The main cell type of cartilage are chondrocytes, previously derived from human mesenchymal stem cells (hMSCs). They are responsible for the production and maintainance of the cartilaginous extracellular matrix (ECM), which consists mainly of collagen and proteoglycans. Signal transmission to or from chondrocytes, generally occurs via interaction with signalling factors connected to the cartilaginous ECM. In this context, proteins of the CCN family were identified as important matricellular and multifunctional regulators with high significance during skeletal development and fracture repair. In this thesis, main focus lies on WISP1/CCN4, which is known as a general survival factor in a variety of cell types and seems to be crucial during lineage progression of hMSCs into chondrocytes. We intend to counter the lack of knowledge about the general importance of WISP1-signalling within the musculoskeletal system and especially regarding cell death and survival by a variety of molecular and cell biology methods. First, we established a successful down-regulation of endogenous WISP1 transcripts within different cell types of the human musculoskeletal system through gene-silencing. Interestingly, WISP1 seems to be crucial to the survival of all examined cell lines and primary hMSCs, since a loss of WISP1 resulted in cell death. Bioinformatical analyses of subsequent performed microarrays (WISP1 down-regulated vs. control samples) confirmed this observation in primary hMSCs and the chondrocyte cell line Tc28a2. Distinct clusters of regulated genes, closely related to apoptosis induction, could be identified. In this context, TRAIL induced apoptosis as well as p53 mediated cell death seem to play a crucial role during the absence of WISP1 in hMSCs. By contrast, microarray analysis of WISP1 down-regulated chondrocytes indicated rather apoptosis induction via MAPK-signalling. Despite apoptosis relevant gene regulations, microarray analyses also identified clusters of differentially expressed genes of other important cellular activities, e.g. a huge cluster of interferon-inducible genes in hMSCs or gene regulations affecting cartilage homeostasis in chondrocytes. Results of this thesis emphasize the importance of regulatory mechanisms that influence cell survival of primary hMSCs and chondrocytes in the enforced absence of WISP1. Moreover, findings intensified the assumed importance for WISP1-signalling in cartilage homeostasis. Thus, this thesis generated an essential fundament for further examinations to investigate the role of WISP1-signalling in cartilage homeostasis and cell death. Den Hauptteil der Zellen im adulten Knorpel stellen die Chondrozyten (Knorpelzellen)dar, welche sich zuvor aus humanen mesenchymalen Stammzellen (hMSCs) entwickelt haben. Sie sind verantwortlich für die Bildung und Aufrechterhaltung der extrazellulären Matrix (ECM) des Knorpelgewebes, welche hauptsächlich aus Kollagen und Proteoglykanen besteht. Signale, die durch Chondrozyten erzeugt oder weitergeleitet werden, finden in der Regel durch Interaktion mit Molekülen der im Knorpel liegenden ECM statt. Mitglieder der CCN-Familie gelten hierbei als bedeutende extrazelluläre Matrixproteine, die bei verschiedenen regulatorischen Prozessen während der Skelettentwicklung und der Frakturheilung eine Rolle spielen. In dieser Doktorarbeit liegt das Hauptaugenmerk auf dem CCN Protein WISP1/CCN4. Dieses Protein gilt bereits in verschiedenen Zellen als ein notwendiger Überlebensfaktor und scheint desWeiteren eine regulatorische Funktion während der Differenzierung von hMSCs in Chondrozyten auszuüben. Die generelle Bedeutung von WISP1 für das muskuloskelettale System ist bislang jedoch weitgehend ungeklärt und soll während dieser Doktorarbeit mittels einer Reihe von molekular- und zellbiologischer Methoden genauer untersucht werden. Hierfür wurde zu Beginn eine erfolgreiche Herunterregulierung endogen hergestellter WISP1 Transkripte mittels Genexpressionshemmung (gene-silencing) in verschiedenen muskuloskelettalen Zellen erzielt. Interessanterweise scheint WISP1 eine bedeutende Rolle für das Überleben dieser Zellen zu spielen, da ein Verlust bei allen untersuchten Zelllinien und primären hMSCs zum Zelltod führte. Um zu Grunde liegende Mechanismen genauer zu untersuchen, wurden daraufhin Microarray Analysen von hMSCs und Tc28a2 Chondrocyten durchgeführt (jeweils WISP1 herunterreguliert vs Kontrollzellen). In diesem Zusammenhang identifizierten bioinformatische Analysen differentielle Expressionen verschiedener apoptoseresponsiver Gene. So scheint eine Apoptoseinduktion über TRAIL und/oder p53 in hMSCs stattzufinden, wohingegen eine starke Regulation des MAPK-Signalweges in Chondrozyten detektiert wurde. Neben diesen Genregulationen, deckten die Analysen ebenso Gengruppen auf, die bei anderen wichtigen zellulären Abläufen eine Rolle spielen. Hier sind in WISP1 herunterregulierten hMSCs u.a. viele differenziell exprimierte Gene zu nennen, die durch Interferone induzierbar sind. In Chondrozyten dagegen scheint eine verringerte WISP1 Expression Genexpressionen zu beeinflussen, welche die Knorpelhomeostase regulieren. Die Ergebnisse, die während dieser Doktorarbeit erzielt wurden, verdeutlichen die Wichtigkeit von WISP1 für das Überleben von primären hMSCs und Chondrozyten. Darüberhinaus verstärken die bioinformatischen Analysen die Annahme, das WISP1 regulatorische Funktionen für die Knorpelhomeostase ausübt. In this study, it could be shown that Arf interacts with Myc and the Myc-associated zinc-finger protein Miz1 to facilitate repression of genes involved in cell adhesion. Formation of a DNA-binding Arf/Myc/Miz1 complex disrupts interaction of Miz1 with its coactivator nucleophosmin and induces local heterochromatinisation, causing cells to lose attachment and undergo anoikis. The assembly of the complex relies on Myc, which might explain why high Myc levels trigger apoptosis and not cell cycle arrest in the Arf response. This mechanism could play an important role in eliminating cells harboring an oncogenic mutation. Arf furthermore induces sumoylation of Miz1 at a specific lysine by repressing the desumoylating enzyme Senp3. A sumoylation-deficient mutant of Miz1 however does not show phenotypic differences under the chosen experimental conditions. Myc can also be modified by Sumo by multisumoylation at many different lysines, which is unaffected by Arf. The exact mechanism and effect of this modification however stays unsolved. N2 - Der Tumorsuppressor Arf wird durch onkogenen Stress induziert und kann entweder einen irreversiblen Zellzyklusarrest oder Apoptose auslösen. In dieser Arbeit konnte gezeigt werden, dass Arf mit Myc und dem Myc-interagierenden Zinkfingerprotein Miz1 assoziiert und dadurch Gene der Zelladhäsion reprimiert. Die Ausbildung eines DNA-bindenden Arf/Myc/Miz1 Komplexes verhindert eine Interaktion von Miz1 mit seinem Koaktivator Nucleophosmin und führt zur lokalen Ausbildung von Heterochromatin, was zum Ablösen der Zellen und schließlich zur Anoikis führt. Die Komplexbildung setzt die Beteiligung von Myc voraus, was erklären könnte warum hohe Mengen an Myc über Arf Apoptose und nicht Zellzyklusarrest auslösen. Dieser Mechanismus könnte eine wichtige Rolle bei der Eliminierung von Zellen mit einer onkogenen Mutation spielen. Arf induziert darüber hinaus die Sumoylierung von Miz1 an einem bestimmten Lysin indem es das desumoylierende Enzyme Senp3 inhibiert. Eine Mutante von Miz1 die nicht mehr sumoyliert werden kann zeigt jedoch in den durchgeführten Untersuchungen keinen anderen Phänotyp als Wildtyp Miz1. These important human pathogens cause diverse diseases including trachoma and wide spread sexually transmitted diseases. Chlamydia establishes membrane bound inclusions in the host cell and loots the host for nutritional requirements. Infections are usually recognized by the host immune system and eliminated systematically, by triggering apoptosis. However, the pathogen Chlamydia has evolved various strategies to prevent the detection as well as protect the invaded cell against apoptosis or any other form of cell death. The evolutionary conservation of cell death regulation has not been investigated in the order Chlamydiales, which also includes Chlamydia-like organisms with a broader host spectrum. The present study was aimed at investigating the apoptotic response of human cells infected with the Chlamydia-like organism Simkania negevensis (Sn). Simkania infected cells exhibited strong resistance to apoptosis induced by intrinsic stress or by the activation of cell death receptors. Apoptotic signaling was blocked upstream of mitochondria since Bax translocation, Bax and Bak oligomerisation and cytochrome c release were absent in these cells. Caspases were differentially regulated upon Sn infection. Caspase-3 and -9 were not activated upon Sn infection and apoptosis induction; whereas caspases-8 was activated in Sn infected cells even without apoptosis induction. This indicates that, Sn utilizes death receptor association independent caspase activation for thriving in the host environment. Infected cells turned on pro-survival pathways like cellular Inhibitor of Apoptosis Proteins (IAP-1/2 and XIAP) and the Akt/PI3K pathway. Sn infection also 20 activated the pro-survival transcription factor NF-кB. Blocking any of these survival pathways sensitized the infected host cell towards apoptosis induction, demonstrating their role in infection-induced apoptosis resistance. The NF-кB mutant cells also showed reduced infectivity of Sn, which indicated an essential role of NF-кB in Sn infection. It was interesting to observe that, Acanthamoeba castellanii, a natural host of Sn, survived maintaining its trophozoite forms after infection with Sn upon starvation. The metacaspases, responsible for encystment could be regulated by Sn upon infection. This suggests an early level of gene regulation indicating how the pathogen evolved its ability to inhibit apoptosis in higher organisms. The resistance to apoptosis pathways subverted in Sn-infected cells was similar but not identical to those modulated by Chlamydia. Together, the data supports the hypothesis of evolutionary conserved signaling pathways to apoptosis resistance as common denominators in the order Chlamydiales. N2 - Vertreter der Ordnung Chlamydiales sind obligat intrazelluläre gram-negative Bakterien mit einer zunehmenden medizinischen Relevanz. Diese Humanpathogene lösen verschiedene Krankheiten aus, unter anderem Blindheit und sexuell übertragbare Krankheiten. Chlamydia führt zur Ausbildung einer Membranumschlossenen Inklusion innerhalb der Wirtszelle und entzieht dem Wirt die nötigen Nährstoffe. Infektionen können vom Immunsystem des Wirts erkannt und systematisch mittels Apoptose bekämpft werden. Chlamydia hat jedoch verschiedene Strategien entwickelt um der Erkennung durch das Immunsystem zu entgehen und um ihre Wirtszelle vor dem Zelltod zu schützen. Ob die Fähigkeit den Zelltod der Wirtszelle zu regulieren evolutionär innerhalb der Ordnung der Chlamydiales, welche auch Chlamydia-ähnliche Organismen mit einem breiteren Wirtsspektrum beinhalten, konserviert ist wurde bislang noch nicht untersucht. In dieser Arbeit wurde das apoptotische Verhalten humaner mit dem Chlamydia-ähnlichen Organismus Simkania negevensis (Sn) infizierter Zellen untersucht. Mit Simkanien infizierte Zellen zeigen eine starke Apoptose-Resistenz, unabhängig ob diese über intrinsischen Stress oder durch die Aktivierung von Todesrezeptoren ausgelöst wurde. Die Apoptose-Signalkaskade wurde vor den Mitochondrien blockiert, da es weder zu einer Bax Translokation, Bax und Bak Oligomerisierung noch zur Cytochrom c Freisetzung in den infizierten Zellen kam. Auch Caspasen zeigten eine veränderte Regulation. Die Caspase-3 und -9 wurden während einer Sn-Infektion und Apoptose-Induktion nicht aktiviert. Die Caspase 8 jedoch ist während der Sn-Infektion unabhängig von einer Apoptose-Induktion aktiv. Diese Tatsache lässt vermuten, dass Sn sich eines von den Todesrezeptoren 22 unabhängigen Mechanismus der Caspaseaktivierung bedienen um in der Wirtzelle zu verbleiben. Infizierte Zellen zeigen eine Aktivierung verschiedener antiapoptotischer Signalwege wie der der Inhibitor of Apoptotis Proteins (IAP1/2 and XIAP)- und den Akt/PI3K Signalweg. Durch eine Sn-Infektion kommt es außerdem zur Aktivierung des Transkriptionsfaktors NF-kB. Wird einer dieser Überlebenssignalwege blockiert zeigen die infizierten Wirtszellen eine erhöhte Sensitivität gegenüber einer Apoptoseinduktion, was wiederum die zentrale Rolle der infektionsvermittelten Apoptoseresistenz aufzeigt. Infektionsversuche mit Sn und dessen natürlichem Wirt Acanthamoeba castellanii zeigten, dass diese im Sn-infizierten Stadium die Aufrechterhaltung ihrer Trophozyten-Form nach Nährstoffmangel überlebten. Die Metacaspasen, welche für die Einkapselung verantwortlich sind konnten durch eine Sn-Infektion reguliert werden. Dies weist auf ein frühes Stadium der Genregulation hin und lässt vermuten wie die Pathogene die Fähigkeit zur Apoptose-Inhibition höherer Organismen erlangt haben könnten. Die durch eine Sn-Infektion vermittelte Resistenz der Wirtszelle gegenüber Apoptose-Signalwegen ist ähnlich, jedoch nicht identisch zu der durch Chlamydia vermittelten Resistenz. It consists of two subunits – GABPa which contains DNA binding domain and GABPb, which provides transcriptional activation domain and nuclear localization signal. GABPa/b complex is essential for transcriptional activation of multiple lineage-restricted and housekeeping genes, several viral genes, and in some cases might function as transcriptional repressor. Large variety of data indicates involvement of GABP in the complex regulation of cell growth, specified by quiescence, stimulation/proliferation, apoptosis and senescence. Expression level of GABPa subunit is rapidly increased when resting cells enter S-phase, and GABPa/b complex is critical to promote the continuity of the cell cycle. Conditional inactivation of GABPa expression in mouse embryonic fibroblasts results in a complete block of proliferation and acquisition of senescence-like phenotype. However, the influence of GABP on the other cell growth determinant – the apoptosis – remains largely obscure. Therefore we aimed to investigate the influence of GABPa/b expression level on the cell growth in vitro. Using siRNA approach we achieved efficient but only transient down-regulation of GABPa expression which precluded further cell growth studies. Persistent increase of the expression of GABPb subunit only resulted in a positive effect on the cell growth speed. Simultaneous conditional overexpression of both GABPa and GABPb subunits though, strongly reduced the growth of the affected cell cultures in reversible and in expression level dependent manner. Interestingly, GABPa/b overexpressing cells did show neither cell cycle arrest nor massive induction of apoptosis. However, more detailed analyses revealed that dampened apoptotic processes were taking place in GABPa/b−overexpressing cells, starting with a prominent activation of caspase-12. Interestingly, activation of downstream effector caspases was rather suppressed explaining a weak increase of apoptotic cells in GABPa/b overexpressing cultures. This effect suggests that the activation of caspase-12 by elevated amounts of exogenous GABPa/b reflects the normal physiological mechanism of caspase-12 regulation. N2 - ZUSAMMENFASSUNG GABP ist ein heterodimerisches Mitglied aus der Familie der Ets- Transkriptionsfaktoren. Es besteht aus zwei Untereinheiten – GABPa, welche die DNA-Bindedomäne enthält, sowie GABPb, welche sowohl die Transkriptions-Aktvierungsdomäne als auch das Kernimportsignal umfasst. GABPa/b ist für die Transkriptions-Aktivierung mehrerer differenzierungstypischer als auch sog. Housekeeping Gene, sowie einiger viraler Gene essentiell und kann, in einigen Fällen, auch als Transkriptionsrepressor fungieren. Eine Vielzahl von Daten deutet darauf hin, dass GABP in der komplexen Kontrolle des Wachstums von Zellen ein wichtige Rolle zukommt. Dies zeigt sich z. B. im Einfluss von GABP auf zelluläre Vorgänge wie der Stimulation/Proliferation, Apoptose und Seneszenz. So steigt z. B. der Spiegel der GABPa Untereinheit rapide an, nachdem ruhende Zellen die G0-Phase verlassen und in die S-Phase eintreten. Der aus beiden Untereinheiten gebildete Komplex ist dann für die Progression der Zellen durch den gesamten Zellzyklus von entscheidender Bedeutung. Die Unterdrückung der Expression der GABPa Untereinheit in embryonalen Mausfibroblasten hingegen führt zu einem vollkommenen Proliferations-Stopp dieser Zellen und induziert in diesen einen Seneszenz-artigen Phänotyp. Andererseits ist über den Einfluss von GABP auf andere wichtige das Zellwachstums beeinflussende Faktoren wie z. B. der Apoptose bislang noch recht wenig bekannt. Daher lag es im Fokus dieser vorliegenden Arbeit, den Einfluss der GABPa/b-Spiegels auf das Zellwachstum in vitro näher zu untersuchen. Mithilfe von siRNA-Ansätzen gelang uns die effiziente Herunterregulierung von GABPa. Diese war jedoch nur von vorübergehender Natur, so dass weitere Studien zum Zellwachstum nicht möglich waren. Die stabile Überexpression der GABPb Untereinheit führte dagegen nur zu einem Anstieg der Zellwachstumsgeschwindigkeit. Wurden jedoch sowohl beide Untereinheit gleichzeitig überexprimiert, so resultierte dies in einer deutlichen, Expressionsspiegel-abhängigen und reversiblen Wachstumshemmung der Zellen. Bemerkenswerterweise zeigte die GABPa/b-überexprimierende Zellpopulation weder einen erhöhten Anteil an G0-Phase noch war eine deutlich ausgeprägte Zunahme der Apoptose-Rate zu verzeichnen. In weiteren Experimenten konnte dennoch eine leichte Erhöhung der Apoptose-Rate in den überexprimierenden Zellen gezeigt werden, was sich durch die deutliche Aktivierung von Caspase-12 belegen ließ. Die Aktivierung von Effektor-Caspasen der Caspase-12 schien allerdings nicht zu erfolgen, was den nur schwach ausgeprägten Charakter der Apoptose zu erklären vermag. Diese Beobachtungen suggerieren, dass die Aktivierung der Caspase-12 durch erhöhte Mengen von exogenem GABPa/b den normalen physiologischen Mechanismus der Caspase-12 Regulation widerspiegelt. In addition to this first wave of NFAT activation, in a second step the occurrence of NFATc1/αA, a short isoform of NFATc1, is strongly induced. Upon primary stimulation of lymphocytes the induction of NFATc1/αA takes place during the G1 phase of cell cycle. Due to an auto-regulatory feedback circuit high levels of NFATc1/αA are kept constant during persistent immune receptor stimulation. Contrary to NFATc2 and further NFATc proteins which dampen lymphocyte proliferation, induce anergy and enhance activation induced cell death (AICD), NFATc1/αA supports antigenmediated proliferation and protects lymphocytes against rapid AICD. Whereas high concentrations of NFATc1/αA can also lead to apoptosis, in collaboration with NF-κB-inducing co-stimulatory signals they support the survival of mature lymphocytes in late phases after their activation. However, if dysregulated, NFATc1/αA appears to contribute to lymphoma genesis and – as we assume – to further disorders of the lymphoid system. While the molecular details of NFATc1/αA action and its contribution to lymphoid disorders have to be investigated, NFATc1/αA differs in its generation and function markedly from all the other NFAT proteins which are expressed in lymphoid cells.
