Hinweise zu finden, die zum besseren Verständnis des Lösungsprozesses führen, ist das Ziel dieser Arbeit. Auf der Grundlage des Hess’schen Satzes wird der Lösungsprozess in Ersatzprozesse zerlegt. Die Teilschritte der Ersatzprozesse werden thermodynamisch, kalorimetrisch und quantenchemisch beschrieben. Freie Energien, Standardenthalpien und Standardentropien der Teilschritte werden erfasst und beurteilt. Weitere physikochemische Größen werden gemessen oder berechnet, die mit den einzelnen Teilschritten in Zusammenhang stehen können. Einfache Naphthalinderivate dienen als Modellsubstanzen. Dies sind zwei einfach und sieben zweifach hydroxylierte Naphthaline, 1-Naphthylamin, 4-Chlor-1-naphthol und 2-Naphthalinthiol. Zum einen wird der Lösungsvorgang als Summe aus Sublimation und anschließender Solvatation betrachtet. Die Freien Sublimationsenergien werden aus dem experimentell ermittelten Sublimationsdampfdruck berechnet. Dies geschieht mittels einer Hochvakuumapparatur oder einer gaschromatographischen Methode. Je mehr polare Gruppen am Grundgerüst sind, desto höher ist der energetische Aufwand, das Molekül zu sublimieren. Die Freie Energie beträgt für Naphthalin 30,5 [kJ·mol-1], für einfach substituierte Naphthaline 40,1 und 45,3 [kJ·mol-1] und für zweifach hydroxylierte Naphthaline 51,8 bis 67,5 [kJ·mol-1]. In ähnlichem Verhältnis stehen auch die Freien Solvatationsenergien. Diese werden mittels AMSOL, eines quantenchemischen semiempirischen Rechenprogramm, für zwei Parametrisierungen AM1 und PM3 berechnet. Die Löslichkeit der Substanzen in Wasser wird ermittelt, und daraus die Freie Lösungsenergie berechnet. Der Unterschied in den Freien Lösungsenergien ist trotz des größeren Energieaufwandes der substituierten Derivate bei der Sublimation um ca. 10 [kJ·mol-1] je polarer Gruppe nicht deutlich. Die Ursache ist die Energiefreisetzung während der Solvatation, die mit zunehmenden polaren Gruppen mehr Energie freisetzt. Gründe dafür sind vermehrte Wasserstoffbrückenbindungen und stärkere Dipol-Dipol-Wechelswirkungen der höher substituierten Naphthaline. Ein den Freien Energien ähnliches Bild ergibt sich bei der Betrachtung der Standardenthalpien. Die Standardsublimationsenthalpie wird aus Sublimationsdrücken bei unterschiedlichen Temperaturen berechnet, die Standardlösungsenthalpie mittels Kalorimetrie und die Standardsolvatationsenthalpie aus der Differenz der beiden vorangegangenen. Adsorptionsmessungen werden durchgeführt, um kalorimetrische Falschmessungen durch Adsorption von Wasser an die Substanzen auszuschließen. Die Ursachen für ein den Freien Energien ähnliches Verhalten sind die gleichen. Eine Ausnahme bildet 1,3-Dihydroxynaphthalin, bei dem die Standardlösungsenthalpie um den Faktor 4 größer ist als bei den restlichen Dihydroxynaphthalinderivaten. Die Standardentropien werden aus den vorher gewonnenen Größen berechnet. Die Standardsublimationsentropien der untersuchten Substanzen liegen zwischen 162 und 350 [J·mol-1·K-1]. Ebenso steigen sie beim Lösevorgang mit Ausnahme von 2-Naphthalinthiol. Dort liegt die Ursache in der mangelnden Hydrophilie, so dass sowohl beim Lösen als auch beim Mischen der Ordnungszustand zunimmt. Bei der Solvatation sinkt die Entropiealler Substanzen mit Ausnahme von 1,3- Dihydroxynaphthalin. Eine Beurteilung aufgrund der Entropien zwischen den Naphthalinen zu machen, ist anhand der Messergebnisse nicht möglich. Die zweite Betrachtungsweise legt dem Lösungsprozess das Schmelzen und anschließende Mischen der Substanz mit dem Lösungsmittel zugrunde. Die Standardschmelzenthalpie wird kalorimetrisch aus Schmelzpunkt, Differenz der Wärmekapazitäten und der Schmelzwärme am Schmelzpunkt ermittelt. Die Standardmischungsenthalpien werden aus der Differenz von kalorimetrisch gemessener Standardlösungsenthalpie und Standardschmelzenthalpie berechnet. Die Standardschmelzenthalpien der Dihydroxynaphthaline sind geringer. Dies hat allerdings in den höheren Schmelzpunkten seine Ursache. Das chlorierte 1-Naphthol liegt mit seinem Schmelzverhalten im Bereich des einfachen 1-Naphthols, so dass das Chloratom scheinbar keine große Rolle im Schmelzprozess spielt. Die Änderungen der Entropien während des Schmelzprozesses werden aus den gleichen Größen wie die Standardschmelzenthalpie gewonnen. Die Mischungsentropie wird aus der Differenz von Schmelz- und Lösungsentropie berechnet. Die Ordnung während des Mischungsvorganges nimmt bei allen untersuchten Substanzen zu außer bei 2-Naphthalinthiol und Naphthalin. Beide hydrophoben Moleküle haben in wässriger Lösung weniger Möglichkeiten sich anzuordnen als in der Schmelze. In die Berechnung der Schmelzentropien gehen die Schmelztemperaturen mit ein. Je höher der Schmelzpunkt ist, desto niedriger ist die Standardschmelzentropie. Die Solvatationsenergien werden mittels AM1- und PM3-Parametrisierung in AMSOL berechnet und mit dem experimentell ermitteltem Wert verglichen. Die Berechnungsmethoden zeigen eine statistisch nicht unterscheidbare gleiche Korrelation von r2 = 0,97 für AM1 und r2 = 0,98 für PM3. Die Berechnung der Freien Solvatationsenergie setzt sich aus einzelnen Teilbeträgen zusammen. Ein Teil ist die freiwerdende Polarisationsenergie, die mit zunehmender Zahl polarer Substituenten ansteigt, da Partialladungen häufiger auftreten und polarisiert werden können. Auch die Oberflächenenergie trägt zur stärkeren Solvatation polar substituierter Naphthaline bei. Van der Waals-Oberflächen und –Volumina der untersuchten Moleküle werden mittels AMSOL berechnet. Die Oberflächen der Dihydroxynaphthaline liegen bei ca. 375 [Ǻ2], die einfach substituierten darunter. Die Volumina der Dihydroxynaphthaline werden zu ca. 146 [Ǻ3] berechnet. Berechnete logarithmierte Oktanol/Wasser-Verteilungskoeffizienten werden mit den experimentell ermittelten verglichen. Zur Berechnung wird eine AM1- und PM3-Parametrisierung verwendet. Beide Parametersätze können nur unzureichend Oktanol/Wasser-Verteilungskoeffizienten berechnen. Die Bestimmtheitsmaße r2 liegen unter 0,79. Modellrechnungen weisen als abschließende Beurteilung auf einzelne Größen hin, die mit Freier Sublimationsenergie, Freier Solvatationsenergie und Freier Lösungsenergie in engerem Zusammenhang stehen. Die Einflussgrößen werden auf maximal drei festgelegt, um eine Zufallskorrelation auszuschließen. Es lässt sich ein Modell für die Freie Sublimationsenergie berechnen, das für AM1-Berechnungen ein Bestimmtheitsmaß von r2 = 0,984 zeigt und die Moleküloberfläche, die Wechselwirkung von Polarisationsenergie und Molekülvolumen und das quadrierte Molekülvolumen mit einbezieht. Für PM3-Berechnungen werden das Molekülvolumen, das Dipolmoment in der Gasphase und die Wechselwirkung von Polarisationsenergie und Molekülvolumen herangezogen, wobei das Bestimmtheitsmaß r2 = 0,986 ist. Die experimentell ermittelte Freie Sublimationsenergie der untersuchten Substanzen kann durch drei berechnete Parameter beschrieben werden. Beim Modell zur Freien Solvatationsenergie werden Polarisationsenergie, Oberflächenenergie und Schmelztemperatur als die entscheidenden Einflussparameter herangezogen. Das Bestimmtheitsmaß r2 erreicht 0,979 für AM1-Berechnungen und 0,983 für PM3-Berechnungen. Das Modell zur Berechnung der Freien Lösungsenergie beruht auf der Polarisationsenergie, der Schmelztemperatur und dem Molekülvolumen bei AM1-Berechnungen. Statt des Molekülvolumens wird bei PM3-Berechnungen als dritte Größe die Schmelzwärme herangezogen. Die Korrelation ist mit r2 = 0,847 bei AM1-Berechnungen und r2 = 0,879 bei PM3-Berechnungen nicht zufällig, aber nicht so deutlich wie bei den beiden anderen Modellierungen. Ausblickend lässt sich festhalten, dass auch 13 Substanzen für eine umfassende theoretische Erfassung der Ersatzprozesse zum Lösungsvorgang nicht ausreichen. Ein wichtiger Schritt wird die Beschreibung der Kristallstruktur und der Gitterenergie sein, um befriedigende Löslichkeitsvorhersagen machen zu können.show moreshow less
  • To a large extent the effectivity of a drug depends on its solubility in water. Poor solubility is characterised by a great positive change of the free energy associated with the dissolution process. It is the aim of this dissertation to contribute to a better understanding of the dissolution process. According to the Hess theorem the dissolution process can be split into a series of alternative processes having the same starting and end points as the direct dissolution. Some steps of these alternative processes can be described by means ofTo a large extent the effectivity of a drug depends on its solubility in water. It is the aim of this dissertation to contribute to a better understanding of the dissolution process. According to the Hess theorem the dissolution process can be split into a series of alternative processes having the same starting and end points as the direct dissolution. Some steps of these alternative processes can be described by means of thermodynamical, calorimetrical and quantum chemical methods. By standardisation the standard free energies, the standard enthalpies as well as the standard entropies are obtained from the corresponding experimentally determined values. Other physical-chemical parameters required to describe some of the alternative processes, e.g. vapour pressure or polarisation energies, were determined experimentally or calculated by quantum chemical methods. Simple naphthalene derivatives served as model substances: two mono- and seven bihydroxylated naphthalenes, 1-naphthylamine, 4-chloro-1-naphthol and 2-naphthalenethiole. In one of these alternative processes dissolution is considered as a sequence of sublimation and solvation. The standard free energies of sublimation were calculated from the vapour pressures of sublimation which were determined by means of a high-vacuum apparatus or by gaschromatography. The standard free energy of sublimation increases with an increasing number of polar groups in a molecule. For naphthalene a value of 30,5 [kJ·mol-1], for monohydroxylated naphthalenes values between 40,1 and 45,3 [kJ·mol-1] were observed. The corresponding values for bihydroxylated naphthalenes ranged from 51,8 to 67,5 [kJ·mol-1]. Similar relations were observed with the standard free energies of solvation. They were calculated using the semiempirical quantum chemical program package AMSOL with the AM1 as well as with the PM3 parametrisations. The standard free energies of dissolution of the various substances is calculated from their solubilities in water which are determined experimentally. Even considering the fact that the introduction of a polar group into a naphthalene molecule increases their free energies of sublimation by about 10 [kJ·mol-1], the differences in the standard free energies of dissolution are still difficult to explain. An increased number of hydrogen bonds and stronger dipole-dipole interactions observed in higher substituted naphthalenes may be potential explanations. The standard enthalpies behave in a similar way as the corresponding standard free energies. The standard dissolution enthalpy can be calculated by standardisation of the dissolution enthalpy determined calorimetrically. The experimentally determined solution enthalpies would be wrong if in the calorimetric cell the substances to be dissolved would adsorb water during the temperature equilibration time. In order to check for such a potential error, water adsorption isotherms were determined for all test substances. The magnitude of the various enthalpies can be explained by the same facts as in the case of the free energies. 1,3-dihydroxynaphthalene is an exception. Its standard dissolution enthalpy four times bigger than the ones of the other dihydroxynaphthalenes. The standard entropies are calculated from the standard free energies and standard enthalpies respectively. The standard sublimation entropies of the substances included in this study range from 162 to 350 [J·mol-1·K-1]. With the exception of 2-naphthalinthiole the standard dissolution entropies are significantly higher than the corresponding sublimation entropies. The sequence formed by the melting of a substance and the subsequent mixing of this melt with a solvent is a second alternative process according to the Hess theorem to describe the dissolution of a substance in a solvent. The standard melting enthalpy is calculated from the molar melting enthalpy determined by means of a calorimeter at the melting temperature of the substances and from the differences of the heat capacities at constant pressure of the solid substance and of its melt. The standard mixing enthalpy is calculated from the standard solution enthalpy and from the standard melting enthalpy. Due to their high melting temperatures the standard melting enthalpies of the dihydroxynaphthalenes are lower than expected. As the melting temperatures of the chlorinated 1-naphthol and of 1-naphthol do not differ markedly one can assume that the chlorine atom has no significant influence on the melting energy. The melting entropy is calculated from the melting enthalpy and the melting temperature. The higher the melting temperature the lower is the standard melting entropy. The mixing entropy is calculated as the difference of the entropies of melting and of solution respectively. With the exception of 2-naphthalenethiole and naphthalene for all substances included in this study the mixing process is associated with an increase in order. These two hydrophobic molecules have less possibilities to arrange themselves in aqueous solutions than in the melt. The solvation energies are calculated by means of AMSOL using the AM1 as well as the PM3 parametrisations. They are compared with the results obtained experimentally. For both sets of parameters the correlation coefficients determined for the results of the quantum chemical calculations are almost identical, r2 = 0,97 for AM1 and r2 = 0,98 for PM3. The solvation energy is considered as consisting of several terms. The total electrostatic energy increases with an increasing number of polar substituents as they allow for a greater number of partial charges and hence for more polarisations. The component accounts for the free energy of formation of a cavity in the solvent and for changes in dispersion interactions and solvent structure that accompany the solvation process. Its contribution to the solvation energy increases for naphthalenes with polar substituents. By means of AMSOL the van der Waals surfaces as well as the van der Waals volumes were calculated. For the dihydroxynaphthalenes surfaces of about 375 Å2 were obtained. The corresponding values for the monosubstituted compounds are somewhat smaller. For the dihydroxynaphthalenes van der Waals volumes of about 146 Å3 were calculated. Octanol-water partition coefficients were calculated using both sets of parameters, AM1 and PM3, as well as determined experimentally. The agreement between the partition coefficients obtained from the quantum chemical calculations and those determined experimentally is rather poor. The regression coefficients r2 are lower than 0,79. In order to gain a better understanding which of the following factors, e.g. polarisation energy, van der Waals surface, van der Waals volume or the melting temperature, take influence on parameters like free sublimation energy, free solvation energy and free solution energy, linear regressions were performed. Only three parameters were included in the linear models in order to avoid a good mathematical fit having no physical meaning. On the basis of the results obtained with AM1 parameters a linear model was found for the free sublimation energy including the interaction between polarisation energy and van der Waals volume, the van der Waals surface and the squared van der Waals volume. The correlation coefficient of this model is given by r2 = 0,984. If the results obtained from calculations with the PM3 are used the free sublimation energy can be described by a model including the interaction between the polarisation energy and the van der Waals volume, the van der Waals volume and the dipole moment in the gas phase. Its correlation coefficient is given by r2 = 0,986. The free sublimation energy as determined experimentally is in a good agreement with the results obtained with this linear model. The attempt to describe the free solvation energy by a linear model showed that good approximations can be obtained by including the polarisation energy, the surface energy as well as the melting point as crucial parameters. Using the results from calculations on a AM1 basis a correlation coefficient r2 = 0,979 was obtained. For corresponding calculations with results on a PM3 basis the correlation coefficient was given by r2 = 0,983. When using results from calculations on the basis of the AM1 parametrisation the free solution energy can be approximated by a linear model with the polarisation energy, the melting temperature and the van der Waals volume as parameters. The correlation coefficient is given by r2 = 0,847. With results obtained from calculations with the PM3 parametrisation in the linear model the van der Waals volume had to be replaced by the heat of melting to obtain a correlation coefficient of r2 = 0,879. Whereas the free energy of sublimation as well as the free energy of solvation could be satisfactorily approximated by a linear three parameter model the approximation is not as good as in the case of the free energy of solution. As a summary it has to be stated that the data gathered for thirteen substances still form a basis being too small for a detailed analysis of their poor water solubility. A closer consideration of the crystal structure and the corresponding lattice energies may contribute to a better understanding of this problem.show moreshow less
