOS and neuronal NOS, nNOS) in plaques, Electron Spin Resonance (ESR) measurements of NO showed significant contribution of eNOS to total circulating and vascular wall NO production. Pharmacological inhibition and genetic deletion of eNOS reduced vascular superoxide production, indicating uncoupling of the enzyme in apoE\(^{-/-}\) vessels. Conclusion: Overt plaque formation, increased vascular inflammation and L/E-interactions are associated with significant reduction of superoxide production in apoE\(^{-/-}\)/eNOS\(^{-/-}\) vessels. Therefore, lack of eNOS does not cause an automatic increase in oxidative stress. Uncoupling of eNOS occurs in apoE\(^{-/-}\) atherosclerosis but does not negate the enzyme's strong protective effects. The experiments revealed that nuclear import and export are conserved among plants and mammals. Further it has been shown that transactivation of a human promoter occurs by human transcription factor NF-\(\kappa\) B in plant cells, demonstrating that the transcriptional machinery is highly conserved in both kingdoms. Functionality was also seen for regulatory elements of NF-\(\kappa\) B such as its inhibitor I\(\kappa\)B isoform \(\alpha\) that negatively regulated the transactivation activity of the p50/RelA heterodimer by interaction with NF-\(\kappa\)B in plant cells. Nuclear export of RelA could be demonstrated by FRAP-measurements so that RelA shows nucleo-cytoplasmic shuttling as reported for RelA in mammalian cells. The data reveals the high level of compatibility of human transcriptional elements with the plant transcriptional machinery. Thus, Arabidopsis thaliana mesophyll protoplasts might provide a new heterologous expression system for the investigation of the human NF-\(\kappa\)B signaling pathways. The system successfully enabled the controlled manipulation of NF-\(\kappa\)B activity. We suggest the plant protoplast system as a tool for reconstitution and analyses of mammalian pathways and for direct observation of responses to e. g. pharmaceuticals. The major advantage of the system is the absence of interference with endogenous factors that affect and crosstalk with the pathway.
