Single crystal X-ray studies on bis[3,4,5,6-tetrabromo-1 ,2-benzenediolato(2- )](pyrrolidiniomethyl)silicate acetonitrile solvate [(C6Br40 2hSiCH2(H)NC4H8 · CH3CN; monoclinic, P2t/c, a = 808.5(4), b = 1533.0(8), c = 2212.6(1) pm, ß = 97.67(2)0 , Z = 4] revealed a zwitterionic structure with a pentacoordinate, formally negatively charged silicon atom and a positively charged ammonium moiety. The silicon atom is surrounded by four oxygen atoms and one carbon atom in a trigonalbipyramidal fashion, with the carbon atom in an equatorial position. The structure is displaced by 7.0% from the trigonal bipyramid towards the square pyramid. The zwitterion and the CH3CN molecule form intermolecular N-H · · · N hydrogen bonds.
Efficient expression systems are required for analysis of gene regulation and function in teleost fish. To develop such systems, a nurober of inducible or constitutive promoter and enhancer sequences of fish or higher vertebrate origin were tested for activity in a variety of fish celllines andin embryos of the Japanese medaka fish (Oryzias latipes) and Xiphophorus. The activity of the different promoterenhancer combinations were quantitated. Considerable differences were found for some constructs if tested in vitro or in vivo. From the data obtained, a set of expression vectors for basic research as weH as for aquaculture purposes were established.
The most commonly used assay to quantitate the response of peripheral T~cells upon stimulation with growth factors is determination of incorporated (JH]TdR. We compared thls test to three other methods: 1. direct countlog of cells with a Coulter type counter as reference assay, 2. a colorimetric assay using the tetrazolium dye 3-[ 4,S-dimethylthiazol-l-yl]-2,5diphenyl tetrazolium (MTT), which is a cheap and increasingly popular non-radioactive method and 3. incorporation of the thymidine analog 5-bromo-2'-deoxyuridine detection with a monoclonal antibody on cytospins. Primary human PHA-blasts from >30 healthy individuals were stimulated with IL-2, IL-4 aod IL-7 and assayed with up to four different methods. We discuss the advantages and disadvantages of the assays used and tbe effects of differences between cell preparations. We observed no significant variations between individuals for the dose dependence, but the relative emctency of IL4 compared to IL-2 and IL-7 was variable. This was probably due to the slower response observed upon stimulation with this factor.

Escherichia coli 0139: K82: H1 strains originating from outbreaks and single cases of oedema disease in pigs were characterized by their genomic restriction fragment length polymorphism (RFLP), their virulence pattern, and by the occurrence as well as the genomic distribution of the determinants for hemolysin (hly) and verotoxins (shiga-like toxins; sltI, sltII). Whereas the RFLPs revealed considerable variation among the E. coli 0139: K82: H1 isolates depending the origin and epidemic source of the strains, the virulence gene slt II was found to be present in nearly all strains in a particular chromosomal region. Similar to RFLPs, the plasmid profiles are useful for epidemiological analysis.
Alkohol: Konsum, Wirkung, Gefahren für die Verkehrssicherheit. Zur Diskussion um neue Grenzwerte

Im ersten Teil werden die Ergebnisse einer umfangreichen Literaturstudie zu Auswirkungen niedriger Alkoholkonzentrationen auf die Fahrsicherheit vorgestellt. Es wird gezeigt, daß bei Konzentration über 0.6 Promille die Leistungsausfälle drastisch ansteigen, dies umso mehr, je mehr es sich um komplexe Handlungen handelt und je näher der Zeitpunkt der Leistungsabgabe dem Trinkende liegt. Diese Ergebnisse decken sich mit Studien aus Verkehrsunfällen, bei denen ebenfalls über 0.7 Promille ein explosionsartiges Ansteigen des Risikos gezeigt wurde. Demgegenüber ist ein deutlich erhöhtes Risiko für den Bereich unter 0.5 Promille nicht nachgewiesen. An Untersuchungen zum Alkoholkonsum wird aufgezeigt, daß die Alkoholtrinker in mehrere Subpopulationen aufzuteilen sind. Dabei wird insbesondere eine Gruppe von Konsumtrinkern identifiziert, deren BAK regelhaft 0.6/0.7 Promille nicht überschreitet. Es wird gezeigt, daß diese Gruppe wahrscheinlich nicht überproportional zu ihrem Auftreten an Unfällen beteiligt ist. Daraus wird die Forderung nach der Einführung eines einzigen, strafrechtlich bewehrten Grenzwerts abgeleitet, der bei etwa 0.7/0.8 Promille liegen sollte.

We have previously reported that in several renal cell types, adenosine receptor agonists inhibit adenylyl cyclase and activate phospholipase C via a pertussis toxin-sensitive G protein. In the present study, in 28A cells, both uf these adenosine receptor-mediated responses were inhibited by 8-cyclopentyl-1,3-dipropylxanthine (DPCPX). a highly selective A1 adenosine receptor antagonist. The binding characteristics of the adenosine A 1 receptor in the 28A renal cell line were studied using the radiolabeled antagonist f:1H]DPCPX to determine whether two separate binding sites could account for these responses. Saturation binding of [: 1H]DPCPX to 28A cell membranes revealed a single class of A1 binding sites with an apparent Kd value of 1.4 nM and maximal binding capacity of 64 fmol/mg protein. Competition experiments with a variety of adenosine agonists gave biphasic displacement curves with a pharmacological profile characteristic of A1 receptors. Comparison of [: 1H]DPCPX competition binding data from 28A cell membranes with rabbit brain membranes, a tissue with well-characterized A1 receptors, reveals that the A 1 receptor population in 28A cells has similar agonist binding affinities to the receptor population in brain but has a considerably lower density. Addition of guanosine ;)' -triphosphate ( 100 ,uM) to 28A cell membranes caused the competition curves to shift from biphasic to monophasic. indicating that the A1 receptors exist in two interconvertible affinity states because of their coupling to G proteins. In the absence of evidence for subpopulations of the A1 receptor, it appears that in 28A cells. A single A1 receptor population. As defined by ligand binding characteristics, couples via one or more pertussis toxin-sensitive guanine nucleotide binding proteins to two different biological signaling mechanisms.
