Especially when treating younger patients with an increased life expectancy, the prosthesis life span is often used up and a complicated revision procedure is necessary. Aseptic loosening of the prosthesis is the most common reason for replacing an implant. The bone marrow contains human mesenchymal stem cells (hMSC) that help maintain and regenerate the connective tissue and the bone itself. Whether these cells play a role in the etiology of aseptic loosening of the prosthesis through a proliferation or differentiation deficit has not yet been conclusively clarified. In the present study, the regenerative capacity of hMSC from two groups of donors was examined and compared: Patients who had undergone aseptic loosening of the prosthesis and who had to undergo revision surgery served as the experimental group. Patients who primarily received a hip or knee joint endoprosthesis formed the control group. To assess the regenerative properties of the hMSC, on the one hand the growth and aging behavior in cell culture and on the other hand the in vitro differentiation capacity was examined.show moreshow less
