N2  - Peroxisomes are ubiquitous organelles with essential functions in numerous cellular processes such as lipid metabolism, detoxification of reactive oxygen species and signaling. Knowledge of the peroxisomal proteome including multi-localized proteins and, most importantly, changes of its composition induced by altering cellular conditions or impaired peroxisome biogenesis and function is of paramount importance for a holistic view on peroxisomes and their diverse functions in a cellular context. In this chapter, we provide a spatial proteomics protocol specifically tailored to the analysis of the peroxisomal proteome of baker's yeast that enables the definition of the peroxisomal proteome under distinct conditions and to monitor dynamic changes of the proteome including the relocation of individual proteins to a different cellular compartment. The protocol comprises subcellular fractionation by differential centrifugation followed by Nycodenz density gradient centrifugation of a crude peroxisomal fraction, quantitative mass spectrometric measurements of subcellular and density gradient fractions and advanced computational data analysis, resulting in the establishment of organellar maps on a global scale.

N2  - Anhand eines geschichtlichen Rückblicks und einer Zusammenfassung jüngerer Emotionstheorien wird die Bedeutung der Emotionen für die Verhaltenstherapie abgeleitet. Die Ursprünge in der verhaltenstherapeutischen Angstbehandlung zeigen, daß für die Verhaltenstherapie seit ihrem Beginn die Veränderung von emotionalen Störungen eine zentrale Aufgabe war. Aus jüngeren Theorien zu diskreten Emotionen und deren Komponenten lassen sich diagnostische und therapeutische Zielrichtungen spezifizieren. Als Komponenten der Emotionen werden die Bewertung der Situation, physiologische Aktivierung, Motivierung, Handlungsvorbereitung, Signal zum Organismus und Signal zur Umgebung erläutert. Von den verschiedenen Funktionen der Emotionen sind die Funktion des Antriebs für unser Verhalten, die Funktion der Koppelung zwischen Reizen und Reaktionen und die verhaltenssteuernde Funktion für die Therapie besonders bedeutsam. Beispiele von verschiedenen Ansätzen auf der somatisch-physiologischen Ebene, der kognitiven Ebene und der Verhaltens-Ebene geben einen Einblick in die Möglichkeiten der Verhaltenstherapiefür die Veränderung negativer emotionaler Zustände. Nach der kognitiven Wende scheint die emotionale Wende in der Verhaltenstherapie angezeigt.
N2  - The relevance of emotions for behavior therapy is derived from a historical view and form a review of current theories on emotions. The origin of behavioral treatment of anxiety reveals thatfrom the beginning, a central task of behavior therapy was the change of emotional disturbances. It is shown that recent theories on discrete emotions and their components allow to derive and specify diagnostic and therapeutic goals. Evaluation of a situation, physiological arousal, motivation, preparation of action, signalling towards the organism and signalling towards the environment are discussed as components of emotions. From the various functions of emotion the function of driving the behavior, the function of coupling between stimuli and responses and the function of regulating the behavior are seen as specially relevant for therapy. The potentials of behavior therapy for changing negative emotional states is shown byexamples of approaches on the somatic-physiologicallevel, the cognitive level, and the behavioral level. After having undergone the cognitive turn it seems appropriate for behavior therapy to also focus on emotions.

N2  - Kaum ein anderer Teil des menschlichen Körpers ist in der Lage, gefühlsmäßige Regungen in derart differenzierter Form auszudrücken wie die Mimik. Über den Zusammenhang zwischen muskulärer Aktivität und Emotionen wird versucht, in einigen Aspekten auch die Beziehung von Stimmungen zur Rückenmuskulatur herzustellen. Vor allem wird jedoch auf die Mimik als Verständigungsmittel eingegangen. Die neuroanatomische und neurophysiologische Basis der Mimik wird dargestellt sowie der Zusammenhang zwischen Stimulus-Situation und Emotionen einerseits und Emotionen und Ausdruck andererseits. Störungen dieser Beziehungen werden anhand von psychiatrischen und neurologischen Erkrankungen erläutert. Aus dem Ausdruck von Konflikten in der Mimik werden einige Bedingungen für psychosomatische Störungen nicht nur im Rückenbereich, sondern auch bei Kopfschmerzzuständen und Streßverhalten abgeleitet.

Generell wendet ein Sprecher nach der Darbietung einer verbalen Aufgabe den Blick von einer Informationsquelle zunehmend ab und wendet ihn erst zum Ende der gesprochenen Antwort wieder zu. Mit größerer Schwierigkeit der Aufgaben erhöht sich die Wahrscheinlichkeit einer Abwendung. Nach diesen und weiteren Ergebnissen scheinen Sprechen und Blickzuwendung zumindest teilweise ähnlichen Regeln zu folgen wie andere simultan ablaufende Tätigkeiten.

N2  - Internal characteristics such as depressed mood, anxiety and general negative emotions are accompanied, particularly during depressive illness, by changes in observable behaviour. Accordingly, the following questions may be examined: are intra-individual changes in speech and gaze behaviour related to changes in the internal psychopathological state? Further, do these changes occur synchronously to changes in the state of subjective well-being? A longitudinal study was made on depressed patients. Their behaviour was observed during standardised interviews and diagnostic-therapeutic discussions held at regu~ lar intervals. Various speech and gaze parameters were examined with respect to their coordination and their relationship to the subjective state of well-being. Considerable variation was found in the temporal relationship amongst these variables. The results are discussed with respect to the relevance of speech parameters and the coordination of verbal and nonverbal behaviour as indicators of the psychopathological condition.

N2  - A panel of simple repetitive oligonucleotide probes has been designed and tested for multilocus DNA fingerprinting in some 200 fungal, plant and animal species as well as man. To date at least one of the probes has been found to be informative in each species. The human genome, however, has been the major target of many fingerprintins studies. Using the probe (CAC)5 or (GTG)5, individualization of all humans is possible except for monozygotic twins. Paternity analyses are now perfonned on a routine basis by the use of multilocus fingerprints, inctuding also cases of deficiency, i.e. where one of the parents is not available for analysis. Depending on the degree of DNA degradation a variety of oligonucleotides are informative, and they have been proven useful in actual case work. Advantages in comparison to other methods including enzymatic DNA amplification techniques (PCR) are evident. Fingerprint patterns of tumors may be changed due to the gain or loss of chromosomes and/or intrachromosomal deletion and amplification events. Locus-specific probes were isolated from the human (CAC)5/( GTG)5 fingerprint with a varying degree of informativeness (monomorphic versus truly hypervariable markers). The feasibility of three different approaches. for the isolation of hypervariable mono-locus probes was evaluated. Finally, one particular mixed simple (gt)n(ga)m repeat locus in the second intron of the HLA-DRB genes has been scrutinized to allow comparison of the extent of exon-encoded (protein-) polymorphisms versus intronie bypervariability of simple repeats: adjacent to a single gene sequence (e.g. HLA-DRB1*0401) many different length alleles were found. Group-specific structures of basic repeats were identified within the evolutionarily related DRB alleles. As a further application it is suggested here that due to the ubiquitous interspersion of their targets, short probes for simple repeat sequences are especially useful tools for ordering genomic cosmid, yeast artificial chromosome and phage banks.
