These droughts affected forest ecosystems. Cascading effects with bark beetle infestations in spruce stands were fatal to vast forest areas in Germany. We present the first assessment of canopy cover loss in Germany for the period of January 2018–April 2021. Our approach makes use of dense Sentinel-2 and Landsat-8 time-series data. We computed the disturbance index (DI) from the tasseled cap components brightness, greenness, and wetness. Using quantiles, we generated monthly DI composites and calculated anomalies in a reference period (2017). From the resulting map, we calculated the canopy cover loss statistics for administrative entities. Our results show a canopy cover loss of 501,000 ha for Germany, with large regional differences. The losses were largest in central Germany and reached up to two-thirds of coniferous forest loss in some districts. Our map has high spatial (10 m) and temporal (monthly) resolution and can be updated at any time. The qualitative visual analysis of digital subtraction angiography (DSA) is dependent on observer experience and prone to interpretation errors. We evaluate the feasibility of 2D-Perfusion Angiography (2D-PA) for objective, quantitative treatment response assessment in CMI. Methods 49 revascularizations in 39 patients with imaging based evidence of mesenteric vascular occlusive disease and clinical signs of CMI were included in this retrospective study. To assess perfusion changes by 2D-PA, DSA-series were post-processed using a dedicated, commercially available software. Regions of interest (ROI) were placed in the pre- and post-stenotic artery segment. In aorto-ostial disease, the inflow ROI was positioned at the mesenteric artery orifice. The ratios outflow to inflow ROI for peak density (PD), time to peak and area-under-the-curve (AUC) were computed and compared pre- and post-interventionally. We graded motion artifacts by means of a four-point scale. Feasibility of 2D-PA and changes of flow parameters were evaluated. Results Motion artifacts due to a mobile vessel location beneath the diaphragm or within the mesenteric root, branch vessel superimposition and inadequate contrast enhancement at the inflow ROI during manually conducted DSA-series via selective catheters owing to steep vessel angulation, necessitated exclusion of 26 measurements from quantitative flow evaluation. The feasibility rate was 47%. In 23 technically feasible assessments, PD\(_{outflow}\)/PD\(_{inflow}\) increased by 65% (p < 0.001) and AUC\(_{outflow}\)/AUC\(_{inflow}\) increased by 85% (p < 0.001). The time to peak density values in the outflow ROI accelerated only minimally without reaching statistical significance. Age, BMI, target vessel (celiac trunk, SMA or IMA), stenosis location (ostial or truncal), calcification severity, plaque composition or the presence of a complex stenosis did not reach statistical significance in their distribution among the feasible and non-feasible group (p > 0.05). Conclusions Compared to other vascular territories and indications, the feasibility of 2D-PA in mesenteric revascularization for CMI was limited. Unfavorable anatomic conditions contributed to a high rate of inconclusive 2D-PA results. In contrast to the well-developed light-gated guanylyl/adenylyl cyclases as optogenetic tools, ideal light-regulated phosphodiesterases are still in demand. Here, we investigated and engineered the RhoPDEs from Salpingoeca rosetta, Choanoeca flexa and three other protists. All the RhoPDEs (fused with a cytosolic N-terminal YFP tag) can be expressed in Xenopus oocytes, except the AsRhoPDE that lacks the retinal-binding lysine residue in the last (8th) transmembrane helix. An N296K mutation of YFP::AsRhoPDE enabled its expression in oocytes, but this mutant still has no cGMP hydrolysis activity. Among the RhoPDEs tested, SrRhoPDE, CfRhoPDE1, 4 and MrRhoPDE exhibited light-enhanced cGMP hydrolysis activity. Engineering SrRhoPDE, we obtained two single point mutants, L623F and E657Q, in the C-terminal catalytic domain, which showed ~40 times decreased cGMP hydrolysis activity without affecting the light activation ratio. The molecular characterization and modification will aid in developing ideal light-regulated phosphodiesterase tools in the future. Declarative and procedural metacognitive competences were assessed in the domain of reading comprehension. Path analyses showed incremental effects of procedural metacognition over and above intelligence on the development of school achievement in gifted students (β = .139). Moreover, declarative metacognition and need for cognition interactively predicted procedural metacognition (β = .169), which mediated their effect on school achievement. It has been discussed as a defect center for a nanographene by Kuck and colleagues. Therefore, extended TBTQ structures are promising models for saturated defect structures in graphene and graphene like molecules and could be used to investigate the role of defects for the electronic properties of graphene. With this motivation, three different pi-extended TBTQ derivatives have been synthesized in this work. Several different Scholl reaction conditions were tried to obtain fully annulated product of hexaphenyl substituted TBTQ. The desired benzannulated TBTQ derivative could not be obtained due to unfavourable electron density in the respective positions of the molecule and increased reactivity of the bay position of the precursor. As an another method for benzannulation is the on-surface synthesis of graphene flakes and can be carried out using electron beams e.g. in a tunneling microscope (STM). According to our previous research, the parent system TBTQ and centro-methyl TBTQ on silver and gold surfaces showed that the gas phase deposition of these molecules gives rise to the formation of highly ordered two-dimensional assemblies with unique structural features. This shows the feasibility for the formation of defective graphene networks starting from the parent structures. Therefore, the same deposition technique was used to deposit Me-TBTQ(OAc)3Ph6, and investigate the molecular self-assembly properties directly on the surface of Cu (111). In summary, the substrate temperature dependent self-assembly of Me-TBTQ(OAc)3Ph6 molecules on Cu(111), shows the following evolution of orientations. At room temperature, molecules form dimers, which construct a higher-coverage honeycomb lattice. Furthermore, one of the acetyl group located in the bay positions of the TBTQ core is cleaved and the remaining two induce the metal-molecule interaction. It was presumed that by increasing the temperature to 393 K, the remaining acetyl and methyl groups would beeliminated from the molecular structure.In addition, the smaller TBTQ-Ph6 molecules preferably lie flat on Cu(111) crystal and allowing the molecules to settle into a C3-symmetry and form a dense hexagonal structure. Mit dieser Motivation wurden in dieser Arbeit drei verschiedene TBTQ-Derivate mit erweitertem -System synthetisiert. Um im Anschluss eine vollständige Annellierung und damit Konjugation des p-Systems zu erreichen, wurden verschiedene Scholl-Reaktionen getestet, um das dreifach anellierte Produkt aus hexaphenylsubstituiertem TBTQ zu erhalten. Das gewünschte benzannulierte TBTQ-Derivat konnte aufgrund der relativ geringen Elektronendichte an den Annellierungspositionen und der erhöhten Reaktivität der Bay-Positionen des Moleküls nicht erhalten werden. Eine weitere Möglichkeit zur Annellierung von Nanographenen besteht in der On-Surface-Synthese. Diese kann mit Elektronenstrahlen z.B. in einem Rastertunnelmikroskop (STM) durchgeführt werden. Nach unseren früheren Untersuchungen, hochauflösende STM-Experimente mit dem Stammsystem TBTQ und centro-methyl TBTQ auf Silber- und Goldoberflächen, dass die Gasphasenabscheidung dieser Moleküle zur Bildung hochgeordneter zweidimensionaler Assemblierte mit einzigartigen strukturellen Eigenschaften führt. Dies zeigt die Möglichkeit zur Bildung von defekthaltigen Graphen-Netzwerken ausgehend von den Stammsystemen. Daher wurde hier die gleiche Abscheidungstechnik verwendet, um für Me-TBTQ(OAc)3Ph6 die molekulare Selbstanordnung auf der Oberfläche von Cu(111) untersuchen. Zusammenfassend zeigt die Selbstorganisation von Me-TBTQ(OAc)3Ph6 auf Cu(111) eine ausgeprägte Temperaturabhängigkeit. Die Moleküle bilden bei 363 K ein höheres Wabengitter aus. SGs form by multivalent interactions between RNAs and RNA-binding proteins and are believed to protect stalled translation initiation complexes from stress-induced degradation. SGs contain hundreds of different mRNAs and proteins, and their assembly and disassembly are tightly controlled by post-translational modifications. The ubiquitin system, which mediates the covalent modification of target proteins with the small protein ubiquitin (‘ubiquitylation’), has been implicated in different aspects of SG metabolism, but specific functions in SG turnover have only recently emerged. Here, we summarize the evidence for the presence of ubiquitylated proteins at SGs, review the functions of different components of the ubiquitin system in SG formation and clearance, and discuss the link between perturbed SG clearance and the pathogenesis of neurodegenerative disorders. We conclude that the ubiquitin system plays an important, medically relevant role in SG biology. Fabry disease (FD) is a lipid disorder disease that leads to accumulations of globotriaosylceramide in different organs, including the heart, which is particularly critical for the patient’s prognosis. Effective treatment options are available if initiated at early disease stages, but many patients are late- or under-diagnosed. Since Coherent anti-Stokes Raman (CARS) imaging has a high sensitivity for lipid/protein shifts, we applied CARS as a diagnostic tool to assess cardiac FD manifestation in an FD mouse model. CARS measurements combined with multivariate data analysis, including image preprocessing followed by image clustering and data-driven modeling, allowed for differentiation between FD and control groups. Indeed, CARS identified shifts of lipid/protein content between the two groups in cardiac tissue visually and by subsequent automated bioinformatic discrimination with a mean sensitivity of 90–96%. Of note, this genotype differentiation was successful at a very early time point during disease development when only kidneys are visibly affected by globotriaosylceramide depositions. Altogether, the sensitivity of CARS combined with multivariate analysis allows reliable diagnostic support of early FD organ manifestation and may thus improve diagnosis, prognosis, and possibly therapeutic monitoring of FD. A previously published questionnaire used in a study published in 2010 was adapted and expanded. The questionnaire was distributed to an international group of experienced CI surgeons. Present results were compared, via descriptive statistics, to those from the 2010 survey. Eighteen surgeons completed the questionnaire. Respondents clearly favored the following: round window insertion, slow array insertion, and the peri- and postoperative use of systematic antibiotics. Insertion depth was regarded as important, and electrode arrays less likely to induce trauma were preferred. The usefulness of dedicated soft-surgery training was also recognized. A lack of agreement was found on whether the middle ear cavity should be flushed with a non-aminoglycoside antibiotic solution or whether a sheath or insertion tube should be used to avoid contaminating the array with blood or bone dust. In conclusion, this paper demonstrates how beliefs about CI soft surgery have changed since 2010 and shows areas of current consensus and disagreement. This study analyzes the mutual influence of (paleo-)relief development and medieval building activity using a geomorphological and geoarchaeological multimethod approach to expand the knowledge of human-environmental interactions during this time. For this purpose, GIS-based terrain analysis and geophysical measurements were conducted and combined with sedimentological information to create 1D-3D models of the subsurface and to assess knowledge of the landscape and relief evolution at various medieval castle and mining sites. The interpretation of all these data led to the answering of numerous site-specific questions on various geomorphological, geoarchaeological, geologic, and archaeological topics that have been explored in this work and have greatly increased our knowledge of each study site. In addition to these key contributions to the archaeological and geomorphological interpretation of individual study sites, a quantification of the anthropogenic influence on the relief development was conducted, a generalized model of the influence was derived, and new methodological and interpretative approaches were developed. Overall, this study links geomorphological/geological and (geo-)archaeological investigations at five medieval sites and delivers important information on human-environmental interactions within the Spessart and beyond. N2 - Mostly taking place in the Spessart, a low mountain range in central Germany, a feud between the counts of Rieneck and the bishops in Mainz escalated during the medieval period. This feud led to many castles being built at both sites, as well as other building activities. Many of these buildings and castles were located at specific and unique relief positions. This study analyzes the mutual influence of (paleo-)relief development and medieval building activity by utilizing a multi-method approach. For this purpose, GIS-based terrain analyses and geophysical measurements were conducted and combined with sedimentological information from invasive methods and outcrops to assess information on the topographic relief at four medieval castle sites and a medieval mining area. From the recent relief, the influence of medieval building activity was revealed and the paleo-relief was derived. The study sites were chosen in conjunction with the Archaeology Project of the Spessart Region (Archäologisches Spessartprojekt), which performed or is planning to perform excavations at these sites. With the help of GIS-based terrain analyses, the main objective was split into a number of site-specific questions. Spatial data were collected with the help of electrical resistivity tomography, ground-penetrating radar, seismic refraction tomography and fluxgate magnetic surveying to answer these questions. The ambiguous, spatial geophysical results were proven using Pürckhauer and Edelman soundings and with the help of boreholes and outcrops. More precise information about the selective soundings was extrapolated using geophysical data. Additionally, with these established methods, a new approach for the description of the relative resistivity distribution within the subsurface was developed. In addition to a quantitative description of the data, additional parameters were derived with the help of a constructed ArcGIS-tool. By interpreting all these data, most of the site-specific questions were answered. These questions are of variable complexity and focus on different geomorphological, geological, geoarchaeological and archaeological topics. For example, the genesis of the rough topography surrounding Wahlmich Castle near Waldaschaff was deciphered. Additionally, mainly boreholes and AMS-14C-datings revealed a connection between the medieval usage of a castle with a mill, including mill ponds, and the sedimentation history in a talweg at Hauenstein Castle. With the help of a geophysical prospection, the location of Mömbris Castle was precisely predicted, which resulted in a targeted excavation off-target the initially suspected areas. At Rannenburg Castle near Alzenau, massive, anthropogenically used loess loam deposits were prospected. Furthermore, at Eisenberg near Steinau an der Straße, important information was derived based on the position and form of iron deposits and collapsed shafts. In addition to all these key contributions for the archaeological and/or geomorphological interpretation of individual study sites, quantification of the anthropogenic influence on the relief was conducted. A generalized model that focuses on the mutual influence of relief development and medieval building activity was derived. Furthermore, new research approaches were developed that focus not only on positive location factors for a specific use, but also on the negative location factors for another type of use. The newly developed approach for describing the relative resistivity distribution can also be transferred to other studies to increase objectivity. Moreover, it is possible to create generalized datasets for individual study sites. Thus, this study links geomorphological/geological and (geo-)archaeological investigations at a medieval site and delivers important information on the mutual influence between the research items of all these disciplines within the Spessart and beyond. N2 - Nachdem im Mittelalter eine Fehde zwischen den Grafen von Rieneck und den Mainzer Bischöfen eskalierte, wurden im Spessart, einem deutschen Mittelgebirge, häufig in sehr spezifischen und ungewöhnlichen Reliefpositionen von beiden Streitparteien diverse Burgen errichtet sowie andere Bautätigkeiten vorgenommen. Die vorliegende Arbeit widmet sich dieser Landschaft, indem sie den wechselseitigen Einfluss von (Paläo-)Reliefentwicklung und mittelalterlicher Bauaktivität mit Hilfe eines Multimethodenansatzes analysiert. Im Rahmen dieser Arbeit wurden an vier mittelalterlichen Burgenstandorten und einem mittelalterlichen Bergbaugebiet GIS-basierte Reliefanalysen und geophysikalische Messungen durchgeführt sowie sedimentologische Informationen aus invasiven Methoden und Aufschlüssen aufgenommen, um das rezente Relief zu erfassen, den Einfluss der mittelalterlichen Bauaktivität darzustellen und das Paläorelief abzuleiten. Die Untersuchungsgebiete wurden mithilfe des Archäologischen Spessartprojektes ausgesucht, welches an den jeweiligen Standorten Ausgrabungen durchgeführt hat oder plant. Mit Hilfe einer GIS-basierten Reliefanalyse wurde die Hauptfragestellung in eine Anzahl standortspezifischer Fragestellungen untergliedert. Für die Beantwortung dieser Fragestellungen wurden mit geoelektrischer Widerstandstomographie, Bodenradar, Refraktionsseismik und Fluxgatemagnetik geophysikalische Daten erhoben. Die mehrdeutigen aber flächenhaften geophysikalischen Daten wurden mit punktuellen Pürckhauer-, Edelman- und Rammkernsondagen genauer definiert; diese präziseren Informationen der punktuellen Sondagen konnten wiederum mit Hilfe der geophysikalischen Daten extrapoliert werden. Zusätzlich zu diesen etablierten Methoden wurde ein neuer Ansatz zur Beschreibung von der relativen elektrischen Widerstandsverteilung im Untergrund entwickelt und neben einer quantitativen Beschreibung der Daten auch zusätzliche Parameter mit Hilfe eines konstruierten ArcGIS-Tools abgeleitet. Durch die übergreifende Interpretation der Daten konnten die aufgeworfenen standortspezifischen Fragestellungen beantwortet werden. Nachdem diese Fragestellungen in ihrer Komplexität variierten und zudem verschiedene geomorphologische, geologische, geoarchäologische und archäologische Themen fokussierten, konnte eine große Bandbreite an Befunden generiert werden. So konnte beispielsweise die Genese einer markanten und rauen Topographie in der Umgebung des Burgstalls Wahlmichs in Waldaschaff entschlüsselt werden. Die Sedimentation in einer Bachaue bei Hauenstein konnte mit Hilfe von Bohrungen und 14C-Datierungen klar zu der mittelalterlichen Nutzung eines Burgstalls mit Mühle und entsprechenden Mühlteichen zugeordnet werden. In Mömbris konnte die Lage der eigentlichen Burganlage mit Hilfe einer geophysikalischen Prospektion präzisiert werden, was zu einer gezielten Ausgrabung außerhalb der eigentlichen Verdachtsflächen geführt hat. An der Rannenburg in der Nähe von Alzenau konnten mächtige, anthropogen genutzte Lössauflagen prospektiert werden und am Eisenberg in der Nähe von Steinau an der Straße konnten wichtige Informationen zur Lage und Form von Erzlagerstätten und Pingen gewonnen werden. Neben all diesen wichtigen Beiträgen zur archäologischen und geomorphologischen Interpretation der einzelnen Standorte konnte der anthropogene Einfluss an jedem Standort quantifiziert werden und ein generalisiertes Modell zur wechselseitigen Einflussnahme von Reliefentwicklung und mittelalterlicher Bebauung abgeleitet werden. Des Weiteren konnten neue Forschungsansätze entwickelt werden, welche nicht nur die Standortvorteile für bestimmte Nutzungen einzelner Standorte fokussieren, sondern auch die Standortnachteile für andere Nutzungsformen betrachten. Außerdem kann der neu entwickelte Ansatz zur Beschreibung der relativen Widerstandsverteilung im Untergrund auch auf zukünftige Fragestellungen übertragen werden, um die Objektivität bei der Betrachtung von verschiedenen Datensätzen zu erhöhen und gegebenenfalls auch generalisierte Datensätze für einzelne Standorte zu erstellen. Somit bildet diese Arbeit ein Bindeglied zwischen der geomorphologischen beziehungsweise geologischen und (geo-)archäologischen Erforschung eines mittelalterlichen Standortes und liefert wichtige Informationen über die wechselseitige Einflussnahme dieser Forschungsgegenstände im Spessart und darüber hinaus. Their construction and destruction were often determined by the struggle for political and economic supremacy in the region and for control over the Spessart's natural resources. Wahlmich Castle is located in a relatively uncommon strategic and geomorphological position, characterized by a fairly remote position and atypical rough relief. In order to reconstruct the local relief development and possible human impact, a multi-method approach was applied combining two-dimensional geoelectrical measurements, geomorphological mapping and stratigraphic-sedimentological investigations. This provides new insights into the influence of landscape characteristics on choices of castle locations. The combined geoelectrical, geomorphological and stratigraphic-sedimentological data show that the rough relief is of natural origin and influenced by regional faulting, which triggered sliding and slumping as well as weathering and dissection of the surface deposits. The rough relief and the lithology permitted intensive land use and building activities. However, the location of the castle offered access to and possibly control over important medieval traffic routes and also represented certain ownership claims in the Aschaff River valley. The economic situation combined with rivalry between different elites led to the castle being built in a geomorphological challenging and strategically less valuable location. Focusing on castles located in rare and challenging geomorphological positions may therefore lead to a better understanding of castle siting in the future. Pathophysiologically, recent positron emission tomography data suggest astroglial activation after experimental myocardial infarction (MI). We analyzed peripheral surrogate markers of glial (and neuronal) damage serially within 12 months after the first ST-elevation MI (STEMI). Serum levels of glial fibrillary acidic protein (GFAP) and neurofilament light chain (NfL) were quantified using ultra-sensitive molecular immunoassays. Sufficient biomaterial was available from 45 STEMI patients (aged 28 to 78 years, median 56 years, 11% female). The median (quartiles) of GFAP was 63.8 (47.0, 89.9) pg/mL and of NfL 10.6 (7.2, 14.8) pg/mL at study entry 0–4 days after STEMI. GFAP after STEMI increased in the first 3 months, with a median change of +7.8 (0.4, 19.4) pg/mL (p = 0.007). It remained elevated without further relevant increases after 6 months (+11.7 (0.6, 23.5) pg/mL; p = 0.015), and 12 months (+10.3 (1.5, 22.7) pg/mL; p = 0.010) compared to the baseline. Larger relative infarction size was associated with a higher increase in GFAP (ρ = 0.41; p = 0.009). In contrast, NfL remained unaltered in the course of one year. Our findings support the idea of central nervous system involvement after MI, with GFAP as a potential peripheral biomarker of chronic glial damage as one pathophysiologic pathway. Thus, detecting and preventing mild cognitive impairment, which is common in patients with HF, is of great importance. Serum biomarkers are increasingly used in neurological disorders for diagnostics, monitoring, and prognostication of disease course. It remains unclear if neuronal biomarkers may help detect cognitive impairment in this high-risk population. Also, the influence of chronic HF and concomitant renal dysfunction on these biomarkers is not well understood. Methods Within the monocentric Cognition.Matters-HF study, we quantified the serum levels of phosphorylated tau protein 181 (pTau) and neurofilament light chain (NfL) of 146 extensively phenotyped chronic heart failure patients (aged 32 to 85 years; 15.1% women) using ultrasensitive bead-based single-molecule immunoassays. The clinical work-up included advanced cognitive testing and cerebral magnetic resonance imaging (MRI). Results Serum concentrations of NfL ranged from 5.4 to 215.0 pg/ml (median 26.4 pg/ml) and of pTau from 0.51 to 9.22 pg/ml (median 1.57 pg/ml). We detected mild cognitive impairment (i.e., T-score < 40 in at least one cognitive domain) in 60% of heart failure patients. pTau (p = 0.014), but not NfL, was elevated in this group. Both NfL (ρ = − 0.21; p = 0.013) and pTau (ρ = − 0.25; p = 0.002) related to the cognitive domain visual/verbal memory, as well as white matter hyperintensity volume and cerebral and hippocampal atrophy. In multivariable analysis, both biomarkers were independently influenced by age (T = 4.6 for pTau; T = 5.9 for NfL) and glomerular filtration rate (T = − 2.4 for pTau; T = − 3.4 for NfL). Markers of chronic heart failure, left atrial volume index (T = 4.6) and NT-proBNP (T = 2.8), were further cardiological determinants of pTau and NfL, respectively. In addition, pTau was also strongly affected by serum creatine kinase levels (T = 6.5) and ferritin (T = − 3.1). Conclusions pTau and NfL serum levels are strongly influenced by age-dependent renal and cardiac dysfunction. These findings point towards the need for longitudinal examinations and consideration of frequent comorbidities when using neuronal serum biomarkers. Serum glial fibrillary acidic protein (GFAP) is an emerging biomarker of cognitive decline in disorders of primary neurodegeneration such as Alzheimer's disease. We evaluated the utility of serum GFAP as a biomarker for cognitive dysfunction and structural brain damage in patients with stable chronic HF. Methods and results Using bead-based single molecule immunoassays, we quantified serum levels of GFAP in patients with HF participating in the prospective Cognition.Matters-HF study. Participants were extensively phenotyped, including cognitive testing of five separate domains and magnetic resonance imaging (MRI) of the brain. Univariable and multivariable models, also accounting for multiple testing, were run. One hundred and forty-six chronic HF patients with a mean age of 63.8 ± 10.8 years were included (15.1% women). Serum GFAP levels (median 246 pg/mL, quartiles 165, 384 pg/mL; range 66 to 1512 pg/mL) did not differ between sexes. In the multivariable adjusted model, independent predictors of GFAP levels were age (T = 5.5; P < 0.001), smoking (T = 3.2; P = 0.002), estimated glomerular filtration rate (T = −4.7; P < 0.001), alanine aminotransferase (T = −2.1; P = 0.036), and the left atrial end-systolic volume index (T = 3.4; P = 0.004). NT-proBNP but not serum GFAP explained global cerebral atrophy beyond ageing. However, serum GFAP levels were associated with the cognitive domain visual/verbal memory (T = −3.0; P = 0.003) along with focal hippocampal atrophy (T = 2.3; P = 0.025). Conclusions Serum GFAP levels are affected by age, smoking, and surrogates of the severity of HF. The association of GFAP with memory dysfunction suggests that astroglial pathologies, which evade detection by conventional MRI, may contribute to memory loss beyond ageing in patients with chronic HF. Intriguingly, components of nuclear paraspeckles like the non-POU domain containing octamer-binding protein (NONO) participate in the repair of DNA double-strand breaks (DSBs). NONO is a multifunctional RNA-binding protein (RBP) that facilitates the retention and editing of messenger (m)RNA as well as pre-mRNA processing. However, the role of NONO in the DDR is poorly understood. Here, we establish a novel human U2OS cell line that expresses NONO fused to the engineered ascorbate peroxidase 2 (U2OS:NONO-APEX2-HA). We show that NONO-APEX2-HA accumulates in the nucleolus in response to DNA damage. Combining viability assays, subcellular localisation studies, coimmunoprecipitation experiments and in vivo proximity labeling, we demonstrate that NONO-APEX2-HA is a stably expressed fusion protein that mimics endogenous NONO in terms of expression, localisation and bona fide interactors. We propose that in vivo proximity labeling in U2OS:NONO-APEX2-HA cells is capable for the assessment of NONO interactomes by downstream assays. U2OS:NONO-APEX2-HA cells will likely be a valuable resource for the investigation of NONO interactome dynamics in response to DNA damage and other stimuli. Cardiovascular diseases and type 2 diabetes are only some of the pathologies associated to overweight. Among the main causes are decreased physical activity and food availability and composition. Diets with high content of fat are energy-dense and their overconsumption leads to an energy imbalance, which ultimately promotes energy storage as fat and obesity. Aberrant activation of signalling cascades and hormonal imbalances are characteristic of this disease and members of the Protein Kinase D (PKD) family have been found to be involved in several mechanisms mediating metabolic homeostasis. Therefore, we aimed to investigate the role of Protein Kinase D2 (PKD2) in the regulation of metabolism. Our investigation initiated with a mice model for global PKD2 inactivation, which allowed us to prove a direct involvement of this kinase in lipids homeostasis and obesity. Inactivation of PKD2 protected the mice from high-fat diet-induced obesity and improved their response to glucose, insulin and lipids. Furthermore, the results indicated that, even though there were no changes in energy intake or expenditure, inactivation of PKD2 limited the absorption of fat from the intestine and promoted energy excretion in feces. These results were verified in a mice model for specific deletion of intestinal PKD2. These mice not only displayed an improved metabolic fitness but also a healthier gut microbiome profile. In addition, we made use of a small-molecule inhibitor of PKD in order to prove that local inhibition of PKD2 in the intestine was sufficient to inhibit lipid absorption. The usage of the inhibitor not only protected the mice from obesity but also was efficient in avoiding additional body-weight gain after obesity was pre-established in mice. Mechanistically, we determined that PKD2 regulates lipids uptake in enterocytes by phosphorylation of Apolipoprotein A4 (APOA4) and regulation of chylomicron-mediated triglyceride absorption. PKD2 deletion or inactivation increased abundance of APOA4 and decreased the size of chylomicrons and therefore lipids absorption from the diet. Moreover, intestinal activation of PKD2 in human obese patients correlated with higher levels of triglycerides in circulation and a detrimental blood profile. In conclusion, we demonstrated that PKD2 is a key regulator of dietary fat absorption in murine and human context, and its inhibition might contribute to the treatment of obesity. Herz-Kreislauf-Erkrankungen und Typ- 2-Diabetes sind nur einige der mit Übergewicht verbundenen Pathologien. Zu den Hauptursachen zählen eine verminderte körperliche Aktivität sowie die Verfügbarkeit und Zusammensetzung von Nahrungsmitteln. Diäten mit hohem Fettgehalt haben eine hohe Energiedichte und ihr übermäßiger Konsum führt zu einem Energieungleichgewicht, das letztendlich die Energiespeicherung als Fett und Fettleibigkeit fördert. Aberrante Aktivierung von Signalkaskaden und hormonelle Ungleichgewichte sind charakteristisch für diese Krankheit, und es wurde festgestellt, dass Mitglieder der Protein Kinase D (PKD) -Familie an mehreren Mechanismen der metabolischen Homöostase beteiligt sind. Daher zielten wir darauf ab die Rolle der Proteinkinase D2 (PKD2) bei der Regulation des Stoffwechsels zu untersuchen. Unsere Untersuchung begann mit einem Mäusemodell für die globale PKD2- Inaktivierung, welches es uns ermöglichte, eine direkte Beteiligung dieser Kinase an der Lipidhomöostase und Fettleibigkeit nachzuweisen. Die Inaktivierung von PKD2 schützte die Mäuse vor fettreicher diätbedingter Fettleibigkeit und verbesserte ihre Reaktion auf Glukose, Insulin und Lipide. Darüber hinaus zeigten die Ergebnisse, dass die Inaktivierung von PKD2 die Absorption von Fett über den Darm begrenzte und die Energieausscheidung im Kot förderte, obwohl sich die Energieaufnahme oder der Energieverbrauch nicht änderten. In the UBA6-FAT10 complex, the C-terminal domain of FAT10 binds to where ubiquitin resides in the UBA1-ubiquitin complex, however, a switch element ensures the alternate recruitment of either modifier. Simultaneously, the N-terminal domain of FAT10 interacts with the 3-helix bundle of UBA6. Therefore, the monitoring of temperature on fruit surface (FST) over the growing period can allow to identify thresholds, above of which several physiological disorders such as sunburn may occur in apple. The current approaches neglect spatial variation of FST and have reduced repeatability, resulting in unreliable predictions. In this study, LiDAR laser scanning and thermal imaging were employed to detect the temperature on fruit surface by means of 3D point cloud. A process for calibrating the two sensors based on an active board target and producing a 3D thermal point cloud was suggested. After calibration, the sensor system was utilised to scan the fruit trees, while temperature values assigned in the corresponding 3D point cloud were based on the extrinsic calibration. Whereas a fruit detection algorithm was performed to segment the FST from each apple. • The approach allows the calibration of LiDAR laser scanner with thermal camera in order to produce a 3D thermal point cloud. • The method can be applied in apple trees for segmenting FST in 3D. Considering this fact, we tested in vitro and in vivo water evaporation rates from the cellulose dressing epicite\(^{hydro}\) when combined with different secondary dressings as well as the resulting wound healing efficacy in a porcine donor site model. The aim of this study was to evaluate how the different rates of water evaporation affected wound healing efficacy. To this end, epicite\(^{hydro}\) primary dressing, in combination with different secondary dressing materials (cotton gauze, JELONET\(^◊\), AQUACEL\(^®\) Extra\(^™\), and OPSITE\(^◊\) Flexifix), was placed on 3 × 3 cm-sized dermatome wounds with a depth of 1.2 mm on the flanks of domestic pigs. The healing process was analyzed histologically and quantified by morphometry. High water evaporation rates by using the correct secondary dressing, such as cotton gauze, favored a better re-epithelialization in comparison with the low water evaporation resulting from an occlusive secondary dressing, which favored the formation of a new and intact dermal tissue that nearly fully replaced all the dermis that was removed during wounding. This newly available evidence may be of great benefit to clinical wound management. It is unknown whether repeated measurements provide additional information for risk prediction. Methods The MonDAFIS (Systematic Monitoring for Detection of Atrial Fibrillation in Patients with Acute Ischemic Stroke) study randomly assigned 3465 acute ischemic stroke patients to either standard procedures or an additive Holter electrocardiogram. Baseline eGFR (CKD‐EPI formula) were dichotomized into values of < versus ≥60 ml/min/1.73 m\(^{2}\). eGFR dynamics were classified based on two in‐hospital values as “stable normal” (≥60 ml/min/1.73 m\(^{2}\)), “increasing” (by at least 15% from baseline, second value ≥ 60 ml/min/1.73 m\(^{2}\)), “decreasing” (by at least 15% from baseline of ≥60 ml/min/1.73 m\(^{2}\)), and “stable decreased” (<60 ml/min/1.73 m\(^{2}\)). The composite endpoint (stroke, major bleeding, myocardial infarction, all‐cause death) was assessed after 24 months. We estimated hazard ratios in confounder‐adjusted models. Results Estimated glomerular filtration rate at baseline was available in 2947 and a second value in 1623 patients. After adjusting for age, stroke severity, cardiovascular risk factors, and randomization, eGFR < 60 ml/min/1.73 m\(^{2}\) at baseline (hazard ratio [HR] = 2.2, 95% confidence interval [CI] = 1.40–3.54) as well as decreasing (HR = 1.79, 95% CI = 1.07–2.99) and stable decreased eGFR (HR = 1.64, 95% CI = 1.20–2.24) were independently associated with the composite endpoint. In addition, eGFR < 60 ml/min/1.732 at baseline (HR = 3.02, 95% CI = 1.51–6.10) and decreasing eGFR were associated with all‐cause death (HR = 3.12, 95% CI = 1.63–5.98). Conclusions In addition to patients with low eGFR levels at baseline, also those with decreasing eGFR have increased risk for vascular events and death; hence, repeated estimates of eGFR might add relevant information to risk prediction. Methods The post hoc analysis included 1080 patients of the investigator-initiated, multicenter prospective Berlin Atrial Fibrillation Registry, designed to analyze medical stroke prevention in AF patients after acute ischemic stroke. Results At stroke onset, an off-label daily dose was prescribed in 61 (25.5%) of 239 NOAC patients with known AF and CHA2DS2-VASc score ≥ 1, of which 52 (21.8%) patients were under-dosed. Under-dosing was associated with age ≥ 80 years in patients on rivaroxaban [OR 2.90, 95% CI 1.05-7.9, P = 0.04; n = 29] or apixaban [OR 3.24, 95% CI 1.04-10.1, P = 0.04; n = 22]. At hospital discharge after the index stroke, NOAC off-label dose on admission was continued in 30 (49.2%) of 61 patients. Overall, 79 (13.7%) of 708 patients prescribed a NOAC at hospital discharge received an off-label dose, of whom 75 (10.6%) patients were under-dosed. Rivaroxaban under-dosing at discharge was associated with age ≥ 80 years [OR 3.49, 95% CI 1.24-9.84, P = 0.02; n = 19]; apixaban under-dosing with body weight ≤ 60 kg [OR 0.06, 95% CI 0.01-0.47, P < 0.01; n = 56], CHA2DS2-VASc score [OR per point 1.47, 95% CI 1.08-2.00, P = 0.01], and HAS-BLED score [OR per point 1.91, 95% CI 1.28-2.84, P < 0.01]. Conclusion At stroke onset, off-label dosing was present in one out of four, and under-dosing in one out of five NOAC patients. Under-dosing of rivaroxaban or apixaban was related to old age. In-hospital treatment after stroke reduced off-label NOAC dosing, but one out of ten NOAC patients was under-dosed at discharge. In this regard, Earth observation (EO) is a valuable tool for monitoring of long term changes over the land surface and its features. Although investigations commonly study environmental change by means of a single EO-based land surface variable, a joint exploitation of multivariate land surface variables covering several spheres is still rarely performed. In this regard, we present a novel methodological framework for both, the automated processing of multisource time series to generate a unified multivariate feature space, as well as the application of statistical time series analysis techniques to quantify land surface change and driving variables. In particular, we unify multivariate time series over the last two decades including vegetation greenness, surface water area, snow cover area, and climatic, as well as hydrological variables. Furthermore, the statistical time series analyses include quantification of trends, changes in seasonality, and evaluation of drivers using the recently proposed causal discovery algorithm Peter and Clark Momentary Conditional Independence (PCMCI). We demonstrate the functionality of our methodological framework using Indo-Gangetic river basins in South Asia as a case study. The time series analyses reveal increasing trends in vegetation greenness being largely dependent on water availability, decreasing trends in snow cover area being mostly negatively coupled to temperature, and trends of surface water area to be spatially heterogeneous and linked to various driving variables. Overall, the obtained results highlight the value and suitability of this methodological framework with respect to global climate change research, enabling multivariate time series preparation, derivation of detailed information on significant trends and seasonality, as well as detection of causal links with minimal user intervention. With the launch of many new insect monitoring schemes to investigate insect communities over large spatial and temporal scales, Malaise traps have become one of the most important tools due to the broad spectrum of species collected and reduced capture bias through passive sampling of insects day and night. However, Malaise traps can vary in size, shape, and colour, and it is unknown how these differences affect biomass, species richness, and composition of trap catch, making it difficult to compare results between studies. We compared five Malaise trap types (three variations of the Townes and two variations of the Bartak Malaise trap) to determine their effects on biomass and species richness as identified by metabarcoding. Insect biomass varied by 20%–55%, not strictly following trap size but varying with trap type. Total species richness was 20%–38% higher in the three Townes trap models compared to the Bartak traps. Bartak traps captured lower richness of highly mobile taxa but increased richness of ground‐dwelling taxa. The white roofed Townes trap captured a higher richness of pollinators. We find that biomass, total richness, and taxa group specific richness are all sensitive to Malaise trap type. Trap type should be carefully considered and aligned to match monitoring and research questions. Additionally, our estimates of trap type effects can be used to adjust results to facilitate comparisons across studies. In order to achieve this, the imaging system must yield an optical resolution of a few micrometers or less, which reduces the measurement area (field of view = FOV) to a few mm at most. If the sample size, however, exceeds the field of view (called local tomography or region of interest = ROI CT), filter problems arise during the CT reconstruction and phase retrieval in holotomography. In this paper, we will first investigate the practical impact of these filter problems and discuss approximate solutions. Secondly, we will investigate the effectiveness of a technique we call “multiscalar holotomography”, where, in addition to the ROI CT, a lower resolution non-ROI CT measurement is recorded. New and rapidly developable strategies for antimicrobial substances, in particular substances highly potent against multidrug-resistant bacteria, are urgently required. Some of the treatment options currently available for multidrug-resistant bacteria are considerably limited by side effects and unfavorable pharmacokinetics. The glycopeptide vancomycin is considered an antibiotic of last resort. Its use is challenged by bacterial strains exhibiting various types of resistance. Therefore, in this study, highly active polycationic peptide-vancomycin conjugates with varying linker characteristics or the addition of PEG moieties were synthesized to optimize pharmacokinetics while retaining or even increasing antimicrobial activity in comparison to vancomycin. The antimicrobial activity of the novel conjugates was determined by microdilution assays on susceptible and vancomycin-resistant bacterial strains. VAN1 and VAN2, the most promising linker-modified derivatives, were further characterized in vivo with molecular imaging and biodistribution studies in rodents, showing that the linker moiety influences both antimicrobial activity and pharmacokinetics. Encouragingly, VAN2 was able to undercut the resistance breakpoint in microdilution assays on vanB and vanC vancomycin-resistant enterococci. Out of all PEGylated derivatives, VAN:PEG1 and VAN:PEG3 were able to overcome vanC resistance. Biodistribution studies of the novel derivatives revealed significant changes in pharmacokinetics when compared with vancomycin. In conclusion, linker modification of vancomycin-polycationic peptide conjugates represents a promising strategy for the modulation of pharmacokinetic behavior while providing potent antimicrobial activity. This includes the discovery of the molecular mechanisms of tumor vascularization, which is critically required for establishment of tumor metastasis. Formation of new blood vessels is the first step in tumor vascularization. Therefore, understanding the molecular and cellular basis of tumor vascularization attracted a significant effort studying in biomedical research. The blood vessels for supplying tumor can be formed by sprouting from pre-existing vessels, a process called angiogenesis, or by vasculogenesis, that is de novo formation of blood vessels from not fully differentiated progenitor cell populations. Vasculogenic endothelial progenitor cells (EPCs) can either be activated from populations in the bone marrow reaching the pathological region via the circulation or they can be recruited from local reservoirs. Neovessel formation influences tumor progression, hence therapeutic response model systems of angiogenesis/vasculogenesis are necessary to study the underlying mechanisms. Although, initially the research in this area focused more on angiogenesis, it is now well understood that both angiogenesis and postnatal vasculogenesis contribute to neovessel formation in adult under both most pathological as well as physiological conditions. Studies in the last two decades demonstrate that in addition to the intimal layer of fully differentiated mature endothelial cells (ECs) and various smaller supplying vessels (vasa vasorum) that can serve as a source for new vessels by angiogenesis, especially the adventitia of large and medium size blood vessels harbors various vascular wall-resident stem and progenitor cells (VW-SPCs) populations that serve as a source for new vessels by postnatal vasculogenesis. However, little is known about the potential role of VW-SPCs in tumor vascularization. To this end, the present work started first to establish a modified aortic ring assay (ARA) using mouse aorta in order to study the contribution of vascular adventitia-resident VW-SPCs to neovascularization in general and in presence of tumor cells. ARA is already established an ex vivo model for neovascularization allows to study the morphogenetic events of complex new vessel formation that includes all layers of mature blood vessels, a significant advantage over the assays that employ monolayer endothelial cell cultures. Moreover, in contrast to assays employing endothelial cells monocultures, both angiogenic and vasculogenic events take place during new vessel formation in ARA although the exact contribution of these two processes to new vessel formation cannot be easily distinguished in conventional ARA. Thus, in this study, a modified protocol for the ARA (mdARA) was established by either removing or keeping the aortic adventitia in place. The mdARA allows to distinguish the role of VW-SPCs from those of other aortic layers. The present data show that angiogenic sprouting from mature aortic endothelium was markedly delayed when the adventitial layer was removed. Furthermore, the network between the capillary-like sprouts was significantly reduced in absence of aortic adventitia. Moreover, the stabilization of new sprouts by assembling the NG2+ pericyte-like cells that enwrapped the endothelial sprouts from the outside was improved when the adventitial layer remained in place. Next, mimicking the tumor-vessel adventitia-interaction, multicellular tumor spheroids (MCTS) and aortic rings (ARs) with or without adventitia of C57BL/6-Tg (UBC-GFP) mice were confronted within the collagen gel and cultured ex vivo. This 3D model enabled analysis of the mobilization, migration and capillary-like sprouts formation by VW-SPCs within tumor-vessel wall-interface in comparison to tumor-free side of the ARs. Interestingly, while MCTS preferred the uptake of single vascular adventitia-derived cells, neural spheroids were directly penetrated by capillary-like structures that were sprouted from the aortic adventitia. In summary, the model established in this work allows to study new vessel formation by both postnatal vasculogenesis and angiogenesis under same conditions. It can be applied in various mouse models including reporter mouse models, e.g. Cxcr1 CreER+/mTmG+/- mice, in which GFP-marked macrophages of the vessel wall were directly observed as they mobilized from their niche and migrated into collagen gel. Another benefit of the model is that it can be used for testing different factors such as small molecules, growth factors, cytokines, and drugs with both pro- and anti-angiogenic/vasculogenic effects. N2 - Die Forschungsarbeiten der letzten Jahrzehnte zur Bekämpfung der Krebserkrankung, einer der dominierenden Krankheiten des 21. Jahrhunderts, haben zu großen Fortschritten, insbesondere im Verständnis bezüglich der Tumormetastasierung geführt. Dies schließt die Prozesse der Tumorvaskularisierung als einen der initialen Schritte der Metastasierung mit ein, die nach wie vor nicht ausreichend geklärt sind. Die Blutgefäßbildung zur Versorgung des Tumorgewebes kann durch die Anagiogenese, die als Einsprießen neuer Gefäße aus den bereits vorhandenen Blutgefäßen definiert wird, oder durch die Vaskulogenese, die als Gefäßneubildung aus Stamm- und Vorläuferzellen beschrieben wird, sichergestellt werden. Noch nicht vollständig ausdifferenzierte endotheliale Vorläuferzellen (EPCs) werden dabei nach dem bisherigen Kenntnistand aus dem Knochenmark rekrutiert und erreichen die Regionen der Gefäßneubildung über die Blutzirkulation und können dort zur de novo Formierung neuer Blutgefäße auch beim Erwachsenen beitragen. Untersuchungen der letzten zwei Dekaden haben gezeigt, dass solche Vorläuferzellen auch aus lokalen Reservoiren, wie z.B. aus der Gefäßwandadventitia der bereits existierenden Blutgefäße mobilisiert werden. Da die Bildung neuer Gefäße einen direkten Einfluss auf die Tumorprogression hat, sind entsprechende Modellsysteme notwendig, um die zugrundeliegenden Mechanismen möglichst präzise zu untersuchen. Obwohl sich die Forschung der Tumorvaskularisierung zunächst auf Prozesse der Angiogenese konzentrierte, ist es mittlerweile ausreichend belegt, dass auch die Vaskulogenese zur Tumorvaskularisierung beiträgt und somit die Tumorprogression beeinflusst. Anhand einer Vielzahl an Studien der letzten zwei Jahrzehnte konnte demonstriert werden, dass sich, neben der Intimaschicht, die vollständig differenzierte Endothelzellen (ECs) enthält und kleineren Gefäßwand-versorgenden Blutgefäßen, der sogenannten Vasa vasorum in der Adventitia der großen Gefäße, auch Populationen gefäßwandresidenter Stamm- und Vorläuferzellen (VW-SPCs) in der äußeren Gefäßwandschicht, nämlich der Adventitia fast aller Gefäßabschnitte nachweisen lassen. Mittels ARA kann die Bildung komplexer vaskulärer Strukturen in Präsenz aller Wandschichten reifer Blutgefäße untersucht werden, was einen wesentlichen Vorteil gegenüber der Verwendung von Endothelzellkulturen in Monolayer bedeutet. Hierbei ist jedoch anzumerken, dass in ARA sowohl angiogene als auch vaskulogene Prozesse zur Gefäßbildung beitragen, aber der genaue Beitrag beider Prozesse schwer oder kaum voneinander unterscheidbar ist. Daher wurde im Rahmen dieser Arbeit ein modifiziertes ARA-Protokoll etabliert (mdARA), in welchem die aortale Adventitia vor dem Beginn des ARA entweder entfernt oder belassen wurde und somit die Rolle der VW-SPCs bei der Gefäßsprossung von den Zellen anderer Aortenwandschichten differenziert studiert werden konnte. Die dabei generierten Daten zeigen, dass sich die angiogene Aussprossung aus dem reifen Aortenendothel nach Entfernung der adventitialen Schicht deutlich verzögerte. Darüber hinaus war das Netzwerk zwischen den kapillarartigen Sprossen in Abwesenheit der Aortenadventitia signifikant reduziert. Mehr noch, das Belassen der adventitialen Wandstruktur führte zu einer verbesserten Stabilisierung neuer Gefäßsprossen. Als sichtbares Korrelat hierfür zeigte sich eine stärkere und bessere Anlagerung der NG2+ Perizyten-ähnlichen Zellen zu den endothelialen Kapillar-ähnlichen Aussprossungen von außen, wie Perizyten an der Kapillarwand in situ. Als nächstes wurden die Aortenringe (ARs) von C57BL/6-Tg (UBC-GFP)-Mäusen mit multizellulären Tumor-Sphäroiden (MCTS) in Kollagengel ko-kultiviert, um die Interaktion zwischen Tumor und Gefäßwand-Adventitia ex vivo nachzuahmen. Dieses 3D Modell ermöglichte die Analyse der Mobilisierung und Migration der VW-SPCs von der aortalen Adventitia sowohl zu der Tumorseite in den Tumor-Gefäßwand-Interfaces als auch zu der tumorfreien Seite der Aortenringe. Interessanterweise wurde die Kapillarsprossung im Tumor-Gefäßwand-Interface an der Grenze zum MCTS gestoppt und die VW-SPCs als Einzelzellen in die MCTS aufgenommen. Demgegenüber wurde auf der tumor-freien Seite der Aortenringe eine deutlich längere Kapillaraussprossung beobachtet. Im Gegensatz zu MCTS resultierte die Ko-Kultivierung der ARs mit neuronalen Spheroiden darin, dass die aus der aortalen Adventitia aussprossenden Kapillar-ähnlichen Strukturen direkt in die neuronalen Spheroide penetrierten. Zusammenfassend berücksichtigt dieses neuartige in vitro 3D-Modell sowohl Angiogenese als auch Vaskulogenese und bietet vielfältige Vorteile, wie zum Beispiel die Kompatibilität zu verschiedenen Mausmodellen einschließlich der Reporter-Mausmodelle, wie z.B. die in dieser Arbeit gezeigte Verwendung der Aorta von Cxcr1 CreER+/mTmG+/- um die GFP-markierten Makrophagen aus der Gefäßwand bei der Gefäßaussprossung studieren zu können. So far, no large cohort study on this topic has been published with data on to age distribution, operative procedures, hospital volume and operative outcome. Methods This is a retrospective analysis of anonymized nationwide hospital billing data (DRG data, 2009-2017). All adrenal surgeries (defined by OPS codes) of patients between the age 0 and 21 years in Germany were included. Results A total of 523 patient records were identified. The mean age was 8.6 ± 7.7 years and 262 patients were female (50.1%). The majority of patients were between 0 and 5 years old (52% overall), while 11.1% were between 6 and 11 and 38.8% older than 12 years. The most common diagnoses were malignant neoplasms of the adrenal gland (56%, mostly neuroblastoma) with the majority being younger than 5 years. Benign neoplasms in the adrenal gland (D350) account for 29% of all cases with the majority of affected patients being 12 years or older. 15% were not defined regarding tumor behavior. Overall complication rate was 27% with a clear higher complication rate in resection for malignant neoplasia of the adrenal gland. Bleeding occurrence and transfusions are the main complications, followed by the necessary of relaparotomy. There was an uneven patient distribution between hospital tertiles (low volume, medium and high volume tertile). While 164 patients received surgery in 85 different “low volume” hospitals (0.2 cases per hospital per year), 205 patients received surgery in 8 different “high volume” hospitals (2.8 cases per hospital per year; p<0.001). Patients in high volume centers were significant younger, had more extended resections and more often malignant neoplasia. In multivariable analysis younger age, extended resections and open procedures were independent predictors for occurrence of postoperative complications. Conclusion Overall complication rate of adrenalectomies in the pediatric population in Germany is low, demonstrating good therapeutic quality. Our analysis revealed a very uneven distribution of patient volume among hospitals. In recent years, with the development of new technologies, it has become possible to study gene expression at single-cell level. This new advances in single-cell RNA-sequencing has revolutionized the way scientists study biological processes. Single-cell RNA-sequencing has been used in different areas to better understand the underlying mechanisms of biological processes. In particular, single-RNA-sequencing is a suitable method to study infectious diseases. Infection is composed of heterogeneous mechanisms on either the host or pathogen side and the best way to understand the heterogeneity of these mechanisms and how they interact with each other is to study infectious diseases at the single-cell level. Studying infection processes at the single-cell level can reveal not only the heterogeneity but also the dynamics of infection and the interplay between the host and pathogen at the molecular level. In this thesis, we implemented and applied different single-cell RNA-seq technologies to better understand infectious diseases. In the present work, we conducted four independent but related research works to shed light on different aspects of infection biology: ● We took advantage of this novel technology to study the consequences of RSV infection on primary human epithelial cells. The primary human epithelial cells were collected from six donors and cultured in air liquid interface (ALI) cell culture inoculated with respiratory syncytial virus (RSV). In this project, we discovered ciliated cells as the susceptible cell types in RSV infection. We applied viral load as an indicator of infection progression and used it to reconstruct the dynamics of host response to RSV infection. Reconstruction of the dynamics of infection revealed many host genes and pathways that were suppressed or induced as a result of RSV infection. Pathways related to innate immune response and interferon response were suppressed during the progression of infection and on the other hand pathways like protein targeting to endoplasmic reticulum and apoptosis were induced. ● We developed a new method which is capable of sequencing the transcriptome of a bacterium at the single-cell level and potentially can help us to characterize the bacterial heterogeneity during the course of infection. In this research project, bacteria were cultured in three different culture conditions namely Late stationary phase, Anaerobic shock and NaCl shock and we used a poly(A)-independent single-cell RNA-sequencing protocol to sequence bacteria at the single-cell level. In this work, we report the faithful capture of growth-dependent gene expression patterns in individual Salmonella and Pseudomonas bacteria. The results of our analysis showed that not only we could capture transcripts across different RNA classes but also our method is capable of discerning the transcriptome of bacteria across different culture conditions. ● We used single-cell RNA-sequencing technology to characterize the immune cells landscape over the course of atherosclerosis. Atherosclerosis is considered a cardiac disease which is highly related to infections and previous infections with bacteria or viruses is considered as a risk factor for atherosclerosis. We performed single-cell RNA sequencing of aortic CD45+ cells extracted from healthy and atherosclerotic aorta of mice. We managed to find certain cell populations which were specifically present in atherosclerotic mice. One of the atheroschelorotic populations was previously undescribed TREM2high macrophages showing enrichment in Trem2 gene expression. This population of macrophages seemed to be involved in functions like lipid metabolism and catabolism and lesion calcification. This work revealed the phenotypic heterogeneity and immune cells landscape of different immune cell populations at different stages of atherosclerosis. Our work paves the way to better describe the relation between different infectious diseases and cardiovascular diseases. ● We developed a web-based platform called Infection Atlas to browse and visualize single-cell RNA-sequencing data. Infection Atlas platform provides a user-friendly interface to study different aspects of infectious diseases at the single-cell level and can potentially promote targeted approaches to intervene in infectious diseases. This platform which is available at infection-atlas.org in the short term provides a user-friendly interface to browse and visualize different aspects of infectious diseases and in the long-term is expected to be a comprehensive atlas of infection in human and mouse across different tissues and different pathogens. Overall, in this thesis we provide a framework to study infectious diseases at the single cell level with providing novel data analysis methods and this thesis paves the way for future studies to study host-pathogen encounters at the single-cell level. Diese Fortschritte in der Einzelzell-RNA- Sequenzierung haben die Art wie Wissenschaftler biologische Prozesse betrachten von Grund auf verändert. Einzelzell-Sequenzierung wird in unterschiedlichen Bereichen angewendet, um die grundlegenden Mechanismen biologischer Prozesse besser zu verstehen. Besonders Einzelzell-Sequenzierung ist eine geeignete Methode, um Infektionskrankheiten zu untersuchen. Infektionen sind durch heterogene Mechanismen auf Wirts- und Erreger Seite gekennzeichnet. Die Rekonstruktion der Infektionsdynamik zeigte viele Wirtsgene und Signalwege, die durch die RSV Infektion unterdrückt oder induziert wurden. Signalwege, die mit der angeborenen Immunantwort und der Interferonantwort assoziiert waren, wurden durch die fortschreitende Infektion unterdrückt und andererseits waren Signalwege, wie die Zielsteuerung von Proteinen zum endoplasmatischen Retikulum und Apoptose induziert. ● Wir haben eine neue Methode entwickelt, die es ermöglicht das Transkriptom eines Bakteriums auf Einzelzell-Niveau zu sequenzieren und potenziell helfen könnte die bakterielle Heterogenität während des Verlaufs einer Infektion zu charakterisieren. In diesem Forschungsprojekt wurden Bakterien unter folgenden drei unterschiedlichen Konditionen angezogen: Späte stationäre Phase, anaerober Schock und Natriumchlorid Schock. Anschließend wendeten wir ein poly(A) unabhängiges Einzelzell-RNA Sequenzier-Protokoll an, um Bakterien auf Einzelzell-Niveau zu sequenzieren. In dieser Arbeit berichten wir die von wachstumsabhängigen Genexpressionsmustern in einzelnen Salmonellen und Pseudomonaden. Hierbei konnten wir bestimmte Zellpopulationen identifizieren, die spezifisch in atherosklerotischen Mäusen vorkommen. Pollination services of cacao are crucial for global chocolate production, yet remain critically understudied, particularly in regions of origin of the species. Notably, uncertainties remain concerning the identity of cacao pollinators, the influence of landscape (forest distance) and management (shade cover) on flower visitation and the role of pollen deposition in limiting fruit set. 2. Here, we aimed to improve understanding of cacao pollination by studying limiting factors of fruit set in Peru, part of the centre of origin of cacao. Flower visitors were sampled with sticky insect glue in 20 cacao agroforests in two biogeographically distinct regions of Peru, across gradients of shade cover and forest distance. Further, we assessed pollen quantities and compared fruit set between naturally and manually pollinated flowers. 3. The most abundant flower visitors were aphids, ants and thrips in the north and thrips, midges and parasitoid wasps in the south of Peru. We present some evidence of increasing visitation rates from medium to high shade (40%–95% canopy closure) in the dry north, and opposite patterns in the semi-humid south, during the wet season. 4. Natural pollination resulted in remarkably low fruit set rates (2%), and very low pollen deposition. After hand pollination, fruit set more than tripled (7%), but was still low. 5. The diversity and high relative abundances of herbivore flower visitors limit our ability to draw conclusions on the functional role of different flower visitors. The remarkably low fruit set of naturally and even hand pollinated flowers indicates that other unaddressed factors limit cacao fruit production. Such factors could be, amongst others, a lack of effective pollinators, genetic incompatibility or resource limitation. Several genetic alterations have been identified in WT over the past years. However, a clear-cut underlying genetic defect has remained elusive. Growing evidence suggests that miRNA processing genes play a major role in the formation of pediatric tumors, including WT. We and others have identified the microprocessor genes DROSHA and DGCR8 as key players in Wilms tumorigenesis. Exome sequence analysis of a cohort of blastemal-type WTs revealed the recurrent hotspot mutations DROSHA E1147K and DGCR8 E518K mapping to regions important for catalyic activity and RNA-binding. These alterations were expected to affect processing of miRNA precursors, ultimately leading to altered miRNA expression. Indeed, mutated tumor samples were characterized by distinct miRNA patterns. Notably, these mutations have been observed almost exclusively in WT, suggesting that they play a specific role in WT formation. The aim of the present work was to first examine the mutation frequency of DROSHA E1147K and DGCR8 E518K in a larger cohort of WTs, and to further characterize these microprocessor gene mutations as potential oncogenic drivers for WT formation. Screening of additional 700 WT samples by allele-specific PCR revealed a high frequency of DROSHA E1147K and DGCR8 E518K mutations, with the highest incidence found in tumors of high-risk histology. DROSHA E1147K was heterozygously expressed in all cases, which strongly implies a dominant negative effect. In contrast, DGCR8 E518K exclusively exhibited homozygous expression, suggestive for the mutation to act recessive. To functionally assess the mutations of the microprocessor complex in vitro, I generated stable HEK293T cell lines with inducible overexpression of DROSHA E1147K, and stable mouse embryonic stem cell (mESC) lines with inducible overexpression of DGCR8 E518K. To mimic the homozygous expression observed in WT, DGCR8 mESC lines were generated on a DGCR8 knockout background. Inducible overexpression of wild-type or mutant DROSHA in HEK293T cells showed that DROSHA E1147K leads to a global downregulation of miRNA expression. It has previously been shown that the knockout of DGCR8 in mESCs also results in a significant downregulation of canonical miRNAs. Inducible overexpression of wild type DGCR8 rescued this processing defect. DGCR8 E518K on the other hand, only led to a partial rescue. Differentially expressed miRNAs comprised members of the ESC cell cycle (ESCC) and let-7 miRNA families whose antagonism is known to play a pivotal role in the regulation of stem cell properties. Along with altered miRNA expression, DGCR8-E518K mESCs exhibited alterations in target gene expression potentially affecting various biological processes. We could observe decreased proliferation rates, most likely due to reduced cell viability. DGCR8-E518K seemed to be able to overcome the block of G1-S transition and to rescue the cell cycle defect in DGCR8-KO mESCs, albeit not to the full extent like DGCR8-wild-type. Moreover, DGCR8-E518K appeared to be unable to completely block epithelial-to-mesenchymal transition (EMT). Embryoid bodies (EBs) with the E518K mutation, however, were still able to silence the self-renewal program rescuing the differentiation defect in DGCR8-KO mESCs. Taken together, I could show that DROSHA E1147K and DGCR8 E518K are frequent events in WT with the highest incidence in high-risk tumor entities. Either mutation led to altered miRNA expression in vitro confirming our previous findings in tumor samples. While the DROSHA E1147K mutation resulted in a global downregulation of canonical miRNAs, DGCR8 E518K was able to retain significant activity of the microprocessor complex, suggesting that partial reduction of activity or altered specificity may be critical in Wilms tumorigenesis. Despite the significant differences found in the miRNA and mRNA profiles of DGCR8 E518K and DGCR8-wild-type mESCs, functional analysis showed that DGCR8 E518K could mostly restore important cellular functions in the knockout and only slightly differed from the wild-type situation. Further studies in a rather physiological environment, such as in a WT blastemal model system, may additionally help to better assess the subtle differences between DGCR8 E518K and DGCR8 wild-type observed in our mESC lines. Together with our findings, these model systems may thus contribute to better understand the role of these microprocessor mutations in the formation of WT. N2 - Der Wilms Tumor (WT), auch Nephroblastom genannt, ist der häufigste Nierentumor im Kindesalter. In den letzten Jahren wurden bereits mehrere genetische Veränderungen in Wilms Tumoren festgestellt. Bisher konnte jedoch keine eindeutige genetische Ursache gefunden werden. Immer mehr Erkenntnisse deuten darauf hin, dass miRNA Prozessierungsgene eine wichtige Rolle bei der Entstehung von pädiatrischen Tumoren, einschließlich von WT, spielen. Uns ist es gelungen die Mikroprozessor-Gene DROSHA und DGCR8 als entscheidende Faktoren in der WT-Entstehung zu identifizieren. Mit Hilfe der Exom-Sequenzierung einer Kohorte blastemreicher Wilms Tumoren konnten die wiederkehrenden Hotspot-Mutationen DROSHA E1147K and DGCR8 E518K gefunden werden. Diese Mutationen betreffen Regionen, die für die katalytische Aktivität und die Bindung von RNA wichtig sind. Bemerkenswerterweise wurden diese Mutationen fast ausschließlich in WT beobachtet, was darauf hindeutet, dass sie eine spezifische Rolle bei der Entstehung von WT spielen. Ziel der vorliegenden Arbeit war es, zunächst die Mutationshäufigkeit von DROSHA E1147K und DGCR8 E518K in einer größeren Kohorte von Wilms Tumoren zu untersuchen und anschließend diese Mikroprozessor-Genmutationen als mögliche onkogene Treiber für die WT-Entstehung näher zu charakterisieren. Das Screening von zusätzlichen 700 WT-Proben mittels allelspezifischer PCR ergab eine hohe Häufigkeit von DROSHA E1147K und DGCR8 E518K Mutationen. Dabei traten sie vermehrt bei Tumoren mit einer Hochrisikohistologie auf. Die DROSHA E1147K Mutation wurde in allen Fällen heterozygot exprimiert, was einen dominant-negativen Effekt impliziert. Im Gegensatz dazu zeigte die DGCR8 E518K Mutation ausschließlich eine homozygote Expression, was darauf hindeutet, dass die Mutation rezessiv wirkt. Um die Mutationen des Mikroprozessorkomplexes in vitro funktionell zu untersuchen, generierte ich stabile HEK293T-Zelllinien mit induzierbarer Überexpression von DROSHA E1147K, und stabile embryonale Mausstammzelllinien mit induzierbarer Überexpression von DGCR8 E518K. Um die in WT beobachtete homozygote Expression nachzustellen, wurden für die Erzeugung der DGCR8-Zelllinien Mausstammzellen mit einem DGCR8-Knockout verwendet. Die induzierbare Überexpression von Wildtyp oder mutiertem DROSHA in HEK293T-Zellen zeigte, dass DROSHA E1147K zu einer globalen Herunterregulierung der miRNA-Expression führt. Es wurde zuvor gezeigt, dass der Knockout von DGCR8 in Mausstammzellen ebenfalls zu einer signifikanten Herunterregulierung kanonischer miRNAs führt. Die induzierbare Überexpression von Wildtyp DGCR8 konnte diesen Prozessierungsdefekt aufheben. DGCR8 E518K führte dagegen nur zu einer partiellen Behebung des Prozessierungsdefekts. Differenziell exprimierte miRNAs umfassten hierbei Mitglieder aus der stammzellspezifischen ESCC-Familie und der let-7-miRNA-Familie, deren Antagonismus bekanntermaßen eine bedeutende Rolle bei der Regulation von Stammzelleigenschaften spielt. Zusammen mit einer veränderten miRNA-Expression zeigten DGCR8-E518K-Zellen Veränderungen in der Zielgenexpression, welche möglicherweise verschiedene biologische Prozesse beeinflussen können. Wir konnten verringerte Proliferationsraten beobachten, höchstwahrscheinlich aufgrund einer verringerten Lebensfähigkeit der Zellen. DGCR8-E518K schien in der Lage zu sein, den Block des G1-S Übergangs zu überwinden und den Zellzyklusdefekt in DGCR8-KO-Zellen zu beheben, wenn auch nicht in vollem Umfang wie Wildtyp DGCR8. Darüber hinaus schien DGCR8-E518K nicht in der Lage zu sein, die epithelial-mesenchymale Transition (EMT) vollständig blockieren zu können. Embryoid-Körper (EBs) mit der E518K Mutation konnten das Selbsterneuerungsprogramm jedoch noch unterdrücken und somit den Differenzierungsdefekt in DGCR8-KO-Zellen beheben. Zusammenfassend konnte ich zeigen, dass DROSHA E1147K und DGCR8 E518K häufige Mutationen in WT sind und dabei am häufigsten in Hochrisiko-Tumoren vorkommen. Jede dieser Mutationen führte in vitro zu einer veränderten miRNA-Expression, was unsere vorherigen Befunde in Tumorproben bestätigte. Während die DROSHA E1147K Mutation zu einer globalen Herunterregulierung kanonischer miRNAs führte, konnte die DGCR8 E518K Mutation die Aktivität des Mikroprozessorkomplexes größtenteils wiederherstellen, was darauf hindeutet, dass eine teilweise Verringerung der Aktivität oder eine veränderte Spezifität bei der WT-Entstehung kritisch sein könnte. Trotz der signifikanten Unterschiede in den miRNA- und mRNA-Profilen von DGCR8-E518K- und DGCR8 Wildtyp-Zellen, ergab die Funktionsanalyse, dass DGCR8 E518K viele wichtige Zellfunktionen im Knockout wiederherstellen konnte und sich nur geringfügig von der Wildtyp-Situation unterschied. Weitere Studien unter physiologischeren Bedingungen, wie beispielsweise in einem WT-Blastem-Modellsystem, könnten zusätzlich helfen, die in unseren Mausstammzelllinien beobachteten feinen Unterschiede zwischen DGCR8 E518K und DGCR8 Wildtyp besser bewerten zu können. Zusammen mit unseren Erkenntnissen könnten diese Modellsysteme somit dazu beitragen, die Rolle dieser Mikroprozessormutationen bei der WT-Entstehung besser zu verstehen. Indeed, using oligomeric fusion proteins of single chain-encoded TNFR2-specific TNF mutants (scTNF80), expansion of regulatory T cells and therapeutic activity could be demonstrated in various autoinflammatory diseases, including graft-versus-host disease (GvHD), experimental autoimmune encephalomyelitis (EAE) and collagen-induced arthritis (CIA). With the aim to improve the in vivo availability of TNFR2-specific TNF fusion proteins, we used here the neonatal Fc receptor (FcRn)-interacting IgG1 molecule as an oligomerizing building block and generated a new TNFR2 agonist with improved serum retention and superior in vivo activity. Methods Single-chain encoded murine TNF80 trimers (sc(mu)TNF80) were fused to the C-terminus of an in mice irrelevant IgG1 molecule carrying the N297A mutation which avoids/minimizes interaction with Fcγ-receptors (FcγRs). The fusion protein obtained (irrIgG1(N297A)-sc(mu)TNF80), termed NewSTAR2 (New selective TNF-based agonist of TNF receptor 2), was analyzed with respect to activity, productivity, serum retention and in vitro and in vivo activity. STAR2 (TNC-sc(mu)TNF80 or selective TNF-based agonist of TNF receptor 2), a well-established highly active nonameric TNFR2-specific variant, served as benchmark. NewSTAR2 was assessed in various in vitro and in vivo systems. Results STAR2 (TNC-sc(mu)TNF80) and NewSTAR2 (irrIgG1(N297A)-sc(mu)TNF80) revealed comparable in vitro activity. The novel domain architecture of NewSTAR2 significantly improved serum retention compared to STAR2, which correlated with efficient binding to FcRn. A single injection of NewSTAR2 enhanced regulatory T cell (Treg) suppressive activity and increased Treg numbers by > 300% in vivo 5 days after treatment. Treg numbers remained as high as 200% for about 10 days. Furthermore, a single in vivo treatment with NewSTAR2 upregulated the adenosine-regulating ectoenzyme CD39 and other activation markers on Tregs. TNFR2-stimulated Tregs proved to be more suppressive than unstimulated Tregs, reducing conventional T cell (Tcon) proliferation and expression of activation markers in vitro. Finally, singular preemptive NewSTAR2 administration five days before allogeneic hematopoietic cell transplantation (allo-HCT) protected mice from acute GvHD. Conclusions NewSTAR2 represents a next generation ligand-based TNFR2 agonist, which is efficiently produced, exhibits improved pharmacokinetic properties and high serum retention with superior in vivo activity exerting powerful protective effects against acute GvHD. This may make genetic material, potentially including relevant environmental adaptations, rapidly available in a gene pool. Consequently, it has been postulated to be an important mechanism for enabling evolutionary rescue, that is the recovery of threatened populations through rapid evolutionary adaptation to novel environments. However, predicting the likelihood of such evolutionary rescue for individual species remains challenging. Here, we use the example of Zosterops silvanus, an endangered East African highland bird species suffering from severe habitat loss and fragmentation, to investigate whether hybridization with its congener Zosterops flavilateralis might enable evolutionary rescue of its Taita Hills population. To do so, we employ an empirically parameterized individual‐based model to simulate the species' behaviour, physiology and genetics. We test the population's response to different assumptions of mating behaviour and multiple scenarios of habitat change. We show that as long as hybridization does take place, evolutionary rescue of Z. silvanus is likely. Intermediate hybridization rates enable the greatest long‐term population growth, due to trade‐offs between adaptive and maladaptive introgressed alleles. Habitat change did not have a strong effect on population growth rates, as Z. silvanus is a strong disperser and landscape configuration is therefore not the limiting factor for hybridization. Our results show that targeted gene flow may be a promising avenue to help accelerate the adaptation of endangered species to novel environments, and demonstrate how to combine empirical research and mechanistic modelling to deliver species‐specific predictions for conservation planning. Histone acetylations by the NuA4/TIP60 complex are required to enable proper incorporation of the histone variant and to promote the recruitment of other complexes and proteins required for transcription initiation. The second key player in H2A.Z-mediated transcription is the chromatin remodelling complex SWR1, which replaces the canonical histone H2A with its variant. By the time this project started little was known about H2A.Z in the unicellular parasite Trypanosoma brucei. Like in other eukaryotes H2A.Z was exclusively found in the transcription start sites of the polycistronic transcription units where it keeps the chromatin in an open conformation to enable RNA-polymerase II-mediated transcription. Previous studies showed the variant colocalizing with an acetylation of lysine on histone H4 and a methylation of lysine 4 on histone H3. Data indicated that HAT2 is linked to H2A.Z since it is required for acetylation of lyinse 10 on histone H4. A SWR1-like complex and a complex homologous to the NuA4/TIP60 could not be identified yet. This study aimed at identifying a SWR1-like remodelling complex in T. brucei and at identifying a protein complex orthologous to NuA4/TIP60 as well as at answering the question whether HAT2 is part of this complex or not. To this end, I performed multiple mass spectrometry-coupled co-Immunoprecipitation assays with potential subunits of a SWR1 complex, HAT2 and a putative homolog of a NuA4/TIP60 subunit. In the course of these experiments, I was able to identify the TbSWR1 complex. Subsequent cell fractionation and chromatin immunoprecipitation-coupled sequencing analysis experiments confirmed, that this complex is responsible for the incorporation of the histone variant H2A.Z in T. brucei. In addition to this chromatin remodelling complex, I was also able to identify two histone acetyltransferase complexes assembled around HAT1 and HAT2. In the course of my study data were published by the research group of Nicolai Siegel that identified the histone acetyltransferase HAT2 as being responsible for histone H4 acetylation, in preparation to promote H2A.Z incorporation. The data also indicated that HAT1 is responsible for acetylation of H2A.Z. According to the literature, this acetylation is required for proper transcription initiation. Experimental data generated in this study indicated, that H2A.Z and therefore TbSWR1 is involved in the DNA double strand break response of T. brucei. The identification of the specific complex composition of all three complexes provided some hints about how they could interact with each other in the course of transcription regulation and the DNA double strand break response. A proximity labelling approach performed with one of the subunits of the TbSWR1 complex identified multiple transcription factors, PTM writers and proteins potentially involved in chromatin maintenance. Overall, this work will provide some interesting insights about the composition of the complexes involved in H2A.Z incorporation in T. brucei. Furthermore, it is providing valuable information to set up experiments that could shed some light on RNA-polymerase II-mediated transcription and chromatin remodelling in T. brucei in particular and Kinetoplastids in general. N2 - Die Histonvariante H2A.Z ist ein Schlüsselelement bei der Transkriptionsregulation in Eukaryoten. Histonacetylierungen die vom NuA4/Tip60 Komplex prozessiert werden, sind für den korrekten Einbau der Variante unerlässlich. Darüber hinaus erlauben diese posttranslationellen Modifikationen die Rekrutierung weiterer Proteine und Komplexe die für die Transkription notwendig sind. Ein weiteres Schlüsselelement der mittels H2A.Z regulierten Transkription ist der Komplex zur Umstrukturierung des Chromatins SWR1, welcher das kanonische Histon H2A gegen seine Variante austauscht. Zu Beginn dieses Projektes war der Wissenstand bezüglich der Histonvariante H2A.Z in dem einzelligen Parasiten Trypanosoma brucei limitiert. Wie in anderen eukaryotischen Organismen wurde die Variante ausschließlich an den Startpunkten der polyzistronischen Transkriptionseinheiten gefunden, an denen es für die Öffnung des Chromatins verantwortlich ist und so die Transkription mittels RNAPolymerase II ermöglicht. Vorangegangene Studien konnten zeigen, dass die Variante mit einer Acetylierung des Lysins 10 im Histon H4 und einer Methylierung des Lysins 4 im Histon H3 co-lokalisiert. Einige Daten lieferten den Hinwies, dass die Histon-Acetyltransferase HAT2 mit H2A.Z in Zusammenhang steht, da diese die Acetylierung des Lysins 10 im Hinston H4 prozessiert. Komplexe die in ihrer Funktion dem SWR1 oder dem NuA4/TIP60 Komplex entsprechen, konnten bisher noch nicht gefunden werden. Die vorliegende Arbeit zielt darauf ab Komplexe zu identifizieren, die in ihrer Funktion dem SWR1 sowie dem NuA4/TIP60 Komplex entsprechen. Zudem soll die Frage geklärt werden ob HAT2 Teil eines möglichen NuA4/TIP60 Komplexes ist. In diesem Zusammenhang habe ich mehrere Massenspektrometrie gekoppelte Co-Immunopräzipitationen mit potenziellen Untereinheiten eines SWR1 Komplexes sowie HAT2 und einem Protein welches otholog zu einer NuA4/TIP60 Untereinheit ist, durchgeführt. Im Verlauf dieser Experimente konnte der SWR1 Komplex in T. brucei (TbSWR1) identifiziert werden. Anschließende Zellfraktionierungen sowie Chromatin Immunopräzipitationen gekoppelte Sequenzanalysen konnten bestätigen, dass der identifizierte Komplex für den Einbau der Histonvariante H2A.Z zuständig ist. Darüber hinaus konnten neben diesem Komplex noch zwei weitere Komplexe identifiziert werden, die jeweils die Histonacetyltransferasen HAT1 und HAT2 als Kernkomponenten enthalten. Im Verlauf meiner Arbeit wurden von der Arbeitsgruppe von Nicolai Siegel Daten publiziert die zeigten, dass die Histonacetyltransferase HAT2, in Vorbereitung auf den Einbau von H2A.Z, für die Acetylierung des Histons H4 verantwortlich ist. Im Gegenzug ist HAT1 für die Acetylierung von H2A.Z notwendig, welche wiederum für die korrekte Initiation der Transkription benötigt wird. Damit entspricht die Funktion der Acetylierung von H2A.Z in T. brucei der in der Literatur beschriebenen Funktion. Experimentelle Daten die im Verlauf dieser Arbeit generiert wurden, lieferten einen Hinweis darauf, dass H2A.Z auch an der Reparatur von DNS Doppelstrangbrüchen beteiligt ist. Die Aufschlüsselung der spezifischen Zusammensetzung aller drei Komplexe gab einige Hinweise darauf, wie sie sowohl während der Transkriptionsregulation als auch der Reparatur von DNS Doppelstrangbrüchen miteinander interagieren. Im Zuge einer molekularen Umgebungskartierung, die mit einer der Untereinheiten des TbSWR1 Komplexes durchgeführt wurde, konnten mehrere Transkriptionsfaktoren und Enzyme zur Histonmodifizierung identifiziert werden. Dabei wurden auch einige Proteine identifiziert, welche möglicherweise mit der Umformung des Chromatins in Zusammenhang stehen. Abschließend ist festzuhalten, dass diese Arbeit einige äußerst interessante Einsichten über die Zusammensetzung der Komplexe, die am H2A.Z Einbau in T. brucei beteiligt sind, liefern konnte. Darüber hinaus stellt sie einige wertvolle Informationen zur Verfügung. Diese könnten zur gezielten Planung von Experimenten genutzt werden, um mehr über RNA-Polymerase II vermittelte Transkription und Chromatin Umstrukturierung in T. brucei im speziellen und in Kinetoplastiden im Allgemeinen zu erfahren. A key player in this cascade is the chromatin remodelling complex SWR1, which replaces the canonical histone H2A with its variant H2A.Z. Transcriptional regulation of polycistronic transcription units in the unicellular parasite Trypanosoma brucei has been shown to be highly dependent on acetylation of H2A.Z, which is mediated by the histone-acetyltransferase HAT2. The chromatin remodelling complex which mediates H2A.Z incorporation is not known and an SWR1 orthologue in trypanosomes has not yet been reported. In this study, we identified and characterised an SWR1-like remodeller complex in T. brucei that is responsible for Pol II-dependent transcriptional regulation. Bioinformatic analysis of potential SNF2 DEAD/Box helicases, the key component of SWR1 complexes, identified a 1211 amino acids-long protein that exhibits key structural characteristics of the SWR1 subfamily. Systematic protein-protein interaction analysis revealed the existence of a novel complex exhibiting key features of an SWR1-like chromatin remodeller. RNAi-mediated depletion of the ATPase subunit of this complex resulted in a significant reduction of H2A.Z incorporation at transcription start sites and a subsequent decrease of steady-state mRNA levels. Furthermore, depletion of SWR1 and RNA-polymerase II (Pol II) caused massive chromatin condensation. The potential function of several proteins associated with the SWR1-like complex and with HAT2, the key factor of H2A.Z incorporation, is discussed. It contains large amounts of carbohydrates besides variable concentrations of amino acids and thus represents an important food source for many pollinators. Its nutrient content and composition can, however, strongly vary within and between plant species. The factors driving this variation in nectar quality are still largely unclear. We investigated factors underlying interspecific variation in macronutrient composition of floral nectar in 34 different grassland plant species. Specifically, we tested for correlations between the phylogenetic relatedness and morphology of plants and the carbohydrate (C) and total amino acid (AA) composition and C:AA ratios of nectar. We found that compositions of carbohydrates and (essential) amino acids as well as C:AA ratios in nectar varied significantly within and between plant species. They showed no clear phylogenetic signal. Moreover, variation in carbohydrate composition was related to family‐specific structural characteristics and combinations of morphological traits. Plants with nectar‐exposing flowers, bowl‐ or parabolic‐shaped flowers, as often found in the Apiaceae and Asteraceae, had nectar with higher proportions of hexoses, indicating a selective pressure to decelerate evaporation by increasing nectar osmolality. Our study suggests that variation in nectar nutrient composition is, among others, affected by family‐specific combinations of morphological traits. However, even within species, variation in nectar quality is high. As nectar quality can strongly affect visitation patterns of pollinators and thus pollination success, this intra‐ and interspecific variation requires more studies to fully elucidate the underlying causes and the consequences for pollinator behaviour. T1 - Trabeculopuncture as a predictive test of distal outflow resistance in canal-based surgery JF - Scientific Reports N2 - We investigated whether trabeculopuncture (TP) could detect distal outflow resistance to predict the outcome of canal-based glaucoma surgery such as ab interno trabeculectomy (AIT). These procedures have a high utilization in open angle glaucoma, but fail in eyes with an unidentified distal outflow resistance. We assigned 81 porcine eyes to two groups: trial (n = 42) and control (n = 39). At 24 h, four YAG-laser trabeculopunctures were placed nasally, followed by a 180° AIT at the same site at 48 h. The proportion of TP responders between both AIT groups was compared. Histology and outflow canalograms were determined. Both post-TP and post-AIT IOPs were lower than baseline IOP (p = 0.015 and p < 0.01, respectively). The success rates of TP and AIT were 69% and 85.7%, respectively. Sensitivity and specificity values of TP as predictive test for AIT success were 77.7% and 83.3%, respectively. The positive and negative predictive values were 96.6% and 38.5%, respectively. We conclude that a 10% reduction in IOP after TP can be used as a predictor for the success (> 20% IOP decrease) of 180° AIT in porcine eyes. We concluded that a 10% reduction in IOP after TP can be used to predict the success (>20% IOP decrease) of AIT in porcine eyes. As porcine eyes share many similarities with human eyes, our findings may have implications on the validity of this test as a predictor for surgical outcomes of AITs in humans. N2 - Wir untersuchten die Trabekulopunktion (TP) zur Vorhersage des Erfolgs einer Ab Interno Trabekulektomie (AIT) an einem ex vivo Schweineaugen-Perfusionsmodell. In this study, our aim was to investigate the putative differential expression of miRNAs in adrenal tissues from different forms of Cushing's syndrome (CS). For this, miRNA-based next-generation sequencing was performed in adrenal tissues taken from patients with ACTH-independent cortisol-producing adrenocortical adenomas (CPA), from patients with ACTH-dependent pituitary Cushing's disease (CD) after bilateral adrenalectomy, and from control subjects. A confirmatory QPCR was also performed in adrenals from patients with other CS subtypes, such as primary bilateral macronodular hyperplasia and ectopic CS. Sequencing revealed significant differences in the miRNA profiles of CD and CPA. QPCR revealed the upregulated expression of miR-1247-5p in CPA and PBMAH (log2 fold change > 2.5, p < 0.05). MiR-379-5p was found to be upregulated in PBMAH and CD (log2 fold change > 1.8, p < 0.05). Analyses of miR-1247-5p and miR-379-5p expression in the adrenals of mice which had been exposed to short-term ACTH stimulation showed no influence on the adrenal miRNA expression profiles. For miRNA-specific target prediction, RNA-seq data from the adrenals of CPA, PBMAH, and control samples were analyzed with different bioinformatic platforms. The analyses revealed that both miR-1247-5p and miR-379-5p target specific genes in the WNT signaling pathway. In conclusion, this study identified distinct adrenal miRNAs as being associated with CS subtypes. A1 - Thorn, Simon T1 - The effect of natural disturbances on forest biodiversity: an ecological synthesis JF - Biological Reviews N2 - Disturbances alter biodiversity via their specific characteristics, including severity and extent in the landscape, which act at different temporal and spatial scales. Biodiversity response to disturbance also depends on the community characteristics and habitat requirements of species. Untangling the mechanistic interplay of these factors has guided disturbance ecology for decades, generating mixed scientific evidence of biodiversity responses to disturbance. Understanding the impact of natural disturbances on biodiversity is increasingly important due to human‐induced changes in natural disturbance regimes. In many areas, major natural forest disturbances, such as wildfires, windstorms, and insect outbreaks, are becoming more frequent, intense, severe, and widespread due to climate change and land‐use change. Conversely, the suppression of natural disturbances threatens disturbance‐dependent biota. Using a meta‐analytic approach, we analysed a global data set (with most sampling concentrated in temperate and boreal secondary forests) of species assemblages of 26 taxonomic groups, including plants, animals, and fungi collected from forests affected by wildfires, windstorms, and insect outbreaks. The overall effect of natural disturbances on α‐diversity did not differ significantly from zero, but some taxonomic groups responded positively to disturbance, while others tended to respond negatively. Disturbance was beneficial for taxonomic groups preferring conditions associated with open canopies (e.g. hymenopterans and hoverflies), whereas ground‐dwelling groups and/or groups typically associated with shady conditions (e.g. epigeic lichens and mycorrhizal fungi) were more likely to be negatively impacted by disturbance. Across all taxonomic groups, the highest α‐diversity in disturbed forest patches occurred under moderate disturbance severity, i.e. with approximately 55% of trees killed by disturbance. We further extended our meta‐analysis by applying a unified diversity concept based on Hill numbers to estimate α‐diversity changes in different taxonomic groups across a gradient of disturbance severity measured at the stand scale and incorporating other disturbance features. We found that disturbance severity negatively affected diversity for Hill number q = 0 but not for q = 1 and q = 2, indicating that diversity–disturbance relationships are shaped by species relative abundances. Our synthesis of α‐diversity was extended by a synthesis of disturbance‐induced change in species assemblages, and revealed that disturbance changes the β‐diversity of multiple taxonomic groups, including some groups that were not affected at the α‐diversity level (birds and woody plants). Finally, we used mixed rarefaction/extrapolation to estimate biodiversity change as a function of the proportion of forests that were disturbed, i.e. the disturbance extent measured at the landscape scale. The comparison of intact and naturally disturbed forests revealed that both types of forests provide habitat for unique species assemblages, whereas species diversity in the mixture of disturbed and undisturbed forests peaked at intermediate values of disturbance extent in the simulated landscape. Hence, the relationship between α‐diversity and disturbance severity in disturbed forest stands was strikingly similar to the relationship between species richness and disturbance extent in a landscape consisting of both disturbed and undisturbed forest habitats. This result suggests that both moderate disturbance severity and moderate disturbance extent support the highest levels of biodiversity in contemporary forest landscapes. These assembled complexes are usually very large, consisting of 10 subunits on an average and include either proteins alone, or proteins and nucleic acids. Consequently, such macromolecular assemblies rather than individual biopolymers perform the vast majority of cellular activities. The faithful assembly of such molecular assemblies is often aided by trans-acting factors in vivo, to preclude aggregation of complex components and/or non-cognate interactions. A paradigm for an assisted assembly of a macromolecular machine is the formation of the common Sm/LSm core of spliceosomal and histone-mRNA processing U snRNPs. The key assembly factors united in the Protein Arginine Methyltransferase 5 (PRMT5) and the Survival Motor Neuron (SMN) complexes orchestrate the assembly of the Sm/LSm core on the U snRNAs. Assembly is initiated by the PRMT5-complex subunit pICln, which pre-arranges the Sm/LSm proteins into spatial positions occupied in the mature U snRNPs. The SMN complex subsequently binds these Sm/LSm units, displaces pICln and catalyses the Sm ring closure on the Sm-site of the U snRNA. The SMN complex consists of the eponoymous SMN protein linked in a modular network of interactions with eight other proteins, termed Gemins 2-8 and Unrip. Despite functional and structural characterisation of individual protein components and/or sub-complexes of this assembly machinery, coherent understanding of the structural framework of the core SMN complex remained elusive. The current work, employing a combined approach of biochemical and structural studies, aimed to contribute to the understanding of how distinct modules within the SMN complex coalecse to form the macromolecular SMN complex. A novel atomic resolution (1.5 Å) structure of the human Gemin8:7:6 sub-complex, illustrates how the peripheral Gemin7:6 module is tethered to the SMN complex via Gemin8’s C-terminus. In this model, Gemin7 engages with both Gemin6 and Gemin8 via the N- and C-termini of its Sm-fold like domain. This highly conserved interaction mode is reflected in the pronounced sequence conservation and identical biochemical behaviour of similar sub-complexes from divergent species, namely S. pombe and C. elegans. Despite lacking significant sequence similarity to the Sm proteins, the dimeric Gemin7:6 complex share structural resemblance to the Sm heteromers. The hypothesis that the dimeric Gemin7:6 functions as a Sm-surrogate during Sm core assembly could not be confirmed in this work. The functional relevance of the structural mimicry of the dimeric Gemin7:6 sub-complex with the Sm heterodimers therefore still remains unclear. Reduced levels of functional SMN protein is the cause of the devastating neurodegenerative disease, Spinal Muscular Atrophy (SMA). The C-terminal YG-zipper motif of SMN is a major hot-spot for most SMA patient mutations. In this work, adding to the existing inventory of the human and fission yeast YG-box models, a novel 2.2 Å crystal structure of the nematode SMN’s YG-box domain adopting the glycine zipper motif has been reported. Furthermore, it could be assessed that SMA patient mutations mapping to this YG-box domain greatly influences SMN’s self-association competency, a property reflected in both the human and nematode YG-box biochemical handles. The shared molecular architecture and biochemical behaviour of the nematode SMN YG-box domain with its human and fission yeast counterparts, reiterates the pronounced conservation of this oligomerisation motif across divergent organisms. Apart from serving as a multimerization domain, SMN’s YG-box also acts as interaction platform for Gemin8. A systematic investigation of SMA causing missense mutations uncovered that Gemin8’s incorporation into the SMN complex is influenced by the presence of certain SMA patient mutations, albeit independent of SMN’s oligomerisation status. Consequently, loss of Gemin8 association in the presence of SMA patient mutations would also affect the incorporation of Gemin7:6 sub-complex. Gemin8, therefore sculpts the heteromeric SMN complex by bridging the Gemin7:6 and SMN:Gemin2 sub-units, a modular feature shared in both the human and nematode SMN complexes. These findings provide an important foundation and a prospective structural framework for elucidating the core architecture of the SMN complex in the ongoing Cryo-EM studies. N2 - Systematische Untersuchungen von zellulären Bestandteilen haben gezeigt, dass viele Proteine nicht isoliert, sondern vielmehr in modularen Komplexen organisiert vorliegen. Mit durchschnittlich zehn Untereinheiten sind diese Komplexe sehr groß, wobei sie entweder ausschließlich aus Proteinen oder aber aus Proteinen und Nukleinsäuren bestehen können. Daher wird der Großteil zellulärer Aktivitäten nicht von einzelnen Biopolymeren, sondern von makromolekularen Komplexen verrichtet. Die Zusammenlagerung dieser Komplexe wird in vivo häufig von Hilfsfaktoren unterstützt, um die Aggregation der Einzelkomponenten und/oder unspezifische Wechselwirkungen zu verhindern. Ein Beispiel für eine derartige Zusammenlagerungshilfe ist die Bildung des Sm/LSm-Cores der mRNA-prozessierenden U snRNPs. Dabei wird die Anlagerung von Sm/LSm Proteinen an die U snRNAs durch eine Anzahl von Hilfsfaktoren orchestriert, die in Protein-Arginin-Methyltransferase 5 (PRMT5)- und dem Survival Motor Neuron (SMN)-Komplexen organisiert sind. Die Zusammenlagerung wird durch die PRMT5-Untereinheit pICln initiiert, die die räumliche Anordnung von Sm/LSm-Proteinen in höher-geordneten Komplexen stabilisiert. Diese werden anschließend auf den SMN-Komplex übertragen, wobei pICln verdrängt und die Verbindung mit der Sm-Seite der U snRNA sichergestellt wird. Der SMN-Komplex besteht aus dem SMN-Protein, das in einem modularen Netzwerk mit acht weiteren Proteinen (Gemins 2-8 und Unrip) interagiert. Auch wenn funktionale und strukturelle Charakterisierungen einzelner Proteinkomponenten und Module dieser Zusammenlagerungs-Maschinerie vorliegen, steht ein tiefergehendes Verständnis des strukturellen Organisation des Gesamt-Komplexes noch aus. In der vorliegenden Arbeit sollte unter Anwendung biochemischer und struktureller Techniken ein Beitrag dazu geleistet werden, die Interaktionen der verschiedenen Komponenten innerhalb des SMN-Komplexes zu verstehen, die so die dreidimensionale Organisation des SMN-Komplexes zu verstehen. Eine neuartige Kristallstruktur des humanen Gemin8:7:6-Subkomplexes bei einer Auflösung von 1.5 Å zeigt, wie der periphere Gemin7:6-Abschnitt durch den C-Terminus von Gemin8 zum SMN-Komplex dirigiert wird. In diesem Modell interagiert Gemin7 sowohl mit Gemin6 als auch Gemin8 über den N- und C-Terminus der Sm-ähnlichen Domäne. Die Annahme, der dimere Gemin7:6-Subkomplex würde als Hilfsfaktor über die direkte Interaktion mit Sm-Proteinen fungieren konnte in der vorliegenden Arbeit nicht bestätigt werden. These declines are more pronounced in areas that are currently biodiversity rich, but economically poor – essentially describing many tropical regions in the Global South where landscapes are dominated by smallholder agriculture. Agriculture is an important driver of biodiversity decline, through habitat destruction and unsustainable practices. Ironically, agriculture itself is dependent on a range of ecosystem services, such as pollination and pest control, provided by biodiversity. Biodiversity on fields and the delivery of ecosystem services to crops is often closely tied to the composition of the surrounding landscape – complex landscapes with a higher proportion of (semi-)natural habitats tend to support a high abundances and biodiversity of pollinators and natural enemies that are beneficial to crop production. However, past landscape scale studies have focused primarily on industrialized agricultural landscapes in the Global North, and context dependent differences between regions and agricultural systems are understudied. Smallholder agriculture supports 2 billion people worldwide and contributes to over half the world’s food supply. Yet smallholders, particularly in sub-Saharan Africa, are underrepresented in research investigating the consequences of landscape change and agricultural practices. Where research in smallholder agriculture is conducted, the focus is often on commodity crops, such as cacao, and less on crops that are directly consumed by smallholder households, though the loss of services to these crops could potentially impact the most vulnerable farmers the hardest. Agroecology – a holistic and nature-based approach to agriculture, provides an alternative to unsustainable input-intensive agriculture. Agroecology has been found to benefit smallholders through improved agronomical and food-security outcomes. Co-benefits of agroecological practices with biodiversity and ecosystem services are assumed, but not often empirically tested. In addition, the local and landscape effects on biodiversity and ecosystem services are more commonly studied in isolation, but their potentially interactive effects are so far little explored. Our study region in northern Malawi exemplifies many challenges experienced by smallholder farmers throughout sub-Saharan Africa and more generally in the Global South. Malawi is located in a global biodiversity hotspot, but biodiversity is threatened by rapid habitat loss and a push for input-intensive agriculture by government and other stakeholders. In contrast, agroecology has been effectively promoted and implemented in the study region. We investigated how land-use differences and the agroecological practices affects biodiversity and ecosystem services of multiple taxa in a maize-bean intercropping system (Chapter 2), and pollination of pumpkin (Chapter 3) and pigeon pea (Chapter 4). Additionally, the effects of local and landscape scale shrub- to farmland habitat conversion was investigated on butterfly communities, as well as the potential for agroecology to mitigate these effects (Chapter 5). Biodiversität und Ökosystemdienstleistungen auf Feldern werden stark durch die umliegende Landschaft beeinflusst - komplexe Landschaften mit einen großen Anteil (halb-)natürlicher haben in der Regel höhere Abudanzen und eine größere Biodiversität von Bestäubern und natürlichen Feinden die vorteilhaft für die landwirtschaftliche Produktion sind. Forschung auf Landschaftebene hat bisher jedoch vorrangig auf die industrialisierte Landwirtschaft in z.B. Europa oder die USA fokussiert und kontextabhängige Unterschiede zwischen Regionen und landwirtschaftlichen Systemen sind nicht ausreichend studiert..Weltweit sind etwa 2 Milliarden Menschen von Subsistenzlandwirtschaft abhängig. Jedoch sind diese Kleinbauern, in der Forschung über die Konsequenzen von Landnutzung und landwirtschaftlichen Managements auf Biodiversität und Ökosystemdienstleistungen unterrepräsentiert, insbesondere Kleinbauern aus Subsahara-Afrika. Die wenigen verfügbaren Studien legend den Fokus oft auf wirtschaftlich wichtige Kulturpflanzen, wie etwa Kakao, und selten auf Kulturpflanzen, die für Ernährungssicherheit der Kleinbauern wichtig sind, obwohl der Verlust der Ökosystemdienstleistungen diese möglicherweise am härtesten trifft. Agroökologie ist eine nachhaltigere Form des landwirtschaftlichen Managements als die konventionelle Landwirtschaft, und will den Einsatz von Agrochemie zu reduzieren und eine holistische Landwirtschaft fördern. Agroökologie steigert die Ernährungssicherheit von Kleinbauern, insbesondere wenn die Bauern viele verschiedene agroökologische Verfahren nutzen. Unsere Studienregion in Nord Malawi spiegelt die viele Herausforderungen der afrikanischen Zusammenfassung Subsistenzlandwirtschaft wider. Malawi liegt in einem Biodiversitäts-Hotspot, jedoch ist diese Biodiversität durch einen schnellen Rückgang natürlicher Lebensräume und durch die Intensivierung der Landwirtschaft stark gefährdet. Dem gegenüber stehen erfolgreicher Ausbau und Umsetzung von Agroökologie in der Region. Das gab mir die Möglichkeit, die Effekte von Landnutzung und Agroökologie auf Biodiversität und Ökosystemdienstleistungen in Malawi zu untersuchen. Dafür habe ich in Mais und Bohnen in Einzel- und Mischkultur 7 taxonomische Gruppen die verschiedene Ökosystemdienstleistungen erbringen erfasst (Kapitel 2) sowie Bestäuber und Bestäubung auf Kürbis (Kapitel 3) und Straucherbsen studiert (Kapitel 4). Zusätzlich habe ich an Schmetterlingen die Effekte von Lebensraumverlust auf der lokalen und auf Landschaftsebene studiert, und untersucht, ob Agroökologie potenziell negative Effekte mindern kann (Kapitel 5). Here we report a method named COMPASS (Critical-Offset-Magnetic-Particle-SpectroScopy), which is based on a critical offset magnetic field, enabling sensitive detection to minimal changes in mobility of MNP ensembles, e.g., resulting from SARS-CoV-2 antibodies binding to the S antigen on the surface of functionalized MNPs. With a sensitivity of 0.33 fmole/50 µl (≙7 pM) for SARS-CoV-2-S1 antibodies, measured with a low-cost portable COMPASS device, the proposed technique is competitive with respect to sensitivity while providing flexibility, robustness, and a measurement time of seconds per sample. Constantly growing and changing demands have led to an enormous decline in ecological key elements and a structural homogenization of most forests. These changes have been accompanied by widespread declines of many forest-dwelling and especially saproxylic, i.e. species depending on deadwood. In order to counteract this development, various conservation strategies have been developed, but they primarily focus on a quantitative deadwood enrichment. However, the diversity of saproxylic species is furthermore driven by a variety of abiotic and biotic determinants as well as interactions between organisms. A detailed understanding of these processes has so far been largely lacking. The aim of the present thesis was therefore to improve the existing ecological knowledge of determinants influencing saproxylic species and species communities in order to provide the basis for evidence-based and adapted conservation measures. In chapter II of this thesis, I first investigated the impact of sun exposure, tree species, and their combination on saproxylic beetles, wood-inhabiting fungi, and spiders. Therefore, logs and branches of six tree species were set up under different sun exposures in an experimental approach. The impact of sun exposure and tree species strongly differed among single saproxylic taxa as well as diameters of deadwood. All investigated taxa were affected by sun exposure, whereby sun exposure resulted in a higher alpha-diversity of taxa recorded in logs and a lower alpha-diversity of saproxylic beetles reared from branches compared to shading by canopy. Saproxylic beetles and wood-inhabiting fungi as obligate saproxylic species were additionally affected by tree species. In logs, the respective impact of both determinants also resulted in divergent community compositions. Finally, a rarefaction/extrapolation method was used to evaluate the effectiveness of different combinations of tree species and sun exposure for the conservation of saproxylic species diversity. Based on this procedure, a combination of broadleaved and coniferous as well as hard- and softwood tree species was identified to support preferably high levels of saproxylic species diversity. The aim of chapter III was to evaluate the individual conservational importance of tree species for the protection of saproxylic beetles. For this, the list of tree species sampled for saproxylic beetles was increased to 42 different tree species. The considered tree species represented large parts of taxonomic and phylogenetic diversity native to Central Europe as well as the most important non-native tree species of silvicultural interest. Freshly cut branches were set up for one year and saproxylic beetles were reared afterwards for two subsequent years. The study revealed that some tree species, in particular Quercus sp., host a particular high diversity of saproxylic beetles, but tree species with a comparatively medium or low overall diversity were likewise important for red-listed saproxylic beetle species. Compared to native tree species, non-native tree species hosted a similar overall species diversity of saproxylic beetles but differed in community composition. In chapter IV, I finally analysed the interactions of host beetle diversity and the diversity of associated parasitoids by using experimentally manipulated communities of saproxylic beetles and parasitoid Hymenoptera as a model system. Classical approaches of species identification for saproxylic beetles were combined with DNA-barcoding for parasitoid Hymenoptera. The diversity of the host communities was inferred from their phylogenetic composition as well as differences in seven functional traits. Abundance, species richness, and Shannon-diversity of parasitoid Hymenoptera increased with increasing host abundance. However, the phylogenetic and functional dissimilarity of host communities showed no influence on the species communities of parasitoid Hymenoptera. The results clearly indicate an abundance-driven system in which the general availability, not necessarily the diversity of potential hosts, is decisive. In summary, the present thesis corroborates the general importance of deadwood heterogeneity for the diversity of saproxylic species by combining different experimental approaches. In order to increase their efficiency, conservation strategies for saproxylic species should generally promote deadwood from different tree species under different conditions of sun exposure on landscape-level in addition to the present enrichment of a certain deadwood amount. The most effective combinations of tree species should consider broadleaved and coniferous as well as hard- and softwood tree species. Furthermore, in addition to dominant tree species, special attention should be given to native, subdominant, silviculturally unimportant, and rare tree species. N2 - Während der letzten Jahrhunderte hat die anthropogene Nutzung das Erscheinungsbild der Waldökosysteme in Europa grundlegend verändert. Stetig wachsende und wandelnde Ansprüche führten zu einem enormen Rückgang ökologischer Schlüsselelemente und einer strukturellen Homogenisierung der meisten Wälder. In der Folge kam es zu Rückgängen vieler waldbewohnender und insbesondere xylobionter, d.h. von Totholz abhängigen, Arten. Um dieser Entwicklung entgegenzuwirken, wurden verschiedene Schutzstrategien entwickelt, welche jedoch vor allem auf eine quantitative Totholzanreicherung abzielen. Die Vielfalt xylobionter Arten wird aber weiterhin durch unterschiedliche abiotische und biotische Einflussfaktoren sowie durch Wechselwirkungen zwischen den Arten beeinflusst. Ein detailliertes Verständnis der genauen Vorgänge fehlt jedoch bislang größtenteils. Ziel der vorliegenden Promotionsarbeit war es deshalb, das diesbezüglich bestehende Wissen zu verbessern, um die Basis für evidenzbasierte und angepasste Naturschutzmaßnahmen zu schaffen. In Kapitel II dieser Arbeit habe ich zunächst den Einfluss der Besonnung und Baumart sowie deren Kombination im Vergleich auf xylobionte Käfer, holzbesiedelnde Pilze und Spinnen untersucht. Für die zugehörige Studie wurden dabei Stämme und Äste von sechs Baumarten bei unterschiedlicher Besonnung in einem experimentellen Ansatz ausgebracht. Der Einfluss der Besonnung und Baumart unterschied sich deutlich zwischen den einzelnen Artengruppen und Totholzdurchmessern. Alle Artengruppen wurden durch die Besonnung beeinflusst, wobei Besonnung im Vergleich zur Beschattung durch Baumkronen bei allen Artengruppen an Stämmen zu einer höheren alpha-Diversität führte und zu einer niedrigeren alpha-Diversität von xylobionten Käfern in Ästen. Xylobionte Käfer und holzbesiedelnde Pilze als obligat xylobionte Arten wurden weiterhin von der Baumart beeinflusst. Für die Artengruppen an Stämmen führten die jeweiligen Auswirkungen von Besonnung und Baumarten ebenfalls zu Unterschieden in der Zusammensetzung der Artgemeinschaften. Abschließend wurden Art-Akkumulationskurven genutzt, um die Effektivität unterschiedlicher Kombinationen aus Baumart und Besonnung für den Erhalt der xylobionten Diversität zu evaluieren. Um eine möglichst hohe Artenvielfalt zu fördern, wurde darauf basierend eine Kombination aus Laub- und Nadelholz einschließlich Weich- und Hartholzarten identifiziert. Ziel meiner Studie in Kapitel III war es den individuellen Beitrag einzelner Baumarten zum Schutz xylobionter Käfer zu identifizieren. Dafür wurde die Zahl untersuchter Baumarten auf 42 erhöht. Die untersuchten Baumarten umfassten dabei große Teile der taxonomischen und phylogenetischen Diversität, die in Mitteleuropa heimisch ist, sowie die wichtigsten, nicht-heimischen Baumarten von waldbaulichem Interesse. Frisch geschnittene Äste wurden für ein Jahr ausgebracht und xylobionte Käfer im Anschluss für zwei aufeinanderfolgende Jahre ausgezüchtet. Im Rahmen der Studie konnte gezeigt werden, dass einige Baumarten, insbesondere Quercus sp., eine besonders hohe Artenvielfalt aufweisen, aber auch Arten mit einer vergleichsweise geringen Gesamtartenzahl für Arten der Roten Liste von Bedeutung sind. Nicht-heimische Baumarten beherbergten insgesamt keine geringere Artenvielfalt von xylobionten Käfern, unterschieden sich aber in der Zusammensetzung ihrer Artgemeinschaften. Die Studie in Kapitel IV analysiert schließlich die Wechselwirkungen zwischen Wirtsdiversität und der Diversität assoziierter Parasitoide unter Verwendung experimentell manipulierter Gemeinschaften von xylobionten Käfern und parasitoiden Hymenopteren als Modellsystem. Klassische Ansätze zur Artidentifizierung für xylobionte Käfer wurden dabei mit DNA-Barcoding für die parasitoiden Hymenopteren kombiniert. Die Vielfalt der Wirtsgemeinschaften wurde aus ihrer phylogenetischen Zusammensetzung sowie Unterschieden in sieben funktionellen Merkmalen abgeleitet. Abundanz, Artenvielfalt und Shannon-Diversität nahmen mit zunehmender Abundanz der Wirte zu. Hingegen zeigten die phylogenetische und funktionelle Ähnlichkeit der Wirtsgemeinschaften insgesamt keinen Einfluss auf die Artgemeinschaften der parasitoiden Hymenopteren. Die Ergebnisse weisen damit klar auf ein abundanz-getriebenes System hin, in dem die generelle Verfügbarkeit und nicht unbedingt die Diversität potentieller Wirte entscheidend ist. Zusammenfassend betont die vorliegende Promotionsarbeit durch die Kombination verschiedener experimenteller Ansätze die generelle Bedeutung der Totholzheterogenität für die Vielfalt xylobionter Arten. Um ihre Effizienz zu steigern, sollten Schutzstrategien für xylobionte Arten neben einer bestimmten Totholzmenge daher generell Totholz verschiedener Baumarten bei unterschiedlicher Besonnung auf Landschaftsebene anreichern. Die effektivsten Baumarten-Kombinationen sollten dabei Laub- und Nadelholz sowie Weich- und Hartholzarten berücksichtigen. Neben den dominierenden Baumarten sollte zudem ein besonderes Augenmerk auf heimischen, subdominanten, wirtschaftlich irrelevanten und seltenen Baumarten liegen. Methods Observational retrospective study of APERIO® (AP) vs. new generation APERIO® Hybrid (APH) (Acandis®, Pforzheim, Germany) stent-retriever device (01/2019–09/2020) for mechanical thrombectomy (MT) in large vessel occlusion (LVO) stroke. Primary effectiveness endpoint was successful recanalization eTICI (expanded Thrombolysis In Cerebral Ischemia) ≥ 2b67, primary safety endpoint was occurrence of hemorrhagic complications after MT. Secondary outcome measures were time from groin puncture to first pass and successful reperfusion, and the total number of passes needed to achieve the final recanalization result. Results A total of 298 patients with LVO stroke who were treated by MT matched the inclusion criteria: 148 patients (49.7%) treated with AP vs. 150 patients (50.3%) treated with new generation APH. Successful recanalization was not statistically different between both groups: 75.7% for AP vs. 79.3% for APH; p = 0.450. Postinterventional hemorrhagic complications and particularly subarachnoid hemorrhage as the entity possibly associated with stent-retriever device type was significantly less frequent in the group treated with the APH: 29.7% for AP and 16.0% for APH; p = 0.005; however, rates of symptomatic hemorrhage with clinical deterioration and in domo mortality were not statistically different. Neither the median number of stent-retriever passages needed to achieve final recanalization, time from groin puncture to first pass, time from groin puncture to final recanalization nor the number of cases in which successful recanalization could only be achieved by using a different stent-retriever as bail-out device differed between both groups. Conclusion In the specific example of the APERIO® stent-retriever device, we observed that further technological developments of the new generation device were not associated with disadvantages with respect to typical observational indicators of safety or effectiveness. However, usually there is no direct comparison between the different restriction forms. Additionally, many cell culture experiments take place under static conditions. In this work, we used a closed perfusion culture in murine L929 cells over a period of 7 days to compare methionine restriction (MetR) and glucose restriction (LowCarb) in the same system and analysed the metabolome by liquid chromatography mass spectrometry (LC-MS). In addition, we analysed the inhibition of glycolysis by 2-deoxy-D-glucose (2-DG) over a period of 72 h. 2-DG induced very fast a low-energy situation by a reduced glycolysis metabolite flow rate resulting in pyruvate, lactate, and ATP depletion. Under perfusion culture, both MetR and LowCarb were established on the metabolic level. Interestingly, over the period of 7 days, the metabolome of MetR and LowCarb showed more similarities than differences. This leads to the conclusion that the conditioned medium, in addition to the different restriction forms, substantially reprogramm the cells on the metabolic level. Profound preoperative panoramic radiography analysis might potentially help predict OAC following tooth extraction. In this exploratory study, we evaluated n = 300 consecutive cases (100 OAC and 200 controls) and trained five machine learning algorithms (VGG16, InceptionV3, MobileNetV2, EfficientNet, and ResNet50) to predict OAC versus non-OAC (binary classification task) from the input images. Further, four oral and maxillofacial experts evaluated the respective panoramic radiography and determined performance metrics (accuracy, area under the curve (AUC), precision, recall, F1-score, and receiver operating characteristics curve) of all diagnostic approaches. Cohen's kappa was used to evaluate the agreement between expert evaluations. The deep learning algorithms reached high specificity (highest specificity 100% for InceptionV3) but low sensitivity (highest sensitivity 42.86% for MobileNetV2). The AUCs from VGG16, InceptionV3, MobileNetV2, EfficientNet, and ResNet50 were 0.53, 0.60, 0.67, 0.51, and 0.56, respectively. Expert 1–4 reached an AUC of 0.550, 0.629, 0.500, and 0.579, respectively. The specificity of the expert evaluations ranged from 51.74% to 95.02%, whereas sensitivity ranged from 14.14% to 59.60%. Cohen's kappa revealed a poor agreement for the oral and maxillofacial expert evaluations (Cohen's kappa: 0.1285). Overall, present data indicate that OAC cannot be sufficiently predicted from preoperative panoramic radiography. The false-negative rate, i.e., the rate of positive cases (OAC) missed by the deep learning algorithms, ranged from 57.14% to 95.24%. Surgeons should not solely rely on panoramic radiography when evaluating the probability of OAC occurrence. Clinical testing of OAC is warranted after each upper-molar tooth extraction. The Archer and the Root Sinus (RS) systems can be used to classify impacted teeth in panoramic radiographs. The Archer classes B-D and the Root Sinus classes III, IV have been associated with an increased risk of OAC following tooth extraction in the upper molar region. In our previous study, we found that panoramic radiographs are not reliable for predicting OAC. This study aimed to (1) determine the feasibility of automating the classification (Archer/RS classes) of impacted teeth from panoramic radiographs, (2) determine the distribution of OAC stratified by classification system classes for the purposes of decision tree construction, and (3) determine the feasibility of automating the prediction of OAC utilizing the mentioned classification systems. Methods: We utilized multiple supervised pre-trained machine learning models (VGG16, ResNet50, Inceptionv3, EfficientNet, MobileNetV2), one custom-made convolutional neural network (CNN) model, and a Bag of Visual Words (BoVW) technique to evaluate the performance to predict the clinical classification systems RS and Archer from panoramic radiographs (Aim 1). We then used Chi-square Automatic Interaction Detectors (CHAID) to determine the distribution of OAC stratified by the Archer/RS classes to introduce a decision tree for simple use in clinics (Aim 2). Lastly, we tested the ability of a multilayer perceptron artificial neural network (MLP) and a radial basis function neural network (RBNN) to predict OAC based on the high-risk classes RS III, IV, and Archer B-D (Aim 3). Results: We achieved accuracies of up to 0.771 for EfficientNet and MobileNetV2 when examining the Archer classification. For the AUC, we obtained values of up to 0.902 for our custom-made CNN. In comparison, the detection of the RS classification achieved accuracies of up to 0.792 for the BoVW and an AUC of up to 0.716 for our custom-made CNN. Overall, the Archer classification was detected more reliably than the RS classification when considering all algorithms. CHAID predicted 77.4% correctness for the Archer classification and 81.4% for the RS classification. MLP (AUC: 0.590) and RBNN (AUC: 0.590) for the Archer classification as well as MLP 0.638) and RBNN (0.630) for the RS classification did not show sufficient predictive capability for OAC. Conclusions: The results reveal that impacted teeth can be classified using panoramic radiographs (best AUC: 0.902), and the classification systems can be stratified according to their relationship to OAC (81.4% correct for RS classification). However, the Archer and RS classes did not achieve satisfactory AUCs for predicting OAC (best AUC: 0.638). Additional research is needed to validate the results externally and to develop a reliable risk stratification tool based on the present findings. However, the number of cases within the studies was limited, and the studies had different measurement conditions. By analyzing data from the National Health and Nutrition Examination Survey III (NHANES III), this study aimed to investigate possible associations between chronic obstructive pulmonary disease (COPD) and periodontitis in the general population. COPD was diagnosed in cases where FEV (1)/FVC ratio was below 70% (non-COPD versus COPD; binary classification task). We used unsupervised learning utilizing k-means clustering to identify clusters in the data. COPD classes were predicted with logistic regression, a random forest classifier, a stochastic gradient descent (SGD) classifier, k-nearest neighbors, a decision tree classifier, Gaussian naive Bayes (GaussianNB), support vector machines (SVM), a custom-made convolutional neural network (CNN), a multilayer perceptron artificial neural network (MLP), and a radial basis function neural network (RBNN) in Python. We calculated the accuracy of the prediction and the area under the curve (AUC). The most important predictors were determined using feature importance analysis. Results: Overall, 15,868 participants and 19 feature variables were included. Based on k-means clustering, the data were separated into two clusters that identified two risk characteristic groups of patients. The algorithms reached AUCs between 0.608 (DTC) and 0.953% (CNN) for the classification of COPD classes. Feature importance analysis of deep learning algorithms indicated that age and mean attachment loss were the most important features in predicting COPD. Conclusions: Data analysis of a large population showed that machine learning and deep learning algorithms could predict COPD cases based on demographics and oral health feature variables. This study indicates that periodontitis might be an important predictor of COPD. Further prospective studies examining the association between periodontitis and COPD are warranted to validate the present results. One of the major inducers of chlamydial persistence is interferon-gamma (IFN-γ) released by immune cells as a mechanism of immune defence. IFN-γ activates the catabolic depletion of L-tryptophan (Trp) via indoleamine-2,3-dioxygenase (IDO), resulting in persistent Ctr. Here, we show that IFN-γ induces the downregulation of c-Myc, the key regulator of host cell metabolism, in a STAT1-dependent manner. Expression of c-Myc rescued Ctr from IFN-γ-induced persistence in cell lines and human fallopian tube organoids. Trp concentrations control c-Myc levels most likely via the PI3K-GSK3β axis. Unbiased metabolic analysis revealed that Ctr infection reprograms the host cell tricarboxylic acid (TCA) cycle to support pyrimidine biosynthesis. Addition of TCA cycle intermediates or pyrimidine/purine nucleosides to infected cells rescued Ctr from IFN-γ-induced persistence. Thus, our results challenge the longstanding hypothesis of Trp depletion through IDO as the major mechanism of IFN-γ-induced metabolic immune defence and significantly extends the understanding of the role of IFN-γ as a broad modulator of host cell metabolism.
