  • In dieser retrospektiven Studie untersuchte ich, ob das profibrotische Zytokin Transforming Growth Faktor beta (TGF Beta) 1 in der Bronchoalveolaeren Lavage Fluessigkeit (BALF) und im Serum von Patienten mit Interstitiellen Lungenkrankheiten (ILD) messbar ist. Ich fokussierte mich auf 61 Sarkoidose Patienten, die radiologisch dem Typ I und dem Typ II/III zugeordnet wurden, sowie 38 Patienten mit idiopathischer pulmonaler Fibrose (IPF) und 22 Kontrollen. Bei der Auswertung der TGF Beta 1 aus der BALF zeigten sich indessen, dass in weniger fibrotischen Erkrankungen wie der Kontrollgruppe und der Sarkoidose Typ I signifikant niedrigere TGF Beta 1 Werte vorhanden waren als bei stärker fibrotischen Erkrankungen wie IPF und Sarkoidose Typ II/III. TGF Beta 1 scheint einen fibrotischen Phänotyp zu charakterisieren. Die TGF Beta 1 Werte in der BALF und im Serum korrelierten nicht signifikant mit der Lungenfunktion der Patienten. Die komplexe Wirkungsweise von TGF Beta 1 bei fibrosierenden Vorgängen wurde durch eine positive Korrelation zu B-Lymphozyten und polymorphkernigen neutrophilen Granulozyten, und eine negative Korrelation zu T-Lymphozyten, T-Helferzellen und Makrophagen deutlich. Das Zytokin TGF Beta 1 ist in der BALF von fibrosierenden Lungenerkrankungen erhoeht und eignet sich moeglicherweise gut als Angriffspunkt für antifibrotische Therapie.show moreshow less
  • In this retrospective study I analyzed the concentration of the profibrotic cytokine TGF Beta 1 in bronchoalveolar lavage fluid (BALF) and serum of patients with ILD and correlated it to clinical parameters. I focused on 61 patients with sarcoid (separated into radiological type I and type II/III), 38 patients with idiopathic pulmonary fibrosis (IPF) and 22 controls. There were no significant differences between the groups concerning TGF Beta 1 in the serum. This suggests that the cytokine is not leaving the lung compartment at a larger scale.In this retrospective study I analyzed the concentration of the profibrotic cytokine TGF Beta 1 in bronchoalveolar lavage fluid (BALF) and serum of patients with ILD and correlated it to clinical parameters. The analysis of TGF Beta 1 in the BALF showed that there were significant lower levels of TGF Beta 1 in less fibrotic diseases like sarcoid type I and control, compared to IPF and sarcoid type II/III. This suggests that TGF Beta 1 level might be dependent on the fibrotic phenotype of ILD. TGF Beta 1 levels in BALF and serum did not correlate with clinical parameters such as pulmonary function tests. There was a significant correlation between TGF Beta 1 levels in BALF and B-lymphocytes, neutrophile granulocytes and a negative correlation with T-lymphocytes, T-helper-cells and macrophages, reflecting the complex proerties of TGF Beta 1 in the context of inflammation and fibrosis. The cytokine TGF Beta 1 is elevated in BALF of patients with fibroproliferative lung disease and may be an attractive target for the development of novel antifibrotic therapies.show moreshow less
