Ein potentielles Antigen für immuntherapeutische Strategien sollte möglichst tumorspezifisch exprimiert sein und es sollte einen Hinweis auf bereits erfolgte Immunantworten im Patienten geben, wie z.B. die Existenz spezifischer Antikörper oder zytotoxischer T-Zellen (CTL). Eine membranständige Lokalisation ist für die Verwendung von Tumorantigenen in Antikörpertherapien notwendig. Während für viele Neoplasien Tumorantigene bekannt sind, wurden für das kutane T-Zell Lymphom (CTCL) bislang nur sehr wenige tumorassoziierte Antigene identifiziert. Die Antigene se57-1, se70-2, cTAGE-1 und GBP-5ta wurden durch serologisches Durchsuchen einer Phagenbank aus Testis- bzw. Tumorgewebe (SEREX-Methode) identifiziert. In der vorliegenden Arbeit wurde die Immunogenität dieser vier Tumorantigene in einem neu entwickelten ELISA mit CTCL-, Parapsoriasis-, Melanom- und Kontrollseren untersucht. se70-2 und cTAGE-1 Protein erkannten nur wenige Patientenseren. Für GBP-5ta konnte dagegen eine signifikant höhere Reaktivität der CTCL-Seren im Vergleich zu den Kontrollseren ermittelt werden. Bei se57-1 waren die CTCL- und die Parapsoriasisseren hoch signifikant verschieden zu den Kontrollseren. Dieses putativ virusinduzierte Antigen sollte in zukünftigen Arbeiten auf seine mögliche Funktion als Entzündungsmarker weiter untersucht werden. Für das CTCL sollten weitere Kombinationen von Tumorantigenen auf ihren diagnostischen Wert in der Serologie getestet werden. Des Weiteren konnten in dieser Arbeit die CTCL assoziierten Antigene se2-2 und die GBP-5 Familie genauer charakterisiert werden: Die Expressionsanalyse von se2-2 Protein und mRNA in verschiedenen Normalgeweben zeigte ein differentielles Expressionsmuster. Im SEREX wurde se2-2 serologisch spezifisch nur von CTCL-Seren erkannt. Möglicherweise wäre se2-2 eine geeignete Zielstruktur für die serologische Diagnostik des CTCL. Aufgrund seiner fehlenden Tumorspezifität ist se2-2 für die Immuntherapie jedoch wenig geeignet. Die neu identifizierte GBP-5 Familie besteht aus mindestens drei Spleißvarianten (GBP-5ta, GBP-5a und GBP-5b), die zwei Proteine, GBP-5ta und GBP-5a/b, kodieren. GBP-5ta ist gegenüber GBP-5a/b C-terminal um 97 AS verkürzt. GBP-5ta mRNA wird differentiell exprimiert, während GBP-5ta Protein PBMC-spezifisch exprimiert wird. In CTCL-Tumorgewebe konnte GBP-5ta nachgewiesen werden, wogegen in Melanomzelllinien fast ausschließlich GBP-5a/b vorliegt. Gegen GBP-5ta konnte eine humorale Immunantwort bei CTCL-Patienten nachgewiesen werden: Im SEREX wurde GBP-5ta nur von CTCL-Patientenseren erkannt. Auch in der ELISA-Methode reagierten signifikant mehr Patientenseren als Kontrollseren mit GBP-5ta. Die höhere Immunogenität von GBP-5ta gegenüber GBP-5a/b im SEREX unterstreicht die Bedeutung der verkürzten Variante. Ob CTL gegen GBP-5ta präsentierende Zellen existieren, wird momentan untersucht. Die GBP-5 Spleißvarianten sind hoch homolog zur Familie der GTPasen, zu denen auch das Onkogen Ras gehört. Das verkürzte Protein von GBP-5ta könnte durch den Verlust der C-terminalen Domäne seine eventuelle anti-proliferierende Funktion verlieren. Ein Knock-out Versuch von GBP-5 könnte die Bedeutung von GBP-5 in der Tumorzelle untersuchen. Darüber hinaus wäre es vielversprechend, die GTPase Aktivität der GBP-5 Varianten in einem GTP-Bindungs-Assay zu überprüfen. GBP-5ta könnte eine mögliche Ursache des unkontrollierten Wachstums der Tumorzelle und somit eine vielversprechende potentielle Zielstruktur für therapeutische Ansätze für das CTCL sein. N2 - Immunotherapies represent a promising concept of tumor-therapies on the basis of well-characterized, tumor-specific antigens. A potential antigen for immunotherapeutic strategies should be preferably tumor-specific expressed and should give a reference to immune responses in the patient, already taken place, as by antibodies or cytotoxic T-cells (CTL). A localization in the membrane is necessarily for antibody therapies. While for many neoplasia tumor antigens are known, the cutaneous t-cell lymphoma (CTCL) so far only very few tumor-associated antigens were identified. The tumor antigens, se57-1, se70-2, cTAGE-1 and GBP-5ta were identified by screening a testis and tumor tissue phage library (SEREX approach). In this work the immunogenicity of this four antigens was investigated in a newly developed ELISA using sera from CTCL, Parapsoriasis and melanoma patients as well as healthy controls. The ELISA results showed that only few patient sera reacted against se70-2 and cTAGE-1. CTCL sera reacted significantly more frequent against GBP-5ta than control sera. se57-1 protein was detected by sera from CTCL and Parapsoriasis patients, but hardly by any control sera. This putativ virus-induced antigen should be further examined for its possible function as inflammation marker. For the CTCL further combinations of tumor antigens should be tested to their diagnostic value. The CTCL associated antigens se2-2 and antigens of the GBP-5 family could be characterized in this work. The expression analysis of se2-2 protein and mRNA in different control tissues showed a differential expression. Secondary screening by SEREX indicated a serological specificity for se2-2. se2-2 could be a suitable target for serological diagnostic of the CTCL but due to its missing expression specificity se2-2 is little suitable for immunotherapy. The newly identified GBP-5 family consists of at least three splicing variants (GBP-5ta, GBP-5a and GBP-5b), coding for two proteins, GBP-5ta and GBP-5a/b. GBP-5ta is C-terminally truncated by 97 aa in comparison to GBP-5a/b. GBP-5ta mRNA is differentially expressed, while GBP-5ta protein is PBMC-specific. GBP-5ta is expressed in CTCL tumor tissue, while in melanoma cell lines almost exclusively GBP-5a/b was found. A humoral immune response in CTCL patients against GBP-5ta could be: SEREX indicated a serological specificity. Accordingly to the ELISA method significantly more patients` sera than control sera reacted against GBP-5ta. The higher immunogenicity of GBP-5ta in comparison to GBP-5a/b underlines the importance of the shortened variant. Whether CTL exist against GBP-5ta epitopes presently examined. The GBP-5 splicing variants are highly homologous to the GTPase superfamily including the ras oncogen. The loss of the C-terminal domain might be one reason why the truncated protein GBP-5ta loses its possible anti-proliferating function. GBP-5 knockout experiments could examine the meaning of GBP-5 in the tumor cell. Beyond that, it would be promising to examine the GTPase activity of the GBP-5 variants in a GTP-binding-assay. GBP-5ta could be a possible cause of the uncontrolled growth of the tumor cell and thus a promising potential target for therapy for the CTCL. A typical accompanying feature is neuroinflammation, the pathogenic impact of which is presently unknown. In this study, the role of inflammatory cells in the pathogenesis was investigated in Palmitoyl-protein thioesterase 1-deficient (Ppt1-/-) and Ceroidlipofuscinosis, neuronal 3-deficient (Cln3-/-) mice, models of the infantile and juvenile forms of NCL, respectively. Focusing predominantly on the visual system, an infiltration of CD8+ cytotoxic Tlymphocytes and an activation of microglia/macrophage-like cells was observed early in disease. To analyze the pathogenic impact of lymphocytes, Ppt1-/- mice were crossbred with mice lacking lymphocytes (Rag1-/-) and axonal transport, perturbation and neuronal survival were scored. Lack of lymphocytes led to a significant amelioration of neuronal disease and reconstitution experiments revealed a crucial role of CD8+ cytotoxic T-lymphocytes. Lack of lymphocytes also caused an improved clinical phenotype and extended longevity. To investigate the impact of microglia/macrophage-like cells, Ppt1-/- and Cln3-/- mice were crossbred with mice lacking sialoadhesin (Sn-/-), a monocyte lineage-restricted cell adhesion molecule important for interactions between macrophage-like cells and lymphocytes. Similar to the lack of lymphocytes, absence of sialoadhesin significantly ameliorated the disease in Ppt1-/- and Cln3-/- mice. Taken together, both T-lymphocytes and microglia/macrophage-like cells were identified as pathogenic mediators in two distinct forms of fatal inherited neurodegenerative storage disorders. These studies expand the concept of secondary inflammation as a common pathomechanistic feature in some neurological diseases and provide novel insights that may be crucial for developing treatment strategies for different forms of NCL. N2 - Die Neuronalen Ceroid Lipofuszinosen (NCL) sind tödlich verlaufende neurodegenerative Erkrankungen, bei denen das visuelle System frühzeitig im Krankheitsverlauf betroffen ist. Eine typische Begleiterscheinung sind Entzündungsreaktionen, deren pathogenetischer Einfluss bisher ungeklärt ist. In dieser Studie wurde die Rolle von Entzündungszellen bei der Pathogenese in Palmitoyl-protein thioestease 1-defizienten (Ppt1-/-) und Ceroid-lipofuscinosis, neuronal 3-defizienten (Cln3-/-) Mäusen untersucht, den jeweiligen Modellen der Infantilen und Juvenilen Formen der NCL. Mit besonderem Augenmerk auf das visuelle System wurde früh in der Krankheit ein Aufkommen von CD8+ zytotoxischen T-Lymphozyten und eine Aktivierung von Mikroglia/Makrophagen-ähnlichen Zellen beobachtet. Um den pathogenetischen Einfluss der Lymphozyten zu klären, wurden Ppt1-/- Mäuse mit Mäusen verkreuzt, welche keine Lymphozyten besitzen (Rag1-/-). Die Abwesenheit von Lymphozyten führte zu einer signifikanten Abmilderung der neuronalen Schädigung und Rekonstitutions-Experimente zeigten, dass CD8+ zytotoxische T-Lymphozyten eine entscheidende Rolle spielen. Die Abwesenheit dieser Lymphozyten führte außerdem zu einem abgemilderten klinischen Phänotyp und einem verlängerten Überleben. Um den Einfluss von Mikroglia/Makrophagen zu untersuchen wurden Ppt1-/- und Cln3-/- Mäuse mit Sialoadhesin-defizienten Mäusen (Sn-/-) verkreuzt. Sn ist ein Monozyten-spezifisches Zelladhäsionsmolekül, das wichtig für Interaktionen zwischen Makrophagen-ähnlichen Zellen und Lymphozyten ist. Ähnlich wie die Abwesenheit von Lymphozyten führte die Abwesenheit von Sialoadhesin zu einer signifikanten Abmilderung der Krankheit in Ppt1-/- und Cln3-/- Mäusen. Zusammengefasst wurden sowohl T-Lymphozyten als auch Mikroglia/Makrophagenähnliche Zellen als pathogenetische Mediatoren in zwei verschiedenen Formen von tödlich verlaufenden erblichen neurodegenerativen Speicherkrankheiten identifiziert. Diese Untersuchungen erweitern das Konzept der sekundären Entzündungsreaktion als verbreitete pathomechanistische Erscheinung in einigen neurologischen Erkrankungen und liefern neue Perspektiven für die Entwicklung von Behandlungsstrategien für verschiedene Formen der NCL. Die Behandlung mit Antikörpern, die murines GM-CSF neutralisieren, zeigte signifikante Behandlungseffekte in Maus-Modellen für Rheumatoide Arthritis, Autoimmune Enzephalomyelitis, entzündliche Erkrankungen der Lunge und Psoriasis. Diese Arbeit beschreibt die Generierung des ersten vollständig humanen Einzelketten (scFv)-Antikörpers, der effektiv humanes GM-CSF (hGM-CSF) neutralisiert. Dieser humane scFv-Antikörper wurde mit Hilfe der Phage-Display-Technologie, ausgehend von einem monoklonalen Ratten-Antikörper mit hGM-CSF-neutralisierender Aktivität, konstruiert. In einem ersten Schritt wurde die VH-Domäne des parentalen Ratten-Antikörpers mit einem humanen leichte Ketten V-Repertoire kombiniert. Nach Phage-Display-Selektion wurde ein dominanter Klon identifiziert, der für ein chimäres Ratte/Human scFv-Fragment kodiert. Dieser scFv-Antikörper neutralisiert hGM-CSF mit einer halbmaximalen inhibitorischen Konzentration (IC50) im nanomolaren Bereich. Die humane leichte Kette dieses Klons wurde dann mit einem humanen schwere Ketten V-Repertoire kombiniert. Um die Bindungsspezifität des Ursprungsantikörpers für den neutralisierenden Bereich auf dem Antigen zu erhalten, enthielt dieses Repertoire die Komplementaritäts-bestimmende Region 3 (CDR3) der parentalen VH-Domäne. Nach Phage-Display-Selektion wurden mehrere scFv-Antikörper identifiziert, die hGM-CSF im niedrig nanomolaren Konzentrationsbereich neutralisierten. Das scFv-Fragment mit der höchsten hGM-CSF-neutralisierenden Aktivität (3077) wurde in verschiedene therapeutisch relevante Antikörperformate überführt. Zum einen wurde das scFv-Fragment über einen zusätzlichen C-terminalen Cystein-Rest an ein verzweigtes, 40 kD-grosses Polyethylenglycol (PEG) –Polymer konjugiert. Zum anderen wurde das scFv-Fragment durch Fusion mit humanen konstanten Immunglobulin-Domänen zu einem ganzen IgG1/kappa-Antikörper vervollständigt. Die so generierten Konstrukte mit identischer Spezifität wurden dann in vitro im Hinblick auf Bindungsaffinität, Neutralisierungsaktivität und Stabilität untersucht. Die Konjugation des PEG-Polymers hatte einen vernachlässigbaren Effekt auf das Neutralisierungspotential des scFv-Fragments in vitro, obwohl sie aufgrund einer verlangsamten Assoziationsrate eine reduzierte Bindungsaffinität verursachte. Der humane IgG1-Antikörper zeigte sowohl eine verbesserte Bindungsaffinität als auch eine erhöhte Neutralisierungsaktivität im Vergleich zum nicht-PEGylierten wie auch zum PEGylierten scFv-Fragment. Wir konnten außerdem zeigen, dass die PEGylierung in der thermischen Stabilität des scFv-Antikörpers einen deutlichen Anstieg um 10°C vermittelte. Aufgrund der hohen Neutralisierungsaktivität und Stabilität des IgG1-Antikörpers 3077 wie auch des PEGylierten scFv-Fragments 3077 sind beide Moleküle - in unterschiedlichen klinischen Anwendungsbereichen - potentielle Kandidaten für eine Behandlung humaner entzündlicher Erkrankungen. N2 - Granulocyte-macrophage colony stimulating factor (GM-CSF) plays a central role in inflammatory processes. Treatment with antibodies neutralizing murine GM-CSF showed significant effects in mouse models of rheumatoid arthritis, lung inflammation, autoimmune encephalomyelitis and psoriasis. This work describes the generation of the first human single-chain antibody (scFv) potently neutralizing human GM-CSF (hGM-CSF). This human scFv fragment was constructed by phage display technology starting from a rat monoclonal antibody with hGM-CSF neutralizing activity. In a first step the VH domain of the parental rat antibody was combined with a human light chain variable (V) repertoire. After phage display selection one dominant clone was identified which encoded a chimeric rat/human scFv antibody neutralizing hGM CSF with a low nanomolar half-maximal inhibitory concentration (IC50). The human light chain of this clone was then combined with a human heavy chain V repertoire. The latter preserved the complementary determining region (CDR) 3 of the parental rat VH domain to retain binding specificity to the neutralizing binding area on the antigen. After phage display selection, several human single-chain antibody fragments were identified that efficiently neutralized human GM-CSF at low nanomolar concentrations. The most potent GM-CSF neutralizing human scFv fragment (3077) was further engineered to different antibody formats with relevance for therapeutic application. On the one hand, the scFv fragment was conjugated to a branched 40 kDa polyethylene glycol (PEG) polymer via an additional C-terminal cysteine residue. On the other hand, the scFv fragment was converted to a human IgG1/kappa-antibody by fusion with human constant immunoglobulin domains. The naked scFv, the PEGylated scFv and the IgG antibody of identical specificity were then compared to each other in terms of binding affinity, neutralizing activity and stability in vitro. PEG conjugation had a neglegible effect on the in vitro neutralizing potential of the scFv fragment although it caused a significant drop in binding affinity due to a reduced on-rate. The human IgG1 antibody variant showed an improved equilibrium dissociation constant as well as neutralizing activity superior to the non-PEGylated and the PEGylated scFv fragment. We also demonstrated that PEGylation mediates a significant increase in thermal stability of about 10°C as compared to the naked scFv fragment. Because of the high neutralizing activity and stability the IgG1 3077 antibody as well as the PEGylated scFv fragment 3077 both are - in different clinical settings - potential candidates to treat pro-inflammatory human diseases.
