In addition, hybridisation between plants is well documented as a source of new weeds, as hybrids gain new characteristics that allow them to become invasive. Corymbia torelliana is an invasive tree with an unusual animal dispersal mechanism: seed dispersal by stingless bees, that hybridizes readily with other species. We examined hybrids between C. torelliana and C. citriodora subsp. citriodora to determine whether hybrids have inherited the seed dispersal characteristics of C. torelliana that allow bee dispersal. Some hybrid fruits displayed the characteristic hollowness, resin production and resin chemistry associated with seed dispersal by bees. However, we did not observe bees foraging on any hybrid fruits until they had been damaged. We conclude that C. torelliana and C. citriodora subsp. citriodora hybrids can inherit some fruit characters that are associated with dispersal by bees, but we did not find a hybrid with the complete set of characters that would enable bee dispersal. However, around 20,000 hybrids have been planted in Australia, and ongoing monitoring is necessary to identify any hybrids that may become invasive. In addition to testing according to current guidelines designed to detect bee mortality, tests are needed to determine possible sublethal effects interfering with the animal's vitality and behavioral performance. Several methods have been used to detect sublethal effects of different insecticides under laboratory conditions using olfactory conditioning. Furthermore, studies have been conducted on the influence insecticides have on foraging activity and homing ability which require time-consuming visual observation. We tested an experimental design using the radiofrequency identification (RFID) method to monitor the influence of sublethal doses of insecticides on individual honeybee foragers on an automated basis. With electronic readers positioned at the hive entrance and at an artificial food source, we obtained quantifiable data on honeybee foraging behavior. This enabled us to efficiently retrieve detailed information on flight parameters. We compared several groups of bees, fed simultaneously with different dosages of a tested substance. With this experimental approach we monitored the acute effects of sublethal doses of the neonicotinoids imidacloprid (0.15-6 ng/bee) and clothianidin (0.05-2 ng/bee) under field-like circumstances. At field-relevant doses for nectar and pollen no adverse effects were observed for either substance. Both substances led to a significant reduction of foraging activity and to longer foraging flights at doses of >= 0.5 ng/bee (clothianidin) and >= 1.5 ng/bee (imidacloprid) during the first three hours after treatment. This study demonstrates that the RFID-method is an effective way to record short-term alterations in foraging activity after insecticides have been administered once, orally, to individual bees. We contribute further information on the understanding of how honeybees are affected by sublethal doses of insecticides. Da die Proteine der Atmungsketten-komplexe sowohl durch die mitochondriale DNA als auch durch das Kerngenom codiert werden, können Mutationen in beiden Genomen die Auslöser dieser Erkrankungen darstellen. Veränderungen der mitochondrialen DNA lassen sich - im Gegensatz zum Kerngenom - bisher nicht korrigieren, weshalb bei einem großen Teil der Erkrankungen nur die Symptome und nicht die Auslöser behandelt werden können. Das grundlegende Problem stellt dabei der Transport der DNA in die Mitochondrien dar. Ziel dieser Arbeit war es, mit Hilfe von physikalischen Transfektionsmethoden exogene DNA in die Mitochondrien menschlicher Kulturzellen einzubringen. Dazu wurden unterschiedliche Vektoren hergestellt, die in Mitochondrien das an die Mitochondrien angepasste grün fluoreszierende mtEGFP exprimieren sollen. Die Expressionsfähigkeit und Prozessierung dieser Konstrukte konnte in in-vitro-Assays mit einem Mitochondrienextrakt nachgewiesen werden. Bei Transfektionsversuchen mit der Gene Gun gelang es erstmals, exogene Plasmid-DNA in die Mitochondrien menschlicher Zellen einzubringen. Das durch die transfizierten Vektoren exprimierte mtEGFP konnte am Fluoreszenzmikroskop eindeutig in den Mitochondrien der Zellen lokalisiert werden. Eine Transfektion mit Hilfe magnetischer Partikel erwies sich jedoch nicht als zielführend, da die die Partikel eine Eigenfluoreszenz aufwiesen, die eine Detektion der mtEGFP-Expression verhinderten. Eine wichtige Voraussetzung für die Transfektion von Mitochondrien durch mechanische Methoden wie die Mikroinjektion ist die reversible Induktion von Megamitochondrien, da sie erst in diesem Zustand penetriert werden können. Durch eine Ansäuerung des Kulturmediums mit Natriumacetat bzw. Essigsäure konnten Mitochondrien erzeugt werden, die beinahe die Größe des Zellkerns aufwiesen und somit ideale Bedingungen für die Mikroinjektion darstellen. Bei den anschließenden Mikroinjektionsversuchen mit den hergestellten mitochondrialen Expressionsvektoren wurden wiederum Zellen mit eindeutig grün fluoreszierenden Mitochondrien gefunden. Zusammenfassend wurden im Rahmen dieser Arbeit erstmalig menschliche Mitochondrien mit exogener DNA transfiziert. Dies stellt einen grundlegenden Schritt für die Entwicklung neuer Therapieformen bei mitochondrialen Myopathien dar. N2 - Mitochondrial dysfunctions play an important role in a variety of neuromuscular and neurodegenerative diseases. As the proteins of the respiratory chain complexes are encoded by the mitochondrial DNA as well as the nuclear genome, mutations in both could trigger solely the diseases. Up to now, changes of the mitochondrial DNA could not be corrected, hence, therapies were designed to decrease symptoms in patients. In this context, the limiting factor to cure these disease relies on the DNA transport into the mitochondria. The aim of this work was to insert exogenous DNA into the mitochondria of human cultured cells by physical transfection methods. A variety of different vectors was constructed to express the mitochondrially adapted green fluorescent mtEGFP within mitochondria. The ability of these constructs to be expressed and processed was proved by in vitro assays using a mitochrondrial extract. In the transfection experiments using the gene gun, we succeeded for the first time to introduce exogenous plasmid DNA into human mitochondria. The mtEGFP expressed by the transfected vectors could definitely be localized to the mitochondria of the cells. Transfections using magnetic particles as mediator could not be used, because the particles exhibit an autofluorescence which prevents a detection of the mtEGFP expression. An essential prerequisite for the transfection of mitochondria by mechanical methods like microinjection is the reversible induction of megamitochondria, since they could not be penetrated as small regular organelles. Acidification of the culture medium with sodium acetate or acetic acid led to mitochondria exhibiting almost the size of the nucleus, thus giving ideal conditions for microinjection. In the applied microinjection experiments using the mitochondrial expression vectors, cells displayed mitochondria with distinct green fluorescence. In summary, human mitochondria were transfected successfully for the first time with mitochondrial expression vectors. This is a fundamental step in the development of new therapies targeting mitochondrial myopathies. However, the transfection methods have to be optimized to achieve higher transfection efficiencies. Die vorliegende Arbeit untersucht die Funktion und Expression von CD28 in Schweine-T-Zellen sowie die Regulierbarkeit der T-Zellaktivierung durch anti-pCD28 mAb. Die Ergebnisse zeigen, dass hierbei vor allem CD4+ und CD8+ T-Zellen differenziert betrachtet werden müssen. Grundsätzlich unterscheiden sich die beiden T-Zellpopulationen in der CD28 mRNA Expression, im Expressionsverhältnis zwischen CD28 mRNA und Protein, sowie im proliferativen Ansprechen auf anti-pCD28mAb. So reagierten CD4+ im Vergleich zu CD8+ T-Zellen auf die kostimulatorische Inkubation mit anti-pCD28 mAb des Klons 3D11 sensibler. In direkt stimulatorischen Ansätzen zeigte sich, dass CD4+ und CD8+ T-Zellen durch unterschiedliche anti-pCD28 mAb differentiell angesprochen werden können. Therefore, our work aimed at investigating the function and expression of CD28 in porcine T cells as well as the applicability of anti-pCD28 mAb as a tool to regulate T cell activation, specifically. Our results indicate that the two major T cell groups, CD4+ and CD8+, have to be considered separately. These cell populations differ in their level of pCD28 mRNA expression, their ratio of CD28 mRNA and protein expression as well as their proliferative responsiveness towards anti-pCD28 mAb. In costimulatory assays, CD4+ compared to CD8+ T cells showed higher sensitivity toward low concentrations of the anti-pCD28 mAb 3D11. Additionally, in direct stimulatory assays, CD4+ and CD8+ T cells can be specifically targeted by different anti-pCD28 mAb. With these assays, a superagonistic anti-pCD28 mAb could not been detected so far. With anti-pCD28 superagonist, promising therapeutic strategies developed in rodents could be tested in pigs as a large animal model, representing the next step on the way towards new therapeutic options for autoim-mune diseases, diseases with strong proinflammatory activity and myocardial infarction. They respond to phosphoantigens (PAgs) like isopentenyl pyrophosphate or (E)-4-hydroxy-3-methyl-but-2-enyl-pyrophosphate (HMBPP) in a butyrophilin 3 (BTN3)-dependent manner and may contribute to the control of mycobacterial infections. These cells were thought to be restricted to primates, but we demonstrated by analysis of genomic databases that TRGV9, TRDV2, and BTN3 genes coevolved and emerged together with placental mammals. Furthermore, we identified alpaca (Vicugna pacos) as species with typical Vγ9Vδ2 TCR rearrangements and currently aim to directly identify Vγ9Vδ2 T cells and BTN3. Other candidates to study this coevolution are the bottlenose dolphin (Tursiops truncatus) and the nine-banded armadillo (Dasypus novemcinctus) with genomic sequences encoding open reading frames for TRGV9, TRDV2, and the extracellular part of BTN3. Dolphins have been shown to express Vγ9- and Vδ2-like TCR chains and possess a predicted BTN3-like gene homologous to human BTN3A3. The other candidate, the armadillo, is of medical interest since it serves as a natural reservoir for Mycobacterium leprae. In this study, we analyzed the armadillo genome and found evidence for multiple non-functional BTN3 genes including genomic context which closely resembles the organization of the human, alpaca, and dolphin BTN3A3 loci. However, no BTN3 transcript could be detected in armadillo cDNA. Additionally, attempts to identify a functional TRGV9/TRGJP rearrangement via PCR failed. In contrast, complete TRDV2 gene segments preferentially rearranged with a TRDJ4 homolog were cloned and co-expressed with a human Vγ9-chain in murine hybridoma cells. These cells could be stimulated by immobilized anti-mouse CD3 antibody but not with human RAJI-RT1Bl cells and HMBPP. So far, the lack of expression of TRGV9 rearrangements and BTN3 renders the armadillo an unlikely candidate species for PAg-reactive Vγ9Vδ2 T cells. This is in line with the postulated coevolution of the three genes, where occurrence of Vγ9Vδ2 TCRs coincides with a functional BTN3 molecule.
