Plasmids (pRSF1-14) containing the cross-hybridizing region, covering in total 13.5 kb of chromosomal DNA, were expressed in vitro in a homologous system. One RSF plasmid directed the synthesis of all three main polypeptides of the R. sphaeroides blc1 complex: the FeS protein, cytochrome b and cytochrome c1• The FeS protein and cytochrome c1 were apparently synthesized as precursor fonns. None of the pRSF plasmids directed the synthesis of the 10-kd polypeptide found in b/c1 complex preparations. Partial sequencing of the cloned region was performed. Several sites of strong homology between R. sphaeroides and eukaryotic polypeptides of the b/c1 complex were identified. The genes encode the three b/c1 polypeptides in the order: (5') FeS protein, cytochrome b, cytochrome c1• The three genes are transcribed to give a polycistronic mRNA of 2.9 kb. This transcriptional unit has been designated the jbc operon; its coding capacity corresponds to the size of the polycistronic mRNA assuming that only the genes for the FeS protein (jbcF), cytochrome b (jbcß) and cytochrome c1 (jbcC) are present. This could indicate that these three subunits constitute the minimal catalytic unit of the b/c1 complex from photosynthetic membranes. The relative rates of subunit synthesis directed by the cloned genes were similar in vitro andin vivo and compared favourably with the subunit stoichiometry of the assembled proton-translocating A TP synthase of E. coli in vivo. The rate of synthesis of subunit c was at least six times that of subunit b and 18 times that of subunit ö. Progressive shortening of the long intercistronic sequence lying upstream of the subunit c gene showed that maximal expression of this gene is dependent upon the presence of a sequence stretching > 20 bp upstream of the Shine-Dalgarno site. This sequence thus acts to enhance the rate of translational initiation. The possibility that similar sequences might perform the same function in other operons of E. coli and bacteriophage A is also discussed. Translation of the subunit b cistron is partially coupled to translation of the preceding subunit c cistron. In conclusion, the expression of all the atp operon genes could be adjusted to accommodate the subunit requirements of A TP synthase assembly primarily by means of mechanisms which control the efficiency of translational initiation and re-initiation at the respective cistron start codons. The biological activity was separable by gel filtration and appeared as a broad peak in the moleeular mass range between 10000 and 50000 kDa. It was found that the suppressive activity released by CTLL neither strictly correlates with their cytotoxic potential nor with their ability to produce immune interferon or Iymphotoxin. SF was shown to elicitits activity in an antigen-nonspeeific manner in that it suppressed the maturation of T lymphocytes responding to both, the appropriate H-Y antigen as weH as to unrelated H_2d alloantigens or to the hapten 2,4,6-trinitrophenyl (TNP). The effect of SF on CTL responses was most pronounced in early phases of primary or secondary MLC. When analyzed for its inhibitory activity on precursor ceHs in populations selected for either Lyt-2- or L3T4- lymphocytes, it was found that SF interfered with the maturation of both subsets. The inhibition of CTL responses elicited by SF could not be reversed by the addition of exogenous interleukin 2. The findtng that SF also inhi. bited the proliferation of some but not a11 antigen-dependent cloned T ceHs with helper or eytc'toxic potential provides evidence that the faetor also may regulate effector lymphl)cytes. In addition, the results support the assumption that SF exerts its effect direetly on the responder rather than the stimulator population, and demonstrate that the development of CTL from their preeursor eeHs is contro11ed at least in part by the eytotoxic effeetor cells themselves via a soluble factor(s) that interferes with distinct stages of T ce11 maturation. The resonance integral found in the case of 1 and 3 amounts to -1.9 eV, that for 2 and 4, to -2.3 eV. The investigations furthermore reveal that the electronic factors which contribute to the higher reactivity of the bicyclobutane compounds amount to 5 kcal/mol. The prostaglandin endoperoxide analogue U-46619 is a selective agonist of TxA2 receptors in many peripheral tissues. In the present study the ·central cardiovascular and ventilatory effects of U-46619 were investigated in rats. In conscious spontaneously hypertensive rats (SHR) U-46619 (1-100 nmol/kg i.c.v.) induced a strong dose-related increase in blood pressure but had no significant effect on heart rate. In conscious normotensive rats (NR) neither blood pressure nor heart rate was significantly affected. Furthermore, U-46619 (0.1-100 nmol/kg i.c.v.) had no significant effect on blood pressure, heart rate or ventilation in urethane-anaesthetised NR . In recent years genetic engineering methods have been used to create strains of microorganisms synthesizing eukaryotic proteins, including hormones and their precursors. The first stage of such developments is the isolation of DNA coding the des~red product. We have accomplished the cloning of the cDNA of a number of polypeptide and peptide hormones of the pituitary of man and domestic animals. The model gene of human calcitonin has also been synthesized and cloned. The obtained genes are being used to develop methods for the microbiological synthesis of human and animal-hormones. Die Ergebnisse lassen sich auf andere disperse nicht-transparente Medien wie dichte Gasatmosphären oder einige Zweiphasengemische übertragen, wenn Nicht-Strahlungsanteile und Gesamt-Energieerhaltung korrekt formuliert werden. Die vorliegenden Untersuchungen konzentrieren sich auf stationäre Randbedingungen und Strahlungsquellen. Die Motivation zu dieser Arbeit ist mindestens zweifach: Die Trennung des totalen Wärmestroms in seine Komponenten, in irgendeinem kontinuierlichen oder dispersen Medium, ist eines der herausfordernden, gleichzeitig schwierigsten physikalischen Probleme bei der Analyse des Wärmetransports; zum zweiten ist es für die Verringerung von Wärmeverlusten (z. B. in thermischen Isolierungen) dringend erforderlich, die einzelnen Komponenten der Wärmeverlustströme zu kennen, um sie einzeln zu minimieren (das geht offensichtlich nur, wenn man den totalen Wärmstrom in seine Komponenten zerlegen kann). Die Trennung kann erfolgreich sein, wenn die optische Dicke des untersuchten Mediums sehr groß ist (das Medium ist dann nicht-transparent). In dieser idealen, in der Energietechnik jedoch häufig auftretenden Situation (und nicht nur dort), liefert das Strahlungsdiffusionsmodell den korrekten Ansatz zur Beschreibung des Strahlungsanteils und dessen Temperaturabhängigkeit. Wegen Energieerhaltung und mit der additiven Näherung erlaubt dieses Ergebnis umgekehrt die Berechnung auch der Nichtstrahlungsanteile im totalen Wärmestrom; diese sind demnach alle gleichzeitig in kalorimetrischen Messungen zugänglich. Damit wird nachfolgende separate Analyse dieser Komponenten mittels geeigneter theoretischer Modelle möglich. Da das Temperaturprofil im Medium alle Wärmestromkomponenten zum totalen Wärmestrom miteinander koppelt, ist für diesen Ansatz die Kenntnis der Temperaturabhängigkeit auch aller Nicht-Strahlungsanteile erforderlich. Neben der kalorimetrischen Methode kann die Bestimmung der Extinktion des dispersen Mediums und hiermit des Strahlungstransports auch mittels Spektroskopie sowie Berechnung nach der strengen Mie-Theorie der Lichtstreuung und mit dem Rosseland-Mittelwert vorgenommen werden. Dadurch wird ein Vergleich möglich zwischen Ergebnissen, die mittels drei voneinander völlig unabhängiger Methoden, nämlich kalorimetrisch, spektroskopisch und analytisch/numerisch erzielt wurden. Die Ergebnisse stimmen überein, wenn das Medium nicht-transparent ist; dieser Nachweis wird in der vorliegenden Habilitationsschrift geführt. Im ersten Teil der Habilitationsschrift wird in breit angelegtem Review die Fachliteratur zum Strahlungstransport bis zum Jahr 1985 diskutiert und Methoden zur Lösung der Strahlungstransportgleichung auch im Fall stark anisotroper Streuung beschrieben. Wegen der Forderung nach Energieerhaltung und mit dem oben genannten Ziel, auch die Nicht-Strahlungskomponenten zu analysieren, muß diese Diskussion die theoretischen Aspekte auch dieser Anteile (hier Gas- und Festkörperkontakt-Wärmetransport) einschließen. Den Schluß des ersten Teils bildet ein Katalog offener Fragen, die im zweiten Teil der Habilitationsschrift angegangen werden. Dort werden mittels experimenteller und analytisch/numerischer Ergebnisse das Strahlungsdiffusionsmodell und seine Anwendbarkeit auf disperse nicht-transparente Medien bestätigt. Die Analysen sind gerichtet auf reine oder mit Infrarot-Trübungsmitteln dotierte Pulver und Faserpapiere; beide sind leicht zugängliche, wohl-definierte Testsubstanzen disperser Medien. Ein wichtiger Teil dieser Untersuchungen enthält Messungen ihrer Wärmeleitfähigkeit unter Vakuum und unter externer mechanischer Druckbelastung. Mit evakuierten, druckbelasteten Faserpapieren wurden Wärmeleitfähigkeiten erzielt, die zu den niedrigsten gehören, die bis 1985 an solchen Medien bei hohen Temperaturen gemessen wurden. Weiter sollen optimale Teilchendurchmesser gefunden werden, mit denen das Extinktionsvermögen solcher Schüttungen signifikant erhöht werden kann. Insbesondere ist eine exotische Vorhersage der Mie-Theorie zu prüfen, nach welcher die Extinktion perfekt elektrisch leitender, langer, extrem dünner Zylinder (unter 50 nm) um Größenordnungen über derjenigen herkömmlicher (nichtleitender) Pulver oder Fasern liegt; hierfür sind Materialproben herzustellen. In der Habilitationsschrift wird aufgezeigt, welcher Weg für diesen Nachweis beschritten werden muß (wenige Jahre nach Vorlage der Habilitationsschrift wurden Gustav Mies und Milton Kerkers Vorhersagen auf diesem Weg mit feinsten metallisierten Glasfasern und mit Nickelfasern in Veröffentlichungen des Autors gemeinsam mit J. Fricke, M. Arduini-Schuster, H.-P. Ebert, R. Caps, D. Büttner und A. Kreh erstmalig bestätigt). N2 - The present thesis for habilitation investigates, in compact form, without claiming completeness, the field of heat, in particular radiative, transfer in disperse media if they are non-transparent. Preference is given to high porosity substances being composed of solid particles. The obtained results can be applied to other disperse media like dense gas atmospheres or some two-phase fluids provided non-radiative components of total heat flow, and corresponding total energy balances, are appropriately modelled. The present investigations are concentrated on stationary boundary conditions and radiation sources. Motivation for this work is at least two-fold: First, splitting of total heat flow into its components is one of the most challenging and difficult problems of experimental physics in any, continuous or disperse, medium, and, secondly, reduction of total heat losses (e. g. in thermal insulations) inevitably requests minimisation steps simultaneously to be taken with all its separated components (this of course works only if separation really is successful). Separation can be successful if the optical thickness of the medium is large (the medium then is non-transparent). In this ideal situation that, nonetheless, frequently arises, not only in energy technology, the radiative diffusion model delivers correct expressions for the radiative heat flow component and its temperature dependency. By conservation of energy, this result together with the additive approximation in turn allows determination, from calorimetric measurements, of also the non-radiative contributions to total heat transfer. The approach thus provides a key to subsequently analyse all heat transfer components by appropriate theoretical models. Since the temperature profile in the medium couples all heat transfer components to total heat flow, knowledge of the temperature dependence of also the non-radiative components is indispensable for this purpose. Besides calorimetric methods, spectral measurements of radiation extinction coefficient, and calculation of spectral extinction properties of disperse media by application of rigorous Mie-theory of scattering and of the Rosseland mean, provide another approach to radiative heat flow and to temperature dependent radiation extinction properties. Accordingly, comparison between results obtained from three different, completely independent methods (calorimetric, spectroscopic and theoretical) to determine extinction coefficients can be made and indeed proves to be successful if the medium is non-transparent; this proof of concept shall be demonstrated in the present thesis. In its first part, this thesis for habilitation presents a general review covering the literature on radiative transfer up to the year 1985 and evaluates methods for solution of the equation of radiative transfer also in case of strongly anisotropic scattering. Because of conservation of energy, and in view of the goal to analyse also the non-radiative heat transfer components, the analysis necessarily must include a description of the theoretical aspects of gaseous and solid/solid contact conduction heat transfer mechanisms. At the end of its first part, a catalogue of open questions is presented that will be tackled in the second part of the thesis. There, experimental and analytical/numerical results that verify the radiation diffusion model and its applicability to disperse media are reported. The analysis is focused on heat transfer in pure and opacified powders and fibrous media; both are easily accessible, disperse sample substances with well defined physical/thermal properties. An important part of these investigations covers thermal conductivity measurements under vacuum and under external mechanical load. Experimental results obtained with load-bearing, evacuated boards of glass fibre paper demonstrate smallest values of thermal conductivity that have been obtained until 1985 in such disperse media at high temperatures. The thesis further deals with the problem of how to substantially increase radiation extinction by optimum particle diameters. In particular, an exotic prediction of Mie theory of scattering shall be confirmed according to which the extinction properties of perfectly electrically conducting, long, extremely thin cylinders (below 50 nm) shall be larger, by orders of magnitude, than those of conventional, non-conducting powders or fibres, and samples shall be prepared. The thesis describes the way how to successfully prove this prediction (few years after submission of the thesis, following the methods suggested here and using finest metallised glass fibres, and with very thin Ni-fibres, Gustav Mie’s and Milton Kerker's predictions for the first time were confirmed in publications of the present author together with J. T1 - Cloning and characterization of genes involved in the production of mannose-resistant, neuraminidase-susceptible (X) fimbriae from an uropathogenic O6:K15:K31 Escherichia coli strain N2 - The Qropathogenic Escherichia coli strain 536 (06:K15:H31) exhibits a mannose-resistant hemagglutination phenotype (Mrh) with bovine erythrocytes and delayed Mrh with human and guinea pig erythrocytes. Neuraminidase treatment of the erythrocytes abolishes mannose resistant hemagglutination, which is typical for X fimbriae. E. coli strain 536 synthesizes two different fimbriae (Fim phenotype) prQtein subunits, 16.5 and 22 kilodaltons in size. In addition the strain shows mannose-sensitive hemagglutination and common type I (Fl) fimbriae. The cosmid clone E. coli K-12(pANN801) and another nine independently isolated Mrh+ cosmid clones derived from a cosmid gene bank of strain 536 express the 16.5-kilodalton protein band, bot not the 22-kilodalton protein, indicating an association of the Mrh+ property with the "16.5-kilodalton fimbriae." All cosmid clones were fimbriated, and they reacted with antiserum produced against Mrh+ fimbriae of the E. coli strain HB101(pANN801) and lacked mannose-sensitive hemagglutination (Fl) funbriae. From the Mrh fim cosmid DNA pANN801, several subclones coding for hemagglutination and X fimbriae were constructed. Subclones that express both hemagglutination and fimbriae and subclones that only code for the hemagglutination antigen were isolated; subclones that only produce fimbriae were not detected. By transposon Tn5 mutagenesis we demonstrated that about 6.5 kilobases of DNA is required for the Mrh+ Fim+ phenotype, and the 1.5- to 2-kilobase DNA region coding for the structural proteiil of the fimbriae has been mapped adjacent to the region responsible for the Mrh+ phenotype. Two different regions can thus be distinguished in the adhesion determinant, one coding for hemagglutination and the other coding for fimbria formation. Transformation of plasmid DNA from these subclones into a Mrh- Fim- mutant of E. coli 536 and into a galE (rough) strain of Salmonella typhimurium yielded transformants that expressed both hemagglutination and fimbria production. These mediators were involved in the induction of inftammatory disease processes and led, for example, to enhancement of vascular permeability, chemotaxis (leukotriene 84 [LTB4]), chemoaggregation, lysosomal enzyme release, and smooth muscle contraction, (LTC4, LTD4 , and LTE4). Washed bacteria (E. coli K-12 Ms+ my=; E. coli 536 Ms+ MR= my=) as weil as their culture supematants were analyzed. Washed E. coli K-12 (Hiy+), unlike Hly- strains, induced high amounts of histamine release from rat mast cells and chemotactic activity from human polymorphonuclear granulocytes. Significant leukotriene releasewas obtained with washed E. coli K-12 my+ strains and their bacterial culture supematants. Leukotriene induction was dependent on the amount of hemolysin activity present in the supematant. However, additional soluble factors should also be considered. The presence of hemolysin appeared to aceeierate and enhance the rate of phagocytosis of bacteria by neutrophUs. When E. coli 536 (MS+ MR= Hly=) strains were analyzed, the simultaneous presence of MR+ pili and hemolysin production led to an increase in histamine release as compared with MR- my+ strains. The genetically cloned MR+ my+ E. coli 536 strain induced higher amounts of IeukotrieDes as compared with the wUd-type strain. Our data soggest a potent role for adhesins and hemolysin as virulence factors in inducing the release of inftammatory mediators. A photolabile derivative of R- \(N^6\)-phenylisopropyladenosine, R-2-azido-\(N^6\)-p-hydroxyphenylisopropyladenosine (R-AHPIA), has been synthesized as a covalent specific Iigand for A\(_1\)-adenosine receptors. In adenylate cyclase studies with membranes of rat fat cells and human platelets, R·AHPIA has adenosine receptor agonist activity with a more than 60-fold selectivity for the A\(_1\)-subtype. It competes for [\(^3\)H].\(N^6\)- phenylisopropyladenosine binding to Arreceptors of rat brain membranes with a Ki value of 1.6 nM. After UV irradiation, R-AHPIA binds irreversibly to the receptor, as indicated by a loss of [\(^3\)H)\(N^6\)-phenylisopropyladenosine binding afterextensive washing; the K; value for this photoinactivation is 1.3 nM. The p-hydroxyphenyl substituent of R-AHPIA can be directly radioiodinated to give a photoaffinity Iabel of high specific radioactivity (\(^{125}\)I-AHPIA). This compound has a KD value of about 1.5 nM as assessed from saturation and kinetic experiments. Adenosine analogues compete for \(^{125}\)I-AHPIA binding to rat brain membranes with an order of potency characteristic for A\(_1\)-adenosine receptors. N\(^6\) - R-(- )-Phenylisopropyladenosine (R-PIA) was 5-fold less potent than NECA. 5'-N-Methylcarboxamidoadenosine (MECA) and 2-chloroadenosine had EC\(_{50}\)-values of 0.29 and 2.6 \(\mu\)mol/l, whereas adenosine and inosine had no effect. The adenosine receptors in guinea pig Iung can therefore be classified as A\(_2\) receptors. Several xanthine derivatives antagonized the NECA-induced increase in cyclic AMP levels. 1,3-Diethyl-8-phenylxanthine (DPX; K\(_i\) 0.14 \(\mu\)mol/l) was the most potent analogue, followed by 8-phenyltheophylline (K\(_i\) 0.55 \(\mu\)mol/l), 3-isobutyl-1-methylxanthine (IBMX; K\(_i\) 2.9 \(\mu\)mol/l) and theophylline (K\(_i\) 8.1 \(\mu\)mol/l). In contrast, enprofylline (1 mmol/1) enhanced basal and NECA-stimulated cyclic AMP formation. In addition, we attempted to characterize these receptors in binding studies with [\(^3\)H]NECA. The K\(_D\) for [\(^3\)H] NECA was 0.25 \(\mu\)mol/l and the maximal number of binding sites was 12 pmol/mg protein. In competition experiments MECA (K\(_i\) 0.14 \(\mu\)mol/l) was the most potent inhibitor of [\(^3\)H] NECA binding, followed by NECA (K\(_i\) 0.19 \(\mu\)mol/l) and 2-chloroadenosine (K\(_i\) 1.4 \(\mu\)mol/l). These results correlate well with the EC\(_{50}\)- values for cyclic AMP formation in lung slices. However, the K\(_i\)-values of R-PIA and theophylline were 240 and 270 \(\mu\)mol/l, and DPX and 8-phenyltheophylline did not compete for [\(^3\)H]NECA binding sites. Therefore, a complete characterization of A\(_2\) adenosine receptors by [\(^3\)H] NECA binding was not achieved. The sequence at the other (right-hand) end (RS) also shows multiple hybridization with the plasmid pHly152 and the chromosome of some Escherichia coli strains but the nucleotide sequence of this region does not reveal the typical properties of an IS element. Similar arrangements in the regions flanking the hly determinant are also found on various Hly plasmids from uropathogenic E. coli strains. Chromosomal hly determinants Iack both flanking sequences (IS2 and RS) in the immediate vicinity of the hly genes. The sequences immediately upstream of the hlyC gene have been determined from several chromosomal hly determinants and compared with the corresponding sequence of the hly determinant of the plasmid pHly152. We show that these sequences, which contain one promoter (left promoter, phlyL) in all hly determinants tested, vary considerably although common sequence elements can still be identified. In contrast, only relatively few nucleotide exchanges have been detected in the adjacent structural hlyC genes. The A + T content of the 200 bp sequence upstream of hlyC is very high (72 mol% A + T) but even the structural hly genes show a considerably higher A + T content (about 60 mol%) than the E. coli chromosome on average (50 mol% A+T) suggesting that the hly determinant may not have originated in E. coli. Bei weiteren Olefinen wurden die 'Y-Ketoketene des Typs 33 als Zwischenprodukte nachgewiesen oder sind aufgrund der Konstitution der Endprodukte wahrscheinlich. Aus 1 a werden mit Norbomen, Norbornadien, Cyc1openten, trans-Cycloocten und Styrol die Sechsring-Enollactone 10, 13, 15, 19 bzw. 21 und 22 erhalten. 10 isomerisiert mit Kalium-tert-butoxid zu 34, und die Methanolyse dieser Enollactone erbringt die diastereomeren Methy]ester 12 bzw. 35. Das y-Ketoketen 16 aus trans-Cycloocten nimmt schneller Wasser und Methanol zu 17 bzw. 18 auf als es zum Enollacton 19 cyclisiert. cis,trans-l,5-Cyc1ooctadien erbringt über die Stufe des nachgewiesenen y-Ketoketens 40 in einer intramolekularen [2 + 2J-Cyc1oaddition den Tricyclus 41. Tetracyclische Verbindungen des Typs 4 gehen aus der Umsetzung von Benzvalen (2) mit 1 hervor. With other alkenes, y-ketoketenes 33 have been observed, or are assumed to be intennediates, based on the structure of the isolated products. Tbe six-mernbered enollactones 10, 13, 15, 19, and 21 and 22 have been obtained with norbomene, norbornadiene, cyclopentene, trans-cyclooctene, and styrene. Cornpound 10 isomerizes to 34 on treatment with potassium tert-butoxide. The rnethanolyses of these enollactones result in the formation of the diastereomeric methyl esters 12 and 35. The yketoketene 16 forrned from trans-cyc1ooctene gives acid 17 and ester 18 with water and methanol, respectively. These reactions are faster than the cyc1ization to enollactone 19. From cis,trans-l,5-cyclooctadiene the more short-lived y-ketoketene 40 is formed. which undergoes intramolecular [2 + 2] cycloaddition, giving rise to the tricyclic cyclobutanone derivative 41. Tetracyclic products of type 4 are generated from benzvalene (2) and 1. The structures and configurations of compounds 10, 19, and 41 have been determined by means of X -ray analyses. In addition, antigen or lectin could induce the same H -Y -specific T -cell clones to secrete factor(s) expressing helper activities similar to that ofthe cloned T cells. It was found that the presence of rec.hIL 2 results in proliferation of mitogen (LPS)-activated B cell blasts. In addition, it is shown that highly enriched murine B cells can be induced by rec.hIL-2 to proliferate and to develop into antibody-secreting cells (PFC) in the presence of antigen (SRBC). When tested for its effect on B cell preparations enriched for resting (small) or activated (blasted) B lymphocytes, it was found that rec.hIL 2 provides signals for both B cell populations to develop into PFC. In contrast, induction of proliferation by the same lymphokine source was only seen in blasted B cells. The data indicate that IL 2 is involved in the generation of B effector cells by directly acting on their precursors thereby providing differentiation as well as proliferation signals. These particles contain the nuclear 28S rRNA and apparently represent the precursor to the large ribosomal subunit in nucleocytoplasmic transit. By immunoelectron microscopy ribocharin has been localized in the granular component of the nucleolus and in interchromatin granules. During mitosis ribocharin-containing particles are associated with surfaces of chromosomes and are recollected in the reconstituting nucleoli in late telophase. We suggest that ribocharin is a specific component of precursor particles of the large ribosomal subunit, which dissociates from the 65S particle before passage through the nuclear envelope, and is reutilized in ribosome biogenesis. Die Ergebnisse besagten im einzelnen, daß Kinder des Vor- und Grundschulalters stärker die Ersatzleistung als das Verschulden in Gut-Böse-Urteilen berücksichtigten und daß Erwachsene beide Informationen gleichstark in ihre Gut-Böse-Urteile einfließen ließen. Bei beiden Gruppen schienen Verschulden und Ersatz jedoch mit unterschiedlicher Absicherung nicht-additiv zusammenzuwirken. Die Ergebnisse enthielten eine Replikation US-amerikanischer Befunde mit der Strafe-Skala und diesem Szenario, wiesen aber darüber hinaus auf einen Entwicklungstrend in der NonAdditivität hin. Außerdem ließen sie Zweifel an der Allgemeingültigkeit von Aussagen der sozialpsychologischen Equity-Theorie aufkommen, da Schilderungen vollständiger Ersatzleistungen in keiner Gruppe unabhängig vom Verschulden beurteilt wurden. Nevertheless, two haemolysin (Hly) plasmids of E. coli belonging to the inc groups incFllI,lv (pSU316) and incIz (pHly152) were able to be introduced into these strains by conjugation and stably maintained. A considerable percentage of the Hly+ transconjugants obtained had lost parts of their O-side chains, a result of selection for the better recipient capability of « semi-rough» variants rather than the direct influence of the Hly+ plasmids themselves. In contrast to the incF1lI1V plasmid pSU316, which exhibited higher conjugation rates with rough recipients, the incIz plasmid pHly152 was accepted best by smooth strains. Transformation with cloned E. coli haemolysin (hly) determinant was inefficient ( <10-8) for smooth strains, but 102-103 times higher for rough recipients, and was increased by the use of Salmonella-modified DNA. The transform ants and transconjugants were relatively stable and showed the same haemolytic activity as the E. coli donor strains. The virulence of the Hly+ smooth, semi-rough and rough S. typhimurium strains was tested in two mouse models, and neither the mortality rate nor the ability to multiply within the mouse spleen was influenced by the hly determinants. T1 - Localization of ribosomal protein S1 in the granular component of the interphase nucleolus and its distribution during mitosis N2 - Using antibodies to various nucleolar and ribosomal proteins, we define, by immunolocalization in situ, the distribution of nucleolar proteins in the different morphological nucleolar subcompartments. In the present study we describe the nucleolar localization of a specific ribosomal protein (51) by immunofluorescence and immunoelectron microscopy using a monoclonal antibody (R5 1-105). In immunoblotting experiments, this antibody reacts specifically with the largest and most acidic protein of the small ribosomal subunit (51) and shows wide interspecies cross-reactivity from amphibia to man. Beside its localization in cytoplasmic ribosomes, this protein is found to be specifically localized in the granular component of the nucleolus and in distinct granular aggregates scattered over the nucleoplasm. This indicates that ribosomal protein 51, in contrast to reports on other ribosomal proteins, is not bound to nascent pre-rRNA transcripts but attaches to preribosomes at later stages of rRNA processing and maturation. This protein is not detected in the residual nucleolar structures of cells inactive in rRNA synthesis such as amphibian and avian erythrocytes. During mitosis, the nucleolar material containing ribosomal protein 51 undergoes a remarkable transition and shows a distribution distinct from that of several other nucleolar proteins. In prophase, the nucleolus disintegrates and protein 51 appears in numerous small granules scattered throughout the prophase nucleus. During metaphase and anaphase, a considerable amount of this protein is found in association with the surfaces of all chromosomes and finely dispersed in the cell plasm. In telophase, protein 51-containing material reaccumulates in granular particles in the nucleoplasm of the newly formed nuclei and, finally, in the re-forming nucleoli. These observations indicate that the nucleolus-derived particles containing ribosomal protein 51 are different from cytoplasmic ribosomes and, in the living cell, are selectively recollected after mitosis into the newly formed nuclei and translocated into a specific nucleolar subcompartment, i.e ., the granular component. It is assumed that the identifying substances are mainly genetically determined and in this paper I shall discuss possible evolutionary origins. The main part of this account is devoted to the presentation of some aspects of the highly developed individual and kin identification and recognition system in the desert isopod Hemilepistus reaumuri - a pure monogamous species in which pairs together with their progeny form strictly exclusive family units. Amongst other things problems of (i) mate choice, (ii) learning to recognize a partner, (iii) avoiding the un adaptive familiarization with aliens are treated. Monogamy under present conditions is for both sexes the only suitable way of maximizing reproductive success; an extremely strong selection pressure must act against every attempt to abandon monogamy under the given ecological conditions. The family «badges» which are certainly always blends of different discriminator substances are extremely variable. This variability is mainly due to genetical differences and is not environmentally caused. It is to be expected that intra-family variabiliry exists in respect of the production of discriminator substances. Since the common badge of a family is the result of exchanging and mixing individual substances, and since the chemical nature of these discriminators requires direct body contacts in order to acquire those substances which an individual does not produce itself, problems must arise with molting. These difficulties do indeed exist and they are aggravated by the fact that individuals may produce substances which do not show up in the common family badge. An efficient learning capability on the one hand and the use of inhibiting properties of newly molted isopods help to solve these problems. In the final discussion three questions are posed and - partly at least - answered; (i) why are families so strictly exclusive, (ii) how many discriminator substances have to be produced to provide a variability allowing families to remain exclusive under extreme conditions of very high population densities, (iii) what is the structure of the family badge and what does an individual have to learn apart from the badge in order not to mistake a family member for an alien or vice versa. MM of Xiphophorus exhibits tightly packed pigment cells with prominent dendritic processes and interdigitations of their plasma membranes. The most impressive feature of MM cells is the occurrence of Iarge lobulated nuclei with numerous nuclear pores and some nuclear pockets. Abundant spheroidal or ellipsoidal melanosomes (diameter 200-650 nm) and vesicular structures are distributed throughout the cellular dendrites, whereas the perinucJear cytoplasm is free of melanosomes. A further characteristic feature of melanoma cells in fish is the occurrence of melanosome complexes (i.e., "compound melanosomes"). These melanosome complexes consist of a few to numerous melanosomes, which are enveloped by a separate rnembrane. Pinocytotic vesicles couJd be demonstrated with distinct differences in frequency and distribution patterns, indicating differences in the metabolic activities of the cells in the same melanoma. Intercellular junctions are lacking in the MM cells. The conventional TEM technique showed clear advantages in the demonstration of intemal architecture of organelles, whereas FE bad considerable potential in respect to the visualization of membrane surface specializations. Die erwarteten Effekte eines aufgabenspezifischen Trainingsprogramms auf den Zusammenhang zwischen Wissen und Verhalten ließen sich nicht sichern. Anhand eines Kausalmodells ließen sich erste empirische Anhaltspunkte für die Bestätigung der sog.
