While diversity gradients have been analysed for numerous single taxa, progress towards general explanatory models has been hampered by limitations in the phylogenetic coverage of past studies. By parallel sampling of 25 major plant and animal taxa along a 3.7 km elevational gradient on Mt. Kilimanjaro, we quantify cross-taxon consensus in diversity gradients and evaluate predictors of diversity from single taxa to a multi-taxa community level. While single taxa show complex distribution patterns and respond to different environmental factors, scaling up diversity to the community level leads to an unambiguous support for temperature as the main predictor of species richness in both plants and animals. Our findings illuminate the influence of taxonomic coverage for models of diversity gradients and point to the importance of temperature for diversification and species coexistence in plant and animal communities. In addition to increased plasma lipid concentrations, irregular/oscillatory shear stress and inflammatory processes trigger atherosclerosis. Inhibitors of the transcription modulatory bromo- and extra-terminal domain (BET) protein family (BETi) could offer a possible therapeutic approach due to their epigenetic mechanism and anti-inflammatory properties. In this study, the influence of laminar shear stress, inflammation and BETi treatment on human endothelial cells was investigated using global protein expression profiling by ion mobility separation-enhanced data independent acquisition mass spectrometry (IMS-DIA-MS). For this purpose, primary human umbilical cord derived vascular endothelial cells were treated with TNFα to mimic inflammation and exposed to laminar shear stress in the presence or absence of the BRD4 inhibitor JQ1. IMS-DIA-MS detected over 4037 proteins expressed in endothelial cells. Inflammation, shear stress and BETi led to pronounced changes in protein expression patterns with JQ1 having the greatest effect. To our knowledge, this is the first proteomics study on primary endothelial cells, which provides an extensive database for the effects of shear stress, inflammation and BETi on the endothelial proteome. The objective of this study was to evaluate the safety and clinical outcomes of rituximab in several standard-of-care-refractory autoimmune diseases (within rheumatology, nephrology, dermatology and neurology) other than rheumatoid arthritis or non-Hodgkin’s lymphoma in a real-life clinical setting. Methods: Patients who received rituximab having shown an inadequate response to standard-of-care had their safety and clinical outcomes data retrospectively analysed as part of the German Registry of Autoimmune Diseases. The main outcome measures were safety and clinical response, as judged at the discretion of the investigators. Results: A total of 370 patients (299 patient-years) with various autoimmune diseases (23.0% with systemic lupus erythematosus, 15.7% antineutrophil cytoplasmic antibody-associated granulomatous vasculitides, 15.1% multiple sclerosis and 10.0% pemphigus) from 42 centres received a mean dose of 2,440 mg of rituximab over a median (range) of 194 (180 to 1,407) days. The overall rate of serious infections was 5.3 per 100 patient-years during rituximab therapy. Opportunistic infections were infrequent across the whole study population, and mostly occurred in patients with systemic lupus erythematosus. There were 11 deaths (3.0% of patients) after rituximab treatment (mean 11.6 months after first infusion, range 0.8 to 31.3 months), with most of the deaths caused by infections. Overall (n = 293), 13.3% of patients showed no response, 45.1% showed a partial response and 41.6% showed a complete response. Responses were also reflected by reduced use of glucocorticoids and various immunosuppressives during rituximab therapy and follow-up compared with before rituximab. A Program for a Fresh Perspective N2 - The nucleus of statehood is situated at the local level: in the village, the neighborhood, the city district. This is where a community, beyond the level of the family, first develops collective rules that are intended to ensure its continued existence. But usually this is not the only level of governance at play. Above it, there are supralocal formations of power, varying in scope from regional networks to empires, which supplement the local orders or compete with them. The premise of this Research Unit is that local forms of self-governance are especially heterogeneous and prominent, wherever supralocal statehood exists in the mode of weak permeation. The central question of our approach is how local forms of self-governance work in this context. We will examine the relations to the state level as well as to other local groups as they develop over time; the scope and spatial contingency of forms of self-governance; their legitimization and the interdependency with the organization and collective identity of those groups which carry them out; finally, we will turn our attention to the significance of self-governance for the configuration of weak statehood. The empirical focus will be at the local level, which has so far been largely neglected in the research on governance beyond the state. In order to achieve this, we will work with case studies that are structured by categories and situated in geographical areas and time periods that lie outside of modern Europe with its particular development of statehood since the Late Middle Ages: in Antiquity, and in the Global South of the present. By incorporating these different time frames, we hope to contribute to overcoming the dichotomy between the modern and pre-modern era, which is often given canonical status. Our goal is to create a comparative analysis of different configurations of order as well as the development of a typology of patterns of local governance. The structure of the empirical comparison itself promises methodological insights, since it will entail recognizing, dealing with, and overcoming disciplinary limitations. Starting with the identification of typical patterns and processes, we hope to gain a better grasp of the mechanisms by which local configurations of order succeed, while at the same time advancing the theoretical debate. This will allow us to make an interdisciplinary contribution to the understanding of fundamental elements of statehood and local governance that are of central importance, especially in the context of weak statehood. The insights we hope to gain by adopting this historical perspective will contribute to understanding a present that is not based exclusively on its own, seemingly completely new preconditions, and will thus significantly sharpen the political analysis of various forms of governance. Primary analyses such as quality control, normalization, statistics and mapping are highly complex and need to be performed by specialists. Thereafter, results are handed back to biomedical researchers, who are then confronted with complicated data lists. For rather simple tasks like data filtering, sorting and cross-association there is a need for new tools which can be used by non-specialists. Here, we describe CrossQuery, a web tool that enables straight forward, simple syntax queries to be executed on transcriptome sequencing and microarray datasets. We provide deepsequencing data sets of stem cell lines derived from the model fish Medaka and microarray data of human endothelial cells. In the example datasets provided, mRNA expression levels, gene, transcript and sample identification numbers, GO-terms and gene descriptions can be freely correlated, filtered and sorted. Queries can be saved for later reuse and results can be exported to standard formats that allow copy-and-paste to all widespread data visualization tools such as Microsoft Excel. CrossQuery enables researchers to quickly and freely work with transcriptome and microarray data sets requiring only minimal computer skills. Furthermore, CrossQuery allows growing association of multiple datasets as long as at least one common point of correlated information, such as transcript identification numbers or GO-terms, is shared between samples. Its invasive success has been attributed to its vigorous resistance against diverse pathogens. This study demonstrates that harmonine ((17R,9Z)-1,17-diaminooctadec-9-ene), which is present in H. axyridis haemolymph, displays broad-spectrum antimicrobial activity that includes human pathogens. Antibacterial activity is most pronounced against fast-growing mycobacteria and Mycobacterium tuberculosis, and the growth of both chloroquine-sensitive and -resistant Plasmodium falciparum strains is inhibited. Harmonine displays gametocytocidal activity, and inhibits the exflagellation of microgametocytes and zygote formation. Because gametocytes are the only parasite stages able to establish an infection in the mosquito, they play a crucial role in spreading the tropical disease. The human-to-mosquito transmission triggers important molecular changes in the gametocytes, which initiate gametogenesis and prepare the parasite for life-cycle progression in the insect vector. Results: To better understand gene regulations during the initial phase of malaria parasite transmission, we focused on the transcriptome changes that occur within the first half hour of parasite development in the mosquito. Comparison of mRNA levels of P. falciparum gametocytes before and 30 min following activation using suppression subtractive hybridization (SSH) identified 126 genes, which changed in expression during gametogenesis. Among these, 17.5% had putative functions in signaling, 14.3% were assigned to cell cycle and gene expression, 8.7% were linked to the cytoskeleton or inner membrane complex, 7.9% were involved in proteostasis and 6.4% in metabolism, 12.7% were cell surface-associated proteins, 11.9% were assigned to other functions, and 20.6% represented genes of unknown function. For 40% of the identified genes there has as yet not been any protein evidence. For a subset of 27 genes, transcript changes during gametogenesis were studied in detail by real-time RT-PCR. Of these, 22 genes were expressed in gametocytes, and for 15 genes transcript expression in gametocytes was increased compared to asexual blood stage parasites. Transcript levels of seven genes were particularly high in activated gametocytes, pointing at functions downstream of gametocyte transmission to the mosquito. For selected genes, a regulated expression during gametogenesis was confirmed on the protein level, using quantitative confocal microscopy. Conclusions: The obtained transcriptome data demonstrate the regulations of gene expression immediately following malaria parasite transmission to the mosquito. Our findings support the identification of proteins important for sexual reproduction and further development of the mosquito midgut stages and provide insights into the genetic basis of the rapid adaption of Plasmodium to the insect vector. In this study, we produced and purified recombinant Pf38 and a fusion protein composed of red fluorescent protein and Pf38 (RFP-Pf38) using a transient expression system in the plant Nicotiana benthamiana. To our knowledge, this is the first description of the production of recombinant Pf38. To verify the quality of the recombinant Pf38, plasma from semi-immune African donors was used to confirm specific binding to Pf38. ELISA measurements revealed that immune responses to Pf38 in this African subset were comparable to reactivities to AMA-1 and \(MSP1_{19}\). Pf38 and RFP-Pf38 were successfully used to immunise mice, although titres from these mice were low (on average 1:11.000 and 1:39.000, respectively). In immune fluorescence assays, the purified IgG fraction from the sera of immunised mice recognised Pf38 on the surface of schizonts, gametocytes, macrogametes and zygotes, but not sporozoites. Growth inhibition assays using \(\alpha Pf38\) antibodies demonstrated strong inhibition \((\geq 60 \% ) \) of the growth of blood-stage P. falciparum. The development of zygotes was also effectively inhibited by \(\alpha Pf38\) antibodies, as determined by the zygote development assay. Collectively, these results suggest that Pf38 is an interesting candidate for the development of a malaria vaccine. In the invasive merozoites, these include members of the PfMSP1 and PfAMA1/RON complexes, which facilitate contact between merozoites and red blood cells. In gametocytes, sexual precursor cells mediating parasite transmission to the mosquito vector, plasma membrane-associated proteins primarily belong to the PfCCp and 6-cys families with roles in fertilization. This study describes a newly identified WD40-repeat protein unique to Plasmodium species that associates with adhesion protein complexes of both merozoites and gametocytes. Methods The WD40-repeat protein-like protein PfWLP1 was identified via co-immunoprecipitation assays followed by mass spectrometry and characterized using biochemical and immunohistochemistry methods. Reverse genetics were employed for functional analysis. Results PfWLP1 is expressed both in schizonts and gametocytes. In mature schizonts, the protein localizes underneath the merozoite micronemes and interacts with PfAMA1, while in gametocytes PfWLP1 primarily accumulates underneath the plasma membrane and associates with PfCCp1 and Pfs230. Reverse genetics failed to disrupt the pfwlp1 gene, while haemagglutinin-tagging was feasible, suggesting a crucial function for PfWLP1 during blood stage replication. Conclusions This is the first report on a plasmodial WD40-repeat protein associating with cell adhesion proteins. Since WD40 domains are known to mediate protein–protein contact by serving as a rigid scaffold for protein interactions, the presented data suggest that PfWLP1 supports the stability of adhesion protein complexes of the plasmodial blood stages. Besides the ultimate goal of inducing sterile protection in vaccinated individuals, the prevention of transmission by so-called transmission blocking vaccines (TBVs) is being regarded as an important feature of an efficient malaria eradication strategy. Recently, Plasmodium falciparum GAP50 (PfGAP50), a 44.6 kDa transmembrane protein that forms an essential part of the invasion machinery (glideosome) multi-protein complex, has been proposed as novel potential transmission-blocking candidate. Plant-based expression systems combine the advantages of eukaryotic expression with a up-scaling potential and a good product safety profile suitable for vaccine production. In this study we investigated the feasibility to use the transient plant expression to produce PfGAP50 suitable for the induction of parasite specific inhibitory antibodies. Results: We performed the transient expression of recombinant PfGAP50 in Nicotiana benthamiana leaves using endoplasmatic reticulum (ER) and plastid targeting. After IMAC-purification the protein yield and integrity was investigated by SDS-PAGE and Western Blot. Rabbit immune IgG derived by the immunization with the plastidtargeted variant of PfGAP50 was analyzed by immune fluorescence assay (IFA) and zygote inhibition assay (ZIA). PfGAP50 could be produced in both subcellular compartments at different yields IMAC (Immobilized Metal Affinity Chromatography) purification from extract yielded up to 4.1 mu g/g recombinant protein per fresh leaf material for ER-retarded and 16.2 mu g/g recombinant protein per fresh leave material for plasmid targeted PfGAP50, respectively. IgG from rabbit sera generated by immunization with the recombinant protein specifically recognized different parasite stages in immunofluorescence assay. Furthermore up to 55 % inhibition in an in vitro zygote inhibition assay could be achieved using PfGAP50-specific rabbit immune IgG. Conclusions: The results of this study demonstrate that the plant-produced PfGAP50 is functional regarding the presentation of inhibitory epitopes and could be considered as component of a transmission-blocking malaria vaccine formulation. Primary analyses such as quality control, normalization, statistics and mapping are highly complex and need to be performed by specialists. Thereafter, results are handed back to biomedical researchers, who are then confronted with complicated data lists. For rather simple tasks like data filtering, sorting and cross-association there is a need for new tools which can be used by non-specialists. Here, we describe CrossQuery, a web tool that enables straight forward, simple syntax queries to be executed on transcriptome sequencing and microarray datasets. We provide deep-sequencing data sets of stem cell lines derived from the model fish Medaka and microarray data of human endothelial cells. In the example datasets provided, mRNA expression levels, gene, transcript and sample identification numbers, GO-terms and gene descriptions can be freely correlated, filtered and sorted. Queries can be saved for later reuse and results can be exported to standard formats that allow copy-and-paste to all widespread data visualization tools such as Microsoft Excel. CrossQuery enables researchers to quickly and freely work with transcriptome and microarray data sets requiring only minimal computer skills. Furthermore, CrossQuery allows growing association of multiple datasets as long as at least one common point of correlated information, such as transcript identification numbers or GO-terms, is shared between samples. For advanced users, the object-oriented plug-in and event-driven code design of both server-side and client-side scripts allow easy addition of new features, data sources and data types. Methods/Principal Findings: We performed a phase II, double blind, multicentre, placebo controlled trial of pioglitazone in ALS patients under riluzole. 219 patients were randomly assigned to receive 45 mg/day of pioglitazone or placebo (one: one allocation ratio). The primary endpoint was survival. Secondary endpoints included incidence of non-invasive ventilation and tracheotomy, and slopes of ALS-FRS, slow vital capacity, and quality of life as assessed using EUROQoL EQ-5D. The study was conducted under a two-stage group sequential test, allowing to stop for futility or superiority after interim analysis. Shortly after interim analysis, 30 patients under pioglitazone and 24 patients under placebo had died. The trial was stopped for futility; the hazard ratio for primary endpoint was 1.21 (95% CI: 0.71-2.07, p = 0.48). Secondary endpoints were not modified by pioglitazone treatment. Pioglitazone was well tolerated. Conclusion/Significance: Pioglitazone has no beneficial effects on the survival of ALS patients as add-on therapy to riluzole. However, subcellular localization of cytoplasmic actin isoforms and proteins involved in the modulation of the cytoplasmic actin networks are elusive. Mena and VASP are important regulators of actin dynamics. Due to the lethal phenotype of mice with combined deficiency in Mena and VASP, however, distinct cardiac roles of the proteins remain speculative. In the present study, we analyzed the physiological functions of Mena and VASP in the heart and also investigated the role of the proteins in the organization of cytoplasmic actin networks. Results: We generated a mouse model, which simultaneously lacks Mena and VASP in the heart. Mena/VASP double-deficiency induced dilated cardiomyopathy and conduction abnormalities. In wild-type mice, Mena and VASP specifically interacted with a distinct αII-Spectrin splice variant (SH3i), which is in cardiomyocytes exclusively localized at Z- and intercalated discs. At Z- and intercalated discs, Mena and β-actin localized to the edges of the sarcomeres, where the thin filaments are anchored. In Mena/VASP double-deficient mice, β-actin networks were disrupted and the integrity of Z- and intercalated discs was markedly impaired. Conclusions: Together, our data suggest that Mena, VASP, and αII-Spectrin assemble cardiac multi-protein complexes, which regulate cytoplasmic actin networks. Conversely, Mena/VASP deficiency results in disrupted β-actin assembly, Z- and intercalated disc malformation, and induces dilated cardiomyopathy and conduction abnormalities. Objective: We aimed to externally validate the proposed asthma prediction method in a German birth cohort. Methods: The MAS-90 study (Multicentre Allergy Study) recorded details on allergic diseases prospectively in about yearly follow-up assessments up to age 20 years in a cohort of 1,314 children born 1990. We replicated the scoring method from the Leicester cohort and assessed prediction, performance and discrimination. The primary outcome was defined as the combination of parent-reported wheeze and asthma drugs (both in last 12 months) at age 8. Sensitivity analyses assessed model performance for outcomes related to asthma up to age 20 years. Results: For 140 children parents reported current wheeze or cough at age 3 years. Score distribution and frequencies of later asthma resembled the Leicester cohort: 9% vs. 16% (MAS-90 vs. Leicester) of children at low risk at 3 years had asthma at 8 years, at medium risk 45% vs. 48%. Performance of the asthma prediction tool in the MAS-90 cohort was similar (Brier score 0.22 vs. 0.23) and discrimination slightly better than in the original cohort (area under the curve, AUC 0.83 vs. 0.78). Prediction and discrimination were robust against changes of inclusion criteria, scoring and outcome definitions. The secondary outcome 'physicians' diagnosed asthma at 20 years' showed the highest discrimination (AUC 0.89). Conclusion: The novel asthma prediction tool from the Leicester cohort, UK, performed well in another population, a German birth cohort, supporting its use and further development as a simple aid to predict asthma risk in clinical settings. Mutations in six spliceosomal proteins, PRPF3, PRPF4, PRPF6, PRPF8, PRPF31 and SNRNP200, cause retinitis pigmentosa (RP), a disease characterized by progressive photoreceptor degeneration. All splicing factors linked to RP are constituents of the U4/U6.U5 tri-snRNP subunit of the spliceosome, suggesting that the compromised function of this particle may lead to RP. Here, we report the identification of the p.R192H variant of the tri-snRNP factor PRPF4 in a patient with RP. The mutation affects a highly conserved arginine residue that is crucial for PRPF4 function. Introduction of a corresponding mutation into the zebrafish homolog of PRPF4 resulted in a complete loss of function in vivo. A series of biochemical experiments suggested that p.R192H disrupts the binding interface between PRPF4 and its interactor PRPF3. This interferes with the ability of PRPF4 to integrate into the tri-snRNP, as shown in a human cell line and in zebrafish embryos. These data suggest that the p.R192H variant of PRPF4 represents a functional null allele. The resulting haploinsufficiency of PRPF4 compromises the function of the tri-snRNP, reinforcing the notion that this spliceosomal particle is of crucial importance in the physiology of the retina. The need for isoform-specific regulation is reflected by the fact that all RAFs not only display a different degree of activity but also perform isoform-specific functions at diverse cellular compartments. Protein-protein-interactions and phosphorylation events are essential for the signal propagation along the Ras-RAF-MEK-ERK cascade. More than 40 interaction partners of RAF kinases have been described so far. Two of the most important regulators of RAF activity, namely Ras and 14-3-3 proteins, are subject of this work. So far, coupling of RAF with its upstream modulator protein Ras has only been investigated using truncated versions of RAF and regardless of the lipidation status of Ras. We quantitatively analyzed the binding properties of full-length B- and C-RAF to farnesylated H-Ras in presence and absence of membrane lipids. While the isolated Ras-binding domain of RAF exhibit a high binding affinity to both, farnesylated and nonfarnesylated H-Ras, the full-length RAF kinases demonstrate crucial differences in their affinity to Ras. In contrast to C-RAF that requires carboxyterminal farnesylated H-Ras for interaction at the plasma membrane, B-RAF also binds to nonfarnesylated H-Ras in the cytosol. For identification of the potential farnesyl binding site we used several fragments of the regulatory domain of C-RAF and found that the binding of farnesylated H-Ras is considerably increased in the presence of the cysteine-rich domain of RAF. In B-RAF a sequence of 98 amino acids at the extreme N terminus enables binding of Ras independent of its farnesylation status. The deletion of this region altered Ras binding as well as kinase properties of B-RAF to resemble C-RAF. Immunofluorescence studies in mammalian cells revealed essential differences between B- and C-RAF regarding the colocalization with Ras. In conclusion, our data suggest that that B-RAF, in contrast to C-RAF, is also accessible for nonfarnesylated Ras in the cytosolic environment due to its prolonged N terminus. Therefore, the activation of B-RAF may take place both at the plasma membrane and in the cytosolic environment. Furthermore, the interaction of RAF isoforms with Ras at different subcellular sites may also be governed by the complex formation with 14-3-3 proteins. 14-3-3 adapter proteins play a crucial role in the activation of RAF kinases, but so far no information about the selectivity of the seven mammalian isoforms concerning RAF association and activation is available. We analyzed the composition of in vivo RAF/14-3-3 complexes isolated from mammalian cells with mass spectrometry and found that B-RAF associates with a greater variety of 14-3-3 proteins than C- and A-RAF. In vitro binding assays with purified proteins supported this observation since B-RAF showed highest affinity to all seven 14-3-3 isoforms, whereas C-RAF exhibited reduced affinity to some and A-RAF did not bind to the 14-3-3 isoforms epsilon, sigma, and tau. To further examine this isoform specificity we addressed the question of whether both homo- and heterodimeric forms of 14-3-3 proteins participate in RAF signaling. By deleting one of the two 14-3-3 isoforms in Saccharomyces cerevisiae we were able to show that homodimeric 14-3-3 proteins are sufficient for functional activation of B- and C-RAF. In this context, the diverging effect of the internal, inhibiting and the activating C-terminal 14-3-3 binding domain in RAF could be demonstrated. Furthermore, we unveil that prohibitin stimulates C-RAF activity by interfering with 14-3-3 at the internal binding site. This region of C-RAF is also target of phosphorylation as part of a negative feedback loop. Using tandem MS we were able to identify so far unknown phosphorylation sites at serines 296 and 301. Phosphorylation of these sites in vivo, mediated by activated ERK, leads to inhibition of C-RAF kinase activity. The relationship of prohibitin interference with 14-3-3 binding and phosphorylation of adjacent sites has to be further elucidated. Taken together, our results provide important new information on the isoform-specific regulation of RAF kinases by differential interaction with Ras and 14-3-3 proteins and shed more light on the complex mechanism of RAF kinase activation. N2 - RAF Protein Kinasen sind essentielle Regulatoren verschiedener zellulärer Prozesse. Unterschiedlich starke Aktivitäten und Lokalisation der drei RAF Isoformen erfordern eine isoform-spezifische Regulation. Der Einfluss von Protein-Protein Interaktionen und Phosphorylierungen ist dabei mitentscheidend für die Signalweiterleitung entlang der Ras-RAF-MEK-ERK Kaskade. Mehr als 40 Interaktionspartner der RAF Kinasen wurden bereits beschrieben von denen zwei der wichtigsten, Ras und 14-3-3 Proteine, Gegenstand der vorliegenden Arbeit sind. Die Interaktion von RAF mit seinem vorgeschaltetem Modulatorprotein Ras wurde bislang nur mit verkürzten RAF-Proteinen und ohne Rücksicht auf den Lipidierungsgrad von Ras untersucht. Wir haben die Bindeeigenschaften von B- und C-RAF in voller, nativer Länge zu farnesyliertem H-Ras in Gegenwart und Abwesenheit von Membranlipiden quantifiziert. Während die isolierte Ras-Bindungsdomäne eine hohe Affinität sowohl zu farnesyliertem als auch nicht-farnesyliertem H-Ras aufweist, zeigen die RAF Proteine in voller Länge entscheidende Unterschiede in ihrem Bindeverhalten zu Ras. C-RAF benötigt für eine effiziente Interaktion mit H-Ras dessen C-terminale Farnesylgruppe, wobei B-RAF auch an nicht-farnesyliertes H-Ras im Cytosol bindet. Um die verantwortliche Farnesylbinderegion zu identifizieren haben wir verschiedene Fragmente der regulatorischen Domäne von C-RAF eingesetzt. Dadurch konnten wir zeigen, dass die Affinität zu farnesyliertem Ras in Gegenwart der sogenannten Cystein-reichen Domäne von RAF beträchtlich erhöht war. In B-RAF ist eine Sequenz von 98 Aminosäuren am N-Terminus verantwortlich für die Ras-Bindung unabhängig von dessen Farnesylierungszustand. Die Deletion dieser Sequenz von B-RAF veränderte die Ras-Bindungseigenschaften sowie die Kinaseaktivität vergleichbar mit C-RAF. Durch Immunfluoreszenzversuche in Säugerzellen konnten darüber hinaus Unterschiede in der Kolokalisation von B- und C-RAF mit Ras beobachtet werden. Zusammenfassend deuten unsere Ergebnisse darauf hin, dass B-RAF, im Gegensatz zu C-RAF, aufgrund seines verlängerten N-Terminus in der Lage ist bereits im Cytosol auch mit unfarnesyliertem Ras zu interagieren, wodurch die Aktivierung von B-RAF sowohl im Cytosol als auch an der Plasmamenbran erfolgen kann. Die Interaktion der RAF-Isoformen mit Ras in unterschiedlichen zellulären Kompartimenten kann aber auch durch die Komplexbildung mit 14-3-3 Proteinen beeinflusst werden. Die 14-3-3 Adapter Proteine spielen eine entscheidende Rolle im Aktivierungszyklus der RAF Proteine. Bislang waren jedoch keine Details bezüglich der Selektivität der sieben 14-3-3 Isoformen aus Säugerzellen hinsichtlich der Assoziation mit und Aktivierung der RAF Kinasen bekannt. Wir haben RAF/14-3-3 Komplexe aus Säugerzellen isoliert und durch Massenspektrometrie analysiert. Dadurch konnten wir zeigen, dass B-RAF mit einer größeren Vielfalt an 14-3-3 Isoformen bindet als C- und A-RAF. In vitro Bindungsversuche mit gereinigten Proteinen bestätigten die höhere Affinität von B-RAF zu allen sieben Säuger-14-3-3 Proteinen. C-RAF dagegen zeigte eine deutlich reduzierte Affinität, während für A-RAF keine Bindung zu den 14-3-3 Isoformen epsilon, sigma, und tau festgestellt wurde. Um diese Isoformspezifität weiter aufzuklären haben wir untersucht, ob sowohl Homo- als auch Heterodimere von 14-3-3 in der Lage sind die RAF-Signaltransduktion zu beeinflussen. Durch die Deletion einer der beiden 14-3-3 Isoformen aus Saccharomyces cerevisiae konnten wir zeigen, dass bereits ein 14-3-3 Homodimer für die korrekte Aktivierung von B- und C-RAF ausreichend ist. In diesem Zusammenhang konnte auch die Rolle der internen, inhibierenden 14-3-3 Bindestelle in RAF gegenüber der C-terminalen, aktivierenden Stelle dargelegt werden. Zusätzlich zeigen wir, dass Prohibitin seinen aktivierenden Einfluss gegenüber C-RAF durch die Beeinträchtigung der 14-3-3 Bindung an der internen Stelle in RAF ausübt. Diese Region in C-RAF ist das Ziel von Phosphorylierungen im Zuge eines negativen Rückkopplungsmechanismus. Durch den Einsatz von Tandem-Massenspektrometrie konnten wir bislang unbekannte Phosphorylierungsstellen an den Serinen 296 und 301 identifizieren deren ERK-vermittelte Phosphorylierung in vivo eine Inaktivierung der C-RAF bewirkt. Der Zusammenhang zwischen der Behinderung der 14-3-3 Anlagerung durch Prohibitin und die Phosphorylierung in unmittelbarer Nachbarschaft bedarf weiterer Untersuchungen. Dabei spielt der Notch-Signaltransduktionsweg eine zentrale Rolle während der Determination von Zellschicksalen und der Zelldifferenzierung. Die primären Zielgene der Notch-Signalkaskaskade bei Vertebraten sind die Hes- sowie die kürzlich identifizierten Hey-Gene. Die Hey-(hairy and E(spl) related with YRPW motif)-Gene kodieren drei hairy/E(spl)/Hes-verwandte basische Helix-Loop-Helix-Transkriptionsfaktoren, die durch eine Orange-Domäne und einen charakteristischen Carboxyterminus gekennzeichnet sind. Während der Embryonalentwicklung werden die Hey-Gene dynamisch in zahlreichen Geweben exprimiert. Ziel dieser Arbeit war es, neue Hey-Interaktionsproteine aus embryonalen Genbanken zu isolieren, die Bindung an weitere bHLH-Transkriptionsfaktoren zu überprüfen und ihre DNA-Bindung zu analysieren. Um die physiologische Hey2-Funktion zu ergründen, wurden Hey2-Knockoutmäuse untersucht. In einem ersten Versuch wurde eine neue Screeningmethode erprobt, bei der Proteinexpressionsfilter mit markierten Hey1-Peptiden nach interagierenden Proteinen durchsucht wurden. Hierbei sind 53 Proteine isoliert worden, jedoch konnte nach eingehenderen Untersuchungen kein relevanter Bindungsspartner beschrieben werden. Für weitere Analysen unter mehr physiologischen Bedingungen wurde das Yeast Two-Hybrid Verfahren für Hey1 und Hey2 etabliert. Das Screening von murinen embryonalen cDNA-Genbanken mit verschiedenen Hey1-Fragmenten führte zur Isolation von mehreren hundert Klonen. Die interessantesten Kandidaten wurden weiteren biochemischen Tests unterzogen, wobei jedoch keine neuen Interaktionspartner verifiziert werden konnten. Mit gezielten direkten Yeast Two-Hybrid und GST-Pulldown Assays für vermutete Kandidaten konnte jedoch die Interaktion von Hey1 bzw. Hey2 mit den bHLH-Proteinen E2-2, E2-5, MyoD und c-hairy1 nachgewiesen werden. Außerdem wurde festgestellt, dass Hey1 und Hey2 Homodimere und Hey1/Hey2-Heterodimere bilden. Die stärkste Interaktion wurde mit dem in der Somitogenese rhythmisch exprimierten c-hairy1-Protein beobachtet. Da Hey2 und c-hairy1 im präsomitischen Mesoderm und in den Somiten coexprimiert werden und starke Heterodimere ausbilden, erscheint es wahrscheinlich, dass beide Proteine gemeinsam die Transkription nachgeschalteter Gene steuern. Diese Interaktionsstudien zeigten außerdem erstmals, dass die Orange-Domäne entscheidend an der Bildung der Dimere beteiligt ist, da durch sie die Dimerisierung in vivo deutlich verstärkt wurde. Schließlich konnte gezeigt werden, dass Hey1 und Hey2, im Gegensatz zu den übrigen hairy-Proteinen, nicht mit dem Corepressor Groucho/TLE1 interagieren. Electrophoretic Mobility Shift Assays ergaben, dass die Hey1- und Hey2-Proteine an eine E(spl)-spezifische E-Box DNA-Sequenz (CACGTG) binden. Auch die interagierenden bHLH-Proteine c-hairy1, E2-2 und E2-5 binden als Homodimere an diese DNA-Sequenz. Im zweiten Teil dieser Arbeit wurde die Hey2-Genfunktion an Hey2-Knockoutmäusen untersucht. Etwa 80 % der homozygoten Mäuse starben wenige Tage nach der Geburt. Sie zeigten eine massive Hypertrophie der Herzventrikel, die wahrscheinlich die Todesursache darstellt. Die lacZ-Expression der untersuchten Organe entsprach der Hey2-Expression im Wildtyp. Es fiel dabei auf, dass es postnatal zu einer Herunterregulation der Hey2-Transkription kommt. Mit Elektrokardiogrammen wurden keine Reizleitungsstörungen bei neugeborenen Hey2-Knockoutmäusen festgestellt. Interessanterweise konnte mit Arteriographien ausgeschlossen werden, dass die Ventrikelhypertophie Folge einer Aortenstenose wie bei der gridlock (zf-Hey2)-Mutante im Zebrafisch ist. Vielmehr führt eine homozygote Hey2-Deletion zu einer Kardiomyopathie in Kombination mit verschiedenene Herzfehlern. Untersuchungen der Hey1- und HeyL-Expression in Hey2-Knockoutembryonen mittels RNA in situ Hybridisierungen zeigten keine Veränderungen im Vergleich mit dem Wildtyp. Daraus kann gefolgert werden, dass Hey1 und HeyL zumindest dort, wo sie nicht mit Hey2 coexprimiert sind, die Hey2-Funktionen nicht kompensieren können. Weitere Erkenntnisse über die Funktionen der Hey-Gene werden sicherlich die Studien an den Doppelknockoutmäusen ergeben. Die bisherigen Ergebnisse zeigen eindeutig, dass die Hey-Gene essentiell für die murine Herzentwicklung sind. Weitere Untersuchungen müssen nun zeigen, welche Rolle diese Gene bei der Entstehung von kongenitalen Herzfehlern des Menschen spielen. N2 - The development from a single fertilized oocyte to a multicellular organism requires complex cellular regulatory mechanisms. During this process the Notch signalling pathway plays a crucial role in determining cell fate and differentiation. Primary target genes of the Notch signalling cascade in vertebrates are Hes and the recently identified Hey genes. Hey (hairy and E(spl) related with YRPW motif) genes encode three hairy/E(spl)/Hes related basic helix-loop-helix transcription factors characterised by an Orange domain and a conserved carboxyterminus. During embryonic development Hey genes are dynamically expressed in various tissues. The goal of this study was to isolate novel Hey-interacting proteins from embryonic cDNA libraries, to analyse the binding to other bHLH transcription factors and to examine their DNA-binding properties. To elucidate the physiological role of Hey2, Hey2 knockout mice were analysed. First, a new screening method was used to identify Hey1-binding proteins from protein expression filters hybridised with labelled Hey1 peptides. Out of 53 candidates isolated no relevant interaction partner could be verified after more detailed examination. For further analysis under more physiological conditions the yeast-two hybrid system was established for Hey1 and Hey2. Screening of murine embryonic cDNA libraries with different Hey1 fragments led to the identification of several hundred clones. The most interesting ones were further analysed with biochemical assays, but no novel interaction partner could be verified. With direct yeast-two hybrid and GST-pulldown assays of candidate proteins an interaction of Hey1 and Hey2 with the bHLH proteins E2-2, E2-5, MyoD and c-hairy1 was found. Furthermore, it could be shown that Hey1 and Hey2 can form homodimers as well as Hey1/Hey2 heterodimers. However, the strongest interaction was seen with c-hairy1, which is dynamically expressed during somitogenesis. Hey2 and c-haiy1 are coexpressed in the presomitic mesoderm and in somites and their strong interaction suggests that both proteins together regulate the transcription of target genes. These interaction studies showed for the first time that the Orange domain is important for dimer formation in vivo, because it strongly increased binding strength. Furthermore, it could be shown that Hey1 and Hey2 do not interact with the corepressor Groucho/TLE1, which is in contrast to all other hairy-related proteins. Electrophoretic mobility shift assays revealed that Hey1 and Hey2 proteins are able to bind an E(spl)-specific E-box DNA sequence (CACGTG). The interacting bHLH proteins c-hairy1, E2-2 and E2-5 could also bind to this sequence as homodimers. The aim of the second part of this study was to examine Hey2-lacZ knockout mice. About 80 % of the homozygous mice died within a few days after birth. They exhibited massive ventricular hypertrophy as the most likely cause of death. Expression of lacZ in the organs analysed was comparable to Hey2 in wildtype. It became clear that Hey2 transcription is downregulated postnatally. Electrocardiography showed no conduction failure or arrythmia in newborn knockouts. Interestingly, it could be ruled out by angiography that ventricular hypertrophy is caused by aortic coarctation as seen in the gridlock (zf-Hey2) zebrafish mutant. However, loss of Hey2 leads to cardiomyopathy combined with various cardiac structural defects. Hey1 and HeyL expression in Hey2 knockout embryos was not altered as shown by RNA in situ hybridisation. Therefore, it can be concluded that Hey1 and HeyL can not compensate Hey2 function, at least in organs where they are not coexpressed. A better understanding of the Hey genes will be achieved by studying double knockout mice. This work clearly shows that Hey genes are essential for murine heart development. Further analysis will reveal if these genes also play a role in congenital heart disease in humans. Hier entwickelt eine Gemeinschaft jenseits der Familie zuerst kollektive Regeln, die ihren Fortbestand sichern sollen. Meist ist aber nicht nur diese Regelungsebene vorhanden. Über ihr stehen überlokale Herrschaftsformationen – von regionalen Verbünden bis zum Imperium –, welche die Ordnungsangebote vor Ort ergänzen oder mit ihnen konkurrieren. Örtliche Selbstregelungen sind, so die Prämisse dieser Forschungsgruppe, dann besonders vielfältig und ausgeprägt, wenn überlokale Staatlichkeit im Modus der schwachen Durchdringung existiert. Wie lokale Selbstregelungen in diesem Kontext funktionieren, ist unsere zentrale Forschungsfrage. Wir untersuchen die Relationen zu den staatlichen Ebenen wie zu anderen lokalen Gruppen in ihrem zeitlichen Verlauf, wir analysieren die Reichweite und die räumliche Bedingtheit von Selbstregelungen, fragen nach ihrer Legitimierung sowie nach der Interdependenz zu Organisation und kollektiver Identität der sie tragenden Gruppen; schließlich wenden wir uns der Bedeutung der Selbstregelungen für die Ordnungsform der schwachen Staatlichkeit zu. Der empirische Fokus liegt auf der lokalen Ebene, die in der bisherigen Forschung zum Regieren jenseits des Staates wenig beachtet wurde. Dazu wird in kategorial strukturierten Fallstudien gearbeitet, die in räumlichen und zeitlichen Bereichen außerhalb der europäischen (Sonder-)Entwicklung von Staatlichkeit seit dem Hochmittelalter situiert sind: in der griechisch-römischen Antike und im Globalen Süden der Gegenwart. Mit der unterschiedlichen Zeitstellung möchten wir zur Überwindung der oft als kanonisch geltenden Dichotomie zwischen Moderne und Vormoderne beitragen. N2 - The nucleus of statehood is situated at the local level: in the village, the neighborhood, the city district. This is where a community, beyond the level of the family, first develops collective rules that are intended to ensure its continued existence. But usually this is not the only level of governance at play. Above it, there are supralocal formations of power, varying in scope from regional networks to empires, which supplement the local orders or compete with them. The premise of the research unit is that local forms of self-governance are especially heterogeneous and prominent, wherever supralocal statehood exists in the mode of weak permeation. The central question of our approach is how local forms of self-governance work in this context. We will examine the relations to the state level as well as to other local groups as they develop over time; the scope and spatial contingency of forms of self-governance; their legitimization and the interdependency with the organization and collective identity of those groups which carry them out; finally, we will turn our attention to the significance of self-governance for the configuration of weak statehood. The empirical focus will be at the local level, which has so far been largely neglected in the research on governance beyond the state. In order to achieve this, we will work with case studies that are structured by categories and situated in geographical areas and time periods that lie outside of modern Europe with its particular development of statehood since the Late Middle Ages: in Antiquity and in the Global South of the present. By incorporating these different time frames, we hope to contribute to overcoming the dichotomy between the modern and pre-modern era, which is often given canonical status. Our goal is a comparative analysis of different configurations of order as well as the development of a typology of patterns of local governance. The structure of the empirical comparison itself promises methodological insights, since it will entail recognizing, dealing with, and overcoming disciplinary limitations. Starting with the identification of typical patterns and processes, we hope to get a better grasp of the mechanisms by which local configurations of order succeed, while at the same time advancing the theoretical debate. This will allow us to make an interdisciplinary contribution to the understanding of fundamental elements of statehood and local governance that are of central importance, especially in the context of weak statehood. Even though it is currently gaining popularity again, the contours of the notion of interdisciplinarity often remain blurred. We therefore explain how the interdisciplinary setup and interconnectedness of the DFG Research Unit 2757 contribute to approaching a complex research agenda that lies beyond the scope of individual disciplines. To this end, we first elucidate the fundamental terms, theories and methods, developed and selected through interdisciplinary reflection, and present the basic ideas of our Research Unit. Furthermore, we describe the organizational structure and explain how the subprojects are connected both in terms of content and methodology. In contrast, brown adipose tissue (BAT) combusts energy to produce heat. Here we show that a small molecule stimulator (BAY 41-8543) of soluble guanylyl cyclase (sGC), which produces the second messenger cyclic GMP (cGMP), protects against diet-induced weight gain, induces weight loss in established obesity, and also improves the diabetic phenotype. Mechanistically, the haeme-dependent sGC stimulator BAY 41-8543 enhances lipid uptake into BAT and increases whole-body energy expenditure, whereas ablation of the haeme-containing \(\beta\)\(_{1}\)-subunit of sGC severely impairs BAT function. Notably, the sGC stimulator enhances differentiation of human brown adipocytes as well as induces 'browning' of primary white adipocytes. Taken together, our data suggest that sGC is a potential pharmacological target for the treatment of obesity and its comorbidities. To date, 10 genes have been identified to be causative for ARCI. NIPAL4 (Nipa‐Like Domain‐Containing 4) is the second most commonly mutated gene in ARCI. In this study, we present a large cohort of 101 families affected with ARCI carrying mutations in NIPAL4. We identified 16 novel mutations and increase the total number of pathogenic mutations in NIPAL4 to 34. Ultrastructural analysis of biopsies from six patients showed morphological abnormalities consistent with an ARCI EM type III. One patient with a homozygous splice site mutation, which leads to a loss of NIPAL4 mRNA, showed additional ultrastructural aberrations together with a more severe clinical phenotype. Our study gives insights into the frequency of mutations, a potential hot spot for mutations, and genotype–phenotype correlations. A1 - Schillberg, Stefan A1 - Reimann, Andreas A1 - Fischer, Rainer T1 - Analysis of a multi-component multi-stage malaria vaccine candidate—tackling the cocktail challenge JF - PLoS ONE N2 - Combining key antigens from the different stages of the P. falciparum life cycle in the context of a multi-stage-specific cocktail offers a promising approach towards the development of a malaria vaccine ideally capable of preventing initial infection, the clinical manifestation as well as the transmission of the disease. To investigate the potential of such an approach we combined proteins and domains (11 in total) from the pre-erythrocytic, blood and sexual stages of P. falciparum into a cocktail of four different components recombinantly produced in plants. After immunization of rabbits we determined the domain-specific antibody titers as well as component-specific antibody concentrations and correlated them with stage specific in vitro efficacy. Using purified rabbit immune IgG we observed strong inhibition in functional in vitro assays addressing the pre-erythrocytic (up to 80%), blood (up to 90%) and sexual parasite stages (100%). Based on the component-specific antibody concentrations we calculated the IC50 values for the pre-erythrocytic stage (17–25 μg/ml), the blood stage (40–60 μg/ml) and the sexual stage (1.75 μg/ml). While the results underline the feasibility of a multi-stage vaccine cocktail, the analysis of component-specific efficacy indicates significant differences in IC50 requirements for stage-specific antibody concentrations providing valuable insights into this complex scenario and will thereby improve future approaches towards malaria vaccine cocktail development regarding the selection of suitable antigens and the ratios of components, to fine tune overall and stage-specific efficacy. Contemporaneously, tVNS can be achieved by stimulating either the auricular branch or the cervical bundle of the vagus nerve, referred to as transcutaneous auricular vagus nerve stimulation(VNS) and transcutaneous cervical VNS, respectively. In order to advance the field in a systematic manner, studies using these technologies need to adequately report sufficient methodological detail to enable comparison of results between studies, replication of studies, as well as enhancing study participant safety. We systematically reviewed the existing tVNS literature to evaluate current reporting practices. Based on this review, and consensus among participating authors, we propose a set of minimal reporting items to guide future tVNS studies. The suggested items address specific technical aspects of the device and stimulation parameters. We also cover general recommendations including inclusion and exclusion criteria for participants, outcome parameters and the detailed reporting of side effects. Furthermore, we review strategies used to identify the optimal stimulation parameters for a given research setting and summarize ongoing developments in animal research with potential implications for the application of tVNS in humans. Finally, we discuss the potential of tVNS in future research as well as the associated challenges across several disciplines in research and clinical practice. One such arcane trick is to allow the finished devices to dwell in time, and the PCE often improves. Herein, a mild postannealing procedure is implemented on coevaporated perovskite solar cells confirming a substantial PCE improvement, mainly attributed to an increased open-circuit voltage (V\(_{OC}\)). From a V\(_{OC}\) of around 1.11 V directly after preparation, the voltage improves to more than 1.18 V by temporal and thermal annealing. To clarify the origin of this annealing effect, an in-depth device experimental and simulation characterization is conducted. A simultaneous reduction of the dark saturation current, the ideality factor (n\(_{id}\)), and the leakage current is revealed, signifying a substantial impact of the postannealing procedure on recombination losses. To investigate the carrier dynamics in more detail, a set of transient optoelectrical methods is first evaluated, ascertaining that the bulk carrier lifetime is increased with device annealing. Second, a drift-diffusion simulation is used, confirming that the beneficial effect of the annealing has its origin in effective bulk trap passivation that accordingly leads to a reduction of Shockley–Read–Hall recombination rates. Therefore, reliable estimates of the performance of monitoring techniques across taxa become important. Using a unique dataset, this study investigates the potential of airborne LiDAR-derived variables characterizing vegetation structure as predictors for animal species richness at the southern slopes of Mount Kilimanjaro. To disentangle the structural LiDAR information from co-factors related to elevational vegetation zones, LiDAR-based models were compared to the predictive power of elevation models. 17 taxa and 4 feeding guilds were modeled and the standardized study design allowed for a comparison across the assemblages. Results show that most taxa (14) and feeding guilds (3) can be predicted best by elevation with normalized RMSE values but only for three of those taxa and two of those feeding guilds the difference to other models is significant. Generally, modeling performances between different models vary only slightly for each assemblage. For the remaining, structural information at most showed little additional contribution to the performance. In summary, LiDAR observations can be used for animal species prediction. However, the effort and cost of aerial surveys are not always in proportion with the prediction quality, especially when the species distribution follows zonal patterns, and elevation information yields similar results. Both interventions can lead to compromised bone perfusion and can ultimately result in infection and necrosis. Treatment usually consists of surgical necrosectomy and prolonged antibiotic therapy, usually through beta-lactams such as ampicillin/sulbactam. The poor blood supply in particular raises the question as to whether this form of antibiosis can achieve sufficient concentrations in the bone. Therefore, we investigated the antibiotic concentration in plasma and bone samples in a prospective study. Bone samples were collected from the necrosis core and in the vital surrounding bone. The measured concentrations in plasma for ampicillin and sulbactam were 126.3 ± 77.6 and 60.2 ± 35.0 µg/mL, respectively. In vital bone and necrotic bone samples, the ampicillin/sulbactam concentrations were 6.3 ± 7.8/1.8 ± 2.0 µg/g and 4.9 ± 7.0/1.7 ± 1.7 µg/g, respectively. These concentrations are substantially lower than described in the literature. However, the concentration seems sufficient to kill most bacteria, such as Streptococci and Staphylococci, which are mostly present in the biofilm of ONJ. We, therefore, conclude that intravenous administration of ampicillin/sulbactam remains a valuable treatment in the therapy of ONJ. Nevertheless, increasing resistance of Escherichia coli towards beta-lactam antibiotics have been reported and should be considered. However, the majority don’t respond to immune therapy. Across different tumor types, pre-existing T cell infiltrates predict response to checkpoint-based immunotherapy. Based on in vitro pharmacological studies, mouse models and analyses of human melanoma patients, we show that the cytokine GDF-15 impairs LFA-1/β2-integrin-mediated adhesion of T cells to activated endothelial cells, which is a pre-requisite of T cell extravasation. In melanoma patients, GDF-15 serum levels strongly correlate with failure of PD-1-based immune checkpoint blockade therapy. Neutralization of GDF-15 improves both T cell trafficking and therapy efficiency in murine tumor models. Thus GDF-15, beside its known role in cancer-related anorexia and cachexia, emerges as a regulator of T cell extravasation into the tumor microenvironment, which provides an even stronger rationale for therapeutic anti-GDF-15 antibody development. Yet, it is unknown to what extent the functional diversity of plant and animal communities controls network assembly along environmental gradients in real-world ecosystems. Here we address this question with a unique dataset of mutualistic bird-fruit, bird-flower and insect-flower interaction networks and associated functional traits of 200 plant and 282 animal species sampled along broad climate and land-use gradients on Mt. Kilimanjaro. We show that plant functional diversity is mainly limited by precipitation, while animal functional diversity is primarily limited by temperature. Furthermore, shifts in plant and animal functional diversity along the elevational gradient control the niche breadth and partitioning of the respective other trophic level. These findings reveal that climatic constraints on the functional diversity of either plants or animals determine the relative importance of bottom-up and top-down control in plant-animal interaction networks.
