One’s own name constitutes a unique part of conscious awareness – but does this also hold true for unconscious processing? The present study shows that the own name has the power to bias a person’s actions unconsciously even in conditions that render any other name ineffective. Participants judged whether a letter string on the screen was a name or a non-word while this target stimulus was preceded by a masked prime stimulus. Crucially, the participant’s own name was among these prime stimuli and facilitated reactions to following name targets whereas the name of another, yoked participant did not. Signal detection results confirmed that participants were not aware of any of the prime stimuli, including their own name. These results extend traditional findings on ‘‘breakthrough’’ phenomena of personally relevant stimuli to the domain of unconscious processing. Thus, the brain seems to possess adroit mechanisms to identify and process such stimuli even in the absence of conscious awareness.
In dieser Dissertation wurde die Exziton- und Ladungsträgerdynamik in halbleitenden und metallischen einwandigen Kohlenstoffnanoröhren (SWNTs) mittels zeitkorreliertem Einzelphotonenzählen (TCSPC) und transienter Absorptionsspektroskopie untersucht. Die Experimente wurden an Tensid- oder DNA-stabilisierten SWNT-Proben in Suspension durchgeführt, in denen durch Dichtegradientenultrazentrifugation (DGU) halbleitende (6,5)-Röhren oder metallische (9,9)-Röhren angereichert wurden. Für die Herstellung der metallischen SWNT-Proben wurde das DGU-Verfahren optimiert. Metallische SWNT-Proben wiesen eine Verunreinigung von etwa 3% halbleitenden SWNTs auf. Von den angereicherten metallischen SWNTs war die (9,9)-Röhre mit einem relativen Anteil von 40% die vorherrschende Chiralität. Für transiente Absorptionsmessungen wurden die metallischen SWNT-Proben zudem durch Filtration aufkonzentriert. Halbleitende (6,5)-Proben wurden mit einem standardmäßig verwendeten Rezept hergestellt. Mit TCSPC-Messungen an (6,5)-Proben wurde erstmals gezeigt, dass halbleitende SWNTs neben der kurzlebigen Fluoreszenz des S1-Exzitons, die auf der ps-Zeitskala abläuft, auch eine langlebig Fluoreszenzkomponente aufweisen. Diese klingt mit t^−1 ab und stammt ebenfalls aus dem S1-Exzitonzustand. Das relative Gewicht der langlebigen Komponente an der Quantenausbeute beträgt (7 ± 2)%. Bei der langlebige Fluoreszenzkomponente handelt es sich um verzögerte Fluoreszenz. Diese entsteht durch die Wiederbesetzung des S1-Zustands aus einem tiefergelegenen Triplettzustand. Der vorherrschende Zerfall des Tripletts skaliert mit t^-0,5 und ist auf das nicht-Fick’sche Diffusionsverhalten der Tripletts zurückzuführen, die an Störstellen gefangen werden und abreagieren. Wird vor dem Übergang in den Grundzustand ein weiteres Triplett eingefangen, so kommt es zu einer Triplett-Triplett-Annihilation, die eine Wiederbesetzung des S1-Zustandes bewirkt. Für die transienten Absorptionsexperimente wurde ein Messaufbau verwirklicht, der Anregung und Abfrage im VIS und NIR Spektralbereich mit einer Zeitauflösung von bis zu 50 fs ermöglicht. Die Detektion des Abfragelichts erfolgt spektral aufgelöst mit einer CCD-Kamera. Der Aufbau ermöglicht Nachweisempfindlichkeiten von bis zu 0,2 mOD bei einer Integrationszeit von einer Sekunde. Durch unterschiedliche Modulation von Anregungs- und Abfragestrahl ist eine Detektion auf der Differenzfrequenz der Modulationen möglich, wodurch Einflüsse des Anregungslichts im Abfragespektrum effizient unterdrückt werden. In transienten Absorptionsexperimenten wurde die Exziton- und Ladungsträgerdynamik der (9,9)-Röhre untersucht. Die transienten Absorptionsdaten wurden mit einer globalen Fitroutine angepasst, der ein Vierniveausystem zugrunde lag. Aus dem globalen Fit sind die Photoanregungsspektren (PAS) - die Beiträge der drei angeregten Niveaus zu den transienten Absorptionsspektren - sowie die Zerfallszeiten zugänglich. Die PAS sind durch die Exzitonresonanz gekennzeichnet. Breite PB-Banden aufgrund der Besetzungsänderung der linearen E00-Bänder sind im Gegensatz zu transienten Absorptionsmessungen an Graphen oder Graphit nicht erkennbar. Die PAS des schnellen und mittleren Zerfalls sind ähnlich und weisen eine starkes PB-Signal bei der Energie des M1-Exzitons der (9,9)-Röhre auf, das von PA-Banden bei höheren undtieferen Energien begleitet wird. Der langsame Zerfall ist hingegen durch eine blauverschobene PB-Bande gekennzeichnet, die nur auf der niederenergetischen Seite mit einem PA-Signal einhergeht. Die Zerfallszeiten nehmen mit steigender Anregungsleistung zu und liegen im Bereich von 30 fs bis 120 fs, 500 fs bis 1000 fs und 40 ps. Die schnelle Zerfallskomponente wird mit der Dissoziation der Exzitonen sowie der Thermalisierung der freien Ladungsträgen in den linearen Leitungsbändern zu einer heißen Ladungsträgerverteilung assoziiert. Die mittlere Zerfallskomponente beschreibt die Abkühlung und Rekombination der freien Elektronen und Löcher. Entscheidender Mechanismus ist hierbei die Streuung an hochenergetischen optischen Phononmoden. Die langsame Zerfallskomponente kann durch langlebige, wahrscheinlich an Störstellen gefangene Ladungsträger erklärt werden, deren elektrische Felder durch den Stark-Effekt das ableitungsähnliche transiente Absorptionsspektrum erzeugen. Mittels transienter Absorptionsmessungen an (6,5)-Röhren wurde aus dem anregungsleistungsabhängigen maximalen PB-Signal des S1-Exzitons die Größe des S1-Exzitons zu (7,2 ± 2,5) nm bestimmt. Aus dem Vergleich der leistungsabhängigen maximalen PB-Signale bei Anregung in das S1- und das S2-Exziton ergibt sich, dass die Konversionseffizienz aus dem S2- in den S1-Zustand 1 ± 0,1 beträgt und innerhalb der experimentellen Zeitauflösung von 60 fs vollständig abläuft. Die Exzitongröße in metallischen (9,9)-Röhren wurde bei Exzitonlebensdauern von 15 fs bis 30 fs zu etwa 7 nm bis 12 nm abgeschätzt.
Das grampositive Bakterium Staphylococcus epidermidis ist ein wesentlicher Bestandteil der kommensalen Flora der Haut und der Schleimhäute des Menschen. Jedoch stellen diese Bakterien eine häufige Ursache nosokomialer Katheter-assozierter Infektionen bei immunsupprimierten Patienten dar. Dies liegt zum einen an der Fähigkeit von S. epidermidis, Biofilm zu bilden. Diese physikalische Barriere schützt die Bakterien vor dem Immunsystem sowie vor Antibiotika. Dabei zählen sie zu den häufigsten Erregern von Infektionen an implantierten Fremdkörpern mit Plastikoberflächen, wie z. B. Venenkathetern, künstlichen Herzklappen oder Gefäßprothesen. Zum anderen stellt die Antibiotikaresistenzentwicklung unter S. epidermidis ein zunehmendes Problem dar. Vor allem die late-onset Sepsis, die durch S. epidermidis als Erreger verursacht werden kann, stellt für Frühgeborene eine Gefahr dar. Ziel der vorliegenden Arbeit war, für S. epidermidis als häufigsten und klinisch bedeutsamen KoNS zu eruieren, ob die zunehmende Dauer des stationären Krankenhausaufenthaltes von sehr kleinen Frühgeborenen mit einer höheren Rate an ica-Präsenz, Biofilmbildung und Antibiotikaresistenz assoziiert ist, sowie die Verbreitungswege und das Reservoir für diese S. epidermidis-Stämme zu identifizieren. Hierzu wurden sequenzielle Isolate von S. epidermidis bei Müttern, Kindern und vom Krankenhauspersonal gewonnen und mittels MLST (Multilocus-Sequence-Typing) klonal typisiert. Sie wurden auf Antibiotikaresistenzen, Biofilmbildung und Präsenz des icaA-Gens, das eine Rolle bei der Biofilmbildung spielt, sowie des mecA-Gens untersucht und mit Isolaten, die aus Blutkulturen oder Venenkatheter des Kindes isoliert wurden, verglichen. Es fiel auf, dass die Isolate der sehr kleinen Frühgeborenen deutlich mehr Virulenzfaktoren, wie z.B. Biofilmbildung, hohe Antibiotikaresistenzraten sowie die Präsenz des mecA- und des icaA- Gens, als die maternalen Stämme besaßen. Im Vergleich mit den Ergebnissen der untersuchten Personalstämme liegt der Verdacht nahe, dass oftmals auch das Personal als Transmitter, vor allem von Klonen mit mehreren Virulenzfaktoren, dient. Das Krankenhaus-Milieu scheint dabei ein ideales Reservoir für die Ausbreitung solcher gefährlichen S. epidermidis-Stämme zu sein.
In der vorliegenden Arbeit wurde die Eignung einer im 3D-Pulverdruckverfahren fabrizierten Trägerstruktur auf Calciumphosphat-Basis (Bruschit) als Zellkultur-Scaffold untersucht. Dazu wurden die Konstrukte in vitro mit osteoblastären Zellen besiedelt und deren Proliferations- und Differenzierungsverhalten über eine Kultivierungsdauer von 12 Tagen analysiert. Als Parameter dienten hierbei die Zellviabilität, die Aktivität des osteoblastären Enzyms Alkalische Phosphatase sowie die Mediumkonzentration von Osteocalcin. Des Weiteren wurde der pH-Wert des Kulturmediums sowie die Konzentrationen der freien Elektrolyte Calcium und Phosphat untersucht. Die Ergebnisse belegen eine gute Zytokompatibilität des Trägermaterials. Diese äußerte sich in einer progredienten Proliferation phänotypisch osteoblastärer Zellen (gemäß Rasterelektronenmikroskopie). Die Zellen exprimierten das ostoblastentypische Enzym Alkalische Phosphatase, welches als früher Differenzierungsmarker gilt. Die Analyse der Osteocalcinproduktion führte aufgrund methodischer Probleme nicht zu verwertbaren Ergebnissen. Die Untersuchung des verbrauchten Zellkulturmediums ergab keine unphysiologischen Schwankungen des pH-Wertes. Jedoch konnten signifikante Veränderungen der Konzentration an freien Calcium und Phosphat-Ionen im Medium festgestellt werden. Diese sind auf die Löslichkeit des Trägermaterials im physiologischen Milieu zurückzuführen. Zusammenfassend konnte mittels vorliegender in vitro Versuche eine geeignete Zytokompatibilität des untersuchten Materials herausgearbeitet werden. Für mögliche klinische Anwendungen zum Knochenersatz sind weitergehende Untersuchungen, insbesondere osteokonduktiver Eigenschaften im orthotopen Implantatlager im Rahmen von in vivo Untersuchungen, erforderlich.
Die Diborane(4) Bis(catecholato)diboran und Bis(pinakolato)diboran können durch homogene und heterogene Katalysatoren durch eine Dehydrokupplungsreaktion ausgehend von Catecholboran und Pinakolboran dargestellt werden. Der effizienteste Katalysator für diese Reaktion ist Platin auf Aluminiumoxid, wobei Umsatzzahlen von maximal 11600 und Umsatzfrequenzen von 444 1/h erreicht werden.
In der vorliegenden Arbeit sollte ein Interview auf die Gütekriterien Objektivität, Validität und Reliabilität überprüft werden. Es sollte untersucht werden, ob das Interview geeignet ist, aktuell vorhandene Symptome der ADHS und der ODD dimensional zu erfassen. Dabei wurden 61 Patienten der Klinik und Poliklinik für Kinder- und Jugendpsychiatrie und Psychotherapie der Universität Würzburg mit ihren Müttern befragt. Die Objektivität wurde überprüft, indem das Interview auf Video aufgezeichnet und nochmals von einem zweiten Beurteiler ausgewertet wurde. Die Summenwerte der beiden Interviewer wurden miteinander korreliert. Die Korrelation war signifikant und ergab einen Wert von rk= ,98. Die Objektivität im Sinne der Beurteilerübereinstimmung kann somit als hoch angesehen werden. Es konnte gezeigt werden, dass die zusammenfassende Beurteilung des Interviewers höher mit der Einschätzung aufgrund der Angaben der Mütter korreliert als mit der Einschätzung aufgrund der Angaben der Kinder. Die Korrelation zwischen der zusammenfassenden Beurteilung und der Einschätzung aufgrund der Angaben der Müttern ergab einen Wert von r= ,98, die Korrelation zwischen der zusammenfassenden Beurteilung und der Einschätzung aufgrund der Angaben der Kindern einen Wert von r= ,57. Die zusammenfassende Beurteilung des Interviewers gründet demnach im Wesentlichen auf den Angaben der Mütter. Die Konstruktvalidität wurde ermittelt, indem das Interview mit anderen diagnostischen Verfahren verglichen wurde. Die Korrelation des Interviews mit ADHS-nahen Konstrukten war signifikant und ergab Werte zwischen rtc= ,48 und rtc= ,70. Die diskriminante Validität wurde durch Korrelation mit ADHS-fernen Konstrukten ermittelt. Der Korrelationskoeffizient betrug rtc= ,27. Die Validität liegt somit im mittleren bis oberen Bereich. Ebenfalls wurde belegt, dass Kinder mit zusätzlicher Störung des Sozialverhaltens einen höheren Gesamtscore im Interview erreichen. Das Interview diskriminiert demnach erwartungsgemäß zwischen ADHS-Patienten mit zusätzlicher Störung des Sozialverhaltens und ADHS-Patienten ohne zusätzliche Störung. Die Überprüfung der Gütekriterien erzielte gute Ergebnisse für Objektivität und Validität. Demnach werden Symptome der ADHS und der oppositionellen Störung des Sozialverhaltens mit dem untersuchten Verfahren in hinreichender Güte erfasst.
ΙΟΥΔΙΘ und IUDITH: Überlegungen zum Verhätlnis der Judit-Erzählung in der LXX und der Vulgata

“Das Buch Judit ist in zwei wesentlich voneinander unterschiedenen Textformen überliefert: einerseits in der griechischen Fassung und den sich daran anschließenden alten Übersetzungen (vetus latina, syr., aeth., sah., arm.), zum anderen in der Fassung der lateinischen Vulgata und, mit dieser durchweg parallel laufend, in mittelalterlichen hebräischen Hss.” – so fasst Helmut Engel die Textsituation des Buches Judit prägnant zusammen. Damit liegen heute zwei unterschiedliche Textformen der Juditerzählung vor: Die griechische Juditerzählung in der Septuaginta und die lateinische Iuditherzählung in der Vulgata. Beide Textfassungen unterscheiden sich deutlich voneinander: Die Vulgata-Fassung ist gegenüber der LXX-Fassung um 20 Prozent kürzer; weil die Vulgata-Fassung aber auch umfangreiche Erweiterungen aufweist, teilen LXX und Vulgata nur ungefähr die Hälfte des Textbestandes. Lange ist man davon ausgegangen, dass hinter der Septuaginta- und der Vulgata-Fassung der Juditerzählung ein hebräischer “Ur”-Text gestanden habe. Diese Annahme, die die Forschung seit dem 19. Jh. prägt, gründet sich im Wesentlichen auf zwei Argumenten:
Erstens sei der griechische Text der Septuaginta so durchgehend von einem hebräischen Sprachduktus geprägt, dass man hier von einem hebräischen Originaltext, der recht wörtlich ins Griechische übertragen worden sei, ausgehen müsse.
Zweitens spreche Hieronymus (ca. 347–420) in seiner praefatio zu seiner lateinischen Übertragung von einem ihm vorliegenden “chaldäischen” Text, der ihm als Vorlage gedient habe.
Beide Argumente werden im Folgenden diskutiert: Im ersten Teil wird auf die in den letzten Jahren neu aufgerollte Frage nach der Originalsprache der LXX-Fassung der Juditerzählung eingegangen; der zweite Teil ist der Analyse der praefatio des Hieronymus gewidmet.
Das Ziel dieser Studie war der Vergleich zweier intraoperativer Applikationsmethoden von Mitomycin C (MMC) im Rahmen der Trabekulektomie. Zu diesem Zweck wurde eine retrospektive Analyse vorgenommen. 100 Patienten hatten intraoperativ eine MMC-Applikation mittels Schwämmchen und 100 Patienten mittels Streifen erhalten. Vor dem Beginn der Studie wurden Labormessungen durchgeführt, um die standartisierte Applikationsdosis von MMC zu bestimmen, die sich jeweils in einem Schwämmchen beziehungsweise in 4 Streifen befand. Die Laborergebnissen zeigten eine wirkliche MMC-Dosis von 17,59±3,15µg für die Schwämmchen und von 12,38 ±1,75µg für die Streifen. Überprüft wurde sowohl die absolute (= complete success, ohne lokale Medikation) als auch die relative Erfolgsrate (= qualified success, mit lokaler Medikation). Beide Methoden zeigten eine zufriedenstellende IOD-senkende Wirkung. Für die Erfolgskriterien ≤21+20%, <16, <12 mmHg gab es keinen statistisch signifikanten Unterschied zwischen den zwei Gruppen weder für die absolute noch für die relative Erfolgsrate 1 Jahr nach der Operation. Für das Erfolgskriterium <18 mmHg gab es einen statistisch signifikanten Unterschied bezüglich der relativen Erfolgsrate zugunsten der Streifen-Gruppe. Zwei Erklärungen dafür sind möglich: Zum einen könnte dies mit der höheren Anzahl von Interventionen (Faden-LKs) in der frühen postoperativen Phase zu tun haben. Zum anderen könnte sich die Behandlung eines größeren Areals mit MMC günstiger auf die Vernarbungsreaktion auswirken und damit auch auf die Erfolgsrate. Die Streifen-Gruppe hatte zudem – trotz weniger MMC – eine statistisch signifikant günstigere Sickerkissen-Morphologie 12 Monate nach der Operation. Auch dies ließe sich durch die größere Applikationsfläche von MMC bei den Streifen erklären. Die Schwämmchen-Gruppe hatte statistisch signifikant mehr Hornhaut-Erosiones in der frühen postoperativen Phase. Auf der anderen Seite hatte die Streifen-Gruppe zu dem Zeitpunkt eine bessere relative Erfolgsrate für die Erfolgskriterien <18 und <12 mmHg. Eine mögliche Erklärung wäre, dass die 8x8-mm-Schwämmchen mehr MMC aufnehmen und näher zur Hornhaut appliziert werden. Im Gegensatz zu den Streifen, die über eine breitere Applikationsfläche in der Peripherie weniger MMC abgeben. Möglich wäre aber auch, dass die Erosiones durch die in der Schwämmchen-Gruppe statistisch signifikant vermehrt vorhandenen Voroperationen erklärbar sind. Diese präoperativen Eingriffe können zu Augentrockenheit führen, welche eine Epitheliopathie der Hornhaut begünstigt. Im Bezug auf die Sickerkissen-Needlings und 5-FU-Injektionen hatten die Schwämmchen-Patienten statistisch signifikant mehr Interventionen als die Streifen-Patienten. Dass die Schwämmchen-Gruppe einen statistisch signifikanten höheren Bedarf an 5-FU-Injektionen zeigte, könnte durch die schlechtere Sickerkissen-Morphologie dieser Patienten erklärt werden. Abschließend sei erwähnt, dass die Schwämmchen-Gruppe statistisch signifikant mehr drucksenkende Augentropfen ein Jahr nach der Operation benötigten, damit der erwünschte IOD postoperativ erreicht werden könnte. Zusammenfassend zeigt die Streifen-Gruppe eine tendenziell etwas günstigere Erfolgsrate als die Schwämmchen-Gruppe und benötigte dafür eine geringere Anzahl von postoperativen Interventionen und weniger MMC. Diese Befunde sprechen für die These, dass die Streifen das MMC effektiver applizieren, indem sie eine größere Fläche behandeln. Aufgrund dieser positiven Tendenz hat die Würzburger Klinik den Einsatz der Schwämmchenapplikation eingestellt.
