Ziel der vorliegenden Arbeit war die Funktionalisierung von Titanoberflächen mit dem Glycosaminoglycan Heparin, um bei Kontakt des Werkstoffs mit Blut die Gerinnungskaskade nicht auszulösen und das Material für Stents (Gefäßstützen) im arteriellen System einsetzbar zu machen. Für die Modifizierungen wurden als Modell der oxidierten Titanoberfläche sowohl oxidierte cp-Titanplättchen als auch TiO2-Pulver verwendet. Heparin kam zum Einsatz, da es sowohl die Hämostase (Blutgerinnung) als auch die Proliferation (Überwucherung) mit glatten Muskelzellen unterdrückt und somit eine Restenose (Wiederverengung) des in die verengte Arterie eingebrachten Stents verhindert. Die kovalente Immobilisierung des Wirkstoffs erfolgte über bifunktionale Spacer (Haftvermittlermoleküle). Spacer waren 3-(Trimethoxysilyl)-propylamin (APMS), N-(2-Aminoethyl)-3-aminopropyltrimethoxysilan (Diamino-APMS) und N1-[3-(Trimethoxysilyl)-propyl]diethylen¬triamin (Triamino-APMS). Der qualitative und quantitative Nachweis der Funktionalisierung von TiO2 mit Haftvermittler bzw. Heparin erfolgte durch schwingungsspektroskopische Methoden, komplexometrische Farbreaktionen sowie der Bestimmung des Zetapotentials im Elektrolytkontakt. Durch die Anbindung von APMS, Di- und Triamino-APMS stieg das Zetapotential von ca. -26 mV auf positive Werte zwischen +41 und +45 mV. Ein Absinken des Zetapotentials belegte die erfolgreiche Anbindung von Heparin (Werte zwischen -39 und -37 mV) an die verschiedenen Haftvermittler, ebenso wie das Vorhandensein der symmetrischen SO3-Valenzschwingung bei 1040 cm-1. Der quantitative Nachweis der immobilisierten Aminogruppen über die Ninhydrinreaktion ergab für die TiO2-Pulver Werte zwischen 17-20 NH2/nm2, wobei die dichteste Funktionalisierung mit APMS und die niedrigste mit Triamino-APMS erzielt werden konnte. Alle Werte lagen im Bereich von Multilayern, da ein Monolayer aus ca. 2 3 NH2/nm2 besteht. Die immobilisierte Menge an Heparin war bei Verwendung von APMS am größten (53.3±3.6 ng/cm2) und bei Triamino-APMS am geringsten (32.1±5.7 ng/cm2). Die biologische Wirksamkeit des gebundenen Heparins wurde über das chromogene Substrat ChromozymTH® bestimmt und verblieb bei Anbindung an den Spacer mit der größten Moleküllänge (Triamino-APMS) mit ca. 70% am wirksamsten. Neben der kovalenten Anbindung des Wirkstoffs an Spacer zielte diese Arbeit auf die Entwicklung von organisch modifizierten, porösen SiO2-Wirkstoffdepots (P-MA-PS; Poly-methacryl¬oxy¬propylpolysilsesquioxane) für Heparin ab, die sowohl als Volumenwerkstoffe als auch zur Modifikation von Titan(dioxid)oberflächen anwendbar wären. Die Matrices wurden ausgehend von MAS (Methacryl¬oxypropyl¬trimethoxysilan) über den Sol-Gel Prozeß anorganisch und anschließend über photochemische Polymerisation zusätzlich organisch vernetzt. Die Quantifizierung des Polymerisationsgrads erfolgte über die Signalintensität der methacrylischen C=C-Doppelbindung bei 1635 cm-1 durch Integration einer Gauß-Funktion. Über den Polymerisationsgrad der organischen Matrix zwischen 0-71% konnte die Freisetzungskinetik von Heparin je nach therapeutischer Anforderung eingestellt werden. Es konnte gezeigt werden, daß hohe Wirkstoff-Beladungen und niedrige Polymerisationsgrade mit einer schnelleren Freisetzung des Heparins korrelierten, die aufgrund der Endlichkeit des Wirkstoffs im Depot einer Kinetik 1. Ordnung unterlag. Die kumulativ freigesetzten Wirkstoffmengen verhielten sich hierbei proportional zur Wurzel aus der Freisetzungszeit, was dem Higuchi-Modell zur Wirkstofffreisetzung aus porösen Matrices mit einem rein Diffusions-kontrollierten Mechanismus entsprach. Die durch Hydrolyse bedingte Degradation der anorganischen Matrix, die UV-VIS-spektroskopisch bei λ = 220 nm gemessen wurde, folgte einer Kinetik pseudo-0. Ordnung. Da das freigesetzte Heparin seine biologische Wirksamkeit beibehielt, sind P-MA-PS Matrices interessant für klinische Anwendungen, wie z.B. für die Beschichtung von Gefäßstützen, die im Blutkontakt stehen.
Die Qualitätskontrolle von pharmazeutisch verwendeten Substanzen ist eine Voraussetzung für die sichere Anwendung von Arzneimitteln. Sie ist eng mit der Bestimmung der Verunreinigungen einer Substanz im Rahmen der Reinheitsanalytik verknüpft. Dabei spielt das Europäische Arzneibuch (Ph. Eur.) eine wichtige Rolle. Die in der vorliegenden Arbeit durchgeführten Untersuchungen zur Bestimmung des Verunreinigungsprofils waren auf die Neuerarbeitung oder Überarbeitung der im Ph. Eur. beschriebenen Prüfungen auf „Verwandte Substanzen“ ausgerichtet. Im Rahmen der Neuerarbeitung einer Monographie für Trimebutin und Trimebutin-Maleat wurde eine RP-HPLC-Methode entwickelt. Neben den bekannten Verunreinigungen war ein weiterer Peak nachweisbar, der N-Desmethyltrimebutin zugeordnet wurde. Die Trennung zwischen N-Desmethyltrimebutin und dem Hauptpeak sowie zwischen zwei bekannten Verunreinigungen ist kritisch. Für beide Peakpaare wurden Systemeignungskriterien festgelegt. Zur Definition von Akzeptanzkriterien in den erarbeiteten Monographieentwürfen wurden Chargen untersucht. Die Quantifizierung erfolgte über die „Externer-Standard“-Methode mit einer Verdünnung der Untersuchungslösung als Referenz. Falls erforderlich, wurden die Korrekturfaktoren in die Berechnung des Gehaltes einbezogen. Die DC-Methode zur Prüfung auf „Verwandte Substanzen“ von Adenosin sollte gegen eine HPLC-Methode ausgetauscht werden. In Übereinstimmung mit Ergebnissen des EDQM-Labors haben die Untersuchungen bestätigt, dass mit einer Ionenpaar-HPLC-Methode die Anwesenheit von Inosin, Guanosin, Uridin und Adenin in Adenosin kontrolliert werden kann. Durch die Festlegung einer Auflösung > 1.5 zwischen Adenin und Inosin als Systemeignungskriterium werden nicht geeignete HPLC-Säulen erkannt. Durch die DC-Prüfung des Ph. Eur. können verschiedene Nucleotide nachgewiesen werden. Die vorgeschlagene HPLC-Methode wurde durch Einführung eines Gradienten so geändert, dass Nucleotide, Nucleoside und Adenin gleichzeitig nachgewiesen werden konnten. Die Analyse der Proben bestätigte die Ergebnisse der DC, dass kleine Mengen Adenosin-5’-monophosphat in den Chargen vorhanden waren. Eine CE-Methode zur Reinheitsprüfung von Glutathion wurde entsprechend der Kommentare zum in Pharmeuropa veröffentlichten Monographievorschlag überarbeitet. Die kritischen Trennungen zwischen dem internen Standard und Cystein sowie zwischen L-γ-Glutamyl-L-cystein und dem Hauptpeak, die durch den Systemeignungstest überprüft werden, waren durch den pH-Wert des Trennpuffers kontrollierbar. Glutathion zeigt beispielhaft, dass das Verunreinigungsprofil fermentativ gewonnener Produkte komplex ist und von den Herstellungs- und Aufreinigungsprozessen abhängt. Gleiches gilt für Aminosäuren, die vermehrt biotechnologisch gewonnen werden. In den Aminosäure-Monographien ist zur Reinheitsprüfung eine DC auf „Ninhydrin-positive Substanzen“ vorgeschrieben. Diese Methode ist auf den Nachweis anderer Aminosäuren, die durch unvollständige Abtrennung bei der Extraktion aus Proteinhydrolysaten stammen können, ausgelegt. Die Analyse von Aminosäuren mittels CE nach Derivatisierung mit FMOC-Cl ermöglicht einen sensitiven und selektiven Nachweis anderer Aminosäuren. Durch den Einsatz von CBQCA als Derivatisierungsreagenz können auch Aminozucker und kleine Peptide erfasst werden. Mit beiden Methoden wurde das Verunreinigungsprofil von Histidin, Isoleucin, Phenylalanin und Serin bestimmt. Während in den Phe- und Ser-Proben keine Verunreinigungen erfasst wurden, wurden in den Ile-Proben nach Derivatisierung mit FMOC-Cl andere Aminosäuren gefunden. Alle untersuchten Aminosäuren zeigten nach Derivatisierung mit CBQCA viele Verunreinigungen. Wegen Peaküberlagerungen konnten Aminosäuren mit dem verwendeten Trennpuffer (25 mM Boratpuffer pH 9.20; 25 mM SDS) nicht bestimmt werden. Durch eine Variation des Puffers (20 mM Tetraboratpuffer pH 9.3; 100 mM SDS) war es möglich, die CBQCA-derivatisierten Aminosäuren zu trennen und zuzuordnen. Im Hinblick auf die Anwesenheit anderer Aminosäuren waren die Ergebnisse der beiden Verfahren vergleichbar. Insbesondere wurden in den Ile-, Phe- und Ser-Proben keine basischen Aminosäuren sowie Cystein gefunden. Deren FMOC-Derivate comigrierten mit einem Peak, der nach Strukturaufklärung mittels NMR-Spektroskopie als Nebenprodukt der Derivatisierungsreaktion mit FMOC-Cl identifiziert wurde. Als Verunreinigungen von biotechnologisch hergestellten Aminosäuren kommen organische Säuren in Frage. Durch eine RP-HPLC unter Zusatz von Heptafluorbuttersäure als Ionenpaarreagenz wurden Aminosäuren und organische Säuren getrennt. Die Detektion erfolgte mittels ELSD. Nach dem Hauptpeak wurden Störsignale detektiert, weshalb sich die Anwendung der Methode zur Reinheitsprüfung als schwierig herausstellte. Die durchgeführten Experimente deuten darauf hin, dass der ELSD mit der großen Substanzmenge überlastet ist und es zur Verschleppung von Substanz kommt.
Körperliche Aktivität ist ein wichtiges Element im Leben von Patienten mit Cystischer Fibrose. Die körperliche Fitness gilt zur Zeit als einer der besten Prädiktoren für die Langzeit-Mortalität bei Patienten mit CF. Bisherige Studien führten ausschließlich überwachtes Training durch. Ziel dieser Arbeit war es zu überprüfen, ob ein unüberwachtes regelmäßiges körperliches Heimtraining bei Mukoviszidosepatienten über 6 Monate durchführbar ist und eine Verbesserung der körperlichen Leistungsfähigkeit ermöglicht. Es wurden 48 Patienten in Trainingsgruppe (n=23) und Kontrollgruppe (n=25) randomisiert. Die Untersuchungen ergaben im Verlauf eine signifikante Verbesserung der maximalen Sauerstoffaufnahme (VO2 max;p=0.0017) und der maximalen Leistungsfähigkeit (W:p=0.0293) bei der Trainingsgruppe im Vergleich zur Kontrollgruppe. Es wurde nachgewiesen, dass unüberwachtes körperliches Training bei CF-Patienten einen Leistungszuwachs bringt und damit Konsequenzen für ihre Lebensqualität und Lebenserwartung hat.
In order to test the effects of environmental factors on different characteristics of plant leaf waxes, barley plants (Hordeum vulgare) were abiotically stress treated (exposure to darkness, heavy metal, high salt concentrations and drought), and biotically stressed by the infection with powdery mildew (Blumeria graminis f.sp. hordei; Bgh). Different wax parameters like amount, chemical composition, and micromorphology of epicuticular wax crystals, were investigated. Etiolated leaves of barley showed distinctly reduced wax amounts and modifications in their relative composition. The alterations of these wax parameters might be a result of a developmental delay, which could have been caused by a decreased availability of energy for cellular processes, due to lack of light. Cadmium exposure led to a 1.5-fold increase of wax amount, while chemical composition was unaffected. In drought- and salt-stressed plants, all investigated leaf wax parameters remained unaltered. In each of the abiotic treatments, the microstructure of epicuticular wax crystals, formed as typical platelets, was not modified. Even after 6d infection with powdery mildew (Bgh), neither locally nor systemically enforced modifications of wax features were revealed.
The analyzed leave surfaces, resulting from these four abiotic and the biotic treatment (phenotypic approach), were compared to altered leaf surfaces’ characteristics of 18 analyzed eceriferum (cer-) wax mutants (genotypic approach). Within the screening, 5 mutants were selected which distinctly differed from the wild-type in wax amount, portions of epi- and intracuticular wax fraction, relative chemical composition, crystal morphology, and surface wettability (hydrophobicity).
Apart from quantitative and qualitative effects on the leaf waxes, environmentally enforced modifications in cuticular waxes might be reflected in molecular processes of wax biogenesis. Therefore, a barley wax-microarray was established. 254 genes were selected, which are putatively involved in processes of de novo fatty acid biosynthesis, fatty acid elongation, and modification, and which are supposed to take part in lipid-trafficking between cell compartments, and transport of wax components to the outer cell surface. The regulations within the expression pattern evoked by the respective treatments were correlated with the corresponding analytical wax data, and the observed molecular effects of a 3d powdery mildew infection were compared with succeeding fungal morphogenesis. Etiolation and cadmium exposition pointed to transcriptional modifications in the de novo fatty acid synthesis, and in the screened, transport-related mechanisms, which correlate with respective alterations in surface wax characteristics. Moderate changes in the gene expression pattern, evoked by drought- and salinity-stress, might give hints for evolved adaptations in barley to such common habitat stresses. Theinvasion of powdery mildew into the epidermal host cells was reflected in the regulation of several genes. Beside other functions, these genes take part in pathogen defense, and intracellular component transport, or they encode transcription factors. The different modifications within the molecular responses evoked by the investigated abiotic treatments, and the effects of powdery mildew infection representing a biotic stressor, were compared between the different treatments.
In order to test the potential impact of different wax parameters on Bgh, conidia germination and differentiation was comparably investigated on leaf surfaces of abiotically stressed wild-type and cer-mutants, isolated cuticles, and further artificial surfaces. The rates of conidial development were similar on each of the leaf surfaces resulting from the abiotic treatments, while a significant reduction of the germination and differentiation success was revealed for the wax mutant cer-yp.949. Compared to the wild-type, developmental rates on isolated cuticles and extracted leaf waxes of the mutant cer-yp.949 indicated a modified embedding of cuticular waxes, and a possibly changed three-dimensional structure of the cer-yp.949 cuticle, which might explain the reduced conidial developmental rates on leaf surfaces of this particular mutant.
Experiments with Bgh conidia on mechanically de-waxed leaf surfaces (selective mechanical removal of the epicuticular leaf waxes with glue-like gum arabic, followed by an extraction of the intracuticular wax portion with chloroform) demonstrated the importance of the wax coverage for the germination and differentiation of the fungal conidia. On all dewaxed leaf surfaces, except those of cer-yp.949, the differentiation success of the germlings was significantly reduced, by about 20% (“wax-effect”). This result was verified through an artificial system with increased conidia developmental rates on glass slides covered with extracted leaf waxes. Further comparative tests with the major components of barley leaf wax, hexacosanol and hexacosanal, showed that the germination and differentiation of powdery mildew conidia not only depends on the different chemistry, but is also influenced by the respective surface hydrophobicity. Compared to hexacosanol, on hexacosanal coated glass surfaces, higher germination and differentiation rates were achieved, which correlated with increased levels of surface hydrophobicity. Developmental rates of conidia on hydrophobic foils demonstrated that hydrophobicity, as a sole surface factor, may stimulate the conidial germination and differentiation processes. Moreover, the survival of conidia on artificial surfaces is determined by additional surface derived factors, e.g. the availability of water, and a pervadable matrix.
Funktionelle Bildgebung der Lunge und des Bronchialkarzinoms mittels Magnetresonanztomographie

Ziel dieser Arbeit war es, die Magnetresonanztomographie (MRT) an der Lunge als Alternative zur traditionellen Lungenbildgebung voranzutreiben. So sollten MRT-Verfahren zur regionalen und quantitativen Lungenfunktionsprüfung für die klinische Routine entwickelt werden. Im Hinblick auf die Strahlentherapie von Patienten mit Bronchialkarzinom sollen funktionelle Lungenareale erkannt werden, um diese während der Bestrahlung optimal schonen zu können. An den zahlreichen Luft-Gewebe-Grenzflächen in der Lunge entstehen Magnetfeldinhomogenitäten. Daraus resultiert ein schneller Zerfall des MRT-Signals in der Lunge. Es wurde in dieser Arbeit ein Ansatz aufgezeigt, um die Ursache für den raschen Signalzerfall, nämlich die unterschiedlichen magnetischen Suszeptibilitäten von Lufträumen und Lungengewebe, zu beseitigen. Durch die intravaskuläre Injektion von paramagnetischen Kontrastmitteln kann die Suszeptibilität des Blutes an die Suszeptibilität der Lufträume angeglichen werden. Durch die Entwicklung einer MR-kompatiblen aktiven Atemkontrolle (MR-ABC) wurde in dieser Arbeit ein weiteres fundamentales Problem der Lungen-MRT adressiert: Die Bewegung während der Datenakquisition. Die MR-ABC detektiert Herzschlag und Atemposition und ist in der Lage die Atembewegung in jeder beliebigen Atemphase reproduzierbar für eine definierte Zeit auszusetzen. Dies wird durch einen Verschluss der Atemluftzufuhr realisiert. Traditionelle Verfahren können zwar ebenfalls die Atemphase detektieren, gestatten jedoch nicht deren Konservierung. Es wurde demonstriert, dass mit der MR-ABC hochauflösende Bilder der Lunge in hoher Bildqualität und durch die Verwendung langer Akquisitionsfenster in relativ kurzer Messzeit erreicht werden können. Eine regionale Lungenfunktionsprüfung ist für die Diagnose und Evaluierung vieler Krankheitsbilder vorteilhaft. In diesem Sinne wird seit einigen Jahren das Potential der Sauerstoff-verstärkten Lungen-MRT erforscht, die auf den paramagnetischen Eigenschaften des molekularen Sauerstoffs basiert. Im Blut gelöster Sauerstoff führt zu einer Verkürzung der T1-Relaxationszeit. Statt diese T1-Verkürzung quantitativ zu bestimmen wird aus praktischen Gründen meist ein T1-gewichteter Ansatz gewählt. In dieser Arbeit wurde jedoch gezeigt, dass nicht-quantitative Verfahren ein erhebliches Risiko zur Falschinterpretation beinhalten. Um Fehldiagnosen zu vermeiden, sollten deshalb prinzipiell quantitative Methoden zur Messung der durch die Sauerstoff-Verstärkung bedingten T1-Verkürzung in der Lunge verwendet werden. Herkömmliche Techniken zur quantitativen T1-Messung benötigen allerdings längere Messzeiten. Deshalb war zur Vermeidung von Bewegungsartefakten bisher die Datenaufnahme im Atemanhaltezustand notwendig. Wiederholtes Atemanhalten von mehreren Sekunden Dauer ist allerdings für einige Patienten sehr belastend. Aus diesem Grund wurden in dieser Arbeit zwei Methoden entwickelt, die eine quantitative Lungenfunktionsprüfung mittels MRT bei freier Atmung der Patienten ermöglichen. Eine gute Sauerstoffversorgung des Tumors wirkt sich positiv auf den Erfolg der Bestrahlung aus. Ein Ansatz zur Verbesserung der Strahlentherapie des Bronchialkarzinoms könnte daher in der Beatmung der Patienten mit hyperoxischen hypercapnischen Atemgasen während der Bestrahlung bestehen. In diesem Zusammenhang könnte die quantitative Messung der T1-Veränderung im Tumor nach Carbogenatmung ein Selektionskriterium darstellen, um diejenigen Patienten zu identifizieren, die von einer Carbogenbeatmung während der Bestrahlung profitieren können. Die Differenzierung zwischen vitalem Tumorgewebe, Nekrosen und atelektatischem Lungengewebe ist von großer Bedeutung bei der Bestrahlungsplanung des Bronchialkarzinoms. Einen neuen Ansatz bildet die in dieser Arbeit vorgestellte Magnetiserungstransfer-MRT. Um einen Magnetisierungstransfer zu erzeugen, wurde ein speziell auf die Bildgebung an der Lunge optimiertes Präparationsmodul entworfen. In Verbindung mit einer schnellen Bildakquisitionstechnik konnte die Magnetisierungstransfer-Lungenbildgebung in einem kurzen Atemstopp durchgeführt werden. Diese Technik wurde an mehreren Patienten mit Bronchialkarzinom evaluiert und die Ergebnisse mit denen der Fluor-Deoxyglykose-Positronen-Emissions-Tomographie (FDG-PET) verglichen. Es wurde festgestellt, dass mit diesem MRT-Verfahren ähnliche diagnostische Erkenntnisse erzielt werden können. Allerdings besitzt die MRT Vorteile im Hinblick auf räumliche Auflösung, Messzeit, Bildqualität, Kosten und Strahlenbelastung. Das erhebliche Potential für die Bestrahlungsplanung des Bronchialkarzinoms durch eine Magnetisierungstransfer-Bildgebung wurde damit nachgewiesen.
Die proximale spinale Muskelatrophie (SMA) stellt eine der häufigsten erblichen Ursachen für den Tod im Kindesalter dar. Die Patienten leiden unter symmetrischer, langsam progredienter Muskelschwäche und in schweren Fällen auch an sensiblen Ausfällen. Die neurodegenerative Erkrankung wird autosomal-rezessiv durch Deletion bzw. Mutationen des SMN1-Gens (survival motor neuron 1-Gens) auf Chromosom 5q13 vererbt. Das SMN-Protein wird ubiquitär exprimiert und findet sich in allen untersuchten Geweben in einem Multiproteinkomplex, dem sogenannten SMN-Komplex, der die Zusammenlagerung von spleißosomalen Komplexen koordiniert. Die Funktion solcher Komplexe ist für alle Zelltypen essentiell. Deshalb stellt sich die Frage, welcher Pathomechanismus für die Erkrankung SMA verantwortlich ist. Die vorliegende Arbeit zeigt, dass die Überlebensraten der Smn–/–;SMN2-Motoneurone 14 Tage alter Mausembryonen gegenüber Smn+/+;SMN2-Motoneuronen (Kontrollen) nicht reduziert waren. Bei der morphologischen Untersuchung der Zellen zum gleichen Entwicklungszeitpunkt zeigten sich jedoch deutliche Unterschiede. Die Axonlängen der Smn-defizienten Motoneurone waren gegenüber Kontrollen signifikant verringert. Das Dendritenwachstum war nicht beeinträchtigt. Die Untersuchung der Wachstumskegel ergab bei den Smn–/–;SMN2 Motoneuronen eine signifikante Verminderung der Fläche gegenüber Kontrollen. Weiterhin zeigten sich Defekte im Zytoskelett. In den Motoneuronen von Kontrolltieren fand sich eine Anreicherung von beta-Aktin in perinukleären Kompartimenten sowie besonders stark in den Wachstumskegeln. Die beta-Aktin-Anreicherung nahm im Verlauf des Axons zu. In Smn–/–;SMN2-Motoneuronen war keine Anreicherung im distalen Axon oder in den Wachstumskegeln detektierbar. Eine gleichartige Verteilungsstörung fand sich für das SMN-Interaktionsprotein hnRNP R (heterogenous nuclear ribonucleoprotein R) und, wie andere Arbeiten zeigen konnten, auch für die beta-Aktin-mRNA, die spezifisch an hnRNP R bindet. In gleicher Weise wurden auch Veränderungen in den sensorischen Neuronen aus den Hinterwurzelganglien 14 Tage alter Mausembryonen untersucht. Bei Smn–/–;SMN2-Mäusen war die Neuritenlänge sensorischer Neurone im Vergleich zur Kontrolle gering, jedoch signifikant verkürzt und die Fläche der Wachstumskegel hochsignifikant verringert. Im Smn–/–;SMN2 Mausmodell für eine schwere Form der SMA fanden sich in den sensorischen Nervenzellen im Vergleich zu den Motoneuronen geringer ausgeprägte, jedoch gleichartige Veränderungen, was auf einen ähnlichen Pathomechanismus in beiden Zelltypen hinweist.
Encoding Redundancy for Task-dependent Optimal Control : A Neural Network Model of Human Reaching

The human motor system is adaptive in two senses. It adapts to the properties of the body to enable effective control. It also adapts to different situational requirements and constraints. This thesis proposes a new neural network model of both kinds of adaptivity for the motor cortical control of human reaching movements, called SURE_REACH (sensorimotor unsupervised learning redundancy resolving control architecture). In this neural network approach, the kinematic and sensorimotor redundancy of a three-joint planar arm is encoded in task-independent internal models by an unsupervised learning scheme. Before a movement is executed, the neural networks prepare a movement plan from the task-independent internal models, which flexibly incorporates external, task-specific constraints. The movement plan is then implemented by proprioceptive or visual closed-loop control. This structure enables SURE_REACH to reach hand targets while incorporating task-specific contraints, for example adhering to kinematic constraints, anticipating the demands of subsequent movements, avoiding obstacles, or reducing the motion of impaired joints. Besides this functionality, the model accounts for temporal aspects of human reaching movements or for data from priming experiments. Additionally, the neural network structure reflects properties of motor cortical networks like interdependent population encoded body space representations, recurrent connectivity, or associative learning schemes. This thesis introduces and describes the new model, relates it to current computational models, evaluates its functionality, relates it to human behavior and neurophysiology, and finally discusses potential extensions as well as the validity of the model. In conclusion, the proposed model grounds highly flexible task-dependent behavior in a neural network framework and unsupervised sensorimotor learning.
Vertebrate and invertebrate visual systems exhibit similarities in early stages of visual processing. For instance, in the human brain, the modalities of color, form and motion are separately processed in parallel neuronal pathways. This basic property is also found in the fly Drosophila melanogaster which has a similar division in color- sensitive and (color blind) motion-sensitive pathways that are determined by two distinct subsets of photoreceptors (the R1-6 and the R7/8 system, respectively). Flies have a highly organized visual system that is characterized by its repetitive, retinotopic organization of four neuropils: the lamina, the medulla, the lobula and the lobula plate. Each of these consists of columns which contain the same set of neurons. In the lamina, axon bundles of six photoreceptors R1-6 that are directed towards the same point in space form columnar structures called cartridges. These are the visual sampling units and are associated with four types of first-order interneuron that receive common input from R1-6: L1, L2, L3 and the amacrine cells (amc, together with their postsynaptic partner T1). They constitute parallel pathways that have been studied in detail at the anatomical level. Little is known, however, about their functional role in processing behaviorally relevant information, e.g. for gaze stabilization, visual course control or the fixation of objects. The availability of a variety of neurogenetic tools for structure-function analysis in Drosophila allowed first steps into the genetic dissection of the neuronal circuitry mediating motion and position detection. In this respect, the choice of the effector turned out to be crucial. Surprisingly, it was found that the clostridial tetanus neurotoxin failed to block mature Drosophila photoreceptor synapses, but caused irreversible damage when expressed during their development. Therefore, the dominant-negative shibire allele shits1 which turned out to be better suited was used for blocking lamina interneurons and thereby analyzing the necessity of the respective pathways. To determine whether the latter were also sufficient for the same behavioral task, the inverse strategy was developed, based on the fact that lamina interneurons express histamine receptors encoded by the ort gene. The specific rescue of ort function in defined channels in an otherwise mutant background allowed studying their sufficiency in a given task. Combining these neurogenetic methods with the optomotor response and object induced orientation behavior as behavioral measures, the aim of the present thesis was to answer the following questions: (a) Which pathways feed into elementary motion detectors and which ones are necessary and/or sufficient for the detection of directional motion? (b) Do pathways exist which specifically mediate responses to unidirectional motion? (c) Which pathways are necessary and/or sufficient for object induced orientation behavior? Some basic properties of the visual circuitry were revealed: The two central cartridge pathways, represented by the large monopolar cells L1 and L2, are key players in motion detection. Under a broad range of stimulatory conditions, the two subsystems are redundant and are able to process motion independently of each other. To detect an impairment when only one of the pathways is intact, one has to drive the system to its operational limits. At low signal to noise ratios, i.e. at low pattern contrast or low background illumination, the L2 pathway has a higher sensitivity. At intermediate pattern contrast, both pathways are specialized in mediating responses to unidirectional motion of opposite stimulus direction. In contrast, neither the L3, nor the amc/T1 pathway is necessary or sufficient for motion detection. While the former may provide position information for orientation, the latter has a modulatory role at intermediate pattern contrast. Orientation behavior turned out to be even more robust than motion vision and may utilize a less sophisticated mechanism, as it does not require a nonlinear comparison of signals from neighboring visual sampling units. The position of objects is processed in several redundant pathways, involving both receptor subsystems. The fixation of objects does not generally require motion vision. However, motion detection improves the fixation of landmarks, especially when these are narrow or have a reduced contrast.
The goal of the project was to establish knock down of mRNA in human mesenchymal stem cells. Since these cells are difficult to transfect, a viral approach is needed to achieve sufficient expression of e. g. shRNA in a high percentage of cells to allow for an efficient silencing of corresponding mRNAs. For this purpose for every gene product of interest, a number of shRNA clones have to be tested to detect an individual shRNA with sufficient efficacy. Lentiviral systems for shRNA approaches have recently become available. The principal advantage of the lentiviral system is that it allows gene silencing in nondividing cells and therefore expands the usefulness of the RNAi-based gene silencing system. Lentivirus-delivered shRNAs are capable of specific, highly stable and functional silencing of gene expression in a variety of cell types. Since the viral transfection of MSCs is a time consuming process that involves transfection of 293 FT cells plus transduction of target cells, for this thesis the following approach was chosen: genes of interest were checked for expression in 293FT cells by RT-PCR. These gene products can be silenced in 293FT cells simply by transfection of shRNA clones and efficacy was subsequently tested by RT-PCR. Beyond this thesis then the project can proceed with effective clones to transduce primary MSCs with individual shRNA clones identified as effective silencing tool in this thesis.
Die Verwendbarkeit des Partizips I als Prädikativ soll laut der einschlägigen Literatur sehr restringiert sein. Die Restriktionen werden aber teilweise sehr unterschiedlich aufgefasst. Dieser Beitrag setzt sich zum Ziel, die Stichhaltigkeit verschiedener semantischer, morphologischer und syntaktischer Kriterien zu überprüfen und die Unzulänglichkeit der bisherigen Regelformulierungen aufzuzeigen.
