One of the main problems we face with PPGL is the lack of molecular markers capable of predicting the development of metastases in patients. Telomere-related genes, such as TERT and ATRX, have been recently described in PPGL, supporting the association between the activation of immortalization mechanisms and disease progression. However, the contribution of other genes involving telomere preservation machinery has not been previously investigated. In this work, we aimed to analyze the prognostic value of a comprehensive set of genes involved in telomere maintenance. For this study, we collected 165 PPGL samples (97 non-metastatic/63 metastatic), genetically characterized, in which the expression of 29 genes of interest was studied by NGS. Three of the 29 genes studied, TERT, ATRX and NOP10, showed differential expression between metastatic and non-metastatic cases, and alterations in these genes were associated with a shorter time to progression, independent of SDHB-status. We studied telomere length by Q-FISH in patient samples and in an in vitro model. NOP10 overexpressing tumors displayed an intermediate-length telomere phenotype without ALT, and in vitro results suggest that NOP10 has a role in telomerase-dependent telomere maintenance. We also propose the implementation of NOP10 IHC to better stratify PPGL patients.
Morbus Fabry betrifft als lysosomale Speicherkrankheit viele Organsysteme durch die Ablagerung von Gb3 in verschiedenen Geweben. Besonders durch die Beteiligung von Nieren und Herz, wird die Lebenszeit von den Patienten häufig verkürzt. Eine Beschreibung konkreter klinischer Symptome, welche auch durch Allgemeinmediziner oder Zahnärzte erkannt werden könnten, könnte eine frühzeitigere Diagnose und damit frühzeitige Therapie ermöglichen. Besonders extraorale gesichtsspezifische Merkmale können von verschiedensten Gruppen von Ärzten erkannt werden.
Die extraorale Auswertung zeigte, wie in der Literatur beschrieben, das Vorkommen von periorbitaler Fülle, prominente Arcus superciliaris, eine kürzere und bullösere Nase. Die Auffälligkeiten waren besonders bei den Männern zu beobachten.
Die intraorale Auswertung wurde in dentale Auffälligkeiten und Ereignisse des Hart- und Weichgewebes eingeteilt. Bei den dentalen Ereignissen zeigte sich eine Diskrepanz zwischen der Kiefergröße und dem Zahnmaterial. So neigte das Patientenkollektiv eher zu einem Breitkiefer, was eine Erklärung für die multiplen Lücken im Frontzahnbereich der Patienten darstellt. An der Mundschleimhaut und perioral konnten vermehrt Angiokeratome und Teleangiektasien festgestellt werden, sowie das vermehrte Vorkommen von Exostosen. Speziell die Zunge der Patienten zeigte auch Auffälligkeiten in Form von einer subjektiven Makroglossie, einer Furchenzunge und Veränderungen der Papillen.
Die Auffälligkeiten in der Mundhöhle und im Kopf-Hals Bereich der Morbus Fabry Patienten sind, wie der Literatur beschrieben, vorhanden, jedoch stellen sie keine Schlüsselrolle in der Diagnose dar, da sie in allen Bereichen nur leichte Abweichungen oder Auffälligkeiten zeigen, welche nicht immer Auftreten und daher schwer zu diagnostizieren sind.
The main goal of this thesis is to elucidate the sense in which recent experimental progress in condensed matter physics, namely the verification of two-dimensional Dirac-like materials and their control in ballistic- as well as hydrodynamic transport experiments enables the observation of a well-known 'high-energy' phenomenon: The parity anomaly of planar quantum electrodynamics (QED\(_{2+1}\)). In a nutshell, the low-energy physics of two-dimensional Quantum Anomalous Hall (QAH) insulators like (Hg,Mn)Te quantum wells or magnetically doped (Bi,Sb)Te thin films can be described by the combined response of two 2+1 space-time dimensional Chern insulators with a linear dispersion in momentum. Due to their Dirac-like spectra, each of those Chern insulators is directly related to the parity anomaly of planar quantum electrodynamics. However, in contrast to a pure QED\(_{2+1}\) system, the Lagrangian of each Chern insulator is described by two different mass terms: A conventional momentum-independent Dirac mass \(m\), as well as a momentum-dependent so-called Newtonian mass term \(B \vert \mathbf{k} \vert^2\). According to the parity anomaly it is not possible to well-define a parity- and U(1) gauge invariant quantum system in 2+1 space-time dimensions. More precisely, starting with a parity symmetric theory at the classical level, insisting on gauge-invariance at the quantum level necessarily induces parity-odd terms in the calculation of the quantum effective action. The role of the Dirac mass term in the calculation of the effective QED\(_{2+1}\) action has been initially studied in Phys. Rev. Lett. 51, 2077 (1983). Even in the presence of a Dirac mass, the associated fermion determinant diverges and lacks gauge invariance. This requires a proper regularization/renormalizaiton scheme and, as such, transfers the peculiarities of the parity anomaly to the massive case.
In the scope of this thesis, we connect the momentum-dependent Newtonian mass term of a Chern insulator to the parity anomaly. In particular, we reveal, that in the calculation of the effective action, before renormalization, the Newtonian mass term acts similarly to a parity-breaking element of a high-energy regularization scheme. This calculation allows us to derive the finite frequency correction to the DC Hall conductivity of a QAH insulator. We derive that the leading order AC correction contains a term proportional to the Chern number. This term originates from the Newtonian mass and can be measured via electrical or via magneto-optical experiments. The Newtonian mass, in particular, significantly changes the resonance structure of the AC Hall conductivity in comparison to pure Dirac systems like graphene.
In addition, we study the effective action of the aforementioned Chern insulators in external out-of-plane magnetic fields. We show that as a consequence of the parity anomaly the QAH phase in (Hg,Mn)Te quantum wells or in magnetically doped (Bi,Sb)Te thin films survives in out-of-plane magnetic fields, violates the Onsager relation, and can therefore be distinguished from a conventional quantum Hall (QH) response. As a smoking-gun of the QAH phase in increasing magnetic fields, we predict a transition from a quantized Hall plateau with \(\sigma_\mathrm{xy}= -\mathrm{e}^2/\mathrm{h}\) to a not perfectly quantized plateau which is caused by scattering processes between counter-propagating QH and QAH edge states. This transition is expected to be of significant relevance in paramagnetic QAH insulators like (Hg,Mn)Te/CdTe quantum wells, in which the exchange interaction competes against the out-of-plane magnetic field.
All of the aforementioned results do not incorporate finite temperature effects. In order to shed light on such phenomena, we further analyze the finite temperature Hall response of 2+1 dimensional Chern insulators under the combined influence of a chemical potential and an out-of-plane magnetic field. As we have mentioned above, this non-dissipative transport coefficient is directly related to the parity anomaly of planar quantum electrodynamics. Within the scope of our analysis we show that the parity anomaly itself is not renormalized by finite temperature effects. However, the parity anomaly induces two terms of different physical origin in the effective Chern-Simons action of a QAH insulator, which are directly proportional to its Hall conductivity. The first term is temperature and chemical potential independent and solely encodes the intrinsic topological response. The second term specifies the non-topological thermal response of conduction- and valence band modes, respectively. We show that the relativistic mass \(m\) of a Chern insulator counteracts finite temperature effects, whereas its non-relativistic Newtonian mass \(B \vert \mathbf{k} \vert^2 \) enhances these corrections. In addition, we are extending our associated analysis to finite out-of-plane magnetic fields, and relate the thermal response of a Chern insulator therein to the spectral asymmetry, which is a measure of the parity anomaly in out-of-plane magnetic fields.
In the second part of this thesis, we study the hydrodynamic properties of two-dimensional electron systems with a broken time-reversal and parity symmetry. Within this analysis we are mainly focusing on the non-dissipative transport features originating from a peculiar hydrodynamic transport coefficient: The Hall viscosity \(\eta_\mathrm{H}\). In out-of-plane magnetic fields, the Hall viscous force directly competes with the Lorentz force, as both mechanisms contribute to the overall Hall voltage. In our theoretical considerations, we present a way of uniquely distinguishing these two contributions in a two-dimensional channel geometry by calculating their functional dependencies on all external parameters. We are in particular deriving that the ratio of the Hall viscous contribution to the Lorentz force contribution is negative and that its absolute value decreases with an increasing width, slip-length and carrier density. Instead, it increases with the electron-electron mean free path in the channel geometry considered. We show that in typical materials such as GaAs the Hall viscous contribution can dominate the Lorentz signal up to a few tens of millitesla until the total Hall voltage vanishes and eventually is exceeded by the Lorentz contribution. Last but not least, we derive that the total Hall electric field has a parabolic form originating from Lorentz effects. Most remarkably, the offset of this parabola is directly characterized by the Hall viscosity. Therefore, in summary, our results pave the way to measure and to identify the Hall viscosity via both global and local measurements of the entire Hall voltage.
In Staphylococcus aureus, de novo methionine biosynthesis is regulated by a unique hierarchical pathway involving stringent-response controlled CodY repression in combination with a T-box riboswitch and RNA decay. The T-box riboswitch residing in the 5′ untranslated region (met leader RNA) of the S. aureus metICFE-mdh operon controls downstream gene transcription upon interaction with uncharged methionyl-tRNA. met leader and metICFE-mdh (m)RNAs undergo RNase-mediated degradation in a process whose molecular details are poorly understood. Here we determined the secondary structure of the met leader RNA and found the element to harbor, beyond other conserved T-box riboswitch structural features, a terminator helix which is target for RNase III endoribonucleolytic cleavage. As the terminator is a thermodynamically highly stable structure, it also forms posttranscriptionally in met leader/ metICFE-mdh read-through transcripts. Cleavage by RNase III releases the met leader from metICFE-mdh mRNA and initiates RNase J-mediated degradation of the mRNA from the 5′-end. Of note, metICFE-mdh mRNA stability varies over the length of the transcript with a longer lifespan towards the 3′-end. The obtained data suggest that coordinated RNA decay represents another checkpoint in a complex regulatory network that adjusts costly methionine biosynthesis to current metabolic requirements.
Die wahrscheinlich größten Probleme des 21. Jahrhunderts sind der Klimawandel und die Sicherstellung der Nahrungsmittelversorgung für eine steigende Zahl an Menschen. Durch die Zunahme von extremen Wetterbedingungen wie Trockenheit und Hitze wird der Anbau konventioneller, wenig toleranter Nutzpflanzen erschwert und die dadurch notwendige, steigende Bewässerung der Flächen führt darüber hinaus zu einer zusätzlichen Versalzung der Böden mit für Pflanzen toxischen Natrium- und Chlorid-Ionen. Kenntnisse über Anpassungsstrategien salztoleranter Pflanzen an Salzstress, aber auch detailliertes Wissen über die Steuerung der Transpiration und damit des Wasserverlusts von Pflanzen sind daher wichtig, um auch künftig ertragreiche Landwirtschaft betreiben zu können. In dieser Arbeit habe ich verschiedene Aspekte der pflanzlichen Stressphysiologie bearbeitet, die im Folgenden getrennt voneinander zusammengefasst werden.
I. Funktionelle Unterschiede der PYR/PYL-Rezeptoren von Schließzellen
Entscheidend für den Wasserstatus von Pflanzen ist die Kontrolle des Wasserverlusts durch Spaltöffnungen (Stomata), die von einem Paar Schließzellen gebildet werden. Externe Faktoren wie Licht, Luftfeuchtigkeit und CO2, sowie interne Faktoren wie das Phytohormon Abszisinsäure (ABA) regulieren über Signalkaskaden die Stomaweite und dadurch den Wasserverlust. Die zugrunde liegenden Signalkaskaden überlappen teilweise. Vor allem der Stomaschluss durch erhöhtes CO2 und ABA weisen viele Gemeinsamkeiten auf und die Identifizierung des Konvergenzpunktes beider Signale ist immer noch aktueller Gegenstand der Forschung. Von besonderem Interesse sind dabei die in Schließzellen exprimierten ABA-Rezeptoren der PYR/PYL-Familie. Denn obwohl bislang nicht nachgewiesen werden konnte, dass CO2 zu einem Anstieg des ABA-Gehalts von Schließzellen führt deuten einige Studien darauf hin, dass die ABA-Rezeptoren selbst am CO2-Signalweg beteiligt sind.
Durch Untersuchungen der Stomareaktion von Arabidopsis ABA-Rezeptormutanten konnte ich in dieser Arbeit zeigen, dass die in Schließzellen exprimierten ABA-Rezeptoren der PYR/PYL-Familie funktionale Unterschiede aufweisen. Fünffach-Verlustmutanten der ABA-Rezeptoren PYR1, PYL2, 4, 5 und 8 (12458) waren in ihrem ABA-induzierten Stomaschluss beeinträchtigt und nur die Komplementation mit PYL2 und in geringerem Maße PYR1 konnte die ABA-Sensitivität wiederherstellen. Die Stomata von 12458-Verlustmutanten waren außerdem insensitiv gegenüber erhöhtem CO2, was auf eine Beteiligung der ABA-Rezeptoren am CO2-induzierten Stomaschluss hindeutet und diese Sensitivität konnte nur durch die Komplementation mit PYL4 oder PYL5, nicht aber mit PYL2 wiederhergestellt werden. Somit konnten in dieser Arbeit erstmals funktionelle Unterschiede der PYR/PYLs beim Stoma-Schluss nachgewiesen werden.
Alle externen und internen Stomaschluss-Signale haben außerdem Einfluss auf die Genexpression der Schließzellen und führen zu individuellen expressionellen Adaptionen. In vorangegangenen Microarray Studien konnte gezeigt werden, dass jeder Stimulus auch die Expression eines distinkten Sets an ABA-Rezeptoren beeinflusst. Im Rahmen dieser Arbeit konnte ich außerdem zeigen, dass die Expression der ABA-Rezeptoren bereits auf kleine Änderungen der ABA-Konzentration der Schließzellen reagiert und dass diese sich außerdem in ihrer Sensitivität gegenüber ABA unterschieden. Geringe Änderungen der ABA-Konzentration von Schließzellen haben demnach Auswirkungen auf deren Rezeptor-zusammensetzung. Darüber hinaus konnte ich zeigen, dass die Rezeptoren die Expression unterschiedlicher nachgeschalteter Gene beeinflussen, was darauf hindeutet, dass Anpassungen des Rezeptorpools durch geringe Änderungen des ABA-Gehalts von Schließzellen schlussendlich auf genexpressioneller Ebene zur längerfristigen Adaption an externe Bedingungen führen und die Rezeptoren auch hier funktional verschieden sind.
II. Stomatäre Besonderheiten der toleranten Dattelpalme (Phoenix dactylifera)
Dattelpalmen kommen natürlicherweise an besonders trockenen und heißen Standorten vor, an denen es aufgrund der harschen Bedingungen nur sehr wenigen Pflanzen möglich ist überhaupt zu wachsen. Ein naheliegender Grund für die herausragende Toleranz dieser Art gegenüber wasserlimitierenden Bedingungen ist eine Anpassung der stomatären Regulation zu Gunsten des Wasserhaushalts.
In dieser Arbeit konnte ich durch vergleichende Untersuchungen der lichtabhängigen Transpiration sowie dem ABA-induzierten Stomaschluss grundlegende Unterschiede in der Stomaphysiologie der Dattelpalmen und der eher sensitiven Modellpflanze Arabidopsis thaliana nachweisen. Blattgaswechselmessungen zeigten, dass Dattelpalmen in der Lage sind die Spaltöffnungen bei niedrigen Lichtintensitäten, bei denen Arabidopsis bereits deutlich geöffnete Stomata aufwies, geschlossen zu halten. Der bedeutendste Unterschied in der Stomaphysiologie von Dattelpalmen und Arabidopsis lag aber im ABA-induzierten Stomaschluss. Während über die Petiole verabreichtes ABA bei Arabidopsis innerhalb von 15 Minuten zu einem vollständigen Stomaschluss führte, konnte ich in dieser Arbeit zeigen, dass der ABA-induzierte Stomaschluss der Datteln nitratabhängig ist. ABA allein führte nur zu einem sehr langsamen Stomaschluss der innerhalb einer Stunde nicht vollständig abgeschlossen war. Nur in Gegenwart von Nitrat führte die ABA-Gabe in den Transpirationsstrom der Fiederblätter der Datteln zu einem schnellen und vollständigen Stomaschluss. In Arabidopsis wird der in Schließzellen vorkommende Anionenkanal AtSLAC1 durch eine über den ABA-Signalweg vermittelte Phosphorylierung aktiviert, was schlussendlich zur Aktivierung spannungsabhängiger Kationenkanäle und zum Ausstrom von Kalium aus den Schließzellen führt. Es konnte gezeigt werden, dass die Nitratabhängigkeit der ABA-Antwort der Schließzellen von Dattelpalmen auf Eigenschaften von PdSLAC1 zurückzuführen ist und dieser Kanal nur in Anwesenheit von extrazellulärem Nitrat aktivierbar ist. Mittlerweile konnte, unter anderem basierend auf diesen Ergebnissen, eine Tandem-Aminosäuresequenz identifiziert werden, die die SLAC-Homologe monokotyler Pflanzen wie der Dattelpalme von der dikotyler Pflanzen unterscheidet und zumindest teilweise für die nitratabhängige Aktivierung des Stomaschlusses vieler monokotyler verantwortlich ist.
III. Die Salztoleranz von Phoenix dactylifera und Chenopodium quinoa
Sowohl Dattelpalmen als auch C. quinoa weisen, verglichen mit den meisten anderen Pflanzen, eine hohe Toleranz gegenüber NaCl-haltigen Böden auf. In dieser Arbeit habe ich die Salztoleranz beider Arten untersucht, um so Strategien zu identifizieren, die diesen Pflanzen diese gesteigerte Toleranz ermöglichen.
Dattelpalmen können natürlicherweise auf salzigen Böden wachsen. Makroskopisch weisen diese Pflanzen aber keine Anpassungen wie bspw. Salzdrüsen auf und bislang ist unklar wie Dattelpalmen mit dem NaCl aus dem Boden umgehen. In dieser Arbeit konnte ich zeigen, dass der Natriumgehalt der Fiederblätter der Datteln durch eine sechswöchige Bewässerung mit 600mM NaCl, was ungefähr der Konzentration von Meerwasser entspricht, nicht zunimmt. Demnach sind Datteln so genannte „Exkluder“, also Pflanzen, die eine übermäßige Natriumaufnahme in photosynthetisch aktives Gewebe vermeiden. Der Natriumgehalt der Wurzeln dagegen nahm unter Salzstress aber zu. Diese Zunahme war allerdings in unterschiedlichen Bereichen der Wurzeln verschieden stark. Flammenphotometrische Messungen ergaben einen vom Wurzelansatz ausgehenden graduellen Anstieg des Natriumgehalts, der an der Wurzelspitze am höchsten war. Darüber hinaus konnte eine Induktion von PdSOS1, einem putativen Na+/H+-Antiporter in diesen unteren, natriumhaltigen Bereichen nachgewiesen werden. Eine hohe SOS1-Aktivität gilt bereits in anderen toleranten Arten als Schlüsselmerkmal für deren Toleranz und die gesteigerte Expression von PdSOS1 deutet auf eine erhöhte Natrium-Exportrate aus der Wurzel zurück in den Boden in diesen unteren Bereichen hin, was schlussendlich den Ausschluss von Natrium vermitteln könnte.
In sensitiven Arten führt Salzstress häufig zu einer Abnahme der Kaliumkonzentration des Gewebes. Interessanterweise war dies weder für das Blatt- noch das Wurzelgewebe der Dattelpalmen der Fall. Der Kaliumgehalt beider Gewebe blieb trotz der Bewässerung der Pflanzen mit Salzwasser konstant. Auf expressioneller Ebene konnte ich darüber hinaus zeigen, dass PdHAK5, ein putativer hochaffiner Kaliumtransporter, der unter Kontrollbedingungen überwiegend in den oberen Wurzelabschnitten exprimiert wurde, durch den Salzstress dort reprimiert wurde. PdKT, ebenfalls ein putatives Kalium-Transportprotein dagegen, wurde nicht durch die Salzbehandlung beeinflusst, was zusammengenommen darauf hindeutet, dass das Aufrechterhalten des Kaliumgehalts bei Salzstress durch die differentielle Regulation verschiedener Kaliumaufnahmesysteme gewährleistet wird. Der effiziente Ausschluss von Natrium zusammen mit dem hohen K+/Na+-Verhältnis könnten demnach Schlüsselmerkmale für die hohe Salztoleranz von Phoenix dactylifera darstellen.
Quinoa ist, ähnlich wie die Dattelpalme, eine salztolerante Nutzpflanze. Im Gegensatz zu Dattelpalmen weist Quinoa allerdings besondere Strukturen auf der Epidermis auf, die so genannten epidermalen Blasenhaare (englisch: epidermal bladder cells, EBCs). Die Funktion dieser ballonartig vergrößerten Zellen als externe Salzspeicher wird seit längerem diskutiert.
Flammenphotometrische Messungen des Natriumgehalts von Quinoa unter Salzstressbedingungen ergaben, dass Quinoa anders als Dattelpalmen, Natrium in die oberirdischen, photosynthetisch aktiven Organe aufnimmt. Auch die Zunahme des Natriumgehalts der EBCs konnte ich nachweisen. Junge Blätter haben eine hohe Dichte an intakten EBCs, was deren Funktion als externe Salzspeicher besonders zum Schutz dieser jungen Blätter nahelegt. mRNA-Sequenzierungen ergaben darüber hinaus, dass die EBCs bereits unter Kontrollbedingungen viele in grundlegende Stoffwechselprozesse involvierte Gene sowie membranständige Transportproteine differentiell exprimieren. Diese Unterschiede im Transkriptom der EBCs zum Blattgewebe zeigen, dass katabole Stoffwechselwege nur eine untergeordnete Rolle in den hochspezialisierten EBCs spielen und deren Stoffwechsel auf dem Import energiereicher Zucker und Aminosäuren basiert.
Mittels qPCR-Messungen und RNA-Sequenzierungen konnte ich die gewebespezifische Expression verschiedener Transportproteine nachweisen, die eine gerichtete Aufnahme von Natrium in EBCs ermöglichen könnten. Besonders die differentielle Expression eines Natriumkanals der HKT1-Familie deutet auf dessen Beteiligung an der Natriumbeladung der EBCs hin. CqHKT1.2 wurde ausschließlich in EBCs exprimiert und die elektrophysiologische Charakterisierung dieses Transportproteins ergab eine spannungsabhängige Natriumleitfähigkeit. Dieser Natriumkanal kann demnach die Natriumaufnahme bei Membranspannungen nahe dem Ruhepotential in die EBCs vermitteln und die Deaktivierung des CqHKT1.2 bei depolarisierenden Membranspannungen kann darüber hinaus einen Efflux von Na+ aus den EBCs verhindern. Auch das Expressionsmuster eines putativen Na+/H+-Antiporters (CqSOS1) der nur sehr gering in EBCs aber deutlich höher in Blattgewebe exprimiert wurde, deutet auf eine indirekte Beteiligung dieses SOS1 an der Beladung der EBCs hin. Bereits charakterisierte SOS1-Proteine anderer Pflanzen zeigten unter physiologischen Bedingungen eine Natriumexport-Aktivität. CqSOS1 könnte demnach den Export von Natrium aus Mesophyll- und Epidermiszellen der Blätter in den Apoplasten vermitteln, welches dann über CqHKT1.2 in die EBCs aufgenommen wird.
Trotz der Natriumaufnahme in die oberirdischen Teile und die EBCs führte die Salzbehandlung ähnlich wie bei den Datteln nicht zu einer Abnahme des bemerkenswert hohen Kaliumgehalts. Mittels qPCR-Untersuchungen konnte ich die Expression verschiedener HAK-Orthologe nachweisen, deren Aktivität die Aufrechterhaltung des Kaliumgehalts unter Salzstress vermitteln könnten. Frühere Studien konnten zeigen, dass Salzstress bei Quinoa wie bei vielen salztoleranten Arten zu einem Anstieg der Konzentration von kompatiblen gelösten Substanzen und besonders von Prolin führt. In dieser Arbeit konnte ich die hohe Expression eines Prolintransporters in EBCs nachweisen, was eher auf einen importbasierten Anstieg der Prolinkonzentration als auf die Synthese innerhalb der EBCs schließen lässt.
Zusammengefasst ergaben der Anstieg des Natriumgehalts der EBCs in Verbindung mit den Ergebnissen der RNA-Sequenzierung und den ergänzenden qPCR Messungen, dass die EBCs von Quinoa bereits unter Kontrollbedingen für die Aufnahme von überschüssigen Ionen unter Salzstress spezialisierte Zellen sind, deren Spezialisierung auf dem Import von energiereichreichen Zucken und anderen Substanzen basiert.
Anthropogenic and environmental drivers shape diversity of naturalized plants across the Pacific

Aim
The Pacific exhibits an exceptional number of naturalized plant species, but the drivers of this high diversity and the associated compositional patterns remain largely unknown. Here, we aim to (a) improve our understanding of introduction and establishment processes and (b) evaluate whether this information is sufficient to create scientific conservation tools, such as watchlists.
Location
Islands in the Pacific Ocean, excluding larger islands such as New Zealand, Japan, the Philippines and Indonesia.
Methods
We combined information from the most up‐to‐date data sources to quantify naturalized plant species richness and turnover across island groups and investigate the effects of anthropogenic, biogeographic and climate drivers on these patterns. In total, we found 2,672 naturalized plant species across 481 islands and 50 island groups, with a total of 11,074 records.
Results
Most naturalized species were restricted to few island groups, and most island groups have a low number of naturalized species. Island groups with few naturalized species were characterized by a set of widespread naturalized species. Several plant families that contributed many naturalized species globally also did so in the Pacific, particularly Fabaceae and Poaceae. However, many families were significantly over‐ or under‐represented in the Pacific naturalized flora compared to other regions of the world. Naturalized species richness increased primarily with increased human activity and island altitude/area, whereas similarity between island groups in temperature along with richness differences was most important for beta diversity.
Main conclusions
The distribution and richness of naturalized species can be explained by a small set of drivers. The Pacific region contains many naturalized plant species also naturalized in other regions in the world, but our results highlight key differences such as a stronger role of anthropogenic drivers in shaping diversity patterns. Our results establish a basis for predicting and preventing future naturalizations in a threatened biodiversity hotspot.
Conventional bivalent IgG antibodies targeting a subgroup of receptors of the TNF superfamily (TNFSF) including fibroblast growth factor-inducible 14 (anti-Fn14) typically display no or only very limited agonistic activity on their own and can only trigger receptor signaling by crosslinking or when bound to Fcγ receptors (FcγR). Both result in proximity of multiple antibody-bound TNFRSF receptor (TNFR) molecules, which enables engagement of TNFR-associated signaling pathways. Here, we have linked anti-Fn14 antibodies to gold nanoparticles to mimic the “activating” effect of plasma membrane-presented FcγR-anchored anti-Fn14 antibodies. We functionalized gold nanoparticles with poly-ethylene glycol (PEG) linkers and then coupled antibodies to the PEG surface of the nanoparticles. We found that Fn14 binding of the anti-Fn14 antibodies PDL192 and 5B6 is preserved upon attachment to the nanoparticles. More importantly, the gold nanoparticle-presented anti-Fn14 antibody molecules displayed strong agonistic activity. Our results suggest that conjugation of monoclonal anti-TNFR antibodies to gold nanoparticles can be exploited to uncover their latent agonism, e.g., for immunotherapeutic applications.
Circadian clocks prepare the organism to cyclic environmental changes in light, temperature, or food availability. Here, we characterized the master clock in the brain of a strongly photoperiodic insect, the aphid Acyrthosiphon pisum, immunohistochemically with antibodies against A. pisum Period (PER), Drosophila melanogaster Cryptochrome (CRY1), and crab Pigment-Dispersing Hormone (PDH). The latter antibody detects all so far known PDHs and PDFs (Pigment-Dispersing Factors), which play a dominant role in the circadian system of many arthropods. We found that, under long days, PER and CRY are expressed in a rhythmic manner in three regions of the brain: the dorsal and lateral protocerebrum and the lamina. No staining was detected with anti-PDH, suggesting that aphids lack PDF. All the CRY1-positive cells co-expressed PER and showed daily PER/CRY1 oscillations of high amplitude, while the PER oscillations of the CRY1-negative PER neurons were of considerable lower amplitude. The CRY1 oscillations were highly synchronous in all neurons, suggesting that aphid CRY1, similarly to Drosophila CRY1, is light sensitive and its oscillations are synchronized by light-dark cycles. Nevertheless, in contrast to Drosophila CRY1, aphid CRY1 was not degraded by light, but steadily increased during the day and decreased during the night. PER was always located in the nuclei of the clock neurons, while CRY was predominantly cytoplasmic and revealed the projections of the PER/CRY1-positive neurons. We traced the PER/CRY1-positive neurons through the aphid protocerebrum discovering striking similarities with the circadian clock of D. melanogaster: The CRY1 fibers innervate the dorsal and lateral protocerebrum and putatively connect the different PER-positive neurons with each other. They also run toward the pars intercerebralis, which controls hormone release via the neurohemal organ, the corpora cardiaca. In contrast to Drosophila, the CRY1-positive fibers additionally travel directly toward the corpora cardiaca and the close-by endocrine gland, corpora allata. This suggests a direct link between the circadian clock and the photoperiodic control of hormone release that can be studied in the future.
Coagulase-negative staphylococci (CoNS) are common opportunistic pathogens, but also ubiquitous human and animal commensals. Infection-associated CoNS from healthcare environments are typically characterized by pronounced antimicrobial resistance (AMR) including both methicillin- and multidrug-resistant isolates. Less is known about AMR patterns of CoNS colonizing the general population. Here we report on AMR in commensal CoNS recovered from 117 non-hospitalized volunteers in a region of Germany with a high livestock density. Among the 69 individuals colonized with CoNS, 29 had reported contacts to either companion or farm animals. CoNS were selectively cultivated from nasal swabs, followed by species definition by 16S rDNA sequencing and routine antibiotic susceptibility testing. Isolates displaying phenotypic AMR were further tested by PCR for presence of selected AMR genes. A total of 127 CoNS were isolated and Staphylococcus epidermidis (75%) was the most common CoNS species identified. Nine isolates (7%) were methicillin-resistant (MR) and carried the mecA gene, with seven individuals (10%) being colonized with at least one MR-CoNS isolate. While resistance against gentamicin, phenicols and spectinomycin was rare, high resistance rates were found against tetracycline (39%), erythromycin (33%) and fusidic acid (24%). In the majority of isolates, phenotypic resistance could be associated with corresponding AMR gene detection. Multidrug-resistance (MDR) was observed in 23% (29/127) of the isolates, with 33% (23/69) of the individuals being colonized with MDR-CoNS. The combined data suggest that MR- and MDR-CoNS are present in the community, with previous animal contact not significantly influencing the risk of becoming colonized with such isolates.
Antimicrobial stewardship measures in cardiac surgery and its impact on surgical site infections

Objective
The goal of this study was to monitor the compliance and impact on a protocol change of surgical antimicrobial prophylaxis in cardiac surgery favouring cefazolin instead cefuroxime, initiated by the hospital’s antimicrobial stewardship team.
Methods
This quality improvement study was performed in a tertiary care hospital in collaboration with the department of cardiothoracic surgery and the hospitals antimicrobial stewardship team following a revision of the standard for surgical antimicrobial prophylaxis including 1029 patients who underwent cardiac surgery. 582 patients receiving cefuroxime and 447 patients receiving cefazolin respectively were compared without altering any other preventative perioperative measures including its postoperative duration of less than 24 h. Adherence and surgical site infections were compiled and analysed.
Results
A complete adherence was achieved. Overall surgical site infections occurred in 37 (3.6%) of the cases, 20 (3.4%) in cefuroxime patients and 17 (3.8%) in cefazolin patients (p value = 0.754). No statistically significant differences could be found in any of the primary endpoints, but there was a trend towards less deep sternal wound infections in the cefazolin group.
Conclusions
The study supports the role of antimicrobial stewardship in cardiac surgery and mirrors the success of a multidisciplinary team aiming to minimize adverse events by optimizing antibiotic use.
