TY - THES A1 - Storz, Oliver T1 - Aufbau eines Rastertunnelmikroskops für Landau Level - Spektroskopie auf topologischen Isolator - Oberflächen T1 - Development of a scanning tunneling microscope for Landau level spectroscopy of topological insulator sufaces N2 - Im Rahmen dieser Arbeit wurde ein Rastertunnelmikroskop (STM) für Messungen bei tiefen Temperaturen und hohen Magnetfeldern konzipiert und aufgebaut. Die Probentemperatur kann dabei auf bis zu 1.4\,Kelvin reduziert werden, was spektroskopische Messungen mit extrem hoher Energieauflösung ermöglicht. Die thermische Verbreiterung spektroskopischer Merkmale liegt somit im Bereich eines Milli-Elektronenvolts, wie durch den Fit der Bandlücke eines supraleitenden Materials demonstriert wird. Ein linearer Bewegungsmechanismus ermöglicht die Positionierung des STM-Körpers innerhalb einer supraleitenden Spule, in der Magnetfelder von bis zu 12.5\,Tesla senkrecht zur Probenoberfläche erzeugt werden können. Das System erlaubt des Weiteren den Wechsel von Spitzen und Proben innerhalb des Kryostaten sowie das Aufdampfen von Einzelatomen auf die kalte Probenoberfläche ohne die Probe aus dem STM zu entfernen. Um den Einfluss mechanischer Vibrationen zu minimieren wurde ein innovatives Feder-Dämpfungssystem entwickelt, dass eine Stabilität des Tunnelkontakts von bis zu einem Pikometer gewährleistet. \\ \noindent Der zweite Teil dieser Arbeit präsentiert die Ergebnisse von STM-Messungen auf Antimon-Tellurid (Sb_{2}Te_{3}). Sb_{2}Te_{3}\, gehört zur relativ neu entdeckten Materialklasse der Topologischen Isolatoren (TI). Diese Verbindungen besitzen auf ihren Oberflächen Zustände mit linearer Dispersion, die durch die Zeitumkehr-Invarianz geschützt werden. Fokus unserer Messungen ist dabei der Einfluss eines magnetischen Feldes auf die Eigenschaften eines derartigen unkonventionellen 2D-Elektronengases. Dazu wurde die Entstehung von Landau Level (LL) innerhalb eines Magnetfelds genau untersucht. Die zwei in dieser Arbeit untersuchten Hauptaspekte sind: \medskip \noindent(i) Die energetische Verbreiterung, die Rückschlüsse auf die Lebensdauer zulässt\\ (ii) Die örtliche Fluktuation. \medskip \noindent Erstaunlicherweise kann die gemessene Verbreiterung der Landau Resonanzen nicht mit gängigen Mechanismen der Lebenszeit-Verbreiterung erklärt werden. Aus diesem Grund wird eine alternative Interpretation basierend auf der Heissenbergschen Unschärferelation vorgestellt, die im guten Einklang mit den von uns gewonnenen Daten steht. Des Weiteren zeigen örtlich aufgelöste Messungen systematische Abweichungen in der Dirac-Geschwindigkeit positiver und negativer Landau Resonanzen. Diese Fluktuationen stehen dabei in direktem Zusammenhang mit Änderungen im lokalen chemischen Potential. Da die physikalischen Ursachen dieser Abweichung im Rahmen dieser Arbeit nicht zweifelsfrei geklärt werden konnten, werden im letzten Teil die zugrundeliegenden Messergebnisse vorgestellt und mögliche Erklärungen des Verhaltens präsentiert. N2 - The scope of this thesis is the design and construction of a scanning tunneling microscope (STM) operating at low temperatures and high magnetic fields. The sample temperature can be reduced to 1.4\,Kelvin which permits to perform spectroscopic measurements with extremely high energy resolution. As demonstrated by fitting the gap of a superconducting material the thermal broadening of spectroscopic features is routinely found to be of the order of one milli-electronvolt. A linear travel mechanism allows to position the STM head inside a superconducting solenoid where magnetic fields up to 12.5\,Tesla can be applied perpendicular to the sample surface. Tips and samples can be exchanged in-situ and single atoms can be directly evaporated onto the cold sample surface without extracting the sample from the STM. To minimize the impact of mechanical vibrations, an innovative spring-damping system has been developed giving the tunneling junction a stability as low as one pico-meter. \\ \noindent The second part of this thesis presents the results of STM measurements on antimony telluride (\Sb_{2}Te_{3}). \Sb_{2}Te_{3}\, belongs to the relatively new class of materials known as topological insulators (TI). These compounds host on their surfaces linearly dispersing states which are protected by time-reversal symmetry. The focus of our measurements is the influence of a magnetic field on the properties of this unconventional 2D electron gas. The evolution of Landau levels (LL) in magnetic fields has been carefully analysed. Two are the main aspects which have been tackled: \medskip \noindent(i) Their energetic broadening, which can be directly linked to the lifetime \\ (ii) Their spacial fluctuation. \medskip \noindent Surprisingly, the energetic broadening of the landau peaks cannot be explained by any of the mechanisms commonly limiting the lifetime. An alternative interpretation based on Heissenberg's uncertainty principle is presented, which is found to be in good agreement with our data. Furthermore spatially resolved experiments reveal systematic deviations of the Dirac velocities for positive and negative LL. These fluctuations are intimately linked to variations of the local chemical potential. As the physical origin of this deviation could not be unambiguously identified, the last part presents the experimental data and suggests possible explanations of this finding. The primary aim of this study was to examine attentional mechanisms relevant for perceiving social cues by means of abnormalities in scanning of facial features in patients with social phobia. In two novel experimental paradigms, patients with social phobia and healthy controls matched on age, gender and education were compared regarding their gazing behavior towards facial cues. The first experiment was an emotion classification paradigm which allowed for differentiating reflexive attentional shifts from sustained attention towards diagnostically relevant facial features. In the second experiment, attentional orienting by gaze direction was assessed in a gaze-cueing paradigm in which non-predictive gaze cues shifted attention towards or away from subsequently presented targets. We found that patients as compared to controls reflexively oriented their attention more frequently towards the eyes of emotional faces in the emotion classification paradigm. This initial hypervigilance for the eye region was observed at very early attentional stages when faces were presented for 150 ms, and persisted when facial stimuli were shown for 3 s. Moreover, a delayed attentional orienting into the direction of eye gaze was observed in individuals with social phobia suggesting a differential time course of eye gaze processing in patients and controls. Our findings suggest that basic mechanisms of early attentional exploration of social cues are biased in social phobia and might contribute to the development and maintenance of the disorder. In the present study, female university freshmen were tested at the start of the first semester and again at the start of the second semester. Attentional bias toward high- or low-calorie food-cues was assessed using a dot-probe paradigm and participants completed the Barratt Impulsiveness Scale. Attentional bias and motor impulsivity interactively predicted change in body mass index: motor impulsivity positively predicted weight gain only when participants showed an attentional bias toward high-calorie food-cues. Attentional and non-planning impulsivity were unrelated to weight change. Results support findings showing that weight gain is prospectively predicted by a combination of weak top-down control (i.e. high impulsivity) and strong bottom-up impulses (i.e. high automatic motivational drive toward high-calorie food stimuli). They also highlight the fact that only specific aspects of impulsivity are relevant in eating and weight regulation. In the present study, we investigated attention mechanisms in fear and anxiety by adapting the NPU-threat test to measure steady-state visual evoked potentials (ssVEPs). We investigated ssVEPs across no aversive events (N), predictable aversive events (P), and unpredictable aversive events (U), signaled by four-object arrays (30 s). In addition, central cues were presented during all conditions but predictably signaled imminent threat only during the P condition. Importantly, cues and context events were flickered at different frequencies (15 Hz vs. 20 Hz) in order to disentangle respective electrocortical responses. The onset of the context elicited larger electrocortical responses for U compared to P context. Conversely, P cues elicited larger electrocortical responses compared to N cues. Interestingly, during the presence of the P cue, visuocortical processing of the concurrent context was also enhanced. The results support the notion of enhanced initial hypervigilance to unpredictable compared to predictable threat contexts, while predictable cues show electrocortical enhancement of the cues themselves but additionally a boost of context processing. Methods We used an analysis of variance model in patients on stable therapy (discovery cohort) to establish critical differences (d(crit)) for the minimum change associated with a significant individual patient response (beyond normal variation) in the PRO measures of pain (0-10), fatigue (0-10), and function (Funktionsfragebogen Hannover questionnaire; 0-100). We then evaluated PRO responses in patients initiating adalimumab in a noninterventional study (treatment cohort). Results In the discovery cohort (n=700), PROs showed excellent long-term retest reliability. The minimum change that exceeded random fluctuation was conservatively determined to be 3 points for pain, 4 points for fatigue, and 16 points for function. In the treatment cohort (n=2,788), 1,483 patients (53.2%) achieved a significant individual therapeutic response as assessed by Disease Activity Score in 28 joints (DAS28)-d(crit) (1.8 points) after 12 months of adalimumab treatment; 68.5% of patients with a DAS28-d(crit) response achieved a significant improvement in pain, whereas approximately 40% achieved significant improvements in fatigue or function. Significant improvements in all 3 PROs occurred in 22.7% of patients; 22.8% did not have any significant PRO responses. In contrast, significant improvements in all 3 PROs occurred in only 4.4% of 1,305 patients who did not achieve a DAS28-d(crit) response at month 12, and 59.1% did not achieve any significant PRO responses. Conclusion The establishment of critical differences in PROs distinguishes true responses from random variation and provides insights into appropriate patient management. This QWoE-methodology was applied to the reproductive toxicity data on diisononylphthalate (DINP), di-n-hexylphthalate (DnHP), and dicyclohexylphthalate (DCHP) to determine if the scientific evidence for adverse effects meets the requirements for classification as reproductive toxicants. The scores for DINP were compared to those when applying the methodology DCHP and DnHP that have harmonized classifications. Based on the quality/reliability scores, application of the QWoE shows that the three databases are of similar quality; but effect scores differ widely. Application of QWoE to DINP studies resulted in an overall score well below the benchmark required to trigger classification. For DCHP, the QWoE also results in low scores. The high scores from the application of the QWoE methodology to the toxicological data for DnHP represent clear evidence for adverse effects and justify a classification of DnHP as category 1B for both development and fertility. The conclusions on classification based on the QWoE are well supported using a narrative assessment of consistency and biological plausibility. Evidence from Transformed Movements JF - PLoS ONE N2 - It has been argued that several reported non-visual influences on perception cannot be truly perceptual. If they were, they should affect the perception of target objects and reference objects used to express perceptual judgments, and thus cancel each other out. This reasoning presumes that non-visual manipulations impact target objects and comparison objects equally. In the present study we show that equalizing a body-related manipulation between target objects and reference objects essentially abolishes the impact of that manipulation so as it should do when that manipulation actually altered perception. Moreover, the manipulation has an impact on judgements when applied to only the target object but not to the reference object, and that impact reverses when only applied to the reference object but not to the target object. A perceptual explanation predicts this reversal, whereas explanations in terms of post-perceptual response biases or demand effects do not. Altogether these results suggest that body-related influences on perception cannot as a whole be attributed to extra-perceptual factors. Fundamentals about conservation laws, Hamilton-Jacobi equations and discontinuous Galerkin methods are presented. In particular, issues in the development of discontinuous Galerkin (DG) methods for the Hamilton-Jacobi equations are discussed. The development of the ALE-DG methods based on the assumption that the distribution of the grid points is explicitly given for an upcoming time level. This assumption allows to construct a time-dependent local affine linear mapping to a reference cell and a time-dependent finite element test function space. In addition, a version of Reynolds’ transport theorem can be proven. For the fully-discrete ALE-DG method for nonlinear scalar conservation laws the geometric conservation law and a local maximum principle are proven. Furthermore, conditions for slope limiters are stated. These conditions ensure the total variation stability of the method. In addition, entropy stability is discussed. For the corresponding semi-discrete ALE-DG method, error estimates are proven. If a piecewise $\mathcal{P}^{k}$ polynomial approximation space is used on the reference cell, the sub-optimal $\left(k+\frac{1}{2}\right)$ convergence for monotone fuxes and the optimal $(k+1)$ convergence for an upwind flux are proven in the $\mathrm{L}^{2}$-norm. The capability of the method is shown by numerical examples for nonlinear conservation laws. Likewise, for the semi-discrete ALE-DG method for nonlinear Hamilton-Jacobi equations, error estimates are proven. In the one dimensional case the optimal $\left(k+1\right)$ convergence and in the two dimensional case the sub-optimal $\left(k+\frac{1}{2}\right)$ convergence are proven in the $\mathrm{L}^{2}$-norm, if a piecewise $\mathcal{P}^{k}$ polynomial approximation space is used on the reference cell. For the fullydiscrete method, the geometric conservation is proven and for the piecewise constant forward Euler step the convergence of the method to the unique physical relevant solution is discussed. N2 - Die vorliegende Arbeit beschäftigt sich mit der Entwicklung und Analyse von arbitrar Lagrangian-Eulerian discontinuous Galerkin (ALE-DG) Methoden mit zeitabhängigen Testfunktionen Räumen für Erhaltungs- und Hamilton-Jacobi Gleichungen. Grundlagen über Erhaltungsgleichungen, Hamilton-Jacobi Gleichungen und discontinuous Galerkin Methoden werden präsentiert. Insbesondere werden Probleme bei der Entwicklung von discontinuous Galerkin Methoden für die Hamilton-Jacobi Gleichungen untersucht. Die Entwicklung der ALE-DG Methode basiert auf der Annahme, dass die Verteilung der Gitterpunkte zu einem kommenden Zeitpunkt explizit gegeben ist. Diese Annahme ermöglicht die Konstruktion einer zeitabhängigen lokal affin-linearen Abbildung auf eine Referenzzelle und eines zeitabhängigen Testfunktionen Raums. Zusätzlich kann eine Version des Reynolds’schen Transportsatzes gezeigt werden. Für die vollständig diskretisierte ALE-DG Methode für nichtlineare Erhaltungsgleichungen werden der geometrischen Erhaltungssatz und ein lokales Maximumprinzip bewiesen. Des Weiteren werden Bedingungen für Limiter angegeben. Diese Bedingungen sichern die Stabilität der Methode im Sinne der totalen Variation. Zusätzlich wird die Entropie-Stabilität der Methode diskutiert. Für die zugehörige semi-diskretisierte ALE-DG Methode werden Fehlerabschätzungen gezeigt. Wenn auf der Referenzzelle ein Testfunktionen Raum, der stückweise Polynome vom Grad $k$ enthält verwendet wird, kann für einen monotonen Fluss die suboptimale Konvergenzordnung $\left(k+\frac{1}{2}\right)$ und für einen upwind Fluss die optimale Konvergenzordnung $\left(k+1\right)$ in der $\mathrm{L}^{2}$-Norm gezeigt werden. To understand genotype–phenotype relationships and to investigate trait architecture, the existence of both high-resolution genotypic and phenotypic data is necessary. Arabidopsis thaliana is a prime model for these purposes. This herb naturally occurs across much of the Eurasian continent and North America. Thus, it is exposed to a wide range of environmental factors and has been subject to natural selection under distinct conditions. Full genome sequencing data for more than 1000 different natural inbred lines are available, and this has encouraged the distributed generation of many types of phenotypic data. To leverage these data for meta analyses, AraPheno (https://arapheno.1001genomes.org) provide a central repository of population-scale phenotypes for A. thaliana inbred lines. AraPheno includes various features to easily access, download and visualize the phenotypic data. Traditional emotional theories assume a linear relationship between these dimensions, with valence determining the direction of a behavior (approach vs. withdrawal) and arousal its intensity or strength. In contrast, according to the valence-arousal conflict theory, both dimensions are interactively related: positive valence and low arousal (PL) are associated with an implicit tendency to approach a stimulus, whereas negative valence and high arousal (NH) are associated with withdrawal. Hence, positive, high-arousal (PH) and negative, low-arousal (NL) stimuli elicit conflicting action tendencies. By extending previous research that used several tasks and methods, the present study investigated whether and how emotional valence and arousal affect subjective approach vs. withdrawal tendencies toward emotional words during two novel tasks. In Study 1, participants had to decide whether they would approach or withdraw from concepts expressed by written words. In Studies 2 and 3 participants had to respond to each word by pressing one of two keys labeled with an arrow pointing upward or downward. Across experiments, positive and negative words, high or low in arousal, were presented. In Study 1 (explicit task), in line with the valence-arousal conflict theory, PH and NL words were responded to more slowly than PL and NH words. In addition, participants decided to approach positive words more often than negative words. In Studies 2 and 3, participants responded faster to positive than negative words, irrespective of their level of arousal. Furthermore, positive words were significantly more often associated with “up” responses than negative words, thus supporting the existence of implicit associations between stimulus valence and response coding (positive is up and negative is down). Its mechanisms of action are divided into antiproliferative and antimetabolic effects. Its influcence on cancer cell metabolism needs further investigation. One objective of this study, therefore, was to further elucidate the antimetabolic action of FWGE. The anticancer compound 2,6-dimethoxy-1,4-benzoquinone (DMBQ) is the major bioactive compound in FWGE and is probably responsible for its anticancer activity. The second objective of this study was to compare the antiproliferative properties in vitro of FWGE and the DMBQ compound. Methods The IC\(_{50}\) values of FWGE were determined for nine human cancer cell lines after 24 h of culture. The DMBQ compound was used at a concentration of 24 μmol/l, which is equal to the molar concentration of DMBQ in FWGE. Cell viability, cell cycle, cellular redox state, glucose consumption, lactic acid production, cellular ATP levels, and the NADH/NAD\(^+\) ratio were measured. Results The mean IC\(_{50}\) value of FWGE for the nine human cancer cell lines tested was 10 mg/ml. Both FWGE (10 mg/ml) and the DMBQ compound (24 μmol/l) induced massive cell damage within 24 h after starting treatment, with changes in the cellular redox state secondary to formation of intracellular reactive oxygen species. Unlike the DMBQ compound, which was only cytotoxic, FWGE exhibited cytostatic and growth delay effects in addition to cytotoxicity. Both cytostatic and growth delay effects were linked to impaired glucose utilization which influenced the cell cycle, cellular ATP levels, and the NADH/NAD\(^+\) ratio. The growth delay effect in response to FWGE treatment led to induction of autophagy. Conclusions FWGE and the DMBQ compound both induced oxidative stress-promoted cytotoxicity. In addition, FWGE exhibited cytostatic and growth delay effects associated with impaired glucose utilization which led to autophagy, a possible previously unknown mechanism behind the influence of FWGE on cancer cell metabolism. The aim of this study was to analyze antimicrobial resistance patterns of the common pathogens occurring at the Bugando Medical Centre (BMC), Mwanza, Tanzania to provide data for antimicrobial stewardship programmes. Methods A total of 3330 microbiological culture results scripts representing non-repetitive specimens reported between June 2013 and May 2015 were retrieved and analyzed for pathogens and their susceptibility patterns using STATA-11 software. Results Out of 3330 specimens, 439 (13.2%) had positive culture. Staphylococcus aureus (n = 100; 22.8%), Klebsiella pneumoniae (n = 65; 14.8%) and Escherichia coli (n = 41; 9.3%) were the most frequently isolated bacteria. Of 78 Staphylococcus aureus tested, 27 (34.6%) were found to be methicillin resistant Staphylococcus aureus (MRSA). Rates of resistance of Klebsiella pneumoniae and Escherichia coli isolates to third generation cephalosporins were 38.5% (25/65) and 29.3% (12/41) respectively. Staphylococcus aureus and Klesbiella pneumoniae were commonly isolated from bloodstream infections while Escherichia coli and Pseudomonas aeruginosa were the predominant isolates from urinary tract and wounds infections respectively. Of 23 Salmonella species isolated, 22 (95%) were recovered from the blood. Nine of the 23 Salmonella species isolates (39%) were found to be resistant to third generation cephalosporins. The resistance rate of gram-negative bacteria to third generation cephalosporins increased from 26.5% in 2014 to 57.9% in 2015 (p = 0.004) while the rate of MRSA decreased from 41.2% in 2013 to 9.5% in 2015 (p = 0.016). Multidrug-resistant gram-negative isolates were commonly isolated from Intensive Care Units and it was noted that, the majority of invasive infections were due to gram-negative bacteria. Conclusion There is an increase in proportion of gram-negative isolates resistant to third generation cephalosporins. The diversity of potential pathogens resistant to commonly prescribed antibiotics underscores the importance of sustained and standardized antimicrobial resistance surveillance and antibiotic stewardship programmes in developing countries. NADH-supported respiration rates are significantly decreased by activated PhotoCORM, mimicking the effect of dissolved CO gas. CO from the PhotoCORM binds to intracellular targets, namely respiratory oxidases in strain EC958 and a bacterial globin heterologously expressed in strain K-12. However, unlike previously characterized CORMs, the PhotoCORM is not significantly accumulated in cells, as deduced from the cellular manganese content. Activated PhotoCORM reacts avidly with hydrogen peroxide producing hydroxyl radicals; the observed peroxide-enhanced toxicity of the PhotoCORM is ameliorated by thiourea. The PhotoCORM also potentiates the effect of the antibiotic, doxycycline. Innovation: The present work investigates for the first time the antimicrobial activity of a light-activated PhotoCORM against an antibiotic-resistant pathogen. A comprehensive study of the effects of the PhotoCORM and its derivative molecules upon illumination is performed and mechanisms of toxicity of the activated PhotoCORM are investigated. Conclusion: The PhotoCORM allows a site-specific and time-controlled release of CO in bacterial cultures and has the potential to provide much needed information on the generality of CORM activities in biology. However, while data in humans is sparse, translation of murine tumor models to the clinic is further hampered by small sample sizes, heterogeneous settings and mixed results concerning tumor growth retardation. The aim was therefore to synthesize the evidence for a growth inhibiting effect of KDs when used as a monotherapy in mice. Methods We conducted a Bayesian random effects meta-analysis on all studies assessing the survival (defined as the time to reach a pre-defined endpoint such as tumor volume) of mice on an unrestricted KD compared to a high carbohydrate standard diet (SD). For 12 studies meeting the inclusion criteria either a mean survival time ratio (MR) or hazard ratio (HR) between the KD and SD groups could be obtained. The posterior estimates for the MR and HR averaged over four priors on the between-study heterogeneity τ\(^{2}\) were MR = 0.85 (95% highest posterior density interval (HPDI) = [0.73, 0.97]) and HR = 0.55 (95% HPDI = [0.26, 0.87]), indicating a significant overall benefit of the KD in terms of prolonged mean survival times and reduced hazard rate. All studies that used a brain tumor model also chose a late starting point for the KD (at least one day after tumor initiation) which accounted for 26% of the heterogeneity. In this subgroup the KD was less effective (MR = 0.89, 95% HPDI = [0.76, 1.04]). Conclusions There was an overall tumor growth delaying effect of unrestricted KDs in mice. Future experiments should aim at differentiating the effects of KD timing versus tumor location, since external evidence is currently consistent with an influence of both of these factors. Methods: Beam's-eye-view (BEV) based fluence warping is a standard adaptation method with disadvantages for strongly varying OAR shapes. 2-Step-adaptation overcomes these difficulties by a deeper analysis of the 3D properties of adaptation processes, but requires separate arcs for every OAR to spare, which makes it impractical for cases with multiple OARs. The authors aim to extend the 2-Step method to arbitrary intensity modulated plan by analyzing the anisotropy of dose contributions. Anisotropy was defined as a second term of Fourier transformation of gantry angle dependent dose contributions. For a cylindrical planning target volume (PTV) surrounding an OAR of varying diameter, the anisotropy and the dose-normalized anisotropy were analyzed for several scenarios of optimized fluence distributions. 2-Step adaptation to decreasing and increasing OAR diameter was performed, and compared to a usual fluence based adaptation method. For two clinical cases, prostate and neck, the VMAT was generated and the behavior of anisotropy was qualitatively explored for deformed organs at risk. # Results: Dose contribution anisotropy in the PTV peaks around nearby OARs. The thickness of the "anisotropy wall" around OAR increases for increasing OAR radius, as also does the width of 2-Step dose saturating fluence peak adjacent to the OAR K. Different optimized beam fluence profiles resulted in comparable radial dependence of normalized anisotropy. A graph is a binary relation between objects, that is, it consists of a set of objects (vertices) and a set of pairs of objects (edges). Networks are common examples of modeling data as a graph. For example, relationships between persons in a social network, or network links between computers in a telecommunication network can be represented by a graph. The clearest way to illustrate the modeled data is to visualize the graphs. The field of Graph Drawing deals with the problem of finding algorithms to automatically generate graph visualizations. The task is to find a "good" drawing, which can be measured by different criteria such as number of crossings between edges or the used area. In this thesis, we study Angular Schematization in Graph Drawing. By this, we mean drawings with large angles (for example, between the edges at common vertices or at crossing points). The thesis consists of three parts. First, we deal with the placement of boxes. Boxes are axis-parallel rectangles that can, for example, contain text. They can be placed on a map to label important sites, or can be used to describe semantic relationships between words in a word network. In the second part of the thesis, we consider graph drawings visually guide the viewer. These drawings generally induce large angles between edges that meet at a vertex. Furthermore, the edges are drawn crossing-free and in a way that makes them easy to follow for the human eye. The third and final part is devoted to crossings with large angles. In drawings with crossings, it is important to have large angles between edges at their crossing point, preferably right angles. Here, surgically harvested specimens from 19 abdominal aortic aneurysm (AAA) and 8 popliteal artery aneurysm (PAA) patients were analyzed for miRNA expression and histologically classified regarding extracellular matrix (ECM) remodeling and inflammation. DIANA-based computational target prediction and pathway enrichment analysis verified our results, as well as previous ones. miRNA-362, -19b-1, -194, -769, -21 and -550 were significantly down-regulated in AAA samples depending on degree of inflammation. Similar or inverse regulation was found for miR-769, 19b-1 and miR-550, -21, whereas miR-194 and -362 were unaltered in PAA. In situ hybridization verified higher expression of miR-550 and -21 in PAA compared to AAA and computational analysis for target genes and pathway enrichment affirmed signal transduction, cell-cell-interaction and cell degradation pathways, in line with previous results. Despite the vague role of miRNAs for potential diagnostic and treatment purposes, the number of candidates from tissue signature studies is increasing. Tissue morphology influences subsequent research, yet comparison of distinct entities of aneurysm disease can unravel core pathways. However, this opens up a new loss pathway from an interfacial charge transfer state to a triplet exciton (TE) state called electron back transfer (EBT), which is detrimental to device performance. To test this hypothesis, we study triplet formation in the high performing PTB7:PC\(_{71}\)BM blend system and determine the impact of the morphology-optimizing additive 1,8-diiodoctane (DIO). Using photoluminescence and spin-sensitive optically detected magnetic resonance (ODMR) measurements at low temperature, we find that TEs form on PC\(_{71}\)BM via intersystem crossing from singlet excitons and on PTB7 via EBT mechanism. For DIO blends with smaller fullerene domains, an increased density of PTB7 TEs is observed. The EBT process is found to be significant only at very low temperature. At 300 K, no triplets are detected via ODMR, and electrically detected magnetic resonance on optimized solar cells indicates that TEs are only present on the fullerenes. We conclude that in PTB7:PC\(_{71}\)BM devices, TE formation via EBT is impacted by fullerene domain size at low temperature, but at room temperature, EBT does not represent a dominant loss pathway. Three epochs of Land-use Land-cover (LULC) maps for 1986, 2001 and 2014 were derived from Landsat images using support vector machines (SVM). Accuracy assessment (AA) of the LULC maps based on the pixel count resulted in overall accuracy of 82%, 92% and 92%, while the AA derived from the error adjusted area (EAA) method stood at 69%, 91% and 91% for 1986, 2001 and 2014, respectively. Two major techniques for detecting changes in the LULC epochs involved the use of binary maps as well as a post-classification comparison approach. Quantitative spatiotemporal analysis was conducted to detect LULC changes with specific focus on the settlement development pattern of Abuja, the federal capital city (FCC) of Nigeria. Logical transitions to the urban category were modelled for predicting future scenarios for the year 2050 using the embedded land change modeler (LCM) in the IDRISI package. Based on the EAA, the result showed that urban areas increased by more than 11% between 1986 and 2001. In contrast, this value rose to 17% between 2001 and 2014. The LCM model projected LULC changes that showed a growing trend in settlement expansion, which might take over allotted spaces for green areas and agricultural land if stringent development policies and enforcement measures are not implemented. In conclusion, integrating geospatial technologies with ancillary datasets offered improved understanding of how urbanization processes such as increased imperviousness of such a magnitude could influence the urban microclimate through the alteration of natural land surface temperature. Urban expansion could also lead to increased surface runoff as well as changes in drainage geography leading to urban floods. Here, we describe the characterization of a multi-incident MS family that nominated a rare missense variant (p.G420D) in plasminogen (PLG) as a putative genetic risk factor for MS. Genotyping of PLG p.G420D (rs139071351) in 2160 MS patients, and 886 controls from Canada, identified 10 additional probands, two sporadic patients and one control with the variant. Segregation in families harboring the rs139071351 variant, identified p.G420D in 26 out of 30 family members diagnosed with MS, 14 unaffected parents, and 12 out of 30 family members not diagnosed with disease. Despite considerably reduced penetrance, linkage analysis supports cosegregation of PLG p.G420D and disease. Genotyping of PLG p.G420D in 14446 patients, and 8797 controls from Canada, France, Spain, Germany, Belgium, and Austria failed to identify significant association with disease (P = 0.117), despite an overall higher prevalence in patients (OR = 1.32; 95% CI = 0.93–1.87). To assess whether additional rare variants have an effect on MS risk, we sequenced PLG in 293 probands, and genotyped all rare variants in cases and controls. This analysis identified nine rare missense variants, and although three of them were exclusively observed in MS patients, segregation does not support pathogenicity. PLG is a plausible biological candidate for MS owing to its involvement in immune system response, blood-brain barrier permeability, and myelin degradation. Moreover, components of its activation cascade have been shown to present increased activity or expression in MS patients compared to controls; further studies are needed to clarify whether PLG is involved in MS susceptibility. Three major production recipes can be distinguished, i.e. alkali glazes, alkali lead glazes, and lead glazes. In the work of Tite (2011), Islamic glazes from Egypt, Iran, Iraq, and Syria are subdivided into four groups of composition, being partly consistent with those of this work. The alkali lime glazes with <2 wt% PbO correspond to the alkali glazes, but with higher content of CaO. The second and third group of low lead alkali and lead alkali glazes (2-10 wt% PbO and 10-35 wt% PbO) can be subsumed to the alkali lead group described here. Tite´s high lead group has PbO contents >35 wt% and is comparable to the lead glazes (>30 wt% PbO) of this study. The lead and the alkali oxides serve as a flux for the lowering the melting point. In the interaction of ceramic body and glaze, primarily an influence from Si, Al, and K is observed in the line scans from the cross section of ceramic and glaze. However, the input of ceramic material doesn’t seem to be critical for the classification of glazes according to their alkali and alkali lead compositions. In every epoch and locality, except of the Ilkhanate dynasty in Iran, lead glaze samples can be verified. This is also observed in previous investigations e.g. from medieval Iraq, Jordan and Iran (McCarthy, 1996; Al-Saad, 2002; Holakooei et al., 2014). In the Moroccan and Bulgarian glazes, lead seems to be the only important flux. In part, the lead flux is supplemented by additional alkali contents. The lack of alkali and alkali lead glazes in Bulgarian and Moroccan glazes (assuming that the Ottoman alkali lead glazes are imported tableware) seems to affect the regions with Roman-influenced history and with geographical distance to the Near East alkali flux tradition. For the alkali lead glazes and alkali glazes, the overall characteristic is sodium dominated, although the absolute soda values are in part surprisingly low. Samples from Bukhara, Takht-i-Suleiman and the Turkish localities have the highest, but still moderate Na2O values up to 15 wt%, compared to other analyses from e.g. India (Gill & Rehren, 2011). The source of the alkali flux is either mineral natron or plant ash. The source can be determined regarding the MgO values, limited to 1.3 wt% in mineral natron and exceeding 2.0 wt% in the case of plant ashes. In the samples of the present study, the K2O component is not suitable for the indication of the flux-relevant alkali source due to its broad scattering. The P2O5 contents are also enhanced in the plant ash compositions but the data set is not sufficient for statistical evaluation. An influence of the ceramic body on the glaze composition is observed only for SiO2, Al2O3, and K2O in quartz frit ceramics with slight K-feldspar content. The earliest Uzbek tableware glazes from the 10th-11th century (Seljuq period) were generally produced using a lead flux. The same applies to part of the Uzbek tile glazes which were produced between the 13th and 16th century. In Iran, glazes from the 12th century (Khwarezmid period) are lead glazes, but also alkali-fluxed glazes with mineral natron characteristics can be found. Although the production of lead-rich glazes was established from the 8th-9th century on in Iraq, Syria, and Egypt (Henshaw, 2010; Tite et al., 2011), alkali glazes are found in almost all regions except of Bulgaria and Morocco. Plant ash-fluxed alkali glazes are found in 13th century glazes from Takht-i-Suleiman. The plant ash flux technology is assumed to be continuously used in Mesopotamia, Iran, and Central Asia (Sayre & Smith, 1974; Henderson, 2009), but it could be shown that a parallel use of mineral natron parallel existed in the alkali glaze production from the 12th-15th century from Uzbekistan to Afghanistan. Mineral natron characteristics are also reported by Mason (2004) for Syrian and Iranian alkali glazes on lustre ware of the 8th-14th century. Tile glazes with partly mineral natron compositions are found in the Mughal architectural glazes from the 14th- 17th century from India (Gill et al., 2014). Alkali and alkali lead tile glazes from Samarkand from the 13th century (Mongolian period) have mineral natron flux characteristics, but samples from the 15th century (Timurid period) show plant ash signature. Alkali fluxed Uzbek glazes from Bukhara from the 16th century (Sheibanid dynasty) are also made by plant ash flux and are subdivided into two groups with high and low sodium oxide content. The Afghan alkali glazes have sodium oxide contents similar to the sodium-poor Uzbek subgroup, which points to a possible exchange of glaze makers or glaze making technology from Uzbekistan and Afghanistan in the 15th-17th century. Regarding the extensive exchange of Timurid craftsmen in Central Asia, this option seems to be even more likely (Golombek, 1996). One sample from the 15th century from Afghanistan with mineral natron reveals that this material was parallel used in these centuries. Concerning the colouring of the glazes, it has to be distinguished between pigments and colouring ions which are incorporated in the glassy matrix. The colouring agents for translucent glazes are cations of various transition metals. As ions, Co2+ (blue), Cu2+ (green in a lead rich matrix), Fe3+ (brown/black), Mn4+ (brown/black) and Mn3+ (violet) are determined by EPMA. For opaque yellow, white, and turquoise glazes, different pigments were used. The crystalline pigments are investigated by a µ XRD2 device with the result of SnO2, SiO2, and PbSiO4 as whitening agents. PbSiO4 and Pb2Sn2O6 are found in the yellow glaze, from which only the lead tin oxide causes the yellow colour. In the black glazes, different Cr-rich pigments, Cu-Cr-Mn-oxides and iron containing clinopyroxenes are found, even in samples of the same period and region. Cr-rich particles are also detected in two turquoise Afghan glazes from the 15th and 16th century. The use of the ions of Fe, Cu, Co, Cr, and Mn seems to be widely common in the Islamic glazes and corresponds to the described colouring agents in e.g. the study of Tite (2011). The use of opacifying SnO2 particles is widespread as it is reported from different Islamic glazes from Iraq, Iran, Egypt, and Syria (Henshaw, 2010; O´Kane, 2011; Tite, 2011). The colouring agents are known already from former, e.g. Egyptian, Roman and pre-islamic periods, but especially SnO2 pigments became increasingly widespread in the Islamic glazing tradition. The use of yellow and black pigments instead varies already within the buildings from Bukhara from Cr crystals and clino-pyroxenes in the mosque Khoja Zainuddin to a Cu-Cr-Mn-oxide in the madrassa Mir-i Arab of the same epoch. Regarding the matrix compositions connected with the colouring, a certain assignment within the different locations and epochs can be seen. It is noticeable that e.g. the content of lead in turquoise glazes in Uzbekistan is in the range of 0.0-9.2 wt% Pb, whereas blue glazes are mostly alkali ones with PbO contents <2.0 wt%. The turquoise glazes show, that this restriction is not influenced by any defaults of availability and processability. The assumption of common addition of lead and tin to the glaze, which is already described for Iranian glazes of the 13th century (Allan et al., 1973) cannot be confirmed by correlations of tin and lead oxide in the compositions. 11.2 Portable XRF measurement With the p-XRF, semi-quantitative information about the major element compositions is generated. The depth of the detectable signals depends on the analysed sample setup. The p-XRF data are collected with the XL3 Hybrid device of the company Analyticon Instruments. In the comparison of p-XRF results of the “mining” program from Uzbek glazes with EPMA results, the same major composition groups can be distinguished. The Moroccan glazes, all lead rich, are measured with the “mining” as well as with the “soil” program, revealing a better performance in the “mining” measurements. The deviations are nevertheless high, because of the high lead contents, which make the calculation of matrix correction difficult. The measurement of the colouring oxides MnO2, CoO, and CuO is satisfying with the internal calibration of the device and even improved with the “mining” program measurement, if compared to the results of the “soil” program. The measurements of glaze imitations lead to better results than that of bulk glass. This can be attributed to the smoother surface texture. In spite of the accuracy limits in the measurements of particular elements in glazes, the classification of flux composition into three groups could be confirmed with the p XRF analysis. The measurement precision is therefore sufficient for the semi-quantitative analysis of the flux characteristic of glazes. Especially for the on-site measurement of large sample quantities on historical buildings, the device is a suitable tool. 11.3 Restoration material The ORMOCER® fulfils the requirements of stability, reversibility, and transparency, which are imposed to a modern restoration material. As pigments, historically coloured glass, cobalt blue, Egyptian blue, lead tin yellow, manganese violet, iron oxide, copper oxide, and cassiterite were used. The metal compounds have higher colour intensities than the pigments of coloured glass. It has to be considered that the proportion of ORMOCER® in the batch must be high enough (70 vol%) to guarantee the ORMOCER® properties of weathering and mechanical stability. The adhesion properties of the ORMOCER® and the homogeneity of the mixture are the best in a fraction of max. 30 vol% particles per ORMOCER®. With integrated particles, the ORMOCER® G materials show homogeneous coatings, whereas the particles in the ORMCOER® E show more agglomeration. In the sedimentation and weathering experiments, the use of an ultrasonic finger in combination with a roller mill is favourable compared to the treatment with bead grinding mill. The treatments with ultrasonic finger and roller mill result in less sedimentation and better adhesion of the dispersions. The treatment of the dispersions in the bead grinding mill does not result in sufficient adhesion, certainly due to the sedimentation behaviour and a congregation of particles on the bottom of the coating. The modification of dispersed nano-particles by 3-methacryl-oxypropyltrimethoxysilan leads to a further homogenization in the sedimentation tests. It is therefore approved for the use in coloured glaze supplements. In weathered coatings of nano-particle compounds, the surface modification shows certainly no enhancement of stability. The treatment of pigmented coatings with an additional layer of pure ORMOCER® results in a bright and transparent appearing, which is closer to the original optical appearance of the glaze. A long-time test application on a historical building will be the next step to validate the suitability of the restoration material. Methods We enriched and re-sequenced continuous genomic regions comprising FTO, MC4R, TMEM18, SDCCAG8, TKNS, MSRA and TBC1D1 in a screening sample of 196 extremely obese children and adolescents with age and sex specific body mass index (BMI) ≥ 99th percentile and 176 lean adults (BMI ≤ 15th percentile). 22 variants were confirmed by Sanger sequencing. Genotyping was performed in up to 705 independent obesity trios (extremely obese child and both parents), 243 extremely obese cases and 261 lean adults. Results and Conclusion We detected 20 different non-synonymous variants, one frame shift and one nonsense mutation in the 7 continuous genomic regions in study groups of different weight extremes. For SNP Arg695Cys (rs58983546) in TBC1D1 we detected nominal association with obesity (pTDT = 0.03 in 705 trios). Eleven of the variants were rare, thus were only detected heterozygously in up to ten individual(s) of the complete screening sample of 372 individuals. Two of them (in FTO and MSRA) were found in lean individuals, nine in extremely obese. In silico analyses of the 11 variants did not reveal functional implications for the mutations. Concordant with our hypothesis we detected a rare variant that potentially leads to loss of FTO function in a lean individual. For TBC1D1, in contrary to our hypothesis, the loss of function variant (Arg443Stop) was found in an obese individual. Functional in vitro studies are warranted. The mucosal inflammation is thought to arise from a standoff between the pathogen's virulence factors and the host's mucosal innate immune defenses, particularly the mucosal NAIP/NLRC4 inflammasome. However, it had remained unclear how this switches the gut from homeostasis to inflammation. This was studied using the streptomycin mouse model. S.Tm infections in knockout mice, cytokine inhibition and –injection experiments revealed that caspase-1 (not -11) dependent IL-18 is pivotal for inducing acute inflammation. IL-18 boosted NK cell chemoattractants and enhanced the NK cells' migratory capacity, thus promoting mucosal accumulation of mature, activated NK cells. NK cell depletion and Prf\(^{-/-}\) ablation (but not granulocyte-depletion or T-cell deficiency) delayed tissue inflammation. Our data suggest an NK cell perforin response as one limiting factor in mounting gut mucosal inflammation. The purpose of this study was to use proton (hydrogen 1)-magnetic resonance spectroscopy (\(^{1}\)H-MRS) at 3Tesla (3 T) field strength to assess potential gender-related differences in myocardial triglyceride content in healthy individuals. Methods: Cardiac MR imaging was performed to enable accurate voxel placement and obtain functional and morphological information. Double triggered (i.e., ECG and respiratory motion gating) \(^{1}\)H-MRS was used to quantify myocardial triglyceride levels for each gender. Two-sample t-test and Mann-Whitney U-test were used for statistical analyses. Results: In total, 40 healthy volunteers (22 male, 18 female; aged >18 years and age matched) were included in the study. Median myocardial triglyceride content was 0.28% (interquartile range [IQR] 0.17–0.42%) in male and 0.24% (IQR 0.14–0.45%) in female participants, and no statistically significant difference was observed between the genders. Furthermore, no gender-specific difference in ejection fraction was observed, although on average, male participants presented with a higher mean ± SD left ventricular mass (136.3 ± 25.2 g) than female participants (103.9 ± 16.1 g). Conclusions: The study showed that \(^{1}\)H-MRS is a capable, noninvasive tool for acquisition of myocardial triglyceride metabolites. Myocardial triglyceride concentration was shown to be unrelated to gender in this group of healthy volunteers. In contrast, antiviral mechanisms initiated by interferon (IFN) signaling predominate in mammals. Whilst mammalian IFN-induced miRNA are known to inhibit specific viruses, it is not known whether host-directed microRNAs, downstream of IFN-signaling, have a role in mediating broad antiviral resistance. By performing an integrative, systematic, global analysis of RNA turnover utilizing 4-thiouridine labeling of newly transcribed RNA and pri/pre-miRNA in IFN-activated macrophages, we identify a new post-transcriptional viral defense mechanism mediated by miR-342-5p. On the basis of ChIP and site-directed promoter mutagenesis experiments, we find the synthesis of miR-342-5p is coupled to the antiviral IFN response via the IFN-induced transcription factor, IRF1. Strikingly, we find miR-342-5p targets mevalonate-sterol biosynthesis using a multihit mechanism suppressing the pathway at different functional levels: transcriptionally via SREBF2, post-transcriptionally via miR-33, and enzymatically via IDI1 and SC4MOL. Mass spectrometry-based lipidomics and enzymatic assays demonstrate the targeting mechanisms reduce intermediate sterol pathway metabolites and total cholesterol in macrophages. These results reveal a previously unrecognized mechanism by which IFN regulates the sterol pathway. The sterol pathway is known to be an integral part of the macrophage IFN antiviral response, and we show that miR-342-5p exerts broad antiviral effects against multiple, unrelated pathogenic viruses such Cytomegalovirus and Influenza A (H1N1). Metabolic rescue experiments confirm the specificity of these effects and demonstrate that unrelated viruses have differential mevalonate and sterol pathway requirements for their replication. This study, therefore, advances the general concept of broad antiviral defense through multihit targeting of a single host pathway. While molecular experiments provide the most reliable annotation of proteins, their relatively low throughput and restricted purview have led to an increasing role for computational function prediction. However, assessing methods for protein function prediction and tracking progress in the field remain challenging. Results We conducted the second critical assessment of functional annotation (CAFA), a timed challenge to assess computational methods that automatically assign protein function. We evaluated 126 methods from 56 research groups for their ability to predict biological functions using Gene Ontology and gene-disease associations using Human Phenotype Ontology on a set of 3681 proteins from 18 species. CAFA2 featured expanded analysis compared with CAFA1, with regards to data set size, variety, and assessment metrics. To review progress in the field, the analysis compared the best methods from CAFA1 to those of CAFA2. Conclusions The top-performing methods in CAFA2 outperformed those from CAFA1. This increased accuracy can be attributed to a combination of the growing number of experimental annotations and improved methods for function prediction. The assessment also revealed that the definition of top-performing algorithms is ontology specific, that different performance metrics can be used to probe the nature of accurate predictions, and the relative diversity of predictions in the biological process and human phenotype ontologies. While there was methodological improvement between CAFA1 and CAFA2, the interpretation of results and usefulness of individual methods remain context-dependent. Compared to Wannier-Mott excitons in inorganic semiconductors, the localized Frenkel excitons in organic emitter materials show weaker interaction with each other but stronger coupling to light, which recently enabled the first realization of a polariton condensate at room temperature. However, this required ultrafast optical pumping, which limits the applications of organic polariton condensates. We demonstrate room temperature polariton condensates of cavity polaritons in simple laminated microcavities filled with biologically produced enhanced green fluorescent protein (eGFP). The unique molecular structure of eGFP prevents exciton annihilation even at high excitation densities, thus facilitating polariton condensation under conventional nanosecond pumping. Condensation is clearly evidenced by a distinct threshold, an interaction-induced blueshift of the condensate, long-range coherence, and the presence of a second threshold at higher excitation density that is associated with the onset of photon lasing. We investigated if it is possible to use auditory stimuli to create a BCI for the Japanese Hiragana syllabary, which has 46 Hiragana characters. Additionally, we investigated if training has an effect on accuracy despite the high amount of different stimuli involved. Able-bodied participants (N = 6) were asked to select 25 syllables (out of fifty possible choices) using a two step procedure: First the consonant (ten choices) and then the vowel (five choices). This was repeated on 3 separate days. Additionally, a person with spinal cord injury (SCI) participated in the experiment. Four out of six healthy participants reached Hiragana syllable accuracies above 70% and the information transfer rate increased from 1.7 bits/min in the first session to 3.2 bits/min in the third session. The accuracy of the participant with SCI increased from 12% (0.2 bits/min) to 56% (2 bits/min) in session three. Reliable selections from a 10 × 5 matrix using auditory stimuli were possible and performance is increased by training. We were able to show that auditory P300 BCIs can be used for communication with up to fifty symbols. This enables the use of the technology of auditory P300 BCIs with a variety of applications. No single satellite sensor has thus far been able to provide consistent time series of high temporal and spatial resolution for such areas. In order to overcome this problem, data fusion algorithms such as the Enhanced Spatial and Temporal Adaptive Reflectance Fusion Model (ESTARFM) have been established and frequently used in recent years to generate high-resolution time series. In order to make it applicable to larger scales and to increase the input data availability especially in cloud-prone areas, an ESTARFM framework was developed in this study introducing several enhancements. An automatic filling of cloud gaps was included in the framework to make best use of available, even partly cloud-covered Landsat images. Furthermore, the ESTARFM algorithm was enhanced to automatically account for regional differences in the heterogeneity of the study area. The generation of time series was automated and the processing speed was accelerated significantly by parallelization. To test the performance of the developed ESTARFM framework, MODIS and Landsat-8 data were fused for generating an 8-day NDVI time series for a study area of approximately 98,000 km\(^{2}\) in West Africa. The results show that the ESTARFM framework can accurately produce high temporal resolution time series (average MAE (mean absolute error) of 0.02 for the dry season and 0.05 for the vegetative season) while keeping the spatial detail in such a heterogeneous, cloud-prone region. We now show that knockin replacement of murine Pgp with its phosphatase-inactive Pgp\(^{D34N}\) mutant is embryonically lethal due to intrauterine growth arrest and developmental delay in midgestation. PGP inactivation attenuated triosephosphate isomerase activity, increased triglyceride levels at the expense of the cellular phosphatidylcholine content, and inhibited cell proliferation. These effects were prevented under hypoxic conditions or by blocking phosphoglycolate release from damaged DNA. Thus, PGP is essential to sustain cell proliferation in the presence of oxygen. Collectively, our findings reveal a previously unknown mechanism coupling a DNA damage repair product to the control of intermediary metabolism and cell proliferation. We recently proposed Sequence Bundles, a motif visualization technique that maintains a one-to-one relationship between sequences and their graphical representation and visualizes covariant sites. We here present Alvis, an open-source platform for the joint explorative analysis of MSAs and phylogenetic trees, employing Sequence Bundles as its main visualization method. Alvis combines the power of the visualization method with an interactive toolkit allowing detection of covariant sites, annotation of trees with synapomorphies and homoplasies, and motif detection. It also offers numerical analysis functionality, such as dimension reduction and classification. Alvis is user-friendly, highly customizable and can export results in publication-quality figures. Die Zielsetzung dieser Arbeit bestand darin, den Einfluss der postovulatorischen Alterung und der ovariellen Alterung im Holstein-Rind (Bos taurus) auf die DNA-Methylierung entwicklungsrelevanter Gene in Eizellen und Embryonen zu untersuchen. Aus Schlachthof-Ovarien wurden Antralfollikeln unterschiedlicher Größe (<2 mm, 3-5 mm und >6 mm) isoliert. Eizellen aus Follikeln der Größe 3-5 mm wurden für 24h (physiologisch) und 48h (gealtert) in vitro gereift (IVM). Die gereiften Oozyten wurden anschließend in vitro fertilisiert und Embryonen im 4-6 Zellstadium generiert. Sowohl in den unreifen Eizellen aus Antralfollikeln unterschiedlicher Größe als auch in den gereiften Oozyten und den Embryonen wurde die Promotormethylierung der Gene bH19, bSNRPN, bZAR1, bDNMT3A, bOCT4, bDNMT3Lo und bDNMT3Ls analysiert. In den daraus isolierten unreifen Eizellen wurde die DNA-Methylierung der Promotorregionen der Gene bTERF2, bREC8, bBCL-XL, bPISD, bBUB1, bDNMT3Lo, bH19 und bSNRPN bestimmt. Als Methode zur Analyse der Promotormethylierung wurde die Limiting Dilution Bisulfit-Sequenzierung angewendet. In unreifen Eizellen aus Antralfollikeln unterschiedlicher Größe (<2 mm, 3-5 mm und >6 mm) konnte ein erhöhtes Auftreten abnormal methylierter Allele in den geprägten Genen bH19 und bSNRPN von Eizellen kleiner Follikel (<2 mm) identifiziert werden. Dieses Ergebnis könnte eine mögliche Ursache einer bereits bekannten und mehrfach beschriebenen geringeren Entwicklungskompetenz von Eizellen kleiner Follikel (<2 mm) auf epigenetischer Ebene darstellen. Die verlängerte Reifungsdauer der IVM-Eizellen hatte eine signifikante Hypermethylierung in der Promotorregion des Gens DNMT3Lo von 48h-gereiften Eizellen zur Folge. Beim Übergang von 48h-gereiften Eizellen zum Embryo konnte eine signifikante Hypomethylierung von CpG7 des stammzellspezifischen Transkripts DNMT3Ls beobachtet werden. Diese CpG-Stelle wies ebenfalls einen signifikanten Anstieg von CpGs mit nicht-eindeutigem Methylierungszustand in unreifen Eizellen mit steigender Follikelgröße auf. Da sich die CpG-Position innerhalb eines Sequenz-Motivs einer Bindungsstelle des Transkriptionsfaktors CREB befindet, könnten die Methylierungsdaten auf eine Interaktion zwischen dem Transkriptionsfaktor CREB und der DNA-Methylierung während der Entwicklung und Reifung der Eizelle sowie der Transition von der Eizelle zum Embryo hindeuten. Die DNA-Methylierungsprofile der untersuchten Gene in unreifen Eizellen aus Kühen unterschiedlichen Alters (9-12 Monate, 3-7 Jahre und 8-11 Jahre) wiesen keine signifikanten Unterschiede zwischen den Altersgruppen auf. Die ovarielle Alterung bei Rindern zwischen 9 Monaten und 11 Jahren zeigte damit keinen Effekt auf die DNA-Methylierung der untersuchten Promotorregionen der Gene bTERF2, bREC8, bBCL-XL, bPISD, bBUB1, bDNMT3Lo, bH19 und bSNRPN. Nach einer simulierten postovulatorischen Alterung durch eine in vitro Reifung für 48h konnte eine Veränderung der DNA-Methylierung der Oozyten-spezifischen (DNMT3Lo) und Stammzell-spezifischen (DNMT3Ls) Promotoren des katalytisch inaktiven Cofaktors von DNMT3A, DNMT3L, beobachtet werden. Die veränderte DNA-Methylierung von DNMT3Ls tritt dabei erst im frühen Embryo in Erscheinung und interagiert vermutlich mit dem Transkriptionsfaktor CREB. Die Veränderungen von DNMT3Lo in Eizellen und DNMT3Ls in den daraus generierten Embryonen lässt vermuten, dass es sich hierbei um eine dynamische Anpassung des Embryos auf äußere Umweltbedingungen der Eizelle über die Methylierung der DNA handelt. N2 - Postovulatory aging and ovarian aging have been identified as key factors in assisted reproductive techniques (ARTs) and have a lasting effect on reproductive success. Postovulatory aging occurs if the mature egg is not fertilized within its physiological time window. On the other hand, ovarian aging describes the decrease in the follicular reserve with increasing age of the female or the ovary, respectively. Both post-ovulatory aging and ovarian aging result in reduced oocyte quality and lower blastocyst rate. The aim of this thesis was to explore the effects of postovulatory aging and ovarian aging in Holstein cattle (Bos taurus) on the DNA methylation of developmentally important genes in oocytes and embryos. Antral follicles of different sizes (<2 mm, 3-5 mm and> 6 mm) were isolated from slaughterhouse ovaries. Female germ cells from middle-sized follicles (3-5 mm) were matured for 24h (physiological conditions) and 48h (aged conditions) in vitro (IVM). The IVM- oocytes were subsequently fertilized in vitro and embryos at the 4-6 cell stage were generated. Promoter methylation of the genes bH19, bSNRPN, bZAR1, bDNMT3A, bOCT4, bDNMT3Lo and bDNMT3Ls was analysed in immature oocytes from antral follicles of different sizes as well as in matured oocytes and the respective embryos. For studying ovarian aging, middle-sized antral follicles were obtained in vivo from animals of different age groups (9-12 months, 3-7 years and 8-11 years). In the extracted immature gametes, the DNA methylation of the promoter regions of bTERF2, bREC8, bBCL-XL, bPISD, bBUB1, bDNMT3Lo, bH19 and bSNRPN was examined. The limiting dilution bisulfite (pyro)sequencing method was applied to determine the promoter methylation of the candidate genes at the single allele level. In immature oocytes from antral follicles of different diameters (<2 mm, 3-5 mm and> 6 mm) an increased occurrence of abnormally methylated alleles of the imprinted genes bH19 and bSNRPN was identified in small follicles (<2 mm). This failure to establish imprinting could be a possible cause of a well-known reduced developmental potential of small follicles (<2 mm) at the epigenetic level. The extended maturation time of the IVM-oocytes resulted in a significant hypermethylation in the promoter region of DNMT3Lo in 48h matured oocytes. After transition from 48h matured oocytes to embryos, a significant hypomethylation of CpG7 of the stem cell specific transcript DNMT3Ls was detected. The same CpG site showed a significant increase of CpGs with unclear methylation state in immature female germ cells with increasing follicular size. This CpG position is located within a potential binding site of the transcription factor CREB. Thus, the methylation data indicates an interaction between the transcription factor CREB and the DNA methylation during development and maturation of oocytes as well as during transition from the oocyte to the embryo. The DNA methylation profiles of the analysed genes in immature oocytes from cows of different age (9-12 months, 3-7 years and 8-11 years) showed no significant differences between age groups. Hence, the ovarian aging in cattle between 9 months and 11 years caused no effect on the DNA methylation of bTERF2, bREC8, bBCL-XL, bPISD, bBUB1, bDNMT3Lo, bH19 and bSNRPN. After a simulated postovulatory aging by in vitro maturation for 48h, a change in the DNA methylation of the oocyte-specific (DNMT3Lo) and stem cell-specific (DNMT3Ls) promoters of the catalytically inactive DNA-methyltransferase DNMT3L was observed. The altered DNA methylation of DNMT3Ls occurs in the early embryo and probably interacts with the transcription factor CREB. The changes of DNMT3Lo in oocytes and DNMT3Ls in the resulting embryos might represent a dynamic adaptation to external environmental conditions. Based on the corneal anatomy and current knowledge of the mechanisms causing eye irritation, different ex vivo and in vitro methods will be presented and discussed with regard to possible limitations and status of regulatory acceptance. Especially atypical or late-onset type 2 phenotypes present a therapeutical dilemma. Methods To determine the clinical impact of the alpha-Galactosidase A (GLA) p.A143T/ c.427G > A variation, we retrospectively analyzed 25 p.A143T patients in comparison to 58 FD patients with other missense mutations. Results p.A143T patients suffering from stroke/ transient ischemic attacks had slightly decreased residual GLA activities, and/or increased lyso-Gb3 levels, suspecting FD. However, most male p.A143T patients presented with significant residual GLA activity (~50 % of reference), which was associated with normal lyso-Gb3 levels. Additionally, p.A143T patients showed less severe FD-typical symptoms and absent FD-typical renal and cardiac involvement in comparison to FD patients with other missense mutations. Two tested female p.A143T patients with stroke/TIA did not show skewed X chromosome inactivation. No accumulation of neurologic events in family members of p.A143T patients with stroke/transient ischemic attacks was observed. Conclusions We conclude that GLA p.A143T seems to be most likely a neutral variant or a possible modifier instead of a disease-causing mutation. Therefore, we suggest that p.A143T patients with stroke/transient ischemic attacks of unknown etiology should be further evaluated, since the diagnosis of FD is not probable and subsequent ERT or chaperone treatment should not be an unreflected option. The circadian system and peptidergic signals are important components of this modulation, but still little is known about the mechanisms and networks by which they interact to regulate feeding and sleep. We show that specific thermogenetic activation of peptidergic Allatostatin A (AstA)-expressing PLP neurons and enteroendocrine cells reduces feeding and promotes sleep in the fruit fly Drosophila. The effects of AstA cell activation are mediated by AstA peptides with receptors homolog to galanin receptors subserving similar and apparently conserved functions in vertebrates. We further identify the PLP neurons as a downstream target of the neuropeptide pigment-dispersing factor (PDF), an output factor of the circadian clock. PLP neurons are contacted by PDF-expressing clock neurons, and express a functional PDF receptor demonstrated by cAMP imaging. Silencing of AstA signalling and continuous input to AstA cells by tethered PDF changes the sleep/activity ratio in opposite directions but does not affect rhythmicity. Taken together, our results suggest that pleiotropic AstA signalling by a distinct neuronal and enteroendocrine AstA cell subset adapts the fly to a digestive energy-saving state which can be modulated by PDF. These terms give rise to additional spin transitions, which would be otherwise forbidden, and lead to a level anticrossing in an external magnetic field. We observe a sharp variation of the photoluminescence intensity in the vicinity of this level anticrossing, which can be used for a purely all-optical sensing of the magnetic field. We achieve dc magnetic field sensitivity better than 100  nT/√Hz within a volume of 3×10\(^{−7}\)mm\(^3\) at room temperature and demonstrate that this contactless method is robust at high temperatures up to at least 500 K. Neben rein komponentenoptimierenden Maßnahmen, wie der Verbesserung der Batterie, ist die Förderung von Eco-Driving, also einer energieeffizienten Fahrweise, ein effektiver Ansatz zur Steigerung der Reichweite. Trainings und visuell dargebotene Eco-Assistenten können Eco-Driving wirksam steigern, schöpfen aber nicht dessen gesamtes Potential aus. Angepasste Pedal- und Motorkennlinien könnten Eco-Driving zusätzlich fördern. Für deren Bewertung sind die Wirksamkeit und Akzeptanz bisher nicht gemeinsam berücksichtig worden oder sie wurden nicht im Elektrofahrzeug evaluiert und validiert. Zu diesen Anpassungen zählen eine Veränderung des Beschleunigungspedals, sodass mit diesem gleichzeitig beschleunigt und rekuperiert werden kann, die Limitierung von Drehmoment und Leistung und der Einsatz eines aktiven Beschleunigungspedals, welches Widerstände abhängig von Fahrzeug- oder Situationsparametern aktiviert. Für diese Arbeit habe ich daher die Pedal- und Motorkennlinien entsprechend angepasst und in ein validiertes Elektroautomodell implementiert. Ziel war es, verschiedene Fahrverhaltensbereiche im Elektrofahrzeug, die Eco-Driving kennzeichnen (energieoptimales Beschleunigen und Verzögern, Einhalten von Geschwindigkeitsbegrenzungen, vorausschauendes Fahren), benutzerfreundlich, akzeptabel und wirksam zu unterstützen. Zu diesem Zweck habe ich vier Probandenstudien im bewegten Fahrsimulator durchgeführt und geeignete Pedal- und Motorkennlinien empirisch bestimmt. In der ersten Studie habe ich untersucht, ob und warum eine Pedalkennlinie zu bevorzugen ist, bei der mit dem Beschleunigungspedal anstelle des Bremspedals rekuperiert wird. Das Ziel der zweiten Studie war es, eine geeignete Rekuperationsstärke für ein kombiniertes Beschleunigungspedal, bei dem mit dem Beschleunigungspedal rekuperiert wird, zu bestimmen. In der dritten Studie habe ich evaluiert, ob die Limitierung der Leistung oder die des Drehmoments zu bevorzugen ist, um das Beschleunigungsverhalten zu optimieren und wie stark die Limitierungen optimaler Weise sein sollten. Basierend auf den Ergebnissen der dritten Studie, habe ich schließlich einen optimierten Limitierungsansatz konzipiert, diesen mit einem aktiven Beschleunigungspedal verglichen und bestimmt, welcher Ansatz zu bevorzugen ist. Aufgrund der Studienergebnisse werden folgende Ansätze für die jeweiligen Eco-Driving-Fahrverhaltensbereiche empfohlen und es werden folgende Gestaltungsempfehlungen abgeleitet: Zur Förderung eines energieeffizienten Beschleunigungsverhaltens ist die Limitierung von Drehmoment und Leistung geeignet. Die Limitierung des Drehmoments ist hierbei besonders wirksam in geringen, die Limitierung der maximalen Leistung in höheren Geschwindigkeitsbereichen. Zu empfehlen sind parallele mittelstarke Limitierungen von maximalem Drehmoment und maximaler Leistung, die Beschleunigungen mit 2.0 m/s² erlauben, bei gleichzeitiger Bereitstellung eines Kick-Downs. Ein aktives Beschleunigungspedal ist insbesondere aus Gründen der Benutzerfreundlichkeit zur Förderung eines energieeffizienten Beschleunigungsverhaltens nur eingeschränkt empfehlenswert. Zur Förderung eines energieeffizienten Verzögerungsverhaltens wird die Implementierung der Rekuperationsfunktion auf dem Beschleunigungspedal anstelle des Bremspedals empfohlen, da dies einerseits ermöglicht, auf hydraulisches Bremsen zu verzichten und gleichzeitig mehr Energie rekuperiert werden kann. Ersteres trägt zu einer hohen Akzeptanz bei, letzteres zu einer günstigen Energiebilanz. Besonders effektiv und akzeptabel ist ein kombiniertes Fahrbremspedal, wenn es eine starke Rekuperation ermöglicht (zwischen -1.7 und -2.1 m/s²). Weiterhin ist ein aktives Beschleunigungspedal, das den geeigneten Zeitpunkt für eine maximal energieeffiziente Verzögerung mit einem kombinierten Fahrbremspedal anzeigt, wirksam, um die rekuperierte Energie zu steigern. Auf diese Weise kann zudem eine vorausschauende Fahrweise unterstützt werden. Hierbei muss jedoch die Funktionalität und Benutzerfreundlichkeit optimiert werden, um eine gesteigerte kognitive Fahrerbeanspruchung und Minderungen der Akzeptanz zu vermeiden. Zur Unterstützung der Einhaltung von Geschwindigkeitsbegrenzungen ist ebenfalls das aktive Beschleunigungspedal geeignet. Der Fahrer sollte hierbei aber die Möglichkeit haben, individuell Grenzwerte einzustellen. Die Verknüpfung eines kombinierten Fahrbremspedals mit einer Limitierung von Drehmoment und Leistung sowie einem aktiven Beschleunigungspedal kann abschließend, unter Berücksichtigung der abgeleiteten Gestaltungsempfehlungen, als effektive und akzeptable Möglichkeit zur Förderung unterschiedlicher Verhaltensbereiche von Eco-Driving bewertet werden. Die erwarteten Synergieeffekte der evaluierten Ansätze in Verbindung mit Eco-Trainings und visuell dargebotenen Eco-Feedback-Assistenten sowie deren Langfristigkeit sollten Gegenstand weiterführender Forschung sein. N2 - The limited range of electric vehicles (EV) is one of the major reasons for the current lack of buying interest. Besides optimizing battery capacity, eco-driving is an effective approach for extending EVs range. Trainings and visually presented assistance systems are effective but do max out the full potential of eco-driving. Yet, adapting accelerator pedal and motor characteristics may additionally support eco-driving. So far, these adaptations were not evaluated concerning both usability and effectiveness simultaneously, however, or they were not evaluated and validated in the context of EVs. The adaptations of interest are first a combination of accelerator and brake pedal, which allows accelerating and regenerative braking by means of only one pedal, second limitations of engine’s maximum torque or power and third an active accelerator pedal which activates forces depending on vehicle or situation parameters. Within the scope of this thesis, I therefore adapted accelerator pedal and motor characteristics respectively and implemented them in a validated electric vehicle model. The aim was to support several eco-driving characteristics (energy efficient accelerating and decelerating, compliance with speed limits, anticipatory driving) in a usable and effective manner. In order to determine suitable accelerator pedal and motor characteristics I thus conducted four experiments in a dynamic simulator. In the first study I evaluated whether recuperating by means of the accelerator or the brake pedal is preferred. The aim of the second study was to determine an appropriate recuperation force when using a combined accelerator pedal which allows recuperating by means of the accelerator pedal. In the third study I evaluated on the one hand whether a limitation of engines torque or power is preferred in order to support energy efficient acceleration and on the other hand how intense limitations should be. Based on the findings of the third study I finally optimized the limitation approach, compared this approach with an active accelerator pedal and evaluated which approach is preferred. Based on the study results, I recommend following adaptations for the respective eco-driving characteristics and derive further design implications: Limiting maximum torque and power is most appropriate for supporting energy efficient acceleration behavior. The limitation of maximum torque is most effective at lower speed range, the limitation of maximum power at higher speed range. I recommend limitating maximum torque and power simultaneously, however, allowing accelerations of 2.0 m/s² and proving a kick-down which deactivates the limitations. Though an active accelerator pedal is effective for supporting energy efficient acceleration behavior it is not recommended for usability reasons. For supporting energy efficient deceleration behavior the implementation of regenerative braking on the accelerator pedal instead of the braking pedal is recommended as it enables dispensing with hydraulic braking on the one hand and increases regenerated energy on the other hand. The former contributes to a high level of acceptance, the latter to a favorable energy balance. In particular strong regenerative braking (between -1.7 and -2.1 m/s²), when using a combined pedal solution, is effective and usable. Furthermore, an active accelerator pedal, which indicates the appropriate time for a maximum energy efficient deceleration with a combined pedal solution, is effective to increase regenerated energy. By this means, also an anticipatory driving style is supported. Nevertheless, accuracy of function and its usability need to be optimized in order to avoid increasing driver workload and decreasing acceptance. Furthermore, an active accelerator pedal is also applicable for supporting compliance with speed limits. However, the possibility of individual customizing of thresholds should be provided. Finally, taking into account the design recommendations, a combination of a combined pedal solution, limitation of maximum torque and power and an active accelerator pedal is a usable and effective approach for supporting various eco-driving characteristics. The expected synergy effects of the evaluated approaches in combination with eco-trainings and visually presented eco-feedback assistance system as well as their long-term effectiveness should be subject of further research. Our previous work has shown that light exposure induces structural neuronal plasticity in the mushroom bodies (MBs), a brain center implicated in processing inputs from sensory modalities. Here, we extended these analyses to the molecular level to unravel light-induced transcriptomic and epigenomic changes in the honey bee brain. We have compared gene expression in brain compartments of 1- and 7-day-old light-exposed honey bees with age-matched dark-kept individuals. We have found a number of differentially expressed genes (DEGs), both novel and conserved, including several genes with reported roles in neuronal plasticity. Most of the DEGs show age-related changes in the amplitude of light-induced expression and are likely to be both developmentally and environmentally regulated. Some of the DEGs are either known to be methylated or are implicated in epigenetic processes suggesting that responses to light exposure are at least partly regulated at the epigenome level. Consistent with this idea light alters the DNA methylation pattern of bgm, one of the DEGs affected by light exposure, and the expression of microRNA miR-932. This confirms the usefulness of our approach to identify candidate genes for neuronal plasticity and provides evidence for the role of epigenetic processes in driving the molecular responses to visual stimulation. Besides, increasing evidence points to ADHD patients showing emotional dysfunctions and concomitant problems in social life. However, systematic research on emotional dysfunctions in ADHD is still rare, and to date most studies lack conceptual differentiation between emotion processing and emotion regulation. The aim of this thesis was to systematically investigate emotion processing and emotion regulation in adult ADHD in a virtual reality paradigm implementing social interaction. Emotional reactions were assessed on experiential, physiological, and behavioral levels. Experiment 1 was conducted to develop a virtual penalty kicking paradigm implying social feedback and to test it in a healthy sample. This paradigm should then be applied in ADHD patients later on. Pleasant and unpleasant trials in this paradigm consisted of hits respectively misses and subsequent feedback from a virtual coach. In neutral trials, participants were teleported to different spots of the virtual stadium. Results indicated increased positive affectivity (higher valence and arousal ratings, higher zygomaticus activations, and higher expression rates of positive emotional behavior) in response to pleasant compared to neutral trials. Reactions to unpleasant trials were contradictory, indicating increased levels of both positive and negative affectivity, compared to neutral trials. Unpleasant vs. neutral trials revealed lower valence ratings, higher arousal ratings, higher zygomaticus activations, slightly lower corrugator activations, and higher expression rates of both positive and negative emotional behavior. The intensity of emotional reactions correlated with experienced presence in the virtual reality. To better understand the impact of hits or misses per se vs. hits or misses with coach feedback healthy participants’ emotional reactions, only 50% of all shots were followed by coach feedback in experiment 2. Neutral trials consisted of shots over the free soccer field which were followed by coach feedback in 50 % of all trials. Shots and feedback evoked more extreme valence and arousal ratings, higher zygomaticus activations, lower corrugator activations, and higher skin conductance responses than shots alone across emotional conditions. Again, results speak for the induction of positive emotions in pleasant trials whereas the induction of negative emotions in unpleasant trials seems ambiguous. Technical improvements of the virtual reality were reflected in higher presence ratings than in experiment 1. Experiment 3 investigated emotional reactions of adult ADHD patients and healthy controls after emotion processing and response-focused emotion regulation. Participants successively went through an ostensible online ball-tossing game (cyber ball) inducing negative emotions, and an adapted version of the virtual penalty kicking game. Throughout cyber ball, participants were included or ostracized by two other players in different experimental blocks. Participants were instructed to explicitly show, not regulate, or hide their emotions in different experimental blocks. Results provided some evidence for deficient processing of positive emotions in ADHD. Patients reported slightly lower positive affect than controls during cyber ball, gave lower valence ratings than controls in response to pleasant penalty kicking trials, and showed lower zygomaticus activations than controls especially during penalty kicking. Patients in comparison with controls showed slightly increased processing of unpleasant events during cyber ball (higher ratings of negative affect, especially in response to ostracism), but not during penalty kicking. Patients showed lower baseline skin conductance levels than controls, and impaired skin conductance modulations. Compared to controls, patients showed slight over-expression of positive as well as negative emotional behavior. Emotion regulation analyses revealed no major difficulties of ADHD vs. controls in altering their emotional reactions through deliberate response modulation. Moreover, patients reported to habitually apply adaptive emotion regulation strategies even more frequently than controls. The analyses of genetic high-risk vs. low-risk groups for ADHD across the whole sample revealed similar results as analyses for patients vs. controls for zygomaticus modulations during emotion processing, and for modulations of emotional reactions due to emotion regulation. To sum up, the virtual penalty kicking paradigm proved to be successful for the induction of positive, but not negative emotions. The importance of presence in virtual reality for the intensity of induced emotions could be replicated. ADHD patients showed impaired processing of primarily positive emotions. Aberrations in negative emotional responding were less clear and need further investigation. Results point to adult ADHD in comparison to healthy controls suffering from baseline deficits in autonomic arousal and deficits in arousal modulation. Deficits of ADHD in the deliberate application of response-focused emotion regulation could not be found. N2 - Die Aufmerksamkeitsdefizit-/hyperaktivitätsstörung (ADHS) ist gekennzeichnet durch Symptome der Unaufmerksamkeit und Hyperaktivität/Impulsivität. Zudem sprechen zunehmende Befunde für emotionale Defizite und damit einhergehende soziale Probleme bei ADHS. Bisher gibt es jedoch kaum systematische Untersuchungen zu emotionalen Defiziten bei ADHS, und die meisten bisherigen Studien trennen nicht klar zwischen den Konzepten der Emotionsverarbeitung und Emotionsregulation. Das Ziel dieser Arbeit war es, Emotionsverarbeitung und Emotionsregulation bei erwachsenen ADHS Patienten in einem Paradigma in virtueller Realität mit sozialer Interaktion zu untersuchen. Emotionale Reaktionen wurden auf Erlebnisebene, physiologischer Ebene und Verhaltensebene erfasst. In Experiment 1 wurde ein virtuelles Elfmeterparadigma mit sozialem Feedback entwickelt und an einer gesunden Stichprobe getestet. Dieses Paradigma sollte später mit ADHS Patienten verwendet werden. Angenehme und unangenehme Versuchsdurchgänge bestanden aus Treffern bzw. gehaltenen Torschüssen und einer darauffolgenden Rückmeldung von einem virtuellen Trainer. In neutralen Durchgängen wurden die Teilnehmer zu verschiedenen Punkten im virtuellen Stadion teleportiert. Die Ergebnisse sprechen für erhöhte positive Affektivität (höhere Valenz- und Arousalratings, höhere Zygomaticusaktivität, mehr positiver emotionaler Ausdruck) durch angenehme im Vergleich zu neutralen Durchgängen. Reaktionen auf unangenehme im Vergleich zu neutralen Durchgängen waren widersprüchlich und sprechen für erhöhte positive und negative Affektivität. Unangenehme vs. neutrale Durchgänge führten zu niedrigeren Valenzratings, höheren Arousalratings, höherer Zygomaticusaktivität, etwas niedrigerer Corrugatoraktivität und mehr positivem und negativem emotionalen Ausdruck. Die Intensität emotionaler Reaktionen korrelierte mit dem Präsenzerleben in der virtuellen Realität. Um den Einfluss von Torschüssen allein vs. Torschüssen mit Trainerrückmeldung auf emotionale Reaktionen bei Gesunden besser zu verstehen, folgte in Experiment 2 nur auf 50 % aller Schüsse eine Trainerrückmeldung. Neutrale Durchgänge bestanden aus freien Schüssen über das Fußballfeld, auf die zu 50 % eine Trainerrückmeldung folgte. Schüsse mit Rückmeldung führten durchgängig zu stärkeren Valenz- und Arousalratings, höherer Zygomaticusaktivität, niedrigerer Corrugatoraktivität, und höheren Hautleitfähigkeits- reaktionen als Schüsse allein. Auch diese Ergebnisse sprechen für die Induktion positiver Emotionen durch angenehme Durchgänge, während die Induktion negativer Emotionen durch unangenehme Durchgänge uneindeutig scheint. Technische Verbesserungen der virtuellen Realität schlugen sich in höherem Präsenzerleben als in Experiment 1 nieder. Experiment 3 untersuchte emotionale Reaktionen von erwachsenen ADHS Patienten und gesunden Kontrollen nach Emotionsverarbeitung und reaktionsfokussierter Emotions- regulation. Versuchsteilnehmer absolvierten nacheinander ein vorgebliches Online-Ballspiel (Cyber Ball) welches negative Emotionen induzierte, und eine weiterentwickelte Version des virtuellen Elfmeterspiels. Während Cyber Ball wurden die Teilnehmer von zwei anderen Mitspielern in verschiedenen Versuchsblöcken entweder eingeschlossen oder ausgeschossen. Die Teilnehmer wurden in verschiedenen Versuchsblöcken instruiert, ihre Emotionen entweder deutlich zu zeigen, nicht zu regulieren, oder zu verbergen. Einige Ergebnisse sprechen für eine defizitäre Verarbeitung positiver Emotionen bei ADHS. Patienten berichteten niedrigeren positiven Affekt als Kontrollen während Cyber Ball, niedrigere Valenzratings als Kontrollen nach angenehmen Elfmeterdurchgängen, und zeigten niedrigere Zygomaticusaktivität als Kontrollen insbesondere während des Elfmeterschießens. Im Vergleich zu Kontrollen zeigten Patienten eine leicht verstärkte Verarbeitung von unangenehmen Ereignissen beim Cyber Ball (höhere Ratings von negativem Affekt v.a. nach Ausschluss), aber nicht beim Elfmeterschießen. Patienten zeigten eine niedrigere Baseline- Hautleitfähigkeit als Kontrollen, sowie beeinträchtigte Hautleitfähigkeitsmodulationen. Im Vergleich zu Kontrollen zeigten Patienten leicht erhöhten Ausdruck von positiven und negativen Emotionen. Emotionsregulationsanalysen zeigten keine bedeutenden Einschränkungen von ADHS vs. Kontrollen im Verändern ihrer emotionalen Reaktionen durch absichtliche reaktionsfokussierte Emotionsregulation. Außerdem gaben Patienten an, gewohnheitsmäßig sogar häufiger als Kontrollen adaptive Emotionsregulationsstrategien anzuwenden. Der Vergleich einer genetischen Hochrisikogruppe mit einer Niedrigrisikogruppe für ADHS über die gesamte Stichprobe zeigte ähnliche Ergebnisse wie die Analysen für Patienten vs. Kontrollen in der Modulation von Zygomaticusaktivität während der Emotionsverarbeitung, sowie in der Modulation emotionaler Reaktionen durch Emotionsregulation. Zusammenfassend lässt sich sagen, dass das virtuelle Elfmeterparadigma geeignet scheint, um positive, aber nicht negative Emotionen zu induzieren. Die Bedeutsamkeit von Präsenzerleben in einer virtuellen Realität für die Intensität von induzierten Emotionen konnte repliziert werden. ADHS Patienten zeigten Beeinträchtigungen v.a. in der Verarbeitung von positiven Emotionen. Abweichungen in der negativen emotionalen Reagibilität waren weniger eindeutig und sollten weiter untersucht werden. Die Ergebnisse deuten darauf hin, dass erwachsene ADHS Patienten im Vergleich zu gesunden Kontrollen eine niedrigere autonome Erregbarkeit bei Baseline und Defizite in der Erregbarkeitsmodulation zeigen. Es zeigten sich keine Defizite von ADHS in der absichtlichen Anwendung von ausdrucksfokussierter Emotionsregulation. Little, however, is known about the effect of adenosine on myeloid cells. Considering that tumor associated macrophages (TAM) and myeloid-derived suppressor cells (MDSC) constitute up to 20 % of OvCA tissue, we investigated the effect of adenosine on myeloid cells and explored a possible contribution of myeloid cells to adenosine generation in vitro and ex vivo. Methods Monocytes were used as human blood-derived myeloid cells. After co-incubation with SK-OV-3 or OAW-42 OvCA cells, monocyte migration was determined in transwell assays. For conversion into M2-polarized “TAM-like” macrophages, monocytes were co-incubated with OAW-42 cells. Ex vivo TAMs were obtained from OvCA ascites. Macrophage phenotypes were investigated by intracellular staining for IL-10 and IL-12. CD39 and CD73 expression were assessed by FACS analysis both on in vitro-induced TAM-like macrophages and on ascites-derived ex situ-TAMs. Myeloid cells in solid tumor tissue were analyzed by immunohistochemistry. Generation of biologically active adenosine by TAM-like macrophages was measured in luciferase-based reporter assays. Functional effects of adenosine were investigated in proliferation-experiments with CD4+ T cells and specific inhibitors. Results When CD39 or CD73 activity on OvCA cells were blocked, the migration of monocytes towards OvCA cells was significantly decreased. In vivo, myeloid cells in solid ovarian cancer tissue were found to express CD39 whereas CD73 was mainly detected on stromal fibroblasts. Ex situ-TAMs and in vitro differentiated TAM-like cells, however, upregulated the expression of CD39 and CD73 compared to monocytes or M1 macrophages. Expression of ectonucleotidases also translated into increased levels of biologically active adenosine. Accordingly, co-incubation with these TAMs suppressed CD4+ T cell proliferation which could be rescued via blockade of CD39 or CD73. Conclusion Adenosine generated by OvCA cells likely contributes to the recruitment of TAMs which further amplify adenosine-dependent immunosuppression via additional ectonucleotidase activity. In solid ovarian cancer tissue, TAMs express CD39 while CD73 is found on stromal fibroblasts. Accordingly, small molecule inhibitors of CD39 or CD73 could improve immune responses in ovarian cancer. Medical rehabilitation increasingly considers occupational issues as determinants of health and work ability. Information on work-related rehabilitation concepts should therefore be made available to healthcare professionals. Objective. To revise a website providing healthcare professionals in medical rehabilitation facilities with information on work-related concepts in terms of updating existing information and including new topics, based on recommendations from implementation research. Method. The modification process included a questionnaire survey of medical rehabilitation centers (n=28); two workshops with experts from rehabilitation centers, health payers, and research institutions (n=14); the selection of new topics and revision of existing text modules based on expert consensus; and an update of good practice descriptions of work-related measures. Results. Health payers’ requirements, workplace descriptions, and practical implementation aids were added as new topics. The database of good practice examples was extended to 63 descriptions. Information on introductory concepts was rewritten and supplemented by current data. Diagnostic tools were updated by including additional assessments. Conclusions. Recommendations from implementation research such as assessing user needs and including expert knowledge may serve as a useful starting point for the dissemination of information on work-related medical rehabilitation into practice. Web-based information tools such as the website presented here can be quickly adapted to current evidence and changes in medicolegal regulations. Methods: Twelve healthy competitive young male cyclists (14.4 ± 0.8 years; 57.9 ± 9.4 ml•min−1•kg−1 peak oxygen uptake) performed one session of high intensity 4 × 4 min intervals (HIIT) at 90–95% peak power output (PPO), each interval separated by 3 min of active recovery, and one high volume session (HVT) consisting of a constant load exercise for 90 min at 60% PPO. Capillary blood from the earlobe was collected under resting conditions, during exercise (d1 = 20 min, d2 = 30 min, d3 = 60 min), and 0, 30, 60, 180 min after the exercise to determine miR-16, -21, -126, and VEGF mRNA. Results: HVT significantly increased miR-16 and miR-126 during and after the exercise compared to pre-values, whereas HIIT showed no significant influence on the miRNAs compared to pre-values. VEGF mRNA significantly increased during and after HIIT (d1, 30′, 60′, 180′) and HVT (d3, 0′, 60′). Conclusion: Results of the present investigation suggest a volume dependent exercise regulation of vascular regulating miRNAs (miR-16, miR-21, miR-126) in children. In line with previous data, our data show that acute exercise can alter circulating miRNAs profiles that might be used as novel biomarkers to monitor acute and chronic changes due to exercise in various tissues. Consequently, TNF not only acts as a macrophage-derived effector molecule but also regulates the activity and viability of macrophages. Here, we investigated the individual contribution of TNFR1 and TNFR2 to TNF-induced cell death in macrophages. Exclusive stimulation of TNFR1 showed no cytotoxic effect whereas selective stimulation of TNFR2 displayed mild cytotoxicity. Intriguingly, the latter was strongly enhanced by the caspase inhibitor zVAD-fmk. The strong cytotoxic activity of TNFR2 in the presence of zVAD-fmk was reversed by necrostatin-1, indicating necroptotic cell death. TNFR1- and TNF-deficient macrophages turned out to be resistant against TNFR2-induced cell death. In addition, the cIAP-depleting SMAC mimetic BV6 also enforced TNF/TNFR1-mediated necroptotic cell death in the presence of zVAD-fmk. In sum, our data suggest a model in which TNFR2 sensitizes macrophages for endogenous TNF-induced TNFR1-mediated necroptosis by the known ability of TNFR2 to interfere with the survival activity of TRAF2-cIAP1/2 complexes. Previous studies on EGC pathophysiology have specifically focused on mucosal glia activation and its contribution to mucosal inflammatory processes observed in the gut of inflammatory bowel disease (IBD) patients. In contrast knowledge is scarce on intestinal inflammation not locally restricted to the mucosa but systemically affecting the intestine and its effect on the overall EGC network. Methods and Results In this study, we analyzed the biological effects of a systemic LPS-induced hyperinflammatory insult on overall EGCs in a rat model in vivo, mimicking the clinical situation of systemic inflammation response syndrome (SIRS). Tissues from small and large intestine were removed 4 hours after systemic LPS-injection and analyzed on transcript and protein level. Laser capture microdissection was performed to study plexus-specific gene expression alterations. Upon systemic LPS-injection in vivo we observed a rapid and dramatic activation of Glial Fibrillary Acidic Protein (GFAP)-expressing glia on mRNA level, locally restricted to the myenteric plexus. To study the specific role of the GFAP subpopulation, we established flow cytometry-purified primary glial cell cultures from GFAP promotor-driven EGFP reporter mice. After LPS stimulation, we analyzed cytokine secretion and global gene expression profiles, which were finally implemented in a bioinformatic comparative transcriptome analysis. Enriched GFAP+ glial cells cultured as gliospheres secreted increased levels of prominent inflammatory cytokines upon LPS stimulation. Additionally, a shift in myenteric glial gene expression profile was induced that predominantly affected genes associated with immune response. Conclusion and Significance Our findings identify the myenteric GFAP-expressing glial subpopulation as particularly susceptible and responsive to acute systemic inflammation of the gut wall and complement knowledge on glial involvement in mucosal inflammation of the intestine. Whereas, preTCR-negative thymocytes exhibit only P2 promoter-derived Nfatc1β expression, preTCR-positive thymocytes express both Nfatc1β and P1 promoter-derived Nfatc1α transcripts. Inducing NFATc1α activity from P1 promoter in preTCR-negative thymocytes, in addition to the NFATc1β from P2 promoter impairs thymocyte development resulting in severe T-cell lymphopenia. In addition, we show that NFATc1 activity suppresses the B-lineage potential of immature thymocytes, and consolidates their differentiation to T cells. Further, in the pTCR-positive DN3 cells, a threshold level of NFATc1 activity is vital in facilitating T-cell differentiation and to prevent Notch3-induced T-acute lymphoblastic leukaemia. Here, we dissect the function of the key regulator of serotonin signaling—the serotonin transporter–in innate olfactory attraction to ethanol in Drosophila melanogaster. We generated a mutated version of the serotonin transporter that prolongs serotonin signaling in the synaptic cleft and is targeted via the Gal4 system to different sets of serotonergic neurons. We identified four serotonergic neurons that inhibit the olfactory attraction to ethanol and two additional neurons that counteract this inhibition by strengthening olfactory information. Our results reveal that compensation can occur on the circuit level and that serotonin has a bidirectional function in modulating the innate attraction to ethanol. Given the evolutionarily conserved nature of the serotonin transporter and serotonin, the bidirectional serotonergic mechanisms delineate a basic principle for how random behavior is switched into targeted approach behavior. Here we describe a novel role for one of these ORs, Olfr1393, as a regulator of renal glucose handling. Olfr1393 is specifically expressed in the kidney proximal tubule, which is the site of renal glucose reabsorption. Olfr1393 knockout mice exhibit urinary glucose wasting and improved glucose tolerance, despite euglycemia and normal insulin levels. Consistent with this phenotype, Olfr1393 knockout mice have a significant decrease in luminal expression of Sglt1, a key renal glucose transporter, uncovering a novel regulatory pathway involving Olfr1393 and Sglt1. In addition, by utilizing a large scale screen of over 1400 chemicals we reveal the ligand profile of Olfr1393 for the first time, offering new insight into potential pathways of physiological regulation for this novel signaling pathway. In blazars, which dominate the extragalactic gamma-ray sky, the jets are aligned close to the observer's line of sight leading to strong relativistic beaming effects of the jet emission. Radio observations especially using very long baseline interferometry (VLBI) provide the best way to gain direct information on the intrinsic properties of jets down to sub-parsec scales, close to their formation region. In this thesis, I focus on the properties of three AGNs, IC 310, PKS 2004-447, and 3C 111 that belong to the small non-blazar population of gamma-ray-loud AGNs. In these kinds of AGNs, the jets are less strongly aligned with respect to the observer than in blazars. I study them in detail with a variety of radio astronomical instruments with respect to their high-energy emission and in the context of the large samples in the monitoring programmes MOJAVE and TANAMI. My analysis of radio interferometric observations and flux density monitoring data reveal very different characteristics of the jet emission in these sources. The work presented in this thesis illustrates the diversity of the radio properties of gamma-ray-loud AGNs that do not belong to the dominating class of blazars. Radiobeobachtungen vor allem mit Very Long Baseline Interferometry (VLBI) bieten die beste Möglichkeit direkt Informationen über die intrinsischen Eigenschaften von Jets in der Nähe ihrer Entstehungsregion auf sub-parsec Skalen zu sammeln. While many comparative genomics studies have focused on gene flux and large scale changes, relatively little attention has been paid to quantifying the effects of single nucleotide polymorphisms and indels on protein function, particularly in bacterial genomics. Results: We present a hidden Markov model based approach we call delta-bitscore (DBS) for identifying orthologous proteins that have diverged at the amino acid sequence level in a way that is likely to impact biological function. We benchmark this approach with several widely used datasets and apply it to a proof-of-concept study of orthologous proteomes in an investigation of host adaptation in Salmonella enterica. We highlight the value of the method in identifying functional divergence of genes, and suggest that this tool may be a better approach than the commonly used dN/dS metric for identifying functionally significant genetic changes occurring in recently diverged organisms. Genetically encoded biosensors based on Förster resonance energy transfer (FRET) are ideally suited for the study of such fast signaling events in single living cells. Here we report on the construction and characterization of three FRET biosensors for the measurement of Gα\(_{i1}\), Gα\(_{i2}\) and Gα\(_{i3}\) activation. To enable quantitative long-term imaging of FRET biosensors with high dynamic range, fluorescent proteins with enhanced photophysical properties are required. Therefore, we use the currently brightest and most photostable CFP variant, mTurquoise2, as donor fused to Gα\(_{i}\) subunit, and cp173Venus fused to the Gγ\(_{2}\) subunit as acceptor. The Gα\(_{i}\) FRET biosensors constructs are expressed together with Gβ\(_{1}\) from a single plasmid, providing preferred relative expression levels with reduced variation in mammalian cells. The Gα\(_{i}\) FRET sensors showed a robust response to activation of endogenous or over-expressed alpha-2A-adrenergic receptors, which was inhibited by pertussis toxin. Moreover, we observed activation of the Gα\(_{i}\) FRET sensor in single cells upon stimulation of several GPCRs, including the LPA\(_{2}\), M\(_{3}\) and BK\(_{2}\) receptor. Furthermore, we show that the sensors are well suited to extract kinetic parameters from fast measurements in the millisecond time range. We present a new experimental set-up that simultaneously measures the temperature rhythm in a queen-less but brood raising mini colony and the walking activity rhythms of singly kept honey bees that have indirect social contact with it. Our approach enables monitoring of individual bees in the social context of a mini colony under controlled laboratory conditions. In a pilot experiment, we show that social contact with the mini colony improves the survival of monitored young individuals and affects locomotor activity patterns of young and old bees. When exposed to conflicting Zeitgebers consisting of a light-dark (LD) cycle that is phase-delayed with respect to the mini colony rhythm, rhythms of young and old bees are socially synchronized with the mini colony rhythm, whereas isolated bees synchronize to the LD cycle. We conclude that the social environment is a stronger Zeitgeber than the LD cycle and that our new experimental set-up is well suited for studying the mechanisms of social entrainment in honey bees. Mouse models in which colitis-like symptoms are induced through challenge with toxins such as oxazolone, dextran sodium sulfate (DSS) or 2,4,6-trinitrobenzenesulfonic acid (TNBS) have been instrumental in understanding the inflammatory processes of UC. However, these neither reflect the heterogeneous symptoms observed in the UC-affected population nor can they be used to test the efficacy of inhibitors developed against human targets where high sequence and structural similarity of the respective ligands is lacking. In an attempt to overcome these problems, we have developed a mouse model that relies on NOD-scid IL2R γnull mice reconstituted with peripheral blood mononuclear cells derived from UC-affected individuals. Upon challenge with ethanol, mice developed colitis-like symptoms and changes in the colon architecture, characterized by influx of inflammatory cells, edema, crypt loss, crypt abscesses and epithelial hyperplasia, as previously observed in immune-competent mice. TARC, TGFβ1 and HGF expression increased in distal parts of the colon. Analysis of human leucocytes isolated from mouse spleen revealed an increase in frequencies of CD1a+, CD64+, CD163+ and TSLPR+ CD14+ monocytes, and antigen-experienced CD44+ CD4+ and CD8+ T-cells in response to ethanol. Analysis of human leucocytes from the colon of challenged mice identified CD14+ monocytes and CD11b+ monocytes as the predominant populations. Quantitative real-time PCR (RT-PCR) analysis from distal parts of the colon indicated that IFNγ might be one of the cytokines driving inflammation. Treatment with infliximab ameliorated symptoms and pathological manifestations, whereas pitrakinra had no therapeutic benefit. Thus, this model is partially reflective of the human disease and might help to increase the translation of animal and clinical studies. Such behavior can be assessed with the open field test (OFT), which is a well-established indicator of animal fear. The detection of similar open field behavior in humans may verify the translational validity of this paradigm. Enhanced thigmotaxis related to anxiety may suggest the relevance of such behavior for anxiety disorders, especially agoraphobia. METHODS: A global positioning system was used to analyze the behavior of 16 patients with agoraphobia and 18 healthy individuals with a risk for agoraphobia (i.e., high anxiety sensitivity) during a human OFT and compare it with appropriate control groups (n = 16 and n = 19). We also tracked 17 patients with agoraphobia and 17 control participants during a city walk that involved walking through an open market square. RESULTS: Our human OFT triggered thigmotaxis in participants; patients with agoraphobia and participants with high anxiety sensitivity exhibited enhanced thigmotaxis. This behavior was evident in increased movement lengths along the wall of the natural open field and fewer entries into the center of the field despite normal movement speed and length. Furthermore, participants avoided passing through the market square during the city walk, indicating again that thigmotaxis is related to agoraphobia. CONCLUSIONS: This study is the first to our knowledge to verify the translational validity of the OFT and to reveal that thigmotaxis, an evolutionarily adaptive behavior shown by most species, is related to agoraphobia, a pathologic fear of open spaces, and anxiety sensitivity, a risk factor for agoraphobia. Ein weniger arbiträres Vorgehen wäre es, experimentalpsychologisch zugrunde liegende Mechanismen zu identifizieren, die allen Arten der Perspektivenübernahme gemein sind, und zu untersuchen, ob eine Manipulation dieser Mechanismen abhängige Maße affektiver, kognitiver und perzeptueller Perspektivenübernahme gleichermaßen beeinflusst. Diesem Ansatz folgend macht die vorliegende Arbeit die Annahme, dass die mentale Selbstrotation des Körperschemas in die Position einer anderen Person, der zentrale Mechanismus visuo-spatialer Perspektivenübernahme, ein gemeinsamer Mechanismus aller Arten der Perspektivenübernahme ist. Entgegen früherer Ansätze wird diese Einheit somit nicht nur über die zentrale gemeinsame Funktionalität aller Arten von Perspektivenübernahme, also dem Verlassen des egozentrischen Referenzrahmens zugunsten einer (visuellen, affektiven oder kognitiven) Fremdperspektive, gerechtfertigt, sondern mit der Annahme eines gemeinsamen zugrundeliegenden Mechanismus. Daraus wird die einfache Hypothese abgeleitet, dass visuo-spatiale Perspektivenübernahme zu psychologischen Konsequenzen führen kann. Dies wurde in 6 Experimenten getestet. In diesen Experimenten mussten die Probanden zunächst immer die visuelle Perspektive einer anderen Person einnehmen. Hierzu sahen die Probanden eine Person, die mit zwei Objekten an einem Tisch sitzt. In jedem Durchgang mussten die Probanden sich entscheiden, mit welcher Hand diese Person eines der beiden Objekte greifen würde. Dabei wurde die Position der Zielperson so manipuliert, dass sie in der Hälfte der Fälle im selben visuo-spatialen Referenzrahmen wie der Proband saß, was Perspektivenübernahme zur Lösung der Aufgabe obsolet machte, während sie sich in den verbleibenden Durchgängen in einem anderen visuo-spatialen Referenzrahmen befand, so dass die Probanden die visuelle Perspektive der Zielperson übernehmen mussten um die Aufgabe korrekt zu lösen. Nach jedem Durchgang wurde dem Ziel dieser visuo-spatialen Aufgabe eine psychologische Eigenschaft zugeschrieben. Dies geschah im Rahmen eines abgewandelten Paradigmas zur Untersuchung der Ankerheuristik. Hierzu wurde den Probanden nach jedem Durchgang der visuo-spatialen Aufgabe eine Schätzfrage gestellt. Zeitgleich wurde die Antwort des Ziels bekannt gegeben. Entsprechend der Haupthypothese, dass visuo-spatiale Perspektivenübernahme psychologische Konsequenzen erzeugen kann, konnte gezeigt werden, dass die Probanden nach visuo-spatialer Perspektivenübernahme in höherem Maße die Gedanken der Zielperson übernahmen. Dies konnte sowohl anhand der absoluten Größe des Ankereffekts, als auch anhand der Differenz zwischen den Urteilen der Probanden und der Zielperson, gezeigt werden. Weitere Experimente schlossen Stichprobeneigenschaften, die verwendeten Stimuli oder die Aufgabenschwierigkeit als Alternativerklärungen für diese Effekte aus. Die beiden letzten Experimente zeigten zudem, dass dieser Effekt spezifisch für alle Konstellationen ist, in denen eine mentale Selbstrotation in die Zielperspektive notwendig war und dass die Übernahme fremder Gedanken mit einem Gefühl von Ähnlichkeit assoziiert war. Zusammengenommen unterstützen die Ergebnisse dieser Arbeit die theoretisch abgeleitete Sicht eines einheitlichen Perspektivenübernahme-Konstrukts und grenzen dieses zusätzlich von verwandten Konstrukten ab. In der abschließenden Diskussion werden die Bedeutung dieser Befunde für die Forschung in den Bereichen Empathie, Theory of Mind, und Perspektivenübernahme und ebenfalls praktische Implikationen der Ergebnisse aufgezeigt. N2 - „Perspective-taking“ is the ability to put yourself into the place of somebody else. Psychological research distinguishes three kinds of perspective-taking, namely, perceptual (visuo-spatial), affective (empathy), and cognitive (theory of mind) perspective-taking. The last two kinds of perspective-taking are often summarized as “psychological perspective-taking”. This dissertation tackles the question of whether these three kinds of perspective-taking should be conceptualized as independent constructs or as facets of one and the same construct. Prior research findings concerning this are equivocal. While some authors consider correlations between the different kinds of perspective-taking as too low for a unitary construct, others interpret correlations of the same magnitude as evidence for this. A less arbitrary way of deciding this would be to identify common mechanisms that underlie all kinds of perspective-taking and to examine whether manipulating these mechanisms in psychological experiments affects measures of perceptual, affective, and cognitive perspective-taking in parallel. In accordance with this reasoning, the present dissertation assumes that the mental self-rotation of the body schema into the physical location of another person, the main mechanism of perceptual perspective-taking, is a common mechanism of all kinds of perspective-taking. Thus, contrary to previous research a unitary construct is not only justified on the grounds of a common central functionality of all kinds of perspective-taking, that is, overcoming one’s egocentrism in favor of an alternative (perceptual, affective or cognitive) point of view, but additionally on the grounds of a common psychological mechanism. From this, the simple hypothesis that inducing visuo-spatial perspective-taking also leads to psychological consequences is derived. This hypothesis was tested in 6 experiments. In these experiments, participants first had to adopt the visual perspective of another person. To this end, they saw a person sitting at a table with two objects. During every trial, participants had to decide which hand the person would have to use in order to grab one of the two objects. Furthermore, the angular disparity between the participant and the target was manipulated in such a way that during half of the trials the target person was within the same visuo-spatial reference frame as the participant and thus no perspective-taking was necessary to solve the task correctly. During the remaining trials, the target person was sitting in another visuo-spatial reference frame so that the participants had to engage in perspective-taking to solve the task correctly. After every such trial, the target person was imbued with a mental state. This was done using an adapted paradigm for the investigation of the anchoring heuristic. Specifically, participants were asked to answer a trivia question and also saw what the target person from the visuo-spatial perspective-taking task was guessing. In line with the hypothesis that visuo-spatial perspective-taking leads to psychological outcomes, too, it was found that participants adopted the thoughts of the target person more strongly after visuo-spatial perspective-taking. This was evident in the absolute size of the anchoring effect, as well as the differences between participant and target estimations. Further experiments ruled out sample and stimulus characteristics and task difficulty as alternative explanations for these effects. The last two experiments furthermore established that the effects were specific to constellations where an embodied self-rotation into the target’s perspective was necessary and that the adoption of the target’s thoughts was associated with feelings of similarity. Taken together, these findings support the theoretically elaborated unitary view of perspective-taking and furthermore distinguish this construct from other related phenomena. In the general discussion, the significance of these findings for research on empathy, theory of mind, and perspective-taking, as well as practical implications are discussed. However, current skin equivalents lack a functional vasculature limiting clinical and research applications. This study demonstrates the generation of a vascularized skin equivalent with a perfused vascular network by combining a biological vascularized scaffold (BioVaSc) based on a decellularized segment of a porcine jejunum and a tailored bioreactor system. Briefly, the BioVaSc was seeded with human fibroblasts, keratinocytes, and human microvascular endothelial cells. After 14 days at the air-liquid interface, hematoxylin & eosin and immunohistological staining revealed a specific histological architecture representative of the human dermis and epidermis including a papillary-like architecture at the dermal-epidermal-junction. The formation of the skin barrier was measured non-destructively using impedance spectroscopy. Additionally, endothelial cells lined the walls of the formed vessels that could be perfused with a physiological volume flow. Due to the presence of a complex in-vivo-like vasculature, the here shown skin equivalent has the potential for skin grafting and represents a sophisticated in vitro model for dermatological research. The goal of the \textit{D. muscipula} genome project is to gain comprehensive insights into the genomic landscape of this remarkable plant. The genome of the diploid Venus flytrap with an estimated size between 2.6 Gbp to 3.0 Gbp is comparatively large and comprises more than 70 % of repetitive regions. Sequencing and assembly of genomes of this scale are even with state-of-the-art technology and software challenging. Initial sequencing and assembly of the genome was performed by the BGI (Beijing Genomics Institute) in 2011 resulting in a 3.7 Gbp draft assembly. I started my work with thorough assessment of the delivered assembly and data. My analysis showed that the BGI assembly is highly fragmented and at the same time artificially inflated due to overassembly of repetitive sequences. Furthermore, it only comprises about on third of the expected genes in full-length, rendering it inadequate for downstream analysis. In the following I sought to optimize the sequencing and assembly strategy to obtain an assembly of higher completeness and contiguity by improving data quality and assembly procedure and by developing tailored bioinformatics tools. Issues with technical biases and high levels of heterogeneity in the original data set were solved by sequencing additional short read libraries from high quality non-polymorphic DNA samples. To address contiguity and heterozygosity I examined numerous alternative assembly software packages and strategies and eventually identified ALLPATHS-LG as the most suited program for assembling the data at hand. Moreover, by utilizing digital normalization to reduce repetitive reads, I was able to substantially reduce computational demands while at the same time significantly increasing contiguity of the assembly. To improve repeat resolution and scaffolding, I started to explore the novel PacBio long read sequencing technology. Raw PacBio reads exhibit high error rates of 15 % impeding their use for assembly. To overcome this issue, I developed the PacBio hybrid correction pipeline proovread (Hackl et al., 2014). proovread uses high coverage Illumina read data in an iterative mapping-based consensus procedure to identify and remove errors present in raw PacBio reads. In terms of sensitivity and accuracy, proovread outperforms existing software. In contrast to other correction programs, which are incapable of handling data sets of the size of D. muscipula project, proovread’s flexible design allows for the efficient distribution of work load on high-performance computing clusters, thus enabling the correction of the Venus flytrap PacBio data set. Next to the assembly process itself, also the assessment of the large de novo draft assemblies, particularly with respect to coverage by available sequencing data, is difficult. While typical evaluation procedures rely on computationally extensive mapping approaches, I developed and implemented a set of tools that utilize k-mer coverage and derived values to efficiently compute coverage landscapes of large-scale assemblies and in addition allow for automated visualization of the of the obtained information in comprehensive plots. Using the developed tools to analyze preliminary assemblies and by combining my findings regarding optimizations of the assembly process, I was ultimately able to generate a high quality draft assembly for D. muscipula. I further refined the assembly by removal of redundant contigs resulting from separate assembly of heterozygous regions and additional scaffolding and gapclosing using corrected PacBio data. The final draft assembly comprises 86 × 10 3 scaffolds and has a total size of 1.45 Gbp. The difference to the estimated genomes size is well explained by collapsed repeats. At the same time, the assembly exhibits high fractions full-length gene models, corroborating the interpretation that the obtained draft assembly provides a complete and comprehensive reference for further exploration of the fascinating biology of the Venus flytrap. Das Ziel des D. muscipula Genomprojekts ist es, neue Einblicke in den genomischen Grundlagen dieser besonderen Pflanze zu gewinnen. Die diploide Venus Fliegenfalle verfügt mit eine geschätzten Größe von 2.6 bp bis 3Gbp über ein vergleichsweise großes Genom, das zudem zu über 70% aus repetitiven Regionen besteht. Sequenzierung und Assembly von Genomen dieser Größenordnung stellen selbst mit neusten technischen und informatischen Methoden eine große Herausforderung dar. Zum ersten mal sequenziert und assembliert wurde das Genom 2011 durch das BGI (Beijing Genomics Institute). Meine Arbeit am Genom der Fliegenfalle begann mit der Analyse des 3.7Gbp großen Assemblies, welches wir vom BGI erhalten haben. Dafür war es notwendig die Qualität der Sequenzierdaten so wie den Assemblierungsprozess selbst zu optimieren, und Programme zu entwickeln, die eine Verbesserung der Daten und eine Analyse der Zwischenergebnisse ermöglichen. So wurden etwa zur neue Bibliotheken von nicht-polymorphen DNA-Proben sequenziert um die Heterogenität im Datensatz zu verringern. Um die Kontinuität der Assemblies zu verbessern und Probleme mit der Heterozygosität der Daten zu lösen habe ich eine Reihe verschiedener Assemblierungsprogramme getestet. Dabei zeigte sich, dass das Programm ALLPATHS-LG am besten geeignet ist für die Assemblierung von D. muscipula Daten. Durch den Einsatz von digitaler Normalisierung konnte ich den Bedarf an Computerressourcen für einzelne Assemblierungen deutlich reduzieren und gleichzeitig die Kontinuität der Assemblies deutlich erhöhen. Zur besseren Auflösung repetitiver Strukturen im Genom, habe ich auf eine neu entwickelte Sequenziertechnologie von PacBio zurückgegriffen, die deutlich länger Sequenzen erzeugt. Um die neuen Daten trotz ihrer hohen Fehlerrate von 15% für Assemblierungen nutzen zu können, entwickelte ich das Korrekturprogramm proovread (Hackl et al., 2014). proovread nutzt kurze Illumina Sequenzen mit hoher Sequenziertiefe um innerhalb eines iterativen Prozess Fehler in PacBio Daten ausfindig zu machen und zu korrigieren. Das Programm erreicht dabei eine bessere Genauigkeit und eine höhere Sensitivität als vergleichbare Software. Darüber hinaus erlaubt sein flexibles Design auch Datensätze in der Größenordung des Fliegenfallengenoms effizient auf großen Rechenclustern zu bearbeiten. Neben dem Assemblierungsprozess an sich, stellt auch die Analyse von Assemblies großer Genome eine Herausforderung dar. Klassische Methoden basieren oft auf der rechenintensiven Berechnung von Alignments zwischen Sequenzierdaten und Assembly. Um vergleichbare Analysen deutlich schneller generieren zu können, habe ich Programme entwickelt die auf der Auswertung von k-mer Häufigkeiten beruhen, und die gewonnenen Ergebnisse in übersichtlichen Graphiken darstellen. Durch Kombination der so gewonnenen Einblicke und der verschiedenen Erkenntnisse bezüglich der Optimierung es Assemblierungsprozesses, war es mir am Ende möglich, ein Assembly von hoher Qualität für das Genom der Venus Fliegenfalle zu rekonstruieren. Dieses habe ich weiter verfeinert, unter anderem durch das Entfernen heterozygoter Sequenzen und durch das Flicken von Lücken mit Hilfe von PacBio Daten. Das so erstelle Assembly besteht aus 86 × 103 Sequenzen und hat eine Gesamtgröße von 1.45Gbp. Der Unterschied zur erwarteten Genomgröße lässt sich dabei gut durch kollabierte repetitive Regionen erklären. Methods and materials: Ten institutions with robotic(2), gimbaled(2), MLC(4) or couch tracking(2) used common materials including CT and structure sets, motion traces and planning protocols to create a lung and a prostate plan. For each motion trace, the plan was delivered twice to a moving dosimeter; with and without real-time adaptation. Each measurement was compared to a static measurement and the percentage of failed points for gamma-tests recorded. Results: For all lung traces all measurement sets show improved dose accuracy with a mean 2%/2 mm gamma-fail rate of 1.6% with adaptation and 15.2% without adaptation (p < 0.001). For all prostate the mean 2%/2 mm gamma-fail rate was 1.4% with adaptation and 17.3% without adaptation (p < 0.001). The difference between the four systems was small with an average 2%/2 mm gamma-fail rate of <3% for all systems with adaptation for lung and prostate. Conclusions: The investigated systems all accounted for realistic tumor motion accurately and performed to a similar high standard, with real-time adaptation significantly outperforming non-adaptive delivery methods. The subtleties in constructing a model which is in agreement with the measured quark masses and mixing for such a low left-right breaking scale are discussed. It is shown that in the constrained version of this model radiative breaking of the gauge symmetries is possible and a SM-like Higgs is obtained. Additional CP-even scalars of a similar mass or even much lighter are possible. The expected mass hierarchies for the supersymmetric states differ clearly from those of the constrained MSSM. In particular, the lightest down-type squark, which is a mixture of the sbottom and extra vector-like states, is always lighter than the stop. A randomized controlled trial design was administered. Twenty‐eight women, who had been treated for cancer were randomly assigned to either a group performing exercise of low‐to‐moderate intensity (LMIE; n = 14) or a group performing high‐intensity interval training (HIIT; n = 14) as part of a 3‐week multimodal rehabilitation program. No adverse events related to the exercise were reported. Work economy improved following both HIIT and LMIE, with improved peak oxygen uptake following LMIE. HIIT reduced mean total body fat mass with no change in body mass, muscle or fat‐free mass (best P < 0.06). LMIE increased muscle and total fat‐free body mass. Total energy expenditure (P = 0.45) did not change between the groups, whereas both improved quality of life to a similar high extent and lessened cancer‐related fatigue. This randomized controlled study demonstrates that HIIT can be performed by female cancer survivors without adverse health effects. Here, HIIT and LMIE both improved work economy, quality of life and cancer‐related fatigue, body composition or energy expenditure. Since the outcomes were similar, but HIIT takes less time, this may be a time‐efficient strategy for improving certain aspects of the health of female cancer survivors. This study investigated the feasibility of non-invasive CXCR4-directed imaging with positron emission tomography/computed tomography (PET/CT) using the radiolabelled chemokine receptor ligand \(^{68}\)Ga-Pentixafor. 15 patients with clinical suspicion on primary or recurrent glioblastoma (13 primary, 2 recurrent tumors) underwent \(^{68}\)Ga-Pentixafor-PET/CT for assessment of CXCR4 expression prior to surgery. O-(2-\(^{18}\)F-fluoroethyl)-L-tyrosine (\(^{18}\)F-FET) PET/CT images were available in 11/15 cases and were compared visually and semi-quantitatively (SUV\(_{max}\), SUV\(_{mean}\)). Tumor-to-background ratios (TBR) were calculated for both PET probes. \(^{68}\)Ga-Pentixafor-PET/CT results were also compared to histological CXCR4 expression on neuronavigated surgical samples. \(^{68}\)Ga-Pentixafor-PET/CT was visually positive in 13/15 cases with SUV\(_{mean}\) and SUV\(_{max}\) of 3.0±1.5 and 3.9±2.0 respectively. Respective values for \(^{18}\)F-FET were 4.4±2.0 (SUV\(_{mean}\)) and 5.3±2.3 (SUV\(_{max}\)). TBR for SUV\(_{mean}\) and SUV\(_{max}\) were higher for \(^{68}\)Ga-Pentixafor than for \(^{18}\)F-FET (SUV\(_{mean}\) 154.0±90.7 vs. 4.1±1.3; SUV\(_{max}\) 70.3±44.0 and 3.8±1.2, p<0.01), respectively. Histological analysis confirmed CXCR4 expression in tumor areas with high \(^{68}\)Ga-Pentixafor uptake; regions of the same tumor without apparent \(^{68}\)Ga-Pentixafor uptake showed no or low receptor expression. In this pilot study, \(^{68}\)Ga-Pentixafor retention has been observed in the vast majority of glioblastoma lesions and served as readout for non-invasive determination of CXCR4 expression. Given the paramount importance of the CXCR4/SDF-1 axis in tumor biology, \(^{68}\)Ga-Pentixafor-PET/CT might prove a useful tool for sensitive, non-invasive in-vivo quantification of CXCR4 as well as selection of patients who might benefit from CXCR4-directed therapy. Based on the recent literature, SST2 antagonists are superior to agonists in tumor uptake. The compound OPS201 is the novel somatostatin antagonist showing the highest SST2 affinity. The aim of this study was to measure the in vivo biodistribution and dosimetry of \(^{177}\)Lu-OPS201 in five anesthetized Danish Landrace pigs as an appropriate substitute for humans to quantitatively assess the absorbed doses for future clinical applications. Results \(^{177}\)Lu-OPS201 was obtained with a specific activity ranging from 10 to 17 MBq/μg. Prior to administration, the radiochemical purity was measured as s > 99.7 % in all cases. After injection, fast clearance of the compound from the blood stream was observed. Less than 5 % of the injected activity was presented in blood 10 min after injection. A series of SPECT/CT and whole-body scans conducted until 10 days after intravenous injection showed uptake mostly in the liver, spine, and kidneys. There was no visible uptake in the spleen. Blood samples were taken to determine the time-activity curve in the blood. Time-activity curves and time-integrated activity coefficients were calculated for the organs showing visible uptake. Based on these data, the absorbed organ dose coefficients for a 70-kg patient were calculated with OLINDA/EXM. For humans after an injection of 5 GBq \(^{177}\)Lu-OPS201, the highest predicted absorbed doses are obtained for the kidneys (13.7 Gy), the osteogenic cells (3.9 Gy), the urinary bladder wall (1.8 Gy), and the liver (1.0 Gy). No metabolites of 177Lu-OPS201 were found by radio HPLC analysis. None of the absorbed doses calculated will exceed organ toxicity levels. Conclusions The \(^{177}\)Lu-OPS201 was well tolerated and caused no abnormal physiological or behavioral signs. In vivo distributions and absorbed doses of pigs are comparable to those observed in other publications. According to the biodistribution data in pigs, presented in this work, the expected radiation exposure in humans will be within the acceptable range. A minor region is found in the centromeric region of the free autosome 3 after denaturing the chromosomal DNA for 3 h or longer. Its exact physical nature deeply influences the properties of the supercurrent. In recent years, considerable efforts have focused on the coupling of superconductors to the surface states of a three-dimensional topological insulator. In such a material, an unconventional induced p-wave superconductivity should occur, with a doublet of topologically protected gapless Andreev bound states, whose energies vary 4π-periodically with the superconducting phase difference across the junction. In this article, we report the observation of an anomalous response to rf irradiation in a Josephson junction made of a HgTe weak link. The response is understood as due to a 4π-periodic contribution to the supercurrent, and its amplitude is compatible with the expected contribution of a gapless Andreev doublet. Our work opens the way to more elaborate experiments to investigate the induced superconductivity in a three-dimensional insulator. In this series, the image quality and value of intraoperative 3D fluoroscopy with intravenous contrast agent for the evaluation of aneurysm occlusion and vessel patency after clip placement was assessed in patients who underwent surgery for intracranial aneurysms. Materials and methods Twelve patients were included in this retrospective analysis. Prior to surgery, a 360° rotational fluoroscopy scan was performed without contrast agent followed by another scan with 50 ml of intravenous iodine contrast agent. The image files of both scans were transferred to an Apple PowerMac® workstation, subtracted and reconstructed using OsiriX® free software. The procedure was repeated after clip placement. Both image sets were compared for assessment of aneurysm occlusion and vessel patency. Results Image acquisition and contrast administration caused no adverse effects. Image quality was sufficient to follow the patency of the vessels distal to the clip. Metal artifacts reduce the assessability of the immediate vicinity of the clip. Precise image subtraction and post-processing can reduce metal artifacts and make the clip-site assessable and depict larger neck-remnants. Conclusion This technique quickly supplies images at adequate quality to evaluate distal vessel patency after aneurysm clipping. Significant aneurysm remnants may be depicted as well. As it does not require visual control of all vessels that are supposed to be evaluated intraoperatively, this technique may be complementary to other intraoperative tools like indocyanine green videoangiography and micro-Doppler, especially for the assessment of larger aneurysms. At the momentary state of this technology, it cannot replace postoperative conventional angiography. However, 3D fluoroscopy and image post-processing are young technologies. Further technical developments are likely to result in improved image quality. Der Großteil dieser Modellstrukturen wurde anschließend hinsichtlich ihrer Hydrolysekinetik in Abhängigkeit der Art der funktionellen Gruppe X (NMe3+, N(H)COOMe, N(Me)COOMe, NH2, N(H)Me, NMe2, Pip, Me), des Abstandes des Substituenten X zu dem Silicium-Atom (α-, β-, γ- und ζ-Position), der Alkoxy-Abgangsgruppe am Silicium-Atom (MeO, iPrO, tBuO) und des pD-Wertes der Reaktionslösung systematisch untersucht. Eine große Herausforderung dieser Studie war die Synthese von β-Amino-funktionalisierten Alkoxysilanen, deren Chemie aufgrund ihrer Labilität bisher kaum erforscht ist. Die einzigen literaturbekannten Vertreter stellten bislang das Trialkoxysilan (EtO)3Si(CH2)2NH2 (1) und sein Dialkoxy-Derivat (EtO)2SiMe(CH2)2NH2 (2) dar, welche durch Reaktion des entsprechenden 2-(Chlorethyl)silans mit Ammoniak unter hohem Druck im Autoklaven zugänglich sind. Unter Verwendung dieser Synthesemethode konnte sowohl die Synthese der Silane 1 und 2 reproduziert, als auch das Trimethoxy-Analogon (MeO)3Si(CH2)2NH2 (3) erstmals dargestellt werden. Darüber hinaus wurde eine Serie von organofunktionellen Monoalkoxysilanen des Typs RORSiMe(CH2)2X und ROSiMe2C(H)MeCH2X (4b–18b) im präparativen Maßstab analyserein dargestellt. Des Weiteren wurden die entsprechenden α-Silane 8a, 11a, 14a und 15a, die γ-Silane 6c, 8c, 11c, 13c–15c und 18c sowie die ζ-Silane 19 und 20 erstmals dargestellt. Weiterhin wurden die bereits literaturbekannten α-Silane 16a–18a und γ-Silane 7c, 16c und 17c für die Verwendung in den Hydrolyseexperimenten synthetisiert. Die Charakterisierung aller im Rahmen dieser Arbeit synthetisierten Verbindungen erfolgte mittels NMR-Spektroskopie (1H-, 13C-, 15N- und 29Si-NMR) und Elementaranalysen (C, H, N) bzw. HRMS-Experimente. Die hydrolytische Spaltung der Si–OC-Bindung in Alkoxy(aminoalkyl)silanen stellt einen technisch sehr wichtigen Schlüsselschritt in der Synthese von Amino-funktionalisierten Polysiloxanen dar. Um den Mechanismus dieser Si–OC-Bindungsspaltung besser zu verstehen, wurden die Alkoxysilane 4b, 4c, 5b, 6b, 6c, 7b, 7c, 8a–8c, 9b, 11a–11c, 12b, 14a–14c, 15a–15c, 16a–16c, 17a–17c, 18a–18c, 19 und 20 hinsichtlich ihrer Hydrolysekinetik in CD3CN/D2O unter sauren und basischen Bedingungen mittels 1H-NMR-Spektroskopie untersucht. Die Ergebnisse dieser Struktur–Reaktivitäts-Studie zeigten, dass die beobachteten unterschiedlichen Hydrolysegeschwindigkeiten das Resultat mehrerer Faktoren sind, wie beispielsweise elektronische und sterische Effekte, der große Einflusses des pD-Wertes und auch intramolekulare N–H∙∙∙O-Wasserstoffbrückenbindungen zwischen der protonierten Amino-Gruppe und der Alkoxy-Abgangsgruppe. Da der Einfluss dieser Effekte auf die Reaktivität der untersuchten α-, β-, γ- und ζ-Silane sehr unterschiedlich ist, kann kein klarer Zusammenhang zwischen der Hydrolysereaktivität und der Positionierung der stickstoff-haltigen funktionellen Gruppe (α-, β-, γ- und ζ-Position) erkannt werden. Die jeweils beobachtete Reaktivität entspricht vielmehr einer Summe aller zuvor genannten Teileffekte. N2 - This thesis deals with the synthesis and properties of a series of organofunctional α-, β-, γ-, and ζ-silanes, with a focus on alkoxy(aminoalky)silanes. The majority of these model compounds were systematically investigated for their hydrolysis kinetics, depending on the functional group X (NMe3+, N(H)COOMe, N(Me)COOMe, NH2, N(H)Me, NMe2, Pip, Me), the spacer between X and the silicon atom (α-, β-, γ-, and ζ-position of the functional group), the alkoxy leaving group at the silicon atom (MeO, iPrO, tBuO), and the pD value of the reaction mixture. One of the major challenges of this study was the synthesis of β-amino-functionalized alkoxysilanes, the chemistry of which was mainly due to stability issues not well established yet. Until now, the trialkoxysilane (EtO)3Si(CH2)2NH2 (1) and its dialkoxyderivative (EtO)2SiMe(CH2)2NH2 (2) were the only representatives of this type described in the literature. They were synthesized by reaction of the respective 2-(chloroethyl)silane and ammonia under high pressure in an autoclave. Using this synthetic method, the synthesis of the silanes 1 und 2 could be reproduced and the trimethoxyanalogue (MeO)3Si(CH2)2NH2 (3) could be prepared for the first time. Furthermore, a series of organofunctional monoalkoxysilanes of the formula type RORSiMe(CH2)2X and ROSiMe2C(H)MeCH2X (4b–18b) was synthesized on a preparative scale in analytically pure form. Also, the corresponding α-silanes 8a, 11a, 14a, and 15a, the γ-silanes 6c, 8c, 11c, 13c–15c, and 18c, and the ζ-silanes 19 and 20 were prepared for the first time. In addition, the well known α-silanes 16a–18a and γ-silanes 7c, 16c, and 17c were synthesized for their use in the hydrolysis experiments. All the compounds synthesized in this study were characterized by NMR spectroscopy (1H, 13C, 15N, and 29Si NMR) and elemental analysis (C, H, N) or HRMS experiments. The hydrolytic cleavage of the Si–OC bond of alkoxy(aminoalkyl)silanes represents a technically very important key step in the synthesis of amino-functionalized polysiloxanes. To get a better understanding of the mechanism of this Si–OC bond cleavage, the alkoxysilanes 4b, 4c, 5b, 6b, 6c, 7b, 7c, 8a–8c, 9b, 11a–11c, 12b, 14a–14c, 15a–15c, 16a–16c, 17a–17c, 18a–18c, 19, and 20 were studied for their hydrolysis kinetics in CD3CN/D2O under acidic and basic conditions, using 1H NMR spectroscopy as the analytical tool. The results of these structure–reactivity studies clearly demonstrate that the different hydrolysis reactivities observed are the result of a number of parameters, such as electronic and steric effects, the strong impact of the pD value, and intramolecular N–H∙∙∙O hydrogen bonds between the protonated amino group and the alkoxy leaving group. These parameters affect the hydrolysis reactivity of the α-, β-, γ-, and ζ-silanes in an unpredictable manner, so that no clear correlation between the hydrolysis reactivity and the position of the nitrogen-containing functional group (α-, β-, γ-, and ζ-position) can be found. The observed reactivity is rather a summation of all the aformentioned parameters. The insight gained from this work allows for a much clearer conceptual understanding of the reactivity of functionalized α-, β-, γ-, and ζ-silanes. These findings are also of great interest for silicone industry as they allow for a more directed application of α-, β-, and γ-aminosilanes for the preparation of the technically important class of amino-functionalized polysiloxanes. We try to fill this gap by adapting Lijphart's typological framework to the Latin American context in the following way. In contrast to previous studies, we treat the type of democracy as an independent variable and include informal factors such as clientelism or informal employment in our assessment of democratic patterns. On this basis, we aim to answer the following questions. First, how did the patterns of democracy evolve in Latin America over the two decades between 1990 and 2010 and what kind of differences can be observed in the region? Second, what are the institutional determinants of the observed changes? We focus on the emergence of new parties because of their strong impact on the first dimension of Lijphart's typology. From our observations we draw the following tentative conclusions: If strong new parties established themselves in the party system but failed to gain the presidency, they pushed the system towards consensualism. Conversely, new parties that gained the presidency produced more majoritarian traits. Probably originating in the 2nd century BCE1, the book tells a legend of how the translation of the Torah into Greek came into being. This shows that translating a holy, canonical text or the first time needed explication. Notably, the translation of the godly nomos (Arist 3) comparatively takes up little space (Arist 301–307). And it has to be noted, that “God” is seldom a topic in the Book of Aristeas. The word (ὁ) θεός “God” is found in only three contexts: in the dialogue between king Ptolemaios and Aristeas (Arist 15–21), in the dialogue of the high priest Eleazar and Aristeas (Arist 121–171; above all 128; 130–141; 155–166; 168) and in the question-and-answer-speech during the symposium at the Ptolemaic royal court between the king and the Jewish scholars (Arist 184–294). In analysing the different statements regarding God, the frame of the narrative is of decisive importance: In the Book of Aristeas, “Aristeas” (Ἀριστέας), who writes in Greek, presents himself as the author, but he is also part of the story. Accordingly, Aristeas is the narrator, who tells the story from his own point of view, and at the same time, he is a character in the ‘world’ of the text. This Aristeas presents himself as a Greek and a Non-Jew (Arist 16; 121–171), who already wrote a book (Arist 6) and plans further publications (Arist 322). In the double-role as narrator of the text and protagonist in the text, Aristeas has to be differentiated from the (real) writer/author of the Book of Aristeas, who possibly was Jewish. That means that the (real, probably Jewish) author of the Book of Aristeas presents (or invents) “Aristeas” and gives him the role of the narrator of his text.3 The author portrays Aristeas as a Greek, non-Jewish character, who is a servant of the royal court. This differentiation between narrator and writer/author is of crucial importance for the question of the different conceptions of God in the Book of Aristeas.
