Interactions of qubits in the qubit ocean are rare. If they happen, they provide a nucleus for a new universe as the qubits become decoherent and freeze-out into defined bit ensembles. Second, we replace inflation by a crystallization event triggered by the nucleus of interacting qubits to which rapidly more and more qubits attach (like in everyday crystal growth) – the crystal unit cell guarantees same symmetries everywhere. Hence, the textbook inflation scenario to explain the same laws of nature in our domain is replaced by the crystal unit cell of the crystal formed. We give here only the perspective or outline of this modified inflation theory, as the detailed mathematical physics behind this has still to be formulated and described. Interacting qubits solidify, quantum entropy decreases (but increases in the ocean around). The interacting qubits form a rapidly growing domain where the n**m states become separated ensemble states, rising long-range forces stop ultimately further growth. After that very early events, standard cosmology with the hot fireball model takes over. Our theory agrees well with lack of inflation traces in cosmic background measurements, but more importantly can explain well by such a type of cosmological crystallization instead of inflation the early creation of large-scale structure of voids and filaments, supercluster formation, galaxy formation, and the dominance of matter: no annihilation of antimatter necessary, rather the unit cell of our crystal universe has a matter handedness avoiding anti-matter. We prove a triggering of qubit interactions can only be 1,2,4 or 8-dimensional (agrees with E8 symmetry of our universe). Repulsive forces at ultrashort distances result from quantization, long-range forces limit crystal growth. Crystals come and go in the qubit ocean. This selects for the ability to lay seeds for new crystals, for self-organization and life-friendliness. The phase space of the crystal agrees with the standard model of the basic four forces for n quanta. It includes all possible ensemble combinations of their quantum states m, a total of n**m states. Neighbor states reach according to transition possibilities (S-matrix) with emergent time from entropic ensemble gradients. However, this means that in our four dimensions there is only one bit overlap to neighbor states left (almost solid, only below h dash liquidity left). However, the E8 symmetry of heterotic string theory has six rolled-up, small dimensions which help to keep the qubit crystal together and will never expand. Finally, we give first energy estimates for free qubits vs bound qubits, misplacements in the qubit crystal and entropy increase during qubit decoherence / crystal formation. Among a variety of experimental SAH models, the double hemorrhage rat model involving direct injection of autologous arterial blood into the cisterna magna has been used most frequently for the study of delayed cerebral vasospasm following SAH in last years. Despite the simplicity of the technique, the second blood injection into the cisterna magna may result in brainstem injury leading to high mortality. Geometric and electronic properties of copper phthalocyanine (CuPc) on top of graphene on hexagonal Ru(0001), rectangular Ru(10-10) and vicinal Ru(1,1,-2,10) surfaces have been studied. First, the Fermi surfaces and band structures of the three Ru surfaces were investigated by high-resolution angle-resolved photoemission spectroscopy. The experimental data and theoretical calculations allow to derive detailed information about the momentum-resolved electronic structure. The results can be used as a reference to understand the chemical and catalytic properties of Ru surfaces. Second, graphene layers were prepared on the three different Ru surfaces. Using low-energy electron diffraction and scanning tunneling microscopy, it was found that graphene can be grown in well-ordered structures on all three surfaces, hexagonal Ru(0001), rectangular Ru(10-10) and vicinal Ru(1,1,-2,10), although they have different surface symmetries. Evidence for a strong interaction between graphene and Ru surfaces is a 1.3-1.7e V increase in the graphene pi-bands binding energy with respect to free-standing graphene sheets. This energy variation is due to the hybridization between the graphene pi bands and the Ru 4d electrons, while the lattice mismatch does not play an important role in the bonding between graphene and Ru surfaces. Finally, the geometric and electronic structures of CuPc on Ru(10-10), graphene/Ru(10-10), and graphene/Ru(0001) have been studied in detail. CuPc molecules can be grown well-ordered on Ru(10-10) but not on Ru(0001). The growth of CuPc on graphene/Ru(10-10) and Ru(0001) is dominated by the Moire pattern of graphene. CuPc molecules form well-ordered structures with rectangular unit cells on graphene/Ru(10-10) and Ru(0001). The distance of adjacent CuPc molecules is 1.5 and 1.3 nm on graphene/Ru(0001) and 1.54 and 1.37 nm on graphene/Ru(10-10). This indicates that the molecule-substrate interaction dominates over the intermolecular interaction for CuPc molecules on graphene/Ru(10-10) and graphene/Ru(0001). Zunächst wurden die Fermioberflächen und Bandstrukturen der drei Rutheniumoberflächen mittels hochauflösender winkelaufgelöster Photoemissions spektroskopie ermittelt. Die experimentellen Daten und theoretischen Berechnungen erlauben es, detaillierte Informationen zur impulsaufgelösten elektronischen Struktur abzuleiten. Die Ergebnisse können als Referenz für ein besseres Verständnis der chemischen und katalytischen Eigenschaften von Rutheniumoberflächen dienen. Als nächstes wurden Graphenschichten auf den drei verschiedenen Rutheniumoberflächen hergestellt. Bei Messungen der Beugung niederenergetischer Elektronen an den Oberflächen sowie mittels Rastertunnelmikroskopie stellte sich heraus, dass Graphen hoch geordnete Strukturen auf allen drei Oberflächen, hexagonalem Ru(0001), rechteckigem Ru(10-10) und vicinalem Ru(1,1,-2,10), bildet, obwohl diese unterschiedliche Symmetrien aufweisen. Ein Hinweis auf eine starke Wechselwirkung zwischen Graphen und den Rutheniumoberflächen ist der Anstieg der Bindungsenergie der Graphen-pi-Bänder um 1.3-1.7 eV im Vergleich zu freistehenden Graphenschichten. Diese Änderung der Energie beruht auf der Hybridisierung zwischen den Graphen-pi-Bändern und den 4d-Elektronen des Rutheniums, wohingegen der Gitterversatz keine große Rolle bei der Bindung zwischen Graphen und Rutheniumoberflächen spielt. Abschließend wurden die geometrischen und elektronischen Strukturen von CuPc auf Ru(10-10), Graphen/Ru(10-10) und Graphen/Ru(0001) im Detail untersucht. CuPc-Moleküle konnten mit hoher Ordnung auf Ru(10-10) abgelagert werden, nicht jedoch auf Ru(0001). Das Wachstum von CuPc auf Graphen/Ru(10-10) und Ru(0001) wird durch die Moirestruktur des Graphens bestimmt. CuPc-Moleküle bilden hoch geordnete Strukturen mit rechteckigen Elementarzellen auf Graphen/Ru(10-10) und Ru(0001). Der Abstand benachbarter CuPc-Moleküle beträgt 1.5 und 1.3 nm auf Graphen/Ru(0001) sowie 1.54 und 1.37 nm auf Graphen/Ru(10-10). The present study evaluates stimulus-dependent structure in spontaneous eye-blink behavior as an objective, corroborative measure for the media use phenomenon of spatial presence. To this end, a mixed methods approach is used in an experimental setting to collect, combine, analyze, and interpret data from standardized participant self-report, observation of participant behavior, and content analysis of the media stimulus. T-pattern detection is used to analyze stimulus-dependent blinking behavior, and this structural data is then contrasted with self-report data. The combined results show that behavioral indicators yield the predicted results, while self-report data shows unpredicted results that are not predicted by the underlying theory. The use of a mixed methods approach offered insights that support further theory development and theory testing beyond a traditional, mono-method experimental approach. The authors present a case of delayed sevoflurane-induced malignant hyperthermia in a 21-year-old male patient that was sufficiently treated by discontinuation of trigger agent application and dantrolene infusion. After surviving an MH episode diagnostic procedures are indicated to increase patient safety. In the presented case, the use of a novel minimal-invasive metabolic test with intramuscular injection of halothane and caffeine successfully confirmed MH susceptibility and hence might be an alternative for invasive in vitro contracture testing in selected cases. Furthermore, to be able to compare neuronal branching patterns across individuals, a common framework is required, which has led to the development of 3D standard brain atlases in most of the neurobiological insect model species. Yet, no bumblebee 3D standard brain atlas has been generated. Here we present a brain atlas for the buff-tailed bumblebee Bombus terrestris using micro-computed tomography (micro-CT) scans as a source for the raw data sets, rather than traditional confocal microscopy, to produce the first ever micro-CT-based insect brain atlas. We illustrate the advantages of the micro-CT technique, namely, identical native resolution in the three cardinal planes and 3D structure being better preserved. Our Bombus terrestris brain atlas consists of 30 neuropils reconstructed from ten individual worker bees, with micro-CT allowing us to segment neuropils completely intact, including the lamina, which is a tissue structure often damaged when dissecting for immunolabeling. Our brain atlas can serve as a platform to facilitate future neuroscience studies in bumblebees and illustrates the advantages of micro-CT for specific applications in insect neuroanatomy. Here we show that a shoot-derived micro RNA, miR2111, systemically steers lateral root initiation and nitrogen responsiveness through its root target TML (TOO MUCH LOVE) in the legume Lotus japonicus, where miR2111 and TML were previously shown to regulate symbiotic infections with nitrogen fixing bacteria. Intriguingly, systemic control of lateral root initiation by miR2111 and TML/HOLT (HOMOLOGUE OF LEGUME TML) was conserved in the nonsymbiotic ruderal Arabidopsis thaliana, which follows a distinct ecological strategy. Mitotane (MT) has been used for treatment of ACC for decades, either alone or in combination with cytotoxic chemotherapy. Even at doses up to 6 g per day, more than half of the patients do not achieve targeted plasma concentration (14–20 mg L\(^{-1}\)) even after many months of treatment due to low water solubility, bioavailability, and unfavorable pharmacokinetic profile. Here a novel MT nanoformulation with very high MT concentrations in physiological aqueous media is reported. The MT‐loaded nanoformulations are characterized by Fourier transform infrared spectroscopy, differential scanning calorimetry, and powder X‐ray diffraction which confirms the amorphous nature of the drug. The polymer itself does not show any cytotoxicity in adrenal and liver cell lines. By using the ACC model cell line NCI‐H295 both in monolayers and tumor cell spheroids, micellar MT is demonstrated to exhibit comparable efficacy to its ethanol solution. It is postulated that this formulation will be suitable for i.v. application and rapid attainment of therapeutic plasma concentrations. In conclusion, the micellar formulation is considered a promising tool to alleviate major drawbacks of current MT treatment while retaining bioactivity toward ACC in vitro. Adequate moisture in the indoor environment helps microbial communities multiply fast. The outdoor air and materials that are brought into the buildings by airflow, sandstorms, animals pets and house occupants endow the indoor dust particles with extra features that impact human health. Assessment of the health effects of indoor dust particles, the type of indoor microbial inoculants and the secreted enzymes by indoor insects as allergens merit detailed investigation. Here, we discuss the applications of next generation sequencing (NGS) technology which is used to assess microbial diversity and abundance of the indoor dust environments. Likewise, the applications of NGS are discussed to monitor the gene expression profiles of indoor human occupants or their surrogate cellular models when exposed to aqueous solution of collected indoor dust samples. We also highlight the detection methods of dust allergens and analytical procedures that quantify the chemical nature of indoor particulate matter with a potential impact on human health. An alternative human-computer interaction para- digm is the maintenance and manipulation of electronic documents, which provides several ad- vantages with respect to the social aspects of knowledge acquisition. Until today it hardly has found any attention as a method for knowledge engineering. This thesis provides a comprehensive discussion of document-centered knowledge acquisition with knowledge markup languages. There, electronic documents are edited by the knowledge authors and the executable knowledge base entities are captured by markup language expressions within the documents. The analysis of this approach reveals significant advantages as well as new challenges when compared to the use of traditional GUI-based tools. Some advantages of the approach are the low barriers for domain expert participation, the simple integration of informal descriptions, and the possibility of incremental knowledge for- malization. It therefore provides good conditions for building up a knowledge acquisition pro- cess based on the mixed-initiative strategy, being a flexible combination of direct and indirect knowledge acquisition. Further it turns out that document-centered knowledge acquisition with knowledge markup languages provides high potential for creating customized knowledge au- thoring environments, tailored to the needs of the current knowledge engineering project and its participants. The thesis derives a process model to optimally exploit this customization po- tential, evolving a project specific authoring environment by an agile process on the meta level. This meta-engineering process continuously refines the three aspects of the document space: The employed markup languages, the scope of the informal knowledge, and the structuring and organization of the documents. The evolution of the first aspect, the markup languages, plays a key role, implying the design of project specific markup languages that are easily understood by the knowledge authors and that are suitable to capture the required formal knowledge precisely. The goal of the meta-engineering process is to create a knowledge authoring environment, where structure and presentation of the domain knowledge comply well to the users’ mental model of the domain. In that way, the approach can help to ease major issues of knowledge-based system development, such as high initial development costs and long-term maintenance problems. In practice, the application of the meta-engineering approach for document-centered knowl- edge acquisition poses several technical challenges that need to be coped with by appropriate tool support. In this thesis KnowWE, an extensible document-centered knowledge acquisition environment is presented. The system is designed to support the technical tasks implied by the meta-engineering approach, as for instance design and implementation of new markup lan- guages, content refactoring, and authoring support. It is used to evaluate the approach in several real-world case-studies from different domains, such as medicine or engineering for instance. We end the thesis by a summary and point out further interesting research questions consid- ering the document-centered knowledge acquisition approach. To determine whether youth soccer players may benefit from the intermittent load and time efficiency of HIIT, we performed a meta-analysis of the relevant scientific literature. Objectives Our primary objective was to compare changes in various physiological parameters related to the performance of youth soccer players in response to running-based HIIT to the effects of other common training protocols (i.e., small-sided games, technical training and soccer-specific training, or high-volume endurance training). A secondary objective was to compare specifically running-based HIIT to a soccer-specific form of HIIT known as small-sided games (SSG) in this same respect, since this latter type of training is being discussed extensively by coaches. Method A systematic search of the PubMed, SPORTDiscus, and Web of Science databases was performed in August of 2017 and updated during the review process in December of 2018. The criteria for inclusion of articles for analysis were as follows: (1) comparison of HIIT to SSG or some other training protocol employing a pre-post design, (2) involvement of healthy young athletes (≤ 18 years old), and (3) assessment of variables related to endurance or soccer performance. Hedges’ g effect size (dppc2) and associated 95% confidence intervals for the comparison of the responses to HIIT and other interventions were calculated. Results Nine studies, involving 232 young soccer players (mean age 16.2 ± 1.6 years), were examined. Endurance training in the form of HIIT or SSG produced similar positive effects on most parameters assessed, including peak oxygen uptake and maximal running performance during incremental running (expressed as Vmax or maximal aerobic speed (MAS)), shuttle runs (expressed as the distance covered or time to exhaustion), and time-trials, as well as submaximal variables such as running economy and running velocity at the lactate threshold. HIIT induced a moderate improvement in soccer-related tests involving technical exercises with the soccer ball and other game-specific parameters (i.e., total distance covered, number of sprints, and number of involvements with the ball). Neuromuscular parameters were largely unaffected by HIIT or SSG. Conclusion The present meta-analysis indicates that HIIT and SSG have equally beneficial impacts on variables related to the endurance and soccer-specific performance of youth soccer players, but little influence on neuromuscular performance. Concerning the latter effect, several mutants of Arabidopsis thaliana with improved capability for \(H^+\) efflux (wat1-1D, overexpression of AKT1 and ost2-1D) are less sensitive to inhibition by ABA than the wild type. This suggested that ABA could inhibit \(H^+\) efflux (\(H^+\)-ATPase) and induce cytosolic acidification as a mechanism of growth inhibition. Measurements to test this hypothesis could not be done in germinating seeds and we used roots as the most convenient system. ABA inhibited the root plasma-membrane H+-ATPase measured in vitro (ATP hydrolysis by isolated vesicles) and in vivo (\(H^+\) efflux from seedling roots). This inhibition involved the core ABA signalling elements: PYR/PYL/RCAR ABA receptors, ABA-inhibited protein phosphatases (HAB1), and ABA-activated protein kinases (SnRK2.2 and SnRK2.3). Electrophysiological measurements in root epidermal cells indicated that ABA, acting through the PYR/PYL/RCAR receptors, induced membrane hyperpolarization (due to \(K^+\) efflux through the GORK channel) and cytosolic acidification. This acidification was not observed in the wat1-1D mutant. The mechanism of inhibition of the \(H^+\)-ATPase by ABA and its effects on cytosolic pH and membrane potential in roots were different from those in guard cells. ABA did not affect the in vivo phosphorylation level of the known activating site (penultimate threonine) of (\(H^+\)-ATPase in roots, and SnRK2.2 phosphorylated in vitro the C-terminal regulatory domain of (\(H^+\)-ATPase while the guard-cell kinase SnRK2.6/OST1 did not. A number of aspects of the calorimeter response to isolated hadrons are explored. After accounting for energy deposited by neutral particles, there is a 5% discrepancy in the modelling, using various sets of GEANT4 hadronic physics models, of the calorimeter response to isolated charged hadrons in the central calorimeter region. The description of the response to anti-protons at low momenta is found to be improved with respect to previous analyses. The electromagnetic and hadronic calorimeters are also examined separately, and the detector simulation is found to describe the response in the hadronic calorimeter well. The jet energy scale uncertainty and correlations in scale between jets of different momenta and pseudorapidity are derived based on these studies. The uncertainty is 2–5% for jets with transverse momenta above 2 TeV, where this method provides the jet energy scale uncertainty for ATLAS. However, no software code exists for DGPS exploiting correction factors at base stations, without relying on double difference information. In order to accomplish it, a methodology is introduced in MATLAB environment for DGPS using C/A pseudoranges on single frequency L1 only to make it feasible for low-cost GPS receivers. Our base station is at accurately surveyed reference point. Pseudoranges and geometric ranges are compared at base station to compute the correction factors. These correction factors are then handed over to rover for all valid satellites observed during an epoch. The rover takes it into account for its own true position determination for corresponding epoch. In order to validate the proposed algorithm, our rover is also placed at a pre-determined location. These terms model the effects of the external stimuli on the molecular network. The usability of this extension is demonstrated with a model of a circadian clock that is extended with certain terms and reproduces data from several experiments at the same time. Once the model including external stimuli is set up, a framework is developed in order to calculate external stimuli that have a predefined desired effect on the molecular network. For this purpose the task of finding appropriate external stimuli is formulated as a mathematical optimal control problem for which in order to solve it a lot of mathematical methods are available. Several methods are discussed and worked out in order to calculate a solution for the corresponding optimal control problem. The application of the framework to find pharmacological intervention points or effective drug combinations is pointed out and discussed. Furthermore the framework is related to existing network analysis tools and their combination for network analysis in order to find dedicated external stimuli is discussed. The total framework is verified with biological examples by comparing the calculated results with data from literature. For this purpose platelet aggregation is investigated based on a corresponding gene regulatory network and associated receptors are detected. Furthermore a transition from one to another type of T-helper cell is analyzed in a tumor setting where missing agents are calculated to induce the corresponding switch in vitro. Next a gene regulatory network of a myocardiocyte is investigated where it is shown how the presented framework can be used to compare different treatment strategies with respect to their beneficial effects and side effects quantitatively. Moreover a constitutively activated signaling pathway, which thus causes maleficent effects, is modeled and intervention points with corresponding treatment strategies are determined that steer the gene regulatory network from a pathological expression pattern to physiological one again. N2 - In dieser Arbeit werden Modelle für molekulare Netzwerke bestehend aus gewöhnlichen Differentialgleichungen durch Terme erweitert, die die Wechselwirkung zwischen dem entsprechenden molekularen Netzwerk und der Umgebung berücksichtigen, in die das molekulare Netzwerk eingebettet ist. Diese Terme modellieren die Effekte von externen Stimuli auf das molekulare Netzwerk. Die Nutzbarkeit dieser Erweiterung wird mit einem Modell der circadianen Uhr demonstriert, das mit gewissen Termen erweitert wird und Daten von mehreren verschiedenen Experimenten zugleich reproduziert. Sobald das Modell einschließlich der externen Stimuli aufgestellt ist, wird eine Grundstruktur entwickelt um externe Stimuli zu berechnen, die einen gewünschten vordefinierte Effekt auf das molekulare Netzwerk haben. Zu diesem Zweck wird die Aufgabe, geeignete externe Stimuli zu finden, als ein mathematisches optimales Steuerungsproblem formuliert, für welches, um es zu lösen, viele mathematische Methoden zur Verfügung stehen. Verschiedene Methoden werden diskutiert und ausgearbeitet um eine Lösung für das entsprechende optimale Steuerungsproblem zu berechnen. Auf die Anwendung dieser Grundstruktur pharmakologische Interventionspunkte oder effektive Wirkstoffkombinationen zu finden, wird hingewiesen und diese diskutiert. Weiterhin wird diese Grundstruktur in Bezug zu existierenden Netzwerkanalysewerkzeugen gesetzt und ihre Kombination für die Netzwerkanalyse diskutiert um zweckbestimmte externe Stimuli zu finden. Die gesamte Grundstruktur wird mit biologischen Beispielen verifiziert, indem man die berechneten Ergebnisse mit Daten aus der Literatur vergleicht. Zu diesem Zweck wird die Blutplättchenaggregation untersucht basierend auf einem entsprechenden genregulatorischen Netzwerk und damit assoziierte Rezeptoren werden detektiert. Weiterhin wird ein Wechsel von einem T-Helfer Zelltyp in einen anderen in einer Tumorumgebung analysiert, wobei fehlende Agenzien berechnet werden um den entsprechenden Wechsel in vitro zu induzieren. For this purpose we extend investigations on the previously discovered tunneling anisotropic magneto resistance effect (TAMR) by scaling the pillar size from 100 µm down to 260 nm. We start in chapter 4 with a theoretical description of the TAMR effect and show experimental data of miniaturized pillars in chapter 5. With such small TAMR probes we are able to locally sense the magnetization on the 100 nm scale. Sub-micron TAMR and anisotropic magneto resistance (AMR) measurements of sub-millimeter areas show that the behavior of macroscopic (Ga,Mn)As regions is not that of a true macrospin, but rather an ensemble average of the behavior of many nearly identical macrospins. This shows that the magnetic anisotropies of the local regions are consistent with the behavior extracted from macroscopic characterization. A fully electrically controllable read-write memory device out the ferromagnetic semiconductor (Ga,Mn)As is presented in chapter 6. The structure consists of four nanobars which are connected to a circular center region. The first part of the chapter describes the lithography realization of the device. We make use of the sub-micron TAMR probes to read-out the magnetization state of a 650 nm central disk. Four 200 nm wide nanobars are connected to the central disk and serve as source and drain of a spin-polarized current. With the spin-polarized current we are able to switch the magnetization of the central disk by means of current induced switching. Injecting polarized holes with a spin angular momentum into a magnetic region changes the magnetization direction of the region due to the p-d exchange interaction between localized Mn spins and itinerant holes. The magnetization of the central disk can be controlled fully electrically and it can serve as one bit memory element as part of a logic device. In chapter 7 we discuss the domain wall resistance in (Ga,Mn)As. At the transition from nanobars to central disk we are able to generate 90° and 180° domain walls and measure their resistance. The results presented from chapter 5 to 7 combined with the preexisting ultracompact (Ga,Mn)As-based memory cell of ref. [Papp 07c] are the building blocks needed to realize a fully functioning programmable logic device. The work of ref. [Papp 07c] makes use of lithographically engineered strain relaxation to produce a structure comprised of two nanobars with mutually orthogonal uniaxial easy axes, connected by a narrow constriction. Measurements showed that the resistance of the constriction depends on the relative orientation of the magnetization in the two bars. The programmable logic device consists of two central disks connected by a small constriction. The magnetization of the two central disks are used as the input bits and the constriction serves as the output during the logic operation. The concept is introduced in the end of chapter 6 and as an example for a logic operation an XOR gate is presented. The functionality of the programmable logic scheme presented here can be straightforwardly extended to produce multipurpose functional elements, where the given geometry can be used as various different computational elements depending on the number of input bits and the chosen electrical addressing. The realization of such a programmable logic device is shown in chapter 8, where we see that the constriction indeed can serve as a output of the logic operation because its resistance is dependent on the relative magnetization state of both disks. Contrary to ref. [Papp 07c], where the individual magnetic elements connected to the constriction only have two non-volatile magnetic states, each disk in our scheme connected to the constriction has four non-volatile magnetic states. Switching the magnetization of a central disk with an electrical current does not only change the TAMR read-out of the respective disk, it also changes the resistance of the constriction. The resistance polar plot of the constriction maps the relative magnetization states of the individual disks. The presented device design serves as an all-electrical, all-semiconductor logic element. It combines a memory cell and data processing in a single monolithic paradigm. N2 - Für die Realisierung eines programmierbaren Logikelements oder beliebiger nanometer großer Bauteile, brauchen wir eine verlässlige Methode, um die Magnetisierungsrichtung lokaler Domänen auzulesen. Dafür erweitern wir die Untersuchungen an TAMR (tunneling magneto resistance) Strukturen und skalieren die Fläche des Tunnelkontakts von 100 µm auf 260 nm. In Kapitel 4 geben wir zunächst eine theoretische Beschreibung des TAMR Effekts und zeigen darauf im folgenden Kapitel 5 experimentelle Daten der miniaturisierten Tunnelkontakte. Mit diesen TAMR-Kontakten ist es möglich die Magnetisierung lokal in einer Grössenordnung von 100 nm zu detektieren. Sub-micron TAMR-Messungen und anisotrope Magnetowiderstandmessungen (AMR) an sub-millimeter Gebieten zeigen, dass das Verhalten von makrokopischen (Ga,Mn)As nicht das eines Makrospins ist, sondern ein Ensembledurchschnitt von vielen fast identischen Makrospins. Dieses Ergebnis ist mit der makroskopischen Beschreibung der lokalen magnetischen Aniotropien konform.\\ Ein rein elektrisch kontrollierbares Read-Write Speicherelement aus dem ferromagnetischen Halbleiter (Ga,Mn)As wird in Kapitel 6 gezeigt. Die Magnetisierung der zentralen Disc kann rein elektrisch kontrolliert werden and als Bit eines Logikelementes verwendet werden. In Kapitel 7 untersuchen wir den Domänenwiderstand in (Ga,Mn)As. Am Übergang von den Streifen zur zentralen Disc ist es möglich 90°- und 180° Domänenwände zu erzeugen und deren Widerstand zu messen.\\ Die Ergebnisse von Kapitel 5 bis 7, kombiniert mit dem bereits existierenden Ergebnissen einer ultrakompakten (Ga,Mn)As-basierenden Speicherzelle von Ref. [Papp 07c], sind die Schlüsselelemente die man zur Realisierung eines programmierbaren Logikelements benötigt. Die Arbeit von Referenz [Papp 07c] nutzt Lithographie induzierte Deformationsrelaxation, um eine Struktur zu erzeugen, die aus zwei senkrechten Streifen besteht und durch eine Verengung verbunden sind. Retroperitoneal lipoblastomas are particularly rare but can grow to enormous size, and preoperative diagnosis is difficult with diverse, mostly malignant differential diagnoses that would lead to aggressive therapy. Since lipoblastoma is a benign tumor that has an excellent prognosis after resection, correct diagnosis is crucial. Case presentation A case of a large retroperitoneal tumor of a 24-month old infant that was clinically suspicious of a malignant tumor is presented. Due to proximity to the right kidney, clinically most probably a nephroblastoma or clear cell sarcoma of the kidney was suspected. Radiological findings were ambiguous. Therefore, the mass was biopsied, and histology revealed an adipocytic lesion. Although mostly composed of mature adipocytes, in view of the age of the patient, the differential diagnosis of a (maturing) lipoblastoma was raised, which was supported by molecular analysis demonstrating a HAS2-PLAG1 fusion. The tumor was completely resected, and further histopathological workup led to the final diagnosis of a 13 cm large retroperitoneal maturing lipoblastoma. The child recovered promptly from surgery and showed no evidence of recurrence so far. Conclusion Although rare, lipoblastoma should be included in the differential diagnoses of retroperitoneal tumors in infants and children, and molecular diagnostic approaches could be a helpful diagnostic adjunct in challenging cases. So far, volumetric brain changes have been proposed as clinical endpoints in the literature, but cognitive scores are still lacking. This study followed disease progression predominantly in language-based performance within 1 year and defined a PPA sum score which can be used in therapeutic interventions. Methods We assessed 28 patients with nonfluent variant PPA, 17 with semantic variant PPA, 13 with logopenic variant PPA, and 28 healthy controls in detail for 1 year. The most informative neuropsychological assessments were combined to a sum score, and associations between brain atrophy were investigated followed by a sample size calculation for clinical trials. Results Significant absolute changes up to 20% in cognitive tests were found after 1 year. Semantic and phonemic word fluency, Boston Naming Test, Digit Span, Token Test, AAT Written language, and Cookie Test were identified as the best markers for disease progression. These tasks provide the basis of a new PPA sum score. Assuming a therapeutic effect of 50% reduction in cognitive decline for sample size calculations, a number of 56 cases is needed to find a significant treatment effect. Correlations between cognitive decline and atrophy showed a correlation up to r = 0.7 between the sum score and frontal structures, namely the superior and inferior frontal gyrus, as well as with left-sided subcortical structures. Conclusion Our findings support the high performance of the proposed sum score in the follow-up of PPA and recommend it as an outcome measure in intervention studies. We introduce the concept of data pools as a smaller‐scale, user‐driven and reasonable alternative to co‐develop remote sensing and forest ecosystem science to validated products, biodiversity indicators and management plans. We demonstrate this concept with the Bohemian Forest Ecosystem Data Pool, which has been established as an interdisciplinary, international data pool within the strictly protected Bavarian Forest and Šumava National Parks and currently comprises 10 active partners. We demonstrate how the structure and impact of the data pool differs from comparable cases. We assessed the international influence and visibility of the data pool with the help of a systematic literature search and a brief analysis of the results. Results primarily suggest an increase in the impact and visibility of published material during the life span of the data pool, with highest visibilities achieved by research conducted on leaf traits, vegetation phenology and 3D‐based forest inventory. We conclude that the data pool results in an efficient contribution to the concept of global biodiversity observatory by evolving towards a training platform, functioning as a pool of data and algorithms, directly communicating with management for implementation and providing test fields for feasibility studies on earth observation missions. A hybrid statistical classifier consisting of stemming and clustering algorithms for comparing lexical frequency profiles of chapters, and deriving dates of revelation has been developed. The classifier is trained using some chapters with known dates of revelation. Then it classifies chapters with uncertain dates of revelation by computing their proximity to the training ones. The results reported here indicate that the proposed methodology yields usable results in estimating dates of revelation of the Quran’s chapters based on their lexical contents. Methods: We clinically and radiologically characterised an acromesomelic chondrodysplasia in an adult woman born to consanguineous parents. We sequenced GDF5 and BMPR1B on DNA of the proposita. We performed 3D structural analysis and luciferase reporter assays to functionally investigate the identified BMPR1B mutation. Results: We extend the genotype-phenotype correlation in the acromesomelic chondrodysplasias by showing that the milder du Pan dysplasia can be caused by a hypomorphic BMPR1B mutation. We show that the homozygous c.91C>T, p.(Arg31Cys) mutation causing du Pan dysplasia leads to a significant loss of BMPR1B function, but to a lesser extent than the previously reported p.Cys53Arg mutation that results in the more severe Grebe dysplasia. Conclusions: The phenotypic severity gradient of the clinically and radiologically related acromesomelic chondrodysplasia spectrum of skeletal disorders may be due to the extent of functional impairment of the ligand-receptor pair GDF5-BMPR1B. Such behavior can be assessed with the open field test (OFT), which is a well-established indicator of animal fear. The detection of similar open field behavior in humans may verify the translational validity of this paradigm. Enhanced thigmotaxis related to anxiety may suggest the relevance of such behavior for anxiety disorders, especially agoraphobia. METHODS: A global positioning system was used to analyze the behavior of 16 patients with agoraphobia and 18 healthy individuals with a risk for agoraphobia (i.e., high anxiety sensitivity) during a human OFT and compare it with appropriate control groups (n = 16 and n = 19). We also tracked 17 patients with agoraphobia and 17 control participants during a city walk that involved walking through an open market square. RESULTS: Our human OFT triggered thigmotaxis in participants; patients with agoraphobia and participants with high anxiety sensitivity exhibited enhanced thigmotaxis. This behavior was evident in increased movement lengths along the wall of the natural open field and fewer entries into the center of the field despite normal movement speed and length. Furthermore, participants avoided passing through the market square during the city walk, indicating again that thigmotaxis is related to agoraphobia. CONCLUSIONS: This study is the first to our knowledge to verify the translational validity of the OFT and to reveal that thigmotaxis, an evolutionarily adaptive behavior shown by most species, is related to agoraphobia, a pathologic fear of open spaces, and anxiety sensitivity, a risk factor for agoraphobia. Meningococcal strains belonging to the sequence type (ST-) 41/44 clonal complex (cc41/44) cause a major proportion of serogroup B meningococcal disease worldwide, but they are also common in asymptomatic carriers. Proteome analysis comparing cc41/44 isolates from invasive disease versus carriage revealed differential expression levels of the outer membrane protein NspA, which binds fH. Deletion of nspA reduced serum resistance and NspA expression correlated with fH sequestration. Expression levels of NspA depended on the length of a homopolymeric tract in the nspA promoter: A 5-adenosine tract dictated low NspA expression, whereas a 6-adenosine motif guided high NspA expression. Screening German cc41/44 strain collections revealed the 6-adenosine motif in 39% of disease isolates, but only in 3.4% of carriage isolates. Thus, high NspA expression is associated with disease, but not strictly required. Our framework is seamlessly integrated into ImageJ, a free image processing package with a vast collection of community-developed biological image analysis tools. Our framework enriches the ImageJ software libraries with methods that greatly reduce the complexity of developing image analysis tools in an interactive 3D visualization environment. In particular, we provide high-level access to volume rendering, volume editing, surface extraction, and image annotation. The ability to rely on a library that removes the low-level details enables concentrating software development efforts on the algorithm implementation parts. Conclusions: Our framework enables biomedical image software development to be built with 3D visualization capabilities with very little effort. In most cases, the excessive activity of N-Myc results from a MYCN amplification. In neuroblastoma, amplification of MYCN is a predictor of poor prognosis and resistance to therapy. The inability to target the N-Myc protein directly necessitates the search for alternative targets. This project aimed at identifying genes specifically required for growth and survival of cells that express high levels of N-Myc using high-throughput shRNA screening combined with next generation sequencing. The identification and analysis of these genes will shed light on functional interaction partners of N-Myc. We screened a shRNA library containing 18,327 shRNAs and identified 148 shRNAs, which were selectively depleted in the presence of active N-Myc. In addition, shRNAs targeting genes that are involved in p53 and ARF turnover and apoptosis were depleted in the cell population during the screen. These processes are known to affect N-Myc-mediated apoptosis. Consequently, these results biologically validated the screen. The 148 shRNAs that showed a significant synthetic lethal interaction with high levels of N-Myc expression were further analysed using the bioinformatics program DAVID. We found an enrichment of shRNAs that target genes involved in specific biological processes. For example, we validated synthetic lethal interactions for genes such as, THOC1, NUP153 and LARP7, which play an important role in the process of RNA polymerase II-mediated transcription elongation. We also validated genes that are involved in the neddylation pathway. In the screen we identified Cullin 3, which is a component of the BTB-CUL3-Rbx1 ubiquitin ligase that is involved in the turnover of Cyclin E. Depletion of cullin 3 and activation of N-Myc was found to synergistically increase Cyclin E expression to supraphysiological levels, inducing S-phase arrest and a strong DNA damage response. Together with results from a proteomics analysis of N-Myc associated proteins, our results lead us to the following hypothesis: In a neuroblastoma cell, the high levels of N-Myc result in a conflict between RNA polymerase II and the replication machinery during S-phase. The newly identified interaction partners of N- Myc are required to solve this conflict. Consequently, loss of the interaction leads to a massive DNA damage and the induction of apoptosis. Dies wurde durch eine Kombination von Hochdurchsatz-RNAi-Screens und Next-Generation-Sequenzierung erreicht. Durch das Screenen einer shRNA-Bibliothek, die insgesamt 18327 shRNAs beinhaltet, konnten 148 shRNAs identifiziert werden, die selektiv nachteilig für das Überleben N-Myc überexpremierender Zellen sind. Die statistische Auswertung der Ergebnisse des Screens zeigte zusätzlich eine Anreichung von shRNAs gegen Gene, die p53-und ARF-abhängig Apoptose vermitteln. Da es bekannt ist, dass diese Gene in der N-Myc-vermittelten Apoptose involviert sind, konnte dadurch der Screen validiert werden. Die weitere Auswertung mit dem bioinformatischen Programm DAVID ergab, dass unter den 148 als synthetisch letal identifizierten shRNAs solche angereichert waren, die gegen Gene spezifischer biologischer Prozesse gerichtet sind. Zum einen wurden Gene wie THOC1, NUP153 und LARP7 validiert, die eine Rolle im Prozeß der Elongation der RNA Polymerase II spielen. Zum anderen konnten Gene validiert werden die einen Beitrag bei der Neddylierung von Proteinen leisten. Durch die Depletion von Cullin 3, ein Bestandteil des BTB-CUL3-Rbx1 Ubiquitin-Ligase-Komplexes, der am Abbau von Cyclin E beteiligt ist, konnte gezeigt werden, dass zusammen mit der Aktivierung von N-Myc eine supraphysiologische Erhöhung von Cyclin E induziert wird. Dies führt zu einem S-Phase Arrest in der Zelle, der die DNA-Schadens-Signalkaskade auslöst. Zusammen mit den Ergebnissen einer Proteomanalyse, bei der neue N-Myc-assoziierte Proteine identifiziert wurden, konnte folgende Hypothese aufgestellt werden: In einer Neuroblastomzelle helfen diese neuen Interaktionspartner den durch die N-Myc Überexpression in der S-phase entstehenden Konflikt zwischen RNA-Polymerase II und Replikationsmaschinerie zu lösen. Der Verlust dieser Interaktion führt zu einer massiven Schädigung der DNA, worauf in der Zelle Apoptose ausgelöst wird. The texturing force itself is represented by the linkers between the zwitterionic building blocks or parts of them. The different length and functionalization of the linkers have a tremendous influence on the mode of self-assembly leading to cyclic dimers, vesicles, layers or solid spheres. Diese Invasion geht langsam aber stetig voran und bedroht kostbare und nicht ersetzbare Ressourcen. Vor diesem Hintergrund wurde ein Gebiet in der direkten Nähe der Großstadt Curitiba als Studienobjekt ausgewählt. In diesem Gebiet, dem Iraí-Becken, wurde während der Untersuchungszeit ein Trinkwasserspeicher geplant und gebaut. Es besteht die große Gefahr, dass das Iraí-Reservoir kontaminiert wird, obwohl das engere Gebiet rundum den See geschützt werden soll. Die Verschmutzungsgefahr geht hauptsächlich von zwei Nebenflüssen aus, die durch mehr oder weniger besiedeltes Gebiet strömen. Im Arbeitsgebiet befinden sich Brunnen, die der Trinkwasserversorgung dienen. Um die negativen Folgen einer möglichen Verschmutzung des Reservoirs abschätzen zu können, wurde ein Grundwasserfließmodell erstellt. Die erforderliche Wasserbilanz und die räumliche Verteilung der Verschmutzungsempfindlichkeit wurde mit dem hydrologischen Modell "MODBIL" abgeschätzt. Weitere Methoden zur Abschätzung der Verschmutzungs-empfindlichkeit wurden angewandt, um die differierenden Ergebnisse der angewendeten unterschiedlichen Methoden mit einander vergleichen und bewerten zu können. Mit dem kalibrierten Grundwasserfließmodell ist mit der gegebenen hydraulischen Situation vor und nach der Konstruktion des Reservoirs, ein einfaches Particle Tracking Transport Modell eingesetzt worden, um mit unterschiedlichen Szenarien die Beeinflussung vom Reservoirwasser auf das Grundwasser zu simulieren. This occupation continues with its slow, silent, although progressive march, threatening precious and irreplaceable resources. From this background an area in the direct vicinity north-east of Curitiba has been studied. In this area a drinking water reservoir was constructed in the time that the study took place in the Iraí-basin. The Iraí-reservoir even though an area around the lake will be protected may be polluted by two tributaries which flow through more or less densely populated areas. In the study area on the same time wells have been constructed. To estimate what the impact may be from the possibly polluted reservoir on the aquifer a groundwater flow model has been constructed. On the same time to estimate the water balance and the spatial distribution of pollution vulnerability the hydrological model MODBIL has been used. Also other methods have been used to estimate the pollution vulnerability to make a comparison and because none of the methods takes every aspect into account. With the calibrated groundwater flow model for the situation before the construction of the Iraí-reservoir and after its construction, simple particle tracking transport models are constructed as scenarios how the water of the aquifer may be influenced. In der psychologischen Forschung unterscheidet man drei Arten der Perspektivenübernahme, nämlich perzeptuelle (visuo-spatiale), affektive (Empathie) und kognitive (Theory of Mind). Die letztgenannten Arten der Perspektivenübernahme werden oft als „psychologische Perspektivenübernahme“ zusammengefasst. Diese Dissertation befasst sich mit der Frage, ob diese verschiedenen Arten der Perspektivenübernahme als theoretisch unterscheidbare Konstrukte oder lediglich als Facetten ein und desselben Konstrukts angesehen werden sollten. Die Befundlage in der psychologischen Fachliteratur ist diesbezüglich nicht eindeutig. Diesem Ansatz folgend macht die vorliegende Arbeit die Annahme, dass die mentale Selbstrotation des Körperschemas in die Position einer anderen Person, der zentrale Mechanismus visuo-spatialer Perspektivenübernahme, ein gemeinsamer Mechanismus aller Arten der Perspektivenübernahme ist. Entgegen früherer Ansätze wird diese Einheit somit nicht nur über die zentrale gemeinsame Funktionalität aller Arten von Perspektivenübernahme, also dem Verlassen des egozentrischen Referenzrahmens zugunsten einer (visuellen, affektiven oder kognitiven) Fremdperspektive, gerechtfertigt, sondern mit der Annahme eines gemeinsamen zugrundeliegenden Mechanismus. Daraus wird die einfache Hypothese abgeleitet, dass visuo-spatiale Perspektivenübernahme zu psychologischen Konsequenzen führen kann. Dies wurde in 6 Experimenten getestet. In diesen Experimenten mussten die Probanden zunächst immer die visuelle Perspektive einer anderen Person einnehmen. Hierzu sahen die Probanden eine Person, die mit zwei Objekten an einem Tisch sitzt. In jedem Durchgang mussten die Probanden sich entscheiden, mit welcher Hand diese Person eines der beiden Objekte greifen würde. Dabei wurde die Position der Zielperson so manipuliert, dass sie in der Hälfte der Fälle im selben visuo-spatialen Referenzrahmen wie der Proband saß, was Perspektivenübernahme zur Lösung der Aufgabe obsolet machte, während sie sich in den verbleibenden Durchgängen in einem anderen visuo-spatialen Referenzrahmen befand, so dass die Probanden die visuelle Perspektive der Zielperson übernehmen mussten um die Aufgabe korrekt zu lösen. Nach jedem Durchgang wurde dem Ziel dieser visuo-spatialen Aufgabe eine psychologische Eigenschaft zugeschrieben. Dies geschah im Rahmen eines abgewandelten Paradigmas zur Untersuchung der Ankerheuristik. Hierzu wurde den Probanden nach jedem Durchgang der visuo-spatialen Aufgabe eine Schätzfrage gestellt. Zeitgleich wurde die Antwort des Ziels bekannt gegeben. Entsprechend der Haupthypothese, dass visuo-spatiale Perspektivenübernahme psychologische Konsequenzen erzeugen kann, konnte gezeigt werden, dass die Probanden nach visuo-spatialer Perspektivenübernahme in höherem Maße die Gedanken der Zielperson übernahmen. Dies konnte sowohl anhand der absoluten Größe des Ankereffekts, als auch anhand der Differenz zwischen den Urteilen der Probanden und der Zielperson, gezeigt werden. Weitere Experimente schlossen Stichprobeneigenschaften, die verwendeten Stimuli oder die Aufgabenschwierigkeit als Alternativerklärungen für diese Effekte aus. Die beiden letzten Experimente zeigten zudem, dass dieser Effekt spezifisch für alle Konstellationen ist, in denen eine mentale Selbstrotation in die Zielperspektive notwendig war und dass die Übernahme fremder Gedanken mit einem Gefühl von Ähnlichkeit assoziiert war. Zusammengenommen unterstützen die Ergebnisse dieser Arbeit die theoretisch abgeleitete Sicht eines einheitlichen Perspektivenübernahme-Konstrukts und grenzen dieses zusätzlich von verwandten Konstrukten ab. In der abschließenden Diskussion werden die Bedeutung dieser Befunde für die Forschung in den Bereichen Empathie, Theory of Mind, und Perspektivenübernahme und ebenfalls praktische Implikationen der Ergebnisse aufgezeigt. N2 - „Perspective-taking“ is the ability to put yourself into the place of somebody else. Psychological research distinguishes three kinds of perspective-taking, namely, perceptual (visuo-spatial), affective (empathy), and cognitive (theory of mind) perspective-taking. The last two kinds of perspective-taking are often summarized as “psychological perspective-taking”. This dissertation tackles the question of whether these three kinds of perspective-taking should be conceptualized as independent constructs or as facets of one and the same construct. Prior research findings concerning this are equivocal. While some authors consider correlations between the different kinds of perspective-taking as too low for a unitary construct, others interpret correlations of the same magnitude as evidence for this. A less arbitrary way of deciding this would be to identify common mechanisms that underlie all kinds of perspective-taking and to examine whether manipulating these mechanisms in psychological experiments affects measures of perceptual, affective, and cognitive perspective-taking in parallel. In accordance with this reasoning, the present dissertation assumes that the mental self-rotation of the body schema into the physical location of another person, the main mechanism of perceptual perspective-taking, is a common mechanism of all kinds of perspective-taking. Thus, contrary to previous research a unitary construct is not only justified on the grounds of a common central functionality of all kinds of perspective-taking, that is, overcoming one’s egocentrism in favor of an alternative (perceptual, affective or cognitive) point of view, but additionally on the grounds of a common psychological mechanism. From this, the simple hypothesis that inducing visuo-spatial perspective-taking also leads to psychological consequences is derived. This hypothesis was tested in 6 experiments. In these experiments, participants first had to adopt the visual perspective of another person. To this end, they saw a person sitting at a table with two objects. During every trial, participants had to decide which hand the person would have to use in order to grab one of the two objects. Furthermore, the angular disparity between the participant and the target was manipulated in such a way that during half of the trials the target person was within the same visuo-spatial reference frame as the participant and thus no perspective-taking was necessary to solve the task correctly. During the remaining trials, the target person was sitting in another visuo-spatial reference frame so that the participants had to engage in perspective-taking to solve the task correctly. After every such trial, the target person was imbued with a mental state. This was done using an adapted paradigm for the investigation of the anchoring heuristic. Specifically, participants were asked to answer a trivia question and also saw what the target person from the visuo-spatial perspective-taking task was guessing. In line with the hypothesis that visuo-spatial perspective-taking leads to psychological outcomes, too, it was found that participants adopted the thoughts of the target person more strongly after visuo-spatial perspective-taking. This was evident in the absolute size of the anchoring effect, as well as the differences between participant and target estimations. Further experiments ruled out sample and stimulus characteristics and task difficulty as alternative explanations for these effects. The last two experiments furthermore established that the effects were specific to constellations where an embodied self-rotation into the target’s perspective was necessary and that the adoption of the target’s thoughts was associated with feelings of similarity. Taken together, these findings support the theoretically elaborated unitary view of perspective-taking and furthermore distinguish this construct from other related phenomena. In the general discussion, the significance of these findings for research on empathy, theory of mind, and perspective-taking, as well as practical implications are discussed. While its transcriptome architecture and posttranscriptional regulation through the RNA-binding proteins Hfq, RsmA, and RsmN have been studied in detail, global information about stable RNA-protein complexes in this human pathogen is currently lacking. Here, we implement gradient profiling by sequencing (Grad-seq) in exponentially growing P. aeruginosa cells to comprehensively predict RNA and protein complexes, based on glycerol gradient sedimentation profiles of >73% of all transcripts and ∼40% of all proteins. As to benchmarking, our global profiles readily reported complexes of stable RNAs of P. aeruginosa, including 6S RNA with RNA polymerase and associated product RNAs (pRNAs). We observe specific clusters of noncoding RNAs, which correlate with Hfq and RsmA/N, and provide a first hint that P. aeruginosa expresses a ProQ-like FinO domain-containing RNA-binding protein. To understand how biological stress may perturb cellular RNA/protein complexes, we performed Grad-seq after infection by the bacteriophage ΦKZ. This model phage, which has a well-defined transcription profile during host takeover, displayed efficient translational utilization of phage mRNAs and tRNAs, as evident from their increased cosedimentation with ribosomal subunits. Additionally, Grad-seq experimentally determines previously overlooked phage-encoded noncoding RNAs. Taken together, the Pseudomonas protein and RNA complex data provided here will pave the way to a better understanding of RNA-protein interactions during viral predation of the bacterial cell. IMPORTANCE Stable complexes by cellular proteins and RNA molecules lie at the heart of gene regulation and physiology in any bacterium of interest. It is therefore crucial to globally determine these complexes in order to identify and characterize new molecular players and regulation mechanisms. Pseudomonads harbor some of the largest genomes known in bacteria, encoding ∼5,500 different proteins. Here, we provide a first glimpse on which proteins and cellular transcripts form stable complexes in the human pathogen Pseudomonas aeruginosa. We additionally performed this analysis with bacteria subjected to the important and frequently encountered biological stress of a bacteriophage infection. We identified several molecules with established roles in a variety of cellular pathways, which were affected by the phage and can now be explored for their role during phage infection. Most importantly, we observed strong colocalization of phage transcripts and host ribosomes, indicating the existence of specialized translation mechanisms during phage infection. The antitoxin activity of the antiborlies was investigated in vivo by either ( l) preincubation of the antibody with w-CgTx at 4°C overnight, or (2) pretreatment with antibody, 30 min before \(\omega\)CgTx injection in a 10:1 antibody/\(\omega\)-CgTx molar ratio. The LD50 dose of \(\omega\)-CgTx in goldfish was 5 nmol/kg ip, and preincubation of monoclonal antibody (50 nmol/kg ip) with \(\omega\)-CgTx (5 nmol/kg ip) significantly (p < 0.05) reduced mortality. Here, we use a metatranscriptomics approach to capture expressed genes in open ocean Tara Oceans stations across four organismal size fractions. The individual sequence reads cluster into 116 million unigenes representing the largest reference collection of eukaryotic transcripts from any single biome. The catalog is used to unveil functions expressed by eukaryotic marine plankton, and to assess their functional biogeography. Almost half of the sequences have no similarity with known proteins, and a great number belong to new gene families with a restricted distribution in the ocean. Overall, the resource provides the foundations for exploring the roles of marine eukaryotes in ocean ecology and biogeochemistry. The yeast-to-hyphae morphogenetic switch is activated by well-established, kinase-based signal transduction pathways that are induced by extracellular stimuli. In order to identify possible inhibitory pathways of the yeast-to-hyphae transition, we interrogated a collection of C. albicans protein kinases and phosphatases ectopically expressed under the regulation of the TETon promoter. Proportionately more phosphatases than kinases were identified that inhibited hyphal morphogenesis, consistent with the known role of protein phosphorylation in hyphal induction. Among the kinases, we identified AKL1 as a gene that significantly suppressed hyphal morphogenesis in serum. Akl1 specifically affected hyphal elongation rather than initiation: overexpression of AKL1 repressed hyphal growth, and deletion of AKL1 resulted in acceleration of the rate of hyphal elongation. Akl1 suppressed fluid-phase endocytosis, probably via Pan1, a putative clathrin-mediated endocytosis scaffolding protein. In the absence of Akl1, the Pan1 patches were delocalized from the sub-apical region, and fluid-phase endocytosis was intensified. These results underscore the requirement of an active endocytic pathway for hyphal morphogenesis. Furthermore, these results suggest that under standard conditions, endocytosis is rate-limiting for hyphal elongation. In both phylogenetic groups, the theoretically possible number of individual antigen receptor specificities can even outnumber that of lymphocytes of a whole organism. Despite fundamental differences in structure and genetics of these antigen receptors, convergent evolution led to functional similarities between the lineages. Jawed vertebrates possess αβ and γδ T‐cells defined by eponymous αβ and γδ T‐cell antigen receptors (TCRs). “Conventional” αβ T‐cells recognize complexes of Major Histocompatibility Complex (MHC) class I and II molecules and peptides. Non‐conventional T‐cells, which can be αβ or γδ T‐cells, recognize a large variety of ligands and differ strongly in phenotype and function between species and within an organism. This review describes similarities and differences of non‐conventional T‐cells of various species and discusses ligands and functions of their TCRs. A special focus is laid on Vγ9Vδ2 T‐cells whose TCRs act as sensors for phosphorylated isoprenoid metabolites, so‐called phosphoantigens (PAg), associated with microbial infections or altered host metabolism in cancer or after drug treatment. While a long-standing research tradition has established two opposing fundamental views of perceptual representation, Structuralism and Gestalt psychology, we test both accounts with respect to action representation: Are multiple actions (characterizing human behavior in general) represented as the sum of their component actions (Structuralist view) or holistically (Gestalt view)? Using a single-/dual-response switch paradigm, we analyzed switches between dual ([A + B]) and single ([A], [B]) responses across different effector systems and revealed comparable performance in partial repetitions and full switches of behavioral requirements (e.g., in [A + B] → [A] vs. [B] → [A], or [A] → [A + B] vs. [B] → [A + B]), but only when the presence of dimensional overlap between responses allows for Gestalt formation. This evidence for a Gestalt view of behavior in our paradigm challenges some fundamental assumptions in current (tacitly Structuralist) action control theories (in particular the idea that all actions are represented compositionally with reference to their components), provides a novel explanatory angle for understanding complex, highly synchronized human behavior (e.g., dance), and delimitates the degree to which complex behavior can be analyzed in terms of its basic components. Depression and anxiety are also highly comorbid and have a strong genetic correlation (r(g) approximate to 1). Cognitive behavioural therapy is a leading evidence-based treatment but has variable outcomes. Currently, there are no strong predictors of outcome. Therapygenetics research aims to identify genetic predictors of prognosis following therapy. We performed genome-wide association meta-analyses of symptoms following cognitive behavioural therapy in adults with anxiety disorders (n = 972), adults with major depressive disorder (n = 832) and children with anxiety disorders (n = 920; meta-analysis n = 2724). We (h(SNP)(2)) and polygenic scoring was used to examine genetic associations between therapy outcomes and psychopathology, personality and estimated the variance in therapy outcomes that could be explained by common genetic variants learning. No single nucleotide polymorphisms were strongly associated with treatment outcomes. No significant estimate of h(SNP)(2) could be obtained, suggesting the heritability of therapy outcome is smaller than our analysis was powered to detect. Polygenic scoring failed to detect genetic overlap between therapy outcome and psychopathology, personality or learning. This study is the largest therapygenetics study to date. Results are consistent with previous, similarly powered genome-wide association studies of complex traits. Later, they differentiate into skeletal muscle, cartilage and dermis. Somitogenesis is regulated by a complex interplay of different pathways. Notch/Delta signaling is one of the pathways well characterized in zebrafish through mutants affected in its different components. Previous work in mouse, chicken and zebrafish has shown that also additional components are required during somitogenesis, most importantly through an FGF and Retinoic acid (RA) gradient, as well as Wnt signaling. However, no zebrafish mutants with defects in these pathways showing specific somite malformations are described. This was explained by functional redundancies among related genes that have resulted from a whole genome duplication which occurred in a teleost fish ancestor 350 million years ago. As distinct duplicates exist in different teleost species, a large scale mutagenesis screen in the medaka (Oryzias latipes) has been performed successfully in Kyoto, Japan. I analyzed nine of the isolated medaka mutants that show variable aspects of somitic phenotypes. This includes a complete or partial loss of somite boundaries (e.g. bms and sne), somites with irregular sizes and shapes (e.g. krz and fsl) or partially fused and enlarged somites (e.g. dpk). Although some of these medaka mutants share characteristics with previously described zebrafish somite mutants, most of the mutants represent unique phenotypes, not obtained in the zebrafish screens. In-situ hybridization analyses with marker genes implicated in the segmentation clock (e.g. her7), establishment of anterior-posterior (A-P) polarity (e.g. mesp) and differentiation of somites (e.g. myf5, lfng) revealed that the medaka mutants can be separated into two classes. Class I shows defects in tailbud formation and PSM prepatterning, and lateron somite boundary formation was impaired in these mutants. A unique member of this class with a novel phenotype is the doppelkorn (dpk) mutant that has single fused or enlarged somites. This phenotype has not been reported till now in zebrafish somite mutants. In-situ analyses on dpk showed that stabilization of the cyclically expressed somitogenesis clock genes must be affected in this mutant. This is accompanied by a disrupted regulation of A-P polarity genes like mesp. This suggests that dpk is a mutant deficient in the wave front, which is necessary for the down-regulation of oscillating genes in the anterior PSM. Furthermore, as the initiation of oscillation of all three cyclic her genes was unaffected in dpk embryos, I could exclude that this mutant in affected in the Notch/Delta pathway. Another mutant that belongs to this class is the samidare (sam) mutant. Morphologically, sam mutants are similar to zebrafish after eight (aei). In both cases, the first 7-9 somites are formed properly, but after this somite formation ceases. Different to the situation in aei, sam mutant embryos presented an additional defect in the mid-hindbrain boundary (MHB) region. Similar MHB defects were described in the zebrafish fgf8 mutant acerebellar (ace). In ace zebrafish mutant, somites were only slightly defective, although FGF signaling has been shown to be important for somite formation in chicken, mouse and zebrafish. This was explained by functional redundancy between fgf8 and fgf24 ligands in the tailbud of zebrafish. Thus, it is interesting to suggest that the sam mutant, based on the parallel defects in somites and MHB, is a potential member of the FGF signaling pathway muatnts. It was shown that FGF plays a crucial role during MHB formation in medaka. In addition, I showed that fgf8 acts non-redundantly during tailbud formation and somitogenesis in medaka. Furthermore, I showed that FGF signaling regulates somite size also in medaka and that fgfr1 is the only FGF receptor expressed in the tailbud and somites. In class II medaka somite mutants, PSM prepatterning appears normal, whereas A-P polarity, boundary formation, epithelialization or the later differentiation of somites appears to be affected. Such mutants have not been isolated so far in zebrafish, mice or chicken. Therefore, medaka class II somite mutants seem to be a novel group of mutants that opens new perspectives to analyze A-P polarity regulation, determination and boundary formation in the presence of a normally functioning clock in the PSM. Identifying the encoding genes for all analyzed medaka somite mutants will contribute to the understanding of the molecular interactions of different signaling pathways involved during somitogenesis, and is expected to result in the identification of new components. N2 - Die Somitogenese stellt einen entscheidenden Prozess bei der Entwicklung von Wirbeltierembryonen dar. Somiten sind transiente Strukturen, die sich im Verlauf der Embryonalentwicklung zu Skelettmuskulatur, Dermis und Wirbelkörper differenzieren. Somiten entstehen in einem sich regelmäßig wiederholenden Zyklus aus Stammzellen des präsomitischen Mesoderms (PSM), einer Wachstumszone am caudalen Ende des Embryos. Ein wichtiger Bestandteil der Somitogenese ist ein molekularer Oszillator, das so genannte „Segmentierungs-Uhrwerk“. Die periodische Segmentierung des präsomitischen Mesoderms wird reguliert durch eine Reihe komplexer Interaktionen von unterschiedlichen Signale wegen. Der Notch/Delta Signalweg spielt dabei eine zentrale Bedeutung, da hierbei Komponenten entdeckt wurden, die während der Somitogenese zyklisch im PSM exprimiert werden. Außer dem Notch/Delta Signalweg spielen auch ein FGF und Retinolsäure Gradient, sowie Wnt Signale eine wichtige Rolle bei der Somitogenese. Trotz mehrerer Mutagenese Screens im Zebrafisch wurden bislang keine Mutanten im FGF oder Wnt Signalweg entdeckt, die einen spezifischen Somiten Defekt besitzen. Die wurde durch eine funktionelle Redundanz unterschiedlicher Gene erklärt, die durch eine Duplikation im Genom von Teleostieren vor 350 Millionen Jahren enstanden ist. Da unterschiedliche Duplikate in verschiedenen Fischspezies existieren, wurde in den letzten Jahren ein grosser Mutagenese Screen bei Medaka (Oryzias latipes) in Kyoto, Japan durchgeführt. In meiner Arbeit habe ich neue Somitogenese Mutanten aus dieser Screen isoliert und Phänotypisch charakterisiert. Die neun isolierten Mutanten zeigten unterschiedliche Somiten Phänotypen. Einige Mutanten hatten wenige oder gar keine Somitengrenzen (z.B bms oder sne), andere hatten unregelmäßige Somiten Formen (z.B. krz oder fsl) oder unterschiedlich große Somiten (z.B dpk). Manche dieser Medaka wiesen Änlichkeiten Mutanten zu im Zebrafisch beschriebenen Somiten Mutanten auf. Die Mehrzahl der Mutanten zeigten jedoch Phänotypen, die bis jetzt noch nicht in Zebrafisch Screens gefunden worden. In-Situ Analysen mit Hilfe unterschiedlicher, neu isolierter Somitenmarker, wie z.B. her7 einem Bestandteil des molekularen Oszillators, mesp einem anterior-posterioren Gen oder den Somitendifferenzierungsgenen lfng oder myf5 erlaubten, die Medaka Mutanten in zwei unterschiedliche Gruppen zuzuordnen. Gruppe I zeigt Defekte in der Bildung der Schwanzknospe und der Musterbildung im PSM. Ein besonderes Beispiel dieser Gruppe ist die Mutante doppelkorn (dpk), die einen bislang nicht beschriebenen Somitenphänotyp besitzt. In-situ Analysen von dpk zeigten, dass zyklische Gene im anterioren PSM dieser Mutante nicht stabilisiert werden und auch A-P Polaritätsgene fehlerhaft reguliert werden. Das deutet darauf hin, dass in der dpk Mutante ein Faktor der sogenannten „Wavefront“ betroffen sein könnte, der wichtig ist für die Regulation von oszillierenden Genen im anterioren PSM ist. Ich konnte zeigen, daß der wichtige Notch/Delta Signalweg in dieser Mutante nicht betroffen ist, weil alle unterschiedlichen zyklischen Gene, her1, her5 und her7, eine normale dynamische initiation ihrer Expression zeigten. Gruppe II Mutanten zeigen Defekte bei der Bildung der Somitengrenzen und Epithelialisierung der Somiten trotz normales, Musterbildung im PSM. Solche Mutanten wurden bislang weder in Zebrafisch, noch in Maus oder Hühnchen gefunden. Deshalb sollten nach der molekularen Identifiezierung der mutierten Gene neue Faktoren erhalten werden, die vor allem für die Regulation später Somitogenese-phasen wichtig sind. GDF5 is a susceptibility gene for osteoarthritis (OA) and mutations in GDF5 are associated with a wide variety of skeletal malformations ranging from complex syndromes such as acromesomelic chondrodysplasias to isolated forms of brachydactylies or multiple synostoses syndrome 2 (SYNS2). Functional studies, including chondrogenesis assays with primary mesenchymal cells, luciferase reporter gene assays and Surface Plasmon Resonance analysis, of the GDF5 W-414R variant in comparison to other GDF5 mutations associated with isolated BDA1 (p.R399C) or SYNS2 (p.E491K) revealed a dual pathomechanism characterized by a gain-and loss-of-function at the same time. On the one hand insensitivity to the main GDF5 antagonist NOGGIN (NOG) leads to a GDF5 gain of function and subsequent SYNS2 phenotype. Whereas on the other hand, a reduced signaling activity, specifically via the BMP receptor type IA (BMPR1A), is likely responsible for the BDA1 phenotype. These results demonstrate that one mutation in the overlapping interface of antagonist and receptor binding site in GDF5 can lead to a GDF5 variant with pathophysiological relevance for both, BDA1 and SYNS2 development. Consequently, our study assembles another part of the molecular puzzle of how loss and gain of function mutations in GDF5 affect bone development in hands and feet resulting in specific types of brachydactyly and SYNS2. These novel insights into the biology of GDF5 might also provide further clues on the pathophysiology of OA.
