Im Rahmen dieser Arbeit wurden verschiedene Inhibitoren der Acetylcholinesterase (AChE) untersucht, die als potentielle Substanzen zur Behandlung von Morbus Alzheimer eingesetzt werden können. Die Hemmwirkung der einzelnen Substanzen wurde mittels Ellman-Test überprüft. Gemeinsames Strukturmerkmal der Substanzklasse, von der im ersten Teil der Arbeit ausgegangen wurde, war das Grundgerüst des AChE-Reaktivators TMB4 [1,1´-Trimethylen-bis(4-Formyl-Pyridiniumbromid)-Dioxim]. Anhand der biologischen Daten konnte beobachtet werden, dass die Art der Substitution die inhibitorische Aktivität der Verbindungen wesentlich beeinflusst. Am wirksamsten von allen Bispyridinium-Derivaten zeigte sich das 2,6-chlorierte Derivat DUO3 (IC50 = 0.34 μM), gefolgt von monobenzyl-substituiertem UNO3, bismethylsubstituiertem TBM und unsubstituiertem TMB4. Weiterhin wurde der Bindungsmodus der DUO-Substanzen im aktiven Zentrum der AChE untersucht. Die Docking-Studien an Substanzen der DUO-Klasse zeigten ein einheitliches Bindungsmodel, welches folgende Wechselwirkungen be-inhaltet: π-π-„stacking“ zwischen dem Benzylring einer DUO-Substanz und dem Trp84 am Grunde der Bindetasche des Enzyms, face-to-face Wechselwirkung (π-π und Kation-π) zwischen dem Pyridiniumring und Trp334 oder Phe331 der aromatischen Furche. Bei 60% der gedockten Strukturen wurde eine face-to-face-Wechselwirkung an der anionischen peripheren Seite (PAS) der AChE-Tasche festgestellt. Weiterhin wurden neue optimierte Inhibitoren entwickelt. Die Bispyridinium-Struktur der DUO-Derivate wurde um den aus der Furche herausragenden Benzylring gekürzt. Als Leitstrukturen dienten die AChE-aktivsten Substanzen DUO3 (2,6-Cl-Derivat) und DUO12 (2-Cl-Derivat) sowie ein bisphthalimidomethyl-substituiertes TMB4-Derivat (WDUO). Die aktivste Verbindung der Pyridinium-Klasse war die Phthalimid-Phenyl-substituierte Substanz 3c (IC50 = 0.073 μM). Ihre inhibitorische Aktivität gegenüber AChE befand sich im Bereich der des Tacrins (IC50 = 0.044 μM). Sie zeigte eine sehr gute AChE-Selektivität; die BChE hemmte sie um den Faktor 34 schwächer. Im Rahmen der vorliegenden Arbeit wurde nicht nur das rationale Design angewandt, um zu viel versprechenden Kandidaten bezüglich klinischen Einsatzes zu gelangen. Auch das Verfahren der „Random Chemistry“ kam zum Einsatz, um neue und interessante Strukturen zu erzeugen, die eventuell bessere Eigenschaften als die Ausgangssubstanz besitzen. Der Grundgedanke dieses Verfahrens liegt in der Ausnutzung der durch gamma-Strahlen (60Co-Quelle) induzierten Radiolyse des Lösungsmittels, welches seine primären Produkte zur chemischen Reaktion mit dem in ihm gelösten Stoff zur Verfügung stellt. Aus den entstandenen Produkten wurden durch spezifische biologische Testung (Inhibition der AChE) positiv reagierende Komponenten herausselektiert. Die Proben wurden zuerst im Ganzen auf ihre Fähigkeit, die AChE zu hemmen, geprüft. Nach der bioaktivitätsgeleiteten Fraktionierung und Subfraktionierung mittels HPLC erwies sich die Tacrin/MeOH-Probe als die, mit dem interessantesten Aktivitätsprofil. Die Charakterisierung der entstandenen Verbindungen bezüglich ihrer Vielfältigkeit erfolgte mittels ESI-Massenspektrometrie und UV-Spektroskopie. Die Substanz mit höchster Hemmwirkung gegenüber AChE (Peak E der Tacrin/MeOH-Probe) wurde nach der Isolierung der Reinheitsprüfung und Strukturaufklärung mittels NMR-, FTIR- und (Tandem)-ESI-Massenspektrometrie zugeführt und auf ihre biologische Wirkung hin untersucht.
Das Ziel dieser Arbeit war die Anwendung intakter Mikroorganismen auf organische Sulfide zur asymmetrischen Synthese von optisch aktiven Sulfoxiden. Die im Vergleich zu den aufwendigen und teueren Reaktionen mit isolierten Enzymen besonders effizienten Rahmenbedingungen bei sogenannten `whole-cell´-Umsetzungen stellten den Grund für die Bemühungen in dem stetig an Bedeutung gewinnenden Arbeitsfeld der Bioorganischen Chemie dar. Die wesentlichen Ergebnisse dieser Studien sind im Folgenden zusammengefasst: 1. Die Mikroorganismen wurden isoliert, singularisiert und kultiviert. Eine Bodenprobe diente als Quelle für eine Vielzahl an Bakterien, Hefen und Pilzen, die mit dem Standardsubstrat Thioanisol auf ihre Fähigkeit zur enantioselektiven Sulfoxidation überprüft wurden. 2. Insgesamt sind sechs Keime nach standardisierten Methoden genotypisch charakterisiert und den entsprechenden Spezies zugeordnet worden. Die beiden Bakterienstämme mit den bei der Sulfoxidation höchsten Enantiomeren-überschüssen (ee-Werten), nämlich Arthrobacter aurescens (bildete das S-Enantiomer) und Pseudomonas frederiksbergensis (lieferte das R-Enantiomer), wurden für nachfolgende Biosynthesen verwendet. Pseudomonas frederiksbergensis war der einzige Stamm, der das R-Enantiomer im Überschuss produzierte. 3. Durch direkte Vergleiche der Biosyntheseleistung der isolierten Bakterien mit kommerziell erhältlichen Referenzstämmen wurde im Fall von Pseudomonas frederiksbergensis gezeigt, dass sich Bodenisolat und zugeordneter Referenz-stamm gegensätzlich enantioselektiv verhalten. Weitere Charakterisierungs-sonden (Farb- und Assimilationsreaktionen, Oberflächenfettsäureverteilung, „Siderophore-Typing“ und direkter rRNA Vergleich) sicherten die Zugehörigkeit beider Bakterienstämme als Pseudomonas frederiksbergensis-Spezies; keinerlei Unterschiede wurden zwischen den beiden Stämmen festgestellt. Zum ersten Mal werden somit zwei natürliche, nicht genetisch manipulierte Stämme von Pseudomonas frederiksbergensis beschrieben, deren Enzymaktivität eine entgegengesetzte Enantioselektivität in der mikrobiellen `whole-cell´ asym-metrischen Sulfoxidation aufweist. 4. In einem umfangreichen Substratscreening sind strukturvariierte organische Sulfide als Substrate zur bakteriellen Sulfoxidation eingesetzt worden. Anhand der ee-Werte wurde der Einfluss der Sulfidstruktur auf den Reaktionsverlauf bestimmt. Generell erwiesen sich Arylalkylsulfide als optimale Substrate für die bakterielle Sulfoxidation mit den isolierten und kommerziell erworbenen Stämmen von Arthrobacter aurescens und Pseuodomonas frederiksbergensis; aliphatische Sulfide wurden zur biokatalytischen Umsetzung nicht akzeptiert. 4a. Elektronenreiche para-Substituenten am Arylsystem ergaben teilweise enantio-merenreine Sulfoxide. 4b. Eine zunehmende Anzahl an Stickstoffatomen im Arylring (N-heterozyklische Grundstruktur) führte zu einer dramatischen Verringerung des ee-Wertes. 4c. Schwefelhaltige Furfuryle und Thiophene wurden nicht als Substrate für die enantioselektive Sulfoxidation akzeptiert. 4d. Der Einsatz schwefelhaltiger Pestizide in der Biokatalyse verlief erfolglos, allerdings wurden die Organophosphorpestizide Fenamiphos® und Fenthion® mit dem aus Sulfoxidationsreaktionen lange bekannten Enzym Chlorperoxidase (CPO) enantiomerenangereichert umgesetzt. 4e. Die biotechnologisch wichtige Anwendung der asymmetrischen Sulfoxidation in der Arzneistoffsynthese -hier versucht mit den Wirkstoffen Omeprazol® und Modafinil®- schlug fehl. 5. Der Einsatz eines Bioreaktors (Fermenter) schuf die Grundlage für künftige asymmetrrische Sulfoxidationen in präparativem Maßstab. Eine Zellzahlstudie mit Pseudomonas frederiksbergensis wurde durchgeführt; ferner erfolgten Bestimmungen der optimalen Fermentationsparameter am Beispiel einfach strukturierter, organischer Sulfide inklusive Blindwerts- und Hemmversuchen. Die toxischen Einflüsse auf die bakteriellen `whole-cell´-Systeme, die vom eingesetzten Sulfid sowohl als auch vom produzierten Sulfoxid verursacht werden, bedürfen besonderer Beachtung bei einer weiteren Bearbeitung dieses Themas. Das vorgestellte, neue Phänomen der asymmetrischen Sulfoxidation mit entgegenge-setzter Enantioselektivität durch zwei geno- und phänotypisch identische Spezies von Pseudomonas frederiksbergensis rechtfertigt eine weitere, intensive Suche nach derartigen, natürlichen Mikroorganismen.
Over the years, hydrogels have been developed and used for a huge variety of different applications ranging from drug delivery devices to medical products. In this thesis, a poly(2-methyl-2-oxazoline) (POx) / poly(2-n-propyl-2-oxazine) (POzi) bioink was modified and analyzed for the use in biofabrication and targeted drug delivery. In addition, the protein fibrinogen (Fbg) was genetically modified for an increased stability towards plasmin degradation for its use as wound sealant.
In Chapter 1, a thermogelling, printable POx/POzi-based hydrogel was modified with furan and maleimide moieties in the hydrophilic polymer backbone facilitating post-printing maturation of the constructs via Diels-Alder chemistry. The modification enabled long-term stability of the hydrogel scaffolds in aqueous solutions which is necessary for applications in biofabrication or tissue engineering. Furthermore, we incorporated RGD-peptides into the hydrogel which led to cell adhesion and elongated morphology of fibroblast cells seeded on top of the scaffolds. Additional printing experiments demonstrate that the presented POx/POzi system is a promising platform for the use as a bioink in biofabrication.
Chapter 2 highlights the versatility of the POx/POzi hydrogels by adapting the system to a use in targeted drug delivery. We used a bioinspired approach for a bioorthogonal conjugation of insulin-like growth factor I (IGF-I) to the polymer using an omega-chain-end dibenzocyclooctyne (DBCO) modification and a matrix metalloprotease-sensitive peptide linker. This approach enabled a bioresponsive release of IGF-I from hydrogels as well as spatial control over the protein distribution in 3D printed constructs which makes the system a candidate for the use in personalized medicine.
Chapter 3 gives a general overview over the necessity of wound sealants and the current generations of fibrin sealants on the market including advantages and challenges. Furthermore, it highlights trends and potential new strategies to tackle current problems and broadens the toolbox for future generations of fibrin sealants.
Chapter 4 applies the concepts of recombinant protein expression and molecular engineering to a novel generation of fibrin sealants. In a proof-of-concept study, we developed a new recombinant fibrinogen (rFbg) expression protocol and a Fbg mutant that is less susceptible to plasmin degradation. Targeted lysine of plasmin cleavage sites in Fbg were exchanged with alanine or histidine in different parts of the molecule. The protein was recombinantly produced and restricted plasmin digest was analyzed using high resolution mass spectrometry. In addition to that, we developed a novel time resolved screening protocol for the detection of new potential plasmin cleavage sites for further amino acid exchanges in the fibrin sealant.
Microbial, mammalian and plant cells produce and contain secondary metabolites, which typically are soluble in water to prevent cell damage by crystallization. The formation of ion pairs, e.g. with carboxylic acids or mineral acids, is a natural blueprint to keep basic metabolites in solution. It was aimed at showing whether the mostly large carboxylates form soluble protic ionic liquids (PILs) with basic natural products resulting in enhanced aqueous solubility. Furthermore, their supramolecular pattern in aqueous solution was studied. Thereby, naturally occurring carboxylic acids were identified being appropriate counterions for natural basic compounds and facilitate the formation of PILs with their beneficial characteristics, like improved dissolution rate and enhanced apparent solubility.

The formation of ion pairs, for example, with carboxylic acids or mineral acids, is a natural blueprint to maintain basic metabolites in solution. Here, we aim at showing whether the mostly large carboxylates form soluble protic ionic liquids (PILs) with the basic natural product papaverine resulting in enhanced aqueous solubility. The obtained PILs were characterized by H-1-N-15 HMBC nuclear magnetic resonance (NMR) and in the solid state using X-ray powder diffraction, differential scanning calorimetry, and dissolution measurements. Furthermore, their supramolecular pattern in aqueous solution was studied by means of potentiometric and photometrical solubility, NMR aggregation assay, dynamic light scattering, zeta potential, and viscosity measurements. Thereby, we identified the naturally occurring carboxylic acids, citric acid, malic acid, and tartaric acid, as being appropriate counterions for papaverine and which will facilitate the formation of PILs with their beneficial characteristics, like the improved dissolution rate and enhanced apparent solubility.
In resent years the rate of biologics (proteins, cytokines and growth-factors) as newly registered drugs has steadily risen. The greatest challenge for pharmaceutical biologics poses its arrival at the desired target location due to e.g. proteolytic and pH dependent degradation, plasma protein binding, insolubility etc. Therefore, advanced drug delivery systems, where biologics are site directed immobilized to carriers mimicking endogenous storage sites such as the extra cellular matrix can enormously assist the application and consequently the release of exogenous administered pharmaceutical biologics. We have resorted to the fibroblast growth factor 2/ heparansulfate/ fibroblast growth factor bindingprotein 1 system as a model.
Phase I deals with the selection and subcloning of a wild type murine FGF-2 construct into the bacterial pHis-Trx vector system for high yields of expression and fast, feasible purification measurements. This first step enables the provision of mFGF-2, which plays a pivotal part as a growth factor in the wound healing process as well as the vascularization of tumors, for future investigations. Therefore, the correct expression of mFGF-2 was monitored via MALDI-MS and SDS-PAGE, whereas the proper folding of the tertiary beta-trefoil structure was assessed by fluorescence spectroscopy. The MTT assay allowed us to ensure that the bioactivity was comparable to sourced FGF-2. In the last step, the purity; a requirement for future binding- and protein-protein interaction assays was monitored chromatographically (RP-HPLC). In addition, a formulation for freeze-drying was developed to ensure protein stability and integrity over a period of 60 days. Altogether, the bacterial expression and purification proved to be suitable, leading to bioactive and stable production of mFGF-2.
In Phase II the expression, purification and characterization of FGFBP1, as the other key partner in the FGF-2/ HS/ FGFBP1 system is detailed. As FGFBP1 exhibits a complex tertiary structure, comprised of five highly conserved disulfide bonds and presumably multiple glycosylation sites, a eukaryotic expression was used. Human embryonic kidney cells (HEK 293F) as suspension cells were transiently transfected with DNA-PEI complexes, leading to expression of Fc-tagged murine FGFBP1. Different PEI to DNA ratios and expression durations were investigated for optimal expression yields, which were confirmed by western blot analysis and SDS-PAGE. LC-MS/MS analysis of trypsin and elastase digested FGFBP1 gave first insights of the three O-glycosylation sites. Furthermore, the binding protein was modified by inserting a His6-tag between the Fc-tag (for purification) and the binding protein itself to enable later complexation with radioactive 99mTc as radio ligand to track bio distribution of administered FGFBP1 in mice. Overall, expression, purification and characterization of mFGFBP1 variants were successful with a minor draw back of instability of the tag free binding protein.
Combining the insights and results of expressed FGF-2 as well as FGFBP1 directed us to the investigation of the interaction of each partner in the FGF-2/ HS/ FGFBP1 system as Phase III. Thermodynamic behavior of FGF-2 and low molecular weight heparin (enoxaparin), as a surrogate for HS, under physiological conditions (pH 7.4) and pathophysiological conditions, similar to hypoxic, tumorous conditions (acidic pH) were monitored by means of isothermal titration calorimetry. Buffer types, as well as the pH influences binding parameters such as stoichiometry (n), enthalpy (ΔH) and to some extent the dissociation constant (KD). These findings paved the way for kinetic binding investigations, which were performed by surface plasmon resonance assays. For the first time the KD of full length FGFBP1 and FGF-2 was measured. Furthermore the binding behavior of FGF-2 to FGFBP1 in the presence of various heparin concentrations suggest a kinetic driven release of bound FGF-2 by its chaperone FGFBP1.
Having gathered multiple data on the FGF-2 /HS /FGFBP1 system mainly in solution, our next step in Phase IV was the development of a test system for immobilized proteins. With the necessity to better understand and monitor the cellular effects of immobilized growth factors, we decorated glass slides in a site-specific manner with an RGD-peptide for adhesion of cells and via the copper(I)-catalyzed-azide-alkyne cycloaddition (CuAAC) a fluorescent dye (a precursor for modified proteins for click chemistry). Human osteosarcoma cells were able to grow an the slides and the fluorescence dye was immobilized in a biocompatible way allowing future thorough bioactivity assay such as MTT-assays and phospho-ERK-assays of immobilized growth factors.
Chronic inflammatory diseases such as rheumatoid arthritis, type 2 diabetes and cardiovascular diseases, are associated with the homeostatic imbalance of one of several physiological systems combined with the lack of spontaneous remission, which causes the disease to persevere throughout patients’ lives. The inflammatory response relies mainly on tissue-resident, pro-inflammatory M1 type macrophages and, consequently, a chance for therapeutic intervention lies in driving macrophage polarization towards the anti-inflammatory M2 phenotype. Therefore, anti-inflammatory cytokines that promote M2 polarization, including interleukin-4 (IL4), have promising therapeutic potential. Unfortunately, their systemic use is hampered by a short serum half-life and dose-limiting toxicity. On the way towards cytokine therapies with superior safety and efficacy, this thesis is focused on designing bioresponsive delivery systems for the anti-inflammatory cytokine IL4.
Chapter 1 describes how anti-inflammatory cytokines are tightly regulated in chronic, systemic inflammation as in rheumatoid arthritis but also in acute, local inflammation as in myocardial infarction. Both diseases show a characteristic progression during which anti-inflammatory cytokine delivery is of variable benefit. A conventional, passive drug delivery system is unlikely to release the cytokines such that the delivery matches the dynamic course of the (patho-)physiological progress. This chapter presents a blueprint for active drug delivery systems equipped with a 24/7 inflammation detector that continuously senses for matrix metalloproteinases (MMP) as surrogate markers of the disease progress and responds by releasing cytokines into the affected tissues at the right time and place. Because they are silent during phases of low disease activity, bioresponsive depots could be used to treat patients in asymptomatic states, as a preventive measure. The drug delivery system only gets activated during flares of inflammation, which are then immediately suppressed by the released cytokine drug and could prevent the steady damage of subclinical chronic inflammation, and therefore reduce hospitalization rates.
In a first proof of concept study on controlled cytokine delivery (chapter 2), we developed IL4-decorated particles aiming at sustained and localized cytokine activity. Genetic code expansion was deployed to generate muteins with the IL4’s lysine 42 replaced by two different unnatural amino acids bearing a side chain suitable for click chemistry modification. The new IL4 muteins were thoroughly characterized to ensure proper folding and full bioactivity. Both muteins showed cell-stimulating ability and binding affinity to IL4 receptor alpha similar to those of wild type IL4. Copper-catalyzed (CuAAC) and strain-promoted (SPAAC) azide–alkyne cycloadditions were used to site-selectively anchor IL4 to agarose particles. These particles had sustained IL4 activity, as demonstrated by the induction of TF-1 cell proliferation and anti-inflammatory M2 polarization of M-CSF-generated human macrophages. This approach of site-directed IL4 anchoring on particles demonstrates that cytokine-functionalized particles can provide sustained and spatially controlled immune-modulating stimuli.
The idea of a 24/7 sensing, MMP driven cytokine delivery system, as described in the introductory chapter, was applied in chapter 3. There, we simulated the natural process of cytokine storage in the extracellular matrix (ECM) by using an injectable solution of IL4 for depot formation by enzyme-catalyzed covalent attachment to ECM components such as fibronectin. The immobilized construct is meant to be cleaved from the ECM by matrix-metalloproteinases (MMPs) which are upregulated during flares of inflammation. These two functionalities are facilitated by a peptide containing two sequences: a protease-sensitive peptide linker (PSL) for MMP cleavage and a sequence for covalent attachment by activated human transglutaminase FXIIIa (TGase) included in the injection mix for co-administration. This peptide was site-selectively conjugated to the unnatural amino acid at IL4 position 42 allowing to preserve wild type bioactivity of IL4. In vitro experiments confirmed the anticipated MMP response towards the PSL and TGase-mediated construct attachment to fibronectin of the ECM. Furthermore, the IL4-peptide conjugates were able to reduce inflammation and protect non-load bearing cartilage along with the anterior cruciate ligament from degradation in an osteoarthritis model in rabbits. This represents the first step towards a minimally invasive treatment option using bioresponsive cytokine depots with potential clinical value for inflammatory conditions.
One of the challenges with this approach was the production of the cytokine conjugate, with incorporation of the unnatural amino acid into IL4 being the main bottleneck. Therefore, in chapter 4, we designed a simplified version of this depot system by genetically fusing the bifunctional peptide via a flexible peptide spacer to murine IL4. While human IL4 loses its activity upon C-terminal elongation, murine IL4 is not affected by this modification. The produced murine IL4 fusion protein could be effectively bound to in vitro grown extracellular matrix in presence of TGase. Moreover, the protease-sensitive linker was selectively recognized and cleaved by MMPs, liberating intact and active IL4, although at a slower rate than expected. Murine IL4 offers the advantage to evaluate the bioresponsive cytokine depot in many available mouse models, which was so far not possible with human IL4 due to species selectivity.
For murine IL4, the approach was further extended to systemic delivery in chapter 5. To increase the half-life and specifically target disease sites, we engineered a murine IL4 variant conjugated with a folate-bearing PEG chain for targeting of activated macrophages. The bioactive IL4 conjugate had a high serum stability and the PEGylation increased the half-life to 4 h in vivo. Surprisingly, the folate moiety did not improve targeting in an antigen-induced arthritis (AIA) mouse model. IL4-PEG performed better in targeting the inflamed joint, while IL4-PEG-folate showed stronger accumulation in the liver. Fortunately, the modular nature of the IL4 conjugate facilitates convenient adaption of PEG chain length and the targeting moiety to further improve the half-life and localization of the cytokine.
In summary, this thesis describes a platform technology for the controlled release of cytokines in response to inflammation. By restricting the release of the therapeutic to the site of inflammation, the benefit-risk ratio of this potent class of biologics can be positively influenced. Future research will help to deepen our understanding of how to perfectly combine cytokine, protease-sensitive linker and immobilization tag or targeting moiety to tackle different diseases.
Intraperitoneal adhesions are fibrous bands that connect tissues in the peritoneal cavity that are usually separated. These adhesions form as a consequence of trauma, inflammation or surgical interventions and often result in severe consequences such as chronic pain, small bowel obstructions or female infertility.
The aim of this thesis was to develop a synthetic barrier device for adhesion prevention made of modified poly(lactide) [PLA]. Solid PLA films (SurgiWrap®) are already successfully in clinical use due to the good biocompatibility and the biodegradability of the material resulting in non-toxic degradation products since lactic acid is naturally part of the metabolic circles of the human body. Considering the brittleness and stiffness of the films, the long degradation time of several months as well as the need for suturing, there is potential for optimization. Through a copolymerization with the hydrophilic poly(ethylene glycol) [PEG], a reduction of the degradation time was intendend. Moreover, the copolymerization should also lead to an improvement of the mechanical properties of the films since PEG acts as plasticizer for PLA. Linear PLA-PEG-PLA triblock copolymers as well as star-shaped PEG-PLA copolymers were synthesized via standard ring opening polymerization to tailor the barrier properties. Besides solid films, solution electrospun meshes from PLA and the synthesized PEG-PLA copolymers were investigated for a potential application as well. Since suturing of a barrier additionally induces adhesion formation, alginate coated membranes were prepared in order to achieve self-adhesiveness. With the intention to reduce infections and consequently inflammation, electrospun meshes and solvent cast films were loaded with the antibacterial drug triclosan and drug release as well as antibacterial efficacy was investigated.
Mechanical tests confirmed that through the variation of the PEG content and branching the mechanical properties can be tailored and are in good accordance with the glass transition temperatures [Tg] of the polymers. Consequently, potentially adequate mechanical properties for surgical handling as well as for the performance within the patient’s body were successfully achieved. Degradation studies revealed that the degradation time was significantly shorter for PEG-PLA membranes than for PLA films and with an appropriate PEG content could be adjusted to the intended time frame. Cell adhesion and viability tests confirmed the non-toxicity of the clinically used PLA films as well as of PEG-PLA films and meshes. With a bioadhesion test the benefit of an alginate coated side towards the pure PLA film concerning self-adhesiveness was successfully demonstrated. Moreover, optical evaluations and a T-peel test of different alginate coated PLA films showed that the cohesion between the chemically different layers was distinctly enhanced by the use of an appropriate PEG-PLA mesh as intermediate cohesion promoting layer. In in vitro release studies with triclosan loaded films a higher release was determined for PEG-PLA than for PLA films. In agar diffusion tests a higher and longer inhibition of staphylococcus aureus growth was observed confirming the release results. Moreover, drug loaded meshes (especially drug loaded after electrospinning) showed enhanced and elongated bacterial inhibition in comparison to films.
Aufgrund ihrer gut dokumentierten, umfangreichen gesundheitsfördernden biologischen Aktivitäten wird den Flavonoiden eine große Bedeutung zugeordnet. Die Ergebnisse von in vitro- und Tierstudien deuten zudem darauf hin, dass diese Verbindungen bei der Prävention und Therapie von Erkrankungen wie Krebs oder Alzheimer Krankheit (AD) positive Effekte zeigen. Zur besseren Charakterisierung der Interaktion von Flavonoiden mit Krebszellen wurden von uns die Cytotoxizität verschiedener Flavonoide auf T-Lymphoblastomzellen untersucht und Strukturelemente identifiziert, welche für einen Flavonoid-induzierten Zelltod relevant sind. Weitere Studien waren der potentiell neuroprotektiven Wirkung von Flavonoiden gewidmet. Die sowohl in neuronalen Zellkulturen als auch in transgenen Alzheimer-Mäusen (TgAPPsw) festgestellte Erhöhung der sAPPα Produktion und Reduktion von Aβ-Bildung wurden mit Aktivitäts- und Expressionssteigerung der α-Sekretase ADAM-10 assoziiert. Um herauszufinden, ob Flavonoide eine neuroprotektive Wirkung zeigen, wurden erste Vorbereitungen für ein Flavonoid-Screening mit einer sowohl hAPP alsauch ADAM-10 stabil transfizierten HT1080 Zellen getroffen. Dies beinhaltete die Suche nach einer potenten siRNA/shRNA und einem effektiven Flavonoid. Im zweiten Teil der Arbeit wurden Experimente durchgeführt, um die Rolle von Heparansulfat (HS) bei der foamyviralen Anbindung an die Wirtszelle zu untersuchen. Foamyviren (FV) sind Spumaviren und gehören zur Familie der Retroviren. Bei unseren Studien wurde die Bindung von FV an Heparin, die Korrelation der Suszeptibilität verschiedener Zellen mit zellulärem HS und die Reduktion der Infektion durch lösliches Heparin sowie durch enzymatischen HS-Abbau festgestellt.
Bile solubilization plays a major role in the absorption of poorly water-soluble drugs. Excipients used in oral drug formulations impact bile-colloidal properties and their molecular interactions. Polymer-induced changes of bile colloids, e.g., by Eudragit E, reduced the flux of the bile interacting drug Perphenazine whereas bile non-interacting Metoprolol was not impacted. This study corroborates these in vitro findings in rats. Eudragit E significantly reduced systemic availability of Perphenazine but not Metoprolol compared to the oral administrations without polymer. This study confirms the necessity to carefully select polymers for bile interacting drugs whereas non-bile interacting drugs are more robust in terms of excipient choice for formulation. The perspective of bile interaction may introduce interesting biopharmaceutical leverage for better performing oral formulations of tomorrow.
Das Ausmaß der Proteinbindung eines Arzneistoffs hat Auswirkungen auf eine Vielzahl pharmakokinetischer Parameter wie z.B. das Verteilungsvolumen, die Metabolisierung oder Ausscheidung. Da nur der freie, ungebundene Anteil eines Arzneistoffs durch bio-logische Membranen diffundieren kann, ist dieser direkt für die pharmakologische Wirkung verantwortlich. Lediglich diese ungebundenen und damit frei beweglichen Arzneistoffmoleküle sind also in der Lage, an Rezeptoren zu binden und damit einen Effekt auszulösen. Je größer der ungebundene Anteil eines Arzneistoffs ist, desto größer kann auch der Effekt sein, den er zu bewirken vermag. Wird nun diese freie Konzentration verändert, so kann sich demzufolge die Wirkintensität des Arzneistoffs verändern. Daraus folgt, dass die Kenntnis über die Proteinbindung speziell für die Dosisfindung eines Arzneistoffs unerlässlich ist. Diese Zusammenhänge veranschaulichen, welche Bedeutung das Ausmaß der Proteinbindung eines Arzneistoffs hat. Für die Bestimmung der Proteinbindung existiert eine Vielzahl von Methoden. Neben der klassischen Gleichgewichtsdialyse werden vor allem Ultrafiltrationstechniken angewendet. Neuere Verfahren stellen verschiedenste kapillarelektrophoretische Methoden dar. Allen Verfahren ist gemein, dass es sich um In-vitro-Methoden handelt. Als einzige In-vivo-Methode hat sich die Mikrodialyse etabliert. Die im Rahmen dieser Arbeit entwickelte Bestimmungsmethode für das Ausmaß der Proteinbindungen stellt eine kontinuierliche Variante der Ultrafiltration dar. Mit Hilfe des in dieser Arbeit beschriebenen Verfahrens ist es im möglich, mit einem einzigen Experiment Proteinbindungen über einen großen Bereich verschiedener Arzneistoff-Protein-Verhältnisse zu berechnen. Dadurch wird die Bestimmung schneller und auch kostengünstiger. Zusätzlich wurde dieses Verfahren teilweise automatisiert, d.h. die Versuche werden programmgesteuert durchgeführt und ein manuelles Eingreifen ist nur noch an wenigen Stellen eines Versuches notwendig. Die Messanlage zur kontinuierlichen Ultrafiltration wurde nach dem Vorbild der Anlage von Oehlmann aufgebaut und im Zuge dessen fortentwickelt. Herzstück ist die Messzelle aus Plexiglas. Sie besteht aus einem Ober- und einem Unterteil. In diesem befindet sich eine Vertiefung für die Rührscheibe. Beim Zusammenbau werden zwischen die beiden Teile die Filterunterstützung und eine Ultrafiltrationsmembran gelegt. Die Ultrafiltrationsmembran hält hierbei aufgrund ihrer Trenngrenze die Proteinmoleküle in der Messzelle zurück und lässt nur freie Arzneistoffmoleküle durch. Ein Dichtungsring sorgt dafür, dass die Zelle, nachdem sie in einem Gestell fixiert wurde, nach außen hin dicht abschließt. Während eines Versuches werden abwechselnd Arzneistofflösung und reine Pufferlösung durch die Messzelle gepumpt. Am Auslass der Ultrafiltrationszelle wird die Absorption gemessen, die zu Absorptions-Zeit-Diagrammen führen. Wird nun der gleiche Versuch durchgeführt, nachdem eine Proteinlösung in die Messzelle injiziert wurde, verschiebt sich diese Kurve aufgrund der Wechselwirkung zwischen Arzneistoff und Proteinlösung nach rechts. Die Fläche zwischen diesen beiden Kurven kann als Maß für die Proteinbindung herangezogen werden. Für die Auswertung der Versuche wird aus den Messdaten errechnet, wie viel Arzneistoff sich zu jedem Zeitpunkt des Versuchs in der Messzelle befunden hat und wie viel ungebunden vorlag und damit am Detektor messbar ist. Da die Konzentration an Arzneistoff in der Messzelle im Laufe eines Versuches kontinuierlich gesteigert wird, erhält man Bindungsdaten über einen weiten Bereich von Arzneistoff-Protein-Verhältnissen. In dieser Arbeit wurden sowohl Versuche mit Rinderserumalbumin (BSA) als auch mit humanem Serumalbumin (HSA) durchgeführt. Die Standardabweichungen der bestimmten Proteinbindungswerte steigen mit fallender Proteinbindung. Damit sind sie nach außen hin neutral und ihre Löslichkeit sinkt. Wie die Ergebnisse zeigen, konnte dennoch das Ausmaß der Proteinbindung für eine Reihe von Fluorochinolonen bestimmt werden. Zusätzlich wurden mit dem Oxazolidinon Linezolid und dem Ketolid Telithromycin weitere Antibiotika vermessen. Die Proteinbindungen aller untersuchten Arzneistoffe gegenüber HSA stimmen mit Daten aus der Literatur überein. Wie bereits erwähnt lassen sich hierbei kleine Abweichungen sowohl mit unterschiedlichen Bestimmungsmethoden als auch mit verschiedenen eingesetzten Proteinen erklären. Die Literaturdaten geben meist die Plasmaproteinbindung eines Arzneistoffes wider und die Messungen in der vorliegenden Arbeit wurden allesamt mit Albumin, sei es vom Mensch oder Rind, durchgeführt. Da jedoch neben dem Albumin auch noch weitere Bestandteile des Plasmas mit den Arzneistoffmolekülen wechselwirken können, sind unterschiedliche Proteinbindungen zu erwarten.
Bestimmung der Plasmaproteinbindung von niedrig affinen Liganden am Beispiel der Ephedra-Alkaloide

Zur Bestimmung der Bindungsaffinität von Liganden zu den Plasmaproteinen, insbesondere Albumin, wurden über die Jahre zahlreiche Methoden entwickelt. Die Grundlage dieser Arbeit war die Bestimmung der Plasmaproteinbindung der Ephedra-Alkaloide unter Verwendung einzelner dieser etablierten Methoden. Aufgrund ihres Anwendungsgebiets als Notfallmedikation bei Anästhesie-bedingter Hypotonie und den damit verbundenen Anforderungen an die Pharmakokinetik, sollten die Ephedra-Alkaloide niedrig-affine Liganden der Plasmaproteine darstellen. In der Literatur und in vorhergehenden Arbeiten wurden für die Ephedra-Alkaloide jedoch sehr unterschiedliche, teilweise der Indikation widersprechende Affinitäten bestimmt. Daher sollte im Rahmen dieser Arbeit das Ausmaß der Plasmaproteinbindung der Ephedra-Alkaloide weiter untersucht und die Affinität zu Albumin bzw. anderen Plasmaproteinen im humanen Serum bestimmt werden. Neben der Affinität sollte auch die Stereoselektivität der Bindung genauer betrachtet werden, die bei der Bindung vieler Wirkstoffe eine Rolle spielt. Als Referenzmethode diente die kontinuierliche Ultrafiltration, die auch schon bei Hörst verwendet wurde.
Folgende Schlussfolgerungen konnten aus den Ergebnissen dieser Arbeit gezogen werden:
1) Die Ergebnisse der kontinuierlichen Ultrafiltration zeigten, dass die Ephedra-Alkaloide, Ephedrin und Pseudoephedrin, ein nur geringes Ausmaß an Plasmaprotein-bindung von 4 – 9 % gegenüber bovinem und humanem Serumalbumin zeigen. Eine deutlich höhere Plasmaproteinbindung von 19 – 37 % konnte hingegen bei der Verwendung von humanem Serum bestimmt werden. Die Affinität von Pseudoephedrin war dabei jeweils geringer als die von Ephedrin.
2) Diese Ergebnisse mit humanem Serum und die Tatsache, dass Albumin vorwiegend saure Stoffe bindet, legen nahe, dass die Ephedra-Alkaloide vermehrt an andere Plasmaproteine in Serum binden. Erste Messergebnisse mit saurem α1 Glykoprotein bestätigen diese Vermutung.
3) Eine Stereoselektivität konnte nur in geringem Maß bei (+) Ephedrin beobachtet werden, wobei der Unterschied nur im Serum signifikant ist. Pseudoephedrin dagegen zeigte keinerlei Stereoselektivität. Diese Beobachtung passt zu den Schlussfolgerungen der Pfeiffer‘schen Regel zur Stereoselektivität einer Bindung.
4) Andere Sympathomimetika mit einer zusätzlichen Phenolgruppe im Molekül zeigen eine ähnlich niedrige Affinität zu Albumin von ca. 10 %. Eine zusätzliche Phenolgruppe scheint die sauren Eigenschaften des Liganden nicht ausreichend zu erhöhen, um die Affinität zu Albumin signifikant zu steigern.
5) Das tertiäre Kohlenstoffatom am Stickstoff des Ephedrins scheint in gewisser Weise an der Bindung zu Albumin beteiligt zu sein. Sympathomimetika mit einer zusätzlichen Methylgruppe an diesem Kohlenstoffatom, wie Ephedrin, Pseudoephedrin und Oxilofrin, zeigen eine größere Streuung der Messergebnisse. Eine zusätzliche Methylgruppe in dieser Position scheint die Bindung daher sterisch zu hindern.
6) Die Ergebnisse der diskontinuierlichen Ultrafiltration bestätigen weitestgehend die Ergebnisse der kontinuierlichen Ultrafiltration
7) Eine Bestimmung des Ausmaßes der Plasmaproteinbindung von niedrig-affinen Stoffen ist mit den anderen orthogonalen Methoden ACE, NMR und iTC nicht möglich. Diese drei verwendeten Methoden trennen nicht wie die klassischen Methoden den gebundenen vom ungebundenen Wirkstoff, sondern beruhen auf einer Veränderung bestimmter Messparameter: bei der ACE die Migrationszeit, bei der NMR-Spektroskopie die chemische Verschiebung der Signale bzw. der Diffusionskoeffizient und bei der iTC die frei werdende Bindungswärme. Bei allen drei Methoden war die Änderung der Messgröße aufgrund der niedrigen Plasmaproteinbindung zu gering, um auswertbar zu sein.
8) Eine Störgröße bei die orthogonalen Methoden war vielfach auch das Albumin selbst bzw. dessen Eigenschaften. Bei der Affinitäts-Kapillarelektrophorese sind physiologische HSA-Konzentrationen wegen des starken Basislinienrauschens nicht messbar. Zudem bewirkt der Albuminzusatz im Trennpuffer eine Viskositätsänderung, die den EOF verlangsamt und so die Messung stört. Bei der NMR-Spektroskopie können wegen der Überlagerung der Signale durch die breiten Albuminbanden weder Veränderungen in der chemischen Verschiebung noch des Diffusionskoeffizienten zuverlässig bestimmt werden. In der iTC erschwerte die Schaumbildung der Lösung, die durch die Oberflächenaktivität des Albumins verursacht wird, die Messung.
In dieser Arbeit konnte somit das Ausmaß der Plasmaproteinbindung der Ephedra-Alkaloide mit verschiedenen Methoden erfolgreich bestimmt werden. Damit bestätigte diese Arbeit, dass die Ephedra-Alkaloide, wie deren Indikation vermuten lässt, zu den niedrig affinen Liganden des Albumins zählen. Um genauer eingrenzen zu können durch welche Plasmaproteine im Blutserum die Ephedra-Alkaloide transportiert werden, sollten die Untersuchungen zum sauren α1-Glykoprotein fortgesetzt und gegebenenfalls durch weitere Bestimmungen mit anderen Plasmaproteinen ergänzt werden.
Die Ergebnisse dieser Arbeit haben auch gezeigt, dass viele der unzähligen Methoden zur Untersuchung der Plasmaproteinbindung bei der Bestimmung von niedrig affinen Liganden ihre Grenzen haben. Nach wie vor sind zur Bestimmung einer niedrigen Bindungsaffinität weiterhin die klassischen Methoden, wie die kontinuierliche Ultrafiltration, Mittel der Wahl. Nicht zuletzt deshalb erfreuen sich diese Methoden auch heute noch großer Beliebtheit.
Shiga Toxin-produzierende Escherichia coli (STEC) verursachen Diarrhöen und hämorrhagische Colitis. Als lebensbedrohliche Komplikation können sie ein hämolytisch-urämisches Syndrom auslösen. Bisher identifizierte Virulenz-faktoren der STEC sind auf Bakteriophagen, Plasmiden und Pathogenitätsinseln kodiert. Bei der Mehrzahl der klinischen STEC-Isolate konnte die Pathogen-itätsinsel LEE (locus of enterocyte effacement) nachgewiesen werden. Der LEE ist stromabwärts des Gens für eine Selenocystein-tRNA (selC) ins Chromosom integriert. Ziel der vorliegenden Arbeit war die Charakterisierung von STEC, denen der LEE fehlt und die funktionelle Analyse von potentiellen Virulenz-assoziierten Genen. Dabei wurde eine Fokussierung auf Gene vorgenommen, die im Bereich der selC-Region vorkommen. Mittels PCR wurde zunächst überprüft, ob bei den LEE-negativen STEC-Stämmen Fremd-DNA in der selC-Region integriert ist. Hierzu wurden 35 LEE-negative STEC getestet. Bei 13 Stämmen konnte eine Insertion von Fremd-DNA gezeigt werden. Von einem dieser Stämme (E. coli 4797/97, Serovar O91:H-) wurde aus einer Genbank ein die selC-Region enthaltendes DNA-Fragment identifiziert. Ein 37,7 kb großes Fragment dieses Cosmids wurde vollständig sequenziert. Die Analyse der gesamten Sequenz ergab 30 offene Leserahmen mit Längen zwischen 95 und 4092 bp. Der G+C-Gehalt betrug 47,4%. An mehreren Stellen wurden Bereiche mit hoher Homologie zu verschiedenen Insertionssequenzen, inverted repeats und Integrasen gefunden. Drei Leserahmen hatten eine hohe Homologie zu bereits bekannten Genen. Hierbei handelt es sich um die iha- , btuB- und espP-Gene von E. coli. Das iha-Gen kodiert für ein Adhärenz-vermittelndes Protein, btuB für den Vitamin B12 Rezeptor und espP für eine Serinprotease. Bei den iha-Adhäsinen und Serinproteasen handelt es sich um potentielle Pathogenitätsfaktoren. Der sequenzierte Bereich hat somit die charakteristischen Eigenschaften einer Pathogenitätsinsel: Die Präsenz von mehreren Pathogenitätsgenen und mobilen Elementen (Insertionselemente, Integrasen), die Lokalisation nahe an tRNA-Genen sowie ein veränderter G+C-Gehalt gegenüber dem Restgenom. Zur weiteren Charakterisierung des espI-Genprodukts wurde espI subkloniert. Bei der Untersuchung von Kulturüberständen zeigte sich, dass der Subklon DH5/pzh4 ein Protein von etwa 110 kDa sezernierte. In weiteren Experimenten konnte gezeigt werden, dass die Serinprotease des Stammes 4797/97 als 140,8 kDa großes Vorläuferprotein synthetisiert und während des anschließenden Exportvorganges sowohl N-terminal als auch C-terminal prozessiert wird. Das C-terminale Ende wirkt als Translokator durch die äußere Membran, wo es nach Abspaltung des reifen EspI-Proteins verbleibt. Das reife Protein weist eine Größe von 110,5 kDa auf. Der hier gezeigte Transportmechanismus ist charakteristisch für sogenannte Autotransporter-Proteine. Trotz der hohen Sequenzhomologie zur IgA1-Protease von Neisseria gonorrhoeae, die als Prototyp der Autotransporter-Proteine gilt, konnte für EspI keine Proteaseaktivität gegenüber IgA gezeigt werden. EspI weist auch hohe Sequenzhomologie zu Exoproteinen von Shigella flexneri (SepA, Mucinase und SigA), EHEC (EspP) und vogelpathogenen E. coli (Tsh) auf, allerdings konnte keines der Substrate dieser Proteasen von EspI gespalten werden. Dagegen konnte eine proteolytische Aktivität gegenüber Schweine-Pepsin A und Apolipoprotein A1 nachgewiesen werden. Die putative Virulenz und räumliche Nähe der Gene espI, btuB und iha macht diese Region zum interessantesten Stück der im Rahmen dieser Arbeit identifizierten neuen Pathogenitätsinsel. Eine PCR-Untersuchung zur Verbreitung dieser Gene zeigte, dass diese Region bei LEE-negativen STEC weit verbreitet ist. Inwieweit diese Pathogenitätsinsel einen Einfluss auf die Virulenz von STEC hat, muss in weiteren Experimenten geklärt werden.
Many (poly‐)phenolic natural products, for example, curcumin and taxifolin, have been studied for their activity against specific hallmarks of neurodegeneration, such as amyloid‐β 42 (Aβ42) aggregation and neuroinflammation. Due to their drawbacks, arising from poor pharmacokinetics, rapid metabolism, and even instability in aqueous medium, the biological activity of azobenzene compounds carrying a pharmacophoric catechol group, which have been designed as bioisoteres of curcumin has been examined. Molecular simulations reveal the ability of these compounds to form a hydrophobic cluster with Aβ42, which adopts different folds, affecting the propensity to populate fibril‐like conformations. Furthermore, the curcumin bioisosteres exceeded the parent compound in activity against Aβ42 aggregation inhibition, glutamate‐induced intracellular oxidative stress in HT22 cells, and neuroinflammation in microglial BV‐2 cells. The most active compound prevented apoptosis of HT22 cells at a concentration of 2.5 μm (83 % cell survival), whereas curcumin only showed very low protection at 10 μm (21 % cell survival).
Azobenzene derivatives with activity against drug‐resistant Candida albicans and Candida auris

Increasing resistance against antimycotic drugs challenges anti‐infective therapies today and contributes to the mortality of infections by drug‐resistant Candida species and strains. Therefore, novel antifungal agents are needed. A promising approach in developing new drugs is using naturally occurring molecules as lead structures. In this work, 4,4'‐dihydroxyazobenzene, a compound structurally related to antifungal stilbene derivatives and present in Agaricus xanthodermus (yellow stainer), served as a starting point for the synthesis of five azobenzene derivatives. These compounds prevented the growth of both fluconazole‐susceptible and fluconazole‐resistant Candida albicans and Candida auris strains. Further in vivo studies are required to confirm the potential therapeutic value of these compounds.
Although the prevalence of substandard and counterfeit pharmaceutical products is a global problem, it is more critical in resource-constrained countries. The national medicines regulatory authorities (MNRA) in these countries have limited resources to cater for regular quality surveillance programmes aimed at ensuring that medicines in circulation are of acceptable quality. Among the reasons explained to hinder the implementation of these strategies is that compendial monographs are too complicated and require expensive infrastructures in terms of environment, equipment and consumables. In this study it was therefore aimed at developing simple, precise, and robust HPLC and HPTLC methods utilizing inexpensive, readily available chemicals (methanol and simple buffers) that can determine the APIs, other API than declared one, and which are capable of impurity profiling. As an outcome of this study, three isocratic and robust HPLC and two HPTLC methods for sulfadoxine, sulfalene, pyrimethamine, primaquine, artesunate, as well as amodiaquine have been developed and validated. All HPLC methods are operated using an isocratic elution mode which means they can be implemented even with a single pump HPLC system and standard C18 columns. The densitometric sulfadoxine/sulfalene and pyrimethamine method utilizes standard TLC plates as well as inexpensive, readily available and safe chemicals (toluene, methanol, and ethyl acetate), while that for artesunate and amodiaquine requires HPTLC plates as well as triethylamine and acetonitrile due to challenges associated with the analysis of amodiaquine and poorly the detectable artesunate. These HPTLC methods can be implemented as alternative to those requiring HPLC equipment e.g. in countries that already have acquired densitometer equipment. It is understood that HPTLC methods are less sensitive, precise and accurate when compared to HPLC methods, but this hindrance can easily be addressed by sending representative samples to third party quality control laboratories where the analytical results are verified using compendial HPLC methods on a regular basis.
It is therefore anticipated that the implementation of these methods will not only address the problem of limited resources required for medicines quality control but also increase the number of monitored targeted antimalarial products as well as the number of resource- constrained countries participating in quality monitoring campaigns. Moreover, the experiences and skills acquired within this work will be applied to other API groups, e. g. antibiotics, afterwards.
Aspekte der Bestimmung des Ausmaßes der Proteinbindung mittels kontinuierlicher Ultrafiltration

Die Plasmaproteine dienen dem Körper u.a. als Transportproteine. Dem Serumal-bumin kommt hierbei die größte Bedeutung zu. Aufgrund seiner Aufgabe weist es eine Reihe unterschiedlicher Bindungsstellen für eine Vielzahl unterschiedlichster Liganden auf, wobei die Bindung von Substanzen sowohl an spezifischen Stellen als auch unspezifisch erfolgen kann. Das Ausmaß dieser Bindungen hat Einfluss auf andere pharmakokinetische Parameter, wie die Bioverfügbarkeit und die Elimination eines Arzneistoffes. Treten mehrere Stoffe in Wechselwirkung mit dem Albumin, so können sich diese gegenseitig in ihrer Bindung beeinflussen. Das Ausmaß der Beeinflussung ist von der Höhe der Bindung der einzelnen Stoffe und dem zugrunde liegenden Bindungsmechanismus abhängig. Die klinische Relevanz der Beeinflussung hängt von der therapeutischen Breite und von zusätzlich auftretenden Wechselwirkungen, der Substanzen wie z.B. wechselseitige Inhibition der Stoffwechselenzyme der Substanzen ab. Für die experimentelle Bestimmung der Proteinbindung stehen eine Reihe von Me-thoden zur Verfügung, die sich im Messprinzip, dem apparativen Aufwand und der Simulation der physiologischen Bedingungen unterscheiden. In der vorliegenden Arbeit wurde die kontinuierliche Ultrafiltration verwendet. Diese stellt die Bedingungen im Körper nach und ermöglicht die Bindungskinetik besser zu betrachten als andere Verfahren, da durch die Titration des Albumins mit der Arzneistofflösung die Wechselwirkung zwischen Arzneistoff und Protein über einen breiten Konzentrationsverlauf beobachtet werden. Die Methode wurde von Heinze etabliert, von Albert weiterentwickelte und jetzt opti-miert. Durch die Konstruktion neuer Messzellen sowie der Entwicklung eines neuen Puffersystems konnte sie für Substanzen zugänglich gemacht werden, die aufgrund ihrer hohen Lipophilie und ihrer starken Adsorption an die Messzellen bisher nicht messbar waren. Darüber hinaus wurden die folgenden Untersuchungen durchgeführt: a) Es wurde die gegenseitige Verdrängung von zwei Arzneistoffen aus der Proteinbindung untersucht. Dabei konnte gezeigt werden, dass sich eine Reihe basischer Arzneistoffe durch saure Arzneistoffe verdrängen ließ, so z.B. Imipramin HCl durch Phenprocoumon. Das dem Phenprocoumon strukturell sehr ähnliche Warfarin rief diese Verdrängung hingegen nicht hervor. Tryptophan, das in HPLC Experimenten die Bindung von Imipramin HCl verringerte [117], hatte im Ultrafiltrationsexperiment keinen Einfluss auf dessen Bindung. Die gegenseitige Beeinflussung zweier Stoffe kann folglich sehr unterschiedlich sein und ist schwer vorhersagbar. Des Weiteren sind die Ergebnisse solcher Bestimmungen stark von der gewählten Methode abhängig und dadurch nur schwer zu vergleichen. Aufgrund der komplexen und uneinheitlichen Wechselwirkungen der basischen und sauren Arzneistoffe in Bezug auf ihre Bindung, kann davon ausgegangen werden, dass basische Arzneistoffe eine Bindestelle an Albumin besitzen. Jedoch lässt sich aus der vorliegenden Messung nicht beurteilen, wie spezifisch basische Stoffe an Albumin binden. b) Es konnte gezeigt werden, dass unterschiedliche Derivate des Acetylcysteins, die kovalent an Cys34 binden können, unterschiedliche Veränderung im Bindungsverhalten von Albumin hervorrufen. So nahm die Bindung des Diphenhydraminhydrochlorids in Gegenwart von äquimolaren Mengen Acetylcystein und Cysteamin ab, während die Anwesenheit von Acetylcysteamin den gegenteiligen Effekt hatte. Dies zeigt, dass der Veränderung der Bindung ein komplexerer Zusammenhang als die reine Belegung des Cys34 zugrunde liegen muss. c) Unterschiede in der Proteinbindung von Ephedrin und seiner Stereoisomere konnten mittels kontinuierlicher Ultrafiltration bestätigt werden. Die Abweichungen in den Ergebnissen in Bezug auf andere Bestimmungsmethoden [130-132] zeigten erneut, wie stark die Messung der Proteinbindung von den Versuchsbedingungen abhängt. d) Für die Messung des antiinfektiven Bisnaphthalimids MT02 wurde eine Messzelle aus PVDF konstruiert, die weniger Wechselwirkungen mit adsorbierenden Arzneistoffen zeigt. Aufgrund der Unbeständigkeit der Substanz unter den Versuchsbedingungen war die Messung dennoch nicht möglich. e) Die Proteinbindung des Naphthylisochinolins GB AP05 konnte mit Hilfe der bereits erwähnten neu konstruierten Messzelle bestimmt werden. Die Bindung war deutlich höher als bei anderen Vertretern dieser Gruppe. Die Ursache hierfür könnte in der Adsorption der Substanz an die Messzellen begründet sein. Da GB AP05 stark an den Kunststoff PMMA binden kann, der aufgrund seiner Beschaffenheit vorwiegend Wasserstoffbrückenbindungen für eine Adsorption bereitstellen kann, ist eine hohe Bindung an Albumin, das eine Vielzahl von Bindestellen mit unterschiedlichen chemischen Gruppen aufweist, nicht unwahrscheinlich. f) Für eine Reihe von Fluorchinoloncarboxamiden wurde das Ausmaß ihrer Protein-bindung bestimmt. Dieses lag in fast allen Fällen bei 80 % oder höher. Die einzigen Ausnahmen, GHQ237Ox und GHQ243Ox, legen den Schluss nahe, dass eine basi-sche Seitenkette in Position 1 und das Fehlen des Fluorsubstituenten in Position 6 die Bindung reduzieren können. Da nicht alle Substanzen in der Pufferlösung gelöst werden konnten, wurde neben dem bereits durch Albert etablierten DMSO-haltigen Puffer ein Polysorbat 20 haltiger entwickelt, in dem hoch lipophile Substanzen mizellar gelöst werden können. Durch Bestimmung der Proteinbindung von Carbamazepin konnte gezeigt werden, dass die Anwesenheit der Mizellen keinen Einfluss auf die Bindung hat, d.h. im Umkehrschluss dass die Methode zur Bestimmung des Ausmaßes der Proteinbindung geeignet ist.
Dietary fatty acids serve as objective biomarkers for the estimation of habitual diet mainly because biomarkers are free of memory bias or inaccuracies of food databases. The aim of the present work encompassed the implementation of a gas chromatographical method coupled with a mass spectrometrical and flame-ionization detector for analysis of fatty acid biomarkers in human biospecimens, their analytical determination and statistical evaluation in two different study populations and different biospecimens as well as the elaboration of adverse reactions to food ingredients with special focus on food allergies and food intolerances in the context of a possible implementation into an application for consumer health. The first aim was the identification of potential influence of fatty acid biomarkers on desaturase and elongase indexes (Δ9DI, Δ6DI, Δ5DI and ELOVLI5), which are factors in type 2 diabetes risk, in breast adipose tissue from healthy women. Influence of further variables on respective indexes was also investigated. 40 samples were investigated and potential variables were either collected by questionnaire or determined. Principle component analysis was applied for fatty acid biomarkers (PCdiet1, PCdiet2 and PCdiet3 representative for the dietary intake of vegetable oils/nuts, fish and partially hydrogenated vegetable oils), endogenous estrogens (PCE1) and oxysterols (PCOxy1). Multiple linear regression models were applied. Δ9DI and Δ6DI were influenced non-significantly and significantly negatively by PCdiet2 supporting a putative beneficial effect of vegetable oils and nuts on type 2 diabetes risk factors. ELOVLI5 and Δ5DI were influenced significantly and non-significantly positively by PCdiet1 supporting a putative beneficial effect of fish consumption on type 2 diabetes risk factors. On the other hand, PCdiet1 also significantly and non-significantly positively influenced Δ9DI and Δ6DI supporting a putative adverse effect of fish biomarkers on type 2 diabetes risk factors. The opposing influences of PCdiet1 suggesting an ambivalent role of dietary intake of fish on investigated indexes. Δ6DI was significantly positively influenced by PCdiet3 and number of pregnancies supporting a putative adverse effect of partially hydrogenated vegetable oils and pregnancies on type 2 diabetes risk factors. Lifestyle factors like smoking significantly and non-significantly influenced Δ9DI and Δ6DI putatively adversely. Δ5DI was influenced significantly positively by estrogen active drugs suggesting a putative beneficial effect on type 2 diabetes risk factors. It must be considered that a variation coefficient of up to 0.44 only explained 44% of variance of the respective indexes, suggesting other influencing factors might play a role. The second aim was the implementation of a gas chromatographical method coupled with a mass spectrometrical and flame-ionization detector for analysis of fatty acid biomarkers in human biospecimens. The method was optimized for separation and detection of 40 fatty acids. Mean recovery for tridecanoic acid was x(tridecanoic acid) = 90.51% and for nonadecanoic acid x(nonadecanoic acid) = 96.21%. Thus, there was no significant loss of fatty acids with shorter and longer carbon chains over the extraction process to be expected. Limit of detections were calculated in adipose tissue samples and ranged from 0.007 to 0.077% of the proportion of the respective fatty acid to total fatty acids. The third aim was the investigation if differentiation between breast glandular and adipose tissue had a relevant impact on the analysis of dietary fatty acid biomarkers or if contamination of breast glandular with breast adipose tissue and vice versa was neglectable for the analysis of dietary fatty acid biomarkers. No statistical significant differences were observed for all investigated fatty acid biomarkers (pentadecanoic-, heptadecanoic-, trans palmitoleic-, eicosapentaenoic-, docosahexaenoic-, linoleic and α-linolenic acid) between breast glandular and adipose tissue. Thus, differentiation between breast glandular and adipose tissue seems not to be necessary for the analysis of fatty acids serving as biomarkers for the intake of specific food groups. Potential influence of mixed breast tissue on fatty acid biomarkers analysis seems to be neglectable. The fourth aim was the determination of fatty acid biomarkers in adipose tissue in another study population from healthy participants. 27 adipose tissue samples were analyzed. Milk and ruminant fat biomarkers exhibited proportions of 0.47% for pentadecanoic acid, 0.34% for heptadecanoic acid and 0.25% for trans palmitoleic acid. Fish fatty acid biomarkers revealed proportions of 0.034% for eicosapentaenoic acid and 0.061% for docosahexaenoic acid. The mean proportion of vegetable oils and nuts biomarkers were 9.58% for linoleic acid and 0.48% for α-linolenic acid in all adipose tissues. Principle component analysis was applied for the fatty acid biomarkers to provide objective markers of habitual diet for this study population. PCdiet1 was mainly characterized by pentadecanoic acid, heptadecanoic acid and trans palmitoleic acid and therefore served as a principle component for the dietary intake of milk and ruminant fat. PCdiet2 and PCdiet3 only exhibited pattern for ω3 and ω6 fatty acids but not for dietary intake of specific food groups and could therefore not used as objective marker. PCdiet1, 2 and 3 explained 82.76% of variance. The last aim of this thesis was the elaboration of adverse reactions to food ingredients with special focus on food allergies and food intolerances in the context of a possible implementation into an application for consumer health. Scientific information on adverse reactions to food ingredients and trigger substances was provided in this thesis and possible implementation strategies were evaluated. For food allergens, which have regulatory requirements in the context of labelling, a strategy was elaborated, where it is necessary to provide information on the list of ingredients, the nexus ’contain’ and the respective food allergen as well as information on the name of the product. For food intolerances, which do not have regulatory requirements, limits were shown in the context of the application. If the elaborated food intolerances shall be implemented into the application, a professional dietary concept has to be developed for every food intolerance because of the complexity of the implementation.
Die oralen Antidiabetika Metformin und Sitagliptin werden überwiegend renal eliminiert, weshalb während der Therapie regelmäßig die Nierenfunktion abgeschätzt werden sollte. Dies geschieht mithilfe von Serumkreatinin-basierten Formeln, zum Beispiel der Gleichung nach Cockcroft-Gault.
Mit dem Ziel, zukünftig eine Möglichkeit für eine vereinfachte Kontrolle der Therapie mit Metformin und/oder Sitagliptin in Kapillarblutproben zu haben, wurde eine Methode zur Bestimmung der Konzentration von Kreatinin, Metformin und Sitagliptin aus Trockenblutproben (Dried Blood Spots, DBS) entwickelt. Als Träger zeigte Blotting Papier die besten Ergebnisse in Bezug auf die Handhabung und die Extraktionseffizienz. Aus einem einzelnen DBS gelang es, Metformin und Kreatinin mittels HPLC-UV und Sitagliptin mittels LC-MS/MS zu quantifizieren. Die flüssigchromatographischen Methoden wurden entsprechend der EMA- und FDA-Kriterien erfolgreich vollvalidiert. Die unteren Nachweisgrenzen (LLOQ) lagen bei 0,2 µg/mL für Metformin, 1,5 µg/mL für Kreatinin und 3 ng/mL für Sitagliptin.
Da Referenzbereiche für Arzneistoffkonzentrationen in der Regel für Serum/Plasma angegeben werden, wurde das Verteilungsverhalten der beiden Antidiabetika zwischen Plasma (cP) und Blutzellen (cBZ) mittels in-vitro Inkubationsversuchen ermittelt. Für Metformin betrug der Verteilungskoeffizient cP/cBZ 4,65 ± 0,73, für Sitagliptin 5,58 ± 0,98. Damit lagen beide Arzneistoffe mehr als 4-fach höher im Plasma als in den Blutzellen vor. Erythrozyten waren zuvor schon als tiefes Kompartiment für Metformin beschrieben worden, für Sitagliptin waren dieses die ersten Daten die zeigten, dass der Arzneistoff ebenfalls eine relevante Verteilung in die Blutzellen zeigt.
In Kooperation mit einer diabetologischen Schwerpunktpraxis wurde eine erste klinische Studie (Basisstudie) durchgeführt, die zum Ziel hatte, aus den DBS die Nierenfunktion abzuschätzen. In DBS von 70 Patienten wurden Metformin, und/oder Sitagliptin sowie Kreatinin quantifiziert. Mit Hilfe der von der Praxis übermittelten Serumkreatinin-konzentration konnte durch den Vergleich mit der Konzentration im Kapillarbut erstmalig ein Korrelationsfaktor bestimmt und verifiziert werden, um die Kapillarblut- in die Serumkonzentration des Kreatinins umzurechnen (F = cKapillarblut/cPlasma = 0,916 ± 0,088). So war es möglich, die Nierenfunktion über die Formel nach Cockcroft und Gault abzuschätzen.
In der Basisstudie fiel auf, dass die Konzentration des Sitagliptins im Blut der Patienten signifikant mit steigendem Hämatokrit korrelierte (Pearson R = 0,396; p < 0,05). Die nähere Untersuchung dieser Beobachtung mittels in-vitro Verteilungsversuchen zeigte eine sehr stark inter-individuell schwankende Verteilung des Sitagliptins zwischen Plasma und den Blutzellen und eine vom Hämatokrit (Hct) linear abhängige Verteilung. In Blut mit einem höheren Hct fand sich mehr Arzneistoff in den Blutzellen als in Blut mit niedrigerem Hct, was die höheren Gesamtkonzentrationen an Sitagliptin im DBS erklärte. Dialyseversuche in-vitro bestätigten, dass die Eliminationszeit mit steigendem Hämatokrit des Blutes anstieg. Damit konnte zum ersten Mal gezeigt werden, dass die Blutzellen ein tiefes Kompartiment für Sitagliptin darstellen.
Eine zweite klinische Studie (Feldstudie) wurde in Kooperation mit 14 öffentlichen Apotheken mit dem Ziel, repräsentative Konzentrationen für die Kapillarblutspiegel der beiden Medikamente unter Alltagsbedingungen zu ermitteln, durchgeführt. In DBS von 84 Patienten wurden wiederum Metformin, Sitagliptin und Kreatinin quantifiziert. Aus den Daten der beiden Studienpopulationen (n = 134) wurde für Metformin eine mittlere Konzentration von 2,22 ± 1,16 µg/mL und für Sitagliptin von 432,20 ± 268,79 ng/mL bestimmt. Mittels populationspharmakokinetischer Methoden konnten für beide Arzneistoffe zum ersten Mal Eliminationshalbwertszeiten (t1/2) aus Kapillarblut für Patienten mit einer Kreatininclearance größer und kleiner als 60 mL/min bestimmt werden. Erwartungsgemäß waren die t1/2 bei besserer Nierenfunktion kürzer, sowohl für Metformin (11,9 h versus 18,5 h) als auch für Sitagliptin (8,4 h versus 13,0 h). Für Sitagliptin waren dies erstmalige klinische Belege für eine ansteigende Eliminationszeit mit sinkender Nierenfunktion.
Die gewonnenen Daten boten zudem Gelegenheit, den literaturbekannten ungünstigen Effekt einer kombinierten Einnahme von Diuretika, NSAIDs, ACE-Inhibitoren und/oder Angiotensinrezeptorantagonisten („target drugs“) auf die Nierenfunktion („triple whammy“) zu betrachten. Tatsächlich korrelierten die Anzahl der eingenommenen „target drugs“ und auch die Dosis der Diuretika mit einer sinkenden Kreatininclearance der Patienten.
Mit vorliegender Arbeit wurden zum einen neue Erkenntnisse über die Pharmakokinetik des Sitagliptins gewonnen, zum anderen wurde die Grundlage geschaffen, um aus einem DBS die Blutspiegel von Metformin und Sitagliptin im Zusammenhang mit der Nierenfunktion zu betrachten. In Zukunft könnte diese Methode für ein Therapeutisches Drug Monitoring der beiden Arzneistoffe eingesetzt werden um dieses für Patienten aufgrund der minimalinvasiven Blutabnahme wesentlich angenehmer zu gestalten.
Marine sponges and their associated bacteria have been proven to be a rich source of novel secondary metabolites with therapeutic usefulness in infection and autoimmunity. This Ph.D. project aimed to isolate bioactive secondary metabolites from the marine sponges Amphimedon compressa, Aiolochroia crassa and Theonella swinhoei as well as from bacteria associated with different Caribbean sponges, specifically actinomycetes and sphingomonads. In this study, amphitoxin was isolated from the crude methanol extract of the sponge A. compressa and it was found to have antibacterial and anti-parasitic activities. Amphitoxin showed protease inhibitory activity when tested against the mammalian protease cathepsin B and the parasitic proteases rhodesain and falcipain-2. Furthermore, miraziridine A was identified in the dichloromethane extract of the sponge T. swinhoei collected offshore Israel in the Red Sea. Miraziridine A, a natural peptide isolated previously from the marine sponge Theonella aff. mirabilis, is a potent cathepsin B inhibitor with an IC50 value of 1.4 g/mL (2.1 M). Secondary metabolites from sponge-derived bacteria were also isolated and identified. A total of 79 strains belonging to 20 genera of the order Actinomycetales and seven strains belonging to two genera of the order Sphingomonadales were cultivated from 18 different Caribbean sponges and identified by 16S rRNA gene sequencing. Seven of these strains are likely to represent novel species. Crude extracts from selected strains were found to exhibit protease inhibition against cathepsins B and L, rhodesain, and falcipain-2 as well as immunomodulatory activities such as induction of cytokine release by human peripheral blood mononuclear cells. The isolates Sphingobium sp. CO105 and Lapillicoccus sp. BA53 were selected for cultivation, extraction and purification of bioactive metabolites based on initial bioactive screening results. The isoalloxazine isolumichrome was isolated from the strain Sphingobium sp. CO105 which inhibited the protease rhodesain with an IC50 of 0.2 M. The strain Lapillicoccus sp. BA53 was found to produce p-aminosalicylic acid methyl ester, which showed activity against the proteases cathepsins B and L, falcipain-2 and rhodesain. These results highlight the significance of marine sponge-associated bacteria to produce bioactive secondary metabolites with therapeutic potential in the treatment of infectious diseases and disorders of the immune system.
Bioassay-guided fractionation of a chloroform extract of Valeriana wallichii (V. wallichii) rhizomes lead to the isolation and identification of caffeic acid bornyl ester (1) as the active component against Leishmania major (L. major) promastigotes (IC50 = 48.8 µM). To investigate the structure-activity relationship (SAR), a library of compounds based on 1 was synthesized and tested in vitro against L. major and L. donovani promastigotes, and L. major amastigotes. Cytotoxicity was determined using a murine J774.1 cell line and bone marrow derived macrophages (BMDM). Some compounds showed antileishmanial activity in the concentration range of pentamidine and miltefosine which are the standard drugs in use. In the L. major amastigote assay compounds 15, 19 and 20 showed good activity with relatively low cytotoxicity against BMDM, resulting in acceptable selectivity indices. Molecules with adjacent phenolic hydroxyl groups exhibited elevated cytotoxicity against murine cell lines J774.1 and BMDM. The Michael system seems not to be essential for antileishmanial activity. Based on the results compound 27 can be regarded as new lead structure for further structure optimization
The chloroform extract of Valeriana wallichii (V. wallichii) rhizomes was investigated to elucidate the structures responsible for reported antileishmanial activity. Besides bornyl caffeate (1, already been reported by us previously), bioassay-guided fractionation resulted in two additional cinnamic acid derivatives 2–3 with moderate leishmanicidal activity. The structure of a novel nepetolactone derivative 4 having a cinnamic acid moiety was elucidated by means of spectral analysis. To the best of our knowledge villoside aglycone (5) was isolated from this plant for the first time. The bioassay-guided fractionation yielded two new (compounds 6–7) and two known valtrates (compounds 8–9) with leishmanicidal potential against Leishmania major (L. major) promastigotes. In addition, β-bisabolol (10), α-kessyl alcohol (11), valeranone (12), bornyl isovalerate (13) and linarin-2-O-methylbutyrate (14) were identified. This is the first report on the isolation of 4'-demethylpodophyllotoxin (15), podophyllotoxin (16) and pinoresinol (17) in V. wallichii. In total thirteen known and four new compounds were identified from the extract and their cytotoxic and antileishmanial properties were evaluated.
Antiinflammatorische Wirkungen und Pharmakokinetik eines standardisierten Kiefernrindenextraktes

In der vorliegenden Arbeit wurde erstmalig die Hemmung des induzierten Abbaus von Matrixproteinen sowie von Gelatine durch Matrixmetalloproteinasen mithilfe des Kiefernrindenextraktes Pycnogenol, einer Auswahl seiner Inhaltsstoffe und seiner Metabolite d-(3,4-Dihydroxyphenyl)-g-valerolacton (M1) und d-(3-Methoxy-4-hydroxyphenyl)-g-valerolacton (M2) untersucht. Beide Metabolite zeigten eine signifikante Hemmwirkung und lagen in der Effektivität ihrer Hemmung auf einer µg/ml-Basis immer leicht über der des Gesamtextraktes. Um die in Frage kommenden Mechanismen der Hemmwirkung gegenüber Matrixmetalloproteinasen aufzuklären, wurde die Bindung des Gesamtextraktes an Hautpulver, sowie an die Matrixproteine Collagen und Elastin untersucht. Es wurde ein Schutz der Substrate vor enzymatischer Degradierung durch MMPs infolge einer Adsorption der Procyanidine abgeleitet. Die Metabolite M1 und M2 schienen auf Grund eines anderen Mechanismus die Aktivität der MMPs zu hemmen. Es konnte gezeigt werden, dass die Hemmwirkung der untersuchten Inhibitoren auf MMP-9 nach Zinkzusatz vollständig aufgehoben wurde. Daher konnte eine direkte Interaktion von beiden Metaboliten auf das Zinkatom des aktiven Zentrums angenommen werden. Die Wirkungen der Metabolite M1 und M2 wurden anschließend auf zellulärer Ebene untersucht. Es wurde getestet, ob sie einen Einfluss auf die Sekretion von MMP-9 aus bakteriellen Lipopolysaccharid (LPS)-stimulierten Monocyten hatten. Als Positivkontrolle wurden antiinflammatorisch wirkenden PPAR-Agonisten, sowie das endogene Glucocorticoid Hydrocortison eingesetzt, deren Hemmwirkung auf die MMP-9-Freisetzung gut belegt war. Die PPAR-Agonisten waren in Bezug auf die MMP-9-Sekretion die wirksamsten Inhibitoren. Mit einer bemerkenswert niedrigen IC50 waren die beiden Metabolite M1 und M2 in ihrer Wirkung equipotent. Im Vergleich zu Hydrocortison konnte sogar gezeigt werden, dass beide Metabolite potentere Inhibitoren darstellten als das körpereigene antiinflammatorisch wirksame Glucocorticoid. In pharmakokinetischen Untersuchungen wurden Plasmaproben von Probanden nach Einmal- (n = 11) und Mehrfach- (n = 5) Einnahme von 300 bzw. 200 mg Pycnogenol mithilfe der HPLC vermessen. Durch diese Untersuchungen sollte festgestellt werden, ob eine Absorption von Extraktbestandteilen, sowie Metabolisierungsreaktionen im Körper stattfinden. Es konnten in den Proben der meisten Probanden erstmalig im Plasma sowohl Extraktbestandteile als auch der Metabolit M1 nachgewiesen werden. Daneben konnten zehn bislang unbekannte Substanzen detektiert werden, die in weiterführenden Arbeiten noch identifiziert werden müssen. Es wurde eine große interindividuelle Variabilität sowohl im Grad der Konjugation als auch bei den im Plasma vorliegenden Konzentrationen der einzelnen Substanzen festgestellt. Nach Einmalgabe des Extraktes wurden verschiedene Gruppen von Substanzen mit frühen, mittleren, späten und interindividuell sehr variablen Plasmaspiegelmaxima nachgewiesen. Es konnten erstmalig nach Pycnogenol-Einnahme pharmakokinetische Parameter der bekannten und im Gesamtextrakt quantifizierbaren Verbindungen errechnet werden. Nachfolgend wurde in den Plasmaproben der Probanden nach Pycnogenol-Einnahme nachgewiesen, dass Wirksubstanzen in Konzentrationen vorhanden waren, die tatsächlich pharmakodynamische Effekte erzielen konnten. Dazu wurde ein neues Versuchskonzept erstellt, bei dem die Hemmung der MMP-9-Sekretion auf zellulärer Ebene mit den Plasmaproben der Studienteilnehmer vor und nach Pycnogenol-Einnahme ex vivo untersucht wurde. Nach Mehrfachgabe bewirkten die verdünnten Plasmaproben der Probanden eine signifikant verminderte MMP-9-Sekretion um im Mittel 25 %. Plasmaproben nach einmaliger Einnahme von 300 mg Pycnogenol riefen schon 30 Minuten nach Extrakt-Einnahme eine Hemmung der MMP-9-Sekretion hervor. Diese Hemmung war bis 14 Stunden nach der Einnahme nachzuweisen. Einen wichtigen Transkriptionsfaktor im Entzündungsgeschehen stellt NF-kB dar. Stimuliert durch verschiedene Agenzien führt NF-kB u.a. zur Induktion von MMP-9. Nach Mehrfachgabe konnte in ex vivo Versuchen mit den Plasmaproben der Studienteilnehmer nach Inkubation mit stimulierten Monocyten eine etwa 15 %ige Hemmung der NF-kB-Aktivierung gezeigt werden. Es konnte am Beispiel von MMP-9 und NF-kB erstmalig gezeigt werden, dass nach Einnahme von Pycnogenol auch in vivo Konzentrationen an Metaboliten bzw. Bestandteilen erreicht werden, die ex vivo in der Lage waren, pharmakodynamische Effekte zu erzielen. Bislang konnte jedoch keine Zuordnung der pharmakokinetisch identifizierten Verbindungen zu den pharmakodynamischen Effekten erfolgen. Die umfassenden in vitro, ex vivo und in vivo Untersuchungen von Bestandteilen und/oder Metaboliten des Pycnogenol-Extraktes in der vorliegenden Arbeit leisten auf molekularer und auf zellulärer Ebene einen grundlegenden Beitrag zum Verständnis der klinischen antiinflammatorischen Effekte des Kiefernrindenextraktes.
Bornyl caffeate (1) was previously isolated by us from Valeriana (V.) wallichii rhizomes and identified as an anti-leishmanial substance. Here, we screened a small compound library of synthesized derivatives 1–30 for activity against schistosomula of Schistosoma (S.) mansoni. Compound 1 did not show any anti-schistosomal activity. However, strong phenotypic changes, including the formation of vacuoles, degeneration and death were observed after in vitro treatment with compounds 23 (thymyl cinnamate) and 27 (eugenyl cinnamate). Electron microscopy analysis of the induced vacuoles in the dying parasites suggests that 23 and 27 interfere with autophagy.
Bei N-Acyl-Ethanolaminphosphaten handelt es sich um eine bislang wenig untersuchte Klasse polarer Substanzen, deren Erforschung aufgrund ihrer strukturellen Analogie zu apolaren, physiologisch wirksamen N-Acyl-Ethanolaminen von Interesse ist. Zu bear-beiten waren analytische Fragestellungen, die auch synthetische Aufgaben beinhalteten, wie Methodenentwicklung und Versuche zur Erfassung von N-Acyl-Ethanolamin-phosphaten in ausgewählten Lebensmitteln sowie strukturelle Studien zur „Bioaktivität“ der Verbindungen. Erstes Ziel der vorliegenden Arbeit war es demzufolge, eine geeig-nete Methode für deren qualitative und quantitative Analytik zu entwickeln. Gleichzei-tig wurden ausgewählte N-Acyl-Ethanolaminphosphate synthetisiert. Aufgrund des literaturbekannten Vorkommens von N-Acyl-Ethanolaminen in Wein wurden für die Lebensmitteluntersuchungen fermentierte Produkte, d.h. drei verschie-dene Sake (Japanischer Reiswein) und ein fermentierter Rotkohl verwendet. Parallel zu diesen Untersuchungen erfolgten auch Studien zur Stabilität der N-Acyl-Ethanolamin-phosphate. Versuchsreihen zur Überprüfung potentieller „Bioaktivität“ umfassten Studien mit al-kalischer Phosphatase, PhospholipaseA2, Lipoxygenase, Xanthinoxidase, β-N-Acetyl-hexosaminidase und dem Cannabinoidrezeptor-1.
Catechine gehören als Flavan-3-ole zur Gruppe der Polyphenole. Aufgrund deren vielfältiger positiver Effekte auf den menschlichen Organismus nehmen sie in der Ernährungsforschung einen hohen Stellenwert ein. Dabei hat man bei den Flavan-3-olen meist nur die in der Natur vorherrschenden Isomere (+)-Catechin und (-)-Epicatechin untersucht, doch auch (-)-Catechin und (+)-Epicatechin sind Naturstoffe. Letztere findet man z.B. in Guarana oder in verarbeiteten Lebensmitteln, wie z.B. Kakao- und Kakaoerzeugnissen. Sie entstehen durch Epimerisierung unter den technologischen Bedingungen beim Rösten der Kakaobohnen und der Alkalisierung der Kakaomasse. Bei der Kakao-Verarbeitung werden ferner auch Catechin-C-Glykoside gebildet. Im ersten Teil dieser Arbeit wurden Stabilitätsstudien mit (+)-Catechin bei unterschiedlichen pH-Werten und Temperaturen durchgeführt. Der zweite Teil dieser Arbeit umfasst Untersuchungen von Catechin-Isomeren und zwei Catechin-C-Glykosiden auf ihren Einfluß auf die Lipoxygenase (LOX)- und Xanthinoxidase (XOD)-Aktivität. Für die Catechin-C-Glykosidbildung ist von uns eine neue Vorstellung zu deren Entstehungsmechanismus im Laufe der Lebensmittelverarbeitung entwickelt worden. Abschließend wurden anhand von Modelling-Studien die Effekte auf die Enzymsysteme erklärt.
An alternative to the time-consuming and error-prone pharmacopoeial gas chromatography method for the analysis of fatty acids (FAs) is urgently needed. The objective was therefore to propose a robust liquid chromatography method with charged aerosol detection for the analysis of polysorbate 80 (PS80) and magnesium stearate. FAs with different numbers of carbon atoms in the chain necessitated the use of a gradient method with a Hypersil Gold C\(_{18}\) column and acetonitrile as organic modifier. The risk-based Analytical Quality by Design approach was applied to define the Method Operable Design Region (MODR). Formic acid concentration, initial and final percentages of acetonitrile, gradient elution time, column temperature, and mobile phase flow rate were identified as critical method parameters (CMPs). The initial and final percentages of acetonitrile were fixed while the remaining CMPs were fine-tuned using response surface methodology. Critical method attributes included the baseline separation of adjacent peaks (α-linolenic and myristic acid, and oleic and petroselinic acid) and the retention factor of the last compound eluted, stearic acid. The MODR was calculated by Monte Carlo simulations with a probability equal or greater than 90%. Finally, the column temperature was set at 33 °C, the flow rate was 0.575 mL/min, and acetonitrile linearly increased from 70 to 80% (v/v) within 14.2 min.
In all the projects presented, it is evident that the selection of suitable separation conditions is only one side of the coin. Equally crucial in the development of methods for the quality assessment of APIs/drugs is the right detection system.
The application of CAD as an alternative to UV detection at low wavelength of the two weak chromophore main degradation products of the very polar, zwitterionic API carbocisteine requires the volatility of the mobile phase. Therefore, as a substitute for the non-volatile ion pairing reagent tetrabutylammonium hydroxide (TBAOH), six different volatile alkylamines as well as a RP/SAX mixed-mode column were evaluated. The best selectivity and separation performance comparable to TBAOH was achieved with the RP/SAX column and a mixture of formic acid and trifluoroacetic acid. For the simultaneous optimisation of the evaporation temperature of the CAD as a function of two chromatographic parameters, a central composite design was chosen and the “desirability function” was subsequently applied for modelling. In addition, column bleeding was investigated with a second RP/SAX column (different batch) with the result that the acetonitrile percentage had to be adjusted and preconditioning by injection of concentrated samples is essential. The final mixed-mode method was finally validated with both columns according to the ICH Q2 (R1) guideline.
Based on this, an MS-compatible method was developed with little effort using an identical RP/SAX column in UPLC dimension for the untargeted analysis by HRMS of two carbocisteine-containing prototype syrup formulations. For a comprehensive characterisation, HRMS and MS/HRMS data were recorded simultaneously by information dependent acquisition mode. Based on the exact masses, isotope patterns and an in silico plausibility check of the fragment spectra, the prediction of the structures of the unknown impurities was possible. In both syrup samples, which had been stored for nine months at 40 °C and 75 % r.h., two additional impurities of carbocisteine (i.e. lactam of the sulfoxides and disulphide between cysteine and thioglycolic acid) were identified by comparison with the corresponding prototype placebo samples using general unknown comparative screening. In addition, the formation of Maillard products by binary mixtures with 13C-labelled sugars was revealed in the sucrose-containing formulation.
For the promising hyphenation of the UV detector with the CAD for the simultaneous detection of all UV-active impurities of the cholesterol-lowering drug simvastatin and the only weak chromophore dihydrosimvastatin, the Ph. Eur. method had to be adapted. Besides replacing phosphoric acid with trifluoroacetic acid, the gradient also had to be adjusted and a third critical peak pair was observed. Based on validation experiments (according to the ICH Q2 (R1) guideline), the suitability of the CAD for sensitive detection (LOQ = 0.0175 % m/m) was proven.
To further investigate the robustness of the adapted method and CAD, a Plackett-Burman design was chosen. None of the factors had a statistically significant effect on the S/N of the CAD in the ranges tested. Regarding the three critical peak pairs, on the other hand, the factors to be controlled were statistically established, so that a targeted correction is possible if the system suitability test is not passed. The idea of employing a hyphenated UV-CAD system was finally applied to the structurally closely related lovastatin and its specified impurity dihydrolovastatin. Here, the CAD showed a significantly better S/N compared to the compendial UV detection at 200 nm.
The suitability of CAD for the analysis of non-volatile fatty acids in polysorbate 80 (PS80) as favourable alternative to the Ph. Eur. GC method (no time-consuming, error-prone and toxic derivatisation) has already been demonstrated. The aim of this project was therefore to develop a robust method with a focus on the AQbD principles, which can be used for the analysis of other excipients with similar fatty acid composition. After the definition of the analytical target profile and a risk assessment by means of an Ishikawa diagram, a suitable C18 column and the chromatographic framework conditions (formic acid concentration and initial/final gradient conditions) were selected after only few preliminary runs. The remaining critical method parameters were then investigated with the help of DoE and RSM. Using the obtained model equations, Monte Carlo simulations were performed to create the method operable design region as a region of theoretical robustness. After validation according to ICH Q2 (R1), the fatty acid composition of a magnesium stearate batch was successfully analysed as a further application example in addition to PS80.
The CAD was able to prove its potential in all the issues investigated in the context of this doctoral thesis. As a cost-effective alternative compared to MS instruments, it thus closes a gap in the quality assessment of APIs or excipients without a suitable chromophore. The easy method transfer to (HR)MS instruments also allows for a unique degree of sample characterisation through untargeted approaches in case of new impurities. For resource- and time-efficient work, the possibilities and limitations of software tools for method development and data evaluation as well as the application of risk-based approaches such as AQbD should also be considered.
In 1998, the aminoglycoside antibiotic gentamicin sulfate caused several cases of deaths in the United States, after the switch from twice- to once-daily application. Endotoxins were discussed as the cause for the adverse effects and sisomicin was identified as the lead impurity; batches containing sisomicin were contaminated with more impurities and were responsible for the fatalities. In 2016, anaphylactic reactions in horses, and later in humans with one fatality, were observed after application of gentamicin sulfate contaminated with histamine. To determine whether histamine was responsible for the 1990s death cases as well, histamine was quantified by means of liquid chromatography–tandem mass spectrometry (LC-MS/MS) in 30 samples of gentamicin sulfate analyzed in previous studies. Furthermore, a relative quantification of sisomicin was performed to check for a correlation between histamine and the lead impurity. A maximum amount of 11.52 ppm histamine was detected, which is below the limit for anaphylactic reactions of 16 ppm, and no correlation of the two impurities was observed. However, the European Medicines Agency recommends a stricter limit with regard to the maximum single dose of gentamicin sulfate to reach a greater gap between the maximum histamine exposition of 4.3 µg and the quantity known to cause hypotension of 7 µg. The low amounts of histamine and the fact that there is no connection with the contamination with sisomicin showed that histamine was not the cause for the death cases in the United States in 1998, and endotoxins remain the most probable explanation.
Aims: The aim of the current study was to establish a simple and yet as much as possible physiologic approach for a simulation of the pulmonary absorption process to compare different inhaled drugs or drug formulations.
Methodology: We designed a dialysis setting that allowed monitoring the drug release from human lung tissue into a continuous-flow plasma compartment. For proof-of-concept experiments we chose the glucocorticoid fluticasone propionate (FP) as model compound. For subsequent experiments we selected a commercially available metered dose inhaler delivering a fixed combination of the short-acting ß2-agonist fenoterol and the muscarinic antagonist ipratropium bromide.
Results: With the novel dynamic dialysis model we observed high drug transport rates from the lung tissue into plasma including an elimination phase. The concentration profile in the plasma compartment of our model system was similar to the plasma concentration courses after inhalation of FP. Compared to FP significantly higher drug fractions of fenoterol and ipratropium bromide were released into plasma and the transfer of ipratropium was more pronounced compared to fenoterol. Again, concentration profiles in plasma were alike to those described in clinical studies.
Conclusion: We suggest that this model is appropriate for rapid assessment of comparative diffusion behaviour of drugs or drug formulations from lung tissue into plasma.
Estrogens, namely 17β-estradiol (E2) and estrone (E1) are considered to play an important role in the initiation and promotion of breast cancer (summarized in Raftogianis et al., 2000), a malignancy responsible for around 500,000 deaths per year (summarized in Ghislain et al., 2016). Two major mechanisms have been postulated to explain the carcinogenic effects of estrogens: (1) the estrogen receptor-mediated stimulation of breast cell proliferation with a concomitant enhanced rate of mutations and (2) the metabolism of hydroxylated estrogens to quinone derivatives which can react with the DNA (Russo and Russo, 2006, summarized in Yager and Davidson, 2006). Nevertheless, as a detoxifying mechanism, E1, E2, and their hydroxylated and methoxylated metabolites are reversibly conjugated into sulfates and glucuronides devoid of biological activity (summarized in Guillemette et al., 2004). Yet, despite the key detoxifying function of these conjugates, the study of their circulating levels face some significant problems: (1) analysis by techniques such as radioimmunoassay lack specificity and accuracy and requires enzymatic/chemical hydrolysis before analysis, being unable to differentiate between sulfates and glucuronides (summarized in Stanczyk et al., 2007, summarized in Wang et al., 2016), (2) very little knowledge in healthy women, which has been identified as a barrier to advance in breast cancer research (summarized in Liu, 2000), and (3) far fewer studies in pre- than in postmenopausal women (summarized in Samavat and Kurzer, 2015). Therefore, to get more insights into the research of breast cancer etiology and prevention, the analysis of circulating levels of estrogens (including metabolites and conjugates) in women without breast cancer through reliable analytical techniques, is required.
Analysis of Drug Impurities by Means of Chromatographic Methods: Targeted and Untargeted Approaches

The presented works aimed on the analysis of new impurities in APIs and medicinal products. Different subtypes of LC were coupled to suitable detection methods, i.e. UV and various MS techniques, depending on the chemical natures of the analytes and the analytical task.
Unexpected impurities in medicinal products and APIs caused several scandals in the past, concomitant with fatalities or severe side effects in human and veterinary patients. The detection of nitrosamines in sartans led to the discovery of nitrosamines in various other drugs, of which the antibiotic rifampicin was analyzed in this work. An examination of the synthesis of rifampicin revealed a high potential for the formation of 4-methyl-1-nitrosopiperazine (MeNP). An LC-MS/HRMS method suitable for the quantification of MeNP was applied in the analysis of drugs collected from Brazil, Comoros, India, Nepal, and Tanzania, where a single dose of rifampicin is used in the post-exposure prophylaxis of leprosy. All batches were contaminated with MeNP, ranging from 0.7-5.1 ppm. However, application of rifampicin containing up to 5 ppm MeNP was recommended by the regulatory authorities for the post-exposure prophylaxis of leprosy.
In the 1990s the aminoglycoside antibiotic gentamicin attracted attention after causing fatalities in the USA, but the causative agent was never identified unequivocally. The related substance sisomicin was recognized as a lead impurity by the Holzgrabe lab at the University of Würzburg: sisomicin was accompanied by a variety of other impurities and batches containing sisomicin had caused the fatalities. In 2016, anaphylactic reactions were reported after application of gentamicin. A contamination of the medicinal products with histamine, an impurity of the raw material fish peptone used upon the production, could be identified as the cause of the adverse effects. Batches of gentamicin sulfate, which had been stored at the University of Würzburg since the earlier investigations, were analyzed regarding their contamination with histamine to determine whether the biogenic amine was responsible for the 1990s fatalities as well. Furthermore, a correlation with the lead impurity sisomicin was checked. Histamine could be detected in all analyzed batches, but at a lower level than in the batches responsible for the anaphylactic reactions. Moreover, there is no correlation of histamine with the lead impurity sisomicin. Hence, the causative agent for the 1990s fatalities was not histamine and remains unknown.
Another source of impurities is the reaction of APIs with excipients, e.g. the esterification of naproxen with PEG 600 in soft gel capsules. The influence of the formulation’s composition on this reaction was investigated by means of LC-UV. Therefore, the impurity naproxen-PEG-ester (NPEG) was synthesized and used for the development of a method suitable for the analysis of soft gel capsule formulations. Different formulations were stressed for 7 d at 60 °C and the relative amount of NPEG was determined. The formation of NPEG was influenced by the concentrations of water and lactic acid, the pH, and the drug load of the formulation, which can easily be explained by the chemistry behind esterification reactions.
Keeping in mind the huge variety of sources of impurities, it might be impossible to predict all potential impurities of a drug substance/product. Targeted and untargeted approaches were combined in the impurity profiling of bisoprolol fumarate. Eight versions of an LC-HRMS method were developed to enable the detection of a maximum number of impurities: an acidic and a basic buffered LC was coupled to MS detection applying ESI and APCI, both in positive in negative mode. MS and MS/MS data were acquired simultaneously by information dependent acquisition. In the targeted approach, potential impurities were derived from a reaction matrix based on the synthesis route of the API, while the untargeted part was based on general unknown comparative screening to identify additional signals. 18 and 17 impurities were detected in the targeted and the untargeted approach, respectively. The molecular formulae were assessed based on the exact mass and the isotope pattern. Theoretical fragment spectra generated by in silico fragmentation were matched with experimental data to estimate the plausibility of proposed/elucidated structures. Moreover, the detected impurities were quantified with respect to an internal standard.
Ziel dieser Arbeit war es, unbekannte (Minor)-Komponenten aus Apfelsaft zu identifizieren und deren Beitrag zur chemopräventiven Wirkung des Saftes zu bestimmen. Deren Motivation war begründet, dass bisher identifizierte niedermolekulare phenolische Verbindungen als Mischung nicht das gleiche chemopräventive Potential wie ein polyphenolangereicherter Apfelsaftextrakt aufgewiesen hatten. Zunächst wurden sieben Apfelsäfte verschiedener Erntejahre sowie 14 polyphenolangereicherte Saftextrakte auf ihre stoffliche Zusammensetzung hin untersucht. Dabei zeigte sich, dass die Polyphenolzusammensetzung der Apfelsäfte und -extrakte sowohl zwischen verschiedenen Produktionsjahren als auch abhängig von der verwendeten Produktionstechnologie stark variierte. Insbesondere der durch enzymatische Tresterverflüssigung erhaltene Saftextrakt (AE03B) wies eine deutliche Anreicherung an Quercetin- und Phloretinglycosiden sowie eine Abreicherung an phenolischen Säuren auf. Die Klärung von trüben Säften hatte keinen signifikanten Einfluss auf die Verteilung niedermolekularer Flavonoide. Tendenziell waren die Extrakte der trüben Säfte leicht an Phloretinglycosiden abgereichert. Im Gegensatz dazu wurde eine leichte Anreicherung der phenolischen Säuren in den untersuchten klaren Säften im Vergleich zu den trüben festgestellt. Der Gehalt hochmolekularer Procyanidine variierte in den untersuchten Säften und Saftextrakten produktions- und sortenbedingt ebenfalls stark. An klaren und trüben Säften einer Charge wurde eine signifikante Abnahme sowohl des Gehaltes als auch des mittleren Polymerisationsgrades durch den Klärungsprozess nachgewiesen. Ferner zeigten Reinfruchtsäfte aus Mostapfelsorten höhere Gehalte und mittlere Polymerisationsgrade als verschnittene Apfelsäfte mit einem Anteil an Tafeläpfeln. Als nicht phenolische Bestandteile wurden in den Säften und Saftextrakten (R)-Oktan-1,3-diol, (R)-5-(Z)-Okten-1,3-diol, 3-Hydroxy-2-pyron und 3-Hydroxy-beta-damascon detektiert. Zwei ausgewählte Saftextrakte aus den Jahren 2004 und 2006 wurden unter Zuhilfenahme unterschiedlicher präparativer Trennmethoden fraktioniert. Im Verlauf der Auftrennung wurden durch präparative Isolierung Quercetin, Qercetin-3-O-glucosid, Quercetin-3-O-galactosid, 3-Hydroxyphloretin-2’-glucosid, 3-Hydroxyphloretin-2’-xyloglucosid, Phloretin-2’-xyloglucosid und Phloretin-2’,4’-diglucosid erhalten. Die erhaltenen Subfraktionen und Reinstoffe wurden in vitro auf ihre Hemmwirkung der Proteintyrosinkinase (PTK) des epidermalen Wachstumsfaktor-Rezeptor (EGFR) untersucht. Darüber hinaus wurden die antioxidativen (DPPH, ORAC, X/XO), die antiinflammatorischen und antihormonellen (COX-1, CYP19) Eigenschaften sowie die Fähigkeit zur Modulation des Fremdstoffmetabolismus (CYP1A, QR) überprüft. Die Procyanidine des Apfels erwiesen sich in Abhängigkeit ihres Polymerisationsgrades als starke Inhibitoren der PTK des EGFR. So zeigten ausschließlich hochpolymere Procyanidine enthaltende Fraktionen (NP.4 und NP.5) eine deutlich stärkere Hemmung als Fraktionen, die nur aus niedermolekularen phenolischen Verbindungen zusammengesetzt waren. Auf die Enzyme Cytochrom P450 1A und Aromatase (CYP19) hatten die Procyanidine ebenfalls einen entscheidenden Einfluss. Die antioxidativen Eigenschaften der Apfelsaftextrakte waren sowohl vom Gehalt polymerer Procyanidine als auch von dem niedermolekularer Polyphenole abhängig. Die für die Apfelsaftextrakte nachgewiesenen Radikalfängereigenschaften und die Hemmung der Superoxidanionbildung wurden für beide Gruppen ebenfalls bestätigt. Dahingegen wurden Peroxylradikale im ORAC-Test fast ausschließlich von niedermolekularen Verbindungen abgefangen. Die 3-Hydroxyphloretin(glycoside) zeigten, mit Ausnahme des Xyloglucosids, am EGFR eine stärkere inhibitorische Wirkung als die entsprechenden Phloretin(glycoside). Die Untersuchung der Enzyme des Fremdstoffmetabolismus zeigte ein uneinheitliches Bild. So war Phloretin ein effektiverer Hemmstoff der CYP1A im Vergleich zum 3-Hydroxyphloretin. Bei den Glycosiden zeigten 3-Hydroxyphloretin-2’-glucosid die etwas bessere Wirkung. Ähnlich verhielt es sich bei der antioxidativen Kapazität. Die 3-Hydroxyphloretin(glycoside) wiesen im DPPH und X/XO-Assay eine größere Aktivität auf. Hydroxylperoxidradikale wurden nur durch das dihydroxylierte freie Aglykon besser abfangen. Weiterhin wurde mit 3-Hydroxy-beta-damascon erstmals ein hochpotenter Induktor der Chinonreduktase in Apfelsaft identifiziert. Die in dieser Arbeit erlangten Erkenntnisse hinsichtlich der neu identifizierten bioaktiven Inhalsstoffe leisten einen Beitrag zum besseren Verständnis der potentiell chemopräventiven Wirkungen von (trüben) Apfelsäften. Ferner ist die Quantifizierung dieser Substanzen in technologisch unterschiedlich behandelten Säften für weiterführende Studien von Relevanz.
Die Permeabilität von Substanzen über Biomembranen erfolgt auf Basis ihrer Größe und Lipophilie, wird jedoch auch zu einem großen Anteil vom aktiven Transport bestimmt. Speziell im menschlichen Verdauungstrakt ist dieser Transportmechanismus neben seinen essentiellen physiologischen Aufgaben, wie den Transport von Nährstoffen, an einer Resistenz gegen exogene Stoffe und Xenobiotika beteiligt, der die Aufnahme in den Organismus über einen Rücktransport in das Darmlumen limitiert. Dabei hat die membranständige Effluxpumpe p-Glykoprotein (p-GP) als ein Baustein dieses Schutzmechanismus auch einen großen Einfluss auf die Arzneimitteltherapie. Über eine Modulierung der Pharmakokinetik von Arzneistoffen beschränkt sie die Aufnahme von Medikamenten und senkt dadurch deren Bioverfügbarkeit. Es wird auch für pflanzliche Inhaltsstoffe aus der Gruppe der Polyphenole ein möglicher Einfluss auf dieses Transportprotein diskutiert. Diese Beeinflussung kann sich entweder in einer Induktion oder einer Inhibition des Proteins äußern, was positive wie negative Effekte haben kann. Eine Hemmung des Transportproteins führt zu einer erhöhten Aufnahme einiger Arzneistoffe, die mit einer erhöhten Bioverfügbarkeit und einer potentiellen Dosissenkung einhergeht. Induziert man p-GP dagegen, so wird es beispielsweise ermöglicht, potentiell schädliche Xenobiotika noch intensiver auszuscheiden und nachteilige Plasmaspiegel zu verhindern. Im Rahmen der vorliegenden Arbeit sollte daher der Einfluss ausgewählter Polyphenole auf die Funktionalität und die Genexpression im CaCo-II-Zellkulturmodell näher untersucht, sowie vorab charakteristische Eigenschaften der pflanzlichen Inhaltsstoffe - Taxifolin, Silibinin, M1, Urolithin A, Urolithin B, Urolithin C, Isourolithin A, racemisches Hydnocarpin D, (+)-Hydnocarpin D, (-)-Hydnocarpin D - vergleichend bestimmt werden. Diese stoffspezifischen Charakteristika umfassten die Zytotoxizität, die Stabilität und die antioxidative Kapazität. Vor allem die Zytotoxizität und die Stabilität sind essentielle Parameter für aussagekräftige Resultate. Die Substanzen waren in der eingesetzten Konzentration von 50 µM mehrheitlich, mit Ausnahme des Hydnocarpins D, nicht-toxisch innerhalb der relevanten Versuchszeiträume, 4 h und 24 h, und den verwendeten Kulturmedien, DMEM-Pest und HBSS. Vor allem im Hinblick auf die Genexpressionsversuche war es die Basis für valide Ergebnisse, den Zeitraum bis 24 h als nicht-toxisch sicherstellen zu können. Hinsichtlich der Stabilität waren nur Taxifolin (27 % Restkonzentration) und der M1 (0 % Restkonzentration) nach 24 h in Zellkulturmedium kritisch. Auf Basis ihrer antioxidativen Kapazität werden pflanzlichen Inhaltsstoffen eine Reihe von gesundheitsförderlichen Merkmalen nachgesagt, weswegen dieser Aspekt für die Testsubstanzen zusätzlich vergleichend evaluiert wurde. Der Eintritt von Pathogenen kann Zusammenfassung 377 zum Beispiel durch oxidative Schädigung des Darmepithels erleichtert werden, was zusätzlich zu einem Effekt auf p-GP durch die Polyphenole unter Umständen positiv beeinflusst werden kann. Taxifolin, der M1 sowie die Urolithine A und C konnten so als antioxidativ aktive Stoffe erstmals vergleichend analysiert und die Resultate sinnvoll zu bestehenden Daten in Relation gesetzt werden. Sie konnten nach antioxidativer Potenz in der Reihenfolge Urolithin C > M1 > Taxifolin > Urolithin A geordnet werden. Zur Analyse des Einflusses der ausgewählten Polyphenole auf die Funktionalität von p-GP sollten Transportversuche über einen CaCo-II-Monolayer mit Rhodamin 123 als Markersubstanz durchgeführt werden. Diese Untersuchungen benötigen typischerweise eine vorbereitende Kulturzeit der Zellen von insgesamt drei Wochen, sodass sich eine Verkürzung dieser Zeitspanne aus Zeitersparnis- und Kostengründen positiv auf den Durchsatz der Versuche auswirken würde. In einem umfassenden Ansatz mit kombinierter Bestimmung der Qualifizierung der Zellschichten im Hinblick auf Qualität des Monolayers (TEER-Messung, Lucifer-Yellow-Transportrate, Fluoreszenzfärbung der Tight-junctions) sowie der Funktionalität und Expression von p-GP gelang der Nachweis, dass 14 Tage hinreichend und sinnvoll waren. Zentraler Bestandteil war in der vorliegenden Arbeit die Identifizierung der Effekte der Urolithine auf sowohl p-GP direkt, als auch auf die Genexpression dieses Transportproteins. Diese Polyphenole werden im menschlichen Verdauungstrakt über einen bakteriellen Metabolismus aus Ellagtanninen und Ellagsäure hergestellt und sind aufgrund ihrer vielfältigen gesundheitsförderlichen Charakteristiken in der Forschung von steigendem Interesse. Hierfür konnten nach unserem Kenntnisstand mit den gewählten Versuchsansätzen neue Erkenntnisse gewonnen werden. In den Transportversuchen mit Rhodamin 123 als Modellsubstrat von p-GP konnten die Urolithine den p-GP-vermittelten Transport positiv beeinflussen. Die Urolithine B (Papp-Ratio 1,98), C (Papp-Ratio 2,15) und das Isourolithin A (Papp-Ratio 1,63) steigerten den Rhodamintransport signifikant und lediglich für Urolithin A (Papp-Ratio 1,45) konnte keine Signifikanz belegt werden. Der Einfluss der Urolithine lag jeweils im Bereich des Modellinduktors Dexamethason. Ebenso konnte eine positive Modulierung der Genexpression nach 24 h detektiert werden. Die Hochregulierungen durch die Urolithine A (zwei- bis dreifach), B (1,4-fach) und C (1,8-fach) waren konsistent und statistisch signifikant. Urolithin A konnte hierbei als potentester Induktor charakterisiert werden, wohingegen sein Isomer Isourolithin A keinerlei signifikante Beeinflussung der Expression zeigte. In diesen Inkubationsversuchen wurde die Eigenschaft zur Erhöhung der Genexpression über den Einfluss auf den p-GP-vermittelten Rhodamintransport bestätigt. Die Urolithine A, B, C und Isourolithin A konnten nach einer Vorinkubation über 24 h und 48 h auch den Transport von Rhodamin 123 nochmals signifikanter zu den klassischen E Zusammenfassung 378 Transportversuchen ohne Vorinkubation steigern. Relevanz hierfür hatte der erste Zeitraum über 24 h, da hier ein deutlicher Anstieg der Rhodamintransportrate zu erkennen war. Nach 48 h stieg der Rhodamintransport nur noch geringfügig an oder ging sogar leicht zurück (Urolithin B). Hinsichtlich der Genexpression konnte nach 48 h nur noch Urolithin C p-GP signifikant hochregulieren, allerdings sind diese Erkenntnisse auf Basis der Zytotoxizität der Substanzen über diesen Zeitraum kritisch zu betrachten. In der Analyse des Effektes der weiteren Polyphenole auf die Genexpression von p-GP konnten für die meisten Stoffe nur zufällige Zusammenhänge hinsichtlich Hoch- und Herunterregulierung bestimmt werden. In den Transportversuchen konnte jedoch (+)-Hydnocarpin (Papp-Ratio 0,48) den Transport in gleichem Ausmaß wie der Modellinhibitor Verapamil (Papp-Ratio 0,48) hemmen. Durch Modifizierung des Versuchsmediums zur Annäherung an physiologischeren Bedingungen (Gallensäuren, pH 6) konnte für manche Substanzen ein deutlich verändertes Verhalten beobachtet werden. Die Rhodamintransportrate nahm unter Einfluss von Urolithin B, Isourolithin A und dem M1 signifikant nun ab und bei Urolithin C signifikant zu. Dies legt nahe, dass mit dem klassischen Transportversuchsmodell lediglich Tendenzen für die Substanzen bestimmt werden können. Weitere Untersuchungen näher an der Physiologie des Verdauungstraktes sind nötig, um ein genaueres Bild des Stoffeinflusses zu gewinnen. Die Frage nach zeitlichem Einsetzen beziehungsweise der Kontinuität des Effektes auf p� GP konnte mit den Urolithinen A, B und C sowie Dexamethason geklärt werden. Eine Substanzexposition von lediglich fünf Minuten war nicht ausreichend, um in den nachfolgenden zwei Stunden einen Effekt zu beobachten. Dies legt eine Reversibilität der zugrundeliegenden Mechanismen und eine notwendige dauerhafte Anwesenheit der Substanzen über die Versuchszeit nahe. Neben Rhodamin 123 wurden noch Transportversuche mit dem Fluorchinolonantibiotikum Ciprofloxacin als Modellsubstanz durchgeführt, da es aufgrund dessen Substratcharakters für p-GP von therapeutischer Relevanz sein kann, wenn das Transportverhalten durch Polyphenole beeinflusst wird. Im Gegensatz zu Rhodamin 123 wurde der Transport von Ciprofloxacin durch die vier Urolithine verringert, was für diese Metabolismusprodukte eine zusätzliche Wirkung auf weitere Transportproteine nahelegt, weil Ciprofloxacin unter anderem auch über BRCP transportiert wird. Mittels des bakteriellen Endotoxins LPS konnte eine Schädigung des CaCo-II-Monolayers erzeugt werden, welche sich über erniedrigte TEER-Werte und einen erhöhten Rhodamintransport nachweisen ließ. Eine Vorinkubation der vier Urolithine war nicht in der Lage, diese Schädigung abzumildern, jedoch nicht komplett zu verhindern. Die TEER- Zusammenfassung 379 Werte konnten zwar wieder etwas gesteigert werden, jedoch maskierte die starke Stimulation dieser Pflanzenstoffe auf p-GP und den damit verbundenen Transport von Rhodamin 123 mögliche positive Effekte auf diese oxidative Stresssituation. Zusammenfassend war es mit der vorliegenden Arbeit erstmals durch systematische vergleichende Untersuchung und Kombination von Charakterisierungsansätzen möglich, eine deutliche Beeinflussung der Genexpression und Funktionalität des p-Glykoproteins durch vor allem die Urolithine aufzuzeigen, was eine Relevanz sowohl des Mikrobioms als auch der Ernährung in der Arzneimitteltherapie nahelegt. Zudem gelang es den klassischen Transportassay durch Verkürzung um eine Woche zu verbessern.
Die muscarinischen Rezeptoren sind ein wichtiger Bestandteil des parasympathischen Nervensystems. Sie gehören zur großen Gruppe der G-Protein-gekoppelten Rezeptoren, die nach ihrer Verwandtschaft in drei große Klassen eingeteilt werden können. Die muscarinischen Rezeptoren gehören zur Klasse A, den rhodopsinähnlichen Rezeptoren. Durch die im Jahr 2000 vorgenommene Aufklärung der hochauflösenden Röntgenkristallstruktur des Rinderrhodpsins und die hohe Aminosäuresequenzähnlichkeit der G-Protein-gekoppelten Rezeptoren hat man eine sehr gute Modellvorstellung über den Aufbau der G-Protein-gekoppelten Rezeptoren. Die Rezeptoren bestehen aus sieben transmembranalen Helices, die von drei intrazellulären und drei extrazellulären Loops stabilisiert werden. Bis heute konnten fünf Rezeptorsubtypen gentechnisch klassifiziert werden, die sich durch ihre Gewebeverteilung und Funktion unterscheiden. Allen Subtypen ist eine hohe Sequenzhomologie im Bereich der orthosteren Bindungsstelle gemeinsam, so dass die Entwicklung von subtyp-spezifischen orthosteren Liganden sehr schwierig ist. Außer der orthosteren Bindungsstelle konnte noch eine weitere Bindungsstelle am muscarinischen Rezeptor identifiziert werden. Diese befindet sich weiter außerhalb im Rezeptor in einem Bereich, der über die fünf Rezeptorsubtypen nicht sehr stark konserviert ist, so dass die Entwicklung von subtyp-spezifischen Liganden möglich ist. An dieser zweiten Bindungsstelle binden allostere Modulatoren. Hierbei handelt es sich um Substanzen, die ohne den orthosteren Liganden keinen Effekt am Rezeptor auslösen, dafür aber die Gleichgewichtsbindung des orthosteren Liganden beeinflussen können. Der Einfluss auf die Gleichgewichtsbindung geschieht wechselseitig und kann positiv, neutral oder negativ kooperativ sein. Zusätzlich üben allostere Modulatoren einen Effekt auf die Dissoziation des orthosteren Liganden aus. Die meisten bisher gefunden allosteren Modulatoren erniedrigen die Dissoziationsgeschwindikeit des orthosteren Liganden vom Rezeptor. Die Summe dieser Eigenschaften machen die allosteren Modulatoren sehr interessant für die Arzneimitteltherapie. Das Ziel dieser Arbeit war die Synthese strukturell neuer allosterer Modulatoren des muscarinischen Rezeptors unter Anwendung des postulierten Pharmakophormodells. Als Ausgangspunkt sollten geländerhelicale Moleküle dienen, die strukturell abgewandelt dieses Pharmakophormodell sehr gut erfüllen. Die geländerhelicalen Moleküle ähneln in ihrem dreidimensionalen Aufbau dem Geländer einer Wendeltreppe. Sie sind durch die Brücken zwischen den aromatischen Bereichen sehr rigide Moleküle, so dass es nur wenige genau definierte Konformationen gibt. Grundsätzlich können drei Atropisomere unterschieden werden, wobei zwei zueinander enantiomer sind. Geplant war die Synthese eine Reihe von tertiären Aminen oder quartären Ammoniumsalzen. Die Synthese der Ausgangsverbindung konnte nach der Vorschrift von Kiupel erfolgen, war aber nur mit geringer Ausbeute möglich. Deshalb wurde dieser Syntheseweg nicht weiterverfolgt. Als Alternative bot sich an, auf die Brücken zwischen den aromatischen Ringen zu verzichten. Die so entstandenen Verbindungen sind weniger rigide und können sich deshalb gegebenenfalls besser an den Rezeptor anpassen. Grundsätzlich können je nach Substitutionsmuster zwei Synthesewege verfolgt werden. Beide Varianten erfüllen das postulierte Pharmakophormodell. Der Aufbau des Grundgerüstes erfolgt mittels einer nickelkatalysierten Grignard-Kupplung. Danach erfolgen eine Wohl-Ziegler-Seitenkettenbromierung und eine Verlängerung der Seitenkette im Sinne einer Alkylierung mittels Malonsäurediethylester und einer Hilfsbase. Anschließend erfolgen die Decarboxylierung und die Umsetzung zum Amid, das zum Amin reduziert werden kann. Betrachtet man die Lage der Pharmakophorelemente so variiert der Abstand der positiv geladenen Stickstoffe je nach Konformation zwischen 5 Å und 15 Å, so dass ein weiter Bereich abgedeckt werden kann. Der Abstand der aromatischen Bereiche bleibt relativ stabil. Die pharmakologische Testung der Verbindungen auf ihre allostere Potenz und Affinität zum muscarinischen Rezeptor erfolgte in der Arbeitsgruppe von Prof. Mohr in Bonn. Hierzu werden Membranhomogenate vom Herzventrikelgewebe des Hausschweines verwendet. Diese enthalten mit großer Prävalenz muscarinische M2-Rezeptoren. Es wurden Gleichgewichtsbindungs- und Dissoziationsexperimente durchgeführt. Bis jetzt sind noch nicht alle Verbindungen getestet worden. Die bisher getesteten Verbindungen weisen alle eine Affinität zum mit [3H]-N-Methylscopolamin besetzten muscarinischen M2-Rezeptor im mikro-molaren Bereich auf. Sie liegen damit im oberen Bereich der bisher synthetisierten allosteren Modulatoren. Das postulierte Pharmakophormodell konnte also mit Hilfe der synthetisierten Substanzen bestätigt werden.
Amber Light Control of Peptide Secondary Structure by a Perfluoroaromatic Azobenzene Photoswitch

The incorporation of photoswitches into the molecular structure of peptides and proteins enables their dynamic photocontrol in complex biological systems. Here, a perfluorinated azobenzene derivative triggered by amber light was site‐specifically conjugated to cysteines in a helical peptide by perfluoroarylation chemistry. In response to the photoisomerization (trans→cis) of the conjugated azobenzene with amber light, the secondary structure of the peptide was modulated from a disorganized into an amphiphilic helical structure.
Das Enzym alpha-Dioxygenase (alpha-DOX) aus Erbsen (Pisum sativum) wurde mit folgenden Zielsetzungen untersucht: Isolierung und Charakterisierung der für die P. sativum alpha-DOX codierenden cDNA, Überproduktion der P. sativum alpha-DOX in Escherichia coli und nachfolgende Isolierung, Untersuchung der Interaktion der P. sativum alpha-DOX mit Fettsäuresubstraten sowie systematische Studie der Expression der P. sativum alpha-DOX während der Keimung und Entwicklung von Erbsenpflanzen. alpha-Dioxygenasen katalysieren in Pflanzen den Initialschritt der alpha-Oxidation von langkettigen Fettsäuren, die über die intermediäre Bildung von (R)-2-Hydroperoxyfettsäuren führt. Folgeprodukte dieser Reaktion sind die entsprechende (R)-2-Hydroxysäure sowie der um ein C-Atom kettenverkürzte Aldehyd. Es wurde die für die alpha-Dioxygenase aus Erbsen codierende cDNA mit einer Gesamtlänge von 2132 bp isoliert, die ein offenes Leseraster von 1929 bp beinhaltet. Sie codiert für ein Protein mit 643 Aminosäuren und einem errechneten Molekulargewicht von ca. 73 kD. Die Pisum sativum alpha-Dioxygenase wurde in E. coli als Fusionsprotein mit einem 6 x His-tag überproduziert und mittels Metallaffinitätschromatographie an Ni-NTA-Agarose isoliert. Studien zur Interaktion der P. sativum alpha-Dioxygenase mit Fettsäuresubstraten umfassten sowohl Versuche zu Anforderungen auf Seiten des Substrats als auch zu potentiellen Interaktionspartnern auf Seiten des Enzym. Es wurde gezeigt, dass für die Reaktion von alpha-Dioxygenasen mit Fettsäuren die freie Carboxylgruppe des Substrats unerlässlich ist. Aufgrund eines Aminosäuresequenzvergleichs zwischen der alpha-Dioxygenase aus Erbsen und PGHS-1 aus O. aries wurden vier Aminosäuren als potentielle Interaktionspartner auf Seiten der alpha-Dioxygenase aus Erbsen ausgewählt. Es handelte sich um die Arginin-Reste Arg-87, Arg-391, Arg-569 und Arg-570. Mit Hilfe der ortsspezifischen Mutagenese wurde gezeigt, dass der Aminosäurerest Arg-570 für die katalytische Aktivität unerlässlich ist. Die Expression der P. sativum alpha-Dioxygenase in keimenden Erbsen und jungen Erbsenpflanzen wurde sowohl in ihrem zeitlichen Verlauf als auch hinsichtlich der Gewebespezifität betrachtet. Die Ergebnisse zeigten, dass Keimung zu einer deutlichen Akkumulation von alpha-Dioxygenase mRNA in Erbsen führte. Auch alpha-Dioxygenase Protein war in großer Menge in keimenden und jungen Erbsenpflanzen vorhanden. Ausgeprägte Gewebespezifität war festzustellen: alpha-DOX mRNA fand sich fast ausschließlich in Wurzeln von Erbsenpflanzen, in Sprossgewebe dagegen war sie kaum vorhanden. Im Gegensatz dazu lag alpha-DOX Protein gleichermaßen in Spross- und in Wurzelgewebe vor. Parallel zur Reifung der Pflanzen nahm die Menge an alpha-DOX mRNA und Protein ab. Alpha-Dioxygenase-Aktivität war bereits in trockenen Samen detektierbar, während der Keimung nahm sie deutlich zu. Im Vergleich von Spross- und Wurzelgewebe war die Aktivität in Wurzeln höher, bezogen sowohl auf das Frischgewicht der Pflanzen als auch auf die Menge an Gesamtprotein (spezifische Aktivität). Die Untersuchungen an Wurzeln zeigten, dass die Aktivität bezogen auf das Frischgewicht der Pflanzen über den betrachteten Zeitraum kaum variierte, während die spezifische Aktivität mit zunehmendem Alter der Pflanzen kontinuierlich zunahm. Dieses Ergebnis deutet darauf hin, dass in Erbsen mehrere alpha-Dioxygenase-Isoenzyme vorhanden sind, so wie man dies für andere höhere Pflanzen bereits postuliert hat. Ein zellprotektiver Effekt von alpha-Dioxygenasen auf Pflanzen während der Interaktion mit Pathogenen ist bekannt. Möglicherweise ist dies auch der Grund für eine verstärkte Expression während der Keimung von Pflanzen. Die bevorzugte Expression in Wurzeln könnte auf eine Funktion als permanentes Schutzsystem gegen Infektion hindeuten.
Each year millions of plastic and reconstructive procedures are performed to regenerate soft tissue defects after, for example, traumata, deep burns or tumor resections. Tissue engineered adipose tissue grafts are a promising alternative to autologous fat transfer or synthetic implants to meet this demand for adipose tissue. Strategies of tissue engineering, especially the use of cell carriers, provide an environment for better cell survival, an easier positioning and supplemented with the appropriate conditions a faster vascularization in vivo. To successfully engineer an adipose tissue substitute for clinical use, it is crucial to know the actual intended application. In some areas, like the upper and lower extremities, only a thin subcutaneous fat layer is needed and in others, large volumes of vascularized fat grafts are more desirable. The use and interplay of stem cells and selected scaffolds were investigated and provide now a basis for the generation of fitted and suitable substitutes in two different application areas.
Complex injuries of the upper and lower extremities, in many cases, lead to excessive scarring. Due to severe damage to the subcutaneous fat layer, a common sequela is adhesion formation to mobile structures like tendons, nerves, and blood vessels resulting in restricted motion and disabling pain [Moor 1996, McHugh 1997]. In order to generate a subcutaneous fat layer to cushion scarred tissue after substantial burns or injuries, different collagen matrices were tested for clinical handling and the ability to support adipogenesis. When testing five different collagen matrices, PermacolTM and StratticeTM showed promising characteristics; additionally both possess the clinical approval. Under culture conditions, only PermacolTM, a cross-linked collagen matrix, exhibited an excellent long-term stability. Ranking nearly on the same level was StratticeTM, a non-cross-linked dermal scaffold; it only exhibited a slight shrinkage. All other scaffolds tested were severely compromised in stability under culture conditions. Engineering a subcutaneous fat layer, a construct would be desirable with a thin layer of emerging fat for cushioning on one side, and a non-seeded other side for cell migration and host integration. With PermacolTM and StratticeTM, it was possible to produce constructs with ASC (adipose derived stem cells) seeded on one side, which could be adipogenically differentiated. Additionally, the thickness of the cell layer could be varied. Thereby, it becomes possible to adjust the thickness of the construct to the surrounding tissue. In order to reduce the pre-implantation time ex vivo and the costs, the culture time was varied by testing different induction protocols. An adipogenic induction period of only four days was demonstrated to be sufficient to obtain a substantial adipogenic differentiation of the applied ASC. Thus, seeded with ASC, PermacolTM and StratticeTM are suitable scaffolds to engineer subcutaneous fat layers for reconstruction of the upper and lower extremities, as they support adipogenesis and are appropriately thin, and therefore would not compromise the cosmesis.
For the engineering of large-volume adipose tissue, adequate vascularization still represents a major challenge. With the objective to engineer vascularized fat pads, it is important to consider the slow kinetics of revascularization in vivo. Therefore, a decellularized porcine jejunum with pre-existing vascular structures and pedicles to connect to the host vasculature or the circulation of a bioreactor system was used. In a first step, the ability of a small decellularized jejunal section was tested for cell adhesion and for supporting adipogenic differentiation of hASC mono-cultures. Cell adhesion and adipogenic maturation of ASC seeded on the jejunal material was verified through histological and molecular analysis. After the successful mono-culture, the goal was to establish a MVEC (microvascular endothelial cells) and ASC co-culture; suitable culture conditions had to be found, which support the viability of both cell types and do not interfere with the adipogenic differentiation. After the elimination of EGF (epidermal growth factor) from the co-culture medium, substantial adipogenic maturation was observed. In the next step, a large jejunal segment (length 8 cm), with its pre-existing vascular structures and arterial/venous pedicles, was connected to the supply system of a custom-made bioreactor. After successful reseeding the vascular structure with endothelial cells, the lumen was seeded with ASC which were then adipogenically induced. Histological and molecular examinations confirmed adipogenic maturation and the existence of seeded vessels within the engineered construct. Noteworthily, a co-localization of adipogenically differentiating ASC and endothelial cells in vascular networks could be observed. So, for the first time a vascularized fat construct was developed in vitro, based on the use of a decellularized porcine jejunum. As this engineered construct can be connected to a supply system or even to a patient vasculature, it is versatile in use, for example, as transplant in plastic and reconstruction surgery, as model in basic research or as an in vitro drug testing system.
To summarize, in this work a promising substitute for subcutaneous fat layer reconstruction, in the upper and lower extremities, was developed, and the first, as far as reported, in vitro generated adipose tissue construct with integrated vascular networks was successfully engineered.
Insulin-like growth factor-I (IGF-I) is a 70-amino acid polypeptide with a molecular weight of approximately 7.6 kDa acting as an anabolic effector. It is essential for tissue growth and remodeling. Clinically, it is used for the treatment of growth disorders and has been proposed for various other applications including musculoskeletal diseases. Unlike insulin, IGF-I is complexed to at least six high-affinity binding proteins (IGFBPs) exerting homeostatic effects by modulating IGF-I availability to its receptor (IGF-IR) on most cells in the body as well as changing the distribution of the growth factor within the organism.1-3 Short half-lived IGF-I have been the driving forces for the design of localized IGF-I depot systems or protein modification with enhanced pharmacokinetic properties. In this thesis, we endeavor to present a versatile biologic into which galenical properties were engineered through chemical synthesis, e.g., by site-specific coupling of biomaterials or complex composites to IGF-I. For that, we redesigned the therapeutic via genetic codon expansion resulting in an alkyne introduced IGF-I, thereby becoming a substrate for biorthogonal click chemistries yielding a site-specific decoration.
In this approach, an orthogonal pyrrolysine tRNA synthetase (PylRS)/tRNAPyl CUA pair was employed to direct the co-translational incorporation of an unnatural amino acid—¬propargyl-L-lysine (plk)—bearing a clickable alkyne functional handle into IGF-I in response to the amber stop codon (UAG) introduced into the defined position in the gene of interest. We summarized the systematic optimization of upstream and downstream process alike with the ultimate goal to increase the yield of plk modified IGF-I therapeutic, from the construction of gene fusions resulting in (i) Trx-plk-IGF-I fusion variants, (ii) naturally occurring pro-IGF-I protein (IGF-I + Ea peptide) (plk-IGF-I Ea), over the subsequent bacterial cultivation and protein extraction to the final chromatographic purification. The opportunities and hurdles of all of the above strategies were discussed. Evidence was provided that the wild-type IGF-I yields were pure by exploiting the advantages of the pHisTrx expression vector system in concert with a thrombin enzyme with its highly specific proteolytic digestion site and multiple-chromatography steps. The alkyne functionality was successfully introduced into IGF-I by amber codon suppression. The proper folding of plk-IGF-I Ea was assessed by WST-1 proliferation assay and the detection of phosphorylated AKT in MG-63 cell lysate. The purity of plk-IGF-I Ea was monitored with RP-HPLC and SDS-PAGE analysis. This work also showed site-specific coupling an alkyne in plk-IGF-I Ea by copper (I)-catalyzed azide-alkyne cycloaddition (CuAAC) with potent activities in vitro. The site-specific immobilization of plk-IGF-I Ea to the model carrier (i.e., agarose beads) resulted in enhanced cell proliferation and adhesion surrounding the IGF-I-presenting particles. Cell proliferation and differentiation were enhanced in the accessibility of IGF-I decorated beads, reflecting the multivalence on cellular performance.
Next, we aimed at effectively showing the disease environment by co-delivery of fibroblast growth factor 2 (FGF2) and IGF-I, deploying localized matrix metalloproteinases (MMPs) upregulation as a surrogate marker driving the response of the drug delivery system. For this purpose, we genetically engineered FGF2 variant containing an (S)-2-amino-6-(((2-azidoethoxy)carbonyl)amino)hexanoic acid incorporated at its N-terminus, followed by an MMPs-cleavable linker (PCL) and FGF2 sequence, thereby allowing site-directed, specific decoration of the resultant azide-PCL-FGF2 with the previously mentioned plk-IGF-I Ea to generate defined protein-protein conjugates with a PCL in between. The click reaction between plk-IGF-I Ea and azide-PCL-FGF2 was systematically optimized to increase the yield of IGF-FGF conjugates, including reaction temperature, incubation duration, the addition of anionic detergent, and different ratios of the participating biopharmaceutics. The challenge here was that CuAAC reaction components or conditions might oxidize free cysteines of azide-PCL-FGF2 and future work needs to present the extent of activity retention after conjugation. Furthermore, our study provides potential options for dual-labeling of IGF-I either by the introduction of unnatural amino acids within two distinct positions of the protein of interest for parallel “double-click” labeling of the resultant plk-IGF-I Ea-plk or by using a combination of enzymatic-catalyzed and CuAAC bioorthogonal coupling strategies for sequentially dual-labeling of plk-IGF-I Ea.
In conclusion, genetic code expansion in combination with click-chemistry provides the fundament for novel IGF-I analogs allowing unprecedented site specificity for decoration. Considerable progress towards IGF-I based therapies with enhanced pharmacological properties was made by demonstrating the feasibility of the expression of plk incorporated IGF-I using E. coli and retained activity of unconjugated and conjugated IGF-I variant. Dual-labeling of IGF-I provides further insights into the functional requirements of IGF-I. Still, further investigation warrants to develop precise IGF-I therapy through unmatched temporal and spatial regulation of the pleiotropic IGF-I.
The International Symposium on Phytochemicals in Medicine and Food (ISPMF2015), organized by the Phytochemical Society of Europe (PSE) and the Phytochemical Society of Asia (PSA), was held June 26-29, 2015, in Shanghai of China. This was the first time that a PSE meeting has been held in Asia and a PSE-PSA joint symposium provided an opportunity for communication between scientists from Europe and Asia and other continents. ISPMF2015 has been jointly sponsored by Fujian Agriculture and Forestry University, Guizhou Medical University, Shanghai Normal University, Yancheng Institute of Technology, Beijing Normal University, and Fudan University. More than 270 scientists from 48 countries attended this meeting and presented their research and opinions on phytochemistry, phytomedicine and phytoneering. The international organizing committee and scientific advisory board of ISPMF 2015 comprised of outstanding scientists from around the globe. Dr. Jianbo Xiao was the chairman of the International Organizing Committee of ISPMF2015 and moderated the open address on June 26.
The organizing committee of ISPMF2015 assembled an exciting and diverse program, featuring 16 sessions including 12 plenary lectures, 20 invited talks, 55 short oral presentations, and more than 130 posters, which were dedicated to creating a podium for exchanging the latest research results in the phytochemicals for food and human health.
Ruxolitinib (RUX) is approved for the treatment of steroid-refractory acute and chronic graft versus host disease (GvHD). It is predominantly metabolized via cytochrome P450 (CYP) 3A4. As patients with GvHD have an increased risk of invasive fungal infections, RUX is frequently combined with posaconazole (POS), a strong CYP3A4 inhibitor. Knowledge of RUX exposure under concomitant POS treatment is scarce and recommendations on dose modifications are inconsistent. A physiologically based pharmacokinetic (PBPK) model was developed to investigate the drug–drug interaction (DDI) between POS and RUX. The predicted RUX exposure was compared to observed concentrations in patients with GvHD in the clinical routine. PBPK models for RUX and POS were independently set up using PK-Sim\(^®\) Version 11. Plasma concentration-time profiles were described successfully and all predicted area under the curve (AUC) values were within 2-fold of the observed values. The increase in RUX exposure was predicted with a DDI ratio of 1.21 (C\(_{max}\)) and 1.59 (AUC). Standard dosing in patients with GvHD led to higher RUX exposure than expected, suggesting further dose reduction if combined with POS. The developed model can serve as a starting point for further simulations of the implemented DDI and can be extended to further perpetrators of CYP-mediated PK-DDIs or disease-specific physiological changes.
Priority tasks of the present thesis were to generate various enantiopure C-3-substituted pyroglutamates as well as C-3-substituted glutamates, and furthermore to ameliorate the serious drawback of the bad atom-economy in the reaction sequence of previously published silylether-mediated procedures. To meet these requirements, the ortho ester functionality (OBO ester) developed by Corey was introduced. According to the plan of synthesis, the starting material, non-racemic (S)-pyroglutamic acid, was converted to the corresponding oxetane ester via a DCC-mediated esterification. The latter was N-protected to provide N-acceptor substituted pyroglutamic acid oxetane esters (Acceptor=Boc,Cbz,CO2Me). After rearrangement with boron trifluoride, the ortho ester derivatives (Acceptor=Cbz,CO2Me) were at hand and exclusively the N-Cbz derivative was converted to the corresponding alpha,beta-unsaturated lactam via a syn-elimination reaction. The formation of the C-3-substituted ortho ester compounds (R=methyl,ethyl,butyl,allyl,phenyl,4-chlorophenyl,biphenyl,naphthyl) was performed via a copper-mediated conjugate addition to the alpha,beta-enone system of the N-Cbz-alpha,beta-unsaturated lactam. The OBO functionality hence was envisaged to support perfect trans selectivity in this cuprate addition to the Michael system of the N-Cbz-alpha,beta-unsaturated lactam. Spectroscopic NMR-data, on the basis of 1H-, 13C- and DEPT spectra, proved the assumption that the C-3-substituted ortho ester derivatives exclusively are trans-configurated, i.e. the alkyl derivatives (R=methyl,ethyl,butyl,allyl) are (2S,3S)-configurated and the aryl derivatives (R=phenyl,4-chlorophenyl,biphenyl,naphthyl) are (2S,3R)-configurated). The C-3-substituted ortho ester derivatives were completely deprotected to yield the C-3-substituted pyroglutamates (R=ethyl,phenyl,4-chlorophenyl,naphthyl). Finally, ring opening reaction via route A-2 lead to the desired enantiopure C-3-substituted glutamates. Alternatively, latter preferably were reacted via route A-1 to yield the C-3-substituted glutamates (R=methyl,ethyl,butyl,phenyl,4-chlorophenyl,naphthyl). Their (2S,3R)-configuration (R=aryl) and (2S,3S)-configuration (R=alky), respectively, unambiguously was proved on the basis of available spectroscopic NMR-data. To ensure this assumption, diastereomeric (2S,3R)-3-methyl glutamic acid (i.e. cis-configurated) examplarily was synthesized via route A-3 and spectroscopic NMR-data was compared to that of (2S,3S)-3-methyl glutamic acid (i.e. trans-configurated). Conclusively, there can be recorded the fact that the serious drawback of the bad atom-economy in the reaction sequence previously used can be circumvented by the introduction of the OBO functionality, so the concept of an improved atom-economy is achieved. Additionally, in comparison to the silyl-ether-mediated synthesis, the OBO functionality provided crystalline ortho ester derivatives, which facilitated their purification as well as characterization.
A closer look at long-established drugs: enantioselective protein binding and stability studies

The aim of this work was to investigate older, established drugs. The extent of the protein binding of chiral ephedra alkaloids to AGP and of ketamine to albumin was determined. Since enantiomers of these drugs are individual available, the focus was on possible enantioselective binding and structural moieties involved in the binding.
Previously published work suggested that ephedrine and pseudoephedrine can bind stereoselectively to proteins other than albumin in serum. For the determination of the extent of protein binding, the established ultrafiltration with subsequent chiral CE analysis was used. To determine the influence of basicity on binding, the drugs methylephedrine and norephedrine were also analyzed. Drug binding to AGP increased with increasing basicity as follows: norephedrine < methylephedrine < ephedrine < pseudoephedrine. pKaff was determined both graphically using the Klotz plot and mathematical indicating a low affinity of the ephedra alkaloids to AGP. Using STD-NMR spectroscopy experiments the aromatic protons and the C-CH3 side chain were shown to be most strongly involved in binding, which could be confirmed by molecular docking experiments in more detail. For all drugs, van der Waals-, π π , cationic interactions, hydrogen bonds, and a formation of a salt bridge were observed. The individual enantiomers showed no significant differences and thus the binding of ephedra alkaloids to AGP is not significant.
In contrast to the ephedra alkaloids, the possible enantioselective binding to albumin was investigated for R and S ketamine. Again, ultrafiltration followed by CE analysis was performed. The binding of ketamine to one main binding site could be identified. A non-linear fit was used for the determination of pKaff. Using the NMR methods STD-NMR, waterLOGSY-NMR, and CPMG-NMRspectroscopy: the aromatic protons as well as the protons of the NCH3 methyl group showed the largest signal intensity changes, while the cyclohexanone protons showed the smallest changes. pKaff was also determined by the change in the chemical shift at different drug-protein ratios. These obtained values confirm the values obtained from ultrafiltration. Based on this, ketamine is classified as a low-affinity ligand to albumin. There were no significant differences between the individual enantiomers and thus the binding of ketamine to albumin is not a stereoselective process.
Using statistical design of experiments an efficient chiral CE method for determining the extent of protein binding of R and S ketamine to albumin was developed and validated according to ICH Q2 (R1) guideline.
The stability of ketamine was also investigated because a yellowish discoloration of an aqueous solution of ketamine developed under heat. XRPD investigations showed the same crystal structure for all batches examined. An untargeted screening using LC HRMS as well as LC UV measurements showed no degradation of ketamine or the presence of impurities in stress and non-stressed ketamine solutions, confirming the stability of ketamine under the stress conditions investigated. The lower the quality of the water used in the stress tests, the more intense the yellow discoloration occurred. The impurity or the mechanism that causes the yellow discoloration could not be identified.
Alzheimer's disease (AD) is a multifactorial disease and the most common form of dementia. There are no treatments to cure, prevent or slow down the progression of the disease. Natural products hold considerable interest for the development of preventive neuroprotectants to treat neurodegenerative disorders like AD, due to their low toxicity and general beneficial effects on human health with their anti-inflammatory and antioxidant features. In this work we describe regioselective synthesis of 7-O-ester hybrids of the flavonoid taxifolin with the phenolic acids cinnamic and ferulic acid, namely 7-O-cinnamoyltaxifolin and 7-O-feruloyltaxifolin. The compounds show pronounced overadditive neuroprotective effects against oxytosis, ferroptosis and ATP depletion in the murine hippocampal neuron HT22 cell model. Furthermore, 7-O-cinnamoyltaxifolin and 7-O-feruloyltaxifolin reduced LPS-induced neuroinflammation in BV-2 microglia cells as assessed by effects on the levels of NO, IL6 and TNFα. In all in vitro assays the 7-O-esters of taxifolin and ferulic or cinnamic acid showed strong overadditive activity, significantly exceeding the effects of the individual components and the equimolar mixtures thereof, which were almost inactive in all of the assays at the tested concentrations. In vivo studies confirmed this overadditive effect. Treatment of an AD mouse model based on the injection of oligomerized Aβ\(_{25-35}\) peptide into the brain to cause neurotoxicity and subsequently memory deficits with 7-O-cinnamoyltaxifolin or 7-O-feruloyltaxifolin resulted in improved performance in an assay for short-term memory as compared to vehicle and mice treated with the respective equimolar mixtures. These results highlight the benefits of natural product hybrids as a novel compound class with potential use for drug discovery in neurodegenerative diseases due to their pharmacological profile that is distinct from the individual natural components.
Die vorliegende Arbeit beschäftigte sich mit der Entwicklung und Synthese von Inhibitoren der Deoxyhypusin-Hydroxylase (DOHH), die einen wichtigen Schritt in der Aktivierung des eukaryotischen Translationsinitiations-Faktors-5A (eIF-5A) katalysiert. Die Hemmung dieses Metalloenzyms durch kleine Moleküle, die mit dem katalytischen Eisenatom im aktiven Zentrum der DOHH einen Chelatkomplex bilden, hat einen antiproliferativen Effekt auf parasitäre Erreger, wie Plasmodien, Trypanosomen und Leishmanien zur Folge. Ausgehend von den antiplasmodial wirksamen Eisenkomplexbildnern und Pyridon-Derivaten Ciclopirox und Mimosin wurden besser wirksame 2,6-Diaryl-4-oxopiperidincarbonsäuremono- und -diester-Derivate abgeleitet, deren 4-Piperidon-Grundgerüst als Leitstruktur für die Entwicklung von antiplasmodialen und antitrypanosomalen Wirkstoffen fungierte. Entsprechend dieser Leitstrukturen gelang im Zuge dieser Arbeit durch verschiedene Modifikationen der Doppel-Mannich-Reaktion die Erstellung einer weitreichenden Bibliothek 52 strukturell diverser 4-Hydroxytetrahydropyridin-3,5-dicarbonsäurediester 1 – 6, darunter auch erstmals Derivate mit t-Butyl-esterfunktionen und 4-Hydroxytetrahydropyridin-3-carbonsäuremonoester 7 – 8. Dabei konnten vor allem Derivate mit der gewünschten nitroaromatischen Substitution in den Positionen 2 und 6 synthetisiert werden. Darüber hinaus wurden vielfältige Strukturabwandlungen dieser Substanzen in Form von verschiedenen 4-Piperidonderivaten ohne Esterfunktionen, deren Oximen sowie von 4-Hydroxychinoloncarbonsäureestern syn-thetisiert. Die hergestellten Derivate wurden In-vitro-Testungen an Plasmodium falciparum, Trypanosoma brucei brucei und Leishmania major unterzogen. Zusätzlich wurde die Zytotoxizität an der Makrophagen-Zelllinie J774.1 ermittelt.
Im Mittelpunkt dieser Arbeit stand die Substanzgruppe der 4-Chinolone, die zum einen über ein intrinsisches antiparasitäres Potenzial gegen Erreger wie Plasmodien, Trypanosomen oder Mykobakterien verfügt und zum anderen über gezielte Substitution auch die Möglichkeit zu strukturellen Modifikationen bietet. Vorrangiges Ziel dieser Arbeit war der Aufbau einer strukturell möglichst diversen Substanzbibliothek und deren sukzessive Testung innerhalb des SFB630. Auf diese Weise sollten neue antiparasitäre Leitstrukturen als Ausgangspunkt für weitere strukturelle Optimierungen erhalten werden. Der Chinolon-Grundkörper sollte hierzu gemäß Gould-Jacobs-Reaktion aufgebaut werden. Zur Synthese diverser Amid-Derivate wurden verschiedene Synthese-strategien verfolgt. Alternativ wurden, ebenfalls über eine nukleophile Substitution (Piperidin-Derivat), in 7-Position modifizierte Verbindungen generiert, die unter Verwendung des Kupplungs-reagenzes PyBOB (Benzotriazol-1-yloxytri-pyrrolidinophosphonium Hexafluorphosphat) in die entsprechenden 1-Alkyl-1,4-dihydro-7-piperidinyl-4-oxo-chinolin-3-carboxamide transformiert wurden. Die in dieser Arbeit generierte Substanzbibliothek wurde anschließend innerhalb des SFB630 getestet. Hierbei zeigte sich, dass die Amidierung der 3-Carbonsäurefunktion eine Steigerung der antimikrobiellen Wirkung gegen Trypanosoma brucei mit sich brachte. Es kristallisierten sich aktive Verbindungen heraus, die erstmals eine Aktivität derartiger Derivate gegen Trypanosomen belegen und so zukünftig als Leitstrukturen für weitere strukturelle Modifizierungen herangezogen werden können. Mit dem in dieser Arbeit angewandten Random-Chemistry-Verfahren sollte in die Suche nach neuen Leitstrukturen gezielt das Zufallsprinzip integriert werden bzw. es sollten neue aktive Verbindungen generiert werden, die über die klassischen kombinatorischen Syntheseschemata bzw. die gängigen Reaktionsmechanismen nur schwer zugänglich sind. Eine Reihe von Fluorchinolon-Derivaten wurden in verschiedenen Lösungsmitteln, meist DMSO mit Zusätzen von Methanol oder Chloroform, gelöst bzw. suspendiert und anschließend einer ionisierenden γ-Strahlung von 500 kGy ausgesetzt. Die Testung mittels HPLC / FCPC generierter Fraktionen ergab zum Teil höhere antitrypanosomale Aktivitäten als die der korrespondie¬renden Ausgangsverbindungen. Eine Aktivität gegen Makrophagen konnte nicht festgestellt werden. Darüber hinaus wurde im Rahmen dieser Arbeit in Kooperation mit Prof. Schneider-Schaulies an der Identifizierung viraler Fusionsinhibitoren ausgewählter Paramyxoviren (Masern-Virus, Nipah-Virus) gearbeitet. Aus einer Ähnlichkeitssuche, basierend auf dem literaturbekannten Masern-Fusionsinhibitor 2-(4-Chlorphenyl)-N-(2-hydroxy-4-nitrophenyl)acetamid (AM-2), konnte die Struktur eines Chinolinamides identifiziert werden, woraufhin die generierte Substanzbibliothek auf antiviral-aktive Verbindungen gescreent werden sollte. Die Kristallstruktur des Nipah-Virus-Fusionsproteins wurde im Jahre 2006 aufgeklärt. Mit diesen Informationen konnte mittels Molecular-Modelling eine Bindetasche innerhalb der HR1-Domäne des F-Proteins identifiziert werden, mit der die erzielten inhibitorischen Aktivitäten gut in Einklang gebracht werden konnten. Diese Bindetasche befindet sich in einem Bereich weitreichender Umstrukturierungsvorgängen: Durch die Einlagerung des Liganden 7-(4-Carbamoyl-piperidin-1-yl)-N-(2,4-dichlorbenzyl)-1-cyclopropyl-6-fluor-4-oxo-1,4-dihydro-chinolin-3-carboxamid, in diese hydrophobe Tasche werden Wechselwirkungen mit den korrespondierenden Aminosäuren in der HR2-Domäne und so auch dessen Anlagerung unterbunden. In 1 μmolarer Konzentration konnte die Fusionsaktivität um 42% reduziert werden, die verwendeten Referenzsubstanz (OX-1) erzielte in selbiger Konzentration keine Wirkung.
Die humane afrikanische Trypanosomiasis (Schlafkrankheit, HAT) wird durch die Parasiten Trypanosoma brucei rhodesiense und Trypanosoma brucei gambiense ausgelöst und führt unbehandelt zum Tod. Wegen begrenzter Therapiemöglichkeiten sowie vernachlässigter Kontrollprogramme ist HAT eine gegenwärtige Bedrohung, was die Suche nach neuen Wirkstoffen notwendig macht. Ausgangspunkt für die Leitstrukturfindung war das 7-Amino-4-chinolon-3-carboxamid IV mit einem IC50-Wert (T. b. brucei) von 1.2 µM. Die 4-Chinolon-3-carboxamid-Grundstrukturen wurden unter Verwendung der Gould-Jacobs- (1-Alkyl-Derivate) bzw. der Grohe-Heitzer-Synthese (1-Aryl-Derivate) aufgebaut und anhand strukturierter Variation der Substituenten in Pos. 1, 3 und 7 die für die antitrypanosomale Wirksamkeit essenziellen Strukturelemente identifiziert: Pos.1: Die Alkylkettenverlängerung von Ethyl zu n-Butyl bewirkte eine stetige Aktivitätssteigerung, welche auch einem Aryl-Rest in dieser Position überlegen war. Pos.3: Benzylamide mit HBD-Funktionen führten zur Aktivitätsabnahme, während HBA-Funktionen und unsubstituierte Reste zur Steigerung der Wirksamkeit, teilweise in den nanomolaren Konzentrationsbereich, beitrugen. Pos.7: Neben cyclischen sek. Aminen wurden auch aliphatische prim. Amine via konventioneller oder Mikrowellen-unterstützter SNAr eingeführt. Dabei zeigten sich die sek. Amine mit einer antitrypanosomalen Aktivität im teilweise submikromolaren Bereich den acyclischen Aminen deutlich überlegen. Vor allem der Morpholin-Rest bewirkte eine sprunghafte Wirksamkeitsverbesserung. Durch Kombination der Einzelresultate konnte schließlich die den Lipinski’s „Rule of 5“ entsprechende Leitstruktur 33 mit vielversprechender antitrypanosomaler Wirksamkeit (IC50 (T. b. brucei) = 47 nM, IC50 (T. b. rhodesiense) = 9 nM) und geringer Zytotoxizität erhalten werden (SI = 19000). Erste Untersuchungen zur Identifikation des Targets der 4-Chinolon-3-carboxamide ergaben folgende Erkenntnisse: Fluoreszenzmikroskopieuntersuchungen zeigten eine deutliche Veränderung der Morphologie des Mitochondriums bei behandelten BSF-T. b. brucei-Zellen. Anhand einer Zellzyklus-Analyse wurde die Beeinträchtigung der Segregation des Kinetoplasten beobachtet, was zu einem Segregationsdefekt führte. Die Topoisomerase (TbTopoIImt) wurde durch ein „Knockdown“-Experiment als Haupt-Target ausgeschlossen. Trotz der bemerkenswerten biologischen Aktivität war eine In-vivo-Untersuchung der Leitstruktur wegen zu geringer Wasserlöslichkeit nicht möglich, welche auf eine hochgeordnete Schichtgitterstruktur zurückzuführen war. Da die Löslichkeit im Wesentlichen eine Funktion der Lipophilie und der intermolekularen Wechselwirkungen ist, wurden zur Verbesserung der Wasserlöslichkeit pharmazeutisch-technische Methoden angewandt sowie chemische Strukturmodifikationen vorgenommen: Es wurde eine Lipid-basierte, selbstemulgierende Formulierung entwickelt. Durch Ausbildung stabiler Emulsionen war 33 bis zu einer Konzentration von 10 mg/ml im Wässrigen löslich und somit für die In-vivo-Untersuchung zugänglich. Nach 4-tägiger peroraler Behandlung von NMRI-Mäusen mit einer wässrigen 1:1-Verdünnung der Formulierung konnte keine In-vivo-Aktivität festgestellt werden. Die Sprühtrocknung von 33 resultierte in amorpher Modifikation, welche in Gegenwart von PVP bzw. Eudragit®L100 stabilisiert wurde. Beide Partikel ermöglichten die Übersättigung von 33 im Wässrigen, was im Fall der Eudragit®L100-Partikel zu 200-facher Löslichkeitssteigerung gegenüber der kristallinen Wirkstoffmodifikation führte und somit die In-vivo-Untersuchung ermöglichte. Zusammen mit ersten Metabolismus-Untersuchungen von 33, welche die Berechnung einer Abbau-Kinetik bzw. Clearance ermöglichte, konnte mittels der Software Simcyp® ein Plasmakonzentrationsprofil der Verbindung 33 (Eudragit®L100-Partikel) erstellt werden. Basierend auf diesem Studiendesign wurde die In-vivo-Untersuchung von 33 an mit T. b. rhodesiense infizierten Mäusen durchgeführt und zeigte nach 8-tägiger Behandlung einen deutlichen Rückgang der Parasitämie. Im Fokus der chemischen Strukturmodifikation stand das Einführen polarer und ionisierbarer Strukturelemente, um 4-Chinolon-3-carboxamid-Derivate mit erhöhter Hydrophilie (logP 1 - 3) bzw. Salz- und Co-Kristall-Strukturen zu erhalten. Sämtliche Strukturvariationen trugen zur Verbesserung der Wasserlöslichkeit und der „drug-like“ Eigenschaften im Vergleich zu Verbindung 33 bei, waren allerdings von Aktivitätsverlusten gegenüber T. b. brucei begleitet. Anhand der „ligand efficiency“- und „lipophilic ligand efficiency“-Analyse wurden schließlich die vielversprechendsten Derivate (94 und 96) für die weitere Untersuchung ausgewählt. Mit IC50 (T. b. rhodesiense)-Werten von 4 nM (94) und 33 nM (96) und geringer Zytotoxizität wurden Selektivitätsindizes bis zu 25000 gefunden, welche jene von 33 übertrafen. Aufgrund einer Löslichkeit im millimolaren Bereich, einer moderaten Membranpermeabilität und einer Plasmastabilität von > 2 h können die Verbindungen 94 und 96 somit als erste Wirkstoffkandidaten angesehen werden. Die vollständige physiko-chemische Charakterisierung wurde mittels eines Sirius-T3-Titrationssystems durchgeführt. Unter Verwendung dieser Parameter wurden für die Derivate 94 und 96 je zwei Plasmakonzentrationsprofile mit der Software Simcyp® simuliert. Basierend auf diesem Studiendesign wurde jeweils die hohe Dosis beider Derivate im Mausmodel (T. b. rhodesiense) untersucht. Während nach 5-tägiger Behandlung mit 94 und 96 bei sämtlichen Tiere keine Parasiten mehr nachweisbar waren, wurde ein leichter Rückfall in beiden Versuchsgruppen an Tag 8 beobachtet. Gegenwärtig wird die Behandlung mit beiden Derivaten fortgesetzt.
