In contrast to c-Myc, a deregulated expression of the MYCN gene is restricted to human neuroendocrine tumours. In most cases, the excessive activity of N-Myc results from a MYCN amplification. In neuroblastoma, amplification of MYCN is a predictor of poor prognosis and resistance to therapy. The inability to target the N-Myc protein directly necessitates the search for alternative targets. This project aimed at identifying genes specifically required for growth and survival of cells that express high levels of N-Myc using high-throughput shRNA screening combined with next generation sequencing. The identification and analysis of these genes will shed light on functional interaction partners of N-Myc.
We screened a shRNA library containing 18,327 shRNAs and identified 148 shRNAs, which were selectively depleted in the presence of active N-Myc. In addition, shRNAs targeting genes that are involved in p53 and ARF turnover and apoptosis were depleted in the cell population during the screen. These processes are known to affect N-Myc-mediated apoptosis. Consequently, these results biologically validated the screen. The 148 shRNAs that showed a significant synthetic lethal interaction with high levels of N-Myc expression were further analysed using the bioinformatics program DAVID. We found an enrichment of shRNAs that target genes involved in specific biological processes. For example, we validated synthetic lethal interactions for genes such as, THOC1, NUP153 and LARP7, which play an important role in the process of RNA polymerase II-mediated transcription elongation. We also validated genes that are involved in the neddylation pathway.
In the screen we identified Cullin 3, which is a component of the BTB-CUL3-Rbx1 ubiquitin ligase that is involved in the turnover of Cyclin E. Depletion of cullin 3 and activation of N-Myc was found to synergistically increase Cyclin E expression to supraphysiological levels, inducing S-phase arrest and a strong DNA damage response.
Together with results from a proteomics analysis of N-Myc associated proteins, our results lead us to the following hypothesis: In a neuroblastoma cell, the high levels of N-Myc result in a conflict between RNA polymerase II and the replication machinery during S-phase. The newly identified interaction partners of N- Myc are required to solve this conflict. Consequently, loss of the interaction leads to a massive DNA damage and the induction of apoptosis. In addition, inhibition or depletion of the essential components of the neddylation pathway also results in an unresolvable problem during S-phase.
In this study, we have analysed the apoptotic effects of the ubiquitous environmental toxin benzo[ a] pyrene (BP) in HaCaT cells and human keratinocytes. Although prolonged exposure to BP was not cytotoxic on its own, a strong enhancement of CD95 (Fas)-mediated apoptosis was observed with BP at concentrations activating the aryl hydrocarbon receptor (AhR). Importantly, the ultimately mutagenic BP-metabolite, that is, (+)-anti-BP-7,8-diol-9,10-epoxide (BPDE), failed to enhance CD95-mediated cell death, suggesting that the observed pro-apoptotic effect of BP is neither associated with DNA adducts nor DNA-damage related signalling. CD95-induced apoptosis was also enhanced by beta-naphtoflavone, a well-known agonist of the AhR that does not induce DNA damage, thus suggesting a crucial role for AhR activation. Consistently, BP failed to sensitise for CD95L-induced apoptosis in AhR knockdown HaCaT cells. Furthermore, inhibition of CYP1A1 and/or 1B1 expression did not affect the pro-apoptotic crosstalk. Exposure to BP did not increase expression of CD95, but led to augmented activation of caspase-8. Enhancement of apoptosis was also observed with the TRAIL death receptors that activate caspase-8 and apoptosis by similar mechanisms as CD95. Together, these observations indicate an interference of AhR signalling with the activity of receptor-associated signalling intermediates that are shared by CD95 and TRAIL receptors. Our data thus suggest that AhR agonists can enhance cytokine-mediated adversity upon dermal exposure.
In der vorliegenden Arbeit konnte die essentielle Beteiligung von Caspasen im Zelltodmodell der AKR 2B-Mausfibroblasten nachgewiesen und ihre Aktivitäten charakterisiert werden. AKR 2B-Mausfibroblasten stellen eine subklonierte und gut charakterisierte Zellinie dar, in der durch Entzug des Serums der Zelltod induziert wird. Während des Zelltods sterben innerhalb von 6h etwa 50 Prozent einer dichtearretierten Kultur. Die überlebenden Zellen bleiben von diesem Mangelzustand für mindestens weitere 48h unbeeinflußt, benötigen aber zum Überleben eine Proteinneusynthese. Der Zelltod zeigt für eine Apoptose typische morphologische Veränderungen der Zelle, obwohl apoptotische Charakteristika, wie die oligonukleosomale Fragmentierung der DNA oder die Aufnahme der zerfallenen Zelle durch benachbarte Zellen, ausbleiben. Mittels unterschiedlicher Methoden konnte die Expression von mRNA aller für den apoptotischen Prozeß bekannten relevanten Caspasen in den AKR 2B-Mausfibroblasten nachgewiesen werden. Die Caspasen-1, -2, -3, -6 und -9 liegen in ihrer zymogenen Form konstitutiv in den Zellen vor. Mit Ausnahme der Caspase-9 konnte die durch Serumentzug induzierte Spaltung dieser Caspasen in Untereinheiten und somit ihre Aktivierung nicht detektiert werden. Die wesentliche Beteiligung dieser Cystein-Proteasen wurde jedoch durch den protektiven Effekt spezifischer Inhibitoren und den Nachweis ihrer spezifischen Aktivität bestimmt. Die Charakterisierung dieser enzymatischen Aktivitäten lieferte Hinweise zur Identität der aktivierten Caspasen. Neben einer konstitutiven VEIDase- und IETDase-Aktivität wird 3h nach Entzug des Serums eine DEVDase maximal aktiviert. Das Gemisch an Caspase-Aktivitäten wird durch eine DEVDase dominiert. Diese Aktivität wird zum größten Teil durch nur ein Enzym gestellt, wie durch eine Affinitätsmarkierung und 2D-Gelelektrophorese gezeigt wurde. KM- und Ki-Wert-Bestimmungen der DEVDase deuten darauf hin, daß dieses Enzym typische Effektoreigenschaften, wie die der Caspase-3, besitzt. Daneben werden Lamine während des Zelltods in AKR 2B-Mausfibroblasten abgebaut, was auf eine aktivierte Caspase-6 hinweist. Die enzymatischen Charakteristika dieser Protease weichen aber von den in AKR 2B-Mausfibroblasten festgestellten Werten deutlich ab, so daß man ihr nur eine untergeordnete Rolle im Caspasen-Gemisch zuordnen kann. Eine mehrfach chromatographische Reinigung der Aktivität bietet die beste Grundlage für eine anschließende Sequenzierung der Caspase mit dem Ziel ihrer Identifizierung. Durch die Expression des viralen Caspase-Inhibitors CrmA konnte eine tragende Rolle der Caspase-8 und damit des Rezeptor-vermittelten Weges in der Initiierung des apoptotischen Programms in AKR 2B-Mausfibroblasten ausgeschlossen werden. Gleiches gilt für den mitochondrial-vermittelten Weg, für dessen Beteiligung, bis auf die Spaltung der Caspase-9, keine Hinweise vorliegen. Der Weg, der zur Aktivierung der DEVDase führt, ist Ziel gegenwärtiger Untersuchungen. Substanzen, die Signalwege aktivieren PDGF-BB, TPA, Forskolin und 8Br-cAMP) oder auch Substanzen, deren Verbindung zu Signalwegen noch weitgehend offen ist, schützen die Zellen vor dem Zelltod. Der protektive Effekt dieser Signalwege konzentriert sich in einem Konvergenzpunkt, der auf noch unbekannte Weise die Aktivierung der Effektor-Caspasen blockiert. Die Identität dieses Konvergenzpunktes und von ihm ausgehenden protektiven Weges ist Ziel weiterer Untersuchungen. So ist es möglicherweise dieser Weg, der zum Überleben von 50 Prozent der AKR 2B-Mausfibroblasten während des Serumentzugs wesentlich beiträgt.
Apoptose ist eine bestimmte Art des programmierten Zelltods. Dieser Prozess erfüllt zahlreiche wichtige physiologische Funktionen. Eine pathologische Dysregulation der Apoptose ist an der Entstehung etlicher Krankheiten beteiligt. Bei der Regulation der Apoptose nehmen die Bcl-2-Familienmitglieder und damit auch das anti-apoptotisches Familienmitglied A1 eine wichtige Stellung ein. Die Stabilität und anti-apoptotische Funktion von A1 wird über den Ubiquitin-Proteasomen-Weg reguliert. Hierbei ist das C-terminale Ende von A1 essentiell. Ausgangspunkt für diese Arbeit war die Hypothese, dass an den C-Terminus von A1 eine bisher unbekannte Ubiquitin-E3-Ligase bindet, verzweigte Ubiquitinketten an Lysinreste des A1-Proteins konjugiert und das Protein dadurch für den proteasomalen Abbau markiert. Durch Mutationen einzelner Lysinreste von A1 sollte untersucht werden, welche dieser Aminosäuren ubiquitinyliert werden. Damit sollte der molekulare Mechanismus, der hinter der Instabilität und der anti-apoptotischen Funktion steckt, weiter charakterisiert werden. In dieser Arbeit wurden insgesamt 11 Mutanten des A1-Proteins hergestellt, bei denen die 11 Lysinreste von A1 gruppenweise zu Argininresten (K-A1-Mutanten) ausge-tauscht wurden. Der Austausch von Lysin zu Arginin wurde gewählt, weil hierdurch die Ladung an der entsprechenden Position des Proteins gleich bleibt, während eine Konjugation von Ubiquitin an Arginin nicht möglich ist. Im Ergebnis zeigte sich, dass das A1-Protein nicht nur an einzelnen, ganz spezifischen Lysinen, sondern an allen oder doch zumindest den meisten seiner 11 Lysine ubiquitinyliert werden kann, denn es ergaben sich bei den K-A1-Mutanten keine signifikanten Unterschiede in Stärke oder Muster ihrer Ubiquitinylierung. Es müssen also nicht einige wenige Lysine notwendigerweise ubiquitinyliert werden, um A1 zu destabilisieren, sondern die Ubiquitinylierung ganz unterschiedlicher Lysine markiert das Protein für den proteasomalen Abbau. Auch hat der Verlust bestimmter Lysingruppen und damit potentieller Ubiquitinylierungsstellen keinen signifikanten Einfluss auf die anti-apoptotische Funktion des A1-Proteins. Zusammengefasst unterstützen die Ergebnisse die Hypothese, dass eine bisher nicht bekannte Ubiquitin-E3-Ligase sowohl die Stabilität als auch die anti-apoptotische Funktion von A1 reguliert, indem sie verzweigte Ubiquitinketten an (fast) alle Lysine des Protein anhängt und das Protein damit für den proteasomalen Abbau markiert.
The molecular pathogenesis of thymomas and thymic arcinomas (TCs) is poorly understood and results of adjuvant therapy are unsatisfactory in case of metastatic disease and tumor recurrence. For these clinical settings, novel therapeutic strategies are urgently needed. Recently, limited sequencing efforts revealed that a broad spectrum of genes that play key roles in various common cancers are rarely affected in thymomas and TCs, suggesting that other oncogenic principles might be important.This made us re-analyze historic expression data obtained in a spectrumof thymomas and thymic squamous cell carcinomas (TSCCs) with a custom-made cDNA microarray. By cluster analysis, different anti-apoptotic signatures were detected in type B3 thymoma and TSCC, including overexpression of BIRC3 in TSCCs. This was confirmed by qRT-PCR in the original and an independent validation set of tumors. In contrast to several other cancer cell lines, the BIRC3-positive TSCC cell line, 1889c showed spontaneous apoptosis after BIRC3 knock-down. Targeting apoptosis genes is worth testing as therapeutic principle in TSCC.
Der Tumornekrosefaktor (TNF) entfaltet seine vielfältigen biologischen Aktivitäten durch die Stimulation der beiden TNF-Rezeptoren TNFR1 und TNFR2. Die TNFR1-vermittelte Signaltransduktion ist in vielen Details gut verstanden, wohingegen die TNFR2-vermittelte Signaltransduktion bis heute kaum untersucht ist. Mit Hilfe einer in unserer Gruppe entwickelten hochaktiven TNFR2-spezifischen TNF-Variante sowie einer bereits länger bekannten TNFR1-spezifischen TNF-Variante wurde in dieser Arbeit die TNF-Signaltransduktion insbesondere im Mutiplen Myelom untersucht. Mit Hilfe der beiden TNF-Varianten konnte gezeigt werden, dass die alleinige Stimulation des TNFR2 die Aktivierung des alternativen NFkappaB-Signalweges vermittelt, wohingegen TNFR1 nicht dazu in der Lage ist. So zeigte sich im Einklang mit der inhibitorischen Funktion des Adapterproteins TRAF2 in der Signaltransduktion des alternativen NFkappaB-Signalweges, dass die TNFR2-Stimulation in einer TRAF2-Depletion resultiert. Dies führt weiterhin zur Akkumulation von NIK und der Prozessierung von p100 zu seiner aktiven Form p52, den klassischen biochemisch nachweisbaren Ereignissen der Aktivierung des alternativen NFkappaB-Signalweges. Aufgrund der Rolle des NFkappaB-Systems im Multiplen Myelom (MM) und der stimulierenden Wirkung des TNFR1 und TNFR2 auf das NFkappaB-System wurde die Expression und Funktion dieser beiden Rezeptoren auf Myelomzelllinien untersucht. Insbesondere wurde analysiert, welchen Effekt eine spezifische Stimulation der beiden TNF-Rezeptoren auf die apoptotische Sensitivität von Myelomzellen hat. Mit einer Ausnahme wiesen alle untersuchten Myelomzelllinien eine eindeutige TNFR2-Oberflächenexpression auf, die TNFR1-Expression hingegen war heterogen. Die TNFR1-Stimulation in den TNFR1-positiven Zelllinien zeigte keinen wesentlichen Einfluss auf die Zellviabilität. Allerdings resultierte eine Vorstimulation mit TNF in einer gesteigerten Sensitivität für den CD95L-induzierten Zelltod, schützte aber gleichzeitig vor der TRAIL-vermittelten Induktion der Apoptose. Der gegenläufige Effekt der TNF-Vorstimulation auf den CD95L- und TRAIL-induzierten Zelltod konnte auf die Hochregulation der CD95-Oberflächenexpression und der gesteigerten Expression des antiapoptotischen cFLIPLong-Proteins zurückgeführt werden. Beide Effekte basieren auf der TNF-induzierten Aktivierung des klassischen NFkappaB-Signalweges. Im CD95L-induzierten Zelltod überkompensierte die Induktion der CD95-Expression offensichtlich die Hochregulation von cFLIPLong und resultierte in gesteigertem Zelltod. Der TRAIL-induzierte Zelltod hingegen wurde durch die TNF-Vorstimulation abgeschwächt, da hier lediglich die durch den klassischen NFkappaB-Signalweg vermittelte gesteigerte Expression des antiapoptotischen cFLIPLong eine Rolle spielte. Desweiteren zeigten die Analysen in dieser Arbeit, dass die TNFR2-Stimulation zu einer Depletion von TRAF2 und z. B. in JJN3-Zellen zu einer Sensitivierung für den TNFR1-induzierten Zelltod führte. Die Ergebnisse dieser Arbeit zeigten in der Summe somit, dass das TNF-TNFR-Signaling durch verschiedene Mechanismen Einfluss auf den Ausgang der extrinsischen Apoptoseinduktion hat, und dass der Effekt von TNF auf das Überleben von MM-Zellen kontextabhängig ist.
Sowohl NFAT- (Nukleäre Faktoren aktivierter T-Zellen) als auch MEF2- (’myosin-enhancer factor 2’) Transkriptionsfaktoren spielen eine wichtige Rolle bei der Differenzierung, Proliferation oder Apoptose vieler eukaryotischer Zellen. Sie sind in einer Vielzahl von Zellen aktiv und können dort die Transkription ihrer Zielgene nach Stimulation der Zellen mit anschließendem Calcium-Einstrom aktivieren. Dabei kooperieren sie oft mit anderen Transkriptionsfaktoren. Kurz vor Beginn dieser Arbeit erschienen zwei Veröffentlichungen, die eine Kooperation von MEF2D mit NFATc2 bei der Aktivierung des nur77-Promotors, der bei der negativen Selektion von T-Zellen aktiv ist, beschrieben. Im Rahmen dieser Arbeit sollte nun das Bindungsverhalten verschiedener NFAT-Proteine an den nur77-Promotor und ihre Kooperationsfähigkeit mit MEF2D bei der Aktivierung des nur77- und anderer Promotoren untersucht werden. Es konnte gezeigt werden, dass verschiedene NFAT-Faktoren direkt an ein Oligonukleotid des nur77-Promotors (von Position -274 bis -247 bp reichend) binden. Diese Bindung wird zwar durch eine Bindung von MEF2-Faktoren an den Promotor unterstützt, ist jedoch nicht wesentlich beeinträchtigt, wenn MEF2-Faktoren aufgrund von einer Mutation in ihrer Bindungssequenz nicht binden können. Dagegen führt eine Mutation der NFAT-Bindungsstelle zu einer aufgehobenen Bindung von NFAT-Faktoren an den Promotor. Desweiteren zeigte sich, dass nicht nur endogenes NFATc2 sondern auch verschiedene NFATc1-Isoformen an den Promotor binden können. In ihrer Fähigkeit mit MEF2D zu kooperieren, zeigte sich kein Unterschied zwischen den getesteten NFAT-Isoformen NFATc1/αA, NFATc1/αC oder NFATc2. So aktivierten in Transfektionsversuchen in Jurkat T-Zellen und 293 T-Zellen alle NFAT-Proteine die Luciferase-Reporter-Gen-Konstrukte gemeinsam mit MEF2D in etwa additiver Weise. Dies konnte an verschiedenen Luciferase-Reporter-Genen (nur77-gesteuert, hFasL-gesteuert und desmin-gesteuert) nachgewiesen werden, was auf eine mögliche Kooperation der verschiedenen NFAT-Faktoren mit MEF2D (und evtl. auch anderen MEF2-Faktoren) nicht nur bei der Apoptose von T-Zellen sondern auch in anderen Zellen hinweist.
C60 fullerene as an effective nanoplatform of alkaloid Berberine delivery into leukemic cells

A herbal alkaloid Berberine (Ber), used for centuries in Ayurvedic, Chinese, Middle-Eastern, and native American folk medicines, is nowadays proved to function as a safe anticancer agent. Yet, its poor water solubility, stability, and bioavailability hinder clinical application. In this study, we have explored a nanosized carbon nanoparticle—C60 fullerene (C60)—for optimized Ber delivery into leukemic cells. Water dispersions of noncovalent C60-Ber nanocomplexes in the 1:2, 1:1, and 2:1 molar ratios were prepared. UV–Vis spectroscopy, dynamic light scattering (DLS), and atomic force microscopy (AFM) evidenced a complexation of the Ber cation with the negatively charged C60 molecule. The computer simulation showed that π-stacking dominates in Ber and C\(_{60}\) binding in an aqueous solution. Complexation with C\(_{60}\) was found to promote Ber intracellular uptake. By increasing C\(_{60}\) concentration, the C\(_{60}\)-Ber nanocomplexes exhibited higher antiproliferative potential towards CCRF-CEM cells, in accordance with the following order: free Ber < 1:2 < 1:1 < 2:1 (the most toxic). The activation of caspase 3/7 and accumulation in the sub-G1 phase of CCRF-CEM cells treated with C\(_{60}\)-Ber nanocomplexes evidenced apoptosis induction. Thus, this study indicates that the fast and easy noncovalent complexation of alkaloid Ber with C\(_{60}\) improved its in vitro efficiency against cancer cells.
Targeted cancer therapy concepts often aim at the induction of adjuvant antitumor immunity or stimulation of tumor cell apoptosis. There is further evidence that combined application of immune stimulating and tumor apoptosis-inducing compounds elicits a synergistic antitumor effect. Here, we describe the development and characterization of bifunctional fusion proteins consisting of a single-chain variable fragment (scFv) domain derived from the CD40-specific monoclonal antibody G28-5 that is fused to the N-terminus of stabilized trimeric soluble variants of the death ligand TNF-related apoptosis-inducing ligand (TRAIL). As shown before by us and others for other cell surface antigen-targeted scFv-TRAIL fusion proteins, scFv:G28-TRAIL displayed an enhanced capacity to induce apoptosis upon CD40 binding. Studies with scFv:G28 fusion proteins of TRAIL mutants that discriminate between the two TRAIL death receptors, TRAILR1 and TRAILR2, further revealed that the CD40 binding-dependent mode of apoptosis induction of scFv:G28-TRAIL is operable with each of the two TRAIL death receptors. Binding of scFv:G28-TRAIL fusion proteins to CD40 not only result in enhanced TRAIL death receptor signaling but also in activation of the targeted CD40 molecule. In accordance with the latter, the scFv:G28-TRAIL fusion proteins triggered strong CD40-mediated maturation of dendritic cells. The CD40-targeted TRAIL fusion proteins described in this study therefore represent a novel type of bifunctional fusion proteins that couple stimulation of antigen presenting cells and apoptosis induction.
To combine the CD27 stimulation inhibitory effect of blocking CD70 antibodies with an antibody-dependent cellular cytotoxicity (ADCC)-independent, cell death-inducing activity for targeting of CD70-expressing tumors, we evaluated here fusion proteins of the apoptosis-inducing TNF family member TRAIL and a single-chain variable fragment (scFv) derived from a high-affinity llama-derived anti-human CD70 antibody (lαhCD70). A fusion protein of scFv:lαhCD70 with TNC-TRAIL, a stabilized form of TRAIL, showed strongly enhanced apoptosis induction upon CD70 binding and furthermore efficiently interfered with CD70-CD27 interaction. Noteworthy, introduction of recently identified mutations that discriminate between TRAILR1 and TRAILR2 binding into the TRAIL part of scFv:lαhCD70-TNC-TRAIL resulted in TRAIL death receptor-specific fusion proteins with CD70-restricted activity.
p53 protects us from cancer by transcriptionally regulating tumor suppressive programs designed to either prevent the development or clonal expansion of malignant cells. How p53 selects target genes in the genome in a context-and tissue-specific manner remains largely obscure. There is growing evidence that the ability of p53 to bind DNA in a cooperative manner prominently influences target gene selection with activation of the apoptosis program being completely dependent on DNA binding cooperativity. Here, we used ChIP-seq to comprehensively profile the cistrome of p53 mutants with reduced or increased cooperativity. The analysis highlighted a particular relevance of cooperativity for extending the p53 cistrome to non-canonical binding sequences characterized by deletions, spacer insertions and base mismatches. Furthermore, it revealed a striking functional separation of the cistrome on the basis of cooperativity; with low cooperativity genes being significantly enriched for cell cycle and high cooperativity genes for apoptotic functions. Importantly, expression of high but not low cooperativity genes was correlated with superior survival in breast cancer patients. Interestingly, in contrast to most p53-activated genes, p53-repressed genes did not commonly contain p53 binding elements. Nevertheless, both the degree of gene activation and repression were cooperativity-dependent, suggesting that p53-mediated gene repression is largely indirect and mediated by cooperativity-dependently transactivated gene products such as CDKN1A, E2F7 and non-coding RNAs. Since both activation of apoptosis genes with non-canonical response elements and repression of pro-survival genes are crucial for p53's apoptotic activity, the cistrome analysis comprehensively explains why p53-induced apoptosis, but not cell cycle arrest, strongly depends on the intermolecular cooperation of p53 molecules as a possible safeguard mechanism protecting from accidental cell killing.
In dieser Arbeit wurde das PVM-Mausmodell verwendet, um die Bedeutung der Typ I und Typ III Interferonantwort für die Pathogenese einer pneumoviralen Infektion zu analysieren. Hierzu wurden zunächst mit Hilfe der reversen Genetik rekombinante PVM-Mutanten hergestellt, bei denen die Gene für die NS-Proteine, welche vermutlich als Interferonantagonisten fungieren, deletiert sind. Die Charakterisierung der Replikationsfähigkeit der rPVM dNS-Mutanten erfolgte in vitro in Interferon-kompetenten und Interferon-inkompetenten Zelllinien. Ein zentraler Schritt innerhalb dieser Charakterisierung war die Untersuchung der Induktion von Interferonen in vivo und in vitro nach Infektion mit den rPVM dNSMutanten, wobei nachgewiesen wurde, dass die NS-Proteine von PVM als Interferonantagonisten fungieren. In allen Interferon-kompetenten Zellkulturen wurde eine Attenuierung von rPVM dNS1, rPVM dNS2 und rPVM dNS1dNS2 bezogen auf rPVM beobachtet. In allen Interferon-inkompetenten Zellkulturen konnte die Attenuierung der rPVM dNS-Mutanten nahezu vollständig revertiert werden. Nach Infektion mit den rPVM dNS-Mutanten wurde in verschiedenen Zelllinien eine Induktion von Typ I und Typ III Interferonen betrachtet, wobei Unterschiede in der Stärke der Interferon-Induktion nach Infektion mit den rPVM dNS-Mutanten vorhanden waren. Zusammenfassend war es möglich, die NS1- und NS2-Proteine von PVM in Analogie zu RSV eindeutig als Antagonisten der Interferonantwort zu identifizieren. Die Untersuchung der protektiven Rolle von Typ I und Typ III Interferonen für die Replikation und Pathogenität von PVM bildete den zweiten Teil dieser Arbeit. Hierzu wurde die Replikationsfähigkeit und Pathogenität der rPVM dNS-Mutanten in verschiedenen Interferon-defizienten Mausstämmen getestet. Die Untersuchungen ergaben eine protektive Rolle von Typ I und Typ III Interferonen bei einer Infektion mit PVM, wobei den Typ I Interferonen ein effektiverer Einfluss zugeordnet werden konnte. Ein Vergleich von Replikation und Virulenz zwischen den verschiedenen Typ I oder Typ III oder Typ I/Typ III Interferonrezeptor-defizienten Mausstämmen belegte eine erhöhte Suszeptibilität der Typ I/Typ III Interferonrezeptor-defizienten Mäuse gegenüber einer Infektion mit den rPVM dNS-Mutanten. Eine vollständige Aufhebung der Attenuierung wurde auch in den Typ I/Typ III Interferonrezeptor-defizienten Mäusen nicht erlangt. Eine anti-apoptotische Funktion der NS-Proteine zusätzlich zu ihrer Wirkungsweise als Interferonantagonisten wurde aufgrund der unvollständigen Revertierung der Pathogenität der rPVM dNS-Mutanten in Typ I/Typ III Interferonrezeptor-defizienten Mäusen vermutet. Der abschließende Teil dieser Dissertation befasste sich mit der Frage, welche Zellen bei einer natürlichen pulmonalen Infektion Interferone in vivo produzieren. In vitro wurde beobachtet, dass überraschenderweise nur sehr wenige virusinfizierte oder uninfizierte Zellen Typ I Interferone bilden. Der Nachweis darüber, welche Zellen während einer pulmonalen Infektion hauptsächlich Interferone in vivo produzieren, war aufgrund der fehlenden Eignung der kommerziell erhältlichen Interferon-Antikörper für intrazelluläre Gewebefärbungen nicht möglich. Dennoch gelang es abschließend durch eine neue Nachweismethode erstmals Zellen mit rezeptorgebundenen Interferon zu identifizieren, wobei es sich um ziliierte Epithelzellen, Alveolarmakrophagen und vermutlich Clarazellen sowie Typ I und Typ II Pneumozyten handelte.
Human leishmaniasis covers a broad spectrum of clinical manifestations ranging from self-healing cutaneous leishmaniasis to severe and lethal visceral leishmaniasis caused among other species by Leishmania major or Leishmania donovani, respectively. Some drug candidates are in clinical trials to substitute current therapies, which are facing emerging drug-resistance accompanied with serious side effects. Here, two cinnamic acid bornyl ester derivatives (1 and 2) were assessed for their antileishmanial activity. Good selectivity and antileishmanial activity of bornyl 3-phenylpropanoate (2) in vitro prompted the antileishmanial assessment in vivo. For this purpose, BALB/c mice were infected with Leishmania major promastigotes and treated with three doses of 50 mg/kg/day of compound 2. The treatment prevented the characteristic swelling at the site of infection and correlated with reduced parasite burden. Transmitted light microscopy and transmission electron microscopy of Leishmania major promastigotes revealed that compounds 1 and 2 induce mitochondrial swelling. Subsequent studies on Leishmania major promastigotes showed the loss of mitochondrial transmembrane potential (ΔΨm) as a putative mode of action. As the cinnamic acid bornyl ester derivatives 1 and 2 had exhibited antileishmanial activity in vitro, and compound 2 in Leishmania major-infected BALB/c mice in vivo, they can be regarded as possible lead structures for the development of new antileishmanial therapeutic approaches.

This study has identified cofilin, an actin binding protein, as a control element in the reorganization of the actin cytoskeleton which is highly relevant for T lymphocyte activation. Cofilin is regulated in its activity by reversible phosphorylation which is inducible by stimulation through accessory receptors such as CD2 and CD28. First it could be demonstrated that accessory receptor triggering induces the transient association of cofilin with the actin cytoskeleton and that only the dephosphorylated form of cofilin possesses the capacity to bind cytoskeletal actin in vivo. PI3-kinase inhibitors block both the dephosphorylation of cofilin and its association with the actin cytoskeleton. Importantly, cofilin, actin, PI3-kinase and one of its substrates, namely phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2) which can bind to cofilin, co-localize within CD2-receptor caps. The cofilin/F-actin interaction has been identified as a crucial regulatory element for receptor cap formation and the strength of signal transduction. To this end, appropriately designed cell permeable non-toxic peptides that are homologous to actin binding motifs of the human cofilin sequence were introduced into untransformed human peripheral blood T lymphocytes. These peptides competitively and dose dependently inhibit the activation induced interaction of cofilin with the actin cytoskeleton in vivo. By this approach it was possible to study, for the first time, the functional consequences of this interaction in immunocompetent T cells. The present data demonstrate that inhibition of the actin/cofilin interaction in human T lymphocytes by means of these cofilin derived peptides abolishes receptor cap formation and strongly modulates functional T cell responses such as T cell proliferation, interleukin-2 production, cell surface expression of CD69, gIFN production, and CD95L expression. Importantly, receptor independent activation by PMA and calcium ionophore circumvents these peptide produced inhibitory effects on lymphocyte stimulation and places the cofilin/actin interaction to a proximal step in the cascade of signaling events following T cell activation via surface signals. The present results are novel since as yet no information existed regarding the molecular elements which link cell surface receptor stimulation directly to the resulting reorganization of the actin cytoskeleton.
Volatile anesthetic-induced preconditioning ( APC) has shown to have cardiac and cerebral protective properties in both pre-clinical models and clinical trials. Interestingly, accumulating evidences demonstrate that, except from some specific characters, the underlying molecular mechanisms of APC-induced protective effects in myocytes and neurons are very similar; they share several major intracellular signaling pathways, including mediating mitochondrial function, release of inflammatory cytokines and cell apoptosis. Among all the experimental results, cortical spreading depolarization is a relative newly discovered cellular mechanism of APC, which, however, just exists in central nervous system. Applying volatile anesthetic preconditioning to clinical practice seems to be a promising cardio- and neuroprotective strategy. In this review, we also summarized and discussed the results of recent clinical research of APC. Despite all the positive experimental evidences, large-scale, long-term, more precisely controlled clinical trials focusing on the perioperative use of volatile anesthetics for organ protection are still needed.
Connecting lysosomes and mitochondria – a novel role for lipid metabolism in cancer cell death

Background
The understanding of lysosomes has been expanded in recent research way beyond their view as cellular trash can. Lysosomes are pivotal in regulating metabolism, endocytosis and autophagy and are implicated in cancer. Recently it was discovered that the lysosomal V-ATPase, which is known to induce apoptosis, interferes with lipid metabolism in cancer, yet the interplay between these organelles is poorly understood.
Methods
LC-MS/MS analysis was performed to investigate lipid distribution in cells. Cell survival and signaling pathways were analyzed by means of cell biological methods (qPCR, Western Blot, flow cytometry, CellTiter-Blue). Mitochondrial structure was analyzed by confocal imaging and electron microscopy, their function was determined by flow cytometry and seahorse measurements.
Results
Our data reveal that interfering with lysosomal function changes composition and subcellular localization of triacylglycerids accompanied by an upregulation of PGC1α and PPARα expression, master regulators of energy and lipid metabolism. Furthermore, cardiolipin content is reduced driving mitochondria into fission, accompanied by a loss of membrane potential and reduction in oxidative capacity, which leads to a deregulation in cellular ROS and induction of mitochondria-driven apoptosis. Additionally, cells undergo a metabolic shift to glutamine dependency, correlated with the fission phenotype and sensitivity to lysosomal inhibition, most prominent in Ras mutated cells.
Conclusion
This study sheds mechanistic light on a largely uninvestigated triangle between lysosomes, lipid metabolism and mitochondrial function. Insight into this organelle crosstalk increases our understanding of mitochondria-driven cell death. Our findings furthermore provide a first hint on a connection of Ras pathway mutations and sensitivity towards lysosomal inhibitors.
Die Stimulation von Lymphozyten durch alleinige Quervernetzung ihrer Antigenrezeptoren führt in Abhängigkeit von der Signalstärke zur Induktion von Apoptose. Dieser Prozess spielt im Rahmen der negativen Selektion von B-Zellen eine wichtige Rolle und kann am Beispiel von WEHI 231 Zellen, einer murinen Lymphomzelllinie modellhaft nachempfunden werden. Die Quervernetzung des B-Zellrezeptors (BZR) in WEHI 231 Zellen führt in Abhängigkeit von der Prozessierung der mitochondrialen Caspase 9 zu Apoptose. Dies kann durch Überexpression des antiapoptotischen Bcl-2 Familienmitglieds A1 verhindert werden. Interessanterweise besitzt A1 im Gegensatz zu Fast allen anderen Bcl-2 Proteinen keinen C-terminalen Bereich, der das Protein in intrazelluläre Membranen wie Mitochondrien und Endoplasmatisches Retikulum verankert. Ob und gegebenenfalls welche Bedeutung das C-terminale Ende für das Protein und dessen Funktion hat wurde in dieser Arbeit untersucht. Eine C-terminale Deletionsmutante wies im Vergleich zum Wildtyp eine höhere Proteinstabilität auf. Die Fusion der letzten 35 Aminosäuren von A1 an eine enzymatisch inaktive Form von Caspase 3 führte zu einem sehr instabilen chimären Protein. Somit scheint der C-Terminus von A1 sowohl notwendig als auch hinreichend für die kurze Halbwertszeit des Proteins zu sein. Die meisten kurzlebigen Proteine werden über den proteasomalen Signalweg degradiert. Dies scheint auch für A1 zu gelten, da seine Halbwertszeit durch den Einsatz eines Proteasomeninhibitors verlängert wurde. In Koexpressionsstudien konnte zudem eine Ubiquitylierung von A1 beobachtet werden, welche bei der stabileren A1 Mutante stark reduziert war. A1 ist jedoch nicht immer instabil. In Anwesenheit des „BH3-only“ Proteins Bim, das A1 direkt binden kann, wird die Halbwertszeit von A1 enorm erhöht. Die antiapoptotische Funktion von A1 scheint sich jedoch nicht nur auf die Neutralisation durch bloße Bindung zu beschränken, da die Deletionsmutante trotz vergleichbarer Interaktion mit Bim einen sehr viel geringeren Schutz gegen Etoposid vermittelter Apoptose verlieh. Zusammenfassend zeigen die Ergebnisse, dass das antiapoptotische Bcl-2 Familienmitglied A1 eine sehr kurze Halbwertszeit besitzt und das der C-Terminus nicht nur die Stabilität sondern auch die antiapoptotische Schutzfunktion für das Protein vermittelt.
The present study reports the synthesis of new purine bioisosteres comprising a pyrazolo[3,4-d]pyrimidine scaffold linked to mono-, di-, and trimethoxy benzylidene moieties through hydrazine linkages. First, in silico docking experiments of the synthesized compounds against Bax, Bcl-2, Caspase-3, Ki67, p21, and p53 were performed in a trial to rationalize the observed cytotoxic activity for the tested compounds. The anticancer activity of these compounds was evaluated in vitro against Caco-2, A549, HT1080, and Hela cell lines. Results revealed that two (5 and 7) of the three synthesized compounds (5, 6, and 7) showed high cytotoxic activity against all tested cell lines with IC50 values in the micro molar concentration. Our in vitro results show that there is no significant apoptotic effect for the treatment with the experimental compounds on the viability of cells against A549 cells. Ki67 expression was found to decrease significantly following the treatment of cells with the most promising candidate: drug 7. The overall results indicate that these pyrazolopyrimidine derivatives possess anticancer activity at varying doses. The suggested mechanism of action involves the inhibition of the proliferation of cancer cells.
Die Bedeutung der Expression des Tumornekrosefaktor-alpha bei Patienten mit kolorektalem Karzinom

Für Tumorprogression müssen entartete Zellen Wege finden, die immunologische Abwehr und die Apoptose zu umgehen. Tumorzellen haben dafür verschiedene, sogenannte Tumor-Escape-Mechanismen entwickelt. Gegenstand dieser Arbeit war in diesem Zusammenhang die Untersuchung des TNF-α-TNF-R1-Systems. Eine signifikante erhöhte TNF-α Protein- und Genexpression konnte in Gewebeproben kolorektaler Karzinome nachgewiesen werden, wobei eine mäßig starke Korrelation beider Analysemethoden ersichtlich war (т = 0.794). Sowohl die immunhistochemische Analyse als auch die Genexpression durch RT-PCR konnten mit Tumorprogression assoziiert werden. Mit erhöhter Expression des Apoptose-induzierenden Zytokins TNF-α durch Tumorzellen konnte darüber hinaus ein signifikant schlechteres Gesamtüberleben der Patienten mit KRK beobachtet werden. In unmittelbarer Umgebung TNF-α exprimierender Tumorzellen wurden zahlreiche TNF-R1+/CD8+ Zellen analysiert, die als Hinweis auf Apoptose in TILs angesehen werden können. Dieser Weg könnte als Tumor-Escape-Mechanismus verstanden werden. Die Verwendung potentieller TNF-α Biologicals (z.B. Etanercept, Infliximab) bleibt jedoch unter dem Aspekt einer geringfügig erhöhten Lymphominzidenz gegenüber der Normalbevölkerung auch als kritisch zu bewerten. Der Einsatz von Anti-TNF-α-Therapien stellt jedoch eine vielversprechende Option bei Patienten mit metastasiertem KRK und Tumorrezidiv dar. Zusammenfassend liefern die Ergebnisse dieser Arbeit zusätzlichen Einblick in die Regulationsmechanismen, die verantwortlich für die Immunsuppression durch kolorektale Karzinome sein können. Diese basiswissenschaftlichen Erkenntnisse stellen eine mögliche Grundlage neuer Behandlungsmöglichkeiten kolorektaler Karzinomen dar, die weiter erforscht werden sollten.
Die Todesrezeptoren der TNF-Familie sind neben der Vermittlung von Apoptosesignalen auch in der Lage, nicht-apoptotische intrazelluläre Signalwege zu beeinflussen. Der Caspase-8-Inhibitor cFLIPlong inhibiert dosisabhängig die Prozessierung der Initiator-Caspase-8 am TRAIL-DISC (death inducing signalling complex) und hemmt die Aktivierung des NF-kappa-B-Signalweges über die Modulation der Rekrutierung und Spaltung des für die NF-kappa-B-Aktivierung notwendigen RIP (receptor interactin protein)am DISC.
