JF - Zeitschrift für Naturforschung B N2 - In the course of systematic studies on sila-substituted drugs the nifedipine-like 1.4-dihydropyridine derivatives 4a, 4b and 4c were prepared and investigated with respect to sila-substitution effects. By X-ray diffraction analyses 4a, 4b and 4c were found to be isostructural. The C/Si-analogues exhibit similar spasmolytic activities (in vitro, guinea pig ileum), comparable with that of nifedipine. However, the compounds differ substantially in their in vivo activity, as measured by the antihypertensive effect on the renal-hypertensive rat. The experimental results are discussed with respect to the carbon/silicon exchange. To facilitate design of an IL-5 antagonist variant or low-molecular weight mimetics only capable of binding to the specific receptor alpha chain, but would lack the ability to attract the receptor common β-chain and thus initiate receptor complex activation it is necessary to gain the information on minimal structural and functional epitopes. Such a strategy was successfully adopted in our group on example of Interleukin 4. To precisely localize minimal structural epitope it is essential to have structure of the ligand in its bound form and especially informative would be structure of complex of the ligand and its specific receptor alpha chain. For this purpose large quantities (tens of milligrams), retaining full biological activity IL-5 and extracellular domain of IL-5 specific receptor α-chain were expressed in a bacterial expression system (E.coli). After successful refolding proteins were purified to 95-99% Stable and soluble receptor:ligand complex was prepared. Each established purification and refolding procedures were subjected to optimization targeting maximal yields and purity. Produced receptor:ligand complex was applied to crystallization experiments. Microcrystals were initially obtained with a flexible sparse matrix screening methodology. Crystal quality was subsequently improved by fine-tuning of the crystallization conditions. At this stage crystals of about 800x150x30µm in size can be obtained. They possess desirable visible characteristics of crystals including optical clarity, smooth facecs and sharp edges. Crystals rotate plane polarized light reflecting their well internal organization. Unfortunately relative slimness and sometimes cluster nature of the produced crystals complicates acquisition of high-resolution dataset and resolution of the structure. With some of obtained crystals diffraction to a resolution up to 4Å was observed. N2 - Das Ziel der vorliegenden Arbeit war, einen Beitrag zum langfristigen Projekt der Entwicklung humaner IL-5 Agonisten/Antagonisten zu leisten, die im tierischen Krankheitsmodell oder in der Zellkultur in die verschiedenen Funktionen des IL-5 eingreifen oder sie inhibieren. Um das Design eines IL-5 Antagonisten oder einer Mimetika mit geringem Molekulargewicht zu vereinfachen, die nur an die spezifische α-Rezeptorkette bindet, nicht jedoch an die gemeinsame β-Kette und somit die Aktivierung des Rezeptorkomplexes initiieren, ist es notwendig, Informationen über minimale strukturelle und funktionelle Epitope zu erhalten. Diese Strategie wurde in unserer Arbeitsgruppe erfolgreich am Beispiel von Interleukin 4 angewandt. Um minimale strukturelle Epitope präzise zu lokalisieren, ist es es notwendig, die Struktur des Liganden in seiner gebundenen Form zu kennen. Besonders informativ wäre die Struktur des Komplexes aus Ligand und spezifischer Rezeptor α-Kette. Zu diesem Zweck wurden große Mengen (einige 10 mg) IL-5 mit vollständiger biologischer Funktionaltät und der extrazellulären Domäne der IL-5 spezifischen Rezeptor α-Kette in einem bakteriellen Expressionssystem (E. coli) exprimiert. Nach erfolgreicher Faltung wurden die Proteine zu einer Reinheit von 95-99% aufgereinigt und ein stabiler und löslicher Rezeptor:Ligandenkomplex erzeugt. Die erfolgreiche Aufreinigung und Faltungsprozedur wurde bezüglich maximaler Menge und Reinheit optimiert. Mit den produzierten Rezeptor:Ligandenkomplexen wurden Kristallisationsexperimente durchgeführt. Zunächst wurden mit einer flexiblen Sparse-Matrix Screening Methode Mikrokristalle erzeugt. Die Qualität der Kristalle wurde dann durch Feinabstimmung der Kristallisationsbedingungen verbessert. In diesem Stadium wurden Kristalle von etwa 800x150x30µm Größe erzeugt, die die gewünschten optischen Eigenschaften wie glatte Oberflächen und scharfe Kanten besaßen. Die Kristalle drehen die Polarisationsebene linear polarisierten Lichtes, was ihren gleichmäßigen Aufbau zeigt. Leider verkompliziert die geringe Dicke und das teilweise Auftreten von Clustern die Aufnahme hochauflösender Daten und damit Auflösung der Struktur. Mit einigen der erhaltenen Kristalle wurde eine Beugung bis zur Auflösung von 4 Å beobachtet. Specifically, we focus on a projection magnification system with a nano focus source. The imaging resolution is quantified with common 2D test structures and validated in 3D applications by means of the Fourier Shell Correlation. As representative application examples from nowadays material research, we show metallization processes in multilayer integrated circuits, aging in lithium battery electrodes, and volumetric of metallic sub-micrometer fillers of composites. Thus, the laboratory system provides the unique possibility to image non-destructively structures in the range of 170–190 nanometers, even for high-density materials. In order to achieve this, the imaging system must yield an optical resolution of a few micrometers or less, which reduces the measurement area (field of view = FOV) to a few mm at most. If the sample size, however, exceeds the field of view (called local tomography or region of interest = ROI CT), filter problems arise during the CT reconstruction and phase retrieval in holotomography. In this paper, we will first investigate the practical impact of these filter problems and discuss approximate solutions. Secondly, we will investigate the effectiveness of a technique we call “multiscalar holotomography”, where, in addition to the ROI CT, a lower resolution non-ROI CT measurement is recorded. This is used to avoid the filter problems while simultaneously reconstructing a larger part of the sample, albeit with a lower resolution in the additional area. I employ pre-defined samples of beamed AGN: the radio-selected MOJAVE and TANAMI samples, as well as the most recent gamma-ray-selected Fermi/LAT 4LAC catalog. The hard X-ray data is extracted from the 105-month all-sky survey maps of the Swift/BAT (Burst Alert Telescope) in the energy band of 20 keV to 100 keV. A great majority of both the MOJAVE and TANAMI samples are significantly detected, with signal-to noise ratios of the sources often just below the X-ray catalog signal thresholds. All blazar sub-types (FSRQs, BL Lacs) and radio galaxies show characteristic ranges of X-ray flux, luminosity, and photon index. Their properties are correlated with the corresponding SED's shape / peak frequency. The LogN-LogS distributions of the samples show a scarcity of blazars in the middle and lower X-ray flux range, indicating differing evolutionary paths between radio and X-ray emission, which is also suggested by the corresponding luminosity functions. Compared to the radio samples, the 4LAC sources are on average significantly less bright in the BAT band since this range often coincides with the spectral gap region between the two big SED emission bumps. Also, the spectral shapes differ notably, especially for the sub-type of BL Lacs. Using the parameter space of X-ray and gamma-ray photon indices, 35 blazar candidate sources can be assigned to either the FSRQ or BL Lac type with high certainty. The reason why many blazars are weak in this energy band can be traced back to a number of factors: the selection bias of the initial sample, differential evolution of the X-rays and the wavelengths in which the sample is defined, and the limited sensitivity of the observing instruments. N2 - In dieser Arbeit charakterisiere ich die Eigenschaften der harten Röntgenstrahlung von Blazaren, aktiven Galaxienkernen mit stark gebeamter Emission, welche in diesem Energiebereich oft schwer detektierbar sind. Dafür verwende ich vordefinierte AGN-Samples: die im Radiobereich ausgewählten MOJAVE- und TANAMI-Samples sowie den im Gammastrahlenbereich definierten Fermi/LAT 4LAC-Katalog. Die Röntgendaten stammen aus dem 105-monatigen All-Sky-Survey des Swift/BAT (Burst Alert Telescope) bei 20 keV bis 100 keV. Der Großteil der MOJAVE- und TANAMI-Samples sind signifikant detektiert, wobei das Signal/Rausch-Verhältnis der Quellen oft knapp unter dem Limit der Röntgenkataloge liegt. Alle Blazar-Subtypen (FSRQs, BL Lacs) und Radiogalaxien zeigen charakteristische Bereiche des Röntgenflusses, der Leuchtkraft und des Photonenindexes. Ihre Eigenschaften korrelieren mit der Form bzw. Peak-Frequenz der entsprechenden SED. Die LogN-LogS-Verteilungen der Samples zeigen einen Mangel an Blazaren im mittleren und unteren Flussbereich, was auf unterschiedliche Entwicklungen zwischen Radio- und Röntgenemission hinweist, bestätigt durch die entsprechenden Leuchtkraftfunktionen. Im Vergleich zu den Radio-Samples sind die 4LAC-Quellen im BAT-Band durchschnittlich deutlich weniger hell, da dieser Bereich oft mit der großen spektralen Lücke der Blazar-SEDs zusammenfällt. Auch die spektralen Formen unterscheiden sich deutlich, insbesondere für BL Lacs. Unter Verwendung des Parameterraums der Röntgen- und Gammastrahlen-Photonenindizes können 35 Blazarkandidaten mit hoher Sicherheit entweder dem FSRQ- oder BL Lac-Typ zugeordnet werden. Der Grund, warum viele Blazare in diesem Energieband schwach sind, kann auf eine Reihe von Faktoren zurückgeführt werden: den Auswahleffekt des ursprünglichen Samples, die unterschiedliche Entwicklung der Röntgenstrahlung und der Wellenlängen, in denen das Sample definiert ist, sowie die begrenzte Empfindlichkeit der Beobachtungsinstrumente. Profound preoperative panoramic radiography analysis might potentially help predict OAC following tooth extraction. In this exploratory study, we evaluated n = 300 consecutive cases (100 OAC and 200 controls) and trained five machine learning algorithms (VGG16, InceptionV3, MobileNetV2, EfficientNet, and ResNet50) to predict OAC versus non-OAC (binary classification task) from the input images. Further, four oral and maxillofacial experts evaluated the respective panoramic radiography and determined performance metrics (accuracy, area under the curve (AUC), precision, recall, F1-score, and receiver operating characteristics curve) of all diagnostic approaches. Cohen's kappa was used to evaluate the agreement between expert evaluations. The deep learning algorithms reached high specificity (highest specificity 100% for InceptionV3) but low sensitivity (highest sensitivity 42.86% for MobileNetV2). The AUCs from VGG16, InceptionV3, MobileNetV2, EfficientNet, and ResNet50 were 0.53, 0.60, 0.67, 0.51, and 0.56, respectively. Expert 1–4 reached an AUC of 0.550, 0.629, 0.500, and 0.579, respectively. The specificity of the expert evaluations ranged from 51.74% to 95.02%, whereas sensitivity ranged from 14.14% to 59.60%. Cohen's kappa revealed a poor agreement for the oral and maxillofacial expert evaluations (Cohen's kappa: 0.1285). Overall, present data indicate that OAC cannot be sufficiently predicted from preoperative panoramic radiography. The false-negative rate, i.e., the rate of positive cases (OAC) missed by the deep learning algorithms, ranged from 57.14% to 95.24%. Surgeons should not solely rely on panoramic radiography when evaluating the probability of OAC occurrence. Clinical testing of OAC is warranted after each upper-molar tooth extraction.
