The most commonly used assay to quantitate the response of peripheral T~cells upon stimulation with growth factors is determination of incorporated (JH]TdR. We compared thls test to three other methods: 1. direct countlog of cells with a Coulter type counter as reference assay, 2. a colorimetric assay using the tetrazolium dye 3-[ 4,S-dimethylthiazol-l-yl]-2,5diphenyl tetrazolium (MTT), which is a cheap and increasingly popular non-radioactive method and 3. incorporation of the thymidine analog 5-bromo-2'-deoxyuridine detection with a monoclonal antibody on cytospins. Primary human PHA-blasts from >30 healthy individuals were stimulated with IL-2, IL-4 aod IL-7 and assayed with up to four different methods. We discuss the advantages and disadvantages of the assays used and tbe effects of differences between cell preparations. We observed no significant variations between individuals for the dose dependence, but the relative emctency of IL4 compared to IL-2 and IL-7 was variable. This was probably due to the slower response observed upon stimulation with this factor.
Bispecific T cell engager (BiTE) display a novel design among the class of bispecific antibodies and hold great promise to fight diverse cancers. BiTE molecules consist of two different binding entities derived from two human IgG antibodies connected by a short peptide linker. Their binding arms are directed against the CD3e chain of the T cell receptor on T cells and against an antigen that is specific for (e.g., CD19 for lymphoma in MT103) or over-expressed on (e.g., EpCAM for epithelial cancer in MT110) tumor cells. Without requirement for pre- or co-stimulation, BiTE molecules efficiently redirect CD3+ T cells towards tumor cells expressing the relevant target antigen. Only a BiTE molecule simultaneously bound to both tumor cell and T cell activates the T cell to exert its cytolytic function resulting in tumor cell death. In T cells stimulated with both BiTE and target cells, elevated levels of caspase activation and increased expression of cytotoxic and signaling proteins are observed. These include cytolytic proteins granzyme B and perforin, activation markers CD69 and CD25 and adhesion molecules CD2 and LFA-1. Activated T cells secrete the usual mix of cytokines, among them pro-inflammatory cytokines IFN-g and TNF-a. The membrane of tumor cells expressing the relevant target antigen is perforated during the attack of BiTE-stimulated effector cells as can be concluded from adenylate kinase release from the cytosol of tumor cells. Ca2+-chelator EGTA completely blocked BiTE-mediated activation of caspases and tumor cell lysis. As perforin is strictly Ca2+-dependent, a major role for this pore-forming protein is assumed for the elimination of tumor cells via BiTE-stimulated T cells. Granzyme B and caspases are main players in BiTE-mediated elimination of tumor cells. Inhibitors of granzyme B or caspases reduce or block, respectively the activation of caspases. However, other signals of apoptosis (cleavage of PARP and fragmentation of DNA) were only reduced by granzyme B inhibitor or caspase inhibitor. Most interestingly, the lytic capacity of BiTE molecules was not impaired by granzyme B inhibitor or caspase inhibitor. It seems that there is no requirement for granzyme B and caspases to be present simultaneously. Instead the data presented provide evidence that they can be replaced one at a time by related proteins. Pre-incubation of effector cells with the glucocorticoids dexamethasone or methylprednisolone resulted in markedly decreased secretion of cytokines by T cells yet only a small reduction in the expression of activation markers and adhesion molecules on T cells and specific lysis of tumor cells upon BiTE stimulation. Soluble factors secreted in an undirected manner by BiTE-stimulated T cells do not mediate tumor cell death by themselves. Bystander cells negative for the antigen that is recognized by the BiTE molecule will not be compromised by BiTE activity. The cytokine TGF-b reduced proliferation as well as granzyme B and perforin expression of BiTE-stimulated T cells. Redirected lysis by BiTE-activated T cells was also decreased under the influence of TGF-b, however lysis was still performed at a reasonable rate (72 % of target cells). TGF-b does not exert a deleterious effect on lytic potential of BiTE-stimulated T cells. The minimal anticipated biological effect level for the BiTE MT110 was determined for the entry of MT110 into phase I clinical studies. Experiments analyzing redirected lysis of tumor cells, expression of activation marker CD25 and cytokine release by T cells revealed a MABEL value of 50 pg/ml for MT110.
Zytotoxische CD8+ T-Lymphozyten spielen in vielen inflammatorischen, aber auch primär neurodegenerativen Erkrankungen eine wichtige Rolle. Daher besitzt die Fragestellung inwiefern CD8+ ZTL Neurone direkt schädigen und ggf. welche mechanistischen Aspekte dieser Schädigung zugrunde liegen, eine hohe Relevanz. Um diese Fragestellung eingehender zu beleuchten, wurde mit dem OT-I-System gearbeitet. Dieses gut vorcharakterisierte CD8+ T-Zell-Modell besitzt den Vorteil, dass diese transgenen Zellen nur eine Peptidsequenz des Ovalbumin (OVA) Protein als spezifisches Antigen erkennen.
Zunächst wurden in der vorliegenden Arbeit Co-Kultivierungs-Experimente durchgeführt. Hierzu wurden akut isolierte murine Hippokampus-Neurone unter verschiedenen Bedingungen mit OT-I Lymphozyten co-kultiviert. Hierbei konnte gezeigt werden, dass unter Antigenpräsentation der Neurone signifikant mehr Neurone in die Apoptose/Nekrose geführt werden, als unter Kontroll-Bedingungen, in denen entweder kein Antigen oder ein Antigen, das nicht von OT-I Lymphozyten erkannt wird, präsentiert wird.
Nachdem die Antigen-abhängigen zytotoxischen Effekte auf Neurone gezeigt werden konnten, wurde mithilfe elektrophysiologischer Techniken die mechanistischen und funktionellen Konsequenzen des direkten neuronalen/OT-I-vermittelten Zellkontakts untersucht. Bei diesem experimentellen Ansatz wurde durch elektrisches Auslenken eines Neurons nach Kontakt mit einem OT-I Lymphozyt die passiven elektrischen Parameter der Neuronenmembran gemessen. In diesen Messungen konnte gezeigt werden, dass nach unmittelbarem Kontakt eines Neurons mit einem OT-I Lymphozyt der neuronale Membranwiderstand reduziert wird bzw. die Leitfähigkeit der Zellmembran erhöht wird. Diese Änderung der neuronalen Membran-Leitfähigkeit findet in einem Zeitraum von 10 min nach dem Zell-Zell-Kontakt statt. Auch hier konnte gezeigt werden, dass dieser Einfluss von OT-I Lymphozyten auf Neurone strikt Antigen-abhängig ist. Zur Untersuchung des Mechanismus der OT-I T-Lymphozyten auf Neurone wurde das Augenmerk auf verschiedene T-Zell-induzierte Apoptosewegegelegt. Es konnte gezeigt werden, dass durch Blockieren der Fas/FasL-Interaktion mittels eines Antikörpers kein Unterschied, weder in der neuronalen Apoptoserate nach Co-Kultivierung, noch eine Änderung der passiven neuronalen Membran-Leitfähigkeit auftritt. Weiterhin wurde die Rolle der von T-Zellen sezernierten Granula Perforin und Granzym B untersucht. Um den Einfluss dieser Granula aufzuklären, wurden OT-I Lymphozyten verwendet, die entweder defizient für Perforin oder Granzym B waren. In diesem experimentellen Ansatz wurde gezeigt, dass ausschließlich Perforin für die Erniedrigung des passiven neuronalen Membran-Widerstandes verantwortlich ist.
Diese Erhöhung der neuronalen Membranleitfähigkeit führte aber nicht direkt zum neuronalen Zelltod. Vielmehr wurde durch die einhergehende Depolarisation des Neurons die elektrische Aktivität der Zelle vermindert, sodass es zu einem sogenannten „electrical silencing“ kommt. Dieser Umstand konnte auch in der Betrachtung der spontanen Netzwerkaktivität von Neuronenkulturen gezeigt werden. Hierfür wurden hoch dichte Neuronenkulturen auf MEA-Chips kultiviert. Mit Hilfe dieser MEA konnten die Summenfeldpotentiale der Neuronenkulturen detektiert werden. Hierbei wurde beobachtet, dass nach Beladung der Neuronen mit dem spezifischen OT-I-Antigen und OT-I Zellen eine Verringerung der spontanen Netzwerkaktivität einhergeht. Auch in diesem Effekt konnte eine Antigen-Spezifität nachgewiesen werden.
Da der Prozess der zellulären Apoptose mit einem Anstieg der intrazellulären Ca2+-Konzentration einhergeht, und Perforin als Ca2+-durchlässiger unselektiver Porenbildner fungiert, wurden zur Überprüfung der Hypothese calcium imaging-Experimente durchgeführt. Analog zu den elektrophysiologischen Messungen wurde gezeigt, dass nach direktem Zell-Zell-Kontakt zwischen Neuron und OT-I Lymphozyt eine Erhöhung der intrazellulären Ca2+-Konzentration zu messen ist. Dass diese Änderung des neuronalen Ca2+-Einstroms durch Perforin-abhängige Membranporen hervorgerufen wird, konnte durch die Verwendung von Perforin-defizienten OT-I Lymphozyten bewiesen werden. Unter Verwendung von Perforin-defizienten OT-I Lymphozyten wurde keine Änderung der neuronalen Ca2+-Konzentration ermittelt. Weiterhin wurde in diesem experimentellen Ansatz gezeigt, dass auch der OT-I-vermittelte neuronale Ca2+-Anstieg strikt Antigen-abhängig ist.Zusammengefasst konnte in dieser Arbeit gezeigt werden, dass MHC-I/Antigen-vermittelte CD8+ Lymphozyten-Interaktion mit einem Neuron zu „electrical silencing“ des Neurons führt. Dieser Prozess ist klar Perforin-abhängig, führt jedoch nicht zum unmittelbaren Zelltod des Neurons.
Cofilin

This study has identified cofilin, an actin binding protein, as a control element in the reorganization of the actin cytoskeleton which is highly relevant for T lymphocyte activation. Cofilin is regulated in its activity by reversible phosphorylation which is inducible by stimulation through accessory receptors such as CD2 and CD28. First it could be demonstrated that accessory receptor triggering induces the transient association of cofilin with the actin cytoskeleton and that only the dephosphorylated form of cofilin possesses the capacity to bind cytoskeletal actin in vivo. PI3-kinase inhibitors block both the dephosphorylation of cofilin and its association with the actin cytoskeleton. Importantly, cofilin, actin, PI3-kinase and one of its substrates, namely phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2) which can bind to cofilin, co-localize within CD2-receptor caps. The cofilin/F-actin interaction has been identified as a crucial regulatory element for receptor cap formation and the strength of signal transduction. To this end, appropriately designed cell permeable non-toxic peptides that are homologous to actin binding motifs of the human cofilin sequence were introduced into untransformed human peripheral blood T lymphocytes. These peptides competitively and dose dependently inhibit the activation induced interaction of cofilin with the actin cytoskeleton in vivo. By this approach it was possible to study, for the first time, the functional consequences of this interaction in immunocompetent T cells. The present data demonstrate that inhibition of the actin/cofilin interaction in human T lymphocytes by means of these cofilin derived peptides abolishes receptor cap formation and strongly modulates functional T cell responses such as T cell proliferation, interleukin-2 production, cell surface expression of CD69, gIFN production, and CD95L expression. Importantly, receptor independent activation by PMA and calcium ionophore circumvents these peptide produced inhibitory effects on lymphocyte stimulation and places the cofilin/actin interaction to a proximal step in the cascade of signaling events following T cell activation via surface signals. The present results are novel since as yet no information existed regarding the molecular elements which link cell surface receptor stimulation directly to the resulting reorganization of the actin cytoskeleton.
T cell activation is supposed to require two signals via engagement of the TCR and a costimulatory molecule. However, the signaling cascade of costimulatory molecules has remained elusive. Here, I provide evidence that CD44 supports proliferation as well as apoptosis mainly, if not exclusively, by enhancing signal transduction via the TCR/CD3 complex. Blockade of CD44 interferes with mounting of an immune response. This has been demonstrated by the significantly decreased IL-2 production of a T helper line, when stimulated in the presence of a competing CD44 receptor globulin. To evaluate the underlying mechanism, CD44 was cross-linked by an immobilized antibody (IM7). Cross-linking of CD44 induces proliferation of peripheral T cells and apoptosis of thymocytes and a T helper line in the presence of subthreshold levels of anti-CD3. CD44-induced proliferation was accompanied by an upregulation of the activation markers CD25 and CD69 and an increased cytokine production. TCR-mediated apoptosis was accompanied by an upregulation of CD95 ligand and CD95 receptor, which could be greatly enhanced by costimulation via CD44. On the level of signal transduction, coligation of CD44 with CD3 resulted in a strong and sustained increase of early tyrosine phosphorylation events and upregulated downstream signal transduction pathways, such as the ras/ERK and the JNK signaling cascades. These pleiotropic effects of CD44 are due to its involvement in the most proximal events in TCR signaling, as demonstrated by a strong increase in the phosphorylation of the TCR z-chain and ZAP-70. Notably, cross-linking of CD44 was binding-site dependent and was only effective when supporting colocalization of the TCR/CD3 complex and CD44. Cross-linking of CD44 via immobilized IM7 also induced profound changes in cell morphology, characterized by strong adhesion, spreading and development of surface extensions, which were dependent on a functional tubulin and actin cytoskeleton. These cytoskeletal rearrangements were mediated by rac1, a small GTPase of the rho subfamily, and src-family kinases, two of which, fyn and lck, were found to be associated with CD44. By cross-linkage of CD44 these kinases were redistributed into so called lipid rafts. It is supposed that for T cell activation a relocation of the TCR/CD3 complex into the same membrane microdomains is required. The data are interpreted in the sense that the costimulatory function of CD44 relies on its cooperativity with the TCR. Most likely by recruitment of phosphokinases CD44 significantly lowers the threshold for the initiation of signaling via the TCR. The requirement for immobilized anti-CD44, the necessity for neighbouring anti-CD3 and the dependence on the binding site of CD44 strongly suggest that the costimulatory mechanism involves cytoskeletal rearrangements, which facilitate recruitment and redirection of src-family protein kinases in glycolipid enriched membrane microdomains.
Die Expression der Hämatopoetischen Progenitor Kinase 1 (HPK1), einem Mitglied der Familie der „Germinal Centre“ Kinasen, ist im adulten Organismus auf die Zellen des hämatopoetischen Systems beschränkt. Die HPK1 wurde ursprünglich als ein Aktivator des JNK-Signalübertragungsweges beschrieben [Hu et al., 1996; Kiefer et al., 1996], und kürzlich wurde eine transiente Aktivierung der HPK1 nach TZR-Stimulation nachgewiesen. Auch wurde eine Assoziation der HPK1 mit dem Linker aktivierter T-Zellen (LAT) und den Adaptorproteinen Nck, Crk, Gads, Grb2, Grap, CrkL sowie SLP-76 gezeigt. Für die Aktivierung der nach TZR-Stimulation in den Lipid-Rafts lokalisierten HPK1 sind sowohl Lck als auch ZAP-70 notwendig [Liou et al., 2000; Liu et al., 2000a; Ling et al., 2001]. Diese Daten legen eine mögliche Funktion von HPK1 bei der TZR-vermittelten Signalübertragung nahe. Trotzdem konnte bisher eine physiologische Rolle der HPK1 im Rahmen der Immunrezeptor-Signalübertragung nicht nachgewiesen werden. In der vorliegenden Arbeit wird dargestellt, dass eine wichtige Funktion der HPK1 in T-Zellen nach TZR-Stimulation, die durch die Förderung des Aktivierungs-induzierten Zelltodes (AICD) vermittelt wird, in der Kontrolle der Termination der Immunantwort und damit Homöostase des Immunsystems besteht. Dies wurde durch retrovirale Überexpression der wildtypischen (wt) HPK1 in murinen CD4+ T-Zellen nachgewiesen, in denen die HPK1 zu einem Anstieg der spontanen und antiCD3-vermittelten Apoptose sowie zu einer gesteigerten Expression des Fas-Liganden (FasL oder auch CD95L) führte. Die Expression einer HPK1-„antisense“ (AS)-RNA in CD4+ T-Zellen bewirkte dagegen eine schwache, jedoch signifikant nachweisbare Hemmung der Apoptose und FasL-Expression. Die Apoptose-Hemmung durch die HPK1-AS-RNA war besonders stark in H2O2-stimulierten EL-4 T-Zellen ausgeprägt, in denen die Überexpression der wt HPK1 den durch reaktive Sauerstoffmetabolite (ROS) induzierten Zelltod verstärkte. Aus diesen Daten folgt, dass die HPK1 die T-Zell-Apoptose reguliert. In H2O2-stimulierten EL-4 T-Zellen führt die HPK1-Expression zu einer verstärkten und anhaltenden Aktivierung der c-Jun N-terminalen Kinase (JNK), die wahrscheinlich an der HPK1-vermittelten Apoptoseinduktion beteiligt ist. Unter den gleichen Bedingungen konnte eine schnelle Spaltung der HPK1 beobachtet werden. Die Überexpression der N- oder C-terminalen Spaltprodukte in CD4+ T-Zellen führte - wie die der Gesamt-HPK1 - zu einem Anstieg des AICD. In Übereinstimmung mit publizierten Daten konnten wir eine Hemmung der NFkB-Aktivität durch das C-terminale HPK1-Peptid nachweisen, das die IkBalpha-Degradation inhibiert. Die erzielten Ergebnisse führten uns in ihrer Gesamtheit zu folgendem Modell: während der Initiationsphase der T-Zell-Stimulierung werden nach schneller, transienter HPK1-Aktivierung pro- und anti-apoptotische Signale durch den JNK- und NFkB-Signalübertragungsweg vermittelt. Durch die Akkumulation der C-terminalen HPK1-Spaltprodukte kommt es später zur Inhibierung der NFkB-Aktivität und damit zu einer Verschiebung des Gleichgewichts zwischen den Überlebens- und Apoptose-stimulierenden Signalen zugunsten des AICD. Allerdings gibt es sicherlich weitere Faktoren und Signalwege, die an der HPK1-vermittelten Kontrolle der T-Zell-Apoptose beteiligt sind und von deren Untersuchung ein detaillierteres Verständnis der HPK1-Physiologie erwartet wird. Die Nukleären Faktoren aktivierter T-Zellen (NFAT´s) gehören zu einer Familie von Transkriptionsfaktoren, denen eine konservierte, ca. 300 Aminosäuren (aa) große DNA-Bindedomäne und eine Calcineurin-Bindedomäne gemeinsam ist. NFATc (auch NFATc1 oder NFAT2 genannt) und NFATp (NFATc2 oder NFAT1) werden in peripheren T-Zellen stark exprimiert und kontrollieren deren Effektorfunktionen u.a. über die Expression von IL-2, IL-3, IL-4, IL-5, IFNgamma und weiterer Lymphokine. Weitere von den NFAT´s kontrollierte Gene sind p21WAF1, der CD40L und der CD95L. Somit scheinen die NFAT´s bei der Zellzyklus-Kontrolle und beim AICD von T-Lymphozyten eine wichtige Rolle zu spielen. Daten unseres Labors zeigten, dass die T-Zell-Aktivierung zu einer massiven Induktion der kurzen Isoform A von NFATc innerhalb von 3-4 h führt [Chuvpilo et al., 1999b], noch vor dem Start des AICD. Dies ließ vermuten, dass sich die biologische Funktion von NFATc/A durch das Fehlen des C-terminalen Peptids von ca. 245 aa, das in allen anderen NFAT-Proteinen einschließlich der längeren Isoform NFATc/C vorhanden ist, unterscheidet. Deshalb sollte in der vorliegenden Arbeit die Induktion und Funktion von NFATc/A in murinen T-Lymphozyten untersucht werden. Es konnte gezeigt werden, dass die Infektion primärer CD4+ T-Lymphozyten mit NFATc/A-exprimierenden rekombinanten Retroviren, im Gegensatz zu der mit NFATc/C- oder NFATp-exprimierenden Retroviren, den AICD unbeeinflusst lässt. Dies deutet darauf hin, dass es durch die massive NFATc/A-Synthese nach Effektor-T-Zell-Aktivierung zur Induktion von Effektor-Funktionen kommt, ohne dass dabei die T-Zell-Apoptose beschleunigt wird. Im Gegensatz dazu üben die langen NFAT-Faktoren wie NFATc/C und NFATp eine pro-apoptotische Wirkung aus.
We examined the regulation of NFATc1 in different lymphomas and observed an inversed correlation between the methylation status and expression of NFATc1. Our data demonstrate that aberrant DNA methylation associated with chromatin remodeling within nfatc1 locus is a major mechanism for the repression of NFATc1 expression, suggesting that the DNA methylation-mediated transcriptional silencing of NFATc1 may be a critical event in the tumorogenesis of ALCLs and cHLs. Furthermore, the DNA methylation of human nfatc1 promoter region could be used as a novel biomarker of tumor progression. Our results indicate a close link between the loss of immunoreceptor signaling and NFATc1 expression in human lymphomas. For both ALCLs and cHLs, defects in immunoreceptor signaling have been described which result in a loss of receptor-mediated gene expression programs (Schwering et al., 2003; Bonzheim et al., 2004; Marafioti et al., 2004). In T cells, one indicator gene of these programs appears to be the nfatc1 gene whose expression is controlled by TCR signals (Chuvpilo et al., 2002a). In contrast, in T cells NFATc1 expression is unaffected by TCR signals, and NFATc2 was found to be expressed at normal levels in ALCLs and cHLs (L.K., unpubl. data). Moreover, the activity of NF-kappaB factors which can bind to certain NFAT binding sites and share a distantly-related DNA binding domain with NFATs is strongly elevated in cHL cells (Bargou et al., 1997; Hinz et al., 2001; Hinz et al., 2002) suggesting that NFATs and NF-kappaBs exert very different effects on generation and maintenance of Hodgkin’s lymhomas. However, it should be mentioned that in Burkitt’s and further B cell lymphomas in which NFATc1 proteins are strongly expressed and controlled by receptor signals (Kondo et al., 2003), they could exert a promoting function in tumor development. The genes of p53 family members p63 and p73 are prominent examples for mammalian genes whose products can act both as oncoproteins and tumor suppressor genes (Hibi et al., 2000; Stiewe and Putzer, 2002), and it is likely that more genes exist which encode both tumor suppressors and oncoproteins. It remains to be shown whether the nfatc1 gene is one of them.
Ziel dieser Arbeit war es, NF-AT1-Gen-defiziente Mauslinien zu erzeugen und die Folgen dieser genetischen Manipulation in vivo zu untersuchen. Die Untersuchung sollte die durch die Gendefizienz erwarteten Mängel während der Entwicklung (Embryogenese) und, im Besonderen, die Auswirkungen auf das Immunsytem und die Entwicklung und Differenzierung der T-Zellen aufzeigen. Zur Untersuchung der genomischen Organisation des Maus-NF-AT1-Gens wurde eine genomische l-Phagen-DNA-Bibliothek "gescreent" (durchgeführt von Dr. E. Jankevics, Universität von Riga, Lettland), die entsprechenden l-Phagen, die das NF-AT1-Gen enthielten, isoliert und die DNA präpariert. Nach Analyse der klonierten Phagen (Subklonierung und Sequenzierung) wurde eine Restriktionskarte der entsprechenden Bereiche erstellt und der "targeting-vector" erstellt. Der "targeting-vector" wurde durch Elektroporation in embryonale Stammzellen (ES-Zellen) eingebracht und die Integration in das Genom ("Homologe Rekombination") durch Southern Blotting- bzw. PCR-Analyse untersucht. Manipulierte ES-Zellklone wurden in C57Bl/6-Blastozysten injiziert, diese in scheinschwangere Ammentiere transferiert und die Nachkommen nach Geburt anhand der Fellfarben klassifiziert. Nachkommen mit einem hohen Anteil hellen Fells wurden mit C57 Bl/6-Tieren verpaart und die Integration des manipulierten Zellklons in die Keimbahn wurde anhand der wildtypischen Fellfarbe und Genotypisierung nachgewiesen. Bezüglich des manipulierten NF-ATp-Gens heterozygote F1-Tiere wurden miteinander verpaart, um eine homozygote NF-ATp-defiziente Mäuse zu erhalten. Die Deletion des NF-ATp-Proteins wurde durch in Western-Blotting-Experimenten und EMSAs ("electrophoretic mobility shift assays") nachgewiesen. Die NF-ATp-/--Tiere zeigten keine augenscheinlichen Veränderungen während der Entwicklung und, bei jungen Tieren, keine offensichtlichen Veränderungen bei der Entwicklung des Immunsystems. In älteren Tieren (> 6 Wochen) war eine Hyperproliferation der Zellen des Immunsystems zu beobachten, was mit einer Splenomegalie, einer verstärkten Bildung von Keimzentren in lymphatischen Organen, vergrößerten Lymphknoten und einer verlangsamten Involution des Thymus einherging. Weitergehende Untersuchungen der Ursache dieser hyperproliferativen Erkrankung offenbarten eine verminderte klonale Deletion nach Aktivierung. Die Ursachen dieses überraschenden Effekts sind wahrscheinlich vielfältig, da NF-AT-Faktoren an der Regulation der Expression vieler Gene beteiligt sind, u.a. des Apoptose-assoziierten CD95-Liganden. Da sich bezüglich der IL-2-Expression keine Unterschiede zwischen NF-ATp-defizienten Tieren und Kontrollen zeigten, jedoch eine erhöhte IL-2-Konzentration im Medium kultivierter NF-ATp-defizienter T-Zellen beobachtet wurde, wurde die Bindung von NF-ATp an putative NF-AT-Bindungssequenzen des CD25-Promotors, die transkriptionelle Aktivierung des Promotors mittels Luciferase-Assays und die Expression der IL-2R-alpha-Kette (CD25) untersucht. Es konnte gezeigt werden, daß (1.) NF-ATp an zwei Regionen des CD25-Promotors bindet, (2.) der CD25-Promotor durch NF-ATp transkriptionell stark stimuliert wird und (3.) T-Zellen NF-ATp-defizienter Tiere nach Stimulation eine verminderte CD25-Expression zeigen. In NF-ATp-defizienten Tieren war die Expression von CD25 moderat reduziert, was eine Erklärung für den abgeschwächten Phänotyp dieser Tiere - im Vergleich zu IL-2- oder CD25-defizienten Tieren - sein kann. Die hyperproliferativen Erkrankungen dieser verschiedenen Mauslinien weisen auf eine Beteiligung der NF-AT-/IL-2-/IL-2R-Signalwege nicht nur während der T-Zell-Aktivierung hin, sondern auch auf eine Beteiligung an Signalwegen, die zur anschließenden Inaktivierung und Apoptose der T-Lymphozyten nötig sind.
Summary Myelin protein zero (P0) is a key myelin component in maintaining the integrity and functionality of the peripheral nervous system. Mutated variants are the cause for several disabilitating peripheral neuropathies such as Charcot-Marie-Tooth disease or Dejerine –Sotas syndrome. Using P0 knockout mice - a mouse model for these diseases - together with their wt counterparts on C57BL/6 background we studied the shaping of the T-cell repertoire specific for P0 in the presence and in the absence of this protein during the ontogeny of T-cells. Our approach was to use a series of overlapping 20-mer peptides covering the entire amino acid sequence of P0. This series of P0 peptides was employed for epitope mapping of the H2-Ab restricted T cell response. Thus, P0 peptide 5 (P0 41-60) in the extracellular domain of P0 was identified as the main immunogenic peptide. The immunogenic peptide containing the core immunodominant determinant in the P0 sequence was employed in studies of tolerance, revealing a highly reactive P0 specific T-cell repertoire in P0 ko mice while in wt mice the high avidity repertoire was inactivated in order to ensure self tolerance. In wild type and heterozygous P0 mice tolerance is not dependent on gene dosage. P0 is a tissue specific antigen whose expression is limited to myelinating Schwann cells. The classical view on tolerance to tissue specific antigens attributed this role to peripheral mechanisms. Driven by the finding that intrathymic expression of tissue-specific antigens is a common occurrence, we confirmed that “promiscuous” expression on thymic stroma holds true also for myelin P0. In addition, using bone marrow chimeras we investigated the capacity of bone marrow derived cells versus nonhematopoietic cells to induce tolerance towards P0. Our findings show that bone marrow derived cells although tolerogenic to some degree are not sufficient to mediate complete tolerance. P0 expression on cells with origin other than bone marrow showed to be sufficient and necessary to induce sound tolerance. We identified one cryptic (P0 peptide 8) and two subdominant epitopes (P0 petides 1, and 3). P0 peptide 8 was reactive in both wt and P0 ko mice. Peptides 1 and 3 were immunogenic in P0 ko but not in wt mice. Several P0 peptides including the immunogenic peptide 5 were involved in direct and adoptive transfer EAN studies. None of them induced clinical signs of EAN. Immunization with P0 peptide 3 did induce inflammation of the peripheral nerves reflected by the infiltration of macrophages and CD3 positive cells. More studies involving highly P0 specific T-cell lines are needed to characterize the P0 induced EAN. Our findings may have direct implications for secondary autoimmunity and inflammation in peripheral nerves developing after correcting the P0 genetic defect by gene therapy in aforementioned diseases.
The neuronal ceroid lipofuscinoses (NCLs) are fatal neurodegenerative disorders in which the visual system is affected in early stages of disease. A typical accompanying feature is neuroinflammation, the pathogenic impact of which is presently unknown. In this study, the role of inflammatory cells in the pathogenesis was investigated in Palmitoyl-protein thioesterase 1-deficient (Ppt1-/-) and Ceroidlipofuscinosis, neuronal 3-deficient (Cln3-/-) mice, models of the infantile and juvenile forms of NCL, respectively. Focusing predominantly on the visual system, an infiltration of CD8+ cytotoxic Tlymphocytes and an activation of microglia/macrophage-like cells was observed early in disease. To analyze the pathogenic impact of lymphocytes, Ppt1-/- mice were crossbred with mice lacking lymphocytes (Rag1-/-) and axonal transport, perturbation and neuronal survival were scored. Lack of lymphocytes led to a significant amelioration of neuronal disease and reconstitution experiments revealed a crucial role of CD8+ cytotoxic T-lymphocytes. Lack of lymphocytes also caused an improved clinical phenotype and extended longevity. To investigate the impact of microglia/macrophage-like cells, Ppt1-/- and Cln3-/- mice were crossbred with mice lacking sialoadhesin (Sn-/-), a monocyte lineage-restricted cell adhesion molecule important for interactions between macrophage-like cells and lymphocytes. Similar to the lack of lymphocytes, absence of sialoadhesin significantly ameliorated the disease in Ppt1-/- and Cln3-/- mice. Taken together, both T-lymphocytes and microglia/macrophage-like cells were identified as pathogenic mediators in two distinct forms of fatal inherited neurodegenerative storage disorders. These studies expand the concept of secondary inflammation as a common pathomechanistic feature in some neurological diseases and provide novel insights that may be crucial for developing treatment strategies for different forms of NCL.
