Diese Arbeit zeigt, dass primäre, HIV-uninfizierte CD4+ Zellen in Kontakt mit HIV-infizierten Zellen zugrundegehen. Zur Aufklärung des Mechanismus dieses Zelltodes wurden einzelne Schritte der Zellfusion zwischen infizierten und uninfizierten Zellen mit Antikörpern und Peptiden blockiert, und die Auswirkung dieser Blockade auf den Zelltod bestimmt.   • The pathomechanism of the destruction of T-helpercells in the course of HIV-infections is still a matter of debate. The objective of this study was to investigate whether uninfected CD4+ primary PBMC are killed when getting in contact with HIV-infected primary PBMC. The destruction of uninfected primary CD4+ cells in contact with HIV-infected cells of ENF-expressing cells was shown with different techniques. Furthermore the individual steps of cell fusion were inhibited using antibodies and peptides directed against CD4, gp120 and gp41. Any block in the fusion process let to an inhibition of cell death indicating that the death of uninfected T-helpercells is a consequence of cell cell fusion. It was also shown that apoptosis can be observed during this cell lysis, however the inhibition of apoptosis did not lead to an inhibition of cell death.show moreshow less
