Background: Multiple sclerosis (MS) may cause variable functional impairment. The discrepancy between functional impairment and brain imaging findings in patients with MS (PwMS) might be attributed to differential adaptive and consolidation capacities. Modulating those abilities could contribute to a favorable clinical course of the disease.
Objectives: We examined the effect of cerebellar transcranial direct current stimulation (c-tDCS) on locomotor adaptation and consolidation in PwMS using a split-belt treadmill (SBT) paradigm.
Methods: 40 PwMS and 30 matched healthy controls performed a locomotor adaptation task on a SBT. First, we assessed locomotor adaptation in PwMS. In a second investigation, this training was followed by cerebellar anodal tDCS applied immediately after the task ipsilateral to the fast leg (T0). The SBT paradigm was repeated 24 h (T1) and 78 h (T2) post-stimulation to evaluate consolidation.
Results: The gait dynamics and adaptation on the SBT were comparable between PwMS and controls. We found no effects of offline cerebellar anodal tDCS on locomotor adaptation and consolidation. Participants who received the active stimulation showed the same retention index than sham-stimulated subjects at T1 (p = 0.33) and T2 (p = 0.46).
Conclusion: Locomotor adaptation is preserved in people with mild-to-moderate MS. However, cerebellar anodal tDCS applied immediately post-training does not further enhance this ability. Future studies should define the neurobiological substrates of maintained plasticity in PwMS and how these substrates can be manipulated to improve compensation. Systematic assessments of methodological variables for cerebellar tDCS are urgently needed to increase the consistency and replicability of the results across experiments in various settings.
Cooperative Breeding in the Ambrosia Beetle Xyleborus affinis and Management of Its Fungal Symbionts

Fungus-farming is known from attine ants, macrotermites, and ambrosia beetles (Scolytinae, Platypodinae). Farming ant and termite societies are superorganismal and grow fungal cultivars in monocultures. Social organization of ambrosia beetle groups and their farming systems are poorly studied, because of their enigmatic life within tunnel systems inside of wood. Ambrosia beetle-fungus symbioses evolved many times independently in both the beetles and their fungal cultivars. Observations suggest that there is evolutionary convergence between these lineages, but also a high variation in the degree of sociality and the modes of fungiculture. Using a laboratory observation technique, I here tried to give insights into the social system and fungus symbiosis of the sugar-cane borer, Xyleborus affinis Eichhoff (Scolytinae: Curculionidae), a currently poorly studied ambrosia beetle. The study revealed a cooperatively breeding system characterized by delayed dispersal of adult daughters, alloparental brood care by larvae and adults, and about half of the totipotent adult daughters laying eggs within the natal nest. Most interesting, there was a tendency of egg-laying females to engage more commonly in mutually beneficial behaviors than non-egg-layers. Fungus gardens covering gallery walls composed of five different filamentous fungi. A Raffaelea isolate was predominant and together with an unidentified fungus likely served as the main food for adults and larvae. Three isolates, a Mucor, a Fusarium and a Phaeoacremonium isolate were most abundant in the oldest gallery part close to the entrance; Mucor, Fusarium and the Raffaelea isolate in diseased individuals. Additionally, there was correlative evidence for some fungal isoaltes influencing beetle feeding and hygienic behaviors. Overall, X. affinis is now the second ambrosia beetle that can be classified as a cooperative breeder with division of labor among and between adults and larvae.
Objectives
The severity of Coronavirus Disease 2019 (COVID-19) is largely determined by the immune response. First studies indicate altered lymphocyte counts and function. However, interactions of pro- and anti-inflammatory mechanisms remain elusive. In the current study we characterized the immune responses in patients suffering from severe COVID-19-induced acute respiratory distress syndrome (ARDS).
Methods
This was a single-center retrospective study in patients admitted to the intensive care unit (ICU) with confirmed COVID-19 between March 14th and May 28th 2020 (n = 39). Longitudinal data were collected within routine clinical care, including flow-cytometry of lymphocyte subsets, cytokine analysis and growth differentiation factor 15 (GDF-15). Antibody responses against the receptor binding domain (RBD) of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) Spike protein were analyzed.
Results
All patients suffered from severe ARDS, 30.8% died. Interleukin (IL)-6 was massively elevated at every time-point. The anti-inflammatory cytokine IL-10 was concomitantly upregulated with IL-6. The cellular response was characterized by lymphocytopenia with low counts of CD8+ T cells, natural killer (NK) and naïve T helper cells. CD8+ T and NK cells recovered after 8 to 14 days. The B cell system was largely unimpeded. This coincided with a slight increase in anti-SARS-CoV-2-Spike-RBD immunoglobulin (Ig) G and a decrease in anti-SARS-CoV-2-Spike-RBD IgM. GDF-15 levels were elevated throughout ICU treatment.
Conclusions
Massively elevated levels of IL-6 and a delayed cytotoxic immune defense characterized severe COVID-19-induced ARDS. The B cell response and antibody production were largely unimpeded. No obvious imbalance of pro- and anti-inflammatory mechanisms was observed, with elevated GDF-15 levels suggesting increased tissue resilience.
Marine natural products have achieved great success as an important source of new lead compounds for drug discovery. The Red Sea provides enormous diversity on the biological scale in all domains of life including micro- and macro-organisms. In this review, which covers the literature to the end of 2019, we summarize the diversity of bioactive secondary metabolites derived from Red Sea micro- and macro-organisms, and discuss their biological potential whenever applicable. Moreover, the diversity of the Red Sea organisms is highlighted as well as their genomic potential. This review is a comprehensive study that compares the natural products recovered from the Red Sea in terms of ecological role and pharmacological activities.
Mycotoxins in agriculturally used plants can cause intoxication in animals and can lead to severe financial losses for farmers. The endophytic fungus Epichloë festucae var. lolii living symbiotically within the cool season grass species Lolium perenne can produce vertebrate and invertebrate toxic alkaloids. Hence, an exact quantitation of alkaloid concentrations is essential to determine intoxication risk for animals. Many studies use different methods to detect alkaloid concentrations, which complicates the comparability. In this study, we showed that alkaloid concentrations of individual plants exceeded toxicity thresholds on real world grasslands in Germany, but not on the population level. Alkaloid concentrations on five German grasslands with high alkaloid levels peaked in summer but were also below toxicity thresholds on population level. Furthermore, we showed that alkaloid concentrations follow the same seasonal trend, regardless of whether plant fresh or dry weight was used, in the field and in a common garden study. However, alkaloid concentrations were around three times higher when detected with dry weight. Finally, we showed that alkaloid concentrations can additionally be biased to different alkaloid detection methods. We highlight that toxicity risks should be analyzed using plant dry weight, but concentration trends of fresh weight are reliable.
Revision Arthroplasty Through the Direct Anterior Approach Using an Asymmetric Acetabular Component

Despite increasing numbers of primary hip arthroplasties performed through the direct anterior approach (DAA), there is a lack of literature on DAA revision arthroplasty. The present study was performed in order to evaluate outcomes and revision rates after revision through the DAA using an asymmetric acetabular component with optional intra- and extramedullary fixation. In a retrospective cohort study, we analyzed prospectively collected data of 57 patients (61 hips, 43 female, 18 male) who underwent aseptic acetabular component revision through the DAA with the abovementioned implant system between January 2015 and December 2017. The mean follow-up was 40 months (12–56). Survival rates were estimated using the Kaplan–Meier method. All complications were documented and functional outcomes were assessed pre- and postoperatively. Kaplan–Meier analysis revealed an estimated five-year implant survival of 97% (confidence interval CI 87–99%). The estimated five-year survival with revision for any cause was 93% (CI 83–98%). The overall revision rate was 6.6% (n = 4). Two patients had to undergo revision due to periprosthetic infection (3.3%). In one patient, the acetabular component was revised due to aseptic loosening four months postoperatively. Another patient suffered from postoperative iliopsoas impingement and was treated successfully by arthroscopic iliopsoas tenotomy. Two (3.3%) of the revised hips dislocated postoperatively. The mean Harris Hip Score improved from 35 (2–66) preoperatively to 86 (38–100) postoperatively (p < 0.001). The hip joint’s anatomical center of rotation was restored at a high degree of accuracy. Our findings demonstrate that acetabular revision arthroplasty through the DAA using an asymmetric acetabular component with optional intra- and extramedullary fixation is safe and practicable, resulting in good radiographic and clinical midterm results.
The clinical picture of depressive disorders is characterized by a plethora of somatic symptoms, psychomotor retardation, and, particularly, anhedonia. The number of patients with residual symptoms or treatment resistance is high. Touch is the basic communication among humans and animals. Its application professionally in the form of, e.g., psychoactive massage therapy, has been shown in the past to reduce the somatic and mental symptoms of depression and anxiety. Here, we investigated the effects of a specially developed affect-regulating massage therapy (ARMT) vs. individual treatment with a standardized relaxation procedure, progressive muscle relaxation (PMR), in 57 outpatients with depression. Patients were given one ARMT or PMR session weekly over 4 weeks. Changes in somatic and cognitive symptoms were assessed by standard psychiatric instruments (Hamilton Depression Scale (HAMD) and the Bech–Rafaelsen–Melancholia–Scale (BRMS)) as well as a visual analogue scale. Furthermore, oral statements from all participants were obtained in semi-structured interviews. The findings show clear and statistically significant superiority of ARMT over PMR. The results might be interpreted within various models. The concept of interoception, as well as the principles of body psychotherapy and phenomenological aspects, offers cues for understanding the mechanisms involved. Within a neurobiological context, the significance of C-tactile afferents activated by special touch techniques and humoral changes such as increased oxytocin levels open additional ways of interpreting our findings.
Neural networks have to capture mathematical relationships in order to learn various tasks. They approximate these relations implicitly and therefore often do not generalize well. The recently proposed Neural Arithmetic Logic Unit (NALU) is a novel neural architecture which is able to explicitly represent the mathematical relationships by the units of the network to learn operations such as summation, subtraction or multiplication. Although NALUs have been shown to perform well on various downstream tasks, an in-depth analysis reveals practical shortcomings by design, such as the inability to multiply or divide negative input values or training stability issues for deeper networks. We address these issues and propose an improved model architecture. We evaluate our model empirically in various settings from learning basic arithmetic operations to more complex functions. Our experiments indicate that our model solves stability issues and outperforms the original NALU model in means of arithmetic precision and convergence.
Delayed natural killer (NK) cell reconstitution after allogeneic stem cell transplantation (alloSCT) is associated with a higher risk of developing invasive aspergillosis. The interaction of NK cells with the human pathogen Aspergillus (A.) fumigatus is mediated by the fungal recognition receptor CD56, which is relocated to the fungal interface after contact. Blocking of CD56 signaling inhibits the fungal mediated chemokine secretion of MIP-1α, MIP-1β, and RANTES and reduces cell activation, indicating a functional role of CD56 in fungal recognition. We collected peripheral blood from recipients of an allograft at defined time points after alloSCT (day 60, 90, 120, 180). NK cells were isolated, directly challenged with live A. fumigatus germ tubes, and cell function was analyzed and compared to healthy age and gender-matched individuals. After alloSCT, NK cells displayed a higher percentage of CD56\(^{bright}\)CD16\(^{dim}\) cells throughout the time of blood collection. However, CD56 binding and relocalization to the fungal contact side were decreased. We were able to correlate this deficiency to the administration of corticosteroid therapy that further negatively influenced the secretion of MIP-1α, MIP-1β, and RANTES. As a consequence, the treatment of healthy NK cells ex vivo with corticosteroids abrogated chemokine secretion measured by multiplex immunoassay. Furthermore, we analyzed NK cells regarding their actin cytoskeleton by Structured Illumination Microscopy (SIM) and flow cytometry and demonstrate an actin dysfunction of NK cells shown by reduced F-actin content after fungal co-cultivation early after alloSCT. This dysfunction remains until 180 days post-alloSCT, concluding that further actin-dependent cellular processes may be negatively influenced after alloSCT. To investigate the molecular pathomechansism, we compared CD56 receptor mobility on the plasma membrane of healthy and alloSCT primary NK cells by single-molecule tracking. The results were very robust and reproducible between tested conditions which point to a different molecular mechanism and emphasize the importance of proper CD56 mobility.
Memory B cells have known to play an important role in the pathogenesis of rheumatoid arthritis (RA). With the emergence of B cell-targeted therapies, the modulation of memory B cells appears to be a key therapeutic target. Human peripheral memory B cells can be distinguished based on the phenotypic expression of CD27 and IgD, characterizing the three major B cell subpopulations: CD27+IgD+ pre-switch, CD27+IgD- post-switch, and CD27-IgD- double-negative memory B cells. We evaluated different memory cell populations for activation markers (CD95 and Ki-67) and chemokine receptors (CXCR3 and 4) expressing B cells in active RA, as well as under IL6-R blockade by tocilizumab (TCZ) and TNF-α blockade by adalimumab (ADA). Memory B cells were phenotypically analyzed from RA patients at baseline, week 12, and week 24 under TCZ or ADA treatment, respectively. Using flow cytometry, surface expression of CD95, intracellular Ki-67, and surface expressions of CXCR3 and CXCR4 were determined. Compared with healthy donors (n = 40), the phenotypic analysis of RA patients (n = 80) demonstrated that all three types of memory B cells were activated in RA patients. Surface and intracellular staining of B cells showed a significantly higher percentage of CD95+ (p < 0.0001) and Ki-67+ (p < 0.0001) cells, with numerically altered CXCR3+ and CXCR4+ cells in RA. CD95 and Ki-67 expressions were highest in post-switch memory B cells, whereas CD19+CXCR3+ and CD19+CXCR4+ expressing cells were substantially higher in the pre-switch compartment. In all subsets of the memory B cells, in vivo IL-6R, and TNF-α blockade significantly reduced the enhanced expressions of CD95 and Ki-67. Based on our findings, we conclude that the three major peripheral memory B cell populations, pre-, post-switch, and double-negative B cells, are activated in RA, demonstrating enhanced CD95 and Ki-67 expressions, and varied expression of CXCR3 and CXCR4 chemokine receptors when compared with healthy individuals. This activation can be efficaciously modulated under cytokine inhibition in vivo.
Regulatory T cells (Tregs) are important for the ontogenetic control of immune activation and tissue damage in preterm infants. However, the role of Tregs for the development of bronchopulmonary dysplasia (BPD) is yet unclear. The aim of our study was to characterize CD4+ CD25+ forkhead box protein 3 (FoxP3)+ Tregs in peripheral blood of well-phenotyped preterm infants (n = 382; 23 + 0 – 36 + 6 weeks of gestational age) with a focus on the first 28 days of life and the clinical endpoint BPD (supplemental oxygen for longer than 28 days of age). In a subgroup of preterm infants, we characterized the immunological phenotype of Tregs (n = 23). The suppressive function of Tregs on CD4+CD25- T cells was compared in preterm, term and adult blood. We observed that extreme prematurity was associated with increased Treg frequencies which peaked in the second week of life. Independent of gestational age, increased Treg frequencies were noted to precede the development of BPD. The phenotype of preterm infant Tregs largely differed from adult Tregs and displayed an overall naïve Treg population (CD45RA+/HLA-DR-/Helios+), especially in the first days of life. On day 7 of life, a more activated Treg phenotype pattern (CCR6+, HLA-DR+, and Ki-67+) was observed. Tregs of preterm neonates had a higher immunosuppressive capacity against CD4+CD25- T cells compared to the Treg compartment of term neonates and adults. In conclusion, our data suggest increased frequencies and functions of Tregs in preterm neonates which display a distinct phenotype with dynamic changes in the first weeks of life. Hence, the continued abundance of Tregs may contribute to sustained inflammation preceding the development of BPD. Functional analyses are needed in order to elucidate whether Tregs have potential as future target for diagnostics and therapeutics.
Exercise and training prescription in endurance-type sports has a strong theoretical background with various practical applications based on threshold concepts. Given the challenges and pitfalls of determining individual training zones on the basis of subsystem indicators (e.g., blood lactate concentration, respiratory parameters), the question arises whether there are alternatives for intensity distribution demarcation. Considering that training in a low intensity zone substantially contributes to the performance outcome of endurance athletes and exceeding intensity targets based on a misleading aerobic threshold can lead to negative performance and recovery effects, it would be desirable to find a parameter that could be derived via non-invasive, low cost and commonly available wearable devices. In this regard, analytics conducted from non-linear dynamics of heart rate variability (HRV) have been adapted to gain further insights into the complex cardiovascular regulation during endurance-type exercise. Considering the reciprocal antagonistic behavior and the interaction of the sympathetic and parasympathetic branch of the autonomic nervous system from low to high exercise intensities, it may be promising to use an approach that utilizes information about the regulation quality of the organismic system to determine training-intensity distribution. Detrended fluctuation analysis of HRV and its short-term scaling exponent alpha1 (DFA-alpha1) seems suitable for applied sport-specific settings including exercise from low to high intensities. DFA-alpha1 may be taken as an indicator for exercise prescription and intensity distribution monitoring in endurance-type sports. The present perspective illustrates the potential of DFA-alpha1 for diagnostic and monitoring purposes as a “global” system parameter and proxy for organismic demands.
Catalytic water splitting is a viable process for the generation of renewable fuels. Here it is reported for the first time that a trinuclear supramolecular Ru(bda) (bda: 2,2′‐bipyridine‐6,6′‐dicarboxylate) catalyst, anchored on multi‐walled carbon nanotubes and subsequently immobilized on glassy carbon electrodes, shows outstanding performance in heterogeneous water oxidation. Activation of the catalyst on anodes by repetitive cyclic voltammetry (CV) scans results in a catalytic current density of 186 mA cm\(^{−2}\) at a potential of 1.45 V versus NHE. The activated catalyst performs water oxidation at an onset overpotential of 330 mV. The remarkably high stability of the hybrid anode is demonstrated by X‐ray absorption spectroscopy and electrochemically, revealing the absence of any degradation after 1.8 million turnovers. Foot of the wave analysis of CV data of activated electrodes with different concentrations of catalyst indicates a monomolecular water nucleophilic attack mechanism with an apparent rate constant of TOFmax (turnover frequency) of 3200 s\(^{−1}\).
In Brassicaceae, tissue damage triggers the mustard oil bomb i.e., activates the degradation of glucosinolates by myrosinases leading to a rapid accumulation of isothiocyanates at the site of damage. Isothiocyanates are reactive electrophilic species (RES) known to covalently bind to thiols in proteins and glutathione, a process that is not only toxic to herbivores and microbes but can also cause cell death of healthy plant tissues. Previously, it has been shown that subtoxic isothiocyanate concentrations can induce transcriptional reprogramming in intact plant cells. Glutathione depletion by RES leading to breakdown of the redox potential has been proposed as a central and common RES signal transduction mechanism. Using transcriptome analyses, we show that after exposure of Arabidopsis seedlings (grown in liquid culture) to subtoxic concentrations of sulforaphane hundreds of genes were regulated without depletion of the cellular glutathione pool. Heat shock genes were among the most highly up-regulated genes and this response was found to be dependent on the canonical heat shock factors A1 (HSFA1). HSFA1-deficient plants were more sensitive to isothiocyanates than wild type plants. Moreover, pretreatment of Arabidopsis seedlings with subtoxic concentrations of isothiocyanates increased resistance against exposure to toxic levels of isothiocyanates and, hence, may reduce the autotoxicity of the mustard oil bomb by inducing cell protection mechanisms.
Acceptance-based regulation of pain, which focuses on the allowing of pain and pain related thoughts and emotions, was found to modulate pain. However, results so far are inconsistent regarding different pain modalities and indices. Moreover, studies so far often lack a suitable control condition, focus on behavioral pain measures rather than physiological correlates, and often use between-subject designs, which potentially impede the evaluation of the effectiveness of the strategies. Therefore, we investigated whether acceptance-based strategies can reduce subjective and physiological markers of acute pain in comparison to a control condition in a within-subject design. To this end, participants (N = 30) completed 24 trials comprising 10 s of heat pain stimulation. Each trial started with a cue instructing participants to welcome and experience pain (acceptance trials) or to react to the pain as it is without employing any regulation strategies (control trials). In addition to pain intensity and unpleasantness ratings, heart rate (HR) and skin conductance (SC) were recorded. Results showed significantly decreased pain intensity and unpleasantness ratings for acceptance compared to control trials. Additionally, HR was significantly lower during acceptance compared to control trials, whereas SC revealed no significant differences. These results demonstrate the effectiveness of acceptance-based strategies in reducing subjective and physiological pain responses relative to a control condition, even after short training. Therefore, the systematic investigation of acceptance in different pain modalities in healthy and chronic pain patients is warranted.
Inflammation and dysregulation of the immune system are hallmarks of several neurodegenerative diseases. An activated immune response is considered to be the cause of myelin breakdown in demyelinating disorders. In the peripheral nervous system (PNS), myelin can be degraded in an autophagy-dependent manner directly by Schwann cells or by macrophages, which are modulated by T-lymphocytes. Here, we show that the NF-κB activator Pleckstrin homology containing family member 5 (Plekhg5) is involved in the regulation of both Schwann cell autophagy and recruitment of T-lymphocytes in peripheral nerves during motoneuron disease. Plekhg5-deficient mice show defective axon/Schwann cell units characterized by myelin infoldings in peripheral nerves. Even at late stages, Plekhg5-deficient mice do not show any signs of demyelination and inflammation. Using RNAseq, we identified a transcriptional signature for an impaired immune response in sciatic nerves, which manifested in a reduced number of CD4\(^+\) and CD8\(^+\) T-cells. These findings identify Plekhg5 as a promising target to impede myelin breakdown in demyelinating PNS disorders.
Published studies on the risk of radiation-induced second primary malignancy (SPM) after radioiodine treatment (RAI) of differentiated thyroid cancer (DTC) refer mainly to patients treated as middle-aged or older adults and are not easily generalizable to those treated at a younger age. Here we review available literature on the risk of breast cancer as an SPM after RAI of DTC with a focus on females undergoing such treatment in childhood, adolescence, or young adulthood. Additionally, we report the results of a preliminary international survey of patient registries from academic tertiary referral centers specializing in pediatric DTC. The survey sought to evaluate the availability of sufficient patient data for a potential international multicenter observational case–control study of females with DTC given RAI at an early age. Our literature review identified a bi-directional association of DTC and breast cancer. The general breast cancer risk in adult DTC survivors is low, ~2%, slightly higher in females than in males, but presumably lower, not higher, in those diagnosed as children or adolescents than in those diagnosed at older ages. RAI presumably does not substantially influence breast cancer risk after DTC. However, data from patients given RAI at young ages are sparse and insufficient to make definitive conclusions regarding age dependence of the risk of breast cancer as a SPM after RAI of DTC. The preliminary analysis of data from 10 thyroid cancer registries worldwide, including altogether 6,449 patients given RAI for DTC and 1,116 controls, i.e., patients not given RAI, did not show a significant increase of breast cancer incidence after RAI. However, the numbers of cases and controls were insufficient to draw statistically reliable conclusions, and the proportion of those receiving RAI at the earliest ages was too low.In conclusion, a potential international multicenter study of female patients undergoing RAI of DTC as children, adolescents, or young adults, with a sufficient sample size, is feasible. However, breast cancer screening of a larger cohort of DTC patients is not unproblematic for ethical reasons, due to the likely, at most slightly, increased risk of breast cancer post-RAI and the expected ~10% false-positivity rate which potentially produced substantial “misdiagnosis.”
Tactile stimulation is less frequently used than visual for brain-computer interface (BCI) control, partly because of limitations in speed and accuracy. Non-visual BCI paradigms, however, may be required for patients who struggle with vision dependent BCIs because of a loss of gaze control. With the present study, we attempted to replicate earlier results by Herweg et al. (2016), with several minor adjustments and a focus on training effects and usability. We invited 16 healthy participants and trained them with a 4-class tactile P300-based BCI in five sessions. Their main task was to navigate a virtual wheelchair through a 3D apartment using the BCI. We found significant training effects on information transfer rate (ITR), which increased from a mean of 3.10–9.50 bits/min. Further, both online and offline accuracies significantly increased with training from 65% to 86% and 70% to 95%, respectively. We found only a descriptive increase of P300 amplitudes at Fz and Cz with training. Furthermore, we report subjective data from questionnaires, which indicated a relatively high workload and moderate to high satisfaction. Although our participants have not achieved the same high performance as in the Herweg et al. (2016) study, we provide evidence for training effects on performance with a tactile BCI and confirm the feasibility of the paradigm.
Using Expansion Microscopy to Visualize and Characterize the Morphology of Mitochondrial Cristae

Mitochondria are double membrane bound organelles indispensable for biological processes such as apoptosis, cell signaling, and the production of many important metabolites, which includes ATP that is generated during the process known as oxidative phosphorylation (OXPHOS). The inner membrane contains folds called cristae, which increase the membrane surface and thus the amount of membrane-bound proteins necessary for the OXPHOS. These folds have been of great interest not only because of their importance for energy conversion, but also because changes in morphology have been linked to a broad range of diseases from cancer, diabetes, neurodegenerative diseases, to aging and infection. With a distance between opposing cristae membranes often below 100 nm, conventional fluorescence imaging cannot provide a resolution sufficient for resolving these structures. For this reason, various highly specialized super-resolution methods including dSTORM, PALM, STED, and SIM have been applied for cristae visualization. Expansion Microscopy (ExM) offers the possibility to perform super-resolution microscopy on conventional confocal microscopes by embedding the sample into a swellable hydrogel that is isotropically expanded by a factor of 4–4.5, improving the resolution to 60–70 nm on conventional confocal microscopes, which can be further increased to ∼ 30 nm laterally using SIM. Here, we demonstrate that the expression of the mitochondrial creatine kinase MtCK linked to marker protein GFP (MtCK-GFP), which localizes to the space between the outer and the inner mitochondrial membrane, can be used as a cristae marker. Applying ExM on mitochondria labeled with this construct enables visualization of morphological changes of cristae and localization studies of mitochondrial proteins relative to cristae without the need for specialized setups. For the first time we present the combination of specific mitochondrial intermembrane space labeling and ExM as a tool for studying internal structure of mitochondria.
The lipids phosphatidylserine (PtdSer) and phosphatidylethanolamine (PtdEth) are normally asymmetrically localized to the cytosolic face of membrane bilayers, but can both be externalized during diverse biological processes, including cell division, cell fusion, and cell death. Externalized lipids in the plasma membrane are recognized by lipid-binding proteins to regulate the clearance of cell corpses and other cell debris. However, it is unclear whether PtdSer and PtdEth contribute in similar or distinct ways to these processes. We discovered that disruption of the lipid flippases that maintain PtdSer or PtdEth asymmetry in the plasma membrane have opposite effects on phagocytosis in Caenorhabditis elegans embryos. Constitutive PtdSer externalization caused by disruption of the major PtdSer flippase TAT-1 led to increased phagocytosis of cell debris, sometimes leading to two cells engulfing the same debris. In contrast, PtdEth externalization caused by depletion of the major PtdEth flippase TAT-5 or its activator PAD-1 disrupted phagocytosis. These data suggest that PtdSer and PtdEth externalization have opposite effects on phagocytosis. Furthermore, externalizing PtdEth is associated with increased extracellular vesicle release, and we present evidence that the extent of extracellular vesicle accumulation correlates with the extent of phagocytic defects. Thus, a general loss of lipid asymmetry can have opposing impacts through different lipid subtypes simultaneously exerting disparate effects.
The internal structures of a moraine complex mostly provide information about the manner in which they develop and thus they can transmit details about several processes long after they have taken place. While the occurrence of glacier–permafrost interactions during the formation of large thrust moraine complexes at polar and subpolar glaciers as well as at marginal positions of former ice sheets has been well understood, their role in the formation of moraines on comparatively small alpine glaciers is still very poorly investigated. Therefore, the question arises as to whether evidence of former glacier–permafrost interactions can still be found in glacier forefields of small alpine glaciers and to what extent these differ from the processes in finer materials at larger polar or subpolar glaciers. To investigate this, electrical resistivity tomography (ERT) and ground-penetrating radar (GPR) surveys were carried out in the area of a presumed alpine thrust moraine complex in order to investigate internal moraine structures. The ERT data confirmed the presence of a massive ice core within the central and proximal parts of the moraine complex. Using GPR, linear internal structures were detected, which were interpreted as internal shear planes due to their extent and orientation. These shear planes lead to the assumption that the moraine complex is of glaciotectonic origin. Based on the detected internal structures and the high electrical resistivity values, it must also be assumed that the massive ice core is of sedimentary or polygenetic origin. The combined approach of the two methods enabled the authors of this study to detect different internal structures and to deduce a conceptual model of the thrust moraine formation.
(1) Background: Prostate-specific membrane antigen (PSMA)-derived tumour volume (PSMA-TV) and total lesion PSMA (TL-PSMA) from PSMA PET/CT scans are promising biomarkers for assessing treatment response in prostate cancer (PCa). Currently, it is unclear whether different software tools for assessing PSMA-TV and TL-PSMA produce comparable results. (2) Methods: \(^{68}\)Ga-PSMA PET/CT scans from n = 21 patients with castration-resistant PCa (CRPC) receiving chemotherapy were identified from our single-centre database. PSMA-TV and TL-PSMA were calculated with Syngo.via (Siemens) as well as the freely available Beth Israel plugin for FIJI (Fiji Is Just ImageJ) before and after chemotherapy. While statistical comparability was illustrated and quantified via Bland-Altman diagrams, the clinical agreement was estimated by matching PSMA-TV, TL-PSMA and relative changes of both variables during chemotherapy with changes in serum PSA (ΔPSA) and PERCIST (Positron Emission Response Criteria in Solid Tumors). (3) Results: Comparing absolute PSMA-TV and TL-PSMA as well as Bland–Altman plotting revealed a good statistical comparability of both software algorithms. For clinical agreement, classifying therapy response did not differ between PSMA-TV and TL-PSMA for both software solutions and showed highly positive correlations with BR. (4) Conclusions: due to the high levels of statistical and clinical agreement in our CRPC patient cohort undergoing taxane chemotherapy, comparing PSMA-TV and TL-PSMA determined by Syngo.via and FIJI appears feasible.
The rating of perceived exertion (RPE) is a subjective load marker and may assist in individualizing training prescription, particularly by adjusting running intensity. Unfortunately, RPE has shortcomings (e.g., underreporting) and cannot be monitored continuously and automatically throughout a training sessions. In this pilot study, we aimed to predict two classes of RPE (≤15 “Somewhat hard to hard” on Borg’s 6–20 scale vs. RPE >15 in runners by analyzing data recorded by a commercially-available smartwatch with machine learning algorithms. Twelve trained and untrained runners performed long-continuous runs at a constant self-selected pace to volitional exhaustion. Untrained runners reported their RPE each kilometer, whereas trained runners reported every five kilometers. The kinetics of heart rate, step cadence, and running velocity were recorded continuously ( 1 Hz ) with a commercially-available smartwatch (Polar V800). We trained different machine learning algorithms to estimate the two classes of RPE based on the time series sensor data derived from the smartwatch. Predictions were analyzed in different settings: accuracy overall and per runner type; i.e., accuracy for trained and untrained runners independently. We achieved top accuracies of 84.8 % for the whole dataset, 81.8 % for the trained runners, and 86.1 % for the untrained runners. We predict two classes of RPE with high accuracy using machine learning and smartwatch data. This approach might aid in individualizing training prescriptions.
Schizophrenia (SCZ) is a severe mental disorder with immense personal and societal costs; identifying individuals at risk is therefore of utmost importance. Genomic risk profile scores (GRPS) have been shown to significantly predict cases-control status. Making use of a large-population based sample from Sweden, we replicate a previous finding demonstrating that the GRPS is strongly associated with admission frequency and chronicity of SCZ. Furthermore, we were able to show a substantial gap in prediction accuracy between males and females. In sum, our results indicate that prediction accuracy by GRPS depends on clinical and demographic characteristics.
Azole resistance of the fungal pathogen Aspergillus fumigatus is an emerging problem. To identify novel mechanisms that could mediate azole resistance in A. fumigatus, we analyzed the transcriptome of a mitochondrial fission/fusion mutant that exhibits increased azole tolerance. Approximately 12% of the annotated genes are differentially regulated in this strain. This comprises upregulation of Cyp51A, the azole target structure, upregulation of ATP-binding cassette (ABC) superfamily and major facilitator superfamily (MFS) transporters and differential regulation of transcription factors. To study their impact on azole tolerance, conditional mutants were constructed of seven ABC transporters and 17 transcription factors. Under repressed conditions, growth rates and azole susceptibility of the mutants were similar to wild type. Under induced conditions, several transcription factor mutants showed growth phenotypes. In addition, four ABC transporter mutants and seven transcription factor mutants exhibited altered azole susceptibility. However, deletion of individual identified ABC transporters and transcription factors did not affect the increased azole tolerance of the fission/fusion mutant. Our results revealed the ability of multiple ABC transporters and transcription factors to modulate the azole susceptibility of A. fumigatus and support a model where mitochondrial dysfunctions trigger a drug resistance network that mediates azole tolerance of this mold.
Objective: We investigated cerebral opioid receptor binding potential in patients with fibromyalgia syndrome (FMS) using positron-emission-tomography (PET) and correlated our results with patients’ systemic interleukin-4 (IL-4) gene expression.
Methods: In this pilot study, seven FMS patients (1 man, 6 women) agreed to participate in experimental PET scans. All patients underwent neurological examination, were investigated with questionnaires for pain, depression, and FMS symptoms. Additionally, blood for IL-4 gene expression analysis was withdrawn at two time points with a median latency of 1.3 years. Patients were investigated in a PET scanner using the opioid receptor ligand F-18-fluoro-ethyl-diprenorphine ([18F]FEDPN) and results were compared with laboratory normative values.
Results: Neurological examination was normal in all FMS patients. Reduced opioid receptor binding was found in mid cingulate cortex compared to healthy controls (p < 0.005). Interestingly, three patients with high systemic IL-4 gene expression had increased opioid receptor binding in the fronto-basal cortex compared to those with low IL-4 gene expression (p < 0.005).
Conclusion: Our data give further evidence for a reduction in cortical opioid receptor availability in FMS patients as another potential central nervous system contributor to pain in FMS.
Obligate human pathogenic Neisseria gonorrhoeae are the second most frequent bacterial cause of sexually transmitted diseases. These bacteria invade different mucosal tissues and occasionally disseminate into the bloodstream. Invasion into epithelial cells requires the activation of host cell receptors by the formation of ceramide-rich platforms. Here, we investigated the role of sphingosine in the invasion and intracellular survival of gonococci. Sphingosine exhibited an anti-gonococcal activity in vitro. We used specific sphingosine analogs and click chemistry to visualize sphingosine in infected cells. Sphingosine localized to the membrane of intracellular gonococci. Inhibitor studies and the application of a sphingosine derivative indicated that increased sphingosine levels reduced the intracellular survival of gonococci. We demonstrate here, that sphingosine can target intracellular bacteria and may therefore exert a direct bactericidal effect inside cells.
Despite its significant overexpression in several malignant neoplasms, the expression of RPS27 in the central nervous system (CNS) is widely unknown. We identified the cell types expressing RPS27 in the CNS under normal and disease conditions. We acquired specimens of healthy brain (NB), adult pilocytic astrocytoma (PA) World Health Organization (WHO) grade I, anaplastic PA WHO grade III, gliomas WHO grade II/III with or without isocitrate dehydrogenase (IDH) mutation, and glioblastoma multiforme (GBM). RPS27 protein expression was examined by immunohistochemistry and double-fluorescence staining and its mRNA expression quantified by RT-PCR. Patients’ clinical and tumor characteristics were collected retrospectively. RPS27 protein was specifically expressed in tumor cells and neurons, but not in healthy astrocytes. In tumor tissue, most macrophages were positive, while this was rarely the case in inflamed tissue. Compared to NB, RPS27 mRNA was in mean 6.2- and 8.8-fold enhanced in gliomas WHO grade II/III with (p < 0.01) and without IDH mutation (p = 0.01), respectively. GBM displayed a 4.6-fold increased mean expression (p = 0.02). Although RPS27 expression levels did not affect the patients’ survival, their association with tumor cells and tumor-associated macrophages provides a rationale for a future investigation of a potential function during gliomagenesis and tumor immune response.
Downregulation of miR-221-3p expression in prostate cancer (PCa) predicted overall and cancer-specific survival of high-risk PCa patients. Apart from PCa, miR-221-3p expression levels predicted a response to tyrosine kinase inhibitors (TKI) in clear cell renal cell carcinoma (ccRCC) patients. Since this role of miR-221-3p was explained with a specific targeting of VEGFR2, we examined whether miR-221-3p regulated VEGFR2 in PCa. First, we confirmed VEGFR2/KDR as a target gene of miR-221-3p in PCa cells by applying Luciferase reporter assays and Western blotting experiments. Although VEGFR2 was mainly downregulated in the PCa cohort of the TCGA (The Cancer Genome Atlas) database, VEGFR2 was upregulated in our high-risk PCa cohort (n = 142) and predicted clinical progression. In vitro miR-221-3p acted as an escape mechanism from TKI in PC3 cells, as displayed by proliferation and apoptosis assays. Moreover, we confirmed that Sunitinib induced an interferon-related gene signature in PC3 cells by analyzing external microarray data and by demonstrating a significant upregulation of miR-221-3p/miR-222-3p after Sunitinib exposure. Our findings bear a clinical perspective for high-risk PCa patients with low miR-221-3p levels since this could predict a favorable TKI response. Apart from this therapeutic niche, we identified a partially oncogenic function of miR-221-3p as an escape mechanism from VEGFR2 inhibition.
Implants elicit an immunological response after implantation that results in the worst case in a complete implant rejection. This biomaterial-induced inflammation is modulated by macrophages and can be influenced by nanotopographical surface structures such as titania nanotubes or fractal titanium nitride (TiN) surfaces. However, their specific impact on a distinct macrophage phenotype has not been identified. By using two different levels of nanostructures and smooth samples as controls, the influence of tubular TiO2 and fractal TiN nanostructures on primary human macrophages with M1 or M2-phenotype was investigated. Therefore, nanotopographical coatings were either, directly generated by physical vapor deposition (PVD) or by electrochemical anodization of titanium PVD coatings. The cellular response of macrophages was quantitatively assessed to demonstrate a difference in biocompatibility of nanotubes in respect to human M1 and M2-macrophages. Depending on the tube diameter of the nanotubular surfaces, low cell numbers and impaired cellular activity, was detected for M2-macrophages, whereas the impact of nanotubes on M1-polarized macrophages was negligible. Importantly, we could confirm this phenotypic response on the fractal TiN surfaces. The results indicate that the investigated topographies specifically impact the macrophage M2-subtype that modulates the formation of the fibrotic capsule and the long-term response to an implant.
Impact of Interparticle Interaction on Thermodynamics of Nano-Channel Transport of Two Species

Understanding the function and control of channel transport is of paramount importance for cell physiology and nanotechnology. In particular, if several species are involved, the mechanisms of selectivity, competition, cooperation, pumping, and its modulation need to be understood. What lacks is a rigorous mathematical approach within the framework of stochastic thermodynamics, which explains the impact of interparticle in-channel interactions on the transport properties of the respective species. To achieve this, stochastic channel transport of two species is considered in a model, which different from mean field approaches, explicitly conserves the spatial correlation of the species within the channel by analysis of the stochastic dynamics within a state space, the elements of which are the channel’s spatial occupation states. The interparticle interactions determine the stochastic transitions between these states. Local flow and entropy production in this state space reveal the respective particle flows through the channel and the intensity of the Brownian ratchet like rectifying forces, which these species exert mutually on each other, together with its thermodynamic effectiveness and costs. Perfect coupling of transport of the two species is realized by an attractive empty channel and strong repulsive forces between particles of the same species. This confines the state space to a subspace with circular topology, in which the concentration gradients as thermodynamic driving forces act in series, and channel flow of both species becomes equivalent. For opposing concentration gradients, this makes the species with the stronger gradient the driving, positive entropy producing one; the other is driven and produces negative entropy. Gradients equal in magnitude make all flows vanish, and thermodynamic equilibrium occurs. A differential interparticle interaction with less repulsive forces within particles of one species but maintenance of this interaction for the other species adds a bypass path to this circular subspace. On this path, which is not involved in coupling of the two species, a leak flow of the species with less repulsive interparticle interaction emerges, which is directed parallel to its concentration gradient and, hence, produces positive entropy here. Different from the situation with perfect coupling, appropriate strong opposing concentration gradients may simultaneously parallelize the flow of their respective species, which makes each species produce positive entropy. The rectifying potential of the species with the bypass option is diminished. This implies the existence of a gradient of the other species, above which its flow and gradient are parallel for any gradient of the less coupled species. The opposite holds for the less coupled species. Its flow may always be rectified and turned anti-parallel to its gradient by a sufficiently strong opposing gradient of the other one.
Super-resolution microscopy has evolved as a powerful method for subdiffraction-resolution fluorescence imaging of cells and cellular organelles, but requires sophisticated and expensive installations. Expansion microscopy (ExM), which is based on the physical expansion of the cellular structure of interest, provides a cheap alternative to bypass the diffraction limit and enable super-resolution imaging on a conventional fluorescence microscope. While ExM has shown impressive results for the magnified visualization of proteins and RNAs in cells and tissues, it has not yet been applied in fungi, mainly due to their complex cell wall. Here we developed a method that enables reliable isotropic expansion of ascomycetes and basidiomycetes upon treatment with cell wall degrading enzymes. Confocal laser scanning microscopy (CLSM) and structured illumination microscopy (SIM) images of 4.5-fold expanded sporidia of Ustilago maydis expressing fluorescent fungal rhodopsins and hyphae of Fusarium oxysporum or Aspergillus fumigatus expressing either histone H1-mCherry together with Lifeact-sGFP or mRFP targeted to mitochondria, revealed details of subcellular structures with an estimated spatial resolution of around 30 nm. ExM is thus well suited for cell biology studies in fungi on conventional fluorescence microscopes.
Data analytics as a field is currently at a crucial point in its development, as a commoditization takes place in the context of increasing amounts of data, more user diversity, and automated analysis solutions, the latter potentially eliminating the need for expert analysts. A central hypothesis of the present paper is that data visualizations should be adapted to both the user and the context. This idea was initially addressed in Study 1, which demonstrated substantial interindividual variability among a group of experts when freely choosing an option to visualize data sets. To lay the theoretical groundwork for a systematic, taxonomic approach, a user model combining user traits, states, strategies, and actions was proposed and further evaluated empirically in Studies 2 and 3. The results implied that for adapting to user traits, statistical expertise is a relevant dimension that should be considered. Additionally, for adapting to user states different user intentions such as monitoring and analysis should be accounted for. These results were used to develop a taxonomy which adapts visualization recommendations to these (and other) factors. A preliminary attempt to validate the taxonomy in Study 4 tested its visualization recommendations with a group of experts. While the corresponding results were somewhat ambiguous overall, some aspects nevertheless supported the claim that a user-adaptive data visualization approach based on the principles outlined in the taxonomy can indeed be useful. While the present approach to user adaptivity is still in its infancy and should be extended (e.g., by testing more participants), the general approach appears to be very promising.
Gait Initiation in Parkinson’s Disease: Impact of Dopamine Depletion and Initial Stance Condition

Postural instability, in particular at gait initiation (GI), and resulting falls are a major determinant of poor quality of life in subjects with Parkinson’s disease (PD). Still, the contribution of the basal ganglia and dopamine on the feedforward postural control associated with this motor task is poorly known. In addition, the influence of anthropometric measures (AM) and initial stance condition on GI has never been consistently assessed. The biomechanical resultants of anticipatory postural adjustments contributing to GI [imbalance (IMB), unloading (UNL), and stepping phase) were studied in 26 unmedicated subjects with idiopathic PD and in 27 healthy subjects. A subset of 13 patients was analyzed under standardized medication conditions and the striatal dopaminergic innervation was studied in 22 patients using FP-CIT and SPECT. People with PD showed a significant reduction in center of pressure (CoP) displacement and velocity during the IMB phase, reduced first step length and velocity, and decreased velocity and acceleration of the center of mass (CoM) at toe off of the stance foot. All these measurements correlated with the dopaminergic innervation of the putamen and substantially improved with levodopa. These results were not influenced by anthropometric parameters or by the initial stance condition. In contrast, most of the measurements of the UNL phase were influenced by the foot placement and did not correlate with putaminal dopaminergic innervation. Our results suggest a significant role of dopamine and the putamen particularly in the elaboration of the IMB phase of anticipatory postural adjustments and in the execution of the first step. The basal ganglia circuitry may contribute to defining the optimal referent body configuration for a proper initiation of gait and possibly gait adaptation to the environment.
Mushroom bodies (MBs) are multisensory integration centers in the insect brain involved in learning and memory formation. In the honeybee, the main sensory input region (calyx) of MBs is comparatively large and receives input from mainly olfactory and visual senses, but also from gustatory/tactile modalities. Behavioral plasticity following differential brood care, changes in sensory exposure or the formation of associative long-term memory (LTM) was shown to be associated with structural plasticity in synaptic microcircuits (microglomeruli) within olfactory and visual compartments of the MB calyx. In the same line, physiological studies have demonstrated that MB-calyx microcircuits change response properties after associative learning. The aim of this review is to provide an update and synthesis of recent research on the plasticity of microcircuits in the MB calyx of the honeybee, specifically looking at the synaptic connectivity between sensory projection neurons (PNs) and MB intrinsic neurons (Kenyon cells). We focus on the honeybee as a favorable experimental insect for studying neuronal mechanisms underlying complex social behavior, but also compare it with other insect species for certain aspects. This review concludes by highlighting open questions and promising routes for future research aimed at understanding the causal relationships between neuronal and behavioral plasticity in this charismatic social insect.
Acromyrmex fracticornis grass-cutting ants construct conspicuous chimney-shaped nest turrets made of intermeshed grass fragments. We asked whether turrets are constructed by merely piling up nearby materials around the entrance, or whether ants incorporate different materials as the turret develops. By removing the original nest turrets and following their rebuilding process over three consecutive days, age-dependent changes in wall morphology and inner lining fabrics were characterized. Micromorphological descriptions based on thin sections of turret walls revealed the building behaviors involved. Ants started by collecting nearby twigs and dry grass fragments that are piled up around the nest entrance. Several large fragments held the structure like beams. As a net-like structure grew, soil pellets were placed in between the intermeshed plant fragments from the turret base to the top, reinforcing the structure. Concomitantly, the turret inner wall was lined with soil pellets, starting from the base. Therefore, the consolidation of the turret occurred both over time and from its base upwards. It is argued that nest turrets do not simply arise by the arbitrary deposition of nearby materials, and that workers selectively incorporate large materials at the beginning, and respond to the developing structure by reinforcing the intermeshed plant fragments over time.
Exon-4 Mutations in KRAS Affect MEK/ERK and PI3K/AKT Signaling in Human Multiple Myeloma Cell Lines

Approximately 20% of multiple myeloma (MM) cases harbor a point mutation in KRAS. However, there is still no final consent on whether KRAS-mutations are associated with disease outcome. Specifically, no data exist on whether KRAS-mutations have an impact on survival of MM patients at diagnosis in the era of novel agents. Direct blockade of KRAS for therapeutic purposes is mostly impossible, but recently a mutation-specific covalent inhibitor targeting KRAS\(^{p.G12C}\) entered into clinical trials. However, other KRAS hotspot-mutations exist in MM patients, including the less common exon-4 mutations. For the current study, the coding regions of KRAS were deep-sequenced in 80 newly diagnosed MM patients, uniformely treated with three cycles of bortezomib plus dexamethasone and cyclophosphamide (VCD)-induction, followed by high-dose chemotherapy and autologous stem cell transplantation. Moreover, the functional impact of KRAS\(^{p.G12A}\) and the exon-4 mutations p.A146T and p.A146V on different survival pathways was investigated. Specifically, KRAS\(^{WT}\), KRAS\(^{p.G12A}\), KRAS\(^{p.A146T}\), and KRAS\(^{p.A146V}\) were overexpressed in HEK293 cells and the KRAS\(^{WT}\) MM cell lines JJN3 and OPM2 using lentiviral transduction and the Sleeping Beauty vector system. Even though KRAS-mutations were not correlated with survival, all KRAS-mutants were found capable of potentially activating MEK/ERK- and sustaining PI3K/AKT-signaling in MM cells.
Amphibians evolved in the Devonian period about 400 Mya and represent a transition step in tetrapod evolution. Among amphibians, high-throughput sequencing data are very limited for Caudata, due to their largest genome sizes among terrestrial vertebrates. In this paper we present the transcriptome from the fire bellied newt Cynops orientalis. Data here presented display a high level of completeness, comparable to the fully sequenced genomes available from other amphibians. Moreover, this work focused on genes involved in gametogenesis and sexual development. Surprisingly, the gsdf gene was identified for the first time in a tetrapod species, so far known only from bony fish and basal sarcopterygians. Our analysis failed to isolate fgf24 and foxl3, supporting the possible loss of both genes in the common ancestor of Rhipidistians. In Cynops, the expression analysis of genes described to be sex-related in vertebrates singled out an expected functional role for some genes, while others displayed an unforeseen behavior, confirming the high variability of the sex-related pathway in vertebrates.
Conventional kiln drying of wood operates by the evaporation of water at elevated temperature. In the initial stage of drying, mobile water in the wood cell lumen evaporates. More slowly, water bound in the wood cell walls evaporates, requiring the breaking of hydrogen bonds between water molecules and cellulose and hemicellulose polymers in the cell wall. An alternative for wood kiln drying is a patented process for green wood dewatering through the molecular interaction of supercritical carbon dioxide with water of wood cell sap. When the system pressure is reduced to below the critical point, phase change from supercritical fluid to gas occurs with a consequent large change in CO2 volume. This results in the efficient, rapid, mechanical expulsion of liquid sap from wood. The end-point of this cyclical phase-change process is wood dewatered to the cell wall fibre saturation point. This paper describes dewatering over a range of green wood specimen sizes, from laboratory physical chemistry studies to pilot-plant trials. Magnetic resonance imaging and nuclear magnetic resonance spectroscopy were applied to study the fundamental mechanisms of the process, which were contrasted with similar studies of conventional thermal wood drying. In conclusion, opportunities and impediments towards the commercialisation of the green wood dewatering process are discussed.
Many modern statistically efficient methods come with tremendous computational challenges, often leading to large-scale optimisation problems. In this work, we examine such computational issues for recently developed estimation methods in nonparametric regression with a specific view on image denoising. We consider in particular certain variational multiscale estimators which are statistically optimal in minimax sense, yet computationally intensive. Such an estimator is computed as the minimiser of a smoothness functional (e.g., TV norm) over the class of all estimators such that none of its coefficients with respect to a given multiscale dictionary is statistically significant. The so obtained multiscale Nemirowski-Dantzig estimator (MIND) can incorporate any convex smoothness functional and combine it with a proper dictionary including wavelets, curvelets and shearlets. The computation of MIND in general requires to solve a high-dimensional constrained convex optimisation problem with a specific structure of the constraints induced by the statistical multiscale testing criterion. To solve this explicitly, we discuss three different algorithmic approaches: the Chambolle-Pock, ADMM and semismooth Newton algorithms. Algorithmic details and an explicit implementation is presented and the solutions are then compared numerically in a simulation study and on various test images. We thereby recommend the Chambolle-Pock algorithm in most cases for its fast convergence. We stress that our analysis can also be transferred to signal recovery and other denoising problems to recover more general objects whenever it is possible to borrow statistical strength from data patches of similar object structure.
The current molecular genetic diagnostic rates for hereditary hearing loss (HL) vary considerably according to the population background. Pakistan and other countries with high rates of consanguineous marriages have served as a unique resource for studying rare and novel forms of recessive HL. A combined exome sequencing, bioinformatics analysis, and gene mapping approach for 21 consanguineous Pakistani families revealed 13 pathogenic or likely pathogenic variants in the genes GJB2, MYO7A, FGF3, CDC14A, SLITRK6, CDH23, and MYO15A, with an overall resolve rate of 61.9%. GJB2 and MYO7A were the most frequently involved genes in this cohort. All the identified variants were either homozygous or compound heterozygous, with two of them not previously described in the literature (15.4%). Overall, seven missense variants (53.8%), three nonsense variants (23.1%), two frameshift variants (15.4%), and one splice-site variant (7.7%) were observed. Syndromic HL was identified in five (23.8%) of the 21 families studied. This study reflects the extreme genetic heterogeneity observed in HL and expands the spectrum of variants in deafness-associated genes.
Metabolic Alterations Caused by Defective Cardiolipin Remodeling in Inherited Cardiomyopathies

The heart is the most energy-consuming organ in the human body. In heart failure, the homeostasis of energy supply and demand is endangered by an increase in cardiomyocyte workload, or by an insufficiency in energy-providing processes. Energy metabolism is directly associated with mitochondrial redox homeostasis. The production of toxic reactive oxygen species (ROS) may overwhelm mitochondrial and cellular ROS defense mechanisms in case of heart failure. Mitochondria are essential cell organelles and provide 95% of the required energy in the heart. Metabolic remodeling, changes in mitochondrial structure or function, and alterations in mitochondrial calcium signaling diminish mitochondrial energy provision in many forms of cardiomyopathy. The mitochondrial respiratory chain creates a proton gradient across the inner mitochondrial membrane, which couples respiration with oxidative phosphorylation and the preservation of energy in the chemical bonds of ATP. Akin to other mitochondrial enzymes, the respiratory chain is integrated into the inner mitochondrial membrane. The tight association with the mitochondrial phospholipid cardiolipin (CL) ensures its structural integrity and coordinates enzymatic activity. This review focuses on how changes in mitochondrial CL may be associated with heart failure. Dysfunctional CL has been found in diabetic cardiomyopathy, ischemia reperfusion injury and the aging heart. Barth syndrome (BTHS) is caused by an inherited defect in the biosynthesis of cardiolipin. Moreover, a dysfunctional CL pool causes other types of rare inherited cardiomyopathies, such as Sengers syndrome and Dilated Cardiomyopathy with Ataxia (DCMA). Here we review the impact of cardiolipin deficiency on mitochondrial functions in cellular and animal models. We describe the molecular mechanisms concerning mitochondrial dysfunction as an incitement of cardiomyopathy and discuss potential therapeutic strategies.
Infants and young children (IYC) remain the most vulnerable population group to environmental hazards worldwide, especially in economically developing regions such as sub-Saharan Africa (SSA). As a result, several governmental and non-governmental institutions including health, environmental and food safety networks and researchers have been proactive toward protecting this group. Mycotoxins, toxic secondary fungal metabolites, contribute largely to the health risks of this young population. In SSA, the scenario is worsened by socioeconomic status, poor agricultural and storage practices, and low level of awareness, as well as the non-establishment and lack of enforcement of regulatory limits in the region. Studies have revealed mycotoxin occurrence in breast milk and other weaning foods. Of concern is the early exposure of infants to mycotoxins through transplacental transfer and breast milk as a consequence of maternal exposure, which may result in adverse health effects. The current paper presents an overview of mycotoxin occurrence in foods intended for IYC in SSA. It discusses the imperative evidence of mycotoxin exposure of this population group in SSA, taking into account consumption data and the occurrence of mycotoxins in food, as well as biomonitoring approaches. Additionally, it discusses the health implications associated with IYC exposure to mycotoxins in SSA.

Die Reaktion zwischen Aryl‐ und Amino(dihydro)boranen und Dibora[2]ferrocenophan 1 führt zur Bildung von 1,3‐trans‐Dihydrotriboranen durch formale Hydrierung und Insertion eines Borylens in die B=B Doppelbindung. Die Aryltriboran‐Derivate unterliegen einer reversiblen Photoisomerisierung zugunsten eines cis‐1,2‐μ‐H‐3‐Hydrotriborans, während eine Hydridabstraktion zu kationischen Triboranen führt, welche die ersten doppelt basenstabilisierten B\(_3\)H\(_4\)\(^+\)‐Analoga darstellen.
Wir berichten über die katalytische Triborierung terminaler Alkine mit B\(_2\)pin\(_2\) (Bis‐(pinakolato)‐dibor) unter Verwendung von einfach zugänglichem Cu(OAc)\(_2\) und P\(^n\)Bu\(_3\). Verschiedene 1,1,2‐Triborylalkene, eine Verbindungsklasse mit potentieller Funktion als Matrix‐Metallo‐Proteinase(MMP‐2)‐Inhibitor, werden direkt in mäßigen bis guten Ausbeuten erhalten. Das Verfahren zeichnet sich durch milde Reaktionsbedingungen, ein breites Substratspektrum und eine gute Verträglichkeit gegenüber funktionellen Gruppen aus. Diese Cu‐katalysierte Reaktion kann im Gramm‐Maßstab durchgeführt werden und liefert die entsprechenden 1,1,2‐Triborylalkene in mäßigen Ausbeuten. Die Verwendung solcher Verbindungen wird anhand weiterer Transformationen der C−B‐Bindungen zur Darstellung eines geminalen Dihalogenborylalkens (F, Cl, Br), eines Monohalogendiborylalkens (Cl, Br) und eines trans‐Diaryldiborylalkens demonstriert, welche bedeutende Synthesebausteine darstellen und bisher nur schwer zugänglich waren.
Anhand der ersten Festkörperstrukturen von Dibortetraiodid (B\(_2\)I\(_4\)) wird gezeigt, dass dieses nicht, wie lange angenommen, analog zu den leichteren Dibortetrahalogeniden B\(_2\)F\(_4\), B\(_2\)Cl\(_4\) und B\(_2\)Br\(_4\) in allen Aggregatzuständen in Form diskreter Moleküle mit planaren, dreifach koordinierten Boratomen vorliegt. Röntgenstrukturanalysen, Festkörper‐NMR‐ und IR‐Messungen zeigen, dass B\(_2\)I\(_4\) im Festkörper in zwei polymeren Konformeren vorkommt, die tetraedrisch koordinierte Boratome enthalten. Anhand von DFT‐Rechnungen werden die IR‐Spektren in Lösung und im Festkörper simuliert und mit den experimentellen Daten verglichen.
Objectives
Embedded in the Collaborative Research Center “Fear, Anxiety, Anxiety Disorders” (CRC‐TRR58), this bicentric clinical study aims at identifying biobehavioral markers of treatment (non‐)response by applying machine learning methodology with an external cross‐validation protocol. We hypothesize that a priori prediction of treatment (non‐)response is possible in a second, independent sample based on multimodal markers.
Methods
One‐session virtual reality exposure treatment (VRET) with patients with spider phobia was conducted on two sites. Clinical, neuroimaging, and genetic data were assessed at baseline, post‐treatment and after 6 months. The primary and secondary outcomes defining treatment response are as follows: 30% reduction regarding the individual score in the Spider Phobia Questionnaire and 50% reduction regarding the individual distance in the behavioral avoidance test.
Results
N = 204 patients have been included (n = 100 in Würzburg, n = 104 in Münster). Sample characteristics for both sites are comparable.
Discussion
This study will offer cross‐validated theranostic markers for predicting the individual success of exposure‐based therapy. Findings will support clinical decision‐making on personalized therapy, bridge the gap between basic and clinical research, and bring stratified therapy into reach. The study is registered at ClinicalTrials.gov (ID: NCT03208400).
The introduction of new therapeutic agents has revolutionized the treatment of metastatic melanoma. The approval of adjuvant anti‐programmed death‐1 monotherapy with nivolumab or pembrolizumab, and dabrafenib plus trametinib has recently set a new landmark in the treatment of stage III melanoma. Now, clinical trials have shown that immune checkpoint blockade can be performed in a neoadjuvant setting, an approach established as a standard therapeutic approach for other tumour entities such as breast cancer. Recent studies suggest that a pathological response achieved by neoadjuvant immunotherapy is associated with long‐term tumour control and that short neoadjuvant application of checkpoint inhibitors may be superior to adjuvant therapy. Most recently, neoadjuvant ipilimumab plus nivolumab in stage III melanoma was reported. With two courses of dose‐optimized ipilimumab (1 mg kg−1) combined with nivolumab (3 mg kg−1), pathological responses were observed in 77% of patients, while only 20% of patients experienced grade 3 or 4 adverse events. However, the neoadjuvant trials employing combined immune checkpoint blockade conducted so far have excluded patients with in transit metastases, a common finding in stage III melanoma. Here we report four patients with in transit metastases or an advanced primary tumour who have been treated with neoadjuvant ipilimumab plus nivolumab according to the OpACIN‐neo trial scheme (arm B). All patients achieved radiological disease control and a pathological response. None of the patients has relapsed so far.
Zum Zusammenhang von Staatlichkeit und lokalen Selbstregelungen: Einige theoretische Überlegungen

Dieses Working Paper widmet sich dem Zusammenhang von Staatlichkeit und lokalen Selbstregelungen. Mittels eines deduktiven Zugangs werden verschiedene theoretische Szenarien dieses Zusammenhangs durchgespielt. Hierbei handelt es sich neben den generellen Zusammenhängen der beiden Regelungsformen um den Umfang der Regelungen, die Beziehungsformen zwischen den Regelungsvarianten, den Voraussetzungen zur Implementierung von Regelungen sowie der Verknüpfung der Regelungsformen mit dem Konzept der Staatlichkeit.
Die Studie erweitert das Forschungsfeld um drei wesentliche Erkenntnisse: Erstens, dass es neben dem postulierten negativen Zusammenhang von staatlichen Regulierungen und lokalen Selbstregelungen noch weitere hypothetisch anzunehmende Zusammenhänge geben kann. Zweitens, dass neben staatlicher Regulierung und lokalen Selbstregelungen auch regelungsfreie Regelungsbedarfe existieren und stärker in den Blick genommen werden sollten. Drittens, dass der Grad der Staatlichkeit unabhängig vom Ausmaß staatlicher Regulierung betrachtet werden sollte.
Erfolgreiche räumliche Orientierung ist für viele Tiere eine alltägliche Herausforderung. Cataglyphis‐Wüstenameisen sind bekannt für ihre Navigationsfähigkeiten, mit deren Hilfe sie nach langen Futtersuchläufen problemlos zum Nest zurückfinden. Wie aber nehmen naive Ameisen ihre Navigationssysteme in Betrieb? Nach mehrwöchigem Innendienst im dunklen Nest werden sie zu Sammlerinnen bei hellem Sonnenschein. Dieser Wechsel erfordert einen drastischen Wandel im Verhalten sowie neuronale Veränderungen im Gehirn. Erfahrene Ameisen orientieren sich vor allem visuell, sie nutzen einen Himmelskompass und Landmarkenpanoramen. Daher absolvieren naive Ameisen stereotype Lernläufe, um ihren Kompass zu kalibrieren und die Nestumgebung kennenzulernen. Während der Lernläufe blicken sie wiederholt zum Nesteingang zurück und prägen sich so ihren Heimweg ein. Zur Ausrichtung ihrer Blicke nutzen sie das Erdmagnetfeld als Kompassreferenz. Cataglyphis‐Ameisen besitzen hierfür einen Magnetkompass, der bislang unbekannt war.
Nearly all classes of coding and non-coding RNA undergo post-transcriptional modification including RNA methylation. Methylated nucleotides belong to the evolutionarily most conserved features of tRNA and rRNA.1,2 Many contemporary methyltransferases use the universal cofactor S-adenosylmethionine (SAM) as methyl group donor. This and other nucleotide-derived cofactors are considered as evolutionary leftovers from an RNA World, in which ribozymes may have catalysed essential metabolic reactions beyond self-replication.3 Chemically diverse ribozymes seem to have been lost in Nature, but may be reconstructed in the laboratory by in vitro selection. Here, we report a methyltransferase ribozyme that catalyses the site-specific installation of 1-methyladenosine (m1A) in a substrate RNA, utilizing O6-methylguanine (m6G) as a small-molecule cofactor. The ribozyme shows a broad RNA sequence scope, as exemplified by site-specific adenosine methylation in tRNAs. This finding provides fundamental insights into RNA’s catalytic abilities, serves a synthetic tool to install m1A in RNA, and may pave the way to in vitro evolution of other methyltransferase and demethylase ribozymes.
Das Ziel der vorliegenden Arbeit war es, intervallartige Substrukturen (Melodieintervalle) auf der Melodiekontur von Säuglingslauten im Längsschnitt über die ersten vier Lebensmonate zu analysieren. Melodieintervalle werden als eine Messgröße für die Kurzzeitvariabilität der Grundfrequenz (F0) und damit der laryngealen Regelleistung angesehen. Es sollte belegt werden, dass gesunde Säuglinge Melodieintervalle regelhaft erzeugen. Dabei war auch die Frage zu beantworten, ob Melodien mit Intervallen häufiger vorkommen als Melodien ohne Intervall über den Untersuchungszeitraum der ersten vier Lebensmonate. Neben Häufigkeitsanalysen sollten auch Analysen temporaler Eigenschaften erfolgen und Frequenzratios (Intervallgrößen) ermittelt werden. Langzeitziel dieser Analysen ist es, potenzielle Risikokinder einer späteren Sprech- und Sprachentwicklungsstörung so früh wie möglich anhand einer nicht-invasiven Vorsprachlichen Diagnostik identifizieren zu können.
Aus einem Gesamtdatenkorpus von 6130 Vokalisationen wurden in einer komplexen Vorselektierungs-Routine mittels audio-visueller Analyse insgesamt 3114 Vokalisationen für die finalen Melodieintervallanalysen als geeignet befunden. Unter Methodenmodifikation zu Vorarbeiten, wie einer herabgesetzten Plateaumindestlänge auf 50 ms und unter Einbezug rhythmisch-segmentierter Vokalisationen in die Analysen wurden die Aufnahmen anhand von Melodie- und Intensitätsdiagrammen sowie semi-automatisch zugehörigen Messroutinen im Cry-Data-Analysis-Program (CDAP) analysiert und Melodieintervalle vermessen. Des Weiteren wurden Analysen der Melodiestrukturkategorien, die den Komplexitätsgrad der Konturen reflektieren, und der Bogenformen (Intonation) durchgeführt, um das Auftreten identifizierter Intervallcodes und deren Muster zu diesen in Bezug zu setzen. Das melodische Einzelintervall (Plateau-Übergang-Plateau) wurde als Modul definiert, das zu Doppelintervallen und noch komplexeren Kombinationen unterschiedlicher Codes und Muster zusammengesetzt wird. Das Repertoire dieser Kombinationsmuster wurde in der Arbeit detailliert aufgezeigt. Ein Modul-Vergleich der steigenden und fallenden Einzelintervalle in Einfach- und Doppelbögen konnte eine hohe Übereinstimmung hinsichtlich spezieller Messgrößen, insbesondere der steigenden Intervalle, belegen. Für die über den Untersuchungszeitraum analysierten Auftrittshäufigkeiten der Intervalle konnte mittels einer verallgemeinerten Schätzgleichung (GEE-Model) eine signifikante Zunahme des Intervallauftretens festgestellt werden, die durch einen nichtlinearen Alterseffekt gekennzeichnet war. Die Intervallkomplexität nahm linear signifikant mit dem Lebensalter zu. Es wurden keine Geschlechtseffekte festgestellt. Als vorherrschende Intervallgröße wurde die kleine Sekunde (Halbton) über den Untersuchungszeitraum gefunden. Die Intervallgrößen und ein Großteil der Analysen der temporalen Messgrößen erfolgte auf signalbasierter Ebene und wurde deskriptiv vergleichend zwischen den Richtungen bzw. den Mustern der Intervalle untersucht und als Referenzwerte in umfangreichen Tabellen berichtet. Die Ergebnisse vorliegender Arbeit belegten durch Analysen unterschiedlicher Messgrößen, wie z.B. dem Komplexitätsgrad der Melodie und deren regelrechter Entwicklung mit dem Alter, dass alle hier analysierten Vokalisationen von Probanden stammten, die sich unauffällig entwickelten. Somit wurde sichergestellt, dass die erarbeiteten Referenzwerte für nachfolgende Studien die Verhältnisse bei gesunden Säuglingen widerspiegeln. Die Ergebnisse vorliegender Arbeit sollen als vorläufige Vergleichswerte für geplante Analysen an Vokalisationen von Säuglingen mit orofazialen Spaltbildungen dienen.
Die therapeutischen Möglichkeiten des metastasierten Prostatakarzinoms (Pca) haben sich durch die neuen Substanzen Docetaxel und Abirateron deutlich verbessert.
Das prostataspezifische Membranantigen (PSMA) stellt für die Diagnose und Therapie des Pca´s einen vielversprechenden Angriffspunkt dar. PSMA wird in Prostatakarzinomzellen überexprimiert und dient als Zielstruktur für nicht-invasives bildgebendes Verfahren und Lutetium-177-PSMA-Radioligandentherapie als Therapieoption.
Der CXCR4-Rezeptor wird an unterschiedlichen Zelltypen und Organen exprimiert. Seine Überexpression wird mit einer Metastasierung und schlechter Prognose assoziiert.
Gallium-68-PSMA PET/CT liefert genaue Kenntnisse bezüglich Ausbreitung und Fortschreiten des Tumorgeschehens.
Die vorliegende Arbeit untersucht die Zusammenhänge zwischen Expression von PSMA und CXCR4 in Verbindung mit etablierten Therapeutika und versucht Wege aufzuzeichnen, welche durch Erhöhung der PSMA-Expression zur verbesserten Sensitivität des PSMA PET/CT führen könnten, wodurch der personalisierte Therapieansatz weiter optimiert werden kann.
Fatigue als ein „überwältigendes Gefühl von Müdigkeit, Energielosigkeit und Erschöpfung” stellt bei Patienten mit MS ein häufig auftretendes und oft im Alltag beeinträchtigendes Symptom dar, das sowohl mit körperlichen als auch mit kognitiven Erschöpfungssymptomen einhergeht. Die objektive Erfassung des Schweregrades der Fatigue beim einzelnen Patienten stellt ein Problem dar, da bisher keine objektiven Messverfahren zur Erfassung der Fatigue existieren. Im klinischen Alltag kommen meist Fragebögen zum Einsatz, die das Ausmaß der subjektiven Beeinträchtigung durch Fatigue im Alltag quantifizieren sollen.
Ziel dieser Arbeit war es, zu untersuchen, inwieweit bestimmte im klinischen Alltag erhobene Parameter Rückschlüsse auf die subjektive Fatigue bei Patienten mit MS erlauben, auch im Hinblick darauf, ob sich einzelne Parameter besonders zur Einschätzung der körperlichen bzw. kognitiven Fatigue eignen. Zudem sollte untersucht werden, ob die untersuchten klinischen Parameter bei bestimmten Patientengruppen besser als bei anderen Rückschlüsse auf die subjektive Fatigue erlauben. Erfasst wurde die subjektive Fatigue durch das Würzburger Erschöpfungsinventar bei Multipler Sklerose (WEIMuS), einer Serie von Fragen, die zwischen körperlicher und kognitiver Fatigue unterscheiden.
Dazu wurden Korrelationsanalysen zwischen der WEIMuS-Gesamtskala bzw. deren Subskalen für körperliche und kognitive Fatigue und EDSS-Wert, MSFC Z-Score einschließlich dessen Subscores und der Zeit des 50-Meter-Gehversuchs durchgeführt. Bezüglich der körperlichen Fatigue ergaben sich zwischen der WEIMuS-Subskala für körperliche Fatigue und dem EDSS sowie der Zeit des 50-Meter-Gehversuchs im Vergleich die stärksten, absolut gesehen als mittelstark zu wertende, Korrelationen. Bezüglich der kognitiven Fatigue ergab sich die stärkste Korrelation zwischen der WEIMuS-Subskala für kognitive Fatigue und dem PASAT3, die allerdings trotzdem als gering zu werten ist. Mit EDSS und 50-Meter-Gehversuch scheinen also zwei objektive klinische Parameter zu existieren, die in einem gewissen Maß auf die subjektive Fatigue rückschließen lassen. Ziel weiterer Untersuchungen wird es sein müssen, einen geeigneten klinischen Parameter zu finden, der bessere Rückschlüsse auf die subjektive kognitive Fatigue erlaubt als der PASAT3.
Zwischen der WEIMuS-Gesamtskala bzw. deren Subskalen für körperliche und kognitive Fatigue und Alter, Geschlecht und Erkrankungsdauer fanden sich bestenfalls geringe Korrelationen, weshalb diese Parameter ungeeignet erscheinen, Aussagen über die subjektive Fatigue zu machen.
Durch die Einteilung der Patienten nach Alter und Geschlecht konnte untersucht werden, inwieweit diese Parameter Einfluss auf die untersuchten Zusammenhänge zwischen klinischen Parametern und subjektiver Fatigue haben. Die Korrelationen zwischen den WEIMuS-Subskalen für körperliche und kognitive Fatigue mit den untersuchten klinischen Parametern waren für junge Patienten überwiegend stärker als für ältere Patienten, insbesondere ältere Männer. Somit scheinen die untersuchten klinischen Parameter bei jüngeren Patienten besser geeignet, Aussagen über die subjektive Fatigue zu machen als bei älteren.
Insgesamt ist festzuhalten, dass EDSS und 50-Meter-Gehversuch insbesondere bei jungen Patienten zu einer besseren objektiven Beurteilbarkeit vor allem der körperlichen Fatigue im klinischen Alltag beitragen können.
Patienten mit einer HIV-Infektion werden mit antiretroviralen Medikamenten therapiert, um die Krankheitsprogression zu verhindern, die HIV-Replikation zu hemmen und klinische HIV-induzierte Symptome zu verbessern. Für HIV-assoziierte neurokognitive Störungen (HAND) als Folge einer Infektion mit HIV gibt es jedoch noch keine ausreichende Therapie. In dieser Arbeit wurden PBMC, Serum- und Liquorproben von Teilnehmern einer placebokontrollierten klinischen Studie analysiert, in der die Wirksamkeit einer Lithium-Therapie bei HIV-Patienten mit HAND untersucht wurde. Die Studienteilnehmer erhielten für sechs Monate zusätzlich zu ihrer antiretroviralen Medikation Lithiumcarbonat bzw. ein Placebo. Die Einschlusskriterien waren eine mindestens sechs Monaten bestehende antiretrovirale Therapie bei unterdrückter Viruslast und eine mittelschwere bis schwere Manifestation der HIV-assoziierten neurokognitiven Störung. Die Studie wurde in Primary Care Kliniken des öffentlichen Sektors in Kapstadt, Südafrika, durchgeführt. Als primärer Wirksamkeitsendpunkt wurde die Veränderung des Global Deficit Scores (GDS) von Beginn der Studie bis zum Ende der 24. Woche festgelegt. Es hatten insgesamt 66 Patienten an der Studie teilgenommen, davon bekamen 32 Patienten das Medikament Lithiumcarbonat und 34 Patienten ein Placebo. Den Patienten wurden zu den Zeitpunkten Baseline (V3) und 24. Woche (V12) Blut- und CSF-Proben entnommen. In der vorliegenden Arbeit wurden die Proben mittles Durchflusszytometrie und ELISA untersucht. Die Daten wurden mit dem GDS sowie weiteren Progressionsmarkern der HIV-Infektion wie Viruslast und CD4+ Zellzahl korreliert. Von den 66 Teilnehmern, die die Studie begonnen haben, schlossen insgesamt 61 Teilnehmer ab, darunter hatten 30 Patienten Lithiumcarbonat und 31 Patienten ein Placebo eingenommen. Das Durchschnittsalter lag bei 40 Jahren und die mittlere CD4+ T-Zellzahl umfasste 500 Zellen pro ml. Es traten sechs schwerwiegende unerwünschte Ereignisse auf, aber keines wurde in Zusammenhang mit dem Studienmedikament betrachtet. Die Patienten mit HAND, die zusätzlich zu einer antiretroviralen Therapie eine medikamentöse Behandlung mit Lithium erhielten, haben dieses klinisch gut vertragen. Jedoch konnte keine Verbesserung in Bezug auf neurokognitive Beeinträchtigungen für die Studienteilnehmer beobachtet werden. In der vorliegenden Arbeit konnte gezeigt werden, dass im zeitlichen Verlauf von V3 zu V12 ein signifikante Verbesserung des GDS innerhalb der Placebo-, sowie innerhalb der Lithium-Gruppe beobachtet werden konnte, wohingegen bei den Gruppen untereinander, Lithium versus Placebo, zu den jeweiligen Zeitpunkten V3 sowie V12, ähnliche Werte gemessen werden konnten und sich daher die Lithium-, und die Placebo-Gruppe nicht signifikant voneinander unterschieden. Aus den in dieser Arbeit gewonnenen Ergebnissen ließ sich zudem ableiten, dass ein signifikanter Abfall der BDNF Konzentration im Plasma zu verzeichnen war. Insgesamt war ein Trend erkennbar, dass Patienten mit blip eine höhere Immunaktivierung zeigten, als Patienten ohne blip. Jedoch hatte die Behandlung mit Lithium keinen Einfluss auf die Immunaktivierung der CD8+ T-Zellen. Des Weiteren zeigte sich unter der Lithium-Therapie ein signifikanter Abfall der CD4+ Zellzahlen.
L-type calcium channels (LTCCs) control crucial physiological processes in cardiomyocytes such as the duration and amplitude of action potentials, excitation-contraction coupling and gene expression, by regulating the entry of Ca2+ into the cells. Cardiac LTCCs consist of one pore-forming α1 subunit and the accessory subunits Cavβ, Cavα2δ and Cavγ. Of these auxiliary subunits, Cavβ is the most important regulator of the channel activity; however, it can also have LTCC-independent cellular regulatory functions. Therefore, changes in the expression of Cavβ can lead not only to a dysregulation of LTCC activity, but also to changes in other cellular functions. Cardiac hypertrophy is one of the most relevant risk factors for congestive heart failure and depends on the activation of calcium-dependent prohypertrophic signaling pathways. However, the role of LTCCs and especially Cavβ in this pathology is controversial and needs to be further elucidated.
Of the four Cavβ isoforms, Cavβ2 is the predominant one in cardiomyocytes. Moreover, there are five different splice variants of Cavβ2 (Cavβ2a-e), differing only in the N-terminal region. We reported that Cavβ2b is the predominant variant expressed in the heart. We also revealed that a pool of Cavβ2 is targeted to the nucleus in cardiomyocytes. The expression of the nuclear Cavβ2 decreases during in vitro and in vivo induction of cardiomyocyte hypertrophy and overexpression of a nucleus-targeted Cavβ2 completely abolishes the in vitro induced hypertrophy. Additionally, we demonstrated by shRNA-mediated protein knockdown that downregulation of Cavβ2 enhances the hypertrophy induced by the α1-adrenergic agonist phenylephrine (PE) without involvement of LTCC activity. These results suggest that Cavβ2 can regulate cardiac hypertrophy through LTCC-independent pathways. To further validate the role of the nuclear Cavβ2, we performed quantitative proteome analyses of Cavβ2-deficient neonatal rat cardiomyocytes (NRCs). The results show that downregulation of Cavβ2 influences the expression of various proteins, including a decrease of calpastatin, an inhibitor of the calcium-dependent cysteine protease calpain. Moreover, downregulation of Cavβ2 during cardiomyocyte hypertrophy drastically increases calpain activity as compared to controls after treatment with PE. Finally, the inhibition of calpain by calpeptin abolishes the increase in PE-induced hypertrophy in Cavβ2-deficient cells. These results suggest that nuclear Cavβ2 has Ca2+- and LTCC-independent functions during the development of hypertrophy. Overall, our results indicate a new role for Cavβ2 in antihypertrophic signaling in cardiac hypertrophy.
Die Entwicklung hochauflösender Fluoreszenzmikroskopiemethoden hat die Lichtmikroskopie revolutioniert. Einerseits ermöglicht die höhere erzielte räumliche Auflösung die Abbildung von Strukturen, die deutlich unterhalb der beugungsbedingten Auflösungsgrenze liegen. Andererseits erhält man durch Einzelmoleküllokalisationsmikroskopiemethoden
wie dSTORM (Direct Stochastic Optical Reconstruction Microscopy) Informationen, welche man für quantitative Analysen heranziehen kann. Aufgrund der sich dadurch bietenden neuen Möglichkeiten, hat sich die hochauflösende Fluoreszenzmikroskopie rasant entwickelt und kommt mittlerweile zur Untersuchung einer Vielzahl biologischer und medizinischer Fragestellungen zum Einsatz. Trotz dieses Erfolgs ist jedoch nicht zu verleugnen, dass auch diese neuen Methoden ihre Nachteile haben. Dazu zählt die Notwendigkeit relativ hoher Laserleistungen, welche Voraussetzung für hohe Auflösung ist und bei lebenden Proben zur Photoschädigung führen kann.
Diese Arbeit widmet sich sowohl dem Thema der Photoschädigung durch Einzelmoleküllokalisationsmikroskopie,
als auch der Anwendung von dSTORM und SIM (Structured Illumination Microscopy) zur Untersuchung neurobiologischer Fragestellungen auf Proteinebene.
Zur Ermittlung der Photoschädigung wurden lebende Zellen unter typischen Bedingungen bestrahlt und anschließend für 20−24 h beobachtet. Als quantitatives Maß für den Grad der Photoschädigung wurde der Anteil sterbender Zellen bestimmt. Neben der zu erwartenden Intensitäts- und Wellenlängenabhängigkeit, zeigte sich, dass die Schwere der Photoschädigung auch von vielen weiteren Faktoren abhängt und dass sich Einzelmoleküllokalisationsmikroskopie bei Berücksichtigung der gewonnenen Erkenntnisse durchaus mit Lebendzellexperimenten vereinbaren lässt.
Ein weiteres Projekt diente der Untersuchung der A- und B-Typ-Glutamatrezeptoren an der neuromuskulären Synapse von Drosophila melanogaster mittels dSTORM. Dabei konnte eine veränderte Anordnung beider Rezeptortypen infolge synaptischer Plastizität beobachtet, sowie eine absolute Quantifizierung des A-Typ-Rezeptors durchgeführt werden.
Im Mittelpunkt eines dritten Projekts standen Cadherin-13 (CDH13) sowie der Glucosetransporter Typ 3 (GluT3), welche beide mit der Aufmerksamkeitsdefizit-Hyperaktivitätsstörung in Verbindung gebracht werden. CDH13 konnte mittels SIM in serotonergen Neuronen, sowie radiären Gliazellen der dorsalen Raphekerne des embryonalen Mausgehirns nachgewiesen werden. Die Rolle von GluT3 wurde in aus induzierten pluripotenten Stammzellen differenzierten Neuronen analysiert, welche verschiedene Kopienzahlvariation des für GluT3-codierenden SLC2A3-Gens aufwiesen. Die Proteine GluT3, Bassoon und Homer wurden mittels dSTORM relativ quantifiziert. Während die Deletion des Gens zu einer erwartenden Verminderung von GluT3 auf Proteinebene führte, hatte die Duplikation keinen Effekt auf die GluT3-Menge. Für Bassoon und Homer zeigte sich weder durch die Deletion noch die Duplikation eine signifikante Veränderung.
Allocation planning describes the process of allocating scarce supply to individual customers in order to prioritize demands from more important customers, i.e. because they request a higher service-level target. A common assumption across publications is that allocation planning is performed by a single planner with the ability to decide on the allocations to all customers simultaneously. In many companies, however, there does not exist such a central planner and, instead, allocation planning is a decentral and iterative process aligned with the company's multi-level hierarchical sales organization.
This thesis provides a rigorous analytical and numerical analysis of allocation planning in such hierarchical settings. It studies allocation methods currently used in practice and shows that these approaches typically lead to suboptimal allocations associated with significant performance losses. Therefore, this thesis provides multiple new allocation approaches which show a much higher performance, but still are simple enough to lend themselves to practical application. The findings in this thesis can guide decision makers when to choose which allocation approach and what factors are decisive for their performance. In general, our research suggests that with a suitable hierarchical allocation approach, decision makers can expect a similar performance as under centralized planning.
Die Häufigkeit der endovaskulären Interventionen (EI) an den unteren Extremitäten zur Behandlung der peripheren arteriellen Verschlusskrankheit (pAVK) hat in den letzten Jahren zugenommen. Bei steigender Inzidenz der pAVK und bei gleichzeitiger Reduzierung der Rate mit operativen Interventionen (OI) profitieren die Patienten von der EI. Außerdem stieg die Anzahl der älteren Patienten, die aufgrund einer pAVK im Krankenhaus einer EI zugeführt wurden. Es zeigt sich eine Erweiterung der Indikationen für eine EI von den Claudicanten hin zu den Patienten mit den Stadien der kritischen Ischämie. Hierdurch werden gerade ältere und morbidere Patienten mit komplexeren endovaskulären Prozeduren konfrontiert. Insbesondere im Krankenhaus ergibt sich die Möglichkeit, den pAVK-Patienten, die bisher eine OI mit einer anästhesiologischen Begleitung erhielten, eine EI in SPA anzubieten. In der Regel werden EI in Lokalanästhesie durchgeführt. Die Anwendung der SPA bei EI ist interessant, da es möglicherweise neben der kompletten Schmerzausschaltung der unteren Extremitäten bei erhaltener Kommunikation zu Synergieeffekten durch die periphere Vasodilatation kommen kann. Als ein seltenes Verfahren zur Verbesserung der peripheren Durchblutung hat dies die CT-gesteuerte temporäre Sympathikolyse nach erfolgter EI gezeigt. Um prospektiv zu untersuchen, ob eine EI in SPA eine Verbesserung des Outcome und des Behandlungsablaufs mit sich bringt, ist zunächst wichtig zu zeigen, dass die Anwendung von SPA bei EI nicht gefährlich ist. Hierzu stehen Daten aus der Allgemein-, Viszeral- und Gefäßchirurgischen Abteilung des Klinikums Main-Spessart zur Verfügung. Im Zeitraum vom 15.12.2009 bis 22.01.2015 wurden bei 59 Patienten EI in SPA durchgeführt.
Bei keinem der 59 Patienten kam es durch die Anwendung der SPA zu anästhesieabhängigen Komplikationen. Als zu erwartende Nebenwirkung trat der durch temporäre Sympathikolyse ausgelöste Blutdruckabfall bei allen Patienten auf. Im Mittel sank der Wert um 24,2 % des Ausgangswerts, bei 13 Patienten (22,0 %) wurde eine Korrektur des Blutdruckabfalls mit Akrinor® durchgeführt und gut beherrscht. Für die Stärke des Blutdruckabfalls konnte keine Signifikanz bei der ASA-Einteilung, der Geschlechterverteilung und dem Alter nachgewiesen werden, somit besteht für eine EI in SPA keine Kontraindikation für den älteren und kränkeren pAVK-Patienten.
Die Ergebnisse dieser Dissertation zeigen, dass die Anwendung der SPA bei EI ein sicheres Verfahren ist. Die SPA kann, gerade bei zu erwartenden komplexen peripheren EI eine für den Patienten und Interventionisten angenehme und sichere Alternative zur Lokalanästhesie sein. Der personelle und materialbedingte Aufwand einer EI in SPA, im Vergleich mit einer EI in Lokalanästhesie, ist dabei deutlich erhöht und scheint übertrieben. Dass die EI in SPA Synergieeffekte mit sich bringt und deshalb sinnvoll ist, kann nur vermutet werden. Dieser Zusammenhang könnte im Rahmen einer prospektiven Studie, die beide Anästhesieverfahren für eine EI vergleicht, gezeigt werden.
Neisseria meningitidis (N. meningitidis) is a human commensal that occasionally causes life-threatening infections such as bacterial meningitis and septicemia. Despite experi-mental evidence that the expression of small non-coding RNAs (sRNAs) as well as the RNA chaperone Hfq affect meningococcal physiology, the impact of RNA-based regula-tion (riboregulation) on fitness and virulence in N. meningitidis is only poorly understood. Therefore, this study addressed these issues using a combination of high-throughput tech-nologies.
A differential RNA-sequencing (dRNA-seq) approach was applied to produce a single-nucleotide resolution map of the primary transcriptome of N. meningitidis strain 8013. The dRNA-seq analysis predicted 1,625 transcriptional start sites including 65 putative sRNAs, of which 20 were further validated by northern blot analysis. By Hfq RNA im-munopreci-pitation sequencing a large Hfq-centered post-transcriptional regulatory net-work comprising 23 sRNAs and 401 potential mRNA targets was identified. Rifampicin stability assays demonstrated that Hfq binding confers enhanced stability on its associat-ed sRNAs. Based on these data, the interactions of two paralogous sRNAs and their cog-nate target mRNA prpB were validated in vivo as well as in vitro. Both sRNAs directly repress prpB encoding a methylisocitrate lyse which was previously shown to be involved in meningococcal colonization of the human nasopharynx.
Besides the well-described RNA chaperone Hfq, FinO-domain proteins have recently been recognized as a widespread family of RNA-binding proteins (RBPs) with regulatory roles in diverse bacteria. They display an intriguing bandwidth of target sites, ranging from a single RNA pair as recognized by plasmid-encoded FinO to the global RNA regu-lons of enterobacterial ProQ proteins. To better understand the intrinsic targeting mode of this RBP family, in vivo targets of the minimal ProQ protein of N. meningitidis were de-termined. In vivo UV crosslinking with RNA deep sequencing (UV-CLIP) identified as-sociations of ProQ with 16 sRNAs and 166 mRNAs encoding a variety of biological functions and thus revealed ProQ as another global RBP in meningococci. It could be shown that meningococcal ProQ predominantly binds to highly structured RNA regions including DNA uptake sequences (DUS) and rho-independent transcription terminators and stabilizes many of its RNA targets as proved by rifampicin stability experiments. As expected from the large suite of ProQ-bound RNAs, proQ deletion globally affects both gene and protein expression in N. meningitidis, changing the expression levels of at least 244 mRNAs and 80 proteins. Phenotypic analyses suggested that ProQ promotes oxida-tive stress tolerance and UV damage repair capacity, both of which are required for full virulence of N. meningitidis.
Together, this work uncovers the co-existence of two major post-transcriptional regulons, one governed by ProQ, the other by Hfq, in N. meningitidis. It further highlights the role of these distinct RBPs and its associated sRNAs to bacterial virulence and indicates that riboregulation is likely to contribute to the way how meningococci adapt to different host niches.
A continuous arms race between the development of novel antibiotics and the evolution of corresponding resistance mechanisms in bacteria has been observed, since antibiotic agents like arsphenamines (e.g. Salvarsan, developed by Paul Ehrlich [1]), sulphonamides (e.g. Prontosil, Gerhard Domagk [2]) and penicillin (Alexander Fleming [3]) were first applied to effectively cure bacterial infections in the early 20th century. The rapid emergence of resistances in contrast to the currently lagging discovery of antibiotics displays a severe threat to human health. Some serious infectious diseases, such as tuberculosis or melioidosis, which were either thought to be an issue only in Third-World countries in case of tuberculosis, or regionally restricted with respect to melioidosis, are now on the rise to expand to other areas. In contrast, methicillin-resistant Staphylococcus aureus (MRSA) is already present in clinical setups all over the world and causes severe infections in immunocompromised patients. Thus, there is an urgent need for new and effective antimicrobial agents, which impair vital functions of the pathogen’s metabolism.
One central metabolic pathway is represented by the bacterial fatty-acid synthesis pathway (FAS II), which is essential for the synthesis of long and branched-chain fatty acids, as well as mycolic acids. These substances play a major role as modulating components of the properties of the most important protective barrier – the cell envelope. The integrity of the bacterial cell wall and the associated membrane(s) is crucial for cell growth and for protection against physical strain, intrusion of antibiotic agents and regulation of uptake of ions and other small molecules. Thus, this central pathway represents a promising target for antibiotic action against pathogens to combat infectious diseases. The last and rate-limiting step is catalysed by the trans-2-enoyl-ACP reductase (ENR) FabI or InhA (in mycobacteria), which has been demonstrated to be a valuable target for drug design and can be addressed, amongst others, by diphenyl ether (DPE) compounds, derived from triclosan (TCL) – the first one of this class which was discovered to bind to ENR enzymes [4, 5].
Based on this scaffold, inhibitors containing different combinations of substituents at crucial positions, as well as a novel type of substituent at position five were investigated regarding their binding behaviour towards the Burkholderia pseudomallei and Mycobacterium tuberculosis ENR enzymes bpFabI and InhA, respectively, by structural, kinetic and in-vivo experiments. Generally, substitution patterns modulate the association and dissociation velocities of the different ENR inhibitors in the context of the two-step slow-onset binding mechanism, which is observed for both enzymes. These alterations in the rapidity of complex formation and decomposition have a crucial impact on the residence time of a compound and hence, on the pharmacokinetic properties of potential drug candidates. For example, the substituents at the 2’-position of the DPE scaffold influence the ground- and transition state stability during the binding process to bpFabI, whereas 4’-substituents primarily alter the transition state [6]. The novel triazole group attached to the 5-position of the scaffold, targeting the hydrophobic part of the substrate-binding pocket in InhA, significantly enhances the energy barrier of the transition state of inhibitor binding [7] and decelerates the association- as well as the dissociation processes. Combinations with different substituents at the 2’-position can enhance or diminish this effect, e.g. by ground-state stabilisation, which will result in an increased residence time of the respective inhibitor on InhA.
Further structural investigations carried out in this work, confirm the proposed binding mode of a customised saFabI inhibitor [8], carrying a pyridone moiety on the DPE scaffold to expand interactions with the protein environment. Structural and preliminary kinetic data confirm the binding of the same inhibitor to InhA in a related fashion. Comparisons with structures of the ENR inhibitor AFN-1252 [9] bound to ENR enzymes from other organisms, addressing a similar region as the pyridone-moiety of the DPE inhibitor, suggest that also the DPE inhibitor bears the potential to display binding to homologues of saFabI and InhA and may be optimised accordingly.
Both of the newly investigated substituents, the pyridone moiety at the 4’-position as well as the 5-triazole substituent, provide a good starting point to modify the DPE scaffold also towards improved kinetic properties against ENR enzymes other than the herein studied and combining both groups on the DPE scaffold may have beneficial effects. The understanding of the underlying binding mechanism is a crucial factor to promote the dedicated design of inhibitors with superior pharmacokinetic characteristics.
A second target for a structure-based drug-design approach is the interaction surface between ENR enzymes and the acyl-carrier protein (ACP), which delivers the growing acyl chain to each distinct enzyme of the dissociated FAS-II system and presumably recognises its respective interaction partner via electrostatic contacts. The interface between saACP and saFabI was investigated using different approaches including crosslinking experiments and the design of fusion constructs connecting the ACP and the FabI subunits via a flexible linker region of varying lengths and compositions. The crosslinking studies confirmed a set of residues to be part of the contact interface of a previously proposed complex model [10] and displayed high crosslinking efficiency of saACP to saFabI when mutated to cysteine residues. However, crystals of the complex obtained from either the single components, or of the fusion constructs usually displayed weak diffraction, which supports the assumption that complex formation is highly transient. To obtain ordered crystals for structural characterisation of the complex it is necessary to trap the complex in a fixed state, e.g. by a high-affinity substrate attached to ACP [11], which abolishes rapid complex dissociation. For this purpose, acyl-coupled long-residence time inhibitors might be a valuable tool to elucidate the detailed architecture of the ACP-FabI interface. This may provide a novel basis for the development of inhibitors that specifically target the FAS-II biosynthesis pathway.
In the framework of the presented doctoral thesis, the plant ubiquitous, non-selective vacuolar cation channel TPC1/SV was electrophysiologically studied in Arabidopsis thaliana mesophyll vacuoles to further enlighten its physiological role in plant stress responses. For this, the hyperactive channel version fou2 (D454N), gaining a non-functional vacuolar calcium sensor, strong retarded growth phenotype and upregulated JA signalling pathway, and eight fou2 reverting WT-like ouf mutants were used. Except of ouf4, all other seven ouf mutants carried a 2nd mutation in the TPC1 gene. Therefore, the TPC1 electrical features of all ouf mutants were electrophysiologically characterized with the patch clamp method and compared with fou2 and WT.
Due to a missense mutation, ouf1 and ouf7 mutants harboured a truncated TPC1 channel protein, resulting in an impaired protein integrity and in turn loss of TPC1 channel activity. Accordingly, ouf1 and ouf7 mimicked the tpc1-2 null mutant with a WT- rather fou2-like phenotype. The ouf2 (G583D D454N) mutant exhibited inactive TPC1 channels, probably because the G583D mutation located in luminal part of the S11 helix caused (i) a shift of the activation threshold to much more positive voltages (i.e. to more than +110 mV) (ii) or channel blockage. As a result of the TPC1 channel inactivity, the ouf2 mutant also imitates the WT-like phenotype of the tpc1-2 null mutant. In the ouf6 mutant (A669V D454N) the 2nd reverting mutation selectively influenced fou2-like SV channel features. Both, the fast activation kinetics and reduced luminal calcium sensitivity were similar in ouf6 and fou2. However, deviations in both, the relative and absolute open channel probability, resulted in strongly reduced (80 %) current density at 0 mM and channel inactivity in the voltage range between -30 mV to +40 mV compared to fou2 and WT. Furthermore, the TPC1 channels in ouf6 exhibited a higher susceptibility to inhibitory luminal Ca2+ than fou2. As a result of these different effects, the TPC1 channel activity almost vanished at high luminal Ca2+ loads, what is very likely the reason that ouf6 lost the fou2-like phenotype. The ouf4 mutation did not change the fou2 TPC1-channel features like fast channel activation, single channel conductance and voltage-dependent gating behaviour. Nevertheless, the TPC1 current density was 80% less in ouf4 than in fou2. Since the TPC1 gene was not the target of the 2nd mutation, it can be assumed that it is modulated via external, yet unknown factor. In the ouf8 mutant the TPC1 channels additionally possess M629I mutation within the selectivity filter II resulting in a 50% decrease in the TPC1 unitary conductance. However, the slightly increased relative open channel probability of the TPC1 channels in ouf8 compared to fou2 appeared to be sufficient to compensate the reduced transport capacity of individual TPC1 channels. As a result, a similar macroscopic outward current density of ouf8 and fou2 was detected in the absence of vacuolar Ca2+. Furthermore, ouf8 mutation did not drastically change the typical fou2 TPC1 channel features such as fast activation, vacuolar calcium insensitivity and voltage dependency. However, a reversible block of the cytosol-directed potassium efflux at increased vacuolar calcium concentration in ouf8 mutant was found. Further inspection of transiently expressed TPC1 channel variants (M629I, M629T) on the single channel level suggest that Met629 of AtTPC1 in the channel pore region is crucial for the unitary channel conductance.
Taken together, current membrane recordings from ouf mutants revealed one common feature: All of them lacked or showed a strongly impaired ability for TPC1-mediated potassium release from the vacuole into the cytosol. Additionally, considering the detected dependence of the vacuolar membrane voltage on TPC1 activity, it thus seems that the TPC1-triggered vacuolar membrane depolarization caused by vacuolar K+ release plays a key role in generation of the fou2-like phenotype. Accordingly, one can conclude that TPC1-dependent vacuolar membrane depolarization and initiation of jasmonate production are likely linked. This statement is supported also by the complete restoration of WT-like plant phenotype and JA signalling in the ouf mutants. Finally, as a control element of the vacuolar membrane voltage TPC1 is probably upstream located in JA signalling pathway and therefore a perfect junction for linking multiple physiological stimuli and response to them.
Im Rahmen der vorgelegten Doktorarbeit wurde der in Pflanzen ubiquitär exprimierte, nicht-selektive vakuoläre Kationenkanal TPC1/SV elektrophysiologisch in Arabidopsis thaliana Mesophyllvakuolen untersucht, um seine physiologische Rolle in der pflanzlichen Stressantwort weiter aufzuklären. Hierfür wurde die hyperaktive Kanalvariante fou2 (D454N), die einen nicht-funktionalen vakuolären Calciumsensor, ein stark verzögertes Pflanzenwachstum und einen hochregulierten Jasmonsäure-Signalweg aufweist, sowie acht ouf Mutanten mit fou2-umkehrenden Phänotyp benutzt. Mit Ausnahme von ouf4 enthalten alle anderen ouf Mutanten eine weitere Mutation im TPC1-Gen. Daher wurden die elektrischen Eigenschaften von TPC1 in allen ouf Mutanten elektrophysiologisch mittels der Patch clamp Technik charakterisiert und mit fou2 und dem Wildtyp verglichen.
Aufgrund einer Missense-Mutation beinhalten die Mutanten ouf1 und ouf7 ein verkürztes TPC1 Protein, woraus eine gestörte Proteinintegrität resultiert und daraus wiederum ein Fehlen der TCP1-Kanalaktivität. Dementsprechend ähneln ouf1 und ouf7 der tpc1-2 Nullmutante mit einem WT- oder eher fou2-artigen Phänotyp. Wahrscheinlich weist die ouf2 (G583D D454N) Mutante einen inaktiven TPC1-Kanal auf, weil die G583D Mutation, die in einem luminalen Teil der S11 Helix sitzt, eine Verschiebung der Aktivierungsschwelle hin zu einer höheren Spannung (z. B. mehr als +110 mV) oder einen Kanalblock verursacht. Als Folge der TPC1 Kanal Inaktivität, ahmt die ouf2 Mutante auch den WT-ähnlichen Phänotyp der tpc1-2 Nullmutante nach. In der ouf6 Mutante (A669V D454N) beeinflusst die zweite Mutation selektiv die fou2-ähnlichen SV-Kanaleigenschaften. Sowohl die schnelle Aktivierungskinetik als auch die verringerte luminale Calciumsensitivität waren denen von ouf6 und fou2 ähnlich. Die Abweichungen in der relativen sowie der absoluten Offenwahrscheinlichkeit resultierten jedoch in einer stark reduzierten (80 %) Stromdichte bei 0 mM luminalem Calcium verglichen mit fou2 und dem WT, sowie einer Kanalinaktivität bei Spannungen zwischen -30 mV und +40 mV. Darüber hinaus zeigten die TPC1 Kanäle in ouf6 eine höhere Anfälligkeit für inhibitorisches, luminales Calcium als die in fou2. Das Ergebnis der beiden unterschiedlichen Effekte ist, dass die TPC1 Kanalaktivität bei einer hohen luminalen Calciumkonzentration fast verschwindet, woraus zu schließen ist, dass ouf6 den fou2-ähnlichen Phänotyp verlor. Die ouf4 Mutation veränderte nicht die fou2 TPC1 Kanaleigenschaften, wie die schnelle Kanalaktivierung, die Einzelkanalleitfähigkeit und das spannungsabhängige Verhalten. Nichtsdestotrotz war die TCP1 Stromdichte in ouf4 um 80 % geringer als in fou2. Da das TPC1 Gen nicht das Ziel der zweiten Mutation war, kann angenommen werden, dass es durch äußere, bisher noch unbekannte Faktoren, reguliert wird. In der ouf8 Mutante haben die TPC1 Kanäle zusätzlich eine M629I Mutation innerhalb des zweiten Selektivitätsfilters, welche in einem 50 % Rückgang der TCP1 Einzelkanalleitfähigkeit resultiert. Jedoch scheint die leicht erhöhte Offenwahrscheinlichkeit der TCP1 Kanäle in ouf8, verglichen mit fou2, ausreichend zu sein, um die reduzierte Transportkapazität der individuellen TPC1 Kanäle zu kompensieren. Schlussfolgernd wurde eine ähnliche makroskopische auswärts gerichtete Stromdichte des ouf8 und des fou2 in Abwesenheit vakuolären Calciums entdeckt. Des Weiteren änderte eine ouf8 Mutation die fou2 TPC1 Kanaleigenschaften wie eine schnelle Aktivierung, vakuoläre Calciuminsensitivität und die Spannungsabhängigkeit nicht drastisch. Jedoch wurde ein reversibler Block des Zytosol-gerichteten Kalium Ausstroms bei erhöhten vakuolären Calcium Konzentrationen in ouf8 gefunden. Eine weitere Betrachtung transient exprimierter TPC1 Kanalvarianten (M629I, M629T) auf Einzelkanalebene weist darauf hin, dass das Met629 des AtTPC1 in der Kanalporenregion entscheidend ist für die Einzelkanalleitfähigkeit.
Zusammengefasst zeigt der über die Membran von ouf Mutanten gemessene Strom eine Gemeinsamkeit: Alle zeigten keinen oder einen stark beeinträchtigten TPC1-vermittelten Kaliumausstrom aus der Vakuole ins Zytosol. Unter Berücksichtigung der beobachteten Abhängigkeit der vakuolären Membranspannung von der TPC1 Aktivität, scheint es, als ob die durch TPC1 angeregte Depolarisation der Vakuolenmembran, welche durch die vakuoläre Kaliumfreisetzung bedingt wird, in der Ausbildung des fou2 Phänotyps eine Rolle spielt. Daraus lässt sich ableiten, dass die TPC1-abhängige Depolarisation der Vakuolenmembran und die Jasmonat Bildung vermutlich verbunden sind. Diese Behauptung wird auch gestützt durch die komplette Wiederherstellung des WT-ähnlichen Pflanzenphänotyps und des Jasmonsäure Signalwegs in den ouf Mutanten. Letztendlich ist TPC1 als kontrollierendes Element der vakuolären Membranspannung wahrscheinlich dem Jasmonsäure Signalweg vorgeschaltet und deswegen ein perfekter Knotenpunkt, der verschiedene physiologische Stimuli und ihre Antworten verbindet.
Die Tumormikroumgebung (TME) spielt eine wichtige Rolle in Bezug auf das Ansprechen von Therapien, Tumorwachstum und die Bildung von Metastasen.
In den letzten Jahren konnte belegt werden, dass Tyrosinkinaseinhibitoren (TKIs) Einfluss auf Zellen des TME haben und vor allem die dort vorherrschenden Zellen, Tumor-assoziierte Makrophagen (TAM), durch die TKIs moduliert werden können. Sie entsprechen meist dem M2-Phänotyp, sezernieren antiinflammatorische Zytokine und sind protumoral, indem sie u.a. die Metastasierung und das Tumorwachstum unterstützen.
Zentrale Targets für die Reprogrammierung von Makrophagen stellen sowohl der NF-κB als auch die Inhibition des CSF1-Rezeptors dar. An diesen beiden Schlüsselstellen wirken u.a. TKIs.
In den durchgeführten Versuchen wurden drei TKIs verwendet – Dasatinib, Src-Kinase-Inhibitor-I, Bosutinib – um die Ergebnisse von Vorarbeiten zu verifizieren und um zu untersuchen, ob ein Klasseneffekt in Bezug auf eine gesteigerte IL-12-Produktion vorliegen könnte.
Ein wichtiger Ansatzpunkt in der Bekämpfung von Tumoren ist die Aktivierung von Immunzellen gegen Tumorzellen, in unserem Fall eine Modulation von TAM in Richtung M1-Makrophagen.
Eine signifikante Änderung des Phänotyps konnte nicht festgestellt werden.
Allerdings wurde eine gesteigerte IL-12-Produktion aller Makrophagensubtypen durch die Inkubation mit Dasatinib- bzw. Src-kinase-inhibitor-I oder Bosutinib gezeigt.
IL-12 ist ein wichtiges proinflammatorisches Schlüsselzytokin des Immunsystems, indem es u.a. NK-Zellen und T-Zellen aktiviert.
Die funktionellen Auswirkungen der verstärkten IL-12-Produktion in Hinblick auf NK-Zellen haben wir untersucht. Eine deutlich verstärkte Aktivierung anhand Aktvierungsmarker von NK-Zellen konnten wir nicht beweisen. Allerdings wurde eine erhöhte Zytotoxizität durch Ko-Kultivierung der NK-Zellen mit den unterschiedlichen Makrophagensubtypen und gleichzeitiger Inkubation mit Dasatinib demonstriert.
Die erhöhte IL-12-Produktion von APCs sowie verringerte IL-10-Produktion und der Einfluss auf andere Immunzellen, hier am Beispiel der NK-Zellen, zeigen u.a. das therapeutische Potential der TKIs als antineoplastisch wirksame Substanz.
Als alleinige Therapie ist deren Wirkungsbereich nach den hier vorliegenden Ergebnissen jedoch noch zu gering. In Kombination mit anderen Therapieoptionen stellen die TKIs allerdings ein mögliches Therapieregime dar. Der genaue Wirkmechanismus und die dadurch entstehenden Veränderungen sind noch genauer zu untersuchen. Ein weiteres Ziel ist in vitro etablierte Ergebnisse auch in die klinische Anwendung einfließen zu lassen.
Der zweite Teil der Arbeit befasste sich mit einem proinflammatorischen Zytokin IL-32γ und dessen Wirkung auf Makrophagen.
Wie bereits auch bei den TKIs wurde der Einfluss des Interleukins auf das Tumormikromilieu und die entsprechenden Auswirkungen untersucht. IL-32γ wirkt nicht nur selbst als proinflammatorisches Zytokin, sondern reguliert eine Vielzahl an weiteren Zytokinen. Der Einfluss von IL-32 auf das Tumormikromilieu und dessen Zellen stellt einen der zentralen Interessenspunkte dar.
In unseren Versuchen konnte unter IL-32γ eine effektivere Antigenpräsentation der Makrophagen – gemessen an einer verstärkten Expression von CD80 und CD86 – gezeigt werden.
Auf der anderen Seite wurde das antiinflammatorische Zytokin, IL-10, von IL-32γ-stimulierten Makrophagen ebenfalls verstärkt sezerniert.
Eine Ko-Kultivierung von Makrophagen und NK-Zellen und gleichzeitige Inkubation mit IL-32γ führte bei NK-Zellen zu einer verstärkten Aktivierung sowie zu einer erhöhten Zytotoxizität. Die Auswirkungen auf NK-Zellen deuten eine antitumorale Wirkung von IL-32γ an.
Das breite Wirkspektrum des Interleukins ist vielversprechend und könnte neue Therapiestrategien eröffnen, wofür allerdings weitere Versuche sowohl in vitro als auch in vivo notwendig sind, um das Interleukin und seine Wirkungen genauestens zu verstehen.
The human body is colonized by trillions of microbes from all three domains of life – eukaryotes, bacteria and archaea. The lower gastrointestinal tract is the most densely colonized part of the body, harbouring a diverse and dynamic community of microbes. While the importance of bacteria in this so-called microbiota is well acknowledged, the role of commensal fungi remains underexplored. The most prominent fungus of the human gastrointestinal microbiota is Candida albicans. This fungus occasionally causes life-threatening disseminated infections in individuals with debilitated immune defences. It is this “pathogenic” facet that has received the most attention from researchers in the past, leaving many aspects of its “commensal” lifestyle understudied. Using gnotobiotic mice as a model system to explore the biology of C. albicans in the mammalian gut, in this dissertation I establish the global response of the host to C. albicans monocolonization as well as the spatial distribution of the fungus in the intestine in the context of co-colonization with single gut bacterial species. The fungus elicited transcriptome changes in murine intestinal tissue, which included the activation of a reactive oxygen species-related defence mechanism and the induction of regulators of the circadian clock circuitry. Both responses have previously been described in the context of a complete bacterial microbiota. Imaging the intestine of animals monocolonized with the fungus or co-colonized with C. albicans and the gut bacteria Bacteroides thetaiotaomicron or Lactobacillus reuteri revealed that the fungus was embedded in a B. thetaiotaomicron-promoted outer mucus layer in the murine colon. The gel-like outer mucus constitutes a unique microhabitat, distinct in microbial composition from the adjacent intestinal lumen. This finding indicates that bacteria can shape the specific microhabitat occupied by the fungus in the intestine. Overall, the results described in this dissertation suggest that gnotobiotic mice constitute a valuable tool to dissect multiple aspects of the interactions among host, commensal fungi and cohabiting bacteria.
Es erfolgte eine elektrochemische Abscheidung von Bruschitschichten auf Titan in Gegenwart von Kristallisationsinhibitoren. Dabei wurden die Kristallisationsinhibitoren Zitronensäure, treta-Natriumdiphosphat-Decahydrat und Phytinsäure verwendet und die entstandenen Schichten mit denen ohne Inhibitorzugabe verglichen. Um das Ausmaß der Inhibierung zu verifizieren, wurde die Masse aller Schichten gemessen, welche für die Inhibition mit Zitronensäure und Phytinsäure abnahm und für Natriumdiphosphat zunahm. Die kristallographische Zusammensetzung der mit und ohne Inhibierung abgeschiedenen Schichten wurde mit Hilfe der Röntgendiffraktometrie bestimmt und zeigte, dass sich reine Bruschitschichten mit unterschiedlichem amorphem Anteil abschieden. Die daraus entstandenen Werte lieferten zugleich die Informationen über die einzelnen Kristallitgrößen innerhalb der Schichten. Über den Einfluss der Inhibitoren auf die Schichtmorphologie gaben rasterelektronenmikroskopische Aufnahmen weiteren Aufschluss. Die Inhibition verursachte teils mit Rissen durchzogene Schichten, deren Kristallformationen sich von Standardelektrolyt unterschieden. Ausgewählte Proben wurden unter verschiedenen Bedingungen desinfiziert bzw. sterilisiert und nachfolgend erneut gewogen und mittels Röntgendiffraktogrammetrie und Rasterelektronenmikroskopie analysiert. Nach der Desinfektion entstanden reine Bruschitschichten, die an Masse verloren aber trotzdem die typischen Kristallformationen zeigten. Die Sterilisation führte zur Umwandlung von Bruschit in Monetit und Hydroxylapatit. Des Weiteren wurde die biologische Reaktion der Schichten auf humane fötale Osteoblasten-Zelllinien zur Überprüfung der Zellverträglichkeit ermittelt. Die entstandenen Ergebnisse waren nicht verwertbar und enthielt sehr hohe Standardabweichungen.
Die zunehmende Antibiotikaresistenz vieler Krankheitserreger ist ein weltweites Problem, welches zu einem klinischen Bedarf an neuen antimikrobiellen Substanzen führt. Sphingolipide einschließlich Ceramide stellen eine vielfältige Gruppe strukturverwandter Lipide dar und bestehen aus einem Sphingosin-Grundgerüst, welches mit einer Fettsäure verbunden ist. Sowohl das Sphingosin-Grundgerüst allein als auch Sphingolipide zeigen eine antibakterielle Wirkung gegenüber einer Vielzahl pathogener Mikroorganismen. Die Intensität der Hemmung hängt von der Sphingolipidstruktur und dem Mikroorganismus ab. Neuere Studien konnten zeigen, dass Sphingosin, Ceramide und Ceramid-Analoga in N. meningitidis aufgenommen werden und eine bakteriostatische oder bakterizide Wirkung zeigen. Jedoch ist die antibakterielle Wirkungsweise noch nicht genau bekannt. Um mehr über den Wirkmechanismus zu erfahren haben wir die ultrastrukturellen Veränderungen von N. meningitidis nach Inkubation mit azido-funktionalisierten Sphingolipiden mit elektronenmikroskopischen Verfahren (transmissionselektronenmikroskopische und rasterelektronenmikroskopische Aufnahmen) untersucht. Mittels korrelativer Licht- und Elektronenmikroskopie (CLEM) konnten wir die azido-funktionalisierten Sphingolipide nach Aufnahme in N. meningitidis lokalisieren. Zum Anfärben der funktionalisierten Sphingolipide wurde die kupferfreie Azid-Alkin-Cyccloaddition verwendet.
Intestinale Infektionen mit pathogenen Protozoen können schwere
Erkrankungsbilder verursachen und stellen eine oftmals vernachlässigte
Problematik, vor allem in Regionen mit geringen Sanitär- und Hygienestandards,
dar. Dies betrifft insbesondere südlich der Sahara gelegene Länder. Hier
existieren leider keine oder nur unzureichende Prävalenzdaten, was zum Teil auch auf die inadäquaten diagnostischen Möglichkeiten zurückzuführen ist. Die
Übertragung erfolgt fäkal-oral über verunreinigte Nahrungsmittel sowie
kontaminiertes Trinkwasser und kann symptomatische und asymptomatische,
akute und chronische Infektionen hervorrufen.
Methoden: Es handelt sich um eine Querschnittsstudie, die 2016 im Rahmen des laufenden SchistoControl-Pilotprojekts auf der Insel Ijinga im Nordwesten Tansanias durchgeführt wurde. Es wurden Stuhlproben von 357 Teilnehmern gesammelt und mikroskopisch auf Trophozoiten oder Zysten von intestinalen Protozoen untersucht. Zusätzlich wurde die Realtime-Polymerasekettenreaktion (qPCR) zur Speziesdifferenzierung verwendet.
Ergebnisse: Basierend auf Mikroskopie und qPCR betrug die Prävalenz von Giardia intestinalis 12% bzw. 15,1%. Basierend auf der Mikroskopie betrug die Prävalenz von Entamoeba histolytica / Entamoeba dispar 26,1%. Durch Speziesdifferenzierung mittels qPCR waren jedoch 21,8% der Studienteilnehmer positiv für E. dispar und keiner positiv für E. histolytica.
Schlussfolgerung: Intestinale Protozoeninfektionen sind in der Bevölkerung des Studienortes häufig. Der Nachweis dieser Infektionen in verschiedenen Altersgruppen weist auf einen geringen Hygienestandard in der Gemeinde hin. Eine Verbesserung der Wasser-, Sanitär-, Hygienestruktur und des Gesundheitsbewusstseins wird zur Kontrolle dieser Infektionen beitragen.
Im metastasierten Melanom sind bei Vorhandensein einer BRAF V600 Mutation zielgerichtete Therapien mit BRAF+MEK-Inhibitoren sowie Immuntherapien (ICB), die Immuncheckpoints wie PD-1 blockieren, zugelassen. Aktuell gibt es keine evidenzbasierte Empfehlung welche Therapie in der Erstlinie im BRAF V600 mutierten Melanom eingesetzt werden sollte. Bis jetzt wurde der Stellenwert PD-1 basierter Immuncheckpoint Blockade in der Zweitlinie nach Progress unter BRAF+MEK-Inhibition nicht beschrieben. Es ist auch unklar, ob die Kombinations-ICB (PD-1 plus CTLA-4 Blockade) mit einer Verbesserung des Ansprechens und Überlebens gegenüber einer PD-1 Monotherapie assoziiert ist, wie für das therapie-naive Melanom beschrieben. Wir haben eine retrospektive, multizentrische Studie durchgeführt um die Wirksamkeit von PD-1 basierten Immuntherapien nach Progress unter zielgerichteter Therapie zu explorieren. In unserer Untersuchung zeigten PD-1 Monotherapie und die kombinierte PD-1 plus CTLA-4 Blockade eine ähnliche Wirksamkeit in Patienten mit BRAFi+MEKi-Resistenz. Die Kombinationstherapie war dagegen mit einem deutlich höheren Risiko für schwerwiegende immunvermittelte Nebenwirkungen im Vergleich zu PD-1 Monotherapie assoziiert. Unsere Daten indizieren, dass eine PD-1 Blockade einer Kombinations-ICB in der Zweitlinie nach Progress unter zielgerichteter Therapie im fortgeschrittenen BRAF V600 mutierten Melanom vorzuziehen ist.
Das Schädel-Hirn-Trauma (SHT) ist ein großes medizinisches Problem. Das Hirnödem mit konsekutiv erhöhten intrakraniellen Drücken ist eine häufige und schwerwiegende Komplikation des schweren SHT. Es ist der signifikanteste Prädiktor für ein schlechtes Outcome. Obwohl Glukokortikoide (GK) die Ausbildung eines Hirnödems bei neuroinflammatorischen Erkrankungen und manchen Hirntumoren reduzieren können, ist diese Substanzklasse im SHT ineffektiv oder sogar schädlich.
Nach controlled cortical impact (CCI) in Mäusen, einem etablierten SHT-Modell in-vivo, zeigte sich ein Zusammenbruch der Blut-Hirn-Schranke (BHS). Des Weiteren wurde der BHS-stabilisierende Effekt der GK nach CCI durch proteasomalen Abbau des Glukokortikoidrezeptors (GR) behindert. Eine Inhibierung des Proteasoms durch den Proteasomeninhibitor Bortezomib zusammen mit einer GK-Therapie mit Dexamethason reduzierte den Abbau des GR und sorgte für eine Restitution der BHS-Integrität.
Unglücklicherweise ließen sich diese Ergebnisse in-vitro nicht auf in Sauerstoff-/Glukosemangel-Modell in der humanen Hirnendothelzelllinie hCMEC/D3 übertragen. Daher konnte für die Kombinationstherapie aus dem Proteasomeninhibitor Bortezomib und Dexamethason kein positiver Effekt gezeigt werden.
Acne inversa (AI), in der englischen Literatur auch als Hidradenitis suppurativa (HS) bezeichnet, ist eine chronische, entzündliche und schmerzhafte Hautkrankheit, die re- zidivierende Knoten, Fisteln, Abszesse und Vernarbungen vor allem in den Intertrigines verursacht. Es zeigen sich bei Betroffenen neben schwerwiegenden somatischen, auch psychologische Komorbiditäten. Das Ziel der vorliegenden wissenschaftlichen Arbeit war es, an einem an AI erkrankten Patientenkollektiv prospektiv systematisch Komor- biditäten, Lebensqualität und psychische Verfassung zu analysieren und auf Korrelatio- nen zu untersuchen, um daraus Handlungsempfehlungen abzuleiten. Mittels dermatolo- gischer und psychologischer Fragebögen mit Fragebögen zur Lebensqualität wurden pseudonymisierte Daten von 110 Studienteilnehmern gewonnen, statistisch aufbereitet und ausgewertet. Es konnte eine statistisch signifikante Korrelation der Visuellen Ana- logskala Schmerz (VAS-Schmerz) mit dem Hospitality Anxiety Depression Scale (HADS) bzw. dem Skindex-29 aufgezeigt werden. Der Zeitraum zwischen Erstsymp- tomen und Diagnosestellung der AI erfolgte im Median nach 6 Jahren. Weiterhin erfuh- ren AI-Patienten häufig eine nicht leitliniengerechte Therapie und zeigten psychische Belastungen anhand von Schlafstörungen, besonderen Stresssituationen und eine damit einhergehende Verschlechterung der AI. Body Mass Index (BMI), „Waist-to-hip-ratio“ und Bluthochdruck waren oftmals erhöht. Die VAS-Schmerz-Skala könnte ergänzend als Instrument zur ersten Quantifizierung der Krankheitsschwere angewandt werden. Internisten, Chirurgen, Allgemeinmediziner, Gynäkologen und Urologen sollten mit der Dermatose AI besser vertraut gemacht werden, um den Patienten eine schnellere, leitli- niengerechte Therapie zukommen zu lassen. AI-Patienten benötigen neben der dermato- logischen eventuell eine psychologische bzw. psychiatrische Therapie, um das Stressni- veau zu senken, was sich auf die Lebensqualität positiv auswirken könnte.
Die nicht-invasive Gefäßdiagnostik stellt einen wichtigen Pfeiler in der Prävention von Herz-Kreislauferkrankungen dar. Während lange Zeit die sonographische Messung der cIMT, als morphologisches Korrelat der Gefäßalterung, als Goldstandard galt, ist in den letzten Jahren in Gestalt der Pulswellenanalyse/PWV-Messung eine Technik weiterentwickelt worden, die, als funktionelles Korrelat der Gefäßalterung, aufgrund der leichteren Durchführbarkeit und geringerer Untersucherabhängigkeit und Kosten vielversprechend ist. So erlaubt die Messung der Pulswelle mittels gewöhnlicher Blutdruckmanschetten, genau wie die cIMT, die Berechnung des individuellen Gefäßalters und die Diagnostik für das Vorliegen eines Endorganschadens der Blutgefäße.
Um die Messergebnisse der beiden Untersuchungen miteinander zu vergleichen, wurden beide in der EUROASPIRE-IV Studie an Patienten mit koronarer Herzkrankheit durchgeführt. Die Auswertung der Messergebnisse der mit dem Vascular Explorer durchgeführten Pulswellenanalyse/PWV-Messung ergab überraschenderweise, dass die Mehrheit der herzkranken Patienten weder eine vaskuläre Voralterung noch einen Endorganschaden der Blutgefäße aufweisen. Im Falle der cIMT-Messung war Gegenteiliges der Fall, was trotz der medikamentösen Therapie der Patienten so zu erwarten war. Weiterhin zeigte sich lediglich eine geringe Korrelation zwischen den Messergebnissen beider Untersuchungen. Die Determinanten der einzelnen Messwerte aus cIMT und Pulswellenanalyse/PWV-Messung waren deckungsgleich mit den in der Literatur beschriebenen Faktoren, wenn auch viele der sonst signifikanten Regressoren das Signifikanzniveau in unserer Auswertung nicht unterschritten.
Eine Limitation der funktionellen Gefäßdiagnostik liegt derzeit darin, dass die Messergebnisse stark von dem verwendeten Messgerät abhängen. Es liegen noch zu wenig Vergleichsstudien vor, um die Messergebnisse, speziell von neueren Geräten wie dem Vascular Explorer, auf andere zu übertragen. Bei der Berechnung des Gefäßalters sollten daher optimalerweise gerätespezifische Normwerte vorliegen, was beim Vascular Explorer nicht der Fall ist. Gleiches gilt für die Verwendung des PWVcf-Grenzwerts für die Diagnose eines Endorganschadens der Blutgefäße.
Analog hat auch die Messung der cIMT gewisse Einschränkungen. So wäre eine weitere Standardisierung der Messorte (A. carotis communis vs Bulbus vs A. carotis interna), zwischen denen sich die durchschnittliche cIMT erheblich unterscheidet, sowie der Messparameter (Minimal- vs Maximal- vs Mittelwert) wünschenswert. Die universelle Anwendung eines cIMT-Grenzwerts zur Diagnose eines Endorganschadens der Blutgefäße ist daher kritisch zu sehen. Dies zeigt sich auch darin, dass in den neuesten Leitlinien der bislang geltende Grenzwert angezweifelt und kein aktuell gültiger Grenzwert mehr genannt wird.
Wir interpretieren unsere Ergebnisse dahingehend, dass unsere Messung der cIMT die zu erwartende pathologische Gefäßalterung bei Patienten mit koronarer Herzkrankheit besser widerspiegelt als die Messung der Pulswelle mit dem Vascular Explorer. Welche der beiden Untersuchungen hinsichtlich der prognostischen Wertigkeit überlegen ist, muss im Rahmen von Längsschnittstudien geklärt werden.
Ebenso wie Tiere verfügen Pflanzen über die Fähigkeit elektrische Signale zu generieren. Dabei repräsentieren elektrische Signale – Membranpotentialänderungen an der Plasmamembran – die frühesten Antworten, welche an Pflanzenzellen im Zuge veränderter externer und intrinsischer Bedingungen beobachtet werden können. Stimuli wie Kälte, Hitze, Verwundung, Herbivorie und Pathogene, aber auch physiologische Prozesse, wie Wachstum und Bestäubung führen zur Änderung des Potentials der Plasmamembran pflanzlicher Zellen. Die meisten dieser Membranpotentialänderungen bestehen aus einer schnellen Depolarisation, gefolgt von einer Repolarisation des Membranpotentials, deren Kinetik, in Abhängigkeit des Stimulus hoch variabel sein kann. Das Wissen über die molekularen Grundlagen der Generierung und Weiterleitung elektrischer Signale in Pflanzen ist im Gegensatz zu Tieren nur wenig verstanden. Eine Ausnahme stellen ‚klassisch-erregbare‘ Pflanzen wie die Venusfliegenfalle oder die Mimose dar. In diesen Pflanzen führt ein Berührungsreiz zur Auslösung eines charakteristischen Aktionspotentials, welches in der Folge zu einer, auf differentiellen Turgoränderungen basierenden, nastischen Bewegung führt. In allen anderen Pflanzen ist die Kinetik der Membranpotentialänderungen sehr variabel, abhängig vom Stimulus und dem physiologischen Zustand der Zellen und – mit Ausnahme der Reaktion auf einen Kältestimulus – lediglich nach langen Latenzzeiten wiederholbar. Dieser Umstand verhindert eine systematische Analyse der molekularen Basis elektrischer Signale in den meisten Pflanzen. Ziel dieser Arbeit war es daher, auf der Basis des Channelrhodopsin-2 (ChR2) aus der Grünalge Chlamydomonas reinhardtii, welches bereits seit 2005 in der Neurobiologie genutzt wird, ein nicht-invasives Werkzeug zur funktionellen Analyse elektrischer Signale in Pflanzen zu etablieren. ChR2 ist ein Blaulicht-aktivierter Kationenkanal, der für seine Funktion all trans-Retinal als Cofaktor benötigt. Im Rahmen dieser Arbeit wurden verschiedene Varianten des ChR2, mit einem Schwerpunkt auf ChR2-C128T und vor allem ChR2-D156C, auch bekannt als ChR2-XXL eingesetzt. ChR2 konnte bereits durch M. Baumann im Rahmen ihrer Dissertation funktionell im transienten Expressionssystem Nicotiana benthamiana dargestellt werden. In der vorliegenden Arbeit wurde das System weiter ausgebaut und die besonders aussichtsreichen ChR2-Varianten nicht nur in N. benthamiana, sondern auch in stabilen Arabidopsis thaliana Linien funktionell charakterisiert. Dabei konnte mit dem ChR2-XXL ein geeignetes optogenetisches Werkzeug zur Untersuchung elektrischer Signale in Pflanzen identifiziert werden. ChR2-XXL bietet die Möglichkeit das Membranpotential durch kurze, 5 s Blaulichtpulse im Mittel um 95 mV zu depolarisieren und im Anschluss die Repolarisationsphase zu untersuchen. Blaulicht-induzierbare, ChR2-XXL-vermittelte Depolarisationen konnten, reproduzierbar und beliebig oft an den gleichen Zellen wiederholt ausgelöst werden. Dadurch ermöglicht ChR2-XXL die bisher nur unzureichend bekannten molekularen Komponenten der Repolarisation des Membranpotentials in Pflanzen zu erforschen. In tierischen Zellen generieren spannungsabhängige Natriumkanäle die Depolarisation, während spannungsabhängige Kaliumkanäle die Depolarisationskinetik bestimmen. Die im Vergleich zu tierischen Zellen veränderten Ionengradienten lassen vermuten, dass die pflanzliche Depolarisation im Wesentlichen durch Ca2+-abhängige Anionenkanäle vermittelt wird, die durch den Efflux von Cl- das Membranpotential depolarisieren. Für die Repolarisation wird zum einen die Beteiligung von auswärtsgleichrichtenden Kaliumkanälen postuliert. Zum anderen wird auch eine Beteiligung der Plasmamembran (PM) H+-ATPasen vermutet, welche gleichzeitig einen essentiellen Beitrag zur Generierung des Ruhepotentials leisten. In der vorliegenden Arbeit wurde es durch den Einsatz von ChR2-XXL möglich, beide potentiellen Komponenten der Repolarisationsphase, Kaliumkanäle und PM H+-ATPasen, erstmals durch eine nicht-invasive, Anionen-unabhängige Methode der Depolarisation zu untersuchen. Durch den Einsatz von Mutanten und Kaliumkanalinhibitoren konnte ein möglicher Beitrag des auswärtsgleichrichtenden Kaliumkanals Arabidopsis thaliana GUARD CELL OUTWARD RECTIFYING K+ CHANNEL (AtGORK) an der Repolarisationsphase in Arabidopsis Mesophyllzellen nahezu ausgeschlossen werden. Der auswärtsgleichrichtende Kaliumkanal GORK öffnet erst bei Membranpotentialen positiv vom Gleichgewichtspotential für Kaliumionen (EK (-118 mV)). Da die ChR2-induzierbaren Depolarisationen ebenso wie viele natürliche Stimuli, diesen Wert kaum erreichen oder nur geringfügig überschreiten, leistet der GORK einen geringfügigen Beitrag bei der Repolarisation. Dies ließ vermuten, dass die Repolarisation von EK bis zum Ruhepotential bei ca. -180 mV dagegen möglicherweise durch die PM H+-ATPasen bewerkstelligt wird. Die Wirkung des PM H+-ATPase Inhibitors Natriumorthovanadat, sowie des PM H+-ATPase Aktivators Fusicoccin auf die Repolarisationsphase konnten diese Hypothese unterstützen. Die Hemmung der PM H+-ATPasen verlangsamte die Repolarisationskinetik während eine Aktivierung der PM H+-ATPasen diese beschleunigte. So wurde es erstmals möglich den genauen Einfluss der PM H+-ATPasen auf Wiederherstellung des Membranpotentials während der Repolarisation in Mesophyllzellen zu studieren. Darüber hinaus wurde beobachtet, dass in Gegenwart des Kaliumkanalblockers Ba2+ die Repolarisation ebenfalls beschleunigt werden konnte. In Übereinstimmung mit dem ‚Pump-and-Leak‘-Modell (Alberts et al. 2002) deutet dies darauf hin, dass schwach einwärtsgleichrichtende Kaliumkanäle, wie der ARABIDOPSIS K+ TRANSPORTER 2 (AKT2) dem PM H+-ATPasen Protonengradienten entgegenwirken und somit das Ruhepotential aus der Summe der bewegten Ladungen von Pumpen und Kaliumkanälen bestimmt wird. Das mögliche Potenzial optogenetischer, Rhodopsin-basierter Werkzeuge für die molekulare Analyse elektrischer Signale, insbesondere unter Einsatz der breiten Palette lichtgesteuerter Pumpen und Kanäle, ihrer spektralen Diversität und ihrer Einkreuzung in ausgewählte Arabidopsis Mutanten wird diskutiert.
ADMM-Type Methods for Optimization and Generalized Nash Equilibrium Problems in Hilbert Spaces

This thesis is concerned with a certain class of algorithms for the solution of constrained optimization problems and generalized Nash equilibrium problems in Hilbert spaces. This class of algorithms is inspired by the alternating direction method of multipliers (ADMM) and eliminates the constraints using an augmented Lagrangian approach. The alternating direction method consists of splitting the augmented Lagrangian subproblem into smaller and more easily manageable parts.
Before the algorithms are discussed, a substantial amount of background material, including the theory of Banach and Hilbert spaces, fixed-point iterations as well as convex and monotone set-valued analysis, is presented. Thereafter, certain optimization problems and generalized Nash equilibrium problems are reformulated and analyzed using variational inequalities and set-valued mappings. The analysis of the algorithms developed in the course of this thesis is rooted in these reformulations as variational inequalities and set-valued mappings.
The first algorithms discussed and analyzed are one weakly and one strongly convergent ADMM-type algorithm for convex, linearly constrained optimization. By equipping the associated Hilbert space with the correct weighted scalar product, the analysis of these two methods is accomplished using the proximal point method and the Halpern method.
The rest of the thesis is concerned with the development and analysis of ADMM-type algorithms for generalized Nash equilibrium problems that jointly share a linear equality constraint. The first class of these algorithms is completely parallelizable and uses a forward-backward idea for the analysis, whereas the second class of algorithms can be interpreted as a direct extension of the classical ADMM-method to generalized Nash equilibrium problems.
At the end of this thesis, the numerical behavior of the discussed algorithms is demonstrated on a collection of examples.
Eine retrospektive Studie anhand von Patientenunterlagen zum Blutverlust, Transfusionshäufigkeit und postoperativen Komplikationen bei Knie-Endo-Prothesen mit systemischer Gabe von 500 mg Tranexamsäure kurz vor Eröffnung der Blutleere! 229 Patienten in der Studiengruppe, von denen 9 eine erlaubte Wiederholungsdosis erhalten hatten, wurden verglichen mit 231 Patienten, die keine Tranexamsäure erhalten hatten. 3 Patienten der Studiengruppe gegenüber 14 Patienten der Kontrollgruppe benötigten eine Bluttransfusion. In der Studiengruppe war der niedrigste postoperative Hb bei 71,2% der Patienten 10 g/dl oder höher. In der Kontrollgruppe war es nur bei 49,4% der Patienten der Fall. Durch die Gabe von Tranexamsäure konnte der durchschnittliche, berechnete Blutverlust bei Männern von etwa 1850 ml auf 1450 ml und bei Frauen von etwa 1300 ml auf etwa 1000 ml gesenkt werden. Die Unterschiede waren signifikant! Im beobachteten Kollektiv konnte keine Steigerung der Rate an Thrombosen oder thrombembolischen Ereignissen festgestellt werden. Das Antifibrinolytikum Tranexamsäure ist in dieser niedrigen Dosierung effektiv und scheint sicher zu sein.
In dieser Arbeit wurde die Herzratenvariabilität (HRV), das Interbeat-Intervall (IBI) sowie die Beziehung zwischen HRV und IBI bei gesunden Probanden, bradykarden Patienten mit einer Mutation im hyperpolarization-activated cyclic nucleotide-gated channel 4-(HCN4) Gen sowie Patienten mit einer Anorexie nervosa (AN) untersucht. Die Haupthypothese lautete, dass die bei Patienten mit AN oft zu beobachtenden Bradykardien durch neurale Mechanismen verursacht werden. Daher wurde angenommen, dass ein Sättigungseffekt der HRV/IBI Beziehung infolge einer anhaltenden parasympathischen Kontrolle des Sinusknotenareals lediglich bei Patienten mit einer AN nachzuweisen ist. In dieser Arbeit konnte ein Sättigungseffekt der HRV/IBI-Beziehung bei Patienten mit einer Anorexia nervosa sowie bei zwei von vier Patienten mit einer HCN4-Mutation nachgewiesen werden. Bei Kontrollen konnten hingegen kaum Zeichen eines Sättigungseffekts der HRV/IBI Beziehung nachgewiesen werden. Es wurde daher geschlussfolgert, dass ein Sättigungseffekt der HRV/IBI Beziehung keine parasympathisch-vermittelte Bradykardie beweist. Es zeigt sich vielmehr, dass die Regulation des HCN4-Gens für die Bradykardie und den festgestellten HRV-Sättigungseffekt bei Patienten mit einer AN verantwortlich sein könnte.
Intuitive Benutzung wird in dieser Arbeit definiert als das Ausmaß, mit dem ein Produkt mental effizient und effektiv genutzt wird, was mit einem starken metakognitiven Gefühl von Flüssigkeit einhergeht. Aktuelle Methoden verfügen nicht über eine ausreichend hohe zeitliche Anwendungseffizienz, um im Industrieprojekt 3D-GUIde effektiv zur Evaluation von Interaktionspatterns für 3D-Creation-Oriented-User-Interfaces (3D-CUIs) eingesetzt werden zu können. Diese Interaktionspatterns beschreiben strukturiert, wie 3D-CUIs als User Interfaces zur Erstellung von dreidimensionalen Inhalten gestaltet werden müssen, um intuitive Benutzung zu unterstützen. In dieser Arbeit werden daher zwei neue Evaluationsmethoden vorgeschlagen: 1) IntuiBeat-F als formative Evaluationsmethode und 2) IntuiBeat-S als summative Evaluationsmethode. Basierend auf Default-Interventionist-Theorien und bestehenden Definitionen intuitiver Benutzung werden die mentale Beanspruchung als zentrales objektives, das metakognitive Gefühl von Flüssigkeit als zentrales subjektives und die Effektivität als zentrales pragmatisches mit intuitiver Benutzung assoziiertes Merkmal identifiziert. Die Evaluation intuitiver Benutzung mithilfe von IntuiBeat-F und IntuiBeat-S ist vielversprechend, da es sich bei beiden Methoden um Inhibition basierende Rhythmuszweitaufgaben handelt und diese somit mentale Beanspruchung objektiv erfassen können. Das Potential beider Methoden wird im Hinblick auf vorherige Forschungsarbeiten zur zeitlich effizienten Evaluation von 3D-CUIs aus der Mensch-Computer-Interaktion und der Psychologie diskutiert. Aus dieser Diskussion werden empirische Forschungsfragen abgeleitet. Die erste Forschungsfrage untersucht die wissenschaftliche Güte von IntuiBeat-S. Im ersten, zweiten und dritten Experiment werden Paare von 3D-CUIs miteinander summativ verglichen (d.h. weniger vs. stärker intuitiv benutzbare User Interfaces). Dabei wird die wissenschaftliche Güte von IntuiBeat-S hinsichtlich der Hauptgütekriterien Objektivität, Reliabilität und Validität beurteilt. Die Ergebnisse zeigen, dass IntuiBeat-S eine hohe wissenschaftliche Güte bei der summativen Evaluation besitzt. Zudem macht es bei der Anwendung von IntuiBeat-S keinen Unterschied, ob der Rhythmus über die Ferse oder den Fußballen eingeben wird, und ob als Stichproben Studierende mit höherer oder geringerer Vorerfahrung bezüglich der Nutzung von 3D-CUIs verwendet werden. Die zweite Forschungsfrage untersucht die wissenschaftliche Güte von IntuiBeat-F. Im vierten, fünften, sechsten und siebten Experiment werden 3D-CUIs einzeln formativ evaluiert (d.h. entweder ein weniger oder stärker intuitiv benutzbares User Interface). Dabei wird die wissenschaftliche Güte von IntuiBeat-F hinsichtlich der Hauptgütekriterien Gründlichkeit, Gültigkeit und Zuverlässigkeit beurteilt. Die Ergebnisse zeigen, dass IntuiBeat-F eine hohe wissenschaftliche Güte bei der formativen Evaluation besitzt. Diese liegt bei strikter Anwendung der Methode (d.h. Berücksichtigung ausschließlich mit der Methode entdeckter Nutzungsprobleme) zwar höher, ist aber bei wenig strikter Anwendung der Methode (d.h. Berücksichtigung auch unabhängig von der Methode entdeckter Nutzungsprobleme) noch ausreichend hoch. Jedoch konnte erst die Entwicklung und Einführung einer zusätzlichen Analysesoftware im Zuge des sechsten und siebten Experiments die wissenschaftliche Güte von IntuiBeat-F hinsichtlich aller drei Hauptgütekriterien demonstrieren, da ohne deren Unterstützung IntuiBeat-F vom Evaluator nicht ausreichend gründlich angewendet wird. Die dritte Forschungsfrage untersucht, wie hoch die zeitliche Anwendungseffizienz beider Methoden als wichtiger Aspekt praktischer Güte im Vergleich zu bereits vorhandenen Evaluationsmethoden für intuitive Benutzung ist. Bezüglich der summativen Evaluation wird im zweiten Experiment eine höhere zeitliche Anwendungseffizienz von IntuiBeat-S im Vergleich zum aktuellen summativen Benchmark, der CHAI-Methode, sowohl bei der Evaluation von weniger als auch bei der von stärker intuitiv benutzbaren 3D-CUIs demonstriert. Auch bezüglich der formativen Evaluation konnten die Ergebnisse der letzten vier Experimente zeigen, dass die zeitliche Anwendungseffizienz von IntuiBeat-F im Vergleich zum aktuellen formativen Benchmark, dem Nutzertest mit retrospektivem Think-Aloud- Protokoll, sowohl bei der Evaluation von weniger als auch stärker intuitiv benutzbaren 3D-CUIs höher liegt. Dieser Unterschied bleibt bestehend, egal ob eine zusätzliche Analysesoftware vom Evaluator verwendet wird oder nicht. Als Ergebnis aller Experimente lässt sich feststellen, dass die wissenschaftliche Güte und die zeitliche Anwendungseffizienz beider Methoden zur Evaluation intuitiver Benutzung von 3D-CUIs mehr als zufriedenstellend beurteilt werden kann. Die Arbeit wird mit einer Diskussion des geleisteten Forschungsbeitrags geschlossen. Dabei werden Anregungen für künftige Forschung aus theoretischer (z.B. Berücksichtigung des Gefühls von Flüssigkeit bei der Evaluation), praktischer (z.B. Untersuchung der Anwendbarkeit beider Methoden in anderen Domänen) und methodischer (z.B. Beurteilung der praktischen Güte beider Methoden anhand anderer Kriterien) Perspektive gegeben.
Angsterkrankungen stellen einen großen Anteil an psychischen Erkrankungen dar und gehen zum Teil mit großem Leidensdruck einher. Da die leitliniengerechte Therapie mit hohen Rückfallraten und ca. 25% Nonrespondern einhergeht, stellt sich die Frage nach alternativen Behandlungsmethoden. Transkranielle Magnetstimulation findet als nichtinvasive Behanslungsmethode zunehmend Anwendung bei neurologischen und psychiatrischen Erkrankungen.
In der vorliegenden randomisierten, kontrollierten Studie wurde die Wirkung der TMS auf den frontotemporalen (FTC) und dorsolateralen präfrontalen Cortex (dlPFC) untersucht. Dazu wurden 42 gesunde Probanden zwischen 18 und 59 Jahren zur Hälfte TMS-stimuliert, die andere Hälfte wurde scheinstimuliert. Vor und nach Stimulation bzw. Placebostimulation wurde die Aktivität von FTC und dlPFC mit Nah-Infrarotspektroskopie (NIRS) während der Durchführung des Verbal Fluency Tasks (VFT) gemessen.
In dieser Studie konnte keine Veränderung der hämodynamischen Gehirnaktivität durch TMS nachgewiesen werden, jedoch äußerten die Probanden der Stimulationsgruppe im Gegensatz zu den Probanden der Placebogruppe, Nebenwirkungen wie Schmerzen oder Muskelzucken verspürt zu haben.
Die während des VFT laufende NIRS zeigte eine signifikant höhere Durchblutung und damit Aktivierung des linken FTC im Seitenvergleich und eine signifikant höhere Aktivierung während der semantischen als bei der phonemischen VFT-Bedingung, analog zu früheren, vergleichbaren Untersuchungen.
Die Frage, ob sich TMS als mögliche Behandlungsmethode bei Angsterkrankungen eignet, lässt sich anhand der hier vorliegenden Studie nicht abschließend beantworten.
Die vorliegende Arbeit befasst sich mit der Synthese, Charakterisierung und Reaktivität von Nebengruppen-Metallkomplexen, die mindestens einen pi-koordinierenden Liganden tragen. Im ersten Abschnitt liegt der Fokus auf heteroleptischen Systemen mit carbocyclischen Liganden, zu deren Synthese die gängige Methodik der Salzeliminierung herangezogen wird. Das Metallierungsverhalten dieser Komplexe, sowie die Reaktivität von Komplexen mit reduktionsstabilen funktionellen Gruppen an den Ligandensystemen wird untersucht. Der zweite Abschnitt behandelt die Redox- und Koordinationseigenschaften des CAAC-stabilisierten 1,4 Diborabenzols, wobei Alkali-Metalle, Gruppe 10 Metalle, Lanthanoide, sowie die Actinoide Thorium und Uran untersucht werden.
In dieser Arbeit wurden die Signalwege an Myokard, Leber und Skelettmuskulatur untersucht, die den Veränderungen des Glucosestoffwechsels im zeitlichen Verlauf bei TAC-induzierter systolischer Herzinsuffizienz zu Grunde liegen können.
Es wurde dafür über eine TAC-Operation am Mausmodell eine Herzinsuffizienz induziert. Erniedrigte Nüchtern-Blutzuckerwerte und ein erhöhter myokardialer Glucoseverbrauch fanden sich als die wesentlichen metabolischen Veränderungen in diesem Mausmodell mit TAC-induzierter Herzinsuffizienz. In Vorarbeiten ergaben die GTTs nach zwei Wochen eine erhöhte Glucoseaufnahme nach TAC, die nicht durch eine verstärkte Insulinsignaltransduktion hervorgerufen wurde, da sich die Insulinkonzentrationen und die Insulinsensitivität zwischen den Gruppen nicht unterschieden. Der Nüchternblutzuckers war bei den TAC-operierten Tieren niedriger als bei den Sham-operierten. In der PET-Bildgebung wurde eine erhöhte Glucoseaufnahme im Herzen bei den TAC-Tieren verglichen mit den Sham-operierten Tieren gezeigt.
Im Rahmen dieser Arbeit wurden nun nach einem bzw. sechs Monaten bei den Versuchsgruppen Western Blot Analysen sowie Glykogenassays durchgeführt. Zudem wurde untersucht, wie sich eine zusätzliche Sucrosefütterung über sechs Monate ab OP auswirkt.
Dabei fand sich eine erhöhte Expression des basalen Glucosetransporters GLUT1 am Myokard, passend zur erhöhten basalen Glucoseaufnahme im PET. Die Expression des insulinabhängigen Transporters GLUT4 hingegen war - wie bereits in anderen Arbeiten zu Hypertrophie und Herzinsuffizienz - vermindert.
Der hepatische und muskuläre Glykogengehalts war nach TAC reduziert, wobei die Depletion der hepatischen Speicher bereits nach einem Monat, im Muskel erst nach sechs Monaten signifikant war.
Die Regulation der muskulären Glykogenreserven wurde in unserem Modell möglicherweise über Akt und Hexokinase II vermittelt. Einen Monat nach TAC war am Skelettmuskel eine vorübergehende Reduktion der Akt-Phosphorylierung zu verzeichnen, was über Aktivierung der Glykogensynthase zur Aufrechterhaltung der muskulären Glykogenspeicher beitragen kann. Sechs Monate nach TAC-OP war die Akt-Expression im Skelettmuskel dann jedoch verstärkt, was zwar einen Kompensationsmechanismus zur Vermeidung einer Muskel-Atrophie darstellt, jedoch durch Inaktivierung der Glykogensynthase sinkende Glykogenreserven bedingt. Gleichzeitig war eine Steigerung der Expression von Hexokinase II zu beobachten, die durch Bildung von Glucose-6-Phosphat die intrazellluläre Glucoseverfügbarkeit kompensiert.
Bei zusätzlicher Sucrosefütterung über sechs Monate zeigte sich im Muskel der TAC-operierten Tiere keine kompensatorische Steigerung von Akt- und HKII-Expression mehr, und auch die Entleerung der muskulären Glykogenspeicher war vollständig abgefangen. Ebenso wurde die Entleerung der hepatischen Glykogenvorräte durch Sucrose-Substitution verhindert, was für eine ausreichende Glucoseverfügbarkeit spricht und die Normalisierung der Glucosetoleranz erklärt. Wichtige Regulatoren der Leber-Glykogensynthese, z.B. GLUT2 und HK IV, oder der zellulären Energie-Homöostase, z.B. AMPK, waren hingegen zu keinem Zeitpunkt wesentlich verändert.
Zusammenfassend konnten somit durch eine Sucrosesubstitution die bei TAC-induzierter Herzinsuffizienz reduzierten Glykogenspeicher in Muskel und Leber erhalten, der systemische Glucosestoffwechsel normalisiert und eine Reduktion der Mortalität erzielt werden. Insgesamt lässt sich feststellen, dass sich Strategien, die zu einer Wiederherstellung der systemischen Glucoseverfügbarkeit beitragen, möglicherweise positiv auf eine Herzinsuffizienztherapie auswirken könnten.
Within the last decades, therapeutic advances, such as antenatal corticosteroids, surfactant replacement, monitored administration of supplemental oxygen, and sophisticated ventilatory support have significantly improved the survival of extremely premature infants. In contrast, the incidence of some neonatal morbidities has not declined. Rates of bronchopulmonary dysplasia (BPD) remain high and have prompted neonatologists to seek effective strategies of non-invasive respiratory support in high risk infants in order to avoid harmful effects associated with invasive mechanical ventilation. There has been a stepwise replacement of invasive mechanical ventilation by early continuous positive airway pressure (CPAP) as the preferred strategy for initial stabilization and for early respiratory support of the premature infant and management of respiratory distress syndrome. However, the vast majority of high risk babies are mechanically ventilated at least once during their NICU stay. Adjunctive therapies aiming at the prevention of CPAP failure and the support of functional residual capacity have been introduced into clinical practice, including alternative techniques of administering surfactant as well as non-invasive ventilation approaches. In contrast, the strategy of applying sustained lung inflations in the delivery room has recently been abandoned due to evidence of higher rates of death within the first 48 h of life.
Moderne Medizinische Rehabilitation versteht sich als der Teil einer Behandlungskette, in dem es insbesondere um die Förderung der Teilhabe geht. Die Ergebnisse der Rehabilitationsmaßnahmen werden durch die Nachsorge bzw. die Umsetzung von Veränderungen in der Zeit nach der Rehabilitation stabilisiert, so dass die Vorbereitung der Nachsorge von besonderer Bedeutung für die Verstetigung ist. Art und Inhalte der Beratung durch Sozialarbeiter unterscheiden sich dabei je nach Problem- und Bedarfslage der Rehabilitanden und dürften innerhalb eine homogenen Patienten-/Indikationsgruppe eine gewisse Ähnlichkeit aufweisen.
Für die vorliegende Arbeit wurde ältere Rehabilitanden (> 65 J.) einer Rehabilitationsklinik mit internistischem und orthopädischem Schwerpunkt acht Wochen nach dem Reha-Aufenthalt in einem halbstrukturierten Telefoninterview gefragt, ob und inwieweit es bisher gelang, die Empfehlungen des Sozialdienstes aus der Rehabilitation umzusetzen.
Im Ergebnis zeigt sich, dass viele Empfehlungen seitens des Sozialdienstes bis zum Zeitpunkt der Nachbefragung von den Rehabilitanden nach Eigenangaben umgesetzt wurden. Vor allem die Teilnahme an Sportprogrammen oder die Unterstützung im Alltag wurden häufig in das alltägliche Leben integriert. Für die untersuchten Einflussvariablen (Geschlecht, Lebensalter, Barthel-Index, Lebensverhältnisse) lassen sich allerdings nur in den wenigsten Fällen signifikante Zusammenhänge nachweisen.
Günstig wäre es sicher auch, wenn gewährleistet wird, dass Patienten in der nachstationären ambulanten Versorgung weiter angehalten werden, um die empfohlene Nachsorge auch effizient umsetzen zu können.
Die Juvenile idiopathische Arthritis (JIA) ist ein chronisch entzündliches Krankheitsbild des Kindesalters mit ungeklärter Ursache. Eine ätiopathogenetische Beteiligung des Immunsystems und im speziellen der T-Zellen ist wahrscheinlich.
Die vorliegende durchflusszytometrische Untersuchung umfasst die Daten von 84 JIA Patienten und 40 altersgleichen Kontrollprobanden und vergleicht die Ergebnisse im transversalen sowie longitudinalen Studiendesign.
Die Phänotypisierung der Lymphozyten zeigte im Vergleich zu gesunden Kontrollprobanden eine abweichende Verteilung der T-Helferzell-Subpopulationen. Die CD4 positiven T-Helferzellen der JIA Patienten setzen sich aus einer Anzahl proportional verminderter differenzierter Zellen und vermehrter naiver Zellen zusammen. Im Zytokinprofil wurde eine reduzierte Th1- sowie Th2-Antwort beobachtet. In allen T-Zell-Subpopulationen der JIA Patienten fand sich RORуt, der Transkiptionsfaktor der Th17-Differenzierung, hochreguliert. Im klinischen Schub der Arthritis zeigte sich eine signifikant erhöhte RORуt-Expression sowie eine vermehrte IL-17 Produktion. Die IL-17 produzierenden Zellen zeigten u.a. einen regulatorischen Phänotyp.
Bei den Stimulationsversuchen von CCR6+ Zellen, die wir als IL-17 produzierende Zellen definieren konnten, wurde eine ausgeprägte Plastizität beobachtet. Mit spezifischen Zytokinstimuli wurden die IL-17 produzierenden Zellen vermehrt zu Th17, Th1 oder regulatorischen T-Zellen polarisiert. Die Polarisierbarkeit unterschied sich zwischen JIA Patienten und Kontrollprobanden. Die CCR6+ Zellen der JIA Patienten zeigten im Vergleich zu den Kontrollprobanden ausgeprägte Polarisierungstendenzen zum Th17-Phänotyp und ließen sich nur gering in Th1 oder Treg konvertieren.
Die Ergebnisse unserer Untersuchung lassen auf eine Dominanz im Th17-Differenzierungsweg schließen. Der Transkriptionsfaktor RORуt scheint vor allem im akuten Krankheitsschub zu prädominieren und die Th17-Differenzierung zu steuern. Eine Gegenregulation von Th1- und Th2-Zellen fehlt.
Zusammenfassend ergeben sich Hinweise darauf, dass Th17-Zellen eine bedeutende Rolle in der Pathogenese der Juvenilen idiopathischen Arthritis spielen. In wieweit man IL-17, den CCR6-Rezeptor oder den Transkriptionsfaktor RORуt als Angriffsziel von krankheitsmodifizierenden Therapeutika nutzen kann, muss in weiteren Studien untersucht werden. Ebenso gilt es herauszufinden, ob die forcierte Polarisierung in Richtung regulatorische T-Zellen oder gegenregulatorisch aktive Th1 und Th2 Zellen eine Therapiestrategie darstellt.
Metall-basierte Antitumorwirkstoffe wie Cisplatin, Carboplatin und Oxaliplatin sind weltweit für die Behandlung verschiedener Krebsarten zugelassen. Resistenzbildung, starke Nebenwirkungen und ein eingeschränktes Spektrum responsiver Tumoren schränken jedoch ihren Anwendungsbereich ein. Daher ist die Suche nach neuen Platinverbindungen mit verbesserten Eigenschaften sowie Antitumor-aktiven Metallkomplexen anderer Metalle ein aktuelles Forschungsthema. Durch die Einbettung der Wirkstoffe in entsprechende Trägermaterialien und eine Freisetzung mit präziser zeitlicher und räumlicher Kontrolle sollten sich zudem die Nebenwirkungen deutlich reduzieren lassen. Im Rahmen dieser Doktorarbeit wurde daher eine Serie von quadratisch-planaren Platin(II)- und Palladium(II)-Komplexen mit N^N^S-Chelatliganden auf der Basis von N-Phenyl-2-(pyridin-2-ylmethylen)hydrazin-1-carbothioamid und N-Phenyl-2-(chinolin-2-ylmethylen)hydrazin-1-carbothioamid synthetisiert, die mit längeren Alkylketten funktionalisiert wurden, um eine hohe Affinität für Lipid-basierte Microbubbles als Träger zu erreichen, aus denen die Metallkomplexe dann unter Einwirkung von Ultraschall freigesetzt werden sollten. Es wurden drei verschiedene Ligandenfamilien ausgehend von der Grundstruktur L = R1-CR2=N-NH-C(S)-NH-R3 synthetisiert, wobei R1 = 2-Pyridyl oder 2-Chinolinyl, R2 = H, CH3, C8H17 oder C10H21 und R3 = CH3 oder C6H5 gewählt wurden. Die Umsetzung der Liganden mit Kaliumtetrachloridoplatinat(II), Natriumtetrachloridopalladat(II) oder [PdCl2(cod)] mit cod = 1,5-Cyclooctadien führte zu neutralen N^N^S-Komplexen [MCl(L)] mit M = Pd, Pt in allgemein guter Ausbeute. Die Kristallstruktur von [PtCl(L)] mit R1 = 2-Pyridyl, R2 = C10H21 und R3 = CH3 bestätigte zudem die quadratisch-planar Koordination des Metalls durch den N^N^S-koordinierten Liganden und ein Chlorid-Anion. Die Verbindungen mit R3 = CH3 zeigen im 195Pt NMR zwei Peaks, was auf das Vorliegen eines Isomerengemischs hindeutet, wobei die Daten vermuten lassen, dass neben der N^N^S-gebundenen Hauptspezies noch eine weitere mit N^N^N-koordiniertem Liganden und freier SH-Gruppe vorliegt. Solche Isomerengemische sind für biologischen Anwendungen ungeeignet, da die Isomere eine unterschiedliche Aktivität aufweisen können. Die anderen Platin(II)-Komplexe zeigen dagegen im 195Pt NMR nur einen Peak und sind somit für Cytotoxizitätsstudien geeignet. Mit Hilfe des MTT-Assays wurden EC50-Werte an verschiedenen Gliablastom-Zellinien für zwölf einheitliche Komplexe bestimmt. Für die potentesten Verbindungen wurden EC50-Werte im unteren mikromolaren Bereich ermittelt (2–9 µM), so dass die Aktivität teilweise sogar die von Cisplatin als Referenzverbindung übertraf. Insbesondere die Variation der aromatischen Oberfläche in den Pyridyl- vs. Chinolinylverbindungen hatte jedoch keinen wesentlichen Einfluss auf die EC50-Werte. Zudem führte eine Verlängerung der R2-Seitenkette bei den Palladium(II)-Verbindungen zu einer niedrigeren Aktivität. Die Verteilungskoeffizienten logP ergaben für alle Verbindungen recht ähnliche positive Werte, was die Lipophilie der Neutralkomplexe belegte. Für eine weitere Strukturvariation wurden außerdem zwei Azido-Komplexe [M(N3)(L)] mit M = Pd, Pt in moderater Ausbeute synthetisiert. Diese wurden dann in einer „iClick“-Reaktion unter sehr milden Bedingungen mit Dimethylacetylendicarboxylat (DMAD) und 4,4,4-Trifluorobut-2-insäureethylester zu den Triazolat-Komplexen [M(triazolateR,R')(L)] umgesetzt. Durch 1H NMR- und 19F NMR-spektroskopische Untersuchungen wurden gezeigt, dass diese teilweise als Isomerengemische vorliegen, da das Triazolat entweder über das N1-, N2- oder N3-Stickstoffatom an das Metall gebunden sein kann. Für eine ausgewählte Verbindung mit M = Pt und DMAD als Alkin wurde die Kinetik der „iClick“-Reaktion mit Hilfe der 1H NMR Spektroskopie untersucht. Die ermittelte Geschwindigkeitskonstante 2. Ordnung k2 = (1.82 ± 0.05).10-1 L mol-1 s 1 ist vergleichbar zum Beispiel der der etablierten strain-promoted azide-alkyne cycloaddition (SPAAC). Für die Einbettung der lipophilen Metallkomplexe in ein Ultraschall-aktivierbares Trägersystem wurden aus Dipalmytoylphosphatidylcholin, 1,2-Dipalmitoyl-sn-glycero-3-phosphat, 1,2-Distearoyl-sn-glycero-3-phosphoethanolamin-N-methoxy(polyethylenglycol)-2000 Ammonium Salz und Komplexlösung unter Zusatz von Octafluopropan gasgefüllte Microbubbles hergestellt. Stabilitätsversuche zeigten, dass die Bläschenzahl selbst unter Normalbedingungen innerhalb von 2 h um 50% abnimmt. Daher sollten die Microbubbles vor jeder Verwendung täglich frisch hergestellt und möglichst unmittelbar danach verwendet werden. Ein unmittelbares Zerplatzen der Bläschen wurde durch Behandlung mit Ultraschall bei 500 Hz erreicht. Die Viabilität der verwendeten GaMG-Zellen wudre unter diesen Bedingungen jedoch nicht beeinträchtigt. Dennoch war die auf die Microbubbles geladene Platin-Konzentration zu niedrig, um mit dem MTT-Assay einen signifikanten Unterschied zwischen beladenen und unbeladenen Bläschen zu erreichen, so dass hier in Zukunft noch weitere Optimierungen erforderlich sein werden.
Die Patientenbestrahlung stellt eine wichtige Therapiesäule in der onkologischen Behandlung von Hirntumoren dar. Das Hauptaugenmerk wird dabei auf die Erreichung einer vorgegebenen Zieldosis im Tumorgebiet und die ausreichende Schonung von sensiblen Strukturen gerichtet. Wir verglichen insgesamt 4 Bestrahlungstechniken untereinander, welche in ihrer Segmentierung und Feldzahl variiert werden können: KoPlanar (Komplettbestrahlung in einer Ebene), KoPlanar+1 (Bestrahlung in einer Ebene mit einem Zusatzfeld in einer anderen Ebene), 2-Ebenen (Bestrahlung auf 2 unterschiedliche Ebenen verteilt), Quasi-Isotrop (Bestrahlung mit Zentralstrahlen in mehreren unterschiedlichen Ebenen). Die Feldzahl kann zwischen wenigen Feldern (9F oder 10F) und vielen Feldern (15F oder 16F) gewählt werden. Die Segmentanzahl wird entweder bei 64 oder 120 Segmenten festgelegt, alternativ wurde eine freie Optimierung der Feldfluenz ermöglicht.
Dabei zeigte die Quasi-Isotrope Technik eindeutige und signifikante Vorteile gegenüber allen anderen Techniken sowohl bei niedrigen als auch hohen Feldzahlen. Die koplanare Bestrahlung schnitt bei unserer Auswertung am schlechtesten ab. Die 2-Ebenen Technik und KoPlanar+1 Technik können bei hohen Feldzahlen als gleichwertig betrachtet werden, bei niedrigen Feldzahlen zeigt die KoPlanar+1 Technik Vorteile. Aus unserer Sicht sollten die unentschiedenen Vergleiche in weiteren Studien untersucht werden, die das Patientengut weiter einengen. Weiterhin wäre eine Erweiterung der Untersuchungen auf die schneller applizierbaren nonkoplanaren Volumetric Arc –Techniken (VMAT) wünschenswert.
N-Glykosylierungen spielen beim Env-Gen eine wichtige Rolle. Sie dienen nicht nur als „Escape-Phänomen“ zur Verhinderung einer Elimination des Virus durch neutralisierende Antikörper. Es hat sich gezeigt, dass bestimmte Menschen sich mit HIV infizieren können, aber es zu keinem Zeitpunkt zu AIDS-typischen Symptomen kommt, ohne die Einnahme antiretroviraler Therapie (ART). Solche Menschen werden als Elite Controller bezeichnet. Ihr Organismus kann selbst die Viruslast in sehr geringen Grenzen halten (< 50 Kopien/ml). Ziel dieser Arbeit ist es, den Einfluss von N-Glykosylierungen in der Entstehung von Elite Controller zu untersuchen und prozentuell eine Tendenz zu schaffen, inwieweit die Glykosylierungsdichte des Env-Proteins entscheidend ist.
Es konnte gezeigt werden, dass eine immunologische Kontrolle auch auf der B-Zellebene stattfinden kann. Als Hinweis dient die geringe Glykosylierungsdichte im Bereich CD4bs und MPER, die indirekt über MHC Klasse II zu einer erhöhten Produktion von Antikörpern führen kann. Bisher wurde bei Elite Controller die T-Zellebene als mögliche immunologische Kontrolle beschrieben, jedoch gibt diese Arbeit hinweise, dass auch eine immunologische Kontrolle mittels Antikörper möglich ist. Die glykosylierten Zielepitope können eine große Hilfe sein für das Aussehen eines späteren Impfstoffs.
Fluorinated compounds are an important motif, particularly in pharmaceuticals, as one-third of the top performing drugs have fluorine in their structures. Fluorinated biaryls also have numerous applications in areas such as material science, agriculture, crystal engineering, supramolecular chemistry, etc. Thus, the development of new synthetic routes to fluorinated chemical compounds is an important area of current research. One promising method is the borylation of suitable precursors to generate fluorinated aryl boronates as versatile building blocks for organic synthesis.
Chapter 1
In this chapter, the latest developments in the synthesis, stability issues, and applications of fluorinated aryl boronates in organic synthesis are reviewed. The catalytic synthesis of fluorinated aryl boronates using different methods, such as C–H, C–F, and C–X (X = Cl, Br, I, OTf) borylations are discussed. Further studies covering instability issues of the fluorinated boronate derivatives, which are accelerated by ortho-fluorine, have been reported, and the applications of these substrates, therefore, need special treatment.
Numerous groups have reported methods to employ highly fluorinated aryl boronates that anticipate the protodeboronation issue; thus, polyfluorinated aryl boronates, especially those containing ortho-fluorine substituents, can be converted into chloride, bromide, iodide, phenol, carboxylic acid, nitro, cyano, methyl esters, and aldehyde analogues. These substrates can be applied in many cross-coupling reactions, such as the Suzuki-Miyaura reaction with aryl halides, the Chan-Evans-Lam C–N reaction with aryl amines or nitrosoarenes, C–C(O) reactions with N-(aryl-carbonyloxy)phthalamides or thiol esters (Liebskind-Srogl cross-coupling), and oxidative coupling reactions with terminal alkynes. Furthermore, the difficult reductive elimination from the highly stable complex [PdL2(2,6-C6F2+nH3-n)2] was the next challenge to be targeted in the homocoupling of 2,6-di-fluoro aryl pinacol boronates, and it has been solved by conducting the reaction in arene solvents that reduce the energy barrier in this step as long as no coordinating solvent or ancillary ligand is employed.
Chapter 2
In this chapter, phenanthroline-ligated copper complexes proved to be efficient catalysts for the Suzuki-Miyaura cross-coupling of highly fluorinated aryl boronate esters (ArF–Bpin) with aryl iodides or bromides. This newly developed method is an attractive alternative to the traditional methods as copper is an Earth-abundant metal, less toxic, and cheaper compared to the traditional methods which commonly required palladium catalysts, and silver oxide that is also often required in stoichiometric amounts. A combination of 10 mol% copper iodide and 10 mol% phenanthroline, with CsF as a base, in DMF, at 130 ˚C, for 18 hours is efficient to cross-couple fluorinated aryl pinacol boronates with aryl iodides to generate cross-coupled products in good to excellent yields. This method is also viable for polyfluorophenyl borate salts such as pentafluorophenyl-BF3K. Notably, employing aryl bromides instead of aryl iodides for the coupling with fluorinated aryl–Bpin compounds is also possible; however, increased amounts of CuI/phenanthroline catalyst is necessary, in a mixture of DMF and toluene (1:1).
A diverse range of π···π stacking interactions is observed in the cross-coupling products partly perfluorinated biaryl crystals. They range from arene–perfluoroarene interactions (2-(perfluorophenyl)naphthalene and 2,3,4-trifluorobiphenyl) to arene–arene (9-perfluorophenyl)anthracene) and perfluoroarene–perfluoroarene (2,3,4,5,6-pentafluoro-2’methylbiphenyl) interactions.
Chapter 3
In this chapter, the efficient Pd-catalyzed homocoupling reaction of aryl pinacol pinacol boronates (ArF–Bpin) that contain two ortho-fluorines is presented. The reaction must be conducted in a “noncoordinating” solvent such as toluene, benzene, or m-xylene and, notably, stronger coordinating solvents or ancillary ligands have to be avoided. Thus, the Pd center becomes more electron deficient and the reductive elimination becomes more favorable. The Pd-catalyzed homocoupling reaction of di-ortho-fluorinated aryl boronate derivatives is difficult in strongly coordinating solvents or in the presence of strong ancillary ligands, as the reaction stops at the [PdL2(2,6-C6F2+nH3-n)2] stage after the transmetalations without the reductive elimination taking place. It is known that the rate of reductive elimination of Ar–Ar from [ML2(Ar)(Ar)] complexes containing group-10 metals decreases in the order Arrich–Arpoor > Arrich–Arrich > Arpoor–Arpoor. Furthermore, reductive elimination of the most electron-poor diaryls, such as C6F5–C6F5, from [PdL2(C6F5)2] complexes is difficult and has been a challenge for 50 years, due to their high stability as the Pd–Caryl bond is strong. Thus, the Pd-catalyzed homocoupling of perfluoro phenyl boronates is found to be rather difficult.
Further investigation showed that stoichiometric reactions of C6F5Bpin, 2,4,6-trifluorophenyl–Bpin, or 2,6-difluorophenyl–Bpin with palladium acetate in MeCN stops at the double transmetalation step, as demonstrated by the isolation of cis-[Pd(MeCN)2(C6F5)2], cis-[Pd(MeCN)2(2,4,6-C6F3H2)2], and cis-[Pd(MeCN)2(2,6-C6F2H3)2] in quantitative yields. Thus, it can be concluded that the reductive elimination from diaryl-palladium complexes containing two ortho-fluorines in both aryl rings, is difficult even in a weakly coordinating solvent such as MeCN. Therefore, even less coordinating solvents are needed to make the Pd center more electron deficient. Reactions using “noncoordinating” arene solvents such as toluene, benzene, or m-xylene were conducted and found to be effective for the catalytic homocoupling of 2,6-C6F2+nH3-nBpin. The scope of the reactions was expanded. Using toluene as the solvent, the palladium-catalyzed homocoupling of ArF–Bpin derivatives containing one, two or no ortho-fluorines gave the coupled products in excellent yields without any difficulties.
DFT calculations at the B3LYP-D3/def2-TZVP/6-311+g(2d,p)/IEFPCM // B3LYP-D3/SDD/6-31g**/IEFPCM level of theory predicted an exergonic process and lower barrier (< 21 kcal/mol) for the reductive elimination of Pd(C6F5)2 complexes bearing arene ligands, compared to stronger coordinating solvents (acetonitrile, THF, SMe2, and PMe3), which have high barriers ( > 33.7 kcal/mol). Reductive elimination from [Pd(ηn-Ar)(C6F5)2] complexes have low barriers due to: (i) ring slippage of the arene ligand as a hapticity change from η6 in the reactant to ηn (n ≤ 3) in the transition state and the product, which led to less σ-repulsion; and (ii) more favorable π-back-bonding from Pd(ArF)2 to the arene fragment in the transition state.
Chapter 4
In this chapter, the efficient Pd-catalyzed C–Cl borylation of aryl chlorides containing two ortho-fluorines is presented. The reactions are conducted under base-free conditions to prevent the decomposition of the di-ortho-fluorinated aryl boronates, which are unstable in the presence of base. A combination of Pd(dba)2 (dba = dibenzylideneacetone) with SPhos (2-dicyclohexylphosphino-2′,6′-dimethoxybiphenyl) as a ligand is efficient to catalyze the C–Cl borylation of aryl chlorides containing two ortho-fluorine substituents without base, and the products were isolated in excellent yields. The substrate scope can be expanded to aryl chloride containing one or no ortho-fluorines and the borylated products were isolated in good to very good yield. This method provides a nice alternative to traditional methodologies using lithium or Grignard reagents.
Die hypertherme intraperitoneale Chemoperfusion (hyperthermic intraperitoneal chemotherapy, HIPEC) ist ein vielversprechendes Verfahren bei Patienten mit limitierter Peritonealkarzinose. Möglicherweise wirken Hitzeschockprotein (HSP)-vermittelte Reparaturmechanismen und proliferative Effekte dem Therapieerfolg jedoch entgegen. Um solche möglichen limitierenden Faktoren zu untersuchen, wurden in einem in vitro-HIPEC-Modell die Auswirkungen von Hyperthermie und Chemotherapie auf die Expression von HSPs und die Proliferationsmarker Ki-67 und PCNA im Kolonkarzinom mittels RT-qPCR und Western Blot untersucht.
Es zeigten sich hochregulierte HSPs, die mit zunehmender Temperatur und durch die Zugabe von Chemotherapie zunahmen. Man kann davon ausgehen, dass hierdurch intrazelluläre antiapoptotische Effekte und Reparaturmechanismen induziert werden. Die Inhibition von HSPs könnte daher zu einer Verbesserung der HIPEC-Therapie führen.
Zum anderen fand sich eine Erhöhung der Proliferationsmarker Ki-67 und PCNA nach der in vitro-HIPEC, was auf ein aggressives Wachstumsverhalten der überlebenden Zellen hindeutet.
Nicotinamide N-methyltransferase (NNMT) is a new regulator of energy homeostasis. Its expression is increased in models of obesity and diabetes. An enhanced NNMT level is also caused by an adipose tissue-specific knockout of glucose transporter type 4 (GLUT4) in mice, whereas the overexpression of this glucose transporter reduced the NNMT expression. Furthermore, the knockdown of the enzyme prevents mice from diet-induced obesity (DIO) and the recently developed small molecule inhibitors for NNMT reverses the DIO. These previous findings demonstrated the exclusive role of NNMT in adipose tissue and further make it to a promising target in obesity treatment. However, the regulation mechanism of this methyltransferase is not yet clarified.
The first part of the thesis focus on the investigation whether pro-inflammatory signals are responsible for the enhanced NNMT expression in obese adipose tissue because a hallmark of this tissue is a low-level chronic inflammation. Indeed, the NNMT mRNA in our study was elevated in obese patients compared with the control group, whereas the GLUT4 mRNA expression does not differ between lean and obese humans. To analyze whether pro inflammatory signals, like interleukin (IL 6) and tumor necrosis factor α (TNF-α), regulate NNMT expression 3T3-L1 adipocytes were treated with these cytokines. However, IL 6, TNF α, and leptin, which is an alternative activator of the JAK/STAT pathway, did not affect the NNMT protein or mRNA level in differentiated 3T3-L1 adipocytes. The mRNA and protein levels were measured by quantitative polymerase chain reaction (qPCR) and western blotting.
In the second part of this study, 3T3-L1 adipocytes were cultivated with varying glucose concentrations to show whether NNMT expression depends on glucose availability. Further studies with activators and inhibitors of AMP-activated protein kinase (AMPK) and mechanistic target of rapamycin (mTOR) signaling pathways were used to elucidate the regulation mechanism of the enzyme.
The glucose deprivation of differentiated 3T3-L1 adipocytes led to a 2-fold increase in NNMT expression. This effect was confirmed by the inhibition of the glucose transports with phloretin as well as the inhibition of glycolysis with 2-deoxyglucose (2-DG). AMPK serves as an intracellular energy sensor and the pharmacological activation of it enhanced the NNMT expression. This increase was also caused by the inhibition of mTOR. Conversely, the activation of mTOR using MHY1485 prevented the effect of glucose deprivation on NNMT. Furthermore, the NNMT up-regulation was also blocked by the different autophagy inhibitors.
Taken together, NNMT plays a critical role in autophagy in adipocytes, because an inhibition of this process prevented the augmented NNMT expression during glucose starvation. Moreover, the effect on NNMT protein and mRNA level depends on AMPK and mTOR. However, pro-inflammatory signals did not affect the expression. Further in vivo studies have to clarify whether AMPK activation and mTOR inhibition as well as autophagy are responsible for the increased NNMT levels in obese adipose tissue. In future this methyltransferase emerges as an awesome therapeutic target for obesity.
Sprachaudiometrische Tests sind subjektive Testverfahren um die Sprachverständlichkeit zu messen. Sie dienen unter anderem zur Indikation für Hörhilfen, zur Verlaufskontrolle und zur Begutachtung. Der deutschsprachige Sprachraum ist von den Freiburger Satztests geprägt. Dennoch gibt es im deutschsprachigen Raum weitere sprachaudiometrische Testverfahren. Der Oldenburger Satztest ist ein moderner Sprachtests, der als Matrixtest syntaktisch korrekte und semantisch inkorrekte Sätze als Sprachmaterial nutzt.
Diese Dissertation untersucht altersabhängige Lerneffekte an einem normalhörenden Probandenkollektiv. Dazu wurden 40 Probanden in homogenen Altersgruppen auf den bereits in der Literatur vielfach beschriebenen Lerneffekt untersucht. Die Dissertation konnte zeigen, dass sich der Lerneffekt in allen Altersgruppen ähnlich verhält. Die Lerneffekte sind damit nicht altersabhängig. Der Lerneffekt zeigte in allen Altersgruppen signifikante Verbesserungen in der fünften durchgeführten Testliste im Vergleich zur ersten durchgeführten Testliste.
Multiple myeloma (MM) is a disease of terminally differentiated B-cells which accumulate in the bone marrow leading to bone lesions, hematopoietic insufficiency and hypercalcemia. Genetically, MM is characterized by a great heterogeneity. A recent next-generation sequencing approach resulted in the identification of a signaling network with an accumulation of mutations in receptor-tyrosine kinases (RTKs), adhesion molecules and downstream effectors. A deep-sequencing amplicon approach of the coding DNA sequence of the six RTKs EPHA2, EGFR, ERBB3, IGF1R, NTRK1 and NTRK2 was conducted in a patient cohort (75 MM samples and 68 corresponding normal samples) of the “Deutsche Studiengruppe Multiples Myelom (DSMM)” to further elucidate the role of RTKs in MM. As an initial approach the detected mutations were correlated with cytogenetic abnormalities and clinical data in the course of this thesis. RTK mutations were present in 13% of MM patients of the DSMM XI trial and accumulated in the ligand-binding and tyrosine-kinase domain. The newly identified mutations were associated with an adverse patient survival, but not with any cytogenetic abnormality common in MM. Especially rare patient-specific SNPs (single nucleotide polymorphism) had a negative impact on patient survival. For a more comprehensive understanding of the role of rare RTK SNPs in MM, a second amplicon sequencing approach was performed in a patient cohort of the DSMM XII trial that included 75 tumor and 184 normal samples. This approach identified a total of 23 different mutations in the six RTKs EPHA2, EGFR, ERBB3, IGF1R, NTRK1 and NTRK2 affecting 24 patients. These mutations could furthermore be divided into 20 rare SNPs and 3 SNVs (single nucleotide variant). In contrast to the first study, the rare SNPs were significantly associated with the adverse prognostic factor del17p.
IGF1R was among the most commonly mutated RTKs in the first amplicon sequencing approach and is known to play an important role in diverse cellular processes such as cell proliferation and survival. To study the role of IGF1R mutations in the hard-to-transfect MM cells, stable IGF1R-knockdown MM cell lines were established. One of the knockdown cell lines (L363-C/C9) as well as a IGF1R-WT MM cell line (AMO1) were subsequently used for the stable overexpression of WT IGF1R and mutant IGF1R (N1129S, D1146N). Overall, an impact on the MAPK and PI3K/AKT signaling pathways was observed upon the IGF1R knockdown as well as upon WT and mutant IGF1R overexpression. The resulting signaling pattern, however, differed between different MM cell lines used in this thesis as well as in a parallel performed master thesis which further demonstrates the great heterogeneity described in MM.
Taken together, the conducted sequencing and functional studies illustrate the importance of RTKs and especially of IGF1R and its mutants in the pathogenesis of MM. Moreover, the results support the potential role of IGF1R as a therapeutic target for a subset of MM patients with mutated IGF1R and/or IGF1R overexpression.
Chapter two reports the catalytic triboration of terminal alkynes with B2pin2 using readily available Cu(OAc)2 and PnBu3. Various 1,1,2-triborylalkenes, a class of compounds which have been demonstrated to be potential Matrix Metalloproteinase-2 (MMP-2) inhibitors, are obtained directly in moderate to good yields. The process features mild reaction conditions, broad substrate scope, and good functional group tolerance were observed. This Cu-catalyzed reaction can be conducted on a gram scale to produce the corresponding 1,1,2-triborylalkenes in modest yields. The utility of these products is demonstrated by further transformation of the C-B bonds to prepare gem-dihaloborylalkenes (F, Cl, Br), monohalodiborylalkenes (Cl, Br), and trans-diaryldiborylalkenes, which serve as important synthons and have previously been challenging to prepare.
A convenient and efficient one step synthesis of 1,1,1-triborylalkanes was achieved via sequential dehydrogenative borylation and double hydroboration of terminal alkynes with HBpin (HBpin = pinacolborane) catalyzed by inexpensive and readily available Cu(OAc)2. This protocol proceeded under mild conditions, furnishing 1,1,1-tris(boronates) with wide substrate scope, excellent selectivity and good functional group tolerance, and is applicable to gram-scale synthesis without loss of yield. The 1,1,1-triborylalkanes can be used in the preparation of α-vinylboronates and borylated cyclic compounds, which are valuable but previously rare compounds. Different alkyl groups can be introduced stepwise via base-mediated deborylative alkylation to produce racemic tertiary alkyl boronates, which can be readily transformed into useful tertiary alcohols.
Chapter 4 reported a NaOtBu-catalyzed mixed 1,1-diboration of terminal alkynes with an unsymmetrical diboron reagent BpinBdan. This Brønsted base-catalyzed reaction proceeds in a regio- and stereoselective fashion affording 1,1-diborylalkenes with two different boryl moieties in moderate to high yields, and is applicable to gram-scale synthesis without loss of yield or selectivity. Hydrogen bonding between the Bdan group and tBuOH is proposed to be responsible for the observed stereoselectivity. The mixed 1,1-diborylalkenes can be utilized in stereoselective Suzuki-Miyaura cross-coupling reactions.
Der primäre Hyperaldosteronismus ist eine der häufigsten Ursachen einer sekundären Hypertonie. Entscheidend für eine adäquate Therapie ist die Subtypen-Differenzierung zwischen Aldosteron produzierenden Adenomen und bilateralen Hyperplasien der Nebennierenrinde. Hierfür wird heutzutage eine selektive Katheterisierung der Nebennierenvenen durchgeführt. Diese Untersuchung ist jedoch sehr aufwendig und geht mit einer hohen Strahlenbelastung einher. Daher wurden im Rahmen dieser Arbeit 11 fluorierte Inhibitoren der Aldosteronsynthase (CYP11B2) auf ihre in-vitro- und in-vivo-Wirksamkeit und Selektivität untersucht. Ziel war es, über eine gezielte Bindung radiofluorierter Tracer an Aldosteron produzierenden Zellen eine nicht invasive Bildgebung zur Subtypendifferenzierung beim primären Hyperaldosteronismus zu entwickeln. Hierbei konnten mehrere vielversprechende Substanzen gefunden werden, die selektiv an CYP11B2 binden und die Aldosteronproduktion hemmen. Diese können somit möglicherweise für die Subtypendifferenzierung beim primären Hyperaldosteronismus eingesetzt werden.
Divalent cations are important second messengers triggering various signal transduction events in platelets. Whereas calcium channel blockers have an established antithrombotic effect and the regulation of Ca2+ homeostasis has been elucidated in platelets, the molecular regulation of Mg2+ and Zn2+ homeostasis has not been investigated so far.
In the first part of the thesis, the role of -type serine-threonine kinase linked to transient receptor potential cation channel, subfamily M, member 7 (TRPM7) in platelets was investigated. Using Trpm7R/R mice with a point mutation deleting the kinase activity, we showed that the TRPM7 kinase regulates platelet activation via immunoreceptor tyrosine-based activation motif (ITAM), hem(ITAM) and protease-activated receptor (PAR) signaling routes. Furthermore, Trpm7R/R mice were protected from in vivo thrombosis and stroke, thus establishing TRPM7 kinase as a promising anti-thrombotic target.
In the second part of the thesis, the role of TRPM7 channel in a megakaryocyte (MK) and platelet-specific knockout mouse, Trpm7fl/fl-Pf4Cre, was investigated. Here, we observed that depending on the type of stimulation, Trpm7fl/fl-Pf4Cre platelets showed either enhanced or inhibited responses. Although Trpm7fl/fl-Pf4Cre mice were thrombocytopenic, no differences to wildtype mice were observed in models of in vivo thrombosis and stroke. The above two studies highlight that inhibition of TRPM7 kinase but not the channel itself (in MKs and platelets) may be a promising anti-thrombotic strategy.
Besides TRPM7, we investigated the role of magnesium transporter 1 (MAGT1) in platelet Mg2+ homeostasis and found that MAGT1 primarily regulates receptor-operated calcium entry (ROCE) in platelets specifically upon GPVI activation. This physiological crosstalk is triggered by protein kinase C (PKC) isoforms. Platelets from Magt1-/y mice hyper-reacted to GPVI and thromboxane A2 (TXA2) receptor stimulation in vitro. Consequently, Magt1-/y platelets were found to be pro-thrombotic in disease models of thrombosis and stroke.
To compare platelet ITAM-signaling to the immune system, we further investigated the role of MAGT1 in T and B cells. We described the primary role of MAGT1 in mice under pathogen-free conditions. Magt1-/y B cells showed dysregulated Mg2+ and Ca2+ homeostasis upon B-cell receptor activation, thereby altering Syk, LAT, phospholipase C (PLC)2 and PKC phosphorylation. In contrast to human MAGT1-deficient T cells, development and effector functions of mouse Magt1-/y T cells showed no alterations.
Finally, in the last part of the thesis, we described methods to measure intracellular free zinc [Zn2+]i in human and mouse platelets with storage pool disease (SPD). We propose to measure the [Zn2+]i status in SPD platelets as a relatively easy diagnostic to screen platelet granule abnormalities.
Chapter 1
N-Heterocyclic olefins (NHOs), relatives of N-heterocyclic carbenes (NHCs), exhibit high nucleophilicity and soft Lewis basic character. To investigate their π-electron donating ability, NHOs were attached to triarylborane π-acceptors (A) giving donor(D)-π-A compounds 1-3. In addition, an enamine π-donor analogue (4) was synthesized for comparison.
UV-visible absorption studies show a larger red shift for the NHO-containing boranes than for the enamine analogue, a relative of a CAAC. The red shifted absorption of NHO-containing boranes indicate smaller energy gaps of NHO-containing boranes than CAAC-containing boranes. Solvent-dependent emission studies indicate that 1-4 have moderate intramolecular charge transfer (ICT) behavior.
Electrochemical investigations reveal that the NHO-containing boranes have extremely low reversible oxidation potentials (e.g., for 3, E1/2ox = –0.40 V vs. Fc/Fc+ in THF) which indicate the electron rich property of NHOs.
Furthermore, TD-DFT calculations were carried out on these four D-π-A boranes. The results show that the LUMOs of 1-4 only show a small difference, but the HOMOs of 1-3 are much more destabilized than that of the enamine-containing 4, which is in agreement with the electrochemical investigations and confirms the stronger donating ability of NHOs.
Chapter 2
Since the beginning of this century, the chemistry of (hetero)arene-fused boroles has attracted increasing interest. (Hetero)arene-fused boroles exhibit strong Lewis acidity, distinct fluorescence properties, strong electron accepting abilities, etc. However, their chemistry been only very briefly reviewed either as part of reviews on “free” boroles or on boron-doped polycyclic aromatic hydrocarbons (PAHs). In this chapter, we addressed the chemistry of (hetero)arene-fused boroles from fundamentals to their widely varying applications. It includes:
1) Synthetic methodology Both historical and recently developed strategies for the synthesis of fused boroles.
2) Stabilities A comparison of different kinetic protection strategies.
3) 9-Borafluorenes with a fluorinated backbone Application as Lewis acids, forming ion pairs with Cp2Zr(CH3)2 and applied as activators for polymerization, activators of H2, and other related applications.
4) Donor-acceptor 9-borafluorenes Applications as F– “turn on” sensors, potential applications as electron accepting units for organic (opto)electronics, bipolar transporting materials, TADF materials, and different functionalization strategies.
5) Heteroarene-fused boroles Enhanced antiaromaticity, unique coordination mode and their interesting properties.
6) Intramolecular dative bonding in 9-borafluorenes Bond-cleavage-induced intramolecular charge transfer (BICT), BICT-induced large Stoke shifts and dual emissions, application as a ratiometric sensor.
7) 9-Borafluorene-based main chain polymers Application in polymer chemistry and their distinct properties, e.g., as a sensor for gaseous NH3.
8) Electrochemistry A comparison of electron-accepting ability of different functionalized fused boroles through electrochemical studies.
9) Chemical reduction of fused boroles Stable radical anions and dianions of fused boroles and their properties.
10) Three-coordinate borafluorenium cations Cationic 9-borafluorenes and their interesting properties, e.g., in THF, reversible thermal colour switching properties.
Finally, a conclusion and outlook regarding the chemistry, properties and applications, and suggestions for areas which require further study was provided.
Chapter 3
Interested in fusing electron-poor arene onto boroles, two electron-poor phenylpyridyl-fused boroles, [TipPBB1]4 and TipPBB2 were prepared. [TipPBB1]4 is a white solid adopting a unique coordination mode, which forming a tetramer with a cavity in both the solid state and solution (1H DOSY). The boron center of TipPBB2 is 4-coordinate in the solid state, evidenced by a solid-state 11B{1H} RSHE/MAS NMR study, but the system dissociates in solution, leading to 3-coordinate borole species.
[TipPBB1]4 exhibits two reduction processes which are attributed to the phenylpyridyl cores. TipPBB2 also exhibits two reduction processes with the first half-reduction potential of E1/2red = –1.94 V. The electron accepting ability of TipPBB2 is largely enhanced and comparable to that of FMesBf. This enhanced electron accepting ability is attributed to the electron withdrawing property of the pyridyl group.
TipPBB2 exhibits concentration- and temperature-dependent dual fluorescence in solution. With the temperature is lowered, the emission intensity decreases (Figure 6.4, left). We suggested that the dual fluorescence is caused by an equilibrium between 3-coordinate TipPBB2 and a weak intermolecular adduct of TipPBB2 via a B–N bond. This hypothesis was further supported by lifetime measurements at different concentrations, low temperature excitation spectra low temperature 1H NMR spectra and lifetime measurements upon addition of DMAP to a solution of TipPBB2 to simulate the 4-coordiante TipPBB2 species. Interestingly, the ratio of the relative percentages of the two lifetimes shows a linear relationship with temperature; thus, TipPBB2 could serve as a fluorescent thermometer.
Furthermore, theoretical studies were carried out on TipPBB2, and two models, ((BMe3)TipPBB1(NMe3) and (BMe3)TipPBB2(NMe3)), which utilize a BMe3 group as the Lewis acid coordinated to pyridine and an NMe3 group as the Lewis base coordinated to the boron center of the borole, were used to simulate the [TipPBB1]4 and intermolecular 4-coordinate TipPBB2, respectively. Theoretical studies indicate that the HOMO of TipPBB2 is located at the Tip group, which is in contrast to its borafluorene derivatives for which the HOMOs are located on the borafluorene cores.
Chapter 4
Two derivatives of phenylpyridyl-fused boroles were prepared via functionalization of the pyridyl groups in two different directions, namely an electron-rich dihydropyridine moiety (compound 10) and an electron-deficient N-methylpyridinium cation (compound 11). Both compounds were fully characterized. The 11B NMR signal of compound 10 was observed at 58.8 ppm in CDCl3, which suggests strong conjugation between the boron atom and dihydropyridine moiety. Compound 11 shows a reversible coordination to THF which was confirmed by NMR studies. Compared to other 2,4,6-triisopropylphenyl protected 9-borafluorenes which only coordinate to CH3CN or DMF, the coordination of the weaker and bulkier THF to compound 11 indicates an extremely electron-deficient boron center in compound 11.
The electron-rich property of the dihydropyridine moiety of compound 10 was confirmed by its oxidation potential (Epc = +0.37 V). Due to the strong conjugation of the dihydropyridine moiety with the boron atom, the reduction potential of compound 10 shifts cathodically and is more negative than –2.5 V. Compound 11 exhibits three reduction processes with the first reversible reduction potential at Ered1/2 = –1.23 V, which is significantly anodically shifted compared to that of its precursor (TipPBB2) or its framework 1-methyl-2-phenylpyridin-1-ium triflate (12). This significantly anodically shifted reduction potential confirms an extremely electron-deficient property of compound 11.
Photophysical studies indicate that the lowest energy transition of compound 10 is more likely a locally-excited (LE) transition and compound 11 exhibits a polarized ground state.
Furthermore, we performed theoretical studies for both compounds. The electron cloud distribution of the HOMO of compound 10 supports the strong conjugation between the boron atom and the dihydropyridine moiety in the ground state. An extremely low LUMO energy was determined by theoretical studies which confirmed the extremely electron-deficient property of compound 11.

Inspired by the enhancement of electron accepting ability with increasing numbers of electron withdrawing groups at boron, we tried to study the properties of a bis(pyridyl)arylboranes. In our attempt to synthesize a bis(pyridyl)arylborane, we obtained a bis(2-pyridyl)methoxyborate Li+ complex which is as a dimer both in solution and the solid state.
In the solid state, compound [16]2 is a dimer containing two bis(2-pyridyl)methoxyborate which are linked by two lithium cations. Each lithium cation coordinates to one methoxy group and two pyridyl groups, one from each of the two bis(2-pyridyl)methoxyborate anions. The parameters of [16]2 were compared with other bis(2-pyridyl)methoxyborate stabilized Pt(IV) complex, bis(2-pyridyl)hydroxylborate stabilized Ru(II) complex and the dimer of EtAl(OMe)(2-pyridyl)2Li.
To confirm the coordination mode in solution, 1H DOSY spectroscopy was carried out in CD2Cl2. The van der Waals radius obtained by 1H DOSY nicely matches with the result from the solid state and thus proves the dimer of 16 is persistent in solution.
Finally, different Lewis acids (e.g., TMSCl, BF3•Et2O, AlCl3, HCl) were used to attempt to detach the methoxy group of [16]2. However, we observed either decomposition or selective cleavage of the Tip group, or no reaction at all, rather than cleavage of the methoxy group from boron.
