Bioactivity-guided isolation supported by LC-HRESIMS metabolic profiling led to the isolation of two new compounds, a ceramide, stylissamide A (1), and a cerebroside, stylissoside A (2), from the methanol extract of the Red Sea sponge Stylissa carteri. Structure elucidation was achieved using spectroscopic techniques, including 1D and 2D NMR and HRMS. The bioactive extract’s metabolomic profiling showed the existence of various secondary metabolites, mainly oleanane-type saponins, phenolic diterpenes, and lupane triterpenes. The in vitro cytotoxic activity of the isolated compounds was tested against two human cancer cell lines, MCF-7 and HepG2. Both compounds, 1 and 2, displayed strong cytotoxicity against the MCF-7 cell line, with IC\(_{50}\) values at 21.1 ± 0.17 µM and 27.5 ± 0.18 µM, respectively. They likewise showed a promising activity against HepG2 with IC\(_{50}\) at 36.8 ± 0.16 µM for 1 and IC\(_{50}\) 30.5 ± 0.23 µM for 2 compared to the standard drug cisplatin. Molecular docking experiments showed that 1 and 2 displayed high affinity to the SET protein and to inhibitor 2 of protein phosphatase 2A (I2PP2A), which could be a possible mechanism for their cytotoxic activity. This paper spreads light on the role of these metabolites in holding fouling organisms away from the outer surface of the sponge, and the potential use of these defensive molecules in the production of novel anticancer agents.
Thalassodendron ciliatum (Forssk.) Den Hartog is a seagrass belonging to the plant family Cymodoceaceae with ubiquitous phytoconstituents and important pharmacological potential, including antioxidant, antiviral, and cytotoxic activities. In this work, a new ergosterol derivative named thalassosterol (1) was isolated from the methanolic extract of T. ciliatum growing in the Red Sea, along with two known first-reported sterols, namely ergosterol (2) and stigmasterol (3), using different chromatographic techniques. The structure of the new compound was established based on 1D and 2D NMR spectroscopy and high-resolution mass spectrometry (HR-MS) and by comparison with the literature data. The new ergosterol derivative showed significant in vitro antiproliferative potential against the human cervical cancer cell line (HeLa) and human breast cancer (MCF-7) cell lines, with IC\(_{50}\) values of 8.12 and 14.24 µM, respectively. In addition, docking studies on the new sterol 1 explained the possible binding interactions with an aromatase enzyme; this inhibition is beneficial in both cervical and breast cancer therapy. A metabolic analysis of the crude extract of T. ciliatum using liquid chromatography combined with high-resolution electrospray ionization mass spectrometry (LC-ESI-HR-MS) revealed the presence of an array of phenolic compounds, sterols and ceramides, as well as di- and triglycerides.
Die Darstellung der Verbindungen 3a (sowie 3b) und 10, die sich vom 1-(4-Chlorphenyl)-1-(2~ diethylaminoethoxy)silacyclohexan (Sila-Chlorphencyclan, II a) ableiten, wird beschrieben. Die Verbindung 3 b wurde pharmakologisch und toxikologisch untersucht. Die biologischen Eigen· schaften von 3b wurden mit denen von Ila (sowie Chlorphencyclan) und seinem Hydrochiarid Ilb verglichen.
Sila-Chlorphencyclan (8b), ein Sila-Analogon des Chlorphencyclans (8a), die Derivate 7 und 9, deren Ammoniumsalze 11, 12, 13 und 14b, das Hydrolyseprodukt 10 sowie die Vorstufen 3-6 wurden erstmalig dargestellt. Die neuen Verbindungen wurden in ihren chemischen und physikalischen Eigenschaften charakterisiert, ihre Struktur wurde sichergestellt. Chlorphencyclan, Sila-Chlorphencyclan und einige seiner Derivate wurden vergleichend pharmakologisch und toxikologisch untersucht.
Identifizierung und Strukturaufklärung von Anthocyanen und ihrer Metabolite erfolgten mit Hilfe der mittels Hochleistungsflüssigchromatographie-Diodenarray-Detektion-Elektro-spray-Tan¬dem¬massen¬spektrometrie (HPLC-DAD-ESI-MS/MS). Quantitative Analysen wurden via HPLC-DAD durchgeführt. Die hierzu erforderlichen Referenzverbindungen wurden mittels präparativer HPLC aus Heidelbeeren isoliert (Reinheit zwischen 85,8% und 99,4%). Der Gehalt an Anthocyanen in den untersuchten Heidelbeerfrüchten lag bei 6 g/kg. Bezüglich der mengen¬mäßigen Verteilung dominierten Delphinidin- und Cyanidin¬glykoside vor den Glykosiden von Malvidin, Petunidin und Peonidin. Als konjugierte Zucker¬reste kamen vor allem Glukose und Galaktose vor, der Gehalt an Arabinosiden war weit geringer. Bei oraler Aufnahme erfolgt ein erster Kontakt der Anthocyane mit Speichel. Daher wurde dessen Wirkung auf die Heidelbeeranthocyane in ex vivo-Studien über einen (unphysio-logisch langen) Zeitraum von bis zu 30 Minuten untersucht. Dabei konnte wurde ins-besondere der Einfluß des pH-Wertes auf die Stabilität der Anthocyane aufgezeigt werden. Zur Simulation des Verhaltens von Anthocyanen im Magen wurden die einzelnen Heidelbeeranthocyane mit künstlichem Magensaft (pH 1,81) über vier Stunden inkubiert. Hier erwiesen sich alle untersuchten Verbindungen als stabil. Die anschließend von uns mit simuliertem Duodenalsekret (pH 7,2) über einen Zeitraum von 24 Stunden durchgeführten Studien zeigten, dass die Anthocyane unterschiedlich starken Modifizierungen unterlagen. Unter den schwach alkalischen Bedingungen wurden vor allem die Glykoside des Delphinidins schnell abgebaut, aber auch die übrigen Anthocyane erwiesen sich unter diesen Bedingungen als nicht stabil; nach 24 h war kein Anthocyan mehr nachweisbar. Um die Metabolisierungsvorgänge der Anthocyane im Dünn- und Dickdarm zu untersuchen, wurden ex vivo-Inkubationen jeweils mit frischem Ileo- bzw. Kolo¬stoma-beutel¬inhalt durchgeführt. Während die Abbaugeschwindigkeit in der ilealen Flüssigkeit vor allem von der pH-Stabilität des Aglykons abhänig war, konnten im Dickdarm einzig die Arabinoside nach einer Stunde noch alle in geringen Konzentrationen identifiziert werden. Die meisten Glukoside und Galaktoside waren zu diesem Zeitpunkt schon vollständig abgebaut. Da im Darm von einer hydrolytischen Spaltung der Anthocyane in Anthocyanidin und Zucker ausgegangen wird, wurde die Metabolisierung von Anthocyanidinen unter physio-logischen pH-Bedingungen untersucht. Neben der jeweiligen Spaltung in das Benzoe¬säure-derivat des B-Ringes sowie Phloroglucinessigsäure traten verschiedene Poly¬merisierungs¬-produkte auf, deren Strukturen nicht aufgeklärt werden konnten. In einer weiteren Versuchsreihe wurde die renale Ausscheidung von Anthocyanen bei Ileostomieprobanden nach oraler Applikation von 300 g Heidelbeeren über einen Zeitraum von acht Stunden untersucht. Es zeigte sich, dass ein Stoma des terminalen Ileums keinen Einfluss auf die Absorption und Metabolisierung der Anthocyane hatte. Die Bilanzierung der Anthocyane im Urin erfolgte als Äquvalente von Malvidin-3-O-glukosid, da nicht alle Anthocyanmetabolite zur Verfügung standen. Der Zeitpunkt der maximalen renalen Anthocyanausscheidung sowie die Menge der ausgeschiedenen Anthocyane waren starken interindividuellen Schwankungen unterworfen. Das Aus¬sscheidungs¬maximum (tmax) lag zwischen 0,5 und zwei Stunden. Bei der ausge¬schiedenen Menge wurden Werte zwischen 0,007% und 0.019% der auf¬ge¬nommenen Anthocyane ermittelt. Aufgrund der literaturbekannten Unterschiede zwischen den in Serum und Urin gefunden Anthocyanmengen ist davon auszugehen, dass es nach Anthocyanverzehr zu Inter-aktionen mit Proteinen in Blut oder Geweben kommt. Mittels Blutfraktionierung wurde das humane Serumalbumin (HSA) als wichtigster Bindungspartner der Anthocyane im Blut identifiziert. Anhand spektroskopischer Methoden war es möglich, die Bindungs¬parameter zu berechnen. Als Bindungsort wurde der hydrophile Eingang der lipophilen Warfarin-Bindungstasche in der Subdomäne IIA des HSA-Moleküls mittels "molecular modelling" identifiziert. Nasschemische Untersuchungen ergaben, dass die Bindung der Anthocyane an HSA diese vor ihrem pH-abhängigen Abbau schützt. Eine signifikante Herab¬setzung der chemischen Abbaugeschwindig¬keit konnte auch für bovines Serumalbumin beobachtet werden. Diese Erkenntnis ließ sich auf andere, mit dem HSA-Molekül nicht strukurverwandte lebensmittelrelevante Albumine übertragen. So zeigten Anthocyane große Stabilität in Milch und Eiklar, wobei die Stabilisierung auf eine Wechselwirkung mit den Proteinen Laktalbumin und Ovalbumin zurückgeführt werden konnte. Die in dieser Arbeit erlangten Erkenntnisse hinsichtlich Absorption, Metabolisierung und systemischer Verfügbarkeit im menschlichen Organismus leisten einen Beitrag zum besseren Verständnis der Wirkungen von Anthocyanen. Die neuen Erkenntnisse der Protein¬bindung sind vor allem für die Bewertung der Verfügbarkeit der Anthocyane in humanem Gewebe relevant.
Melanoma formation in the teleost Xiphophorus is caused by a dominant genetic locus, Tu. This locus includes the Xmrk oncogene, which encodes a receptor tyrosine kinase. Tumor induction is. suppressed in wild-type fish by a tumor suppressor locus, R. Molecular genetic analyses revealed that the Tu locus emerged by nonhomologaus recombination of the Xmrk proto-oncogene with a previously uncharacterized sequence, D. This event generated an additional copy of Xmrk with a new promoter. Suppression of the new Xmrk promoter by R in parental fish and its deregulation in hybrids explain the genetics of melanoma formation in Xiphophorus.
The melanoma inducing locus of Xiphophorus encodes a tumorigenic version of a novel putative receptor tyrosine kinase (Xmrk). To elucidate the mechanism of oncogenic activation of Xmrk, we compared the structure and expression of two oncogenic loci with the corresponding proto-oncogene. Only minor structural alterations were found to be specific for the oncogenic Xmrk genes. Marked overexpression of the oncogene transcripts in melanoma, which are approximately 1 kb shorter than the proto-oncogene transcript, correlates with the malignancy of the tumors. The tumor transcripts are derived from an alternative transcription start site that is used only in the oncogenic loci. Thus, oncogenic activation of the melanoma inducing Xmrk gene appears primarily to be due to novel transcriptional control and overexpression.
The high failure rate of new drug candidates in preclinical or clinical studies due to hepatotoxicity represents a considerable problem in the drug development. Hence, there is an urgent need to develop new approaches for early and reliable prediction of drug-induced hepatotoxicity that enables a better identification of drug candidates with high potential for toxicity at early stages of drug development. Therefore, the aim of this work was to improve the prediction of drug-induced liver injury in preclinical studies through evaluation of more reliable and sensitive biomarkers of hepatotoxicity and a better understanding of the underlying mechanistic basis for drug-induced toxicity. First, the ability of a set of potential markers (NGAL, thiostatin, clusterin, PON1) to detect early signs of liver injury was assessed in rats treated with drug candidates that were dropped from further development, in part due to toxic adverse effects in the liver. In summary, PON1 and clusterin were not consistently altered in response to liver injury and thus provide no additive information to the traditional liver enzymes in detecting drug-induced hepatotoxicity. In contrast, thiostatin and NGAL were increased in serum and urine of treated animals in a time- and dose-dependent manner. These changes correlated well with mRNA expression in the target organ and generally reflected the onset and degree of drug-induced liver injury. Receiver-operating characteristics analyses supported serum thiostatin, but not NGAL, as a better indicator of drug-induced hepatobiliary injury than conventional clinical chemistry parameters, such as ALP, ALT and AST. Although thiostatin, an acute phase protein expressed in a range of tissues, may not be specific for liver injury, our results indicate that thiostatin may serve as a sensitive, minimally-invasive diagnostic marker of inflammation and tissue damage in preclinical safety assessment. In the second part of this work, combined application of genomics profiling technology and RNAi to inhibit the pharmacological target of a drug candidate BAY16, a glucagon receptor (GCGR) antagonist, was used to determine if interference with the pharmacological target plays a role in the toxic response to BAY16, and to narrow down those molecular changes that are associated with toxicity, and not the pharmacological action of BAY16. In contrast to Bay 16, which was found to be cytotoxic at concentrations of 75 µM, silencing of the glucagon receptor did not affect cell viability in primary rat hepatocytes. Thus, it can be concluded that hepatotoxicity of Bay 16 was not related to the drugs inhibitory effect on the glucagon receptor in vitro and in vivo. These findings were supported by the fact that most of BAY16-induced changes in gene expression occurred independently of the pharmacological modulation of GCGR. These off-target effects include altered xenobiotic metabolism, oxidative stress, increased fatty acid synthesis, and alterations in cholesterol and bile acid metabolic processes. Although it was not possible to draw a final conclusion about the mechanism of BAY16 hepatotoxicity, changes in these molecular mechanisms appear contribute to progression of hepatic injury. With regard to drug safety assessment in preclinical studies, the utilization of siRNA technology in vitro represents a new approach to improve mechanistic understanding of the nature of drug’s toxicity, being either chemically mediated or due to primary or secondary pharmacological mode of action.
Optimal open-loop control, i.e. the application of an analytically derived control rule, is demonstrated for nanooptical excitations using polarization-shaped laser pulses. Optimal spatial near-field localization in gold nanoprisms and excitation switching is realized by applying a shift to the relative phase of the two polarization components. The achieved near-field switching confirms theoretical predictions, proves the applicability of predefined control rules in nanooptical light–matter interaction and reveals local mode interference to be an important control mechanism.
Radiationless energy transfer is at the core of diverse phenomena, such as light harvesting in photosynthesis\(^1\), energy-transfer-based microspectroscopies\(^2\), nanoscale quantum entanglement\(^3\) and photonic-mode hybridization\(^4\). Typically, the transfer is efficient only for separations that are much shorter than the diffraction limit. This hampers its application in optical communication and quantum information processing, which require spatially selective addressing. Here, we demonstrate highly efficient radiationless coherent energy transfer over a distance of twice the excitation wavelength by combining localized and delocalized\(^5\) plasmonic modes. Analogous to the Tavis-Cummings model, two whispering-gallery-mode antennas\(^6\) placed in the foci of an elliptical plasmonic cavity\(^7\) fabricated from single-crystal gold plates act as a pair of oscillators coupled to a common cavity mode. Time-resolved two-photon photoemission electron microscopy (TR 2P-PEEM) reveals an ultrafast long-range periodic energy transfer in accordance with the simulations. Our observations open perspectives for the optimization and tailoring of mesoscopic energy transfer and long-range quantum emitter coupling.
The aim of the present study was to design different dosage forms as carrier systems to deliver sorafenib to the lung of BXB-23 transgenic mice using different routes of administration. Three dosage forms were used one of them was an oil-in-water emulsion and the oral route was chosen for this experiment. The other delivery system was a liposome preparation for intratracheal instillation. In this case the oral route was considered as a control experiment. The last dosage form was PLGA microspheres. Before sorafenib administration it was important to develop a HPLC method to assess sorafenib absorption after its administration and to determine its concentrations in mouse serum. The HPLC method allowed sorafenib quantification in small volumes (30 µl) of mouse serum and tissues. The developed HPLC method was validated resulting in satisfactory selectivity, good linearity, good accuracy and precision over the concentration range examined. Sorafenib was successfully incorporated in a fat emulsion (o/w) using a traditional method resulting in a white homogenous emulsion and no particle aggregation was observed. Sorafenib exhibited antitumor activity on the lung adenoma in BXB-23 transgenic mice when administered orally (2 mg sorafenib per mouse) in the emulsion preparation. The determined effect was an approximately 29 % reduction in the tumor area of the adenoma foci and a proliferation reduction. In order to improve the pharmacological effects of sorafenib on the lung adenoma in BXB-23 mice, the targeting of sorafenib directly to the site of action (the lung) was an attractive concept. For this purpose the intratracheal route was used. Since sorafenib administration by instillation required incorporation of sorafenib in a dosage form suitable for its lipophilic nature, a liposome suspension was the second dosage form used. A lyophilization method was employed for sorafenib liposome preparation utilizing dilauroylphosphatidylcholine (DLPC) which is safe and tolerable for the lung. Incorporation of sorafenib in the liposomes did not influence the particle size and its distribution. The sorafenib liposomes showed high encapsulation efficiency, good stability at 4 °C for one month and satisfactory in vitro release properties and inhibited Raf-1 mediated activation of ERK in cell culture assay. In a pharmacokinetic experiment sorafenib loaded liposomes were instilled directly into the lung. The results revealed that a significant level of sorafenib was achieved in the lung tissues after 2 hours and then reduced after 48 h and remained nearly constant for one week. On the other hand, only traces of sorafenib were found in the mice serum up to 48 h. Subsequently, the pharmacological activity of sorafenib (1 mg per mouse) was studied when delivered in a liposomal suspension intratracheally to treat the lung adenoma of BXB-23 mice. The data of this experiment demonstrated that sorafenib intratracheal instillation resulted in a reduction of tumor area of adenoma foci (67 %) and an elevation of the percent of apoptotic cells. In contrast, prolongation of the treatment period did not further enhance sorafenib activity on the lung adenoma. This previous finding suggested a development of multidrug resistance (MDR) by the adenoma foci cells against sorafenib instillation, which was examined by immunohistochemistry staining. The percent of MDR positive cells was higher after two and three weeks sorafenib liposome instillation treatment than that after one week treatment. The last dosage form used for sorafenib was microspheres, which were prepared by emulsion-diffusion-evaporation method using biodegradable PLGA 50:50 resulting in a white lyophilized powder. The system was characterized physicochemically and revealed a good microspheres yield, high encapsulation efficiency, a homogenous particle size distribution and slow in vitro release of sorafenib. The other strategy studied in the present research project was gene delivery to target the lung bearing tumor of BXB-23 mice using a non-viral vector (polyethylenimine). Polyethylenimine (PEI) was used to investigate its efficiency in transfecting lung bearing tumor of BXB-23 mice model and its ability to transfect the adenoma foci cells. LacZ, which encodes Beta-galactosidase was used in the present study as a reporter gene and was complexed with PEI before delivered intravenously. A high LacZ expression in the alveolar region with some expression in the adenoma foci was observed. On contrary, a low LacZ expression in the alveoli and in the adenoma foci was achieved after instillation of the same polyplex intratracheally.
A liquid chromatography tandem mass spectrometry method for the analysis of ten kinase inhibitors (afatinib, axitinib, bosutinib,cabozantinib, dabrafenib, lenvatinib, nilotinib, osimertinib, ruxolitinib, and trametinib) in human serum and plasma for theapplication in daily clinical routine has been developed and validated according to the US Food and Drug Administration andEuropean Medicines Agency validation guidelines for bioanalytical methods. After protein precipitation of plasma samples withacetonitrile, chromatographic separation was performed at ambient temperature using a Waters XBridge® Phenyl 3.5μm(2.1×50 mm) column. The mobile phases consisted of water-methanol (9:1, v/v) with 10 mM ammonium bicarbonate as phase A andmethanol-water (9:1, v/v) with 10 mM ammonium bicarbonate as phase B. Gradient elution was applied at a flow rate of 400μL/min. Analytes were detected and quantified using multiple reaction monitoring in electrospray ionization positive mode. Stableisotopically labeled compounds of each kinase inhibitor were used as internal standards. The acquisition time was 7.0 min perrun. All analytes and internal standards eluted within 3.0 min. The calibration curves were linear over the range of 2–500 ng/mLfor afatinib, axitinib, bosutinib, lenvatinib, ruxolitinib, and trametinib, and 6–1500 ng/mL for cabozantinib, dabrafenib, nilotinib,and osimertinib (coefficients of correlation≥0.99). Validation assays for accuracy and precision, matrix effect, recovery,carryover, and stability were appropriate according to regulatory agencies. The rapid and sensitive assay ensures high throughputand was successfully applied to monitor concentrations of kinase inhibitors in patients.
G protein-coupled receptor research looks out for new technologies to elucidate the complex
processes of receptor activation, function and downstream signaling with spatiotemporal
resolution, preferably in living cells and organisms. A thriving approach consists in making use
of the unsurpassed properties of light, including its high precision in space and time, noninvasiveness
and high degree of orthogonality regarding biological processes. This is realized
by the incorporation of molecular photoswitches, which are able to effectively respond to light,
such as azobenzene, into the structure of a ligand of a given receptor. The muscarinic
acetylcholine receptors belong to class A GPCRs and have received special attention in this
regard due to their role as a prototypic pharmacological system and their therapeutic potential.
They mediate the excitatory and inhibitory effects of the neurotransmitter acetylcholine and
thus regulate diverse important biological processes, especially many neurological functions in
our brain.
In this work, the application of photopharmacological tool compounds to muscarinic receptors
is presented, consisting of pharmacophores extended with azobenzene as light-responsive
motif. Making use of the dualsteric concept, such photochromic ligands can be designed to bind
concomitantly to the orthosteric and allosteric binding site of the receptor, which is
demonstrated for BQCAAI (M1) and PAI (M2) and may lead to subtype- and functionalselective
photoswitchable ligands, suitable for further ex vivo and in vivo studies.
Moreover, photoswitchable ligands based on the synthetic agonist iperoxo were investigated
extensively with regard to their photochemical behavior and pharmacological profile, outlining
the advantages and challenges of using red-shifted molecular photoswitches, such as tetraortho-
fluoro azobenzene. For the first time on a GPCR it was examined, which impact the
different substitution pattern has on both the binding and the activity on the M1 receptor. Results
show that substituted azobenzenes in photopharmacological compounds (F4-photoiperoxo and
F4-iper-azo-iper) not just represent analogs with other photophysical properties but can exhibit
a considerably different biological profile that has to be investigated carefully.
The achievements gained in this study can give important new insights into the binding mode
and time course of activation processes, enabling precise spatial and temporal resolution of the
complex signaling pathway of muscarinic receptors. Due to their role as exemplary model
system, these findings may be useful for the investigation into other therapeutically relevant
GPCRs.
Assessing particle deposition in a representative in vitro model of the rat respiratory tract

The aim of this thesis was to develop an in vitro model (IVR) of the rat lung for the purpose of investigating the deposition of drug particles in the rat airways. The model attempted to account for the affect of drug product characteristics and physiological parameters on deposition in the lungs. In addition, the model outputs were compared with in vivo lung deposition results from live rats and in silico predictions using published computer model of lung deposition in pre-clinical species.
Initial work focussed on developing an aerosol exposure system capable of dosing small rodent to a range of airborne test materials. The system consists of two main parts; a fluidised bed aerosol generator and connection of the generator output to a nose only exposure chamber capable of accommodating 12 small animals in a single layer. In addition, an aerodynamic particle spectrometer (APS) was installed for continuously measuring the size distribution and airborne concentration of aerosol particles generated in the exposure chamber. System validation showed acceptable degree of variation of the test material tested, Fluorescent Microspheres (FMS) throughout the exposure chamber (CV < 15.0%). Particle size (MMAD ± GSD) using the APS was shown to be stable throughout the exposure periods.
The IVR model developed in this project was based on a number of euthanased (n=7), female Sprague-Dawley rats (weight: 372 ± 56 g), which underwent high-resolution micro-CT scans. The physical model consisted of five sub sections; Extra-Thoracic region containing the snout and nasophyarynx, trachea-bronchial region containing the trachea, bronchi, and bronchioles. All sections of the model were attached to one another in numerical order and housed within a containment unit. At the rear end of the cast, a flexible diaphragm was attached in order to collect the fraction of inhaled particles exiting the TB section and possibly reaching the lung, referred to as the Post-TB section.
A study was conducted to assess the influence of inhalation parameters such as the breathing frequency and tidal volume on total and regional dose distribution using FMS as test material. The major finding of this study was the demonstration of the model sensitivity to changes in breathing parameters especially respiratory frequency, where the data showed increased deposition in the peripheral regions of the model with decreased respiratory frequency. Other studies assessed the effect of particle characteristics on deposition on the IVR model, such as particle size, dose increase and formulation changes.
The results assessing particle size effect showed a slightly higher deposition levels for the 4µm sized particles versus 2µm sized particles in the head region; 90.8 ± 3.6% and 88.2 ± 6.6%. However, this difference did not reach statistical significance (P> 0.05) probably due to the polydispersity of aerosolised FMS particles. In addition, the regional deposition analysis showed an increased lung peripheral deposition with the smaller particles. In addition, the model was shown to be sensitive to changes in formulation composition mediated by inclusion of MgSt.
The next stage of work was to validate the model in terms of comparison with lung deposition for in vivo rats. For lung deposition comparison, the absolute amount deposited in the IVR lung model (expressed as µg/kg) was shown to have a reasonably strong correlation with in vivo lung concentration measures (µg/kg); R2= 0.66, P < 0.05. Compounds were predicted well and within 2-folds of the measured lung deposition values. However, knowing the variability in biological systems and the multiple components required to estimate lung doses, predictions within 2-fold of the measured values would seem reasonable
In terms of comparison with in silico model predictions using MPPD, similar deposition levels were noted between the two models, particularly when the data was expressed as percentage of total particles inhaled. The data showed the highest deposition levels were noted in the head region (> 80%) and less than 5.0% deposition for the peripheral lung fractions.
With regards to using the IVR model to assess the relationship between dose, particle size and efficacy, an in vivo study using FP with different particle sizes (2.0 and 4.0 µm) but same doses ( 100 and 1000 µg/kg). This study demonstrated that exposure of rat to FP powder resulted in a dose-dependent inhibition of neutrophils in BAL fluids. However, a clear difference in neutrophils suppression was demonstrated for equivalent doses but different particle sizes of FP, where the smaller FP particles (2.0 µm) induced a greater level of neutrophils suppression in comparison with larger FP particles (4.0 µm). In addition, a reasonably good correlation for the relationship between lung deposition in the IVR model and a neutrophils suppression level was demonstrated. Furthermore this data support the hypothesis that regional deposition is an important determinant in efficacy. Therefore, this suggests that the IVR model may be a useful as a tool to describe in vivo efficacy with in vitro data. However, further studies should be conducted to evaluate the validity of this model and relationship.
The IVR model has a number of important limitations. First, the model is based on scans up to generation four of the rat respiratory tract as this represented the limits of the micro-CT scanning technology at the time of this study. Therefore deposition in the deeper region of the lung may not be reflected precisely in the IVR model. Second, the regional deposition data generated using the model tended to show an overestimation of deposition in head region and an underestimation of deposition in the peripheral regions of the lung, in comparison with in vivo lung deposition data. Third, the current model does not take into account lung clearance. However, the amount of the drug present in the in vivo lungs is dependent on numerous physiological processes such as dissolution, passive or active absorption into the systemic circulation, binding to lung tissue and mucociliary clearance. Consequently, the results generated using this IVR model for drug molecules with high lung clearance rate should be treated with some caution.
Future work extending this research could go in a number of directions. In this research, a representative model of the rat respiratory tract was constructed from analysis of imaging data from a number of euthanised Sprague-Dawley rats. This model represented the “average respiratory tract” in terms of dimensions of Sprague-Dawley rats. However, there is considerable variability in the airway dimensions between rats. This variability encompasses a number of factors such as the strains of rats, sex and age, and disease state. Thus, it may be possible to produce a small number of airway models to represent small and large rats and scaled to represent the extrathoracic and peripheral regions based on literature reports of their dimensions in different rat populations. This approach will then enable the effect of intersubject airway dimensions for different rat populations on aerosol deposition to be thoroughly examined.
In addition, due to the limitation of the micro-CT technology used to construct the physical IVR model, detailed morphology only up to generation 4 were captured. However, recent advances in MRI technology, such as the use of in situ-MRI based scanning technology have enabled rat airway morphometry to be extended to 16 airway generation. This coupled with improvements in the resolutions of rapid-prototyping process means it may be possible to construct a rat model that reflects the in vivo lung morphology more accurately, and thus enable greater understanding of the link between aerosol deposition and airway geometry.
In conclusion, a model cast of the rat lung was developed and validated to allow the deposition of inhaled particles in the rat lung to be investigated. The model may be used to estimate the lung concentration in vivo rats in preference to exposure concentration measurements based on filter samples which have been shown to be a poor indicator of the lung concentration immediately after exposure. In addition, the model has the potential to be used along with live rats in an inhalation rig in pulmonary pharmaceutics research and may facilitate in development of inhaled formulations to target specific regions within the lung as well as screening of inhaled drugs in preclinical setting.
In the present report, well-defined WO3 nanorods (NRs) and a rGO–WO\(_3\) composite were successfully synthesized using a one-pot hydrothermal method. The crystal phase, structural morphology, shape, and size of the as-synthesized samples were studied using X-ray diffraction (XRD) and transmission electron microscopy (TEM) measurements. The optical properties of the synthesized samples were investigated by Raman, ultraviolet-visible (UV-Vis) and photoluminescence (PL) spectroscopy. Raman spectroscopy and TEM results validate the formation of WO\(_3\) (NRs) on the rGO sheet. The value of the dielectric constant (ε′) of WO3 NRs and rGO–WO\(_3\) composite is decreased with an increase in frequency. At low frequency (2.5 to 3.5 Hz), the value of ε′ for the rGO–WO3 composite is greater than that of pure WO\(_3\) NRs. This could be due to the fact that the induced charges follow the ac signal. However, at higher frequency (3.4 to 6.0), the value of ε′ for the rGO–WO\(_3\) composite is less compared to that of the pure WO3 NRs. The overall decrease in the value of ε′ could be due to the occurrence of a polarization process at the interface of the rGO sheet and WO3 NRs. Enhanced interfacial polarization in the rGO–WO\(_3\) composite is observed, which may be attributed to the presence of polar functional groups on the rGO sheet. These functional groups trap charge carriers at the interface, resulting in an enhancement of the interfacial polarization. The value of the dielectric modulus is also calculated to further confirm this enhancement. The values of the ac conductivity of the WO\(_3\) NRs and rGO–WO\(_3\) composite were calculated as a function of the frequency. The greater value of the ac conductivity in the rGO–WO\(_3\) composite compared to that of the WO\(_3\) NRs confirms the restoration of the sp:\(^{++}\) network during the in situ synthesis of the rGO–WO\(_3\) composite, which is well supported by the results obtained by Raman spectroscopy.
Die Bedeutung der cAMP- und cGMP-abhängigen Proteinkinase für die Hemmung der Plättchenaktivierung und -aggregation ist gut beschrieben. Zahlreiche fundamentale Plättchenantworten wie die Erhöhung der intrazellulären Calciumkonzentration, die Exposition von Adhäsionsrezeptoren und die Aktinpolymerisation können durch die Cyclonukleotid vermittelte Kinasenaktivierung fast vollständig gehemmt werden. Die Vielfalt der cGMP bindenden Proteine und deren synergistische Interaktion mit cAMP vermittelten Signalwegen deuten auf eine Reihe von cGMP Zielproteinen hin. Vor kurzem wurde die zentrale Bedeutung einer Gi-Protein Stimulation für die Plättchenaktivierung und –aggregation gezeigt. In dieser Dissertation wurde daher der Frage nachgegangen, ob Signalmoleküle, die an Gi-Protein vermittelten Effekten beteiligt sind, einen Angriffspunkt für cAMP/cGMP-abhängige Proteinkinasen darstellen. Zu diesem Zweck wurden die Effekte erhöhter cGMP Spiegel und die selektive Aktivierung der cGMP-abhängigen Proteinkinase auf die adrenerge und purinerge Rezeptor vermittelte Erniedrigung stimulierter cAMP Konzentrationen untersucht. In unseren Versuchen konnte erstmalig gezeigt werden, dass eine Erhöhung der intrazellulären cGMP Konzentration Gi-Protein vermittelte Signale hemmt. Dieses erfolgt nicht auf Grund einer cGMP stimulierten Aktivierung von cyclonukleotidabbauenden Phosphodiesterasen, sondern auf Grund einer Aktivierung der cGMP-abhängigen Proteinkinase. In Anbetracht der essentiellen Bedeutung der Gi-Protein Stimulation für die Plättchenaktivierung stellt dies einen wichtigen Mechanismus dar, wie das aus dem Endothel freigesetzte NO über cGMP die Thrombozytenfunktion hemmt. Klinisch bedeutsame Substanzen wie Clopidogrel oder Ticlopidin imitieren diesen in vivo Effekt des NO, indem sie extrazellulär über eine Rezeptorhemmung Gi-Protein Stimulation verhindern. (Aktas et al., Biochem Pharmacol 2002; 64: 433-439) Dipyridamol und im Besonderen die Kombination von Dipyridamol und niedrig dosierter Acetylsalicylsäure sind in der Sekundärprävention des Schlaganfalles sehr gut wirksam. Jedoch sind die hierfür zu Grunde liegenden biochemischen Mechanismen noch nicht vollständig aufgeklärt. Da für das Dipyridamol eine in vitro Hemmung der cGMP-spezifischen Phosphodiesterase 5 (PDE 5) nachgewiesen ist, wurde in dieser Arbeit untersucht, ob Dipyridamol in therapeutisch relevanten Konzentrationen die NO/cGMP vermittelte Effekte auf die Plättchenfunktion unter ex vivo Bedingungen verstärkt. Die Phosphorylierung von VASP (VAsodilator-Stimulated Phosphoprotein) diente dabei als Meßparameter NO/cGMP Signale in Thrombozyten mit Hilfe von Antikörpern und Western Blot Technik zu quantifizieren. Die Sekretion von Serotonin aus Thrombozyten und die Aktivität der Thromboxansynthase wurden durch die fluorimetrische Bestimmung derivatisierten Serotonins bzw. des Synthaseprodukts Malondialdehyd quantifiziert. Endotheliale Faktoren wie NO oder PG-I2 erhöhen cGMP bzw. cAMP, die zu einer Plättchenhemmung und gleichzeitigen VASP Phosphorylierung führen. In in vitro Versuchen potenzierte Dipyridamol in einer therapeutisch relevanten Konzentration (3,5 µmol/l) nur die cGMP vermittelte, aber nicht die cAMP vermittelte VASP Phosphorylierung. Darüber hinaus konnte Dipyridamol (3,5 µmol/l) die Hemmung von Plättchenfunktionen wie der Serotoninsekretion und die Aktivität der Thromboxansynthase durch einen NO Donor klar verstärken. Schließlich steigerte Dipyridamol die NO vermittelte VASP Phosphorylierung auch in Thrombozyten von Probanden, die vorher Dipyridamol eingenommen hatten. Unter therapeutisch relevanten Bedingungen verstärkt also Dipyridamol NO/cGMP Signalwege und damit die Hemmung von Thrombozyten. Dieser Befund bekräftigt die Vorstellung, dass die Verstärkung endothelialer NO/cGMP Effekte auf Thrombozyten eine wichtige Komponente der Dipyridamol Wirkung unter in vivo Bedingungen darstellt. (Aktas et al., Stroke 2003; 34(3): 764-769) Die Stimulation von Thrombozyten führt u.a. zu einer Sekretion von Plättchenaktivatoren wie Thrombin, Thromboxan A2, ADP oder Serotonin aus dem Zellinnern. Durch diesen Prozess der Degranulierung können nun weitere Thrombozyten aktiviert werden. Die Sekretion stellt somit einen wichtigen, verstärkenden Schritt in der Aktivierung von Thrombozyten während der Hämostase dar. Diese Arbeit zeigt, dass in Thrombozyten eine Gi-Protein Aktivierung nicht nur wie bisher angenommen eine initiale Sekretion durch Gq verstärkt und aufrecht erhält, sondern der eigentliche Stimulus ist, der die Degranulierung von Thrombozyten auslöst. Die Stimulation Gq vermittelter Signalwege ist nur insofern erforderlich, als diese das Auslösen der Sekretion durch eine Aktivierung von Gi-Proteinen ermöglichen. Die Stimulierung beider G-Proteine ist daher essentiell für die thrombozytäre Sekretion. Zudem konnte die Phospholipase D als ein neuer Effektor des P2Y12 nachgewiesen werden, deren Stimulierung wahrscheinlich zur Degranulierung von Thrombozyten führt. Dieser Mechanismus könnte der Entscheidende sein, der der essentiellen Rolle des Gi-Proteins bei der Stimulation der Sekretion und der Aktivierung von Thrombozyten zu Grunde liegt und könnte ein neues Licht auf die Wirkweise des Clopidogrels und des Ticlopidins werfen, die irreversibel an den P2Y12 Rezeptor binden. In den starren Molekülen 1- 10 reagieren die benachbarten parallelen C = C- und N = N-Bindungen nahezu quantitativ unter Photocyclisierung lU den l,2-Diazetidinen 11-10, deren Struktur spektroskopisch und für 13 durch Kristallstrukturanalyse bewiesen wird. Die in Abwesenheit der C = C-Bindung beobachtete Photo-Denitrogenierung unterbleibt selbst bei den empfindlichen Derivaten des 2,3-Diazabicyclo[2.2.11heptens. Photocyclisierung von 6 mit lwei zur N=N· Bindung benachbarten C=C-Bindungen tritt nur mit der Norbornendoppelbindung ein.
Tropische Infektionskrankheiten wie Malaria, Leishmaniose oder auch die Afrikanische Trypanosomiase sind aufgrund von zunehmenden Resistenzen der Erreger, globaler Erwärmung, aber auch von Versäumnissen in der Vergangenheit bei der kontinuierlichen Weiterentwicklung bestehender sowie der Erforschung neuer Medikamente auch im 21. Jahrhundert noch eine große Bedrohung für Millionen von Menschen. Die Suche nach neuartigen Wirkstoffen und deren Weiterentwicklung zu potenziellen Medikamenten ist daher zwingend erforderlich. Insbesondere Produkte des Sekundärstoffwechsels wie etwa die Alkaloide bilden wichtige Grundlagen als Leitstrukturen für pharmazeutische Wirkstoffe. Eine solche Klasse phytochemischen Ursprungs sind die Naphthylisochinolin-Alkaloide mit interessanten strukturellen Eigenschaften sowie pharmakologischen Wirksamkeiten. Einige Vertreter zeigen ausgeprägte In-vitro-Aktivitäten gegen protozoische Erreger wie Plasmodien, Leishmanien und Trypanosomen. Besonders die neuartige Unterklasse ionischer N,C-verknüpfter Naphthylisochinolin-Alkaloide, wie z.B. Ancistrocladinium A und Ancistrocladinium B, zeichnen sich durch gute antileishmaniale Wirkungen aus. In Vorarbeiten zeigten erste Studien zu Struktur-Aktivitäts-Beziehungen (SAR-Studien) mit vereinfachten N,C-gekuppelten Arylisochinolinen, dass sich durch gezielte Strukturvariation die Aktivität gegen einen Erreger verbessern lässt. Zusätzlich wurde mit ersten Untersuchungen zum Wirkmechanismus dieser interessanten Verbindungen begonnen. Darüber hinaus ermöglicht die kontinuierliche Verbesserung der analytischen Methoden inzwischen die schnelle und gezielte Suche nach neuen Verbindungen aus der Natur. Durch die Anwendung von Online-Analyse-Verfahren, wie z.B. die Kopplung von HPLC mit NMR und MS, gelingt die Aufklärung der Konstitution von Substanzen direkt aus Extrakten. Ziel der vorliegenden Arbeit war die Verbesserung der biologischen Aktivitäten der N,C-verknüpften Arylisochinoline durch strukturelle Derivatisierung sowie Beiträge zur Aufklärung des Wirkmechanismus mittels markierter Verbindungen. Zusätzlich sollten Naturstoffe unter Verwendung moderner HPLC-Kopplungstechniken untersucht und strukturell aufgeklärt werden.
Das Ausmaß der Proteinbindung eines Arzneistoffs wirkt sich auf viele unterschiedliche pharmakokinetische Parameter aus. So wird beispielsweise das Verteilungsvolumen, die Metabolisierung oder die Elimination des entsprechenden Stoffes durch die Höhe seiner Proteinbindung beeinflusst. Da nur der im Plasma frei vorliegende Anteil eines Arzneistoffs in der Lage ist biologische Membranen zu überwinden, können auch nur die freien Arzneistoffmoleküle eine pharmakologische Wirkung an Rezeptoren oder Enzymen auslösen. Dementsprechend ist auch die Intensität der hervorgerufenen Wirkung von der Größe des ungebundenen Anteils eines Arzneistoffs abhängig. Aufgrund dieser Zusammenhänge ist klar, dass die Proteinbindung eines Arzneistoffes letztendlich Einfluss auf die Dosisfindung hat. Zur Ermittlung der Proteinbindung stehen viele unterschiedliche Methoden, wie beispielsweise die HPLC, Kapillarelektrophorese, Ultrazentrifugation, Gleichgewichtsdialyse und Ultrafiltration zur Verfügung. In der vorliegenden Arbeit wurde die kontinuierliche Ultrafiltration zur Ermittlung der Proteinbindung von Arzneistoffen angewendet. Hier wird die Proteinbindung nicht nur anhand einer bestimmten Arzneistoff- bzw. Albuminkonzentration gemessen, sondern über einen weiteren Bereich von Wirkstoff-Protein-Verhältnissen beobachtet. Des Weiteren ist der apparative Aufwand im Vergleich zu vielen anderen Methoden als geringer einzustufen. Im Rahmen dieser Arbeit wurde, die auf der von Heinze[122] entwickelte Messanlage weiter optimiert und eine zweite Anlage mit einem Diodenarraydetektor aufgebaut. Für letztere musste eine Software-Anpassung vorgenommen werden. Folgende Projekte wurden durchgeführt: 1) Um den In-vivo-Bedingungen nahe zu kommen, wurde bei der Bestimmung der Proteinbindung der Sartane nicht nur BSA und HSA verwendet, sondern erstmals auch humanes Plasma. Die Plasmamessungen der Sartane verliefen insgesamt problemlos, allerdings ist eine erfolgreiche Messung stark von der Qualität des eingesetzten Plasmas abhängig, wie Messungen der Naphthylisochinoline gezeigt haben. Im Vergleich mit HSA und Plasma ergaben die Messungen der Sartane mit bovinem Serumalbumin geringfügig erniedrigte Proteinbindungswerte. Insgesamt sind alle Ergebnisse sehr gut mit den Literaturwerten vergleichbar. 2) Das Ausmaß der Proteinbindung von Naphthylisochinolinen war bislang unbekannt und lag im Bereich von ca. 30-70%. Erneut waren die Resultate aus den Messungen von BSA und HSA nahezu gleich. 3) Am Beispiel der Interaktion zwischen Phenprocoumon und Phenylbutazon wurden zwei unterschiedliche Ansätze getestet, um die Verdrängung aus der Proteinbindung zu simulieren. Die erste Methode entsprach hierbei einer Konkurrenz der beiden interagierenden Stoffe um die Proteinbindungsstellen. Durch den Einfluss des Phenylbutazon verringerte sich die Proteinbindung des Phenprocoumon um 1%, was allerdings als statistisch nicht signifikant betrachtet werden kann. Im zweiten Ansatz, der eine direktere Verdrängung aus der Proteinbindung simulieren sollte, fiel die Proteinbindung des Phenprocoumon gegenüber den Einzelmessungen um 2,5% ab. Unter physiologischen Konzentrationsverhältnissen sank sich die Proteinbindung des Phenprocoumon auf 93,3%. Der freie Anteil erhöhte sich dementsprechend von 1% auf 6,7%. Somit konnte der Einfluss des Phenylbutazon auf die Proteinbindung des Phenprocoumon erfolgreich nachgewiesen werden. Die unveränderte Proteinbindung des Phenylbutazon im inversen Ansatz und die ermittelten pK-Werte bestätigen diese Interaktion. Grundsätzlich ist es also möglich mit der kontinuierlichen Ultrafiltration solche Interaktionen zu simulieren. 4) Zuletzt sollte der Frage nachgegangen werden, ob es mit der kontinuierlichen Ultrafiltration auch möglich ist die Proteinbindung von wasserunlöslichen Stoffen, nämlich den Aziridinen, in Gegenwart steigender Mengen DMSO, zu bestimmen. Die erhaltenen Ergebnisse wurden mit Literaturwerten ohne DMSO-Zusatz verglichen. Abgesehen von Candesartan, das eine lineare Korrelation zwischen DMSO-Gehalt der Wirkstofflösung und Absinken der Proteinbindung zeigte, konnte kein Zusammenhang zwischen der DMSO-Konzentration und der gemessenen Proteinbindung festgestellt werden. Die Mittelwerte lagen im Bereich der Literaturwerte. Insgesamt zeigten alle Versuchsreihen, dass die kontinuierliche Ultrafiltration eine ausgezeichnete, schnelle und robuste Screeningmethode zur Bestimmung des Ausmaßes der Proteinbindung bekannter und neuer Wirkstoffe darstellt.
The work presented in this thesis was mainly targeted at exploring the capabilities of evaporation based LC detectors as well as further alternatives for the control of impurities in substances not exhibiting a suitable chromophore for UV-detection. In the course of the work carried out, several new methods for the identification, impurities control and composition testing of APIs were elaborated. An evaporation based detector that entered into the field of pharmaceutical analysis in the recent years was the Evaporative Light Scattering Detector (ELSD). However, non-reproducible spikes were reported when injecting concentrated test solutions as they are usually required for the control of impurities. The reasons, for the appearance of these spikes as well as possibilities for their avoidance were explored in a systematic study. Moreover, the dependence of the detector sensitivity on different eluent composition, eluent flow-rate and ELSD settings was investigated. In the course of the revision of the Ph.Eur. monographs for aspartic acid and alanine, a C18 reversed phase ion-pair LC method using 1 mmol/L of perfluoroheptanoic acid as an ion-pair reagent and a charged aerosol detector (CAD) was developed and fully validated for the purity control of Asp. The method was capable of separating the organic acids and major amino acids known to occur as process related impurities. With a slight modification, the method was also applicable for the purity control of Ala. Based on the developed LC-CAD method for the impurity control of alanine, a comparative study of the performance characteristics of different evaporation based LC detectors, i.e. ELSD, CAD and the recently developed Nano Quantity Analyte Detector (NQAD) was carried out. Additionally, an MS detector and qNMR were included in this study. It was found that the control of impurities in Alanine at an ICH conform level could be ensured using LC coupled to CAD, MSD and NQAD detection as well as by the use of qNMR. In terms of performance, prize and ease of use CAD and NQAD were found to be the most suitable alternatives. In terms of repeatability and sensitivity, the CAD appeared slightly superior to the NQAD. The quality of streptomycin sulfate is not sufficiently controlled by the current Ph.Eur. monograph in that an appropriate test for the control of the related substances is missing. A study was carried out to develop a C18 reversed phase ion-pair LC method using pentafluoropropionic acid as an ion-pair reagent and a CAD for the identification and control of the related substances. The developed method allowed the separation of 21 impurities from streptomycin. Moreover, coupling of the method to MS allowed the identification of the separated impurities. The method was shown to be sufficiently sensitive to control the related substances with a disregard limit of 0.1% as it is normally applied in the Ph.Eur. for products derived from fermentation. Currently, the aescin content of horse-chestnut standardized dry extract is determined using a complex and laborious photometric determination. A more selective LC-UV assay determination for beta-aescin has been proposed for the Ph.Eur. draft monograph of horse-chestnut standardized dry extract. Possibilities were explored to further improve the LC-method using detection by CAD. It was demonstrated that by the use of a modified LC-CAD method several problems related to the differences in the UV-response of the various components contained in the active aescin fraction could be eliminated. Moreover the proposed reference standard strategy was reviewed. Eventually, it was demonstrated on the example of two different clusters of pharmacologically active peptides how low energy collision induced dissociation mass spectrometry (low energy CID-MS) can successfully be used for identification testing in pharmacopoeial monographs. In this respect, the combination of a direct confirmation of the molecular mass via the m/z-ratio of the molecule ions with structural sequence information obtained by low energy CID-MS experiments was found to deliver a higher degree of certainty of the identity of a given substance than the set of tests currently described in the monographs. A significant gain in efficiency and throughput and important reduction of the amount of sample consumed during testing were identified as being additional advantages of this approach. Taken together, it could be demonstrated on various examples how recent technological advancements in the field of analytical chemistry can contribute to improve the quality control of APIs.
Basierend auf dem Auslauftrichter nach DIN ISO 4324 wurde ein neuartiges Gerät zur Bestimmung der Fließeigenschaften von Schüttgütern entwickelt. Der modifizierte Auslauftrichter zerstört mit Hilfe eines speziellen Rührwerkzeuges ausflussverhindernde Schüttgutbrücken. Durch Charakterisierung des Ausflussverhaltens eines Modellschüttgutes (Aerosil 200/Maisstärke) konnten verschiedene Prozessparameter identifiziert werden, die eine Abhängigkeit von unterschiedlichen Fließeigenschaften des Modellschüttgutes aufweisen. Mit Hilfe des modifizierten Auslauftrichters wurden im weiteren Teil dieser Arbeit binäre Mischungen aus einem Fließregulierungsmittel und Maisstärke auf ihr Fließverhalten untersucht. Hierdurch konnte eine Aussage über das fließregulierende Potential der Nanomaterialien erhalten werden. Es zeigte sich, dass die Primärpartikelgröße, die Aggregatfestigkeit und der hydrophile/hydrophobe Charakter der jeweiligen Nanomaterialien einen entscheidenden Einfluss auf das fließregulierende Potential der Nanomaterialien besitzten.
Nach wie vor ist die Zahl der Malaria-Neuerkrankungen mit ca. 500 Millionen Menschen weltweit sehr besorgniserregend. Durch zunehmende Resistenzen der Erreger gegen zahlreiche Arzneimittel wird die Situation zusätzlich verschärft. Daher ist die Suche und Entwicklung neuartiger Medikamente wichtiger denn je. Die Natur ist immer noch das größte Reservoir an neuen Wirkstoffen, welche als Leitstrukturen für Arzneistoffe fungieren. In den letzten Jahren wurde eine große Zahl neuartiger, biologisch aktiver Naturstoffe identifiziert und hinsichtlich ihres Potenzials für eine pharmazeutische Weiterentwicklung analysiert. Die Naphthylisochinolin-Alkaloide gehören zu solch einer vielversprechenden Wirkstoffklasse, da sie sich v.a. durch ihre hervorragenden pharmakologischen Eigenschaften auszeichnen. Kürzlich wurden die ersten N,C-gekuppelten Naphthyldihydroisochinolin-Alkaloide, Ancistrocladinium A und B, entdeckt. Diese Verbindungen weisen als strukturelle Besonderheit eine außergewöhnliche Iminium-Aryl-Achse auf und besitzen zudem exzellente anti-infektive Aktivitäten, insbesondere gegen den Erreger der Orientbeule, Leishmania major. Ziel der vorliegenden Dissertation war die Synthese neuartiger sterisch gehinderter und strukturell vereinfachter Naphthylisochinoline für weiterführende Struktur-Aktivitäts-Beziehungen (SAR-Studien) und die stereochemische Analyse dieser Verbindungen. Zudem sollte eine Syntheseroute zu den neuartigen dimeren C,C-gekuppelten Naphthylisochinolin-Alkaloiden Shuangancistrotectorin A und B entwickelt werden.
The one electron oxidation potential of ten TAAs with all permutations of Cl , OMe- and Me-substituents in the three p-positions were determined by CV. The half wave potential of the first oxidation wave correlates linearly with the number of Cl- and OMe-substituents. AM1-CISD derived values of the absorption energies are in good agreement with the experiments but differ strongly for the oscillator strengths as well as for neutral compounds and their corresponding mono radical cations. The small solvent dependence of the experimental UV/Vis spectra in CH2Cl2 and MeCN reflects a minor charge transfer character of the electronic transitions. The UV/Vis/NIR spectra of the series of TAAs and their corresponding radical cations and the AM1 computations reveal that even small substituents may lead to strong symmetry breaking and to a modified electronic structure. The spectroscopic properties of a series of four bis-TAA donor-bridge-donor X-B-X dimers, composed of two asymmetric TAA chromophores (monomers) were investigated. UV/vis-, fluorescence and transient absorption spectra were recorded and compared with those of the corresponding X-B monomers. The excited states of the dimers are described as MV states which show, depending on the chemical nature of the bridge, a varying amount of interactions. It was found that superradiant emission only proceeds in the case of weak and medium coupling. Whether the first excited state potential energy surface of the dimers is a single minimum or a double minimum potential depends on the solvent polarity and the electronic coupling. In the latter case, the dimer relaxes in a symmetry broken CT state. The [2.2]paracyclophane bridged dimer is an example for a weakly coupled system, because the spectroscopic behavior is very similar to the corresponding p xylene monomer. In contrast, anthracene as well as p-xylene bridges mediate a stronger coupling and reveal a significant cooperative influence on the optical properties. A series of [2.2]paracylophane bridged bis-TAA MV radical cations X-B-X+ were analyzed by a GMH three-level model which takes two transitions into account: the IV-CT band and the bridge band. From the GMH analysis, one can conclude that the [2.2]paracyclophane moiety is not the limiting factor which governs the intramolecular charge transfer. The electronic interactions are of course smaller than direct conjugation but from the order of magnitude of the couplings of the [2.2]paracyclophane MV species it can be assumed that this bridge is able to mediate significant through-space and through-bond interactions. From the exponential dependence of the electronic coupling V between the two TAA localized states on the distance r between the two redox centers, it was inferred that the HT proceeds via superexchange mechanism. The analysis reveals that even significantly longer conjugated bridges should still mediate significant electronic interactions, because the decay constant of a series of conjugated MV species is small. The absorption properties of a series of bis-TAA-[2.2]paracyclophane dications X+-B-X+ were presented. The localized and the CT transitions of these dications are explained and analyzed by an exciton coupling model which also considers the photophysical properties of the monomeric TAA radical cations. Together with AM1-CISD calculated transition moments, experimental transition moments and transition energies of the bis-TAA dications were used to calculate electronic couplings by a GMH approach. These couplings are a measure for interactions of the excited MV CT states. The modification of the diabatic states reveals similarities of the GMH three-level model and the exciton coupling model. Comparison of the two models shows that the transition moment between the excited mixed-valence states of the dimer equals the dipole moment difference of the ground and the excited bridge state of the corresponding monomer. Thianthrenophane (1) has a cavity which offers enough room to potentially enable endohedral coordination to small ions or molecules. For the complexation of silver(I) perchlorate, the complex stability constants of thianthrenophane logK1=5.45 and of thianthrene logK2=9.16 were determined by UV/Vis titration. Single competition transport experiments with ten metal salts demonstrate a very high selectivity of thianthrenophane as a carrier for silver(I) and a distinctly higher transport rate compared to carriers such as thianthrene and 14-ane-S4. Although the X-ray crystal structure analysis of the polymeric [Ag(1)]ClO4 shows an exohedral coordination to silver(I), the formation of an endohedral [Ag(1)]+ complex is suggested to be the explanation for the unusual carrier selectivity of silver(I) by 1 in bulk liquid membrane.

The process leading to susceptibility for neoplasia is represented by the disintegration of gene systems that normally protect the fish from neoplasia. Hybridization is the most effective process that leads to disintegration of the protection gene systems. Environmental factors may complete disintegration and thus may trigger neoplasia. It is discussed whether the findings on Xiphophorus may also apply to humans.

The capacity of Xiphophorus to develop neoplasia can be formally assigned to a "tumor gene" (Tu), which appears to be a normal part of the genome of all individuals. The wild fish have evolved population-specific and cell type-specific systems of regulatory genes (R) for Tu that protect the fish from neoplasia. Hybridization of members of different wild populations in the laborstory followed by treatment of the hybrids with carcinogens led to disintegration of the R systems permitting excessive expression of Tu and thus resulting in neoplasia. Certain hybrids developed neoplasia even spontaneously. Observations on the genuine phenotypic effect of the derepressed Tu in the early embryo indicated an essential normal function of this oncogene in cell differentiation, proliferation and cell-cell communication. Tu appeared to be indispensable in the genome but may also be present in accessory copics. Recently, c-src, the cellular homolog of the Rous sarcoma virus oncogene v-src, was detected in Xiphophorus. The protein product of c-src, pp60c-src, was identified and then examined by its associated kinase activity. This pp60c-src was found in all individuals tested, but, depending on the genotype, its kinase activity was different. The genetic characters of c-src, such as linkage relations, dosage relations, expression, etc., correspond to those of Tu. From a systematic study which showed that pp60c-src was present in all metazoa tested ranging from mammals down to sponges, we concluded that c-src has evolved with the multicellular organization of animals. Neoplasia of animals and humans is a characteristic closely related to this evolution. Our data showed that small aquariurn fish, besides being used successfully because they are time-, space-, and money-saving systems for carcinogenicity testing, are also highly suitable for basic studies on neoplasia at the populational, morphological, developmental, cell biological, and molecular levels.
Untersuchungen zur 2H/1H- und 13C/12C-Isotopenfraktionierung bei der Biogenese von Aromastoffen

Für die Authentizitätsbewertung achiraler Aromastoffe ist die gaschromatographische Isotopenverhältnismessung mittels massenspektrometrischer Analyse ein etabliertes Verfahren. Diese Technik ermöglicht es, über geeignete Datenbanken authentischer Referenzproben gesicherte Aussagen hinsichtlich deren Herkunft aus natürlicher oder synthetischer Quelle zu treffen. Zunehmend ins Interesse rückt allerdings auch die Frage, ob es mittels Techniken der Stabilisotopenanalytik ebenso möglich ist, das breite Feld der legislativ als „natürlich“ deklarierten Aromastoffe analytisch weiter in deren Herkunft aus biotechnologischer oder natürlicher („ex plant“) Quelle aufzutrennen. Zwar kann dieser Fragestellung prinzipiell über die Erweiterung bestehender Stabilisotopen-Datenbanken mit authentischen Proben nachgegangen werden, sie scheitert jedoch häufig an der limitierten Verfügbarkeit authentischer biotechnologischer Referenzen oder der eingeschränkten Kenntnis über die der Produktion „natürlicher“ Aromastoffe zugrundeliegenden Verfahrenstechniken. Eine mögliche Vorgehensweise zur Umgehung dieses Sachverhalts stellt daher die in Anlehnung an beschriebene biotechnologische Verfahren im Labormaßstab durchgeführte Produktion ausgewählter und somit auch authentischer Referenz-Aromastoffe dar. Diese Methode hat zudem den Vorteil, dass gegebenenfalls zusätzliche Informationen über mögliche Isotopenfraktionierungen in solchen Systemen ermittelt werden können, welche sich nicht nur zur Authentizitätsprüfung als nützlich erweisen können, sondern auch zur stetig wachsenden Grunderkenntnis über Isotopenfraktionierungen in biologischen Systemen beitragen. Ziel der vorliegenden Arbeit war es daher, der geschilderten Fragestellung bezüglich ausgewählter Aromastoffe aus den Gruppen der C6-Aldehyde und -Alkohole („Grünnoten“) sowie der Gärungsalkohole nachzugehen. Zu diesem Zweck erfolgten zum einen im Labormaßstab die biogenetische Bildung von C6-Aldehyden und -Alkoholen ausgehend von den ungesättigten Fettsäuren Linol- und Linolensäure, ferner wurden parallel Edukte, Intermediate und Produkte isoliert und hinsichtlich ihrer Stabilisotopengehalte durch Bestimmung der Delta-2H(V-SMOW)- und Delta-13C(V-PDB)-Werte untersucht. Zum anderen sind auf fermentativem Wege ausgehend von unterschiedlichen Kohlenhydratquellen die Gärungsalkohole 2-Phenylethanol und 2-Methyl-1-propanol dargestellt worden. Des weiteren galt es, die bei den Gärungsalkoholen resultierende Datenlage dahingehend zu prüfen, ob sich diese über eine Korrelation der Delta-2H(V-SMOW)- und Delta-13C(V-PDB)-Werte dazu eignet, eine Authentizitätsbewertung dieser Aromastoffe hinsichtlich natürlicher oder synthetischer Herkunft zu ermöglichen.
Mittels Laserblitz-Photolyse wurden die Triplettlebenszeiten sowie die Löschraten der Triplettzustände verschiedener Acetophenonderivate durch dG, 8-oxodG, DNA, molekularen Sauerstoff und die Ketone selbst bestimmt. Für AP-OAc, AP und BP wurden Triplettlebensdauern von 7-9 µs gemessen, während die Triplettzustände von AP-OH und AP-OtBu aufgrund alpha Spaltung deutlich kurzlebiger waren (ca. 1 µs); die alpha Spaltung konnte EPR-spektroskopisch durch Spinabfangexperimente mit DMPO und TEMPO belegt werden. Im Fall von AP-OMe wurde weder dessen Triplettzustand noch die Bildung von Radikalen detektiert, was auf einer schnell ablaufenden Norrish-Typ-II-Spaltung beruht. Aufgrund dieses photochemischen Verhaltens wurden die Ketone (mit Ausnahme von AP-OMe) in zwei Gruppen klassifiziert, nämlich die „Gruppe A“-Ketone (keine Radikalbildung) und die „Gruppe B“-Ketone (Radikalbildner). Während die „Gruppe A“-Ketone gegenüber niedrigen Konzentrationen von DNA (62.5 µM) inaktiv waren, verursachten die bei der Bestrahlung der „Gruppe B“-Ketone generierten Peroxylradikale, neben wenigen direkt induzierten Strangbrüchen, hauptsächlich die Guaninoxidationsprodukte 8-oxoGua und guanidinfreisetzende Produkte (GRP). Erst wenn die DNA-Konzentration zehnfach erhöht wird (625 µM), tritt bei der Photolyse der „Gruppe A“-Ketone auch DNA-Oxidation durch einen Elektronentransfer von der Guaninbase auf das angeregte Keton ein. Ein analoger Konzentrationseffekt wurde auch in der dG-Oxidation beobachtet, bei niedrigen Substratkonzentrationen sind nur die radikalbildenden „Gruppe B“-Ketone aktiv. Die Tatsache, dass in der dG-Oxidation durch die „Gruppe A“-Ketone kein 8-oxodG detektiert wurde, wurde auf dessen effiziente Oxidation durch dG•+-Radikalkationen zurückgeführt. Die „Gruppe B“-Ketone sind in Abwesenheit von O2 gegenüber dG und DNA oxidativ inaktiv, da die in der alpha Spaltung generierten kohlenstoffzentrierten Radikale keine Peroxylradikale bilden können. Die „Gruppe A“-Ketone sind gegenüber DNA in Abwesenheit wie auch in Anwesenheit von Sauerstoff genauso reaktiv, da der Elektronentransfer von DNA zum Keton unabhängig von Sauerstoff ist. Um mechanistische Einblicke in die oxidative DNA-Schädigung zu erlangen, wurden photochemische Modellstudien mit dem Nukleosid dG sowie 8-oxodG durchgeführt, wobei zusätzlich Spiroiminodihydantoin gebildet wird. Bis vor kurzem wurde die Struktur dieses Oxidationsproduktes als 4-HO-8-oxodG angenommen, dass zuerst in der dG Oxidation mit Singulettsauerstoff (1O2) beobachtet wurde. Weder Spiroiminodihydantoin noch 4 HO-8-oxodG sind als authentische Verbindungen bekannt, so dass eine zweifelsfreie Strukturaufklärung die Bestimmung der Konnektivität der markierten Positionen erforderte. Diese Zuordnung erfolgte mittels eines SELINQUATE-NMR Spektrums, mit dem schlüssig die 4 HO-8-oxodG-Struktur ausgeschlossen wurde. Wie alle „Gruppe B“-Ketone sind auch alle „Gruppe A“-Ketone in Abwesenheit von O2 mit Ausnahme von AP-OAc gegenüber dG inert. Dies ist ein Beleg dafür, dass der Elektronentransferschritt von dG zum Keton in Abwesenheit von Sauerstoff (im Gegensatz zur DNA-Oxidation) reversibel ist und daher keine Oxidation möglich ist, wenn die Ketylradikale nicht durch O2 abgefangen werden. Das aus AP-OAc gebildete Ketylradikal besitzt als einziges einen effektiven unimolekularen Deaktivierungsweg, nämlich die Acetation-abspaltung, so dass die Reversibilität nicht mehr möglich ist.
Im Rahmen der vorliegenden Arbeit wurden Dihydroborane (H2BR) sowie Dihalogenborane (X2BR) mit Übergangsmetall-Lewis-Basen umgesetzt und die Reaktivität der auf diese Weise erhaltenen Übergangsmetall–Bor-Komplexe eingehend untersucht. So wurde eine Serie neuer Borylkomplexe des Typs trans-[Pt{B(Br)R‘}Br(PR3)2] dargestellt und mit Salzen schwach-koordinierender Anionen umgesetzt. Diese Studien sollten die Triebkraft für die Bildung kationischer Borylenkomplexe näher beleuchten. Die experimentellen Ergebnisse zeigen, dass eine Substitution in ortho-Position des borgebundenen Arylliganden für den notwendigen [1,2]-Halogenshift vom Bor- zum Platinzentrum und somit zur Realisierung einer Pt=B-Mehrfachbindung unabdingbar ist. Demnach reagieren Komplexe mit para-substituierten Arylliganden bei Halogenidabstraktion aus Borylkomplexen zu T-förmigen, kationischen Borylplatinkomplexen, während die Duryl-substituierten Analoga unter [1,2]-Halogenwanderung in kationische Borylenplatinkomplexe überführt werden. Neben dem Substitutionsmuster des borgebundenen Arylliganden wurde auch der Einfluss des Phosphanliganden untersucht.
Die Molekülstrukturen der Borylkomplexe 2 und 4 im Festkörper zeigen grundlegende Unterschiede im strukturellen Aufbau. Der Durylsubstituent ist in 2 im Vergleich zur (Ph-4-tBu)-Einheit in 4 deutlich aus der {Br2–Pt–B–Br1}-Ebene herausgedreht (2: Pt–B–C1–C2: 31.4(1); 4: 4.3(7)°), was vermutlich einen [1,2]-Halogenshift in 2 begünstigt. Die Pt–B-Bindungen der kationischen Borylenkomplexe 6 (1.861(5) Å) und 7 (1.863(5) Å) sind deutlich kürzer als im neutralen Borylkomplex 2 (2.004(4) Å), was ein eindeutiger Beleg für den Mehrfachbindungscharakter der Pt–B-Bindungen in 6 und 7 ist. Demzufolge scheint der sterische Anspruch des borgebundene Arylsubstituenten entscheidend für den Reaktionspfad bei Halogenidabstraktionen und somit für die Bildung kationischer Borylenplatinkomplexe zu sein, während diesen Studien zu Folge der Einfluss der Ligandensphäre am Platinzentrum eher eine untergeordnete Rolle spielt.
Des Weiteren gelang die Synthese der neuartigen heteroleptischen Platinkomplexe [Pt(cAACMe)(PiPr3)] (13) und [Pt(cAACMe)(PCy3)] (14) durch Umsetzung von [Pt(PCy3)2] und [Pt(PiPr3)2] mit dem cyclischen (Alkyl)(Amino)Carben cAACMe (Schema 34, A), bzw. durch Umsetzung von [Pt(nbe)2(PCy3)] (Schema 34, B) mit cAACMe. Die Darstellung des literaturbekannten homoleptischen Komplexes [Pt(cAACMe)2] (11) konnte durch Reaktion von [Pt(nbe)3] mit cAACMe deutlich vereinfacht werden bei gleichzeitiger Steigerung der Ausbeute (96%, Literatur: 79%). Die ungewöhnlich intensiv orangene Farbe dieser Verbindungsklasse geht laut DFT-Rechnungen auf die elektronische Anregung aus dem HOMO in das LUMO zurück, wobei hauptsächlich die π-Wechselwirkungen zwischen den Platin- und Carbenkohlenstoffatomen des cAACMe-Liganden beteiligt sind (DFT-Rechnungen von Dr. Mehmet Ali Celik). Auch in ihren strukturellen Eigenschaften sind sich 11 - 14 sehr ähnlich, wohingegen deutliche Unterschiede in deren Elektrochemie und Reaktivität beobachtet wurden. So konnte für 11 eine quasi-reversible Oxidationswelle (E1/2 = –0.30 V gegen [Cp2Fe]/[Cp2Fe]+ in THF) bestimmt werden, während die heteroleptischen Komplexe 13 und 14 (Epa = –0.09 V; –0.11 V) sowie deren Vorläufer [Pt(PCy3)2] und [Pt(PiPr3)2] (Epa = 0.00 V; +0.12 V) irreversible Oxidationswellen zeigen. Demnach kann 13 und 14 im Vergleich zu [Pt(PCy3)2] und [Pt(PiPr3)2] ein größeres Reduktionsvermögen zugeordnet werden. Reaktivitätsstudien zeigen, dass der homoleptische Komplex 11 inert gegenüber vielen Substraten wie z.B. Boranen, Diboranen(4) und Lewis-Säuren ist. Im Gegensatz dazu haben sich die heteroleptischen Komplexe 13 und 14 als deutlich reaktiver erwiesen, womit diese eine Mittelstellung zwischen 11 und der Spezies [Pt(PR3)2] einnimmt.
Die Umsetzung von [Pt(cAACMe)(PiPr3)] (13) mit BBr3 und Br2BPh lieferte die Borylkomplexe 18 und 19, welche vollständig charakterisiert wurden. Die Reaktivität von 13 und 14 gegenüber den Lewis-Säuren GaCl3 und HgCl2 zeigt ebenfalls Analogien zu der von Bis(phosphan)platinkomplexen. Reaktion mit GaCl3 führte hierbei zur Bildung der MOLP-Komplexe [(cAACMe)(PiPr3)Pt→GaCl3] (21) und [(cAACMe)(PCy3)Pt→GaCl3] (22), während die oxidative Addition der Hg–Cl-Bindung an das Platinzentrum von 14 im Komplex [PtCl(HgCl)(cAACMe)(PiPr3)] (23) resultierte. Die Synthese von 23 gelang auch durch Umsetzung mit Kalomel unter Abscheidung eines Äquivalentes elementaren Quecksilbers.
Ein weiterer Schwerpunkt dieser Arbeit lag auf der Übergangsmetall-vermittelten Dehydrokupplung von Dihydroboranen. Die Umsetzung von [Pt(cAACMe)(PiPr3)] (13) mit BBr3 und Br2BPh lieferte die Borylkomplexe 18 und 19, welche vollständig charakterisiert wurden. Die Reaktivität von 13 und 14 gegenüber den Lewis-Säuren GaCl3 und HgCl2 zeigt ebenfalls Analogien zu der von Bis(phosphan)platinkomplexen. Reaktion mit GaCl3 führte hierbei zur Bildung der MOLP-Komplexe [(cAACMe)(PiPr3)Pt→GaCl3] (21) und [(cAACMe)(PCy3)Pt→GaCl3] (22), während die oxidative Addition der Hg–Cl-Bindung an das Platinzentrum von 14 im Komplex [PtCl(HgCl)(cAACMe)(PiPr3)] (23) resultierte. Die Synthese von 23 gelang auch durch Umsetzung mit Kalomel unter Abscheidung eines Äquivalentes elementaren Quecksilbers.
Ein weiterer Schwerpunkt dieser Arbeit lag auf der Übergangsmetall-vermittelten Dehydrokupplung von Dihydroboranen. Vor Beginn dieser Reaktivitätsstudien wurde zunächst eine vereinfachte Syntheseroute für Dihydroborane entwickelt. Durch Umsetzung von Cl2BDur mit HSiEt3 konnte auf diese Weise der Syntheseaufwand deutlich verringert und die Ausbeute an H2BDur von 74% auf 98% deutlich gesteigert werden. Zur Dehydrokupplung wurden neben Gold-, Rhodium- und Iridiumkomplexen auch Platinkomplexe mit H2BDur umgesetzt. Die Untersuchungen mit Gold- und Rhodiumverbindungen erwiesen sich hierbei als erfolglos und die Umsetzung der Iridiumpincerkomplexe [(PCP)IrH2] 26 und 27 (tBuPCP, AdPCP) mit H2BDur lieferte die Boratkomplexe 28 und 29 mit κ2-koordinierten {H2BHDur}-Liganden. Analog konnte bei Umsetzung von 26 mit H2BThx der Boratkomplex 30 spektroskopisch beobachtet, jedoch nicht isoliert werden. Bei den Komplexen 28 - 30 handelt es sich um die ersten κ2-σ:σ-Dihydroboratkomplexe mit sterisch anspruchsvollen Arylsubstituenten. Neben den Iridiumpincerkomplexen wurde auch der Komplex [Cp*IrCl2]2 mit H2BDur umgesetzt. Die Bildung des Boratkomplexes 34 ist mit einem [1,2]-Shift eines Chloratoms von Iridium auf das Borzentrum verbunden.
Die Reaktivität von H2BDur gegenüber [Pt(PCy3)2] zeigte eine starke Abhängigkeit hängt von der Stöchiometrie. Bei der 1:1-Umsetzung konnten sowohl die farblosen Verbindungen trans-[(PCy3)2PtH2] und Cy3P→BH2Dur (48) isoliert werden, als auch die beiden dunkelroten Verbindungen [(Cy3P)3Pt3(2-B2Dur2)] (36) und [{(PCy3)Pt}4(2-BDur)2(4-BDur)] (37), kristallographisch untersucht werden.
Der B–B-Abstand im π-Diborenkomplex 36 (1.614(6) Å) deutet eindeutig auf die Gegenwart einer B=B-Doppelbindung hin, wobei das Diboren side-on gebunden an zwei der drei Platinatome des Pt3-Gerüsts koordiniert ist. Die Zusammensetzung von 36 und 37 konnte auch durch Elementaranalysen bestätigt werden.
Die Bildung von 36 und 37 deuten auch darauf hin, dass bei dieser Art der Dehydrokupplung multimetallische Wechselwirkungen eine wichtige Rolle für die Stabilisierung der borzentrierten Liganden spielen. So konnten bei der Reaktion von [Pt(PCy3)2] mit zwei Äquivalenten H2BDur neben Cy3P→BH2Dur (48) auch zwei weitere zweikernige Platinverbindungen isoliert und vollständig charakterisiert werden. Erhitzen der Reaktionslösung auf 68°C für 170 Minuten führte hierbei zur Bildung von [{(Cy3P)Pt}2(μ-BDur)(ƞ2:(μ-B)-HB(H)Dur)] (38) mit zwei verbrückenden borzentrierten Liganden, einem Borylen- (BDur) und einem Boranliganden (BH2Dur), welche im 11B{1H}-NMR Spektrum bei δ = 101.3 und δ = 32.8 ppm detektiert wurden. Die Röntgenstrukturanalyse von 38 lässt einen signifikanten σ-BH-Hinbindungsanteil des Boranliganden zu einem der Platinzentren vermuten, was einen anteiligen Pt2→B-Bindungscharakter andeutet. Dieser Befund konnte auch durch DFT-Rechnungen von Dr. William Ewing bestätigt werden.
Die Studien haben auch gezeigt, dass die Bildung von 38 über eine Zwischenstufe verläuft, den hypercloso-Cluster [{(Cy3P)HPt}2(μ-H){μ:ƞ2-B2Dur2(μ-H)}] (39) mit einer tetraedrischen {Pt2B2}-Einheit, zwei terminalen Pt–H-Bindungen sowie je einen die Pt–Pt- bzw. B–B-Bindung verbrückenden Hydridliganden. 39 erwies sich als anfällig gegenüber H2-Eliminierung und lagert bei Raumtemperatur innerhalb von Tagen, bzw. bei 68°C innerhalb einer Stunde unter B–B-Bindungsbruch quantitativ in 38 um, welche selbst keinen direkten Bor–Bor-Kontakt mehr aufweist.
Auf Grundlage der beschriebenen Resultate wurde zudem ein einfacher Zugang zu zweikernigen Platinkomplexen entwickelt. Demnach gelang es, den literaturbekannten zweikernigen Komplex [Pt2(μ:ƞ2-dppm)3] (50) (dppm = Ph2PCH2PPh2) durch Umsetzung von [Pt(nbe)3] mit dppm in guten Ausbeuten zu synthetisieren. Des Weiteren wurde die Reaktivität von 50 gegenüber verschiedenen Lewis-Säuren untersucht. Ein Großteil dieser Umsetzungen war mit der Bildung von schwer löslichen Feststoffen verbunden, weshalb lediglich bei der Reaktion mit Br2BPh und Br2BMes geringe Mengen an definiertem Produkt isoliert und durch Röntgenstrukturanalyse charakterisiert werden konnten. Demnach führte die Umsetzung von 50 mit Br2BPh oder Br2BMes zur oxidativen Addition beider B–Br-Bindungen an je eines der Platinzentren und der Bildung der verbrückenden Borylenplatinkomplexe 51 und 52. NMR-spektroskopische Studien deuteten eine analoge Reaktivität von Br2BDur und Br2BFc an, wobei die Komplexe 53 und 54 noch nicht vollständig charakterisiert werden konnten.
Es wird die Reaktivität eines hochgespannten und reaktiven [1],[1]-Disilamolybdänocenophanes gegenüber ungesättigten Substraten, E–H Funktionen und E–E-Bindungen untersucht. Die Produkte wurden mittels spektroskopischer Methoden in Lösung sowie im Festkörper identifiziert und charakterisiert. Weiterhin wird die Reaktivität eines [2]-Disilamolybdänocenophanes ebenfalls gegenüber ungesättigten Substraten und E–E-Bindungen sowie gegenüber Pt(0)-Verbindungen erforscht. Die erhaltenen Komplexe wurden sowohl im Festkörper, als auch in Lösung spektroskopisch untersucht und charakterisiert.
Oxygen-centered radicals are important intermediates in photobiological, mechanistic and synthetic studies. The majority of precursors of reactive oxyl radicals are labile and thus delicate to handle. Therefore N-(alkoxy)-pyridinethiones and N-(Alkoxy)-thiazolethiones have attracted attention as "mild'' photochemical source of alkoxyl radicals, in the last few years. A disadvantage of the pyridine compounds, is their sensibility to daylight. Despite of their similarities, both molecules behave surprisingly different, if photolyzed in the absence of trapping reagents. The pyridinethione compounds undergo highly efficient radical chain reactions under such conditions while the corresponding thiazolethiones react surprisingly sluggish and give rise to several unwanted side products. The properties of both compounds should be understood and optimized in the frame of this work. Additionally new compounds should be suggested that can also be applied in the photochemical alkoxyl radical generation. Some background information about the generation and application of alkoxyl radicals is provided in chapter 2. Electronic excitations and UV/vis spectroscopy together with a description of quantum chemical approaches that are able to calculate such phenomena are outlined in chapter 3. Chapter 4 deals with the description of the vertical excitation spectra. During the validation CASSCF, CASPT2, TD-DFT and RI-CC2 were tested with respect to their ability to describe the vertical excitations in both compounds. The CASPT2 approach gives accurate descriptions of the electronic excitation spectra of all compounds. The time-dependent DFT results are very sensitive on the choice of the functional and a validation of the results should be always done. On the basis of these computations the spectroscopic visible absorption bands of both compounds were assigned to a pi-->pi* transition in the thiohydroxamic acid functionality. In chapter 5 the mechanism of the thermally and the photochemically induced N,O homolysis in both compounds is unveiled. The near UV-induced N,O homolysis will start from the S2 state. The expected relaxation from the S2- to the S1-state and the dissociation process is expected to be very fast in the case of the thiazolethione compound. The potential surfaces of the pyridine compound in contrast point to a slower N,O bond dissociation. Due to the resulting faster dissociation process the excess energy which results from the photochemical activation is quenched only to small amounts. The maximal possible excess energy of the fragments is lower and a quenching is much more likely in the case of the pyridinethione compounds. This explaines the different reactivities of both compounds. For the also already successfully applied precursor system N-(alkoxy)-pyridineones the computed dissociation paths show courses that clearly predict a slow bond dissociation process. Chapter 6 deals with the tuning of the initial excitation wave length of the known pyridinethiones und thiazolethiones. In the first part the effects of substituents on the thiazolethione heterocycle was examined. The UV/vis spectra of 4 and 5 substituted thiazolethiones can be interpreted like the spectrum of the parent compound. The second part of chapter 6 deals with the identification of a substitution pattern on the pyridine heterocycle which induces a blue shift of the photo active band. The computations showed that electron rich and electron poor substituents result the same effects on the electronic excitation spectra. These substituent effects are additive, but the steric orientation of the substituents has to be taken into account. Chapter 7 describes a computer aided design of new alkoxyl radical precursors. Combining the advantages of both compounds the radical formation should be initiated by an irradiation with light at about 350 nm, and the amount of side products during the radical formation process should be small. To achieve this 18 test candidates were obtained by a systematic variation of the parent compound of the thiazolethione precursor. To identify the promising new precursor systems a screening of the lower electronic excitations of all resulting 18 systems was performed with TD-DFT. For promising systems the N,O or P,O dissociation paths, respectively, were analyzed according to the developed model. N-(methoxy)-azaphospholethione and N-(methoxy)-pyrrolethione seem to be the most promising candidates. The computations predict a strong absorption at about 350 nm respectively 320 nm. Due to the amounts of maximal excess energy and the shapes of the potential surfaces of the N,O bond dissociation paths their reactivity should resemble more the behavior of the pyridinethiones.
This thesis describes the inclusion of dynamical effects in the theoretical calculation of Electron Paramagnetic Resonance (EPR) spectroscopic parameters. The studies were performed using Density Functional Theory (DFT) methodology and a perturbation-theoretical approach to g-tensor calculations. Hydrogen atoms trapped in octasilasesquioxane cages display unexpectly high, positive g-values. Computational simulation of these systems successfully reproduced the positive g-values and found them to arise from spin-orbit coupling around the oxygen nuclei. Dynamical effects were estimated by calculating the potential well in which the hydrogen atom moves. Semiquinone radical anions are important bioradicals that play a role in photosynthesis and respiration. The simplest and most prototypical, benzosemiquinone anion, was simulated both in the gas phase and in aqueous solution by Car-Parrinello Molecular Dynamics (CPMD). The neutral benzoquinone was also simulated for comparison. The solvation environments of both the anionic and neutral molecules were analysed and compared. EPR parameters were calculated for the semiquinone, providing the first example of full inclusion of dynamic effects in g-tensor calculation. The effects of different solvation interactions on the g-tensor and hyperfine interactions were extensively examined. Additionally, static calculations (i.e., calculations not incorporating any dynamical effects) were performed. Comparison between these (and prior computational studies) and the dynamical system allowed an assessment of the effects of dynamics on solvation and EPR parameters. Ubisemiquinone radical anion, one of the most widely-occurring semiquinone radicals, was simulated in the aqueous phase using CPMD. The solvation environment was analysed and EPR parameters were calculated. The motion of the side-chain, and its effects on solvation and EPR parameters, were examined.
Bei N-Acyl-Ethanolaminphosphaten handelt es sich um eine bislang wenig untersuchte Klasse polarer Substanzen, deren Erforschung aufgrund ihrer strukturellen Analogie zu apolaren, physiologisch wirksamen N-Acyl-Ethanolaminen von Interesse ist. Zu bear-beiten waren analytische Fragestellungen, die auch synthetische Aufgaben beinhalteten, wie Methodenentwicklung und Versuche zur Erfassung von N-Acyl-Ethanolamin-phosphaten in ausgewählten Lebensmitteln sowie strukturelle Studien zur „Bioaktivität“ der Verbindungen. Erstes Ziel der vorliegenden Arbeit war es demzufolge, eine geeig-nete Methode für deren qualitative und quantitative Analytik zu entwickeln. Gleichzei-tig wurden ausgewählte N-Acyl-Ethanolaminphosphate synthetisiert. Aufgrund des literaturbekannten Vorkommens von N-Acyl-Ethanolaminen in Wein wurden für die Lebensmitteluntersuchungen fermentierte Produkte, d.h. drei verschie-dene Sake (Japanischer Reiswein) und ein fermentierter Rotkohl verwendet. Parallel zu diesen Untersuchungen erfolgten auch Studien zur Stabilität der N-Acyl-Ethanolamin-phosphate. Versuchsreihen zur Überprüfung potentieller „Bioaktivität“ umfassten Studien mit al-kalischer Phosphatase, PhospholipaseA2, Lipoxygenase, Xanthinoxidase, β-N-Acetyl-hexosaminidase und dem Cannabinoidrezeptor-1.
Synthesis of (RS)-5-amino-3-aryl (methyl)-pentanoic acid hydrochlorides, 3 aminomethyl-5-chloro-benzoic acid hydrochloride and (RS)-4-amino-3-(4`-ethynyl(iodo)-phenyl)-butanoic acid hydrochlorides have been accomplished. The aim of their synthesis was to evaluate their GABABR agonist activity and to derive a model which will correlate their structure with the observed pEC50. The GABABR agonist activity of the prepared compounds has been determined in functional assay based on calcium measurement in vitro using tsA cells transfected with GABAB1b/GABAB2/Gαq-z5. Reviews on the neurotransmitter receptors (ligand-gated ion channel receptors and G protein-coupled receptors), their agonists and antagonists have been given in the general part of this work. A detailed discussion on the strategy followed for the synthesis of the designed compounds as well as the starting materials and intermediates has been described and illustrated in Schemes 2-6. The synthesized compounds were evaluated for their GABABR agonist activity. Furthermore, these compounds were docked in the available 3D homology model of GABABR using the program FlexiDock implemented in SYBYL software. Subsequently, we derived a predictive model which correlates the experimentally determined pEC50 with the calculated binding energy of certain baclofen analogues and homologues. In addition, we used the program DISCO (DIStance COmparisons) implemented in SYBYL software to find the pharmacophore features of GABAB agonists.
Die vorliegende Arbeit liefert einen Beitrag zur Synthese und zum Reaktionsverhalten enantiomerenangereicherter Silyllithium- und Silylmagnesiumverbindungen. Dabei sind mehrere Punkte von Bedeutung: die Aufklärung der Struktur im Festkörper wie in Lösung, die Bestimmung der Selektivitäten und des stereochemischen Verlaufs hoch enantiomerenangereicherter polarer Silylmetallverbindungen in Umsetzungen mit Elektrophilen, die konfigurative Stabilität hoch enantiomerenangereicherter Silyllithium- und Silylmagnesiumverbindungen sowie die Aufklärung ungewöhlicher NMR-Verschiebungen. Im Rahmen dieser Arbeit durchgeführte quantenchemische Berechnungen haben sich vor allem auf die beiden letzten Punkte konzentriert. Insgesamt wurden folgende Themenkomplexe bearbeitet: 1.) Synthese einer hoch enantiomerenangereicherten Silyllithiumverbindung 2.) Reaktionsverhalten der hoch enantiomerenangereicherten Silyllithiumverbindung 2 gegenüber Elektrophilen 3.) Konfigurative Stabilität der hoch enantiomerenangereicherten Silyllithiumverbindung 2 4.) Weitere Untersuchungen zur Darstellung von Silyllithiumverbindungen 5.) Untersuchungen zur Struktur von Silyllithiumverbindungen im Festkörper und in Lösung 6.) Quantenchemische Analysen der Ursachen der NMR-Verschiebungen von Disilenen und heteroatom-substituierten Silanen
Zeil dieser Arbeit war die Identifikation von Proteinen, die mit Bestandteilen des für die DNA-Replikation essentiellen präreplikativen Komplexes in der Maus wechselwirken. Hierbei konnten Interkationen des Heterochromatin Proteins 1a mit den Replikationsfaktoren ORC1, ORC2 und CDC6 sowohl in Two Hybrid-Studien als auch in Immunpräzipitationen gezeigt werden. Darüberhinaus konnten signifikante Kolokalisationen dieser Proteine mit HP1a an heterochromatischen Regionen in murinen NIH3T3-Zellen nachgewiesen werden. Ebenfalls konnte hier erstmals eine Lokalisation von HP1a am Centrosom demonstriert werden. Ein siRNA-vermittelter Knock Down von HP1a zeigte jedoch keinen direkten Einfluß auf die Replikation. Es konnte hingegen gezeigt werden, daß ein Knock Down von HP1a in signifikatnen Defekten in der Cytokinese und einer deutlich verlangsamten Zellproliferation resultiert. So konnten häufig multinukleäre Zellen und eine Arretierung in der G1-Phase beobachtet werden. Weiterhin wurde der Einfluß der Phosphorylierung von HP1a durch die Casein Kinase II mithilfe von Phosphorylierungsmutanten untersucht. Im Gegensatz zu Drosophila-Zellen zeigten sich in murinen Zellen jedoch keine Auswirkungen dieser Mutationen auf die Lokalisation von HP1a an Heterochromatin. Wieterhin konnten Interaktionen des Replikationsinhibitors Geminin mit den Replikationsproteinen ORC1, ORC2 und CDC7 sowohl im Two Hybrid-System als auch in Immunpräzipitationen gezeigt werden, die unterschiedliche Zellzyklusabhängigkeiten aufwiesen. In murinen NIH3T3-Zellen zeigte eine Knock Down von Geminin jedoch im Gegensatz zu anderen Zellinien keinen Einfluß auf die Replikation. In weiteren Teilen dieser Arbeit konnten Interaktionen des Retinoblastoma Proteins mit ORC2 und MCM7 sowohl in Two Hybrid- als auch in Immunpräzipitations-Experimenten gezeigt werden. Darüberhinaus wies Pescadillo Interaktionen mit den Replikationsproteinen ORC2, ORC6, MCM2, MCM3, MCM6 und CDC45 im Two Hybrid-System und Interaktionen mit MCM2 und MCM3 in Biolumineszenz-Resonanzenergietransfer-Experimenten auf. Eine Kolokalisation von Pescadillo und ORC6 in den Nukleoli läßt auf eine Funktion beider Proteinen bei der Ribosomen Biogenese schließen. Es konnten ebenfalls Interaktionen der Untereinheit E1 des humanen Papillomavirus Subtyp 11 mit den Replikationsfaktoren ORC2,3,4,5,6, MCM2, MCM3, MCM6, CDC6, CDC7, CDT1, HP1a, Rb und Pescadillo im Two Hybrid-System beobachtet werden.
The enhancement of electronic and optical properties of semiconductor nanostructures is known as a direct consequence of the spatial confinement of carriers. However, the physics of quantum confinement is still not entirely understood. This work focuses on a qualitative study of quasi-zero dimensional II-VI semiconductor nanostructures (quantum dots QDs). In particular, commercially available as-received and heat treated CdSxSe1-x QDs embedded in a dielectric matrix were investigated by means of linear and nonlinear spectroscopy techniques. Low wavenumber Raman in off-resonance scattering regime was applied in order to obtain key-properties of the nanocrystals, such as the QD's size and the distribution of the QD's size inside the inhomogeneous broadening. Moreover, by careful selection of the polarization geometries, different acoustic vibrational modes could be evidenced. In comparison to the bulk, 3D confinement of carriers leads to modifications in the energy distribution in a QD and as a consequence, the intensity of the acoustical phonons is enhanced. However, only 2 acoustic vibrational modes (labelled l=0 and l=2) are Raman-active, which were selectively excited using linear polarized laser light in parallel- and cross-polarized excitation geometries. The QD's size was determined using the dependence of the frequency of the acoustic vibrational mode on the diameter of the vibrating particle, whereas the QD's size distribution was estimated from the normalized full width at the half of the maximum (FWHM) of the symmetric acoustic vibrational mode. In order to study relaxation mechanisms, which in quantum confined systems occur on a ps time scale, ultrafast spectroscopy techniques using laser pulses in the fs range must be employed. To this purpose, fs-FWM and fs-PPT measurements were performed on CdS0.6Se0.4 QDs of 9.1 nm in diameter, embedded in a glass matrix. The laser pulses employed in these experiments were circularly polarized, careful selection of the polarization geometries making different nonlinear processes available to study. It was shown that the relaxation of polarization selection rules depend strongly on the symmetry of the nanocrystals under discussion. The investigated nanocrystals belong to the symmetry group C2v or lower and their hexagonal crystal shape could be evidenced. The relaxation of selection rules was explained in the framework of the 4-level system, including a ground state, two exciton states and a biexcitons state. The appearance of FWM and PPT signals in forbidden polarization geometries was shown to be due to exciton state splitting due to lowering of the QD’s symmetry and due to the strong Coulomb interaction between carriers belonging to the same nanocrystal. Moreover, the significant difference in the origin of the gratings created by two pulses having the same and opposite polarizations, respectively. The intensity of the FWM signals should be the square of the intensity of the PPT signals and therefore the PPT measurements were employed as a check method for the results yielded by the FWM technique. The efficiency of circularly polarized femtosecond FWM spectroscopy techniques was proved once more in the investigation of heat treated CdSe QDs embedded in a dielectric matrix. The role of non-phonon energy relaxation mechanisms in the exciton ground and excited state of the QDs ensemble was extensively studied. Moreover, the dependence of the crystal shape asymmetry on the particle size and on the growth conditions could be estimated. It was shown, that the most efficient procedure to grow high quality nanocrystals is a longer heat treating at lower temperatures. In this case, the particles have more time to "nucleate" and to adopt a more "symmetric" shape. Further, the relaxation of excitons was extensively investigated. It was shown, that the electron intraband dynamics depend strongly on the Coulomb interaction between electrons and holes. Even at low excitation density, the Auger processes cannot be ignored. Auger autoionization of excitons followed by capture of carriers in surface states and deep traps in the dielectric matrix slow down the exciton relaxation process leading to an exciton lifetime ranging on a ps time scale. The relaxation of excitons from higher lying energy levels occurs also on two paths. At the beginning of the relaxation process (t31 < 400 fs), Auger-like thermalization of carriers is responsible for relaxation of the electron from 1pe into its 1se state, while the hole relaxes rapidly through its dense spectrum of states in the valence band. This process is immediately followed by capturing of carriers in deep traps, situated at the semiconductor-dielectric heterointerface. The traps are a consequence of the QD's asymmetry: the more and the deeper the traps, the higher the asymmetry of the nanocrystals (the band offset  is larger). This work presents a complete characterization of CdSSe QDs embedded in a glass matrix. The most important properties of the nanocrystals like QD's size and size distribution inside the inhomogeneous broadening were determined by means of low wavenumber Raman spectroscopy. In order to draw a full picture of these nanoparticles further complementary nonlinear spectroscopy techniques were used. Invaluable conclusions were available as a result of TI-FWM techniques applied in the framework of transient grating on 3D confined nanocrystals embedded in a glass matrix. The polarized the TI-FWM measurements were successfully performed on different QDs ensembles in order to determine symmetry properties and to describe the ultrafast relaxation mechanisms. This work brings additional contribution concerning the preparation of high quality QDs by presenting the effect of different growth conditions on the QDs symmetry, thus indicating a way for efficient manufacturing of nanocrystals.
A modular synthesis of both difurooxa‐ and difuroazadiborepins from a common precursor is demonstrated. Starting from 2,2′‐bifuran, after protection of the positions 5 and 5’ with bulky silyl groups, formation of the novel polycycles proceeds through opening of the furan rings to a dialkyne and subsequent re‐cyclization in the borylation step. The resulting bifuran‐fused diborepins show pronounced stability, highly planar tricyclic structures, and intense blue light emission. Deprotection and transformation into dibrominated building blocks that can be incorporated into π‐extended materials can be performed in one step. Detailed DFT calculations provide information about the aromaticity of the constituent rings of this polycycle.
Die vorliegende Arbeit beschreibt die Entwicklung von SiO2-basierten nano- und mikroskalig strukturierten Schichten, welche eine Temperaturstabilität > 500 °C sowie eine Strukturtreue > 90 % aufzeigen. Diese wurden unter Verwendung von hybridpolymeren, partikelmodifizierten Sol-Gel-Vorstufen via UV-basierter Nanoimprintlithographie (UV-NIL) hergestellt. Der Einfluss chemischer und verfahrenstechnischer Syntheseparameter auf die anorganische Netzwerkbildung der flüssigen Vorstufe (hybridpolymerer Binder), die Polymerisation und Verdichtung der hybridpolymeren Sol-Gel-Schichten während der thermischen Härtung sowie deren Strukturierung via UV-NIL wurden im Detail untersucht und die erhaltenen Zusammenhänge mit den resultierenden Materialeigenschaften korreliert. Dabei dienten die Kenntnisse hinsichtlich des Schrumpf- und Verdichtungsverhaltens von planaren Schichten als Basis für die daraus hergestellten, strukturierten Schichten.
Für die Lösung der quantenmechanischen Bewegungsgleichungen, die komplexe, molekulare Systeme beschreiben, sind effiziente und verlässliche Näherungsverfahren erforderlich. Die Dichtefunktionaltheorie (DFT) stellt für die Behandlung der Elektronenwechselwirkung in vielen Fällen den besten Kompromiss zwischen Effizienz und Genauigkeit dar. Im Rahmen der DFT wird die gesamte nicht-klassische Elektron-Elektron-Wechselwirkung im so genannten Austausch-Korrelationsfunktional angenähert. Viele solcher Näherungen sind semi-empirischer Natur, andere wurden ausschließlich von physikalischen Überlegungen abgeleitet. In globalen Hybridfunktionale wird ein konstanter Anteil der integrierten DFT-Austauschenergiedichte durch exakten Austausch aus der Hartree-Fock Näherung ersetzt. Das populärste Funktional B3LYP enthält 20 % exakten Austausch und mehrere empirische Parameter. Der optimale Prozentsatz hängt allerdings sehr stark von den zu berechnenden Systemen und molekularen Eigenschaften ab. Eine Lösung dieses Problems sollten lokale Hybridfunktionale liefern, in denen die Beimischung der exakten Austauschenergiedichte über eine lokale Mischfunktion (LMF) gesteuert wird und daher positions- und molekülabhängig ist. In dieser Arbeit wird ein semi-empirischer Ansatz für die Entwicklung neuer lokaler Hybridfunktionale verfolgt: während die Energiedichten unverändert aus etablierten Näherungen zum Austauschkorrelationsfunktional übernommen werden, stehen parametrisierte LMFs im Zentrum der Untersuchungen. Die verschiedenen LMFs beinhalten neben mindestens einem empirischen Parameter eine Variable die vom Quotienten der von-Weizsäcker kinetischen Energiedichte und der korrelierten kinetischen Energiedichte (sogenannte t-LMFs) bzw. dem reduzierten Dichtegradienten (bezeichnet als t-LMFs) abhängt. Weitere LMFs werden durch zusätzliche Berücksichtigung der Spinpolarisation erhalten. Alle Parameter werden an Atomisierungsenergien bzw. Reaktionsbarrieren bekannter molekularer Testsätze gefittet. Durch Visualisierung der LMFs können zusätzlich Einblicke in den physikalischen Hintergrund und in Möglichkeiten der Weiterentwicklung gewonnen werden. Es wurde beispielsweise beobachtet, dass entlang einer gedehnten Bindung höhere Werte der LMF und damit größere Beimischungen exakter Austauschenergie in Übergangszuständen einhergehen. Dieser Effekt ist für t-LMFs am ausgeprägtesten und korreliert mit besseren Ergebnissen für Reaktionsbarrieren mit lokalen Hybridfunktionalen, die auf einer t-LMF basieren. Bis auf wenige Ausnahmen leiten sich die lokalen Hybridfunktionale in dieser Arbeit aus dem Austausch- und Korrelationsfunktional der lokalen Dichtenäherung (LSDA) ab und enthalten keine Gradientenkorrektur im Sinne der GGA (generalized gradient approximation). Die neuen Funktionale wurden zunächst nicht-selbstkonsistent in eine Entwicklerversion des quantenchemischen Programmpaketes Turbomole implementiert. Das bedeutet, für gegebene Molekülorbitale bzw. eine gegeben Elektronendichte kann lediglich die Gesamtenergie berechnet werden. Dies ist eine anerkannte Näherung, die vor allem für die Optimierung der Parameter eine große Zeitersparnis darstellt. Um letztlich orbitalabhängige, molekulare Eigenschaften berechnen zu können wird neben der Gesamtenergie auch noch das zugehörige lokale Hybridpotential benötigt. Für die selbstkonsistente Implementierung wird die funktionale Ableitung der Austauschkorrelationsenergie nach den Orbitalen bestimmt. Daraus resultierend müssen neben den üblichen lokalen Austauschkorrelationspotentialtermen auch Integrale berechnet werden, die das mit der LMF gewichtete nicht-lokale exakte Austauschpotential enthalten. Die entsprechenden Terme kann man, genauso wie die exakte Austauschenergiedichte an sich, nicht analytisch berechnen. Früheren Ansätzen folgend wurden sie in der vorliegenden Arbeit in einer Basissatzentwicklung angenähert, wobei der Einfachheit halber die atomaren Basisfunktionen verwendet wurden. Um die Genauigkeit dieser sogenannten RI (resolution of the identity)-Näherung validieren zu können und auch schon im Hinblick auf die Anpassung einer Hilfsbasis, wurde darüber hinaus die numerische Berechnung aller Integrale, die das exakte Austauschpotential und die entsprechende Energiedichte enthalten, implementiert. Unter Verwendung der RI-Näherung ist der Rechenaufwand lokaler Hybride vergleichbar mit dem globaler Hybridfunktionale: Während die formale Skalierung in Abhängigkeit der Systemgröße gleich ist, ergab sich ein etwas höherer Vorfaktor für die lokalen Hybride. Verschiedene Literaturbekannte Testsätze mit Atomisierungsenergien, Reaktionsbarrieren, Dissoziationsenergien oder Gleichgewichtsabständen, die teilweise einige Schwächen bisheriger Dichtefunktionalnäherungen aufdecken, wurden berücksichtigt. Für die 223 Atomisierungsenergien des G3 Testsatzes stellen alle unsere Funktionale eine signifikante Verbesserung gegenüber B3LYP dar. Atomisierungsenergien sind insofern ein sensibler Test, da alle Bindungen gebrochen werden und Fehlerkompensation eine untergeordnete Rolle spielt. Vor allem lokale Hybridfunktionale, deren LMFs neben der kinetischen Energiedichte explizit von der Spinpolarisation abhängen, lieferten hervorragende Resultate. Obwohl im Vergleich zu Atomisierungsenergien für die korrekte Berechnung von Reaktionsbarrieren im Allgemeinen mehr exakter Austausch benötigt wird, sind unsere Funktionale auch für zwei Testsätze mit jeweils 38 Reaktionsbarrieren besser als B3LYP. Zwar kann mit einem globalen Hybrid mit 50 % exaktem Austausch eine geringere Abweichung von den Richtwerten erzielt werden, aber ein solches Funktional ist für thermochemische Daten unzureichend. Hier wurde erstmals gezeigt, dass lokale Hybridfunktionale ohne Gradientenkorrektur sowohl für Thermochemie als auch für Kinetik zufrieden stellende Ergebnisse liefern können. Das Dissoziationsverhalten symmetrischer Radikalkationen stellt für die hier diskutierten Dichtefunktionale nach wie vor eine Herausforderung dar: Die Dissoziationsenergien von sieben Modellsystemen werden mit unseren Funktionalen stark überschätzt und Gleichgewichtsabstände unterschätzt. Insgesamt sind die Werte nur marginal besser als mit B3LYP. Neben Eigenschaften von Hauptgruppenverbindungen wurden zudem Übergangsmetalldimere und -monohydride untersucht. Für erstere ist eine gute Beschreibung dynamischer sowie statischer Elektronenkorrelation ausschlaggebend. In den Hydriden andererseits dominiert mit gängigen Dichtefunktionalen die unphysikalische Selbstwechselwirkung eines Elektrons mit sich selbst. Für die 3d-Übergangsmetalldimere sind die getesteten Funktionale genauso gut wie B3LYP und für die Hydride etwas besser. Atomare s-d Transferenergien von 3d Übergangsmetallen verbleiben auch für unsere lokalen Hybridfunktionale, die insgesamt schlechtere Ergebnisse erzielen als B3LYP, noch problematisch. Das hierfür geeignetste lokale Hybridfunktional basiert auf einer s-LMF und beinhaltet LYP Korrelation. Für die isotropen Hyperfeinkopplungskonstanten (HFCCs) kleiner Hauptgruppenverbindungen wurden zufriedenstellende Ergebnisse (ähnlich wie B3LYP) mit einem t-LMF basierten lokalen Hybrid erzielt. Die RI Näherung zum lokalen Hybridpotential wurde dem numerisch exakten Potential für die Berechnung von Gesamtenergien, isotrope HFCCs und Orbitalenergien für verschiedene Basissätze gegenübergestellt. Wie erwartet ist der Fehler für Gesamtenergien mit der RI-Näherungen vergleichsweise gering, vor allem relativ zu den verbleibenden Abweichungen von experimentellen Energien. Der Vergleich der mittleren absoluten Abweichung von experimentellen Werten für 26 isotrope HFCCs zeigt sogar für mittelgroße und kontrahierte IGLO Basissätze nur geringe Unterschiede zwischen dem RI-Potential und dem numerisch exakten lokalen Hybridpotential. Die Analyse der HFCCs einzelner Moleküle und der Orbitalenergien des CN Moleküls offenbart allerdings, dass Ungenauigkeiten aufgrund der RI-Näherung hier eine größere Rolle spielen, vor allem wenn zu kleine atomare Basissätze verwendet werden. Von den untersuchten lokalen Hybriden stellen sich einige als hervorragende Kandidaten für die Berechnung thermochemischer und kinetischer Eigenschaften heraus. Jeweils unterschiedliche Funktionale erzielen darüber hinaus mit den besten bekannten Funktionalen vergleichbare Ergebnisse für isotrope Hyperfeinkopplungskonstanten und ausgewählte Eigenschaften kleiner Übergangsmetallverbindungen. Die in dieser Arbeit präsentierten lokalen Hybridfunktionale stellen daher einen wichtigen Schritt in der Entwicklung universeller Näherungen zum Austauschkorrelationsfunktional dar. Zur akkuraten Beschreibung molekularer Eigenschaften von Übergangsmetallkomplexen und dem Dissoziationsverhalten von Radikal-Kation-Dimeren neben Thermochemie und Kinetik, werden in Zukunft wohl komplexere LMFs benötigt. Um konkurrenzfähige lokale Hybride mit gradientenkorrigierter Austausch- und Korrelationsenergiedichte zu entwickeln, müssen darüber hinaus weitere Studien zum Einfluss des abweichenden Eichursprungs der miteinander kombinierten Austauschenergiedichten durchgeführt werden. Eine andere Möglichkeit ist die Entwicklung speziell abgestimmter Korrelationsfunktionale für lokale Hybride. Außerdem sollte die Qualität der RI-Näherung zum lokalen Hybridpotential detaillierter untersucht werden. Hierfür könnten zum Beispiel Ionisierungsenergien und Elektronenaffinitäten herangezogen werden. Um zusätzliche Abweichungen oder sogar fälschlicherweise "zu gute" Ergebnisse bei Validierungsrechnungen zu vermeiden, sollten Hilfsbasen für die Entwicklung des nicht-lokalen exakten Austauschpotentials implementiert und optimiert werden. Einer der nächsten Implementierungsschritte sollte auch Gradienten bezüglich der Kernkoordinaten beinhalten, um die Validierung der neuen lokalen Hybridfunktionale auf Strukturoptimierungen auszuweiten.
The thesis consists of two major parts. The first part contains a theoretical-experimental study of confocal micro-Raman spectroscopy on hybrid polymer coatings and an application of this spectroscopic method on PDMS-membranes. The theoretical-experimental study includes the application of a model that describes the influence of the refraction effect on the focus length on confocal Raman experiments, and the development of a new model that additionally takes into account the effect of diffraction on the focus dimensions. A parallel comparison between these two theoretical approaches and experimental data has been also drawn and a better agreement between theory and experiment was observed, when both refraction and diffraction effects were considered. Further, confocal resonance micro-Raman spectroscopy has been applied to characterise the diffusion processes of pharmacologically relevant molecules (b-carotene dissolved in dimethylsulfoxide) through a polydimethylsiloxane (PDMS)-membrane. The diffusion rate as a function of the measurement depth and diffusion time as well as the concentration gradient under a steady flux have been determined. The measurements shown that the confocal micro-Raman technique is a powerful tool to investigate the kinetics of diffusion processes within a membrane before the steady state has been reached. The second part of the thesis contains infrared and Raman spectroscopic studies of copper and iron doped B2O3-Bi2O3 glass systems. These studies were performed to obtain specific data regarding their local structure and the role played by dopant ions on boron and bismuthate units. The changes of B2O3 and Bi2O3 structural units due to the relaxation of the amorphous structure, which was induced in these samples by the thermal treatment, were also evidenced.
Novel dyes were prepared by simple “click CuAAC” attachment of a triarylborane–alkyne to the azide side chain of an amino acid yielding triarylborane dye 1 which was conjugated with pyrene (dye 2) forming a triarylborane–pyrene FRET pair. In contrast to previous cationic triarylboranes, the novel neutral dyes interact only with proteins, while their affinity to DNA/RNA is completely abolished. Both the reference triarylborane amino acid and triarylborane–pyrene conjugate bind to BSA and the hDPP III enzyme with high affinities, exhibiting a strong (up to 100-fold) fluorescence increase, whereby the triarylborane–pyrene conjugate additionally retained FRET upon binding to the protein. Furthermore, the triarylborane dyes, upon binding to the hDPP III enzyme, did not impair its enzymatic activity under a wide range of experimental conditions, thus being the first non-covalent fluorimetric markers for hDPP III, also applicable during enzymatic reactions with hDPP III substrates.
