- Die opportunistische humanpathogene Hefe Candida glabrata ist zum zweithäufigsten Erreger einer Candidose avanciert. Infektionen mit Candida glabrata sind schwierig zu behandeln und bergen eine hohe Sterblichkeit bei Risikopatienten in sich. Dies liegt vor allem in der geringen Sensitivität von C. glabrata gegenüber den etablierten Azol-Derivaten begründet. Deswegen ist man bei C. glabrata, wie auch bei anderen pathogenen Pilzen, auf der Suche nach geeigneten Zielstrukturen für neue Antimykotika. Die 1-3-ß-D-Glucan-Synthetase ist ein pilzspezifisches Enzym, welches beim Menschen nicht vorkommt. Sie ist am Zellwandaufbau der Hefen beteiligt und wird durch die FKS Gene codiert. Die 1-3-ß-D-Glucan-Synthetase-Hemmer sind wirksam gegenüber Pilzen, die gegenüber anderen Antimykotika resistent sind. Dies führte zur Entwicklung der Echinocandine, echinocandin ähnlicher Lipopeptide und Pneumocandine, wovon einige in klinische Versuche aufgenommen wurden. In dieser Arbeit wurde ein FKS homologes Gen von C. glabrata identifiziert, kloniert und sequenziert. Die erhaltene Gensequenz des gefundenen FKS-Homologs von Candida glabrata wurde in der GenBank des National Center for Biotechnological Information veröffentlicht.…

In common with other fungal pathogens, there is a lack of defined C. glabrata targets which can be exploited for the rational design of novel antifungal drugs.
