  • Das Ziel der vorliegenden Arbeit war die Charakterisierung der molekularen Grundlagen der Proepikardentwicklung im Huhn. Das Proepikard (PE) stellt die Vorläuferpopulation des Koronargefäßsystems dar. Es wurden zwei Schwerpunkte bearbeitet, zum einen die Rolle von einem Mitglied der TGFß-Superfamilie (BMP, bone morphogenetic protein) während der Induktionsphase des PE und zweitens die molekularen Mechanismen, welche der links/rechts Asymmetrie der PE Entwicklung zugrunde liegen. Die Expressionsmuster von TBX18, WT1, CFC weisen diese alsDas Ziel der vorliegenden Arbeit war die Charakterisierung der molekularen Grundlagen der Proepikardentwicklung im Huhn. Es wurden zwei Schwerpunkte bearbeitet, zum einen die Rolle von einem Mitglied der TGFß-Superfamilie (BMP, bone morphogenetic protein) während der Induktionsphase des PE und zweitens die molekularen Mechanismen, welche der links/rechts Asymmetrie der PE Entwicklung zugrunde liegen. Die Expressionsmuster von TBX18, WT1, CFC weisen diese als proepikardiale Marker aus. BMP4 wird ebenfalls im PE exprimiert, wenngleich wesentlich schwächer als BMP2 im benachbarten Sinusmyokard. Durch in vitro Experimente mit proepikardialen Primärkulturen wurde festgestellt, dass die Zugabe von rekombinatem BMP2 einen Teil der Zellen zu Kardiomyozyten differenzieren lässt. Daher wird angenommen, dass ein Teil der proepikardialen Zellen konzentrationsabhängig auf Wachstumsfaktoren reagieren und in der Lage sind, die epikardiale Linie zu verlassen und ein myokardiales Schicksal anzunehmen. Im zweiten Teil der Arbeit wurde die Rolle des NODAL-PITX2-Signalweges in Bezug auf die Unilateralität des Proepikards im Hühnchen analysiert. Dazu wurden Manipulationen des Hensenschen Knotens im HH Stadium 4 durchgeführt, mit dem Ziel linksseitige Markergene auf der rechten Seite ektopisch zu induzieren. Keine dieser Manipulationen, als auch eine virale Überexpression von PITX2 in sinuatrialen Progenitoren in vivo, liess eine Veränderung der TBX18-Lateralität erkennen, obwohl die PITX2-Expression randomisiert war. Dagegen führte die Inhibition des asymmetrischen Ionenfluxes sowie der Apoptose im Primitivstreifen zu einer völligen Randomisierung der TBX18-Expression, sowie unter anderem der Entwicklung bilateraler Proepikardien. Die Induktion bilateraler TBX18-Expressionsdomänen war ebenfalls durch ektopisches FGF8 auf der linken Seite des Knotens zu beobachten. Der Verlust der rechtsseitigen FGF Signalgebung im Knoten resultierte in einem Verlust der rechtsseitigen TBX18-Expression im Sinus. Daraus kann geschlossen werden, dass proepikardiale Progenitoren nicht nur durch den asymmetrischen Ionenflux sowie Apoptose im Primitivstreifen in ihrer Lateralität festgelegt werden, sondern auch von rechtsseitigen FGF-Signalen im Knoten und unabhängig vom linkseitigen NODAL-PITX2-Signalweg lateralisiert werden.show moreshow less
  • The proepicardium (PE) is a progenitor population of cells, which gives rise to the coronary blood vessel system of the heart. The aim of this study was to unravel the underlying mechansims of the induction of proepicardial development in the chick. Two main topics were hereby addressed, (1) investigation of the role of BMP (bone morphogenetic protein), a member of the TGFß-superfamily, during the induction of the PE and (2) the molecular mechanisms, which underlie the asymmetrical development of the PE. The analysis of TBX18, CFC and WT1The proepicardium (PE) is a progenitor population of cells, which gives rise to the coronary blood vessel system of the heart. The aim of this study was to unravel the underlying mechansims of the induction of proepicardial development in the chick. Two main topics were hereby addressed, (1) investigation of the role of BMP (bone morphogenetic protein), a member of the TGFß-superfamily, during the induction of the PE and (2) the molecular mechanisms, which underlie the asymmetrical development of the PE. The analysis of TBX18, CFC and WT1 expression shows that these genes serve as proepicardial markers. BMP4 is also expressed in the PE, but significantly weaker than BMP2 that was known to be expressed in the sinus myocardium. Studies of proepicardial explants revealed, that treatment with recombinant BMP2 leads to differentiation into contractile cardiomyocytes. One can assume that proepicardial cells react after stimulation with growth factors like BMP2 by leaving the epicardial lineage and adopting a myocardial fate. In the second topic, the role of the NODAL-PITX2 pathway in the asymmetrical PE development was analyzed. Manipulations of Hensens node at stage HH 4 were performed to induce leftsided marker gene expression on the right side. However, none of these manipulations as well as a forced overexpression of PITX2 in sinuatrial progenitors had an influence on the expression of TBX18 in the right sinus. In contrast, inhibition of asymmetrical ionflux and prevention of apoptosis in the primitive streak randomized TBX18 expression and lead to the induction of bilateral PE anlagen. The applicaton of FGF8 on the left side of the node also lead to induction of bilateral TBX18 expression domains and the loss of FGF on the right side was followed by a loss of rightsided TBX18 expression. Presumably, the laterality of proepicardial cells is not only determined by an early asymmetrical ionflux and apoptosis in the primitive streak, but also via rightsided FGF signaling in the node which acts independently of the leftsided NODAL-PITX2 pathway.show moreshow less
