  • Goal of this thesis was the biophysical characterization of the binding component of epsilon-toxin and epsilon-toxin-mutants via artificial lipid-bilayer-experiments. We had to identify the component of the epsilon-toxin which is responsible for channel forming in order to be able to define the channel forming domain. Therefore, we tested four epsilon-toxin-mutants which had been marked in this potential pore domain through point mutation. We called them epsilon-toxin 700, 701, 702 and 703. During the comparison of the wild type and the mutants it turned out that the results of the single channel conductance, of the selectivity and of the bias dependence had changed. Hence we could prove that the point mutations had been carried out exactly in that area of the toxin which must be responsible for channel forming.show moreshow less
