Background: Animal models have implicated an integral role for coagulation factors XI (FXI) and XII (FXII) in thrombus formation and propagation of ischemic stroke (IS). However, it is unknown if these molecules contribute to IS pathophysiology in humans, and might be of use as biomarkers for IS risk and severity. This study aimed to identify predictors of altered FXI and FXII levels and to determine whether there are differences in the levels of these coagulation factors between acute cerebrovascular events and chronic cerebrovascular disease (CCD). Methods: In this case-control study, 116 patients with acute ischemic stroke (AIS) or transitory ischemic attack (TIA), 117 patients with CCD, and 104 healthy volunteers (HVs) were enrolled between 2010 and 2013 at our University hospital. Blood sampling was undertaken once in the CCD and HV groups and on days 0, 1, and 3 after stroke onset in patients with AIS or TIA. Correlations between serum FXI and FXII levels and demographic and clinical parameters were tested by linear regression and analysis of variance. Results: The mean age of AIS/TIA patients was 70 ± 12. Baseline clinical severity measured with NIHSS and Barthel Index was 4.8 ± 6.0 and 74 ± 30, respectively. More than half of the patients had an AIS (58%). FXI levels were significantly correlated with different leukocyte subsets (p < 0.05). In contrast, FXII serum levels showed no significant correlation (p > 0.1). Neither FXI nor FXII levels correlated with CRP (p > 0.2). FXII levels were significantly higher in patients with CCD compared with those with AIS/TIA (mean ± SD 106 ± 26% vs. 97 ± 24%; univariate analysis: p < 0.05); these differences did not reach significance in multivariate analysis adjusted for sex and age. FXI levels did not differ significantly between study groups. Sex and age were significantly associated with FXI and/or FXII levels in patients with AIS/TIA (p < 0.05). In contrast, no statistical significant influence was found for treatment modality (thrombolysis or not), pre-treatment with platelet inhibitors, and severity of stroke. Conclusions: In this study, there was no differential regulation of FXI and FXII levels between disease subtypes but biomarker levels were associated with patient and clinical characteristics. FXI and FXII levels might be no valid biomarker for predicting stroke risk.
The chromosomes of the turnip-tailed gecko Thecadactylus rapicauda from the Falcón State in northern Venezuela were examined by means of conventional staining, a variety of banding techniques and in situ hybridization with an 18S + 28S rDNA probe. In female specimens, C-banding analyses detected a cryptic W sex chromosome-associated interstitial heterochromatic segment which is absent in the Z sex chromosome. These ZW sex chromosomes are considered to be in a nascent stage of morphological differentiation and are absent in T. rapicauda collected in Guatemala. The amount, location and fluorochrome affinities of constitutive heterochromatin, the position of the nucleolus organizer region, and the genome sizes of female and male individuals were determined. The previously published cytogenetic data on T. rapicauda are discussed.
Enteric pathogens often cycle between virulent and saprophytic lifestyles. To endure these frequent changes in nutrient availability and composition bacteria possess an arsenal of regulatory and metabolic genes allowing rapid adaptation and high flexibility. While numerous proteins have been characterized with regard to metabolic control in pathogenic bacteria, small non-coding RNAs have emerged as additional regulators of metabolism. Recent advances in sequencing technology have vastly increased the number of candidate regulatory RNAs and several of them have been found to act at the interface of bacterial metabolism and virulence factor expression. Importantly, studying these riboregulators has not only provided insight into their metabolic control functions but also revealed new mechanisms of post-transcriptional gene control. This review will focus on the recent advances in this area of host-microbe interaction and discuss how regulatory small RNAs may help coordinate metabolism and virulence of enteric pathogens.
Introduction: Since 1996, the preferred approach for positioning the active middle-ear implant Vibrant Soundbridge© is a mastoidectomy and a posterior tympanotomy. With this device, placement of the floating mass transducer (FMT) on the long incus process is the standard method for treatment of mild-to-severe sensorineural hearing loss in the case of normal middle-ear anatomy. The aim of this study was to determine the vibrational effectiveness of FMT placement at the short incus process. Materials and Methods: An extended antrotomy and a posterior tympanotomy were performed in 5 fresh human temporal bones. As a control for normal middle-ear function, the tympanic membrane was stimulated acoustically and the vibration of the stapes footplate and the round-window (RW) membrane were (sequentially) measured by laser Doppler vibrometry. Vibration responses for coupling of an FMT to the long incus process (standard coupling) were compared to those for coupling to the short incus process. Results: Apart from narrow frequency bands near 3 and 9 kHz for the stapes footplate and RW membrane, respectively, the velocity responses presented no significant differences between standard coupling of the FMT and coupling to the short incus process. Conclusion: Coupling the FMT to the short incus process may be a viable alternative in cases where the surgical approach is limited to an extended antrotomy. A reliable technique for attachment to the short incus process has yet to be developed.
Background: Dose requirements of erythropoietin-stimulating agents (ESAs) can vary considerably over time and may be associated with cardiovascular outcomes. We aimed to longitudinally assess ESA responsiveness over time and to investigate its association with specific clinical end points in a time-dependent approach. Methods: The German Diabetes and Dialysis study (4D study) included 1,255 diabetic dialysis patients, of whom 1,161 were receiving ESA treatment. In those patients, the erythropoietin resistance index (ERI) was assessed every 6 months during a median follow-up of 4 years. The association between the ERI and cardiovascular end points was analyzed by time-dependent Cox regression analyses with repeated ERI measures. Results: Patients had a mean age of 66 ± 8.2 years; 53% were male. During follow-up, a total of 495 patients died, of whom 136 died of sudden death and 102 of infectious death. The adjusted and time-dependent risk for sudden death was increased by 19% per 5-unit increase in the ERI (hazard ratio, HR = 1.19, 95% confidence interval, CI = 1.07-1.33). Similarly, mortality increased by 25% (HR = 1.25, 95% CI = 1.18-1.32) and infectious death increased by 27% (HR = 1.27, 95% CI = 1.13-1.42). Further analysis revealed that lower 25-hydroxyvitamin D levels were associated with lower ESA responsiveness (p = 0.046). Conclusions: In diabetic dialysis patients, we observed that time-varying erythropoietin resistance is associated with sudden death, infectious complications and all-cause mortality. Low 25-hydroxyvitamin D levels may contribute to a lower ESA responsiveness.
Regeneration of calvarial defects with Escherichia coli-derived rhBMP-2 adsorbed in PLGA membrane

Objective: Escherichia coli-derived recombinant human bone morphogenetic protein-2 (E-BMP-2) has been shown to be as effective as mammalian cell-derived BMP-2. However, several in vitro and in vivo experiments are still necessary to validate the effectiveness of E-BMP-2 due to the difference in synthesis process, mainly related to protein nonglycosylation. The objective of this study was to investigate whether biodegradable polylactide-co-glycolide (PLGA) membrane is a suitable carrier for E-BMP-2 delivery for bone regeneration of critical-sized defects in rat calvaria. Materials and Methods: First, the osteoinductive effect of E-BMP-2 was confirmed in vitro in mouse bone marrow stromal cells by analysis of osteocalcin mRNA levels, and calcium deposition was detected by alizarin red staining. Before in vivo experiments, the release profile of E-BMP-2 from PLGA membranes was determined by ELISA. E-BMP-2 (0, 1, 5 and 10 μg/μl) was applied for ectopic and orthotopic bone formation and was analyzed by X-ray, micro-CT and histology. Results: Release-profile testing showed that PLGA membrane could retain 94% of the initially applied E-BMP-2. Ectopic bone formation assay revealed that combination of E-BMP-2/PLGA membrane strongly induced bone formation. Stronger osteoinductivity with complete repair of critical-sized defects was observed only with PLGA membranes adsorbed with 5 and 10 μg/μl of E-BMP-2, whereas no bone formation was observed in the groups that received no membrane or 0-μg/μl dose of E-BMP-2. Conclusion: PLGA membrane was shown to be a suitable carrier for sustained release of E-BMP-2, and the E-BMP-2/PLGA membrane combination was demonstrated to be efficient in bone regeneration in a model of critical-sized defects.
Mitotic and meiotic chromosomes of 5 species of the reptile genus Gonatodes are described by means of conventional staining, banding analyses and in situ hybridization using a synthetic telomeric DNA probe. The amount, location and fluorochrome affinities of constitutive heterochromatin, the number and positions of nucleolus organizer regions, and the patterns of telomeric DNA sequences were determined for most of the species. The karyotypes of G. falconensis and G. taniae from northern Venezuela are distinguished by their extraordinarily reduced diploid chromosome number of 2n = 16, which is the lowest value found so far in reptiles. In contrast to most other reptiles, both species have exclusively large biarmed (meta- and submetacentric) chromosomes. Comparison of the karyotypes of G. falconensis and G. taniae with those of other Gonatodes species indicates that the exceptional 2n = 16 karyotype originated by a series of 8 centric fusions. The karyotypes of G. falconensis and G. taniae are further characterized by the presence of considerable amounts of (TTAGGG)<sub>n</sub> telomeric sequences in the centromeric regions of all chromosomes. These are probably not only relics of the centric fusion events, but a component of the highly repetitive DNA in the constitutive heterochromatin of the chromosomes. The genome sizes of 4 Gonatodes species were determined using flow cytometry. For comparative purposes, all previously published cytogenetic data on Gonatodes and other sphaerodactylids are included and discussed.
The mitotic chromosomes of 11 species from the anuran families Centrolenidae and Allophrynidae were analyzed by means of conventional staining, banding techniques, and in situ hybridization. The amount, location, and fluorochrome affinities of constitutive heterochromatin, the number and positions of nucleolus organizer regions, and the patterns of telomeric DNA sequences were determined for most of the species. The karyotypes were found to be highly conserved with a low diploid chromosome number of 2n = 20 and morphologically similar chromosomes. The sister group relationship between the Centrolenidae and Allophrynidae (unranked taxon Allocentroleniae) is clearly corroborated by the cytogenetic data. The existence of heteromorphic XY♂/XX♀ sex chromosomes in an initial stage of morphological differentiation was confirmed in Vitreorana antisthenesi. The genome sizes of 4 centrolenid species were determined using flow cytometry. For completeness and for comparative purposes, all previously published cytogenetic data on centrolenids are included.
Background: Numerous birth cohorts have been initiated in the world over the past 30 years using heterogeneous methods to assess the incidence, course and risk factors of asthma and allergies. The aim of the present work is to provide the stepwise proceedings of the development and current version of the harmonized MeDALL-Core Questionnaire (MeDALL-CQ) used prospectively in 11 European birth cohorts. Methods: The harmonization of questions was accomplished in 4 steps: (i) collection of variables from 14 birth cohorts, (ii) consensus on questionnaire items, (iii) translation and back-translation of the harmonized English MeDALL-CQ into 8 other languages and (iv) implementation of the harmonized follow-up. Results: Three harmonized MeDALL-CQs (2 for parents of children aged 4-9 and 14-18, 1 for adolescents aged 14-18) were developed and used for a harmonized follow-up assessment of 11 European birth cohorts on asthma and allergies with over 13,000 children. Conclusions: The harmonized MeDALL follow-up produced more comparable data across different cohorts and countries in Europe and will offer the possibility to verify results of former cohort analyses. Thus, MeDALL can become the starting point to stringently plan, conduct and support future common asthma and allergy research initiatives in Europe.

Franz von Kutschera über ungegenständliche Erfahrungen und ihre (religions-)philosophische Bedeutung

Zusammenfassung
Der Beitrag diskutiert im Anschluss an Franz von Kutschera die (religions-)philosophische und theologische Bedeutung ungegenständlicher Erfahrungen. Es handelt sich um Erfahrungen, die sich nicht oder nur bedingt als Erfahrungen eines Subjekts von einem Objekt beschreiben lassen. Besonders in den Blick genommen wird die Relevanz ungegenständlicher Erfahrungen sowohl für die Semantik religiöser Rede als auch für die Epistemologie religiöser Überzeugungen. In diesem Zusammenhang wird argumentiert, dass die kognitive Relevanz dieser Erfahrungen davon abhängt, ob sie innerhalb eines weltanschaulichen Paradigmas einen Beitrag zu einem kohärenten und umfassenden Verständnis der uns begegnenden Wirklichkeit leisten.
Abstract
Constructing evidence constitutes a practice to establish the speaker's authority at Prime Minister's Question Time (PMQT), a weekly half-hour session in the British House of Commons. Here the verb see constitutes a resource for both the questioning Leader of the Opposition (LO) and Members of Parliament (MP) as well as for the responding Prime Minister (PM) to claim first-hand perceptual experience. This paper takes an integrated approach, offering a combined analysis of the grammatical formatting, semantics and pragmatics of the verb see in the context of evidential moves at PMQT. It shows how the verb see is functional in referring to the perceptual basis of a claim made and how its grammatical formatting is reflective of the contingencies of the local interactional context. The analysis is grounded in 32 sessions of PMQT (ca. 16 hrs of video-recordings). The results can be summarised as follows: 1) The evidential function of the verb is achieved through its context-specific grammatical formatting and semantics. 2) The reference to the perceptual basis of a claim evoked by see may co-occur with epistemic qualification and evaluative expressions. 3) The formatting of the verb may be indexical of the political relationship between the questioner and the responding PM.
Autophagy is a central process behind the cellular remodeling that occurs during differentiation of Leishmania, yet the cargo of the protozoan parasite's autophagosome is unknown. We have identified glycosomes, peroxisome-like organelles that uniquely compartmentalize glycolytic and other metabolic enzymes in Leishmania and other kinetoplastid parasitic protozoa, as autophagosome cargo. It has been proposed that the number of glycosomes and their content change during the Leishmania life cycle as a key adaptation to the different environments encountered. Quantification of RFP-SQL-labeled glycosomes showed that promastigotes of L. major possess ~20 glycosomes per cell, whereas amastigotes contain ~10. Glycosome numbers were significantly greater in promastigotes and amastigotes of autophagy-defective L. major Δatg5 mutants, implicating autophagy in glycosome homeostasis and providing a partial explanation for the previously observed growth and virulence defects of these mutants. Use of GFP-ATG8 to label autophagosomes showed glycosomes to be cargo in ~15% of them; glycosome-containing autophagosomes were trafficked to the lysosome for degradation. The number of autophagosomes increased 10-fold during differentiation, yet the percentage of glycosome-containing autophagosomes remained constant. This indicates that increased turnover of glycosomes was due to an overall increase in autophagy, rather than an upregulation of autophagosomes containing this cargo. Mitophagy of the single mitochondrion was not observed in L. major during normal growth or differentiation; however, mitochondrial remnants resulting from stress-induced fragmentation colocalized with autophagosomes and lysosomes, indicating that autophagy is used to recycle these damaged organelles. These data show that autophagy in Leishmania has a central role not only in maintaining cellular homeostasis and recycling damaged organelles but crucially in the adaptation to environmental change through the turnover of glycosomes.
The molecular basis of male infertility is poorly understood, the majority of cases remaining unsolved. The association of aberrant sperm DNA methylation patterns and compromised semen parameters suggests that disturbances in male germline epigenetic reprogramming contribute to this problem. So far there are only few data on the epigenetic heterogeneity of sperm within a given sample and how to select the best sperm for successful infertility treatment. Limiting dilution bisulfite sequencing of small pools of sperm from fertile donors did not reveal significant differences in the occurrence of abnormal methylation imprints between sperm with and without morphological abnormalities. Intracytoplasmic morphologically selected sperm injection was not associated with an improved epigenetic quality, compared to standard intracytoplasmatic sperm injection. Deep bisulfite sequencing (DBS) of 2 imprinted and 2 pluripotency genes in sperm from men attending a fertility center showed that in both samples with normozoospermia and oligoasthenoteratozoospermia (OAT) the vast majority of sperm alleles was normally (de)methylated and the percentage of epimutations (allele methylation errors) was generally low (<1%). However, DBS allowed one to identify and quantify these rare epimutations with high accuracy. Sperm samples not leading to a pregnancy, in particular in the OAT group, had significantly more epimutations in the paternally methylated GTL2 gene than samples leading to a live birth. All 13 normozoospermic and 13 OAT samples leading to a child had <1% GTL2 epimutations, whereas one (7%) of 14 normozoospermic and 7 (50%) of 14 OAT samples without pregnancy displayed 1–14% GTL2 epimutations.

10-jahres Ergebnisse einer prospektiven, randomisierten klinischen Studie.
Verfahren:
Diese Studie vergleicht die Ergebnisse einer 10-Jahres Nachuntersuchung nach arthroskopisch unterstütztem Ersatz des vorderen Kreuzbandes mittels zweier verschiedener autologer Transplantate. Zwischen November 1998 und November 1999 wurden in der Orthopädischen Universitätsklinik König-Ludwig-Haus in Würzburg 62 Studienteilnehmer (45 Männer, 17 Frauen) mit Ruptur des vorderen Kreuzbandes operiert. Bei 31 Patienten erfolgte die ACL-Ersatzplastik mittels Lig. Patellae Transplantat (BTB-Transplantat), bei der anderen Hälfte mittels Semitendinosussehnentransplantat. Die Gruppenzusammensetzung zeigte vor dem Verletzungszeitpunkt keinen signifikanten Unterschied bezüglich Alter, Geschlecht und Aktivitätsniveau. Alle Patienten erhielten die gleiche postoperative Behandlung mit Don-Joy Schiene (Ext/Flex 0/0/90), Teilbelastung bis 20 kg während der ersten 6 Wochen und externe Rehabilitationsmaßnahmen. Nach über 10 Jahren konnten 47 (24 LP/23 ST) Patienten vollständig klinisch untersucht und bei 9 (5 LP/4 ST) weiteren zumindest standardisierte Fragebögen ausgewertet werden. 2 Studienteilnehmer (beide ST) schieden wegen einer Transplantatruptur, ein weiterer Patient (ebenfalls ST) aufgrund einer postoperativen Kniegelenksinfektion vorzeitig aus der Studie aus.
Ergebnisse:
Die klinische Untersuchung ergab bezüglich der verwendeten Parameter IKDC- und Lysholm-Score, Tegener-Aktivitätsskala, KT-1000-Arthrometer-Messung und radiologischer Kontrolle keine signifikanten Unterschiede zwischen den beiden Gruppen. Auch in den verwendeten Fragebögen mit Evaluation der subjektiven Kniefunktion, möglicher Schmerzen im Kniegelenk, Entnahmestellenmorbidität und der Zufriedenheit mit dem Operationsergebnis konnten keine signifikanten Gruppenunterschiede festgestellt werden.
In der vorliegenden Studie fanden sich somit auch über 10 Jahre nach vorderem Kreuzbandersatz keine relevanten Unterschiede oder Vorteile für eines der beiden untersuchten autologen Sehnentransplantate.
Die autoimmune Thyreoiditis nach Hashimoto stellt aktuell eine der häufigsten Autoimmunerkrankungen eines Organs und die häufigste Ursache der Hypothyreose dar. Die Hashimotothyreoiditis (HT) weist eine hohe Prävalenz und Inzidenz auf. Es existieren Hinweise, dass die Inzidenz der HT aus noch nicht geklärten Gründen gestiegen sein könnte.
Die Kohlenhydrate Fruktose, Laktose und Sorbitol werden in der Lebensmittelproduktion umfassend eingesetzt. Insbesondere die industrielle Verwendung sowie der weltweite Konsum von Fruktose und Laktose unterlagen in den letzten Jahrzehnten einer rasanten Steigerung, obwohl ein hoher Prozentsatz der Bevölkerung zur Malassimilation jener Kohlenhydrate prädisponiert ist.
In einer internistischen Praxis (Praxis Frau Dr. med. I. Heckl, Bad Homburg) zeigte sich, dass HT-Patienten trotz verifizierter Euthyreose vermehrt über gastrointestinale Symptome berichteten. Unter anderem wurden eine bakterielle Fehlbesiedelung des Dünndarmes und eine Zöliakie ausgeschlossen. In der weiteren Abklärung durch die Praxis Dr. I. Heckl wurde eine deutliche Häufung der Malassimilation der Kohlenhydrate Fruktose, Laktose oder Sorbitol unter euthyreoten HT-Patienten ersichtlich. In Abhängigkeit von einer konsequenten Nahrungsumstellung normalisierten sich regelmäßig das Befinden der Patienten sowie die sonographischen, die serologischen und die laborchemischen Marker der HT, sodass man einen ursächlichen Zusammenhang empirisch vermuten konnte.
Im Rahmen einer prospektiven Studie sollte dieser neu beobachtete Zusammenhang zwischen der HT und der Kohlenhydratmalassimilation in der Klinik und Poliklinik für Nuklearmedizin der Universität Würzburg untersucht werden.
In einem unizentrischen Fall-Kontroll-Studiendesign wurden 45 euthyreote HT-Patienten und 38 schilddrüsengesunde Kontrollpersonen auf das Vorliegen einer Kohlenhydratmalassimilation mittels des Wasserstoffatemtests (H2-Atemtest) untersucht. Alle Probanden erhielten einen Fruktose-H2-Atemtest sowie einen Laktose-H2-Atemtest inklusive einer kapillären Blutglukosemessung. Im Falle eines positiven Ergebnisses des Fruktose-H2-Atemtests wurde ein Glukose-H2-Atemtest zum Ausschluss einer bakteriellen Fehlbesiedelung des Dünndarmes durchgeführt. Lieferte der Fruktose-H2-Atemtest ein negatives Ergebnis, so folgte ein H2-Atemtest mit Sorbitol. Das Auftreten gastrointestinaler Symptome während der Testdurchführung wurde dokumentiert. Symptomfragebögen und semiquantitative Ernährungsfragebögen im retrospektiven Design dienten der Erfassung alltäglicher Symptome und Ernährungsgewohnheiten. Blutproben dienten der Messung von Schilddrüsenhormonen, Schilddrüsenautoantikörpern, Gewebstransglutaminase-Antikörpern und Antiparietalzell-Autoantikörpern.
Unter den euthyreoten HT-Patienten konnte ein signifikant häufigeres Auftreten der Fruktose- sowie der Laktosemalassimilation im Vergleich zu den schilddrüsengesunden Kontrollpersonen demonstriert werden. Die Fruktosemalassimilation wurde bei den HT-Patienten mit 48,9% signifikant häufiger als in der Kontrollgruppe nachgewiesen (p=0,035). Im Kontrollgruppenkollektiv hatte eine Fruktosemalassimilation lediglich bei 26,3% der Probanden bestanden. Eine Laktosemalassimilation wurde bei den HT-Patienten mit 42,2% signifikant häufiger als im Kontrollkollektiv diagnostiziert, welches in 21,1% der Fälle eine Laktosemalassimilation aufwies (p=0,04). Insgesamt lag eine Fruktose- und / oder Laktosemalassimilation bei 73,3% der HT-Patienten und bei 42,1% der Kontrollgruppenprobanden vor. Somit vertrugen nur 26,7% der Fallgruppe, jedoch 57,9% der Kontrollgruppe beide Kohlenhydrate (p=0,004). Hinsichtlich der Prävalenz der Sorbitolmalassimilation oder eines positiven Glukose-H2-Atemtestes kam kein signifikanter Unterschied zur Darstellung. Die Auswertung der Ernährungsfragebögen zeigte für beide Kollektive eine vergleichbare durchschnittliche Konsummenge der jeweiligen Kohlenhydrate auf.
Gastrointestinale Symptome waren während des Laktose-H2-Atemtests sowie während des Fruktose-H2-Atemtests jeweils in der Fallgruppe signifikant häufiger anzutreffen als in der Kontrollgruppe. Auch im Hinblick auf das Alltagsleben beschrieben die euthyreoten HT-Patienten signifikant häufiger unter den folgenden Symptomen zu leiden: Weicher Stuhlgang, Oberbauchschmerzen, Meteorismus, laute Darmgeräusche, „Kugelbauch“, Sodbrennen, Schleimauflagerungen des Stuhlgangs, Obstipation, Müdigkeit, postprandiale Kraftlosigkeit, Depressionen, Heißhunger auf Süßes, Migräne, Konzentrationsmangel und eine vermehrte Infektanfälligkeit.
Zur Kausalität des hier erstmals beschriebenen Zusammenhangs existieren mehrere Hypothesen. Die Einteilung der HT-Patienten gemäß ihrer Schilddrüsenautoantikörper-Titer in Subkollektive ergab keinen Hinweis auf einen Einfluss der Aktivität des Autoimmungeschehens auf die Häufigkeit der Kohlenhydratmalassimilation. Es steht zur Diskussion, ob die HT zur Entstehung einer Kohlenhydratmalassimilation führen, oder ob eine vorbestehende Kohlenhydratmalassimilation, im Sinne eines neu identifizierten Risikofaktors, zur Genese einer HT prädisponieren könnte.
In der vorliegenden Studie konnte erstmalig eine signifikante Häufung der Kohlenhydratmalassimilation bei euthyreoten HT-Patienten aufgezeigt werden. Vor dem Hintergrund der weitreichenden lebensmittelindustriellen Verwendung und des hohen Konsums der Kohlenhydrate Fruktose, Laktose und Sorbitol sowie der hohen Prävalenz und Inzidenz der HT, ergibt sich eine hohe Relevanz des hier nachgewiesenen Zusammenhangs. In der differenzialdiagnostischen Abklärung gastrointestinaler Beschwerden bei euthyreoten HT-Patienten nimmt die hier beschriebene Assoziation zwischen der HT und der Kohlenhydratmalassimilation einen besonderen Stellenwert ein. Die Kohlenhydratmalassimilation verkörpert einen neuen, sowohl klinisch, als auch potentiell pathogenetisch relevanten Aspekt der Hashimotothyreoiditis.
The Staphylococcus aureus two component system (TCS) sae governs expression of numerous virulence factors, including Eap (extracellular adherence protein), which in turn among other functions also mediates invasion of host cells. The sae TCS is encoded by the saePQRS operon, with saeS coding for the sensor histidine kinase (SaeS) and saeR encoding the response regulator (SaeR). The saeRS system is preceded by two additional open reading frames (ORFs), saeP and saeQ, which are predicted to encode a lipoprotein (SaeP) and a membrane protein (SaeQ), respectively. Earlier, we have shown that SDS-containing subinhibitory concentrations of biocides (Perform®) and SDS alone activate sae transcription and increase cellular invasiveness in S. aureus strain Newman. The effect is associated with an amino acid exchange in the N-terminus of SaeS (L18P), specific to strain Newman.
In this work, the role of whether the two additional genes, saePQ coding for the accessory proteins SaeP and SaeQ, respectively, are involved in SDS-mediated saeRS was investigated. It could demonstrated that the lack of the SaeP protein resulted in an increased saeRS transcription without SDS stress in both SaeSL/P variants, while the SDS effect was less pronounced on sae and eap expression compared to the Newman wildtype, suggesting that the SaeP protein represses the sae system. Also, SDS-mediated inductions of sae and eap transcription along with enhanced invasion were found to be dependent on presence of the SaeSP variant in Newman wildtype. On the other hand, the study also shows that the saePQ region of the sae operon is required for fully functional two-component system saeRS under normal growth conditions, but it is not involved in SDS-mediated activation of the saeS signaling and sae-target class I gene, eap.
In the second approach, the study investigates whether SDS-induced sae expression and host cell invasion is common among S. aureus strains not carrying the (L18P) point mutation. To demonstrate this strain Newman, its isogenic saeS mutants, and various S. aureus isolates were analysed for sae, eap expression and cellular invasiveness. Among the strains tested, SDS exposure resulted only in an increase of sae transcription, Eap production and cellular invasiveness in strain Newman wild type and MRSA strain ST239-635/93R, the latter without an increase in Eap. Interestingly, the epidemic community-associated MRSA strain, USA300 LAC showed a biphasic response in sae transcription at different growth stages, which, however, was not accompanied by increased invasiveness. All other clinical isolates investigated displayed a decrease of the parameters tested. While in strain Newman the SDS effect was due to the saeSP allele, this was not the case in strain ST239-635/93R and the biphasic USA300 strains. Also, increased invasiveness of ST239-635/93R was found to be independent of Eap production. Furthermore, to investigate the global effect of SDS on sae target gene expression, strain Newman wild-type and Newman ∆sae were treated with SDS and analyzed for their transcription profiles of sae target genes using microarray assays. We could show that subinhibitory concentrations of SDS upregulate and downregulate gene expression of several signaling pathways involved in biosynthetic, metabolic pathways as well as virulence, host cell adherence, stress reponse and many hypothetical proteins.
In summary, the study sheds light on the role of the upstream region saePQ in SDS-mediated saeRS and eap expression during S. aureus SDS stress. Most importantly, the study also shows that subinhibitory SDS concentrations have pronounced strain-dependent effects on sae transcription and subsequent host cell invasion in S. aureus, with the latter likely to be mediated in some strains by other factors than the known invasin Eap and FnBP proteins. Moreover, there seems to exist more than the saeSP-mediated mechanism for SDS-induced sae transcription in clinical S. aureus isolates. These results help to further understand and clarify virulence and pathogenesis mechanisms and their regulation in S. aureus.
Das „Phänomen“ Shopping Center beschäftigt geographische Handelsforscher und Praktiker seit Jahren gleichermaßen. Dies liegt nicht zuletzt an vielfältigen Prozessen der raum-zeitlichen Ausbreitung, welche auf globaler Ebene zunehmende Internationalisierung, auf der regionalen Ebene rücken vermehrt mittelgroße und kleinere Städte in den Fokus der Projektentwickler und auf lokaler Ebene ist eine Verlagerung des Ansiedlungsgeschehens von Stadtrandlagen zu innerstädtischen Zentren zu beobachten.
Auf Akteursseite spielen Projektentwickler und Management eine wichtige Rolle, um Shopping Center zu erfolgreichen Investmentprodukten zu machen, während die räumliche Planung bei der Bewertung von Projektanträgen und bei der Einordnung in städtische Ansiedlungsstrategien und Leitbilder zu berücksichtigen hat, dass Shopping Center eine besondere Raumkonfiguration in den Stadtlandschaften darstellen - mit Auswirkungen auf verschiedene Aspekte der wirtschaftlichen Entwicklungen und des sozialen Lebens. Ihre Verträglichkeit mit bestehenden Einzelhandelsstrukturen, ihre Wirkungen auf die Stadtentwicklung sowie auf Kaufkraftströme mit geeigneten Methoden und unter Berücksichtigung aktueller Prozesse zu untersuchen, ist Anliegen der Forschung und Praxis.
Diesem Anliegen widmet sich der vorliegende Sammelband, wobei der Großteil der Beiträge auf den Vorträgen einer Fachsitzung im Rahmen des Deutschen Geographentages 2013 in Passau basiert.
Was bedeutet eine Änderung der Kontrastmittelaufnahme in niedrig malignen Gliomen bei Kindern?

Bei niedriggradigen Gliomen WHO I° und II° ist das Kontrastmittelverhalten variabel und nicht mit der Prognose assoziiert. Andererseits wurde bei spontanen Regressionen von niedriggradigen Gliomen bei Patienten sowohl mit als auch ohne eine Neurofibromatose Typ I eine Abnahme der Kontrastmittelaufnahme berichtet. Vielleicht deswegen verleitet eine Zunahme des Enhancements oder ein neu aufgetretenes Enhancement nicht selten zur Diagnose einer Tumorprogression. Es stellt sich also die Frage, ob eine Kontrastmittelaufnahme bei LGGs mit einer Größenzunahme somit einem Tumorprogress assoziiert ist.
Es lässt sich eine überdurchschnittliche Assoziation einer Änderung der Kontrastmittelaufnahme mit dem Wachstumsverhalten bei LGGs nachweisen. Allerdings ist das Maß für eine Progression oder Regression weiterhin die Größenänderung des Tumors. Auch eine neue Kontrastmittelaufnahme innerhalb eines Tumors hat keine Bedeutung für das aktuelle Staging. Sie findet sich zwar häufiger bei Wachstum aber auch bei regredienten Tumoren.
Flatworm parasites (platyhelminths) cause serious infection diseases in humans, such as schistosomiasis and hydatid disease, mainly prevalent in developing countries. However, the current repertoire of drug armamentarium used to combat flatworm infections is limited. For instance, praziquantel is the only drug available for mass treatment of Schistosoma infections. In contrast to their hosts, flatworm parasites possess a distinct redox arrangement of redox pathways in which the selenoenzyme thioredoxin glutathione reductase (TGR) controls the overall redox homeostasis. Interference with this enzyme leads to parasite death. Hence, this key redox enzyme seems to be a new promising drug target against flatworm infections.
Because most flatworms are difficult to cultivate in the laboratory (e.g. Echinococcus granulosus experimental infection in mice takes about 10 month to develop into cysts), this work was focused on Mesocestoides vogae (syn. corti), a non-human flatworm parasite which is an interesting laboratory model to study other flatworm infections: it is very rare in humans, can be easily manipulated both in vivo and in vitro and grows extremely fast in mice. With the aim to assess TGR inhibitors as possible drugs to treat flatworm infections, the thioredoxin and glutathione pathways of M.vogae were studied. Here, the objectives were to study whether the biochemical pathways that maintain the redox homeostasis in M. vogae conform to the general biochemical scenario proposed for other platyhelminth parasites.
Here, it was proven that M. vogae extracts possess both thioredoxin and glutathione reductase activities. The thioredoxin and glutathione reductase activities were partially purified from total extracts by a combination of ammonium sulfate precipitation, anion exchange and hydroxyapatite chromatography. Both activities co-purified in all steps which strongly indicates the existence of TGR rather than a single TR and GR. Furthermore partially purified activities could be inhibited by the organogold compound auranofin, a known TGR inhibitor. Moreover, the glutathione reductase activity displays hysteresis (a peculiar kinetic behavior) at high concentrations of oxidised glutathione, a feature typical of flatworm TGRs, but not of conventional GR. Although M. vogae activities could not be purified to homogeneity, the overall results strongly indicate that this flatworm possesses TGR and lacks conventional GR and TR.
Furthermore the thiadiazole WPQ75 and the N-oxide VL16E (a furoxan derivate) were identified as inhibitors of TGR activity of M.vogae at a 10 µM concentration. These inhibitors were able to kill M.vogae larval worms in vitro as well as in experimental infection in mice.
Due to the existence of TGR activity in M.vogae, the possibility to inhibit this activity with recently discovered inhibitors of flatworm TGR and the successes achieved by testing these inhibitors both in vitro and in vivo, it is strongly evident that M. vogae would be an excellent model to assess TGR inhibitors in flatworm infections.
Aminoborylenkomplexe der Gruppe 6 [(OC)5M=BN(SiMe3)2] (M = Cr, Mo, W) reagieren mit Übergangsmetallkomplexen unter Transfer der Boryleneinheit bzw. in Transmetallierungsreaktionen und bilden dabei neuartige Borylenkomplexe. In dieser Dissertation wird die Synthese, Charakterisierung und Reaktivität der auf diesem Wege dargestellten Verbindungen - unter anderem Hydridoborylenkomplexe, Bis(borylen)komplexe und borylensubstituierte MOLPs - beschrieben.
CD40-restringierte Aktivierung der TRAIL-Todesrezeptoren durch bifunktionelle rekombinante Proteine

Der Ligand TRAIL wurde 1997 aufgrund seiner hohen Sequenzhomolgie ge-genüber dem TNFL CD95L entdeckt (28 %). Allerdings besitzt TRAIL, anders als die Liganden CD95L und TNF, die bemerkenswerte Eigenschaft vor allem in veränderten Zellen Apoptose zu induzieren, während gesunde Zellen davor bewahrt werden. Die TRAIL-induzierte Apoptose wird durch die apoptoseinduzierenden Todesrezeptoren TRAILR1 und TRAILR2 vermittelt. Allerdings bindet und aktiviert lösliches TRAIL hauptsächlich den Todesrezeptor TRAILR1, während membrangebundes TRAIL sowohl TRAILR1 als auch TRAILR2 gut aktiviert. In den letzten Jahren wurden verschiedene Methoden entwickelt, um die Bioaktivität löslicher TNFL zu steigern. Hierzu zählen z.B.: Stabilisierung der trimeren Molekülanordnung über die TNC-Domäne, Oligomerisierung des Flag-getaggten Liganden mithilfe des monoklonalen Antikörpers M2, sowie Generierung einer artifiziellen, antigenabhängigen Membranständigkeit. In dieser Arbeit wurde der Oberflächenrezeptor CD40 zur Immobilisierung des generierten Fusionsproteins scFv:CD40-Flag-TNC-TRAIL genutzt. In verschieden Experimenten konnten mit scFv:CD40-Flag-TNC-TRAIL in CD40-exprimierenden Zellen starke Apoptoseinduktion ermittelt werden. Charakteris-tische Kennzeichen und Spaltprodukte der Apoptose konnten ausschließlich in CD40-positiven Tumorzellen detektiert werden. Dabei wurde in allen Versuchen die für die Apoptoseinduktion benötigte Konzentration des Konstrukts mithilfe des Proteinsyntheseinhibitors CHX um das 10- bis 100-fache verringert. Es konnte auch gezeigt werden, dass in CD40-positiven Zellen, nach Stimulation mit scFv:CD40-Flag-TNC-TRAIL, nicht-apoptotische Signalwege verstärkt aktiviert werden. Dies war auf die agonistische Aktivität des monoklonalen Antikörperfragments scFv:CD40 zurückzuführen. Die Antikörperdomäne war folglich nicht nur zur effizienten Aktivierung der TRAIL-Todesrezeptoren mittels Immobilisierung fähig, sondern konnte zusätzlich zur Stimulation des Immunsystems genutzt werden. Zusammenfassend konnte gezeigt werden, dass der lösliche, schwach aktive Ligand TRAIL mittels Oberflächenimmobilisierung über Antigen-Antikörper-Wechselwirkungen in einen hochaktiven Liganden mit lokal begrenzter Toxizität überführt werden kann.
Mithilfe dieses Fusionsproteins ist es somit möglich die selektive Toxizität von TRAIL durch Steigerung seiner Aktivität effizient zu nutzen. Zusätzlich kann durch die Antigenbindung der Wirkungsbereich weiter eingegrenzt werden (CD40-positive Tumoren), wodurch unerwünschte Nebenwirkungen reduziert oder sogar ausgeschaltet werden können. Das in Tumoren oft heruntergefahrene Immunsystem kann CD40-abhängig stimuliert werden, um somit auch Tumorzellen in apoptoseresistenten Stadien zu eliminieren. Basierend auf diesen Ergebnissen können in der Zukunft weitere Studien zur Therapie von TRAIL-resistenten, CD40-exprimierenden Tumoren fortgeführt werden.
Die primordialen Keimzellen (PGCs) sind die einzigen Zellen des Embryos, die die genetische Information von einer Generation an die nächste weiter geben können. Es wurde gezeigt, dass in allen bislang untersuchten Knochenfischen die Anzahl der Urgeschlechtszellen während der Embryonalentwicklung der erste sichtbare Unterschied zwischen Männchen und Weibchen ist. Daraus ergibt sich die Frage, ob die Anzahl der primordialen Keimzellen das Geschlecht bestimmt, oder ob die somatischen Zellen je nach sexueller Identität die Urgeschlechtszellen zur Proliferation anregen. Um zu untersuchen, wie die Anzahl der
Urgeschlechtszellen mit der Geschlechtsdetermination zusammenhängt, habe ich in dieser Arbeit die Anzahl der Urgeschlechtszellen manipuliert und deren Schicksal im Verlauf der Embryonalentwicklung verfolgt. Weiterhin untersuchte ich, in wieweit die Temperatur einen Einfluss auf die Geschlechtsbestimmung hat und ob sie Auswirkungen auf die Anzahl
und die Wanderung der Urgeschlechtszellen hat beim Medaka hat.
Durch meine Experimente, in denen ich die Fische während der Embryonalentwicklung bei verschiedenen Temperaturen hielt, konnte ich zeigen, dass beim Medaka der genetische Geschlechtsbestimmungsmechanismus durch erhöhte Temperatur überschrieben werden kann. Die Temperaturerhöhung in der Embryonalentwicklung führt zu einer Weibchen‐zu‐Männchen
Geschlechtsumkehr. Dabei wird die Anzahl der primordialen Keimzellen im Vergleich zu den Kontrollen reduziert. Zudem wird durch die höhere Temperatur das autosomale dmrt1a viel früher angeschaltet, wa sauf einen alternativenSignalweg deutet, der die männliche Geschlechtsentwicklung in XX geschlechtsumgewandelten Tieren steuert.
Das Syndrom Herzinsuffizienz ist in Deutschland einer der häufigsten Beratungsanlässe in einer allgemeinmedizinischen Praxis und der häufigste Grund für eine stationäre Krankenhausaufnahme. Die Inzidenz ist weiter steigend. Die häufigste Todesursache dieser Patienten ist der plötzliche Herztod, meist auf Grund maligner Herzrhythmusstörungen. Es gibt verschiedene Kriterien, anhand derer das Risiko des Einzelnen, potenziell tödliche Herzrhythmusstörungen zu erleiden, individuell abgewägt werden kann. Eine zuverlässige Vorhersage ist nach wie vor schwierig. Neben der Symptomatik des Patienten und der Vorgeschichte spielen hier vor allem Ejektionsfraktion und EKG-Veränderungen eine Rolle. Zahlreiche Publikationen haben den Parametern STV-RR und STV-QT einen unabhängigen Stellenwert in der Risikostratifizierung bei Patienten mit chronischer Herzinsuffizienz zugesprochen. Diese beiden Werte können aus der Analyse 30 aufeinanderfolgender Herzaktionen berechnet werden und spiegeln die zeitliche Dispersion des RR-Intervalls bzw. der QT-Zeit wider.
Ziel dieser Arbeit war es, diese beiden Marker an einem bislang nicht etablierten Herzinsuffizienzmodell zu evaluieren. Dies ist zum einen interessant, da STV-RR und STV-QT im Rahmen des Monitorings intensivpflichtiger Patienten eine kardiale Dekompensation möglicherweise frühzeitig anzuzeigen vermögen, zum anderen lässt dieses Modell unter Umständen auch Rückschlüsse auf die Ätiologie dieser EKG-Veränderungen zu.
Zu diesem Zweck wurde bei primär herzgesunden Schafen ein hypertensives Lungenödem induziert und die EKG-Parameter wurden mit dem Ausmaß der kardialen Dekompensation korreliert. Eine Gruppe gematchter Vergleichstiere wurde unter denselben Versuchsbedingungen mit Ausnahme der Ödeminduktion untersucht. Insgesamt wurden die Daten von 15 Schafen evaluiert, von denen 10 der Experiment- und 5 der Kontrollgruppe zugeordnet wurden. Es wurden sowohl intraindividuelle (Baseline- versus Endwerte) als auch interindividuelle Änderungen der EKG-Parameter ausgewertet. Die Vergleiche basierten dabei vornehmlich auf relativen Änderungen, da die Tiere zum Teil mit deutlich unterschiedlichen Baseline-Werten in den Versuch gestartet waren. Nicht alle Experimenttiere erreichten das maximal angestrebte Ausmaß der Dekompensation (130 % des Ausgangswertes des EVLW). Daher nahm die Anzahl der in die Auswertung miteinfließenden Tiere mit zunehmendem EVLW-Level ab. Zusätzlich wurde eine rein zeitbezogene Analyse durchgeführt.
Es galt zu prüfen, ob sich im Rahmen der hypertensiven Entgleisung mit Hypervolämie und konsekutiver kardialer Dekompensation signifikante Änderungen von STV-RR oder STV-QT in Abhängigkeit vom Ausmaß des Lungenödems ergeben. Die Analyse der aus dem Schafmodell gewonnenen Daten ergab keine statistisch signifikanten Änderungen was STV-QT betrifft, zudem konnte keine Korrelation zwischen STV-QT und EVLW nachgewiesen werden. Das gesamte Experiment-Kollektiv betrachtet, fand sich auch kein signifikanter Abfall von STV-RR im Rahmen des Versuchs. Die Tiere einzeln betrachtet fand sich jedoch bei allen mit Ausnahme von Nr. 77 ein signifikanter Abfall des Parameters nach Anstieg von EVLW auf 130 % des Ausgangswerts. Das Kollektiv ohne Nr. 77 zeigte einen signifikanten Abfall von STV-RR (p = 0,001) bei einem EVLW Level von 130 %. Das gesamte Kollektiv (15 Schafe) betrachtet, ließ sich eine signifikante Korrelation von STV-RR mit EVLW (p = 0,0003) und dem RR-Intervall nachweisen (p = 1,5 x10-11).
Diese Ergebnisse lassen die Schlussfolgerung zu, dass sich am gesunden Myokard im Rahmen einer akuten kardialen Dekompensation im Tiermodell keine verlässlichen, pathologischen Veränderungen von STV-QT ableiten lassen. Bezüglich STV-RR waren die Ergebnisse insofern nicht überraschend, als dass mit zunehmender Herzfrequenz bekanntermaßen die Herzfrequenzvariabilität abnimmt. Hier wäre ein Vergleich mit einer entsprechend tachykarden Kontrollgruppe nötig, um festzustellen, ob die registrierte Abnahme von STV-RR über die Grenzen des Normalen hinausgeht. Alternativ wäre auch eine erneute Evaluation in demselben Setting denkbar, jedoch mit Induktion einer strukturellen Veränderung des Myokards im Vorfeld zum Akutversuch. Da die Untersuchung an primär herzgesunden Tieren durchgeführt wurde, stellen diese Ergebnisse in keiner Weise den Stellenwert von STV-QT und STV-RR im Zusammenhang mit CHF und SCD in Frage. Vielmehr stützen sie die Theorie, dass strukturelle Veränderungen am Myokard, z.B. im Rahmen des kardialen Remodellings, für die entsprechenden pathologischen EKG-Veränderungen ursächlich sind. Ob die beiden Parameter sinnvoll beim Monitoring von Intensivpatienten zum Einsatz kommen könnten, lässt sich mit den erhaltenen Daten nicht abschließend beantworten. Unter den gegeben Versuchsbedingungen erfolgte die kardiale Dekompensation so rasch, dass die Herzfrequenz sich als sensitivster und zuverlässigster Parameter erwies, die akute Kreislaufbelastung anzuzeigen. Interessant bleibt die Frage, wie sich die beiden Parameter bei primär herzkranken Patienten im Rahmen einer kardialen Dekompensation verhalten, die sich - insbesondere wenn sie langsam und stetig erfolgt - erst spät auf einen klassischen Vitalparameter wie die Herzfrequenz auswirken kann.
Embryonale Stammzellen (ESCs) sind durch zwei charakteristische Eigenschaften definiert. Neben einer kontinuierlichen Selbsterneuerungskapazität weisen ESCs die Fähigkeit auf, in alle Zelltypen der drei Keimblätter differenzieren zu können. Diese Eigenschaften werden unter anderem durch ein Netzwerk wichtiger Pluripotenzfaktoren als auch durch epigenetische Mechanismen reguliert, welche die Transkription von Pluripotenz- und Differenzierungsgenen kontrollieren.
In murinen ESCs sind an der Repression von Differenzierungsgenen auch Polycomb group (PcG) Proteine beteiligt. Diese Proteine bauen zwei Chromatin-modifizierende Komplexe auf, die als Polycomb repressive complex 1 bzw. 2 (PRC1 bzw. PRC2) bezeichnet werden. Nach dem klassischen Modell der Polycombfunktion, katalysieren PRC1 und PRC2 gemeinsam zwei charakteristische Histonmodifikationen, die zur Repression PRC-spezifischer Zielgene beitragen. Zahlreiche Studien in den letzten Jahren belegen, dass der Proteinaufbau der PRC1 Komplexe stark variieren kann, wobei die Familie der Polycomb group RING finger (Pcgf) Proteine eine wichtige Rolle spielt. In diesem Zusammenhang definieren einzelne Pcgf Paraloge (Pcgf1 – 6) verschiedene PRC1 Varianten (PRC1.1 – 1.6), die Komplex-spezifische Bindestellen im Genom aufweisen. Diese Erkenntnisse lassen auf unterschiedliche Mechanismen der PRC1 Varianten und Pcgf Paralog-spezifische Funktionen schließen, die zum jetzigen Zeitpunkt nur wenig erforscht sind.
Für manche Pcgf Paraloge sind wichtige Rollen in verschiedenen Stammzelltypen und während der iPS Reprogrammierung bekannt. Pcgf1 (Nspc1), Pcgf2 (Mel18) und Pcgf4 (Bmi1) zeigen eine Funktion in verschiedenen adulten Stammzellen. Pcgf4 spielt darüber hinaus eine wichtige Rolle in der murinen iPS Reprogrammierung. Für Pcgf6 (Mblr) wird eine Pluripotenz-assoziierte Funktion angenommen, denn Pcgf6 ist das einzige Pcgf Paralog, das eine erhöhte Expression in murinen ESCs aufweist, die jedoch im Verlauf der ESC-Differenzierung absinkt. Außerdem zeigen murine Pcgf6 KD ESCs eine verminderte Expression der Pluripotenzgene Oct4, Sox2 und Nanog, eine De-Repression mesodermaler und Testes-spezifischer Gene als auch eine erhöhte Tendenz zur hämatopoetischen Differenzierung. Wie genau Pcgf6 an der Regulation dieser Prozesse in murinen ESCs beteiligt ist, ist nicht bekannt.
In der hier vorliegenden Dissertation wurde die Funktion von Pcgf6 in der murinen iPS Reprogrammierung untersucht. Da bereits für Pcgf4 eine Rolle in der Reprogrammierung somatischer Zellen gezeigt wurde und Pcgf6 eine erhöhte Expression in ESCs aufweist, wurde auch für Pcgf6 eine Funktion in der iPS Reprogrammierung angenommen. Zunächst konnte in dieser Arbeit gezeigt werden, dass Pcgf6 während der iPS Reprogrammierung verstärkt exprimiert wird und in iPS Zellen eine ESC-ähnliche Expression aufweist. Darüber hinaus konnte Pcgf6 in Kombination mit Oct4, Klf4 und c-Myc spezifisch den Transkriptionsfaktor Sox2 in der iPS Reprogrammierung ersetzen. Zudem wurden für OPKM-induzierte iPS Zellen charakteristische Eigenschaften pluripotenter Zellen nachgewiesen. Außerdem konnte eine Rolle von Pcgf6 als Enhancer-Faktor für die iPS Reprogrammierung ausgeschlossen werden, da die Überexpression von Pcgf6 zusammen mit den OSKM Faktoren keine additiven Effekte auf die Reprogrammierungseffizienz erzielte. Im Gegensatz dazu führte der Knockdown (KD) von Pcgf6 in embryonalen Mausfibroblasten (MEFs) zu verminderten Effizienzen nach OSKM Reprogrammierung. Darüber hinaus handelte es sich bei der Mehrheit der AP+ Kolonien, die unter Pcgf6 KD Konditionen entstanden, um partiell-reprogrammierte iPS Zellen.
Zusammengefasst zeigen die Ergebnisse der hier vorliegenden Arbeit, dass Pcgf6 ein neuer und essentieller Faktor der iPS Reprogrammierung ist, der in Kombination mit Oct4, Klf4 und c-Myc spezifisch den Transkriptionsfaktor Sox2 ersetzen kann.
Die Cholecystektomie gilt mit jährlich über 190.000 Operationen in Deutschland als Standardeingriff. Goldstandard für symptomatische Cholecystolithiasis ist die laparoskopische Multiport-Cholecystektomie, jedoch etabliert sich die „narbenlose“ Single-Port Cholecystektomie seit den letzten Jahren stetig als Alternative. Als Komplikation können sich nach beiden Operationsverfahren an Stellen der intraoperativen Faszieninzision Narbenhernien bilden. Die aktuelle Datenlage zur Inzidenz der Narbenhernien zeigt auf, dass eine längere Faszieninzision, wie sie bei der SLC-Methode primär vorliegt, mit einer höheren Trokarhernieninzidenz einhergehen könnte. Deshalb sollten die Operationsverfahren MLC und SLC diesbezüglich anhand des Patientenkollektivs und unter Berücksichtigung der prädispositionierenden Faktoren verglichen werden.
Die Untersuchung erfasst 161 Patienten, die im Zeitraum von Juli 2010 bis Dezember 2011 an der chirurgischen Universitätsklinik Würzburg an der Gallenblase mittels konventioneller 4-Port-Cholecystektomie (MLC) bzw. der wiederverwendbaren Single-Port Methode mittels X-Cone® System (SLC) operiert wurden. Patienteneigene Daten, Risikofaktoren, Operationsverfahren und intraoperative sowie postoperative Befunde des Kollektivs wurden evaluiert. Im Follow-up zwischen zwölf und 24 Monaten postoperativ erfolgten eine sonografische und körperliche Untersuchung auf Narbenhernien sowie eine Befragung anhand eines nicht-standardisierten Erhebungsbogens.
Es zeigen sich im Beobachtungszeitraum des Follow-ups keine signifikanten Unterschiede bezüglich der Hernienbildung, weder im Gesamtkollektiv (MLC-Hernien 9,6%, SLC-Hernien 7%; p=0,772) noch in der Subgruppe (MLC-Hernien 9,6%, SLC-Hernien 3,85%, p=0,165). Jedoch traten tendenziell mehr Hernien bei den MLC-Patienten auf. Die SLC-Patienten sind im Mittel signifikant jünger, weisen eine kürzere OP-Dauer auf, haben einen geringeren BMI, weisen einen geringeren ASA-Score auf und hospitalisierten kürzer als die MLC-Patienten. Weiterhin befinden sich signifikant mehr Frauen in der SLC-Gruppe. Die SLC-Hernienpatienten sind alle weiblich (MLC-Hernienpatienten: 40% weiblich), tendenziell jünger, weisen eine geringere OP-Dauer auf, haben einen geringeren BMI, weisen einen niedrigeren ASA-Score auf und hospitalisierten kürzer als die MLC-Hernienpatienten (je ns).
Die Hernienpatienten sind auf die Gruppe bezogen, der sie zugehörig sind (SLC oder MLC) tendenziell älter, wurden länger operiert, haben einen höheren BMI und weisen eine längere Hospitalisationszeit auf (je ns). Die meisten Hernienpatienten litten an chronischer Cholecystitis (75% SLC; 70% MLC), nur eine SLC-Hernienpatienten zeigte eine Wundheilungsstörung auf. Bei zwei (20%) MLC-Hernienpatienten wurde intraoperativ eine Herniotomie einer bereits bestehenden Hernie durchgeführt. Drei Hernien (30%) in der MLC-Gruppe waren zum Follow-up Zeitpunkt schon operiert, eine SLC-Hernienpatientin (25%) wurde schon operiert, leidet jedoch an einer Rezidivhernie. In der SLC-Gruppe traten mehr Konversionen (Reduced-Port) als in der MLC-Gruppe (Konversion zum offenen Verfahren) auf. Derselbe Trend zeigt sich bei den Hernienpatienten. Gründe für eine Entscheidung für das SLC-Verfahren sind vor allem Kosmetik und die Notwendigkeit von weniger Inzisionen.
Prozentual traten mit 9,6% mehr Hernien in der MLC-Gruppe als in der SLC-Gruppe (7% im Gesamtkollektiv bzw. 3.85% in der Subgruppe) auf. Die Hernieninzidenz liegt somit je etwas über den in der Literatur berichteten Prozentangaben (MLC 0 – 5,21%; SLC 0 – 4,76%). Jedoch liegen längere Follow-up Zeiten und eine palpatorisch- sonografische Untersuchung mit hoher Diagnosequote in der vorliegenden Arbeit vor, während in der Literatur hierzu oft keine Angaben zu finden sind. Es wurden potenzielle Risikofaktoren für die Entstehung einer Narbenhernie diskutiert: Geschlecht, hoher BMI, Wundinfektion, höherer ASA-Score und Nahtmaterial konnten nicht als prädisponierende Faktoren dargestellt werden. Lediglich höheres Alter stellte sich als potenzieller Risikofaktor dar. Abdominelle Voroperationen lagen bei den Hernienpatienten lediglich als Unterbauchoperationen vor, die nicht in das Narbengebiet der Cholecystektomie fallen und somit nicht als Risikofaktor betrachtet werden können. Auch die längere umbilikale Faszieninzision konnte nicht klar als Prädisposition dargestellt werden. Die Hernienneigung zeigt sich folglich multifaktoriell. Nicht alle in der Literatur beschriebenen Risikofaktoren konnten bestätigt werden. Die beiden Verfahren vergleichend, stellte sich bei SLC eine signifikant kürzere OP-Dauer dar, während in der Literatur meist gegensätzliche Angaben zu finden sind. Die Sicherheit des Verfahrens stellte sich gegeben dar, es traten keine biliären Komplikationen auf. Prozentual wurden geringfügig mehr Konversionen (Reduced Port bei SLC bzw. offenes Verfahren bei MLC) bei SLC durchgeführt, jedoch sind diese Komplikationen so nicht direkt vergleichbar, da eine Konversion zum offenen Verfahren eine schwerwiegendere Änderung des OP-Verfahrens, als nur ein zusätzlicher Trokar, darstellt. Das SLC-Verfahren zeigte sich auch bei Adipösen als durchführbar. Auch die Konversionsrate bei SLC konnte nicht in Verbindung mit Adipositas gebracht werden: Die zu Reduced-Port konvertierten Patienten waren alle nicht-adipös .Der BMI ist nicht alleiniges Schlüsselkriterium bei der Entscheidung für SLC oder MLC, auch die „body composition“ muss beachtet werden. Insgesamt waren die SLC Patienten in der vorliegenden Studie am zufriedensten mit dem OP-Ergebnis. Die kosmetischen Aspekte und weniger Schnitte waren Beweggründe für die Entscheidung von SLC. Das kosmetische Ergebnis nach SLC ist dem nach MLC überlegen, was auch die Literatur bestätigt. Die Sicherheit des Verfahrens hat stets oberste Priorität: Das SLC-Verfahren muss vom Operateur gut beherrscht werden, dieser muss eine Lernkurve durchlaufen haben, folglich eine gewisse Erfahrung haben und so das Risiko biliärer Komplikationen möglichst vermeiden können. Die eigenen Erfahrungen im Rahmen der Studie decken sich meist mit der Literatur, das SLC-Verfahren erweist sich als etabliert. Vor allem das hervorragende kosmetische Ergebnis der SLC-Operation ist ein Vorteil für den Patienten. Da sich das SLC-Verfahren in der vorliegenden Arbeit als sicher darstellen konnte, weniger Hernien auftraten, keine sonstigen Komplikationen (Konversion von SLC zum offenen Verfahren) vorlagen und aufgrund der Überlegenheit des kosmetischen Ergebnisses stellt SLC eine gute Alternative zu MLC dar.
Wir untersuchen zunächst das Hubbard-Modell des anisotropen Dreiecksgitters als effektive Beschreibung der Mott-Phase in verschiedenen organischen Verbindungen mit dreieckiger Gitterstruktur. Um die Eigenschaften am absoluten Nullpunkt zu bestimmen benutzen wir die variationelle Cluster Näherung (engl. variational cluster approximation VCA) und erhalten das Phasendiagramm als Funktion der Anisotropie und der Wechselwirkungsstärke. Wir finden für schwache Wechselwirkung ein Metall. Für starke Wechselwirkung finden wir je nach Stärke der Anisotropie eine Néel oder eine 120◦-Néel antiferromagnetische Ordnung. In einem Bereich mittlerer Wechselwirkung entsteht in der Nähe des isotropen Dreiecksgitters ein nichtmagnetischer Isolator. Der Metall-Isolator-Übergang hängt maßgeblich von der Anisotropie ab, genauso wie die Art der magnetischen Ordnung und das Erscheinen und die Ausdehnung der nichtmagnetischen Isolatorphase.
Spin-Bahn Kopplung ist der ausschlaggebende Parameter, der elektronische Bandmodelle in topologische Isolatoren wandelt. Spin-Bahn Kopplung im Allgemeinen beinhaltet auch den Rashba Term, der die SU(2) Symmetrie vollständig bricht. Sobald man auch Wechselwirkungen berücksichtigt, müssen sich viele theoretische Methoden auf die Analyse vereinfachter Modelle beschränken, die nur Spin-Bahn Kopplungen enthalten, welche die U(1) Symmetrie erhalten und damit eine Rashba Kopplung ausschließen. Wir versuchen diese bisher bestehende Lücke zu schließen und untersuchen das Kane-Mele Hubbard (KMH) Modell mit Rashba Spin-Bahn Kopplung und präsentieren eine systematische Analyse des Effekts der Rashba Spin-Bahn Kopplung in einem korrelierten zweidimensionalen topologischen Isolator. Wir wenden die VCA auf dieses Problem an und bestimmen das Phasendiagramm mit Wechselwirkung durch die Berechnung der lokalen Zustandsdichte, der Magnetisierung, der Einteilchenspektralfunktion und der Randzustände. Nach einer ausführlichen Auswertung des KMH-Modells, bei erhaltener U(1) Symmetrie, finden wir auch für endliche Wechselwirkung, dass eine zusätzliche Rashba Kopplung zu neuen elektronischen Phasen führt, wie eine metallische Phase und eine topologische Isolatorphase ohne Bandlücke in der lokalen Zustandsdichte, die aber eine direkte Bandlücke für jeden Wellenvektor besitzt.
Für eine Klasse von 5d Übergangsmetallen untersuchen wir ein KMH ähnliches Modell mit multidirektionaler Spin-Bahn Kopplung, das wegen seiner Relevanz für die Natrium-Iridate (engl. sodium iridate) als SI Modell bezeichnet wird. Diese intrinsische Kopplung bricht die SU(2) Symmetrie bereits vollständig und dennoch erhält man wegen der speziellen Form für starke Wechselwirkung wieder einen rotationssymmetrischen Néel-AFM Isolator. Der topologische Isolator des SIH-Modells ist adiabatisch mit dem des KMH-Modells verbunden, jedoch sind die Randströme hier nicht mehr spinpolarisiert.
Wir verallgemeinern das Konzept der Klein-Transformation, das bereits erfolgreich auf Spin-Hamiltonians angewandt wurde, und wenden es auf ein Hubbard-Modell mit rein imaginären spinabhängigen Hüpfen an, das im Grenzfall unendlicher Wechselwirkung in das Kitaev-Heisenberg Modell übergeht. Dadurch erhält man ein Modell des Dreiecksgitters mit reellen spinunabhängigen Hüpfen, das aber eine mehratomige Einheitszelle besitzt. Für schwache Wechselwirkung ist das System ein Dirac Halbmetall und für starke Wechselwirkung erhält man eine 120◦-Néel antiferromagnetische Ordnung. Für mittlere Wechselwirkung findet man aber einen relativ großen Bereich in dem eine nichtmagnetische Isolatorphase stabil ist. Unsere Ergebnisse deuten auf die mögliche Existenz einer Quanten Spinflüssigkeit hin.
The consequences of habitat change for human well-being are assumed to be especially extreme in Burkina Faso. The country is located in a highly drought-sensitive zone of West Africa, and small‐scale subsistence farmers may be especially affected if losses of biodiversity lead to changes in ecosystem functioning; many depend on more or less degraded lands for agricultural production.
The overall aim of the present thesis consequently was to characterize the functional traits of soil-organisms which are crucial for a productive and balanced soil environment in the study region – termites and ants. They are true ecosystem engineers whose activity alters the habitat. Through soil-turnover in the course of constructing biogenic structures of varying size and nature (mounds, nests, galleries, soil-sheetings, foraging-holes), they bioturbate huge amounts of soil masses and exert massive effects on soil structure, positively influencing the fertility, stability, aeration and water infiltration rate into soils; and they provide habitats for other species. In sub-Saharan Africa, ants and termites are the only active soil macrofauna during the long dry season; in the sub-Sahel zone of Burkina Faso, termites even represent the only active, quantitatively remarkable decomposers all year round. Since no information was available about the actual diversity of the focal arthropods, I divided the thesis in two main parts: In the first part, a baseline study, I assessed the local termite and ant fauna, and investigated their quantitative and qualitative response to changing habitat parameters resulting from increasing human impact (‘functional response traits’). In the second and applied part, I addressed the impact of the biogenic structures which are important for the restoration of degraded soils (‘functional effect traits’).
Two traditional agricultural systems characteristic for the study region were selected. Each system represented a land-use intensification gradient comprising four distinct habitats now differing in the magnitude of human intervention but formerly having the same initial state. The first disturbance gradient, the temporal cross-section of a traditional soil water conservation technique to restore degraded heavily encrusted, barren soil named Zaï in Ouahigouya (Yatenga province, sub-Sahel zone); the second disturbance gradient, an agriculture type using crop rotation and fallow as nutrient management techniques near Fada N’Gourma (Gourma province, North-Sudanese zone).
No standard protocol existed for the assessment of termite and ant diversity in semi-arid (agro-) ecosystems; two widely accepted standard protocols provided the basis for the newly revised and combined rapid assessment protocol ‘RAP’: the ALL protocol for leaf litter ants of Agosti and Alonso (2000), and the transect protocol for termites in tropical forests of Jones and Eggleton (2000). In each study site, three to four replicate transects were conducted during the rainy seasons (2004—2008).
The RAP-protocol turned out to be very effective to characterize, compare and monitor the taxonomic and functional diversity of termites and ants; between 70% and 90% of the estimated total species richness were collected on all levels (transects, habitats, regions). Together in both regions, 65 ant species (25 genera) and 39 termite species (13 genera) were collected. These findings represent the first records for Burkina Faso. The data indicate a high sensitivity of termites and ants to land-use intensification. The diversity strongly decreased with increasing anthropogenic impact in the North-Sudan region. In total, 53 ant species (23 genera) and 31 termite species (12 genera) were found. Very promising results concerning the recovery potential of the soil-arthropods’ diversity were gathered in the Zaï system. The diversity of both taxa strongly increased with increasing habitat rehabilitation – in total, 41 ant species (16 genera) and 33 termite species (11 genera) were collected. For both taxa significant differences could be noted in the shape of the density variations along the gradient. For instance termites: Fungus-growers showed the greatest adaptability to different management practices. The greatest variations between the habitats were observed in soil and grass-feeding termites. Whole functional groups were missing in heavily impacted habitats, e.g. soil-, grass-, and wood-feeders were absent in the degraded site in the sub-Sahel zone. Several environmental parameters could be identified which significantly explained a great part of the variations in the composition of the arthropods’ communities; they indicate the importance of the habitats’ structural complexity (vegetation structure) and concomitant effects on diurnal temperature and moisture fluctuations, the availability of food sources, and the soil-structure. The diversity of termites in the sub-Sahel region was strongly correlated with the crown-cover percentages, the topsoils’ sand-content, and the availability of litter; in the North-Sudan region with the cumulated woody plant basal area, the topsoils’ clay- and organic matter-content. The parameters identified for ant communities in the Zaï system, were the height of trees, the topsoils’ clay-content and air humidity; in the North-Sudan region the habitats’ crown-cover percentages, the quantity of litter and again the height of trees.
In the second part of the thesis, I first rapidly assessed the (natural) variations in the amount of epigeal soil-structures along the two disturbance gradients in order to judge the relative importance of termites and ants for soil-turnover. The results illustrated impressively that a) in all study sites, termites were the main bioturbators while ant structures were of minor importance for soil turn-over; b) earthworms and grass-feeding termites contributed significantly to soil turn-over in the more humid North-Sudan region; and c) the bioturbated soil mass varied between seasons and years, however, the relative importance of the different taxa seemed to be fairly constant. In the sub-Sahel zone, fungus-growing Odontotermes and Macrotermes species fully take over the important function of bioturbation, leading to the transport of huge amounts of fine-textured soil material to the surface; with increasing habitat restoration, coarse fragments decreased in the upper horizons and became concentrated deeper along the soil profile.
Consequently, in the applied part, I concentrated on the bioturbation activity of fungus-growing termites in the four main stages of the Zaï system: crusted bare soil (initial stage), millet field, young and old forest. In each of the four Zaï sites nine experimental blocks (each comprising four plots of 1m2) were used to stimulate the foraging activity of fungus-growing termites with different, locally available organic materials (Aristida kerstingii hay, Bombax costatum wooden blocks, compost and a control without any organic amendment). The experiment was conducted twice for the duration of four weeks (rainy season 2005, dry season 2006). The plots were regularly checked and the increase of the area covered by sheetings chronologically followed. After four weeks a) all sheeting-soil was collected, air dried and separately weighed according to the different genera, and b) the foraging-holes were counted and their diameter measured. Additionally, c) ponded water infiltration was measured in selected plots, and d) the physicochemical properties of sheeting-soil were analyzed. In case of complete consumption of the offered hay during the experimental 4-weeks-duration, the same procedure (a, b) was followed before adding new hay to the respective plot.
The comparison between the different plots, sites and seasons revealed clearly that hay was the most attractive bait; for each gram of hay removed, Odontotermes brought about 12 g soil to the surface, Macrotermes 4 g. Odontotermes was the only genus attracted by organic material to the degraded area, and was therefore the decisive primary physical ecosystem engineer in the Zaï system, initiating the restoration process. The mass of soil bioturbated in the course of foraging increased strongly from the degraded, barren towards the most rehabilitated reforested site. Combining all 36 experimental plots per Zaï stage, Odontotermes bioturbated 31.8 tons of soil per hectare and month dry season in the degraded area, and 32.4 tons ha-1 mon-1 in the millet fields; both genera moved 138.9 tons ha-1 mon-1 in the young and 215.5 tons ha-1 mon-1 in the old Zaï forest. Few comparable figures were found in the literature. In northern Burkina Faso, both genera constructed 20 tons of sheetings ha-1 mon-1 after mulching with a straw-wood mixture (Mando & Miedema 1997), and in Senegal, around 10 tons ha-1 mon-1 were moved in heavily foraged plots (Rouland et al. 2003). Within a site, soil turn-over and the number of foraging holes created was always highest in hay, followed by compost, then by wood and in the end control. The fungus-growers’ foraging-activity was leading to an enormous increase in surface pore space – after one month of induced foraging activity in hay-plots, the median number of foraging-holes increased from 142 m-2 in the degraded site up to 921 m-2 in the old Zaï forest. The creation of subterranean galleries and macropores significantly increased the water infiltration rate by a mean factor 2–4.
Laboratory analyses revealed that sheeting-soil differed strongly from the respective control soil as well as between the seasons, the food-type covered, and the two genera. Odontotermes-sheetings differed in more parameters than Macrotermes-sheetings, and dry season sheetings differed in more parameters (and more strongly) than rainy season sheetings. In the present study, soil organic matter, carbon and nitrogen contents were significantly increased in all dry season sheetings; in the rainy season mainly in those built on compost. Texture analysis pointed out that both genera used topsoil and soil from deeper horizons in varying mixture ratios, thereby supporting findings of Jouquet et al. (2006).
To summarize, the present thesis contributes to a better understanding of the functional response traits of termites and ants to changing environmental parameters resulting from increasing human impact. The RAP-protocol represents an easy-to-learn and very effective method to representatively characterize, compare and monitor the taxonomic and functional diversity of termites and ants. The experiment has provided conclusive evidence of the importance of the consideration of fungus-growing termites (particularly Odontotermes and Macrotermes species) when aiming to restore infertile, degraded and crusted soils and to maintain a sustainable agricultural production in the Sahel‐Sudanese zone of West Africa.
Molecular and developmental characterization of the Echinococcus multilocularis stem cell system

The metacestode larva of Echinococcus multilocularis is the causative agent of alveolar echinococcosis (AE), one of the most dangerous zoonotic diseases in the Northern Hemisphere. Unlike “typical” metacestode larvae from other tapeworms, it grows as a mass of interconnected vesicles which infiltrates the liver of the intermediate host, continuously forming new vesicles in the periphery. From these vesicles, protoscoleces (the infective form for the definitive host) are generated by asexual budding. It is thought that in E. multilocularis, as in other flatworms, undifferentiated stem cells (so-called germinative cells in cestodes and neoblasts in free-living flatworms) are the sole source of new cells for growth and development. Therefore, this cell population should be of central importance for the progression of AE.
In this work, I characterized the germinative cells of E. multilocularis, and demonstrate that they are indeed the only proliferating cells in metacestode vesicles. The germinative cells are a population of undifferentiated cells with similar morphology, and express high levels of transcripts of a novel non-autonomous retrotransposon family (ta-TRIMs). Experiments of recovery after hydroxyurea treatment suggest that individual germinative cells have extensive self-renewal capabilities. However, germinative cells also display heterogeneity at the molecular level, since only some of them express conserved homologs of fgfr, nanos and argonaute genes, suggesting the existence of several distinct sub-populations. Unlike free-living flatworms, cestode germinative cells lack chromatoid bodies. Furthermore, piwi and vasa orthologs are absent from the genomes of cestodes, and there is widespread expression of some conserved neoblast markers in E. multilocularis metacestode vesicles. All of these results suggest important differences between the stem cell systems of free-living flatworms and cestodes.
Furthermore, I describe molecular markers for differentiated cell types, including the nervous system, which allow for the tracing of germinative cell differentiation. Using these molecular markers, a previously undescribed nerve net was discovered in metacestode vesicles. Because the metacestode vesicles are non-motile, and the nerve net of the vesicle is independent of the nervous system of the protoscolex, we propose that it could serve as a neuroendocrine system. By means of bioinformatic analyses, 22 neuropeptide genes were discovered in the E. multilocularis genome. Many of these genes are expressed in metacestode vesicles, as well as in primary cell preparations undergoing complete metacestode regeneration. This suggests a possible role for these genes in metacestode development. In line with this hypothesis, one putative neuropeptide (RGFI-amide) was able to stimulate the proliferation of primary cells at a concentration of 10-7 M, and the corresponding gene was upregulated during metacestode regeneration.
Techniken des computergestützten Wirkstoffdesigns spielen eine wichtige Rolle bei der Entwicklung neuer Wirkstoffe. Die vorliegende Arbeit befasst sich sowohl mit der Entwicklung als auch mit der praktischen Anwendung von Methoden des strukturbasierten Wirkstoffdesigns. Die Arbeit glieder sich daher in zwei Teile.
Der erste Teil beschäftigt sich mit der Entwicklung von empirischen Scoring-Funktionen, die eine Schlüsselrolle im strukturbasierten computergestützen Wirkstoffdesign einnehmen. Grundlage dieser Arbeiten sind die empirischen Deskriptoren und Scoring-Funktionen aus dem SFCscore-Programmpaket.
Dabei wurde zunächst untersucht, wie sich die Zusammensetzung der Trainingsdaten auf die Vorhersagen von empirischen Scoring-Funktionen auswirkt. Durch die gezielte Zusammenstellung eines neuen Trainingsdatensatzes wurde versucht, die Spannweite der Vorhersagen zu vergrößern, um so vor allem eine bessere Erkennung von hoch- und niedrig-affinen Komplexen zu erreichen. Die resultierende Funktion erzielte vor allem im niedrig-affinen Bereich verbesserte Vorhersagen.
Der zweite Themenkomplex beschäftigt sich ebenfalls mit der verbesserten Separierung von aktiven und inaktiven Verbindungen. Durch den Einsatz der Machine Learning-Methode RandomForest wurden dazu Klassifizierungsmodelle abgeleitet, die im Unterschied zu den klassischen Scoring-Funktionen keinen genauen Score liefern, sondern die Verbindungen nach ihrer potentiellen Aktivität klassifizieren.
Am Beispiel des mykobakteriellen Enzyms InhA konnte gezeigt werden, dass derartige Modelle den klassischen Scoring-Funktionen im Bezug auf die Erkennung von aktiven Verbindungen deutlich überlegen sind.
Der RandomForest-Algorithmus wurde im nächsten Schritt auch verwendet, um eine neue Scoring-Funktion zur Vorhersage von Bindungsaffinitäten abzuleiten. Diese Funktion wurde unter dem Namen SFCscoreRF in das SFCscore-Programmpaket implementiert. Die Funktion unterschiedet sich in einigen wesentlichen Punkten von den ursprünglichen SFCscore-Funktionen.
Zum einen handelt es sich beim RF-Algorithmus um eine nicht-lineare Methode, die im Unterschied zu den klassischen Methoden, die zur Ableitung von Scoring-Funktionen eingesetzt werden, nicht von der Additivität der einzelnen Deskriptoren ausgeht. Der Algorithmus erlaubt außerdem die Verwendung aller verfügbaren SFCscore-Deskriptoren, was eine deutlich umfassendere Repräsentation von Protein-Ligand-Komplexen als Grundlage des Scorings ermöglicht. Für die Ableitung von SFCscoreRF wurden insgesamt 1005 Komplexe im Trainingsdatensatz verwendet. Dieser Datensatz ist somit einer der größten, die bisher für die Ableitung einer empirischen Scoring-Funktion verwendet wurden.
Die Evaluierung gegen zwei Benchmark-Datensätze ergab deutlich bessere Vorhersagen von SFCscoreRF im Vergleich zu den ursprünglichen SFCscore-Funktionen. Auch im internationalen Vergleich mit anderen Scoring-Funktion konnten für beide Datensätze Spitzenwerte erreicht werden.
Weitere ausgiebige Testungen im Rahmen einer Leave-Cluster-Out-Validierung und die Teilnahme am CSAR 2012 Benchmark Exercise ergaben, dass auch SFCscoreRF Performanceschwankungen bei der Anwendung an proteinspezifischen Datensätzen zeigt - ein Phänomen, dass bei Scoring-Funktionen immer beobachtet wird. Die Analyse der CSAR 2012-Datensätze ergab darüber hinaus wichtige Erkenntnisse im Bezug auf Vorhersage von gedockten Posen sowie über die statistische Signifikanz bei der Evaluierung von Scoring-Funktionen.
Die Tatsache, dass empirische Scoring-Funktionen innerhalb eines bestimmten chemischen Raums trainiert wurden, ist ein wichtiger Faktor für die protein-abhängigen Leistungsschwankungen, die in dieser Arbeit beobachtet wurden. Verlässliche Vorhersagen sind nur innerhalb des kalibrierten chemischen Raums möglich. In dieser Arbeit wurden verschiedene Ansätze untersucht, mit denen sich diese ``Applicability Domain'' für die SFCscore-Funktionen definieren lässt. Mit Hilfe von PCA-Analysen ist es gelungen die ``Applicability Domain'' einzelner Funktionen zu visualisieren. Zusätzlich wurden eine Reihe numerischer Deskriptoren getestet, mit den die Vorhersageverlässlichkeit basierend auf der ``Applicability Domain'' abgeschätzt werden könnte. Die RF-Proximity hat sich hier als vielversprechender Ausgangspunkt für weitere Entwicklungen erwiesen.
Der zweite Teil der Arbeit beschäftigt sich mit der Entwicklung neuer Inhibitoren für das Chaperon Hsp70, welches eine vielversprechende Zielstruktur für die Therapie des multiplen Myeloms darstellt.
Grundlage dieser Arbeiten war eine Leitstruktur, die in einer vorhergehenden Arbeit entdeckt wurde und die vermutlich an einer neuartigen Bindestelle in der Interface-Region zwischen den beiden großen Domänen von Hsp70 angreift.
Die Weiterentwicklung und Optimierung dieser Leitstruktur, eines Tetrahydroisochinolinon-Derivats, stand zunächst im Vordergrund. Anhand detaillierter Docking-Analysen wurde der potentielle Bindemodus der Leitstruktur in der Interfaceregion von Hsp70 untersucht. Basierend auf diesen Ergebnissen wurde eine Substanzbibliothek erstellt, die von Kooperationspartnern innerhalb der KFO 216 synthetisiert und biologisch getestet wurde. Die Struktur-Wirkungsbeziehungen, die sich aus diesen experimentellen Daten ableiten lassen, konnten teilweise gut mit den erstellten Docking-Modellen korreliert werden. Andere Effekte konnten anhand der Docking-Posen jedoch nicht erklärt werden. Für die Entwicklung neuer Derivate ist deswegen eine umfassendere experimentelle Charakterisierung und darauf aufbauend eine Verfeinerung der Bindungsmodelle notwendig.
Strukturell handelt es sich bei Hsp70 um ein Zwei-Domänen-System, dass verschiedene allostere Zustände einnehmen kann. Um die Auswirkungen der daraus folgenden Flexibilität auf die Stabilität der Struktur und die Bindung von Inhibitoren zu untersuchen, wurden molekulardynamische Simulationen für das Protein durchgeführt.
Diese zeigen, dass das Protein tatsächlich eine überdurchschnittlich hohe Flexibilität aufweist, die vor allem durch die relative Bewegung der beiden großen Domänen zueinander dominiert wird. Die Proteinkonformation die in der Kristallstruktur hscaz beobachtet wird, bleibt jedoch in ihrer Grundstruktur in allen vier durchgeführten Simulationen erhalten. Es konnten hingegen keine Hinweise dafür gefunden werden, dass die Mutationen, welche die für die strukturbasierten Arbeiten verwendete Kristallstruktur im Vergleich zum Wildtyp aufweist, einen kritischen Einfluss auf die Gesamtstabilität des Systems haben.
Obwohl die Interface-Region zwischen NBD und SBD also in allen Simulationen erhalten bleibt, wird die Konformation in diesem Bereich doch wesentlich durch die Domänenbewegung beeinflusst und variiert. Da dieser Proteinbereich den wahrscheinlichsten Angriffspunkt der Tetrahydroisochinolinone darstellt, wurde der Konformationsraum detailliert untersucht. Wie erwartet weist die Region eine nicht unerhebliche Flexibilität auf, welche zudem, im Sinne eines ``Induced-Fit''-Mechanismus, durch die Gegenwart eines Liganden (Apoptozol) stark beeinflusst wird. Es ist daher als sehr wahrscheinlich anzusehen, dass die Dynamik der Interface-Region auch einen wesentlichen Einfluss auf die Bindung der Tetrahydroisochinolinone hat. Molekuardynamische Berechnungen werden deswegen auch in zukünftige Arbeiten auf diesem Gebiet eine wichtige Rolle spielen.
Die Analysen zeigen zudem, dass die Konformation der Interface-Region eng mit der Konformation des gesamten Proteins - vor allem im Bezug auf die relative Stellung von SBD und NBD zueinander - verknüpft ist. Das untermauert die Hypothese, dass die Interface-Bindetasche einen Angriffspunkt für die Inhibtion des Proteins darstellt.
Object six Degrees of Freedom (6DOF) pose estimation is a fundamental problem in many practical robotic applications, where the target or an obstacle with a simple or complex shape can move fast in cluttered environments. In this thesis, a 6DOF pose estimation algorithm is developed based on the fused data from a time-of-flight camera and a color camera. The algorithm is divided into two stages, an annealed particle filter based coarse pose estimation stage and a gradient decent based accurate pose optimization stage. In the first stage, each particle is evaluated with sparse representation. In this stage, the large inter-frame motion of the target can be well handled. In the second stage, the range data based conventional Iterative Closest Point is extended by incorporating the target appearance information and used for calculating the accurate pose by refining the coarse estimate from the first stage. For dealing with significant illumination variations during the tracking, spherical harmonic illumination modeling is investigated and integrated into both stages. The robustness and accuracy of the proposed algorithm are demonstrated through experiments on various objects in both indoor and outdoor environments. Moreover, real-time performance can be achieved with graphics processing unit acceleration.
Recently a new state of matter was discovered in which the bulk insulating state in a material is accompanied by conducting surface or edge states. This new state of matter can be distinguished from a conventional insulator phase by the topological properties of its band structure which led to the name "topological insulators". Experimentally, topological insulator states are mostly found in systems characterized by a band inversion compared to conventional systems. In most topological insulator systems, this is caused by a combination of energetically close bands and spin orbit coupling. Such properties are found in systems with heavy elements like Hg and Bi. And indeed, the first experimental discovery of a topological insulator succeeded in HgTe quantum wells and later also in BiSb bulk systems.
Topological insulators are of large interest due to their unique properties: In 2-dimensional topological insulators one dimensional edge states form without the need of an external magnetic field (in contrast to the quantum Hall effect). These edge states feature a linear band dispersion, a so called Dirac dispersion. The quantum spin Hall states are helical edge states, which means they consist of counterpropagating oppositely spin polarized edge channels. They are therefore of great potential for spintronic applications as well as building blocks for new more exotic states like Majorana Fermions. 3-dimensional topological insulators feature 2-dimensional surface states with only one Dirac band (also called Dirac cone) on each surface and an interesting spin texture where spin and momentum are locked perpendicular to each other in the surface plane. This unique surface band structure is predicted to be able to host several exotic states like e.g. Majorana Fermions (in combination with superconductors) and magnetic monopole like excitations.
This PhD thesis will summarize the discovery of topological insulators and highlights the developments on their experimental observations. The work focuses on HgTe which is up to now the only topological insulator material where the expected properties are unambiguously demonstrated in transport experiments. In HgTe, the topological insulator properties arise from the inversion of the Gamma_6 and Gamma_8 bands. The band inversion in HgTe is due to a combination of a high spin orbit splitting in Te and large energy corrections (due to the mass-velocity term) to the energy levels in Hg. Bulk HgTe, however, is a semimetal, which means for the conversion into a topological insulator a band gap has to be opened. In two dimensions (HgTe quantum well structures) this is achieved via quantum confinement, which opens a band gap between the quantum well subbands. In three dimensions, strain is used to lift the degeneracy of the semimetallic Gamma_8 bands opening up a band gap.
The thesis is structured as follows:
- The first chapter of this thesis will give a brief overview on discoveries in the field of topological insulators. It focuses on works relevant to experimental results presented in the following chapters. This includes a short outline of the early predictions and a summary of important results concerning 2-dimensional topological insulators while the final section discusses observations concerning 3-dimensional topological insulators.
- The discovery of the quantum spin Hall effect in HgTe marked the first experimental observation of a topological insulator. Chapter 2 will focus on HgTe quantum wells and the quantum spin Hall effect.
Above a critical thickness, HgTe quantum wells are predicted to host the quantum spin Hall state, the signature of a 2-dimensional topological insulator. HgTe quantum wells exhibiting low carrier concentrations and at the same time high carrier mobilities are required to be able to measure the quantum spin Hall effect. The growth of such high quality HgTe quantum wells was one of the major goals for this work. Continuous optimization of the substrate preparation and growth conditions resulted in controlled carrier densities down to a few 10^10 cm^-2. At the same time, carrier mobilities exceeding 1 x 10^6 cm^2/Vs have been achieved, which provides mean free paths of several micrometers in the material. Thus the first experimental evidence for the existence of the quantum spin Hall edge states succeeded in transport experiments on microstructures: When the Fermi energy was located in the bulk band gap a residual quantized resistance of 2e^2/h was found. Further experiments focused on investigating the nature of transport in this regime. By non-local measurements the edge state character could be established. The measured non-local resistances corresponded well with predictions from the Landauer-Büttiker theory applied to transport in helical edge channels.
In a final set of experiments the spin polarization of the edge channels was investigated. Here, we could make use of the advantage that HgTe quantum well structures exhibit a large Rashba spin orbit splitting. In systems with a large Rashba spin orbit splitting a spin accumulation is expected to occur at the edge of the sample perpendicular to a current flow. This so-called spin Hall effect was then used as a spin injector and detector. Using split gate devices it was possible to bring spin Hall and quantum spin Hall state into direct contact, which enabled an all electrical detection of the spin polarization of the quantum spin Hall edge channels.
- HgTe as a 3-dimensional topological insulator will be presented in chapter 3. Straining the HgTe layer enables the observation of topological insulator behavior. It was found that strain can be easily implemented during growth by using CdTe substrates. CdTe has a slightly larger lattice constant than HgTe and therefore leads to tensile strain in the HgTe layer as long as the growth is pseudomorphic. Magnetotransport studies showed the emergence of quantum Hall transport with characteristic signatures of a Dirac type bandstructure. Thus, this result marks the first observation of the quantum Hall effect in the surface states of a 3-dimensional topological insulator.
Transport experiments on samples fitted with a top gate enabled the identification of contributions from individual surfaces. Furthermore, the surface state quantum Hall effect was found to be surprisingly stable, perturbations due to additional bulk transport could not be found, even at high carrier densities of the system.
- Chapters 4 - 6 serve as in depth overviews of selected works: Chapter 4 presents a detailed overview on the all electrical detection of the spin Hall effect in HgTe quantum wells. The detection of the spin polarization of the quantum spin Hall effect is shown in chapter 5 and chapter 6 gives a detailed overview on the quantum Hall effect originating from the topological surface state in strained bulk HgTe.
The investigations discussed in this thesis pioneered the experimental work on the transport properties of topological insulator systems. The understanding of the fundamental properties of topological insulators enables new experiments in which e.g. the inclusion of magnetic dopants or the interplay between topological insulator and superconductors can be investigated in detail.
Die Entwicklung von Clustern ist in den vergangenen zwei Dekaden zu einem äußerst beliebten Ziel der regionalen Wirtschaftsförderung geworden. Dieser Trend wird seitens der Wissenschaft recht kritisch betrachtet. Sie befürchtet, dass die Clusterförderung den jeweiligen Kontext zu wenig beachtet und sich zudem auf wenige Instrumente beschränkt, ohne alle Anknüpfungspunkte, die ihr die Clustertheorie bietet, auszuschöpfen. Allerdings muss sich die Wissenschaft eingestehen, dass sie den Anschluss an die Förderung verloren hat und sich daher mit einer fundierten Beratung schwer tut.
Die vorliegende Arbeit hat das Ziel diese Wissenslücke zu verkleinern, indem sie Clusterplattformen untersucht, die häufig die zentralen Umsetzungsorganisationen der clusterorientierten Wirtschaftsförderung sind. Im Zentrum stehen dabei die Fragen, wie diese Plattformen arbeiten und welche Möglichkeiten und Begrenzungen sie haben, um eine den Clustertheorien entsprechende Förderung zu betreiben. Untersuchungsgegenstand sind die Clusterplattformen des bayerischen Förderprogramms Cluster-Offensive Bayern.
Als theoretische Grundlage zur Analyse der Clusterplattformen wurde der Neo-Institutionalismus gewählt. Dieser soziologischen Theorie zufolge werden die Handlungen von Organisationen durch die Akteure in ihrem Umfeld bestimmt, auf deren Legitimitätszuweisungen sie angewiesen sind. Für den vorliegenden Fall heißt das, dass sich die Clusterplattformen in ihren Handlungen an die Erwartungen der zu fördernden Unternehmen und der politischen Auftraggeber anpassen müssen. Das wird dazu führen, dass die Plattformen keine theoretisch optimale Förderung betreiben können. Die Frage ist schließlich, welche Elemente der Clustertheorien sie gut und welche sie weniger gut fördern können. Um das zu beantworten, werden die Erwartungen der einzelnen Akteure auf der Basis von qualitativen Experten Interviews identifiziert und ihre Auswirkungen auf ausgewählte Elemente der Clustertheorien untersucht und diskutiert.
Die Untersuchung zeigt, dass die Anpassungen der Clusterplattformen an die Erwartungen der Akteure in ihrem Umfeld in der Tat sehr stark sind, was generell zu einer sehr kontextspezifischen Förderung führt. Allerdings wird von Maßnahmen Abstand genommen, die den partikularen Erwartungen widersprechen, obwohl sie für den Gesamtcluster bedeutsam sein können. Andere Aspekte der Clustertheorien, die von den Akteuren allgemein als sehr bedeutsam angesehen werden, sind hingegen im Werkzeugkasten der Plattformen überrepräsentiert. Bei vielen weiteren Elementen beeinflussen jedoch ganz praktische Umstände die Handlungsmöglichkeiten der Clusterplattformen. Grundsätzlich schöpfen die Clusterplattformen ihre Möglichkeiten dennoch weitestgehend aus. Für eine umfassende clustertheoretisch orientierte Wirtschaftsförderung ist daher vor allem für die Einbeziehung von weiteren Akteuren oder Programmen zu plädieren, welche die Handlungsdefizite der Clusterplattformen ausgleichen können.
