Inhibition of coronavirus (CoV)‐encoded papain‐like cysteine proteases (PL\(^{pro}\)) represents an attractive strategy to treat infections by these important human pathogens. Herein we report on structure‐activity relationships (SAR) of the noncovalent active‐site directed inhibitor (R)‐5‐amino‐2‐methyl‐N‐(1‐(naphthalen‐1‐yl)ethyl) benzamide (2 b), which is known to bind into the S3 and S4 pockets of the SARS‐CoV PL\(^{pro}\). Moreover, we report the discovery of isoindolines as a new class of potent PL\(^{pro}\) inhibitors. The studies also provide a deeper understanding of the binding modes of this inhibitor class. Importantly, the inhibitors were also confirmed to inhibit SARS‐CoV‐2 replication in cell culture suggesting that, due to the high structural similarities of the target proteases, inhibitors identified against SARS‐CoV PL\(^{pro}\) are valuable starting points for the development of new pan‐coronaviral inhibitors.
Cholinesterases are important biological targets responsible for regulation of cholinergic transmission, and their inhibitors are used for the treatment of Alzheimer’s disease. To design new cholinesterase inhibitors, of different structure-based design strategies was followed, including the modification of compounds from a previously developed library and a fragment-based design approach. This led to the selection of heterodimeric structures as potential inhibitors. Synthesis and biological evaluation of selected candidates confirmed that the designed compounds were acetylcholinesterase inhibitors with \(IC_{50}\) values in the mid-nanomolar to low micromolar range, and some of them were also butyrylcholinesterase inhibitors.
Whereas most currently used antibiotics act by interfering with essential bacterial processes, a smaller group of antibacterials disturbs the integrity of the cell membrane. Since fatty acids are a vital component of membrane phospholipids, the type-II fatty acid biosynthesis pathway (FAS-II) of bacteria constitutes a promising drug target. The front-line anti-tuberculosis prodrug isoniazid blocks the FAS-II pathway in M. tuberculosis thereby leading to morphological changes and finally to cell lysis. When it became evident that the enoyl-ACP reductase in the FAS-II pathway is the target of the activated isoniazid, several programs were initiated to develop novel inhibitors directed against this protein in different pathogens. The S. aureus enoyl-ACP reductase (saFabI) is of particular interest since three promising drug candidates inhibiting this homologue have reached clinical trials. However, despite these prospects, no crystal structures of saFabI were publicly available at the time the present work was initiated. Thus, one major goal of this thesis was the generation of high-resolution atomic models by means of X-ray crystallography. The development of a highly reproducible approach to co-crystallize saFabI in complex with NADP+ and diphenyl ether-based inhibitors led to crystal structures of 17 different ternary complexes. Additional crystallographic experiments permitted the view into two apo-structures and two atomic models of saFabI in complex with NADPH and 2-pyridone inhibitors. Based on the established saFabI structure, molecular dynamics (MD) simulations were performed to improve our understanding of the conformational mobility of this protein. Taken together, these investigations of the saFabI structure and its flexibility served as an ideal platform to address important questions surrounding substrate and inhibitor recognition by this enzyme. Intriguingly, our saFabI structures provide several vastly different snapshots along the reaction coordinate of ligand binding and hydride transfer, including the closure of the flexible substrate binding loop (SBL). The extraordinary mobility of saFabI was confirmed by MD simulations suggesting that conformational motions indeed play a pivotal role during substrate delivery and turnover. A water chain linking the active site with a water-basin inside the homo-tetrameric enzyme was found likely to be crucial for the closure and opening of the SBL and, thus, for the catalyzed reaction. Notably, the induced-fit ligand binding process involves a dimer-tetramer transition, which could be related to the observed positive cooperativity of cofactor and substrate binding. Overall, saFabI displays several unique characteristics compared to FabI proteins from other organisms that might be necessary for the synthesis of branched-chain fatty acids, which in turn are required for S. aureus fitness in vivo. This finding may explain why S. aureus is sensitive to FAS-II inhibitors even in the presence of exogenous fatty acids. Accordingly, saFabI remains a valid drug target and our structures can be used as a molecular basis for rational drug design efforts. In fact, binding affinity trends of diphenyl ether inhibitors and, more importantly, the correlated residence times could be rationalized at the molecular level. Furthermore, the structure of saFabI in complex with the 2-pyridone inhibitor CG400549 revealed unique interactions in the wider binding crevice of saFabI compared to other FabI homologues explaining the narrow activity spectrum of this clinical candidate with proven human efficacy. In summary, these studies provide an ideal platform for the development of new, effective saFabI inhibitors as exemplified by the promising 4-pyridone PT166. In the context of this dissertation, crystal structures of the condensing enzyme KasA in complex with several analogs of the naturally occurring inhibitor thiolactomycin have been solved.
The inhibitory glycine receptors are one of the major mediators of rapid synaptic inhibition in the mammalian brainstem, spinal cord and higher brain centres. They are ligand-gated ion channels that are mainly involved in the regulation of motor functions. Dysfunction of the receptor is associated with motor disorders such as hypereklepxia or some forms of spasticity. GlyR is composed of two glycosylated integral membrane proteins α and β and a peripheral membrane protein of gephyrin. Moreover, there are four known isoforms of the α-subunit (α1-4) of GlyR while there is a single β-subunit. Glycine receptors can be homomeric including α subunits only or heteromeric containing both α and β subunits. To date, strychnine is the ligand that has the highest affinity as glycine receptor ligand. It acts as a competitive antagonist of glycine that results in the inhibition of Cl- ions permeation and consequently reducing GlyR-mediated inhibition.
For a long time, the details of the molecular mechanism of GlyRs inactivation by strychnine were insufficient due to the lack of high-resolution structures of the receptor. Only homology models based on structures of other cys-loop receptors have been available. Recently, 3.0 Å X-ray structure of the human glycine receptor- α3 homopentamer in complex with strychnine, as well as electro cryo-microscopy structures of the zebra fish α1 GlyR in complex with strychnine and glycine were published. Such information provided detailed insight into the molecular recognition of agonists and antagonists and mechanisms of GlyR activation and inactivation.
Very recently, a series of dimeric strychnine analogs obtained by diamide formation of two molecules of 2-aminostrychnine with diacids of different chain length was pharmacologically evaluated at human α1 and α1β glycine receptors. None of the dimeric analogs was superior to strychnine.
The present work focused on the extension of the structure-activity relationships of strychnine derivatives at glycine receptors
All the synthesized compounds were pharmacologically evaluated at human α1 and α1β glycine receptors in a functional FLIPRTM assay and the most potent analogs were pharmacologically evaluated in a whole cell patch-clamp assay and in [3H]strychnine binding studies.
It was reported that 11-(E)-isonitrosostrychnine displayed a 2-times increased binding to both α1 and α1β glycine receptors which prompted us to choose the hydroxyl group as a suitable attachment point to connect two 11-(E)-isonitrosostrychnine molecules using a spacer. In order to explore the GlyR pocket tolerance for oxime extension, a series of oxime ethers with different spacer lengths and sterical/lipophilic properties were synthesized biologically evaluated. Among all the oxime ethers, methyl, allyl and propagyl oxime ethers were the most potent antagonists displaying IC50 values similar to that of strychnine. These findings indicated that strychnine binding site at GlyRs comprises an additional small lipophilic pocket located in close proximity to C11 of strychnine and the groups best accommodated in this pocket are (E)-allyl and (E)-propagyl oxime ethers.
Moreover, 11-aminostrychnine, and the corresponding propionamide were prepared and pharmacologically evaluated to examine the amide function at C11 as potential linker.
A series of dimeric strychnine analogs designed by linking two strychnine molecules through amino groups in position 11 with diacids were synthesized and tested in binding studies and functional assays at human α1 and α1β glycine receptors. The synthesized bivalent ligands were designed to bind simultaneously to two α-subunits of the pentameric glycine receptors causing a possibly stronger inhibition than the monomeric strychnine. However, all the bivalent derivatives showed no significant difference in potency compared to strychnine. When comparing the reference monomeric propionamide containing ethylene spacer to the dimeric ligand containing butylene spacer, a 3-fold increase in potency was observed. Since the dimer containing (CH2)10 spacer length was found to be equipotent to strychnine, it is assumed that one molecule of strychnine binds to the receptor and the ‘additional’ strychnine molecule in the dimer probably protrudes from the orthosteric binding sites of the receptor.
During the last decades the number of biologics increased dramatically and several biopharmaceutical drugs such as peptides, therapeutic proteins, hormones, enzymes, vaccines, monoclonal antibodies and antibody-drug conjugates conquered the market. Moreover, administration and local delivery of growth factors has gained substantial importance in the field of tissue engineering. Despite progress that has been made over the last decades formulation and delivery of therapeutic proteins is still a challenge. Thus, we worked on formulation and delivery strategies of therapeutic proteins to improve their biological performance.
Phase I of this work deals with protein stability with the main focus on a liquid protein formulation of the dimeric fusion protein PR-15, a lesion specific platelet adhesion inhibitor. In order to develop an adequate formulation ensuring the stability and bioactivity of PR-15 during storage at 4 °C, a pH screening, a forced degradation and a Design of Experiments (DoE) was performed. First the stability and bioactivity of PR-15 in 50 mM histidine buffer in relation to pH was evaluated in a short-term storage stability study at 25 °C and 40 °C for 4 and 8 weeks using different analytical methods. Additionally, potential degradation pathways of PR-15 were investigated under stressed conditions such as heat treatment, acidic or basic pH, freeze-thaw cycles, light exposure, induced oxidation and induced deamidation during the forced degradation study. Moreover, we were able to identify the main degradation product of PR-15 by performing LC/ESI-MS analysis. Further optimization of the injectable PR 15 formulation concerning pH, the choice of buffer and the addition of excipients was studied in the following DoE and finally an optimal PR-15 formulation was found.
The growth factors BMP-2, IGF-I and TGF-β3 were selected for the differentiation of stem cells for tissue engineering of cartilage and bone in order to prepare multifunctionalized osteochondral implants for the regeneration of cartilage defects.
Silk fibroin (SF) was chosen as biomaterial because of its biocompatibility, mechanical properties and its opportunity for biofunctionalization. Ideal geometry of SF scaffolds with optimal porosity was found in order to generate both tissues on one scaffold.
The growth factors BMP-2 and IGF-I were modified to allow spatially restricted covalent immobilization on the generated porous SF scaffolds. In order to perform site-directed covalent coupling by the usage of click chemistry on two opposite sides of the scaffold, we genetically engineered BMP-2 (not shown in this work; performed by Barbara Tabisz) and IGF-I for the introduction of alkyne or azide bearing artificial amino acids. TGF β3 was immobilized to beads through common EDC/NHS chemistry requiring no modification and distributed in the pores of the entire scaffold.
For this reason protein modification, protein engineering, protein immobilization and bioconjugation are investigated in phase II. Beside the synthesis the focus was on the characterization of such modified proteins and its conjugates. The field of protein engineering offers a wide range of possibilities to modify existing proteins or to design new proteins with prolonged serum half-life, increased conformational stability or improved release rates according to their clinical use.
Site-directed click chemistry and non-site-directed EDC/NHS chemistry were used for bioconjugation and protein immobilization with the aim to underline the preferences of site-directed coupling.
We chose three strategies for the incorporation of alkyne or azide functionality for the performance of click reaction into the protein of interest: diazonium coupling reaction, PEGylation and genetic engineering. Azido groups were successfully introduced into SF by implementation of diazonium coupling and alkyne, amino or acid functionality was incorporated into FGF-2 as model protein by means of thiol PEGylation. The proper folding of FGF-2 after PEGylation was assessed by fluorescence spectroscopy, WST-1 proliferation assay ensured moderate bioactivity and the purity of PEGylated FGF-2 samples was monitored with RP-HPLC. Moreover, the modification of native FGF-2 with 10 kDa PEG chains resulted in enhanced thermal stability.
Additionally, we genetically engineered one IGF-I mutant by incorporating the unnatural amino acid propargyl-L-lysine (plk) at position 65 into the IGF-I amino acid sequence and were able to express hardly verifiable amounts of plk-IGF-I. Consequently, plk-IGF-I expression has to be further optimized in future studies in order to generate plk-IGF-I with higher yields.
Bioconjugation of PEGylated FGF-2 with functionalized silk was performed in solution and was successful for click as well as EDC/NHS chemistry. However, substantial amounts of unreacted PEG-FGF-2 were adsorbed to SF and could not be removed from the reaction mixture making it impossible to expose the advantages of click chemistry in relation to EDC/NHS chemistry. The immobilization of PEG-FGF-2 to microspheres was a trial to increase product yield and to remove unreacted PEG-FGF-2 from reaction mixture. Bound PEG-FGF-2 was visualized by fluorescence imaging or flow cytometry and bioactivity was assessed by analysis of the proliferation of NIH 3T3 cells. However, immobilization on beads raised the same issue as in solution: adsorption caused by electrostatic interactions of positively charged FGF-2 and negatively charged SF or beads. Finally, we were not able to prove superiority of site-directed click chemistry over non-site-directed EDC/NHS.
The skills and knowledge in protein immobilization as well as protein characterization acquired during phase II helped us in phase III to engineer cartilage tissue in biofunctionalized SF scaffolds.
The approach of covalent immobilization of the required growth factors is relevant because of their short in vivo half-lives and aimed at controlling their bioavailability. So TGF-β3 was covalently coupled by means of EDC/NHS chemistry to biocompatible and biostable PMMA beads. Herein, we directly compared bioactivity of covalently coupled and adsorbed TGF-β3. During the so-called luciferase assay bioactivity of covalent coupled as well as adsorbed TGF-β3 on PMMA beads was ensured. In order to investigate the real influence of EDC/NHS chemistry on TGF-β3’s bioactivity, the amount of immobilized TGF-β3 on PMMA beads was determined. Therefore, an ELISA method was established. The assessment of total amount of TGF-β3 immobilized on the PMMA beads allowed as to calculate coupling efficiency. A significantly higher coupling efficiency was determined for the coupling of TGF-β3 via EDC/NHS chemistry compared to the reaction without coupling reagents indicating a small amount of adsorbed TGF-β3. These results provide opportunity to determine the consequence of coupling by means of EDC/NHS chemistry for TGF β3 bioactivity. At first sight, no statistically significant difference between covalent immobilized and adsorbed TGF-β3 was observed regarding relative luciferase activities. But during comparison of total and active amount of TGF-β3 on PMMA beads detected by ELISA or luciferase assay, respectively, a decrease of TGF-β3’s bioactivity became apparent. Nevertheless, immobilized TGF β3 was further investigated in combination with SF scaffolds in order to drive BMSCs to the chondrogenic lineage. According to the results obtained through histological and immunohistochemical studies, biochemical assays as well as qRT-PCR of gene expression from BMSCs after 21 days in culture immobilized TGF-β3 was able to engineer cartilage tissue. These findings support the thesis that local presentation of TGF β3 is superior towards exogenous TGF β3 for the development of hyaline cartilage. Furthermore, we conclude that covalent immobilized TGF β3 is not only superior towards exogenously supplemented TGF-β3 but also superior towards adsorbed TGF-β3 for articular hyaline cartilage tissue engineering. Diffusion processes were inhibited through covalent immobilization of TGF-β3 to PMMA beads and thereby a stable and consistent TGF-β3 concentration was maintained in the target area.
With the knowledge acquired during phase II and III as well as during the studies of Barbara Tabisz concerning the expression and purification of plk-BMP-2 we made considerable progress towards the formation of multifunctionalized osteochondral implants for the regeneration of cartilage defects. However, further studies are required for the translation of these insights into the development of multifunctionalized osteochondral SF scaffolds.
In der vorliegenden Arbeit werden Studien zu verschiedenen Aspekten der Stimulation von humanen Vg9Vd2-T-Lymphozyten vorgestellt. Ein Schwerpunkt war die Charakterisierung der Erkennung von Bisphosphonaten durch Vg9Vd2-T-Zellen. Weder die Alkylmonophosphonate Ethyl- und Propylphosphonat noch 3-Aminopropylphosphonat bewirkten eine Stimulation von Vg9Vd2 T-Lymphozyten. Anscheinend ist also für die gd-T-Zell-stimulierende Aktivität von Aminobisphosphonaten das Vorhandensein sowohl der Methylenbisphosphonat-Gruppe als auch des Amino-Stickstoffs entscheidend. Es wurden verschiedene Pamidronat-Derivate untersucht, die sich durch Substituenten am Stickstoffatom unterscheiden. Die meisten dieser Verbindungen konnten Vg9Vd2-T-Zellen aktivieren, die Art der Substituenten hatte aber großen Einfluss darauf, welche Konzentration des jeweiligen Bisphosphonats für eine gd-T-Zell-Antwort nötig war. Besonders negativ auf die gd-T-Zell-stimulierende Aktivität wirkte sich aus, wenn das Stickstoffatom Teil einer Säureamidbindung war. Das lässt darauf schließen, dass die Gegenwart einer positiven Ladung (durch Protonierung des Stickstoffatoms) von Bedeutung für die Bioaktivität dieser Verbindungen ist. Beim Vergleich verschiedener Bisphosphonate mit stickstoffenthaltenden Heteroaromaten (Fünfringe mit ein bis drei Stickstoffatomen) zeigte sich, dass sowohl die Position des basischen Stickstoffatoms im Ring als auch Art und Position von Ringsubstituenten Einfluss auf deren gd-T-Zell-stimulierende Aktivität haben. Es ergaben sich Hinweise, dass sich eine gesteigerte Neigung eine positive Ladung in der Seitenkette zu tragen bei diesen Verbindungen genau wie bei Aminobisphosphonaten günstig auf das gd-T-Zell-aktivierende Potential auswirkt. Durch Behandlung mit Zoledronat wurde die monozytäre Zelllinie THP-1 stimulierend für Vg9Vd2-T-Zellen. Auch weitere Zelllinien und Lymphozyten des peripheren Bluts (PBL) konnten nach Vorinkubation mit Zoledronat Vg9Vd2-T-Zellen aktivieren. Dabei genügten bei den PBL deutlich geringere Zoledronat-Konzentrationen um einen Effekt zu erzielen als bei den untersuchten Zelllinien. Für die indirekte Stimulation von Vg9Vd2-T-Zellen durch Zoledronat mittels THP-1 Zellen oder PBL war Zell-Zell-Kontakt zwischen den präsentierenden Zellen und den gd-T-Zellen Voraussetzung. Die Anwesenheit von alkalischer Phosphatase hatte keine Auswirkungen auf die gd-T-Zell-Aktivierung durch Zoledronat. Dies spricht dafür, dass Vg9Vd2-T-Zellen Oberflächenstrukturen auf anderen Zellen erkennen und dass freie Phosphoantigene bei der gd-T-Zell-Stimulierung durch Stickstoff enthaltende Bisphosphonate keine Rolle spielen. Wurde die Vorinkubation von THP-1 Zellen mit Zoledronat in Gegenwart von Saponin, einem Detergenz das die Durchlässigkeit der Zellmembran reversibel erhöht, durchgeführt, reichten deutlich niedrigere Konzentrationen des Bisphosphonats aus, um die THP-1 Zellen stimulierend für gd-T-Lymphozyten zu machen. Das ist ein Hinweis darauf, dass für die Aktivierung von Vg9Vd2-T-Zellen durch Zoledronat intrazelluläre Vorgänge in den „antigenpräsentierenden“ Zellen verantwortlich sein könnten. Beim Vergleich verschiedener N-BPs hinsichtlich ihrer Aktivität gegenüber gd-T-Zellen und ihrer antiresorptiven Potenz ergab sich eine erstaunlich gute Korrelation. Dies könnte darauf hinweisen, dass beide Effekte durch den gleichen Mechanismus – die Hemmung der Farnesylpyrophosphat-Synthase – zustande kommen. Die Gegenwart von Farnesol oder Geranylgeraniol während der Vorinkubation von THP-1 Zellen mit Zoledronat verringerte deren gd-T-Zell-stimulierendes Potential nicht, so dass vielleicht nicht die Verarmung an längerkettigen Isoprenoiden sondern die Anreicherung von Vorläufern zu Veränderungen in den Zellen führt, die diese schließlich erkennbar für Vg9Vd2-T-Zellen machen. Zusätzlich wurden im Rahmen dieser Arbeit einige Versuche zu bakteriellen Phosphoantigenen durchgeführt. Mittels einer selektiven HPLC-MS/MS-Methode gelang uns in einer E. coli-Probe der Nachweis eines Vg9Vd2-T-Zell-stimulierenden Pyrophosphats mit der molekularen Masse von 3-Formyl-1-butyl-pyrophosphat, einem Phosphoantigen das in Mykobakterien gefunden worden war (Belmant und Mitarbeiter, 1999). Eine weitere Strukturaufklärung war aufgrund der äußerst geringen Konzentration des Phosphoantigens nicht möglich. 2-C-Methyl-D-erythritol-2,4-cyclodiphosphat (MEcPP) ist ein Zwischenprodukt des 2-C-Methylerythritol-4-phosphat- (MEP) Wegs der Isoprenoidbiosynthese. Es wird in manchen Bakterien – z. B. Corynebacterium ammoniagenes – bei oxidativem Stress verursacht durch Benzylviologen (BV) akkumuliert. Es konnte gezeigt werden, dass Extrakte aus C. ammoniagenes, die in Gegenwart von BV kultiviert wurden, in einem höherem Maße Vg9Vd2-T-Zellen stimulieren als Proben von Bakterien, die ohne BV-Zusatz wuchsen; MEcPP wirkte selbst nicht als Phosphoantigen. Dies war ein weiterer Hinweis darauf, dass bakterielle Phosphoantigene mit dem MEP-Weg assoziiert sind.
Alzheimer′s disease (AD) is a neurological disorder with still no preventive or curative treatment. Flavonoids are phytochemicals with potential therapeutic value. Previous studies described the flavanone sterubin isolated from the Californian plant Eriodictyon californicum as a potent neuroprotectant in several in vitro assays. Herein, the resolution of synthetic racemic sterubin (1) into its two enantiomers, (R)‐1 and (S)‐1, is described, which has been performed on a chiral chromatographic phase, and their stereochemical assignment online by HPLC‐ECD coupling. (R)‐1 and (S)‐1 showed comparable neuroprotection in vitro with no significant differences. While the pure stereoisomers were configurationally stable in methanol, fast racemization was observed in the presence of culture medium. We also established the occurrence of extracted sterubin as its pure (S)‐enantiomer. Moreover, the activity of sterubin (1) was investigated for the first time in vivo, in an AD mouse model. Sterubin (1) showed a significant positive impact on short‐ and long‐term memory at low dosages.
Bioprinting has emerged as a valuable threedimensional (3D) biomanufacturing method to fabricate complex hierarchical cell-containing constructs. Spanning from basic research to clinical translation, sterile starting materials are crucial. In this study, we present pharmacopeia compendial sterilization methods for the commonly used bioink component alginate. Autoclaving (sterilization in saturated steam) and sterile filtration followed by lyophilization as well as the pharmacopeia non-compendial method, ultraviolet (UV)-irradiation for disinfection, were assessed. The impact of the sterilization methods and their effects on physicochemical and rheological properties, bioprinting outcome, and sterilization efficiency of alginate were detailed. Only sterile filtration followed by lyophilization as the sterilization method retained alginate's physicochemical properties and bioprinting behavior while resulting in a sterile outcome. This set of methods provides a blueprint for the analysis of sterilization effects on the rheological and physicochemical pattern of bioink components and is easily adjustable for other polymers used in the field of biofabrication in the future.
Unser Ziel war es, einen neuen Zugang zu den sehr ähnlichen Cassia- und Microcos-artigen Piperidinalkaloiden zu finden. Aus dem „chiral pool“ wurde der L-Zucker Rhamnose als Ausgangsmaterial gewählt. Diese wurde zunächst in einen Azidoaldehyd, einem flexiblen Synthesebaustein zur Darstellung von Cassia- und Microcos-Piperidinalkaloiden, umgesetzt. Die Lücke zu den Piperidinalkaloiden wurde mit Hilfe der Tandem Wittig-[3+2]-Cycloaddition geschlossen. Das Reaktionsprodukt ist ein Triazolin, welches in einen Diazoester isomerisiert wurde. Eine nachfolgende Stickstoffextrusion ergab ein vinyloges Urethan. Mit Hilfe einer hochgradig steroselektiven katalytischen Hydrierung konnten verschiedene Homopipecolinsäuremethylester erhalten werden. über einen identischen Weg wurden auch verschiedene Homopipecolinsäuren mit Cassia- und Microcos-Grundkörper dargestellt. Anhand eines Modellsystems wurde zunächst das Konzept der Tandem Wittig-[3+2]-Cycloadditions-Reaktion zu einer Tandem HWE-[3+2]-Cycloadditions-Reaktion erweitert. In nur einem Ansatz gelangen so gleichzeitig der Aufbau des Piperidingrundkörpers und das Anbringen der gewünschten Seitenkette. Dieses neue Konzept wurde nun zur Synthese des natürlichen (-)-Cassins verwendet. Analog zur Tandem Wittig-[3+2]-Cycloaddition entsteht ein vinyloges Amid nach Stickstoffextrusion. Durch Hydrierung und nachfolgende Barton-McCombie Desoxygenierung wurde eine direkte Vorstufe des Cassins erhalten. Durch saure Hydrolyse konnten in einem Ansatz sämtliche Schutzgruppen entfernt und der Naturstoff (-)-Cassin (1) erfolgreich dargestellt werden.
Das Ziel dieser Arbeit war es, eine im Arbeitskreis entwickelte Methode zur Darstellung von unterschiedlich substituierten -Aminopiperidinen zu etablieren und zu verifizieren, indem unser Konzept einer Tandemreaktion zum Einsatz kommen sollte. Diese Reaktionssequenz sollte anschließend zur Totalsynthese von Microconin, einem aus Microcos paniculata isoliertem Alkaloid, genutzt werden. Den ersten Schritt in Richtung -Aminopiperidinderivate machte die Veresterung von L-Pyroglutaminsäure. Nach der Reduktion des Methylesters erfolgte die Aktivierung der Hydroxyfunktion des Alkohols in Form des Tosylats. Die Azideinführung resultierte aus einer nucleophilen Substitutionsreaktion, anschließend daran wurde der Lactam-Stickstoff mit Boc2O und einer katalytischen Menge DMAP geschützt. Das Lactam ist durch den Elektronenakzeptor aktiviert, so dass die Ringöffnung zum Methylester unter sehr milden Bedingungen und ohne weiteren Reinigungsschritt erfolgen konnte. Die Aminofunktion musste mit einer zweiten Schutzgruppe blockiert werden, die anschließende selektive Reduktion mit DiBAl-H in trockenem Ether verlief problemlos und lieferte mit dem Aldehyd das Edukt für Olefinierungen mittels verschiedener Wittig-Reaktionen. Dafür wurden stabilisierte Phosphonate hergestellt und in einer HWE-Reaktion mit dem Aldehyd umgesetzt. Die resultierenden elektronenarmen Olefine gingen dann die intramolekulare Cycloaddition mit dem Azidsubstituenten ein und bauten so den Grundkörper der -Aminopiperidinderivate in einer Reaktionssequenz auf, die wir als Tandem Wittig-[3+2]-Cycloaddition bezeichnen. Die Bildung der primären Triazoline erfolgte stereoselektiv, die Geschwindigkeit der Cycloaddition hing sowohl vom konjugierten Olefinsubstituenten als auch vom vicinalen Substituenten der Azidfunktion ab. Die Konfigurationsbestimmung erfolgte mittels NMR-Spektroskopie durch Analyse der Kopplungskonstanten und NOE-Messungen. Die asymmetrische Induktion der Cycloaddition konnte direkt für die Stereochemie am Piperidinring genutzt werden, indem, nach basischer Triazolin/Diazoamin Isomerisierung, gleich das Diazoamin hydriert wurde. Die Hydrierung der vinylogen Urethane, den Produkten aus der rhodiumkatalysierten Stickstoffextrusion, lieferte ein Diastereomerengemisch, wobei das Verhältnis der Diastereomere hauptsächlich vom Substitutionsgrad des exocyclischen Amins abhängig war. Überraschenderweise fand beim Sulfontriazolin keine Isomerisierung zum Diazoamin statt, daher musste für die Darstellung der Sulfonylmethyl--aminopiperidine eine alternative Route über ein Ketosulfon beschritten werden. Die Synthese von Microconin begann mit der Desoxygenierung von L-Rhamnose durch die sog. Fischer-Zach-Reaktion. Das Rhamnal wurde in einer drei Stufen Eintopfreaktion erhalten und mittels Perlinhydrolyse in den offenkettigen Aldehyd umgewandelt. Die Aktivierung der Hydroxyfunktion als Mesylat resultierte in einer äußerst empfindlichen Verbindung, die nur durch Verwendung des Lindlar-Katalysators mit zufrieden stellenden Ergebnis zum aliphatischen Aldehyd reduziert werden konnte. Eine bimolekulare nucleophile Substitutionsreaktion lieferte bei der Azideinführung zur Schlüsselverbindung sowohl die benötigte funktionelle Gruppe als auch die benötigte Inversion der Konfiguration. Die Tandem HWE-[3+2]-Cycloadditions-Reaktion führte auch bei dem Sulfontriazolin in eine Sackgasse, weshalb wieder eine alternative Syntheseroute eingeschlagen werden musste. Ausgehend von derselben Schlüsselverbindung gelang dies durch eine zinnkatalysierte Umsetzung mit stabilisierten Diazomethan zum Ketosulfon. Der Aufbau des Piperidin-Heterocyclus konnte dann wieder über eine intramolekulare Imin-Bildung des intermediären Amins mit dem Keton erzielt werden. Die diastereoselektive Hydrierung verlief unter Wasserstoffaddition von der sterisch weniger gehinderten -Seite und Ausbildung des all cis Substitutionsmusters. Nach dem erfolgreichen Aufbau des Heterocyclus mussten noch die beiden Heteroatome methyliert werden. Dabei wurden die besten Ergebnisse am Ringstickstoff mit der reduktiven Aminierung erzielt. Bei der anschließenden Abspaltung der Acetylgruppe zeigte sich erstmals, dass das Substitutionsmuster am Piperidinring nicht konfigurationsstabil war, da neben dem erwarteten Alkohol auch das Diastereomer isoliert wurde. Die genaue Ursache für die Epimerisierung nach der N-Methylierung konnte nicht geklärt werden. Die Einführung des Methoxy-Substituenten am Grundkörper erfolgte über eine Williamsonschen Ethersynthese. Bei den Versuchen zur Kupplung des Grundkörpers mit der Seitenkette 2,4-Nonadienal erwies sich der Zusatz von HMPT als förderlich. Weiterhin konnte die Ausbeute an -Hydroxysulfonen durch das Erwärmen der Reaktionsmischung gesteigert werden. In den Folgeschritten der Julia-Olefinierung blieben die Versuche zur Ausbildung der dreifach ungesättigten Struktureinheit in der Seitenkette des isolierten Naturstoffs jedoch erfolglos.
Stachys officinalis galt von der Antike bis zur frühen Neuzeit als wichtiges Phy-topharmakon. Zu den bedeutendsten Indikationen zählen die Anwendung bei Er-krankungen der Atemwege, des Gastrointestinaltrakts, der Harnwege und der Ein-satz als Analgetikum bei Schmerzen verschiedenster Art. Über den Zeitraum von fast 1800 Jahren zählte die Pflanze fest zum Arzneischatz. Erst Ende des 18. Jahrhunderts nahm ihre Bedeutung als Arzneimittel ab. Mit dem Beginn des 20. Jahrhundert verschwand sie fast vollständig aus der Phytotherapie. Heute wird sie nur noch selten im Bereich der homöopathischen Medizin oder volksheilkundlich eingesetzt. In der vorliegenden Arbeit wird Stachys officinalis durch ihre 2000 jährige Ge-schichte als Arzneipflanze begleitet. Dabei werden die Indikationen verschiedener Quellen mit aktuellem pharmazeutischem Wissen verglichen und beurteilt. Durch diese Auswertung wird geklärt, ob die jeweiligen Indikationsnennungen pharma-zeutisch nachvollziehbar sind oder nicht. Als Vergleichsweke dienen Hagers En-zyklopädie der Arzneistoffe und Drogen´ und Dr. Duke´s Phytochemical and Ethnobotanical Databases´. , Die inhaltsstoffbezogenen Angaben dieser Wer-ke werden durch weitere aktuelle pharmazeutische Erkenntnisse über die phar-makologischen Eigenschaften der Pflanzeninhaltsstoffe von Stachys officinalis ergänzt. Auf Grundlage der zugeordneten Pflanzeninhaltsstoffe und ihrer individu-ellen Wirkungen wird die Plausibilität der Indikationsangaben der Quellen unter-sucht. Im Rahmen dieser Arbeit werden die Indikationsnennungen von 34 Kräuterbü-chern, Lexika und anderer Quellen betrachtet. Dabei werden die vielfältigen Indi-kationsnennungen zu 142 normierten Indikationen zusammengefasst und ausge-wertet. Unter Berücksichtigung der Angaben beider oben genannter Vergleichs-werke erscheinen 59% der 142 normierten Indikationen für Stachys officinalis plausibel und sind mit der Anwesenheit definierter Pflanzeninhaltsstoffe zu erklä-ren. Es wird deutlich, dass viele Nennungen Erkrankungen des Gastrointestinaltraktes, der Atemwege und der Leber betreffen. Die analgetischen Indikationsangaben sind wenig einheitlich, betreffen aber häufig Schmerzen des Kopfes und des Be-wegungsapparats. Zusammengefasst haben die verschiedenen analgetischen Indikationsangaben allerdings einen bemerkenswert hohen Anteil von 11% an den insgesamt 801 Indikationsnennungen. Die vorliegende Arbeit gibt 13 Hauptindikationen für Stachys officinalis an. Diese sind mit Ausnahme der Indikationen Bei Milzerkrankungen´ und Bei Epilepsie´ durch die Wirkungen benannter Pflanzeninhaltsstoffe nachzuvollziehen und phar-mazeutisch plausibel. Es gelingt, aus der Fülle der Indikationsnennungen der his-torischen Quellen die meistgenannten belegbaren Anwendungen hervorzuheben. Die Untersuchung der Überlieferungsgeschichte macht deutlich, dass die antiken Autoren, wie zum Beispiel Dioskurides und Plinius, einen großen Einfluss auf die Indikationsangaben von Stachys officinalis haben. Viele spätere Werke orientieren sich an diesen antiken Quellen und nehmen deren Indikationsangaben auf. Ab dem 18. Jahrhundert nimmt der Umfang der Beschreibungen von Stachys officina-lis und ihre Bedeutung als Phytopharmakon deutlich ab. Auf Grund der hohen Plausibilität der 13 Hauptindikationen sowie der analgeti-schen Anwendungen scheint es nicht gerechtfertigt, dass Stachys officinalis voll-ständig aus dem Arzneischatz verschwunden ist.
The stability of Trp in pure solutions and in parenteral AA formulations was evaluated with regard to typically used manufacturing processes, storage conditions and primary packaging. Therefore, thorough stability studies on Trp solutions were conducted beforehand. The applied stressing method, i.e. steam sterilization by autoclave, are chemically seen relatively mild but showed to be efficient to induce Trp degradation in the presence of oxygen. Subsequent identification, separation and characterization were challenging due to similar substance properties, numerous stereoisomers and pairs of diastereomers found amongst them. However, the identified o-aminoacetophenone compounds, Kyn and NFK, are associated with photo reactivity and have photo-oxidizing properties. Thus, best possible protection from UV-light, together with strict oxygen expulsion, are the most important criteria to impede Trp degradation after autoclaving.
The identification of Trp degradation products was assisted by the compilation of a substance library, which included manifold reported and chemically plausible Trp degradation substances. The substances were classified for priority and their early or late-stage occurrence. The large number of possible substances and stereoisomers was narrowed down with the information retrieved from LC-UV/MS experiments. However, final identification was achieved by the synthesis of proposed substances as references. The following eight substances were characterized as Trp degradation substances: Kyn, NFK and three pairs of diastereomers R,R/R,S DiOia, R,R/R,S Oia and cis/trans PIC. Fig. 33 shows the proposed degradation pathway and demonstrates the close chemical relationship, which may be an explanation for the conversion of some substances into each other during the storage period. The proposed pathway brings together the results of different Trp stability and stressing studies, respectively [89, 94, 97, 98, 103, 133]. To our knowledge, the simultaneous formation of the identified degradation substances has not been reported before and especially not under the stressing conditions applied.
The application of a traditional RP-HPLC method was compared to two developed IP-HPLC methods and a RP-HPLC methods using a modified perfluorinated column. Orthogonal analyses methods and especially the combination of UV and MS detection are necessary in order to indicate potentially undetected degradation substances. Main evaluation criteria were the separation performance, analyses time, reproducibility and feasibility. The best results upon assessment of all Trp degradation products, in both; pure Trp solutions and pharmaceutical formulations, were obtained by a traditional RP-HPLC. The optimized method was validated according to ICH guidelines Q2(R1) and meets the criteria of a stability-indicating HPLC-UV method. The validated method has a sufficient separation performance with an adequate selectivity indicating the Trp degradation substances next to each other and next to other AAs in finished pharmaceutical formulations.
The detailed knowledge of Trp degradation and the method presented may be transferred practically to the pharmaceutical industry processing Trp-containing products. In general, the findings might contribute to the quality management of such pharmaceutical products during
manufacturing and storage. Additionally, the study results provide basic information for the establishment of an impurity consideration following the ICH guidelines Q3B (R2) (impurities in new drug products) for products containing Trp. However, further development of the method applying more sophisticated detectors or more potent HPLC techniques like e.g. UHPLC and the implication of more sensitive (MS) detectors like ToF-MS would be advantageous with regard to economic and practical aspects.
Successful formulation development of novel, particularly organic APIs of low molecular weight as candidates for ground-breaking pharmaceutical products is a major challenge for the pharmaceutical industry because of the poor aqueous solubility of most of these compounds.
The hit identification strategies of drug development in use today apply high throughput screening techniques for the investigation of thousands of substances. This approach led to a systematical increase in molecular weight and lipophilicity and a decrease of water solubility of lead compounds reaching market access.
The high lipophilicity causes an excellent permeability of the compounds which favours the absorption process from the small intestine, but it causes a decrease of water-solubility. It becomes evident that an adequate aqueous solubility is necessary for absorption of the API from the gastrointestinal fluids into the systemic circulation and hence for efficacy of the pharmaceutical product. Only an dissolved API is getting absorbed and becomes efficacious. The precipitated proportion is resigned directly. Therefore, the development of an individual formulation aligning the physicochemical characteristics is necessary for every API to produce supersaturated solutions in the small intestine and to reach an adequate bioavailability after absorption into the systemic circulation.
In this thesis a specific formulation development was investigated for two exemplary poorly water-soluble APIs to replace the empirical approach often used today. The basic tyrosine-kinase inhibitor imatinib and six different acetylated amino acids were transferred into ILs. As compared to the free base and the mesylate salt, which is marketed by Novartis AG as Gleevec®, the dissolution rate as well as the supersaturation time was increased significantly. By changing the mesylate anion with its potential genotoxic risks, the total toxicity of the drug product could be decreased. The amorphous ILs proved adequate stability under forcing conditions and there was no recrystallization of the free base observed. The amorphous character of the ILs caused an increased amount of water vapour sorption which can be compensated by special packaging materials. Taken together, the presentation of imatinib as an IL is intended for oral administration as a tablet and can cause a reduction of dose because of the increased solubility. Therefore, the occurrence of side effects can be reduced as compared to Gleevec®. If there is actually an increased bioavailability to observe, has to be proved by the execution of animal trials.
The novel NOX inhibitor VAS3947 is intended for the treatment of endothelial dysfunctions causing diseases like heart failure and stroke. The compounds poor aqueous solubility hindered further clinical development so far and make the drug candidate to remain in a very early stage of the drug development process. Therefore, different formulation concepts were evaluated in this study:
An amorphous solid dispersion prepared from VAS3947 and Eudragit® L100 by means of spray drying was able to increase the dissolution rate and solubility of the compound significantly, but with the accomplished kinetic solubility being in the low µM range it is not possible to reach therapeutic plasma concentrations.
In contrast, the incorporation into cyclodextrins resulted in an 760-fold increased solubility. Different cyclodextrins were evaluated. Especially the lipophilic derivatives of the β-cyclodextrin showed to be the most adequate excipients. The incorporation of the API into the cyclodextrin cavity was proved by means of NMR spectroscopy. Additionally, a formulation of VAS3947 and hydroxypropyl-β-cyclodextrin was prepared. This formulation is intended for the intravenous application during animal trials, which have to be conducted to get to know the pharmacokinetics of VAS3947. This formulation reached a concentration of 1 mg/mL spending striking protection of VAS3947 against degradation.
Presentation of VAS3947 as a microemulsion system led also to increase the aqueous solubility of the compound, but not in the same extent as the cyclodextrin formulation. Beside the formulation development a physicochemical characterization was performed to get to know important parameters such as log P and pKa values of VAS3947. An HPLC method was developed and validated to analyse the extent of solubility improvement.
A major issue of the compound VAS3947 and all related triazolopyrimidine derivatives, developed by Vasopharm GmbH, is the insufficient chemical stability because of presence of a hemiaminal moiety in the chemical structure. Stability investigations and an extensive biopharmaceutical characterization confirm the hindering of further clinical development by insufficient drug stability and high cytotoxicity. Poor aqueous solubility is an additional disadvantage which can be handled by a concerted formulation development.
Es gibt viele Hinweise, dass G-Protein-gekoppelte Rezeptoren bei ihrer Aktivierung durch einen Agonisten ligandenselektive Konformationen eingehen. Ein tatsächlichen Beleg hierfür konnte bisher in lebenden Zellen noch nicht erbracht werden. Zu diesem Zweck wurde in dieser Arbeit ein Fluoreszenz-Resonanz-Energie-Transfer (FRET)-basierter Ansatz gewählt, um ligandenselektive Konformationen in der dritten intrazellulären Schleife des α2a-adrenergen Rezeptors (α2a-AR) in lebenden Zellen darzustellen. Dazu wurden Rezeptorsensoren erstellt, welche jeweils ein CFP am Ende des C-Terminus trugen und in der dritten intrazellulären Schleife an verschiedenen Stellen mit einem Tetracysteinmotiv versehen wurden. Drei Konstrukte wurden verglichen, die das Tetracysteinmotiv N-terminal in der Nähe der Transmembrandomäne V (I3-N), in der Mitte der dritten intrazellulären Schleife (I3-M) beziehungsweise C-terminal in der Nähe der Transmembrandomäne VI (I3-C) trugen. Die drei Rezeptorsensoren unterschieden sich hinsichtlich ihrer Ligandenbindung sowie ihrer G-Proteinaktivierung nicht vom Wildtyp α2a-AR. Durch das Tetracysteinmotiv ist es möglich, den Rezeptor spezifisch mit dem niedermolekularen Fluorophor FlAsH (fluorescein arsenical hairpin binder) zu markieren, welcher als Akzeptor für den Donor CFP in FRET-Experimenten dient. Die Änderung des FRET-Signals zwischen den beiden Fluorophoren, das durch den vollen Agonist Norepinephrin ausgelöst wurde, war bei allen drei Rezeptorsensoren vergleichbar. Der starke partielle Agonist Clonidin war ebenfalls in der Lage, in allen drei Konstrukten ein ähnliches FRET-Signal hervorzurufen. Dagegen zeigte der partielle Agonist Dopamin an dem Konstrukt I3-N ein signifikant schwächeres Signal, als an I3-C. Die schwachen partiellen Agonisten Octopamin und Norphenephrin konnten an den Konstrukten I3-N und I3-M keine Änderung des FRET-Signals bewirken, wobei an I3-C eine deutliche Signaländerung detektiert wurde. Dies legt nahe, dass die Transmembrandomäne V bei der Aktivierung des Rezeptors eine kleinere Bewegung eingeht als die Transmembrandomäne VI, und bestätigt damit ein auf Röntgenstrukturanlysen basierendes Modell der Rezeptorbewegung. Außerdem wurden die Aktivierungskinetiken für die Agonisten Norepinephrin und Dopamin verglichen. Hierbei konnte gezeigt werden, dass die durch Norepinephrin ausgelöste Bewegung an allen beobachteten Punkten gleich schnell war. Im Gegensatz dazu aktivierte Dopamin I3-C und I3-M ca. 1,5-mal langsamer, als Norepinephrin. Für das I3-N Konstrukt wurde sogar eine 3-mal langsamere Aktivierung gemessen. Diese Daten zeigen, dass unterschiedliche Agonisten in der dritten intrazellulären Schleife spezifische Konformationen auslösen können. Die Untersuchungen zur Rezeptorbewegung im ersten Teil dieser Arbeit wurde mit dem kleinen Fluorophor FlAsH in Kombination mit einer großen GFP-Variante durchgeführt. Im zweiten Teil dieser Arbeit wurde eine Methode entwickelt, bei der es möglich ist Proteine spezifisch mit beiden kleinen Fluorophoren FlAsH und ReAsH in einer lebenden Zelle zu markieren. Hierfür wurden zwei Tetracysteinmotive, CCPGCC und FLNCCPGCCMEP, gewählt, an die beide kleine Fluorophore kovalent binden. Durch Verdrängungsexperimente mit BAL konnte gezeigt werden, dass FlAsH für beide Motive eine dreifach höhere Affinität besitzt, als ReAsH. Dabei besitzt das FLNCCPGCCMEP-Motiv jedoch eine dreifach höhere Affinität zu dem jeweiligen Fluorophor besitzt als CCPGCC. Durch Ausnutzung dieser Affinitätsunterschiede konnte ein Protokoll entwickelt werden, mit dem es möglich ist, beide Motive in einer Zelle zu markieren. Dabei werden zunächst beide Motive mit ReAsH markiert. Durch anschließendes Waschen mit einer geeigneten Konzentration von BAL wird das ReAsH ausschließlich von der CCPGCC-Sequenz verdrängt, wohingegen die FLNCCPGCCMEP-Sequenz mit ReAsH markiert bleibt. Die nun unbesetzte CCPGCC-Sequenz kann dann anschließend mit FlAsH markiert werden, ohne dabei die Bindung des ReAsH an die FLNCCPGCCMEP-Sequenz zu beeinflussen. Um die Funktionalität dieses Protokolls zu überprüfen, sollten zwei verschiedene Proteine mit unterschiedlicher subzellulärer Lokalisation in einer lebenden Zelle spezifisch mit jeweils einem kleinen Fluorophor markiert werden. Hierzu wurden ein PTH-Rezeptor, in dem im C-Terminus die FLNCCPGCCMEP-Sequenz eingebracht wurde, mit ReAsH und ein β-Arrestin-2, dem die CCPGCC-Sequenz eingebracht wurde, in Zellen co-exprimiert und gemäß dem Protokoll mit FlAsH und ReAsH markiert. Beide Proteine konnten spezifisch markiert werden, wobei der mit ReAsH markierte PTH-Rezeptor eine deutliche Lokalisation in der Zellmembran zeigte. Durch sequentielle Exzitation konnte in der gleichen Zelle das zytosolisch lokalisierte, mit FlAsH markierte β-Arrestin-2 detektiert werden. Wurden die so markierten Zellen mir 1 µM PTH stimuliert, wurde das FlAsH-markierte β-Arrestin-2 an die Zellmembran rekrutiert. Somit konnte durch die Entwicklung dieses Protokolls eine duale spezifische Markierung von Proteinen mit zwei kleinen Fluorophoren zu innerhalb einer Zelle erreicht werden.
An important kinetic parameter for drug efficacy is the residence time of a compound at a drug target, which is related to the dissociation rate constant koff. For the essential antimycobacterial target InhA, this parameter is most likely governed by the ordering of the flexible substrate binding loop (SBL). Whereas the diphenyl ether inhibitors 6PP and triclosan (TCL) do not show loop ordering and thus, no slow-binding inhibition and high koff values, the slightly modified PT70 leads to an ordered loop and a residence time of 24 minutes. To assess the structural differences of the complexes from a dynamic point of view, molecular dynamics (MD) simulations with a total sampling time of 3.0 µs were performed for three ligand-bound and two ligand-free (perturbed) InhA systems. The individual simulations show comparable conformational features with respect to both the binding pocket and the SBL, allowing to define five recurring conformational families. Based on their different occurrence frequencies in the simulated systems, the conformational preferences could be linked to structural differences of the respective ligands to reveal important determinants of residence time. The most abundant conformation besides the stable EI* state is characterized by a shift of Ile202 and Val203 toward the hydrophobic pocket of InhA. The analyses revealed potential directions for avoiding this conformational change and, thus, hindering rapid dissociation: (1) an anchor group in 2'-position of the B-ring for scaffold stabilization, (2) proper occupation of the hydrophobic pocket, and (3) the introduction of a barricade substituent in 5'-position of the diphenyl ether B-ring.
Serum half-life elongation as well as the immobilization of small proteins like cytokines is still one of the key challenges for biologics. This accounts also for cytokines, which often have a molecular weight between 5 and 40 kDa and are therefore prone to elimination by renal filtration and sinusoidal lining cells. To solve this problem biologics are often conjugated to poly(ethylene glycol) (PEG), which is the gold standard for the so called PEGylation. PEG is a synthetic, non-biodegradable polymer for increasing the hydrodynamic radius of the conjugated protein to modulate their pharmacokinetic performance and prolong their therapeutic outcome. Though the benefits of PEGylation are significant, they also come with a prize, which is a loss in bioactivity due to steric hindrance and most often the usage of heterogeneous bioconjugation chemistries. While PEG is a safe excipient in most cases, an increasing number of PEG related side-effects, such as immunological responses like hypersensitivity and accelerated blood clearance upon repetitive exposure occur, which highlights the need for PEG alternative polymers, that can replace PEG in such cases.
Another promising method to significantly prolong the residence time of biologics is to immobilize them at a desired location. To achieve this, the transglutaminase (TG) Factor XIIIa (FXIIIa), which is an important human enzyme during blood coagulation can be used. FXIIIa can recognize specific peptide sequences that contain a lysine as substrates and link them covalently to another peptide sequence, that contains a glutamine, forming an isopeptide bond. This mechanism can be used to link modified proteins, which have a N- or C-terminal incorporated signal peptide by mutation, to the extracellular matrix (ECM) of tissues.
Additionally, both above-described methods can be combined. By artificially introducing a TG recognition sequence, it is possible to attach an azide group containing peptide site-specifically to the TG, recognition sequence. This allows the creation of a site-selective reactive site at the proteins N- or C-terminus, which can then be targeted by cyclooctyne functionalized polymers, just like amber codon functionalized proteins.
This thesis has focused on the two cytokines human Interferon-α2a (IFN-α2a) and human, as well as murine Interleukin-4 (IL-4) as model proteins to investigate the above-described challenges. IFN-α2a has been chosen as a model protein because it is an approved drug since 1986 in systemic applications against some viral infections, as well as several types of cancer. Furthermore, IFN-α2 is also approved in three PEGylated forms, which have different molecular weights and use different conjugation techniques for polymer attachment. This turns it into an ideal candidate to compare new polymers against the gold standard PEG. Interleukin-4 (IL-4) has been chosen as the second model protein due to its similar size and biopotency. This allows to compare found trends from IFN-α2a with another bioconjugate platform and distinguish between IFN-α2a specific, or general trends. Furthermore, IL-4 is a promising candidate for clinical applications as it is a potent anti-inflammatory protein, which polarizes macrophages from the pro-inflammatory M1 state into the anti-inflammatory M2 state.
Red fruit oil (RFO) can be extracted from fruits of Pandanus conoideus, Lam., an endogenous plant of Papua, Indonesia. It is a commonly used essential original traditional medicine. By applying a newly developed quantitative \(^1\)H NMR (qNMR) spectroscopy method for quality assessment, a simultaneous determination of the saponification value (SV), acid value (AV), ester value (EV), and iodine value (IV) in RFO was possible. Dimethyl sulfone (DMSO\(_2\)) was used as an internal standard. Optimization of NMR parameters, such as NMR pulse sequence, relaxation delay time, and receiver gain, finally established the \(^1\)H NMR-based quantification approach. Diagnostic signals of the internal standard at δ = 2.98 ppm, SV at δ = 2.37–2.20 ppm, AV at δ = 2.27–2.20 ppm, EV at δ = 2.37–2.27 ppm, and IV at δ = 5.37–5.27 ppm, respectively, were used for quantitative analysis. The method was validated concerning linearity (R\(^2\) = 0.999), precision (less than 0.83%), and repeatability in the range 99.17–101.17%. Furthermore, this method was successfully applied to crude RFO, crude RFO with palmitic and oleic acid addition, and nine commercial products. The qNMR results for the respective fat values are in accordance with the results of standard methods, as can be seen from the F- and t-test (< 1.65 and < 1.66, respectively). The fundamental advantages of qNMR, such as its rapidity and simplicity, make it a feasible and existing alternative to titration for the quality control of RFO.
Polymer-Biokonjugationen, vornehmlich mit dem Goldstandard PEG, führen zu einer verbesserten Pharmakokinetik, beeinflussen aber auch die konformative Stabilität von Proteinen. Bisherige Mutationsstudien, in denen überwiegend (Asn)PEG4 -Konjugate der Beta-faltblattstrukturreichen, humanen Pin 1 WW-Domäne untersucht wurden, postulieren auf einer Proteindesolvatation beruhende Stabilisierungsmechanismen: eine Stärkung intramolekularer Salzbrücken und NH-pi-Bindungen, sowie entropisch günstige Wasserverdrängungen um apolare Aminosäuren und Hydroxylgruppen. Ziel dieser Arbeit ist es, die Protein-Polymer-Dynamik auf molekularer Ebene zu charakterisieren, um damit rationale Ansätze zum Design neuer Biokonjugate voranzutreiben und mögliche PEG-Alternativen zu etablieren. Hierzu wurde eine Vielzahl an Deskriptoren mittels Molekulardynamik-Simulationen der WW-Konjugate gewonnen und mit publizierten Stabilitätsdaten in multivariaten Regressions- und logistischen Klassifikationsmodellen korreliert. Die gewonnenen QSPR-Modelle decken im Vergleich zu einer bereits publizierten, kristallstrukturbasierten Richtlinie einen größeren und strukturell vielfältigeren Datensatz an Konjugaten ab und zeigen gleichzeitig, auch für ein Konjugat der Src SH3-Domäne, eine deutlich verbesserte Leistung. Die Modelldeskriptoren beschreiben sowohl eine Modulation der Solvatation als auch Protein-Polymer-Interaktionen. Metadynamik-Simulationen zeigten zudem die Polymerdynamik während einer partiellen Proteinentfaltung auf. Mithilfe weiterer Simulationen von Konjugaten des alpha-helikalen Her2-Affibodys wurde die Dynamik von PEG und verschiedener Alternativen (LPG, PEtOx, PMeOx) systematisch studiert. PEG interagierte mit positiv geladenen Lysinen und Argininen in der Nähe hydrophober Aminosäuren. LPG zeigte zusätzliche Wechselwirkungen der Hydroxylgruppen mit Aspartaten und Glutamaten. POx-Polymere interagierten mit Phenylalaninen, Tyrosinen und über Carbonylgruppen mit HB-Donatoren. Größere Konjugate (10 - 50 kDa PEG/LPG/PEtOx) des antiviralen Biologikums Interferon-alpha2a wurden mittels gaußbeschleunigter MDs und einer CG-Simulation analysiert. Charakteristische Wechselwirkungspartner stimmten mit den Beobachtungen zu Oligomer-Konjugaten überein. In Einklang mit experimentellen Daten der Kooperationspartner zu den 10-kDa-Varianten deuteten zusätzliche Constrained-Network-Analysen, welche die Proteinflexibilität evaluieren, auf eine thermische Destabilisierung hin. Die Bioaktivität der untersuchten Konjugate wurde weiterhin erfolgreich mit den Gyrationsdurchmessern der modellierten Strukturen korreliert.
Naturally occurring compounds such as sesquiterpenes and sesquiterpenoids (SQTs) have been shown to modulate GABA\(_{A}\) receptors (GABA\(_{A}\)Rs). In this study, the modulatory potential of 11 SQTs at GABA\(_{A}\)Rs was analyzed to characterize their potential neurotropic activity. Transfected HEK293 cells and primary hippocampal neurons were functionally investigated using electrophysiological whole-cell recordings. Significantly different effects of β-caryophyllene and α-humulene, as well as their respective derivatives β-caryolanol and humulol, were observed in the HEK293 cell system. In neurons, the concomitant presence of phasic and tonic GABA\(_{A}\)R configurations accounts for differences in receptor modulation by SQTs. The in vivo presence of the γ\(_{2}\) and δ subunits is important for SQT modulation. While phasic GABA\(_{A}\) receptors in hippocampal neurons exhibited significantly altered GABA-evoked current amplitudes in the presence of humulol and guaiol, negative allosteric potential at recombinantly expressed α\(_{1}\)β\(_{2}\)γ\(_{2}\) receptors was only verified for humolol. Modeling and docking studies provided support for the binding of SQTs to the neurosteroid-binding site of the GABA\(_{A}\)R localized between transmembrane segments 1 and 3 at the (\(^{+}\)α)-(\(^{-}\)α) interface. In sum, differences in the modulation of GABA\(_{A}\)R isoforms between SQTs were identified. Another finding is that our results provide an indication that nutritional digestion affects the neurotropic potential of natural compounds.
Plant extracts from Cecropia genus have been used by Latin-American traditional medicine to treat metabolic disorders and diabetes. Previous reports have shown that roots of Cecropia telenitida that contains serjanic acid as one of the most prominent and representative pentacyclic triterpenes. The study aimed to isolate serjanic acid and evaluate its effect in a prediabetic murine model by oral administration. A semi-pilot scale extraction was established and serjanic acid purification was followed using direct MALDI-TOF analysis. A diet induced obesity mouse model was used to determine the impact of serjanic acid over selected immunometabolic markers. Mice treated with serjanic acid showed decreased levels of cholesterol and triacylglycerols, increased blood insulin levels, decreased fasting blood glucose and improved glucose tolerance, and insulin sensitivity. At transcriptional level, the reduction of inflammation markers related to adipocyte differentiation is reported.
μ‐Opioid receptors (μ‐ORs) play a critical role in the modulation of pain and mediate the effects of the most powerful analgesic drugs. Despite extensive efforts, it remains insufficiently understood how μ‐ORs produce specific effects in living cells. We developed new fluorescent ligands based on the μ‐OR antagonist E‐p‐nitrocinnamoylamino‐dihydrocodeinone (CACO), that display high affinity, long residence time and pronounced selectivity. Using these ligands, we achieved single‐molecule imaging of μ‐ORs on the surface of living cells at physiological expression levels. Our results reveal a high heterogeneity in the diffusion of μ‐ORs, with a relevant immobile fraction. Using a pair of fluorescent ligands of different color, we provide evidence that μ‐ORs interact with each other to form short‐lived homodimers on the plasma membrane. This approach provides a new strategy to investigate μ‐OR pharmacology at single‐molecule level.
Polyphenols exert beneficial effects in type 2 diabetes mellitus (T2DM). However, their mechanism of action remains largely unknown. Endothelial Akt-kinase plays a key role in the pathogenesis of cardiovascular complications in T2DM and therefore the modulation of its activity is of interest. This work aimed to characterize effects of structurally different polyphenols on Akt-phosphorylation (pAkt) in endothelial cells (Ea.hy926) and to describe structure-activity features. A comprehensive screening via ELISA quantified the effects of 44 polyphenols (10 µM) on pAkt Ser473. The most pronounced inhibitors were luteolin (44 ± 18%), quercetin (36 ± 8%), urolithin A (35 ± 12%), apigenin, fisetin, and resveratrol; (p < 0.01). The results were confirmed by Western blotting and complemented with corresponding experiments in HUVEC cells. A strong positive and statistically significant correlation between the mean inhibitory effects of the tested polyphenols on both Akt-residues Ser473 and Thr308 (r = 0.9478, p = 0.0003) was determined by immunoblotting. Interestingly, the structural characteristics favoring pAkt inhibition partially differed from structural features enhancing the compounds’ antioxidant activity. The present study is the first to quantitatively compare the influence of polyphenols from nine different structural subclasses on pAkt in endothelial cells. These effects might be advantageous in certain T2DM-complications involving over-activation of the Akt-pathway. The suggested molecular mode of action of polyphenols involving Akt-inhibition contributes to understanding their effects on the cellular level.
Purpose
Knowledge on Ruxolitinib exposure in patients with graft versus host disease (GvHD) is scarce. The purpose of this prospective study was to analyze Ruxolitinib concentrations of GvHD patients and to investigate effects of CYP3A4 and CYP2C9 inhibitors and other covariates as well as concentration-dependent effects.
Methods
262 blood samples of 29 patients with acute or chronic GvHD who were administered Ruxolitinib during clinical routine were analyzed. A population pharmacokinetic model obtained from myelofibrosis patients was adapted to our population and was used to identify relevant pharmacokinetic properties and covariates on drug exposure. Relationships between Ruxolitinib exposure and adverse events were assessed.
Results
Median of individual mean trough serum concentrations was 39.9 ng/mL at 10 mg twice daily (IQR 27.1 ng/mL, range 5.6-99.8 ng/mL). Applying a population pharmacokinetic model revealed that concentrations in our cohort were significantly higher compared to myelofibrosis patients receiving the same daily dose (p < 0.001). Increased Ruxolitinib exposure was caused by a significant reduction in Ruxolitinib clearance by approximately 50%. Additional comedication with at least one strong CYP3A4 or CYP2C9 inhibitor led to a further reduction by 15% (p < 0.05). No other covariate affected pharmacokinetics significantly. Mean trough concentrations of patients requiring dose reduction related to adverse events were significantly elevated (p < 0.05).
Conclusion
Ruxolitinib exposure is increased in GvHD patients in comparison to myelofibrosis patients due to reduced clearance and comedication with CYP3A4 or CYP2C9 inhibitors. Elevated Ruxolitinib trough concentrations might be a surrogate for toxicity.
In recent decades, our planet has undergone dramatic environmental changes resulting in the loss of numerous species. This contrasts with species that can adapt quickly to rapidly changing ambient conditions, which require physiological plasticity and must occur rapidly. The Western honeybee (Apis mellifera) apparently meets this challenge with remarkable success, as this species is adapted to numerous climates, resulting in an almost worldwide distribution. Here, coordinated individual thermoregulatory activities ensure survival at the colony level and thus the transmission of genetic material. Recently, we showed that shivering thermogenesis, which is critical for honeybee thermoregulation, depends on octopamine signaling. In this study, we tested the hypothesis that the thoracic neuro-muscular octopaminergic system strives for a steady-state equilibrium under cold stress to maintain endogenous thermogenesis. We can show that this applies for both, octopamine provision by flight muscle innervating neurons and octopamine receptor expression in the flight muscles. Additionally, we discovered alternative splicing for AmOARβ2. At least the expression of one isoform is needed to survive cold stress conditions. We assume that the thoracic neuro-muscular octopaminergic system is finely tuned in order to contribute decisively to survival in a changing environment.
1 Verlängerung der kardialen Repolarisationsdauer unter psychiatrischer Medikation bei gleichzeitigem genetischen Basisrisiko
Vielen Psychopharmaka wird eine repolarisationsverlängernde Wirkung zugeschrieben. Diese unerwünschte Arzneimittelwirkung, erkennbar an einer Verlängerung des QT-Intervalls im Elektrokardiogramm, ist in den vergangenen Jahren, aufgrund des Zusammenhanges mit lebensbedrohlichen Torsades-de-Pointes-Tachyarrhythmien, in den Fokus der klinischen Forschung gerückt. Aufgrund dieser Nebenwirkung werden viele gut wirksame Arzneimittel einer erneuten eingehenden Nutzen-Risiko-Analyse unterzogen und in manchen Fällen führte dies zu einer Limitierung der pharmakologischen Möglichkeiten.
Als Hauptmechanismus für eine Psychopharmaka-induzierte QT-Zeit-Verlängerung gilt die Blockade von kardialen Kaliumkanälen. Aber auch genetische Veränderungen unterschiedlicher kardialer Ionenkanäle gelten als Risikofaktoren, ebenso wie Effekte anderer ionenabhängiger Signalwege. Da Patienten mit genetischer Prädisposition ein defacto erhöhtes Risiko für eine pharmakologisch induzierte QT-Zeit-Verlängerung aufweisen, spricht man von reduzierter Repolarisationsreserve, mit erhöhtem Basislinienrisiko für kardiale Nebenwirkungen.
Ziel war es, über einen additiven genetischen Risikoscore eine Quantifizierung individueller Vulnerabilität zu erreichen und zu zeigen, dass dieses Risiko durch die Kontrolle von Medikamenten-Serumspiegeln modulierbar sein kann.
Aus einer prospektiven Studie, mit 2062 an endogener Psychose leidenden Patienten des Zentrums für Psychische Gesundheit des Universitätsklinikums Würzburg, wurden 392 Patienten (mittleres Alter bei Studieneinschluss 41,0 ± 15,0 Jahre, 36,2 % Frauen) rekrutiert. Primäres Einschlusskriterium für die angeknüpfte, retrospektive Studie war das Vorliegen einer Serumspiegelbestimmung der psychiatrischen Medikation binnen drei Tagen vor oder nach einer elektrokardiographischen Untersuchung (N = 392). Die den Einschlusskriterien entsprechenden 392 Patienten wurden daraufhin auf 62 Einzelpolymorphismen, die in Verbindung mit einer verlängerten QT-Zeit stehen, getestet und die Ergebnisse mit den patientenspezifischen Daten aus den elektrokardiographischen Untersuchungen korreliert.
Des Weiteren wurden, basierend auf vier großen Publikationen des internationalen „Cardiac Safety Consortium“ (77-79, 148), bekannte polygene Risikoscores, die diese Risikopolymorphismen enthalten, anhand des eigenen Patientenkollektivs berechnet und durch Korrelation mit der QT-Zeit überprüft. Diese Scores funktionieren jeweils nach einem Additionsmodell, bei dem nach unterschiedlicher Gewichtung das individuelle Risiko, das durch das Vorhandensein eines bekannten Risikopolymorphismus quantifizierbar wird, zu einem Gesamtrisiko aufsummiert wird.
Darüber hinaus ist das Patientenkollektiv auf einen Zusammenhang zwischen dem Serumspiegel der psychiatrischen Medikation und der QT-Zeit geprüft worden. Dazu wurde das Gesamtkollektiv in medikamentenspezifische Subgruppen unterteilt (Amitriptylin (N = 106), Clomipramin (N = 48), Doxepin (N = 53), Mirtazapin (N = 45), Venlafaxin (N = 50), Aripiprazol (N = 56), Clozapin (N = 127), Haloperidol (N = 41), Olanzapin (N = 37), Perazin (N = 47), Quetiapin (N = 119) und Risperidon (N = 106)).
Abschließend wurden die Subkollektive in einem kombinierten Rechenmodell daraufhin geprüft, ob Zusammenhänge zwischen den genetischen Risikoscores nach Strauss et al. (148) mit dem jeweiligen Medikamenten-Serumspiegel auf die QT-Zeit bestehen.
13 der 62 untersuchten Einzelpolymorphismen zeigten einen signifikanten Zusammenhang mit einer verlängerten Repolarisationsdauer. Ebenfalls korrelieren polygene Risikoscores einer verlängerten kardialen Repolarisation und erklären einen dabei signifikanten Anteil der Varianz. Die Ergebnisse der Literatur, bezüglich der Scores nach Pfeufer et al. (77) (R = 0,124, p = 0,014; N = 392), nach Noseworthy et al. (79) (R = 0,169; p = 0,001; N = 392), sowie nach Strauss et al. (148) (R = 0,199; p = 0,000; N = 392) konnten anhand des eigenen Kollektives reproduziert werden, wohingegen der Score von Newton-Cheh et al. (78) keinen signifikanten Zusammenhang mit der QT-Zeit zeigte (R = 0,029; p = 0,568; N = 392).
In der Subgruppenanalyse konnte ein stark vom Serumspiegel abhängiger, verlängernder Effekt auf die QT-Zeit für die Arzneistoffe Amitriptylin, Nortriptylin, Clomipramin, und Haloperidol nachgewiesen werden. Die Analyse der mit Amitriptylin behandelten Patienten (N = 106) ergab für Nortriptylin (F (1,104) = 5.986; p = .016, R = .233), als auch für den Summenspiegel aus Amitriptylin und Nortriptylin (F (1,104) = 4.408, p = .038, R = .202) einen signifikanten, nach Cohen einen mittelstarken Zusammenhang mit der QT-Zeit. Starke Effekte auf die QT-Zeit wurden im Zusammenhang mit den Serumspiegeln der Medikamente Clomipramin (F (1,46) = 39.589, p < .001, R = .680, N = 48) und Haloperidol (F (1,39) = 12.672, p = .001, korrigiertes R2= .245, N = 41) errechnet.
Ein kombiniertes Rechenmodell, das sowohl den Einfluss des jeweiligen Serumspiegels, als auch des genetischen Risikoscores nach Strauss et al. (148) berücksichtigte, erlaubte bei diesen Arzneistoffen eine signifikant höhere Varianzaufklärung der QT-Zeit, als die jeweiligen Effekte für sich genommen.
Die QT-Zeit gilt als erwiesenermaßen genauso abhängig von der individuellen genetischen Ausstattung, wie auch von Serumspiegeln potentiell als QT-verlängernd eingestufter Medikamente. Diese Effekte scheinen additiv verknüpfbar, so dass das von Roden et al. entwickelte Konzept der reduzierten Repolarisationsreserve (54) als bestätigt gelten darf. Die jeweiligen Einzeleffekte vom genetischen Risiko, sowie der Medikation haben zusammen einen größeren Einfluss auf die gemessenen QT-Zeit als für sich alleine genommen. Durch die Genetik lässt sich somit tatsächlich eine grobe vorab-Risikoabschätzung treffen. Dies könnte nach sorgfältiger Nutzen-Risiko-Analyse durch Kontrollen des EKGs und des Serumspiegels moduliert werden und somit vielfältigere therapeutische Möglichkeiten erhalten.
2 Entwicklung und Validierung einer Dried-Blood-Spot-Methode zum therapeutischen Drug Monitoring von Clozapin und Quetiapin
Die Technik der Extraktion und Analyse von Stoffen aus getrocknetem Blut ist bereits seit den 1960er Jahren bekannt, wurde bis zur jüngeren Vergangenheit aber eher zu diagnostischen Zwecken angewendet. Durch Fortschritte in der Analytik im Sinne ausgefeilterer Chromatographie und sensitiverer Detektion wurde das Verfahren der Dried-Blood-Spot-Analytik auch für die Spiegelbestimmung von Arzneistoffen interessant. So wurden auch im Bereich des Therapeutischen Drug Monitorings bereits Methoden, beispielsweise für Antibiotika, Antiepileptika, Virostatika und in jüngerer Zeit auch Antidiabetika publiziert. Die Vorteile in der Probenhandhabung und durch geringeren Aufwand bei der Blutentnahme sowie geringeres Probenentnahmevolumen werden durch weitere Fortschritte im Bereich der Analytik vordergründiger.
Ziel war es, ein Extraktionsverfahren zu entwickeln und zu validieren, dass die gemeinsame Quantifizierung der häufig verabreichten Antipsychotika Clozapin und Quetiapin aus einem einzelnen getrockneten Blutstropfen ermöglicht.
Die Extraktion mit einer Mischung aus 99 % Acetonitril und 1 % 1 M Salzsäure und anschließender HPLC-Analyse mit Säulenschaltung und photometrischer Detektion wurde nach den Richtlinien der Gesellschaft für toxikologische und forensische Chemie (GTFCh) (146) validiert. Somit gilt diese Methode in der Praxis als anwendbar und dürfte, nach Überprüfung der therapeutischen Bereiche für kapillares Vollblut im Vergleich zu den bereits definierten Bereichen für venöse entnommene Serumproben, Eingang in die klinische Praxis finden.
Background
Reproducibility of reported antibacterial activities of plant extracts has long remained questionable. Although plant-related factors should be well considered in serious pharmacognostic research, they are often not addressed in many research papers. Here we highlight the challenges in reproducing antibacterial activities of plant extracts.
Methods
Plants with reported antibacterial activities of interest were obtained from a literature review. Antibacterial activities against Escherichia coli and Klebsiella pneumoniae were tested using extracts’ solutions in 10% DMSO and acetone. Compositions of working solutions from both solvents were established using LC-MS analysis. Moreover, the availability of details likely to affect reproducibility was evaluated in articles which reported antibacterial activities of studied plants.
Results
Inhibition of bacterial growth at MIC of 256–1024 μg/mL was observed in only 15.4% of identical plant species. These values were 4–16-fold higher than those reported earlier. Further, 18.2% of related plant species had MICs of 128–256 μg/mL. Besides, 29.2% and 95.8% of the extracts were soluble to sparingly soluble in 10% DMSO and acetone, respectively. Extracts’ solutions in both solvents showed similar qualitative compositions, with differing quantities of corresponding phytochemicals. Details regarding seasons and growth state at collection were missing in 65% and 95% of evaluated articles, respectively. Likewise, solvents used to dissolve the extracts were lacking in 30% of the articles, whereas 40% of them used unidentified bacterial isolates.
Conclusion
Reproducibility of previously reported activities from plants’ extracts is a multi-factorial aspect. Thus, collective approaches are necessary in addressing the highlighted challenges.
Poor or variable oral bioavailability is of major concern regarding safety and efficacy for the treatment of patients with poorly water-soluble drugs (PWSDs). The problem statement of this work involves a pharmaceutical development perspective, the physicochemical basis of the absorption process and physiological / biopharmaceutical aspects. A methodology was developed aiming at closing the gap between drug liberation and dissolution on the one hand and the appearance of drug in the blood on the other. Considering what is out of control from a formulation development perspective, a clear differentiation between bioavailability and bioaccessibility was necessary. Focusing on the absorption process, bioaccessibility of a model compound, a poorly soluble but well permeable weak base, was characterized by means of flux across artificial biomimetic membranes. Such setups can be considered to reasonably mimic relevant oral absorption resistances in vitro in terms of diffusion through an unstirred water layer (UWL) and a lipidic barrier. Mechanistic understanding of the driving force for permeation was gained by differentiating drug species and subsequently linking them to the observed transfer rates using a bioaccessibility concept. The three key species that need to be differentiated are molecularly dissolved drug, drug associated in solution with other components (liquid reservoir) and undissolved drug (solid reservoir). An innovative approach to differentiate molecularly dissolved drug from the liquid reservoir using ultracentrifugation in combination with dynamic light scattering as control is presented. A guidance for rational formulation development of PWSDs is elaborated based on the employed model compound. It is structured into five guiding questions to help drug formulation scientists in selecting drug form, excipients and eventually the formulation principle. Overall, the relevance but also limitations of characterizing bioaccessibility were outlined with respect to practical application e.g. in early drug formulation development.

Purpose: A new PET radiotracer \(^{18}\)F-AF78 showing great potential for clinical application has been reported recently. It belongs to a new generation of phenethylguanidine-based norepinephrine transporter (NET)-targeting radiotracers. Although many efforts have been made to develop NET inhibitors as antidepressants, systemic investigations of the structure–activity relationships (SARs) of NET-targeting radiotracers have rarely been performed. Methods: Without changing the phenethylguanidine pharmacophore and 3-fluoropropyl moiety that is crucial for easy labeling, six new analogs of \(^{18}\)F-AF78 with different meta-substituents on the benzene-ring were synthesized and evaluated in a competitive cellular uptake assay and in in vivo animal experiments in rats. Computational modeling of these tracers was established to quantitatively rationalize the interaction between the radiotracers and NET. Results: Using non-radiolabeled reference compounds, a competitive cellular uptake assay showed a decrease in NET-transporting affinity from meta-fluorine to iodine (0.42 and 6.51 µM, respectively), with meta-OH being the least active (22.67 µM). Furthermore, in vivo animal studies with radioisotopes showed that heart-to-blood ratios agreed with the cellular experiments, with AF78(F) exhibiting the highest cardiac uptake. This result correlates positively with the electronegativity rather than the atomic radius of the meta-substituent. Computational modeling studies revealed a crucial influence of halogen substituents on the radiotracer–NET interaction, whereby a T-shaped π–π stacking interaction between the benzene-ring of the tracer and the amino acid residues surrounding the NET binding site made major contributions to the different affinities, in accordance with the pharmacological data. Conclusion: The SARs were characterized by in vitro and in vivo evaluation, and computational modeling quantitatively rationalized the interaction between radiotracers and the NET binding site. These findings pave the way for further evaluation in different species and underline the potential of AF78(F) for clinical application, e.g., cardiac innervation imaging or molecular imaging of neuroendocrine tumors.
Die vorliegende Arbeit befasst sich mit der Suche nach neuen Leitstrukturen im Bereich der 4-Chinolone mit dem Verfahren der Random Chemistry. Die zugrunde liegende Idee hinter diesem Verfahren besteht darin, neue, potentiell wirksamere Strukturen durch die zufällige Erzeugung einer Substanzbibliothek zu erhalten. Die Substanzbibliothek entsteht dabei durch die Kombination bzw. Reaktion einer mäßig aktiven Ausgangsverbindung mit einem Initiator, der eine Zufallsreaktion in Gang setzt. Vorangegangene Untersuchungen dazu belegten, dass bei der Nutzung von γ-Strahlen als auslösende Spezies vornehmlich durch Radiolyse des Lösungsmittels erzeugte Radikale für die Bildung neuartiger Substanzen verantwortlich waren. Diese als Initiator dienenden Radikale können auch mit herkömmlichen Radikalstartern wie dem Fentons Reagenz erzeugt werden. Fentons Reagenz erweist sich dabei als deutlich kostengünstigere und einfacher zu handhabende Alternative, die ohne jeglichen technischen Aufwand genutzt werden kann. Durch die Reaktion von Wasserstoffperoxid und Eisen(II)-Ionen entstehen Hydroxylradikale, die als hochreaktive Spezies durch die Addition an Doppelbindungen oder durch die Abstraktion von Wasserstoffradikalen und die dadurch ausgelösten Sekundärreaktionen eine Vielzahl neuer Verbindungen erzeugen. Zur Leitstruktursuche wurden als Ausgangssubstanzen zum einen ein Vertreter der 6-Fluor-7-(piperazin-1-yl)-chinolone (1) und zum anderen das 4-Chinolon-Grundgerüst (2) gewählt, um sowohl den Einfluss möglicher Strukturvariationen am Chinolongrundkörper als auch an möglichen Substituenten zu ermitteln. Aus den nach der Umsetzung mit Fentons Reagenz entstandenen Substanzbibliotheken konnten zunächst Fraktionen erhalten werden, die bei der Testung gegen Trypanosoma brucei brucei Aktivitäten mit geringeren IC50-Werten als die Ausgangschinolone zeigten. Die Strukturen der mittels HPLC isolierten Reinsubstanzen wurden aufgeklärt und ihre biologische Wirkung ermittelt. Auf diese Weise konnten ein Chinolonderivat mit N-(2-Aminoethyl)formamid-Rest in Position 7 (Frk1-2c) sowie ein Derivat mit (2-((Methyl-amino)methoxy)ethyl)amino-Rest in Position 7 und einer Hydroxylgruppe in Position 6 (Frk1-1c) identifiziert werden, die eine antitrypanosomale Aktivität mit einem IC50-Wert von 20.69 μM bzw. 17.29 μM aufweisen. Neusynthetisierte Chinolone mit einem amidofunktionalisiertem Piperazinring in Position 7 zeigten eine noch bessere Aktivität gegen Trypanosoma brucei brucei, welche durch Amidierung der Carbonsäure in 3-Stellung noch weiter gesteigert werden konnte. Die Suche nach neuen Leitstrukturen mit Hilfe von Fentons Reagenz offenbarte, dass ein Großteil der in den Substanzbibliotheken gebildeten neuen Strukturen literaturbekannten Metaboliten der Fluorchinolone ähnelt. Da annähernd 50% der möglichen Wirkstoffkandidaten an einer zu geringen Bioverfügbarkeit oder aufgrund ihrer toxischen Metabolite scheitern, ist es sinnvoll bereits in einem möglichst frühen Forschungsstadium auch Parameter wie Resorption, Distribution, Metabolismus, Elimination und Toxikologie (ADMET-Parameter) einer Substanz zu berücksichtigen bzw. zu ermitteln. Deshalb wurde in einem weiteren Teil dieser Arbeit untersucht, ob Fentons Reagenz zur Erzeugung des metabolischen Profils neuer biologisch aktiver Substanzen dienen kann, um damit frühzeitig auch auf mögliche Toxizitäten der Metabolite einer Substanz schließen zu können. Dazu wurden die drei bekannten Antiinfektiva, Cinoxacin, Ciprofloxacin und Linezolid mit Fentons Reagenz umgesetzt und die entstandenen Substanzbibliotheken nach literaturbekannten Metaboliten der jeweiligen Ausgangssubstanz gescreent. Bei einer ausreichenden Löslichkeit der Ausgangssubstanz konnten so vor allem die Metabolite erzeugt werden, die durch Hydroxylierung, Oxidation oder Hydrolyse auch bei der Verstoffwechslung durch Cytochrom-P450 erhalten werden. Da der Nachweis der Bildung literaturbekannter Metabolite durch Fentons Reagenz gelang, ist es nun möglich, einen Teil der potenziellen Metabolite neuer biologisch aktiver Substanzen bereits im Vorfeld, ohne die Nutzung von humanem Lebergewebe zu identifizieren.
Heart failure is one of the growing causes of death especially in developed countries due to longer life expectancy. Although many pharmacological and instrumental therapeutic approaches have been introduced for prevention and treatment of heart failure, there are still limitations and challenges. Nuclear cardiology has experienced rapid growth in the last few decades, in particular the application of single photon emission computed tomography (SPECT) and positron emission tomography (PET), which allow non-invasive functional assessment of cardiac condition including neurohormonal systems involved in heart failure; its application has dramatically improved the capacity for fundamental research and clinical diagnosis. In this article, we review the current status of applying radionuclide technology in non-invasive imaging of neurohormonal system in the heart, especially focusing on the tracers that are currently available. A short discussion about disadvantages and perspectives is also included.
Quantitative Bestimmungen Anhand verschiedener Substanzen konnte im Rahmen dieser Arbeit gezeigt werden, dass die NMR-Spektroskopie in der Lage ist, Verunreinigungen von Arzneistoffen zu quantifizieren. Für das Antidepressivum Fluvoxamin ist im Arzneibuch eine Ionenpaarchroma-tographie vorgeschrieben, um die Verunreinigung des wirksamen E-Isomers durch das Z-Isomer zu quantifizieren. Ionenpaarchromatographischen Methoden mangelt es häufig an der Robustheit. Eine quantitative Auswertung der NMR-Spektren einer Mischung beider Isomere ist ohne aufwändige Probenvorbereitung möglich. In den 1H-NMR-Spektren der Mischung sind die Signale der Was-serstoffe beider Isomere an Position 2 gut voneinander getrennt. Werden diese quantitativ ausgewertet, dann ist es nach Optimierung insbesondere hinsichtlich der T1-Relaxationszeit möglich, den Anteil des Z-Isomers auf 0,2 % zu begrenzen. Auch für die Bestimmung der Abbauprodukte des Perphenazinenantats konnte gezeigt werden, dass die qNMR eine geeignete Methode darstellt. Perphenazine-nantat kann durch Esterhydrolyse gespalten werden. Zur Auswertung der 1H-NMR-Spektren wird der Vergleich der Integralflächen der Signale der Wasserstoffe an Position 21 des Perphenazins mit dem zusammenfallenden Signal der Wasserstoffe an Position 11 beider Substanzen herangezogen. Es konnte sowohl Perphenazin als Abbauprodukt des Esters als auch Perphena-zinenantat in Perphenazin quantifiziert werden. Zusätzlich kann der Bereich der aromatischen Wasserstoffe zu einer Aussage über die Oxidation genutzt werden. Bei der Oxidation des Schwefels im Phenothiazinring zum Sulfoxid und zum Sul-fon ändern sich die chemischen Verschiebungen der Wasserstoffkerne in diesem Ringsystem. Dadurch wird eine halbquantitative Aussage ermöglicht. Schließlich konnten die beiden Epimere Chinin und Chinidin jeweils als Verunrei-nigung des anderen Chinaalkaloides quantifiziert werden. Auch in diesem Fall lie-gen in den 1H-NMR-Spektren in DMSO-d6 von Mischungen dieser beiden Verbin-dungen Signale weit genug auseinander, um eine Quantifizierung zu ermöglichen. In beiden Fällen, der Bestimmung von Chinidin in Chinin und von Chinin in Chini-din konnte dies auf einem Niveau von 2,5% geschehen, was den Anforderungen der Arzneibücher entspricht. Gentamicinsulfat Die 1H-NMR-Spektroskopie wurde ebenfalls zur Charakterisierung der Zusam-mensetzung des Antibiotkums Gentamicin eingesetzt. Gentamicin, das fermentativ aus Micromonospora purpurea gewonnen wird, besteht aus verschiedenen Haupt- und Nebenkomponenten, deren Zusammensetzung je nach Fermentationsbedingungen schwankt. Nach einer Reihe von Todesfällen im Zusammenhang mit der Anwendung des Antibiotikums Gentamicin in den USA wurde vermutet, dass diese auf verschiede-ne Verunreinigungen zurückzuführen sind. In der aktuellen Arzneibuch-Monographie wird eine HPLC-Methode beschrieben, die zwar die Hauptkomponenten quantifizieren kann, aber nicht alle Nebenkomponenten gut abtrennt. Auch ist die gesamte Elutionszeit sehr lang, so dass spät eluierende Substanzen breite Peaks zeigen. Außerdem ist der benutzte gepulste amperometrische Detektor sehr empfindlich und die Methode insgesamt daher wenig robust. Unter Zuhilfenahme von ein- und zweidimensionalen Standardmesstechniken sowie selektiver TOCSY-Messungen konnten alle Signale in den 1H- und 13C-NMR-Spektren der Haupt- und Nebenkomponenten von Gentamicin vollständig zugeordnet werden. Dabei zeigte sich, dass der Bereich der anomeren Wasserstoffe sehr gut geeignet ist, Aussagen über die Reinheit und über das Verhältnis der Hauptkomponenten zueinander treffen zu können. In dem in der Abbildung gezeigten Ausschnitt aus einem 400 MHz-1H-NMR-Spektrum ist eine Integration der H20-Signale der Hauptkomponenten aufgrund mangelnder Trennung nicht möglich. Diese ist jedoch in 600 MHz-Spektren möglich. Auf diese Weise können die Verhältnisse der Hauptkomponenten zueinander bestimmt werden. Die so erhaltenen Ergebnisse zeigen eine sehr gute Übereinstimmung mit den aus einer MEKC-Trennung erhaltenen Daten. Das zeigt die sehr gute Ergänzung dieser beiden Methoden. Insgesamt wurden für diese Arbeit über 40 Gentamicin-Proben verschiedener Hersteller untersucht, miteinander verglichen und in verschiedene Gruppen einge-teilt. Als Leitverunreinigung hat sich dabei Sisomicin erwiesen. Daneben konnte der Vergleich der Verunreinigungsprofile Hinweise auf Handelswege geben. Unter den untersuchten Proben waren auch diejenigen, zu den Todesfällen führten. Die-se konnten den stark verunreinigten Gruppen zugeordnet werden.
Das Ziel der vorliegenden Arbeit war die Klärung der Fragestellung, ob sich die quantitative NMR-Spektroskopie zur Bestimmung von Identität, Reinheit und Gehalt von Arzneistoffen eignet, und wie sich Präzision und Empfindlichkeit dieser Methode im Vergleich zu etablierten chromatographischen Verfahren verhalten. Die quantitative Untersuchung der drei strukturell jeweils verwandten Mehrkomponentengemische Codergocrinmesilat, Clomifencitrat und Flupentixoldihydrochlorid bewies eindrucksvoll die Eignung der 1H-NMR-Spektroskopie als orthogonale, analytische Messmethode im Vergleich zu validierten HPLC-Arzneibuchmethoden. Die im Rahmen einer Validierung der 1H-NMR-Methode ermittelten Ergebnisse erfüllten bezüglich Präzision und Richtigkeit die an eine im pharmazeutischen Bereich eingesetzte analytische Methode gestellten Anforderungen; zudem wurden weitere Prüfparameter wie Selektivität, Linearität, Robustheit und Stabilität verifiziert. Externe-Standard-Experimente wie "Zwei-Röhrchen-Methode" und ERETIC-Verfahren bestätigten die quantitativen Ergebnisse der Internen Standardisierung; jedoch wurde hier -- insbesondere für die ERETIC-Technik -- eine höhere Fehleranfälligkeit und somit eine größere Streuung der Einzelergebnisse beobachtet. Am Beispiel von Codergocrinmesilat und Flupentixoldihydrochlorid konnte zudem die Eignung anderer NMR-aktiver Kerne wie 13C und 19F für die quantitative Analyse von komplexen Substanzgemischen aufgezeigt werden. Das Potential der 1H-NMR-Spektroskopie für die Reinheitsprüfung von Arzneistoffen wurde am Beispiel der Aminosäure L-Alanin aufgezeigt. Die zu erwartenden Verunreinigungen Glutamin-, Asparagin-, Äpfel- und Fumarsäure konnten im Gegensatz zu den "veralteten" Prüfmethoden des Europäischen Arzneibuches eindeutig identifiziert und quantifiziert werden; mit einer Bestimmungsgrenze von <= 0.03% wurden die Vorgaben der ICH-Richtlinie Q3A(R2) erfüllt. Die deutliche Übereinstimmung der NMR-spektroskopisch ermittelten Ergebnisse einer quantitativ untersuchten Alanin-Modellmischung mit einer für den Routinebetrieb geeigneten HPLC-Methode unter Einsatz verschiedener Detektoren wie CAD, NQAD, ELSD und MS, sowie der Vergleich wichtiger Prüfparameter wie Linearität und Nachweisgrenze bestätigten die Eignung der 1H-NMR-Spektroskopie im Rahmen der routinemäßig durchgeführten Qualitätskontrolle. Die Aufdeckung von Arzneimittelfälschungen mit Hilfe der NMR-Spektroskopie wurde im Rahmen dieser Arbeit anhand der zwei aktuellen Fallbeispiele Heparin und Glycerin näher untersucht. Die in Zusammenhang mit dem Heparin-Skandal verantwortliche Kontaminante OSCS konnte neben Dermatansulfat und weiteren natürlich vorkommenden Glykosaminoglykan-Verunreinigungen im 1H-NMR-Spektrum eindeutig identifiziert und auf 0.1% OSCS bzw. 0.5% Dermatansulfat begrenzt werden. Eine präzise und richtige quantitative Bestimmung der beiden Glykosaminoglykane wurde über die N-Acetyl-Resonanzen mit Hilfe der Signalhöhenbestimmung und dem Standard-Additionsverfahren ermöglicht; deutliche Abweichungen vom "wahren" Gehalt wurden hingegen, bedingt durch starke Signalüberlagerungen, nach Flächenvergleich beobachtet. Weitere Verunreinigungen, insbesondere Lösungsmittelrückstände, die während des Extraktions- und Reinigungsprozesses des Heparins eingesetzt werden, konnten ebenfalls über charakteristische Resonanzen identifiziert und mit Hilfe der Internen-Standard-Methode quantitativ erfasst werden. Eine umfangreiche Untersuchung von 145 Heparin-API-Mustern mittels NMR-Spektroskopie und weiteren, neuentwickelten Verfahren wie HPLC, CE, IR- und Raman-Spektroskopie konnte die Eignung der entwickelten 1H-NMR-Methode bestätigen. Potentielle Glycerin-Kontaminanten wie Diethylenglycol und Ethylenglycol konnten ebenso wie eine weitere, natürlich vorkommende Verunreinigung, Propylenglycol, mittels 1H- und 13C-NMR-Spektroskopie identifiziert und quantifiziert werden. Beide Methoden erfüllten die in der USP beschriebenen Anforderungen, die für pharmazeutisch eingesetztes Glycerin jeweils höchstens 0.1% Diethylenglycol bzw. Ethylenglycol erlaubt. Während die quantitative Reinheitsprüfung beim Einsatz der 1H-NMR-Spektroskopie mit einer Messdauer im Bereich von etwa 30 min für den Routineeinsatz geeignet ist, ist die entwickelte quantitative 13C-NMR-Methode beim Einsatz von Spektrometern geringer Magnetfeldstärke aufgrund einer geringen Nachweisempfindlichkeit und der NOE-Problematik für den Routinebetrieb nur bedingt anwendbar. Abschließend kann zusammengefasst werden, dass die untersuchten Beispiele die NMR-Spektroskopie als in hohem Maße geeignet für die quantitative Analyse von Arzneimitteln ausweisen.
The aim of the present work was to determine the breast adipose tissue composition regarding fatty acids, cholesterol and (aut)oxidation products of cholesterol in women without breast cancer and to identify associated variables. Thus the necessary methods were optimized and validated where required and the breast adipose tissues of women without breast cancer were collected and analyzed.
The gas chromatography with flame ionization detection was optimized for detection and separation of 37 relevant fatty acids. Fifty breast adipose tissues were analyzed using the optimized method. 26 fatty acids were detected in breast adipose tissues. The median proportion of saturated (sum of 11 fatty acids), monounsaturated (sum of 5 fatty acids), polyunsaturated (sum of 9 fatty acids) and one trans fatty acid were 34.6%, 53.2%, 12.1% and 0.3% respectively. Moreover, absolute levels of pentadecanoic acid (median: 0.37 mg/g, range: 0.08 - 1.31 mg/g), elaidic acid (median: 0.50 mg/g, range: 0.09 - 1.92 mg/g), linolenic acid (median: 0.88 mg/g, range: 0.10 - 3.06 mg/g) and docosahexaenoic acid (median: 0.31 mg/g, range: 0.04 - 1.80 mg/g) were determined in breast adipose tissues for the first time. These four fatty acids are indicative for consumption of dairy products, processed fats, vegetable oils such as flax seed oil and fish respectively.
Furthermore, for the investigation of cholesterol in breast adipose tissues a gas chromatography was optimized and validated. The accuracies of the method in three independent spiked samples with low, medium and high levels of cholesterol were 99.1 ± 10.1%, 87.0 ± 11.2%, and 103.4 ± 4.6% with precisions of 2.1, 2.1, and 0.8% respectively. Using external calibration with internal standard cholesterol was quantified in samples (median: 1.1 mg/g, range: 0.7 - 1.5 mg/g).
In order to detect (aut)oxidation products of cholesterol, gas chromatography coupled triple quadrupole mass spectrometry was optimized and validated. The accuracy was between 81.6% and 115.7% and precisions for low, medium and high oxy-cholesterols levels were below 10.0%. The quantitative determination of (aut)oxidation products of cholesterol was established using external calibration with an internal standard. The most abundant oxy-cholesterol was 5,6β-Epoxy- (median: 147.2 ng/g, range: 25.7 – 624.2 ng/g), followed by 5,6α-Epoxy- (median: 34.6 ng/g, range: 9.9 – 124.7 ng/g), 7-Keto- (median: 19.1 ng/g, range: 7.9 – 220.6 ng/g), 7α-Hydroxy- (median: 10.2 ng/g, range: 3.8 – 111.3 ng/g) and 7β-Hydroxy-Cholesterol (median: 3.5 ng/g, range: 1.0 – 45.6 ng/g) respectively. Median oxy-cholesterol/cholesterol ratios ranged from 0.0001 (5,6β-Epoxy-Cholesterol) to 0.000003 (7β-Hydroxy-Cholesterol).
Finally the associations between fatty acids, cholesterol and oxy-cholesterol were investigated using Spearman’s rank correlation. Absolute levels of elaidic acid were positively correlated with levels of linolenic and docosahexaenoic acid (R = 0.79, 0.68, p < 0.01). Absolute levels of linolenic acid were positively associated with levels of docosahexaenoic acid (R = 0.81, p < 0.01). Moreover, relative proportions of saturated fatty acids capric, myristic, palmitic and stearic acid were negatively correlated with oleic acid (R = -0.36, -0.71, -0.65, -0.39, p < 0.05). Tissue levels of cholesterol were not correlated with levels of 5,6α/β-Epoxy-Cholesterols but were negatively associated with that of 7α-Hydroxy-, 7β-Hydroxy- and 7-Keto-Cholesterol (R = -0.29, -0.32, -0.29 p = 0.04, 0.02, 0.04). Levels of 7-Keto- and 7-Hydoxy-Cholesterol were strongly correlated with each other (R = 0.81, 0.91, p < 0.01) and, weaker, with 5,6α/β-Epoxy-Cholesterols (R = 0.60-0.70, p < 0.01). 5,6α/β-Epoxy-Cholesterols were associated positively with each other (R = 0.90, P < 0.01). Total oxy-cholesterol, 7β-Hydroxy-Cholesterol, and 5,6β-Epoxy-Cholesterol levels were correlated with relative proportions of elaidic acid (R = 0.30, 0.30, and 0.31 respectively, p = 0.04, 0.03, 0.03, respectively), whereas no correlation was observed between levels of oxy-cholesterols and relative proportion of pentadecanoic acid, linolenic acid and docosahexaenoic acid.
Furthermore, Spearman’s rank correlation was performed to investigate the relationship of fatty acids, cholesterol and oxy-cholesterol with age and body mass index. The relative proportions of total saturated fatty acids were negatively correlated with age (R = -0.47, p < 0.01) and body mass index (R = -0.29, p = 0.05). A positive significant correlation was observed between proportions of oleic acid and body mass index (R = 0.32, p = 0.02). There was no correlation between levels of cholesterol and body mass index or age. Likewise, no correlations of oxy-cholesterol levels with age or body mass index were observed.
In sum, in this work the quantification methods of cholesterol and oxy-cholesterol were validated. The validation data met the criteria according to the FDA guideline. Using the validated methods the absolute levels of cholesterol and oxy-cholesterols were determined in breast adipose tissue of human females for the first time.
Das Ziel vorliegender Arbeit war die Entwicklung einer HPLC-MS(/MS)-Methode, die im Rahmen der pharmazeutischen Qualitätskontrolle zur direkten Quantifizierung von Aminosäuren (AS) in Infusionslösungen angewendet werden kann. Die Zielset-zung schloss eine Validierung innerhalb der für die Zweckbestimmung vorgesehenen Grenzen ein. Im Rahmen der Methodenentwicklung wurde das ESI-MS/MS-Fragmentierungs-muster von 21 Aminosäuren, von 20 stabil-isotopenmarkierten Aminosäuren, die als interne Standards verwendet wurden, sowie von einigen weiteren Substanzen bestimmt. Nach Kenntnis von Precursor- und Produktionen erstellte man eine SRM-Methode zur spezifischen MS/MS-Analyse. Dabei wurden durch das jeweilige Frag-mentierungsmuster bedingte Interferenzen bei den zu untersuchenden Aminosäuren bestimmt, die bei der zu erarbeitenden HPLC-MS-Methode beachtet werden mussten. Die Methodenentwicklung zur HPLC-MS-Analytik von underivatisierten AS umfasste mit der RP-HPLC unter Verwendung eines Ionenpaarreagenzes (IP) und der hydrophilen Interaktionschromatographie (HILIC) zwei verschiedene chromatographi-sche Ansätze. Bei der Anwendung der RP-HPLC ergaben sich Probleme. Die Verwendung eines IP, im vorliegenden Fall TDFHA (Tridecafluorheptansäure), führte zu langen Equilibrierungs-, Re-Equilibrierungs- und Spülzeiten und damit bei zwar relativ kurzer HPLC-Laufzeit zu einem aber insgesamt hohen Zeitaufwand. Gleich-zeitig war die LC-MS-Anlage auf diese Anwendung fixiert, da das Ionenpaarreagenz das Gerät stark verschmutzte und dadurch andere Analysen erheblich störte. Zudem waren die Retentionszeiten der Analyten trotz langer Equilibrierungszeiten schlecht reproduzierbar, so dass eine solche Methode im Rahmen der pharmazeutischen Qualitätskontrolle schwer validierbar wäre. Weiterführende Untersuchungen erfolgten daher nicht. In nachfolgenden Studien mit der HILIC wurden verschiedene Einflussparameter (Anteil organischer Phase im Fließmittel, pH-Wert des Fließmittels, Temperatur der Säule, Pufferkonzentration im Fließmittel, Gradientenelution) auf die Trennung der AS an einer ZIC®-HILIC-Säule untersucht. Durch Optimierung der Parameter wurde so eine HILIC-HPLC-Methode entwickelt, bei der 21 AS und 20 ihrer isotopen-markierten Referenz-AS innerhalb von 20 min eluierten. Diejenigen AS, bei denen im Rahmen der Fragmentierungsstudien Interferenzen aufgrund gleicher bzw. ähnlicher Massen der Precursor- bzw. Produktionen aufgetreten waren, wurden chroma-tographisch getrennt. Gleichzeitig hat sich die SIM-Analyse als anwendbar erwiesen. Die Anwendung des spezifischeren SRM-Modus und damit der Tandem-Massenspektrometrie war nicht erforderlich. Im Rahmen der nachfolgenden Studien zur Validierung ergab sich, dass die entwickelte Methode über einen weiten Bereich eine lineare Abhängigkeit zwischen Konzentrations- und Messwerten zeigte. Für drei der 21 AS (NAcCys, NAcTyr, Pro) wurde die quadratische Regression mit dem Anpassungstest nach Mandel als geeig-neteres Regressionsmodell ermittelt. Bei Untersuchungen zur Wiederfindung wurde ein Einfluss der Matrix-Lösung der Infusionslösung festgestellt, der zu Abweichungen hinsichtlich des Quotienten AreaAS / AreaIS führte, so dass eine Quantifizierung innerhalb der geforderten Grenzen bei Kalibrierung über reine Standardlösungen nicht möglich war. Die Validierung wurde daher nachfolgend in der Matrixlösung durchgeführt. Dabei wurde gezeigt, dass mit der entwickelten HILIC-HPLC-MS-Methode Aminosäuren in Infusionslösungen mit hoher Präzision und Richtigkeit bestimmt werden können. Neun der 21 untersuchten AS konnten im Bereich von 30% - 350%, zehn weitere im Bereich von 50% - 350% innerhalb der zur Gehaltsbestimmung von pharmazeutischen Formulierungen vorgeschriebenen Grenzen (Wiederfindung Einzelbestimmung: 98% -102.0%, Mittelwert einer Dreifachbestimmung: 98.5% – 101.5%) quantifiziert werden. Für His und Phe gelang allerdings keine Quantifizierung innerhalb der Akzeptanzkriterien, wobei der Grund hierfür in weiteren Studien geklärt werden müsste. Mit der entwickelten Methode ist damit eine gleichzeitige Quantifizierung verschiedener AS-Infusionslösungsformulierungen möglich, die sich bei gleicher Matrix in der Konzentration an AS unterscheiden. Beispielsweise seien hier die Formulierungen „Aminoplasmal® E 5% / 10% /15%“ genannt, die mit der validierten Methode erfassbar sind. Die Probenvorbereitung beschränkt sich dabei auf den Zusatz der IS-Formulierung zur Infusionslösung und einen Verdünnungsschritt. Die Quanti-fizierung erfolgt über eine 5-Punkt-Kalibriergerade, die aus einer AS- und IS-Standardmischung, nach Zusatz der einfach herzustellenden Elektrolyt-Matrix, erstellt wird. Die Analysenzeit der HPLC-MS-Methode beträgt einschließlich Equilibrie-rungszeit 35 min und ist damit deutlich kürzer als die 120 min, die bei der nach wie vor zur AS-Analytik allgemein gebräuchlichen Ionenaustauschchromatographie mit Ninhydrin-Nachsäulenderivatisierung anzusetzen sind.

Therefore, quality and composition of 88 different batches of Albendazole, Mebendazole and Praziquantel locally collected from randomly selected facilities in Western Burkina Faso, Southeast Côte d’Ivoire, Southwest Ghana and Northwest Tanzania were analysed.

Products were then screened with the GPHF Minilab, consisting of tests of mass uniformity, disintegration times and thin-layer chromatography (TLC). Confirmatory tests were performed according to international pharmacopoeiae, applying assays for dissolution profiles and high-performance liquid chromatography (HPLC).
Despite minor irregularities, appearance of the products did not hint at falsified medicines. However, 19.6 % of the brands collected in Ghana and Tanzania were not officially licensed for sale. Mass uniformity was confirmed in 53 out of 58 brands of tablets. 41 out of 56 products passed disintegration times; 10 out of the 15 failing products did not disintegrate at all.
TLC results did not reveal any falsifications or pronounced dosing errors. HPLC findings confirmed the TLC results despite shifted specification limits: ten of the 83 tested batches contained less than 90 %, none more than 110 % label claim. However, no more than 46.3 % (31 / 67) of the tablet batches assayed passed the respective criteria for dissolution.
In the four study countries, no falsified anthelminthic medicine was encountered. The active pharmaceutical ingredient was not found to either exceed or distinctively fall below specification limits. Galenic characteristics as most critical criteria however, especially dissolution profiles, revealed substantial deficits.

Der Gruppe der Macrogole sowie den darauf basierenden Abkömmlingen, den Macrogolfettalkoholethern, Macrogolfettsäureestern und Polysorbaten, kommt in der modernen Galenik eine wichtige Rolle zu. Dienten sie vormals nur als gewöhnliche Emulgatoren, so finden sie heutzutage vor allem im Bereich der gezielten Wirkstofffreisetzung, der Erhöhung der Bioverfügbarkeit sowie als Löslichkeitsvermittler komplexer Systeme Anwendung. Diese vielschichtigen Anwendungsgebiete erfordern, auch aufgrund der polydispersen Strukturen der Macrogole, eine reproduzierbare und aussagekräftige Analytik.
Das Europäische Arzneibuch (Ph. Eur.) bietet zur Charakterisierung der Hilfsstoffe eine Handvoll Messgrößen, die sog. Fettkennzahlen, die eine Größenordnung vorhandener funktioneller Gruppen liefern. Zu diesen gehören Werte wie Hydroxylzahl, Iodzahl, Peroxidzahl oder Säurezahl. Diese bieten zwar einen Überblick über den Größenbereich der mittleren Kettenlängen oder einen möglichen Abbau der Strukturen, beispielsweise durch Autoxidation, jedoch geben sie keine Auskunft über die Polymerverteilung. Insbesondere diese kann jedoch, je nach Herstellungsweise, stark variieren. Außerdem ist die Methodik der Fettkennzahlenbestimmungen aufgrund der strikten Reaktionsabläufe und zahlreicher Reaktionsschritte einerseits sehr zeitaufwändig und andererseits anfällig für Fehler.
Die HPLC hat, insbesondere aufgrund der Automation, bereits seit Jahren den Status des Goldstandards in der pharmazeutischen Analytik inne. Gekoppelt mit der UV-Detektion bietet sie für zahlreiche Wirkstoffe die Möglichkeit zur schnellen, einfachen und robusten Analyse. Im Bereich der Hilfsstoffe verbreitet sich die HPLC-Analytik langsamer, da viele Hilfsstoffe keinen Chromophor aufweisen. Eine Anwendung der hochsensitiven Massenspektrometrie wäre zwar zur Detektion geeignet, würde sich für die Routineanwendung jedoch als zu komplex und kostenintensiv gestalten. Doch mit der Entwicklung der Aerosol-basierten Detektoren wie dem ELSD (evaporative light scattering detector), dem CAD (charged aerosol detector) und dem NQADTM (nano quantity aerosol detector) wurde auch für nicht-chromophore Substanzen ein Einsatz der HPLC möglich.
Die vorliegende Arbeit befasste sich mit der Entwicklung einer HPLC-CAD-Methode, die eine möglichst große Bandbreite der Macrogole und der darauf basierenden Hilfsstoffe erfassen kann. Die Trennung erfolgte an einer C18-Trennsäule. Es wurde eine Gradienten-Methode entwickelt, die aus mehreren linearen Gradientenstufen zusammengesetzt wurde, um verschiedene Kettenlängen der Polymere besser voneinander zu trennen. Als mobile Phasen dienten Wasser und Acetonitril, denen jeweils 0.1 % Ameisensäure zugesetzt wurden.
Es konnten Macrogole im Bereich PEG 300 bis PEG 3000 mit akzeptabler Auflösung aufgetrennt werden. Diese Ergebnisse wurden für PEG 300 – 1500 mittels Massenspektrometrie verifiziert. Es konnten fünf gesättigte und zwei ungesättigte Fettsäuren, sowie zwei Fettalkohole verschiedener Kettenlängen voneinander getrennt werden. Es wurden 13 Macrogol-basierte Hilfsstoffe mit der entwickelten Methode untersucht und erfolgreich getrennt. Die Macrogolfettalkoholether, -stearate und Polysorbate wurden insoweit aufgetrennt, dass die Polymerverteilung beobachtet werden konnte.
Freie PEGs in den Hilfsstoffen wurden getrennt und identifiziert. Anhand dieser konnten unterschiedliche Herstellungsweisen zugeordnet werden. Abhängig von der mittleren Kettenlänge der verarbeiteten PEGs konnten teilweise die freien Fettsäuren bzw. -alkohole von den Estern bzw. Ethern getrennt und identifiziert werden. Im Bereich der kürzeren mittleren Kettenlängen wurden die freien Fettsäuren und -alkohole von den Estern und Ethern überlagert.
Macrogolglycerolhydroxystearat (Cremophor® RH40) wurde in seine Komponenten aufgetrennt, mit Ausnahme der linearen Monoester, die mit den freien PEGs partiell koeluierten und die Glyceroltriester, die Größenausschlusseffekte zeigten.
Die Methode wurde für Stabilitätsuntersuchungen der ungesättigten Fettsäuren, Öl- und Linolsäure, eingesetzt. Hierzu wurden diese Säuren in Lösung chemisch (Wasserstoffperoxid) und thermisch (60 °C) gestresst und in bestimmten Zeitabständen analysiert. Es zeigte sich ein zeit- und temperaturabhängiger Abbau. Die teilweise Zuordnung der Abbauprodukte erfolgte durch Bestimmung des m/z mittels Massenspektrometrie. Die Methode war geeignet, um das Ausmaß eines oxidativen Abbaus von der Hauptsubstanz zu trennen und strukturell einzuordnen.
Generell bietet die Methode eine gute Basis, die eine Vielzahl an Substanzgruppen erfassen und charakterisieren kann. Sie bietet eine Ergänzung der Fettkennzahlen, die einen verringerten Arbeitsaufwand mit sich bringt. Für spezifischere Betrachtungen (Langzeitstabilität, verwandte Substanzgruppen) stellt sie einen guten Ausgangspunkt dar.
Pseudodistomin E : Versuche zur Totalsynthese über das Konzept der Tandem Wittig-[3+2]-Cycloaddition

Die Einleitung gibt einen kurzen Überblick über die Bedeutung von Piperidinalkaloiden und im speziellen wird kurz auf das pharmakologische Potential mariner Piperidinalkaloide eingegangen. Anschließend wird die Substanzklasse der „Pseudodistomine“ vorgestellt, gefolgt von einer Übersicht bereits literaturbekannter Synthesemöglichkeiten. Das primäre Ziel der vorliegenden Arbeit war die Entwicklung eines neuen stereoselektiven Zugangs zum all cis substituierten Grundkörper der Pseudodistomine C und E über die Kaskade einer Tandem Wittig-[3+2]-Cycloaddition. Weiterhin sollten Möglichkeiten ausgelotet werden, um hieran die Seitenkette des Pseudodistomins E aufbauen zu können, um erstmals eine Totalsynthese dessen zu ermöglichen.
Background
Genetic code expansion has developed into an elegant tool to incorporate unnatural amino acids (uAA) at predefined sites in the protein backbone in response to an amber codon. However, recombinant production and yield of uAA comprising proteins are challenged due to the additional translation machinery required for uAA incorporation.
Results
We developed a microtiter plate-based high-throughput monitoring system (HTMS) to study and optimize uAA integration in the model protein enhanced green fluorescence protein (eGFP). Two uAA, propargyl-L-lysine (Plk) and (S)-2-amino-6-((2-azidoethoxy) carbonylamino) hexanoic acid (Alk), were incorporated at the same site into eGFP co-expressing the native PylRS/tRNAPyl CUA pair originating from Methanosarcina barkeri in E. coli. The site-specific uAA functionalization was confirmed by LC-MS/MS analysis. uAA-eGFP production and biomass growth in parallelized E. coli cultivations was correlated to (i) uAA concentration and the (ii) time of uAA addition to the expression medium as well as to induction parameters including the (iii) time and (iv) amount of IPTG supplementation. The online measurements of the HTMS were consolidated by end point-detection using standard enzyme-linked immunosorbent procedures.
Conclusion
The developed HTMS is powerful tool for parallelized and rapid screening. In light of uAA integration, future applications may include parallelized screening of different PylRS/tRNAPyl CUA pairs as well as further optimization of culture conditions.
Ziel der vorliegenden Arbeit war es, den Umfang der gastrointestinalen Absorption und Metabolisierung von mit der Nahrung aufgenommenen Polyphenolen in vivo zu ermitteln. Darüber hinaus sollte deren systemische Verfügbarkeit anhand von humanen Serum- und Urinproben bestimmt werden. Lebensmittel der Wahl war dabei Apfelsaft. Die Identifizierung und Strukturaufklärung der Polyphenole und ihrer Metabolite erfolgte mittels Hochleistungsflüssigchromatographie-Diodenarray-Detektion (HPLC-DAD), HPLC-Elektrospray-Tandemmassenspektrometrie (HPLC-ESI-MS/MS) sowie Kapillargaschromatographie-Massenspektrometrie (HRGC-MS). Quantitative Analysen wurden mittels HPLC-DAD durchgeführt; für die Bestimmung der Polyphenolgehalte in Urinproben sowie von D-(-)-Chinasäure wurde die HPLC-ESI-MS/MS im Single Reaction Monitoring (SRM) Modus eingesetzt. Zur Etablierung der Polyphenolanalytik und zur Auswahl eines für die Studien geeigneten Saftes wurden die Polyphenolprofile verschiedener Presssäfte aus Most- und Tafeläpfeln sowie kommerziell erhältlicher Apfelsäfte ausgewertet. Für die Säfte aus Tafeläpfeln wurden Polyphenolmengen zwischen 154 und 178 mg/L bestimmt, wohingegen die Säfte aus Mostäpfeln Gehalte zwischen 261 und 970 mg/L aufwiesen. Bei den Säften des Handels wiesen die naturtrüben Apfelsäfte mit 182 bis 459 mg/L höhere Polyphenolgehalte auf als die klaren Produkte (120 - 173 mg/L). Bei oraler Aufnahme kommen die Polyphenole zuerst mit Speichel in Kontakt. Umsetzungen mit zentrifugiertem Speichel führten zu keiner Modifikation der Substanzen. In Gegenwart von nativem Speichel wurden für die ß-glycosidisch gebundenen Flavonoidglycoside hydrolytische Abbaureaktionen in Abhängigkeit der Struktur ihres Zuckerrestes beobachtet. Nach Antibiotikumzugabe wurden deutlich geringere Abbauraten ermittelt. Die Hydrolyse erfolgt demnach hauptsächlich durch Enzyme der bakteriellen Mundflora. Im Weiteren gelangen die Polyphenole über die Speiseröhre in den stark sauren Magen. Zur Überprüfung ihrer Stabilität wurden die Apfelpolyphenole mit künstlichem Magensaft (pH 1,81) über vier Stunden inkubiert. Einzig für Procyanidin B2 wurde ein nahezu vollständiger Abbau nachgewiesen. Nach Passage des Magens erreichen die Polyphenole das neutrale bis leicht alkalische Duodenum. Die Inkubation erfolgte mit simuliertem Duodenalsekret (pH 7,2) über einen Zeitraum von 24 Stunden. Für 5-Kaffeoylchinasäure wurde eine 37%ige Abnahme beobachtet. Dabei wurden 3- und 4-Kaffeolychinasäure, Kaffeesäure, D-(-)-Chinasäure sowie Kaffeesäuremethylester generiert. Vergleichbare Ergebnisse wurden bei der Inkubation von 4-p-Cumaroylchinasäure erhalten. Kaffeesäure unterlag einer 26,3%igen Umsetzung zu Ferulasäure, Dihydrokaffeesäure und Kaffeesäuremethylester. Bei den monomeren Flavan-3-olen wurden mittels HPLC-Analytik an chiraler Phase Epimerisierungen nachgewiesen. Procyanidin B2 war nach vier Stunden nur noch in Spuren erfassbar. Quercetin wurde vollständig in Phloroglucin, 3,4-Dihydroxybenzoesäure und 2,4,6-Trihydroxybenzoesäure gespalten. Um die Verfügbarkeit der Polyphenole im Dickdarm zu untersuchen, wurde eine Interventionsstudie mit naturtrübem Apfelsaft bei Probanden mit einem Stoma des terminalen Ileums durchgeführt. Nach oraler Aufnahme von einem Liter Saft wurde der Ileostomaausfluss über einen Zeitraum von acht Stunden gesammelt. In den Ileostomabeuteln wurden zwischen Null und 33,1% der einzelnen aus dem Apfelsaft aufgenommenen phenolischen Substanzen wiedergefunden. Der ausgeschiedene Anteil der Flavonoidglycoside war dabei abhängig von der Struktur des jeweiligen Zuckerrestes. Als Metabolite waren D-(-)-Chinasäure, 1- und 3-Kaffeoylchinasäure, Phloretin und dessen 2´-O-Glucuronid sowie die Methylester der Kaffee- und p-Cumarsäure nachweisbar. Für die höhermolekularen Procyanidine wurden Wiederfindungen von 90,3% sowie deren partieller Abbau ermittelt. Die systemische Verfügbarkeit der Polyphenole sowie ihre renale Ausscheidung wurden in zwei weiteren Humanstudien mit gesunden Probanden untersucht. Nach Konsum von einem Liter naturtrüben Apfelsaft erfolgten Blutabnahmen über einen Zeitraum von acht Stunden; Urin wurde über einen Zeitraum von 24 Stunden untersucht. Die Bilanzierung der Apfelpolyphenole erfolgte sowohl vor als auch nach enzymatischer Hydrolyse. Kaffeesäure, 5-Kaffeoylchinasäure, 4-p-Cumaroylchinasäure, (-)-Epicatechin, Phloretin und Quercetin waren sowohl im Serum als auch im Urin detektierbar. Insgesamt wurden 5,3% (Serum) bzw. 23% (Urin) der mit dem Saft aufgenommenen phenolischen Verbindungen wiedergefunden. Davon waren im Urin 19,5% in Form hydroxylierter phenolischer Säuren nachweisbar.
Der Piperidin-Heterozyklus kann als wichtiger, multifunktionaler Arzneistoffbaustein angesehen werden, da eine große Anzahl derzeit eingesetzter Arzneistoffe den Piperidin-Derivaten zuzuordnen ist. Dabei kommen diese Substanzen bei einer Vielzahl verschiedenster Indikationen zum Einsatz. Aus diesem Grund wurden im Zuge dieser Arbeit ebenfalls Piperidin-Derivate synthetisiert, und zwar zum einen 2,6-Diaryl-4-oxo-piperidin-3,5-dicarbonsäurediester und 2,6-Diaryl-4-oxo-piperidin-3-carbonsäuremethylester, die auf ihre antiproliferativen Eigenschaften an Protozoen untersucht werden sollten, und zum anderen Spiropiperidinderivate, die als Liganden des Opioidrezeptors ORL1 synthetisiert worden sind. Die synthetisierten Spiropiperidin-Derivate basieren auf der Leitverbindung Ro 64-6198, einem selektiven und hochaffinen Agonisten am ORL1-Rezeptor, welcher als viertes Mitglied der Opioidrezeptor-Familie zugeordnet wurde. Die bisherigen pharmakologischen Untersuchungen konnten ein breites Wirkprofil seines endogenen Liganden Nociceptin aufdecken. Da jedoch aus der Literatur gerade im Bereich der Schmerzmodulation teilweise kontroverse Ergebnisse vorliegen und nur wenig über die Wirkmechanismen bekannt ist, ist die Synthese selektiver Agonisten und Antagonisten notwendig. Ziel dieser Arbeit war es, Derivate der Leitverbindung zu synthetisieren. Die wesentlichste Änderung stellte die Substitution des Piperidin-Grundgerüstes durch Alkylseitenketten dar. Die pharmakologischen Untersuchungen am ORL1-Rezeptor sind jedoch bislang noch nicht abgeschlossen. Unter den Infektionskrankheiten stellt vor allem Malaria eine große Belastung für die hauptsächlich in tropischen Gebieten lebende Bevölkerung dar. Das gleiche gilt für Trypanosomeninfektionen (afrikanische Schlafkrankheit und Chagas-Erkrankung). Das Hauptproblem in der Therapie dieser Infektionen besteht in der zunehmenden Resistenzbildung der Erreger gegenüber den derzeit eingesetzten Arzneistoffen. Die Aufklärung des Polyaminstoffwechsels von Protozoen bietet einen neuen Ansatzpunkt, denn die Unterbrechung dieses Metabolismus durch gezielte Hemmung der beteiligten Enzyme kann die Vermehrung der Protozoen verhindern. Polyamine wie Putrescin, Spermin und Spermidin spielen bei der Zellteilung und -proliferation von Eukaryonten eine maßgebliche Rolle. Gleiches gilt für den durch Metabolisierung des Spermidins aktivierten “eukaryotic initiaton factor“ (eIF5A). Dessen Aktivierung verläuft über die beiden Enzyme Deoxyhypusinsynthase (DHS) und Deoxyhypusin-hydroxylase (DHH). Für die Pflanzenaminosäure L-Mimosin und das Fungizid Ciclopirox ist an Plasmodien bereits eine inhibitorische Wirkung der Deoxyhypusinhydroxylase in vitro und damit verbunden die Hemmung des Plasmodienwachstums nachgewiesen. Beide entfalten ihre Wirkung über die Chelatisierung des im Enzym vorliegenden Metall-Ions Fe(II)/Fe(III). Da nur L-Mimosin in vivo eine inhibitorische Aktivität zeigt, wurde dieses als Leitstruktur für die zu synthetisierenden 2,6-Diaryl-4-oxo-piperidin-3,5-dicarbonsäurediester und 2,6-Diaryl-4-oxo-piperidin-3-carbonsäuremethylester herangezogen. Im Zuge dieser Arbeit konnten diverse Derivate beider Verbindungstypen synthetisiert werden, deren inhibitorische Aktivität in vitro an Plasmodium falciparum und Trypanosoma brucei brucei und deren Zytotoxizität an Makrophagen getestet wurden. Die Synthese erfolgte in beiden Fällen über eine Mannichreaktion. Die IC50-Werte dieser an Trypanosoma brucei brucei untersuchten Verbindungen liegen im Bereich der Aktivität der derzeit bei Trypanosomeninfektionen eingesetzten Arzneistoffe Eflornithin-HCl und Nifurtimox für die Verbindungen 10a-10n bzw. Suramin-Na und Nifurtimox für 11a-11d. Somit stellen die Monoester-Verbindungen die potentere Substanzklasse dar. Die an Plasmodium falciparum getesteten und als inhibitorisch aktiv identifizierten 2,6-Diaryl-4-oxo-piperidin-3,5-dicarbonsäurediestern sind die Derivate 10h-10k. Unter den 2,6-Diaryl-4-oxo-piperidin-3-carbonsäuremethylestern konnte 11c als aktive Verbindung identifiziert werden. Diese Monoester-Verbindung weist im Vergleich zu den aktiven Diester-Derivaten eine 10-fach höhere Potenz auf. Daher ist anzunehmen, dass die Monoester-Derivate auch an Plasmodien die aktivere Substanzklasse darstellen. Die Verbindungen 10h-10k wurden wegen ihrer guten In-vitro-Aktivität an Plasmodium falciparum weiter untersucht. Allerdings konnte in den In-vivo-Versuchen an Plasmodium berghei-infizierten Mäusen keine Hemmung der Parasitämie festgestellt werden.
Cabozantinib (CAB) is a receptor tyrosine kinase inhibitor approved for the treatment of several cancer types. Enterohepatic recirculation (EHC) of the substance is assumed but has not been further investigated yet. CAB is mainly metabolized via CYP3A4 and is susceptible for drug–drug interactions (DDI). The goal of this work was to develop a physiologically based pharmacokinetic (PBPK) model to investigate EHC, to simulate DDI with Rifampin and to simulate subjects with hepatic impairment. The model was established using PK-Sim® and six human clinical studies. The inclusion of an EHC process into the model led to the most accurate description of the pharmacokinetic behavior of CAB. The model was able to predict plasma concentrations with low bias and good precision. Ninety-seven percent of all simulated plasma concentrations fell within 2-fold of the corresponding concentration observed. Maximum plasma concentration (C\(_{max}\)) and area under the curve (AUC) were predicted correctly (predicted/observed ratio of 0.9–1.2 for AUC and 0.8–1.1 for C\(_{max}\)). DDI with Rifampin led to a reduction in predicted AUC by 77%. Several physiological parameters were adapted to simulate hepatic impairment correctly. This is the first CAB model used to simulate DDI with Rifampin and hepatic impairment including EHC, which can serve as a starting point for further simulations with regard to special populations.
Considerable effort has previously been invested in a light‐controlled inhibition of the enzyme acetylcholinesterase (AChE). We found that a novel azobenzene‐based bistacrine AChE inhibitor switched faster than the known dithienylethene based bistacrine and inverted the photo‐controlled interactions of the photoisomers compared to its dithienylethene congener. Furthermore, we have optimized a previously described light‐controlled tacrine‐based AChE inhibitor. Isomerization upon irradiation with UV light of the novel inhibitor was observed in aqueous medium and showed no fatigue over several cycles. The cis‐enriched form showed an 8.4‐fold higher inhibition of hAChE compared with its trans‐enriched form and was about 30‐fold more active than the reference compound tacrine with a single‐digit nanomolar inhibition. We went beyond proof‐of‐concept to discover photoswitchable AChE inhibitors with pharmacologically desirable nanomolar inhibition, “cis‐on” effect, and pronounces differences between the photoisomers.
Bisherige Projekte zur Pharmazeutischen Betreuung haben sich auf Erwachsene konzentriert. Daraus ergab sich die Fragestellung, inwiefern Kinder und Jugendliche von einer Pharmazeutischen Betreuung profitieren. In der vorliegenden Arbeit wurde diese Frage an der Modellkrankheit Asthma bronchiale untersucht. Zur Pharmazeutischen Betreuung von erwachsenen Asthmatikern lagen bereits einige Studien vor. Kinder wurden jedoch noch in keiner der Untersuchungen, die in Europa durchgeführt wurden, eingeschlossen. Folglich eröffnete sich die Möglichkeit, erstmals einen Beitrag zur Pharmazeutischen Betreuung von asthmakranken Kindern und Jugendlichen im Rahmen von öffentlichen Apotheken in Deutschland zu liefern. Der Effekt der Pharmazeutischen Betreuung von Kindern und Jugendlichen mit Asthma bronchiale wurde im Rahmen einer Studie untersucht, die in Kooperation mit Offizinapothekern durchgeführt wurde. Hauptzielkriterium der Studie war die gesundheitsbezogene Lebensqualität der asthmakranken Kinder und Jugendlichen sowie deren Eltern. Des Weiteren wurden klinische, ökonomische und intermediäre Zielkriterien erfasst. Die Pharmazeutische Betreuung erfolgte gemäß eines für die Untersuchung erarbeiteten, modifizierten TOM-Schemas. Die Treffen zwischen Apotheker und Patient fanden alle sechs bis acht Wochen über einen Zeitraum von einem Jahr statt. Es konnte eine repräsentative Patientenpopulation von 28 Kindern und Jugendlichen mit Asthma bronchiale rekrutiert werden. Da keine Kontrollgruppe erhoben werden konnte, wurde die Untersuchung als Prä-Post Studie ausgewertet. Die angestrebte Patientenzahl von 35 Patienten wurde allerdings nicht erreicht, weshalb die Untersuchung als Pilotstudie angesehen werden muss. Dennoch konnte ein statistisch und klinisch signifikanter Effekt der Pharmazeutischen Betreuung auf die gesundheitsbezogene Lebensqualität der Eltern gezeigt werden: Der Gesamtwert des PACQLQ verbesserte sich von einem Median (Interquartil) von 5,1 (3,9–6,3) an der Basislinie auf 6,2 (5,1-6,8) (p=0,016) nach einem Jahr Pharmazeutischer Betreuung. Der für diesen Fragebogen definierte Schwellenwert für eine klinisch signifikante Verbesserung wurde von acht der vierzehn Eltern erreicht oder überschritten. Für die gesundheitsbezogene Lebensqualität der asthmakranken Kinder und Jugendlichen, die mit einem generischen Fragebogen erfasst wurde, wurde keine statistisch signifikante Entwicklung beobachtet. Die Asthmakontrolle befand sich mit einem medianen Wert von 0,7 Punkten per ACQ schon an der Basislinie auf einem für Kinder und Jugendliche mit Asthma bronchiale untypisch hohem Niveau. Entsprechend konnte für diesen Endpunkt kein statistisch signifikanter Effekt der Pharmazeutischen Betreuung festgestellt werden. Bei der vorliegenden Studie handelt es sich um die erste Erhebung zur Pharmazeutischen Betreuung von Asthmatikern, in der die gesundheitsbezogene Lebensqualität der Eltern der asthmakranken Kinder bzw. Jugendlichen erfasst wurde. Dagegen sollte für die Messung der gesundheitsbezogenen Lebensqualität der asthmakranken Kinder und Jugendlichen auf einen krankheitsspezifischen Fragebogen zurückgegriffen werden, da diese veränderungssensitiver sind als generische Fragebögen. Für Studien zur Pharmazeutischen Betreuung von Asthmatikern ist aufgrund der Ergebnisse der vorliegenden Studie zu überlegen, ob anhand des seit kurzem vorliegenden Schwellenwertes für den ACQ schwerpunktmäßig Patienten mit nicht ausreichend kontrolliertem Asthma rekrutiert werden sollten. Ein weiteres Ziel der Arbeit bestand darin, den potentiellen Stellenwert des 8-iso Prostaglandin F2 alpha, das als Marker für die Bestimmung des Entzündungsgrades diskutiert wird, für die Therapieoptimierung im Rahmen von Studien zur Pharmazeutischen Betreuung von asthmakranken Kindern und Jugendlichen zu evaluieren. Voraussetzung für die Verwendung eines Markers im Rahmen von Studien bzw. von Routinediagnostik und –monitoring bei Kindern ist, ein möglichst nicht-invasiv zugängliches Spezimen zu finden, in dem der Marker zuverlässig bestimmt werden kann. Hierfür wurden im Rahmen einer Querschnittsstudie erstmals von gesunden Kindern sowie von Kindern mit Asthma bzw. Cystischer Fibrose Atemkondensat, Speichel, Serum und Urin gesammelt. Die Konzentrationen des 8-iso Prostaglandin F2 alpha, die bestimmt wurden, waren im Bereich des Detektionslimits des verwendeten ELISA und es stellte sich heraus, dass die Ergebnisse nicht reproduzierbar waren. Zudem war im Rahmen der Pilotstudie keine Korrelation zwischen den Konzentrationen in den verschiedenen Spezimen nachweisbar und es bestand kein offensichtlicher Unterschied zwischen den Konzentrationen bei gesunden und erkrankten Kindern. Folglich erscheint die Erfassung des 8-iso Prostaglandin F2 alpha für Studien zur Pharmazeutischen Betreuung von asthmakranken Kindern und Jugendlichen zum jetzigen Zeitpunkt weniger geeignet. Als Alternative ist die Messung des exhalierten NO in Erwägung zu ziehen.
Pharmakokinetische und molekularpharmakodynamische Aspekte inhalativ angewandter Glucocorticoide

In der vorliegenden Arbeit wurden vielfältige pharmakokinetische bzw. molekularpharmakodynamische Aspekte topisch angewandter Glucocorticoide untersucht und deren klinische Relevanz diskutiert. Um die Potenz der Glucocorticoide vergleichen zu können, wurden deren relative Rezeptoraffinitäten zum humanen Glucocorticoidrezeptor mit Dexamethason als Referenzsubstanz bestimmt. Dabei zeigte sich, dass Glucocorticoide der neueren Generation wie Mometasonfuroat und Fluticasonpropionat sich in ihrer Assoziationskinetik unterschieden, während die Dissoziationskinetik vergleichbar war. Damit wurde erstmals für Mometasonfuroat eine relative Rezeptoraffinität berechnet, die einen unmittelbaren Vergleich mit weiteren in der Literatur publizierten Daten anderer Glucocorticoide erlaubte. Diese relative Rezeptoraffinität ist die höchste aller soweit bekannten inhalativen Glucocorticoide. In Kompetitionsversuchen wurde erstmals gezeigt, dass die in vitro nachgewiesenen Metabolite des Mometasonfuroats eine signifikante Rezeptoraffinität aufwiesen. Die Potenz dieser Metabolite ist dabei vergleichbar mit der von anderen inhalativ angewandten Corticosteroiden, wie Flunisolid und Triamcinolonacetonid. Mometasonfuroat konnte damit als das einzige inhalative Glucocorticoid charakterisiert werden, dessen Metabolite affiner als Dexamethason sind. Weiterführend zu den Erkenntnissen zur Rezeptorkinetik des Mometasonfuroats und Fluticasonpropionats wurde erstmals die vergleichende Kinetik der mRNA-Induktion auf zellulärer Ebene untersucht. Diese wurde am Beispiel des Glucocorticoid-regulierten CD163 in humanen Monocyten verfolgt. Es wurde eine Methode adaptiert, die schließlich eine Quantifizierung der CD163-mRNA mittels Real-time-PCR ermöglichte. Es zeigte sich, dass die Anzahl der neu gebildeten mRNA-Kopien direkt von der Rezeptoraffinität und der Konzentration des Mometasonfuroats und Fluticasonpropionats abhängig war. Die unterschiedliche Assoziationsgeschwindigkeit der beiden Glucocorticoide spiegelte sich jedoch nicht in einer schnelleren Induktion der CD163-mRNA wider. Neben der spezifischen Rezeptorbindung werden Glucocorticoide auch unspezifisch an Gewebe gebunden. Es wurden erstmals umfassende In-vitro-Versuche zur Bindung des Mometasonfuroats, Ciclesonids und seines aktiven Metaboliten an humanes Lungengewebe durchgeführt. Die Adsorption der Glucocorticoide an humanes Lungengewebe verlief schnell und war vergleichbar hoch. Die wesentlichen Unterschiede zwischen den Substanzen beruhten dabei auf den nach Equilibrierung mit Plasma im Gewebe verbleibenden Konzentrationen. Es stellte sich heraus, dass Mometasonfuroat im Vergleich zu Fluticasonpropionat, Ciclesonid und dessen aktivem Metabolit Desisobutyryl-Ciclesonid die niedrigste Gewebeaffinität aufwies (Fluticasonpropionat > Ciclesonid > Desisobutyryl-Ciclesonid > Mometasonfuroat). Es konnte eine gute Übereinstimmung der in vitro Bindungsverhältnisse an Lungengewebe mit klinischen In-vivo-Daten aufgezeigt werden. Aus den zuvor in der Literatur beschriebenen Untersuchungen ließen sich zwar Hinweise auf die hepatische Metabolisierung des Mometasonfuroats entnehmen, doch Ergebnisse einer möglichen Metabolisierung bzw. der Stabilität in humanem Spezimen von therapeutischer Relevanz lagen soweit nicht vor. Somit wurde die Stabilität dieses Glucocorticoids im humanen Lungengewebe und Plasma systematisch untersucht. Dabei wurde eine unbekannte Substanz detektiert, die mittels HPLC-MS/MS, 1H- und 13C-NMR eindeutig als das 9,11-Epoxid des Mometasonfuroats identifiziert wurde. Kompetitionsversuche am humanen Glucocorticoidrezeptor zeigten, dass die relative Rezeptoraffinität des 9,11-Epoxy-Mometasonfuroats hoch war und wiederum vergleichbar mit der Bindungsaffinität des Triamcinolonacetonid oder Flunisolid. Das Auflösungsverhalten inhalativ angewandter Glucocorticoide in Bronchialflüssigkeit spielt eine entscheidende Rolle für deren weitere Verteilungskinetik und Wirkungen. Es wurde erstmals eine Methode entwickelt, mit der neben Morphologie und Größe der Partikel aus kommerziell erhältlichen Arzneiformen auch das Auflösungsverhalten in Bronchialsekret untersucht werden konnte. Durch den Einsatz der Raster-Elektronen-Mikroskopie wurde eine maximale Sensitivität zur Beobachtung der Partikelveränderungen bis zu einer Größe von 0,5 µm gewährleistet. Am Beispiel des Beclomethasondipropionats wurde gezeigt, dass die Arzneistoffpartikel aus verschiedenen Dosieraerosolen nach einer raschen Auflösung rekristallisieren. Erst diese Rekristallisation ist offenbar die Grundlage für eine nachfolgende langsame Auflösung und Umverteilung des Arzneistoffs ins Plasmakompartiment. Da diese neuartigen Beobachtungen in vollkommener Übereinstimmung mit pharmakokinetischen Daten nach Inhalation dieses Glucocorticoids stehen, darf angenommen werden, dass sich vergleichbare Vorgänge in vivo abspielen.
The inhaled pharmacotherapy is fundamental in the management of obstructive lung diseases such as asthma bronchiale or chronic obstructive pulmonary disease. In this context short- and long-acting β2-agonists play a prominent role as relieve and control medication. Regarding the risk-benefit profile of an inhaled drug, the pattern of pulmonary deposition and the rate and extent of absorption into systemic circulation are essential parameters. New developments of drugs are characterized by high lung retention and improved efficacy. The aim of the present thesis was the parallel evaluation the pharmacokinetic (PK) and -dynamic (PD) properties of inhaled β2-agonists employing an isolated human lung perfusion model (IPL). The short-acting β2-agonist salbutamol and the newly developed ultra long-acting β2-agonist GW597901 were chosen for the analysis of pulmonary drug absorption and bronchodilation. In a pharmacokinetic enabling study an established human IPL setting was modified to monitor the pharmacokinetics of the β2-agonists by measuring the concentrations in perfusion fluid, lung tissue and BAL samples obtained during and after the experiments. The IPL model revealed differences in the pulmonary absorption behaviour of GW597901 and salbutamol. The lipophilic compound GW597901 was distributed to a lower extent into the perfusion fluid compared to the more hydrophilic compound salbutamol. The analyzed time profiles of nebulized salbutamol in the perfusate were consistent to with a clinical study if considering experimental conditions as the actual deposited doses and the differing volume of distribution. Thus, the suitability of the IPL model for the PK analysis of inhaled β2-agonists was confirmed. In a PK/PD study the human ex vivo model was employed for the first time for the evaluation of the clinical relevant bronchodilating effect induced by inhaled β2-agonists in addition to the analysis of their pharmacokinetics. Thereby the focus was to determine the onset and extent of bronchodilation. A new method was established to monitor changes in lung function parameters due to pharmacodynamic interventions over the duration of the experiment that allowed permanent online recording of the ventilation volume and lung mechanic parameters. Bronchial challenges with aerolised MCh were performed successfully in isolated ventilated human lung lobes, even though the responder rate was lower than expected despite high administered doses. The administration of the short acting agent salbutamol led to an immediate onset of action recognized as a sudden increase of the ventilation volumes. The bronchodilation following the application of GW597901 was observed delayed after about 6 min. Monitored lung function parameters considerably improved by both β2 - agonists in the IPL setting but not significantly different. Thus, in regard of the different applied doses GW597901 had a higher intrinsic activity and bronchodilating potency than salbutamol. The concentrations of salbutamol and GW597901 in the perfusate determined in the PK/PD study were significantly lower than those observed in the pharmacokinetic enabling study, while the tmax values and the course of the distribution profiles remained similar. Most likely, the application of nebulized MCh prior to the administration of the β2 - agonists had a substantial influence on their pharmacokinetic behaviour. It is yet not clear whether pharmacodynamic effects or molecular competition processes for the passage to the systemic circulation or both influenced the redistribution of the β2 - agonists as seen in the PK/PD study. The potential clinical relevance of this observation has to be further investigated. The development of pulmonary edema during the experiment was one limitation of the IPL model. For the determination of the onset of edema formation four potential biochemical markers, specifically surfactant-protein A (SP-A), angiotensin-converting enzyme (ACE), urea and lactate dehydrogenase, were measured in perfusion fluids. In this context, an ELISA method for the quantification of human SP-A in biological matrices was successfully established. The investigations showed that the concentrations of SP-A and ACE in the perfusate increased over time as a sign for lung tissue damage and correlated with the degree of edema formation. For the first time the IPL model was used for the evaluation of potential pulmonary edema marker and the results have shown that it is valuable tool for further investigations in this field. In conclusion, the pharmacokinetic and pharmacodynamic characterization of GW597901 and salbutamol was successfully achieved using the IPL model. This ex vivo methodology may contribute to further insights and understanding of the complex pharmacokinetic processes of inhaled β2 – agonists in the lung.
Protein-protein interactions play a crucial role in the development of drug delivery devices for the increasingly important biologicals, including antibodies, growth factors and cytokines. The understanding thereof might offer opportunities for tailoring carriers or drug proteins specifically for this purpose and thereby allow controlled delivery to a chosen target. The possible applications range from trigger-dependent release to sustained drug delivery and possibly permanently present stimuli, depending on the anticipated mechanism.
Silk fibroin (SF) is a biomaterial that is suitable as a carrier for protein drug delivery devices. It combines processability under mild conditions, good biocompatibility and stabilizing effects on incorporated proteins.
As SF is naturally produced by spiders and silkworms, the understanding of this process and its major factors might offer a blueprint for formulation scientists, interested in working with this biopolymer. The natural process of silk spinning covers a fascinating versatility of aggregate states, ranging from colloidal solutions through hydrogels to solid systems. The transition among these states is controlled by a carefully orchestrated process in vivo. Major players within the natural process include the control of spatial pH throughout passage of the silk dope, the composition and type of ions, and fluid flow mechanics within the duct, respectively. The function of these input parameters on the spinning process is reviewed before detailing their impact on the design and manufacture of silk based drug delivery systems (DDS). Examples are reported including the control of hydrogel formation during storage or significant parameters controlling precipitation in the presence of appropriate salts, respectively. The review details the use of silk fibroin to develop liquid, semiliquid or solid DDS with a focus on the control of SF crystallization, particle formation, and drug-SF interaction for tailored drug load.
Although we were able to show many examples for SF drug delivery applications and there are many publications about the loading of biologics to SF systems, the mechanism of interaction between both in solution was not yet extensively explored. This is why we made this the subject of our work, as it might allow for direct influence on pharmaceutical parameters, like aggregation and drug load.
In order to understand the underlying mechanism for the interaction between SF and positively charged model proteins, we used isothermal titration calorimetry for thermodynamic characterization. This was supported by hydrophobicity analysis and by colloidal characterization methods including static light scattering, nanoparticle tracking analysis and zeta potential measurements. We studied the effects of three Hofmeister salts – NaCl (neutral), NaSCN (chaotropic) and Na2SO4 (cosmotropic) – and the pH on the interaction of SF with the model proteins in dependence of the ratio from one to another. The salts impacted the SF structure by stabilizing (cosmotropic) or destabilizing (chaotropic) the SF micelles, resulting in completely abolished (cosmotropic) or strongly enhanced (chaotropic) interaction. These effects were responsible for different levels of loading and coacervation when varying type of salt and its concentration. Additionally, NaCl and NaSCN were able to prolong the stability of aqueous SF solution during storage at 25°C in a preliminary study.
Another approach to influence protein-protein interactions was followed by covalent modification. Interleukin-4 (IL-4) is a cytokine driving macrophages to M2 macrophages, which are known to provide anti-inflammatory effects. The possibility to regulate the polarization of macrophages to this state might be attractive for a variety of diseases, like atherosclerosis, in which macrophages are involved. As these cases demand a long-term treatment, this polarization was supposed to be maintained over time and we were planning to achieve this by keeping IL-4 permanently present in an immobilized way. In order to immobilize it, we genetically introduced an alkyne-carrying, artificial amino acid in the IL-4 sequence. This allowed access to a site-specific click reaction (Cu(I)-catalyzed Huisgen azide-alkyne cycloaddition) with an azide partner. This study was able to set the basis for the project by successful expression and purification of the IL-4 analogue and by proving the availability for the click reaction and maintained bioactivity. The other side of this project was the isolation of human monocytes and the polarization and characterization of human macrophages. The challenge here was that the majority of related research was based on murine macrophages which was not applicable to human cells and the successful work was so far limited to establishing the necessary methods.
In conclusion, we were able to show two different methods that allow the influence of protein-protein interactions and thereby the possible tailoring of drug loading. Although the results were very promising for both systems, their applicability in the development of drug delivery devices needs to be shown by further studies.
Salt formation is a routinely used strategy for poorly water-soluble drugs and traditionally performed with small inorganic counterions. High energy crystal lattices as well as effects on the local pH within the aqueous boundary layer during dissolution drive the increased dissolution rate and apparent solubility. Ionic liquids however, by definition low melting ionic salts with often large organic counterions, combine an increased dissolution rate with solubilization of the drug by the counterion itself. Long lasting supersaturation profiles of increased kinetic solubility were reported for several drugs formulated as ionic liquids increasing their overall bioavailability. Furthermore, aggregation and micellization between highly lipophilic compounds and amphiphilic bile acids was described before, demonstrating the capabilities of the human body itself to utilize solubilization of poorly water-soluble compounds. Development of novel counterions not only tailoring the desired physicochemical properties e.g. dissolution rate of the parent drug but adding – in a best-case scenario synergistic – pharmacological activity has been driven forward in the last years. However, salt formation can only be applied for ionizable i.e. acidic or basic compounds. While co-crystals can be used as a nonionized alternative, their formation is not always successful leading to an urgent need for other formulation strategies. In these lines, development of 2D and 3D printing techniques has been ongoing for the last decades and their pharmaceutical application has been demonstrated. The versatile nature and commercial availability allow a decentralized production further elaborating this technique for a highly flexible and patient-oriented supply with medication.
This thesis focuses on the theoretical background and potential application of salt formation in the pharmaceutical development of a drug candidate. The first section presents the current knowledge and state of the art in preparation of low melting ionic liquids i.e. salts and is translated to the in vitro investigation of molecular interaction between the poorly water-soluble drug imatinib and components of the human intestinal fluid in the second section. Development of novel antibiotic counterions and assessment of their potential use in pharmaceutical formulations with fluoroquinolones is described in the last two sections.
Chapter I describes the application of low melting ionic liquids in pharmaceutical formulation and details their development in the last two decades from versatile organic solvents in chemical synthesis towards amorphous strategies for drug delivery. The chapter gives a general overview on molecular structure and physicochemical properties of several drug containing ionic liquids and details the mechanisms which attribute to a typically fast dissolution, increased aqueous solubility as well as enhanced permeation which was reported in several publications.
Chapter II translates the increased aqueous solubility of drugs by an organic counterion to the human gastrointestinal tract with taurocholate and lecithin as main drivers for the solubilization of highly lipophilic and poorly water-soluble drugs. Investigation of the interaction of imatinib – a poorly water-soluble weak base – with fasted- and fed state simulated intestinal fluids revealed a complex interplay between the components of the intestinal fluid and the drug. Mixed vesicles and micelles were observed in concentration dependent aggregation assays and revealed differences in their size, molecular arrangement as well as composition, depending on the tested drug concentration. Overall, the study outlines the effective interaction of weakly basic drugs with taurocholate and lecithin to minimize recrystallization during intestine passage finally leading to favorable supersaturation profiles.
Chapter III focuses on the development of novel antibiotic counterions which potentially move the evolution of ionic liquids from a pharmaceutical salt with tailored physicochemical properties to a synergistic combination of two active pharmaceutical ingredients. The natural occurring anacardic acid derived from the cashew nut shell inspired a series of antibacterial active acidic compounds with increasing alkyl chain length. Their physicochemical properties, antibacterial activity, bacterial biofilm inhibition and cytotoxicity were detailed and in vivo activity in a Galleria mellonella model was assessed. This group of anacardic acid derivatives is synthetically accessible, easily modifiable and yielded two compounds with favorable activity and physicochemical profile for further drug development.
Chapter IV outlines the potential application of anacardic acid derivatives in pharmaceutical formulations by salt formation with fluoroquinolone antibiotics as well as novel techniques such as 2D/3D printing for preparation of drug imprinted products. Despite anacardic acid derivatives demonstrated promising physicochemical properties, salt formation with fluoroquinolone antibiotics was not feasible. However, 2D/3D printed samples with anacardic acid derivative alone or in combination with ciprofloxacin demonstrated physical compatibility between drug and matrix as well as antibacterial activity against three S. aureus strains in an agar diffusion assay. Conclusively, drug printing can be applied for the herein tested compounds, but further process development is necessary.
In summary, preparation of low melting ionic liquids, salts or co-crystals is an appropriate strategy to increase the aqueous solubility of poorly water-soluble drugs and tailor physicochemical properties. The counterion itself solubilizes the drug and furthermore potentially interferes with the complex micellar environment in the human intestine. However, salt formation as routinely used formulation strategy is not feasible in every case and development of alternative techniques is crucial to hurdle challenges related to unfavorable physicochemical properties. The outlined techniques for 2D/3D drug printing provide versatile production of drug products while extending the design space for novel drug development.
Therapeutic drug monitoring (TDM) is increasingly relevant for an individualized antibiotic therapy and subsequently a necessary tool to reduce multidrug-resistant pathogens, especially in light of diminishing antimicrobial capabilities. Critical illness is associated with profound pharmacokinetic and pharmacodynamic alterations, which challenge dose finding and the application of particularly hydrophilic drugs such as β-lactam antibiotics. Methods: Implementation strategy, potential benefit, and practicability of the developed standard operating procedures were retrospectively analyzed from January to December 2020. Furthermore, the efficacy of the proposed dosing target of piperacillin in critically ill patients was evaluated. Results: In total, 160 patients received piperacillin/tazobactam therapy and were subsequently included in the study. Of them, 114 patients received piperacillin/tazobactam by continuous infusion and had at least one measurement of piperacillin serum level according to the standard operating procedure. In total, 271 measurements were performed with an average level of 79.0 ± 46.0 mg/L. Seventy-one piperacillin levels exceeded 100 mg/L and six levels were lower than 22.5 mg/L. The high-level and the low-level group differed significantly in infection laboratory parameters (CRP (mg/dL) 20.18 ± 11.71 vs. 5.75 ± 5.33) and renal function [glomerular filtration rate (mL/min/1.75 m2) 40.85 ± 26.74 vs. 120.50 ± 70.48]. Conclusions: Piperacillin levels are unpredictable in critically ill patients. TDM during piperacillin/tazobactam therapy is highly recommended for all patients. Although our implementation strategy was effective, further strategies implemented into the daily clinical workflow might support the health care staff and increase the clinicians' alertness.
