Der Pathomechanismus des T-Helferzellverlusts bei der HIV-Infektion ist bisher ungeklärt. Diese Arbeit zeigt, dass primäre, HIV-uninfizierte CD4+ Zellen in Kontakt mit HIV-infizierten Zellen zugrundegehen. Zur Aufklärung des Mechanismus dieses Zelltodes wurden einzelne Schritte der Zellfusion zwischen infizierten und uninfizierten Zellen mit Antikörpern und Peptiden blockiert, und die Auswirkung dieser Blockade auf den Zelltod bestimmt. Dabei zeigte sich dass die Blockade jedes Schritts der Fusion von nicht-infizierten und infizierten Zellen den Zelltod der uninfizierten Zellen verhindert. Weiter wurde gezeigt, dass im Verlauf des Zelluntergangs Apoptose auftritt, diese aber für den Zellverlust keine notwendige Voraussetzung ist.
In der vorliegenden Arbeit wurden die in-vivo-Effekte eines „superagonistischen“ mAks mit Spezifität für den kostimulierenden Rezeptor CD28 der Ratte untersucht. Dieser Antikörper unterscheidet sich von konventionellen CD28-spezifischen mAk durch seine Fähigkeit, T Zellen auch ohne Ligation des TZR und damit polyklonal zu aktivieren. Die so ausgelöste Expansion der T Zellen verläuft so schnell und effizient wie eine durch Antigen gesteuerte Proliferation; die überproportionale Vermehrung anti-inflammatorischer regulatorischer T Zellen während der initialen Expansionsphase ist vermutlich für das Ausbleiben toxischer Effekte im Zuge der polyklonalen TZellvermehrung verantwortlich.
The transcriptional repressor-Blimp-1 terminates differentiation of B lymphocytes as well as myeloid cells. Our data show that Blimp-1 is highly expressed in freshly isolated murine primary T lymphocytes, particularly its minor splice variant. Ectopic expression of Blimp-1 by retroviral transduction neither dramatically altered secretion of IFN-ã or IL-4 nor did it induce the ability to suppress as regulatory T cells. However, induction of Blimp-1 resulted in not only a significant reduction in the production of IL-2 but also an inability to proliferate as well as in the reduced viability. These results demonstrate that Blimp-1 might mark end stages of lineage differentiation in T cells.
In dieser Arbeit wurde die Zytolyse, die durch HIV-spezifische zytotoxische T-Lymphozyten (CTL) bewirkt wird, untersucht. Grundsätzlich erscheinen in der Abwehr viraler Infekte sowohl CD4+ wie CD8+ CTL. In chronisch HIV-Infizierten sind in der Regel lediglich CD8+ CTL zu finden. HIV-spezifische CD4+ CTL können aber zum Beispiel in Impfstudien aus Probandenblut isoliert werden. Sie erwiesen sich als zytolytisch aktiv und waren außerdem, wie in dieser Arbeit erstmals gezeigt werden konnte, auch an einer Fusions-vermittelten Lyse beteiligt. Diese ist Kalzium-unabhängig, auch mit heterologen Zellen möglich und die Expression des HIV-gp120 an der Zelloberfläche der Zielzellen ist essentiell, während die Präsentation von gp120 Epitopen nicht ausreicht. Die Konsequenz dieser Lyseform ist die apoptotische Zerstörung aller beteiligten Zellen. CD4+ HIV-spezifische CTL können über diesen Lysemechanismus bereits in der akuten Phase der Infektion einer negativen Selektion unterliegen, was ein Grund für das Fehlen der HIV-spezifischen CD4+ CTL in chronisch Infizierten darstellen kann. HIV-spezifische CD8+ CTL konnten aus einem HIV-positiven Patienten isoliert werden. Insgesamt wurden fünf Klone näher charakterisiert. Zwei erkannten jeweils ein Epitop aus unterschiedlichen Bereichen des extrazellulären Anteils des Glykoproteins, einer einen Bereich des p17-Anteils und zwei dasselbe Epitop im p24-Anteil des gag-Proteins. Dieses Epitop, plaziert innerhalb der Aminosäuren 71 bis 85, konnte hier zum erstenmal für die Präsentation mit HLA B51 beschrieben werden. In der spezifisch induzierten Lyse wiesen alle CD8+ CTL-Klone zwei Lysemechanismen auf, einen Perforin-vermittelten und einen CD95-vermittelten Anteil. Die CD95-vermittelte Lyse war stets prozentual geringer, und zusätzlich, sowohl Klon- wie Zielzell-spezifisch, unterschiedlich stark ausgeprägt . In HIV-infizierten primären Zellen ließ sich kein CD95-Anteil signifikant nachweisen, was darauf hinweist, daß dieser Lysemechanismus wahrscheinlich keine Rolle bei der Zerstörung infizierter Zellen im peripheren Blut spielt. In anderen Geweben kann sich die Situation unterschiedlich darstellen. Desweiteren konnte in dieser Arbeit nachgewiesen werden, daß das Vakzinia-Virus B13R-Genprodukt die CD95-vermittelte Apoptose hemmt. Daher muß bei der Untersuchung apoptotischer Vorgänge, die Wahl der eingesetzten Vektoren genauestens bedacht werden.
Identification of rat NKT cells and molecular analysis of their surface receptor mediated activation

Summary: Originally, NKT cells have been defined by their expression of T-cell receptor (TCR) and NK cell markers NKRP1A in human and NK1.1 (NKRP1C) in mouse. Most of these cells express CD1d-restricted TCR with a characteristic rearrangement- Va24JaQ/Vb11 in human and Va14Ja18/Vb8.2 in mouse, and have been implicated in playing an important role in first line defence and immunoregulation. The subject of this thesis was the characterisation of the hypothetical rat NKT cell population. In the mouse system, CD1d-restricted NK1.1+ T cells represented around 30% of intrahepatic and around 3% of splenic lymphocytes and could be visualised by staining with a-GalCer-loaded mouse CD1d tetramer. Rat NKRP1A+TCR+ cells, similar to mouse NKT cells, were predominantly expressed in the liver. However, their frequency was around 5 fold lower than the frequency of mouse intrahepatic lymphocytes. F344 rat NKT cells, in contrast to mouse CD4+ or DN NK1.1+ T lymphocytes, were of CD8 rather than CD4 phenotype, and did not bind to mCD1d-a-GalCer-tetramer. Since human hepatic CD1d-restricted Va24JQ+ T cells are not as frequent as their mouse counterparts and may express CD8- a marker not expressed by mouse CD1d-restricted cells, it is possible that the phenotype of F344 rat NKT cells corresponds more to the phenotype of human than mouse NKT cells. Similar to mouse NKT cells, F344 rat liver- and spleen-derived lymphocytes were able to produce IL-4 and IFN-g; when stimulated with the synthetic ligand a-GalCer in vitro. Therefore, the lack of binding of rat lymphocytes to mouse CD1d tetramer could not be due to their inability to respond to a-GalCer. To better characterise the reactivity of rat NKRP1A+TCR+ cells to a-GalCer, the rat invariant TCR was analysed. RT-PCR of liver lymphocytes with Va14-specific primers and subsequent cloning revealed a much weaker PCR signal for rat lymphocyte cDNA than for mouse cDNA. Furthermore the analysis of rat AV14JA18 sequences showed that the rat Va14+TCR invariant could be rearranged not only with AJ18 but also with other AJ segments. The low number of clones with in frame Va14Ja18 rearrangement could suggest that only a small proportion of liver lymphocytes were CD1d restricted NKT cells. Mouse and human NKT cells are able to recognise a-GalCer presented by the CD1d-b2 microglobulin complex, leading to their activation, proliferation and cytokine secretion. In order to compare the capacity of mouse and rat CD1d to present a-GalCer, rat CD1d was cloned. Sequence analysis and functional tests in vitro confirmed the structural and functional homology of rat CD1d with mouse CD1d. In parallel, to characterise the reactivity of rat NKRP1A+TCR+ cells to a-GalCer, rat Va14+TCR invariant was cloned and expressed in the TCR- T cell hybridoma BWr/mCD28. Rat Va14TCR+CD28+ transgenic cells secreted IL-2 upon aTCR/CD3 antibody stimulation, but were not specific for a-GalCer. Such cells were also negative in staining with mCD1d-a-GalCer tetramer. The lack of reactivity to a-GalCer and the lack of binding to mouse tetramer were probably caused by amino acid alterations, particularly at position 72 (51 according to IMTG nomenclature) of cloned rat TCRinv. Reversal of these “alterations” using molecular biology techniques was performed but the expression of this TCR on the surface of BWr/mCD28 cells could not be achieved. In contrast to rat TCRinv, mouse Va14+TCR was fully functional and was specific for mouse CD1d tetramer. KT12 hybridoma and BWr/mCD28 cells expressing mouse TCRinv, when stimulated with a-GalCer presented by primary CD1d+ cells or rCD1d transgenic cell lines, produced IL-2 in an Ag- and CD1d-dependent manner. Transgenic lines expressing TCR comprising mouse Va14 and rat Vb8.4 responded to a-GalCer presented by rat and mouse CD1d, and bound mCD1 tetramer. By contrast, cell lines expressing TCR comprising mouse Va14 and rat Vb8.2 responded only to a-GalCer presented by rCD1d and bound weakly to mCD1d tetramer. This suggests that germ line encoded regions of the b-chain (CDR2 or CDR4) bind to species-specific determinants of CD1d. The cytokine secretion of the cell lines was inhibited by anti-CD80 mAb, indicating the importance of CD80-CD28 costimulation in their activation. To check whether rat NKT cells may exist in other rat strains, the frequency and functions of NKRP1A+TCR+ in F344 and LEW rat were compared. F344 and LEW, two rat strains expressing different allelic CD1d forms, varied slightly in the level of CD1d expression, as assessed by staining with a newly generated CD1d specific monoclonal antibody. By contrast, these rat strains differed in terms of a-GalCer recognition. NKRP1A+TCR+ cells were less frequent in LEW than in F344 rats, and did not respond to a-GalCer or the analogue OCH in vitro, a result which is of special interest considering the susceptibility of LEW but not F344 rats to experimentally induced organ specific autoimmune diseases. In summary, the rat and mouse CD1d-invariant TCR systems show a high degree of structural and functional homology, but it seems that invariant NKT cells in rat, similar to such cells in human, occur at lower frequency than in mice. TCR transgenic cell line species-specific patterns of CD1d a-GalCer reactivity will provide a valuable tool for the mapping of CD1d/TCR contacts. Also monoclonal antibodies specific for rat and mouse CD1d, generated in this study, provide valuable tools to determine CD1d protein expression in various rat tissues and will help to better characterise functions of CD1d-restricted rat T cells.
Die Infektion mit dem Borna Disease Virus (BDV) ruft bei einem weiten Spektrum von Warmblütern ein teilweise progredientes, immunmediiertes neurologisches Syndrom hervor. BDV zeichnet sich durch ein einzelsträngiges RNA Genom negativer Polarität, einen ausgeprägten Neurotropismus und einen nicht-lytischen Replikationszyklus aus, der in viraler Persistenz mündet. In vitro Experimente zeigten kürzlich einen virostatischen Effekt des Guanosinanalogs Ribavirin gegenüber BDV. Ziel der vorliegenden Studie war es, den therapeutischen Nutzen einer intrathekalen Ribavirinapplikation bei akuter Bornascher Erkrankung (BD) im Rattenmodell zu untersuchen. Toxikologische und pharmakokinetische Studien ergaben eine maximal verträgliche tägliche Dosis von 2,5 mg/kg KG. Drei Wochen nach intranasaler Virusinokulation wurde Ribavirin in einer Dosis von 0, 1,25 und 2,5 mg/kg KG/Tag für 7 Tage intrathekal appliziert. Hierdurch gelang es, die klinische Symptomatik akuter BD dosisabhängig zu reduzieren. Die Bestimmung der Konzentration viraler RNA, Proteine, sowie infektiöser Partikel in zentralnervösem Gewebe ergab jedoch keine signifikante Reduktion. Anschließende immunhistologische Untersuchungen konnten eine quantitative Reduktion von T-Lymphozyten und Mikrogliazellen in Hirnparenchym behandelter Tiere nachweisen. Sowohl CD4+, als auch CD8+ T-Lymphozyten sind wesentlich an der progressiven Neurodestruktion im Rahmen von BD beteiligt. Auch für Mikrogliazellen wird eine kausale Beteiligung an der Pathogenese von BD postuliert. Der klinisch protektive Effekt der zentralen Ribavirinapplikation scheint in dem vorliegenden Modell nicht auf eine Inhibition der Virusreplikation, sondern auf die Suppression der neuropathogenen Immunantwort des Wirtsorganismus zurückzuführen zu sein. Wie in anderen Studien wiederholt beschrieben, konnte virale RNA im Blut infizierter Lewis-Ratten detektiert werden. Die Virämie bei hochdosiert behandelten Tieren ist deutlich ausgeprägter. Diese Beobachtung ist vermutlich durch eine systemische Infektion unter insuffizienter Immunkontrolle zu erklären. Hieraus ergeben sich auch Implikationen für die diagnostische Nutzbarkeit des peripheren Nukleinsäurenachweises zur Erkennung von BDV Infektionen. Die intrathekale Ribavirinapplikation ist der erste therapeutische Ansatz für BD, der zu einer Linderung des akuten Krankheitsbildes ohne gleichzeitige verstärkte Virusreplikation führt.
Diese Dissertation befaßt sich mit der Frage, ob der Transkriptionsfaktor Blimp-1 ebenso wie sein humanes Äquivalent PRDI-BF1 die Interferonproduktion reprimiert und somit die Virusvermehrung in Zellen erleichtert. An Blimp-1 exprimierenden L929-Zellen wurde die Interferonproduktion mit Hilfe des RNase Protection Assay auf mRNA-Ebene quantifiziert und die Virusvermehrung im Plaquetest untersucht. In beiden Fällen bestand kein signifikanter Unterschied zu einer Kontrollpopulation ohne Blimp-1-Expression, so daß die funktionelle Äquivalenz der beiden Transkriptionsfaktoren angezweifelt werden kann.
Hintergrund : Die HIV-Infektion ist durch eine schwere Beeinträchtigung sowohl der zellulären wie der humoralen Immunantwort gekennzeichnet. Dies kann auch Auswirkungen auf die bei mikrobiellen Infektionen auftretende Aviditätsreifung haben. Wir haben die Kinetik der Anti-HIV-Antikörperavidität bei HIV-Patienten studiert und untersucht, inwieweit die Aviditätsentwicklung mit dem Krankheitsverlauf der Infektion, der CD4-Zellzahl und der HI-Viruslast korreliert und ob der Aviditätsverlauf durch eine hochaktive, antiretrovirale Therapie (HAART) beeinflusst wird. Methoden : Bei insgesamt 39 HIV-Patienten wurden die Anti-gp41-, Anti-gp120- und Anti-p55-IgG-Avidität von Proben aus den Jahren 1991-2001 in ein- bis dreijährigem Abstand bestimmt. Auf der Basis des Infektionsverlaufes wurden die Patienten in vier Gruppen eingeteilt : 1. Langzeitüberlebende (LTS; n=12); 2. Patienten mit rascher Progression der HIV-Infektion vor der HAART-Ära (PROG; n=11); 3. Patienten, die mit HAART behandelt wurden (HAART; n=13); 4. Patienten in der Serokonversionsphase (SKP; n=3). Die Aviditätsbestimmung erfolgte mit Hilfe eines Festphasen-Enzymimmunoassays und 4 M Harnstoff als Elutionsreagenz. Ergebnisse : Unabhängig vom klinischen Status zeigten alle Patienten hohe (>60%) Aviditätswerte für Anti-gp41-IgG. Lediglich bei einem der drei Patienten in der Serokonversionsphase war eine Zunahme der Anti-gp41-IgG-Avidität feststellbar. Die Aviditätswerte für Anti-gp120-IgG bzw- Anti-p55-IgG lagen zwischen 25 und 91 % bzw. 14 und 106 %. Ein Vergleich der Aviditätswerte zwischen den Kollektiven LTS, PROG und HAART ergab keine signifikanten Unterschiede. Die Höhe der CD4-Zellzahl und der HI-Viruslast hatte keinen nachweisbaren Effekt auf den Aviditätsindex. Der Vergleich der Aviditätsdifferenzen für den Zeitraum vor HAART-Beginn mit denen für den Zeitraum während der HAART-Applikation wies keine signifikanten Unterschiede auf. Schlussfolgerungen : Obwohl bei einigen Patienten signifikante Änderungen der Aviditätswerte im Verlauf nachgewiesen werden konnten, ergab sich kein Hinweis darauf, dass die Aviditätsentwicklung durch den Krankheitsverlauf, den Immunstatus oder durch HAART beeinflusst wird.
Summary: In the present work, two important negative regulators of T cell responses in rats were examined. At the molecular level, rat CTLA-4, a receptor important for deactivating T cell responses, was examined for the expression pattern and in vitro functions. For this purpose, anti-rat CTLA-4 mAbs were generated. Consistent with the studies in mice and humans, rat CTLA-4 was detectable only in CD25+CD4+ regulatory T cells in unstimulated rats, and was upregulated in all activated T cells. Cross-linking rat CTLA-4 led to the deactivation of anti-TCR- and anti-CD28 stimulated (costimulation) T cell responses such as reduction in activation marker expression, proliferation, and cytokine IL-2 production. Although T cells stimulated with the superagonistic anti-CD28 antibody alone without TCR engagement also increased their CTLA-4 expression, a delayed kinetics of CTLA-4 upregulation was found in cells stimulated in this way. The physiological relevance of this finding needs further investigation. At the cellular level, rat CD25+CD4+ regulatory T cells were examined here in detail. Using rat anti-CTLA-4 mAbs, the phenotype of CD25+CD4+ regulatory T cells was investigated. Identical to the mouse and human Treg phenotype, rat CD25+CD4+ T cells constitutively expressed CTLA-4, were predominantly CD45RC low, and expressed high level of CD62L (L-selectin). CD25+CD4+ cells proliferated poorly and were unable to produce IL-2 upon engagement of the TCR and CD28. Furthermore, rat CD25+CD4+ cells produced high amounts of anti-inflammatory cytokine IL-10 upon stimulation. Importantly, freshly isolated CD25+CD4+ T cells from naïve rats exhibited suppressor activities in the in vitro suppressor assays. In vitro, CD25+CD4+ regulatory T cells proliferated vigorously upon superagonistic anti-CD28 stimulation and became very potent suppressor cells. In vivo, a single injection of CD28 superagonist into rats induced transient accumulation and activation of CD25+CD4+ regulatory T cells. These findings suggest firstly that efficient expansion of CD25+CD4+ cells without losing their suppressive effects (even enhance their suppressive activities) can be achieved with the superagonistic anti- CD28 antibody in vitro. Secondly, the induction of disproportional expansion of CD25+CD4+ cells by a single injection of superagonistic anti-CD28 antibody in vivo implies that superagonistic anti-CD28 antibody may be a promising candidate in treating autoimmune diseases by causing a transient increase of activated CD25+CD4+ T cells and thus tipping ongoing autoimmune responses toward selftolerance.
