Es handelt sich hierbei um einen klassischen und einen kombinatorischen Ansatz. Der klassische Ansatz basiert auf einer Struktur, deren Aktivität per Zufall entdeckt wurde. In Screenings von synthetisierten Strukturanaloga, gestützt durch virtuelle Docking-Experimente am aktiven Zentrum der Cysteinproteasen, wurden Struktur-Aktivitäts-Beziehungen erarbeitet. Bei der kombinatorischen Methode wurde zunächst ein peptidischer Inhibitor entworfen, der durch Festphasensynthese an einem geeigneten Harz synthetisiert wurde. Durch den kombinatorischen Einsatz von Aminosäuren konnte auf diese Weise unter enormer Zeitersparnis eine Bibliothek von 150 Inhibitoren synthetisiert werden. In einem Screening dieser harzgebundenen Inhibitoren wurden anschließend die potentesten Inhibitoren identifiziert. Die Aktivität der gefundenen Inhibitoren aus beiden Ansätzen an den protozoischen Erregern wurde durch in-vitro-Experimente an Plasmodien und Trypanosomen untersucht. Beim klassischen Ansatz wurde eine neue Substanzklasse entwickelt, die sehr gute Hemmeigenschaften an beiden Cysteinproteasen mit IC50-Werten im niedrigen mikromolaren Bereich zeigten. Außerdem besaßen sie eine hohe in-vitro-Aktivität gegenüber den Erregern im gleichen Konzentrationsbereich. Einige der Inhibitoren zeigten keine Zytotoxizität an Makrophagen. Aus dem klassischen Ansatz konnten also hochaktive Substanzen mit geringer Zytotoxizität entwickelt werden, deren Einsatz als Wirkstoffe gegen Malaria oder der Schlafkrankheit denkbar ist. Für den kombinatorischen Ansatz wurde zur Inhibitoroptimierung eine Screeningmethode für Falcipain-2 und Rhodesain direkt an einem geeigneten Harz zur Festphasensynthese entwickelt. Neu bei dieser Screeningmethode war es, dass erstmals ein quantitatives Screening einer Inhibitorbibliothek möglich sein sollte, und nicht nur die besten Inhibitoren identifiziert werden können. Aus den Ergebnissen der Festphasenscreenings an beiden Proteasen wurden 14 besonders interessante Inhibitoren der Bibliothek ausgewählt und synthetisiert. Diese Verbindungsklasse zeigte ebenfalls sehr gute Ergebnisse an den isolierten Enzymen, in den mikrobiologischen Tests an den Erregern jedoch fielen alle Ergebnisse vergleichsweise schlechter aus. Die schlechte Löslichkeit, die Bioverfügbarkeit und der Metabolismus durch den Erreger der peptidischen Inhibitoren schienen von großer Bedeutung zu sein. Der bearbeitete kombinatorische Ansatz lieferte eine neuartige Screeningmethode, die auch auf andere Targets anwendbar ist.show moreshow less

That was a classical and a combinatorial method. The classical approach is based on a compound with a high activity against falcipain-2 which was discovered by chance. The structure-reactivity relations were to be developed in screenings ofThe main goal of the thesis “Development of cysteine protease inhibitors – a classical and a combinatorial approach for inhibitor optimization” was the optimization of new inhibitors of falcipain-2 and rhodesain as new potential agents against malaria and the sleeping sickness by two different methods. That was a classical and a combinatorial method. The classical approach is based on a compound with a high activity against falcipain-2 which was discovered by chance. The structure-reactivity relations were to be developed in screenings of analogous compounds and by computational docking experiments in the active site of the cysteine proteases. The combinatorial method was a completely different procedure. Initially a peptidic inhibitor had to be designed that could be introduced via solid phase synthesis on a suitable resin. By the combinatorial variation of amino acids 150 inhibitors could be obtained in a very short time. In a screening of these resin-bound compounds the active members of the library were identified. The antiplasmodial and antitrypanosomal activity of the inhibitors of both approaches were determined by biological in vitro investigations with the protozoic pathogens. The classical approach resulted in substances that showed excellent inhibitory properties of both cysteine proteases with IC50 values in the lower micromolar range. Additionally, they have a high in-vitro activity against the pathogens in the same concentration region. Some of the inhibitors showed no cytotoxicity on macrophages. From the classical approach we derived highly active substances with little cytotoxicity which are promising agents against malaria or the sleeping sickness. For the combinatorial approach, first a new screening method for falcipain-2 and rhodesain performed directly on a resin for solid phase synthesis was developed. Advantages of this new screening method are that it is possible to screen the library quantitatively. The results of the solid phase screenings with both proteases led to the choice of 14 notably interesting inhibitors and their synthesis. All results from the microbiological tests with the pathogens were worse compared to the results from the assays with the isolated enzymes. The bad solubility, the bioavailability and the metabolism by the pathogen played a role in inhibition. Compounds In summary a new screening method was developed and applied.
