N2  - Die Behandlung der Ovarial- und Endometriumkarzinomzellen mit Perifosin ruft eine Inhibition der AKT-Phosphorylierung und eine damit verbundene Hemmung der Zellproliferation hervor. In PTEN-defizienten Zellen, die mutmaßlich eine stärkere konstitutive AKT-Aktivität aufweisen, waren die zytostatischen Effekte von Perifosin stärker ausgeprägt. In allen Zelllinien wurde gezeigt, dass der Zelltod unabhängig von einer Inhibition der Caspasen erfolgen kann. Die Kombinationsbehandlung von Endometrium- und Ovarialkarzinomzelllinien mit Perifosin und Cisplatin demonstrierte additive Effekte. Daher scheint Perifosin ein guter Kandidat für eine Phase II Studie bei Patientinnen mit fortgeschrittenem Endometrium- und Ovarialkarzinomen zu sein und zwar sowohl als Monopräparat als auch in Kombination mit Platin-Derivaten.
N2  - The treatment Ovarial-and Endometriumkarzinomzellen with Perifosin causes an Inhibition of the AKT-Phosphorylierung and an inhibition linked with it of the cell proliferation. In PTEN-defizienten the cells which show presumably a stronger constitutive act activity the zytostatischen effects were stronger coined by Perifosin. In all cell lines it was shown that the cell death can occur regardless of an Inhibition of the Caspasen. The combination treatment from Endometrium-and Ovarialcarcinomcelllines with Perifosin and Cisplatin demonstrated additive effects. Thus, perifosine appears to be a good candidate for phase II studies in patients with advanced endometrial cancers, both as single agent therapy or in combination with platinum derivatives.
