Mit Hilfe kinetischer Messungen ermittelte man die Aktivierungsparameter für das System Ja~ 3b. - Aus 2 und den Abfangreagenzien p-Methoxystyrol, 1,1-Diphenylethylen sowie 1-Phenylpropen gingen mit bescheidenen Ausbeuten die Titelverbindungen 6a, b, 7 bzw. 8 hervor. Analoa zu 2 wurde sein l-Methylderivat 13 aus 6,6-Dibrom-1-methylbicyclo[3.1.0]hexan (9) durch Methyllithium freigesetzt. The fate of several nucleolar proteins representing the three main structural subcomponents of the nucleolus was examined by immunofluorescence and electron microscopy. The results show that the RNA polymerase I antibodies do not interfere with normal mitotic progression or the early steps of nucleologenesis, i.e. , the aggregation of nucleolar material into prenucleolar bodies. However,they inhibit the telophasic coalescence of the prenucleolar bodies into the chromosomal nucleolar organizer regions, thus preventing the formation of new nucleoli. These prenucleolar bodies show a fibrillar organization that also compositionally resembles the dense fibrillar component of interphase nucleoli . We conclude that during normal nucleologenesis the dense fibrillar component forms from preformed entities around nucleolar organizer regions, and that this association seems to be dependent on the presence of an active form of RNA polymerase I. The transcriptional state of the lampbrush chromosomes was experimentally modulated by incubating the oocytes for various time periods in medium containing actinomycin D. The observations show that the structure of the lateral loops changes rapidly in response to alterations in transcriptional activity. During decreasing transcriptional activity and reduced packing density of transcripts, the chromatin axis first condensed into nucleosomes and then into an approximately 30 nm thick higher order chromatin fiber. Packaging of the loop axis into supranucleosomal structures may contribute to the foreshortening and retraction of the loops observed during inhibition of transcription and in later stages of meiotic prophase. The increasing packing density of the DNA during the retraction process of the loops could also be visualized by immunofluorescence microscopy using antibodies to DNA. The dependence of the loop chromatin structure on transcriptional activity is discussed in relation to current views of mechanisms involved in gene activation. These exoproteins derive from the two gram-negative bacteria Escherichia coli and Aeromonas hydrophila and from the gram-positive pathogen Listeria monocytogenes. The hemolysin of E. coli is determined by an 8-kilobase (kb) region that includes four clustered genes (hlyC, hlyA, hlyB, and hlyD). This hemolysin determinant is part either of large transmissible plasmids or of the chromosome. The genes located chromosomally are found predominantly in E. coli strains that can cause pyelonephritis and/or other extraintestinal infections. A detailed analysis of the chromosomal hly determinants of one nephropathogenic E. coli strain revealed the existence of specific, large chromosomal insertions 75 kb and lOO kb in size that carry the hly genes but that also influence the expression of other virulence properties, i.e., adhesion and serum resistance. The direct involvement of E. coli hemolysin in virulence could be demonstrated in several model systems. The genetic determinants for hemolysin (cytolysin) formation in , A. hydrophila (aerolysin) and L. monocytogenes (listeriolysin) are less complex. Both cytolysins seem to be encoded by single genes, although two loci (aerB and aerC) that affect the expression and activity of aerolysin have been identified distal and proximal to the structural gene for aerolysin (aerA). Cytolysin-negative mutants of both bacteria were obtained by site-specific deletion and/or transposon mutagenesis. These mutants show a drastic reduction in the virulence of the respective bacteria. A crude cell extract was partiaUy dissociated into fimbriae and an adhesin-enriched fraction by heating to 7O°C. From the latter, adhesin was purified to apparent homogeneity (by fast protein liquid chromatography, immunoblot, and NaDodSO\(_4\)/PAGE) by differential ammonium sulfate precipitation, dissociation in 8 M guanidine hydrochloride, and high-resolution anion-exchange chromatography in 8 M urea. The purified adhesin formed an aggregate of M\(_r\)\(\approx\)10\(^6\) that was made up of one type of 12-kDa polypeptide (fimbrillin is 16.5 kDa). It had pI value of 4.7 (fimbriae has a pI value of 6). Adhesin and fimbrillin had different amino add compositions. The purified adhesins agglutinated human and bovine erythrocytes with the same speclfkity as the whole bacteria; purified fimbriae were not adhesive. Monoclonal anti-adhesin and anti-fimbriae antibodies were obtained. Monoclonal antiadhesin, but none of the anti-fimbriae, antibodies inhibited the agglutination of erythrocytes. The anti-adhesive antibodies were used in immuno-gold electron microscopy to localize adhesin exclusively on the fimbriae, with a possible preference to their tips. Similar relationships were found between coreid bugs and Crematogaster sp. and Anoplolepis longipes, on bamboo in the same area. The ants recruit to groups of the bugs and feed on the liquid, sugar-rich faeces of the larvae, stimulating release of the honeydew by tactile signals. They protect all stages of the bugs from disturbance by biting and by the use of defensive secretions. Phloem-feeding bugs in the families Plataspidae and Coreidae need long sty lets to pierce the thick bark of their host tree. The different methods of accommodating the resting stylets in these two families are described. In actinomycin D and 5,6-dichloro-{j-D-ribofuranosylbenzimidazoletreated Vero cells, staining was restricted to the fibrillar and not the granular regions. By double immunofluorescence, specific rabbit anti-RNA polymerase I antibodies stained the same fibrillar structures in drug-segregated nucleoli as scleroderma sera. Scleroderma sera immunoprecipitated 13 polypeptides from (35S)methionine-labeled HeLa cell extract with molecular weights ranging from 210,000 to 14,000. Similar polypeptides were precipitated by rabbit anti-RNA polymerase I antibodies, and their common identities were confirmed in immunoabsorption experiments. Microinjection of purified IgG from a patient with speckled nucleolar staining effectively inhibited ribosomal RNA transcription. Such covalent labelling of the receptor led to a concentration-dependent reduction of cellular cyclic AMP levels. This persistent effect of covalent labeHing occurred with an IC50 value of 9 nM, as compared to an IC50 value of 0.9 nM for the direct reduction of cyclic AMP Ievels by the ligand. The difference in the IC5o values can be explained by assuming spare receptors. This hypothesis was verified in binding studies using [ 3HJPIA as a radioligand. R-AHPIA inhibited binding of [3H)PIA to intact fat cells with a K1 value of about 20 nM, which is about 20 tim es high er than the corresponding IC50 value of cyclic AMP reduction. The resulting risk estimates likewise span an enormous range (nine orders of magnitude ): dietary ureas and aromatic amines combined with high nitrite concentration could pose as great a risk as the intake of preformed N-nitrosodimethylamine in the diet. In contrast, the risk posed by the in-vivo nitrosation of primary and secondary amines is probably negligible. T1 - In-vitro assays to detect alkylating and mutagenic activities of dietary components nitrosated in situ N2 - Nitrosation of dietary components has been combined with the 4-(para-nitrobenzyl)pyridine (NBP) colorimetric test for screening alkylating agents and with the Ames test for the detection of mutagenic activity. This allowed the investigation of short-hved nitrosation products of dietary components which generate electrophilic degradation products requiring no metabolic activation (natural amino acids and some derivatives, ureas, guanidines, primary alkyl and aryl amines). In a first system, precursor, nitrous acid and NBP were present simultaneously. All amino acids tested, except glutamic acid and glutamine, gave positive results. The reactivities spanned more than three orders of magnitude, with the aromatic amino acids and methionine the most active; two primary amines, tryptamine and histamine, were also strongly reactive. All guanidines tested, except the amino acid arginine, gave negative results. A second system consisted of two phases: NBP was added only after destruction of residual nitrite and adjustment of the pH to neutrality. This system was useful for the study of ureas, which are stable in acid but not in neutral media. The range of responses covered more than two orders of magnitude. Most amino acids and primary amines also gave positive results, but could be assessed only after analysing the kinetics of the competing reactions and choosing appropriate reaction times. In a third system, Salmonella typhimurium strain TA1OO replaced NBP. Representatives of the class of amino acids, ureas, the primary amine tryptamine, and aniline became higbly mutagenic upon nitrosation. Methylguanidine was only weakly mutagenic under the present assay conditions. The results indicate that further studies with unstable nitrosation products of dietary components are required to understand more thoroughly the role of endogenous nitrosation in gastric cancer. When used in immunocytochemical experiments on tissue sections and permeabilized cultured cells, this antibody detects DNA-containing structures, even when the DNA is present in very small amounts. Examples of high resolution detection include the DNA present in amplified extrachromosomal nucleoli, chromomeres of lampbrush chromosomes, mitochondria, chloroplasts and mycoplasmal particles. In immunoelectron microscopy using the immunogold technique, the DNA was localized in distinct substructures such as the "fibrillar centers" of nucleoli and certain stromal centers in chloroplasts. The antibody also reacts with DNA of chromatin of living cells, as shown by microinjection into cultured mitotic cells and into nuclei of amphibian oocytes. The potential value and the limitations of immunocytochemical DNA detection are discussed. Die an je 32 Zweit- und Viertkläßlern gewonnenen Befunde: lassen sich insgesamt so interpretieren, daß die jüngeren Kinder kaum etwas über angemessene Verhaltensweisen bei freien Reproduktionsaufgaben wissen; sie ziehen perzeptuelle Organisationsstrategien konzeptuellen (taxonomischen) Organisationsstrategien vor, profitieren tatsächlich jedoch signifikant mehr von konzeptuellen Strategien. The stages of decay were correlated with the temperature dependent development of blow fly maggots affecting the abundance of competing saprophageous beetles and of carnivors preying upon maggots of different size classes. By using ( 4.) microhabitats (spatial subdivision of a carcass) as further niche dimension, the dustering of speries using similar food resources was domonstrated in a niche overlap dendrogram. 2. The quantitative effect of predators on blow fly maggots was investigated both in field and laboratory experiments. Predation upon maggots rcduces their scramble competition, resulting in a higher pupal weight. Thus, the reproductive succcss of the blow flies seems to be buffered by the developmental flexibility of the calliphorids. The numerical effect of predators and parasitoids on the blow fly pupae was also quantified. 3. In a case study on the staphylinid beetle, Aleochara curtula, we investigated the diffenntial abundance of sexes. The ratio at which the males and females arrive at the carcass is balanced. Here the beetles feed and copulated. Females depart into the vicinity of the carrion much earlier than males, thus shifting the sex ratio to a maale bias. In the surroundings the females deposit their eggs, and the parasitoid first instar larvae search for scattered blow fly pupae. The temporal and spatial distribution of both sexes of A. curtula is thus not only affected by the food allocation of the adults, but a.lso by limiting resources of mating end egg laying sites as well as larval hosts. Zu Beginn wird das LISREL-Modell unter den Aspekten der simultanen Analyse von Kovarianzund Mittelwertstrukturen und der Modelltestung diskutiert. Anschließend werden anhand eines Illustrationsbeispiels aus der pädagogisch-psychologischen Forschung verschiedene Vorgehensweisen der Kreuzvalidierung einander gegenübergestellt und an empirischen Daten demonstriert. Es zeigt sich, daß die Obernahme exakter numerischer Parameterwerte sowohl aufgrund inhaltlicher wie auch formaler Argumente unangemessen streng erscheint. N2 - This study focuses on the problem of how to judge the adequacy of theoretical models on the basis of statistical hypothesis testing procedures. In particular, problems of testing multivariate structural equation models (LISREL-models) are discussed. In a first step, the possibilities of simultaneously analyzing covariance and mean structures and of testing LISREL models are discussed . Next, data from a study conducted in the field of educational psychology are used to illustrate different procedures of cross-validating LISREL models. As it turns out, the usual restriction to base the model on equivalent numerical parameter values appears to be inappropriately rigorous. Thus, the suggestion is that cross-validations of structural equation models should rely on approximations of interval restrictions instead. Die Inventar-Profile der entstehenden Gruppen werden erneut multidimensional und multivariat verglichen. Von den zwei Bedeutungen des "Anti" im Anti-Typ-Begriff bewährt sich die geometrische Interpretation des "Anti" an den Profilen nicht. Die Interpretation "konsistenter Andersartigkeit" dagegen kann auf der Grundlage der Bayes-statistischen Klassifikation beibehalten werden. Die Untersuchung des Anti-Typ-Begriffs lohnt möglicherweise auch in anderen Bereichen. N2 - The concept "Anti-Type" is analyzed theoretically and empirically. The data were from standardized comments on personal problems related to therapy-motivation and from responses to a multivariate personality-inventory of 187 arrested juvenile delinquents and 361 police-trainees. In the first section of the analytical part hypotheses on types and anti-types oftherapy-motivation are obtained with the Konfigurationsfrequenzanalyse (KFA). Additionally the inventory-profiles of the obtained typological groups are scaled with MDS and tested with MANOVA. In the second section the KFA-typological hypotheses are cross-examined with a Bayes-statistical classification approach by separating a "restgroup" from the typological groUPS. Again, the inventory-profiles of these distinct groups are scaled with MDS and tested with MANOVA. As a consequence, the geometrical interpretation of "anti" does not seem appropriate on the basis of the profiles. However, the interpretation of "anti" as showing consistent differences of behavior compared with type-configurations and with the restgroup is supported on the basis of the Bayes-statistical method. With regard to other variables the presented conclusions about the validity of the "anti-type" in this particular realm may not be valid. Thus, the concept of "AntiType" may be a worthwhile research subject with other variables also.
