Dendritic cells (DCs) and T cells are present within atherosclerotic lesions and function as central players in the initiation and modulation of adaptive immune responses. In previous years, we have studied the functional contribution of distinct DC subsets in disease development, namely, that of CCL17-expressing DCs as well as that of plasmacytoid DCs that play specialized roles in disease development. This review focuses on important findings gathered in these studies and dissects the multifaceted contribution of CCL17-expressing DCs and pDCs to the pathogenesis of atherosclerosis. Furthermore, an outlook on future challenges faced when studying DCs in this detrimental disease are provided, and hurdles that will need to be overcome in order to enable a better understanding of the contribution of DCs to atherogenesis are discussed, a prerequisite for their therapeutic targeting in atherosclerosis. Das entscheidende Ereignis in der Pathogenese dieser Entität stellt eine chromosomale Translokation dar, welche zur Entstehung eines chimären Transkriptionsfaktors, meist EWS-FLI1, führt. Unsere Absicht war es, die Mechanismen zu verstehen, die letztlich zur Metastasierung von Ewing-Sarkomen mit der damit verbundenen, infausten Prognose führen. Die Mitglieder der Zyxin-Proteinfamilie sind in vielfältige zelluläre Funktionen involviert. Hierbei nehmen sie, teilweise funktionell redundant, Einfluss auf zytoplasmatische und nukleäre Prozesse. Durch Analyse von öffentlich verfügbaren Microarraydaten konnten wir belegen, dass lediglich das Protein TRIP6 (thyroid receptor interacting protein 6) aus der Familie in EFT deutlich überexprimiert ist. Dieses Protein ist, neben seiner Funktion in der Organisation des Zytoskeletts, auch nukleär als Kotranskriptionsfaktor und als Element der Telomerprotektion tätig. Vielfach wurde eine Implikation des multifunktionellen Adaptorproteins in maligne Prozesse dokumentiert. Die Überexpression von TRIP6 in EFT ist jedoch unabhängig von EWS-FLI1. Eine Bindung von EWS-FLI1 an eine putative Bindungsstelle im Promotor von TRIP6 konnte nicht nachgewiesen werden. Die Analyse von Microarrays nach TRIP6-Knockdown in EFT-Zelllinien identifizierte mehrere Gensets, welche mit Proliferation und Invasivität assoziiert sind und die nach TRIP6-Knockdown vermindert exprimiert werden. Die für Malignome pathogenetisch relevanten Zielgene Radixin, CD164 und CRYZ konnten als Zielgene des Kotranskriptionsfaktors TRIP6 durch qRT-PCR und Western Blot bestätigt werden. Durch RNA-Interferenz-mediierte Verminderung der Proteinmenge von TRIP6 in EFT kam es zu einer deutlich reduzierten Klonogenität und Migration der Zellen in vitro. Nach induzierbarem TRIP6-Knockdown konnte eine verminderte Tumorigenität und hepatische Metastasierung von hierfür generierten EFT-Einzelzellklonen in vivo beobachtet werden. Zusammengefasst deuten diese Daten auf eine Rolle von TRIP6 in der Pathogenese der EFT und insbesondere beim Prozess der Metastasierung hin. Somit legen diese Ergebnisse eine weitere Evaluierung von TRIP6 als Biomarker oder molekulare Zielstruktur für therapeutische Ansätze in EFT nahe. N2 - Ewing’s sarcoma (ES) is the second most common bone-associated malignancy in children that is driven by the fusion oncogene EWS/FLI1. Despite the great propensity of ES toward early metastasis, recent evidence showed that EWS/FLI1 expression reduces migration and invasiveness of ES cells. However, we strived for exploring the underlying mechanisms of how ES maintains its basal migratory and invasive properties ultimately contributing to metastasis. We focused on the Zyxin-protein family comprising key players in actin remodeling, migration, and invasion. By interrogation of published microarray data, we observed that of all seven Zyxin-proteins only TRIP6 (thyroid receptor interacting protein 6) is highly overexpressed in ES. Besides its effects on cytoskeletal organization, TRIP6 is a nucleocytoplasmic shuttling protein involved in telomere protection, apoptosis, chemo-resistance, and transcriptional control. Initial experiments indicate that TRIP6 expression is independent of EWS/FLI1. However, RNAi-mediated knockdown of TRIP6 in ES cells significantly reduced clonogenicity and migration, whereas proliferation and adhesion were not reduced. As similar phenotypes have been described in Naxos disease and Carvajal syndrome, respectively, the genes for plakoglobin (JUP) and desmoplakin (DSP) were screened for mutations using direct genomic sequencing. A novel homozygous 2 bp deletion was identified in an alternatively spliced region of DSP. The deletion 5208_5209delAG led to a frameshift downstream of amino acid 1,736 with a premature truncation of the predominant cardiac isoform DSP-1. This novel homozygous truncating mutation in the isoform-1 specific region of the DSP C-terminus caused Carvajal syndrome comprising severe early-onset heart failure with features of non-compaction cardiomyopathy, woolly hair and an acantholytic form of palmoplantar keratoderma in our patient. Congenital hair abnormality and manifestation of the cutaneous phenotype in toddler age can help to identify children at risk for cardiac death. While the role of monocyte subset heterogeneity has already been well investigated in coronary artery disease (CAD), the knowledge about monocytes and their heterogeneity in peripheral artery occlusive disease (PAOD) still is limited. Therefore, we aimed to investigate monocyte subset heterogeneity in patients with PAOD. Peripheral blood was obtained from 143 patients suffering from PAOD (Rutherford stage I to VI) and three monocyte subsets were identified by flow cytometry: CD14\(^{++}\)CD16\(^{-}\) classical monocytes, CD14\(^{+}\)CD16\(^{++}\) non-classical monocytes and CD14\(^{++}\)CD16\(^{+}\) intermediate monocytes. Additionally the expression of distinct surface markers (CD106, CD162 and myeloperoxidase MPO) was analyzed. Proportions of CD14\(^{++}\)CD16\(^{+}\) intermediate monocyte levels were significantly increased in advanced stages of PAOD, while classical and non-classical monocytes displayed no such trend. Moreover, CD162 and MPO expression increased significantly in intermediate monocyte subsets in advanced disease stages. Likewise, increased CD162 and MPO expression was noted in CD14\(^{++}\)CD16\(^{-}\) classical monocytes. These data suggest substantial dynamics in monocyte subset distributions and phenotypes in different stages of PAOD, which can either serve as biomarkers or as potential therapeutic targets to decrease the inflammatory burden in advanced stages of atherosclerosis. One third of these patients will additionally develop pulmonary metastases. Methods 137 consecutive patients with CRLM, were analyzed regarding survival data, clinical, histological data and treatment. Results were stratified according to the occurrence of pulmonary metastases and metastases resection. Results 39% of all patients with liver resection due to CRLM developed additional lung metastases. 44% of these patients underwent subsequent pulmonary resection. Patients undergoing pulmonary metastasectomy showed a significantly better five-year survival compared to patients not qualified for curative resection (5-year survival 71.2% vs. 28.0%; p = 0.001). Interestingly, the 5-year survival of these patients was even superior to all patients with CRLM, who did not develop pulmonary metastases (77.5% vs. 63.5%; p = 0.015). Patients, whose pulmonary metastases were not resected, were more likely to redevelop liver metastases (50.0% vs 78.6%; p = 0.034). However, the rate of distant metastases did not differ between both groups (54.5 vs.53.6; p = 0.945). Conclusion The occurrence of colorectal lung metastases after curative liver resection does not impact patient survival if pulmonary metastasectomy is feasible. Those patients clearly benefit from repeated resections of the liver and the lung metastases. To prevent nephrotoxicity, positively charged amino acids (AA) are co-infused. The aim of this study was to correlate the risk for therapy-related hyperkalaemia with the total amount of AA infused. Methods Twenty-two patients undergoing PRRT with standard activities of 177Lu-DOTATATE/-TOC were monitored during two following treatment cycles with co-infusion of 75 and 50 g of AA (L-arginine and L-lysine), respectively. Mean serum levels of potassium and other parameters (glomerular filtration rate [GFR], creatinine, blood urea nitrogen [BUN], phosphate, chloride, lactate dehydrogenase) prior to, 4 h and 24 h after AA infusion were compared. Results Self-limiting hyperkalaemia (>5.0 mmol/l) resolving after 24 h occurred in 91% (20/22) of patients in both protocols. Potassium levels, BUN, creatinine, GFR, phosphate, chloride and LDH showed a similar range at 4 h after co-infusion of 75 or 50 g of AA, respectively (p > 0.05). Only GFR and creatinine levels at 24 h varied significantly between the two co-infusion protocols (p < 0.05). Conclusions Hyperkalaemia is a frequent side effect of AA infusion in PRRT. Varying the dose of co-infused amino acids did not impact on the incidence and severity of hyperkalaemia. Ideally, this application requires antibodies with high sensitivity and specificity. The present work evaluates the cross-reactivity of commercially available cyclic nucleotide analogs with two non-radioactive and one radioactive cAMP and cGMP immunoassay. Most of the tested cyclic nucleotide analogs showed low degree competition with the antibodies; however, with Rp-cAMPS, 8-Br-cGMP and 8-pCPT-cGMP, a strong cross-reactivity with the corresponding cAMP and cGMP, respectively, immunoassays was observed. The determined EIA-binding constants enabled the measurement of the intracellular cyclic nucleotide concentrations and revealed a time- and lipophilicity-dependent cell membrane permeability of the compounds in the range of 10–30% of the extracellular applied concentration, thus allowing a more accurate prediction of the intracellular analog levels in a given experiment. Zum ersten Mal war es auch möglich, funktionale Konsequenzen für STAT3 nach Stimulierung des BMP-Rezeptors 1a zu dokumentieren. Nach Belegung des BMP-Rezeptors 1a mit dem mutierten BMP2-A34D wird STAT3 trotz Aktivierung durch Phosphorylierung an Tyrosin 705 im Zytoplasma von Maus Stammzellen festgehalten. Beide Vorgaenge werden hauptsaechlich durch den Thrombozyten-Rezeptor Integrin alphaIIb-beta3 vermittelt. Nach Bindung des Liganden Fibrinogen aendert sich die Rezeptor-Konformation, Integrine assoziieren und ein intrazellulaeres Signalnetzwerk wird aktiviert, welches die Organisation des Aktin-Zytoskeletts steuert. Diese Zytoskelett-Reorganisationen sind Grundlage für zellulaere Adhaesions- und Aggregations-Prozesse. Die Signalvermittlung vom Integrin zum Zytoskelett wird durch die Protein-Tyrosinkinase Src eingeleitet, deren Aktivitaetszustand den Signalweg reguliert. Bei der Src-Aktivierung wird Tyrosin 418 durch Autokatalyse phosphoryliert. Die Kinase muss jedoch wieder inaktiviert werden. Dies übernimmt in Plaettchen ausschliesslich die Tyrosinkinase Csk (C-terminale Src Kinase) durch Phosphorylierung von Tyrosin 529 im C-terminalen Ende des Proteins. Die Csk-vermittelte Inaktivierung von Src stellt den entscheidenden Kontrollschritt des alphaIIb-beta3-vermittelten Signalwegs dar. Obwohl bekannt ist, dass die Src-Aktivierung bei der Zelladhaesion an den Zellraendern der Lamellipodien geschieht und man den Mechanismus und die Kinetik der Src-Csk Interaktion genauer versteht, ist bislang immer noch unbekannt, wo und wie Src inaktiviert wird bzw. welche Rolle der Src-Inaktivierung genau zukommt. FRET (Fluoreszenz-Resonanz-Energie-Transfer) ist ein physikalischer Effekt, mit dem Interaktionen beliebiger fluoreszenzmarkierter Proteine mikroskopisch detektiert werden koennen. Diese Technik wurde genutzt, um die Src-Csk-Interaktion waehrend der alphaIIb-beta3-vermittelten Fibrinogen-Adhaesion in einer etablierten Thrombozyten-Modellzelllinie (A5-CHO) direkt visualisierbar zu machen. Es zeigten sich starke Src-Csk Interaktionen (FRET-Signale) an den Zellraendern aktiver Lamellipodien und zusaetzlich in Fokalkontakten, wo beide Proteine mit Vinculin, einem Fokalkontakte-Marker, co-lokalisierten. Die Proteininteraktionen folgten einem hochdynamischen Ablauf. Nach der Akkumulation der Src-Csk Komplexe an den Zellraendern wanderten sie in Abstaenden von 2-3 Minuten nach innen, fragmentierten und bildeten schliesslich stabile Fokal-Adhaesionen. FRET-Signale an den Zellraendern fanden sich vor allem in ruhenden Lamellipodien bzw., waehrend des Lamellipodien-Rückzugs, in wachsenden Lamellipodien traten die FRET-Signale dort dagegen nicht auf. In unabhaengigen biochemischen Tests im Zeitfenster der FRET-Beobachtungen wurde ein spezifischer Anstieg der Src-Tyr529-Phosphorylierung (Inaktivierung) und eine parallele Abnahme der Src-Tyr418-Phosphorylierung (Aktivierung) gemessen. Weiterführende Ergebnisse lieferten Versuche mit Src- und Csk-Mutanten. Die Co-Expression von Wildtyp-Src mit Kinase-inaktivem CskK222R hatte weder einen Effekt auf die Adhaesion und Ausbreitung der Zellen noch auf die Praesenz von FRET, es aenderte sich jedoch drastisch die zellulaere Verteilung der FRET-Signale sowie das Wachstum und die Form der Lamellipodien. Die Co-Expression von Wildtyp-Csk mit konstitutiv aktivem SrcY529F verursachte dagegen eine stark verringerte Adhaesionsfaehigkeit und Hemmung der Lamellipodien-Bildung. Die Fokal-Adhaesionspunkte in diesen Zellen waren sehr schwach und ueberdimensioniert und lagen ungeordnet verteilt in der Adhaesionsebene. Zusaetzlich verursachte SrcY529F eine starke Ueberaktivierung des Zytoskeletts und das fast vollstaendige Verschwinden der FRET-Signale. Die ermittelten Daten zeigen, dass die enge Kontrolle der Src-Aktivitaet durch Csk eine bedeutende Rolle für die funktionelle Zell-Adhaesion and -Ausbreitung spielt. Co-Immunpraezipitations-Resultate und Messungen der Menge an markiertem Protein in Zellen, in welchen FRET detektierbar war, untermauern zusaetzlich unsere These, zum ersten Mal die Src-Regulation durch Csk in lebenden Zellen direkt beobachtbar gemacht zu haben. Dieser neue FRET-Ansatz kann auch als Reporter-System für Prozesse der Src-Inaktivierung in anderen Signalwegen und Zellen angewendet werden. Das Messprinzip kann weiterhin auf das Studium der Inaktivierung weiterer Mitglieder der Familie der Src-Kinasen (in verschiedensten Signalwegen) erweitert werden. N2 - Platelet adhesion and thrombus formation required for functional hemostasis depends on integrin receptor mediated “outside-in” signaling to the cytoskeleton. Integrin alphaIIb-beta3 is the major integrin on the platelet surface and acts as a specific receptor for the plasma protein fibrinogen. Fibrinogen binding causes clustering of integrins within the plasma membrane activating the protein tyrosine kinase Src (signal initiation) by phosphorylation of tyrosine 418. Src, however, is negatively regulated by another tyrosine kinase, Csk (C-terminal Src kinase), which phosphorylates tyrosine 529. Although, in adhering cells, it is believed that Src is getting activated at lamellipodia leading edges, neither the cellular location nor the dynamics and exact role of Src inactivation is known to date. Here, we studied Src inactivation during alphaIIb-beta3-dependent adhesion to fibrinogen in the established platelet model cell line A5-CHO. Using a live cell FRET (fluorescence resonance energy transfer) microscopy technique with CFP and YFP label molecules (cyan and yellow fluorescent protein), we were able to image highly dynamic Src-Csk interactions at the leading edges of active lamellipodia. Every 2-3 minutes, signals detecting Src-Csk interactions (complexes) appeared at the cell periphery before they begin to move inward in the cell and reorganize while lamellipodia start to protrude (grow). FRET signals were also found in small accumulations at the fringe and also further to the centre of the adhesion plane (focal complexes and adhesions). Src and Csk co-localize with vinculin (a focal adhesion marker) within these regions. During the runtime of FRET observation a specific increase in Src-Tyr529 phosphorylation with a parallel decrease in Src-Tyr418 phosphorylation was observed supporting the idea that Src inactivation occurs within the cells. The role of Src-Csk interaction was studied in further detail using Src and Csk mutants. The data revealed that co-expression of inactive CskK222R did not alter the presence of FRET signals, but fundamentally changed its distribution within the cell. Furthermore it caused lamellipodia shape changes and a tendency of constant lamellipodia protrusion. Co-expression of constitutively active SrcY529F in turn caused a severe adhesion and spreading dysfunction. Adherent cells showed very weak, disorganized and oversized focal adhesions, a hyper-activated cytoskeleton (visible in fast-changing membrane blebs) and absence of FRET signals. Results from immunoprecipitation analyses and protein level determination within cells, in which FRET was detectable, further supported that we were able, for the first time, to directly visualize Src (and integrin) regulation by Csk control in live cells. The results show that Src control by Csk is ultimately required for lamellipodia and focal adhesion function and thus for cell anchorage and spreading. The novel FRET-approach reported here can be readily applied to other integrin and signaling pathways including the study of closely related Src family kinases (SFKs). Results may also contribute to a better understanding of the processes of tumor formation. Its expression is increased in many malignant tumors. However, little is known about the physiological role of the protein. In the present study, we investigated the expression and function of LASP1 in normal skin, melanocytic nevi and malignant melanoma. In normal skin, a distinct LASP1 expression is visible only in the basal epidermal layer while in nevi LASP1 protein is detected in all melanocytes. Melanoma exhibit no increase in LASP1 mRNA compared to normal skin. In melanocytes, the protein is bound to dynamin and mainly localized at late melanosomes along the edges and at the tips of the cell. Knockdown of LASP1 results in increased melanin concentration in the cells. Collectively, we identified LASP1 as a hitherto unknown protein in melanocytes and as novel partner of dynamin in the physiological process of membrane constriction and melanosome vesicle release.
