T1  - A deletion map of the WAGR region on chromosome II
N2  - The WAGR (Wilms tumor, aniridia, genitourinary anomalies, and mental retardation) region has been assigned to chromosome 11p13 on the basis of overlapping constitutional deletions found in affected individuals. We have utilized 31 DNA probes which map to the WAGR deletion region, together with six reference loci and 13 WAGR-related deletions, to subdivide this area into 16 intervals. Specific intervals have been correlated with phenotypic features, leading to the identification of individual subregions for the aniridia and Wilms tumor loci. Delineation, by specific probes, of multiple intervals above and below the critical region and of five intervals within the overlap area provides a framework map for molecular characterization of WAGR gene loci and of deletion boundary regions.

N2  - A long-range restriction map of part of the short arm of ehromosome 11 including the WAGR region has been constructed using pulsed-field gel electrophoresis and a number of infrequently cutting restriction enzymes. A total of 15.4 Mbp has been mapped in detall, extending from proximal 11p14 to the distal part of 11p12. The map localizes 35 different DNA probes and reveals at least nine areas with features eharaeteristle of BTF islands, some of which may be candidates for the different loci underlying the phenotype of the WAGR syndrome. This map will furthermore allow screening of DNA from individuals with WAGR-related phenotypes and from Wilms tumors for associated chromosomal rearrangements.

T1  - The genomic organization and expression of the WT1 gene
N2  - The Wilms tumor gene WTl, a proposed tumor suppressor gene, has been identifled based on its location within a homozygous deletion found in tumor tissue. The gene encodes a putative transcription factor containing a Cys/His zinc finger domain. The critical homozygous deletions, however, are rarely seen, suggesting that in many cases the gene may be inactivated by more subtle alterations. To facilitate the seareh for smaller deletions and point mutations we have established the genomic organization of the WTl gene and have determined the sequence of all 10 exons and flanking intron DNA. The pattern of alternative splicing in two regions has been characterized in detail. These results will form the basis for future studies of mutant alleles at this locus.

T1  - The potassium channel gene HK1 maps to human chromosome 11p14.1, close to the FSHB gene
N2  - Transiently activating (A-type) potassium (K) channels are important regulators of action potential and action potential firing frequencies. HK1 designates the firsthuman cDNA that is highly homologous to the rat RCK4 cDNA that codes for an A-type K-channel. The HK1 channel is expressed in heart. By somatic cell hybrid analysis, the HK1 gene has been assigned to human chromosome 11p13-pl4, the WAGR deletion region (Wilms tumor, aniridia, genito-urinary abnormalities and mental retardation). Subsequent pulsed field gel (PFG) analysis and comparison with the well-established PFG map of this region localized the gene to 11p14, 200-600 kb telomeric to the FSHB gene.

N2  - The combined use of qualitative and quantitative analysis of I I p I 3 polymorphic markers tagether with chromosomal in situ suppression hybridization (CISS) with biotin labeled probes mapping to I I p allowed us to characterize a complex rearrangement segregating in a family. We detected a pericentric intrachromosomal insertion responsible (or recurrence of del( I I )(p 13p 14) in the family: an insertion of band I I p 13-p 14 carrying the genes for predisposition to Wilms' tumor, WT I, and for aniridia, AN2, into the long arm of chromosome I I in II q 13-q 1<4. Asymptomatic balanced carriers were observed over three generations. Classical cytogenetics had failed to detect this anomaly in the balanced carriers, who were first considered to be somatic mosaics for del( II )(p 13). Two of these women gave birth to children carrying a deleted chromosome II. most likely resulting from the loss of the I I p 13 band inserted in I I q. Although in both cases the deletion encompassed exactly the same maternally inherited markers, there was a wide Variation in clinical expression. One child, with the karyotype 46,XY,del(ll)(pllpl4), presented the full-blown WAGR syndrome with anlridia, mental retardation, Wilms' tumor, and pseudohermaphroditism, but also had proteinuria and glomerular sclerosis reminiscent of Drash syndrome. In contrast, the other one, a girl with the karyotype 46,XX,del( I I )(p I 3), only had aniridia. Although a specific set of mutational sites has been observed in Drash patients, these findings suggest that the loss of one copy of the WTI gene can result in similar genital and kidney abnormalities.

N2  - Wilms' tumor is a childhood nephroblastoma that is postulated to arise through the inactivation of a tumor suppressor gene by a two-hit mechanism. A candidate II p 13 Wilms' tumor gene, WTI, has been cloned and shown to encode a zinc finger protein. Patients with the WAGR syndrome (Wilms' tumor, aniridia, genitourinary abnormalities, and mental retardation) have a high risk of developing Wilms' tumor and they carry constitutional deletions of one chromosome II allele encompassing the WTI gene. Analysis of the remaining WTI allele in a Wilms' tumor from a WAGR patient revealed the deletion of a single nucleotide in exon 7. This mutation likely played a key role in tumor formation, as it prevents translation of the DNA-binding zinc finger domain that is essential for the function of the WTI polypeptide as a transcriptional regulator.

H a bicydo[3.1.0]hexane system is part of a rigid skeleton, the cydopropane ring exerts spedfk: 'Y substituent eflects of two ldnds. In the case of the bicyclobexane boat form an upfield shift of the C-3 signal is observed and in tbe esse of the chair form a downfield shift of 15-20 ppm. Compared to the corresponding cydopentanes the double bond in strained cydopentenes causes downfield shifts of the C-4 absorption. 1bis eftect increases witb increasing strain, reaching 8 45.9 ppm maximum in benzvalene. Hence it is tbe only known bicydo[l.l.O]butane baving 8 reversed order of carbon shieldings. The downfield shifts are e:xplained by means of simple orbital interaction schemes.

T1  - A WAGR region gene between PAX-6 and FSHB expressed in fetal brain
N2  - Developmental delay or mental retardation is a frequent component of multi-system anomaly syndromes associated with chromosomal deletions. Isolation of genes involved in the mental dysfunction in these disorders should define loci important in brain formation or function. We have identified a highly conserved locus in the distal part of 11 p 13 that is prominently expressed in fetal brain. Minimal expression is observed in a number of other fetal tissues. The gene maps distal to PAX-6 but proximal to the loci for brain-derived neurotrophic factor (BDNF) and the beta subunit of follicle stimulating hormone (FSHB), within a region previously implicated in the mental retardation component of some WAGR syndrome patients. Within fetal brain, the corresponding transcript is prominent in frontal, motor and primary visual cortex as weil as in the caudate-putamen. The characteristics of this gene, including the striking evolutionary conservation at the locus, suggest that the encoded protein may function in brain development.

N2  - In cats anesthetized with alpha-chloralose, extracellular recordings were made from fine afferent units belonging to the medial articular nerve (MAN) of the knee joint. The excitatory and sensitizing effects on articular afferents of serotonin (5-HT) applied intra-arterially close to the joint were examined. The joints were either normal or an experimental arthritis had been induced some hours before the recording session. Bolus injections of 1.35-135 p,g 5-HT excited about 43% of group 111 (CV: 2.5-20 m/sec) and 73% of group IV units (CV: < 2.5 mjsec) from normal joints. The latency was usually between 10 and 30 sec, and the duration and size of the responses were dose-dependent. Fast group 111 units (CV: > 16 mjsec) and group li units (CV: > 20 m/sec) were never excited by 5-HT. Repetitiveadministration led to pronounced tachyphylaxis of the 5-HT response. Inflammation induced an enhanced sensitivity of group III articular afferent units to close intra-arterial application of 5-HT. In particular the total duration of each response was considerably prolonged (4-10 min against 1-2 min under normal conditions). At the same time the tachyphylaxis seen under normal conditions was gteatly reduced. In contrast, group IV articular afferent units did not become sensitized to 5-HT in the course of inflammation. In normal joints 5-HT did not sensitize fineafferent units for movement-induced responses. However, after inflammation, a distinct sensitization to such movements by 5-HT application could be observed bothin group 111 and group IV fiber ranges. The sensitization had a short time course not exceeding 7 min. The tonic component of the movement-induced response was more enhanced than the phasic one. The bolus application of 5-HT led to temporary vasoconstriction of the knee joint vessels. This vasoconstriction was especially pronounced in inflamed joints and impeded the access of subsequently applied substances to the terminal regions of the afferent units under observation. lt is concluded that the present results support the notion that 5-HT may participate in the mediation of pain from inflamed tissue such as an arthritic joint by exciting and sensitizing fine afferent units. During inflammation group 111 units are particularly sensitive to 5-HT and, thus, may carry the bulk of the 5-HT-induced nociceptive messages.

N2  - The total nerve cell numbers in the right and in the left human entorhinal areas have been calculated by volume estimations with the Cavalieri principle and by cell density determinations with the optical disector. Thick gallocyanin-stained serial frozen sections through the parahippocampal gyrus of 22 human subjects (10 female, 12 male) ranging from 18 to 86 years were analysed. The laminar composition of gallocyanin (Nissl)-stained sections could easily be compared with Braak's (1972, 1980) pigmentoarchitectonic study, and Braak's nomenclature of the entorhinal laminas was adopted. Cellsparse laminae dissecantes can more clearly be distinguished in Nissl than in aldehydefuchsin preparations. These cell-poor dissecantes, lamina dissecans extema (dis-ext), lamina dissecans 1 (dis-1) and lamina dissecans 2 (dis-2), were excluded from nerve cell nurober determinations. An exact delineation of the entorhinal area is indispensable for any kind of quantitative investigation. We have defined the entorhinal area by the presence of pre-alpha ceil clusters and the deeper layers of lamina principalis externa (pre-beta and gamma) separated from lamina principalis interna (pri) by lamina dissecans 1 (dis-1). The human entorhinal area is quantitatively characterized by a left-sided (asymmetric) higher pre-alpha cell number and an age-related nerve cell loss in pre as well as pri layers. At variance with other CNS cortical and subcortical structures, the neuronal number of the entorhinal area appears to decrease continuously from the earliest stages analysed, although a secular trend has to be considered. The asymmetry in pre-alpha cell number is discussed in the context of higher human mental capabilities, especially language.

The increase in harmonic mean thickness of the basal laminae is statistically significant (α<=0.05) and compensates for a decrease in thickness of capillary endothelium. Consequently, the total barrier mass and thickness of cerebellar cortical capillaries in senile animals is higher than in young individuals.

N2  - The distribution of lipofuscin in the perikarya of Purkin je cells of vermal and hemispheric lobules has been determined quantitatively in 7 rats, 30-38 months old, by the point-counting method. On the basis of morphologically and statistically significant differences a pigmentarchitectonics of the cerebellar cortex is established. The Purkinje cells of lobule VIa (Larsell 1952) are extremely lipofuscin-rich. The Purkinje cells of the hemispheres, lobules V, Vlb + c and VII contain considerable amounts of a finely granular lipofuscin, the Purkinje cells of lobules I-III and VIII- IXa a globular type of lipofuscin. The Purkinje cells of sublobule XI d c and X are lipofuscin-poor cells. Three types of lipofuscin ha ve been identified in the light microscope.

N2  - The ultrastructure of autofluorescent, PAS-positive lipofuscin in Purkinje, granule, Golgi epithelial, basket and stellate, microglial and perivascular cells in the cerebellar cortex of senescent rats is described. The membrane- bounded pigment is composed ofthree elements: 1) electron-lucent homogeneaus droplets, 2) a granular matrix and 3) intensely osmiophilic patches. The proportians ofthese three components vary between cell types and one can grossly differentiate a neuronal and a gliallipofuscin. The lipofuscin granules of stellate and perivscular cells are different from lipofuscin of other cerebellar neurons and glia. lt can be concluded from these morphological observations that each cerebellar cell type has its distinct lipofuscin.

N2  - The quantitative postnatal changes of the cerebella of 65 Wistar rats aged 2-120 days have been examined. The cerebellar volume increases in two phases: The first phase lasts from birth to the seventh postnatal week. The second phase begins ten weeks post parturn and lasts for a Ionger period than the first phase. The cerebellar surface increases continuously from birth to the end of the seventh week. The volume of the external granular layer is maximal when the organ grows rapidly. The external granular layer has nearly disappeared 24 days after birth; the volume of the interaal granular layer is maximal at this time. Later on, the volume and the width of the interaal granular layer decrease. Myelinization of the cerebellar fibers and growth of the molecular layer run parallel to this decrease. The second late, but protracted growth of the cerebellum, ten weeks after birth, is due to an increase of the molecular and medullary layer. These findings are in good accord with histological, histochemical, and ultrastructural observations of other authors.

N2  - CATIO~IC drugs of different types and structures (antihistaminics, antiarrhythmics, sedatives, opiates, cytostatics and antibiotics, for example) are excreted in mammals by epithelial cells of the renal proximal tubules and by hepatocytes in the liver<sup>1-4</sup>. In the proximal tubules, two functionally disparate transport systems are involved which are localized in the basolateral and luminal plasma membrane and are different from the previously identified neuronal monoamine transporters and A TP-dependent multidrug exporting proteins<sup>1-3,5-12</sup>. Here we report the isolation of a complementary DNA from rat kidney that encodes a 556-amino-acid membrane protein, OCT1, which has the functional characteristics of organic cation uptake over the basolateral membrane of renal proximal tubules and of organic cation uptake into hepatocytes. OCTl is not homologous to any other known protein and is found in kidney, liver and intestine. As OCTl translocates hydrophobic and hydrophilic organic cations of different structures, it is considered to be a new prolotype of polyspecific transporters that are important for drug elimination.

T1  - Molecular cloning of a new bombesin receptor subtype expressed in uterus during pregnancy
N2  - The homology screening approach has been used to clone a new member of the guanine-nucleotidebinding-protein-coupled receptor superfamily from guinea pig uterus. The cloned cDNA encodes a 399-amino-acid protein and shows the highest amino acid similarity to members of the bombesin receptor family; 52% and 47% similarity to the gastrin-releasing-peptide (GRP) receptor and the neuromedin-B receptor, respectively. Bindingexperiments with the stably transfected LLC-PK<sub>1</sub> cell line expressing the new receptor protein confmned the bombesin-like nature of the cloned receptor. The relative order ofligand affinity, GRP = neuromedin C >> neuromedin B, suggests that the cloned cDNA represents the GRP subtype rather than the neuromedin-B subtype of bombesin receptors. Northern-blot analysis of mRNA species from several guinea-pig tissues showed that the mRNA for the new bombesin receptor subtype is expressed mainly in uteri of pregnant animals.

N2  - A cDNA encoding guinea-pig uterine substance P (SP) receptor has been isolated using the homology screening approach. Northern blot analysis reveals that the corresponding mRNA, of approx. 4.8 kb, is expressed in all tissues tested, but predominantly in the uteri of non-pregnant animals; during pregnancy its expression is reduced. The guinea-pig SP receptor was expressed in COS-7 cells and demonstrated relative Iigand affinity in the order: SP >> neurokinin A > neurokinin B.

N2  - This article gives an overview of possibilities for the assessment offacial behavior. With regard to validity, results from a longitudinal study of 36 depressed patients and nine controls as weil as often schizophrenic patients and their relatives will be referred to. These results are used to illustrate the following principles which have to be taken into account when studying facial behavior: a) communication strongly facilitates facial expression, b) activation of facial behavior follows the "principle of least effort", and c) the principle of individual specificity applies to the association of nonverbal behavior and mood states. Making allowance for these principles has, among others, consequences a) for situations or conditions under which to asses behavior (specifically conditions of communication), b) for data analysis (e.g., dealing with frequent and rare events), and c) for empirical or experimental strategies (e.g., aggregation of single-case longitudinal comparisons). From the results on facial behavior during depression it can be concluded that the nonverbal reaction tendencies of endogenous and neurotic depressed patients differ. Moreover, the differential behavioral pattems observed cast doubt on the assumption of a homogeneity of affects in depression. Taking into account the conditions which govern it, facial behavior has proved to be a valid and, especially, a differential indicator for pathoIogic affective states and their changes. Given the fact that a psychiatric illness generally incorporates emotional problems it is more than surprising that little attention has been paid to the systematic study of emotional behavior. Some of the reasons for this will be clarified in the following.
N2  - Dieser Beitrag gibt eine Übersicht über die Möglichkeiten zur Erfassung mimischen Verhaltens. Zur Bewertung der Validität werden Ergebnisse aus einer Verlaufsuntersuchung an 36 depressiven Patienten und neun Kontrollpersonen sowie von zehn schizophrenen Patienten und ihren Angehörigen herangezogen. Diese Ergebnisse sollen die folgenden Prinzipien, die bei der Untersuchung mimischen Verhaltens zu berücksichtigen sind, illustrieren: a) Kommunikation erleichtert in hohem Maße das Auftreten mimischen Ausdrucks, b) die Aktivierung mimischen Verhaltens folgt dem "Prinzip des geringsten Aufwandes", und c) auf die Beziehung zwischen nonverbalem Verhalten und Stimmungszuständen trifft das Prinzip der Individuenzpezifität zu. Aus diesen Prinzipien ergeben sich Konsequenzen a)für die Situationen oder Bedingungen, unter denen Verhalten zu erfassen ist (speziell Bedingungen der Kommunikation), b) für die Datenanalyse (z. B. die statistische Behandlung häufiger und seltener Ereignisse) und c) für empirische oder experimentelle Strategien (z. B. die Aggregation von Einzelfall-Verlaufsbeobachtungen). Aus den Ergebnissen zum mimischen Verhalten in der Depression kann geschlossen werden, daß sich endogen und neurotisch depressive Patienten hinsichtlich ihrer nonverbalen Reaktionstendenzen unterscheiden. Darüber hinaus stellen die differentiellen Verhaltensmuster die Annahme einer Affekthomogenität in der Depression in Frage. Berücksichtigt man die Bedingungen, die das mimische Verhalten steuern, so erweist es sich als valider und insbesondere differentieller Indikator für pathologische affektive Zustände und deren Veränderungen. Angesichts der Tatsache, daß eine psychiatrische Erkrankung im allgemeinen emotionale Probleme beinhaltet, so ist es mehr als erstaunlich, daß bisher dem Studium des unmittelbaren emotionalen Ausdrucksverhaltens so wenig Aufmerksamkeit geschenkt wurde. Einigen Gründen hierfür wollen wir im folgenden nachgehen.

N2  - HIV infection of CD4+ peripheral blood lymphocytes leads to a loss of MHC dass I molecules on the surface of the infected cells as detectable by monodonal antibody staining and flow cytometry. Incubation of the infected cells at 26 oe or treatment at 37 oe with peptides leads to upregulation of MHC dass I to levels equal to those found on uninfected cells cultured und er the same conditions. The data suggest that, after HIV infection, the mechanisms responsible for peptide generation, peptide transport and thus stable association between peptides and MHC dass I molecules are severely affected.

N2  - The He (I) photoelectron spectra of 2-bicyclo[2.1.l]hexene (1), 2,3-bis(methylene)bicyclo[2.1.l]hexane (3), and 3,4-bis(methylene)tricyclo[3.l.O.0\(^{2.6}\)]hexane (4) have been investigated. The assignment given is based on a ZDO model and semiempirical calculations. Tagether with the PE data of benzvalene (2), the reported data allow a comparison between 1-2 and 3-4. This yields a measure of the interactions between 8 cyclobutane or 8 bicyclobutane moiety and a double bond system within a ZDO model. The resonance integral found in the case of 1 and 3 amounts to -1.9 eV, that for 2 and 4, to -2.3 eV. The investigations furthermore reveal that the electronic factors which contribute to the higher reactivity of the bicyclobutane compounds amount to 5 kcal/mol.

N2  - In den Reaktionen von Tetracyanethylen (TCNE) und 5,6-Dichlor-2,3-dicyan-p-benzochinon mit Benzvalen haben wir kürzlich die ersten Beispiele für die lange gesuchte einstufige 1,4-Cycloaddition eines Alkens an ein Vinylcyclopropan gcfunden(I~J. Sie ist als [(.,2.+.2s)+ 112J-Prozeß der Dicls-Alder-Addition nahe verwandtllbl. Allerdings entsteht das betreffende TCNE-Addukt, ein Dihydrosemibullvalen-Derivat, nur in einer Ausbeute von wenigen Prozent. Die Hauptprodukte gehen aus einer Zwitterionischen Zwischenstufe hervor, die durch Anlagerung von TCNE an die Benzvalen-n-Bindung resultiert.

N2  - Die Arbeit beschreibt im ersten Teil die Auswirkung der Sandstein- und Eisenkrustenverkarstung auf das Plateaurelief des östlichen Niger. Angesprochen werden die abflußlosen Karstdepressionen auf den Plateaudachflächen, die Rolle intensiver Verkarstung bei der Herauspräparierung von Sandsteinsäulen an Hängen, von Plateauspornen und Inselbergen, die Mitwirkung bei der Talbildung sowie Einsturzdolinen an Hängen. Im zweiten Teil wird die Mikromorphologie von Karsthöhlenüberzügen aus der Endphase der phreatischen Karstlösung beschrieben. Oberflächennahe Körner weisen starke Korrosion auf. Phosphor in der wiederausgefällten Matrix und eingekieselte Bakterien deuten auf eine organische Komponente bei der Ausbildung des tertiären Silikatkarsts.
N2  - In its first part the paper describes the morphological effects of silicate karstification on the plateau landscape of eastern Niger. The description centers on the endorheic depressions dotting the plateau surfaces, the role of intensive karstification in the formation of slope pillars, plateau spurs and inselbergs, its contribution to valley formation, as well as on collapse structures on slopes. The second part describes the micromorphology of the lining of cave walls. Near-surface grains are heavily corroded. Reprecipitated matrix containing phosphorous and silicified bacteria on quartz grains point towards an organic element in the Tertiary formation of the silicate karst.

T1  - Immunoregulation by mouse T cell clones III. Cloned H-Y-specific cytotoxic T cells secrete a soluble mediator(s) that inhibits cytotoxic responses by acting on both Lyt-2\(^-\) and L3T4\(^-\)- lymphocytes
N2  - In this study we report that cloned Thy-l +, L3T4-, Lyt-l-, Lyt-2+, H-Y-specific and H-2Db-restricted cytotoxic T ce11 lines (CTLL) when indueed by lectin or antigen secrete a soluble mediator(s) (SF) that inhibits proliferation and generation of cytotoxic lymphocytes (CTL) in mixed lymphocyte cultures (MLC). The biological activity was separable by gel filtration and appeared as a broad peak in the moleeular mass range between 10000 and 50000 kDa. It was found that the suppressive activity released by CTLL neither strictly correlates with their cytotoxic potential nor with their ability to produce immune interferon or Iymphotoxin. SF was shown to elicitits activity in an antigen-nonspeeific manner in that it suppressed the maturation of T lymphocytes responding to both, the appropriate H-Y antigen as weH as to unrelated H_2d alloantigens or to the hapten 2,4,6-trinitrophenyl (TNP). The effect of SF on CTL responses was most pronounced in early phases of primary or secondary MLC. When analyzed for its inhibitory activity on precursor ceHs in populations selected for either Lyt-2- or L3T4- lymphocytes, it was found that SF interfered with the maturation of both subsets. The inhibition of CTL responses elicited by SF could not be reversed by the addition of exogenous interleukin 2. The findtng that SF also inhi. bited the proliferation of some but not a11 antigen-dependent cloned T ceHs with helper or eytc'toxic potential provides evidence that the faetor also may regulate effector lymphl)cytes. In addition, the results support the assumption that SF exerts its effect direetly on the responder rather than the stimulator population, and demonstrate that the development of CTL from their preeursor eeHs is contro11ed at least in part by the eytotoxic effeetor cells themselves via a soluble factor(s) that interferes with distinct stages of T ce11 maturation. These findings again emphasize the expression of multiple functions by CTL and indieate their possible role du ring the course of an immune response by their capability to eliminate target cells and to secrete a soluble product(s) that mediates feedback contro!.

N2  - The cycloadducts 6 and 7 of tricyc1o[4.1.0.0 2 ,7)hepta- 3,4-diene (~) with styrene and 1,3-butadiene rearrange to unusual products on thermolysis, namely the cycloheptatriene derivatives ~ and 10. 1-0xa-3,4-cyclohexadiene (20) is generated smoothly from 6,6-dichloro-3-oxabicyclo[3.1.0]hexane (22) and n-butyllithium. 1-0xa-2,3-cyclohexadiene (11) is formed from 6-exo-bromo-6-endo-fluoro-2-oxabicyclo[ 3.1.0]hexane (30) and methyllithium. In the presence of activated olefins, this reaction provides an efficient route to 28 and 33 - 38, the trapping products of 21. Interestingly, [2+2]-cycloadditions do not take place at the same double bond of 21 as [4+2]-cycloadditions. The reactions of 1,3-cyclopentadiene and indene with bromofluorocarbene afford 6-exo-bromo-6-endo-fluorobicyclo[3.1.0]hex-2-ene (50) and its benzo derivative ~, respectively. On treatment of these compounds with methyl lithium in the presence of styrene, the interception products 53 and 47 of 1,2,4-cyc10- hexatriene (44) and its benzo derivative 43, respectively, are formed in good yields.

N2  - We have previously shown that during an infection with Leishmania major, susceptible BALB/c mice, as opposed to mice of a resistant strain (C57BLl6), are primed by lipopolysaccharide for the production of high levels of tumor necrosis factor-\(\alpha\) (TNF-\(\alpha\)) which is known to be a potent maerophage M\(\Phi\) stimulator in other parasitic diseases. In the present study we investigated whether TNF-\(\alpha\) activates M\(\Phi\) for killing of L. major parasites. In the absence of interferon-y (IFN-\(\gamma\)) or lipopolysaccharide, TNF-\(\alpha\) (0.025-25000 U/ml) failed to activate peritoneal exudate M\(\Phi\) from BALB/c mice for killling of L. major amastigotes. In the presence of suboptimal doses of IFN-\(\gamma\) (5 or 10 Vlml), however, TNF-\(\alpha\) mediated a rapid elimination of intracellular parasites, which was highly significant compared to IFN-\(\gamma\) alone. Tbe combination of TNF with interleukin 4, in contrast, was inactive in this respect and allowed survival of intracellular parasites. From these data we conelude that the presence of IFN-\(\gamma\) is crucial for TNF-\(\alpha\)-mediated killing of L. major parasites by M\(\Phi\). Disease progression in susceptible mice therefore seems to be a consequence of a deficiency of IFN-\(\gamma\) and a predominance of interleukin 4 rather than the result of an excess amount of TNF-\(\alpha\).

N2  - Tbe human foamy virus (HFV) is a recently characterized member ofthe spumavirus family. Although no diseases have been unequivocally associated with HFV infection, expression of HFV regulatory genes in transgenie mice induces a characteristic aeute neuro degenerative disease and a myopathy. To better eharaeterize the sequenee of events leading to disease, and to gain a better understanding of the underlying pathogenetic meehanisms, we have analyzed in detail the transgene expression pattern during development. Transcription of a construet containing all regulatory elements and aneillary genes of mv was analyzed by in situ hybridization and was shown to occur in two distinct phases. At midgestation, low but widespread expression was first deteeted in eells of extraembryonie tissues. Later, various tissues originating from embryonie mesoderm, neuroeetoderm, and neural erest transeribed the transgene at moderate levels. However, expression deereased dramatically during late gestation and was suppressed shortly after birth. After a latency period of up to 5 weeks, transeription of the transgene resumed in single eelJs distributed irregularly in the central nervous system and in the skeletal museIe. By the age of 8 weeks, an increasing number of eells displayed much higher expression levels than in embryonie Iife and eventually underwent severe degenerative ehanges. These findings demonstrate that HFV transgene expression is differentially regulated in development and that HFV cytotoxicity may be dose-dependent. Such biphasic pattern of expression differs from that of murine retroviruses and may be explained by the specificity of HFV regulatory elements in combination with cellular faetors. Future studies of this model system should, therefore, provide novel insights in the mechanisms controlling retrovirallatency.

N2  - Bei 46 Typ-I-Diabetikern im Alter zwischen 15 und 44 Jahren wurden die Auswirkungen der viszeralen Deafferentierung bei autonomer Diabetesneuropathie auf körperliche Befindlichkeit und Emotionalität untersucht. Die Erfassung des Ausprägungsgrad der autonomen Neuropathie erfolgte mit vegetativen Funktionstest. Diabetiker ohne Neuropathie (N = 14), mit leichter Neuropathie (N = 23) und mit schwerer Neuropathie (N = 9) wurden miteinander verglichen. Gering ausgeprägte autonome Neuropathie führt zu vegetativen Funktionsstörungen, ist aber gleichzeitig mit einer verminderten Wahrnehmung körperlicher Beschwerden - erfaßt mit der Freiburger Beschwerdenliste - verbunden. Im Streß- und Copingverhalten (SCOPE) und in bestimmten Persönlichkeitsdimensionen (FPI) finden sich dabei Hinweise auf eine verminderte emotionale Reaktivität. Patienten mit fortgeschrittener autonomer Neuropathie haben zumeist ausgeprägte diabetische Folgekrankheiten (diabetische Retinopathie, Makro- und Mikroangiopathie etc.), scheinen aber die damit verbundenen körperlichen Beeinträchtigungen und emotionalen Belastungen nicht stärker als Diabetiker ohne Neuropathie wahrzunehmen. Die autonome Deafferentierung bei Diabetikern scheint somit sowohl zu einer gestörten Wahrnehmung körperlicher Beschwerden wie auch zu einer beeinträchtigten emotionalen Reaktionsfähigkeit zu führen.
N2  - Bei 46 Typ-I-Diabetikern im Alter zwischen 15 und 44 Jahren wurden die Auswirkungen der viszeralen Deafferentierung bei autonomer Diabetesneuropathie auf körperliche Befindlichkeit und Emotionalität untersucht. Die Erfassung des Ausprägungsgrad der autonomen Neuropathie erfolgte mit vegetativen Funktionstest. Diabetiker ohne Neuropathie (N = 14), mit leichter Neuropathie (N = 23) und mit schwerer Neuropathie (N = 9) wurden miteinander verglichen. Gering ausgeprägte autonome Neuropathie führt zu vegetativen Funktionsstörungen, ist aber gleichzeitig mit einer verminderten Wahrnehmung körperlicher Beschwerden - erfaßt mit der Freiburger Beschwerdenliste - verbunden. Im Streß- und Copingverhalten (SCOPE) und in bestimmten Persönlichkeitsdimensionen (FPI) finden sich dabei Hinweise auf eine verminderte emotionale Reaktivität. Patienten mit fortgeschrittener autonomer The perception of visceral activity is impaired in diabetics with autonomie neuropathy. This should affect the perception of somatic complaints and the emotional state of these patients. The degree of auronomic neuropathy was examined with three physiological tests for autonomie functions. Differences were evaluated between diabetics without neuropathy (N = 14), with mild neuropathy (N = 23) and with severe neuropathy (N = 9). It was found that mild autonomie neuropathy is related to a small increase in somatic problems (resulting from diabetes). In spite of this, autonomie deafferentation leads to a de~rease in somatic complaints (Freiburger Beschwerden Liste (FBL» and to a reduction in emotionality (assessed with personality questionaires (SCOPE and FPI». Patients with severe autonomie neuropathy exhibit frequent somatic problems. The somatic complaints and the emotionality of these patients are comparable with diabetics without neuropathy. The results show that autonomie deafferentation in diabetics is related to a reduced perception of somatic complaints and a decrease in the emotionality of these patients.

A cocarcinogenic effect of ethanol in the liver is therefore unlikely to be due to an effect on the metabolic activation and inactivation processes governing the formation of DNA-binding AFBI metabolites.

N2  - The total cortical and striatal neurone and glial numbers were estimated in five cases of Huntington's disease (three males, two females) and five ageand sex-matched control cases. Serial 500-l-lm-thick gallocyanin-stained frontal sections through the left hemisphere were analysed using Cavalieri's principle for volume and the optical disector for cell density estimations. The average cortical neurone number of five controls (mean age 53±13 years, range 36-72 years) was 5.97x 109±320x 106 , the average number of small striatal neurones was 82 X 106± 15.8 X 106• The left striatum (caudatum, putamen, and accumbens) contained a mean of 273 X 106±53 X 106 glial cells (oligodendrocytes, astrocytes and unc1assifiable glial profiles). The mean cortical neurone number in Huntington's disease patients (mean age 49±14 years, range 36-75 years) was diminished by about 33 % to 3.99x109±218x106 nerve cells (P ::;:::: 0.012, MannWhitney V-test). The mean number of small striatal neurones decreased tremendously to 9.72 X 106 ± 3.64 X 106 (-88 % ). The decrease in total glial cells was less pronounced (193 X 106±26 X 106) but the mean glial index, the numerical ratio of glial cells per neurone, increased from 3.35 to 22.59 in Huntington's disease. Qualitatively, neuronal loss was most pronounced in supragranular layers of primary sensory areas (Brodmann's areae 3,1,2; area 17, area 41). Layer HIc pyramidal cells were preferentially lost in association areas of the temporal, frontal, and parietal lobes, whereas spared layer IV granule cells formed a conspicuous band between layer IH and V in these fields. Methodological issues are discussed in context with previous investigations and similarities and differences of laminar and lobar nerve cellloss in Huntington's disease are compared with nerve cell degent-ration in other neuropsychiatric diseases.

N2  - To investigate the influence of inflammatory cytokines on the potential of peripheral nerves to regenerate, we analyzed the effect of interferon-y (lFN-y) and tumor necrosis factor-a (TNF-a) on the ability of immortalized Schwann cells to mediate outgrowth of neurites from primary DRG neurons. We found that IFN-y and TNF-a synergistically inhibited the neurite outgrowth-promoting properties of the Schwann cells by spedfically dowllregulating myelin-associated glycoprotein (MAG) at the levels of mRNA and cell surface protein by approximately 60%. Antibodies to MAG inhibited the outgrowth of neurites on Schwann cells to the same extent as treatment with the two cytokines. Since MAG appears to be involved in both neurite outgrowth and myelination, our findings may provide evidence for a mechanism, by wh ich inflammatory cytokines interfere with Schwann cell-neuron interactions.

The inhibitory effect of EDTA can be reversed with micro molar amounts of ZnCI2• ZnCh in micromolar concentrations also inhibits Ap.A hydrolase and stimulates amino acid-dependent Ap.A synthesis, suggesting that Zn2+ is modulating intracellular Ap.A pools. Serum addition to GI-arrested cells enhances uptake of Zn, whereas serum depletion leads to a fivefold decrease of the rates of zinc uptake. These results are discussed by regarding Zn2+ as a putative 'second messenger' of mitogenic induction and Ap.A as a possible 'third messenger' and trigger of DNA synthesis.

N2  - Vorgestellt werden Lösungsformen in karbonatfreien Sandsteinen und Eisenkrusten der östlichen Republik Niger. Diese Formen haben eine außerordentlich weite Verbreitung und sind wesentliche Elemente der Landschaft. Beschrieben werden sowohl auf den Dachflächen der südsaharisch/sahelischen Schichtstufen entwickelte Depressionen mit subterraner Entwässerung als auch an Wadi hängen und steilen Stufenabschnitten gekappte, horizontal bis schräg verlaufende Höhlen und Röhren. Viele dieser Lösungsformen sind zumindest in Resten noch von einer Eisen- oder Kieselkruste ausgekleidet, die u.a. als Indiz für eine Entstehung der Formen im Grundwasserbereich angesehen werden kann. Die Anlage der Karstformen wäre demnach noch vor die Herauspräparierung der Stufen und Inselberge im ausgehenden Tertiär zu stellen. Die Karstformen in nichtkarbonatischen Gesteinen gleichen bis ins Detail denen des Karbonatkarstes. Eine initiale Formung durch äolische oder fluviatile Prozesse oder durch Piping kann ausgeschlossen werden.
N2  - The authors describe subaerial and subterranean solution features in noncarbonatic sandstones and iron crusts from eastern Niger. They are of common occurence and are an important element of the landscape. The paper discusses karst depressions of various sizes on all the plateau surfaces of the saharo-sahelian scarpland and mesa landscape, together with their subterranean network of tubes and caves. In some tubes relicts of an iron or silica coating are preserved, one evidence among other, morphological ones that the passageways developed when completely filled with groundwater. The system originated during the Miocene and became dismembered during Pliocene scarpland and inselberg formation. The noncarbonatic karst forms closely resemble those of carbonate karst. Aeolian and fluvial processes, including piping, can be excluded as having created these landforms.

N2  - Seven monoclonal antibodies were raised against the immunoglobulin-like extracellular domain of PO (POED), the major protein of peripheral nervous system myelin. Mice were immunized with purified recombinant rat PO-ED. After fusion, 7 clones (POI-P07) recognizing either recombinant, rat, mouse, or human PO-ED were selected by ELlS A and were characterized by Western blot, immunohistochemistry, and a competition assay. Antibodies belonged to the IgG or IgM class, and P04-P07, reacted with PO in fresh-frozen and paraffin-embedded sections of human or rat peripheral nerve, but not with myelin proteins of the central nervous system of either species. Epitope specificity of the antibodies was determined by a competition enzyme-linked immunosorbent assay (ELISA) and a direct ELlS A using short synthetic peptides spanning the entire extracellular domain of PO. These assays showed that POl and P02 exhibiting the same reaction pattern in Western blot and immunohistochemistry reacted with different distant epitopes of PO. Furthermore, the monoclonal antibodies P05 and P06 recognized 2 different epitopes in close proximity within the neuritogenic extracellular sequence of PO. This panel of monoclonal antibodies, each binding to a different epitope of the extracellular domain of PO, will be useful for in vitro and in vivo studies designed to explore the role of PO during myelination and in demyelinating diseases of the peripheral nervous system.

Auslöser dieser umfassenden Neugestaltung war die Reform der gymnasialen Oberstufe (G8). Bereits vor dem G8 waren Schüler eine wichtige Zielgruppe der UB. Im Beitrag werden speziell die Angebote und Veränderungen vorgestellt, die sich durch die Oberstufenreform ergeben haben. Berichtet wird auch über Kooperationen zu anderen Bibliotheken, die in der Zukunft noch ausgebaut werden sollen.
N2  - The cooperation of the University Library (UB) of Würzburg with schools, students and teachers has changed thoroughly during the past year. The cause of this comprehensive reorganisation was the reform of the senior years of secondary school, the so-called G8 reform. Students used to be an important target group of the library already before the reform. The article introduces in particular the services and the changes brought about by the school reform. It furthermore reports about the cooperation with various other libraries, which is to be intensified in the future.

Drei Arten, C. humifusa, C. maculata und C. prostrata, konnten nachgewiesen werden. Bevorzugte Wuchsorte sind neben Bahnhöfen, botanischen Gärten und Gärtnereien vor allem Friedhöfe. Auf den untersuchten 126 Friedhöfen des Main-Spessart-Kreises wuchsen die Chamaesyce-Arten auf Kies- und Sandwegen, in Pflaster- und Plattenfugen, auf Gräbern und Beeten. C. humifusa wurde auf 27, C. maculata auf 46 und C. prostrata auf einem der 126 Friedhöfe des Untersuchungsgebietes gefunden.
N2  - The occurrence and distribution of the genus Chamaesyce in 126 cemeteries of the district of Main-Spessart (area of 1322 km2) were investigated from 2003 until 2007. Three species, C. humifusa, C. maculata and C. prostrata, were recorded. Habitats of naturalized Chamaesyce species are train stations, botanical gardens, nurseries and, mainly, cemeteries. In the 126 cemeteries investigated, Chamaesyce species grow preferably on gravel or sand walks and also in pavement gaps, on graves and flower beds. In the study area C. humifusa was found in 27 and C. maculata in 46 cemetries. C. prostrata was observed in one cemetery only.
