  • The afferent function (excitability) and efferent function (release of neuropeptides) of primary afferent nerve fibres is based on Ca2+-influx. Aim of the present study was to examine the presence of L-and N-type Ca2+-channels at sensory nerve fibres in the mouse knee joint capsule. Specific fluorescent labelled channel blocker and antisera against these channel subtypes were combined with an immunhistochemical staining for calcitonin gene-related peptide (CGRP), a neuropeptide that is wildly distributed in primary afferents. There was nearly complete colocalisation of calcitonin gene-related peptide immunoreactivity and the binding of conotoxin GVIA or BODIPY- verapamil demonstrating the presence of N-type and L-type Ca2+-channels, respectively.
