Das Masernvirus (MV) kann in Erkrankten eine schwere, langanhaltende Immunsuppression verursachen, wodurch Infektionen mit opportunistischen Pathogenen begünstigt werden. Diese basiert auf einer Paralyse der hämatopoetischen Zellen, welche das Virus durch Kontakt eines viralen Glykoproteinkomplexes zu einem unbekannten RezeptorX auf der Zell- Oberfläche induzieren kann. Kerncharakterisitika hiervon sind unter anderem die Herabregulation der Akt-Kinase-Phosphorylierung, die Inhibition der zellulären Proliferation und die Aktivierung der neutralen Sphingomyelinase 2 (NSM2).
In einem kinetischen Phosphoproteom konnten zwei potentielle Interaktionsrezeptoren des MV identifiziert werden: CD43 und P2X3. Das hochglykosylierte Oberflächenmolekül CD43 ist auf hämatopoetischen Zellen ubiquitär exprimiert und reguliert in T-Zellen deren Überleben, Proliferation, Aktivierung, Migration und Adhäsion. P2X3 wird in hämatopoetischen Zellen nur in geringem Maße exprimiert. Seine funktionelle Bedeutung ist in diesem Kompartiment nicht bekannt. Beide Kandidaten wurden mittels CRISPR/Cas9 Verfahren einzeln oder kombiniert aus Jurkat-T-Zellen ablatiert, welche nachfolgend nach MV-Kontakt hinsichtlich der oben erwähnten MV-modulierten Parameter getestet wurden. Zusätzlich wurden iso- und allosterische P2X3-Inhibitoren an primären und Jurkat-T-Zellen verwendet, um dessen Rolle in Ca2+-Mobilisierung und Proliferation nach T-Zell-Rezeptor Co-Stimulation zu analysieren.
Die genetische Depletion beider Rezeptor-Kandidaten verringerte die Effekte des MV auf alle getesteten Parameter signifikant, was darauf hindeutet, dass beide Proteine entscheidend an der T-Zell-Suppression beteiligt sind. Während die isosterische Inhibition von P2X3 keinen Effekt hatte, wurde die Proliferation primärer T-Zellen durch dessen allosterische Inhibition vor Co-Stimulation fast verdoppelt und die Effizienz der Ca2+-Mobilisierung in Jurkat- und primären T-Zellen signifikant erhöht. In P2X3-depletierten Jurkat-Zellen hingegen war die Ca2+-Mobilisierung nach Stimulation signifikant geringer als in WT-Zellen.
In dieser Arbeit konnten zwei wichtige Mediatoren der MV induzierten T-Zell-Suppression identifiziert werden. Vor allem P2X3, dessen Expression, Regulation und funktionelle Bedeutung im hämatopoetischen Kompartiment noch nicht erforscht wurde, könnte ein vielversprechender Kandidat für eine antivirale Therapie darstellen, da ein klinisch getesteter P2X3-Inhibitor bereits verfügbar ist.
After priming in Peyer's patches (PPs) and mesenteric lymph nodes (mLN) T- cells infiltrate the intestine through lymphatic draining and homing through the bloodstream. However, we found that in mouse models of acute graft-versus-host disease (GvHD), a subset of alloreactive T-cells directly migrates from PPs to the adjacent intestinal lamina propria (LP), bypassing the normal lymphatic drainage and vascular trafficking routes. Notably, this direct migration occurred in irradiated and unirradiated GvHD models, indicating that irradiation is not a prerequisite for this observed behavior.
Next, we established a method termed serial intravascular staining (SIVS) in mouse models to systematically investigate the trafficking and migration of donor T- cells in the early stages of acute GvHD initiation. We found that the direct migration of T-cells from PPs to LP resulted in faster recruitment of cells after allogeneic hematopoietic cell transplantation (allo-HCT). These directly migrating T-cells were found to be in an activated and proliferative state, exhibiting a TH1/TH17-like phenotype and producing cytokines such as IFN-γ and TNF-α. Furthermore, we observed that the directly migrating alloreactive T-cells expressed specific integrins (α4+, αE+) and chemokine receptors (CxCR3+, CCR5+, and CCR9+). Surprisingly, blocking these integrins and chemokine-coupled receptors did not hinder the direct migration of T- cells from PPs to LP, suggesting the involvement of alternative mechanisms. Previous experiments ruled out the involvement of S1PR1 and topographical features of macrophages, leading us to hypothesize that mediators of cytoskeleton reorganization, such as Coro1a, Dock2, or Cdc42, may play a role in this unique migration process.
Additionally, we observed that directly migrating T-cells created a local inflammatory microenvironment, which attracts circulating T-cells. Histological analysis confirmed that alloreactive PPs-derived T-cells and bloodborne T-cells colocalized. We employed two experimental approaches, including either photoconversion of T-cells in PPs or direct transfer of activated T-cells into the vasculature, to demonstrate this colocalization. We hypothesize that cytokines released by migrating T-cells, such as IFN-γ and TNF-α, may play a role in recruiting T-cells from the vasculature, as inhibiting chemokine-coupled receptors did not impair recruitment.
Die akute Graft-versus-Host Erkrankung (GvHD), insbesondere die Darm GvHD, stellt weiterhin eine der Hauptursachen für Mortalität und Morbidität nach allogener SZT dar. Aktivierte, alloreaktive Spender T-Zellen infiltrieren dabei über die Blutbahn die intestinale Lamina Propria. Erst kürzlich konnten wir zeigen, dass neben der vaskulären Migration ein Teil der Spender T-Zellen auch direkt aus den PP in die angrenzende Lamina Propria migrieren. Um Faktoren, die diese direkte Migration fördern, zu untersuchen und die direkt migrierenden T-Zellen genauer zu charakterisieren, verwendeten wir ein MHC-inkompatibles Mausmodell zur Induktion einer akuten GvHD.
Durch RNA Sequenzierung und Massenspektrometrie lasermikrodissezierter Darmschleimhautproben konnte eine starke Expression der Chemokine CXCL9, CXCL10, CXCL11, CCL3, CCL4 und CCL5 während der akuten intestinalen GvHD aufgezeigt werden. Neben CCL4 und XCL1 wiesen verschiedene Faktoren der T-Zellaktivierung, wie CD3ζ, LAT, Lck und ZAP70, sowie Faktoren der zytoskelettalen Reorganisation, wie Dock2, Coro1α und Parvin-γ, eine vermehrte Expression insbesondere nahe der PP auf. Die Expression der migrationsfördernden Faktoren Coro1α und Parvin-γ in Spender T-Zellen nahe der PP konnte anschließend mittels histologischen Immunfluoreszenzfärbungen bestätigt werden. Durchflusszytometrische Analysen konnten weiterhin eine vermehrte Expression von CCR5, CCR9 und Intgerin α4β7 auf den vornehmlich Tbet+ Spender T-Zellen nahe der PP nachweisen. Funktionelle in vitro Migrationsversuche zeigten abschließend, dass in vivo aktivierte Spender T-Zellen eine gerichtete Migration in Richtung auf CXCL11 und zu späterem Zeitpunkt auch auf CCL4 vollziehen können.
Zusammenfassend zeigt diese Arbeit die Bedeutung zahlreicher Chemokine für das sequenzielle T-Zell-Homing während der akuten intestinalen GvHD. Neben der insbesondere durch Faktoren der zytosekeletalen Reorganisation vermittelten amoeboiden Migration kann auch eine mesenchymale Fortbewegung über Faktoren wie CCR5, CCR9 und Integrin α4β7 die direkte Migration der T-Zellen fördern. Den direkt migrierenden vornehmlich TH1 polarisierten Zellen folgen weitere, CD27 und Integrin αLβ2 exprimierende, zytotoxische T-Zellen aus der Blutbahn. Die direkt migrierenden Zellen könnten als Initiator und Potentiator der intestinalen T-Zell Infiltration wirken und müssen für zukünftige therapeutische Strategien nicht nur der Darm GvHD, sondern der intestinalen Inflammation im Allgemeinen mitberücksichtigt werden.
In Ratten und Mäusen aktiviert der superagonistische anti-CD28 monoklonale Antikörper (CD28SA) vorzugsweise regulatorische T-Zellen. In niedriger Dosierung führt CD28SA zu einer fast ausschließlichen Aktivierung von regulatorischen T-Zellen (Tregs). Diese Beobachtung konnte inzwischen auch für menschliche Zellen in Zellkultur bestätigt werden.
In gesunden und freiwilligen Testpersonen deutet die Zytokin-Antwort nach Applikationen von niedrigen CD28SA-Dosen darauf hin, dass sich diese Beobachtung auch in-vivo bewahrheitet. Eine Gabe von CD28SA in niedriger Dosierung, die zu einer exklusiven Aktivierung von regulatorischen T-Zellen führt, könnte somit in der Behandlung von Autoimmunkrankheiten oder von entzündlichen Erkrankungen eingesetzt werden.
Eine mechanistische Erklärung für dieses Phänomen blieb lange Zeit unklar. Die CD28SA-vermittelte T-Zell-Aktivierung ist abhängig von der Verstärkung von basalen tonischen Signalen, die T-Zellen über ihren T-Zell-Rezeptor erhalten. Diese Tatsache führte zu der Hypothese, dass die schwachen, tonischen Signale, die konventionelle CD4+ T-Zellen in Abwesenheit ihrer spezifischen Antigene über den T-Zell-Rezeptor erhalten, ein stärkeres CD28 Signal für ihre Aktivierung benötigen als die selbstreaktiven regulatorischen T-Zellen, die ein stärkeres Selbstpeptid-TCR Signal erhalten.
In dieser Arbeit konnte gezeigt werden, dass die Blockade von MHC-Klasse-II-Molekülen in Mäusen, in-vitro und in-vivo, den Vorteil der regulatorischen T-Zellen gegenüber den konventionellen T-Zellen bezüglich der Antwort auf niedrige CD28SA Dosierungen, aufhebt.
Trotz zahlreicher Fortschritte im Verständnis der Funktionsweise des kostimulatorischen Rezeptors CD28 in Mensch, Maus, Ratte und Makake ist nach wie vor wenig hierüber in Bezug auf das Tiermodell Schwein bekannt. Die vorliegende Arbeit untersucht die Funktion und Expression von CD28 in Schweine-T-Zellen sowie die Regulierbarkeit der T-Zellaktivierung durch anti-pCD28 mAb. Die Ergebnisse zeigen, dass hierbei vor allem CD4+ und CD8+ T-Zellen differenziert betrachtet werden müssen. Grundsätzlich unterscheiden sich die beiden T-Zellpopulationen in der CD28 mRNA Expression, im Expressionsverhältnis zwischen CD28 mRNA und Protein, sowie im proliferativen Ansprechen auf anti-pCD28mAb. So reagierten CD4+ im Vergleich zu CD8+ T-Zellen auf die kostimulatorische Inkubation mit anti-pCD28 mAb des Klons 3D11 sensibler. In direkt stimulatorischen Ansätzen zeigte sich, dass CD4+ und CD8+ T-Zellen durch unterschiedliche anti-pCD28 mAb differentiell angesprochen werden können. Eine superagonistische Funktion konnte für CD4+ T-Zell aktivierende anti-pCD28 mAb in den bisherigen Versuchen noch nicht beobachtet werden. Letzteres ist hierbei vor allem für den Transfer von vielversprechenden Therapiestrategien vom Kleintier- zum Großtiermodell auf dem Weg zur Entwicklung neuer Therapieoptionen für Autoimmunerkrankungen, Erkrankungen mit starker proinflammatorischer Aktivität und dem Myokardinfarkt von Bedeutung.
The defense against invading pathogens is, amongst other things, mediated via the action of antibodies. Class-switched antibodies and antibodies of high affinity are produced by plasma cells descending from germinal center B (GCB) cells. GCB cells develop in the germinal center (GC), a specialized microstructure found in the B-cell follicle of secondary lymphoid organs. GCB-cell maturation and proliferation are supported by follicular T- helper (Tfh) cells. On the other hand, follicular regulatory T (Tfr) cells control this process in quantity and quality preventing, for instance, the formation of autoantibodies directed against endogenous structures. The development of GCB, Tfh and Tfr cells essentially depends on the migration into the GC, which is mediated via the expression of the chemokine receptor CXCR5.
One transcription factor highly expressed in follicular T cells, comprising Tfh and Tfr cells, is NFATc1. Tfr cells additionally express the transcriptional repressor Blimp-1, which is not expressed in Tfh cells. We found that NFATc1 is transactivating Cxcr5 via response elements in the promoter and enhancer in vitro. Blimp-1 binds to the same elements, transactivating Cxcr5 expression in cooperation with NFATc1, whilst mediating Cxcr5- repression on its own. In Tfr cells Blimp-1 suppresses CXCR5 expression in the absence of NFATc1. Blimp-1 itself is necessary to restrict Tfr-cell frequencies and to mediate Tfr- cell function as in mice with Blimp-1-ablated Tregs high frequencies of Tfr cells do not reduce GCB- or Tfh cell frequencies. NFATc1 and Blimp-1 double deficient Tfr cells show additional loss of function, which becomes visible in clearly expanded antibody titers.
To evaluate the function of NFATc1 in Tfr cells, we not only deleted it, but also overexpressed a constitutive active form of NFATc1/aA (caNFATc1/aA) in regulatory T cells (Tregs). The latter is leading to an upregulation of CXCR5 per cell, without changing Tfh or Tfr-cell frequencies. However, the high density of surface CXCR5 enhances the migration of Tfr cells deep into the GC, which results in a tighter control of the antigen- specific humoral immune response. Additionally, caNFATc1/aA increases the expression of genes coding for Tfr effector molecules like Il1rn, Il10, Tigit and Ctla4. Interestingly, this part of the transcriptional change is dependent on the presence of Blimp-1. Furthermore, Blimp-1 regulates the expression of multiple chemokine receptor genes on the background of caNFATc1/aA.
In contrast, when caNFATc1/aA is overexpressed in all T cells, the frequencies of Tfh- and GCB cells are dominantly reduced. This effect seems to stem from the conventional T- cell (Tcon) side, most probably originating from increased secretion of interleukin-2 (IL- 2) via the caNFATc1/aA overexpressing Tcons. IL-2 is known to hinder the germinal center reaction (GCR) and it might in its abundance not be neutralizable by Tfr cells.
Taken together, NFATc1 and Blimp-1 cooperate to control the migration of Tfr cells into the GC. Tfr cells in the GC depend on NFATc1 and Blimp-1 to perform their proper function. Overexpression of caNFATc1 in Tregs strengthens Tfr function in a Blimp-1-dependent manner, whilst overexpression of caNFATc1 in all T cells dominantly diminishes the GCR.
Modulation CD4+ humaner Treg- und Tconv-Zellen durch Inhibition der sauren Sphingomyelinase in vitro

Die saure Sphingomyelinase (ASM) stellt durch die Umwandlung von Sphingomyelin in Ceramid und Phosphorylcholin ein zentrales, fein reguliertes Enzym im Sphingolipidmetabolismus dar. Dadurch nimmt es Einfluss auf verschiedene zelluläre Mechanismen wie Signalvermittlung, Endo- und Exozytose und Zellaktivierung. Dementsprechend weitreichend ist auch die Bedeutung der ASM bei verschiedenen Krankheiten wie Arteriosklerose, Depression oder Neoplasien. Auch auf das Immunsystem, insbesondere auf die Signalvermittlung durch T-Zellen innerhalb des adaptiven Immunsystems, nimmt die saure Sphingomyelinase Einfluss. Aufbauend auf früheren Forschungsarbeiten zur pharmakologischen und genetischen Hemmung der ASM im Mausmodell untersuchten wir, welche Auswirkungen die Hemmung dieses Enzyms in humanen Zellkulturen auf die Population regulatorischer und konventioneller T-Zellen haben. Hierzu verwendeten wir die beiden selektiven Serotonin-Wiederaufnahmehemmer Sertralin und Citalopram; zwei antidepressiv wirksame Medikamente, die durch eine Verdrängung der ASM von der lysosomalen Membran eine hemmende Wirkung ausüben. Wir konnten zeigen, dass diese beiden Substanzen sowohl in Maus-T-Zellen, als auch in humanen T-Zellen, in der Lage sind, die Aktivität der sauren Sphingomyelinase zu inhibieren. Durch Kultivierung von Immunzellen der Maus zusammen mit den Inhibitoren konnte darüber hinaus eine Erhöhung der Treg-Zellfrequenz erreicht werden. Verschiedene Zellkulturexperimente mit humanen PBMCs zeigten weiterhin, dass unter gewissen Umständen so auch eine Vermehrung regulatorischer T-Zellen im Menschen möglich ist, und dass dies mutmaßlich durch Einbindung der ASM im CD3/CD28-Signalweg bedingt ist. In mit AntiCD3-Antikörper stimulierten experimentellen Ansätzen kam es jedoch nur bei einzelnen Individuen, die als Responder identifiziert werden konnten, zu einer Treg-Zellvermehrung. Umgekehrt kam es durch externe Zugabe von C6-Ceramid zu einer Verringerung des Anteils an regulatorischen T-Zellen. Des Weiteren wurden verschiedene Veränderungen im Expressionsverhalten von Treg- und Tconv-Zellen bezüglich CD25, CD69 und CTLA-4 in Anwesenheit der ASMInhibitoren beobachtet. Weiterhin bestätigte sich, dass die pharmakologische Hemmung der sauren Sphingomyelinase auch Auswirkungen auf die Effektorfunktion von T-Zellen hat. Während die Proliferation der Zellen weitgehend unbeeinträchtigt blieb, kam es zu einer verringerten Sekretion der Zytokine IFN-gamma, TNF, IL-5 und IL-10. In ihrer Gesamtheit sprechen diese Ergebnisse dafür, dass Inhibitoren der sauren Sphingomyelinase begünstigend auf Krankheitsgeschehen mit überschießender oder dysregulierter Aktivität des Immunsystems einwirken könnten. Immunmodulatorischen Wirkungen durch Inhibition der ASM erklären möglicherweise auch Einflüsse auf das Immunsystem, die für verschiedene Antidepressiva beschrieben wurden. Insgesamt ist die Bedeutung der sauren Sphingomyelinase innerhalb der Regulation des adaptiven Immunsystems jedoch noch ein weitgehend ungeklärtes Thema mit vielen offenen Fragen. Daher ist auch in Zukunft weitere klinische und experimentelle Forschung erforderlich, um zu klären, welchen Einfluss dieses Enzyms auf Immunzellen hat und wie sich dieser auch klinisch anwenden lässt.
In this thesis, three species were investigated for the conservation of two non-conventional T cell systems, the CD1d/ iNKT cell system and the BTN3/ Vγ9Vδ2 T cell system. Non-conventional T cells are αβ or γδ T cells that do not fit into the classical mode of antigen recognition and adaptive responses. These T cells recognize antigens different from classical peptide antigens and are not restricted to the polymorphic MHC molecules but rather to non-polymorphic antigen-presenting molecules. The iNKT cell subset is restricted by the lipid antigen-presenting molecule CD1d and carries out immunomodulatory functions by rapid cytokine secretion. The molecular basis of this system, the semi-invariant iNKT TCR chains and CD1d were proven to be expressed and compared to homologs in human and rodents. Cotton rats possess multiple members of the AV14 and BV8 family and only one isoform of CD1d which is comparable to findings in the rat.
Moreover, the reactivity of primary cells to glycolipid antigens could be shown, and an iNKT
cell-like population was detected in primary cells using newly developed cotton rat CD1d oligomers. These were also applied to test the capacity of CD1d to present typical glycolipid
antigens to iNKT TCR transductants. In addition, expression of cotton rat iNKT TCR α and β chains in TCR-negative cell lines was used to show successful pairing and detection of glycolipids in the context of CD1d. In summary, the conservation of a functional CD1d/iNKT cell system in the cotton rat could be shown, and tools were developed to study this cell subset in the course of infectious diseases. The Vγ9Vδ2 T cell subset is the major γδ T cell subset in human peripheral blood and has the unique ability to contribute to immune surveillance by detecting pyrophosphorylated metabolites of isoprenoid synthesis that indicate cell stress, transformation or infection. Up to this date, phosphoantigen-reactive γδ T cells have only been shown in primate species. However, evidence for the existence and functional conservation of the genes implied in the BTN3/Vγ9Vδ2 T cell system was found in several placental mammal species,
and two candidate species were chosen for further investigation. The nine-banded armadillo, a valuable model for leprosy research, was shown to possess homologous genes to TRGV9, TRDV2 and BTN3. In this study, the expression of productive rearrangements of TRDV2 gene segments could be shown in peripheral blood samples, but no evidence was found for the expression of a functional TRGV9 rearrangement or BTN3 molecules. Moreover, determinants of phosphoantigen-reactive Vγ9Vδ2 T cells and functional BTN3 molecules were found to still be prevalent in armadillo genes. This makes the armadillo an interesting model to study the structural determinants that allow phosphoantigen recognition by a functional Vγ9Vδ2 T cell subset although this species is merely a witness for a functional system in a placental mammal ancestor. In contrast, alpacas were shown to express functional Vγ9Vδ2 T cells which conserved many features of the human counterpart. Expression of Vγ9Vδ2 pairings could be shown by single-cell PCR and functional phosphoantigenreactive pairings were observed. This phosphoantigen reactivity was also shown in PBMC cultures with a newly developed antibody specific for alpaca Vδ2Jδ4 chains. Moreover, a more detailed study of the alpaca TCR repertoire showed similarities to “γδ high” species like
camelids and cattle which possess an extended family of TRDV genes. The γ and δ loci of alpaca
TCR genes were drafted based on genomic information and cDNA studies and provide an overview for more detailed studies. Conservation of phosphoantigen recognition by the single BTN3 molecule of alpacas was shown in 293T knock out cell lines, and BTN3 detection on PBMCs was investigated with a newly developed alpaca BTN3-specific antibody. These findings prove the existence of a functional BTN3-dependent phosphoantigen-reactive Vγ9Vδ2 T cell subset and provide a basis for the future study of this cell system in a non-primate species. Moreover, as the first non-primate candidate species with the BTN3/Vγ9Vδ2 T cell system the alpaca is an important outgroup for research in this field. The use of a single BTN3 variant in contrast to three human isoforms that work together renders the alpaca a unique and to this date indispensable model for Vγ9Vδ2 T cells.
In conclusion, this study provides an overview of the applicability of new animal models in the
study of the non-conventional T cell subsets iNKT cells and Vγ9Vδ2 T cells and leads the way for a better understanding of structural and functional relationships.
T cell infiltration into the intestine occurs after priming and activation in the mesenteric lymph nodes and Peyer’s patches and subsequent trafficking via the blood circulation. We hypothesized that additionally to the vascular trafficking route, a fraction of T cells in the Peyer’s patches directly migrate into the adjacent lamina propria of the small intestine. To test this hypothesis, we employed a mouse model of acute Graft-versus-Host Disease to study the direct T cell migration from the Peyer’s patches to the adjacent lamina propria.
First, we analyzed the border of Peyer’s patches on histological sections and found that the Peyer’s patch is not enclosed by a capsule or basement membrane. Thus, the tissue architecture allows for direct access to the surrounding tissue. With whole-mount light sheet fluorescence microscopy we quantified a three-dimensional gradient of T cells around Peyer’s patches on day 2.5 and day 3 after transplantation. This gradient evened out at day 4 and day 6 when high numbers of T cells started to evenly infiltrate the intestine from the blood circulation. We confirmed that gradient-forming T cells around Peyer’s patches resided within the tissue parenchyma of the lamina propria and not inside lymphatic vessels.
To positively prove that the recently activated donor T cells around Peyer’s patches have egressed directly from that patch, we established a protocol for intravital photoconversion of T cells inside Peyer’s patches. 12 h after photoconversion inside a single Peyer’s patch, photoconverted T cells resided only around this particular Peyer’s patch and not elsewhere in the small intestine. This indicated that the T cells did not infiltrate via the blood but migrated to the adjacent lamina propria of the small intestine. Dynamic intravital two-photon microscopy revealed that these T cells next to the Peyer’s patch migrated in a random pattern. This suggested that these cells did not follow a positive chemoattractive gradient once they had reached the lamina propria. Laser-capture microdissection combined with RNA sequencing of the mucosa near the Peyer’s patch identified a wide range of migration-promoting factors. These included chemokines, co-stimulatory receptors and migration-associated intracellular molecules, which are candidates to promote this direct migration from Peyer’s patches.
Altogether, we demonstrate for the first time that additionally to the vascular trafficking route, a fraction of T cells migrates directly from the Peyer’s patch to the surrounding mucosa. This mechanism implies so far unrecognized regional specification of Peyer’s-patch-primed T cells. Our findings may impact treatment strategies to avoid intestinal inflammation or foster immunity after oral vaccination.
Infektionen durch C. albicans auf den Schleimhäuten sind eine häufige Erkrankung bei Patienten mit einer Schwächung der T-Zellimmunität. Blutstrominfektionen mit der Hefe C. albicans (Candidämie) stellen, vor allem bei Patienten auf Intensivstationen, eine nach wie vor bedrohliche Komplikation mit hoher Letalität dar.
Das pH-regulierte Antigen 1 (Pra1) ist ein Protein, das von C. albicans produziert wird, auf der Oberfläche des Pilzes gebunden vorkommt und auch vom Pilz in den Überstand sezerniert wird. Im humanen System bindet das Protein an T-Zellen an das Oberflächenprotein CD46. Es ist des Weiteren bekannt, dass das Pra1 an bestimmte Immunzellen der Maus (Monozyten und Phagozyten) binden kann. Eine Bindung an T-Zellen der Maus ist bisher nicht beschrieben. Eine genaue Charakterisierung der Interaktion von Pra1 mit Immunzellen der Maus ist interessant, da die Maus als biologischer Modellorganismus zur Erforschung der Infektion mit C. albicans dient. In dieser Arbeit konnte gezeigt werden, dass rekombinantes Pra1 (rPra1) auch an Maus-CD4+ T-Zellen binden kann.
Es wurden Einflussfaktoren auf die gefundene Bindung von Pra1 an CD4+ T- Zellen gesucht. Als ein Einflussfaktor wurde Zink identifiziert. Pra1 kann an freies Zink binden und durch Zugabe von ZnCl2 während der Inkubation von Pra1 mit T-Zellen kann das Signal von gebundenem Pra1 an CD4+ T-Zellen erhöht werden. Aspf2, ein Protein aus Aspergillus fumigatus mit großer Homologie zu Pra1, kann nicht an diese Zellen binden.
Im in-vivo-Experiment mit Tieren, die mit C. albicans infiziert wurden, konnte kein wildtypisches sezerniertes Pra1 gebunden an T-Zellen nachgewiesen werden. Zellkulturüberstände von C. albicans zeigten nach Inkubation in vitro mit T-Zellen ein Signal für gebundenes Pra1 an CD4+ T-Zellen.
Die Bindungskinetik von Pra1 an T-Zellen zeigte eine über die Zeit der Inkubation konstante Zunahme des Signals von zellgebundenem rPra1 an CD4+ T-Zellen. In der off-Kinetik fand sich eine Abnahme des Signals über die Zeit bis an die Grenze der Nachweisbarkeit.
Der Bindungspartner von Pra1 auf T-Zellen konnte nicht identifiziert werden. Die strukturell und funktionell verwandten Oberflächenproteine Crry, CD59a und CD55 wurden auf Bindungsfähigkeit an T-Zellen in entsprechenden Knockout- Mäusen getestet, konnten jedoch als Rezeptor für Pra1 ausgeschlossen werden. Durch die Bindung von sezerniertem Pra1 an neutrophile Granulozyten wird die Fähigkeit dieser Zellen zur Phagozytose eingeschränkt. Die Bindung von Pra1 an CD4+ T-Zellen führt zur Kostimulation der T-Zellen, also zur verstärkten Zellaktivierung und Proliferation. Durch die Zugabe von 10 μM Zinkchlorid wird die kostimulatorische Aktivität von Pra1 verstärkt.
Während der Zellaktivierung von Effektor-Memory-CD4+ T-Zellen reduziert rPra1 die Sekretion von IFN-γ. Diese Reduktion von IFN-γ-produzierenden Zellen entsteht nicht durch einen Einfluss von Pra1 während der Zellaktivierung von naiven CD4+ T-Zellen zu Th1-Zellen und auch nicht durch die Auslösung von Apoptose in IFN-γ-produzierenden Th1-Zellen. Die Bindung von Pra1 an CD4+- T-Zellen, die über den T-Zell-Rezeptor aktiviert werden, reduziert in vitro die Sekretion des Zytokins. Zusätzlich werden weitere Zytokine in ihrer sezernierten Menge reduziert wie IL-2 und TNF-α.
