Kardiomyopathien sind Erkrankungen des Herzmuskels, die mit einer kardialen Funktionsstörung einhergehen. Formen, die ohne erkennbare Ursache zu einer progredienten Dilatation und Reduktion der Kontraktilität des linken Ventrikels führen, werden als idiopathische dilatative Kardiomyopathie (DCM) bezeichnet. Sie ist der Hauptgrund für schwere Herzinsuffizienz und die damit assoziierten Einschränkungen der Lebensqualität bei jungen Erwachsenen. Neben der beeinträchtigten kardialen Funktion weisen diese Patienten oftmals auch Veränderungen im Bereich der humoralen und zellulären Immunität auf. Ein Teil der Patienten entwickelt Autoantikörper, die sich gegen den kardialen β1-adrenergen Rezeptor richten und ihn ähnlich wie der natürliche Ligand Adrenalin aktivieren. Hieraus resultiert eine chronische Überstimulation des Rezeptors, die über eine initiale Hypertrophie dann zu einer eingeschränkten Pumpfunktion führt.
Nachdem sich die Therapie der Antikörper-vermittelten Immunkardiomyopathie bisher auf die Behandlung der Herzinsuffizienz und die Kontrolle der Herz-insuffizienzsymptome beschränkt, könnten β1-ECII-homologe Peptide als Antikörper-Fänger bei Antikörper-positiven Patienten nun einen kausalen Therapieansatz darstellen. In diesem Zusammenhang wurden ein aus 25 Aminosäuren bestehendes zyklisches Peptid, eine aus 18 Aminosäuren bestehende Zyklopeptid-Mutante und ihre jeweiligen linearen Äquivalente im Rattenmodell auf Antikörper-neutralisierende Effekte und potentielle therapeutische Wirksamkeit getestet. Das Rattenmodell ist hierfür besonders geeignet, da die Aminosäuresequenz der funktionell wichtigen zweiten extrazellulären Domäne des β1-adrenergen Rezeptors (β1-ECII) bei Mensch und Ratte absolut identisch ist.
Auf immunologischer Ebene konnte der Titer der krankheitsinduzierenden β1-ECII-Antikörper bereits nach der ersten Applikation zyklischer Peptide relevant gesenkt werden und nahm im weiteren Verlauf der Behandlung kontinuierlich ab. Nach Zyklopeptidgabe kam es am Herzen zu einer Reduktion des linksventrikulären Durchmessers und zu einer fast vollständigen Normalisierung der anatomischen Proportionen. Auf die Morphologie der Myozyten selbst und auch den Kollagengehalt des Gewebes hatte die Zyklopeptidtherapie keinen wesentlichen Einfluss. Die funktionellen Eigenschaften des Herzens ließen sich durch die Neutralisation stimulatorischer β1-ECII-Antikörper mittels intravenöser Zyklopeptidapplikation deutlich verbessern: Die Verkürzungsfraktion des linken Ventrikels und der Herzindex als Parameter für die kardiale Leistungsfähigkeit konnten durch die Behandlung wieder weitgehend normalisiert werden.
Diese im Tiermodell erzielten Ergebnisse lassen einen therapeutischen Effekt der Zyklopeptide vermuten. Der Ansatz einer spezifisch gegen Antikörper gerichteten Therapie zur Behandlung von Patienten mit β1-Antikörper-positiver Herzinsuffizienz erscheint daher vielversprechend.
The discovery of the Giant Magneto Resistance (GMR) effect in 1988 by Albert Fert [Baib 88] and Peter Grünberg [Bina 89] led to a rapid development of the field of spintronics and progress in the information technology. Semiconductor based spintronics, which appeared later, offered a possibility to combine storage and processing in a single monolithic device. A direct result is reduced heat dissipation. The observation of the spin Seebeck effect by Ushida [Uchi 08] in 2008 launched an increased interest and encouraged research in the field of spin caloritronics. Spintronics is about the coupling of charge and spin transport. Spin caloritronics studies the interaction between heat and spin currents. In contrast to spintronics and its variety of applications, a particular spin-caloritronic device has not yet been demonstrated. However, many of the novel phenomena in spin caloritronics can be detected in most spintronic devices. Moreover, thermoelectric effects might have a significant influence on spintronic device operation. This will be of particular interest for this work. Additional knowledge on the principle of coupling between heat and spin currents uncovers an alternative way to control heat dissipation and promises new device functionalities.
This thesis aims to further extend the knowledge on thermoelectrics in materials with strong spin-orbit coupling, in this case the prototypical ferromagnetic semiconductor (Ga,Mn)As. The study is focused on the thermoelectric / thermomagnetic effects at the interface between a normal metal and the ferromagnetic (Ga,Mn)As. In such systems, the different interfaces provide a condition for minimal phonon drag contribution to the thermal effects. This suggests that only band contributions (a diffusion transport regime) to these effects will be measured.
Chapter 2 begins with an introduction on the properties of the studied material system, and basics on thermoelectrics and spin caloritronics. The characteristic anisotropies of the (Ga,Mn)As density of states (DOS) and the corresponding magnetic properties are described. The DOS and magnetic anisotropies have an impact on the transport prop- erties of the material and that results in effects like tunneling anisotropic magnetores- istance (TAMR) [Goul 04]. Some of these effects will be used later as a reference to the results from thermoelectric / thermomagnetic measurements. The Fingerprint tech- nique [Papp 07a] is also described. The method gives an opportunity to easily study the anisotropies of materials in different device geometries.
Chapter 3 continues with the experimental observation of the diffusion thermopower of (Ga,Mn)As / Si-doped GaAs tunnel junction. A device geometry for measuring the diffusion thermopower is proposed. It consists of a Si - doped GaAs heating channel with a Low Temperature (LT) GaAs / (Ga,Mn)As contact (junction) in the middle of the channel. A single Ti / Au contact is fabricated on the top of the junction. For transport characterization, the device is immersed in liquid He. A heating current technique is used to create a temperature difference by local heating of the electron system on the Si:GaAs side. An AC current at low frequency is sent through the channel and it heats the electron population in it, while the junction remains at liquid He temperature (experimentally con- firmed). A temperature difference arises between the heating channel and the (Ga,Mn)As contact. As a result, a thermal (Seebeck) voltage develops across the junction, which we call tunnelling anisotropic magneto thermopower (TAMT), similar to TAMR. TAMT is detected by means of a standard lock-in technique at double the heating current frequency (at 2f ). The Seebeck voltage is found to be linear with the temperature difference. That dependence suggests a diffusion transport regime. Lattice (phonon drag) contribution to the thermovoltage, which is usually highly nonlinear with temperature, is not observed.
The value of the Seebeck coefficient of the junction at 4.2 K is estimated to be 0.5 µV/K.
It is about three orders of magnitude smaller than the previously reported one [Pu 06]. Subsequently, the thermal voltage is studied in external magnetic fields. It is found that the thermopower is anisotropic with the magnetization direction. The anisotropy is explained with the anisotropies of the (Ga,Mn)As contact. Further, switching events are detected in the thermopower when the magnetic field is swept from negative to positive fields. The switchings remind of a spin valve signal and is similar to the results from previous experiments on spin injection using a (Ga,Mn)As contacts in a non-local detection scheme. That shows the importance of the thermoelectric effects and their possible contribution to the spin injection measurements. A polar plot of the collected switching fields for different magnetization angles reveals a biaxial anisotropy and resembles earlier TAMR measurements of (Ga,Mn)As tunnel junction. A simple cartoon model is introduced to describe and estimate the expected thermopower of the studied junction. The model yields a Fermi level inside of the (Ga,Mn)As valence band. Moreover, the model is found to be in good agreement with the experimental results.
The Nernst effect of a (Ga,Mn)As / GaAs tunnel junction is studied in Chapter 4. A modified device geometry is introduced for this purpose. Instead of a single contact on the top of the square junction, four small contacts are fabricated to detect the Nernst signal. A temperature difference is maintained by means of a heating current technique described in Chapter 3. A magnetic field is applied parallel to the device plane. A voltage drop across two opposite contacts is detected at 2f. It appears that a simple cosine function with a parameter the angle between the magnetization and the [100] crystal direction in the (Ga,Mn)As layer manages to describe this signal which is attributed to the anomalous Nernst effect (ANE) of the ferromagnetic contact. Its symmetry is different than the Seebeck effect of the junction. For the temperature range of the thermopower measurements the ANE coefficient has a linear dependence on the temperature difference (∆T). For higher ∆T, a nonlinear dependence is observed for the coefficient. The ANE coefficient is found to be several orders of magnitude smaller than any Nernst coefficient in the literature. Both the temperature difference and the size of the ANE coefficient require further studies and analysis. Switching events are present in the measured Nernst signal when the magnetic field is swept from positive to negative values. These switchings are related to the switching fields in the ferromagnetic (Ga,Mn)As. Usually, there are two states which are present in TAMR or AMR measurements - low and high resistance. Instead of that, the Nernst signal appears to have three states - high, middle and low thermomagnetic voltage. That behaviour is governed not only by the magnetization, but also by the characteristic of the Nernst geometry.
Chapter 5 summarizes the main observations of this thesis and contains ideas for future work and experiments.
Es werden Daten zur Entwicklung von 99 Kindern zwischen 13 und 36 Monaten, die in deutschen Kindertagesstätten betreut wurden, für die Bereiche Motorik, Sprache vorgestellt. Die Erhebung beruht auf dem Konzept des Pikler-Instituts in Budapest/Ungarn, mit dem die Entwicklung von mehreren tausend Kindern dokumentiert wurde. In den vorläufigen Schlussfolgerungen werden die aktuellen Diskussionen zur Tagesbetreuung von Kleinkindern berücksichtigt.
Der Wnt Signalweg spielt eine entscheidende Rolle in der Embryogenese durch Steuerung der Proliferation, Apoptose, Differenzierung und der Festlegung der Körperachsen im frühen Embryo. Eine Fehlregulation des Signalwegs durch Mutationen in einem der Proteine und Gene dieser hochkomplexen Signalkaskade kann fatale Folgen haben, und ist ein erster Schritt auf dem Weg der Krebsentstehung. Dabei spielt das Protein β-Catenin eine Schlüsselrolle im kanonischen Zweig des Wnt Signalwegs. Durch Steuerung seiner Konzentration im Zytoplasma wird die Expression seiner direkten Zielgene reguliert, da β-Catenin im aktiven Signalweg als Co-Transkriptionsfaktor agiert. Durch Sichtbarmachung dieses Proteins durch fluoreszierende Reportergenkonstrukte kann der Aktivitätsstatus des Wnt Signalwegs in der Zelle beobachtet werden. Das ermöglicht zum einen genaue Analysen des Signalwegs, wie zum Beispiel das Studium des Zusammenspiels mit anderen Signalwegen. Vor allem aber erlaubt es die gezielte Suche nach Wnt-Signalwegs-modulierenden Substanzen als potentielle Wirkstoffe in der Krebsmedikamentenentwicklung. In der vorliegenden Arbeit wurden mehrere Reportergenkonstrukte für die stabile Transfektion von Zelllinien entwickelt und hinsichtlich eines möglichen Einsatzes sowohl in der Forschung, als auch in Wirkstoffscreenings validiert. Dies umfasst sowohl mehrere Reporter mit β-Catenin als Fusionsprotein, als auch Wnt-Promoter-regulierte eGFP-Reporter, die den Akitvitätsstatus des Wnt-Signalwegs anzeigen. Mit Hilfe dieser Reporter konnten Untersuchungen zur Wirkung des Wnt-Signalwegs auf die Morphologie von transfizierten und nicht-transfizierten MDCK-Zellen durchgeführt werden. Überdies wurde ein promotorregulierter eGFP-Reporter konstruiert, mit welchem transfizierte Zellen mit aktiviertem Wnt-Signalweg aus einem Zellpool gefischt werden können. Diese Methode ist sowohl für den Einsatz in kultivierten Zelllinien, als auch in der Diagnostik nach der Transfektion primärer Zellen geeignet. Auf Grundlage der neuen Zelllinien wurde weiterhin ein neuer Screeningansatz für potentielle Wnt-Signalwegsinhibitoren entwickelt, der auf dem Ausbleichen der Fluoreszenz in einem Well einer Multiwell-Kulturplatte beruht.
In this work the synthesis of dendritic macromolecules and small redox cascades was reported and studies of their energy and electron transfer properties discussed.
The chromophores in the dendrimers and the redox cascades are linked via triazoles, which were built up by CuAAC. Thereby, a synthetic concept based on building blocks was implemented, which allowed the exchange of all basic components. Resulting structures include dendrimers composed exclusively of TAAs (G1–G3), dendrimers with an incorporated spirobifluorene core (spiro-G1 and spiro-G2) and the donor-acceptor dendrimer D-A-G1, in which the terminal groups are exchanged by NDIs.
Furthermore, a series of model compounds was synthesised in order to achieve a better understanding of the photophysical processes in the dendrimers.
A modification of the synthetic concept for dendrimers enabled the synthesis of a series of donor-acceptor triads (T-Me, T-Cl and T-CN) consisting of two TAA donors and one NDI acceptor unit. The intermediate TAA chromophore ensured a downhill redox gradient from the NDI to the terminal TAA, which was proved by cyclic voltammetry measurements. The redox potential of the intermediate TAA was adjusted by different redox determining substituents in the “free” p-position of the TAA. Additionally, two dyads (Da and Db) were synthesised which differ in the junction of the triazole to the TAA or the NDI, respectively. In these cascades a nodal-plane along the N-N-axes in the NDI and a large twist angle between the NDI and the N-aryl substituent guaranteed a small electronic coupling.
The photophysical investigations of the dendrimers focused on the homo-energy transfer properties in the TAA dendrimers G1–G3. Steady-state emission spectroscopy revealed that the emission takes place from a charge transfer state. The polar excited state resulted in a strong Stokes shift of the emission, which in turn led to a small spectral overlap integral between the absorption of the acceptor and the emission of the donor in the solvent relaxed state. According to the Förster theory, the overlap integral strongly determines the energy transfer rate. Fluorescence up-conversion measurements showed a strong and rapid initial fluorescence anisotropy decay and a much slower decrease on the longer time scale. The experiment revealed a fast energy transfer in the first 2 ps followed by a much slower energy hopping. Time resolved emission spectra (TRES) of the model compound M indicated a solvent relaxation on the same time scale as the fast energy transfer.
The Förster estimation of energy transfer rates in G1 explains fast energy transfer in the vibrotionally relaxed state before solvent relaxation starts. Thereby, the emission spectrum of G1 in cyclohexane served as the time zero spectrum. Thus, solvent relaxation and fast energy transfer compete in the first two ps after excitation and it is crucial to discriminate between energy transfer in the Franck-Condon and in the solvent relaxed state. Furthermore, this finding demonstrates that fast energy transfer occurs even in charge transfer systems where a large Stokes shift prevents an effective spectral overlap integral if there is a sufficient overlap integral in before solvent relaxation.
Energy transfer upon excitation was also observed in the spiro dendrimers spiro-G1 and spiro-G2 and identified by steady-state emission anisotropy measurements. It was assumed that the energy in spiro-G1 is completely distributed over the entire molecule while the energy in spiro-G2 is probably distributed over only one individual branch. This finding was based on a more polarised emission of spiro-G2 compared to spiro-G1. This issue has to be ascertained by e.g. time resolved emission anisotropy measurements in further energy transfer studies.
Concerning the electron transfer properties of TAA-triazole systems the radical cations of G1–G2, spiro-G1 and spiro-G2 and of the model compound M were investigated by steady-state absorption spectroscopy. Experiments showed that the triazole bridge exhibits small electronic communication between the adjacent chromophores but still possesses sufficient electronic coupling to allow an effective electron transfer from one chromophore to the other.
Due to the high density of chromophores, their D-A-D structure and their superficial centrosymmetry, the presented dendrimers are prospective candidates for two-photon absorption applications.
The dyads, triads and the donor-acceptor dendrimer D-A-G1 were investigated regarding their photoinduced electron transfer properties and the effects that dominate charge separation and charge recombination in these systems.
The steady-state absorption spectra of all cascades elucidated a superposition of the absorption characteristics of the individual subunits and spectra indicated that the chromophores do not interact in the electronic ground state.
Time resolved transient absorption spectroscopy of the cascades was performed in the fs- and ns-time regime in MeCN and toluene as solvent. Measurements revealed that upon with 28200 cm-1 (355) nm and 26300 cm-1 (380 nm), respectively, an electron is transferred from the TAA towards the NDI unit yielding a CS state. In the triads at first a CS1 state is populated, in which the NDI is reduced and the intermediate TAA1 is oxidised. Subsequently, an additional electron transfer from the terminal TAA2 to TAA1 led to the fully CS2 state. Fully CS states of the dyads and triads exhibit lifetimes in the ns-time regime. In contrast for Db in MeCN, a lifetime of 43 ps was observed for the CS state together with the population of a 3NDI state. The signals of the other CS states decay biexponentially, which is a result of the presence of the 1CS and the 3CS states. While magnetic field dependent measurements of Db did not show an effect due to the large singlet-triplet splitting, T-CN exhibited a strong magnetic field dependence which is an evidence for the 1CS/3CS assignment. Further analysis of the singlet-triplet dynamics are required and are currently in progress.
Charge recombination occurred in the Marcus inverted region for compounds solved in toluene and in the Marcus normal region for MeCN as solvent. However, a significant inverted region effect was observed only for Db. Triads are probably characterised by charge recombination rates in the inverted and in the normal region near to the vertex of the Marcus parabola. Hence the inverted region effect is not pronounced and the rate charge recombination rates are all in the same magnitude. However, compared to the charge recombination rate of Db the enlarged spatial distance between the terminal TAA and the NDI in the fully CS2 states in the triads resulted in reduced charge recombination rates by ca. one order of magnitude.
More important than a small charge recombination rate is an overall lifetime of the CS states and this lifetime can significantly be enhanced by the population of the 3CS state. The reported results reveal that a larger singlet-triplet splitting in the dyads led to a CS state lifetime in the us time regime while a lifetime in the ns-time regime was observed in cases of the triads. Moreover, the singlet-triplet splitting was found to be solvent dependent in the triads, which is a promising starting point for further investigations concerning singlet-triplet splitting.
The donor-acceptor dendrimer D-A-G1 showed similar characteristics to the dyads. The generation of a CS state is assumed due to a clear NDI radical anion band in the transient absorption spectrum. Noteworthy, the typical transient absorption band of the TAA radical cation is absent for D A-G1 in toluene. Bixon-Jortner analysis yielded a similar electronic coupling in D-A-G1 compared to the dyads. However, the charge recombination rate is smaller than of Db due to a more energetic CS state, which in the inverted region slows down charge recombination. In combination a singlet-triplet splitting similar to the dyads prolongs the CS state lifetime up to 14 us in diluted solution. Both effects result in an even better performance of D-A-G1 concerning energy conversion. D A-G1 is therefore a promising key structure for further studies on light harvesting applications. In a prospective study a second generation donor-acceptor dendrimer D-A-G2 might be an attractive structure accessible by “click reaction” of 13 and 8. D-A-G2 is expected to exhibit a downhill oriented gradient of CS states as assumed from the CV studies on G1–G3.
In this work the synthesis, the spectroscopic and electrochemical investigation as well as some applications of a broad diversity of indolenine squaraine dyes were presented. This diversity was based on two parent squaraine dyes, one standard trans-configured compound (M1) and one in which one central oxygen atom was replaced by a dicyanomethylene moiety (M2), which increased the acceptor strength and induced a cis-configuration. The variety of synthesised dyes included functionalised squaraine monomers, donor- and acceptor-substituted monomeric model squaraines, donor- and acceptor-squaraine copolymers, pure squaraine homopolymers, a squaraine-squaraine copolymer, as well as some conjugated cyclic oligomers.
In order to be able to synthesise all these different kinds of dyes, several bromine and boronic ester derivatives were synthesised, which enabled the use of the Suzuki cross coupling reaction, to generate model dyes and copolymers. In addition, the bromine derivatives were used to carry out the Yamamoto homocoupling reaction to the respective homopolymers and macrocycles.
The absorption maximum of unsubstituted reference dye M1 was found at ~ 15500 cm–1, while that of M2 was red-shifted to ~ 14300 cm–1 due to the increased acceptor strength of the central unit. The extinction coefficients were in the order of ~ 300000 M–1 cm–1 and ~ 200000 M–1 cm–1, respectively. It was found that the implementation of functional groups (M3–M9), additional electron donors (M10–M19) or acceptors (M20–M22) at the periphery lead to bathochromic shifts of the absorption depending on the strength of either - and/or -donating properties of the substituents.
For the bis- and triarylamine substituted dyes M10–M13 and the dibrominated dyes M5 and M7 the electronic structure of the mono- and diradical (di)cations was explored using the interplay of cyclic voltammetry, spectroelectrochemistry, and DFT calculations. It was demonstrated that the monoradical cations still show a cyanine-like character and are delocalised Robin-Day class III species due to the low redox potential of the squaraine bridge between the additional amine redox centres. To the best of my knowledge, this made M13+∙, with an N-N-distance of 26 bonds between the additional redox centres to the longest bis(triarylamine) radical cation that is completely delocalised. For the diradical dications, the situation was of larger complexity. The computed most stable energetic state of the dianisylamine-substituted dyes turned out to be a broken-symmetry state with almost equal contributions of an open-shell singlet and triplet state. In addition, it was shown that the HOMO–1→HOMO transition dominated the absorption spectra of the diradical dications where the trans-/cis-configuration of the squaraines had a direct impact due to symmetry reasons.
Based on the donor–squaraine model compounds M10–M19, a series of donor–squaraine copolymers was synthesised (P7–P12) in order to further red shift and broaden the low energy absorption band. However, these effects were only of marginal extent. Both the optical and the electrochemical derived band gaps were barely lowered compared to the respective monomeric model dyes. This was assigned to an increased squaraine-squaraine distance and resulting lower exciton coupling between the squaraine chromophores due to the bridging units. In addition, according to semiempirical calculations the bridges were twisted out of the squaraine plane what reduced conjugational effects between the chromophores. To sum up, the idea to insert additional electron rich bridging units in order to create copolymers with broad and red-shifted absorption did not fully work out for the presented systems.
The addition of strong electron accepting NDI units at the periphery resulted in M21, the most unique monomeric model squaraine in this work. The common picture of a sharp low energy squaraine absorption completely altered due to the addition of the NDIs and a rather broad and solvent dependent low energy absorption was found. Spectroelectrochemical experiments and semiempirical calculations showed that this band is a superposition of the common squaraine HOMO→LUMO transition and a partial squaraine→NDI charge transfer transition. The latter was lost upon oxidation of the squaraine and the absorption spectrum of the monocation of M21 was found to be nearly a 1:1 image of a pure squaraine monocation. Both the monomeric model M21 and the respective copolymer P13 showed low electrochemically obtained band gaps of 1.05–1.20 eV, which were the lowest of all squaraines in this work. For both dyes, transient absorption measurements in the fs-time regime revealed the ultrafast formation of a CS state via an intermediate CT state within a few ps. Besides, charge recombination to the ground state also occured within a few ps. In the polymer, there was barely any further energy or charge transfer within the excited state lifetime and therefore the CS state was confined on adjacent squaraine-NDI pairs and did not further travel along the polymer strand.
The Ni-mediated Yamamoto homocoupling reaction was applied for the synthesis of the homopolymers (P1–P5). In contrast to the donor–squaraine copolymers, those polymers revealed strongly red-shifted and broad absorption in the red to NIR region in addition to a sharp fluorescence. These features could be explained to originate mainly from the exciton coupling of localised excited states and the presence of different superstructures in solution. For the polymers P1 and P2, an elongated J-type polymer chain caused the strong lowest energy absorption band whereas a zig-zag type arrangement of the single chromophores lead to transitions into both low and high energy excited states of the excitonic manifold. For the polymers P3 and P4, several polymer fractions of different size were investigated. Here, also an elongated chain with J-type character induced the lowest energy absorption band whereas a helical H-type arrangement caused transitions to higher energies of the excitonic manifold. The fractions to which these structures were formed depended on the chain length and the solvent. In thin film measurements, it was shown that the initially in solution formed superstructures were partly retained in the thin film but could be altered by annealing procedures. A control of the superstructures should enable the controlled tuning of the optical properties. Despite the strong interaction of the chromophores in the excited state, the redox potentials of the homopolymers barely differed to those of the respective reference dyes, indicating negligible electronic interaction in the ground state.
In addition squaraine-squaraine copolymer P6, consisting of alternating parent dyes M1 and M2, was synthesised. Likewise to the homopolymers, a broad and red-shifted absorption was observed. This was explained by exciton coupling theory, which was extended to also suit alternating copolymers. In toluene, an extraordinary narrow and intense lowest energy absorption band was observed. This exchange narrowing might be a result of a highly ordered J-type structure of the polymer especially in this solvent because it was not found in others. The features of the polymer may be compared to typical J-aggregates formed from monomeric cyanine molecules for example and the polymer used as model for excitonic interactions in an alternating copolymer. Transient absorption measurements revealed a strong energy dependence of the decay traces of the copolymer, most strikingly at early decay times. This was assigned to the occurrence of multiple excitations of one polymer strand (due to the large extinction coefficients of the polymer) and resulting exciton-exciton annihilation. Due to the large exciton diffusion constants that were estimated, the static exciton-exciton annihilation was the rate limiting process of the decay, in contrast to other conjugated polymers, where in thin film measurements the decay was diffusion controlled.
To sum up, for the polymers consisting of exclusively squaraine chromophores, it was shown that the exciton coupling of single chromophores with strong transition dipole moments was a fruitful way to tune the absorption spectra.
As a side product of some of the polycondensation reactions, unprecedented cyclic conjugated oligomers such as the triarylamine-bridged dimer Dim1, the cyclic homotrimers Tri1–Tri3, and the tetramer Tet1 were obtained by recycling GPC in low yields. Especially the cyclic trimers showed unusual absorption and even more extraordinary fluorescence properties. They showed multiple fluorescence bands in the NIR that covered a range from ~ 8000–12500 cm–1 (800–1250 nm). First hints from theoretical calculations indicated that the trimer was not fully planar but comprised a mixture of both planar and bent single squaraine chromophores. However, final results of the calculations were still missing at the time of writing.
In the last part of this work, the application of some monomeric and polymeric squaraines in binary and ternary bulk heterojunction solar cells was demonstrated. Also the utilisation as a dopant in a polymer matrix in an OLED device was shown. The homopolymers P1–P4 were tested in the binary BHJ solar cells revealing poor performances and especially very low short circuit currents. The utilisation of the polymers P3 and P4 that carried the dicyanomethylene group resulted in higher open circuit voltages due to the lower LUMO energy levels but still an overall poor performance. Neither for the different alkyl chains nor for the size of the polymers was a trend observed. In the ternary BHJ solar cells, small amounts of either monomer M14 or polymers P1A, P4–1 or P13 were added to a P3HT/PCBM system in order to generate an additional pathway for charge or energy transfer that should result in a better device performance. However, for none of the tested squaraines, improved solar cells could be built. In similarity to the binary solar cells, the short circuit currents were lower compared to a P3HT/PCBM reference device. These low short circuit currents indicated that the morphology of the squaraine dyes was the major limitation in those devices. It is possible that the dimethyl groups at the indolenine hindered a favoured alignment of the compounds that would allow decent charge transport. In the squaraine doped OLED the squaraine M6 worked rather well as an NIR emitter. Already at low dye loads the fluorescence of the host polymer SY-PPV was completely quenchend and emission from the squaraine was observed. For electroluminescence measurements, a lower dye load (0.5 wt.%) compared to the photoluminescence measurements was sufficient, indicating that apart from FRET additional quenching mechanisms were at work in the electrically driven devices such as charge carrier dynamics.
Dockingbasierte Ansätze zählen zu den wichtigsten Komponenten im virtuellen Screening. Sie dienen der Vorhersage der Ligandposition und -konformation in der Bindetasche sowie der Abschätzung der Bindungsaffinität zum Protein. Bis heute stellt die korrekte Identifizierung proteingebundener Ligandkonformationen ein noch nicht vollständig gelöstes Problem für Scoringfunktionen dar. Der erste Teil der vorliegenden Arbeit ist daher der Entwicklung computergestützter Methoden zur Bewertung von Docking-Lösungen gewidmet.
Der Fokus eines ersten Teilprojektes lag auf der Berücksichtigung der Absättigung vergrabener Wasserstoffbrückenakzeptoren (HBA) und -donoren (HBD) bei der Bewertung von Docking-Lösungen. Nicht-abgesättigte vergrabene HBA und HBD stellen einen der Bindungsaffinität abträglichen Beitrag dar, der bis dato aufgrund fehlender Struktur- bzw. Affinitätsdaten in Scoringfunktionen vernachlässigt wird. Im Rahmen der vorliegenden Arbeit wurde auf der Basis einer detaillierten Untersuchung zur Häufigkeit vergrabener nicht-abgesättigter HBA und HBD in hochaufgelösten Protein-Ligand-Komplexen des Hartshorn-Datensatzes eine empirische Filterfunktion („vnaHB“-Filterfunktion) entwickelt, die unerwünschte Ligandbindeposen erkennt und von der Bewertung mittels Scoringfunktionen ausschließt. Der praktische Nutzen der empirischen Filterfunktion wurde für die Scoringfunktionen SFCscore und DSX anhand vorgenerierter Docking-Lösungen des Cheng-Datensatzes untersucht. Die Häufigkeitsuntersuchung zeigt, dass eine Absättigung vergrabener polarer Gruppen in Protein-Ligand-Komplexen für eine hochaffine Protein-Ligand-Bindung notwendig ist, da vergrabene nicht-abgesättigte HBA und HBD nur selten auftreten. Eine vollständige Absättigung durch entsprechende Proteinpartner wird für ca. 48 % der untersuchten Komplexe beobachtet, ca. 92 % weisen weniger als drei hauptsächlich schwache, nicht-abgesättigte HBA bzw. HBD (z. B. Etherfunktionen) auf. Unter Einbeziehung von Wassermolekülen in die Häufigkeitsanalyse sind sogar für ca. 61 % aller Komplexe alle wasserstoffbrückenbindenden Gruppen abgesättigt. Im Gegensatz zu DSX werden für SFCscore nach Anwendung der empirischen Filterfunktion erhöhte Erfolgsraten für das Auffinden einer kristallnahen Pose (≤ 2.0 Å Abweichung) unter den am besten bewerteten Docking-Posen erzielt. Für die beste SFCscore-Funktion (SFCscore::229m) werden Steigerungen dieses als „Docking Power“ bezeichneten Kriteriums für die Top-3-Posen (Erfolgsrate für die Identifizierung einer kristallnahen 2.0 Å Pose unter den besten drei Docking-Lösungen) von 63.1 % auf 64.2 % beobachtet.
In einem weiteren Teilprojekt wurden repulsive Protein-Ligand-Kontakte infolge sterischer Überlappungen der Bindungspartner bei der Bewertung von Docking-Lösungen berücksichtigt. Die adäquate Einbeziehung solcher repulsiver Kontakte im Scoring ist für die Identifizierung proteingebundener Ligandkonformationen entscheidend, jedoch aufgrund fehlender Affinitäts- bzw. Strukturdaten problematisch. Im Rahmen der vorliegenden Arbeit wurde auf der Basis des Lennard-Jones-Potentiales des AMBER-Kraftfeldes zunächst ein neuer Deskriptor zur Beschreibung repulsiver Kontakte („Clash“-Deskriptor) entwickelt und zur Untersuchung der Häufigkeit ungünstiger Protein-Ligand-Kontakte in hochaufgelösten Protein-Ligand-Komplexen des Hartshorn-Datensatzes herangezogen. Eine aus der Häufigkeitsverteilung abgeleitete empirische Filterfunktion („Clash“-Filterfunktion) wurde anschließend der Bewertung von Docking-Lösungen des Cheng-Datensatzes mittels der Scoringfunktionen SFCscore und DSX vorgeschaltet, um unerwünschte Ligandbindeposen auszuschließen. Die Häufigkeitsuntersuchung zeigt, dass vorwiegend schwache repulsive Kontakte in Protein-Ligand-Komplexen auftreten. So werden in 75 % der Komplexe des Hartshorn-Datensatzes abstoßende Potentiale unter 0.462 kcal/mol beobachtet. Zwar betragen die ungünstigen Beiträge pro Komplex für 50 % aller Strukturen ca. 0.8 kcal/mol bis 2.5 kcal/mol, jedoch können diese auf Ungenauigkeiten der Kristallstrukturen zurückzuführen sein bzw. durch günstige Protein-Ligand-Wechselwirkungen kompensiert werden. Die Anwendung der „Clash“-Filterfunktion zeigt signifikante Verbesserungen der Docking Power für SFCscore. Für die beste SFCscore-Funktion (SFCscore::frag) werden Steigerungen der Erfolgsraten für das Auffinden einer kristallnahen Pose unter den drei am besten bewerteten Docking-Lösungen von 61.4 % auf 86.9 % erzielt, was an die Docking Power der bis dato besten Scoringfunktionen aus der Literatur (z. B. DSX, GlideScore::SP) heranreicht (Docking Power (DSX): 92.6 %; Docking Power (GlideScore::SP): 86.9 %). Die „Clash“-Filterfunktion allein ist auch der Kombination der „Clash“- und der „vnaHB“-Filterfunktion überlegen.
Ein weiterer Schwerpunkt der vorliegenden Arbeit wurde auf die Einbeziehung von Decoy-Daten (Struktur- und Affinitätsdaten schwach affiner und inaktiver Liganden) im Zuge der Entwicklung computergestützter Methoden zur Bewertung von Docking-Lösungen gelegt. Dadurch soll eine adäquate Berücksichtigung ungünstiger Beiträge zur Bindungsaffinität ermöglicht werden, die für die Richtigkeit und Zuverlässigkeit ermittelter Vorhersagen essentiell ist. In der vorliegenden Arbeit wurden binäre Klassifizierungsmodelle zur Bewertung von Docking-Lösungen entwickelt, die die Einbeziehung von Decoy-Daten ohne die Verfügbarkeit von Affinitätsdaten erlauben. Der Random-Forest-Algorithmus (RF), SFCscore-Deskriptoren, der neu entwickelte „Clash“-Deskriptor, und die Decoy-Datensätze von Cheng und Huang (Trainingsdaten) bilden die Grundlage des leistungsfähigsten Klassifizierungsmodells. Der praktische Nutzen des „besten“ RF-Modells wurde nach Kombination mit der Scoringfunktion DSX anhand der Docking Power für das Auffinden einer kristallnahen Pose auf Rang 1 am unabhängigen Cheng-/Huang- (Komplexe, die nicht in den Trainingsdaten enthalten sind) und CSAR-2012-Testdatensatz untersucht. Gegenüber einer alleinigen Anwendung von DSX werden an beiden Testdatensätzen weitere Verbesserungen der Docking Power erzielt (Cheng-/Huang-Testdatensatz: DSX 84.24 %, RF 87.27 %; CSAR-2012-Testdatensatz: DSX 87.93 %, RF 91.38 %). Das „beste“ Modell zeichnet sich durch die zuverlässige Vorhersage richtig-positiver Docking-Lösungen für einige wenige Komplexe aus, für die DSX keine kristallnahe Ligandkonformation identifizieren kann. Ein visueller Vergleich der jeweils am besten bewerteten RF- und DSX-Pose für diese Komplexe zeigt Vorteile des RF-Modells hinsichtlich der Erkennung für die Protein-Ligand-Bindung essentieller Wechselwirkungen. Die Untersuchung der Bedeutung einzelner SFCscore-Deskriptoren für die Klassifizierung von Docking-Lösungen sowie die Analyse der Misserfolge nach Anwendung des Modells geben wertvolle Hinweise zur weiteren Optimierung der bestehenden Methode. Hinsichtlich der zu bewertenden Eigenschaften ausgeglichenere Trainingsdaten, Weiterentwicklungen bestehender SFCscore-Deskriptoren sowie die Implementierung neuer Deskriptoren zur Beschreibung bis dato nicht-berücksichtigter Beiträge zur Bindungsaffinität stellen Ansatzpunkte zur Verbesserung dar.
Der zweite Teil der vorliegenden Arbeit umfasst die Anwendung dockingbasierter Methoden im Rahmen der Entwicklung neuer Inhibitoren des „Macrophage Infectivity Potentiator“-(MIP)-Proteins von Legionella pneumophila und Burkholderia pseudomallei.
Das MIP-Protein von Legionella pneumophila stellt einen wichtigen Virulenzfaktor und daher ein attraktives Zielprotein für die Therapie der Legionellose dar. Im Rahmen der vorliegenden Arbeit erfolgten systematische Optimierungen des Pipecolinsäure-Sulfonamides 1, des bis dato besten niedermolekularen MIP-Inhibitors (IC50 (1): 9 ± 0.7 µM). Nach Hot-Spot-Analysen der Bindetasche wurden Docking-Studien zur Auswahl aussichtsreicher Kandidaten für die Synthese und Testung auf MIP-Inhibition durchgeführt. Die Ergebnisse der Hot-Spot-Analysen zeigen günstige Wechselwirkungsbereiche für Donorgruppen und hydrophobe Substituenten in meta-Position sowie Akzeptorgruppen in para-Position des Benzylringes von 1 auf. Die Einführung einer Nitrofunktion in para-Position des Benzylringes von 1 (2h) resultiert in einer erhöhten MIP-Inhibition (IC50 (2h): 5 ± 1.5 µM), was wahrscheinlich auf die Ausbildung einer zusätzlichen Wasserstoffbrücke zu Gly116 zurückzuführen ist. Selektivitätsverbesserungen gegenüber dem strukturverwandten humanen FKBP12-Protein werden insbesondere für das para-Aminoderivat von 1 (2n) erzielt (Selektivitätsindex (1): 45, Selektivitätsindex (2n): 4.2; mit Selektivitätsindex = IC50 (MIP)/IC50 (FKBP12)). Der Ersatz des hydrophoben Trimethoxyphenylrestes von 1 durch einen Pyridinring (2s) führt zu einer verbesserten Löslichkeit bei vergleichbarer MIP-Inhibition.
Das MIP-Protein von Burkholderia pseudomallei spielt eine wichtige Rolle in der Pathogenese der Melioidose und stellt daher ein attraktives Zielprotein für die Entwicklung neuer Arzneistoffe dar. In der vorliegenden Arbeit erfolgten Optimierungen des bis dato besten niedermolekularen MIP-Inhibitors 1. Ausgehend von einem Strukturvergleich von Burkholderia pseudomallei MIP mit Legionella pneumophila MIP und einer Hot-Spot-Analyse der Burkholderia pseudomallei MIP-Bindetasche wurden Docking-Studien zur Auswahl aussichtsreicher Kandidaten für die Synthese und Testung auf MIP-Inhibition durchgeführt. Der Strukturvergleich zeigt eine hohe Homologie beider Bindetaschen. Größere konformelle Änderungen werden lediglich für den von Ala94, Gly95, Val97 und Ile98 geformten Bindetaschenbereich beobachtet, was unterschiedliche Optimierungsstrategien für 1 erforderlich macht. Günstige Wechselwirkungsbereiche der Burkholderia pseudomallei MIP-Bindetasche finden sich einerseits für Donorgruppen oder hydrophobe Substituenten in para-Position des Benzylringes (Region A) von 1, andererseits für Akzeptor- bzw. Donorgruppen in para- bzw. meta-/para-Position des Trimethoxyphenylringes (Region B). Anhand von Docking-Studien konnten sowohl für Variationen in Region A als auch in Region B aussichtsreiche Kandidaten identifiziert werden. Initiale MIP-Inhibitionsmessungen der bis dato synthetisierten Derivate deuten auf erhöhte Hemmungen im Vergleich zu 1 hin. Der Ersatz des hydrophoben Trimethoxyphenylrestes von 1 durch einen Pyridinring führt auch hier zu vergleichbarer MIP-Inhibition bei verbesserter Löslichkeit. Derzeit sind weitere Synthesen und Testungen aussichtsreicher Liganden durch die Kooperationspartner geplant. Die Ergebnisse der Inhibitionsmessungen sollen deren Nutzen als MIP-Inhibitoren aufzeigen und wertvolle Informationen für weitere Zyklen des strukturbasierten Wirkstoffdesigns liefern.
The transport of optically excited states, called excitons, as well as their conversion into charges define the two major steps allowing for the operation of organic photovoltaic (OPV) devices. Hence, a deep understanding of these processes, the involved mechanisms as well as possible loss channels is crucial for further improving the efficiency of organic solar cells. For studying the aforementioned processes spectroscopic methods like absorption and emission measurements are useful tools. As many of the processes take place on a sub-nanosecond (ns) timescale ultrafast spectroscopic methods are required. Due to this reason two experiments based on a femtosecond laser system were built and employed in this work, namely picosecond (ps) time-resolved photoluminescence (PL) and transient absorption (TA) spectroscopy.
By analyzing the PL decay dynamics in the prototypical organic semiconductor rubrene, the feasibility of a new approach for improving the efficiency of organic solar cells by harvesting triplet excitons generated by singlet fission was examined. Singlet fission describes a process where two triplet excitons are generated via a photoexcited singlet exciton precursor state if the energy of the two triplets is comparable with the energy of the singlet. For this purpose the influence of characteristic length scales on the exciton dynamics in different rubrene morphologies exhibiting an increasing degree of confinement was analyzed. The results show that the quenching at interfacial states efficiently suppresses the desired fission process if these states are reached by excitons during migration. Since interfacial states are expected to play a significant role in thin film solar cells and are easily accessible for the migrating excitons, the results have to be considered for triplet-based OPV.
While the aforementioned approach is only investigated for model systems so far, the efficiency of disordered organic bulk heterojunction (BHJ) solar cells could be significantly enhanced in the last couple of years by employing new and more complex copolymer donor materials. However, little is known about the photophysics and in particular the excitation dynamics of these systems. By carrying out a systematic optical study on the prominent copolymer PCDTBT and its building blocks we were able to identify the nature of the two characteristic absorption bands and the coupling mechanism between these levels. The latter mechanism is based on an intrachain partial charge transfer between two functional subunits and our time-resolved measurements indicate that this coupling governs the photophysical properties of solar cells based on these copolymers. The efficient coupling of functional subunits can be seen as a key aspect that guarantees for the success of the copolymer approach.
Another important issue concerns the optimization of the morphology of BHJ solar cells. It arises from the discrepancy between the exciton diffusion length \mbox{($\approx$ 10 nm)} and the absorption length of solar irradiation ($\approx$ 100 nm). Due to this reason, even for devices based on new copolymer materials, processing parameters affecting the morphology like annealing or employing processing additives are of major importance. In our combined optical, electrical and morphological study for solar cells based on the high-efficient copolymer PBDTTT-C we find a direct correlation between additive content and intermixing of the active layer. The observed maximum in device efficiency can be attributed to a morphology guaranteeing for an optimized balance between charge generation and transport. Our results highlight the importance of understanding the influence of processing parameters on the morphology of the BHJ and thus on the efficiency of the device.
Ziel dieser Studie war es, mögliche Veränderungen des psychischen Befindens sowie Ausprägungen der Persönlichkeitsdimensionen Extraversion und Neurotizismus im Verlauf der Übungen der Progressiven Muskelrelaxation zu untersuchen. Weiterhin sollte der Einfluss der Persönlichkeitsmerkmale auf die Veränderung des Befindens überprüft werden.
Im theoretischen Teil der Arbeit wurde die Entstehung und Wirkungsweise der PR, Eysencks Theorien und die bisherige Literatur zum Thema dargestellt. Zum Einen konnte festgesellt werden, dass die PR als Teil unterschiedlicher Therapieintervention Einfluss auf das Befinden und die Persönlichkeitsmerkmale hat. Zum Anderen wurde deutlich, dass die Wirkung der PR auf das psychische Befinden und Persönlichkeitsausprägung durch verschiedene, sich gegenseitig bedingende Faktoren, beeinflusst wird. Folgende Hypothesen wurden aufgestellt:
Im Verlauf eines Kurses der PR verbessert sich das psychische Befinden. Zugleich nimmt der Neurotizismuswert ab und der Extraversionswert zu. Außerdem hängt das Ausmaß der Veränderung des psychischen Befindens mit der Persönlichkeits-ausprägung zu Beginn der Durchführung der PR zusammen. Introvertierte und emotional Labile sollen deutlicher von der Wirkung der PR profitieren als Extrovertierte und emotional Stabile.
93 Probanden füllten zu vier Messzeitpunkten vor, während und nach der PR Fragebögen (Eysenck-Persönlichkeits-Inventar, Form A und Eigenschaftswörterliste, Normalform) aus.
Die Ergebnisse zeigen keine Veränderung des Extraversionswertes, wohingegen es nach Beendigung der Übungen zu einer signifikanten Besserung der emotionalen Stabilität kam, was sich in einer Verringerung des Neurotizismuswertes äußerte. Es konnte während und nach Durchführung der PR eine signifikante Verringerung des negativen und Zunahme des positiven Befindens festgestellt werden. Eine Abhängigkeit der Veränderung des Befindens von den Persönlichkeitsdimensionen ließ sich nicht nachweisen. Die Annahme, dass vor allem emotional Labile und Introvertierte eine Verbesserung des Befindens zeigen, ließ sich nicht bestätigen.
Unter Beachtung, dass die tatsächlich involvierten Daten etwa der Hälfte der insgesamt an der Studie teilgenommenen Probanden entsprechen sowie unter Berücksichtigung des Studiendesigns (Ein-Gruppen-Prä-Post-Studie ohne Kontrollgruppe), kann man abschließend formulieren, dass die PR zur Verbesserung der Stimmungslage und Erlangung einer psychischen Stabilität beiträgt, unabhängig von der Ausprägung des Extraversions- und Neurotizismusgrades.
Zahnverluste können zu erheblichen Beeinträchtigungen der Phonetik, Ästhetik und Funktionalität führen. Daher muss Zahnersatz das natürliche Gebiss funktionell ersetzen, möglichst ohne als künstlich empfunden zu werden. Die dentale Implantologie als moderne Therapieform zur Behandlung verloren gegangener Zähne, weist heutzutage hohe Erfolgsraten auf. Dennoch ist die Suche nach Misserfolgsursachen unerlässlich, um auch langfristig eine Therapiesicherheit bieten zu können.
Die vorliegende Untersuchung hatte das Ziel, den Einfluss von Erkrankungen wie Diabetes mellitus, Nikotinabusus und kardiovaskulären Erkrankungen auf die Implantattherapie und deren Erfolg zu untersuchen.
1904 Patienten, welche in einem Zeitraum von 15 Jahren (9.6.1995 - 16.7.2010) in der Zahnarztpraxis Dr. med. Robert Böttcher mit Implantaten verschiedener Implantatsysteme versorgt wurden, wurden in dieser Studie untersucht.
Das Patientengut bestand aus 1107 (58,1%) Frauen und 797 (41,9%) Männern. Im Mittel wurden pro Patient 3 Implantate gesetzt. Das mittlere Alter der 1904 Patienten bei Implantation lag bei 52 Jahren (Minimum: 15 Jahre, Maximum: 86 Jahre). 58% der Implantate (n=3608) wurden bei Frauen eingesetzt. 42% der Implantate (n=2609) wurden bei Männern inseriert.
Bei der Analyse implantatabhängiger Faktoren stellte sich der Parameter „Indikation“ und auch die Interaktion des Parameters Indikation „2b“ und der Einheildauer als signifikant dar. Erstaunlicherweise haben Implantate, welche in „Schaltlücken“ inseriert wurden, ein zunächst über 65000-fach höheres Risiko explantiert zu werden. Dieses Risiko nimmt allerdings mit zunehmender Einheildauer signifikant ab. So ist es nach einem Monat nur noch 1478-fach erhöht, nach 2 Monaten nur noch 33-fach erhöht.
Ein Erklärungsansatz für das bessere Überleben der Implantate im zahnlosen Unterkiefer liegt in der dort zu findenden Knochenqualität mit einem deutlich höheren Anteil an Knochenkompakta und der daraus resultierenden erhöhten Widerstandsfähigkeit. Die Indikation „Schaltlücke“ wurde in der vorliegenden Untersuchung zudem vor allem im Seitenzahnbereich versorgt. Hier kommt es zu einer höheren Kaubelastung und anatomisch bedingt ist in der Seitenzahnregion mit einer geringeren Restknochenhöhe zu rechnen. Zudem kommt es durch den frühen Verlust der Molaren (6-Jahres-Molar) zu einer vertikalen und horizontalen Knochenatrophie, was wiederum bei der Implantation in dieser Position Augmentationsmaßnahmen erfordern kann. Diese sind erwartungsgemäß mit einem höheren Verlustrisiko assoziiert. Weiterhin kann durch Mangel an fixierter Gingiva eine Optimierung der Weichgewebssituation in Form einer Vestibulumplastik erforderlich sein. Dies kann mit einem erhöhten Risiko einer Periimplantitis und schließlich einem Implantatverlust einhergehen.
In der Literatur fanden sich ähnliche Ergebnisse bei Shatkin et al. (2007). Hier kam es zu den höchsten Verlusten in der posterioren Maxillaregion. Auch Machtei et al. (2007) und Johns et al. (1992) ermittelten die höchste Verlustrate im Bereich der oberen ersten Molaren. [81, 102]
Auch die Einheildauer und die Interaktion der Einheildauer mit der Zeit erwiesen sich in der vorliegenden Arbeit als signifikante Variablen. Es erstaunt jedoch nicht, dass sich das Risiko eines Implantatverlustes mit zunehmender Einheildauer, in dieser Studie um den Faktor 3,89 pro Monat Einheildauer, reduziert. Gerade bei zusätzlichen Maßnahmen wie Osteoplastiken oder der gesteuerten Geweberegeneration werden die Einheilzeiten auf bis zu zwölf Monate verlängert um eine optimale Osseointegration zu gewährleisten.
Ein offen-transgingivaler Einheilmodus zeigte gegenüber dem gedeckt-subgingivalen Einheilmodus mit etwa 3% Verlust nach 10 Jahren ein signifikant schlechteres Überleben.
Additive Implantatoberflächen zeigten im Vergleich zu subtraktiven Oberflächen signifikant bessere 10-Jahresüberlebensraten. Die Verlustraten lagen bei letztgenannten bei bis zu 5% nach 10 Jahren.
Erwartungsgemäß wurden in dieser Studie Augmentationsmaßnahmen und zunehmende Implantatanzahl als Risikofaktor einer Explantation identifiziert. So haben augmentierte Implantate ein 2,4-fach höheres Risiko explantiert zu werden. Das relative Risiko eines Implantatverlustes nimmt mit zunehmender Implantatanzahl um den Faktor 1,18 zu.
Implantatregion, -durchmesser und -länge, wie auch der Implantationszeitpunkt und die Knochenqualität konnten nicht als Risikoparameter identifiziert werden.
Bei der Untersuchung patientenabhängiger Faktoren erwies sich das Patientenalter als signifikanter Einflussfaktor auf den Implantaterfolg. So kam es bei Patienten im höheren Lebensalter seltener zu Implantatverlusten. Pro Lebensjahr sank das Risiko eines Implantatverlustes um den Faktor 0,9. Dies lässt sich durch ein größeres Gesundheitsbewusstsein im zunehmenden Lebensalter erklären. Zudem bestand bei jungen Patienten in dieser Untersuchung ganz überwiegend eine Indikation zur Einzelzahnversorgung beispielsweise bei Nichtanlagen der oberen seitlichen Schneidezähne, multiplen Nichtanlagen oder nach Unfällen (Fahrrad, Skater, Schlitten).
Diese Indikationen erfordern häufig Augmentationsmaßnahmen, teilweise auch Knochenblocktransplantationen, was insgesamt wiederum zu einem erhöhten Implantatverlustrisiko führt.
Das Patientengeschlecht hatte keinen Einfluss auf den Implantaterfolg.
Wie erwartet, konnte im untersuchten Patientengut ein negativer Einfluss des Rauchens auf den Implantaterfolg aufgezeigt werden.
In der Literatur wird dem Nikotinabusus nahezu einheitlich ein negativer Einfluss auf das Implantatüberleben zugeschrieben.
Eine diabetische Stoffwechsellage hatte in unserem Patientenkollektiv keinen nachweisbaren Einfluss auf den Implantaterfolg.
Hingegen wurde bei Patienten mit kardiovaskulären Erkrankungen nach 10 Jahren eine signifikant geringere Implantatüberlebenswahrscheinlichkeit nachgewiesen.
Die Hypothese der Arbeit, dass Risikofaktoren zu Komplikationen bei der dentalen Implantation führen und somit das Implantatüberleben beeinflussen, konnte in dieser Untersuchung bestätigt werden.
Insgesamt ist in Anbetracht des Patienten-individuellen Risikos eine spezifische Risiko-Nutzen-Abwägung erforderlich, da eine implantatprothetische Versorgung der Patienten mit definierten Risikofaktoren nicht generell kontraindiziert ist.
Patienten mit chronischer Nebenniereninsuffizienz (NNI) haben ein hohes Risiko, eine lebensbedrohliche Nebennierenkrise (NNK) zu erleiden. Hauptauslöser sind insbesondere gastrointestinale und fieberhafte Infekte, sowie Unfälle, Operationen, psychische und physische Belastungssituationen. Häufig entwickeln sich dabei NNK innerhalb weniger Stunden und können rasch zu einem letalen Verlauf führen, bevor den betroffenen Patienten suffiziente ärztliche Hilfe gewährleistet werden kann.Zur Prävention solcher NNK, werden nebenniereninsuffiziente Patienten wiederholt in der Eigenanpassung ihrer Glucocorticoiddosis in Belastungssituationen geschult. Sie erhalten einen Notfallausweis und werden zusätzlich mit einem Notfallkit, bestehend aus einer Ampulle Hydrocortison für Injektionszwecke, ausgestattet. Da Patienten jedoch nach wie vor (zum Beispiel im Rahmen von Gastroenteritiden durch ungenügende enterale Resorption des Hydrocortisons) an NNK versterben, sind eine Verbesserung der Krisenprävention und des Notfallmanagements dringend anzustreben.Um den Patienten in Krisensituationen ein höheres Maß an Unabhängigkeit zu ermöglichen, erfolgen daher mittlerweile Schulungen in der Eigeninjektion von Hydrocortison. Aktuell besteht hier eine Zulassung für die intramuskuläre (i.m.) Verabreichung. Es ist jedoch anzunehmen, dass die i.m. Eigeninjektion für viele Patienten eine große Hemmschwelle darstellt und daher in Krisensituationen nicht konsequent genug angewandt wird. Die subkutane (s.c.) Verabreichung von Hydrocortison wäre eine für die Patienten leicht durchzuführende und äquivalente Alternative. Erfahrungsgemäß besteht bezüglich einer s.c. Applikation von Medikamenten eine höhere Akzeptanz , da diese in vielen anderen Bereichen, wie z. B. der s.c. Selbstinjektion von Insulin bei Diabetikern oder der Injektion von Heparin in der postoperativen Phase oder bei der Injektion von Hormonpräparaten im Rahmen einer Hypophyseninsuffizienz, bereits regelmäßig Anwendung findet und bei vielen Patienten bekannt ist.
Ziel der Studie war es daher, die Pharmakokinetik und die Sicherheit der s.c. Hydrocortisonapplikation für einen routinemäßigen Einsatz in Krisensituationen im Vergleich zur i.m. Gabe zu evaluieren. Für die Studie wurden zwölf Patienten mit einer chronischen NNI eingeschlossen. Ihnen wurde an drei verschiedenen Untersuchungstagen s.c. Kochsalzlösung (Kontrollintervention), sowie s.c. und i.m. 100 mg Hydrocortison injiziert. An jedem Untersuchungstag, wurden Speichelproben sowie über eine venöse Verweilkanüle Blutproben zu 15 Zeitpunkten (0 bis 240 min nach Injektion) entnommen und asserviert. Anhand der pharmakokinetischen Profile zeigte sich, dass sowohl die i.m. als auch die s.c. Applikation von Hydrocortison zu suffizienten Wirkspiegeln (> 36 µg/dl) von Cortisol im Serum führt. Wie erwartet zeigte sich bei der i.m. Injektion von Hydrocortison ein etwas schnellerer Anstieg der Cortisolkonzentration. Andererseits ergab sich nach der s.c. Injektion von Hydrocortison eine längere mittlere Verweildauer im Blut. Des Weiteren wurde nach s.c. Applikation eine stärkere Abhängigkeit der Serumcortisolprofile vom BMI der Patienten beobachtet. Schwerwiegende unerwünschte Ereignisse traten nicht auf. Leichte unerwünschte Ereignisse, ließen sich vielmehr auf die Injektion an sich, als auf die applizierte Substanz Hydrocortison zurückführen und waren überwiegend bei der i.m. Applikation zu beobachten. Die Beurteilung der Patientenzufriedenheit anhand eines Fragebogens zeigte deutlich, dass die Patienten die s.c. gegenüber der i.m. Eigeninjektion bevorzugen würden. Weiterhin wurde deutlich, dass die Bedrohung durch NNK, für einige Patienten auch eine Einschränkung der Lebensqualität bedeutet und dass sie sich durch die Möglichkeit der Eigeninjektion sicherer fühlen würden. Alle Patienten würden sich nach Studienteilnahme eine Eigeninjektion von Hydrocortison zutrauen. Diese Ergebnisse zeigen, dass die s.c. Applikation von Hydrocortison eine geeignete Alternative zur i.m. Injektion darstellt; besonders für Patienten mit einem normwertigen BMI und im Falle von beginnenden NNK, bevor es evtl. zur Beeinträchtigung der Zirkulation im Schock und einer möglicherweise unzureichenden Resorption bei einer Minderdurchblutung des Fettgewebes kommt. Da die Patienten die s.c. der i.m. Applikation deutlich vorziehen würden, kann davon ausgegangen werden, dass nebenniereninsuffiziente Patienten die s.c. Eigeninjektion konsequenter einsetzten würden. Durch eine geringere Hemmschwelle gegenüber der s.c. Gabe und einem folglich vermehrten Einsatz der Eigeninjektion bei beginnenden Krisen, könnte die Zahl der NNK und deren Mortalität gesenkt werden. Allerdings ist die i.m. Verabreichung von Hydrocortison auch weiterhin als eine sehr gute und schneller wirksamere Art der Eigeninjektion zu betrachten, die den Patienten in erster Linie empfohlen werden kann.
Zur Krisenprävention sollte zukünftig, zusätzlich zum Notfallausweis und der generellen Aufklärung, der Schwerpunkt auch auf eine individuelle Schulung im richtigen Umgang mit der Eigeninjektion von Hydrocortison in Notfallsituationen gelegt werden. Des Weiteren sollte generell neben der Optimierung der Substitutionstherapie auch die Verbesserung des Krisenmanagements mit im Fokus der NNI-Forschung stehen.
Beim Siciliano – dem Forschungsgegenstand dieser Arbeit – handelt es sich um einen musikalischen Typ oder Topos, der sich etwa durch den 6/8-Takt, den punktierten Rhythmus und ein langsames Tempo auszeichnet. In der heutigen musikhistorischen Literatur ist die Annahme verbreitet, der Siciliano habe lediglich in der ersten Hälfte des 18. Jahrhunderts große Popularität genossen, sei aber nach 1750 langsam in Vergessenheit geraten. Der Verfasser ist jedoch durch eine Untersuchung von 184 Siciliano-Kompositionen, die zwischen ca. 1750 und 1820 entstanden sind, zu dem Schluss gekommen, dass die gängige These nicht mehr aufrechtzuerhalten ist: Das kompositorische Repertoire – darunter finden sich nicht nur „große Namen“, sondern auch zahlreiche Komponisten, die nur zu ihren Lebzeiten beliebt waren – macht deutlich, dass der Siciliano auch nach der Jahrhunderthälfte eine bedeutende Rolle gespielt hat. Darüber hinaus wurde festgestellt, dass der Begriff des Siciliano auf der kompositionspraktischen Ebene ziemlich flexibel und keinesfalls einheitlich verstanden wurde. Es scheint gerade seine ausgesprochene Heterogenität zu sein, die diesen Topos wesentlich auszeichnet.
Vor diesem Hintergrund widmet sich der zweite Teil der Arbeit den Siciliano-Kompositionen von Joseph Haydn (1732–1809), einem der repräsentativen Komponisten des klassischen Stils. Dabei wurde vor allem ihr Stellenwert sowohl in Haydns Gesamtschaffen als auch in der Geschichte des Siciliano untersucht. Haydn greift während seines gesamten Schaffens und gattungsübergreifend auf diesen Topos zurück und hinterließ 36 instrumentale Siciliani, die zumeist in langsamen Sätzen mehrsätziger Werkzyklen vorkommen. Besonders ab den ausgehenden 1760er Jahren lässt sich in den zentralen Gattungen Haydns künstlerische Absicht erkennen, die Ausdrucksmöglichkeiten des Siciliano-Topos systematisch auszuloten. Auffallend ist unter anderem die Einführung verschiedener Variations- techniken, die Haydn in seinen anderen Werken entwickelt hat. Daraus resultiert dort eine Wechselwirkung zwischen der „Gemeinsprache“ des Topos und Haydns Personalstil. Aus einer stilanalytischen Untersuchung dieser Werke geht hervor, dass Haydns Auseinandersetzung mit dem Siciliano eine Aktualisierung und Weiterentwicklung der seit dem Beginn des 18. Jahrhunderts bestehenden Tradition bedeutet, jedoch keineswegs deren „Ver- steinerung“.
Die vorliegende Arbeit darf als die erste umfassende Untersuchung zum Siciliano-Topos nach 1750 gelten. Zugleich setzt sich insbesondere der zweite Dissertationsteil intensiv mit der in der herkömmlichen Haydn-Literatur vernachlässigten Frage nach dem Kompositionsstil in Haydns langsamem Satz auseinander. In diesem Sinne ist es ein zentrales Anliegen dieser Arbeit, zwei wesentliche Forschungslücken zu füllen.
Bei Melanomen handelt es sich um die gefährlichste Form von Hautkrebs mit der höchsten Mortalitätsrate. Deshalb sind Untersuchungen dieser Hautkrebsart von immenser Bedeutung. Es ist bekannt, dass der AP-1-Transkriptionsfaktorkomplex eine große Rolle für Melanomentstehung und -progression spielt. In der vorliegenden Arbeit wurde die Funktion der AP-1 Komponente FOSL1 in Melanomen untersucht.
Hierbei konnte zunächst ermittelt werden, dass die FOSL1 Expression im humanen Melanom durch den MAPK-Signalweg vermittelt wird und von den Onkogenen BRAF und NRAS abhängig ist. Dies wird auch durch die Tatsache unterstützt, dass die Stabilität von FOSL1 durch MAPK reguliert wird.
Des Weiteren konnte gezeigt werden, dass FOSL1 in vielen Melanomzellen die Proliferation verstärkt und auch an Migration beteiligt ist. Da diese Prozesse zur Krebsprogression beitragen, deutet dies darauf hin, dass FOSL1 bei der Melanomentwicklung eine wichtige Funktion besitzt. Weiterhin konnten SLUG, SNAI3, IL6 und MMP14 als FOSL1-Zielgene identifiziert werden, deren Regulierbarkeit durch FOSL1, jedoch abhängig von der jeweiligen Zelllinie war. Somit konnte mit dieser Arbeit gezeigt werden, dass FOSL1 nicht nur, wie zuvor für Brustkrebszellen beschrieben, an Migration beteiligt ist, sondern auch zur Proliferation humaner Melanome beiträgt. Zukünftige Arbeiten werden zeigen, ob die identifizierten Gene für die FOSL1-vermittelte Migration und Proliferation verantwortlich sind.
Bacterial mastitis is caused by invasion of the udder, bacterial multiplication and induction of
inflammatory responses in the bovine mammary gland. Disease severity and the cause of disease are
influenced by environmental factors, the cow’s immune response as well as bacterial traits. Escherichia coli (E. coli) is one of the main causes of acute bovine mastitis, but although pathogenic E. coli strains can be classified into different pathotypes, E. coli causing mastitis cannot unambiguously be distinguished from commensal E. coli nor has a common set of virulence factors
been described for mastitis isolates. This project focussed on the characterization of virulence-
associated traits of E. coli mastitis isolates in comprehensive analyses under conditions either
mimicking initial pathogenesis or conditions that E. coli mastitis isolates should encounter while entering the udder. Virulence-associated traits as well as fitness traits of selected bovine mastitis or faecal E. coli strains were identified and analyzed in comparative phenotypic assays. Raw milk whey was introduced to
test bacterial fitness in native mammary secretion known to confer antimicrobial effects.
Accordingly, E. coli isolates from bovine faeces represented a heterogeneous group of which some
isolates showed reduced ability to survive in milk whey whereas others phenotypically resembled
mastitis isolates that represented a homogeneous group in that they showed similar survival and
growth characteristics in milk whey. In contrast, mastitis isolates did not exhibit such a uniform phenotype when challenged with iron shortage, lactose as sole carbon source and lingual
antimicrobial peptide (LAP) as a main defensin of milk. Reduced bacterial fitness could be related to LAP suggesting that bacterial adaptation to an intramammary lifestyle requires resistance to host
defensins present in mammary secretions, at least LAP.
E. coli strain 1303 and ECC-1470 lack particular virulence genes associated to mastitis isolates. To find out whether differences in gene expression may contribute to the ability of E. coli variants to cause mastitis, the transcriptome of E. coli model mastitis isolates 1303 and ECC-1470 were analyzed to
identify candidate genes involved in bacterium-host interaction, fitness or even pathogenicity during bovine mastitis.
DNA microarray analysis was employed to assess the transcriptional response of E. coli 1303 and
ECC-1470 upon cocultivation with MAC-T immortalized bovine mammary gland epithelial cells to
identify candidate genes involved in bacterium-host interaction. Additionally, the cell adhesion and invasion ability of E. coli strain 1303 and ECC-1470 was investigated. The transcriptonal response to the presence of host cells rather suggested competition for nutrients and oxygen between E. coli and MAC-T cells than marked signs of adhesion and invasion. Accordingly, mostly fitness traits that may also contribute to efficient colonization of the E. coli primary habitat, the gut, have been utilized by the mastitis isolates under these conditions. In this study, RNA-Seq was employed to assess the bacterial transcriptional response to milk whey.
According to our transcriptome data, the lack of positively deregulated and also of true virulence-associated determinants in both of the mastitis isolates indicated that E. coli might have adapted by other means to the udder (or at least mammary secretion) as an inflammatory site. We identified traits that promote bacterial growth and survival in milk whey. The ability to utilize citrate promotes fitness and survival of E. coli that are thriving in mammary secretions. According to our results, lactoferrin has only weak impact on E. coli in mammary secretions. At the same time bacterial determinants involved in iron assimilation were negatively regulated, suggesting that, at least during the first hours, iron assimilation is not a challenge to E. coli colonizing the mammary gland. It has been hypothesized that cellular iron stores cause temporary independency to extracellular accessible iron. According to our transcriptome data, this hypothesis was supported and places iron uptake
systems beyond the speculative importance that has been suggested before, at least during early
phases of infection. It has also been shown that the ability to resist extracytoplasmic stress, by oxidative conditions as well as host defensins, is of substantial importance for bacterial survival in mammary secretions.
In summary, the presented thesis addresses important aspects of host-pathogen interaction and
bacterial conversion to hostile conditions during colonization of the mastitis inflammatory site, the mammary gland.
Schmerzen sind ein Hauptsymptom der Entzündung. Während der Entzündungsreaktion führt die Freisetzung von Zytokinen und Chemokinen zur Einwanderung von Leukozyten in das entzündete Gewebe. Durch die Freisetzung weiterer proalgetischer Mediatoren tragen Leukozyten zur Sensitivierung des Nozizeptors bei und verursachen damit die Schmerzentstehung.
In Verhaltensexperimenten verursacht intraplantare Injektion des Monozyten-rekrutierenden Chemokins CCL2 bei Wistar Ratten eine Hyperalgesie. Gleichzeitige Injektion von CCL2 mit dem Enzym Katalase oder dem Superoxiddismutasemimetikum TEMPOL verhindert die Entwicklung der CCL2-induzierten Hyperalgesie. Dark Agouti Ratten mit einer verringerten Aktivität der NADPH-Oxidase, aufgrund eines Polymorphimus im Gen ncf1, entwickeln keine CCL2-induzierte Hyperalgesie.
In dieser Arbeit wurde die Bedeutung von Monozyten/Makropagen und reaktiven Sauerstoffradikalen für die Entstehung der CCL2-induzierten Hyperalgesie untersucht. In vitro wurde die Bildung von reaktiven Sauerstoffradikalen in humanen Monozyten und Peritonealmakrophagen aus Wistar und Dark Agouti Ratten nach Stimulation mit CCL2 untersucht. In vivo wurde die Bildung des Lipidperoxidationsproduktes 4-HNE im Pfotengewebe von Wistar und Dark Agouti Ratten nach CCL2 Injektion untersucht.
More warning Advanced Driver Assistance Systems (ADAS) will be integrated into the European vehicles in the coming years, due to the ongoing progress on the way to automated driving and Euro NCAP requirements. Furthermore, upcoming technologies like Car-2-X will extend the sensory horizon of ADAS and enable the possibility to warn drivers earlier against various hazards than today. Regarding this progress, increasing numbers of different ADAS warnings will be communicated to the driver. In this context, an important question is how multiple ADAS warnings can be integrated into the Human Machine Interface (HMI) of vehicles and which warning elements are needed to ensure warning acceptance, efficiency and understandability seen from the driver’s point of view.
Two driving simulator studies were conducted and the effects of specific warning elements examined to develop a concept for the integration of upcoming warning ADAS, which focuses on early collision warnings. The implemented early warnings were defined with a warning onset of approximately two seconds before the last possible warning onset. Main questions were whether and how drivers profit from warning direction cues and/or warning object cues for their response to a hazard, and how these cues affect the acceptance of an integrated warning ADAS approach. Furthermore, it was analyzed whether a generalized warning can be used for a cluster of different ADAS concerning the group “warning of collisions”. Therefore critical scenarios in rural and urban surroundings were evaluated, including frontal and lateral (intersections) scenarios. Unnecessary warnings and false alarms have also been taken into account.
The results indicate that early warning direction cues have a high potential to assist drivers with an ADAS warning cluster which covers warning of collisions. In contrast, warning object cues seem to be less important for the drivers’ performance and acceptance regarding early collision warnings. According to these findings, this thesis provides recommendations which warning elements should be included into future ADAS warnings in favor of an integrated warning approach.
Malignant melanoma is the most severe form of all skin cancers with a particular poor prognosis once metastases have developed. Angiogenesis, the formation of new blood vessels, is a prominent feature of human melanoma, which have angiogenic activity already early in development. This is at least partly ascribed to the action of MAPK- and PI3K pathways which are hyperactivated in most melanoma. Animal models which combine in depth in vivo examinations with the opportunity to perform small molecular screens are well suited to gain a more detailed insight into how this type of cancer modulates its angiogenic program. Here, a first transgenic melanoma angiogenesis model was established in the fish species Oryzias latipes (Japanese medaka). In this model, tumors are generated by the pigment cell-specific expression of the oncogenic receptor tyrosine kinase Xmrk. Xmrk is a mutated version of the fish Egfp. Furthermore, to get an angiogenesis model, a medaka line with endothelial cell specific GFP expression was used. By using crosses between these Xmrk- and GFP transgenic fishes, it was shown that angiogenesis occurs in a reactive oxygen species- and NF-κB-dependent manner, but was hypoxia-independent. It was observed that blood vessel sprouting and branch point formation was elevated in this model and furthermore that sprouting could even be induced by single transformed cells. The mouse melanocytes expressing the oncogenic receptor tyrosine kinase Xmrk as well human melanoma cells, which display various oncogenic alterations, produced pro-angiogenic factors, most prominently angiogenin, via NF-κB signaling. Furthermore, inhibiting NF-κB action prevented tumor angiogenesis and even led to the regression of existing tumor blood vessels. In summary, the present medaka melanoma angiogenesis model displays a high sensitivity for angiogenesis detection and is perfectly suited as in vivo model for the testing of anti-angiogenesis inhibitors, as exemplified by the NF-kappaB inhibitor.
Furthermore, results indicate that it might be a promising anti-tumor strategy to target signaling pathways such as the NF-κB pathway which are able to induce angiogenesis-dependent as well as -independent pro-tumorigenic effects.
Aurora B is a mitotic kinase that is essential for cell division. Because it is mutated or overexpressed in a range of cancer types, it has been suggested as a novel therapeutic target. Currently chemical inhibitors against Aurora B are in various phases of clinical trials for treatment of solid tumors and leukemia. Information regarding the molecular requirements for the reported phenotypes of Aurora B inhibition such as cell cycle arrest, activation of the tumor suppressor p53 and its target p21 are not well understood.
In this study, I investigated the requirements for p21 induction after Aurora B inhibition. I found that p38 is phosphorylated and activated when Aurora B is inhibited. Experiments with chemical inhibitors against p38 indicate that p38 is required for p21 induction and cell cycle arrest in response to Aurora B inhibition. p53 induction after impairment of Aurora B function and the recruitment of p53 to its binding site in the p21 gene promoter occur independently of p38 signaling. Instead, I found that p38 is required for the enrichment of the elongating RNA Polymerase II in the coding region of the p21 gene. Furthermore, p38 is required for formation of the full-length p21 mRNA transcript. These data indicate that p38 promotes the transcriptional elongation of p21 gene in response to Aurora B inhibition. In further experiments I could show that the p21 causes cell cycle arrest due to a decrease in E2F-dependent transcription by promoting the dephosphorylation of the retinoblastoma protein.
Using synchronized cells I could show that the induction of p21 in response to Aurora B inhibition requires transition through an aberrant mitosis and does not occur in cells that are arrested in interphase. Interestingly, p38, p53 and p21 are already induced by partial inhibition of Aurora B, which results in aneuploidy but not in cytokinesis failure and in tetraploidy. This supports the notion that activation of p38-p53-p21 signaling correlates with aneuploidy but not with tetraploidy or binucleation. Partial inhibition of Aurora B also leads to increased generation of reactive oxygen species (ROS), which are required for the activation of p38, p21 and cell cycle arrest. Based on these observations I propose the following model: Inhibition of Aurora B leads to chromosome missegregation resulting in aneuploidy. This results in increased generation of ROS (reactive oxygen species) possibly through proteotoxic stress caused by an imbalance of protein synthesis in aneuploid cells. ROS triggers the activation of p38, which then stimulates the transcriptional elongation of p21 resulting in cell cycle arrest.
Aneuploidy, proteotoxic stress and oxidative stress are hallmarks of cancer cells. Based on my results reported in this study, I suggest that the combination of Aurora B inhibitors with drugs that specifically target aneuploid cells might be a novel strategy for cancer therapy, as this is a lethal combination for proliferation of cancer cells.
Die Afrikanische Union und die Europäische Union unterhalten eine hoch institutionalisierte Sicherheitspartnerschaft. Die im Jahre 2007 beschlossene Gemeinsame Afrika-EU-Strategie stellt für die interregionalen Beziehungen der Afrikanischen Union mit der Europäischen Union im Allgemeinen und für die sicherheitspolitische Zusammenarbeit im Besonderen eine Zäsur dar. Kooperieren die Afrikanische Union und die Europäische Union partnerschaftlich, also auf Augenhöhe, um mehr Sicherheit für beide Kontinente zu erreichen? Um diese Frage zu beantworten werden auf Grundlage des interorganisationalen Modells von Biermann (2009) in seiner Weiterentwicklung von Scheuermann (2012) verschiedene kooperationsfördernde und -hemmende Faktoren herausgearbeitet. Mit Hilfe dieses Rahmens werden dann die Entwicklung der interregionalen Beziehung und die mit der Strategie implementierten Mechanismen analysiert, um so fünf spezifische Problemmuster zu identifizieren, die eine Partnerschaft auf Augenhöhe in der Praxis konterkarieren können: Das Selbstverständnis der EU, die Perzeption der EU durch die AU, die unterschiedliche Perzeption von Sicherheit, die divergenten institutionellen Kapazitäten und der gravierende Ressourcenmangel. In welcher Weise sich diese Problemmuster auswirken, wird abschließend in den Fallbeispielen zur Finanzierung der Friedensmission AMISOM und zum konträren Verhalten der beiden Regionalorganisation in der Libyenkrise deutlich.
In dieser Dissertation wurden zwei Aspekte der Wechselwirkung von Laserpulsen mit Molekülen betrachtet: Erstens wurden numerische Algorithmen, die auf der zeitabhängigen Störungstheorie basieren, zur Berechnung von quantenmechanischen Wellenfunktionen analysiert. Zweitens wurden Effekte der absoluten Phase (Carrier envelope phase = CEP) von Laserpulsen bei der Laseranregung molekularer Systeme analysiert. In den Analysen zum ersten Aspekt wurden zwei verschiedene Algorithmen - in dieser Arbeit als simple und improved algorithm bezeichnet - verwendet, und die Normabweichung von mit diesen Algorithmen berechneten Wellenfunktionen untersucht. Es konnte gezeigt werden, dass diese Normabweichung für beide Algorithmen in zwei unterschiedliche Beiträge zerlegt werden kann. Der erste Normabweichungsbeitrag tritt aufgrund der numerischen Diskretisierung der Zeit auf und verschwindet, wenn der Zeitschritt, der die Dauer der Intervalle für diese Diskretisierung angibt, gegen Null geht. Man kann den ersten Normabweichungsbeitrag mit exzellenter Genauigkeit berechnen und seine Eigenschaften, die sich für die beiden Algorithmen erheblich unterschieden, eingehend analysieren. Der zweite Normabweichungsbeitrag tritt dadurch auf, dass die zeitabhängige Störungstheorie nicht normerhaltend ist, und geht daher gegen Null, wenn die Störungsordnung gegen unendlich geht. Dieser zweite Beitrag ist außerdem in guter Näherung unabhängig vom Zeitschritt und für beide Algorithmen näherungsweise gleich. Des Weiteren kann man das Verhalten des zweiten Normabweichungsbeitrags im Gegensatz zum ersten Beitrag nur qualitativ beschreiben. Für die Analyse zum zweiten Themengebiet dieser Arbeit, den CEP-Effekten, wurde betrachtet, ob CEP-Effekte auch für Laserpulse beliebiger Länge auftreten können. Über eine analytische Betrachtung erkennt man, dass dies für ein Zweiniveausystem nur dann der Fall ist, wenn beide Zustände vor Beginn der Wechselwirkung des Systems mit dem Laserpuls besetzt sind. Man kann aus diesem Ergebnis folgern, dass für einen Laserpuls, der zwei elektronische Zustände eines Moleküls über Einphotonenübergänge koppelt, in der Regel kein CEP-Effekt für beliebige Längen dieses Pulses auftritt. Der Grund dafür ist, dass vor der Wechselwirkung eines molekularen Systems mit einem Laserpuls für dieses üblicherweise nur der elektronische Grundzustand besetzt ist. In dieser Arbeit wird gezeigt, dass dieses Problem durch ein spezielles Zweipulsschema für die Anregung eines molekularen Systems gelöst werden kann. Für dieses Pulsschema wird ein erster Puls verwendet, der zeitlich so kurz ist, dass Wellenpakete in mehreren elektronischen Zuständen angeregt werden. Der nachfolgende zweite Laserpuls ist spektral schmal, und seine zeitliche Länge kann beliebig groß gewählt werden. Man erhält für dieses Pulsschema Observablen, die von der CEP des zweiten Pulses, aber nicht von der CEP des ersten Pulses abhängen; somit ist ein CEP-Effekt nachweisbar. Derartige Observablen sind geometrische Asymmetrien für Zerfallsprodukte von Photodissoziationsreaktionen. Insbesondere unterscheidet sich das hier vorgestellte Pulsschema von anderen Zweipulsschemata, für welche Observablen von der Differenz der CEPs beider Pulse abhängen, aber nicht von der CEP einer der beiden Pulse allein.
HINTERGRUND: Um den Erfolg einer antiretroviralen HIV-Therapie zu kontrollieren und Therapieversagen suffizient aufzudecken ist die quantitative Messung der HIV-1 RNA der Bestimmung der CD4-Zellzahl sowie der Beurteilung anhand der Klinik des Patienten überlegen. Die Viruslastbestimmung ist jedoch ein sehr teures Verfahren, sodass Kosten und Infrastruktur häufig die begrenzende Komponente für die Anwendung darstellen. Das gilt besonders für arme Länder, in denen die technische Entwicklung des Gesundheitssystems wenig fortgeschritten ist. In aktuellen Studien konnte gezeigt werden, dass das Poolen von Blutproben, bei einer an die klinische Relevanz angepassten Schwelle, ein effizientes Verfahren ist, um die ART mittels Viruslastbestimmung zu überwachen. Dies gilt, wenn die Therapieversagerquote niedrig ist. In Ländern, in denen der Kostenreduktion der Viruslastbestimmung eine besondere Wichtigkeit zukommt, sind die Durchseuchungsrate und die Therapieversagerquote jedoch oft hoch.
METHODEN: Es wurde das Poolen von Blutplasmaproben, DBS und DPS bei einer Population mit hoher Therapieversagerquote untersucht. Ein Pool wurde jeweils aus fünf Einzelproben angefertigt. Zur Auswertung der Pools aus flüssigen Plasmaproben wurde ein Algorithmus zur Vereinfachung der Dekonvolution angewandt.
ERGEBNISSE: Die Effizienz der Methode bei DPS (n=185) und flüssigem Plasma (n=385) lag zwischen 34,29% und 47,57%. Die Anwendung des Poolingsystems an DBS (n=100) erwies sich als wenig rentabel. Die Effizienz beim Gebrauch von DPS war am höchsten. Die Kosten pro Test konnten hier bei einem negativen Vorhersagewert von 95% und minimalem technischen Aufwand auf fast die Hälfte (21 US$, statt 40 US$ pro Assay) reduziert werden.
SCHLUSSFOLGERUNG: Durch die Kombination von Vorauswahlkriterien und Minipoolmethode können die Kosten für das virologische Monitoring der antiretroviralen Therapie auch in Entwicklungsländern gesenkt werden, ohne wesentliche Verluste in Genauigkeit und Validität der Methode verzeichnen zu müssen. DPS stellen eine erfolgsversprechende Möglichkeit dar, die Viruslastbestimmung auch in abgelegen Regionen ohne entsprechenden Laboratorien zu ermöglichen.
MicroRNAs are endogenous ≈22 nt long non coding RNA molecules that modulate gene expression
at the post transcriptional level by targeting mRNAs for cleavage or translational repression.
MicroRNA-mRNA interaction involves a contiguous and perfect pairing within complementary
sites usually in the 3’ UTR of the target mRNA. Heart failure is associated with myocyte
hypertrophy and death, due to compensatory pathological remodeling and minimal functional repair
along with microRNA deregulation.
In this study, we identified candidate microRNAs based on their expression strength in
cardiomyocytes and by their ability to regulate hypertrophy. Expression profiling from early and
late stages of heart failure showed several deregulated microRNAs. Of these microRNAs, miR-378
emerged as a potentially interesting microRNA that was highly expressed in the mouse heart and
downregulated in the failing heart. Antihypertrophic activity of miR-378 was first observed by
screening a synthetic miR library for morphologic effects on cardiomyocytes, and validated in vitro proving the tight control of hypertrophy by this miR. We combined bioinformatic target prediction analysis and microarray analysis to identify the targets of miR-378. These analyses suggested that factors of the MAP kinase pathway were enriched among miR-378 targets, namely MAPK1 itself (also termed ERK2), the insulin-like growth factor receptor 1 (IGF1R), growth factor receptor bound protein 2 (GRB2) and kinase suppressor of ras 1 (KSR1). Regulation of these targets by miR-378 was then confirmed by mRNA and protein expression analysis. The use of luciferase reporter constructs with natural or mutated miR-378 binding sites further validated these four proteins as direct targets of miR-378. RNA interference with MAPK1 and the other three targets prevented the prohypertrophic effect of antimiR-378, suggesting that they functionally relate to miR-378. In vivo restoration of disease induced loss of miR-378 in a pressure overload mouse model of hypertrophy using adeno associated virus resulted in partial attenuation cardiac hypertrophy and significant improvement in cardiac function along with reduced expression of the four targets in heart.
We conclude from these findings that miR-378 is an antihypertrophic microRNA in cardiomyocytes, and the main mechanism underlying this effect is the suppression of the MAP kinase-signaling pathway on four distinct levels. Restoration of disease-associated loss of miR-378 through cardiomyocyte-targeted AAV-miR-378 may prove as an effective therapeutic strategy in myocardial disease.
Einen Satz syntaktisch zu analysieren heißt, ein Satzverstehen nach bestimmten Kriterien offenzulegen. Jeder Satzanalyse geht ein grundsätzliches, voranalytisch verborgenes Verstehen ebenso voraus, wie der Prozess der Analyse ein solches Verstehen konkretisiert und ausdrücklich festzurrt: Ohne Verstehen keine Analyse. Für Analysen, die ein Satzverstehen syntaktisch explizieren sollen, benötigt man einen theoretischen Hintergrund, der die Analysewerkzeuge bereitstellt: Ohne Beschreibungsmittel keine Analyse. Schließlich braucht jede Analyse auch noch eine Darstellungsmethode: Ohne Festhalten der Ergebnisse kein Zugriff auf die Analyse.
Der vorliegende Band versteht sich als eine Explikation von Satzverstehen anhand eines konkreten, valenzorientierten Beschreibungsinventars. Im Vordergrund stehen dabei die Visualisierungen der Analyse-Ergebnisse durch Baumgraphen. Über 100 Bäumchen sind hier zu einem Wald versammelt. Damit ist insbesondere (aber nicht nur) für Studierende, die nach dieser Methode Sätze analysieren, eine Möglichkeit gegeben, einen valenz- und dependenzgrammatischen Zugriff auf verschiedene syntaktisch beschreibbare Phänomene authentischen Sprachvorkommens zu erproben. Darüber hinaus stellt der „Würzburger Wald“ unter Beweis, dass die in diesem Band intensiv angewandte Analysemethode ein stabiles Instrument für adäquate syntaktische Analysen sprachlicher Einheiten aus der freien Wildbahn journalistischer Textproduktion ist.
Der Lagerungsplagiocephalus (sekundärer Plagiocephalus) ist in seiner ansteigenden Inzidenz seit Anfang der Neunziger Jahre ein von verschiedenen Autoren beschriebenes Phänomen. Zunehmend verzeichnet wurde er nach der von dem US Public Health Service im Jahre 1992 veröffentlichten "Back to Sleep Campaign". Zahlreiche kinderärztliche Vereinigungen (wie bspw. die „American Academy of Pedriatrics (APP)“) empfahlen ab diesem Zeitpunkt eine Rückenschlafposition für Neugeborene als SIDS-Prophylaxe (Sudden Infant
Death Syndrome). Der sekundäre Plagiocephalus ist eine Form der Schädeldeformität, dessen Ätiologie auf externe Einflüsse zurückzuführen ist. Durch seinen ähnlichen Phänotypus zur Differentialdiagnose der Craniosynostosen (primäre Plagiocephali) müssen diagnostisch frühzeitig
Verwechslungen ausgeschlossen werden, da sich die therapeutischen Maßnahmen grundlegend unterscheiden. In Fällen schwerer lagerungsbedingter Schädelasymmetrien besteht neben manuellen
Therapieformen die Möglichkeit einer Kopforthesentherapie, die der funktionellen Wachstumssteuerung des Neurokraniums dient.
Aufgrund der aktuellen Diskussion über die Wirksamkeit und Effektivität der
Kopforthesentherapie, der Unstimmigkeit der Krankenkassen zur Kostenübernahme und des großen Interesses der Mitarbeiter bezüglich der Qualität ihrer Arbeit entstand eine Analyse der subjektiven Beurteilung der Beratung, der Betreuung und des Behandlungsprozesses. Auch das Produkt (der Kopforthese) und das Behandlungsergebnis dieser Therapieform wurde durch die Eltern von 83 mit Lagerungsplagiocephalus behandelter Kinder am Cranio Facialen Centrum des Universitätsklinikums Würzburg (CFCW) beurteilt. Des Weiteren sollten die Gründe der Motivation zur Therapieaufnahme und Faktoren einer Reduktion der Besorgnis bzw. Steigerung der Zufriedenheit der Eltern analysiert werden.
Als Erhebungsinstrument dieser Studie diente ein modifizierter Elternfragebogen zur Beurteilung der Behandlung ihres Kindes (FBBm). Die Fragebögen wurden an die Eltern von 33 weiblichen und 60 männlichen Lagerungspagiocephalus-Patienten versandt, die eine abgeschlossene
Kopforthesentherapie zwischen den Jahren 2010 und Januar 2012 verzeichneten.
Die Rücklaufquote der Fragebögen betrug 89,25%. Das Ergebnis der Analyse, zur Motivation der Eltern ein Spezialzentrum aufzusuchen, zeigte, dass diese bei einer erheblichen Anzahl der befragten Eltern aus ihrer seelischen Belastung entsprang, welche sich in Form von Selbstvorwürfen, Ängsten und Sorgen aufgrund der Schädeldeformität ihres Kindes äußerte. Das Ergebnis der Analyse zur Prozessqualität zeigte diesbezüglich eine exzellente Bewertung der befragten Eltern. Die Mitarbeiter des CFCW wurden in der Ausführung ihrer Kommunikationsmethodik
(Gesprächsgestaltung), ihrer Handlungsabläufe und ihrer Betreuung (Hilfestellung) durchweg positiv bewertet. Das Ergebnis der Analyse zur Produktqualität zeigte eine sehr große Akzeptanz und Toleranz im Gebrauch des Therapiemittels. Die Kopforthese wurde in ihrer Wirksamkeit positiv
bewertet, entsprach jedoch nicht vollständig den Qualitätsanforderungen der befragten Eltern. Das äußerst beeindruckende Ergebnis der Analyse zur Ergebnisqualität zeigte eine 96,3%ige Zufriedenheit der Eltern mit dem Behandlungsergebnis ihres Kindes durch Kopforthesentherapie. 96,4% der Befragten waren der Meinung, dass die Helmtherapie für ihr Kind hilfreich war
und sich die Kopfform zudem deutlich besserte.
Weitere Studien zur Zufriedenheit der Eltern mit einem geeigneten Produkt, dessen Defizite behoben wurden und welches den Ansprüchen und Erwartungen der Eltern gerecht wird, sollten zukünftig erfolgen. Eine frühere und umfangreichere Aufklärung der Therapievarianten, insbesondere der Kopforthesentherapie, in der Bevölkerung erscheint sinnvoll, um Eltern und Krankenkassen ihre Verunsicherung bezüglich einer Therapiezustimmung zu nehmen.
With the introduction of OpenFlow by the Stanford University in 2008, a process began in the area of network research, which questions the predominant approach of fully distributed network control. OpenFlow is a communication protocol that allows the externalization of the network control plane from the network devices, such as a router, and to realize it as a logically-centralized entity in software. For this concept, the term "Software Defined Networking" (SDN) was coined during scientific discourse.
For the network operators, this concept has several advantages. The two most important can be summarized under the points cost savings and flexibility. Firstly, it is possible through the uniform interface for network hardware ("Southbound API"), as implemented by OpenFlow, to combine devices and software from different manufacturers, which increases the innovation and price pressure on them. Secondly, the realization of the network control plane as a freely programmable software with open interfaces ("Northbound API") provides the opportunity to adapt it to the individual circumstances of the operator's network and to exchange information with the applications it serves. This allows the network to be more flexible and to react more quickly to changing circumstances as well as transport the traffic more effectively and tailored to the user’s "Quality of Experience" (QoE).
The approach of a separate network control layer for packet-based networks is not new and has already been proposed several times in the past. Therefore, the SDN approach has raised many questions about its feasibility in terms of efficiency and applicability. These questions are caused to some extent by the fact that there is no generally accepted definition of the SDN concept to date. It is therefore a part of this thesis to derive such a definition. In addition, several of the open issues are investigated. This Investigations follow the three aspects: Performance Evaluation of Software Defined Networking, applications on the SDN control layer, and the usability of SDN Northbound-API for creation application-awareness in network operation.
Performance Evaluation of Software Defined Networking: The question of the efficiency of an SDN-based system was from the beginning one of the most important. In this thesis, experimental measurements of the performance of OpenFlow-enabled switch hardware and control software were conducted for the purpose of answering this question. The results of these measurements were used as input parameters for establishing an analytical model of the reactive SDN approach. Through the model it could be determined that the performance of the software control layer, often called "Controller", is crucial for the overall performance of the system, but that the approach is generally viable. Based on this finding a software for analyzing the performance of SDN controllers was developed. This software allows the emulation of the forwarding layer of an SDN network towards the control software and can thus determine its performance in different situations and configurations. The measurements with this software showed that there are quite significant differences in the behavior of different control software implementations. Among other things it has been shown that some show different characteristics for various switches, in particular in terms of message processing speed. Under certain circumstances this can lead to network failures.
Applications on the SDN control layer: The core piece of software defined networking are the intelligent network applications that operate on the control layer. However, their development is still in its infancy and little is known about the technical possibilities and their limitations. Therefore, the relationship between an SDN-based and classical implementation of a network function is investigated in this thesis. This function is the monitoring of network links and the traffic they carry. A typical approach for this task has been built based on Wiretapping and specialized measurement hardware and compared with an implementation based on OpenFlow switches and a special SDN control application. The results of the comparison show that the SDN version can compete in terms of measurement accuracy for bandwidth and delay estimation with the traditional measurement set-up. However, a compromise has to be found for measurements below the millisecond range.
Another question regarding the SDN control applications is whether and how well they can solve existing problems in networks. Two programs have been developed based on SDN in this thesis to solve two typical network issues. Firstly, the tool "IPOM", which enables considerably more flexibility in the study of effects of network structures for a researcher, who is confined to a fixed physical test network topology.
The second software provides an interface between the Cloud Orchestration Software "OpenNebula" and an OpenFlow controller. The purpose of this software was to investigate experimentally whether a pre-notification of the network of an impending relocation of a virtual service in a data center is sufficient to ensure the continuous operation of that service. This was demonstrated on the example of a video service.
Usability of the SDN Northbound API for creating application-awareness in network operation: Currently, the fact that the network and the applications that run on it are developed and operated separately leads to problems in network operation. SDN offers with the Northbound-API an open interface that enables the exchange between information of both worlds during operation. One aim of this thesis was to investigate whether this interface can be exploited so that the QoE experienced by the user can be maintained on high level. For this purpose, the QoE influence factors were determined on a challenging application by means of a subjective survey study. The application is cloud gaming, in which the calculation of video game environments takes place in the cloud and is transported via video over the network to the user. It was shown that apart from the most important factor influencing QoS, i.e., packet loss on the downlink, also the type of game type and its speed play a role. This demonstrates that in addition to QoS the application state is important and should be communicated to the network. Since an implementation of such a state conscious SDN for the example of Cloud Gaming was not possible due to its proprietary implementation, in this thesis the application “YouTube video streaming” was chosen as an alternative. For this application, status information is retrievable via the "Yomo" tool and can be used for network control. It was shown that an SDN-based implementation of an application-aware network has distinct advantages over traditional network management methods and the user quality can be obtained in spite of disturbances.
Theories of attention deficit hyperactivity disorder (ADHD) aetiology have placed a focus on impaired behavioural inhibition presumably leading to executive function (EF) deficits. Neuroimaging studies report neurophysiological findings consistent with these hypothesised impairments, and investigations of functional brain activation from a network perspective report hypoactivation in the frontoparietal network as well as hyperactivation in the dorsal attention network. Studies investigating the acute effects of stimulant medication on EF show an improvement on behavioural EF measures including working memory. In addition, methylphenidate (MPH) was shown to up-regulate the task-positive/ frontoparietal network in children and adolescents with ADHD. So far, there are only few studies investigating the impact of ADHD on behavioural and neurophysiological EF measures as well as the effect of several weeks of stimulant medication in adult patients.
The importance of the catechol-O-methyltransferase (COMT) enzyme for subcortical and cortical dopaminergic and noradrenergic functioning furthermore led to studies investigating a potential interactive impact of COMT genotype and ADHD on neuropsychological functioning, with a particular focus on working memory. The results of these studies were very heterogeneous. In addition, as none of the studies compared the results of ADHD patients to those of a healthy control group, possible differential effects of COMT in patients and healthy controls could not be examined.
The aim of this dissertation was to investigate selective attention properties of the central executive component during a working memory task and to transfer this task to fMRI. A third study then aimed to investigate the effects of adult ADHD (aADHD), MPH, and COMT genotype on working memory with a particular focus on activation of the task-positive network during the analysis of the fMRI data.
The first study (EEG) could replicate and extend the results from previous research. This study could furthermore connect the overall activation in frontal areas to suppression efficiency in posterior visual areas as well as establish the impact of hyperactive/ impulsive ADHD symptoms on task performance. The second study (fMRI) allowed the successful transfer of the paradigm to fMRI, and the further replication and extension of previous findings. In addition, this study showed the sensitivity of the task to the effects of the COMT genotype. The third study (fMRI) was one of the first studies that exploratorily investigated the effects COMT in a sample of aADHD patients and a comparable healthy control group. This study showed an interactive effect of these two factors on neuropsychological measures as well as on fMRI activation during a classic n-back working memory task. In addition, this task led to more activation in the task-positive network of the aADHD group compared to a healthy control group in the absence of performance differences, pointing towards compensatory activation in the aADHD group. Furthermore, activation in the frontal cortex was increased in patients taking MPH compared to a placebo. The fMRI data from the selective attention task moreover showed decreased activation in the right DLPFC of the patient group, which was associated with reduced suppression efficiency across all participants. The clinical effect of MPH in the third study was visible but did not reach significance, which is probably attributable to a lack of experimental power.
The studies in this dissertation could successfully replicate and extend previous findings. A goal for future studies should be the further investigation of the interactive effects of COMT genotype and aADHD on neuropsychological test results and fMRI activation, but also on medication response and adverse effects. In this context, the adaptation of a network perspective during the analysis of fMRI data seems to be the best way to detect existing between-group differences.
Besides caloric restriction, a diet rich in fruits and vegetables is believed to delay the ageing process thus providing a powerfull tool in preventive medicine. To investigate underlying interactions between food ingredients and genes simple models, such as the red flour beetle Tribolium castaneum, appear especially constructive. Here we show that 1 % of a grape seed extract containing 30 % of procyanidins, significantly increases the survival of T. castaneum at 42 °C when added to flour as a dietary source. The beneficial effects of grape seed extract could not be reproduced by supplementing flour with single catechins of which the oligomeric procyanidins consist. We identified previously stress resistance genes responsible for a survival extension by dietary ingredients and show here by the use of RNA-interference that a knockdown of transcripts encoding homologues of Nrf-2 or Jnk-1 block the effects of grape seed extract on survival. Interestingly, grape seed extract under knockdown of Foxo-1 caused a significant survival reduction, stressing the hormetic response as underlying the survival extension by the dietary interventions.
In conclusion, our studies provide evidence that a procyanidin-rich extract is able to extend the survival of the model organism T. castaneum. Catechin monomers, however, appear not to mediate the effects. The active ingredients, moreover, need the presence of stress resistance factors, and here especially of Foxo-1, in order to promote their preventive activities with regard to degenerations.
In attempting to solve the regular inverse Galois problem for arbitrary subfields K of C (particularly for K=Q), a very important result by Fried and Völklein reduces the existence of regular Galois extensions F|K(t) with Galois group G to the existence of K-rational points on components of certain moduli spaces for families of covers of the projective line, known as Hurwitz spaces.
In some cases, the existence of rational points on Hurwitz spaces has been proven by theoretical criteria. In general, however, the question whether a given Hurwitz space has any rational point remains a very difficult problem. In concrete cases, it may be tackled by an explicit computation of a Hurwitz space and the corresponding family of covers.
The aim of this work is to collect and expand on the various techniques that may be used to solve such computational problems and apply them to tackle several families of Galois theoretic interest. In particular, in Chapter 5, we compute explicit curve equations for Hurwitz spaces for certain families of \(M_{24}\) and \(M_{23}\).
These are (to my knowledge) the first examples of explicitly computed Hurwitz spaces of such high genus. They might be used to realize \(M_{23}\) as a regular Galois group over Q if one manages to find suitable points on them.
Apart from the calculation of explicit algebraic equations, we produce complex approximations for polynomials with genus zero ramification of several different ramification types in \(M_{24}\) and \(M_{23}\). These may be used as starting points for similar computations.
The main motivation for these computations is the fact that \(M_{23}\) is currently the only remaining sporadic group that is not known to occur as a Galois group over Q.
We also compute the first explicit polynomials with Galois groups \(G=P\Gamma L_3(4), PGL_3(4), PSL_3(4)\) and \(PSL_5(2)\) over Q(t).
Special attention will be given to reality questions. As an application we compute the first examples of totally real polynomials with Galois groups \(PGL_2(11)\) and \(PSL_3(3)\) over Q.
As a suggestion for further research, we describe an explicit algorithmic version of "Algebraic Patching", following the theory described e.g. by M. Jarden. This could be used to conquer some problems regarding families of covers of genus g>0.
Finally, we present explicit Magma implementations for several of the most important algorithms involved in our computations.
Attention-deficit/hyperactivity disorder (ADHD) is a highly prevalent childhood-onset neurodevelopmental disorder that involves a substantial risk of persisting into adolescence and adulthood. A number of genome-wide screening studies in ADHD have been conducted in recent years, giving rise to the discovery of several variants at distinct chromosomal loci, thus emphasising the genetically complex and polygenic nature of this disorder. Accordingly, promising novel candidate genes have emerged, such as the gene encoding the glucose transporter isoform 3 (SLC2A3) and the gene encoding the latrophilin isoform 3 (LPHN3).
In this thesis, both genes were investigated in form of two separated projects. The first focused on SLC2A3 polymorphisms associated with ADHD and their potential physiological impact. For this purpose, gene expression analyses in peripheral cell models were performed as well as functional EEG measurements in humans. The second project concerned the murine gene Lphn3 including the goal of developing a mouse line containing a genetically modified Lphn3 with conditional knockout potential. In this respect, a specific DNA vector was applied to target the Lphn3 gene locus in murine embryonic stem (ES) cells as a prerequisite for the generation of appropriate chimeric mice.
The results of the first project showed that SLC2A3 duplication carriers displayed increased SLC2A3 mRNA expression in peripheral blood cells and significantly altered event-related potentials (ERPs) during tests of cognitive response control and working memory, possibly involving changes in prefrontal brain activity and memory processing. Interestingly, ADHD patients with the rs12842 T-allele, located within and tagging the SLC2A3 gene, also exhibited remarkable effects during these EEG measurements. However, such effects reflected a reversed pattern to the aforementioned SLC2A3 duplication carriers with ADHD, thus indicative of an opposed molecular mechanism. Besides, it emerged that the impact of the aforementioned SLC2A3 variants on different EEG parameters was generally much more pronounced in the group of ADHD patients than the healthy control group, implying a considerable interaction effect. Concerning the second project, preliminary results were gathered including the successful targeting of Lphn3 in murine ES cells as well as the production of highly chimeric, phenotypically unremarkable and
mostly fertile mouse chimeras. While germline transmission of the modified Lphn3 allele has not yet occurred, there are still several newborn chimeric mice that will be tested in the near future.
In conclusion, the findings suggest that SLC2A3 variants associated with ADHD are accompanied by transcriptional and functional changes in humans. Future research will help to elucidate the molecular network and neurobiological basis involved in these effects and apparently contributing to the complex clinical picture of ADHD. Moreover, given the increasing number of publications concerning latrophilins in recent years and the multitude of research opportunities provided by a conditional knockout of Lphn3 in mice, the establishment of a respective mouse line, which currently is in progress, constitutes a promising approach for the investigation of this gene and its role in ADHD.
TRAIL is a member of TNF superfamily and mediates apoptosis by binding to two DRs, TRAILR1 and TRAILR2. Despite the fact that there are other TRAILRs, TRAILR1 and TRAILR2 receive the major research interest due to their ability to trigger apoptosis and their possible use as targets in tumor therapy. Due to the potential advantages of TRAILR1- or TRAILR2-specific targeting, we investigated recently published TRAIL DR-specific mutants, one conferring specificity for TRAILR1 (TRAILmutR1) and one for TRAILR2 (TRAILmutR2). It was well proved in this work that TRAILmutR1 shows specific binding to TRAILR1 and no specific binding to TRAILR2. TRAILmutR2 vice versa shows specific binding to TRAILR2 and no significant binding to TRAILR1. Moreover, these mutants were able to induce caspase activation and cell death in a TRAILR1/2-specific manner. Moreover, the enhancement of TRAILR2-induced apoptosis by secondary oligomerization of soluble wild-type TRAIL was confirmed for the TRAILR2-specifc TRAIL mutant and similar findings were made with the TRAILR1-specific TRAIL mutant.
The soluble form of TRAIL exhibits weak apoptotic activity as compared to transmembrane TRAIL. Therefore, there is the challenge in clinical research to improve the activity of soluble TRAIL. A second strategy besides the above mentioned oligomerization to improve soluble TRAIL activity is anchoring of the molecule to the cell surface, e.g. through the genetic fusion with a scFv domain recognizing a cell surface antigen. In this work, we generated fusion proteins of TRAIL, TRAILmutR1 and TRAILmutR2 with a scFv recognizing CD40 (scFv:G28). Initially, we analyzed the functionality of both the TRAIL domain and the scFv:G28 domain of the corresponding fusion proteins. TRAIL functionality was well proved through its ability to induce cell death in TRAIL sensitive cells such as Jurkat cells, provided that scFv:G28-TRAIL fusion proteins were oligomerized by anti-Flag mAb M2. Concerning the scFv:G28 domain, the fusion proteins showed enhanced binding affinity to cell lines expressing CD40 as compared to their parental CD40-negative cells. Consistent with previous studies investigating TRAIL fusion proteins with other cell surface antigen-targeting scFvs, the scFv:G28 fusion proteins with TRAIL, TRAILmutR1 and TRAILmutR2 showed enhanced induction of cell death in a CD40-dependent manner. Moreover, our results revealed that these fusion proteins have a significant paracrine apoptotic effect on CD40-negative bystander cells upon anchoring to CD40-positive cells which are TRAIL resistant. Thus, the current work provides for the first time scFv fusion proteins of TRAIL and TRAILR1- and TRAILR2-specific TRAIL mutants with CD40-restricted activity. These fusion proteins provide the advantage of attenuating the off-target effects and the potential side effects of per se highly active TRAIL variants on one hand due to the CD40-binding dependent enhancement of activity and on the other hand due to the differential use of TRAILR1 and TRAILR2.
CD40 represents a tumor associated marker which is expressed on many tumor cells but also on immune cells. Therefore, the last part of this work focused on the analysis of the ability of scFv:G28-TRAIL fusion proteins to induce CD40 signaling both in tumor cells and also in immune cells. It turned out that the scFv:G28-TRAIL fusion proteins are able to induce CD40 signaling in CD40-positive tumor cells but especially also in immune cells such as iDCs leading to their maturation and further activation of immune responses.
Taken together, this work provides novel bifunctional scFv-TRAIL fusion proteins which combine the induction of apoptosis via TRAIL DR with stimulation of CD40 signaling which possibly enhances antitumor immunity.
The synaptonemal complex (SC) is a highly conserved structure in sexually reproducing organism. It has a tripartite, ladder-like organization and mediates the stable pairing, called synapsis, of the homologous chromosomes during prophase of meiosis I. Failure in homolog synapsis result in aneuploidy and/or apoptosis of the developing germ cells.
Since 1956, the SC is subject of intense research and its presence was described in various species from yeast to human. Its structure was maintained during millions of years of evolution consist-ing of two parallel lateral elements (LEs), joined by numerous transverse filaments (TFs) which run perpendicular to the LEs and an electron dense central element (CE) in the middle of the SC. Individual protein components, however, were characterized only in few available model organ-isms, as for example Saccharomyces cerevisiae, Arabidopsis thaliana, Drosophila melanogaster, Ceanorhabditis elegans and Mus musculus. Rather unexpectedly, these characterizations failed to detect an evolutionary homology between the protein components of the different SCs. This fact challenged the general idea of a single origin of the SC in the evolution of meiosis and sexual reproduction.
This thesis now addressed itself to the task to unravel the discrepancy between the high conser-vation of the SC structure and its diverse and apparently non-homologous protein composition, focusing on the animal kingdom. It is the first study dealing with the evolution of the SC in Meta-zoa and demonstrates the monophyly of the mammalian SC components in metazoan species. The thesis demonstrates that at least four out of seven murine SC proteins emerged in Eumeta-zoa at the latest and have been likewise part of an ancient SC as it can be found in the present-day cnidarian species Hydra. This SC displays the common organization and already possesses the minimal protein kit corresponding to the three different structural domains: LEs, TFs and the CE. Additionally, the individual phylogenies of the murine SC proteins revealed the dynamic evolu-tionary history of the ancient SC. Further components were added during the diversification of Bilateria and vertebrates while ancestral proteins likely duplicated in the vertebrate lineage and diversified or got lost in the branch leading to ecdysozoan species. It is hypothesized that the apparently non-homologous SC proteins in D. melanogaster and C. elegans actually do derive from the ancient SC proteins but diversified beyond recognition during the fast evolution of Ar-thropoda and Nematoda.
The study proposes Hydra as an alternative invertebrate model system for meiosis and SC re-search to the standard organisms D. melanogaster and C. elegans. Recent results about the cni-darian SC as well as the possible application of standard methods is discussed and summarized in the concluding section.
Adenosinrezeptoren werden auf nahezu allen Körperzellen exprimiert und übernehmen dort vielfältige und wichtige Funktionen. Auch auf diversen Tumorzelllinien konnten bereits Adenosinrezeptoren nachgewiesen und – je nach Subtyp – mit Pro- oder Anti-tumor-Effekten in Zusammenhang gebracht werden.
In dieser Arbeit wurden Gebärmutterhalskrebszellen sowie endometriale und triple-negative Brustkrebszellen auf Expression und mögliche Funktionen von Adenosinrezep-toren untersucht. Da spezifische Antikörper bis heute nicht verfügbar sind, wurde ein pharmakologischer Ansatz mit subtypspezifischen Agonisten und Antagonisten gewählt.
In Radioliganden-Bindungsassays, konnte nachgewiesen werden, dass sich auf der Zer-vixkarzinom-Zelllinie SiHa und der Brustkrebs-Zelllinie HCC1806 Adenosinrezeptoren des Subtyps A1 befinden. Die endometrialen Krebszelllinien Ishikawa und HEC-1-A exprimieren Rezeptoren vom Subtyp A1 und A2A. A3-Adenosinrezeptoren wurden auf keiner der untersuchten Zelllinien gefunden.
Der Nachweis von A2B-Rezeptoren kann mit dem Radioliganden-Bindungsassay nicht erbracht werden, da bislang kein Radioligand bekannt ist, der eine ausreichende Affini-tät besitzt, um diesen Subtyp zweifelsfrei nachweisen zu können.
Obwohl die Mehrheit der untersuchten Zelllinien Adenosinrezeptoren exprimiert, konnte ein signifikanter Effekt auf die Adenylatcyclase bei Stimulation der auf den Zellen vorhandenen Adenosinrezeptoren nur bei den HEC-1-A-Zellen festgestellt werden. Auch auf funktionelle A2B-Rezeptoren fand sich im Adenylatcyclaseassy kein Hinweis.
Im durchgeführten Kristallviolettassay zeigte sich ein proapoptotischer Effekt auf Ishi-kawa- und HEC-1-A-Zellen bei hohen Adenosin-Konzentrationen (100 µM). Die im BrdU-Assay gemessene Proliferationsrate hingegen änderte sich nach Vorbehandlung mit Adenosin nicht. Das metabolisch stabilere NECA (in Kombination mit ADA) hatte im Kristallviolettassay einen stärkeren Einfluss auf die Apoptoserate der jeweiligen Zelllinie als Adenosin und auch im BrdU-Assay sank die Menge an inkorporiertem BrdU. Ein Synergismus zwischen Stimulation von Adenosinrezeptoren und diversen Todesliganden bzw. Chemotherapeutika konnte nicht nachgewiesen werden.
Freies extrazelluläres Adenosin kann auch aus dem Abbau von ATP generiert werden, wenn Zellen die Ektonukleotidasen CD39 und CD73 exprimieren. Aufgrund der im-munsuppressiven Wirkung von Adenosin können diese Enzyme T-Zell- und NK-Zellantworten im Mikromilieu von Tumoren hemmen. Die durchflusszytometrische Analyse von HEC-1-A- und Ishikawa-Zellen zeigte zwar, dass die Expression von CD39 und CD73 nach Stimulation der Adenosinrezeptoren unverändert blieb. Die Ex-pression von Enzymen, lässt aber vermuten, dass die Zellen in vivo von Adenosin profi-tieren könnten. Angesichts der in vitro Daten, die allenfalls einen wachstumshemmen-den Effekt von Adenosin zeigten, könnte die vorrangige Wirkung von Adenosin im Tumormikromilieu tatsächlich auf der Inhibition von Immunantworten beruhen. Mög-licherweise würden die Rezeptoren dann in erster Linie als Sensoren dienen.
Weitere Forschungsarbeit wird helfen, die Rolle der Adenosinrezeptoren im Tumorge-schehen vollständig zu verstehen und möglicherweise für die Krebstherapie nutzbar zu machen.
Die vorliegende Arbeit setzt sich mit den europa- und verfassungsrechtlichen Rechtsfragen der Einführung sogenannter Eurobonds auseinander.
Einleitend ordnet der Autor das Konzept der Eurobonds in den Kontext der Finanzkrise ein, um anschließend eine Begriffsklärung vorzunehmen. Dabei erläutert er auch die veränderbaren „Bausteine“ der Eurobonds und skizziert die Vielzahl bereits diskutierter Modelle.
Im zweiten Abschnitt des Hauptteils widmet sich der Autor sodann den europarechtlichen Rechtsfragen der Eurobonds. Im Fokus stehen hier insbesondere die Erörterung der denkbaren Kompetenzgrundlagen und die Frage der Vereinbarkeit mit der sogenannten No-Bail-Out-Klausel des Art. 125 Abs. 1 AEUV. Dabei gelangt er zu dem Befund der Unvereinbarkeit von Eurobonds jeglicher Art mit dem Telos des Art. 125 Abs. 1 AEUV. Als Konsequenz wird auf die Notwendigkeit einer Vertragsänderung hingewiesen, wobei der Autor für die Durchführung des ordentlichen Vertragsänderungsverfahrens gem. Art. 48 Abs. 2-5 EUV plädiert.
Im dritten Abschnitt wendet sich der Autor den verfassungsrechtlichen Rechtsfragen zu. Dabei wirft er ausgehend von der Integrationsverantwortung der deutschen Staatsorgane die Frage der Vereinbarkeit von Eurobonds mit der Budgethoheit des Bundestages auf. So gelangt er schließlich zu dem Befund, dass die Einführung von Eurobonds mit Blick auf das Budgetrecht im Grundsatz möglich ist. Im Anschluss hieran werden die verfassungsrechtlichen Grenzen der Übernahme von Gewährleistungen und die sich daraus ergebenden Folgen für die Einführung von Eurobonds diskutiert.
In einem Resümee werden Ergebnisse der juristischen Analyse mit einem kurzen Seitenblick auf die US-amerikanische Geschichte zusammengefasst.
Evolution of Vγ9Vδ2 T-cells

Human Vγ9Vδ2 T cells are the major subset of blood γδ T cells and account for 1-5% of blood T cells. Pyrophosphorylated metabolites of isoprenoid biosynthesis are recognized by human Vγ9Vδ2 T cells and are called as phosphoantigens (PAg). Isopentenyl pyrophosphate (IPP) and (E)-4-Hydroxy-3-methyl-but-2-enyl pyrophosphate (HMBPP) are among the few well studied PAg. IPP is found in all organisms while HMBPP is a precursor of IPP found only in eubacteria, plants and apicomplexaen parasite. Interestingly, the PAg reactive Vγ9Vδ2 T cells are so far identified only in human and higher primates but not in rodents. Hence, Vγ9Vδ2 T cells are believed to be restricted to primates. With regard to PAg recognition, a Vγ9JP recombined TCRγ chain and certain CDR3 motifs of the TCR chain are mandatory. The BTN3A1 molecule is essential for a response to PAg. BTN3 is a trans-membrane protein belonging to butyrophilin family of proteins. Though BTN3A1 was found to be essential for PAg presentation, the exact molecular basis of PAg presentation still remains unclear.
This thesis presents new data on the evolution of Vγ9Vδ2 TCR and its ligands (BTN3) as well as the genetic basis of PAg presentation to Vγ9Vδ2 TCR.
The comprehensive analysis of genomic database sequences at NCBI and other public domain databases revealed for the first time that Vγ9, Vδ2 and BTN3 genes emerged and co-evolved along with the placental mammals. Vγ9, Vδ2 and BTN3 genes are scattered across mammalian species and not restricted to primates. But interestingly, all three genes are highly conserved between phylogenetically distinct species. Moreover, the distribution pattern of Vγ9, Vδ2 TCR genes and BTN3 genes suggests a functional association between these genes representing the TCR - ligand relationship. Alpaca (Vicugna pacos), a member of the camelid family, is one among the 6 candidate non-primate species which were found to possess functional Vγ9, Vδ2 and BTN3 genes.
From peripheral lymphocytes of alpaca, Vγ9 chain transcripts with a characteristic JP rearrangement and transcripts of Vδ2 chains with a CDR3 typical for PAg-reactive TCR were identified. The transduction of αβ TCR negative mouse thymoma BW cells with alpaca Vγ9 and Vδ2 TCR chains resulted in surface expression of the TCR complex as it was deduced from detection of cell surface expression of mouse CD3. Cross-linking of alpaca Vγ9Vδ2 TCR transductants with anti-CD3ε led to IL-2 production which confirmed that alpaca Vγ9 and Vδ2 TCR chains pair to form a functional TCR. Besides the conservation of human like Vγ9 and Vδ2 TCR chains, alpaca has conserved an orthologue for human BTN33A1 as well. Interestingly, the predicted PAg binding sites of human BTN3A1 was 100% conserved in deduced amino acid sequence of alpaca BTN3A1. All together alpaca is a promising candidate for further studies as it might have preserved Vγ9Vδ2 T cells to function in surveillance of stress and infections.
This thesis also provides the sequence of Vγ9Vδ2 TCR of African green monkey (Chlorocebus aethiops), which was previously unknown. Moreover, our data indicates the lack of any species specific barrier which could hinder the PAg presentation by African monkey derived COS cells to human Vγ9Vδ2 TCR and vice versa of human cells to African green monkey Vγ9Vδ2 TCR which was in contradiction to previously reported findings.
Apart from the above, the thesis also presents new data on the genetic basis of PAg presentation to Vγ9Vδ2 T cells, which revealed that human chromosome 6 is sufficient for the presentation of exogenous and endogenous PAg. By employing human/mouse somatic hybrids, we identified the role of human chromosome 6 in PAg presentation and in addition, we observed the lack of capacity of human chromosome 6 positive hybrids to activate Vγ9Vδ2 TCR transductants in the presence of the alkylamine sec-butylamine (SBA). Investigation of Chinese hamster ovary (CHO) cells containing the human chromosome 6 also yielded similar results. This suggests that aminobisphosphonates (zoledronate) and alkylamines employ different mechanisms for activation of Vγ9Vδ2 T cells although both have been described to act by inhibition of farnesyl pyrophosphate synthase activity which is known to increase intracellular levels of the IPP.
In conclusion, this thesis suggests that Vγ9, Vδ2 and BTN3 genes controlling Vγ9Vδ2 TCR- ligand relationship emerged and co-evolved along with placental mammals; and also identified candidate non-primate species which could possess Vγ9Vδ2 T cells. Furthermore, it suggests alpaca as a promising non-primate species to investigate the physiological function of Vγ9Vδ2 T cells. With respect to PAg antigen presentation it was shown that chromosome 6 is essential and sufficient for exogenous and endogenous PAg presentation. Moreover, the alkylamine SBA and aminobisphosphonate zoledronate may engage different cellular mechanism to exert inhibition over IPP consumption. The thesis raises interesting questions which need to be addressed in future: 1) What are the environmental and evolutionary factors involved in preservation of Vγ9Vδ2 T cells only by few species? 2) What could be the functional nature and antigen recognition properties of such a conserved T cell subset? 3) What is the genetic and molecular basis of the differential capacity of human chromosome 6 bearing rodent-human hybridoma cells in activating Vγ9Vδ2 T cells in presence of SBA and aminobisphosphonates?
The subject of this thesis is the controllability of interconnected linear systems, where the interconnection parameter are the control variables. The study of accessibility and controllability of bilinear systems is closely related to their system Lie algebra. In 1976, Brockett classified all possible system Lie algebras of linear single-input, single-output (SISO) systems under time-varying output feedback. Here, Brockett's results are generalized to networks of linear systems, where time-varying output feedback is applied according to the interconnection structure of the network. First, networks of linear SISO systems are studied and it is assumed that all interconnections are independently controllable. By calculating the system Lie algebra it is shown that accessibility of the controlled network is equivalent to the strong connectedness of the underlying interconnection graph in case the network has at least three subsystems. Networks with two subsystems are not captured by these proofs. Thus, we give results for this particular case under additional assumption either on the graph structure or on the dynamics of the node systems, which are both not necessary. Additionally, the system Lie algebra is studied in case the interconnection graph is not strongly connected. Then, we show how to adapt the ideas of proof to networks of multi-input, multi-output (MIMO) systems. We generalize results for the system Lie algebra on networks of MIMO systems both under output feedback and under restricted output feedback. Moreover, the case with generalized interconnections is studied, i.e. parallel edges and linear dependencies in the interconnection controls are allowed. The new setting demands to distinguish between homogeneous and heterogeneous networks. With this new setting only sufficient conditions can be found to guarantee accessibility of the controlled network. As an example, networks with Toeplitz interconnection structure are studied.
Kinasen der SRC-Familie (SFKs) sind sowohl in Wachstum und Metastasierung von Tumor- und Leukämiezellen als auch an prominenter Stelle in vielgestaltige Signalwege aller Immunzellen involviert. Eine Hemmung von SFKs ist damit ein vielversprechendes Mittel zur Therapie maligner Erkrankungen, kann aber darüber hinaus auch sehr effektiv zur Immunmodulation genutzt werden. Für den zur Therapie von CML und AML zugelassenen Tyrosinkinaseinhibitor (TKI) Dasatinib (Handelsname Sprycel®), für den unter anderem SFKs die Hauptziele darstellen, wurden, neben der antitumoralen Wirkung, sowohl immunsuppressive als auch immunstimulierende Effekte beschrieben. Aus diesem Grund könnte Dasatinib ein für die Modulation von Immunantworten sehr interessantes Hilfsmittel darstellen. In der vorliegenden Arbeit werden die hemmenden und fördernden Einflüsse von Dasatinib auf zwei Typen von Immunzellen genauer untersucht, um so die Auswirkungen einer Dasatinib-Behandlung auf Zellen des Immunsystems besser zu verstehen und sich das immunmodulatorische Potenzial von Dasatinib besser nutzbar machen zu können.
Der erste Teil der Arbeit beschäftigt sich mit der Untersuchung möglicher kombinatorischer Effekte zwischen Dasatinib und dem Glucocorticoid Dexamethason auf verschiedene Subsets von T-Zellen vor dem Hintergrund eines potentiellen Einsatzes der Kombination bei der allogenen Hämatopoetischen Stammzelltransplantation (HSCT) zur Separation von Graft-versus-Leukemia (GvL)-Effekten und der Graft-versus-host Disease (GvHD). Während keine kombinatorischen Effekte bei der Aktivierung von T-Zellen auftraten, ergaben sich bei der Untersuchung des Einflusses auf die Proliferation besonders in CD8+ T-Zellen additive Effekte durch die Kombination. Die Proliferation naiver T-Zell-Subsets wurde bereits durch die beiden Einzelsubstanzen alleine stark gehemmt. Dagegen waren Memory T-Zell-Subsets deutlich unempfindlicher, allerdings konnte durch eine Kombination von Dexamethason und Dasatinib auch die Proliferation dieser Memory Subsets effektiv gehemmt werden. Hierbei zeigten sich bei CD8+ Memory Subsets die deutlichsten synergistischen Effekte. Da eine Kombination in stärkerem Maße auch CD8+ gegenüber CD4+ Memory Subsets hemmt und diese Subsets unterschiedliche Rollen in der Induktion von GvL-Effekten und der Auslösung einer GvHD zu spielen scheinen, ist eine Steigerung der GvL-Effektivität durch die Medikamenten-Kombination bei gleichzeitiger Minimierung eines GvHD-Risikos in Zusammenhang mit anderen publizierten Ergebnissen durchaus denkbar. Weil eine starke Hemmung von virus-spezifischen T-Zellen nur bei sehr hohen Konzentrationen auftrat, ist zudem das Risiko einer Virus-Reaktivierung, die ein großes Problem bei einer HSCT darstellt, eher als gering einzuschätzen.
Der zweite Teil der Arbeit befasst sich mit dem Einfluss von Dasatinib auf aus Monozyten generierte Dendritische Zellen (moDCs) mit einem Fokus auf der Beeinflussung ihrer Migration. Während eine Behandlung mit Dasatinib nur sehr geringe Auswirkungen auf die Ausreifung der moDCs und die Expression von kostimulatorischen Molekülen hatte, führte eine Dasatinib-Behandlung zu einer Zeit- und Dosis-abhängigen Verringerung der Zytokinsekretion (IL-10 und IL-12). Im Gegensatz dazu hatte Dasatinib keinen Einfluss auf die phagozytotische Aktivität der moDCs und auf ihre Fähigkeit, Virus-spezifische T-Zell-Antworten auszulösen. Dasatinib zeigte dagegen einen deutlich steigernden Einfluss auf die Migration von moDCs gegen einen CCL19-Gradienten im Transwell-Assay, ohne die Expression des CCL19-Rezeptors CCR7 zu beeinflussen. Da ähnliche Migrations-steigernde Effekte auch bei einer Behandlung mit dem spezifischen SFK-Inhibitor SKI-1 auftraten, eine Behandlung mit Nilotinib, einem TKI der nicht auf SFKs wirkt, im Gegensatz dazu aber zu einer Hemmung der Migration führte, liegt es nahe dass die Migrations-steigernde Wirkung von Dasatinib über SFKs vermittelt wird. Dasatinib führte zu einer deutlichen Inhibierung der Phosphorylierung der inhibitorischen Immunrezeptoren Siglec-9 und Siglec-3 (CD33) ohne ihre Expressionslevel zu beeinflussen. Eine mit spezifischen Antikörpern durchgeführte Blockierung dieser Immunrezeptoren, deren ITIM-Domänen mutmaßlich von SFKs phosphoryliert werden, hatte eine deutliche Steigerung der Migration und eine verringerte Phosphorylierung von Siglec-9, Siglec-3 und SHP-2 zur Folge. Letztere ist eine Phosphatase, die nach Bindung an phosphorylierte ITIM-Domänen von Rezeptoren wie den Siglecs verschiedene Zielmoleküle dephosphoryliert. Die Ergebnisse dieser Arbeit legen nahe, dass die Migrations-steigernde Wirkung von Dasatinib über eine Hemmung von SFKs und daraus resultierend auf dem Wegfall eines inhibitorischen Signalwegs erfolgt. Diese Steigerung der Migration könnte in der Tumor-Therapie von großem Nutzen sein, da bei einer Vakzinierung mit autologen DCs, die mit Tumor-assoziierten Antigenen stimuliert wurden, die schlechte Einwanderung in die Lymphknoten eines der Hauptprobleme darstellt. Zur Überwindung dieses Problems könnte Dasatinib ein sehr effektives Hilfsmittel darstellen und die Therapie-Effizienz deutlich verbessern. Da Dasatinib aber auch eine ganze Reihe weiterer, sehr vielfältiger Einflüsse auf alle Arten von Immunzellen ausübt, scheint eine Verwendung spezifischer blockierender α-Siglec-Antikörper auf Grund geringerer Nebenwirkungen im Vergleich zu Dasatinib möglicherweise sogar noch deutlich besser geeignet zu sein, das Migrationsverhaltens Dendritischer Zellen positiv zu beeinflussen. Die Verwendung gegen Siglec-Rezeptoren gerichteter Antikörper als Adjuvantien könnte somit zu einem erfolgreicheren Einsatz der Vakzination mit Dendritischen Zellen in der Tumor-Therapie führen.
The auditory system is an exquisitely complex sensory organ dependent upon the synchronization of numerous processes for proper function. The molecular characterization of hereditary hearing loss is complicated by extreme genetic heterogeneity, wherein hundreds of genes dispersed genome-wide play a central and irreplaceable role in normal hearing function. The present study explores this area on a genome-wide and single gene basis for the detection of genetic mutations playing critical roles in human hearing.
This work initiated with a high resolution SNP array study involving 109 individuals. A 6.9 Mb heterozygous deletion on chromosome 4q35.1q35.2 was identified in a syndromic patient that was in agreement with a chromosome 4q deletion syndrome diagnosis. A 99.9 kb heterozygous deletion of exons 58-64 in USH2A was identified in one patient. Two homozygous deletions and five heterozygous deletions in STRC (DFNB16) were also detected. The homozygous deletions alone were enough to resolve the hearing impairment in the two patients. A Sanger sequencing assay was developed to exclude a pseudogene with a high percentage sequence identity to STRC from the analysis, which further solved three of the six heterozygous deletion patients with the hemizygous, in silico predicted pathogenic mutations c.2726A>T (p.H909L), c.4918C>T (p.L1640F), and c.4402C>T (p.R1468X). A single patient who was copy neutral for STRC and without pathogenic copy number variations had compound heterozygous mutations [c. 2303_2313+1del12 (p.G768Vfs*77) and c.5125A>G (p.T1709A)] in STRC. It has been shown that STRC has been previously underestimated as a hearing loss gene. One additional patient is described who does not have pathogenic copy number variation but is the only affected member of his family having hearing loss with a paternally segregating translocation t(10;15)(q26.13;q21.1).
Twenty-four patients without chromosomal aberrations and the above described patient with an USH2A heterozygous deletion were subjected to a targeted hearing loss gene next generation sequencing panel consisting of either 80 or 129 hearing-relevant genes. The patient having the USH2A heterozygous deletion also disclosed a second mutation in this gene [c.2276G>T (p.C759F)]. This compound heterozygous mutation is the most likely cause of hearing loss in this patient. Nine mutations in genes conferring autosomal dominant hearing loss [ACTG1 (DFNA20/26); CCDC50 (DFNA44); EYA4 (DFNA10); GRHL2 (DFNA28); MYH14 (DFNA4A); MYO6 (DFNA22); TCF21 and twice in MYO1A (DFNA48)] and four genes causing autosomal recessive hearing loss were detected [GJB2 (DFNB1A); MYO7A (DFNB2); MYO15A (DFNB3), and USH2A]. Nine normal hearing controls were also included. Statistical significance was achieved comparing controls and patients that revealed an excess of mutations in the hearing loss patients compared to the control group. The family with the GRHL2 c.1258-1G>A mutation is only the second family published worldwide with a mutation described in this gene to date, supporting the initial claim of this gene causing DFNA28 hearing loss. Audiogram analysis of five affected family members uncovered the progressive nature of DFNA28 hearing impairment. Regression analysis predicted the annual threshold deterioration in each of the five family members with multiple audiograms available over a number of years.
Das Ziel dieser Dissertation besteht darin, die für sein Künstlerleben relevanten Aspekte der Biografie Sándor (Alexander)von Wagners darzustellen. Dabei bewegen wir uns auf einem noch kaum erforschten Gebiet und bieten gleichsam die nötigen Anhaltspunkte für weitere Forschungsbeiträge. Des Weiteren sind die Werke Wagners zu betrachten. Insbesondere die anhand der Biographie belegbaren Einflüsse auf seine Werke und die von seinen Werken ausgehende Wirkung sind dabei zu berücksichtigen, um anhand der Darstellung von Wagners Position, peripher auch diejenige osteuropäischer Künstler, im gesamteuropäischen Kontext aufzudecken.
Im Mittelpunkt der vorliegenden Dissertation steht der künstlerische Werdegang Sándor (Alexander) von Wagners. Er stammte aus Ungarn, ließ sich später in München als professioneller Kunstmaler nieder und arbeitete von 1866 bis 1869 als Hilfslehrer und von 1869 bis 1910 als ordentlicher Professor für Maltechnik an der Münchner Kunstakademie. In dieser Abhandlung werden die für die künstlerische Entwicklung wesentlichen Aspekte seines Lebenslaufes dargestellt:
- die künstlerische Laufbahn Wagners unter besonderer Betrachtung seiner künstlerischen Vorbereitung und Studien sowie seiner Karriere in München
- Grundzüge seiner Werke und Einflüsse, die Wagners Lehrer und Professoren auf sein künstlerisches Schaffen ausübten
- Wagners kunstpädagogische Tätigkeit an der Königlichen Kunstakademie in München
- sein Privatleben in München
Daneben liefert diese Untersuchung eine Liste mit Wagners Studenten und legt ein vorläufiges Werkverzeichnis vor. Vollständigkeit war auf Grund der prekären Forschungslage im Rahmen einer einzelnen Dissertation nicht anzustreben, sondern es galt, eine erste grundlegende Basis zu schaffen, deren Ergänzung weiteren Forschungsbemühungen vorbehalten bleiben muss. Diese Arbeit will damit als Anregung zu weiteren Untersuchungen dienen.
Fragestellung: Allgemeine und mütterspezifische Kontextfaktoren in Zusammenhang mit der Kindererziehung können zu einer mütterspezifischen Problemkonstellation führen, die zur Ausbildung oder Aufrechterhaltung von Gesundheitproblemen beiträgt (vgl. Medizinischer Dienst des Spitzenverbandes Bund der Krankenkassen e.V., 2012). Als Vorsorge- und Rehabilitationsmaßnahmen sind für diesen Bereich Mutter-Kind-Maßnahmen etabliert und eine Qualitätssicherung gesetzlich vorgeschrieben. In dieser Studie wurde untersucht, wie die Prozessqualität von den Müttern wahrgenommen und beschrieben wird.
Material und Methoden: Als Material dienten 147 Erfahrungsberichte aus dem
Internet von Müttern, die an einer Mutter-Kind-Maßnahme teilgenommen hatten.
Die Auswertung erfolgte durch eine qualitative Inhaltsanalyse nach Mayring (2008). Es wurde ein induktives Verfahren gewählt, um eine materialgeleitete Bildung der Kategorien zu erreichen. Kodiert wurden Textstellen, die sich auf Merkmale der Prozessqualität bezogen sowie Bewertungen der Patientinnen von globalen Aspekten und der Maßnahme im Ganzen (Fazit). Insgesamt wurden 2424 Textstellen kodiert, die in 157 Kategorien erfasst und in neun Oberbereiche eingeteilt wurden. Die Strukturierung des Kategoriensystems orientiert sich an dem „idealen Prozess“, der durch eine Expertengruppe unter der Leitung der Universität Würzburg formuliert wurde. Es erfolgte eine Analyse der Nennungshäufigkeiten der Kategorien, der Verteilung auf negative und positive Kategorien, der Bewertungskategorien und der Richtung der Bewertung, sowie des Fazits. Es wurden absolute und relative Häufigkeiten berechnet.
Ergebnisse: Die Mütter beschreiben überdurchschnittlich häufig Merkmale, die in die Phasen Anreise und Hoteleigenschaften kodiert wurden. Besonders häufig werden positive Kategorien in der Phase Anreise genannt, wohingegen viele negative Merkmale in der Phase Hoteleigenschaften beschrieben werden. Die Mütter zeigen sich insgesamt zufrieden mit der Maßnahme (79%), die ärztliche Betreuung und das Freizeitangebot werden allerdings überdurchschnittlich negativ bewertet.
Schlussfolgerung: Diese Studie konnte zeigen, dass Mütter einen anderen Schwerpunkt in der Prozessqualität setzen als Experten. Prozessmerkmale, die sich auf die Hoteleigenschaften beziehen, sind ihnen besonders wichtig. Hier besteht möglicherweise Verbesserungsbedarf. Es sollte geprüft werden, ob eventuell eine vermehrte Berücksichtigung des Bereichs Hoteleigenschaften in zukünftigen Qualitätssicherungsverfahren im Bereich der Prozessqualität sinnvoll wäre, um die Sicht der Mütter zu integrieren. Das Verfahren und das Kategoriensystem sollten an anderen Stichproben validiert werden, um mögliche Schwächen in der Methodik auszuschließen. Eine Überprüfung der Ergebnisse durch andere Erhebungsmethoden ist ebenfalls wünschenswert.
