Untersuchung des Epsilon-Toxins und Mutanten des Epsilon-Toxins aus Clostridium perfringens auf Kanalbildung

- Ziel dieser Arbeit war die biophysikalische Charakterisierung der Bindekomponente des Epsilon-Toxins und der Epsilon-Toxin-Mutanten mit Hilfe künstlicher Lipid-Bilayer-Messungen. Es galt herauszufinden, welcher Teil des Epsilon-Toxins für die Kanalbildung verantwortlich ist, um somit die kanalbildende Domäne definieren zu können. Um diese ermitteln können, wurden in der Arbeit vier Epsilon-Toxin Mutanten untersucht, bei denen in dieser potentiellen Porendomäne Punktmutationen eingefügt worden sind. Im Vergleich von Wildtyp und Mutanten hat sich gezeigt, dass sich die Werte der Einzelkanalleitfähigkeit, der Selektivität und der Spannungsabhängigkeit verändert haben. Damit wurde bewiesen, dass die Punktmutationen genau in dem Bereich des Toxins durchgeführt wurden, der für die Kanalbildung verantwortlich sein muss.…
- Goal of this thesis was the biophysical characterization of the binding component of epsilon-toxin and epsilon-toxin-mutants via artificial lipid-bilayer-experiments. We had to identify the component of the epsilon-toxin which is responsible for channel forming in order to be able to define the channel forming domain. Therefore, we tested four epsilon-toxin-mutants which had been marked in this potential pore domain through point mutation. We called them epsilon-toxin 700, 701, 702 and 703.
