These effects have been studied most extensively in humans and mice. In this work, we used whole genome metagenomics to investigate the relationship between the gut metagenomes of dogs, humans, mice, and pigs. Results We present a dog gut microbiome gene catalog containing 1,247,405 genes (based on 129 metagenomes and a total of 1.9 terabasepairs of sequencing data). Based on this catalog and taxonomic abundance profiling, we show that the dog microbiome is closer to the human microbiome than the microbiome of either pigs or mice. To investigate this similarity in terms of response to dietary changes, we report on a randomized intervention with two diets (high-protein/low-carbohydrate vs. lower protein/higher carbohydrate). We show that diet has a large and reproducible effect on the dog microbiome, independent of breed or sex. Moreover, the responses were in agreement with those observed in previous human studies. Conclusions We conclude that findings in dogs may be predictive of human microbiome results. In particular, a novel finding is that overweight or obese dogs experience larger compositional shifts than lean dogs in response to a high-protein diet. While the task of queen and drones is only reproduction, all other tasks are accomplished by sterile female worker bees. Different tasks are mostly divided by worker bees of different ages (temporal polyethism). Young honeybees perform tasks inside the hive like cleaning and nursing. Older honeybees work at the periphery of the nest and fulfill tasks like guarding the hive entrance. The oldest honeybees eventually leave the hive to forage for resources until they die. However, uncontrollable circumstances might force the colony to adapt or perish. For example, the introduced Varroa destructor mite or the deformed wing virus might erase a lot of in-hive bees. On the other hand, environmental events might kill a lot of foragers, leaving the colony with no new food intake. Therefore, adaptability of task allocation must be a priority for a honeybee colony. In my dissertation, I employed a wide range of behavioral, molecular biological and analytical techniques to unravel the underlying molecular and physiological mechanisms of the honeybee division of labor, especially in conjunction with honeybee malnourishment. The genes AmOARα1, AmTAR1, Amfor and vitellogenin have long been implied to be important for the transition from in-hive tasks to foraging. I have studied in detail expression of all of these genes during the transition from nursing to foraging to understand how their expression patterns change during this important phase of life. My focus lay on gene expression in the honeybee brain and fat body. I found an increase in the AmOARα1 and the Amforα mRNA expression with the transition from in-hive tasks to foraging and a decrease in expression of the other genes in both tissues. Interestingly, I found the opposite pattern of the AmOARα1 and AmTAR1 mRNA expression in the honeybee fat body during orientation flights. Furthermore, I closely observed juvenile hormone titers and triglyceride levels during this crucial time. Juvenile hormone titers increased with the transition from in-hive tasks to foraging and triglyceride levels decreased. Furthermore, in-hive bees and foragers also differ on a behavioral and physiological level. For example, foragers are more responsive towards light and sucrose. I proposed that modulation via biogenic amines, especially via octopamine and tyramine, can increase or decrease the responsiveness of honeybees. For that purpose, in-hive bees and foragers were injected with both biogenic amines and the receptor response was quantified 1 using electroretinography. In addition, I studied the behavioral response of the bees to light using a phototaxis assay. Injecting octopamine increased the receptor response and tyramine decreased it. Also, both groups of honeybees showed an increased phototactic response when injected with octopamine and a decreased response when injected with tyramine, independent of locomotion. Additionally, nutrition has long been implied to be a driver for division of labor. Undernourished honeybees are known to speed up their transition to foragers, possibly to cope with the missing resources. Furthermore, larval undernourishment has also been implied to speed up the transition from in-hive bees to foragers, due to increasing levels of juvenile hormone titers in adult honeybees after larval starvation. Therefore, I reared honeybees in-vitro to compare the hatched adult bees of starved and overfed larvae to bees reared under the standard in-vitro rearing diet. However, first I had to investigate whether the in-vitro rearing method affects adult honeybees. I showed effects of in-vitro rearing on behavior, with in-vitro reared honeybees foraging earlier and for a shorter time than hive reared honeybees. Yet, nursing behavior was unaffected. Afterwards, I investigated the effects of different larval diets on adult honeybee workers. I found no effects of malnourishment on behavioral or physiological factors besides a difference in weight. Honeybee weight increased with increasing amounts of larval food, but the effect seemed to vanish after a week. These results show the complexity and adaptability of the honeybee division of labor. They show the importance of the biogenic amines octopamine and tyramine and of the corresponding receptors AmOARα1 and AmTAR1 in modulating the transition from inhive bees to foragers. Furthermore, they show that in-vitro rearing has no effects on nursing behavior, but that it speeds up the transition from nursing to foraging, showing strong similarities to effects of larval pollen undernourishment. However, larval malnourishment showed almost no effects on honeybee task allocation or physiology. It seems that larval malnourishment can be easily compensated during the early lifetime of adult honeybees. N2 - Honigbienen gehören zu den wenigen Spezies, die in eusozialen Gemeinschaften leben. Die eierlegende Königin und die männlichen Drohnen dienen nur der Fortpflanzung. Alle anderen Arbeiten von den sterilen Arbeiterinnen ausgeführt werden. Die Arbeitsteilung wird meistens anhand des Alters der Bienen organisiert. Junge Arbeiterinnen bleiben im Inneren der Kolonie und führen beispielsweise Putzarbeiten und Ammentätigkeiten aus. Mit zunehmendem Alter verlagern sich ihre Tätigkeiten immer mehr in Richtung des Nestausgangs wo sie, unteranderem als Wächterbienen, den Stockeingang bewachen. Die ältesten Honigbienen verlassen das Nest, um Honig, Pollen, Wasser oder Propolis zu sammeln, bis sie am Ende sterben. Allerdings können unvorhersehbare Ereignisse dazu führen, dass sich die Kolonie anpassen muss, um nicht unterzugehen. Krankheiten wie der Flügeldeformationsvirus oder die, durch den Menschen eingeführte, Varroa destructor Milbe können auf einen Schlag eine große Zahl an Bienen auslöschen. Des Weiteren können beispielsweise starke Unwetter dafür sorgen, dass etliche Sammlerinnen auf ihrem Sammelflug sterben und die Kolonie ohne neuen Nektar oder Pollen zurückgelassen wird. Es liegt auf der Hand, dass eine starre Arbeitsverteilung nicht ausreicht, um solchen Umständen entgegenzuwirken und, dass eine gewisse Flexibilität notwendig ist. In meiner Dissertation habe ich eine weitreichende Anzahl an verhaltensbiologischen und molekularbiologischen Techniken verwendet, um die molekularen und physiologischen Mechanismen der Arbeitsteilung bei Honigbienen aufzuklären, vor allem im Bezug auf den Übergang von Ammenbienen zu Sammlerinnen. Es ist seit langer Zeit bekannt, dass die Gene AmOARα1, AmTAR1, Amfor und Vitellogenin beim Übergang von Ammenbienen zu Sammlerinnen von zentraler Bedeutung sind. Deshalb habe ich die Expression dieser Gene, sowohl im Gehirn als auch im Fettkörper, in genau diesem Zusammenhang betrachtet und die unterschiedlichen Veränderungen der Expressionsmuster während dieser wichtigen Phase im Leben einer Honigbiene analysiert. Ich konnte zeigen, dass sowohl die mRNA Expression des AmOARα1 und des Amforα beim Übergang von Ammenbienen zu Sammlerinnen anstieg, während die Expression der anderen Kandidatengene im gleichen Zeitraum sowohl im Gehirn als auch im Fettkörper abfiel. Interessanterweise zeigten die Expressionsmuster des AmOARα1 und des Am3 TAR1, während der Orientierungsflüge, genau in die entgegengesetzte Richtung. Zusätzlich habe ich mir bei denselben Bienen auch den Juvenilhormongehalt in der Hämolymphe und die Menge an Triglyceriden im Fettkörper angeschaut. Der Juvenilhormongehalt nahm schlagartig zu, als die Bienen mit dem Sammeln begannen. Die Menge an Triglyceriden nahm allerdings von Ammenbienen, über Bienen während des Orientierungsfluges zu Sammlerinnen konstant ab. Des Weiteren war bereits bekannt, dass sich Ammenbienen und Sammlerinnen nicht nur auf genetischer, sondern auch auf verhaltensbiologischer und physiologischer Ebene voneinander unterscheiden. Zum Beispiel sind Sammlerinnen empfindlicher für Licht und Saccharose. Ich stellte die Hypothese auf, dass die Empfindlichkeit von Honigbienen für solche Schwellen durch biogene Amine, insbesondere Oktopamin und Tyramin, moduliert werden kann. Oktopamin sollte die Empfindlichkeit von Bienen erhöhen, wohingegen Tyramin diese verringern sollte. Hierfür injizierte ich Stockbienen und Sammlerinnen beide biogenen Amine und analysierte die Rezeptorantwort mit einem Elektroretinogramm (ERG) und die Lichtempfindlichkeit in einer Phototaxisarena. Oktopamininjektion führte dazu, dass die Rezeptorantwort im ERG erhöht wurde und dass beide Gruppen eine erhöhte Lichtempfindlichkeit aufwiesen. Tyramin hatte in beiden Experimenten genau den gegenteiligen Effekt. Allerdings kann der Ammen-Sammlerinnen-Übergang nicht nur durch biogene Amine moduliert werden, auch die Ernährung hat einen großen Einfluss. Zum Beispiel fangen unterernährte Honigbienen eher an zu sammeln als satte Honigbienen. Des Weiteren sollte auch die larvale Unterernährung bereits einen Einfluss auf die spätere Arbeitsteilung haben, da man bei Arbeiterinnen, die im Larvenstadium bereits unterernährt waren, eine erhöhte Menge an Juvenilhormon festgestellt hatte. Dies sieht man auch beim Übergang von Ammenbienen zu Sammlerinnen. Deshalb nutzte ich eine Methode zur artifiziellen Aufzucht von Honigbienen, um die Standarddiät, die diese normalerweise erhalten, zu variieren. Allerdings musste ich zuerst den Effekt der in-vitro Aufzucht auf im Stock aufgezogene Honigbienen untersuchen. Ich konnte zeigen, dass die artifizielle Aufzucht das Sammelverhalten erwachsener Honigbienen signifikant beeinflusste, während das Ammenverhalten der in-vitro aufgezogenen Bienen nicht beeinflusst wurde. Artifiziell aufgezogene Honigbienen begannen, im Vergleich zu normalen Bienen, früher zu sammeln und sammelten für eine kürzere Zeit. Danach zog ich unterernährte, normal ernährte und überfütterte Honigbienen in-vitro 4 auf. Ich fand Unterschiede im Gewicht zwischen den Behandlungsgruppen. Unterernährte Bienen waren die leichtesten und überfütterte Bienen wogen am meisten. Dieser Unterschied verschwand aber über die Zeit. Des Weiteren konnte ich keinen Einfluss der Ernährung auf das Ammenverhalten oder das Sammelverhalten zeigen. Dieser Ergebnisse zeigen sowohl die Komplexität als auch das Anpassungsvermögen der Arbeitsteilung von Honigbienen. Sie zeigen, dass sowohl die beiden biogenen Amine Oktopamin und Tyramin, als auch die dazugehörigen Rezeptoren AmOARα1 und AmTAR1 bei der Modulation des Ammen-Sammlerinnen-Übergangs eine große Rolle spielen. Des Weiteren zeigen die Ergebnisse des Vergleichs von artifiziell und im Stock aufgezogenen Bienen, starke Gemeinsamkeiten zu einer larvalen Unterernährung mit Pollen. Jedoch scheint eine allgemeine larvale Unterernährung kaum einen Effekt auf den AmmenSammlerinnen-Übergang zu haben. Diese scheint während der ersten Lebenstage von Honigbienen relativ leicht kompensiert werden zu können. While for a long time, a diet as low in nitrates as possible was advocated on the basis of epidemiological data suggesting a cancer-promoting effect of nitrate-rich diets, more recent findings show that dietary nitrate, after its conversion to nitrite by nitrate-reducing bacteria of the oral microbiota, is an indispensable alternative source for the formation of nitric oxide (NO), which comprises a key element in the physiology of a variety of central body functions such as blood pressure control, defense against invading bacteria and maintenance of a eubiotic microbiota in the gut and oral cavity. This compact narrative review aims to present the evidence supported by clinical and in vitro studies on the ambivalent nature of dietary nitrates for general and oral health and to explain how the targeted adjuvant use of nitrate-rich diets could open new opportunities for a more cause-related control of caries and periodontal disease. Results from the Women’s Health Initiative JF - Journal of the American Heart Association N2 - Background Presence of clonal hematopoiesis of indeterminate potential (CHIP) is associated with a higher risk of atherosclerotic cardiovascular disease, cancer, and mortality. The relationship between a healthy lifestyle and CHIP is unknown. Methods and Results This analysis included 8709 postmenopausal women (mean age, 66.5 years) enrolled in the WHI (Women's Health Initiative), free of cancer or cardiovascular disease, with deep‐coverage whole genome sequencing data available. Information on lifestyle factors (body mass index, smoking, physical activity, and diet quality) was obtained, and a healthy lifestyle score was created on the basis of healthy criteria met (0 point [least healthy] to 4 points [most healthy]). CHIP was derived on the basis of a prespecified list of leukemogenic driver mutations. The prevalence of CHIP was 8.6%. A higher healthy lifestyle score was not associated with CHIP (multivariable‐adjusted odds ratio [OR] [95% CI], 0.99 [0.80–1.23] and 1.13 [0.93–1.37]) for the upper (3 or 4 points) and middle category (2 points), respectively, versus referent (0 or 1 point). Across score components, a normal and overweight body mass index compared with obese was significantly associated with a lower odds for CHIP (OR, 0.71 [95% CI, 0.57–0.88] and 0.83 [95% CI, 0.68–1.01], respectively; P‐trend 0.0015). Having never smoked compared with being a current smoker tended to be associated with lower odds for CHIP. Conclusions A healthy lifestyle, based on a composite score, was not related to CHIP among postmenopausal women. However, across individual lifestyle factors, having a normal body mass index was strongly associated with a lower prevalence of CHIP. These findings support the idea that certain healthy lifestyle factors are associated with a lower frequency of CHIP. Status, Verbreitung, Ökologie und Verhalten N2 - Although wild chimpanzees (Pan troglodytes) have been studied intensely for more than 50 years, there are still many aspects of their ecology and behavior that are not well understood. Every time that a new population of chimpanzees has been studied, new behaviors and unknown aspects of their ecology have been discovered. All this accumulated knowledge is helping us to piece together a model of how could last human and chimpanzee common ancestors have lived and behaved between seven and five million years ago. Comoé chimpanzees had never been studied in depth, until we started our research in October 2014, only a few censuses had been realized. The last surveys prior our work, stated that the population was so decimated that was probably functionally extinct. When we started this research, we had to begin with a new intensive survey, using new methods, to ascertain the real status and distribution of the chimpanzees living in Comoé National Park (CNP). During the last five years, we have realized a deep study aiming to know more about their ecology and behavior. We combined transects and reconnaissance marches (recces) with the use of camera traps, for the first time in CNP, obtaining a wealth of data that is not fully comprised in this dissertation. With this research, we determined that there is a sustainable continuous population of Western chimpanzees (Pan troglodytes verus) in CNP and the adjacent area of Mont Tingui, to the West, with a minimum of 127 weaned chimpanzees living in our main 900 km2 study area, SW of CNP. We found that this population is formed by a minimum of eight different chimpanzee communities, of which we studied seven, four of them more in detail. These chimpanzees spent much more time in the forest than in the savanna habitats. We also found that Comoé chimpanzees consumed at least 58 different food items in their dit, which they obtained both from forest and savanna habitats. Another finding was that insectivory had an important role in their diet, with at least four species of ants, three of termites and some beetle larvae. These chimpanzees also hunted at least three species of monkeys and maybe rodents and duikers and occasionally consumed the big land snails of genus Achatina. We found that, during the fruit scarcity period in the late rainy season, they intensely consumed the cambium of Ceiba pentandra, as fallback food, much more than the bark or cambium of any other tree species. Another interesting finding was that all the chimpanzees in the studied area realized this particular bark-peeling behavior and had been repeatedly peeling the trees of this species for years. This did not increase tree mortality and the damage caused to the trees was healed in two years, not reducing the growth, thus being a sustainable use of the trees. We found that Comoé chimpanzees produced and used a great variety of tools, mainly from wooden materials, but also from stone and herbaceous vegetation. Their tool repertory included stick tools to dip for Dorylus burmeisteri ants, to fish for Camponotus and Crematogaster ants, to dip for honey, mainly from Meliponini stingless bees, but sometimes from honey bees (Apis mellifera). It also included the use of stick tools to fish termites of Macrotermes subhyalinus and Odontotermes majus (TFTs), to dip for water from tree holes and investigatory probes for multiple purposes. Additionally, these chimpanzees used leaf-sponges to drink from tree holes and to collect clayish water from salt-licks. They also used stones to hit the buttresses of trees during displays, the so called accumulative stone throwing behavior and probably used stones as hammers, to crack open hard-shelled Strichnos spinosa and Afraegle paniculata fruits and Achatina snails. The chimpanzees also used objects that are not generally accepted as animal tools, for being attached to the substrate, with different purposes: they drummed buttresses of trees with hands and/or feet to produce sound during male displays and they pounded open hard-shelled fruits, Achatina snails and Cubitermes termite mounds on stone or root anvils. We finally measured the stick tools and found significant differences between them suggesting that they were specialized tools made specifically for every purpose. We studied more in detail the differences between apparently similar tools, the honey dipping tools and the water dipping tools, often with brushes made at their tips to collect the fluids. These last tools were exclusive from Comoé and have not been described at any other site. We found that total length, diameter and brush length were significantly different, suggesting that they were specialized tools. We concluded that Comoé chimpanzees had a particular culture, different from those of other populations of Western chimpanzees across Africa. Efficient protection, further research and permanent presence of research teams are required to avoid that this unique population and its culture disappears by the poaching pressure and maybe by the collateral effects of climate change. N2 - Obwohl wild lebende Schimpansen (Pan troglodytes) seit mehr als 50 Jahren intensiv untersucht werden, gibt es noch zahlreiche Aspekte ihrer Ökologie und ihres Verhaltens, die nicht gut verstanden werden. Jedes Mal, wenn eine neue Population von Schimpansen studiert wurde, wurden neue Verhaltensweisen und unbekannte Aspekte ihrer Ökologie entdeckt. All dieses gesammelte Wissen hilft uns, ein Modell zu erstellen, wie lange die gemeinsamen Vorfahren von Menschen und Schimpansen vor sieben bis fünf Millionen Jahren gelebt und sich verhalten haben könnten. Als wir im Oktober 2014 mit unserer Forschung begannen waren die Comoé-Schimpansen, bis auf einige Populations-Zensus von Schimpansen in der Elfenbeinküste, noch nie eingehend untersucht worden. Die letzte Zählung bevor unsere Arbeit began ergab, dass die Schimansenpopulation so stark dezimiert war, dass sie als funktionell ausgestorben erarchtet werden konnte. Zum Beginn unserer Forschung, führten wir zuerst mit neusten Methoden einen neuen detailierten Zensus durch, um den tatsächlichen Status und die Verteilung der im Comoé-Nationalpark (CNP) lebenden Schimpansen zu ermitteln. In den folgenden fünf Jahren haben wir zudem eine umfassende Studie durchgeführt, um mehr über ihre Ökologie und ihr Verhalten zu erfahren. Wir haben im CNP erstmals systematische Transekte und Datenerhebungen mittels Kamerafallen kombiniert, um eine Fülle von Erkenntnissen zu erhalten, die in dieser Dissertation nicht vollständig enthalten sind. Wir stellten fest, dass es in CNP und dem westlich angrenzenden Gebiet des Mont Tingui nach wie vor eine nachhaltige und kontinuierliche Population westlicher Schimpansen (Pan troglodytes verus) existiert, wobei mindestens 131 adulte (entwöhnte) Schimpansen in unserem 900 km² großen Hauptuntersuchungsgebiet südwestlich des CNP leben. Diese Population besteht aus mindestens acht verschiedenen Schimpansengruppen, von denen wir sieben untersuchten, vier davon genauer. Wir konnten zeigen dass diese Schimpansen deutlich mehr Zeit im Wald als in den angrenzenden Savannenhabitaten verbringen. Wir stellten fest, dass Comoé- Schimpansen mindestens 58 verschiedene Futtermittel aus Wald- als auch aus Savannenhabitaten nutzen. Zudem spielt der Konsum von Insekten, bestelhend aus mindestens vier Ameisen-, drei Termiten- und verschiedenene Käferlarven eine wichtige Rolle in ihrem Ernährungsreportoire. Die Comoé-Schimpansen jagen zudem mindestens drei Affena sowie möglicherweise Nagetiere und Duiker, und fraßen gelegentlich die großen Schnecken der Gattung Achatina. Wir fanden heraus, dass sie den typischen Mangel an reifen Fuechten in der \späten Regenzeit durch den intensiven Konsum der Rinde (Kambium) von Ceiba pentandra kompensieren. Alle Schimpansen im untersuchten Gebiet zeigten dieses besondere Verhalten, bei dem sie die Rinde von Ceiba Bäumen schälen. Wir konnte zeigen, dass die Schimpansen diese Bäume seit Jahren wiederholt geschält hatten, was offenbar den Bäumen keinen nachhaltigen Schaden zugefügte. Innerhalb von zwei Jahren ware die Schäden geheilt and das Wachstum nicht verringert, was schlussfolgern lässt dass die Nutzung der Baumrinde nachhaltig ist. Wir fanden heraus, dass Comoé-Schimpansen eine Vielzahl von Werkzeugen aus Vegetation aber auch Steinen herstellten und verwendeten. Das Werkzeugrepertoire umfasste Stöckchen zur Gewinning von on Ameisen der Art Dorylus burmeisteri, sowie Ameisen der Gattungen Camponotus und Crematogaster, aber auch von Bienenhonig produziert von der stachellosen Gattung Melipoa sowie von Apis mellifera. Die Schimpansen nutzen ausserdem Pflanzenwerkzeuge zum Termitenfischen von Macrotermes subhyalinus und Odontotermes majus, um an das Wasser in Baumvertiefungen zu gelangen, sowie für diverse andere Untersuchungszwecke. Zusätzlich verwenden die Comoé-Schimpansen Blattschwämme, um aus Baumlöchern zu trinken und lehmiges Wasser von den Salzlecken zu sammeln. Im Rahmen ihres Imponierverhaltens schleuden sie Steine an die Brettwurzeln spezieller grosser Bäume, ein neu entdecktes Verhalten das als akkumulatives Steinerfen bezeichnet wird. Es ist wahrscheinlich dass sie Steine auch als Hammerwerkzeuge nutzen, um hartschalige Früchte wie Strichnos spinosa und Afraegle paniculata sowie grosse Landschnecken aufzubrechen. Die Schimpansen verwenden Gegenstände auch in anderen Zusammenhängen, die nicht unbedingt als Werkzeuggebrauch definiert werden können: Sie trommlen im Rahmen vom männlichen Imponierverhalten laut mit Händen und Füßen auf die Brettwurzeln von Bäumen, und zerschmettern harte Früchte, Schneckenhäuser und Cubitermes-Termitenhügel auf Ambossen aus Gestein oder Wurzeln. Wir haben signifikante Unterschiede beim Vermessen der Stabwerkzeuge festgestellt, was darauf hindeutet, dass es sich um Spezialwerkzeuge handelt, die speziell für verschiedene Zwecke hergestellt werden. Wir haben insbesondere die Unterschiede zwischen scheinbar ähnlichen Pinselwerkzeugen für den Konsum von Flüssigkeiten (H zu verhindern, sowie die möglichen Nebeneffekte des Klimawandels zu dokumentieren.onig, Wasser) genauer untersucht. Diese Pinselwerkzeuge der Comoé-Schimpansen sind offenbar einzigartig und bislang nicht in der Literatur beschrieben. Gesamtlänge, Durchmesser und Bürstenlänge weichen je nach Verwendungszweck der Pinsel erheblich voneinander ab, was darauf hindeutet, dass es sich um Spezialwerkzeuge handelt. Wir schlussfolgern, dass die Kultur der Comoé-Schimpansen einzigartig innerhalb der der westlichen Schimpansen ist. We aimed to evaluate the relationship between adherence to a Dietary Approaches To Stop Hypertension‐style dietary pattern and the risk of incident AAAs. Methods and Results Dietary intake was assessed via a 66‐item food frequency questionnaire at baseline (1987–1989) and at visit 3 (1993–1995) in 13 496 participants enrolled in the ARIC (Atherosclerosis Risk in Communities) study without clinical AAA (mean age, 54 years). A dietary scoring index based on food times was constructed to assess self‐reported adherence to a dietary approaches to stop hypertension‐style dietary pattern. Participants were followed for incident clinical AAAs using hospital discharge diagnoses, Medicare inpatient and outpatient diagnoses, or death certificates through December 31, 2011. Cox proportional hazards models with covariate adjustment were used to estimate hazard ratios with 95% confidence intervals. During a median follow‐up of 23 years, there were 517 incident AAA cases. Individuals with a Dietary Approaches To Stop Hypertension‐style diet score in the highest quintile had a 40% lower risk of hospitalization for AAA than those in the lowest quintile (hazard ratio\(_{Q5}\) vs \(_{Q1}\): 0.60; 95% confidence intervals: 0.44, 0.83; P\(_{trend}\)=0.002). In detailed analyses, higher consumption of fruits, vegetables, whole grains, low‐fat dairy, and nuts and legumes was related to a lower risk for AAA. Conclusions Greater adherence to a Dietary Approaches To Stop Hypertension‐style dietary pattern was associated with lower risk for AAA. Higher consumption of fruits, vegetables, whole grains, low‐fat dairy as well as nuts and legumes may help to decrease the burden of AAAs. These changes can be associated with abnormalities in social behaviour, hippocampus-dependent cognitive functions, and metabolism. Female C57BL/6J mice were fed with a regular chow or with a WD containing 0.2% of cholesterol and 21% of saturated fat for three weeks. WD-treated mice exhibited increased social avoidance, crawl-over and digging behaviours, decreased body-body contacts, and hyperlocomotion. The WD-fed group also displayed deficits in hippocampal-dependent performance such as contextual memory in a fear conditioning and pellet displacement paradigms. A reduction in glucose tolerance and elevated levels of serum cholesterol and leptin were also associated with the WD. The peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PPARGC1a) mRNA, a marker of mitochondrial activity, was decreased in the prefrontal cortex, hippocampus, hypothalamus, and dorsal raphe, suggesting suppressed brain mitochondrial functions, but not in the liver. This is the first report to show that a WD can profoundly suppress social interactions and induce dominant-like behaviours in naïve adult mice. The spectrum of behaviours that were found to be induced are reminiscent of symptoms associated with autism, and, if paralleled in humans, suggest that a WD might exacerbate autism spectrum disorder. Honey bee larvae are directly exposed to transgenic products by the consumption of GM pollen. But most ERA studies only consider responses of adult bees, although Bt-proteins primarily affect the larval phases of target organisms. We adopted an in vitro larvae rearing system, to assess lethal and sublethal effects of Bt-pollen consumption in a standardized eco-toxicological bioassay. The effects of pollen from two Bt-maize cultivars, one expressing a single and the other a total of three Bt-proteins, on the survival and prepupae weight of honey bee larvae were analyzed. The control treatments included pollen from three non-transgenic maize varieties and of Heliconia rostrata. Three days old larvae were fed the realistic exposure dose of 2 mg pollen within the semi-artificial diet. The larvae were monitored over 120 h, until the prepupal stage, where larvae terminate feeding and growing. Neither single nor stacked Bt-maize pollen showed an adverse effect on larval survival and the prepupal weight. In contrast, feeding of H. rostrata pollen caused significant toxic effects. The results of this study indicate that pollen of the tested Bt-varieties does not harm the development of in vitro reared A. mellifera larvae. To sustain the ecosystem service of pollination, Bt-impact on A. mellifera should always be a crucial part of regulatory biosafety assessments. We suggest that our approach of feeding GM pollen on in vitro reared honey bee larvae is well suited of becoming a standard bioassay in regulatory risk assessments schemes of GM crops. Atherosclerosis is regarded as a chronic inflammatory disease of the vessel wall and DCs can be found in non-inflamed and diseased arteries. We here performed a systematic analyses of DCs subsets during atherogenesis. Our data indicate that distinct DC subsets can be localized in the vessel wall. In C57BL/6 and low density lipoprotein receptor-deficient (Ldlr−/−) mice, CD11c+ MHCII+ DCs could be discriminated into CD103− CD11b+F4/80+, CD11b+F4/80− and CD11b−F4/80− DCs and CD103+ CD11b−F4/80− DCs. Except for CD103− CD11b− F4/80− DCs, these subsets expanded in high fat diet-fed Ldlr−/− mice. Signal-regulatory protein (Sirp)-α was detected on aortic macrophages, CD11b+ DCs, and partially on CD103− CD11b− F4/80− but not on CD103+ DCs. Notably, in FMS-like tyrosine kinase 3-ligand-deficient (Flt3l−/−) mice, a specific loss of CD103+ DCs but also CD103− CD11b+ F4/80− DCs was evidenced. Aortic CD103+ and CD11b+ F4/80− CD103− DCs may thus belong to conventional rather than monocyte-derived DCs, given their dependence on Flt3L-signalling. CD64, postulated to distinguish macrophages from DCs, could not be detected on DC subsets under physiological conditions, but appeared in a fraction of CD103− CD11b+ F4/80− and CD11b+ F4/80+ cells in atherosclerotic Ldlr−/− mice. The emergence of CD64 expression in atherosclerosis may indicate that CD11b+ F4/80− DCs similar to CD11b+ F4/80+ DCs are at least in part derived from immigrated monocytes during atherosclerotic lesion formation. Our data advance our knowledge about the presence of distinct DC subsets and their accumulation characteristics in atherosclerosis, and may help to assist in future studies aiming at specific DC-based therapeutic strategies for the treatment of chronic vascular inflammation. Here, we investigated the role of the co-inhibitory receptor programmed cell death-1 (PD-1) in T cell activation and CD4(+) T cell polarization towards pro-atherogenic or atheroprotective responses in mice. Mice deficient for both low density lipoprotein receptor and PD-1 (Ldlr(-/-)Pd1(-/-)) displayed striking increases in systemic CD4(+) and CD8(+) T cell activation after 9 weeks of high fat diet feeding, associated with an expansion of both pro-atherogenic IFNγ-secreting T helper 1 cells and atheroprotective Foxp3+ Tregs. Importantly, PD-1 deficiency did not affect Treg suppressive function in vitro. Notably, PD-1 deficiency exacerbated atherosclerotic lesion growth and entailed a massive infiltration of T cells in atherosclerotic lesions. In addition, aggravated hypercholesterolemia was observed in Ldlr(-/-)Pd1(-/-) mice. In conclusion, we here demonstrate that although disruption of PD-1 signaling enhances both pro- and anti-atherogenic T cell responses in Ldlr(-/-) mice, pro-inflammatory T cell activation prevails and enhances dyslipidemia, vascular inflammation and atherosclerosis.
