Analysis of Drug Impurities by Means of Chromatographic Methods: Targeted and Untargeted Approaches

The presented works aimed on the analysis of new impurities in APIs and medicinal products. Different subtypes of LC were coupled to suitable detection methods, i.e. UV and various MS techniques, depending on the chemical natures of the analytes and the analytical task.
Unexpected impurities in medicinal products and APIs caused several scandals in the past, concomitant with fatalities or severe side effects in human and veterinary patients. The detection of nitrosamines in sartans led to the discovery of nitrosamines in various other drugs, of which the antibiotic rifampicin was analyzed in this work. An examination of the synthesis of rifampicin revealed a high potential for the formation of 4-methyl-1-nitrosopiperazine (MeNP). An LC-MS/HRMS method suitable for the quantification of MeNP was applied in the analysis of drugs collected from Brazil, Comoros, India, Nepal, and Tanzania, where a single dose of rifampicin is used in the post-exposure prophylaxis of leprosy. All batches were contaminated with MeNP, ranging from 0.7-5.1 ppm. However, application of rifampicin containing up to 5 ppm MeNP was recommended by the regulatory authorities for the post-exposure prophylaxis of leprosy.
In the 1990s the aminoglycoside antibiotic gentamicin attracted attention after causing fatalities in the USA, but the causative agent was never identified unequivocally. The related substance sisomicin was recognized as a lead impurity by the Holzgrabe lab at the University of Würzburg: sisomicin was accompanied by a variety of other impurities and batches containing sisomicin had caused the fatalities. In 2016, anaphylactic reactions were reported after application of gentamicin. A contamination of the medicinal products with histamine, an impurity of the raw material fish peptone used upon the production, could be identified as the cause of the adverse effects. Batches of gentamicin sulfate, which had been stored at the University of Würzburg since the earlier investigations, were analyzed regarding their contamination with histamine to determine whether the biogenic amine was responsible for the 1990s fatalities as well. Furthermore, a correlation with the lead impurity sisomicin was checked. Histamine could be detected in all analyzed batches, but at a lower level than in the batches responsible for the anaphylactic reactions. Moreover, there is no correlation of histamine with the lead impurity sisomicin. Hence, the causative agent for the 1990s fatalities was not histamine and remains unknown.
Another source of impurities is the reaction of APIs with excipients, e.g. the esterification of naproxen with PEG 600 in soft gel capsules. The influence of the formulation’s composition on this reaction was investigated by means of LC-UV. Therefore, the impurity naproxen-PEG-ester (NPEG) was synthesized and used for the development of a method suitable for the analysis of soft gel capsule formulations. Different formulations were stressed for 7 d at 60 °C and the relative amount of NPEG was determined. The formation of NPEG was influenced by the concentrations of water and lactic acid, the pH, and the drug load of the formulation, which can easily be explained by the chemistry behind esterification reactions.
Keeping in mind the huge variety of sources of impurities, it might be impossible to predict all potential impurities of a drug substance/product. Targeted and untargeted approaches were combined in the impurity profiling of bisoprolol fumarate. Eight versions of an LC-HRMS method were developed to enable the detection of a maximum number of impurities: an acidic and a basic buffered LC was coupled to MS detection applying ESI and APCI, both in positive in negative mode. MS and MS/MS data were acquired simultaneously by information dependent acquisition. In the targeted approach, potential impurities were derived from a reaction matrix based on the synthesis route of the API, while the untargeted part was based on general unknown comparative screening to identify additional signals. 18 and 17 impurities were detected in the targeted and the untargeted approach, respectively. The molecular formulae were assessed based on the exact mass and the isotope pattern. Theoretical fragment spectra generated by in silico fragmentation were matched with experimental data to estimate the plausibility of proposed/elucidated structures. Moreover, the detected impurities were quantified with respect to an internal standard.
Zur Bestimmung von geringen Wirkstoffkonzentrationen in biologischen, speziell human-biologischen Matrizes wie Blut, Urin oder Mikrodialysat bedarf es einer Analysentechnik, die den Wirkstoff mit einem Höchstmaß an Selektivität, Spezifität und Präzision bestimmen kann. Daneben muß die verwendetete Methode eine hohe Geschwindigkeit aufweisen und sehr robust sein, da bei der heutigen marktwirtschaftlichen Lage Analysensysteme eine optimale Auslastung erfahren müssen. Aus diesem Grund ist der Umbau oder die Umstellung der Methode von einem zum anderen Wirkstoff ohne nennenswerten Zeitverlust ein maßgeblicher Faktor. Als Technik, die diese Anforderungen optimal erfüllt, hat sich in den letzten Jahren die LC-MS/MS-Technik etabliert. Sie ist den bislang überwiegenden Methoden, wie GC-MS-Techniken oder HPLC-UV-Detektion bzw. Fluoreszenztechniken in Bezug auf die oben genannten Parameter deutlich überlegen. In der vorliegenden Arbeit wurden für die Wirkstoffe Cisaprid, SC-72393, Haloperidol, Linezolid, Methotrexat und Ketoprofen LC-MS/MS-Methoden entwickelt oder via unabhängiger Laborvalidierung auf die lokalen Gegebenheiten transferiert und zur Bestimmung pharmakokinetischer Parameter zur Anwendung gebracht. Ziel der Methodenentwicklung war es die hohe Selektivität und Empfindlichkeit des Detektors zu nutzten, um bei geringen Probenvolumina eine Bestimmungsgrenze zu erreichen, die es ermöglichte ausreichend viele Meßwerte zu bestimmen, um die pharmakokinetischen Parameter der Wirkstoffe zu berechnen. Zusätzlich wurde eine Maximierung des Probendurchsatzes und eine Minimierung des personellen und materiellen Aufwandes angestrebt ohne dabei einen Qualitätsverlust der Methode zu erleiden. Eine gelungene Methoden-entwicklung bedurfte daher der Optimierung der Probenaufarbeitung, die sich neben den chemisch-physikalischen Eigenschaften des Wirkstoffes hauptsächlich an der Menge der zur Verfügung stehenden Probe orientierte. Das chromato-graphische System hingegen hing weitestgehend von den chemischen Eigenschaften des Analyten und von den massenspektroskopischen Bedingungen ab, die verdampfbare Puffer im Fließmittel erforderten. Diese drei zu optimierenden Teilbereiche, die miteinander interagieren, wurden jeweils sorgsam aufeinander abgestimmt, um eine Methode zu entwickeln, die die zu erwartenden Wirkstoffkonzentrationen in der jeweiligen Matrix sicher und robust bis hin zum Quantifizierungslimit bestimmen konnte.
Die Fähigkeit sich an die Rotation der Erde und den daraus resultierenden Tag- und Nacht-Rhythmus anzupassen, basiert auf einer komplexen Regulation verschiedener physiologischer Prozesse. Auf molekularer Ebene liegt diesen Prozessen eine Orchestration von Uhr-Genen zugrunde – auch als innere Uhr bezeichnet – die einen aktivierenden bzw. reprimierenden Einfluss auf die Expression einer Vielzahl weiterer Gene hat. Ausgehend von dieser Regulation lassen sich auf unterschiedlichsten Ebenen tageszeitabhängige, wiederkehrende Rhythmen beobachten.
Während diese wiederkehrenden Rhythmen auf einigen Ebenen bereits gut erforscht und beschrieben sind, gibt es weitere Ebenen wie den Metabolismus, über die das Wissen bisher noch begrenzt ist.
So handelt es sich bei Drosophila beispielsweise um den Organismus, dessen innere Uhr auf molekularer Ebene wahrscheinlich mit am besten charakterisiert ist. Dennoch ist bisher nur wenig über Stoffklassen bekannt, deren Metabolismus durch die innere Uhr kontrolliert wird.
Zwar konnte bereits gezeigt werden, dass sich eine gestörte innere Uhr auf die Anlage der Energiespeicher auswirkt, inwiefern dies allerdings einen Einfluss auf dem intermediären Stoffwechsel hat, blieb bisher weitgehend unerforscht. Auch die Frage, welche Metaboliten wiederkehrende, tageszeitabhängige Rhythmen aufweisen, wurde bisher nur für eine begrenzte Anzahl Metaboliten untersucht.
Bei der hier durchgeführten Arbeit wurden deshalb zunächst die globalen Metabolit-Profile von Fliegen mit einer auf molekularer Ebene gestörten inneren Uhr (per01) mit Fliegen, die über eine funktionale Uhr verfügen (CantonS), zu zwei Zeitpunkten verglichen. Um die Anzahl der zeitgleich untersuchten Gewebe und somit die Komplexität der Probe zu reduzieren, wurden hierfür die Köpfe von den Körpern der Fliegen getrennt und separat analysiert. Beide Körperteile wurden sowohl auf kleine hydrophile als auch auf hydrophobe Metaboliten hin mittels UPLC-ESI-qTOF-MS untersucht. Die anschließend durchgeführte, statistische Analyse brachte hervor, dass sich Unterschiede zwischen den beiden Fliegenlinien besonders in den Spiegeln der essentiellen Aminosäuren, den Kynureninen, den Pterinaten sowie den Spiegeln der Glycero(phospho)lipiden und Fettsäureester zeigten. Bei den Lipiden zeigte sich, dass die Auswirkungen weniger ausgeprägt für die Anlage der Speicher- und Strukturlipide als für die Intermediate des Lipidabbaus, die Diacylglycerole (DAGs) sowie die Acylcarnitine (ACs), waren.
Um zu bestätigen, dass die inneren Uhr tatsächlich einen regulatorischen Einfluss auf die ausgemachten Stoffwechselwege hat, wurden anschließend die Spiegel aller Mitglieder darauf hin untersucht, ob diese wiederkehrende, tageszeitabhängige Schwankungen aufweisen. Hierfür wurden Proben alle zwei Stunden über drei aufeinanderfolgende Tage genommen und analysiert, bevor mittels JTK_CYCLE eine statistische Analyse der Daten durchgeführt und die Metaboliten herausgefiltert wurden, die ein rhythmisches Verhalten bei einer Periodenlänge von 24h zeigten. Hierbei bestätigte sich, dass besonders die Mitglieder des intermediären Lipidmetablismus hiervon betroffen waren. So konnten zwar auch für einige Aminosäuren robuste Rhythmen ausgemacht werden, besonders ausgeprägt waren diese jedoch erneut bei den DAGs und den ACs. Die abschließende Untersuchung letzterer unter Freilaufbedingungen (DD) sowie in per01 brachte hervor, dass die ausgemachten Rhythmen unter diesen Bedingungen entweder nicht mehr detektiert werden konnten oder deutlich abgeschwächt vorlagen. Lediglich zwei kurzkettige ACs zeigten auch unter DD-Bedingungen statistisch signifikante Rhythmen in ihren Spiegeln. Dies spricht dafür, dass neben der Regulation durch die innere Uhr weitere Faktoren, wie beispielsweise das Licht, eine entscheidende Rolle zu spielen scheinen.
In this thesis eight robust and reliable LC-MS/MS methods were developed and validated to analyze atorvastatin, clopidogrel, furosemide, itraconazole, loratadine, naproxen, nisoldipine and sunitinib in human plasma. The active metabolites 2-hydroxyatorvastatin, 4-hydroxyatorvastatin, hydroxyitraconazole, descarboethoxy-loratadine, 4-hydroxynisoldipine and N-desethylsunitinib were also included in the corresponding methods. Due to the different physical, chemical and pharmacokinetic properties of the analytes a wide spectrum regarding sample preparation techniques, chromatography and mass spectrometric detection was covered. Protein precipitation methods were developed for furosemide, itraconazole, naproxen, nisoldipine and sunitinib. Liquid-liquid extraction methods were developed for atorvastatin, clopidogrel and loratadine. Criteria to choose protein precipitation or liquid-liquid extraction were the final plasma concentrations of the drugs, which are mainly dependant on the dose, bioavailability and t1/2 and of course cost-effectiveness. Altogether, the methods have a concentration range from 0.001 ng/mL (LLOQ of clopidogrel) to 50000 ng/mL (highest calibration point for naproxen), covering 5 x 107 orders of magnitude. The runtime of the methods ranged from 2 to 4 minutes, facilitating a high sample throughput. All developed methods were validated according to recent guidelines as they were used to analyze sampes from clinical trials. Excellent linearity, intra-day and inter-day precision and accuracy were observed in the validated calibration ranges. Hemolyzed, lipemic and different batches of human plasma as well as sample dilution did not affect the determiantion of the analytes. Clopidogrel, loratadine, nisoldipine and sunitinib and if available their metabolites were subjected to a matrix effect test, resulting in no influence of different batches of human plasma on the analytical methods. Noteworthy is clopidogrel that shows a slight effect on one of the two used mass spectrometers. However, that effect was reproducible and did therefore not affect clopidogrel determination. No evidence of instability during chromatography, extraction and sample storage processes for all analytes except 4-hydroxyatorvastatin was found, for which a significant decrease was observed after three months. During incurred sample reanalysis of study samples 95 % of the samples were within ±15 % with respect to the first analysis. Moreover, the atorvastatin, loratadine and clopidogrel method were compared on two generations of triple quadrupole mass spectrometers, the API 3000™ and the API 5000™. The new ion source and the changes in the ion path of the API 5000™ provided higher sensitivity, the extend depending on the substance. However, the API 3000™ had very good precision in the performed system comparison. The validated methods showed excellent performance and quality data during routine sample analysis of eight clinical trials. Moreover, they are suitable for high sample throughput due to their short run times.
Grundsätzlich sollte in dieser Arbeit der Einfluss der Glutamin-Restriktion als eine Form der Aminosäure-Restriktion auf das Proliferationsverhalten, den Stoffwechsel sowie die Morphologie der murinen Fibroblastenzelllinie L929 untersucht werden. Es sollte dabei auch gezeigt werden, ob sich Indizien für die Induktion eines LEM finden lassen. Weiterhin sollten die Polyamine Spermidin, Spermin, Putrescin und N-Acetylputrescin als CRMs diskutiert und ihr Einfluss auf das Proliferationsverhalten an sieben Zelllinien herausgestellt werden. Der antiproliferative Effekt der Polyamine Spermidin und Spermin auf das Wachstum der untersuchten Zelllinien konnte in dieser Arbeit bestätigt werden, wobei sich Spermin als potenter erwies als Spermidin. Eine durch Spermidin/Spermin induzierte Autophagie konnte in den Western Blots nicht signifikant nachgewiesen werden. Putrescin und N-Acetylputrescin zeigten nur leichte Wirkungen bei hoher Dosis (10 mM). Während Putrescin einen leicht antiproliferativen Effekt hatte, zeigte N-Acetylputrescin eine leicht proliferationssteigernde Wirkung. Die Autophagie-Induktion durch Methionin-Restriktion konnte durch den Nachweis Autophagie-assoziierter Proteine in den durchgeführten Western Blots bestätigt werden. Um regelmäßig von den positiven Effekten der Autophagie zu profitieren, wäre eine pflanzenbasierte Ernährungsweise sinnvoll und gut durchführbar, da Methionin hauptsächlich in tierischen Lebensmitteln und nur geringfügig in pflanzlichen Nahrungsmitteln vorkommt. Ein völliger Verzicht auf Methionin ist nicht empfehlenswert, da die Aminosäure essentiell ist und somit über die Nahrung aufgenommen werden muss. Um das Methionin-Level konstant, aber niedrig zu halten, erscheint eine Bedarfsdeckung über pflanzliche Nahrung als ideal. Die an den Zelllinien L929 und HeLa durchgeführten Proliferationsstudien zeigten, dass die Zellen selbst nach fünftägiger Glutamin- Restriktion geringfügig weiter proliferierten und keinerlei Merkmale des Zelltodes aufwiesen. Die massenspektrometrische Analyse der Modellzelllinie L929, welche über einen Zeitraum von fünf Tagen einer Glutamin-Restriktion ausgesetzt war, zeigte deutliche metabolische Veränderungen bei den über 150 untersuchten Metaboliten, die auf die Induktion eines LEM schließen lassen. Es konnte ein metabolischer Fingerabdruck nach 48 h für die Zelllinie L929 unter Glutamin-Restriktion definiert werden, der zukünftig als Referenz bei der Testung potentieller CRMs herangezogen werden kann.
Der Nachweis von oxidativen Stressmarkern hat bei der Untersuchung von Krankheiten wie Diabetes, Krebs und Hypertonie an großer Bedeutung gewonnen. Vor allem 8-Oxo-2’-desoxyguanosin (8-oxodG) wird gezielt mit verschiedenen Methoden gemessen und als Marker für oxidativen Stress herangezogen. Daneben haben 8 Oxoguanin (8-oxoGua), als Produkt aus der Basenexzisionsreparatur der DNA, sowie 8-Oxoguanosin (8-oxoGuo), als Biomarker für oxidativ geschädigte RNA, bisher weniger Aufmerksamkeit bekommen. Das Renin-Angiotensin Aldosteron System (RAAS) spielt eine wichtige Rolle in der Regulierung des Blutdrucks. Im Falle einer Hypertonie werden Angiotensin II (Ang II) und Aldosteron (Aldo) über einen langen Zeitraum in erhöhter Konzentration ausgeschüttet. Dieser Umstand bewirkt eine nicht physiologische Wirkung der Hormone des RAAS, welche zu einer Induktion von oxidativem Stress führt. Die Zielsetzung dieser Arbeit ist es, die oxidative Schädigung, ausgelöst durch Ang II und Aldo, in der DNA und der RNA in vitro und in vivo nachzuweisen und dabei speziell den Biomarker 8-oxodG zu untersuchen.
In-vitro-Experimente wurden mit LLC PK1-Zellen, einer Schweinenierenzelllinie, durchgeführt. Ang II und Aldo lösten einen dosisabhängigen Anstieg der DNA Schäden in LLC PK1 Zellen aus. Eine Zeitabhängigkeit wurde für die ersten 30 Minuten gezeigt. Für die restliche Zeit (4 h) blieb der nachgewiesene DNA Schaden konstant. Der FPG Comet-Assay und die immunzytochemische Färbung zeigten jeweils eine signifikante Zunahme von 8-oxodG in LLC-PK1-Zellen an, während die HPLC MS/MS Messung nur geringe Veränderungen nachwies. Das FPG Enzym erkennt neben 8-oxodG auch andere oxidierte Purine und sorgte so für eine Überbestimmung des DNA-Schadens. Bei der immunzytochemischen Färbung entsteht die Überbestimmung durch Kreuzreaktionen des 8 oxodG Antikörpers mit oxidierten Strukturen in der DNA. Der Vorteil beider Analysemethoden ist die direkte Messung von Schädigungen in der Zelle, während die HPLC-MS/MS eine Isolierung der Nukleinsäuren voraussetzt. Bei diesem Schritt kann es zur Oxidation der Marker für oxidativen Stress kommen, welche einen genauen Nachweis erschwert.
In vivo-Versuche hatten zum Ziel, die oxidativen Stressmarker 8-oxoGua, 8-oxodG und 8-oxoGuo im Urin nachzuweisen. Die Behandlung der C57BL/6-Mäuse und Sprague Dawley-Ratten (SD-Ratten) mit den Hormonen des RAAS zeigten einen Anstieg des Blutdrucks, erhöhte DNA Schäden durch oxidativen Stress sowie erhöhte Exkretionsraten der oxidativen Stressmarker. Durch eine Inhibierung des Angiotensin II-Typ1- oder Mineralkortikoidrezeptors sowie die Mutation des Gens AT1a konnte gezeigt werden, dass die Schädigungen unabhängig vom Blutdruck sind. Zudem konnte gezeigt werden, dass neben NOX4 auch andere NADPH Oxidasen für den oxidativen Stress verantwortlich sein müssen. Eine Aktivierung des Nrf2 Signalweges in den SD-Ratten hat Einfluss auf die Wirkung von Aldo.
Die Exkretionsrate der oxidativen Biomarker im 20-h-Urin der behandelten Tiere zeigen, wie sich das Gleichgewicht zwischen DNA-Reparatur und oxidativem Stress verändert. Da 80 % der DNA in RNA umgeschrieben werden, ist der Nachweis von 8 oxoGuo in den Fokus gerückt. In der praktischen Anwendung kann mit der Messung von 8 oxodG und 8-oxoGuo ein Krankheits- oder Heilungsprozess auf nicht invasive Weise verfolgt werden. Der Nachweis von 8-oxodG und 8-oxoGuo in den Nukleinsäuren stellt einen Einstieg für die Grundlagenforschung dar, da sie nur eine Momentaufnahme der Nukleinsäureschädigung in der Zelle zeigen. Meist findet eine Überbestimmung, ausgelöst durch die Messmethode, statt. In Gewebeproben kann eine Unterbestimmung vorliegen, falls nicht alle Zelltypen vom oxidativen Stress betroffen sind. Daher sollte es ein vorrangiges Ziel sein, ein stabileres Oxidationsprodukt des Guanins nachzuweisen, um das Gleichgewicht der DNA-Oxidation und Reparatur besser zu verstehen.
Furan wird in einer Vielzahl von Speisen durch Hitzebehandlung gebildet und ist kanzerogen in der Leber von Ratte und Maus. Durch die hohe Flüchtigkeit von Furan ist eine Expositionsabschätzung auf Basis der Kontamination von Lebensmitteln nur bedingt möglich. Ein alternativer Ansatz dazu ist die Identifizierung von Furanmetaboliten als Expositionsbiomarker. Nach der Aufnahme wird Furan zunächst zum Dialdehyd cis-2-Buten-1,4-dial oxidiert. cis-2-Buten-1,4-dial besitzt mehrere elektrophile Strukturelemente, welche eine Reaktion mit Protein und DNS wahrscheinlich machen und damit zur bekannten Toxizität von Furan beitragen können. Es stellt sich in diesem Zusammenhang die Frage, ob eine Reaktion mit Protein die Reaktion mit der DNS verhindern kann und somit keine direkt gentoxischen Effekte auftreten. Für ein kanzerogenes Agens ohne direkte gentoxische Wirkung kann eine Schwellendosis unterhalb derer kein DNS-Schaden auftritt diskutiert werden. Für eine fundierte Risikobewertung bezüglich der Aufnahme von Furan über die Nahrung ist dies unabdingbar. In der vorliegenden Arbeit wurde nach der oralen Gabe von Furan im Urin von Fischer 344 Ratten nach Metaboliten gesucht. Eine Kontrollgruppe erhielt nur die Trägersubstanz Öl. Das vor und nach Exposition über jeweils zwei 24 Stunden Perioden gesammelte Urin wurde mittels einer Tandemmassenspektrometrie-Methode analysiert. Die Methode bestand aus einem Full-Scan und einer darüber gesteuerten Aufzeichnung eines Fragmentionenspektrums. Die Full-Scan-Daten wurden mit Hilfe der Hauptkomponentenanalyse untersucht. In der ersten Sammelperiode nach der Behandlung konnten durch die erste Hauptkomponente die behandelten von den unbehandelten Tieren getrennt werden. Aus den für die Trennung relevanten Verbindungen konnten fünf Biomarker strukturell aufgeklärt werden. In einer weiteren Tierstudie an Ratten und Mäusen wurde die Kinetik und die Dosis-Wirkungs-Beziehung der identifizierten Biomarker untersucht. Die gezielte LC-MS/MS-Analyse der Urine auf die identifizierten Biomarker hin zeigte, dass in der Ratte alle und in der Maus alle bis auf einen dosisabhängig anstiegen. Die Kinetik der Ausscheidung lieferte wertvolle Hinweise auf die Entstehung der Biomarker. Die Ausscheidung der Biomarker mit Lysinstruktur erfolgte über mehr als 72 Stunden. Dies war ein Hinweis auf eine Freisetzung aus Protein. Die Ausscheidung der restlichen Verbindungen erfolgte ausschließlich in den ersten 24 Stunden. Die in der Literatur vorhandenen Daten zur Gentoxizität von Furan und cis-Buten-1,4-dial sind unschlüssig und unvollständig. In der vorliegenden Arbeit wurde cis-2-Buten-1,4-dial im Ames Stamm TA104 und in L5178Y Mauslymphomzellen auf Mutagenität und Gentoxizität untersucht. Durch starke Zytotoxizität war der Konzentrationsbereich auf 4.5 µmol/Platte limitiert. Innerhalb dieses Bereich konnte mit der Vorinkubationsvariante des Ames-Tests keine Mutagenität beobachtet werden. Die L5178Y Mauslymphomzellen wurden mit Standardprotokollen für den Mikrokern-Test, Kometen-Test und den Thymidinkinase-Test untersucht. Der Konzentrationsbereich von cis-2-Buten-1,4-dial erstreckte sich bis 100 µM, konnte aber auf Grund der starken Zytotoxizität nur bis 25 µM ausgewertet werden. Dennoch konnte bereits in diesem Bereich ein 1.7- bzw. 2.2-facher Anstieg im Kometen- bzw. Thymidinkinase-Test beobachtet werden. Verglichen mit der Positivkontrolle Methylmethansulfonat hatte cis-2-Buten-1,4-dial bei einer deutlich höheren Zytotoxizität eine ähnliche Potenz bezüglich der Mutagenität und Gentoxizität. Um das DNS-vernetzende Potential von cis-2-Buten-1,4-dial zu bestimmen wurde eine Variante des Kometen-Tests verwendet. Es wurde dabei untersucht, ob die Vorbehandlung von Zellen mit cis-2-Buten-1,4-dial die durch γ-Strahlung induzierbaren Kometen reduzieren kann. Während die Positivkontrolle Glutaraldehyd die Kometen tatsächlich verringerte, blieb dieser Effekt bei cis-2-Buten-1,4-dial aus. Im Gegenteil, bei einer Konzentration von ≥100 mM konnte durch die Zunahme von Zellen mit beginnender Apoptose ein Anstieg der Kometen beobachtet werden. Obwohl cis-2-Buten-1,4-dial sehr deutliche gentoxische und mutagene Effekte zeigte, beschränkte die hohe Zytotoxizität den auswertbaren Bereich. Möglicherweise kann diese Problematik einen Teil der unschlüssigen Ergebnisse erklären, sicher ist jedoch, dass für die Untersuchung der Mechanismen der Toxizität und Kanzerogenität ein Beitrag von nicht gentoxischen Effekten diskutiert werden muss.
Development and validation of LC-MS/MS methods to determine PK/PD parameters of anti-infectives

In the present thesis the development and validation of bioanalytical LC-MS/MS methods for the quantification of erythromycin A, erythromycin ethylsuccinate, roxithromycin, clarithromycin, 14 hydroxy clarithromycin, flucloxacillin, piperacillin and moxifloxacin in human plasma and human urine (piperacillin) is introduced. All methods were applied to analyze human plasma and urine samples from clinical trials and therefore, have been validated according to international guidelines. The methods were reliable in these studies and fulfilled all regulatory requirements known at the time of the study conduct.
Moreover, the validation data of the macrolides were compared on three different mass spectrometers (API III Plus, API 3000™, API 5000™). The new innovations in the ion source (horizontal versus vertical electrospray), the ionpath (skimmer, QJet) and the diameter of the orifice resulted in better sensitivity and a larger linearity range for the majority of the analytes. Sensitivity was improved up to a factor of 12 (for clarithromycin) between API III Plus to API 3000™ and up to a factor of 8 (for erythromycin and roxithromycin) between API 3000™ and API 5000™, keeping the accuracy and precision data at about the same level. The high sensitivity was a benefit for example for the flucloxacillin study, because concentrations from all subject samples were detectable up to approximately eight half-lives, i.e. no concentrations needed to be reported below the quantification limit. Also the linearity range were extended from two orders of magnitude to up to four orders of magnitude, which increases the likelihood to allow to analyze all samples from a pharmacokinetic study in the same run.
This is especially useful if a large concentration range needs to be analysed, for example, if the method shall be applied in an ascending dose study. Then, all low concentrations from the beginning of the study can be determined, as well as all high concentrations, without the need to dilute and analyse single samples repeatedly.
The pharmacokinetic data were compared to previously reported literature data and correlated graphically with MIC values of popular microorganisms which might be a starting point for further PK/PD investigations.
The PK/PD theory is a very helpful tool for prediction of the efficacy of given drugs against certain micro-organisms. Depending on the pharmacodynamic processes, e. g. the mode of action, three classes of drugs have been identified.
In the same way this applies to adverse effects, which need to be minimised by reducing plasma concentrations. These coherences are not well-investigated, yet, and are not discussed further in this thesis.
Still, a lot of research has to be done in this interdisciplinary field to minimise uncertainty in single values, like an AUC/MIC. These include:
Improve accuracy and precision of bioanalytical methods determining total and free concentration data in biological matrices for calculation of AUC and Cmax
These parameters are related to the MIC in pharmacodynamic considerations. Since the determination of the MIC often underlies significant variations and also differences between microbiological laboratories, the determination of concentrations of anti-infectives is particular important, being achievable by scientific exact techniques. Finally, from the volume of distribution of antibiotics can be used to derive information about intracellular concentrations and effectivity of antiinfectives.
Posttranslational modifications (PTMs) play a crucial role in many cellular processes. They are reversible, dynamic, and highly regulated events that alter the properties of proteins and increase their functional diversity. The identification and quantification of PTMs are critical for deciphering the molecular mechanisms of PTMs-related biological processes and disease treatment and prevention. Two of the most common and important PTMs that regulate many protein functions are acetylation and phosphorylation.
An important role of acetylation is the regulation of DNA/RNA-protein interactions. A prominent example for this are histones, whose tail regions are lysine-rich and can be highly acetylated at their N-terminal domain. In spite of the utmost importance of this PTM, methods that allow the accurate measuring the site-specific acetylation degree are missing. One of the challenges in quantifying the acetylation degree at an individual lysine residue of the histones N-termini is the occurrence of multiple lysines in close proximity. Herein, we describe the development of the ”Fragment Ion Patchwork Quantification,” a new mass spectrometry-based approach for the highly accurate quantification of sites-pecific acetylation degrees. This method combines 13C1-acetyl derivatization on the protein level, proteolysis by low-specificity proteases and quantification on the fragment ion level. Acetylation degrees are determined from the isotope patterns of acetylated b and y ions. We have shown that this approach allows determining the site-specific acetylation degrees of all lysine residues for all core histones of Trypanosoma brucei. In addition, we demonstrate the use of this approach to identify the substrate sites of histone acetyltransferases and to monitor the changes in acetylation of the histones of canonical nucleosome and transcription start site nucleosomes.
Phosphorylation is one of the most common and most important PTMs. The analysis of the human genome showed that there are about 518 kinases and more than 500,000 phosphorylation sites are believed to exist in the cellular proteome. Protein phosphorylation plays a crucial role in signaling many different cell processes, such as intercellular communication, cell growth, differentiation of proliferation and apoptosis. Whereas MS-based identification and relative quantification of singly phosphorylated peptides have been greatly improved during the last decade, and large-scale analysis of thousands of phosphopeptides can now be performed on a routine-base, the analysis of multi-phosphorylated peptides is still lagging vastly behind. The low pKa value of phosphate group and the associated negative charge are considered the major source of the problems with the analysis of
multi-phosphorylated peptides. These problems include the formation of phosphopeptide-metal complexes during liquid chromatography (e.g. Fe 3+), which leads to a drastic deterioration of the chromatographic properties of these peptides (peak tailing), the decreased ionization efficiencies of phosphorylated peptides compared to their unphosphorylated counterparts, the labile nature of phosphate during CID/HCD fragmentation, and the unsuitability of low-charged phosphopeptides for ETD fragmentation are the most important factors that hinder phosphorylation analysis by LC-MS/MS. Here we aimed to develop a method for improving the identification of multi-phosphorylated peptides as well as the localization of phosphorylation sites by charge-reversal derivatization of the phosphate groups. This method employs a carbodiimide-mediated phosphoramidation to converted the phosphates to stable aromatic phosphoramidates. This chemical modification of phosphosite(s) reversed the negative charge of the phosphate group(s) and increased the number of the positive charges within the phosphopeptide. This modification prevented the formation of phosphopeptide-metal ion complexes that dramatically decreases or completely diminishes the signal intensity of protonated phosphopeptides, specifically multi-phosphorylated peptides. Furthermore, the increased net charge the (phospho-)peptides made them suitable for ETD fragmentation, which generated a high number of fragment ions with high intensities that led to a better phosphopeptide identification and localization of phosphosite(s) with high confidence.
